The determination of ethanol in fermented substrates is an important parameter for monitoring the production of distilled beverage samples. The correct measurement of its content has a direct impact on the profitability of the process. In this work, a diffusive micro-distillation device (DMDD) is proposed that allows the determination of ethanol directly in the fermented or distilled beverages samples. The DMDD consists of a 5 mL plastic test tube containing a reagent solution of potassium dichromate and sulfuric acid, inserted into another 50 mL polyethylene tube containing the sample. This set is heated in a water bath for 15 min at 80 °C, providing the ethanol diffusion, which reacts with the receptor solution contained in the test tube. The chromium (III) produced by the oxidation reaction, is spectrophotometrically quantified at 589 nm. The proposed procedure has a linear range between 1 and 12% (v/v) with R2 = 0.999 and RSD = 3.8% and results in agreement with those obtained by the distillation-densitometry official method.

These are considered one of the main pathogens, causing significant economic losses, such as in peppers and guarana. The current forms of control include the use of resistant cultivars, sanitary pruning and fungicides. However, even with the use of some methods of controlling these cultures, the crops are not free of anthracnose. Additionally, excessive application of fungicides increases the resistance of pathogens to agrochemicals and cause harm to human health and the environment. In order to find natural antifungal agents against guarana anthracnose, endophytic fungi were isolated from Amazon guarana. The compounds piliformic acid and cytochalasin D were isolated by chromatographic techniques from two Xylaria spp., guided by assays with Colletotrichum gloeosporioides. The isolated compounds were identified by spectrometric techniques, as NMR and mass spectrometry. This is the first report that piliformic acid and cytochalasin D have antifungal activity against C. gloeosporioides with MIC 2.92 and 2.46 mu mol mL(-1) respectively. Captan and difenocona-zole were included as positive controls (MIC 16.63 and 0.02 mu mol mL(-1), respectively). Thus, Xylaria species presented a biotechnological potential and production of different active compounds which might be promising against anthracnose disease. One sorghum commercial genotype (MASSA 03) and nine ICRISAT high-lysine genotypes from India were analyzed for storage protein content, distribution profile, and soluble amino acid concentrations. Storage proteins fraction were extracted and separated by SDS-PAGE. Soluble amino acids contents were determined by HPLC. Variations in intensity and appearance and disappearance of protein bands were observed among the sorghum genotypes suggesting genetic variability. Amino acid profile also indicated large variations in the amino acid concentrations. The high lysine and threonine soluble concentrations observed in the seeds of the sorghum genotypes encouraged the use of these genotypes as potential food source due to the better balanced amino acids profile.

Saccharomyces cerevisiae hexokinase-less strains were produced to study the production of ethanol and fructose from sucrose. These strains do not have the hexokinases A and B. Twenty-three double-mutant strains were produced, and then, three were selected for presenting a smaller growth in yeast extract-peptone-fructose. In fermentations with a medium containing sucrose (180.3 g L(-1)) and with cell recycles, simulating industrial conditions, the capacity of these mutant yeasts in inverting sucrose and fermenting only glucose was well characterized. Besides that, we could also see their great tolerance to the stresses of fermentative recycles, where fructose production (until 90 g L(-1)) and ethanol production (until 42.3 g L(-1)) occurred in cycles of 12 h, in which hexokinase-less yeasts performed high growth (51.2% of wet biomass) and viability rates (77% of viable cells) after nine consecutive cycles.

The goal of this study was to develop conditional flocculant yeasts for use in the alcohol fermentation industry. Promoters were constructed to completely repress gene transcription in the presence of sugar and to support strong expression after the exhaustion of this compound. A fusion of regulatory regions of the ADH2 promoter with the FLO5 core promoter was constructed to regulate the FLO5 gene. This construct was inserted into multicopy plasmids and transformed into laboratory strains of Saccharomyces cerevisiae, whereby the transformed cells were selected by sedimentation from the bulk medium after sugar exhaustion, without decreasing ethanol production. The ADH2-FLO5 region was converted into an integrative cassette to disrupt the CAN1 gene in industrial yeast strains. Transformed cells became resistant to canavanine and demonstrated conditional flocculation. Although ethanol production was significantly decreased in the industrial transformants, this development reveals a promising technology for the substitution of centrifugation in industrial ethanol production.

