In this investigation, the X-ray photoelectron spectroscopic technique has been used to examine the surface chemistry of C.I. Leuco Sulphur Black 1 dyed cotton fabric, which has been after-treated with a tannin-based commercial product, Bayprotect CL, and laundered in accordance with ISO 105 C06 and C09 washing protocols. The dyeings were evaluated for colour strength, wash fastness and colour loss. Surface chemical compositions of the dyeings were analysed to relate visual changes with surface chemical changes. Alterations in percentage atomic concentrations of carbon, nitrogen, oxygen and sulphur were compared for untreated, after-treated, unlaundered and laundered dyed samples. After-treated and laundered dyed samples exhibited higher content of surface sulphur and lower surface oxidation, as compared to the untreated counterparts. The S (2p) spectra of after-treated and laundered samples demonstrated reduced over-oxidised (S6+) species at 168 eV. There was 5% and 4% less surface oxidation for ISO 105 C06 and C09 washing systems, respectively.

Purpose Sulphur dyes provide an inexpensive medium to dye cellulosic fibres with heavy shade depths. They offer moderate to good fastness to light and wet treatments. However, owing to the environmental hazard produced by the use of sodium sulphide, the practical implication of these dyes is steadily decreasing. Moreover, these dyes are prone to oxidation causing pronounced fading on exposure to laundering. This paper aims to present the green processing of sulphur dyes by using a biodegradable reducing agent in place of sodium sulphide to dye cotton fabrics. The study also proposes after-treatments with tannin to improve the fastness properties of the dyeings. Design/methodology/approach In this study, dyeings were produced on cotton fabric with a range of C.I. Leuco Sulphur dyes, which were reduced with sodium sulphide and glucose. Sulphur dyeings were after-treated with an environment-friendly tannin-based product (Bayprotect CL (BP)); subsequently, the after-treated samples were evaluated for colour strength, wash, light and rubbing fastness. Findings A novel after-treatment method was developed, which substantially improved the wash fastness of C.I. Leuco Sulphur Black 1 dyeing to ISO 105 C06/C09 washing. However, the degree of this improvement varied for the other sulphur dyes used. The surface morphology and the possible mechanisms for the improved fastness properties were also discussed. Research limitations/implications The effect of after-treatment was significant for improving the wash fastness of sulphur black dyeings in particular, while the effect on other colours was minor. Significant improvements were observed for light and wet rub fastness for most of the dyeings, which signifies the importance of tannins as a finishing agent. Practical implications It is observed that the tannin-based product, BP, is found to provide the photoprotective effect by improving the lightfastness of the dyeings. Future research may involve the exploration of various tannins as a finishing agent to sulphur dyeings. Originality/value This novel finishing technique is found significant for improving the wash fastness of sulphur black 1 dyeings for both the reducing systems. Improvements were also observed for light and wet rub fastnesses for most of the dyeings.

Introduction: SARS-CoV2, first reported in December 2019 in Wuhan as COVID-19 causing respiratory illness, rapidly evolved into a pandemic owing to its very high infectivity. There is insufficient evidence about if and how smoking affects the risk of COVID-19 infection, and the reports on whether smoking increases or reduces the risk of respiratory infections, are contradictory. Therefore, the current study was designed to determine the effects of nicotine consumption on the infectivity of COVID-19. Methods: We performed in silico computer simulation-based study. The structures of SARS-CoV2spike ectodomain, and its receptor ACE2, were obtained from PDB. The structure of nicotine and its metabolites NNK and NNAL were obtained from the PubChem chemical database. After optimization, they were interacted using AutoDock 4.2, to see the effect of nicotine, NNK, or NNAL presence on the docking of viral spike protein to its receptor ACE2. Results: ACE2 vs spike protein interaction results were used as a control (ZDOCK score 1498.484, with four hydrogen bonds). The NNK+ACE2 vs spike protein docking formed 10 hydrogen bonds with the highest ZDOCK score of 1515.564. NNAL+ ACE2 vs spike protein interaction formed eleven hydrogen bonds with the ZDOCK score of 1499.371. Nicotine+ACE2 vs spike protein docking showed the lowest ZDOCK score of 1496.302 and formed 8 hydrogen bonds. Whereas, NNK+spike vs ACE2 interaction had a ZDOCK score of 1498.490 and formed eight hydrogen bonds. NNAL+spike vs ACE2 docking formed eleven hydrogen bonds with a ZDOCK score of 1498.482. And Nicotine+spike vs ACE2 interaction showed a ZDOCK score of 1498.488 and formed 9 hydrogen bonds. Conclusions: The binding of nicotine to either spike of virus or its receptor ACE2 is not affecting the viral docking with the receptor. But binding of NNK, a metabolite of nicotine, is facilitating the viral docking with its receptor indicating that smoking may increase the risk of COVID-19 infection.

Sulfur dyed cellulosic goods exhibit limited wash fastness against oxidative laundering; particularly to washing powder formulations containing activated bleaches. This study presents a novel two-bath exhaust finishing process to improve the wash fastness of Sulfur Black 1 dyed cotton fabrics reduced with two different reducing agents—sodium sulfide and a commercial glucose-based Diresul reducing agent D. The finishing process involves sequential aftertreatments with a cationic fixative and synthetic tanning agent (syntan). A Box-Behnken design with three variables: concentrations of cationic fixative and syntan, and application temperature of syntan (at three different levels) was studied to understand the effects of these variables on the color retention of the dyed cotton fabrics. Color retention of the sulfur dyeings was improved using the newly developed finishing process.

Flexible and ultrasensitive biosensing platforms capable of detecting a large number of trinucleotide repeats (TNRs) are crucial for future technology development needed to combat a variety of genetic disorders. For example, trinucleotide CGG repeat expansions in the FMR1 gene can cause Fragile X syndrome (FXS) and Fragile X-associated tremor/ataxia syndrome (FXTAS). Current state-of-the-art technologies to detect repeat sequences are expensive, while relying on complicated procedures, and prone to false negatives. We reasoned that two-dimensional (2D) molybdenum sulfide (MoS2) surfaces may be useful for label-free electrochemical detection of CGG repeats due to its high affinity for guanine bases. Here, we developed a low-cost and sensitive wax-on-plastic electrochemical sensor using 2D MoS2 ink for the detection of CGG repeats. The ink containing few-layered MoS2 nanosheets was prepared and characterized using optical, electrical, electrochemical, and electron microscopic methods. The devices were characterized by electron microscopic and electrochemical methods. Repetitive CGG DNA was adsorbed on a MoS2 surface in a high cationic strength environment and the electrocatalytic current of the CGG/MoS2 interface was recorded using a soluble Fe(CN)6-3/-4 redox probe by differential pulse voltammetry (DPV). The dynamic range for the detection of prehybridized duplexes ranged from 1 aM to 100 nM with a 3.0 aM limit of detection. A detection range of 100 fM to 1 nM was recorded for surface hybridization events. Using this method, we were able to observe selectivity of MoS2 for CGG repeats and distinguish nonpathogenic from disease-associated repeat lengths. The detection of CGG repeat sequences on inkjet printable 2D MoS2 surfaces is a forward step toward developing chip-based rapid and label-free sensors for the detection of repeat expansion sequences.

