A 37-year-old African–British man was referred to our hospital for detailed examination because of persistent fever, swelling and pain in both ankle joints, and blurred vision for two months. Inguinal lymph node biopsy showed a large number of epithelioid granulomas without necrosis. Granulomatous anterior uveitis, nephropathy, high serum angiotensin-converting enzyme activity, and high serum-soluble interleukin-2 receptor were observed, and the diagnosis of systemic sarcoidosis was made. His serum creatinine was 1.4 mg/dL and hematuria, leukocyturia, and urine protein were also seen. The renal biopsy finding was mesangial proliferative glomerulonephritis, with no findings of granuloma formation or tubular interstitial nephritis. Immunofluorescence staining showed deposition of IgG, C3, and C1q in the mesangial region. IgG3 was dominant in subclass staining. There was no monoclonality on kappa and lambda staining. Electron microscopy showed predominant deposition in the mesangial region with some subepithelial and endothelial deposition. His hematuria and leukocyturia disappeared with steroid therapy, suggesting sarcoidosis-related nephropathy. A case of systemic sarcoidosis with mesangial proliferative glomerulonephritis showing predominant deposition of IgG in the mesangial region is presented. No cases of such histological findings have been reported so far, and it is necessary to analyze further cases to clarify the pathogenic significance of the renal biopsy findings observed in this case.

Objective To avoid risk of infections associated with dental implants, thermal oxidation processes for practical dental Ti alloys were studied for both high bonding strength and antibacterial properties in visible light. Methods Two-step thermal oxidation, comprising carburization (first step of treatment: in Ar-1%CO gas) and subsequent oxidation (second step of treatment: in air), was conducted on commercially pure (CP) Ti, Ti-6Al-4V (Ti64), and Ti-6Al-7Nb (Ti67) alloys to form TiO2 layers. Their bonding strengths and antibacterial properties against Escherichia coli (E. coli) in visible light (λ ≥ 400 nm) were evaluated. Results TiO2 layers formed on each metal were composed of anatase and/or rutile. Anatase fraction and carbon concentration in the layers decreased with increasing temperature in the second step of treatment. Antibacterial properties of the TiO2 layers were dependent on the temperature in the second step of treatment. An approximate antibacterial activity value of 2 (killing ∼99% bacteria) was obtained when the temperatures in the second step of treatment were 673 and 773 K for CP Ti, 773 K for Ti64, and 773 and 873 K for Ti67. It was found that the TiO2 layer must contain carbon and be anatase-rich to exhibit excellent antibacterial properties. Bonding strength between the substrate and TiO2 layers formed at 773 K in the second step of treatment exceeded 80 MPa and was independent of substrate type. Significance TiO2 layers, possessing both high bonding strength and excellent antibacterial properties, were successfully formed on practical dental Ti alloys via two-step thermal oxidation.

Tumor human epidermal growth factor receptor 2 (HER2) status is defined by either protein expression using immunohistochemistry (IHC) or gene amplification using fluorescence in situ hybridization (FISH). Approximately 20% of HER2-positive breast cancer is HER2 IHC 2+/FISH-positive. Unlike trastuzumab, it has not been studied whether the response to trastuzumab emtansine (T-DM1) differs according to HER2-positive status. We retrospectively identified and reviewed medical records of all patients with HER2-positive advanced breast cancer (ABC) who received T-DM1 in our hospital from October 2013 to December 2016. We compared the objective response rate (ORR) and progression-free survival (PFS) between patients in the HER2 IHC 3+ group and those in the HER2 IHC 2+/FISH-positive group. A total of 39 patients (IHC 3+: n = 32; IHC 2+/FISH-positive: n = 7) were analyzed. Nineteen (48.7%), 13 (33.3%), and 29 (74.4%) patients had received at least one prior chemotherapy, more than three lines of chemotherapy, and prior pertuzumab for ABC, respectively. ORR was significantly higher in the IHC 3+ group than in the IHC 2+/FISH-positive group (53.3% vs. 0%, P = 0.024). Median PFS was 7.9 months in the IHC 3+ group versus 3.9 months in the IHC 2+/FISH-positive group (hazard ratio 0.68; 95% confidence interval 0.28-1.69, P = 0.408). Among the HER2-positive ABC patients treated with T-DM1, ORR was significantly worse in HER2 IHC 2+/FISH-positive than in HER2 IHC 3+ patients. Median PFS tended to be shorter in patients with HER2 IHC 2+/FISH-positive.

