The urchin-like nanoparticles of semiconducting oxides have been attracting considerable attention as they can significantly improve reactivity owing to their enlarged surface areas. Herein, we report a simple and scalable synthesis method of ZnO nanomaterials via a microwave-assisted drying process. Commercial ZnO powders (~300 nm) and expanded graphite were used as starting materials to prepare urchin-like ZnO nanoparticles with nanoscale legs (diameter ~20 nm) by triggering the rapid phase transformation, which was induced by a repeated microwave irradiation in an alumina crucible for a few minutes. The possible mechanisms to trigger this morphological and dimensional change are described in detail. The high-yield production and superior reproducibility completed in shortened processing times enable this synthetic route to be widely applicable for the massive production of nanostructured oxide particles.

In this study, reduced graphene oxide (RGO) nanosheets were synthesized and then functionalized with Pt nanoparticles (NPs) for gas sensing studies at room temperature. Pt-functionalized RGOs (Pt-RGOs) were irradiated by a 2 MeV electron beam with different doses of 100 and 500 kGy (Pt-RGO-100 and Pt-RGO-500) to study the role of electron beam irradiation (EBI) on properties and NO2 gas sensing characteristics. Different characterization techniques verified the generation of structural defects after EBI. Additionally, NO2 sensing tests at room temperature revealed that the Pt-RGO-100 sensor had the highest response to NO2 gas as well as the shortest response time. We surmise that the improved sensing signal with respect to NO2 gas for Pt-RGO-100 sensor was related to the role of oxygen functional groups. However, decreases in the response for Pt-RGO-500 sensor were related to the creation of reactive oxygen gases that could be adsorbed on the active defects of the RGO by the interaction of electron beams with oxygen species in the ambient air, leading to a decrease of the effective defect sites and decreases of the sensing response. This work suggests progress towards an improved performance of RGO-based gas sensors that can work at low temperature, by using EBI as a clean tool applied at room temperature with Pt-functionalization.

A new gas sensor working in room temperature, which is compatible with silicon fabrication technology is presented. Porous silicon nanowires (NWs) were synthesized by metal-assisted chemical etching method and then TeO₂ NWs branches were attached to their stem by thermal evaporation of Te powders in the presence of air. Afterwards TeO₂ branched porous Si NWs were functionalized by Pt via sputtering followed by low temperature thermal annealing. Scanning electron microscopy, transmission electron microscopy and energy-dispersive X-ray spectroscopy collectively confirmed successful formation of TeO₂ branched porous Si NWs functionalized by Pt nanoparticles. Their gas sensing properties in the presence of CO, C6H6 and C7H8 were tested at room temperature, for Si wafer, pristine porous Si NWs, pristine TeO₂ branched porous Si NWs, and Pt functionalized TeO₂ branched porous Si NWs sensors. Pt functionalized TeO₂ branched porous Si NWs have higher responses to all tested gases than the other sensors. The origin of high response is discussed in detail. This new room temperature gas sensor can open a new aperture for development of gas sensors with minimum energy consumption which are compatible with silicon fabrication technology.

Branched nanowires (b-NWs) have been used for advanced applications because of their nanoscale dimensions, high surface area, and scalable synthetic methods. Herein, for the first time, we report the fabrication of Au-functionalized ZnO-branched SnO2 NWs for enhanced gas sensing application. As-fabricated NWs were structurally and morphologically investigated by X-ray diffraction, scanning electron microscopy as well as transmission electron microscopy to reveals the crystallinity and morphology of NWs. By comparing the gas sensing properties of Au-functionalized b-NWs, b-NWs, and pristine SnO2 NWs gas sensors, in terms of their sensitivity and cross-sensitivity towards NO2, we demonstrate the superior sensing properties of Au-functionalized b-NWs sensors. In particular the Au-functionalized b-NWs sensor showed a response (Rg/Ra) of 13.07 to 10 ppm NO2 gas which was higher than those of b-NWs, and pristine SnO2 NWs gas sensors. In addition to the special architecture of sensors (i.e. ZnO b-NWs on SnO2 NWs), which produces a large number of hetero- and homojunctions (a branching effect), Au acts as a catalyst to further improve the sensing characteristics of this particular sensor (a functionalization effect). According to the obtained results, we believe that metal oxide gas sensors based on noble metal-functionalized b-NWs, which use a facile synthesis procedure and produce enhanced sensing performance, will play a vital role in the future of the sensor industry.