The mitochondrial manganese superoxide dismutase (MnSOD) enzyme protects lungs against oxidative stress by neutralizing the free radical superoxide produced in the respiratory function. This has relevance to asthma. Therefore, it is of interest to describe the potential effect of MnSOD Ala16Val genetic polymorphism to asthma risk. Known data in this context is inconclusive in nature. The possible link between MnSOD Ala16Val polymorphism and asthma is explored using sequence meta-analysis. Data from the pooled analysis of MnSOD Ala16Val polymorphism using five genetic models i.e., allelic (Val vs. Ala: p=0.846; OR=1.033, 95% CI=0.742 to 1.440) is discussed. Homozygous (Val Val vs. Ala Ala: p=0.517; OR=1.307, 95% CI=0.582 to 2.932) and heterozygous (Val Ala vs. Ala Ala: p=0.307; OR=1.138, 95% CI=0.888 to 1.459) data using the described models are documented. Data from the dominant model (Val Val + Val Ala vs. Ala Ala: p=0.301; OR=1.289, 95% CI=0.797 to 2.085) and the recessive model (Val Val vs. Val Ala + Ala Ala: p=0.761; OR=0.924, 95% CI=0.555 to 1.538) analyses for several ethnic subgroups in this context is reported.

PurposeSulphur dyes are the most highly consumed colourants for cellulosic substrates owing to their reasonable cost and acceptable fastness. However, the use of noxious conventional reducing agent, sodium sulphide and impaired wash fastness against oxidative bleaching is gradually decreasing the market of these dyes. As the need for “Green” goods and services is raising public awareness, this paper aims to use a glucose-based biodegradable reducing agent in place of sodium sulphide to dye cotton fabrics with a range of commercial sulphur dyes. The study also proposes an aftertreatment method to improve the fastness properties of the dyeing.Design/methodology/approachThe paper investigated the impact of a newly developed aftertreatment method on the fastness properties of dyeing. This involved the sequential application of a cationic fixing agent (Tinofix ECO) and tannin (Bayprotect CL) on the coloured fabrics and subsequent evaluation of colour strength, washing, light and rubbing fastness.FindingsThe effect of aftertreating the dyed cotton was found to significantly improve the light and wet rub fastness. The surface morphology of the dyeing remained unaffected as depicted by the absence of any finish residues.Research limitations/implicationsThe protective effect of the cation–tannin aftertreatments was examined with a view to providing the necessary commercial performance; however, it was established that the dry rub fastness was either reduced or remained unaffected and the wash fastness to International Organization for Standardization 105 C09 was also marginal.Originality/valueThis finishing technique is novel and can be found useful for manufacturing sulphur-dyed products with the improved light and wet rub fastness.

Sulphur‐dyed cellulosic textiles are susceptible to washing powders containing activated peracid formulations resulting in obvious fading during repeated laundering. This study presents a novel one‐bath/two‐stage exhaust finishing process to improve the oxidative wash fastness of CI Leuco Sulphur Black 1‐dyed cotton fabric. The effect of the combined Tinofix ECO and Bayprotect Cl after‐treatment was to impart better colour fastness to oxidative International Organisation for Standardisation (ISO) 105 CO9 washing of the dyed cotton compared with those dyed fabrics after‐treated with Tinofix ECO alone. In addition, while the dyed fabrics rub and light fastness performances were unchanged by the after‐treatments, some improvement in the tensile strength of the combined Tinofix ECO and Bayprotect Cl after‐treated fabrics was observed.

In this study, cotton fabric and nylon fabric were dyed with a range of commercial sulphur dyes and the light and wash fastness of the coloured fabrics was investigated. The effect of after‐treating the coloured cotton and nylon fabrics with a tannin‐based commercial product, Bayprotect Cl, in the presence or absence of sodium sulphate in the treatment bath, was found to significantly improve the light fastness of the sulphur‐dyed cotton, and the photoprotective effect was partially stable to ISO 105‐C06 washing. In addition, the tannin‐based after‐treatment also improved the colour stability of the dyed fabrics to the perborate‐based ISO 105‐C06 washing. The possible mechanisms for the improved fastness properties are also discussed. The application of sulphur dyes to nylon is potentially commercially useful but has been limited because of the reported poor light fastness of the dyeings. The photoprotective effect of the tannin‐based after‐treatment was investigated with a view to providing the necessary commercial performance. However, it was established that on this fibre, the light fastness improvement was marginal, and the associated wash fastness to oxidative bleach‐based ISO 105‐C06 washing was limited.

Purpose DNA damage is a continuous process occurring within the cells caused by intrinsic and extrinsic factors, but it gets repaired regularly. If the DNA repair process is faulty, the incidences of damages/mutations can accumulate in cells resulting in cell transformation. It is hypothesized that the negative variations in DNA repair pathways in even at one point viz. genetic, translational or posttranslational stage may fairly be crucial for the beginning and development of carcinogenesis. Therefore, we investigated the potential of tobacco specific nitrosamines (TSNs) related carcinogens to interact with the enzymes involved in DNA repair mechanisms in the current study. Methods The derivatives of cigarettes' smoke like NNK and NNAL are very well known and recognized carcinogens. Therefore, almost 120 enzymes playing crucial role in the DNA repair process have been analysed for their reactivity with NNK and NNAL. Results The molecular docking study helped to screen out, 07 possible DNA repair enzyme targets for NNK, and 12for NNAL. Present study revealed the loss of activity of DNA repair enzymes in the presence of NNK and NNAL, and this accumulation may induce the tendency of DNA damage which can lead the transformation of exposed normal cells in to cancerous cells. This study also demonstrated the protective potential of nanoparticles like SWCNTs/MWCNTs against TSN's induced toxicity; here SWCNT against NNK (-17.16 Kcal/Mol) and MWCNT against NNK -17.01 Kcal/Mol were showing maximum binding affinities than the known biomolecular target of NNK 1UGH (Uracil-DNA glycosylase,-7.82Kcal/Mol). Conclusion CNTs can be applied as chemo-preventive agents against environmental and tobacco induced carcinogens owing to their scavenging potential and warrants for in vivo and in vitro experimental validation of the results obtained from the present study. .

DNA repeat expansion sequences cause a myriad of neurological diseases when they expand beyond a critical threshold. Previous electrochemical approaches focused on the detection of trinucleotide repeats (CAG, CGG, and GAA) and relied on labeling of the probe and/or target strands or enzyme-linked assays. However, detection of expanded GC-rich sequences is challenging because they are prone to forming secondary structures such as cruciforms and quadruplexes. Here, we present label- free detection of hexanucleotide GGGGCC repeat sequences, which cause the leading genetic form of frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS). The approach relies on capturing targets by surface-bound oligonucleotide probes with a different number of complementary repeats, which proportionately translates the length of the target strands into charge transfer resistance (R(CT)) signal measured by electrochemical impedance spectroscopy. The probe carrying three tandem repeats transduces the number of repeats into R(CT) with a 3× higher calibration sensitivity and detection limit. Chronocoulometric measurements show a decrease in surface density with increasing repeat length, which is opposite of the impedance trend. This implies that the length of the target itself can contribute to amplification of the impedance signal independent of the surface density. Moreover, the probe can distinguish between a control and patient sequences while remaining insensitive to non-specific Huntington’s disease (CAG) repeats in the presence of a complementary target. This label-free strategy might be applied to detect the length of other neurodegenerative repeat sequences using short probes with a few complementary repeats.

