The preparation of pH-responsive nanocontainers by typical silane modification of the mesoporous silica nanoparticle (MSN) surface is usually high-cost, complex, and time-consuming, which remains a great challenge for effective corrosion protection of magnesium alloy. Here, a new strategy to construct pH-responsive nanocontainers (MSN-MBT@LDH) is demonstrated. The nanocontainers consist of corrosion inhibitor (2-mercaptobenzothiazole, MBT) loaded MSN core and layered double hydroxide (LDH) nanosheet shell serving as gatekeepers. The successful loading of MBT and encapsulation by LDH nanosheets were confirmed by a series of characterization such as scanning transmission electron microscopy coupled with energy dispersive X-ray spectroscopy (STEM-EDS) and N2 adsorption/desorption isotherms. The pH-responsive feature of the nanocontainers was demonstrated by determination of the MBT concentration in buffer solutions with different pH values. A smart corrosion protection system on Mg alloy is obtained by incorporating the synthesized nanocontainers into a self-assembled nanophase particle (SNAP) coating. The electrochemical tests and visual observations show that the hybrid coating has the best barrier properties and robustness in corrosion protection in NaCl corrosive solutions in comparison with the control coatings. The present method simplifies the synthesis processes of nanocontainers and eliminates the potential detrimental effect of excess gatekeepers on the coating. The findings provide new insights into the preparation of scalable nanocontainers. The self-healing coatings are expected to have widespread applications for corrosion protection of Mg alloy and other metals.

Aflatoxin B1 (AFB1) induced intestinal epithelial damage in rodent models, which indicates that long-term exposure to AFB1 may cause chronic gut disorders. In this study, we tested the hypothesis that AFB1-induced adverse effects on gut is mediated by gut-microbiota, which is partially reflected by the changes of fecal microbiome and metabolome. F344 rats were orally exposed to AFB1 of 0, 5, 25, and 75 µg kg−1 body weight for 4 weeks and fecal samples were collected. An ion-fragmentation-spectrum-based metabolomics approach was developed to investigate the fecal microbiota-associated metabolic changes in fecal samples. We found that AFB1 inhibited the hepatic and intestinal metabolism of bile constituents. As compared with the controls, bile acid synthesis-associated cholesterols in rats treated with 25 µg kg−1 (the middle-dose group) were significantly decreased in the fecal samples, for example, lathosterol (45% reduction), cholesterol ester (21% reduction), chenodeoxycholic acid (20% reduction), dihydroxycholesterol (55% reduction), hydroxycholesterol (20% reduction), and 5-cholestene (29% reduction). Although disease-associated lipids were not detectable in the feces of the control group, they were found in AFB1-treated groups, including diglyceride, monoacylglyceride, 19,20-dihydroxy-docosapentaenoic acid, and phosphatidylethanolamine. Metabolisms of carbohydrates and production of short-chain fatty acids were remarkedly decreased in all treated groups. Parkinson's disease (PD), the typical neurodegenerative disease, is characterized by the progressive loss of dopaminergic neurons in the substantia nigra (SN). However, no therapeutic agent used currently could slow down neuronal cell loss so as to decelerate or halt the progression of PD. Traditional Chinese medicine (TCM) has been utilized to treat the dysfunction of the autonomic nervous system. Wen-Shen-Yang-Gan decoction (WSYGD) has a good effect on the clinical treatment of PD with constipation. However, it is not clear which ingredients and what mechanism are responsible for the therapeutic effect. In this study, the pharmacodynamic study of WSYGD in PD mice was applied. Concurrently, a novel method for the identification of metabolic profiles of WSYGD has been developed. Finally, we found that WSYGD could protect the PD mice induced by rotenone. The underlying mechanism of the protective effect may be related to the reduction of the DA neurons apoptosis via reducing inflammatory reaction. By virtue of UPLC-MS and chemoinformatics method, 35 prototype compounds and 27 metabolites were filtered out and tentatively characterized. In conclusion, this study provides an insight into the metabolism of WSYGD in vivo to enable understanding of the metabolic process and therapeutic mechanism of PD.

First, the neurobehavioral symptoms in mice of each group were evaluated, and the monoamine neurotransmitters in the striatum in each group were measured with a high-performance liquid phase. Immunofluorescence double staining was adopted to observe the expression of tyrosine hydroxylase (TH), α-synuclein, and LC3. The transmission electron microscope was used to observe the changes of ultrastructure in substantia nigra and the formation of autophagosomes. Then, the expressions of TH, α-synuclein, Beclin 1, LC3, P62, mTOR, and the up-stream protein AKT were detected by Western blot.Results: When compared with the model group, the neurobehavioral function significantly improved in the echinacoside group (P < 0.01), together with increased expression of TH, DA, and DOPAC in the brain (P < 0.01). In the echinacoside group, while the expressions of Beclin 1 and LC3-II increased (P < 0.01), the expression levels of P62 and α-synuclein decreased significantly (P < 0.01). Echinacoside could up-regulate the expression level of the survival signal p-AKT/AKT and decrease the expression of mTOR.Conclusion: Echinacoside could increase autophagy by inhibiting the expression of mTOR, thereby promoting the clearance of α-synuclein and the degradation of the autophagy substrate P62 and exerting the neuroprotective effect.Keywords: Parkinson’s disease, MPTP, echinacoside, α-synuclein, P62, autophagy, mTOR

Synechococcus accounts for a considerable proportion of oceanic carbon fixation and has close relation with heterotrophic bacteria at sub-micrometre scales. However, the relationship between Synechococcus and heterotrophic bacteria and their impacts on oceanic carbon flow is not well understood. Here, we separately co-cultured the heterotrophic bacterial community with two different axenic Synechococcus strains, i.e. Synechococcus sp. PCC7002 and Synechococcus sp. CCMP1334. The dissolved organic carbon (DOC) released by Synechococcus promoted the rapid growth of heterotrophic bacteria. Due in part to the slight differences in the Synechococcus-derived DOC composition, the starting heterotrophic bacterial community was reshaped by the two Synechococcus strains into significantly divergent community structures, suggesting that different heterotrophic bacterial communities may gather around different Synechococcus strains and interact with each other in the oceans. Interestingly, the two Synechococcus strains also released some very stable humic-like DOC components that resisted use by heterotrophic bacteria and were gradually accumulated in seawater, implying that in addition to photosynthetic carbon fixation, Synechococcus may also play an important role in long-term ocean carbon sequestration by producing relatively recalcitrant DOC. Moreover, the interactions of Synechococcus and the heterotrophic bacterial communities could increase aggregate formation and particle sinking, which can likely enhance the contribution of Synechococcus to the magnitude of the sinking biological carbon pump in the oceans.