An unstable energy-unbalanced state such as superheating or supercooling is often unexpectedly observed because a factor of energy depends not only on the temperature but is a product of temperature (T) and entropy (S). Thus, at the same temperature, if the entropy is different, the total energy of the system can be different. In such cases, the temperature-change-rate cannot match the entropy-change-rate, which results in a hysteresis curve for the temperature/entropy relationship. Due to the difference between the temperature- and entropy-change-rates, properties of a material, such as the boiling and freezing points, can be extended from point to area. This study confirmed that depending on the heating rate, tin powders exhibit different melting points. Given the contemporary reinterpretation of many energy-non-equilibrium phenomena that have only been discussed on the basis of temperature, this study is expected to contribute to the actual expansion of scientific/engineering applications.

We present a highly sensitive and selective NO2 sensor based on Bi2O3 branched SnO2 nanowires (NWs). SnO2 NWs were first synthesized by a vapor-liquid-solid method, were coated with an Au layer, and Bi2O3 branches were grown on their stems by the same procedure used for pure Bi powders. The fabricated sensor showed a high response (Rg/Ra) of 56.92 to 2 ppm of NO2 gas at an optimal temperature. Furthermore, its response to other interfering gases such as ethanol, acetone, toluene, and benzene, was less than 1.55, which demonstrated excellent selectivity of the sensor towards NO2 gas. For comparison and to better understand the sensing mechanism, a pristine SnO2 NWs sensor was also tested. The superior sensing properties of the branched NW sensor relative to the pristine sensor were mainly attributed to the high surface area of the sensor resulting from Bi2O3 branching, as well as the formation of homo-and heterojunctions (Bi2O3-SnO2). In addition, several factors including the presence of Au contributed to the excellent selectivity to NO2 gas. Based on the results obtained in this work, we believe that the present sensor with an easy fabrication method, along with its high sensitivity and selectivity towards NO2, can be used for the detection of NO2 gas in real applications.

We present a dual sensitization gas sensor for selective detection of either H2S or C2H5OH gases, based on ZnO/CuO nanoparticle (NP)-decorated MWCNTs. First, Cu-Zn layers of three different thickness (3, 6, 9 nm) were successfully deposited on the surfaces of CNTs by a sputtering process and subsequently converted to their corresponding oxides by thermal annealing. The gas sensing characteristics of metal oxide decorated-MWCNTs were studied in the presence of three gases, namely NO2, H2S, and C2H5OH. The best sensing properties were obtained when the Cu-Zn deposition layer was 6 nm thick. The optimized sensor showed extraordinary responses to both H2S and C2H5OH gases at working temperatures of 100 °C and 200 °C, respectively. Therefore, selectivity was tuned by selection of the working temperature. The sensing mechanisms of the metal oxide-decorated CNTs sensors are discussed in detail. The approach described, namely co-decoration of the surfaces of MWCNTs with different metal oxides, will be of great utility to researchers who wish to fabricate dual sensitive gas sensors.

In this study, we synthesized porous (porosity: ~16%, average pore size: ~60 nm) ZnO nanosheets (thickness: ~80 nm) using a conventional solvothermal method to investigate NO2 gas sensing properties. Porous ZnO nanosheets triggered the detection of NO2 gas with high sensitivity. Responses of 2.93 – 0.5 ppm and 74.68 – 10 ppm NO2 gas at 200 °C were observed in the porous ZnO nanosheet-based gas sensor. In addition, improved sensing properties with high selectivity to NO2 gas, reasonable stability, and high response even in the presence of water vapor molecules were obtained. We found that the enhanced NO2 gas response of the porous ZnO nanosheet-based gas sensor was due to the synergetic effects of the high surface area, ZnO/ZnO homojunctions, and structural defects. We developed a highly sensitive NO2 gas sensor with improved reliability using morphologically engineered ZnO, which was prepared via a simple and scalable chemical-synthesis route.

In this study, a peculiar configuration for a gas sensor consisting of vertically aligned silicon nanowires (VA-Si NWs) synthesized by metal-assisted chemical etching (MACE) is reported. Si NWs were prepared via a facile MACE method and subsequent thermal annealing. Etching was performed by generation of silver nanoparticles (Ag NPs) and subsequent etching in HF/H2O2 aqueous solution; the growth conditions were optimized by changing the process parameters. Highly vertically oriented arrays of Si NWs with a straight-line morphology were obtained, and a top–top electrode configuration was applied. The VA-Si NW gas sensor showed good sensing performance, and the VA-Si NWs exhibited a remarkable response (Rg/Ra = 11.5 ∼ 17.1) to H2 gas (10–50 ppm) at 100 °C which was the optimal working temperature. The formation mechanism and gas sensing mechanism of VA-Si NWs are described. The obtained results can suggest new approaches to making inexpensive, versatile, and portable sensors based on Si NWs having a novel top–top electrode structure that are fully compatible with well-developed Si technologies.

