An eosauropterygian skeleton found in the Middle Triassic (upper Anisian) Gutenstein Formation of the Fatric Unit (Demänovská dolina Valley, Low Tatra Mountains, Slovakia) represents the earliest known occurrence of marine tetrapods in the Western Carpathians. The specimen represents a partly articulated portion of the postcranial skeleton (nine dorsal vertebrae, coracoid, ribs, gastral ribs, pelvic girdle, femur and one zeugopodial element). It is assigned to the Pachypleurosauria, more precisely to theSerpianosaurus–Neusticosaurusclade based on the following combination of features: (1) small body size; (2) morphology of vertebrae, ribs and femur; (3) tripartite gastral ribs; and (4) microanatomy of the femur as revealed by μCT. Members of this clade were described from the epicontinental Germanic Basin and the Alpine Triassic (now southern Germany, Switzerland, Italy), and possibly from Spain. This finding shows that pachypleurosaur reptiles attained a broader geographical distribution during the Middle Triassic, with their geographical range reaching to the Central Western Carpathians. Pachypleurosaurs are often found in sediments formed in shallow, hypersaline carbonate-platform environments. The specimen found here occurs in a succession with vermicular limestones in a shallow subtidal zone and stromatolitic limestones in a peritidal zone, indicating that pachypleurosaurs inhabited hypersaline, restricted carbonate ramps in the Western Carpathians.

Mycobacterium goodii, a rapidly growing nontuberculous mycobacterium, is an emerging pathogen in nosocomial infections. Its inherent resistance patterns make it a challenging organism to treat, and delays in identification can lead to poor outcomes. We present a case of cardiac device pocket infection with M. goodii, complicated by both antibiotic resistance and drug reactions that highlight the challenges faced by clinicians trying to eradicate these infections. We also present a brief review of the English literature surrounding this disease, including a table of all reported cases of M. goodii infections and their outcomes to act as guide for clinicians formulating treatment plans for these infections. A clear understanding of diagnostic methods and treatment caveats is essential to curing infections caused by these organisms.

Optical calorimetry is a powerful technique for the characterization of porous materials within only a few minutes (e.g. specific surface area, adsorption capacity). In the current work, optical calorimetry is presented to be a versatile tool for the pore size characterization of activated carbons. Therefore, measurements were performed with six different test gases (N2O, C2H6, C3H8, n-C4H10, i-C4H10, SF6) in the optical calorimeter InfraSORP at ambient conditions. By combining the results of optical calorimetric measurement for each adsorptive, a pore size distribution (PSD) can be estimated in the range of 0.4–6 nm which is in accurate accordance with the PSD of reference CO2 (273 K) and N2 (77 K) physisorption experiments. While common physisorption experiments can easily take a few days, the PSD by using the optical calorimetric screening is obtained within roughly 1 h.

The effect of different substituents on the structural tolerance of benzene-1,3,5-tribenzoate (BTB) based MOFs was studied. For this purpose, three new BTB derivatives ((NH2)3-BTB, tri((tert-butoxycarbonyl)-amino)-BTB; tri((S)-2-acetoxy-2-phenylacetamido)-BTB) were synthesized and utilized in MOF synthesis using synthetic conditions established for eight known MOFs. Three new isoreticular compounds (NH2-DUT-6, NHBoc-DUT-25, mand-DUT-25) could be identified. Furthermore, two compounds with network topologies differing from the parent compound comprising NHBoc-BTB and Cu-paddlewheels as nodes were found: Cu2(H2O)2(H(NHBoc)-BTB)2 (1), a dense structure containing layers with sql topology, and Cu3(H2O)(NHBoc-BTB)2(bipy) (bipy – 4,4′-bipyridine, DUT-109), with crosslinked iab-network. The porosity of DUT-109 was proven by liquid phase adsorption of ethyl cinnamate from n-heptane. In general, a significant influence of the BTB substituent on the framework formation could be demonstrated.