DNA is strongly adsorbed on oxidized graphene surfaces in the presence of divalent cations. Here, we studied the effect of DNA adsorption on electrochemical charge transfer at few-layered, oxygen-functionalized graphene (GOx) electrodes. DNA adsorption on the inkjet-printed GOx electrodes caused amplified current response from ferro/ferricyanide redox probe at concentration range 1 aM–10 nM in differential pulse voltammetry. We studied a number of variables that may affect the current response of the interface: sequence type, conformation, concentration, length, and ionic strength. Later, we showed a proof-of-concept DNA biosensing application, which is free from chemical immobilization of the probe and sensitive at attomolar concentration regime. We propose that GOx electrodes promise a low-cost solution to fabricate a highly sensitive platform for label-free and chemisorption-free DNA biosensing.

We present a PNA-based microprobe sensing platform to detect nucleic acid repeat mutations by electrochemical impedance spectroscopy. The microprobe platform discriminated Huntington’s disease-associated CAG repeats in cell-derived total RNA. This sensitive, label-free, and PCR-free detection strategy has the potential to detect a plethora of length mutation disorders.

BACKGROUND Seborrheic keratosis is a benign skin tumour. None of the common therapies is satisfactory. This study compared the efficacy of 30% hydrogen peroxide versus 50% trichloroacetic acid in treatment of seborrheic keratoses. METHODS This unblinded, comparative interventional study was conducted on 85 patients of seborrheic keratosis. Lesions were graded using physician's lesion assessment (PLA) scale (0, clear; 1, nearly clear; 2≤1 mm thick; 3≥1 mm thick). Patients were randomly assigned into two groups: A and B. Five target seborrheic keratosis lesions with PLA score of three were selected in each patient. Group A was treated with 30% hydrogen peroxide and group B with 50% trichloroacetic acid. Both groups received treatment every week up to 5 weeks or till complete resolution of lesions. Clinical photographs of all patients were taken every 2 weeks. RESULTS Both the groups were comparable in terms of age and sex. At the end of 6 weeks, lesions were cleared completely in 41.8% of patients in group A and in 23.8% in group B. Temperature monitoring and thermal management of CMOS circuits / sensors is often required for long term circuit reliability and sensors temperature compensations. The ideal solution is to co-fabricate the temperature sensors along with the CMOS circuits and other sensors using CMOS materials and the same CMOS processes. In this paper we therefore, report on the fabrication and thermal characterization of such thin film temperature sensors. The thin film Resistance Temperature Detectors (RTDs) are made of single crystal p-type silicon on a silicon substrate using a Silicon on Insulator (SOI) Complementary Metal Oxide Semiconductor (CMOS) process. The influence of thin film length and width on the response of RTDs has been investigated. Furthermore, the effects of aluminum (Al) and tungsten (W) metallization used for power tracks on the response of RTDs have been studied. To assess the performance of the RTDs based on SOI CMOS p-type silicon, in addition to sensitivity and TCR, their power consumption, linearity, hysteresis, repeatability, reproducibility and expected life span have also been evaluated. The p-type silicon RTDs consume very less power (i.e. 0.9 μW only) and demonstrate a sensitivity of 13.88 Ω / oC, which is much higher than the previously reported CMOS RTDs and widely used non-CMOS platinum RTDs. The RTDs also exhibit a linear response (98.9% fsd), an excellent repeatability and significantly low hysteresis with maximum variation of 0.2% between forward and reverse cycle. These RTDs also boast good reproducibility, within the wafer and wafer to wafer, with maximum sensitivity variation of 1.3% between different tested samples. The expected life span of the RTD is > 10 years. These inexpensive RTDs are therefore, good candidates for use as temperature sensors on CMOS circuits / sensors system.

Survivin (IAP proteins) remains an important target for anticancer drug development as it is reported to be over-expressed in tumor cells to enhance resistance to apoptotic stimuli. The study focuses on virtual screening of marine compounds inhibiting survivin, a multifunctional protein, using a computational approach. Structures of compounds were prepared using ChemDraw Ultra 10. Software and converted into its 3D PDB structure and its energy was minimized using Discovery Studio client 2.5. The target protein, survivin was retrieved from RCSB PDB. Lipinski's rule and ADMET toxicity profiling was carried out on marine compounds and the filtered compounds were further promoted for molecular docking analysis and interaction studies using AutoDock Tools 4.0. Molecular docking results revealed that analog (AP 4) of Aplysin, showed very promising inhibitory potential against survivin with a binding energy of -8.75 kcal/mol and Ki 388.28 nM as compared to its known inhibitor, Celecoxib having binding energy of -6.65 kcal/mol and Ki 13.43 μM. AP 4. The analog depicted similarity in pattern when compared to standard. The result proposes AP 4, is an effective molecule exhibiting prominent potential to inhibit survivin and thus promoting apoptosis in tumor cells.

Trinucleotide repeat (TNR) sequences introduce sequence-directed flexibility in the genomic makeup of all living species leading to unique non-canonical structure formation. In humans, the expansions of TNR sequences are responsible for almost 24 neurodegenerative and neuromuscular diseases because their unique structures disrupt cell functions. The biophysical studies of these sequences affect their electrophoretic mobility and spectroscopic signatures. Here, we demonstrate a novel strategy to characterize and discriminate the TNR sequences by monitoring their capillary flow in the absence of an external driving force using wax-on-plastic microchannels. The wax-on-plastic microfluidic system translates the sequence-directed flexibility of TNR into differential flow dynamics. Several variables were used to characterize sequences including concentration, single- vs. double-stranded samples, type of repeat sequence, length of the repeat sequence, presence of mismatches in duplex, and presence of metal ion. All these variables were found to influence the flow velocities of TNR sequences as these factors directly affect the structural flexibility of TNR at the molecular level. An overall trend was observed as the higher flexibility in the TNR structure leads to lower capillary flow. After testing samples derived from relevant cells harboring expanded TNR sequences, it is concluded that this approach may transform into a reagent-free and pump-free biosensing platform to detect microsatellite expansion diseases.

