Increased adoption of crops with stacked traits conferring glyphosate and dicamba resistance, and recent confirmation of kochia [Bassia scoparia (L.) A.J. Scott] biotypes resistant to these herbicides in Alberta and Saskatchewan, warrant surveillance of herbicide-resistant kochia in Manitoba. A randomized–stratified survey of 315 sites in Manitoba was conducted in the fall of 2018. Overall, 58% of the kochia populations tested were glyphosate-resistant, while 1% were dicamba-resistant. This survey documents rapid increase in glyphosate-resistant kochia over a five-year time frame, and also confirms the first cases of kochia in Manitoba with dicamba resistance alone and in combination with glyphosate resistance.

Recent confirmation of dicamba-resistant kochia [Bassia scoparia (L.) A.J. Scott] in Alberta warrants investigation of resistance to other commonly used synthetic auxin herbicides like fluroxypyr. A randomized-stratified survey of 305 sites in Alberta was conducted in 2017 to determine the status of fluroxypyr-resistant kochia. Overall, 13% of the kochia populations were fluroxypyr-resistant. Only 4% of the populations were both fluroxypyr- and dicamba-resistant, indicating that different mechanisms may confer resistance to these herbicides. When combined with estimates of dicamba resistance, about 28% of the kochia populations sampled in Alberta in 2017 were resistant to at least one synthetic auxin herbicide.

Recent confirmations of glyphosate-resistant Russian thistle (Salsola tragus L.) in Montana, Washington, and Oregon, warrant greater surveillance of herbicide-resistant Russian thistle in western Canada. A randomized-stratified survey of 315 sites in Manitoba was conducted in 2018 to determine the incidence of herbicide resistance in Russian thistle and other weeds sampled post-harvest. Russian thistle populations were collected from 14 of the 315 sites surveyed. None of these populations exhibited resistance to acetolactate synthase inhibitors (tribenuron/thifensulfuron), synthetic auxins (2,4-D ester or fluroxypyr), or glyphosate. This Manitoba survey of herbicide-resistant Russian thistle serves as a baseline for future surveillance efforts.

Herbicide-resistant (HR) kochia is a growing problem in the Great Plains region of Canada and the United States. Resistance to up to four herbicide sites of action, including photosystem II inhibitors, acetolactate synthase inhibitors, synthetic auxins, and the 5-enolpyruvylshikimate-3-phosphate synthase inhibitor glyphosate have been reported in many areas of this region. Despite being present in the United States since 1993/1994, auxinic-HR kochia is a recent and growing phenomenon in Canada. This study was designed to characterize 1) the level of resistance and 2) patterns of cross-resistance to dicamba and fluroxypyr in 12 putative auxinic-HR kochia populations from western Canada. The incidence of dicamba-resistant individuals ranged among populations from 0% to 85%, while fluroxypyr-resistant individuals ranged from 0% to 45%. In whole-plant dose-response bioassays, the populations exhibited up to 6.5-fold resistance to dicamba and up to 51.5-fold resistance to fluroxypyr based on visible injury 28 d after application. Based on plant survival estimates, the populations exhibited up to 3.7-fold resistance to dicamba and up to 72.5-fold resistance to fluroxypyr. Multiple patterns of synthetic auxin resistance were observed, in which one population from Cypress County, Alberta, was resistant to dicamba but not fluroxypyr, whereas another from Rocky View County, Alberta, was resistant to fluroxypyr but not dicamba based on single-dose population screening and dose-response bioassays. These results suggest that multiple mechanisms may confer resistance to dicamba and/or fluroxypyr in Canadian kochia populations. Further research is warranted to determine these mechanisms. Farmers are urged to adopt proactive nonchemical weed management practices in an effort to preserve efficacy of the remaining herbicide options available for control of HR kochia.

Physicochemical descriptors commonly used to define "drug-likeness" and ligand efficiency measures are assessed for their ability to differentiate marketed drugs from compounds reported to bind to their efficacious target or targets. Using ChEMBL version 26, a data set of 643 drugs acting on 271 targets was assembled, comprising 1104 drug-target pairs having ≥100 published compounds per target. Taking into account changes in their physicochemical properties over time, drugs are analyzed according to their target class, therapy area, and route of administration. Recent drugs, approved in 2010-2020, display no overall differences in molecular weight, lipophilicity, hydrogen bonding, or polar surface area from their target comparator compounds. Drugs are differentiated from target comparators by higher potency, ligand efficiency (LE), lipophilic ligand efficiency (LLE), and lower carboaromaticity. Overall, 96% of drugs have LE or LLE values, or both, greater than the median values of their target comparator compounds.