In this study, multi-walled carbon nanotubes (MWCNTs) were decorated by the Cu2O/CuO nanoparticles for gas sensing. For this purpose, Cu layer with different thicknesses of 3, 6, and 9 nm was coated on multi-walled carbon nanotubes using sputtering technique for different times of 1, 2, and 3 min at 25°C, respectively, followed by annealing at 500°C to produce isolated Cu2O/CuO islands. The synthesized products were fully characterized and their expected morphology, chemical composition and phases were confirmed. The gas sensors were fabricated and the optimal sensing temperatures for H2S sensing was found to be relatively low (150 °C). With control of the size of the Cu2O/CuO nanoparticles, very high sensor response ((Rg – Ra)/Ra x 100) of about 1244 % to 1 ppm H2S gas was obtained, with response time and recovery time of 219 and 77 s, respectively. Since sensor response, response time, and recovery time became significantly higher, shorter, and shorter, respectively, by the Cu2O/CuO functionalization, the associated mechanisms were explained in regard to the selective sensitivity to H2S gas. In fact, Cu2O/CuO decorations not only increased the effective surface area for sensing studies, but also they acted as effective catalyst for H2S gas. By considering the energy bands, among the multi-walled carbon nanotubes-comprising interfaces, the multi-walled carbon nanotubes/CuS heterointerfaces were effective in the enhancing the sensing behavior.

The typical methods used to improve the sensing properties of metal oxide nanowire-based gas sensors include decoration of a metal catalyst on the surface of nanowires or generation of surface defects. In this study, we suggest a process for preparing a Pd metal catalyst surrounded in SnO2-x with a large number of structural defects on the surface of SnO2 nanowires for gas sensing applications. Initially, SnO2 nanowires were prepared using a simple vapor-liquid-solid method and Pd-embedded SnO2-x-decoration was achieved by flame chemical vapor deposition. Gas sensing studies demonstrated the promising effects of Pd-embedded SnO2-x-decoration. The underlying sensing mechanism was studied in detail and explained in terms of generation of oxygen defects, catalytic activity of Pd, and formation of heterojunctions in the sensing material. The results of the present study clearly demonstrate the usefulness of this simple and fast approach to significantly enhance the gas sensing properties of metal oxide nanowires.

Microwave (MW) irradiation has obtained extensive importance in the field of synthesis and treatment of nanoparticles, because of its faster, cleaner and cost effectiveness than the other conventional and wet chemical methods. In this study, ZnO/graphene nanocomposites were prepared and subsequently post-treated by MW irradiation. Responses of the MW irradiated ZnO/graphene nanocomposites sensors were tested towards various gases including NO2, ethanol, acetone, toluene and CO and the results were compared with those of pristine ZnO and ZnO/graphene sensors without MW irradiation. It was demonstrated that the MW irradiated sensor had much higher response particularly to NO2 gas along with superior selectivity and shorter response/recovery times in comparison to unirradiated ZnO/graphene and pristine ZnO sensors. The possible underlying mechanism of this behavior is discussed in detail, mainly in terms of the MW-induced surface defects and the generation of finer ZnO nanoparticles. The results obtained demonstrated the beneficial effect of MW irradiation for enhancing the NO2-gas sensing behavior of ZnO/graphene nanocomposites, opening a new door not only to a novel synthesis of semiconductors/graphene nanocomposites, but also to a cost-effective way of improving their sensing capabilities.

In this work, ZnO-decorated multi-walled carbon nanotube (MWCNT) nanocomposites prepared using an in-situ method involving the thermal evaporation of Zn powders in the presence of MWCNTs. The gas sensing characteristics of the MWCNT/ZnO nanocomposites are studied, and results for the material characterizations for the synthesized nanocomposites confirm the formation of well-distributed ZnO nanoparticles onto MWCNTs, creating MWCNT/ZnO nanocomposites. The gas sensing properties of the MWCNT/ZnO nanocomposite gas sensor, such as response, cross-sensitivity, and response-recovery time, are investigated and compared with a bare MWCNT sensor. The decoration of ZnO nanoparticles greatly improves the gas sensing properties of bare MWCNTs. We discussed the possible mechanisms for the enhancement of sensing capabilities. The results suggest that decoration of n-type semiconducting oxide materials, such as ZnO in the form of nanoparticles, is a promising strategy for improvement of gas sensing properties of p-MWCNTs.