Fabrication of inexpensive and flexible electronic and electrochemical sensors is in high demand for a wide range of biochemical and biomedical applications. We explore hand fabrication of CNT modified AgNPs electrodes using wax-on-plastic platforms and their application in electrochemical immunosensing. Wax patterns were printed on polyethylene terephthalate-based substrates to laydown templates for the electrodes. Hand painting was employed to fabricate a silver conductive layer using AgNPs ink applied in the hydrophilic regions of the substrate surrounded by wax. CNT was drop cast on top of the working electrodes to improve their electrochemical signal. The device layers were characterized by scanning electron microscopy. The electrochemical performance of the hand fabricated AgNPs and CNT/AgNPs electrodes was tested using cyclic voltammetry, differential pulse voltammetry, and amperometry. The electrochemical response of CNT/AgNPs electrodes was relatively faster, higher, and more selective than unmodified AgNPs sensing electrodes. Finally, the hand-painted CNT/AgNPs electrodes were applied to detect carcinoembryonic antigen (CEA) by measuring the end-product of immunoassay performed on magnetic particles. The detection limit for CEA was found to be 0.46 ng/mL.

The importance of sulphur dyes are realized due to the fact that they offer an economical method of achieving dyeing of good color strength and acceptable fastness on cellulosic substrates. However, the main limitations associated with this class of dye are the lack of availability of bright colors and it’s fading on exposure to laundering with detergent and perborates/peroxides formulations. In order to meet the growing requirements of customers regarding the higher wash fastness, garment traders are in turn demanding their suppliers to achieve these properties against single and multiple washes. In this study, sulphur black 1 dyed cotton fabric has been aftertreated with Fixapret CP, Bayprotect Cl and CC (Choline Chloride) with various sequential combinations to improve the wash fastnesss of the dyed fabric against the aggressive ISO-1O5 CO9 washing system. Sequential application of Bayprotect Cl followed by CC was found to offer a significant improvement causing a 12% reduction in percentage colour loss relating to the untreated fabric, which was comparable to the independent application of Fixapret CP with minimum deleterious effects on the hue, chroma and tensile strength of the fabric.

We report a novel bimetallic iron oxide/nickel oxide metal–organic framework (FeO/NiO MOF) that has been synthesized by using two different linkers; 2, 5-dihydroxy terephthalic acid (DHTA) and Pyrazine and also its composites are prepared with reduced graphene oxide from 1 to 5 wt% rGO FeO/NiO MOF and afterwards characterized via several important techniques like FTIR, XRD, SEM and EDX for examination of their morphological and structural properties. The synthesized series of bimetallic MOFs composites were later used as catalyst for methanol oxidation, tested via cyclic voltammetry (CV) in 3 M Methanol and 1 M NaOH on modified GCE. The electrochemical results illustrate that electrocatalytic response of the MOF boosts astonishingly with gradual loadings of rGO from (1–4 wt%) and among all prepared series 4 wt% rGO-FeO/NiO MOF shows the highest current density of 486.14 mA/cm2 at potential of 0.85 V with scan rate of 50 mV/cm2. This superlative response of prepared catalyst makes it a promising catalytic material for methanol oxidation in DMFC.

Expression of various cytochrome P450s (CYPs) in mammalian brain cells is well documented. However, such studies are hampered in neural/glial cells of human origin due to nonavailability of human brain cells. To address this issue, we investigated the expression and inducibility of CYP2C8 and CYP3A4 and their responsiveness against cyclophosphamide (CPA) and organophosphorus pesticide monocrotophos (MCP), a known developmental neurotoxicant in human neural (SH-SY5Y) and glial (U373-MG) cell lines. CPA induced significant expression of CYP2C8 and CYP3A4 in both types of cells in a time-dependent manner. Neural cell line exhibited relatively higher constitutive and inducible expression of CYPs than the glial cell line. MCP exposure alone could not induce the significant expression of CYPs, whereas the cells preexposed to CPA showed a significant response to MCP. Similar to the case of CPA induced expressions, neural cells were found to be more vulnerable than glial cells. Our data indicate differential expressions of CYPs in cultured human neural and glial cell lines. The findings were synchronized with protein ligand docking studies, which showed a significant modulatory capacity of MCP by strong interaction with CYP regulators-CAR and PXR. Similarly, the known CYP inducer CPA has also shown significant high docking scores with the two studied CYP regulators. We also observed a significant induction in reactive oxygen species (ROS), lipid peroxides (LPO), micronucleus (MN), chromosomal aberration (CA), and reduction in reduced glutathione (GSH) and catalase following the exposure of MCP. Moreover, the expressions of apoptotic markers such as caspase-3, caspase-9, Bax, and p53 were significantly upregulated, whereas the levels of antiapoptotic marker, Bcl2, was downregulated after the exposure of MCP in both cell lines. These findings confirm the involvement of ROS-mediated oxidative stress, which subsequently triggers apoptosis pathways in both human neural (SH-SY5Y) and glial (U373-MG) cell lines following the exposure of MCP.

Nanotechnology, a science dealing with particles at nano scale, is currently used in many fields including environmental management and medicine for welfare of human being. The economic development and quality of life have been improved through nanotechnology. The Polycyclic aromatic hydrocarbons (PAHs) and other toxicants have higher affinity to scaveng by nanopartilces. The structural properties and surface chemistry of nanoparticles are the players, further, extremely high surface area to volume ratio results in multiple enhancement of many beneficial properties. Hence, we have followed a methodology to compare the binding efficiency of nanoparticles and cigarette smoke carcinogens with selected enzymes involved in DNA repair pathways. The molecular interactions have been accomplished using PatchDock server and interestingly got significant interacting results for our hypothesis. PatchDock results showed nanoparticles could be able to trap cigarette smoke carcinogens efficiently in the cellular system. The highest obtained binding efficiency between 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) versus Single wall carbon nanotube (SWcNT) is 2632 score in contrast with NNK versus Human MDC1 BRCT T2067D in complex (PDB ID: 3K05) shows 2454 score, which means NNK could interact with SWcNT more efficiently than 3K05. Another part of the study shows that the highest binding efficiency 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) versus SWcNT = 2746 score and NNAL versus Titanium dioxide (TiO2) Rutile = 2110 score in contract with NNAL versus Human Thymine DNA Glycosylase(PDB ID: 2RBA) shows 1696 score. It is also signified that NNAL interact with SWcNT and TiO2 rutile more efficiently than 2RBA. The results clearly signifying that SWcNT/TiO2 are binding with NNK/NNAL more efficiently than biomolecules.

The requirement and need of novel techniques to speed up the sanitization of polluted and adulterated sites and reduction in the expenses of these methods is a growing concern. The application of nanoparticles, predominantly the iron nanoparticles, as a pioneering and inventive technique to decontaminate the adulterated sites has received attention and consideration in recent times. Though, all over the world, many research studies have been carried on nanoparticles, diminutive level of knowledge is realized about their performance, actions, and conduct in the soil and in aquatic habitats, their adsorption on soil mineral particles, and communication with soil microbes.