Guanidinoglycosides are a class of non-cytotoxic molecular transporters capable of delivering high molecular weight bioactive cargos into cells at low nanomolar concentrations. Efficient bioconjugation with guanidinoglycosides has been previously demonstrated by utilizing a guanidinoneomycin decorated with a reactive but also unstable N-hydroxysuccinimmide ester-containing linker. Herein we report the synthesis, chemistry, and application of a new, stable guanidinoneomycin derivative armed with a highly specific maleimide moiety which allows for thiol-maleimide click chemistry, a highly popular bioconjugation strategy, widening the field of application of these intriguing and useful delivery vehicles.

Supplemental Digital Content is available in the text., Abnormal uteroplacental remodeling leads to placental hypoperfusion, causing fetal growth restriction and pregnancy-related hypertension, which are associated with endothelial dysfunction and markers of reduced vascular NO bioavailability and oxidative stress. Tetrahydrobiopterin (BH4) is a redox cofactor for eNOS (endothelial NO synthase) with a required role in NO generation. Using mice models and human samples, we investigated the physiological requirement for endothelial cell BH4 in uteroplacental vascular adaptation and blood pressure regulation to pregnancy. In pregnant mice, selective maternal endothelial BH4 deficiency resulting from targeted deletion of Gch1 caused progressive hypertension during pregnancy and fetal growth restriction. Maternal endothelial cell Gch1 deletion caused defective functional and structural remodeling in uterine arteries and in spiral arteries, leading to placental insufficiency. Using primary endothelial cells isolated from either normal or hypertensive pregnancies, we found that hypertensive pregnancies are associated with reduced endothelial cell BH4 levels, impaired eNOS activity, and reduced endothelial cell proliferation, mediated by reduced GTPCH (GTP cyclohydrolase 1) protein. In rescue experiments, high blood pressure and fetal growth restriction in pregnant endothelial cell Gch1 deficient mice was not rescued by oral BH4 supplementation, due to systemic oxidation of BH4 to dihydrobiopterin. However, the fully reduced folate, 5-methyltetrahydrofolate prevented BH4 oxidation, reduced blood pressure to normal levels, and normalized fetal growth. We identify a critical requirement for maternal endothelial cell BH4 biosynthesis in uteroplacental vascular remodeling in pregnancy. Restoration of endothelial cell BH4 with reduced folates identifies a novel therapeutic target for the prevention and treatment of pregnancy-related hypertension such as preeclampsia.

Pepper yellow leaf curl virus (PYLCV) is a threat to chili plants and can significantly reduce yields. This study aimed as a pilot project to detect PYLCV by analyzing compounds emitted by chili plants using gas chromatography-mass spectrometry (GC-MS). The samples investigated in this research were PYLCV-infected and PYLCV-undetected chili plants taken from commercial chili fields. The infection status was validated by using a polymerase chain reaction (PCR) test. A headspace technique was used to extract the volatile organic compounds emitted by plants. The analysis of GC-MS results began with pre-processing, analyzing sample compound variability with a boxplot analysis, and sample classification by using a multivariate technique. Unsupervised multivariate technique principal component analysis (PCA) was performed to discover whether GC-MS could identify PYLCV-infected or not. The results showed that PYLCV-infected and PYLCV-undetected chili plants could be differentiated, with a total percent variance of the first three principal components reaching 91.32%, and successfully discriminated between PYLCV-infected and PYLCV-undetected chili plants. However, more comprehensive studies are needed to find the potential biomarkers of the infected plants.

This report updates the incidence of herbicide-resistant (HR) weeds across western Canada from the last report covering 2007 to 2011. This third round of preharvest surveys was conducted in Saskatchewan in 2014 and 2015, Manitoba in 2016, and Alberta in 2017, totaling 798 randomly selected cropped fields across 28 million ha. In addition, we screened 1,108 weed seed samples submitted by prairie growers or industry between 2012 and 2016. Of 578 fields where wild oat seed was collected, 398 (69%) had an HR biotype: 62% acetyl-CoA carboxylase inhibitor (WSSA Group 1) resistant, 34% acetolactate synthase inhibitor (Group 2) resistant, and 27% Group 1+2 resistant (vs. 41%, 12%, and 8%, respectively, in the previous second-round surveys from 2007 to 2009). The sharp increase in Group 2 resistance is the result of reliance on this site of action to manage Group 1 resistance and the resultant increased selection pressure. There are no POST options to control Group 1+2–HR wild oat in wheat or barley. The rise of Group 2 resistance in green foxtail (11% of sampled fields) and yellow foxtail (17% of Manitoba fields), which was not detected in the previous survey round, parallels the results for wild oat resistance. Various Group 2–HR populations of broadleaf weeds were confirmed, with cleavers and field pennycress being most abundant. Results of submission-sample testing reflected survey results. Although not included in this study, a postharvest survey in Alberta in 2017 indicated widespread Groups 2, 4 (dicamba), and 9 (glyphosate) resistance in kochia and Group 2 resistance in Russian thistle. These surveys bring greater awareness of HR weeds to growers and land managers at local and regional levels, and highlight the urgency to preserve herbicide susceptibility in our key economic weed species.