Aims In contrast to the membrane bound adenylyl cyclases, the soluble adenylyl cyclase (sAC) is activated by bicarbonate and divalent ions including calcium. sAC is located in the cytosol, nuclei and mitochondria of several tissues including cardiac muscle. However, its role in cardiac pathology is poorly understood. Here we investigate whether sAC is involved in hypertrophic growth using two different model systems. Methods and results In isolated adult rat cardiomyocytes hypertrophy was induced by 24 h β1-adrenoceptor stimulation using isoprenaline (ISO) and a β2-adrenoceptor antagonist (ICI118,551). To monitor hypertrophy cell size along with RNA/DNA- and protein/DNA ratios as well as the expression level of α-skeletal actin were analyzed. sAC activity was suppressed either by treatment with its specific inhibitor KH7 or by knockdown. Both pharmacological inhibition and knockdown blunted hypertrophic growth and reduced expression levels of α-skeletal actin in ISO/ICI treated rat cardiomyocytes. To analyze the underlying cellular mechanism expression levels of phosphorylated CREB, B-Raf and Erk1/2 were examined by western blot. The results suggest the involvement of B-Raf, but not of Erk or CREB in the pro-hypertrophic action of sAC. In wild type and sAC knockout mice pressure overload was induced by transverse aortic constriction. Hemodynamics, heart weight and the expression level of the atrial natriuretic peptide were analyzed. In accordance, transverse aortic constriction failed to induce hypertrophy in sAC knockout mice. Mechanistic analysis revealed a potential role of Erk1/2 in TAC-induced hypertrophy. Conclusion Soluble adenylyl cyclase might be a new pivotal player in the cardiac hypertrophic response either to long-term β1-adrenoceptor stimulation or to pressure overload.

The gate opening mechanism in the highly flexible MOF Ni2 2,6 ndc 2dabco DUT 8 Ni , DUT Dresden University of Technology with unprecedented unit cell volume change was elucidated in detail using combined single crystal X ray diffraction, in situ XRD and EXAFS techniques. The analysis of the crystal structures of closed pore cp and large pore lp phases reveals a drastic and unique unit cell volume expansion of up to 254 , caused by adsorption of gases, surpassing other gas pressure switchable MOFs significantly. To a certain extent, the structural deformation is specific for the guest molecule triggering the transformation due to subtle differences in adsorption enthalpy, shape, and kinetic diameter of the guest. Combined adsorption and powder diffraction experiments using nitrogen 77 K , carbon dioxide 195 K , and n butane 272.5 K as a probe molecules reveal a one step structural transformation from cp to lp. In contrast, adsorption of ethane 185 K or ethylene 169 K results in a two step transformation with the formation of intermediate phases. In situ EXAFS during nitrogen adsorption was used for the first time to monitor the local coordination geometry of the metal atoms during the structural transformation in flexible MOFs revealing a unique local deformation of the nickel based paddle wheel node

Amyloid-beta (Aβ) oligomers are thought to be causative for the development and progression of Alzheimer's disease (AD). Starting from the Aβ oligomer eliminating d-enantiomeric peptide D3, we developed and applied a two-step procedure based on peptide microarrays to identify D3 derivatives with increased binding affinity and specificity for monomeric Aβ(1-42) to further enhance the Aβ oligomer elimination efficacy. Out of more than 1000 D3 derivatives, we selected seven novel d-peptides, named ANK1 to ANK7, and characterized them in more detail in vitro. All ANK peptides bound to monomeric Aβ(1-42), eliminated Aβ(1-42) oligomers, inhibited Aβ(1-42) fibril formation, and reduced Aβ(1-42)-induced cytotoxicity more efficiently than D3. Additionally, ANK6 completely inhibited the prion-like propagation of preformed Aβ(1-42) seeds and showed a nonsignificant tendency for improving memory performance of tg-APPSwDI mice after i.p. application for 4 weeks. This supports the hypothesis that stabilization of Aβ monomers and thereby induced elimination of Aβ oligomers is a suitable therapeutic strategy.

A new instrumentation for in situ X-ray powder diffraction allows to follow structural changes in the crystal structure of flexible coordination polymers during adsorption and desorption of gases. The system is based on closed cycle He-cryostat that ensures the high temperature stability in the range of 5.5–450 K. The automated gas dosing system facilitates the physisorption measurements in the pressure range of 0.01–130 kPa. The system is implemented on KMC-2 and MAGS beamlines at Helmholtz Zentrum Berlin fur Materialien und Energie. Using new instrumentation, the adsorption of CO2 and n-butane at 273 K on a flexible two-dimensional coordination polymer ELM-11 (ELM – Elastic Layer Material) was investigated.