We report a novel method to significantly improve the C 6 H 6 -sensing performance of Si nanowires through the combination of TeO 2 branches and Pd sensitization. The morphological investigation revealed that TeO 2 branches were densely formed on the stem nanowires (NWs). The sensor responses of the Pd-functionalized branched NWs exhibited superior sensor responses of 55.19 to C 6 H 6 gas. In particular, Pd nanoparticles enhanced the sensor response to C 6 H 6 gas most efficiently, increasing the sensor response by 173.5%. Possible mechanisms for the sensing of the Pd-decorated branched nanowires will be associated with resistance modulation along the branch TeO 2 nanowires (including catalytic Pd effects), at the networked homojunctions between the branch TeO 2 nanowires, at the boundaries of the TeO 2 nanograins, and at the Pd/TeO 2 heterojunctions.

Herein, we present the effect of electron-beam irradiation (EBI) on the gas-sensing properties of Pd-functionalized reduced graphene oxide (RGO). Scanning electron microscopy, transmission electron microscopy, and X-ray photoelectron spectroscopy were used to characterize the synthesized products. The samples were irradiated using electron beams at doses of 0 (Pd-RGO-0), 100 (Pd-RGO-100), and 500 kGy (Pd-RGO-500), and the NO2 gas-sensing properties were investigated. It was found that irradiation by electron beams has a critical effect on the gas-sensing properties of the samples, and the Pd-RGO-500 sensor showed the highest response to NO2 gas. In particular, the response of the unirradiated sensor and the sensor irradiated at doses of 100 and 500 kGy to 10 ppm NO2 were 1.027, 1.045, and 1.047, respectively. The response times of Pd-RGO-0, Pd-RGO-100, and Pd-RGO-500 to 10 ppm NO2 gas were 389, 335, and 345 s, respectively. The corresponding recovery times for these sensors were 808, 766, and 816 s, respectively. The increased numbers of oxygen functional groups and high-energy defects were the main reasons for the increased gas response. This study will eventually lead to increased performance levels of RGO-based sensors that use EBI, as this technology can introduce changes into materials in a non-contact, clean, and powerful manner.

Nanodiamond (ND) was successfully synthesized using single-walled carbon nanotubes (SWCNTs) as a pure solid carbon source by means of a spark plasma sintering process. Raman spectra and X-ray diffraction patterns revealed the generation of the cubic diamond phase by means of the SPS process. Lattice-resolved TEM images confirmed that diamond nanoparticles with a diameter of about ∼10 nm existed in the products. The NDs were generated mainly through the gas-phase nucleation of carbon atoms evaporated from the SWCNTs.

Thin Spray-on Liners(TSLs) based on polymer materials have been considered as an alternative to shotcrete and wire mesh in relatively fair rock conditions, and used in mines since 1990s. Nevertheless, Few experimental studies on their mechanical properties necessary for the evaluation of their bearing capacities as a support member have been carried out. In this study, tensile and bond strengths of two kinds of TSLs with different material compositions were measured at the age of 7 days. In addition, two kinds of bending tests proposed by EFNARC (2008) to simulate representative failure mechanisms of TSLs were carried out on the same materials and curing age as in tension and pull-out tests. From the tests, tensile strength of a TSL is shown to increase as its content of polymer is higher. In contrast, its bond strength seems to be in inverse proportion to its polymer content. Especially, the TSL material in which a cementitious component is included with relatively smaller polymer content shows a faster hardening characteristic which results in higher resistance to de-bonding between a TSL and a substrate. As a result, it is shown that the performance of TSLs might be dependent upon its corresponding polymer content.

Type I interferons (IFNs) play an important role in antiviral immunity as well as immunopathogenesis of diverse chronic viral infections. However, the precise mechanisms regulating the multifaceted effects of type I IFNs on the immune system and pathological inflammation still remain unclear. In order to assess the immunological dynamics associated with rapid viral clearance in chronic hepatitis C patients during the acute phase of type I IFN therapy, we analyzed multiple parameters of virological and immunological responses in a cohort of 59 Korean hepatitis C patients who received pegylated IFN-alpha and ribavirin (IFN/RBV). Most of the Korean patients had favorable alleles in the IFN-lambda loci for responsiveness to IFN/RBV (i.e., C/C in rs12979860, T/T in rs8099917, and TT/TT in rs368234815). Rapid virological response (RVR) was determined mainly by the hepatitis C virus genotype. Among the cytokines analyzed, higher plasma levels of IL-17A and FGF were observed in non-RVR patients infected with viral genotype 1 and IP-10 was consistently elevated in RVR group infected with genotype 2 during the early phase of antiviral therapy. In addition, these three cytokines were correlated each other, suggesting a functional linkage of the cytokines in antiviral responses during IFN/RBV therapy. A low baseline frequencies of regulatory T cells and.d T cells, but high level of group 2 innate lymphoid cells, in peripheral bloods were also significantly associated with the RVR group, implicating a potential role of the cellular immunity during the early phase of IFN/RBV therapy. Therefore, the immunological programs established by chronic hepatitis C and rapid disruption of the delicate balance by exogenous type I IFN might be associated with the subsequent virological outcomes in chronic hepatitis C patients.