A survey was conducted in Alberta in 2017 to determine the distribution and abundance of acetolactate synthase (ALS) inhibitor and glyphosate resistance in Russian thistle. Of 45 populations tested, 31 (62%) were ALS inhibitor resistant. No populations exhibited resistance to glyphosate. This survey serves as a baseline to monitor future incidence of resistance in this weed.

A randomized stratified survey was conducted in Alberta in 2017 to determine the distribution and abundance of multiple-resistant [acetolactate synthase (ALS) inhibitor, glycine, and synthetic auxin] kochia. All populations were ALS inhibitor resistant, with glyphosate and dicamba resistance confirmed in 50% and 18% of populations, respectively. Ten percent of populations exhibited resistance to all three mode-of-action herbicides.

We report the determination of ammonia (NH3) diffusive sampling rates for six different designs of commercial diffusive samplers (CEH ALPHA sampler, Gradko diffusion tube, Gradko DIFRAM-400, Passam ammonia sampler, and ICS Maugeri Radiello radial sampler (blue and white turbulence barriers)), together with the validation test results for a pumped sampler (CEH DELTA denuder). The devices were all exposed in the UK's National Physical Laboratory's (NPL) controlled atmosphere test facility (CATFAC). For each of the seven diffusive sampler exposure tests there were traceable concentrations of ammonia (in the range 3–25 μg m−3) generated under well-defined conditions of temperature, relative humidity and wind speed, which are applicable to a variety of ambient monitoring environments. The sampler exposure time at each concentration was 28 days, except for the radial devices, which were exposed for 14 days. The work relied on the dilution of newly developed stable Primary Standard Gas Mixtures (PSMs) prepared by gravimetry in passivated gas cylinders as a method of improving the metrological traceability of ammonia measurements. The exposed diffusive samplers were sent blind to the participants for analysis and the reported NH3 concentrations were then compared against the known reference concentration. From the results for each sampler type a diffusive sampling rate was calculated and compared against the rate used routinely by the participants. Some measurement results were in good agreement with the known traceable reference concentration (particularly for one diffusive sampler design (ALPHA)), while other devices exhibited over-reading and under-reading (each with a clear bias). The new diffusive sampling rates determined in the laboratory study were then applied to measurements in a field comparison campaign, and this was found to deliver an improvement in agreement between the different devices deployed.

Excessive expansion of adipose tissue in obesity typically leads to overflow and accumulation of lipids in other tissues, causing fatty liver disease and atherosclerosis. The intracellular protein, phosphoprotein enriched in astrocytes (PEA)-15 has been linked to metabolic disease but its role in lipid storage has not been examined. To delineate the role of PEA-15 in adipose tissue, we placed PEA-15−/− mice on a high fat diet. These mice developed increased body weight and greater white adipose tissue expansion compared to high fat diet-fed wild type mice. This was due to increased adipocyte cell size in PEA-15−/− mice consistent with greater lipid storage capacity. Surprisingly, PEA-15−/− mice exhibited improvements in whole body insulin sensitivity, lower hepatic weight and decreased serum triglycerides indicating a protective phenotype. To determine effects on atherosclerosis, PEA-15−/− mice were crossed with the ApoE−/− mice on a high fat diet. Strikingly, these mice were protected from atherosclerosis and had less hepatic lipid accumulation despite increased adiposity. Therefore, we reveal for the first time that PEA-15 plays a novel role in regulating the expansion of adipose tissue. Decreasing PEA-15 expression increases the sequestering of lipids in adipose tissue, protecting other tissues in obesity, thereby improving metabolic health.

Anthelmintic resistance threatens the sustainability of sheep production globally. Advice regarding strategies to reduce the development of anthelmintic resistance incorporates the outcomes of modelling exercises. Further understanding of gastrointestinal nematode species diversity, and population dynamics and genetics (which may vary between species) is required to refine these models; and field studies combining faecal egg outputs, species composition and resistance genetics are needed to calibrate them. In this study, faecal samples were taken from ewes and lambs on a commercial farm in south-eastern Scotland at approximately 3 t-4 week intervals between spring and autumn over a period of 4 years. Faecal egg counts were performed on these samples, and L3 were collected from pooled coprocultures. Deep amplicon sequencing was used to determine both the species composition of these L3 and the proportions of benzimidazole-resistant single nucleotide polymorphisms in the isotype-1 β-tubulin locus of the predominant species, Teladorsagia circumcincta L3. Despite consistent management throughout the study, the results show variation in gastrointestinal nematode species composition with time and between age groups, that was potentially associated with weather conditions. The F200Y benzimidazole resistance mutation is close to genetic fixation in the T. circumcincta population on this farm. There was no evidence of variation in isotype-1 β-tubulin single nucleotide polymorphisms frequency between age groups, and no genetic evidence of reversion to benzimidazole susceptibility, despite targeted benzimidazole usage. This study highlights the need to include speciation when investigating gastrointestinal nematode epidemiology and anthelmintic resistance, and serves as an example of how genetic data may be analysed alongside species diversity and faecal egg counts, when markers for other anthelmintic classes are identified.