The interconversion of monomers, oligomers, and amyloid fibrils of the amyloid-β peptide (Aβ) has been implicated in the pathogenesis of Alzheimer disease. The determination of the kinetics of the individual association and dissociation reactions is hampered by the fact that forward and reverse reactions to/from different aggregation states occur simultaneously. Here, we report the kinetics of dissociation of Aβ monomers from protofibrils, prefibrillar high molecular weight oligomers previously shown to possess pronounced neurotoxicity. An engineered binding protein sequestering specifically monomeric Aβ was employed to follow protofibril dissociation by tryptophan fluorescence, precluding confounding effects of reverse or competing reactions. Aβ protofibril dissociation into monomers follows exponential decay kinetics with a time constant of ∼2 h at 25 °C and an activation energy of 80 kJ/mol, values typical for high affinity biomolecular interactions. This study demonstrates the high kinetic stability of Aβ protofibrils toward dissociation into monomers and supports the delineation of the Aβ folding and assembly energy landscape.

Carbide-derived carbon (CDC) monoliths (DUT-38) with a distinctive macropore network are physically activated using carbon dioxide as oxidizing agent. This procedure is carried out in a temperature range between 850 and 975 °C with durations ranging from 2 to 6 h. Resulting materials show significantly increased specific surface areas as high as 3100 m 2 /g and total (micro/meso) pore volumes of more than 1.9 cm 3 /g. The methane (214 mg/g at 80 bar/25 °C), hydrogen (55.6 mg/g at 40 bar/−196 °C), and n -butane (860 mg/g at 77 vol.%/25 °C) storage capacities of the activated CDCs are significantly higher as compared to the non-activated reference material. Moreover, carbon dioxide activation is a suitable method for the removal of metal chlorides and chlorine residuals adsorbed in the pores of CDC after high temperature chlorination. The activation does not influence the hydrophobic surface properties of the CDCs as determined by water adsorption experiments. The macropore network and the monolithic shape of the starting materials can be fully preserved during the activation procedure. n -Butane breakthrough studies demonstrate the materials applicability as an efficient hydrophobic filter material by combining excellent materials transport with some of the highest capacity values that have ever been reported for CDCs.

Mixed ligand approach could be successfully applied to design porous three-dimensional coordination polymers, involving 1,3-bis(imidazol-1-yl)benzene (1,3-bib) as ligand. Five novel Co(II) compounds with different dimensionality have been synthesized. Utilizing only 1,3-bib in the solvothermal reaction with Co(II) salts, only molecular dimer Co2(bib)2Cl4 or 1D polymer Co2(1,3-bib)2(NO3)2 could be synthesized. Copolymerization technique using additional linear dicarboxylates 1,4-benzenedicarboxylate (1,4-bdc) or 2,6-naphthalenedicarboxylate (2,6-ndc) facilitates the formation of 2D layered coordination polymers Co(1,3-bib)(1,4-bdc) and Co(1,3-bib)(2,6-ndc). Utilizing 4,4′-biphenyldicarboxylate (4,4′-bpdc) as a co-linker, a 3D metal–organic framework Co3(1,3-bib)(4,4′-bpdc)3 (DUT-58, DUT – Dresden University of Technology), possessing two interpenetrated uninodal 8-connected frameworks, has been obtained. DUT-58 exhibits a specific surface area of 1208 m2/g and adsorbs 27 mg/g hydrogen (maximum excess value at 30 bar and 77 K), 0.18 g/g n-butane (at 293 K) and 72 mg/g methane (maximum excess value at 60 bar and 298 K).