A simple DNA isolation method was developed with routine chemicals that yields high quality and integrity preparations when compared to some of the most well known protocols. The method described does not require the use of lysing enzymes, water bath and the DNA was obtained within 40 minutes The amount of nucleic acid extracted (measured in terms of absorbancy at 260 nm) from strains of Xanthomonas spp., Pseudomonas spp. and Erwinia spp. was two to five times higher than that of the most commonly used method.

The herbicide Metolachlor (α-chloroacetamide group) inhibits the growth of Saccharomyces cerevisiae on complete, minimal, and non-fermentative media. Spontaneous and induced resistant mutants showed monogenic segregation patterns. Among the resistant clones, 70% were recessives, 16.4% were partially dominants and 13.4% were dominants. The spontaneous partially dominant mutation Mtc1 was mapped on linkage group XV at 33.3 cM from ade2 and 31.7 cM from his3, in a region that is characterized by the presence of several resistant genes. The recessive mutation mtc2 was located on chromosome IV. Although all the mutants had the ability to grow in the presence of the herbicide, they remained affected in their respiration efficiency, indicating two different mechanism of action of Metholachor on yeast cells.

No. of bitstreams: 0 Previous issue date: 2004-01-01 The aim of the present research was to evaluate the protein polymorphism degrees among forty-eight C. albicans isolates from fourteen anatomical sites of clinical patients by polyacrylamide gel electrophoresis (SDS-PAGE) and numerical analyzes, in order to identify subspecies and their similarities in some infectious niches. Cell cultures were grown in YEPD medium, collected by centrifugation, and washed in cold saline solution. The whole-cell proteins were extracted by cell disruption using glass beads and submitted to SDS-PAGE technique. After electrophoresis, the protein bands were stained with coomassie-blue and analyzed by statistics package NTSYS-pc version 1.70 software. Similarity matrixes and dendrograms were generated by application of similarity coefficient of simple matching and UPGMA algorithm, respectively. The results obtained showed several C. albicans subtypes and their similarity degrees (80% to 100%). Such data showed that same patients may be infected by two or more C. albicans subtypes in certain anatomical sites (i.e. only in oral cavity of immunocompromised patients, blood, or tracheal secretion), or yet, two or more patients can be infected in identical anatomical sites (i.e. bronchial washing, urine, oral cavity, tracheal secretion, vaginal secretion, and healthy saliva) with a same C. albicans subtype. However, two or more patients also can show infections in corresponding sites (i.e. oral cavity of immunocompromised patients, blood, oropharyngeal secretion, oral cavity, tracheal secretion, vaginal secretion, and healthy saliva) by different C. albicans subtypes. Besides, two or more patients also can be infected with identical or different C. albicans subtypes in different anatomical sites (i.e.1. identical subtypes in vaginal secretion, tracheal secretion, and urine; abdominal secretion and spittle; drainage and oral cavity; catheter and healthy saliva - i.e.2. subtypes different in bronchial washing, oropharyngeal secretion, pulmonary secretion, oral cavity of immunocompromised patients, and blood). Complementary studies involving C. albicans sample isolated from several anatomical sites of immunocompetent or immunocompromised patients (before, during and after specifics therapies) and their families or hospital workers must be done in order to establish the sources of C. albicans colonization. The whole-cell proteins profile performed by SDS-PAGE associated with computer-assisted numerical analysis may provide preliminary criteria for taxonomic and epidemiological studies of such microorganisms. Brazil is one of the many countries that forbids the use of anabolic compounds, which generates difficulties on monitoring its use, once it has one of the biggest cattle herd. Therefore, several anabolic compounds are used, including trenbolone acetate. With the agreement of "Ministerio da Agricultura, Pecuaria e Abastecimento", an ELISA based test was done with the production of polyclonal antibodies in rabbits and testing in steers urine which received trenbolone acetate implants. The test showed to be cheap, easy and reliable to use in bovine urine to determine alfa- trenbolone, trenbolone acetate major metabolite, released in the animals urine until 60 days after implant was been injected. The results in comparison to the ones obtained by the commercial kit used by the Brazilian "Ministerio da Agricultura, Pecuaria e Abastecimento" were similar, with no significant differences.