Ammonia and ammonium have received less attention than other forms of air pollution, with limited progress in controlling emissions at UK, European and global scales. By contrast, these compounds have been of significant past interest to science and society, the recollection of which can inform future strategies. Sal ammoniac ( nūshādir , nao sha ) is found to have been extremely valuable in long-distance trade ( ca AD 600–1150) from Egypt and China, where 6–8 kg N could purchase a human life, while air pollution associated with nūshādir collection was attributed to this nitrogen form. Ammonia was one of the keys to alchemy—seen as an early experimental mesocosm to understand the world—and later became of interest as ‘alkaline air’ within the eighteenth century development of pneumatic chemistry. The same economic, chemical and environmental properties are found to make ammonia and ammonium of huge relevance today. Successful control of acidifying SO 2 and NO x emissions leaves atmospheric NH 3 in excess in many areas, contributing to particulate matter (PM 2.5 ) formation, while leading to a new significance of alkaline air, with adverse impacts on natural ecosystems. Investigations of epiphytic lichens and bog ecosystems show how the alkalinity effect of NH 3 may explain its having three to five times the adverse effect of ammonium and nitrate, respectively. It is concluded that future air pollution policy should no longer neglect ammonia. Progress is likely to be mobilized by emphasizing the lost economic value of global N emissions ($200 billion yr −1 ), as part of developing the circular economy for sustainable nitrogen management. Production of hydroxyl (OH) radicals is frequently dominated by the photolysis of tropospheric ozone (O3). However, photolysis of nocturnal radical reservoirs, such as nitrous acid (HONO) and nitryl chloride (ClNO2), also produces radicals (OH and Cl atoms) that contribute to the oxidising capacity of the local atmosphere, and initiate many radical-chain reactions that lead to the formation of harmful secondary pollutants. Photolysis of nitric acid (HNO3) is also a minor radical production mechanism. In this paper, locally representative photolysis rate constants (j-values) for these molecules are shown to be critical for quantifying and understanding the rate of radical production in a local atmosphere. The first long-term 4-π filter radiometer dataset in the UK (21 November 2018–20 November 2019) available for direct atmospheric model validation is reported. Measurements were made at Auchencorth Moss, a Scottish rural background site, and j(NO2) is used to generate a measurement-driven adjustment factor (MDAF) for calculated j-values that accounts for local changes in meteorological variables without significantly increasing computational cost. Modelled clear-sky j-values and actinic flux for Auchencorth Moss were generated using the Tropospheric Ultraviolet and Visible radiation model (TUV; v.5.3.1). Applying the MDAF metric resulted in the calculated photolytic production rate of OH radicals, from all sources considered, being ~40 % lower over the year. Photolysis of HONO resulted in an increased rate of OH production compared to that from O3 in low-light conditions, such as sunrise and sunset (Solar Zenith Angle > 80°). Hydroxyl radical production from HONO photolysis exceeded that from O3 consistently throughout the day during the winter and autumn (by a factor of 5 and 2.1, respectively). Radical production rates from HONO and ClNO2 reached maximum values during the early morning hours of summer (06:00–09:00 UTC), with OH produced at a rate of 1.06 × 106 OH radicals cm−3 s−1, and Cl radicals at 3.20 × 104 Cl radicals cm−3 s−1, with the MDAF metric applied. This first application of the MDAF j-values demonstrates an efficient measurement and computational approach to improve modelling of the local atmospheric photochemistry that drives NO2, O3 and PM pollution levels. The incorporation of local radiation measurements in measurement networks, and the consequent greater spatial resolution of locally-relevant photolysis coefficients in model photolysis parameterisations, will improve the accuracy of assessment of air pollution and policy-intervention impacts.

The practices and tactics employed in successful optimizations are examined, judged from the trajectories of ligand efficiency and property evolution. A wide range of targets is analyzed, encompassing a variety of hit finding methods (HTS, fragments, encoded library technology) and types of molecules, including those beyond the rule of five. The wider employment of efficiency metrics and lipophilicity control is evident in contemporary practice and the impact on quality demonstrable. What is clear is that while targets are different, successful molecules are almost invariably among the most efficient for their target, even at the extremes. Trajectory mapping, based on principles rather than rules, is useful in assessing quality and progress in optimizations while benchmarking against competitors and assessing property-dependent risks.