This study aimed to explore the protective effects of Wenshen-Yanggan decoction on dopaminergic (DA) neuron injury in a rotenone-induced mouse model with chronic Parkinson’s disease (PD) and explore its mechanism of action. Ultraperformance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was used to measure the content of six main components in the Wenshen-Yanggan decoction. The chronic PD mouse model was established by treating 10-month-old healthy wild C57BL/6 male mice with rotenone 30 mg/kg/day for 28 days in succession. The pole test and rotarod test were applied to detect the rescue effect of Wenshen-Yanggan decoction in high, medium, and low dosages, respectively, on PD-like behaviors in mice with chronic PD. The protective effect of Wenshen-Yanggan decoction on the mesencephalic nigrostriatal DA neuron injury was determined employing tyrosine hydroxylase (TH) immunofluorescence staining. Enzyme-linked immunosorbent assay (ELISA) was adopted to measure the inflammatory cytokines in serum, including TNF-α (tumor necrosis factor-alpha), IFN-γ (interferon gamma), NF-κB (nuclear factor kappa-B), and IL-1β (interleukin-1 beta). Western blotting was performed to quantify the expression of phosphorylated c-Jun N-terminal kinase (p-JNK), cleaved caspase-3, B-cell lymphoma-2 (Bcl-2), and NF-κB in the brain. Our results showed that the Wenshen-Yanggan decoction in high, medium, and low dosages reduced the turning time of mice P<0.01,P<0.01, and P<0.05. The high and medium dosages shortened the total climbing time of PD mice in the pole test P<0.01 and P<0.05. Meanwhile, the high, medium, and low dosages increased the rod-standing time of PD mice in the rotarod test P<0.01,P<0.05, and P<0.05. Besides, the decoction reversed the decrease in TH-positive neurons induced by rotenone, upregulated TH protein expression, and downregulated the α-syn expression in the PD model. Moreover, the decoction in high dosage significantly inhibited the expression of p-JNK, cleaved caspase-3, and NF-κB in the midbrain of PD mice P<0.05,P<0.05, and P<0.01, upregulated the expression of Bcl-2 P<0.05, and decreased the content of TNF-α, IFN-γ, NF-κB, and IL-1β in the serum P<0.001,P<0.001,P<0.001, and P<0.001. Taken together, the Wenshen-Yanggan decoction could protect mice from rotenone-induced chronic PD, which might be related to the reduction of the DA neuron apoptosis via suppressing the inflammatory reaction and the neuronal apoptosis pathway.

From April to September 2018, five sampling sites were selected in Lianyungang City for volatile organic compounds (VOCs) analysis, including two sampling sites in the urban area (Lianyungang City Environmental Monitoring Supersite and Mine Design Institute), one sampling site in the industrial area (Deyuan Pharmaceutical Factory), and two sampling sites from the suburb (Hugou Management Office and YuehaiLou). The results showed that the mean VOCs concentration followed this pattern: industrial area (36.06 ± 12.2 µg m−3) > urban area (33.47 ± 13.0 µg m−3) > suburban area (27.68 ± 9.8 µg m−3). The seasonal variation of the VOCs trend in the urban and suburban areas was relatively consistent, which was different from that in industrial areas. The concentration levels of VOCs components in urban and industrial areas were relatively close, which were significantly higher than that in suburban areas. The possible sources and relative importance of VOCs in Lianyungang City atmosphere were measured by the characteristic ratio of toluene/benzene (T/B), ethane/acetylene (E/E) and isopentane/TVOCs. The contribution of traffic sources to the VOCs in Lianyungang City was significant (T/B ~ 2), and there were obvious aging phenomena in the five sampling sites (E/E > 4). The ratio of isopentane/TVOCs in the contribution of gasoline volatilization sources in urban and suburban areas was significantly bigger than that in industrial areas. According to the maximum incremental reactivity (MIR) method, aromatics (40.32–58.09%) contributed the most to ozone formation potential (OFP) at the five sampling sites. The top 10 OFP species showed that controlling n-hexane and aromatics, such as benzene, toluene, xylene, and trimethylbenzene in Lianyungang City can effectively control ozone generation. Nineteen typical VOCs components were selected and the sources of VOCs from five sampling points were analyzed by the principal component analysis (PCA) model. The sources of VOCs in different areas in Lianyungang were relatively consistent. Five sources were analyzed at the two sampling sites in the urban area: industrial emission + plants, vehicle exhaust, fuel evaporation, combustion and industrial raw materials. Four sources were analyzed in the industrial area: industrial emission + plants, vehicle exhaust, fuel evaporation and combustion. Five sources were analyzed at the two sampling sites in the suburban area: industrial emission + plants, vehicle exhaust, fuel evaporation, combustion and solvent usage.

Cubic perovskite-type Sm0.3Sr0.7Nb0.08Co0.92O3−δ (SSNC) demonstrated excellent oxygen reduction reaction (ORR) activity at 600 °C due to a high concentration and high mobility of oxygen vacancies, which were derived from the low average cobalt valence and cubic structure in SSNC at room temperature.

Autophagy serves a crucial role in the etiology of kidney diseases, including drug-induced renal impairment, inherited kidney disease, diabetic nephropathy and aristolochic acid nephropathy (AAN) and is, therefore, a potential target for treatment. We previously demonstrated that rapamycin could attenuate AAN in mice; however, the underlying mechanism remains to be elucidated. Therefore, whether the renal protective effect of rapamycin (an autophagy activator) is related to autophagy in aristolochic acid (AA)-treated mice was of particular interest. The pathophysiological roles of rapamycin were investigated in AA-induced nephrotoxicity in mice and the mechanisms of rapamycin action were explored by evaluating the modulation of autophagy in rapamycin-treated mice and cultured renal tubular epithelial cells. Supplementation with rapamycin reversed AA-induced kidney injury in mice and improved AA-induced autophagy damage in vivo and in vitro. Mechanistically, rapamycin inhibited the renal expression of phosphorylated (p-)mammalian target of rapamycin (mTOR) and p-ribosomal S6 protein kinase 1, which in turn activated renal autophagy and decreased apoptosis, probably by removing AA-elicited damaged mitochondria and misfolded proteins. The findings of the present study demonstrated that rapamycin protects against AA-induced nephropathy by activating the mTOR-autophagy axis and suggested that rapamycin may be a promising pharmacological target for the treatment of AAN.

Urban air pollution has received increasing attention in recent years. To investigate the interaction between several heavy metal elements and the degree of atmospheric pollution, the leaves of three evergreen tree species—Chinese juniper (Juniperus chinensis L.), dragon juniper (Juniperus chinensis L. ‘Kaizuca’), and cedar (Cedrus deodara (Roxb. ex D.Don) G.Don)—were collected from main road intersections in the urban area of Tianjin, China. Two different treatments—water washing (WW) and ethanol washing (EW)—were used, and the contents of Cu, Mn, Cd, and Zn were measured in both washed and unwashed (UW) leaves. It was found that the heavy metal contents within Chinese juniper and dragon juniper were ranked as Mn &gt, Zn &gt, Cu &gt, Cd, and the metal accumulation index (MAI) value was higher for dragon juniper. For the three plants, water washed off 5.36% to 58.58% of the total heavy metals in the needles, while ethanol washed off 16.08% to 71.60% of the total. Both washes were more effective for Cu, Zn, and Cd, and especially for the element Cd. Ethanol could clean off 38.64% to 71.60% of the total Cd from the leaves. Ethanol had a better elution effect compared to water, and the trend of the Cd content in the leaves of the three plants showed a change after the use of different washing methods, which suggests that the water washing may have masked the real difference.