Angiosarcomas are highly aggressive tumors of endothelial origin, which carry a poor prognosis. Fenofibrate is a hypolipidemic drug, which acts by activating the transcription factor PPARα. It has also been widely reported to have ‘anti-cancer’ activity. The current study investigated its effect in a murine VEGF-dependent angiosarcoma cell-line, MS1 VEGF. The study utilised assays to monitor cell proliferation and viability, apoptosis, cell cycle progression, mitochondrial membrane potential, changes in protein expression, and changes in miRNA expression using microarrays. Fenofibrate showed potent anti-proliferative action in MS1 VEGF angiosarcoma cells, without inducing apoptosis. It enriched cells in G2/M cell cycle phase and hyperpolarised mitochondria. Other PPARα activators failed to mimic fenofibrate action. Inhibitors of PPARα and NFκB failed to reverse the inhibitory effect of fenofibrate and their combination with fenofibrate was cytotoxic. Fenofibrate downregulated the expression of key VEGF-effector proteins, including Akt, ERK, Bcl-2 and survivin, and a chemical inhibitor screen discovered relevance of these proteins to cell proliferation. A miRNA microarray revealed that fenofibrate differentially regulated cellular miRNAs with known roles in cancer and angiogenesis. The data raise the possibility that fenofibrate could be useful in angiosarcoma therapy, especially considering its well-established clinical safety and tolerability profile.

Efforts to develop preventatives against HIV infection through sexual route have identified, among many, algal lectins as the potent molecules for scaffolding HIV entry inhibition. Algal lectin scytovirin (SVN) from Scytonema varium, a cyanobacterium, has anti-HIV effects with the potential for use in sculpting HIV neutralization. We created a recombinant strain of human vaginal L. plantarum for extracellular expression of recombinant (r)SVN. The rSVN protein containing culture supernatant was analyzed for its binding with HIV-1 gp160, and for inhibiting infection with primary R5 and X4 HIV-1 strains in TZM-bl cells. The rSVN protein extant in recombinant L. plantarum culture supernatant binds to HIV-1 gp160 and reduces the HIV-induced cytopathic effect to nearly 56.67% and 86.47% in R5 and X4 HIV-1 infected TZM-bl cells, respectively. The fortified L. plantarum may be explored for its use as a live virucide in vaginal mucosa of high risk women to prevent HIV entry.

The B-cell lymphoma-2 (Bcl-2) family proteins have been attributed to be the key regulators in programmed cell death and apoptosis with a prominent role in human cancer. Understanding the fundamental principles of cell survival and death have been the main cornerstone in cancer drug discovery for identification of novel anticancer agents. In this context the Bcl-2 family of anti-and pro-apoptotic proteins provide an excellent opportunity for development of anticancer agents, as blocking the Bcl-2 or Bcl-XL functionally promotes apoptosis in tumor cells and also sensitize them to chemo- and radiotherapies. The present study reports the identification of novel Aplysin analogs as BCL-2 inhibitors from a sequential virtual screening approach using drug-like, ADMET, docking, pharmacophore filters and molecular dynamics simulation. We identified promising Aplysin analogs that have a potential to be Bcl-2 inhibitors just like the standard drug Obatoclax. One of the compound analog 11 was identified to be a promising inhibitor of Bcl-2 in the docking, pharmacophore and simulation based models.The molecular modeling information provided here can be vital in designing of the novel Bcl-2 inhibitors.

The fabrication of cost effective paper-based analytical devices by wax printing has recently become popular, by and large, using cellulose filter papers. Paper-based devices need higher temperature to form hydrophobic barrier across paper substrate, rely on large working channels (≥500 μm) for liquid handling, and exhibit lower efficiency (∼50%) of sample mobility. Such limitations confine applications of wax based fabrication. In this work, we report printability, fidelity, and application of wax micropatterns on a non-cellulosic, non-fibrous, and non-porous polyethylene terephthalate based substrate (mPET). Resolution of wax printing on mPET was found to be 120 μm for line and 60 μm for channel micropatterns. The wax micropatterns can sustain heat and retain their structural integrity at melting temperature of wax and above (≥120 °C). In application, wax microwells were patterned on the new substrate in a high throughput fashion, which formed a suitable niche for mouse embryonic stem cell (mESC) culture either to maintain self-renewal or direct differentiation. This study will open a new direction in wax printing applications not only as a low-cost but a multipurpose fabrication tool.

Dopamine (DA) is a classical neurotransmitter and dysfunction in its synaptic handling underlies many neurological disorders, including addiction, depression, and neurodegeneration. A key to understanding DA dysfunction is the accurate measurement of dopamine uptake by dopaminergic neurons. Current methods that allow for the analysis of dopamine uptake rely on standard multiwell-plate based ELISA, or on carbon-fibre microelectrodes used in in vivo recording techniques. The former suffers from challenges associated with automation and analyte degradation, while the latter has low throughput and is not ideal for laboratory screening. In response to these challenges, we introduce a digital microfluidic platform to evaluate dopamine homeostasis in in vitro neuron culture. The method features voltammetric dopamine sensors with limit of detection of 30 nM integrated with cell culture sites for multi-day neuron culture and differentiation. We demonstrate the utility of the new technique for DA uptake assays featuring in-line culture and analysis, with a determination of uptake of approximately ∼32 fmol in 10 min per virtual microwell (each containing ∼200 differentiated SH-SY5Y cells). We propose that future generations of this technique will be useful for drug discovery for neurodegenerative disease as well as for a wide range of applications that would benefit from integrated cell culture and electroanalysis.

Cigarette smoke derivatives like NNK (4-(Methylnitrosamino)-1-(3-pyridyl)-1-butanone) and NNAL (4-(methylnitrosamino)-1-(3-pyridyl)-1-butan-1-ol) are well-known carcinogens. We analyzed the interaction of enzymes involved in the NER (nucleotide excision repair) pathway with ligands (NNK and NNAL). Binding was characterized for the enzymes sharing equivalent or better interaction as compared to +Ve control. The highest obtained docking energy between NNK and enzymes RAD23A, CCNH, CDK7, and CETN2 were -7.13 kcal/mol, -7.27 kcal/mol, -8.05 kcal/mol and -7.58 kcal/mol respectively. Similarly the highest obtained docking energy between NNAL and enzymes RAD23A, CCNH, CDK7, and CETN2 were -7.46 kcal/mol, -7.94 kcal/mol, -7.83 kcal/mol and -7.67 kcal/mol respectively. In order to find out the effect of NNK and NNAL on enzymes involved in the NER pathway applying protein-protein interaction and protein-complex (i.e. enzymes docked with NNK/NNAL) interaction analysis. It was found that carcinogens are well capable to reduce the normal functioning of genes like RAD23A (HR23A), CCNH, CDK7 and CETN2. In silico analysis indicated loss of functions of these genes and their corresponding enzymes, which possibly might be a cause for alteration of DNA repair pathways leading to damage buildup and finally contributing to cancer formation.

Most of the variables measured in scientific investigations or engineering applications depend, by varying degrees, on temperature. This necessitates the simultaneous measurement of temperature along with the variable of interest in order to perform high fidelity temperature compensated measurements. Silicon diode based temperature sensors (or silicon thermodiodes) have the advantages of being low cost, having an absolute temperature measurement capability as well as providing the option of on-chip integration with electronic circuits and a wide temperature measurement range. Leveraging these advantages, engineers and scientists have used silicon thermodiodes in numerous and diverse applications. This paper identifies the common temperature measuring techniques, and focuses on the use and advantages offered by silicon diodes operated as temperature sensors in different drive modes. Finally it explores the published literature for summarizing the application areas where such sensors have been utilized successfully in recent years.