Mixed mono- and tributyrate glycerides have been used for effective delivery of butyrate to the gut to benefit broilers. However, limited information is available on the efficacy of butyrate glycerides individually and in combination with different levels and feeding schedules. The present study has first investigated the effects of monobutyrin at inclusion levels of zero, 500, 1,000, 2,000, and 3,000 ppm on the performance of broilers, and second, the effects of its combination with tributyrin. In the monobutyrin trial, there were no overall significant differences in average daily gain or feed efficiency. However, 2,000 ppm birds had significantly decreased abdominal fat deposition compared to controls (P ≤ 0.05), and the breast muscle deposition increased in a dose-response manner to the supplementation of monobutyrin (P ≤ 0.05). The combination trial tested 5 treatment groups: control, 500 ppm tributyrin + 500 ppm monobutyrin (5T5M), 500 ppm tributyrin + 500 ppm monobutyrin staggered (5T5Ms), 500 ppm tryibutyrin + 2,000 ppm monobutyrin (5T20M), or 500 ppm tributyrin + 2,000 ppm monobutyrin staggered (5T20Ms). In staggered groups, birds were fed tributyrin for one wk followed by 2 wk of monobutyrin, after which the feed was butyrate glyceride free. The non-staggered groups had constant inclusions levels through the 5 weeks. There were no significant differences in average daily gain or feed efficiency among groups. At 5 wk of age, all treatment groups except for 5T5Ms had significantly lower relative abdominal fat weight compared to control birds (P ≤ 0.05), although 5T5Ms birds demonstrated a trend for a decrease (P = 0.095). Relative breast muscle weight was significantly increased only in 5T5M birds over control birds at 5 wk of age (P ≤ 0.05). Serum biochemistry revealed significant changes in factors relating to muscle growth and fat deposition (P ≤ 0.05). These results indicate a consistent shift in lipid metabolism with the addition of butyrate glycerides and that the deposition of breast muscle may be highest with the incorporation of butyrate glycerides at a moderate level for the duration of development.

International audience; In this study, we present the first long-term N2O eddy covariance dataset measured from a working farm. The eddy covariance method was used over a four year period to measure fluxes of the greenhouse gas nitrous oxide (N2O) from an intensively managed grazed grassland, to which regular applications of ammonium nitrate or urea fertilisers were spread, for two years each at the field site. The mean emission factors (EFs) reported for ammonium nitrate and urea fertiliser applications in this study over a period of 30 days after fertilisation, were 0.90 and 1.73% of the nitrogen applied, respectively, with EFs of individual events ranging between 0.13 and 5.71%. Our study accurately quantifies emission factors for multiple events and showing unambiguously that large-scale variability is real. EFs do indeed vary from one fertiliser event to another, even at the same site with the same fertiliser type under similar environmental conditions. This makes distinguishing EFs between different fertiliser types for the purposes of developing emission mitigation policy very difficult.

Live-cell flow cytometry is increasingly used among cell biologists to quantify biological processes in a living cell culture. This protocol describes a method whereby live-cell flow cytometry is extended upon to analyze the multiple functions of P2X7 receptor activation in real-time. Using a time module installed on a flow cytometer, live-cell functionality can be assessed and plotted over a given time period to explore the kinetics of calcium influx, transmembrane pore formation, and phagocytosis. This simple method is advantageous as all three canonical functions of the P2X7 receptor can be assessed using one machine, and the gathered data plotted over time provides information on the entire live-cell population rather than single-cell recordings often obtained using technically challenging patch-clamp methods. Calcium influx experiments use a calcium indicator dye, while P2X7 pore formation assays rely on ethidium bromide being allowed to pass through the transmembrane pore formed upon high agonist concentrations. Yellow-green (YG) latex beads are utilized to measure phagocytosis. Specific agonists and antagonists are applied to investigate the effects of P2X7 receptor activity. Individually, these methods can be modified to provide quantitative data on any number of calcium channels and purinergic and scavenger receptors. Taken together, they highlight how the use of real-time live-cell flow cytometry is a rapid, cost-effective, reproducible, and quantifiable method to investigate P2X7 receptor function.

Clustering – the automated grouping of similar data – can provide powerful and unique insight into large and complex data sets, in a fast and computationally efficient manner. While clustering has been used in a variety of fields (from medical image processing to economics), its application within atmospheric science has been fairly limited to date, and the potential benefits of the application of advanced clustering techniques to climate data (both model output and observations) has yet to be fully realised. In this paper, we explore the specific application of clustering to a multi-model climate ensemble. We hypothesise that clustering techniques can provide (a) a flexible, data-driven method of testing model–observation agreement and (b) a mechanism with which to identify model development priorities. We focus our analysis on chemistry–climate model (CCM) output of tropospheric ozone – an important greenhouse gas – from the recent Atmospheric Chemistry and Climate Model Intercomparison Project (ACCMIP). Tropospheric column ozone from the ACCMIP ensemble was clustered using the Data Density based Clustering (DDC) algorithm. We find that a multi-model mean (MMM) calculated using members of the most-populous cluster identified at each location offers a reduction of up to ∼ 20 % in the global absolute mean bias between the MMM and an observed satellite-based tropospheric ozone climatology, with respect to a simple, all-model MMM. On a spatial basis, the bias is reduced at ∼ 62 % of all locations, with the largest bias reductions occurring in the Northern Hemisphere – where ozone concentrations are relatively large. However, the bias is unchanged at 9 % of all locations and increases at 29 %, particularly in the Southern Hemisphere. The latter demonstrates that although cluster-based subsampling acts to remove outlier model data, such data may in fact be closer to observed values in some locations. We further demonstrate that clustering can provide a viable and useful framework in which to assess and visualise model spread, offering insight into geographical areas of agreement among models and a measure of diversity across an ensemble. Finally, we discuss caveats of the clustering techniques and note that while we have focused on tropospheric ozone, the principles underlying the cluster-based MMMs are applicable to other prognostic variables from climate models.