A combination of topological rules and quantum chemical calculations has facilitated the development of a rational metal-organic framework (MOF) synthetic strategy using the tritopic benzene-1,3,5-tribenzoate (btb) linker and a neutral cross-linker 4,4'-bipyridine (bipy). A series of new compounds, namely [M(2)(bipy)](3)(btb)(4) (DUT-23(M), M = Zn, Co, Cu, Ni), [Cu(2)(bisqui)(0.5)](3)(btb)(4) (DUT-24, bisqui = diethyl (R,S)-4,4'-biquinoline-3,3'-dicarboxylate), [Cu(2)(py)(1.5)(H(2)O)(0.5)](3)(btb)(4) (DUT-33, py = pyridine), and [Cu(2)(H(2)O)(2)](3)(btb)(4) (DUT-34), with high specific surface areas and pore volumes (up to 2.03 m(3) g(-1) for DUT-23(Co)) were synthesized. For DUT-23(Co), excess storage capacities were determined for methane (268 mg g(-1) at 100 bar and 298 K), hydrogen (74 mg g(-1) at 40 bar and 77 K), and n-butane (99 mg g(-1) at 293 K). DUT-34 is a non-cross-linked version of DUT-23 (non-interpenetrated pendant to MOF-14) that possesses open metal sites and can therefore be used as a catalyst. The accessibility of the pores in DUT-34 to potential substrate molecules was proven by liquid phase adsorption. By exchanging the N,N donor 4,4'-bipyridine with a substituted racemic biquinoline, DUT-24 was obtained. This opens a route to the synthesis of a chiral compound, which could be interesting for enantioselective separation.

A zinc-based metal-organic framework Zn(2)(adb)(2)(dabco)·4.5 DMF (K) (DUT-30(Zn), DUT = Dresden University of Technology, adb = 9,10-anthracene dibenzoate, dabco =1,4-diazabicyclo[2.2.2]octane, DMF = N,N-dimethylformamide) was synthesized using a solvothermal route. This MOF exhibits six crystallographic guest dependent phases. Two of them were characterized via single crystal X-ray analysis. The as-synthesized phase K crystallizes in the orthorhombic space group Fmmm, with a = 9.6349(9), b = 26.235(3), and c = 28.821(4) Å and consists of two interpenetrated pillar-layer networks with pcu topology. When the substance loses 0.5 DMF molecules per formula unit, a phase transition from the kinetic phase K to a thermodynamic phase T occurs. Zn(2)(adb)(2)(dabco)·4 DMF (T) crystallizes in the tetragonal space group I4/mmm, with a = 19.5316(8) and c = 9.6779(3) Å. During the evacuation the DUT-30(Zn) undergoes again the structural transformation to A. The activated compound A shows the gate pressure effect in the low pressure region of nitrogen physisorption isotherm and has a BET surface area of 960 m(2 )g(-1) and a specific pore volume of 0.43 cm(3) g(-1). Furthermore, DUT-30(Zn) exhibits a hydrogen storage capacity of 1.12 wt % at 1 bar, a CO(2) uptake of 200 cm(3) g(-1) at -78 °C and 0.9 bar, and a n-butane uptake of 3.0 mmol·g(-1) at 20 °C. The N(2) adsorption process was monitored in situ via X-ray powder diffraction using synchrotron radiation. A low temperature induced transformation of phase A to phase V could be observed if the compound was cooled under vacuum to -196 °C. A further crystalline phase N could be identified if the framework was filled with nitrogen at -196 °C. Additionally, the treatment of activated phase A with water leads to the new phase W.

Recently, we have described the metal-organic framework Ni(2)(2,6-ndc)(2)(dabco), denoted as DUT-8(Ni) (1) (DUT = Dresden University of Technology, 2,6-ndc = 2,6-naphthalenedicarboxylate, dabco = 1,4-diazabicyclo[2.2.2]octane). Upon adsorption of molecules such as nitrogen and xenon, this material exhibits a pronounced gate-pressure effect which is accompanied by a large change of the specific volume. Here, we describe the use of high-pressure in situ (129)Xe NMR spectroscopy, i.e., the NMR spectroscopic measurements of xenon adsorption/desorption isotherms and isobars, to characterize this effect. It appears that the pore system of DUT-8(Ni) takes up xenon until a liquid-like state is reached. Deeper insight into the interactions between the host DUT-8(Ni) and the guest atom xenon is gained from ab initio molecular dynamics (MD) simulations. van der Waals interactions are included for the first time in these calculations on a metal-organic framework compound. MD simulations allow the identification of preferred adsorption sites for xenon as well as insight into the breathing effect at a molecular scale. Grand canonical Monte Carlo (GCMC) simulations have been performed in order to simulate adsorption isotherms. Furthermore, the favorable influence of a sample pretreatment using solvent exchange and drying with supercritical CO(2) as well as the influence of repeated pore opening/closure processes, i.e., the "aging behavior" of the compound, can be visualized by (129)Xe NMR spectroscopy.