The contamination of heavy metals, a class of naturally occurring and persistent toxicants, has become a major public health concern due to increasing industrial and anthropogenic activities. The use of COPAS Biosort, a flow cytometer capable of measuring thousands of nematodes in minutes via high-throughput assays, has been widely applied in C. elegans studies for assessing toxicity of individual metals; however, such application yet to be seen for metals or other chemical mixtures. In the present protocol, we investigated toxic effects of individual metals, Cd, Pb, and Mn, as well as their binary and ternary mixtures, using nematode C. elegans. The toxic outcomes, including effects on growth, reproduction, and feeding behavior, were measured using high-throughput platform analysis (COAPS Biosort).

Then, immunohistochemistry and Western blot analysis are adopted to measure the expression of tyrosine hydroxylase (TH) and α-synuclein in the substantia nigra (SN). The content of dopamine (DA) and other neurotransmitters in the brain is detected by high-performance liquid chromatography. The expression of nerve growth factors and inflammatory factors in SN in mice in each group is measured by quantitative polymerase chain reaction. Finally, the expression of oxidative stress-related parameters in each group is measured.Results: Compared with the model group, the pole-climbing time among mice in the moderate and high-dose ECH groups is significantly reduced (P < 0.01). The rotarod staying time, as well as fore and hind-limb strides, shows a significant increase (P < 0.01), as does spontaneous activity (P < 0.01). Moreover, the expression levels of TH, DA, glial cell line-derived neurotrophic factor, and brain-derived neurotrophic factor in SN in mice show significant increases in these two groups (P < 0.01). The content of superoxide dismutase, catalase, and glutathione peroxidase indicates significant increases in the low, moderate, and high-dose ECH groups (P < 0.01), and the content of MDA was reduced (P < 0.01). In the high-dose ECH group, the expression of interleukin (IL) 6 and tumor necrosis factor-α is significantly reduced (P < 0.01), while the expression of IL-10 shows a marked increase (P < 0.01) alongside a decrease in the expression of α-synuclein (P < 0.01).Conclusion: Echinacoside improves neurobehavioral symptoms in PD mice and significantly increases the expression of TH and DA. Polypeptide N-acetylgalactosaminyl transferase-6 (GALNT6), a member of the N-acetyl-D-galactosamine transferase family, was shown to be over-expression in mammary cancer and could be used as a biomarker. However, its roles and underlying mechanisms in the pathogenesis of breast cancer are still unclear. In this study, we reported that GALNT6 was up-expression in breast cancer, and it was not associated with tumor stage. The expression level of GALNT6 and menopause status was associated with patient survival. Biological function results illustrated that knockdown of GALNT6 inhibited proliferation, migration and invasion of MDA-MB-231 cells, and increased cell apoptosis. Knockdown of GALNT6 in breast cancer cell attenuated the protein expression of PCNA, cyclin D1, C-myc and β-catenin, and increased the expression of E-cadherin, caspase 3 and cleaved PARP1. Cell fractionation assay showed that knockdown of GALNT6 reduced the levels of β-catenin and MUC1-C in nucleus. Simultaneously knockdown of GALNT6 and β-catenin significantly reduced the level of C-myc. Co-IP experiments indicated that GALNT6 interacted with MUC1-N, β-catenin interacting with MUC1-C in breast cancer cells. Together, our study reveals that GALNT6 promotes tumorigenicity and metastasis through β-catenin/MUC1-C signaling pathway.

The Pr3+-doped NaY(MoO4)2 crystals were prepared by the hydrothermal method, and sodium citrate was used as the chelating agent. The structure, morphology, and relationship between optical properties and temperature were measured. The X-ray diffraction results showed that the crystal structure is body-centered tetragonal, and the addition of sodium citrate did not distinctly alter the phosphor structure. From 298 to 483 K, under a 270 nm excitation, 1D2-3H4, 3P0-3H4, and 3P0-3F2 transitions have different temperature dependencies, and two groups of fluorescence intensity ratio (1D2-3H4 and 3P0-3H4, 1D2-3H4 and 3P0-3F2) can be defined to characterize the temperature. The morphological characteristics of the sample were obtained by the detection of field emission scanning electron microscope. The experiment showed that the relative sensitivity changed as a function of the sample morphology. The maximum relative sensitivity of the synthesized phosphors (Cit3−/Re3+ = 2) reached 1.00% K−1 at 313 K, and the relative sensitivity remained near the maximum value when the temperature was close to human body temperature (308–315 K). The Pr3+-doped NaY(MoO4)2 phosphor is a high-performance material that can be employed for refined calibration and temperature measurements according to the two-group schema defined by fluorescence intensity ratio. Thus, it show utilization potentiality in self-calibration optical thermometers and temperature measurements including humans and special environment.

Parkinson's disease (PD) is a common chronic neurodegenerative disease and greatly affects the quality of PD patients' life. Current symptomatic treatment of PD is limited. There are no effective treatment and drugs that could radically cure PD. Increasing experimental evidence has proven a causal relationship between alpha-synuclein (α-synuclein, α-syn) and the neuropathology of Parkinson's diseases, although the exact pathophysiological role of α-synuclein is not fully clarified. Previous studies showed that monomers and polymers of α-synuclein were secreted from damaged nerve cells via exocytosis and occupied healthy nerve cells via endocytosis, which afford evidence for the prion-like role of α-synuclein. Autophagy is the known mechanism for eukaryotic cells to degrade protein polymers and damaged organelles that proteasome does not cope with. Therefore, promoting the clearance of α-synuclein by enhancing autophagy in neuronal cells could be a promising treatment in the early stage of PD. SIRT1 is a potent regulator of autophagy, because it deacetylates a mass of important transcription factors such as Forkhead Box subgroup O (FoxO) transcription factors family. SIRT1's action relates to FoxO, because FoxO transcription factors are involved in various molecular pathways underlying neuronal protection and autophagy. Moreover, Sirt1 deacetylates proautophagic proteins such as Atg5, Atg7, and Atg8. Echinacoside (ECH) is the main active ingredient of a widely used Chinese herb cistanche, which has been proven to elicit neuroprotective effects in models of neurodegenerative diseases. In this study, we found that ECH could improve PD-like symptoms in MPTP-lesioned mouse model. We further showed that the underlying mechanism of the action of ECH was associated with enhancing autophagy in neurons via bind to Sirt1 directly and affect FoxO expression. Our study demonstrated ECH as a potential therapeutic agent against PD.