Very short‐lived substances (VSLS), including dichloromethane (CH2Cl2), chloroform (CHCl3), perchloroethylene (C2Cl4), and 1,2‐dichloroethane (C2H4Cl2), are a stratospheric chlorine source and therefore contribute to ozone depletion. We quantify stratospheric chlorine trends from these VSLS (VSLCltot) using a chemical transport model and atmospheric measurements, including novel high‐altitude aircraft data from the NASA VIRGAS (2015) and POSIDON (2016) missions. We estimate VSLCltot increased from 69 (±14) parts per trillion (ppt) Cl in 2000 to 111 (±22) ppt Cl in 2017, with >80% delivered to the stratosphere through source gas injection, and the remainder from product gases. The modeled evolution of chlorine source gas injection agrees well with historical aircraft data, which corroborate reported surface CH2Cl2 increases since the mid‐2000s. The relative contribution of VSLS to total stratospheric chlorine increased from ~2% in 2000 to ~3.4% in 2017, reflecting both VSLS growth and decreases in long‐lived halocarbons. We derive a mean VSLCltot growth rate of 3.8 (±0.3) ppt Cl/year between 2004 and 2017, though year‐to‐year growth rates are variable and were small or negative in the period 2015–2017. Whether this is a transient effect, or longer‐term stabilization, requires monitoring. In the upper stratosphere, the modeled rate of HCl decline (2004–2017) is −5.2% per decade with VSLS included, in good agreement to ACE satellite data (−4.8% per decade), and 15% slower than a model simulation without VSLS. This study evaluated the interactive effects of combined phosphorus (P) and potassium (K) additions on canopy nitrogen (N) retention (CNR) and subsequent canopy leaching at a long-term N manipulation site on Whim bog in south Scotland. Ambient deposition is 8 kg N ha-1 year-1 and an additional 8, 24, and 56 kg N ha-1 year-1 of either ammonium (NH4+) or nitrate (NO3-) with or without P and K has been applied over 11 years. Throughfall N deposition below Calluna vulgaris and foliar N and P concentrations were assessed. Results showed that 60% for low dose and 53% for high dose of NO3- contrasting with 80% for low dose and 38% for high dose of NH4+ onto Calluna was retained by Calluna canopy. The CNR was enhanced by P and K addition in which 84% of NO3 - and 83% of NH4+ for high dose were retained. CNR for NO3- increased the canopy leaching of dissolved organic N (DON) and associated organic anions. NH4+ retention increased canopy leaching of magnesium and calcium through ion exchange. Even over 11-years N exposure without P and K, foliage 29 N:P ratio of Calluna did not increase, suggesting that N exposure did not lead to N saturation of Calluna at Whim bog. Our study concluded that increases in P and K availability enhance CNR of Calluna, but accelerate the associated canopy leaching of DON and base cations, depending on foliar N status.