Cyclotrimerization of bifunctional acetyl compounds is used to obtain highly porous organic frameworks. Syntheses in solution induced by silicon tetrachloride result in highly disperse powders while syntheses in molten 4-toluene sulfonic acid result in polymeric monoliths with a hierarchical pore structure containing micro- and macropores allowing for direct impregnation of textiles with a porous polymer. The materials show specific BET surface areas up to 895 m2 g−1 and large pore volume (1.99 cm3 g−1) combined with a highly hydrophobic character. The amorphous materials are thermally stable below 300 °C in air and show no decomposition effects in aqueous environment. These outstanding properties in combination with the opportunity to generate shapes of any kind desired for an application render the materials as highly promising for application in air filtration systems and individual protection, as well as gas storage and separation.

The n -butane adsorption capacity and heat of adsorption for the metal–organic frameworks Cu 3 (btc) 2 (btc = benzene-1,3,5-tricarboxylate) and MIL-101 was determined at three different temperatures (293, 303, and 313 K) under dynamic conditions at atmospheric pressure with a micro-balance combined with a Tian-Calvet micro-calorimeter. The adsorption rate was studied at the adsorption temperature of 293 K. The n -butane adsorption properties were compared with activated carbon Sorbonorit 3. For low n -butane concentration the microporous Cu 3 (btc) 2 has a higher storage capacity than the mesoporous material MIL-101. For high n -butane concentrations it is vice versa. The saturation capacity at 293 K for Cu 3 (btc) 2 is 3.7 mmol g −1 and for MIL-101 remarkable 11.2 mmol g −1 . The storage capacity and adsorption rate depends on the alkane concentration. For higher n -butane concentration MIL-101 has a higher adsorption rate than Cu 3 (btc) 2 independent on the amount of adsorbed n -butane.

In this study, we characterized unexpected side-products in a commercially synthesized peptide with the sequence RPRTRLHTHRNR. This so-called peptide D3 was selected by mirror phage display against low molecular weight amyloid-β-peptide (Aβ) associated with Alzheimer's disease. Capillary electrophoresis (CE) was the method of choice for structure analysis because the extreme hydrophilicity of the peptide did not allow reversed-phase liquid chromatography (RPLC) and hydrophilic interaction stationary phases (HILIC). CE-MS analysis, applying a strongly acidic background electrolyte and different statically adsorbed capillary coatings, provided fast and efficient analysis and revealed that D3 unexpectedly showed strong ion-pairing with sulfuric acid. Moreover, covalent O-sulfonation at one or two threonine residues was identified as a result of a side reaction during peptide synthesis, and deamidation was found at either the asparagine residue or at the C-terminus. In total, more than 10 different species with different m/z values were observed. Tandem-MS analysis with collision induced dissociation (CID) using a CE-quadrupole-time-of-flight (QTOF) setup predominantly resulted in sulfate losses and did not yield any further characteristic fragment ions at high collision energies. Therefore, direct infusion Fourier transform ion cyclotron resonance (FT-ICR) MS was employed to identify the covalent modification and discriminate O-sulfonation from possible O-phosphorylation by using an accurate mass analysis. Electron transfer dissociation (ETD) was used for the identification of the threonine O-sulfation sites. In this work, it is shown that the combination of CE-MS and FT-ICR-MS with ETD fragmentation was essential for the full characterization of this extremely basic peptide with labile modifications.

The aim of this study is to assess the convenience of exploring longterm synergistic effects when multiple AD therapeutics of different mechanisms, fundamental and symptomatic, are combined. Methods: As a symptomatic drug, we selected donepezil, which have significant clinical benefits by activating undamaged cholinergic systems. From our previous study, as a fundamental drug, we prepared a small molecule KMSB600 that disassembles neurotoxic Ab aggregates back to inert Ab monomers and reverses the behavioral deficits in 2xTG-AD mice (APP/ PS1). During the oral co-administration, we performed Y-maze tasks every week for 4 months and performed fear conditioning tasks after the last trial of Y-maze. Ab plaques were visualized by thioflavin-S staining. Results: Co-administration of donepezil and KMSB600 significantly improved the deficits in the both behavior tests and histology study, compared to individual administration of donepezil or KMSB600 at the same dosages. Conclusions: Here, we report that co-administration of donepezil and KMSB600 significantly enhanced cognitive deficits in aged-AD mice in a complementary manner. Our findings imply that the cure of AD may require cocktail therapy of multiple drugs with different modes of action: cholinergic system improvement and amyloidogenic pathology reduction.