Different parts of Prunus persica as fruits, flowers, leaves and kernels have been consumed with dietary and therapeutic purposes traditionally. During fruit production, remarkable amount of leaves which can hold important bioactive groups as phenolics, have been left unutilized. the aim of this study was to investigate cytotoxic, antimicrobial and nitric oxide inhibitory activities of supercritical carbondioxide extracts of Prunus persica leaves. Among studied cell lines, supercritical carbon dioxide extract which was processed at 150 bar, 60 degrees C, and 6% co-solvent ethanol, exhibited remarkable cytotoxic activity against HeLa, MPanc-96 and MCF-7 cell lines with IC50 values of 12.22 mu g/ml, 28.17 mu g/ml and 35.51 mu g/ml respectively, whereas IC50 value of conventional solvent extract was above 50 mu g/ml. Minimum inhibitory concentration values determined for antibacterial and antifungal activities against Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecalis, Enterococcus faecium and Candida albicans were found as 62.50 mu g/ml. Strong nitric oxide inhibition was achieved with IC50 of 9.30 mu g/ml. the promising results revealed that Prunus persica leaves may have remarkable potential as supplement both for drug and food industries. This study is the first report revealing cytotoxic, antimicrobial and nitric oxide inhibitory activity of supercritical carbon dioxide extract of Prunus persica leaves.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection induces severe pneumonia and is the cause of a worldwide pandemic. Coronaviruses, including SARS-CoV-2, have RNA proofreading enzymes in their genomes, resulting in fewer gene mutations than other RNA viruses. Nevertheless, variants of SARS-CoV-2 exist and may induce different symptoms; however, the factors and the impacts of these mutations are not well understood. We found that there is a bias to the mutations occurring in SARS-CoV-2 variants, with disproportionate mutation to uracil (U). These point mutations to U are mainly derived from cytosine (C), which is consistent with the substrate specificity of host RNA editing enzymes, APOBECs. We also found the point mutations which are consistent with other RNA editing enzymes, ADARs. For the C-to-U mutations, the context of the upstream uracil and downstream guanine from mutated position was found to be most prevalent. Further, the degree of increase of U in SARS-CoV-2 variants correlates with enhanced production of cytokines, such as TNF-a and IL-6, in cell lines when compared with stimulation by the ssRNA sequence of the isolated virus in Wuhan. Therefore, RNA editing is a factor for mutation bias in SARS-CoV-2 variants, which affects host inflammatory cytokines production.

Morus nigra L. is a beneficial food due to rich phenolic components. While aiming higher yields for bioactive constituents, reduction in terms of raw material, solvent, time and energy gained more importance to provide a sustainable life for human and nature. Microwave assisted extraction (MAE) of Morus nigra fruits was optimized in order to elicit process parameters maximizing bioactive metabolites and tyrosinase inhibitory activity. Spectrophotometry and UPLC-DAD-ESI-MS/MS systems were utilized for quantitative analysis of total phenol, flavonoid and anthocyanin contents. Optimum conditions for MAE were determined as 500 W, 35% ethanol, 10 min yielding 12.63 mg/g cya-3-glu equiv. anthocyanin and IC50 value of 1.60 mg/ml for tyrosinase inhibitory activity. Microwave extracts prevailed better outcomes compared to conventional extraction methods (10.93 mg/g content with IC50 of 2.81 mg/ml). MAE could be considered as an advanced technique to obtain extracts from Morus nigra fruits with higher bioactive content and activity.