O etanol contribui significativamente para que a matriz energética do país se apresente extremamente favorável quanto à participação da energia renovável. A demanda por este biocombustível é crescente e tecnologias que permitam a sua produção de forma sustentável é de suma importância, como a fermentação com alto teor de etanol, já empregada em alguns países. Linhagens de leveduras com tolerância a múltiplos estresses muito contribuíram para a implantação de tal tecnologia no Brasil. Neste contexto se insere o presente trabalho, o qual busca linhagens de leveduras capazes de suportar os estresses impostos por uma fermentação com alto teor alcoólico. Para tal, 3 entre as melhores linhagens industriais de Saccharomyces cerevisiae atualmente disponíveis (CAT-1, PE-2 e SA- 1), foram utilizadas num programa para a seleção de híbridos para tolerância múltipla aos estresses etanólico, osmótico, ácido, além de outros, inerentes à fermentação com alto teor de etanol. Estas linhagens foram esporuladas e dissecadas para obtenção de células haplóides, as quais foram submetidas a cruzamentos entre- e intra-linhagens. Foram conduzidos cruzamentos massais (aleatórios) cujos produtos foram submetidos à evolução adaptativa em meios com os fatores estressantes em intensidades crescentes no transcorrer de 80 gerações. Ao final da evolução buscou-se variantes prevalentes na população de híbridos, os quais foram submetidos a novos procedimentos seletivos com imposições de várias condições estressantes. Igualmente foram conduzidos cruzamentos direcionados entre haplóides mediante micromanipulação, sendo tais híbridos submetidos ao mesmo procedimento seletivo nos meios estressantes. Assim, a partir de 230 haplóides das 3 linhagens, 174 isolados (120 oriundos dos cruzamentos massais e 54 dos cruzamentos direcionados) foram pré-selecionados pela maior tolerância aos meios seletivos, tendo os seus cariótipos estabelecidos mediante a cariotipagem eletroforética. Os isolados com maior tolerância (27) foram novamente avaliados em fermentações com reciclo de células e sob condições de elevado teor alcoólico (até 14,5% v/v) em mosto de melaço e água. Em todas as etapas da seleção os isolados foram comparados com as linhagens parentais (CAT-1, PE-2 e SA-1), sendo que ao final do processo seletivo destacou-se a linhagem 35B (híbrido entre CAT-1 e PE-2) com atributos fermentativos superiores aos exibidos pelos parentais. Tais atributos fermentativos contemplaram parâmetros bioquímicos, fisiológicos e tecnológicos (rendimento em etanol, viabilidade celular, crescimento em biomassa, formação de glicerol e teores celulares de carboidratos de reserva glicogênio e trealose). Os resultados permitem sugerir que devido às características fermentativas desejáveis do híbrido 35B, o mesmo possa ser empregado no processo industrial para ser avaliado como uma promissora linhagem a conduzir a fermentação com alto teor alcoólico. Ethanol contributes significantly to the energy country matrix, which presents itself as extremely favorable to the share of renewable energy. The demand for this biofuel is increasing and technologies for its production in a sustainable way is of paramount importance such as fermentation with very high gravity, already used in some countries. Yeast strains tolerant to multiple stresses greatly contributed to the deployment of such technology in Brazil. In this context the present work is inserted, which seeks yeast strains capable of withstanding the stresses imposed by high ethanol content fermentation. In order that, three of the best industrial strains of Saccharomyces cerevisiae currently available (CAT-1, PE-2 and SA-1), were used in a program to select hybrids with tolerance towards multiples stresses: ethanolic, osmotic and acid, besides other factors involved in a high ethanol content fermentation. These strains were sporulated and dissected to obtain haploid cells, which were submitted to inter- and intra-strains crossings. Hybrids from polycrossings (random crossings) were subjected to an adaptative evolution in media with increasing stressing action over the course of 80 generations. At the end of evolution, prevalent variants were sought in the hybrids population, and submitted to new selective procedures with several stressing conditions. In the same way, directed crossings (between identified haplois) were performed by micromanipulation, and the resulting hybrids were subjected to the same selective procedure. Therefore, from 230 haploid from the 3 parent strains, 174 were isolated (120 from polycrossings and 54 from directed crossings) and pre-selected for higher tolerance in selective media; moreover their karyotypes were established by electrophoretic karyotyping. Strains showing greater tolerance (27) were again evaluated during cell recycling fermentations with high ethanol content (up to 14.5% v/v) using must formulated with water and molasses. At all stages, the isolates were compared with the parental strains (CAT-1, PE-2 and SA-1), and at the end of the selection process, the strain 35B (hybrid between CAT-1 and PE-2) standed out with fermentative attributes higher than the parentals. The fermentative performance was assessed by biochemical, physiological and technological parameters (ethanol efficiency, cell viability, biomass gain, glycerol formation and cellular levels of reserve carbohydrates - glycogen and trehalose). The results suggest that due to desirable fermentation traits, the hybrid 35B, could be used as starter in industrial fermentation process with high ethanol content.