The aggregation of Tau into paired helical filaments is involved in the pathogenesis of several neurodegenerative diseases, including Alzheimer disease. The aggregation reaction is characterized by conformational conversion of the repeat domain, which partially adopts a cross-beta-structure in the resulting amyloid-like fibrils. Here, we report the selection and characterization of an engineered binding protein, beta-wrapin TP4, targeting the Tau repeat domain. TP4 was obtained by phage display using the four-repeat Tau construct K18DeltaK280 as a target. TP4 binds K18DeltaK280 as well as the longest isoform of human Tau, hTau40, with nanomolar affinity. NMR spectroscopy identified two alternative TP4-binding sites in the four-repeat domain, with each including two hexapeptide motifs with high beta-sheet propensity. Both binding sites contain the aggregation-determining PHF6 hexapeptide within repeat 3. In addition, one binding site includes the PHF6* hexapeptide within repeat 2, whereas the other includes the corresponding hexapeptide Tau(337-342) within repeat 4, denoted PHF6**. Comparison of TP4-binding with Tau aggregation reveals that the same regions of Tau are involved in both processes. TP4 inhibits Tau aggregation at substoichiometric concentration, demonstrating that it interferes with aggregation nucleation. This study provides residue-level insight into the interaction of Tau with an aggregation inhibitor and highlights the structural flexibility of Tau.

Background Chagas disease is most often transmitted to humans by Trypanosoma cruzi infected triatomine bugs, and remains a significant cause of morbidity and mortality in Central and South America. Control of Chagas disease has relied mainly on vector eradication. However, development of insect resistance has prompted us to develop a paratransgenic strategy to control vectorial transmission of T. cruzi. Here, the potential role of recombinant endoglucanases as anti-trypanosomal agents for paratransgenic application is examined. The surface of T. cruzi is covered by a thick coat of mucin-like glycoproteins that have been proposed to play a role in the binding of T. cruzi to the membrane surface of the vector gut. We hypothesize that disruption of these glycoconjugates could arrest parasite development in the vector and abort the transmission cycle. In this work, we examine the effects of recombinant Arthrobacter luteus β-1, 3-glucanase expressed via Rhodococcus rhodnii on T. cruzi Sylvio II strain. Methods and results The coding sequence for β-1, 3-glucanase was cloned in-frame to a heterologous promoter/signal sequence from the Mycobacterium kansasii alpha antigen gene resident in an E. coli/R. rhodnii shuttle vector. The resulting construct was confirmed by sequencing, and electroporated into R. rhodnii. Expression products from positive clones were purified from log phase cultures followed by dialysis into physiological buffers. Lysates and media were quantitated by ELISA against rabbit antibody specific to β-1,3-glucanase. Glucanase-positive samples were applied to live T. cruzi parasites in culture and viability accessed by spectrophotometric and fluorescent microscopic measurements. R. rhodnii-expressed β-1,3-glucanase exhibited toxicity against T. cruzi compared to controls when applied at 5 and 10% of the total culture volume. The decrease in cell viability ranged from a maximum of 50% for the media treatments to 80% for the filtered lysates. Conclusions These results suggest that recombinant β-glucanase could be a powerful addition to the arsenal of effector molecules for paratransgenic control of Chagas disease. In future studies, the ability of β-glucanase to function in combination with other effector molecules will be explored. Dual targeting of T. cruzi should not only slow resistance but also permit synergistic or additive lethal effects on T. cruzi.