We have identified an enzyme, galactolipase (ckGL), which hydrolyzes the acyl ester bond of galactolipids such as digalactosyldiacylglycerol (DGDG), in the microalga Chlorella kessleri. Following purification of the enzyme to electrophoretic homogeneity from cell-free extract, the maximum activity toward DGDG was observed at pH 6.5 and 37 °C. ckGL was Ca2+-dependent enzyme and displayed an apparent molecular mass of approx. 53 kDa on SDS-PAGE. The substrate specificity was in the order: DGDG (100%) > monogalactosyldiacylglycerol ≈ phosphatidylglycerol (~ 40%) > sulfoquinovosyldiacylglycerol (~ 20%); the enzyme exhibited almost no activity toward glycerides and other phospholipids. Gas chromatography analysis demonstrated that ckGL preferably hydrolyzed the sn-1 acyl ester bond in the substrates. The genomic DNA sequence (5.6 kb) containing the ckGL gene (designated glp1) was determined and the cDNA was cloned. glp1 was composed of 10 introns and 11 exons, and the 1608-bp full-length cDNA encoded a mature ckGL containing 475 amino acids (aa), with a presequence (60 aa) containing a potential chloroplast transit peptide. Recombinant functional ckGL was produced in Escherichia coli. Although the deduced aa sequence of ckGL contained the typical GXSXG motif of serine hydrolases together with conserved histidine and aspartate residues which would form part of the catalytic triad of α/β-hydrolases, ckGL showed no significant overall similarity with known lipases including GLs from Chlamydomonas reinhardtii and Aspergillus japonicus, indicating that ckGL is a novel GL. ckGL, with high specificity for DGDG, could be applicable to food processing as an enzyme capable of improving material textures. International audience; Peptide-based inhibitors hold promising potential in the development of antiviral therapy. Here, we investigated the antiviral potential of fragmented viral proteins derived from ribonucleoprotein (RNP) components of the human respiratory syncytial virus (HRSV). Based on a mimicking approach that targets the functional domains of viral proteins, we designed various fragments of nucleoprotein (N), matrix protein M2-1 and phosphoprotein (P) and tested the antiviral activity in an RSV mini-genome system. We found that the fragment comprising residues 130–180 and 212–241 in the C-terminal region of P (81 amino acid length), denoted as P Fr, significantly inhibited the polymerase activity through competitive binding to the full-length P. Further deletion analysis of P Fr suggested that three functional domains in P Fr (oligomerization, L-binding and nucleocapsid binding) are required for maximum inhibitory activity. More importantly, a purified recombinant P Fr displayed significant antiviral activity at low nanomolar range in RSV-infected HEp-2 cells. These results highlight P as an important target for the development of antiviral compounds against RSV and other paramyxoviruses.

Chromium (Cr) is commonly added into various metal alloys to improve some mechanical properties such as corrosion resistance, strength, and workability. However, Cr is also known to be a metal allergen for some individuals. Metal allergy is a T cell-mediated disease with symptoms of inflammation and swelling that involve inflammatory cytokines and prostaglandins. Hence, suppressing these inflammation paths by using COX-2 inhibitor might be useful in treating Cr allergy. In this study, mice were used with Cr-induced allergy challenge model. The mice were injected with celecoxib once per day for 7 days one hour after the challenge. Footpad samples were stained with haematoxylin and eosin (H&E), and lymphocytes were isolated from popliteal lymph nodes for the flow cytometric analysis. The results show that both prostaglandin E2 (PGE2), a known mediator of inflammation, and cyclooxygenases (COX)-2 have important roles in the development of Cr allergy. Further, COX-2 inhibitor, celecoxib, was effective in relieving swelling and inflammation in Cr-allergic mice concordant with suppression of IFN-γ production by CD8+ T cells and T cell accumulation in the lymph nodes. Therefore, the inhibition of COX-2 may be a therapeutic target for Cr allergy, and additional molecules in the PGE2 signalling pathway may also be an effective therapeutic target for the treatment of metal allergy.