Our recent metagenomics analysis has uncovered remarkable modifying effects of green tea polyphenols (GTP) on gut-microbiota community structure and energy conversion related gene orthologs in rats. How these genomic changes could further influence host health is still unclear. In this work, the alterations of gut-microbiota dependent metabolites were studied in the GTP-treated rats. Six groups of female SD rats (n=12/group) were administered drinking water containing 0%, 0.5%, and 1.5% GTP (wt/vol). Their gut contents were collected at 3 and 6 months and were analyzed via high performance liquid chromatography (HPLC) and gas chromatography (GC)-mass spectrometry (MS). GC-MS based metabolomics analysis captured 2668 feature, and 57 metabolites were imputatively from top 200 differential features identified via NIST fragmentation database. A group of key metabolites were quantitated using standard calibration methods. Compared with control, the elevated components in the GTP-treated groups include niacin (8.61-fold), 3-phenyllactic acid (2.20-fold), galactose (3.13-fold), mannose (2.05-fold), pentadecanoic acid (2.15-fold), lactic acid (2.70-fold), and proline (2.15-fold); the reduced components include cholesterol (0.29-fold), cholic acid (0.62-fold), deoxycholic acid (0.41-fold), trehalose (0.14-fold), glucose (0.46-fold), fructose (0.12-fold), and alanine (0.61-fold). These results were in line with the genomic alterations of gut-microbiome previously discovered by metagenomics analysis. The alterations of these metabolites suggested the reduction of calorific carbohydrates, elevation of vitamin production, decreases of bile constituents, and modified metabolic pattern of amino acids in the GTP-treated animals. Changes in gut-microbiota associated metabolism may be a major contributor to the anti-obesity function of GTP.

In this study, male F344 rats were orally exposed to aflatoxin B1 (AFB1) at 0, 5, 25, and 75 μg/kg for 4 weeks. Rat feces were collected from 2 to 4 weeks following exposure and were assessed for gut-microbiota-dependent metabolites. Gut-microbiota-related organic acids were quantitated in the feces using 2-nitrophenylhydrazine derivatization coupled HPLC-profiling method which was validated and showed good reliability, accuracy and sensitivity. After 2-week exposure, AFB1 significantly reduced the levels of fecal short-chain fatty acids (SCFAs) with an over 70% reduction in the high-dose group (75 μg/kg). Mixed-effects model revealed an inverse correlation between AFB1 dose and fecal levels of SCFAs, but no significant time effect was found. When compared with the control, oral exposure to middle-dose AFB1 (25 μg/kg) resulted in remarkable elevations of fecal cholic acid (2.18-fold), linoleic acid (cis-9, cis-12-18:2) (11.3-fold), pentadecanoic acid (15: 0) (3.68-fold), pyruvic acid (4.56-fold), and 3-phenyllactic acid (3.74-fold), but deoxycholic acid level was reduced by 41% in the low-dose group (5 μg/kg). These results demonstrated the disruptions of several important gut-microbiota metabolic pathways, including the synthesis of SCFAs, pyruvic acid related pathways, metabolisms of amino acids, bile acids and long-chain fatty acids, which may further affect host digestive efficiency, energy supply, intestinal immunity, production of neurotransmitters, and enterohepatic cross-talk. Our study suggests that the impairment of gut-microbiota-dependent metabolism may contribute to pathological mechanisms of AFB1-induced adverse health effects.

Populations consuming aflatoxin (AF) and fumonisin (FN)-contaminated foods may be at increased risk for hepatocellular carcinoma (HCC) and developmental disorders; consequently, development of intervention strategies to reduce AF/FN-induced liver disease and adverse health effects in humans could be very useful. Calcium montmorillonite clay (NovaSil) has been shown to absorb AF in vitro, in multiple animal models, as well as in human studies. In the present study, we aimed to evaluate whether uniform particle size NovaSil (UPSN) possessed an ability to modulate the co-carcinogenic potentials of aflatoxin B1 (AFB1) and fumonisin B1 (FB1) in F344 rats. Sequential treatment of FB1 following AFB1 synergistically induces preneoplastic alterations as well as liver damage, indicating that AFB1 acts as an initiator while FB1 as a promoter in the carcinogenesis model, confirming findings from previous studies. The enterosorbent agent UPSN clay at dose of up to 0.5% in diet was shown to be effective in modulating the toxicity and carcinogenicity of co-exposure to AFB1 and FB1, as demonstrated by significant reduction in number and size of hepatic GST-P+ foci, in alterations indicative of liver toxicity, and in levels of AFB1 and FB1 biomarkers.

A PM2.5-capable aerosol chemical speciation monitor (ACSM) was deployed in urban Nanjing, China for the first time to measure in-situ non-refractory fine particle (NR-PM2.5) composition from October 20 to November 19, 2015 along with parallel measurements of submicron aerosol (PM1) species by a standard ACSM. Our results show that the NR-PM2.5 species (organics, sulfate, nitrate, and ammonium) measured by the PM2.5-ACSM are highly correlated (r2 > 0.9) with those measured by a Sunset Lab OC/EC Analyzer and a Monitor for AeRosols and GAses (MARGA). The comparisons between the two ACSMs illustrated similar temporal variations in all NR species between PM1 and PM2.5, yet substantial mass fractions of aerosol species were observed in the size range of 1–2.5 μm. On average, NR-PM1–2.5 contributed 53 % of the total NR-PM2.5 with sulfate and secondary organic aerosols (SOA) being the two largest contributors (26 % and 27 %, respectively). Rapid formation and thereafter growth of secondary inorganic aerosols (SIA) were observed under fog processing in NH3-rich environments. Positive matrix factorization of organic aerosol showed similar temporal variations in both primary and secondary OA between PM1 and PM2.5 although the mass spectra were slightly different due to more thermal decomposition on the capture vaporizer of PM2.5-ACSM. We observed an enhancement of SOA under high relative humidity conditions, which is associated with simultaneous increases in particle surface area, gas-phase species (NO2, SO2, and NH3) concentrations and aerosol water content driven by anthropogenic SIA. These results likely indicate an enhanced reactive uptake of SOA precursors upon aqueous particles. Therefore, reducing anthropogenic NOx, SO2, and NH3 emissions might not only reduce SIA but also SOA burden during haze episodes in China.

Fusarium graminearum is the main worldwide causal agent of Fusarium head blight grain disease. The production of mycotoxin caused by Fusarium head blight epidemics of wheat affects the flour and malt products, which results in heavy economic losses to farmers. In this paper, an inactivation procedure for F. graminearum F7875 spores was tested and found to follow a first-order kinetic model. We characterized the inactivation of F7875 by aqueous chlorine dioxide (ClO 2 ) under different concentrations (5–15 mg L −1 ). The results suggest that aqueous ClO 2 significantly enhanced inactivation of F7875 at high concentration (15 mg L −1 ) compared to lower concentration of 5 and 10 mg L −1 . The time required for a five-log reduction of F7875 spores was calculated based on the established models. Observed by scanning electron microscope, the ClO 2 destroys the structure of the outer coats of F7875, the surface of F7875's hyphal appeared smooth prior to treatment, but after treatment with 30 mg L −1 aqueous ClO 2 , it appeared rugged and wrinkled. In conclusion, our study shows that treatment with aqueous ClO 2 is an effective, fast, and safe method to control F. graminearum in grain production.