Metal–organic frameworks (MOFs), hybrid materials built up from metal clusters and organic linkers, have shown a huge potential for a wide range of applications. In recent years, MOFs have set new records in terms of specific surface areas and pore volumes and therefore are highly suitable as storage materials for small and large molecules. The development of new materials is crucial for the improvement of storage devices, but MOFs are also ideal candidates for functionalization. One functionalization strategy is the integration of complex organic linker molecules containing secondary functional groups. However, this approach is synthetically demanding and not general, because functional donor atoms may affect the linker connectivity resulting in unexpected network topologies. A second powerful strategy is postsynthetic modification of the framework. In this case, the range of functions is restricted due to the limited stability of MOFs against aggressive chemicals. A modular and more versatile approach may be the encapsulation of functional guest molecules into the MOF material. However, for these systems leaching is critical. A crucial requirement in all cases is also the accessibility of MOF functionalities for guest molecules. In this context, a large pore size is highly beneficial. However, the development of such mesoporous frameworks is challenging, because expanded frameworks are often more fragile leading to a collapse of the framework during removal of included guests molecules. A very effective concept to achieve robust frameworks lies in the creation of hierarchical pore structures by combination of two different linker molecules. The most prominent examples of such copolymerization approach are UMCM-1/ 2/ 3, DUT-6 and DUT-23 or MOF210. In the case of DUT-23, auxiliary linker was used successfully to avoid interpenetration and to enhance the robustness, which resulted in highly porous structures. To date, this concept was restricted only to the combination of triand ditopic linkers. On the other hand, DUT-10(M) (M= Zn, Cu, Co) compounds based on the tetratopic N,N,N’,N’benzidinetetrabenzoate (benztb) ligand and the paddlewheel secondary building unit (SBU) undergo structural change upon solvent removal. Porous carbon materials are crucial components in catalysis, gas storage, electronics, and biochemistry. A hierarchical pore architecture in these materials is essential to achieve high surface areas combined with advanced mass transport kinetics. Widely used approaches for the generation of microor mesopores are activation and nanocasting. In contrast, macroporous carbon materials are primarily obtained by carbonization of polymeric precursor gels or replication of larger templates. A relatively new class of microand mesoporous carbon material with tunable porosity are carbide-derived carbon materials (CDCs). High-temperature chlorination of carbides leads to selective removal of metalor semi-metal atoms and allows control over the pore size of the resulting CDCs in a subngstrcm range by changing synthesis conditions or the carbide precursor. These materials have been studied for applications in gas storage and as electrode materials in supercapacitors because of their high specific surface areas. Recently, metal etching from pyrolyzed pre-ceramic components (polysilsesquioxanes or polysilazanes) was found to be a useful route towards carbide-derived carbon materials with enhanced porosity and gas-storage properties. A significant step towards ultrahigh specific surface area combined with a hierarchical mesoporous–microporous system was achieved using nanocasting of silica templates (SBA-15 or KIT-6) with polycarbosilane precursors and subsequent chlorine treatment of the resulting ordered mesoporous silicon carbides. These ordered mesoporous CDCs offer specific surface areas as high as 2800 mg 1 and total pore volumes of up to 2 cmg . Their mesostructure can be easily controlled by changing the silica hard template, resulting in excellent performance in protein adsorption, gas storage, and as electrodes for supercapacitors. However, such carbon materials are available only as nonstructured micrometer-sized powders and cannot be shaped into films without the addition of binders or the use of high mechanical stress, leading to structural deformation. Chlorine treatment of mechanically mixed Si/SiC precursors was found to be a useful route towards monolithic CDC with a hierarchical pore system. The presence of a free metal phase in the precursor system provides the opportunity to introduce a secondary macroporosity of 3 mm sized channels with a volume of 0.23 cmg 1 along with the microporous carbide-derived carbon material system. The introduction of large transport pores in polymerbased CDCs might be an alternative way to form materials that combine high surface areas with efficient fluid transport. The current literature describes a variety of routes for the production of highly macroporous ceramics from precursor polymers with controllable cell and window sizes. In particular, direct blowing of polycarbosilanes was found to be a useful approach for the generation of silicon carbide foams that might be suitable materials for the production of hierarchical CDCs. In the following, we describe a novel synthesis route for monolithic carbide-derived carbon materials, including micro-, meso-, and macroporous structures with extremely high specific surface area. They can be obtained by hightemperature chlorination of macroporous polymer-derived silicon carbide (SiC-PolyHIPE). A soft-templating approach starting from a high internal phase emulsion (HIPE) was used with an external oil phase consisting of liquid polycarbosilane SMP-10 and the cross-linker paradivinylbenzene. Using Span-80 as surfactant to stabilize the internal water phase, the application of oxidic or carbon hard templates and the corresponding template removal under harsh conditions is no longer necessary. After cross-linking the polymer chains, the resulting PolyHIPEs were pyrolyzed to silicon carbides at maximum temperatures of 700, 800, and 1000 8C and subsequently converted into CDCs by chlorine treatment at the maximum pyrolysis temperature (Supporting [*] M. In the following a new class of highly porous carbon structures is applied in gas storage. Ordered mesoporous carbide derived carbons (OM-CDC) were synthesized by chlorination of mesostructured silicon carbide ceramics (OM-SiC). Resulting OM-CDC structures were characterized by nitrogen physisorption methods and small angle X-ray scattering demonstrating high specific surface areas and bimodal pore size distributions by varying the synthesis and chlorination conditions. The adsorption properties could be further enhanced by reductive hydrogen treatment. Storage capacities for mobile applications dependant on the synthesis conditions were investigated in high pressure hydrogen and methane adsorption with extraordinary high uptakes compared to micro- and mesoporous reference materials. The synthesis and structural flexibility of the metal–organic frameworks M2(2,6-ndc)2(dabco) (DUT-8(M), M = Ni, Co, Cu, Zn; 2,6-ndc = 2,6-naphthalenedicarboxylate, dabco = 1,4-diazabicyclo[2.2.2]octane) as well as their characterization by gas adsorption, 129Xe NMR and 13C MAS NMR spectroscopy are described. Depending on the integrated metal atom the compounds show reversible (DUT-8(Ni), DUT-8(Co)), non-reversible (DUT-8(Zn)) or no (DUT-8(Cu)) structural transformation upon solvent removal and/or physisorption of several gases. DUT-8(Co) exhibits a similar structural transformation by solvent removal and adsorption behavior as observed for DUT-8(Ni). DUT-8(Zn) undergoes an irreversible structural change caused by solvent removal. The non-flexible copper containing MOF reveals the best performance concerning porosity and gas storage capacities within the DUT-8 series. Xenon adsorption studies combined with 129Xe NMR spectroscopy are used to study the flexibility of the DUT-8 compounds. 129Xe chemical shift and line width strongly depend on the metal atom. Solid-state 13C NMR spectroscopy has been applied in order to further characterize the organic parts of the DUT-8 frameworks. While DUT-8(Ni) exhibits narrow, well-resolved lines in its “as made” state, the signals of DUT-8(Co) are broadened and shifted over an unusually wide chemical shift range (−72 to 717 ppm). No detectable signals are found in DUT-8(Cu) indicating significantly changed internal dynamics compared to DUT-8(Ni) and DUT-8(Co).