Long-term additions of nitrogen (N) to peatlands have altered bryophyte growth, species dominance, N content in peat and peat water, and often resulted in enhanced Sphagnum decomposition rate. However, these results have mainly been derived from experiments in which N was applied as ammonium nitrate (NH4NO3), neglecting the fact that in polluted areas, wet deposition may be dominated either by NO3- or NH4+. We studied effects of elevated wet deposition of NO3- vs. NH4+ alone (8 or 56 kg N ha(-1) yr(-1) over and above the background of 8 kg N ha(-1) yr(-1) for 5 to 11 years) or combined with phosphorus (P) and potassium (K) on Sphagnum quality for decomposers, mass loss, and associated changes in hummock pore water in an ombrotrophic bog (Whim). Adding N, especially as NH4+, increased N concentration in Sphagnum, but did not enhance mass loss from Sphagnum. Mass loss seemed to depend mainly on moss species and climatic factors. Only high applications of N affected hummock pore water chemistry, which varied considerably over time. Overall, C and N cycling in this N treated bog appeared to be decoupled. We conclude that moss species, seasonal and annual variation in climatic factors, direct negative effects of N (NH4+ toxicity) on Sphagnum production, and indirect effects (increase in pH and changes in plant species dominance under elevated NO3- alone and with PK) drive Sphagnum decomposition and hummock C and N dynamics at Whim. DYRK1A has emerged as a potential target for therapies of Alzheimer's disease using small molecules. On the basis of the observation of selective DYRK1A inhibition by firefly d-luciferin, we have explored static and dynamic structural properties of fragment sized variants of the benzothiazole scaffold with respect to DYRK1A using X-ray crystallography and NMR techniques. The compounds have excellent ligand efficiencies and show a remarkable diversity of binding modes in dynamic equilibrium. Binding geometries are determined in part by interactions often considered "weak", including "orthogonal multipolar" types represented by, for example, F-CO, sulfur-aromatic, and halogen-aromatic interactions, together with hydrogen bonds that are modulated by variation of electron withdrawing groups. These studies show how the benzothiazole scaffold is highly promising for the development of therapeutic DYRK1A inhibitors. In addition, the subtleties of the binding interactions, including dynamics, show how full structural studies are required to fully interpret the essential physical determinants of binding.

Magnesium deficiency is prevalent in women of childbearing age in both developing and developed countries. The need for magnesium increases during pregnancy, and the majority of pregnant women likely do not meet this increased need. Magnesium deficiency or insufficiency during pregnancy may pose a health risk for both the mother and the newborn, with implications that may extend into adulthood of the offspring. The measurement of serum magnesium is the most widely used method for determining magnesium levels, but it has significant limitations that have both hindered the assessment of deficiency and affected the reliability of studies in pregnant women. Thus far, limited studies have suggested links between magnesium inadequacy and certain conditions in pregnancy associated with high mortality and morbidity, such as gestational diabetes, preterm labor, preeclampsia, and small for gestational age or intrauterine growth restriction. This review provides recommendations for further study and improved testing using measurement of red cell magnesium. Pregnant women should be counseled to increase their intake of magnesium-rich foods such as nuts, seeds, beans, and leafy greens and/or to supplement with magnesium at a safe level.

The ability of Sphagnum moss to efficiently intercept atmospheric nitrogen (N) has been assumed to be vulnerable to increased N deposition. However, the proposed critical load (20kgNha(-1)yr(-1)) to exceed the capacity of the Sphagnum N filter has not been confirmed. A long-term (11years) and realistic N manipulation on Whim bog was used to study the N filter function of Sphagnum (Sphagnum capillifolium) in response to increased wet N deposition. On this ombrotrophic peatland where ambient deposition was 8kgNha(-1)yr(-1), an additional 8, 24, and 56kgNha(-1)yr(-1) of either ammonium (NH4(+)) or nitrate (NO3(-)) has been applied for 11years. Nutrient status of Sphagnum and pore water quality from the Sphagnum layer were assessed. The N filter function of Sphagnum was still active up to 32kgNha(-1)yr(-1) even after 11years. N saturation of Sphagnum and subsequent increases in dissolved inorganic N (DIN) concentration in pore water occurred only for 56kgNha(-1)yr(-1) of NH4(+) addition. These results indicate that the Sphagnum N filter is more resilient to wet N deposition than previously inferred. However, functionality will be more compromised when NH4(+) dominates wet deposition for high inputs (56kgNha(-1)yr(-1)). The N filter function in response to NO3(-) uptake increased the concentration of dissolved organic N (DON) and associated organic anions in pore water. NH4(+) uptake increased the concentration of base cations and hydrogen ions in pore water though ion exchange. The resilience of the Sphagnum N filter can explain the reported small magnitude of species change in the Whim bog ecosystem exposed to wet N deposition. However, changes in the leaching substances, arising from the assimilation of NO3(-) and NH4(+), may lead to species change.