Calcium signaling has been implicated in various steps in bacterial pathogenesis. Here, we investigated the role of Ca(2+) signaling in intracellular invasion of non-phagocytic host cells infected with Orientia tsutsugamushi, the causative agent of scrub typhus. The bacteria induced a transient Ca(2+) increase in HeLa cells immediately after infection and the source of the mobilized Ca(2+) appears to be intracellular stores, not the extracellular milieu, as determined using extracellular (EGTA) or intracellular (BAPTA-AM) Ca(2+) chelators. O. tsutsugamushi rapidly induced activation of PLC-γ1, as indicated by tyrosine phosphorylation. PLC-γ1 activity increased within 1 min of infection and returned to the basal level by 5 min. Pretreatment of host cells with inhibitors of PLC-γ1 (U73122) or IP3R channel activity (2-APB) significantly blocked bacterial entry without affecting bacterial attachment. In addition, these chemical inhibitors were effective in suppressing not only the activation of ERK MAP kinase but also the expression of the chemokine MCP-1. Taken together, Ca(2+) signaling induced by O. tsutsugamushi may play a crucial role in bacterial pathogenesis including inflammatory reactions as well as intracellular invasion into non-phagocytic host cells.

Dissolving microneedles (DMNs) have been widely studied in medical applications due to their pain-free administration, superior efficiency, and safe drug delivery. In skin vaccination, preserving the activity of the encapsulated antigen is an important consideration, as antigen activity is lost during DMN fabrication because of various stress factors. These stress factors vary between fabrication methods and each method affects the antigen's activity to different degrees. In this study, the activity of encapsulated antigens delivered by DMNs is compared between two recently developed DMN fabrication methods; droplet-born air blowing (DAB) and centrifugal lithography (CL) for a model scrub typhus vaccine antigen, ScaA. Although the in vitro analysis of ScaA-loaded DMNs (ScaA-DMNs) does not show any differences in physical properties depending on the fabrication methods, the immunogenicity of the CL-produced ScaA-DMN is significantly higher based on cytokine measurement and humoral immunity. DAB and CL differ in their solidification conditions, suggesting that solidification factors critically affect the encapsulated antigen's activity. ScaA-DMNs may also be stably stored for 4 weeks at room temperature. In conclusion, CL is a superior DMN fabrication method compared with DAB, and this study proves that DMN is feasible and practical for skin vaccination.

Novel luminescence dyes having benzoxazole 3 and benzthiazole 4 moieties were synthesized and characterized. Zinc (II) complex dyes 5 and 6 were consecutively prepared and their corresponding light emitting properties were also studied with respect to the photoluminescence (PL) functions of the dyes 3 and 4 . The strong light emitting properties and their resulting PL spectra were observed from the dyes 3 and 5 , namely dyes having benzoxazole moieties. To expect the molecular conformation, theoretical modeling calculation were performed to determine the molecular interatomic distance and dihedral angle due to the limitation of single crystal formation from polymeric dye molecules.

Five different types of 2D–π–A (vinylcyanoacetate)pyran derivatives, each having two donor (2D) and one acceptor (A) groups showed significantly large Stokes shifts. Their fluorescence bands shifted to longer wavelengths with an increase in solvent dielectric constant. The π-conjugated moiety, such as a carbazolyl group, as an electron donor did not impart red-emission saturation. A locked ring structure of the electron donor group led to narrowing of the full width at half maximum (fwhm) and to increased dipole moment in the ground state; in addition, bulky donor groups resulted in molecular stability and restrained electronic perturbation on the excited state. An EL device based on the dyes displayed bright and saturated red light with a high luminance of about 2000 cd/m2 at a current density of 160 mA/cm2 at 16 V.

A novel, bolaform spiroxazine in which the hydrophobic chain comprised a hexamethylene unit and a 1,4-diazabicyclo[2,2,2]octane unit was synthesized. Alternate multilayer films of the spiroxazine and polystyrenesulfonate (PSS) were prepared using layer-by-layer deposition. UV–vis absorbance measurements revealed that the bilayer deposition process was linear and highly reproducible. The photoinduced ionic conductivity response of the SO/PSS multilayer was also studied.