A new tool (Infrasorb-12) for the screening of porosity is described, identifying high surface area materials in a very short time with high accuracy. Further, an example for the application of the tool in the discovery of new cobalt-based metal-organic frameworks is given.

The synthesis and structure of a new flexible metal-organic framework Ni(2)(2,6-ndc)(2)(dabco) (DUT-8(Ni), DUT = Dresden University of Technology, 2,6-ndc = 2,6-naphthalenedicarboxylate, dabco = 1,4-diazabicyclo[2.2.2]octane) as well as its characterization by gas adsorption and (129)Xe NMR spectroscopy is described. The compound shows reversible structural transformation without loss of crystallinity upon solvent removal and physisorption of several gases. Xenon adsorption studies combined with (129)Xe NMR spectroscopy turn out to be favorable methods for the detection and characterization of the so-called "gate-pressure" effect in this novel MOF material. The linewidth and chemical shift of the (129)Xe NMR signal are shown to be very sensitive parameters for the detection of this structural transition from a narrow pore system with low porosity to a wide pore state. The transition and threshold temperature is clearly detected.

The synthesis of polyHIPEs with bimodal pore-size distribution and very high specific surface areas up to 1210 m2 g−1 is reported. Monolithic polyHIPE precursors were swollen, and subsequently the polymeric network was crosslinked in the expanded state by internal and external electrophiles under catalysis of a Lewis acid leading to hypercrosslinked polymers of the Davankov type. The materials have a high n-butane uptake (34 wt% at 298 K) and moderate hydrogen storage capacity (2.02 wt% at 19.5 bar and 77.4 K). According to water adsorption isotherms, the inner surface area of the systems is highly hydrophobic shifting the adsorption branch to relative humidities of 0.8. The hierarchical organization of the system with transport macropores and highly microporous walls leads to an enhanced n-butane adsorption.