O presente trabalho teve por objetivo avaliar a atividade celulolítica de fungos isolados de bagaço de cana-de-açúcar e serapilheira, atuantes sobre bagaço de cana-de-açúcar e farelo de arroz como substratos. Dez isolados fúngicos de amostras de bagaço de cana-de-açúcar e material vegetal em decomposição foram avaliados quanto à atividade celulolítica, tendo como referências os fungos Trichoderma reesei QM9414 e T. reesei RUTC30. A atividade celulolítica foi estimada pela capacidade hidrolítica do extrato enzimático dos fungos crescidos em bagaço de cana-deaçúcar, com e sem pré-tratamento térmico e com diferentes proporções de farelo de arroz. A atividade celulolítica foi estimada tanto em papel de filtro, para celulase total, como a carboximetilcelulose sódica, para endoglucanase. Os isolados identificados por análise molecular através da região 26S rDNA foram: Aspergillus giganteus (S1), Aspergillus fumigatus (S2 e F4), Trichoderma viride/Trichoderma hamatum (S3 e S6), Trichoderma koningiopsis (TPB), Paecilomyces variotti (F1), Moniliophthora perniciosa (F2), Acremonium cellulyticus/Penicillium verruculosum (G3) e Trichoderma sp RA305 (MAD). O Trichoderma reesei QM9414 apresentou a maior atividade tanto para celulase total como para endoglucanase. O isolado F1 não diferiu estatisticamente para atividade de endoglucanase em relação à cepa QM9414 em substrato contento somente bagaço. O isolado MAD não diferiu estatisticamente para atividade de celulase total e endoglucanse em relação à cepa QM9414 em substrato contendo 10% de farelo de arroz. Os isolados TPB e S2 apresentaram as maiores atividades para endoglucanase em bagaço de cana-de-açúcar tratado. Tais resultados mostram que a biodiversidade em nichos, como o bagaço de cana-de-açúcar e materiais vegetais em decomposição, podem fornecer linhagens de fungos celulolíticos com grande potencial biotecnológico. The objective of this work was to evaluate the cellulolytic activity of fungi isolated from sugarcane bagasse and decaying plant material, growing on sugarcane bagasse and rice bran as substrate. Ten isolates from sugarcane bagasse and decayed plant material were evaluated for their cellulolytic activity as compared to the fungi Trichoderma reesei QM9414 and T. reesei RUTC30. The cellulolytic activity was estimated by the hydrolytic capacity of the enzymatic extract of fungi grown on sugarcane bagasse, with and without pre-heat treatment and with different proportions of rice bran. It was used as substrates both filter paper, for total cellulolytic activity, and sodium carboxymethyl cellulose, for endoglucanase activity. The isolates identified by molecular analysis using 26S rDNA region were: Aspergillus giganteus (S1), Aspergillus fumigatus (S2 and F4), Trichoderma viride/Trichoderma hamatum (S3 and S6), Trichoderma koningiopsis (TPB), Paecilomyces variotti (F1), Moniliophthora perniciosa (F2), Acremonium cellulyticus/Penicillium verruculosum (G3) and Trichoderma sp RA305 (MAD). Trichoderma reesei QM9414 showed the highest total cellulolytic activity and endoglucanase activity among all isolates. Isolate F1 was not statistically different for endoglucanase activity in relation to strain QM9414 using bagasse as substrate. Isolate MAD was not statistically different for total cellulose activity and endoglucanase activity in relation to strain QM9414 using substrate containing 10% of rice bran. Isolates TPB and S2 had the highest endoglucanase activities using sugarcane bagasse treated as substrate. These results show that biodiversity in niches, such as bagasse and decayed plant material, can provide strains of cellulolytic fungi with great biotechnological potential.