An initial investigation of the fabrication of a novel biosensor utilizing toll-like receptor 5 (TLR5) has been conducted. The detection assay using this sensor platform has been carried out using two complementary electrochemical techniques. The electrochemical properties of the modified bare gold surface following TLR5 immobilization were characterized. The electrochemical response to changes in the sensor film resistance and electron charge-transfer permittivity triggered by independent exposures to flagellins from Salmonella typhimurium (S. typhimurium) and Bacillus subtilis (B. subtilis) were examined and observed. The quantified film resistance data gathered using electrochemical impedance spectroscopy (EIS) over a macroscopic scale are in significant agreement with the corresponding electron charge-transfer permittivity measured locally by scanning electrochemical microscopy (SECM). Unlike other sensors that exploit pathogen recognition elements, TLR5 biosensors have the potential to carry out broad-spectrum detection of flagellated bacterial pathogens in near real time. This broad-spectrum detection platform is a significant step toward the development of fast, inexpensive clinical tools for early warning diagnoses and immediate on-site treatment.

Fullerenes have attracted considerable attention due to their unique chemical structure and potential applications which has opened wide venues for possible human exposure to various fullerene types. Therefore, in depth knowledge of how fullerene may interfere with various cellular processes becomes quite imperative. The present study was designed to investigate how the presence of fullerene affect the binding of DNA with different enzymes involved in replication process. Different fullerenes were first docked with DNA and then binding scores of different enzymes was analyzed with fullerene docked DNA. C30, C40 & C50 once docked with DNA, reduced the binding score of primase, whereas no significant change in the binding score was observed with the helicase, ssb protein, dna pol δ, dna pol e, ligase, DNA clamp, and topoisomerases. On the contrast, the binding score of RPA14 decreases in fluctuating manner while interacting with increasing molecular weight of fullerene bound single-stranded DNA complex. The study revealed the affect of fullerene family interacting with DNA on the binding pattern of enzymes involved in replication process. Study suggests that the presence of most of fullerenes may not affect the activity of these enzymes necessary for replication process whereas C30, C40 & C50 may disrupt the activity of primase, (strating point for DNA polymerase) its docking score decreases from 13820 to 10702.

The role of niacin's metabolite, nicotinamide adenine dinucleotide (NAD), in DNA repair via base-excision repair pathway is well documented. We evaluated if niacin deficiency results in genetic instability in normal human fetal lung fibroblasts (MRC-5), and further, does it leads to enhanced accumulation of cigarette smoke-induced genetic damage? MRC-5 cells were grown discretely in niacin-proficient/deficient media, and exposed to nicotine-derived nitrosamine ketone (NNK, a cigarette smoke carcinogen). Niacin deficiency abated the NAD polymerization, augmented the spontaneous induction of micronuclei (MN) and chromosomal aberrations (CA) and raised the expression of 10 genes and suppressed 12 genes involved in different biological functions. NNK exposure resulted in genetic damage as measured by the induction of MN and CA in cells grown in niacin-proficient medium, but the damage became practically marked when niacin-deficient cells were exposed to NNK. NNK exposure raised the expression of 16 genes and suppressed the expression of 56 genes in cells grown in niacin-proficient medium. NNK exposure to niacin-deficient cells raised the expression of eight genes including genes crucial in promoting cancer such as FGFR3 and DUSP1 and suppressed the expression of 33 genes, including genes crucial in preventing the onset and progression of cancer like RASSF2, JUP, and IL24, in comparison with the cells grown in niacin-proficient medium. Overall, niacin deficiency interferes with the DNA damage repair process induced by chemical carcinogens like NNK, and niacin-deficient population are at the higher risk of genetic instability caused by cigarette smoke carcinogen NNK.

Caspase-3 has been identified as a key mediator of neuronal apoptosis. The present study identifies caspase-3 as a common player involved in the regulation of multineurodegenerative disorders, namely, Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD), and amyotrophic lateral sclerosis (ALS). The protein interaction network prepared using STRING database provides a strong evidence of caspase-3 interactions with the metabolic cascade of the said multineurodegenerative disorders, thus characterizing it as a potential therapeutic target for multiple neurodegenerative disorders.In silicomolecular docking of selected nonpeptidyl natural compounds against caspase-3 exposed potent leads against this common therapeutic target. Rosmarinic acid and curcumin proved to be the most promising ligands (leads) mimicking the inhibitory action of peptidyl inhibitors with the highest Gold fitness scores 57.38 and 53.51, respectively. These results were in close agreement with the fitness score predicted using X-score, a consensus based scoring function to calculate the binding affinity. Nonpeptidyl inhibitors of caspase-3 identified in the present study expeditiously mimic the inhibitory action of the previously identified peptidyl inhibitors. Since, nonpeptidyl inhibitors are preferred drug candidates, hence, discovery of natural compounds as nonpeptidyl inhibitors is a significant transition towards feasible drug development for neurodegenerative disorders.

Human aldose reductase (hAR) is the key enzyme in sorbitol pathway of glucose utilization and is implicated in the etiology of secondary complications of diabetes, such as, cardiovascular complications, neuropathy, nephropathy, retinopathy, and cataract genesis. It reduces glucose to sorbitol in the presence of NADPH and the major cause of diabetes complications could be the change in the osmotic pressure due to the accumulation of sorbitol. An activated form of hAR (activated hAR or ahAR) poses a potential obstacle in the development of diabetes drugs as hAR-inhibitors are ineffective against ahAR. The therapeutic efficacy of such drugs is compromised when a large fraction of the enzyme (hAR) undergoes conversion to the activated ahAR form as has been observed in the diabetic tissues. In the present study, attempts have been made to employ systems biology strategies to identify the elementary nodes of human polyol metabolic pathway, responsible for normal metabolic states, followed by the identification of natural potent inhibitors of the activated form of hAR represented by the mutant C298S for possible antidiabetic applications. Quantum Mechanical Molecular Mechanical docking strategy was used to determine the probable inhibitors of ahAR. Rosmarinic acid was found as the most potent natural ahAR inhibitor and warrants for experimental validation in the near future.

Arsenic is the most toxic metalloid present in the natural environment in both organic and inorganic arsenic forms. Inorganic arsenic is often more hazardous than the organic form. Arsenite and arsenate compounds are the major inorganic forms which are toxic causing severe human health dysfunction including cancer. Excretion of arsenic from the system is found elusive. Therefore, it is of interest to screen channel proteins with the arsenic complex in the different combination of arsenic, GSH (glutathione) and arsenic, selenium using docking methods. The mode of arsenic removal. The complex structure revealed the mode of arsenic binding efficiency with the receptor aquaporine 9 and ABCC1 channel protein. This provides insights to understand the mechanism of arsenic efflux. These inferences find application in the design, identification and development of novel nutracetucal or any other formulation useful in the balance of arsenic efflux.