In this study, we prepared a novel hetero-bi-functional chromic dye and evaluated the photochromic and electrochromic properties. With based on spiroxazine as a photochromic part and 4,4′-bipyridinyl derivative, namely, viologen as an electrochromic part within the single dye molecular structure, this novel multi-functional chromic dye showing both photochromism and electrochromism was synthesized and characterized. The absorption spectra of the colored viologen part after electrocoloration are shown: the prominent peak appeared at 610 nm and was colored blue. Further absorption spectral increases were determined using successive UV irradiation.

In the previous study, we synthesized and characterized the novel hetero-bi-functional chromic dye having spiroxazine moiety as a photochromic part and viologen moiety as an electrochromic part within the single dye molecular structure. This novel multi-functional chromic dye showed photochromic and electrochromic properties and DMSO solution. When UV irradiation and electric energy-on/off states were repeated, these photochromism and electrochromism properties were represented with a fairly good reversibility. First-order kinetics indicates that the open-to-close reaction of spiroxazine occurred more rapidly than the redox system reaction of viologen. Structural change of the spiroxazine to the merocyanine form was in difficulties due to the inclusion complex between dye and β-cyclodextrin, which displayed the dramatic decrease of absorption peaks.

As the basic approach to improve the emission performance under cold start engine operation to meet stringent emission regulations, the effects of thin wall catalysts and spark timing retard on total hydrocarbon (THC) emission characteristics were investigated by engine performance and vehicle emissions tests. From this study, the effects of cell density on back pressure and engine performance were studied for thin-wall catalysts. The light-offtime reduction of the thin-wall catalysts was also demonstrated through vehicle emission tests. The effect of spark timing retard from minimum spark advance for best torque on THC emission reduction under the cold-start condition was also studied quantitatively using a fast flame ionization detector and a flame visualization technique. As the spark timing is retarded, THC emission at the exhaust manifold is effectively reduced regardless of the air-fuel ratio. From flame visualization, as the spark timing is retarded, the flame propagation speed becomes slower and the duration of the main flame is longer. It was also found that the reduction in THC emission at the beginning of the engine start is essential to meet the more stringent emission regulations. As a result, the adoption of a high-cell-density catalyst (900 cells/2.0 mil) and the spark timing retard technique (spark advance retard case, after top dead centre 8 ° crank angle) makes it possible to meet the super-ultra-low-emission vehicle emission regulation if effectively combined along with a metallic catalyst and exhaust gas-flow-optimized exhaust manifold.

Cell surface heparan sulfate proteoglycans (HSPGs) play a critical role in the cellular invasion of intracellular bacteria and are presumed to have a role in the infection of host cells by Orientia tsutsugamushi. Previously, we showed that O. tsutsugamushi infection decreased markedly after treating host cells with heparinase, which suggests that HSPGs play an important role in oriential infection. We tested oriential infection in REF-Syn4 cells over-expressing syndecan-4, and in REF-Syn4AS cells in which the expression of syndecan-4 was down regulated by transfecting with anti-sense syndecan-4 cDNA. Oriential infection was found to be dependent on the expression level of syndecan-4 on the cell surface. Furthermore, the infectivity of O. tsutsugamushi was specifically reduced by treating O. tsutsugamushi with the purified recombinant core protein of syndecan-4 (Syn4E). These results suggest that the core protein of syndecan-4 and the heparin/heparan sulfate chain of syndecan play an important role in oriential infection by O. tsutsugamushi.

Objectives: We performed a prospective study to evaluate the intestinal colonization of Oxalobacter formigenes and its relationship with urinary oxalate levels in patients with calcium oxalate stone disease. Methods: One hundred and three patients with calcium oxalate urolithiasis, ranging in age from 21 to 73 years (mean age, 47 years) who were followed from August 2000 to September 2001 participated in this study. Fresh stool and 24-hour urine samples were collected. Genus specific oligonucleotide sequences corresponding to the homologous regions residing in the oxc gene were designed. In order to quantify O. formigenes in clinical specimens, a quantitative-PCR-based assay system utilizing a competitive DNA template as an internal standard was developed. Urine volume, pH, creatinine, oxalate, calcium, magnesium, phosphate, citrate and uric acid were measured. Results: Intestinal Oxalobacteria were detected in 45.6% ( n =47) of calcium oxalate stone patients by PCR. In stone formers who tested negative for Oxalobacteria , the average urinary oxalate level was 0.36mmol/day, and this compared to 0.29mmol/day for those patients that tested positive for Oxalobacteria ( p 7 (0–4.1×10 8 ), and the level of 24hours urine oxalate significantly decreased with increasing level of colony forming units of O. formigenes ( r =−0.356, p =0.021). Conclusion: Our results support the concept that O. formigenes is important in maintaining oxalate homeostasis and that its absence from the gut may be the risk of calcium oxalate urolithiasis.