This work evaluates chronic safety in middle-aged ovariectomized rats supplemented with different dosages of green tea polyphenols (GTP) in drinking water. The experiment used 6-mo-old sham (n = 39) and ovariectomized (OVX, n = 143) female rats. All sham (n = 39) and 39 of the OVX animals received no GTP treatment and their samples were collected for outcome measures at baseline, 3 mo, and 6 mo (n = 13 per group for each). The remaining OVX animals were randomized into 4 groups receiving 0.15%, 0.5%, 1%, and 1.5% (n = 26 for each) of GTP (wt/vol), respectively, in drinking water for 3 and 6 mo. No mortality or abnormal treatment-related findings in clinical observations or ophthalmologic examinations were noted. No treatment-related macroscopic or microscopic findings were noted for animals administered 1.5% GTP supplementation. Throughout the study, there was no difference in the body weight among all OVX groups. In all OVX groups, feed intake and water consumption significantly decreased with GTP dose throughout the study period. At 6 mo, GTP intake did not affect hematology, clinical chemistry, and urinalysis, except for phosphorus and blood urea nitrogen (increased), total cholesterol, lactate dehydrogenase, and urine pH (decreased). This study reveals that the no-observed-adverse-effect level (NOAEL) of GTP is 1.5% (wt/vol) in drinking water, the highest dose used in this study.

The recent development of selective oxidation of aromatic sulfides with molecular oxygen was highlighted. The sulfoxides and sulfones could be obtained by simply switching the reaction media, i.e., bis(2-butoxyethyl)ether (BBE) or poly(ethylene glycol)dimethyl ether (PEGDME). The application of the high-boiling-point polyether as an initiator and green media can eliminate the need of large quantities of additives and volatile solvents. This strategy represents an economic and eco-friendly method that could find potential applications.

International audience; A single particle soot photometer (SP2) was deployed in urban Nanjing, located in the Yangtze River Delta (China), to investigate the mixing state and sources of ambient refractory black carbon (rBC) from 26 January to 25 February 2014, along with an in-situ measurement of submicron aerosol chemical species by an aerodyne aerosol chemical speciation monitor (ACSM). The results showed that anthropogenic activities associated with firework emissions can be a significant source for rBC-containing particles during the period of the Chinese New Year, resulting from the evident peaks of rBC at midnight. During the residual periods, namely regular day (RD), the diurnal cycles of rBC presented two typical peaks that can be attributed to a synergistic influence of local traffic emissions and boundary layer changes throughout a day. Three coating factors, including organics, sulfate, and nitrate (-rich), were resolved using a positive matrix factorization (PMF) approach to explain the potential contribution of non-rBC coatings (i.e., organics, sulfate, and nitrate) to the coating thickness of rBC-containing particles. As the results show, organic aerosols (OAs) might be a major contributor to the coating thickness of rBC-coating particles during the whole period. The relative coating thickness (a ratio between coated particle size to BC core) exhibited a positive relationship with sulfate, indicative of the favorable coating factor during the episode caused by firework emissions. Source apportionment of rBC was performed via a multiple linear regression between the total rBC mass and each ACSM-PMF factor (rBC-ACSM-PMF). On average, biomass burning emissions accounted for 43%, being the largest contributor during the RD period, whereas local traffic emissions played a major role during the new year time.

Background Wheat straw is a rich resource worldwide. Straw return is an effective strategy to alleviate soil-borne diseases on monoculture watermelon. Previous studies focus on soil structure, physical and chemical properties; however, little is known about the molecular responses on host plant. Results No significant difference on the population of Fusarium oxysporum f.sp. niveum race 1(Fon1) in rhizosphere soil was found between control (no addition of wheat straw) and the treated groups (addition of 1% (T1) or 2% (T2) wheat straw). RNA-Seq analysis showed that 3419 differentially expressed genes were clustered into 8 profiles. KEGG analysis revealed that phenylpropanoid biosynthesis and plant hormone signal transduction were involved in wheat straw induced response in monoculture watermelon. Genes in lignin biosynthesis were found to be upregulated, and the lignin and auxin contents were higher in T1 and T2 compared to the control. Lignin was also enriched and the Fon1 population decreased in watermelon roots treated with wheat straw. The enzyme activities of phenylalanine ammonia-lyase and peroxidase were increased. Conclusions Our data suggest that the addition of wheat straw enhances the defense response to Fon1 infection in watermelon through increasing lignin and auxin biosynthesis.

Background Consumption of moldy food has previously been identified as a risk factor for esophageal squamous cell carcinoma (ESCC) in high-risk countries; however, what contributing roles these dietary carcinogenic mycotoxins play in the etiology of ESCC are largely unknown. Methods A mycotoxin biomarker-incorporated, population-based case-control study was performed in Huaian area, Jiangsu Province, one of the two high-risk areas in China. Exposure biomarkers of aflatoxins (AF) and fumonisins (FN) were quantitatively analyzed using HPLC-fluorescence techniques. Results Among the cases (n = 190), the median levels of AF biomarker, serum AFB1-lysine adduct, and FN biomarker, urinary FB1, were 1.77 pg/mg albumin and 176.13 pg/mg creatinine, respectively. Among the controls (n = 380), the median levels of AFB1-lysine adduct and urinary FB1 were 1.49 pg/mg albumin and 56.92 pg/mg creatinine, respectively. The objective of this study is to determine the occurrence and level of aflatoxins (AFs) contamination in freshly harvested maize for human consumption in rural Kenya. Maize kernels and freshly milled maize flour (n = 338) were collected from households in Siaya and Makueni counties. The importance of phenolic compounds for responding to various environmental conditions has been widely emphasized. However, the role of interactions between polyphenols and ecological factors, especially C, N, and P stoichiometry was little studied. Here, 15 sites across five provinces of Pinus bungeana in temperate regions of China were studied. The results showed that the higher values of total phenolic contents (TPC) of leaf and litter were distributed among the north distribution area of P. bungeana, lower values were in the south, whereas soil TPC were contrary to leaf and litter TPC. The stepwise regression, path analysis and decision index of path analysis for leaf TPC and ecological factors showed that altitude had the most direct impact on leaf TPC. Moreover, the principal determinants of leaf, litter and soil TPC were soil C/P ratios, longitude, and soil N/P ratios, respectively. In addition, the leaf, litter and soil TPC of P. bungeana were limited by soil C/N ratios, mean annual temperature, and soil P, respectively. Overall, our study provided evidence that ecological factors affected strongly the leaf, litter and soil TPC of P. bungeana.