Cell cycle is maintained almost all the times and is controlled by various regulatory proteins and their complexes (Cdk+Cyclin) in different phases of interphase (G1, S and G2) and mitosis of cell cycle. A number of mechanisms have been proposed for the initiation and progression of carcinogenesis by abruption in cell cycle process. One of the important features of cancer/carcinogenesis is functional loss of these cell cycle regulatory proteins particularly in CDKs and cyclins. We hypothesize that there is a direct involvement of these cell cycle regulatory proteins not only at the genetic level but also proteins level, during the initiation of carcinogenesis. Therefore, it becomes significant to determine inconsistency in the functioning of regulatory proteins due to interaction with carcinogen 4-(Methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK). Hence, we investigated the interaction efficiency of NNK, against cell cycle regulatory proteins. We found a different value of ΔG (free energy of binding) among the studied proteins ranging between -3.29 to -7.25 kcal/mol was observed. To validate the results, we considered Human Oxy-Hemoglobin at 1.25 Å Resolution, [PDB_ID:1HHO] as a +ve control, (binding energy -6.06 kcal/mol). Finally, the CDK8 (PDB_ID:3RGF) and CDK2 (PDB_ID:3DDP) regulatory proteins showing significantly strong molecular interaction with NNK -7.25 kcal/mol, -6.19 kcal/mol respectively were analyzed in details. In this study we predicted that CDK8 protein fails to form functional complex with its complementary partner cyclin C in presence of NNK. Consequently, inconsistency of functioning in regulatory proteins might lead to the abruption in cell cycle progression; contribute to the loss of cell cycle control and subsequently increasing the possibility of carcinogenesis.

Digital microfluidics (DMF) has emerged as a popular format for implementing quantitative immunoassays for diagnostic biomarkers. All previous reports of such assays have relied on optical detection; here, we introduce the first digital microfluidic immunoassay relying on electrochemical detection. In this system, an indium tin oxide (ITO) based DMF top plate was modified to include gold sensing electrodes and silver counter/pseudoreference electrodes suitable for in-line amperometric measurements. A thyroid stimulating hormone (TSH) immunoassay procedure was developed relying on magnetic microparticles conjugated with primary antibody (Ab1). Antigen molecules are captured followed by capture of a secondary antibody (Ab2) conjugated with horseradish peroxidase enzyme (HRP). HRP catalyzes the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) which can be detected amperometrically. The limit of detection of the technique (2.4 μIU mL(-1)) is compatible with clinical applications; moreover, the simplicity and the small size of the detector suggest utility in the future for portable analysis.

To develop naphthyridine derivatives as anticancer candidates, pharmacokinetic (PK) evaluations of 10 novel derivatives of 1,4-dihydro-4-oxo-1-proparagyl-1,8-naphthyridine-3-carboxamide, with potent anticancer activity were done using in vitro ADME (absorption, distribution, metabolism, excretion) and pharmacokinetic--pharmcodynamic (PK/PD) assays. Only derivatives 5, 6, 9 and 10 showed better metabolic stability, solubility, permeability, partition coefficient and cytochrome P450 (CYP) inhibition values. PK of derivatives 5, 6, 9 and 10 in rat showed comparable PK profile for derivative 5 (C0 = 6.98 µg/mL) and 6 (C0 = 6.61 µg/mL) with no detectable plasma levels for derivatives 9 and 10 at 5.0 mg/kg i.v. dose. PK/PD assay of derivatives 5 and 6 in tumor-bearing mice (TBM) showed comparable PK but tumor plasma index (TPI) of derivative 6 (4.02) was better than derivative 5 (2.50), suggesting better tumor uptake of derivative 6. Derivative 6, as lead compound, showed highest tumor growth inhibition (TGI) value of 33.6% in human ovary cancer xenograft model.

Aggregation of aspartate-β-semialdehyde dehydrogenase (ASD) was analyzed by applying modified Lumry-Eyring with nucleated polymerization (LENP) model. Intrinsic nucleation time scales were determined. In absence of glycerol, ASD undergoes concentration and time-dependent polymerization into low-molecular weight soluble aggregates and thereafter condensation into insoluble aggregates. In the presence of increasing solvent glycerol concentration, the aggregation becomes more and more nucleation dominated, with slower polymerization to low-molecular weights soluble aggregates, without any condensation into insoluble aggregates. Effective nucleus size as well as the number of monomers in each irreversible growth event were sensitive to the changes in solvent glycerol concentration. Glycerol-directed diminution of aggregation appears to be largely due to the inhibition of rearrangement (decreased nucleation rearrangement rate coefficient, K r,x ) because of compaction induced due to preferential hydration, thus, preventing the soluble aggregates from locking into irreversible soluble nuclei. Appreciably decreased K r,x (as compared to nucleation dissociation constant, K d,x ), appears to be responsible for increased nucleus size at higher solvent glycerol concentration. This study explains how modified LENP model can be applied to determine the predominant mechanism responsible for the diminution of aggregation by polyhydric alcohols (glycerol).

Imaging of toll-like receptor microarrays was achieved using scanning electrochemical microscopy with the successful integration of two ferrocene derivatives in order to enhance the background contrast. This investigation has resulted in the novel fabrication of a tuneable, multiplex, broad-spectrum bacterial sensor for the interrogation of conserved microbial stimuli.

The first sequence-dependent study of DNA films containing metal-mediated base pairs was performed to investigate the charge transfer resistance (RCT) of metal-modified DNA. The imidazole (Im) deoxyribonucleoside was chosen as a highly AgI-specific ligandoside for the formation of Im–AgI–Im complexes within the duplexes. This new class of site-specifically metal-modified DNA films was characterized by UV, circular dichroism (CD), and X-ray photoelectron spectroscopy (XPS). The electrochemical properties of these systems were investigated by means of electron impedance spectroscopy and scanning electrochemical microscopy. Taken together, these experiments indicated that the incorporation of AgI ions into the DNA films leads to reduced electron transfer through the DNA films. A simple device was proposed that can be switched reversibly between two distinct states with different charge transfer resistance.

We have performed the interaction analysis of cigarette smoke carcinogens with the enzymes involved in DNA repair mechanisms. Cigarette smoke’s derivatives like 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) are well known carcinogens. The binding efficiency of carcinogens with enzymes obtained from docking methods were ranging from +36.96 to −7.47 kcal/mol. Binding efficiency was characterized for the enzymes sharing equivalent or better interaction as compared to positive control. Also we have analyzed the interaction pattern of NNK and NNAL with DNA. The present study suggests that NNK and NNAL may alter the DNA repair machinery that could initiate the progression of tumor leading to cancer. Computational method explores the toxicological characteristics of these enzymes and also opening an opportunity for researchers.

