Epidermal growth factor (EGF) infused over 3-14 days into fetal sheep of 110-125 days gestation resulted in a number of morphological and endocrine changes. Striking hypertrophy of the skin, wool follicles and their accessory structures was seen, together with a reduction in the ratio of secondary to primary follicles and degenerative changes in wool fibres associated with shedding of fibres. Adrenal, thyroid, liver and kidney weights were increased while thymus weight was decreased. The increase in adrenal size resulted from cortical hypertrophy and was associated with increased cortisol secretion. Thyroid hypertrophy was accompanied by an increase in colloid stores, decreased plasma thyroxine and reverse triiodothyronine (T3) concentrations, unchanged plasma T3 and thyroid-binding globulin and raised thyrotropin (TSH) levels. Thyrotropin receptor affinity and content per gram of tissue were unchanged. Fetal and maternal plasma prolactin and growth hormone levels, and fetal plasma placental lactogen levels, were unchanged, although there was a significant rise in maternal plasma placental lactogen concentrations with high doses of EGF. Other maturational parameters such as switching from fetal to adult haemoglobin and liver glycogen content were unaffected.

SUMMARYThe wool-severing properties of 2·8% aqueous solution of calcium thioglycollate were increased by 76% by dissolving the depilatory agent in a KCl-NaOH buffer of pH 12·5. The optimum Ca-thioglycollate concentration for defleecing was 3·6%. One ml of this solution defleeced an area of 12·1 cma.A comparison of five buffers and water as solvents for 3·6% Ca-thioglycollate indicated that the area of skin defleeced was directly related to solution pH and the stubble remaining appeared to be inversely related to solution pH. A KCl-NaOH buffer of pH 12·0 provided the optimum relationship between area defleeced (10·2 cm2/ml applied) and stubble (ca. 2 mm wool) remaining.Single site applications of 0·1–1·0 ml of defleecing agent at skin level were compared and it was shown that the application of 0·1 ml aliquots would require 320 ml solution to defleece an average-sized sheep.Preliminary evaluation of 6% formaldehyde buffered to pH 6 and a carbowax-citric acid solution of pH 2 showed that protection of fibres against dissolution by Cathioglycollate solutions can be sustained for 21 days. Pre-treatment with these compounds could therefore produce a protected band of wool immediately above the proposed site of wool dissolution.The practical implications of these findings are discussed with regard to a wool harvesting programme.

SUMMARY A radioimmunoassay technique for measuring the thyrotrophin (TSH) concentration of sheep and cattle plasma is described. The sensitivity of the assay allowed the measurement of 1–50 ng TSH/ml unextracted plasma. Cross-reaction with ovine luteinizing hormone, prolactin and growth hormone was very low. The average recovery of added TSH was 103 ± 4·1 (s.e.m.)% and the between-assay coefficient of variation was 13·8%. The normal plasma TSH levels of sheep and cattle were approximately 2·5 ng/ml (5 mu. bovine TSH/100 ml). Foetal sheep had plasma TSH concentrations of approximately 3·2 ng/ml during the last 20 days of gestation. Levels of TSH in the circulation decreased abruptly after hypophysectomy of the foetal lamb and a decline in the plasma thyroxine (T4) concentrations was also apparent within 24 h of the operation. However, thyroidectomy of adult and foetal sheep did not increase plasma TSH concentrations until almost all the T4 had been cleared from the circulation. The injection of T4 into thyroidectomized sheep rapidly reduced plasma TSH concentrations to normal values. However, the continued injection of T4 did not further reduce TSH concentration. The injection of T4 or triiodothyronine into normal sheep was also without effect on plasma TSH concentrations.

Urea supplementation of lambing Merino ewes increased milk yield and lamb growth and survival rates in both pen and paddock studies. These productivity gains occurred in a semi-arid environment characterized by poor reproductive efficiency in Merinos. In pens, urea supplenlents fed to lactating ewes at the rate of c. 12 g/day stimulated intake of poor quality pasture hay (0.9 %nitrogen) by c. 30% and increased total nitrogen intake by c. 100%. Milk yield was improved by c. 50% and lamb growth rate by c. 140%. Sorghum grain supplements stimulated milk yield and lamb growth to a lesser extent than urea. The correlations found in pen studies between nitrogen intake and milk yield of ewes (r � 0.85; P < 0.05), and between ewe milk yield and lamb growth (r % 0.80; P < 0.01), prompted further investigations under paddock conditions. Ewes grazing dry pastures and supplemented with urea (8-10 glday) via drinking water yielded more milk (11-32%) than unsupplemented counterparts. Similarly, there were improvements in lamb growth (80-100%) and survival (c. 10%) in the supplemented groups.