Adding sodium citric as chelator, NaY(MoO4)2: Pr3+, Tb3+ phosphors were prepared by hydrothermal method. XRD (X-ray diffraction) is employed to confirm the microscopic arrangement of the samples, and the test data show that phosphors are body – centered tetragonal structure. The FE-SEM (Field Emission Scanning Electron Microscope) results indicate that the addition of sodium citrate has a certain effect on the morphology of the samples. It was found from the measured temperature-dependent emission spectrum that Pr3+ and Tb3+ has different quenching trends, and temperature can be characterized by FIR (fluorescence intensity ratio) of Pr3+ ion and Tb3+ ion. Moreover, the experimental results display that with the rise of the molar ratio of Cit3−/Re3+, the sensitivity of phosphors gradually decreases, but the relative sensitivity gradually increases. The relative sensitivity of obtained samples has a higher value of 3.00% K−1 (Cit3−/Re3+ = 2). From the CIE chromaticity coordinates, it displays that the luminous color of phosphors changed continuously with the change of temperature.

Dried blood spots (DBS) were proposed as potentially viable method for exposure assessment of environmental toxicants in infant and young children. For this study, we validated an experimental protocol to quantify AFB1-lysine adduct in DBS samples of AFB1-treated F344 rats, as well as samples from human field study. Significant dose-response relationships in AFB1-lysine adduct formation were found in DBS samples of rats treated with single- and repeated-dose AFB1. AFB1-lysine levels in DBS samples were highly correlated with corresponding serum sample levels. The Person coefficients were 0.997 for the single-dose exposure, and 0.996 for the repeated-dose exposure. Levels of AFB1-lysine adduct had also good agreement between DBS and serum samples as shown by Bland-Altman plot analysis. For human field study samples (n = 36), a Pearson correlation coefficient of 0.784 was found between AFB1-lysine adduct levels of DBS and corresponding serum samples. Bland-Altman plots showed the distribution of the log differences between DBS and serum AFB1-lysine levels are within 95% confidence intervals. These results showed AFB1-lysine adduct levels in DBS cards and serum samples from animals and human samples are comparable, and the DBS technique and analytical protocol is a good means to assess AFB1 exposure in infant and children populations.

Deoxynivalenol (DON) is a secondary metabolite produced by Fusarium graminearum in grains, food and contaminated feed, which can lead to many adverse health effects to humans and livestock. The degradation of DON in different contaminated grains oxidated by saturated aqueous ozone (80 mg L−1) was monitored by ultra-high-performance liquid chromatography, tandem-quadrupole-detection mass spectrometry (UPLC–TQD-MS). Results suggest that ozone has a significant effect on DON reduction in solution. When 80 mg L−1 gaseous ozone was used to treat 10 mg L−1 of DON solution, the degradation rate of DON reached 83% within 7 min, while the respective detoxification rates of contaminated wheat, corn and bran by saturated aqueous ozone (80 mg L−1) were 74.86%, 70.65% and 76.21 in 10 min. Ozone at 80 mg L−1 was applied on various DON solution concentrations at 1 mg L−1, 10 mg L−1 and 20 mg L−1 in ultra-pure water. In this paper, the degradation procedure for DON is calculated and described by a first-order kinetic equation. At lower levels (20 mg L−1) of aqueous ozone, intermediate degradation products were observed by ultra-ne quickly and effectively degrades DON and toxicity in various contaminated grains in a matter of minutes. Therefore, ozonation is projected to be an effective, fast, and safe method for DON degradation.

Understanding how engineered nanoparticles (ENPs) interact with natural organic acids is important to ecological risk assessment of ENPs, but this interaction remains poorly studied. Here, we investigate the dispersion stability, ion release, and toxicity of yttrium oxide nanoparticles (nY2O3) suspensions after exposure to two low molecular weight natural organic acids (LOAs), namely benzoic acid and gallic acid. We find that in the presence of LOAs the nY2O3 suspensions become more stable with surface zeta potential more positive or negative, accompanied by small agglomerated size. LOA interaction with nY2O3 is shown to promote the release of dissolved yttrium from the nanoparticles, depending on the concentrations of LOAs. Toxic effects of the nY2O3 suspensions incubated with LOAs on Scenedesmus obliquus as a function of their mixture levels show three types of signs: stimulation, inhibition, and alleviation. The mechanism of the effects of LOAs on the nY2O3 toxicity may be mainly associated with the degree of agglomeration, particle-induced oxidative stress, and dissolved yttrium. Our results stressed the importance of LOA impacts on the fate and toxicity of ENPs in the aquatic environment.

The decontamination of Aflatoxin B1 (AFB1) by immobilized cells of a new mutant strain, prepared on a base of HSCAS (hydrated sodium calcium aluminosilicate), was studied. Novel strains were induced by UV irradiation, from which 50 were screened according to their degradation efficacy on AFB1, compared with the wild strain (FS-Z1). The FS-UV1 strain exhibited highest degradation efficacy, which was confirmed by 18SrDNA to be Aspergillus niger. The results indicate that both immobilized cells and this mutant strain which are incubated for 48 h at 30 °C, would considerably remediate AFB1 in nutrient broth culture, by 95.32% and 82.43%, respectively. By the application of samples of contaminated cottonseed meal, with results of 93.46%∼96.82%, the degradation rate was also validated. The results of Ames test indicate the mutagenic activity of treated AFB1 is greatly abated, with treated controls. The Application of LC-q-TOFMS (liquid-chromatography, quadrupole, time-of-flight mass spectrometry) deduces the structure and molecular formulas of the degradation products. In the vivo study, the damages of photomicrographic evidence are decreased in kidney and liver and the serum biochemical parameters is improved, in response to preventative treatment with immobilized cells. This is the application of HSCAS-prepared, immobilized A. niger cells to degrade AFB1 of contaminated samples. The investigation in this paper offers a novel path for economical, time-saving biodegradation of AFB1 in foods and feeds.

Nickel is the most prevailing metal allergen with the highest sensitization rate among the “TOP 25” contact allergens and can affect about 15% of the human population. It is an essential trace metal in plants, animals, and humans. However, the environmental levels of nickel are considerably higher than what is needed for human life. Exposure to high levels of nickel can lead to skin allergies, lung fibrosis, and carcinogenesis. Few existing studies have closely examined the toxicity of nickel, let alone investigated the effective detoxification pathways. Here, we developed a high-throughput screening platform to comprehensively evaluate the nickel toxicity in wild-type C. elegans and explore the underlying detoxification mechanisms in transgenic nematodes. We demonstrated that nickel exerted multiple toxic effects on growth, brood size, feeding, and locomotion in C. elegans. Of which, brood size is the most sensitive endpoint. Nickel was found to first bind to phytochelatin (PC) after entering the worms’ body and this PC-Ni complex was further transported by the ABC transporter, CeHMT-1, into the coelomocytes for further detoxification. Our study also demonstrated that the high-throughput screening platform is a promising system for evaluation and investigation of the ecological risks of heavy metals.