A lignin peroxidases-producing Serratia liquefaciens was used for bioremediation of pulp and paper (P&P) mill effluent. The treatment led to reduction of chemical oxygen demand (COD), colour, lignin and phenolic content by 84%, 72%, 61% and 95%, respectively. The effluent detoxification was studied by genotoxicity assays using Allium cepa L. (onion) root tip cells. Genotoxicity studies included measuring mitotic index (MI), chromosomal aberrations (CA) and nuclear abnormalities (NA) in root tip cells following treatment with 25, 50, 75 and 100% (v/v) of effluent. The root tip cells grown in untreated effluent showed a significant decrease in MI from 69% (control) to 32%, 27%, 22% and 11% at 25%, 50%, 75% and 100% effluent concentration, respectively. This indicated that the untreated effluent was highly cytotoxic in nature. Further, root tip cells, when treated with different concentrations of effluent showed various CA and NA including c-mitosis, stickiness, chromosome loss, chromosome break, anaphase bridge, multipolar anaphase, vagrant chromosomes, micronucleated and binucleated cells. The MI observed in root tip cells grown in bacterial treated effluents at similar concentrations (25, 50, 75 and 100% v/v) showed an increase of 33%, 36%, 42% and 66%. CA showed a substantial decrease and in some instances, complete absence of CA was also observed. The findings suggest that S. liquefaciens culture could be a potential bacterial culture for bioremediation of P&P mill effluent, as it is effective in substantial lowering of pollutants load as well as reduces the cytotoxic and genotoxic effects of effluent.

The increased inhaled application of titanium dioxide nanoparticles (TiO(2) NPs) increases the potential pulmonary health risks. The present investigations were carried out to study the TiO(2) NPs-induced apoptosis, oxidative stress and genotoxicity in the human lung cancer cell line, A549, a widely used cell system for pulmonary toxicity studies. Tetrazolium bromide salt and lactate dehydrogenase release assays were used to study the cytotoxicity. The genotoxicity studies were carried out using cytokinesis block micronucleus assay. Apoptosis was confirmed by the formation of apoptotic bodies and altered expression (messenger RNA (mRNA) and protein) of markers such as P(53), P(21), Bax, Bcl(2) and cleaved caspase-3. Cells exposed to TiO(2) NPs (10 and 50 μg/ml) for 6-24 h shows significant induction in oxidative stress, that is, the production of reactive oxygen species and malondialdehyde and decrease in the activity of catalase and glutathione. TiO(2) NPs exposure also induces the formation of apoptotic bodies and micronucleus as marker of genotoxicity. A significant up-regulation in the expression of apoptosis markers such as P(53), P(21) and cleaved caspase-3 was observed, while the levels were down-regulated for Bcl(2) at both mRNA and protein levels. TiO(2) NPs exposure could not pose significant effects on Bax expression. Data indicate that nano-TiO(2) induces oxidative stress, genotoxicity and apoptosis in human lung cancer cell line, A549. Our result also identifies the mechanisms involved in TiO(2) NP-induced changes in A549 cells. Perhaps, reporting for the first time, the association of TiO(2) NPs-induced genotoxicity and apoptosis at transcriptional and translational level in the human lung cancer cell line, A549 cells.

Nucleic acids play a critical role in life as we know it. It contains the necessary information required for the structure and function of a living organisms. Metal ions play a critical role in stabilizing conformations. In the well-known double helix structure of DNA, metal ions stabilize a particular conformation that ensures storage and propagation of genetic information. Metal ions, however, can interact with various sites on nucleic acids. Moreover, metal coordination can have a tremendous impact on the structure, conformation, stability and the electronic properties of the nucleic acids. The interactions are controlled by the relative affinity of metal ion coordination to the negatively charged phosphodiester backbone versus binding to other donor sites located in the nucleobases. The canonical Watson–Crick base pairs (A-T and G-C) as well as non-canonical base pairs (Hoogsteen and wobble) and mismatched pairs are often sites for metal ion interactions. In this review, an overview will be provided of the structure of different forms of nucleic acids (DNA and RNA) and the impact of different metal ions on their stability and structure. In addition, the recent applications of metal-DNA interactions in nanotechnology, biosensor and bioelectronics will also be discussed along with some therapeutic applications of metal complexes.

Plasma polymerization of styrene was used to modify the hydrophilic surfaces of halloysite nanotubes to investigate the reinforcement effect of new hydrophobic surfaces on the dynamic mechanical properties of halloysite–polystyrene nanocomposites at various degrees of loading. Pristine and plasma-modified halloysite–polystyrene nanocomposites were synthesized by solution blending and characterized using various techniques. The storage moduli and glass transition temperatures of the composites were analyzed using dynamic mechanical thermal analysis. It was found that the rubbery moduli increased substantially after plasma coating with styrene due to the compatibility of the new hydrophobic halloysite surfaces with the polystyrene polymer matrix. In addition, the rubbery moduli increased with increasing amount of clay. The glass transition temperatures of the composites did not change significantly with the amount of clay. The strengths of the plasma-modified composites were also compared with those of recently reported halloysite–epoxy resin composites. Copyright © 2010 Society of Chemical Industry

Pharmacokinetics o f Florfenicol was established in the plasma of poultry. Florfenicol was given as multiple dose 30 mg.kg−1 orally daily for 5 days in poultry. The mean values of half-life, body clearance (ClB) and mean volume of distribution (Vdarea) were 8.39 and 7.91 h; 0.5 and 0.29 l.h−1.kg−1 and 4.8 and 2.70 l.kg−1 after first and last dose, respectively. On the basis of depletion study for Florfenicol in poultry, a withdrawal time of 72 h was calculated.

Dopamine is linked to gastrointestinal functions. However, its exact nature in stress-induced gastric pathology is still not clear. In the present study, an attempt has been made to identify the effects of dopamine in stress-induced gastric ulcers, and concurrent alterations in various ulcer-influencing factors such as plasma corticosterone levels, gastric mucosal PGE(2) content and proton pump activity. The dopamine D(1) receptor agonist (A 68930) and antagonist (SCH 23390), and D(2) receptor agonist (quinpirole) and antagonist (sulpiride) were used to evaluate their effects on acute stress (single immobilization for 150 min) and chronic unpredictable stress (two different types of stressors for 7 days) induced gastric ulcers in rats. Acute and chronic unpredictable stress significantly increased the gastric ulcer severity, adrenal hypertrophy and corticosterone levels, while gastric mucosal dopamine levels were decreased. Pretreatment of sulpiride (60 mg/kg) significantly reverted the acute stress-induced alterations, while A 68930 (0.25mg/kg) significantly restored the acute and chronic unpredictable stress-induced alterations. In contrast, administration of SCH 23390 (0.1-0.5mg/kg) and quinpirole (0.1-0.5mg/kg) failed to alter acute stress-induced alterations. Further, A 68930 and sulpiride showed different response on proton pump inhibition under in-vitro condition. A 68930 (10-50 microg/ml) inhibited the gastric H(+) K(+)-ATPase activity comparable to positive control omeprazole, while sulpiride (10-50 microg/ml) had no effect. A 68930 also normalized the decreased gastric PGE2 content observed during chronic unpredictable stress. The histopathological evaluation of gastric mucosal tissue supported the observations regarding the gastroprotective effect of sulpiride during acute stress and of A 68930 during both acute and chronic unpredictable stress conditions. Our results provide important insights into the mechanism of dopamine-regulated pathways, which cause an overall pathophysiology of gastric stress ulcers and implicating the importance of D(1) agonist in ulcer protection. Thus, current study highlights the need to evaluate anti-stress and anti-ulcer agents in terms of their ability to modulate dopaminergic transmissions.