O aumento das atividades industriais intensificou os problemas com poluição ambiental por metais pesados e principalmente por cádmio, por ser dentre os metais pesados, muito utilizado em vários processos industriais e por ser extremamente tóxico em baixas concentrações, além de causar sérios problemas de saúde. Dentre vários processos utilizados na remoção de cádmio de efluentes industriais, um dos mais promissores é a biossorção que envolve a utilização de microrganismos na recuperação e remoção de metais de efluentes. Saccharomyces cerevisiae é um dos organismos que apresentam grande capacidade de biossorção de metais pesados, além de ser um resíduo facilmente obtido em grandes quantidades como subproduto de processos industriais. Visando o tratamento de resíduos líquidos contaminados com cádmio, foram determinados alguns parâmetros ideais para a biossorção de cádmio de soluções artificiais com a utilização de Saccharomyces cerevisiae vivas e mortas por calor. Dentre estes parâmetros foram estudados a concentração de biomassa, concentração do metal, pH e tempo de residência ideal para concentrações de sal entre 10 e 100 mg.L-1. A recuperação do metal pode chegar a 100% e viabilizar o uso de biomassa morta e principalmente de reutilização de biomassa das indústrias alimentícias e alcooleira. The increase of the technological activities intensified the problems with environmental pollution with heavy metals and mainly with cadmium. The attention for this metal is due to the extensive use and for being extremely toxic at low dosages and causing serious problems of human health. Among several process used in the removal of cadmium of effluents one of the most promising it is the biosorption that involves the use of microorganisms in the recovery and removal of effluent metals. Saccharomyces cerevisiae is one of the organisms that show great capacity of biosorption of heavy metals, besides being easily obtained in great amounts as residue of industrial process. Seeking the treatment of polluted liquid residues with cadmium, we determined some ideal parameters for the biosorption of cadmium in artificial solutions with the use of viable and non-living (by heat) Saccharomyces cerevisiae. Among the parameters we studied the biomass concentrations, metal concentrations, pH and ideal time of residence for solutions in concentrations of salt between 10 - 100 mg Cd.L-1. The recovery of the metal can arrive up to 100% and makes possible to use the died biomass and mainly the reuse of biomass from food and sugar-cane alcohol industries.

A Cryptomeria japonica D. Don. "elegans" é uma conífera pertencente à família Taxodiaceae, que apresenta crescimento rápido e boas respostas à fertilização. Dez variações nas concentrações de nitrato e amônio presentes no meio de cultura MS foram realizadas no cultivo 'in vitro' de brotações obtidas a partir de explantes primários de ápices caulinares durante 90 dias. A cada 30 dias foram avaliados, a taxa de crescimento relativo e o incremento em massa vegetal seca. Ao final de 90 dias, foram realizadas análises dos teores de proteínas solúveis totais, aminoácidos solúveis totais, carboidratos não-estruturais totais e eletroforese de proteínas totais em gel de poliacrilamida. Os tratamentos que exibiram resultados mais satisfatórios foram utilizados em um experimento com diferentes relações auxina/citocinina, onde foram avaliadas a produção de brotações e a altura média das mesmas aos 30, 60 e 90 dias de cultivo. As concentrações de nitrato e amônio entre 25 e 31mmol.L -1 e até 5mmol.L -1, respectivamente, foram as mais efetivas para o crescimento e desenvolvimento do material vegetal cultivado 'in vitro'. Durante o experimento com diferentes concentrações de ANA e BAP, foi possível observar que a produção de brotações, etapa essecial para um processo de micropropagação eficiente, foi superior quando as combinações entre nitrato e amônio foram 22,5 + 2,5mmol.L -1 e 25,0 + 5,0mmol.L -1. Cryptomeria japonica D. Don. "elegans" is a fast-growing tree belonging to the Taxodiaceae and it is considered a responsive species 'in vitro'. The aim of this work was to evaluate the effect of ten different concentrations of nitrate and ammonium in morphogenesis 'in vitro' of Cryptomeria japonica D. Don. "elegans" explants. Analysis of relative growth rate, total soluble proteins, total soluble aminoacids, total non structural carbohydrates and electrophoresis of total proteins was performed in order to observe the role of different nitrogen sources on the growth and development of the explants cultivated 'in vitro'. The concentratios of nitrate and ammonium that showed the best results were selected and used in an experiment with different concentrations of NAA and BAP to observe the shoot production. Each experiment was analised at the 30 th , 60 th and 90 th days of culture. The biochemical analysis was performed only at 90 th day of cultive. Concentrations from 26 to 31mmol.L -1 of NO3 - and lower than 5mmol.L -1 of NH4 + were the most effective for growth and development of the explants. However, the experiment with different concentrations of NNA and BAP showed that the best shoot production was achieved on 22,5mmol.L -1 of NO3- + 2,5mmol.L -1 of NH4 + and on 25,0mmol.L -1 of NO3 - + 5,0mmol.L -1 of NH4 + and with 2,0mg.L -1 of BAP + 1,0mg.L -1 of NNA.