Green tea polyphenols (GTPs) were found to boost mammal energy conversion by modulating gut-microbial community structure, gene orthologs and metabolic pathways. Here we examined the metabolites present in the gut-microbiota-dependent mitochondrial tricarboxylic acid (TCA) cycle and urea cycle using hydrophilic interaction liquid chromatography (HILIC)-heated electrospray ionization (HESI)-tandem liquid chromatogram mass spectrometry (LC-MS). Six groups (n=12) of Sprague-Dawley rats (6-mo, ~250 g) were administered with water containing 0%, 0.5%, and 1.5% GTPs (wt/vol or g/dL). Gut-content samples were collected at 3- and 6-mo. Untargeted metabolomics detected 2177 features, with 91 features demonstrating significant dose- and time-dependencies on the GTPs treatment. Targeted metabolomics analysis revealed remarkable changes of 39 metabolites in the mitochondrial TCA cycle and urea cycle, including argininosuccunic acid (0.9-fold vs control), dihydrouracil (1.14-fold vs control), fumaric acid (1.19-fold vs control), malic acid (2.17-fold vs control), citrulline (1.86-fold vs control), and succinic acid (0.4-fold vs control). The untargeted metabolomics data were mined using bioinformatics approaches, such as analysis of variance-simultaneous component analysis (ASCA), enrichment pathway analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway mapping analysis. The results of 16S rRNA survey, metagenomics analysis, and metabolomics analysis were extrapolated and integrated using databases of Integrated Microbial Genomes and Microbiomes (IMG/M) and KEGG. Our analysis demonstrates that GTPs enhance energy conversion by boosting mitochondrial TCA cycle and urea cycle of gut-microbiota in rats. This metabolic modulation is achieved by enriching many gene orthologs, following the increase of beneficial microbials in families C. Ruminococcaceae, C. Lachnospiraceae and B. Bacteroidaceae.

Humic acids are one of the main organic matters in sediments and contribute importantly to the marine biogeochemical cycles. Extracellular electron transfer is a ubiquitous natural process and has potentials to change the macrostructure of humic acids which can act as an electron shuttle. By setting up marine sediment microbial fuel cells, the present study revealed that enhanced extracellular electron transfer process could increase the content of C and H, but decrease the O content in humic acids, which could result in an increased aromaticity and decreased polarity of humic acids, whereas no significant changes occurred to the humification degree of the humic acids. Specific bacterial groups as potential exoelectrogens including Proteobacteria (especially Pseudomonas strains) and Firmicutes were enriched under enhanced extracellular electron transfer process, indicating that they were active to exchange electrons and might play important roles during the changes of humic acids, while the relative abundance of Verrucomicrobia and Bacteroidetes was reduced during these processes. The results of the present research shed lights on the relation between exoelectrogens and the transformation of humic acids in coastal sediment, while the microbial process and mechanisms behind it require further study.

Although it is well-recognized that inflammation enhances leukocyte bactericidal activity, the underlying mechanisms are not clear. Here we report that PRL2 is sensitive to oxidative stress at inflamed sites. Reduced PRL2 in phagocytes causes increased respiratory burst activity and enhances phagocyte bactericidal activity. PRL2 (Phosphatase Regenerating Liver 2) is highly expressed in resting immune cells, but is markedly downregulated by inflammation. in vitro experiments showed that PRL2 was sensitive to hydrogen peroxide (H2O2), a common damage signal at inflamed sites. In response to infection, PRL2 knockout (KO) phagocytes were hyper activated, produced more reactive oxygen species (ROS) and exhibited enhanced bactericidal activity. Mice with PRL2 deficiency in the myeloid cell compartment were resistant to lethal listeria infection and cleared the bacteria more rapidly and effectively. Moreover, in vitro experiments demonstrated that PRL2 binds to GTPase Rac and regulates ROS production. Rac GTPases were more active in PRL2 (KO) phagocytes than in wild type cells after bacterium infection. Our findings indicate that PRL2 senses ROS at inflamed sites and regulates ROS production in phagocytes. This positive feedback mechanism promotes bactericidal activity of phagocytes and may play an important role in innate anti-bacterial immunity.

The present study aimed to investigate the expression of microRNA (miRNA) 26a in blood mononuclear cells and serum in neonatal sepsis, as well as its role in the disease pathogenesis. In total 28 cases of neonatal sepsis were included in the study. The mRNA expression levels of miRNA-26a and interleukin (IL)-6 in the blood mononuclear cells and serum samples were detected by reverse transcription-quantitative polymerase chain reaction. The protein expression of IL-6 was detected by western blot analysis and ELISA. The in vitro septic environment was simulated by lipopolysaccharide (LPS) in THP-1 cells, and the expression of miRNA-26a and IL-6 were determined. Interaction between miRNA-26a and IL-6 was confirmed by a dual-luciferase reporter assay. Compared with the control group, the mRNA and protein expression levels of IL-6 in the blood mononuclear cells and serum samples from the neonates with sepsis were significantly elevated, while the expression of miRNA-26a was significantly decreased. In addition, similar results were observed in the LPS-induced septic models in THP-1 cells. Furthermore, the results of the dual-luciferase reporter assay demonstrated that IL-6 was the direct target of miRNA-26a. The expression of IL-6 was significantly upregulated in the blood mononuclear cells and serum in neonatal sepsis, which may be associated with the downregulation of miRNA-26a. miRNA-26a may regulate the disease pathogenesis and immune responses.

Nowadays, quantitative structure–activity relationship (QSAR) methods have been widely performed to predict the toxicity of compounds to organisms due to their simplicity, ease of implementation, and low hazards. In this study, to estimate the toxicities of substituted aromatic compounds to Tetrahymena pyriformis, the QSAR models were established by the multiple linear regression (MLR) and radial basis function neural network (RBFNN). Unlike other QSAR studies, according to the difference of functional groups (−NO2, −X), the whole dataset was divided into three groups and further modeled separately. The statistical characteristics for the models are obtained as the following: MLR: n = 36, R2 = 0.829, RMS (root mean square) = 0.192, RBFNN: n = 36, R2 = 0.843, RMS = 0.167 for Group 1; MLR: n = 60, R2 = 0.803, RMS = 0.222, RBFNN: n = 60, R2 = 0.821, RMS = 0.193 for Group 2; MLR: n = 31 R2 = 0.852, RMS = 0.192; RBFNN: n = 31, R2 = 0.885, RMS = 0.163 for Group 3, respectively. The results were within the acceptable range, and the models were found to be statistically robust with high external predictivity. Moreover, the models also gave some insight on those characteristics of the structures that most affect the toxicity.