Little is known about the micronutrient status of women and children in the Democratic Republic of the Congo, which is critical for the design of effective nutrition interventions. We recruited 744 mother-child pairs from South Kivu (SK) and Kongo Central (KC). We determined hemoglobin (Hb), serum zinc, vitamin B12, folate, ferritin, soluble transferrin receptor (sTfR), retinol binding protein (RBP), C-reactive protein, and α-1 acid glycoprotein concentrations. Anemia prevalence was determined using Hb adjusted for altitude alone and Hb adjusted for both altitude and ethnicity. Anemia prevalence was lower after Hb adjustment for altitude and ethnicity, compared to only altitude, among women (6% vs. 17% in SK; 10% vs. 32% in KC), children 6–23 months (26% vs. 59% in SK; 25% vs. 42% in KC), and children 24–59 months (14% vs. 35% in SK; 23% vs. 44% in KC), respectively. Iron deficiency was seemingly higher with sTfR as compared to inflammation-adjusted ferritin among women (18% vs. 4% in SK; 21% vs. 5% in KC), children 6–23 months (51% vs. 14% in SK; 74% vs. 10% in KC), and children 24–59 months (23% vs. 4% in SK; 58% vs. 1% in KC). Regardless of indicator, iron deficiency anemia (IDA) never exceeded 3% in women. In children, IDA reached almost 20% when sTfR was used but was only 10% with ferritin. Fluxes of carbon monoxide (CO) were measured using a fast-response quantum cascade laser absorption spectrometer and the eddy covariance method at a long-term intensively grazed grassland in southern Scotland. Measurements lasted 20 months from April 2016 to November 2017, during which normal agricultural activities continued. Observed fluxes followed a regular diurnal cycle, peaking at midday and returning to values near zero during the night, with occasional uptake observed. CO fluxes correlated well with the meteorological variables of solar radiation, soil temperature and soil moisture content. Using a general additive model (GAM) we were able to gap fill CO fluxes and estimate annual fluxes of 0.38 ± 0.046 and 0.35 ± 0.045 g C m−2 y−1g C m−2 y−1 for 2016 and 2017, respectively. If the CO fluxes reported in this study are representative of UK grasslands, then national annual emissions could be expected to be in the order of 61.91 (54.3–69.5) Gg, which equates to 3.8% (3.4–4.3%) of the current national inventory total., Highlights • A long term dataset of CO fluxes measured from agricultural grasslands. • A potentially large source of CO emissions is quantified from UK grasslands. • CO emissions from grassland can be modelled using meteorological measurements.

The increasing use of intensive agricultural practices can lead to damaging consequences for the atmosphere through enhanced emissions of air pollutants. However, there are few direct measurements of the surface–atmosphere exchange of trace gases and water-soluble aerosols over agricultural grassland, particularly of reactive nitrogen compounds. In this study, we present measurements of the concentrations, fluxes and deposition velocities of the trace gases HCl, HONO, HNO 3 , SO 2 and NH 3 , and their associated water-soluble aerosol counterparts Cl − , NO 2 − , NO 3 − , SO 4 2− , NH 4 + as determined hourly for one month in May–June 2016 over agricultural grassland pre- and post-fertilisation. Measurements were made using the Gradient of Aerosols and Gases Online Registration (GRAEGOR) wet–chemical two–point gradient instrument. Emissions of NH 3 peaked at 1460 ng m −2 s −1 three hours after fertilisation, with an emission of HONO peaking at 4.92 ng m −2 s −1 occurring five hours after fertilisation. Apparent emissions of NO 3 − aerosol were observed after fertilisation which, coupled with a divergence of HNO 3 deposition velocity ( V d ) from its theoretical maximum value, suggested the reaction of emitted NH 3 with atmospheric HNO 3 to form ammonium nitrate aerosol. The use of the conservative exchange fluxes of tot-NH 4 + and tot-NO 3 − indicated net emission of tot-NO 3 − , implying a ground source of HNO 3 after fertilisation. Daytime concentrations of HONO remained above the detection limit (30 ng m −3 ) throughout the campaign, suggesting a daytime source for HONO at the site. Whilst the mean V d of NH 4 + was with 0.93 mm/s in the range expected for the accumulation mode, the larger average V d for Cl − (3.65 mm/s), NO 3 − (1.97 mm/s), SO 4 2− (1.89 mm/s) reflected the contribution of a super-micron fraction and decreased with increasing PM 2.5 / PM 10 ratio (a proxy measurement for aerosol size), providing direct evidence of a size-dependence of aerosol deposition velocity for aerosol chemical compounds.

Butyrate can modulate the immune response and energy expenditure of animals and enhance intestinal health. The present study investigated changes in the intestinal microbiota composition and serum metabolites of young broilers in response to 3,000 ppm butyrate in the form of butyrate glycerides (BG) via pyrosequencing of bacterial 16S rRNA genes and nuclear magnetic resonance (NMR). The dietary treatment did not affect the alpha diversity of intestinal microbiota, but altered its composition. Thirty-nine key operational taxonomic units (OTUs) in differentiating cecal microbiota community structures between BG treated and untreated chickens were also identified. Bifidobacterium was, in particular, affected by the dietary treatment significantly, showing an increase in not only the abundance (approximately 3 fold, P ≤ 0.05) but also the species diversity. The (NMR)-based analysis revealed an increase in serum concentrations of alanine, low-density and very low-density lipoproteins, and lipids (P ≤ 0.05) by BG. More interestingly, the dietary treatment also boosted (P ≤ 0.05) serum concentrations of bacterial metabolites, including choline, glycerophosphorylcholine, dimethylamine, trimethylamine, trimethylamine-N-oxide, lactate, and succinate. In conclusion, the data suggest the modulation of intestinal microbiota and serum metabolites by BG dietary treatment and potential contribution of intestinal bacteria to lipid metabolism/energy homeostasis in broilers.