Copper dissolution and electrochemical behavior have been investigated in order to find out which parameters are critical and important during the two major copper removal processes for chemical cleaning of nuclear steam generator and to evaluate safety aspects and effectiveness of these processes. Hydrogen peroxide was very effective for the process using EDTA-based solution at 38 °C to control the corrosion potential of copper into an optimum potential range from −0.3 to +0.2 V for copper sludge dissolution. Corrosion rates of carbon steel SA 285 Gr.C and Alloy 600 were very small in this potential range. The process using EDA-based solution at 60 °C was effective to dissolve copper sludge if the corrosion potential of copper could be controlled above −0.3 V. However, it was very difficult under the laboratory conditions to raise its corrosion potential to this range by air blowing and stirring.

The effect of temperature on magnetite dissolution and corrosion behavior was evaluated in ethylenediaminetetraacetic acid (EDTA)-based solvents for iron oxide chemical cleaning of nuclear steam generators at temperatures ranging from 93 to 150 °C. Magnetite dissolution was drastically accelerated as the temperature was raised to 150 °C, while the amount of corrosion was controlled well within an allowable limit. Periodic venting and excessive EDTA concentration in the solvents were expected to be very effective in dissolving hard tube sheet sludge piles. The linear polarization (LP) method was also suitable to monitor the changes of corrosion rates and process conditions such as venting.

The influence of hydrazine, pH and inhibitor concentration were evaluated with respect to the dissolution effectiveness and material corrosion when using the iron oxide removal solvent composed of ethylenediaminetetraacetic acid (EDTA), N 2 H 4 , NH 4 OH and the corrosion inhibitor CCI-801 at 150 °C. The optimum EDTA solvent chemistry at 150 °C was found to be 1% N 2 H 4 , pH 7 and 1% corrosion inhibitor. This formula is quite similar to the conventional 200 °F (93 °C) process.

Reactive oxygen species (ROS) play an important role in cellular signaling as second messengers. However, studying the role of ROS in physiological redox signaling has been hampered by technical difficulties in controlling their generation within cells. Here, we utilize two inert components, a photosensitizer and light, to finely manipulate the generation of intracellular ROS and examine their specific role in activating dendritic cells (DCs). Photoswitchable generation of intracellular ROS rapidly induced cytosolic mobilization of Ca(2+), differential activation of mitogen-activated protein kinases, and nuclear translocation of NF-κB. Moreover, a transient intracellular ROS surge could activate immature DCs to mature and potently enhance migration in vitro and in vivo. Finally, we observed that intracellular ROS-stimulated DCs enhanced antigen specific T-cell responses in vitro and in vivo, which led to delayed tumor growth and prolonged survival of tumor-bearing mice when immunized with a specific tumor antigen. Therefore, a transient intracellular ROS surge alone, if properly manipulated, can cause immature DCs to differentiate into a motile state and mature forms that are sufficient to initiate adaptive T cell responses in vivo.

Background: Orientia tsutsugamushi, the causative agent of scrub typhus, is an obligate intracellular bacterium. Previously, a large number of genes that encode proteins containing eukaryotic protein-protein interaction motifs such as ankyrin-repeat (Ank) domains were identified in the O. tsutsugamushi genome. However, little is known about the Ank protein function in O. tsutsugamushi. Methodology/Principal Findings: To characterize the function of Ank proteins, we investigated a group of Ank proteins containing an F-box-like domain in the C-terminus in addition to the Ank domains. All nine selected ank genes were expressed at the transcriptional level in host cells infected with O. tsutsugamushi, and specific antibody responses against three Ank proteins were detected in the serum from human patients, indicating an active expression of the bacterial Ank proteins post infection. When ectopically expressed in HeLa cells, the Ank proteins of O. tsutsugamushi were consistently found in the nucleus and/or cytoplasm. In GST pull-down assays, multiple Ank proteins specifically interacted with Cullin1 and Skp1, core components of the SCF1 ubiquitin ligase complex, as well as the eukaryotic elongation factor 1 alpha (EF1 alpha). Moreover, one Ank protein co-localized with the identified host targets and induced downregulation of EF1 alpha potentially via enhanced ubiquitination. The downregulation of EF1 alpha was observed consistently in diverse host cell types infected with O. tsutsugamushi. Conclusion/Significance: These results suggest that conserved targeting and subsequent degradation of EF1 alpha by multiple O. tsutsugamushi Ank proteins could be a novel bacterial strategy for replication and/or pathogenesis during mammalian host infection.