A Gram-stain-negative, aerobic, rod-shaped, motile bacterium with a subpolar flagellum, designated strain CCMM005T, was isolated from offshore seawater at Qingdao, China. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain CCMM005T belonged to the genus Salinisphaera and exhibited highest 16S rRNA gene sequence similarity to Salinisphaera dokdonensis CL-ES53T (96.9 %). It showed lower sequence similarities (94.9–96.4 %) with all other representatives of the genus Salinisphaera . Optimal growth occurred in the presence of 4 % (w/v) NaCl, at 30 °C and at pH 7.0. The polar lipids of strain CCMM005T consisted of phosphatidylethanolamine, phosphatidylcholine, phosphatidylglycerol, one unidentified phosphoglycolipid and one unidentified phospholipid. The predominant isoprenoid quinone was Q-8. The major fatty acids were C19 : 0cyclo ω8c, C18 : 0 and C18 : 1ω7c. The DNA G+C content of strain CCMM005T was 65.3 mol%. On the basis of data from this polyphasic study, strain CCMM005T is considered to represent a novel species of the genus Salinisphaera , for which the name Salinisphaera aquimarina sp. nov. is proposed. The type strain is CCMM005T (=MCCC 1K03246T=KCTC 52640T).

In tumors, vascular endothelial growth factor (VEGF) contributes to angiogenesis, vascular permeability, and tumorigenesis. In our previous study, we found that α-solanine, which is widespread in solanaceae, has a strong anti-cancer effect under normoxia. However, it is unknown whether α-solanine has a similar effect under hypoxia. We used cobalt chloride (CoCl2) to mimic hypoxia in vitro. HIF-1α, which is almost undetectable under normoxia, was significantly increased. Simultaneously, another regulator of VEGF, STAT3, was also significantly activated by CoCl2. We utilized α-solanine in co-culture with CoCl2. α-solanine decreased the expression of VEGF and loss of E-cadherin. α-solanine also suppressed the activation of phospho-ERK1/2 (p-ERK1/2), HIF-1α, and STAT3 signaling. The results provide new evidence that α-solanine has a strong anti-cancer effect via the ERK1/2-HIF-1α and STAT3 signaling pathways and suggest that it may be a potential new drug.

To quantify regional sources contributing to submicron particulate matter (PM1) pollution in haze episodes, on-line measurements combining two modeling methods, namely, positive matrix factorization (PMF) and backward Lagrangian particle dispersion modeling (LPDM), were conducted for the period of one month in urban Nanjing, a city located in the western part of Yangtze River Delta (YRD) region of China. Several multi-day haze episodes were observed in December 2013. Long-range transport of biomass burning from the southwestern YRD region largely contributed to PM1 pollution with more than 25% of total organics mass in a lasting heavy haze. The LPDM analysis indicates that regional transport is a main source contributing to secondary low-volatility production. The high-potential source regions of secondary low-volatility production are mainly located in areas to the northeast of the city. High aerosol pollution was mainly contributed by regional transport associated with northeastern air masses. Such regional transport on average accounts for 46% of total NR-PM1 with sulfate and aged low-volatility organics being the largest fractions (>65%).

Aflatoxins B1 (AFB1), deoxynivalenol (DON), fumonisin B1 (FB1), T-2 toxin (T-2), and zearalenone (ZEA) are the major foodborne mycotoxins of public health concerns. In the present study, the multiple toxic endpoints of these naturally-occurring mycotoxins were evaluated in Caenorhabditis elegans model for their lethality, toxic effects on growth and reproduction, as well as influence on lifespan. We found that the lethality endpoint was more sensitive for T-2 toxicity with the EC50 at 1.38 mg/L, the growth endpoint was relatively sensitive for AFB1 toxic effects, and the reproduction endpoint was more sensitive for toxicities of AFB1, FB1, and ZEA. Moreover, the lifespan endpoint was sensitive to toxic effects of all five tested mycotoxins. Data obtained from this study may serve as an important contribution to knowledge on assessment of mycotoxin toxic effects, especially for assessing developmental and reproductive toxic effects, using the C. elegans model.

To investigate the composition, sources and evolution processes of submicron aerosol during wintertime, a field experiment was conducted during December 1–31, 2013 in urban Nanjing, a megacity in Yangtze River Delta of China. Non-refractory submicron aerosol (NR-PM1) species were measured with an Aerodyne Aerosol Chemical Speciation Monitor. NR-PM1 is dominated by secondary inorganic aerosol (55%) and organic aerosol (OA, 42%) during haze periods. Six OA components were identified by positive matrix factorization of the OA mass spectra. The hydrocarbon-like OA and cooking-related OA represent the local traffic and cooking sources, respectively. A highly oxidized factor related to biomass burning OA accounted for 15% of the total OA mass during haze periods. Three types of oxygenated OA (OOA), i.e., a less-oxidized OOA (LO-OOA), a more-oxidized OOA (MO-OOA), and a low-volatility OOA (LV-OOA), were identified. LO-OOA is likely associated with fresh urban secondary OA. MO-OOA likely represents photochemical products showing a similar diurnal cycle to nitrate with a pronounced noon peak. LV-OOA appears to be a more oxidized factor with a pronounced noon peak. The OA composition is dominated by secondary species, especially during haze events. LO-OOA, MO-OOA and LV-OOA on average account for 11%, (18%), 24% (21%) and 23% (18%) of the total OA mass for the haze (clean) periods respectively. Analysis of meteorological influence suggested that regional transport from the northern and southeastern areas of the city is responsible for large secondary and low-volatility aerosol formation.

This trial was designed to compare the efficacy and safety between epirubicin (E) and carboplatin (C) in combination with paclitaxel (P) and trastuzumab (H) in neoadjuvant setting. In 13 Chinese cancer centers, 100 patients with HER2-positive, locally advanced breast cancer were 1:1 randomized to receive medication as follows: trastuzumab and paclitaxel weekly combined with carboplatin weekly for PCH group, or epirubicin every 3 weeks for PEH group. Patients were given 4 to 6 cycles of chemotherapy. The primary endpoint was pathologic complete response (pCR) rate, which was no significant difference in PCH and PEH regimen (39.1% vs. 48.8%; p=0.365). However, PEH regimen achieved higher pCR in luminal-B (HER2-poitive) subgroup (55.0% vs. 24.0%; p = 0.033), but not in ERBB2+ subgroup (42.9% vs. 57.1%; p = 0.355). PEH regimen showed a favorable efficacy in PIK3CA mutated subgroup (69.2% vs.23.5%, p=0.012). No significant difference was observed in the subgroup analysis of TP53 mutation status, PTEN expression, FCGR2A SNP and FCGR3A SNP. Both regimens as neoadjuvant chemotherapy achieve similar efficacy and safety. PEH might improve pCR rate, especially in the luminal-B subtype and PIK3CA mutation subtype. PEH is feasible and less likely to increase the incidence of acute cardiac events compared to PCH.