Using a newly developed in situ affinity-labeling method of nucleotide-binding proteins (NTPoxi technique) we discovered that the human T-cell receptor-associated CD3-zeta protein might bind GTP/GDP. To further characterize GTP/GDP binding to CD3-zeta, murine T-cell lines expressing zeta zeta homodimers or CD3-zeta/Fc epsilon R1 gamma heterodimers were used. Specific GTPoxi labeling of CD3-zeta was found in all murine T cells in which a complete CD3-zeta polypeptide chain was expressed, including cells in which CD3-zeta was disulfide bridged to the Fc epsilon R1 gamma chain. In murine T cells the kinetics of labeling of CD3-zeta was similar to that of small G-proteins. Upon activation of murine T cells a slight but significant increase in GTPoxi labeling of CD3-zeta was detected. Whether all 3 so-called 'Reth motifs' (zeta A, zeta B and/or zeta C) were necessary for the binding of GTP/GDP was addressed by using cells expressing truncated CD3-zeta molecules. Whereas truncated CD3-zeta, in which zeta A and part of zeta B were deleted, was still able to bind GTP, upon deletion of all 3 Reth motifs cross-linking by the GTPoxi method became impossible. Regardless of whether this implies a direct or indirect binding of GTP/GDP to CD3-zeta, these nucleotides and their hydrolysis must play an important role in T-cell activation through the TCR/CD3 complex.

It has been proposed that during T cell receptor antigen recognition, CD4- or CD8-p56lck molecules interact with the T cell antigen receptor-CD3 complex (TCR-CD3) to phosphorylate various undefined substrates, which then initiate signal transduction through the TCR-CD3 complex. The ability of CD4 to modulate the TCR-CD3-induced increase in intracellular Ca2+, [Ca2+]i, and substrate tyrosine phosphorylation was studied in mutants of the human leukemic T cell line HPB-ALL characterized by their low expression of the TCR-CD3 complex on the cell surface. In TCR-CD3low cells, in which CD3-zeta was found to be associated with the TCR-CD3 complex, cross-linking CD3 with CD4 resulted in a profile of calcium mobilization, CD3-zeta, and phospholipase C-gamma 1 tyrosine phosphorylation similar to that observed in HPB-ALL cells, although the magnitude of generalized substrate tyrosine phosphorylation appeared to be smaller, as compared with wild-type cells. Responses were weak or absent when CD3 was cross-linked alone. In contrast, in a mutant in which association of CD3-zeta 2 with the TCR-CD3 was defective, cross-linking of CD3 with CD4 had a weaker effect on any of the activation parameters tested. These experiments showed that the presence of CD3-zeta 2 in the TCR-CD3 complex is of critical importance for the ability of CD4 to enhance early transducing signals inside the cell. The data also suggest that CD4-associated protein tyrosine kinase p56lck could up-regulate defective CD3-mediated induction of phospholipase C activity by increasing tyrosine phosphorylation of phospholipase C-gamma 1.

Role of transmembrane heparan sulfate proteoglycans on invasion of Orientia tsutsugamushi into host cells was investigated. Pretreatment with heparan sulfate and heparin inhibited the infection of O. tsutsugamushi for L cell, mouse fibroblast, whereas other glycosaminoglycans had little effect. These same treatments were also shown to reduce the infection in a dose-dependent manner, and enzymatic treatment of cells with heparitinase, but not chondroitinase ABC, inhibited the infection. In addition, mutant cell lines of Chinese hamster ovarian cell defective in heparan sulfate synthesis but not chondrotin sulfate synthesis and defective in all glycosaminoglycan synthesis showed marked reduction in susceptibility to infection by O. tsutsugamushi. Also mutant cell lines, which express heparan sulfate proteoglycans at low level, showed intermediate level of infectivity. Finally O. tsutsugamushi bind to(35)S-labelled heparin. Collectively, these findings provide strong evidence that heparan sulfate proteoglycans contribute to the attachment of O. tsutsugamushi to the cells.

Polymethine dyes have been synthesized using bis-dimethylaminophenylethylene compound. These dyes can be converted to the colorless leuco form by the treatment with alkali. Interestingly, these leuco dyes showed reversible color formation properties with following acid addition. The feasibility of the use as thermochromic indicator was investigated using bisphenol A and 1-hexadecanol.

The inclusion complex properties of the multichromic dye in DMSO with three different type of cyclodextrins, namely α-, β- and γ-cyclodextrin were studied by absorption spectra measurements. The first order fading rates were also calculated by decoloration properties of the three inclusion complex conditions. Futhermore, the inclusion complex of different sizes of cyclodextrins was compared, which showed the evident decoloration data for interaction of the multichromic dye and cyclodextrins.