Hypericum perforatum L. (Hypericaceae) has been widely prescribed for mild to moderate depression following the release of promising results in clinical trials. However, it is known that its constituents may be affected by milieu. The stability complexities of the constituents of H. perforatum have gained interest in recent years. The aim of the present study was to examine the impact of storage conditions on H. perforatum total extract simultaneously under different storage conditions. Temperature, humidity, and light conditions were evaluated. Comparative analyses of methanol extracts were conducted using high performance liquid chromatography-diode array detection for chlorogenic acid, rutin, hyperoside, isoquercitrin, quercitrin, quercetin, amentoflavone, pseudohypericin, hyperforin, and hypericin. Analysis and extraction were performed using a validated method. The fluctuation of the constituents of the plant extract has been demonstrated. Among these components, chlorogenic acid was the most stable. Hyperforin, hypericin, and pseudohypericin were more stable than the flavonoids at -20°C, in the 6th month. As estimated, decay was lowest at -20°C and highest at 40°C-75% relative humidity for the analyzed constituents. Except for hyperforin, light protection decreased the breakdown of components within 4 months. However, at the 6th month, equivalent changes were seen for all constituents. Degradation of the constituents at -20°C indicates the importance of stability tests in analysis studies covering time and storage conditions. © 2015, Food and Drug Administration, Taiwan. AURKC, a member of the Aurora kinase gene family, is highly expressed in testis but is either moderately expressed or repressed in most somatic cells. Varying expression of AURKC has been observed in human cancers, but the underlying mechanisms of differential expression have been investigated only to a limited extent. We investigated the role of promoter CpG methylation in the regulation of AURKC gene expression in human cancer cells, in relation to a recently reported AURKC transcription repressor PLZF/ZBTB16, implicated in transformation and tumorigenesis. AURKC and PLZF/ZBTB16 expression profiles were investigated in reference to CpG methylation status on the AURKC promoter experimentally, and also in The Cancer Genome Atlas (TCGA) dataset involving multiple cancer types. AURKC promoter showed dense to moderate hypermethylation correlating with low to moderate expression of the gene in normal somatic cells and cancer cell lines, while testis with high expression revealed marked hypo-methylation. Treatment with the demethylating agent, 5-aza-dC, but not the histone deacetylase (HDAC) inhibitor, TSA, led to elevated expression in cancer cell lines, indicating that promoter DNA methylation negatively regulates AURKC expression. High expression of PLZF in PLZF-transfected cells treated with 5-aza-dC only partially repressed expression of AURKC despite 5-aza-dC also inducing elevated PLZF expression. Analyses of the TCGA data showed differential expression of AURKC in multiple cancer types and stronger correlation of AURKC expression with CpG methylation compared to PLZF levels. These findings demonstrate that differential promoter CpG methylation is an important mechanism regulating AURKC expression in cancer cells.

This is the first paper to reveal the formation mechanism of the abnormal growth of nickel silicide that causes leakage-current failure in complementary metal-oxide- semiconductor (CMOS) devices by using advanced transmission electron microscope (TEM) techniques: electron tomography and spatially-resolved electron energy-loss spectroscopy (EELS). We reveal that the abnormal growth of Ni silicide results in a single crystal of NiSi2 and that it grows toward Si directions along (111) planes with the Ni diffusion through the silicon interstitial sites. In addition, we confirm that the abnormal growth is related to crystal microstructure and crystal defects. These detailed analyses are essential to understand the formation mechanism of abnormal growths of Ni silicide.

This study investigates the preventive effect of caffeic acid phenethyl ester (CAPE) on pancreatic damage induced by vancomycin (VCM) in rats. Rats were equally divided into three groups: group I (control), group II (only VCM-treated group) and group III (VCM + CAPE-treated groups). VCM was intraperitoneally administered at a dose of 200 mg kg−1twice daily for 7 days. CAPE was administered orally at 10 µM mL−1 kg−1 dose once daily for 7 days. The first dose of CAPE administration was performed 24 h prior to VCM injection. Blood and pancreas tissue samples were removed and collected after the study. Serum alkaline phosphatase (ALP), amylase, γ-glutamyl transferase (GGT) and lipase activities were determined. Pancreas tissue samples were evaluated with the light microscope. Group II significantly increased serum ALP, amylase, GGT and lipase activities when compared with the control group. Group III significantly decreased serum ALP, amylase, GGT and lipase activities when compared with group II. In histopathological examination, it has been observed that there was a significant pancreatic damage in group II. CAPE exerted prominent structural protection against VCM-induced pancreatic damage and this effect was statistically significant. CAPE caused a marked reduction in the extent of pancreatic damage. We have concluded that it may play an important role in the VCM-induced pancreatic damage and reduce the pancreatic damage both at the biochemical and histopathological aspects.

The RNA polymerase of influenza A virus is a heterotrimeric complex of PB1, PB2 and PA subunits that is required for transcription and replication of the viral genome. Here, we demonstrate a differential requirement of the PA subunit for binding to the vRNA and cRNA promoters — specifically, PA is more important for binding to the cRNA than the vRNA promoter. Furthermore, five point mutations were identified in the L163–I178 region of PA, which resulted in an inhibition of polymerase activity when provided with a cRNA compared to vRNA promoter. Cross-linking studies suggested that this inhibition was due to a reduction in promoter binding of the mutant polymerases to the cRNA promoter. We conclude that the L163–I178 region of PA is directly or indirectly involved in cRNA promoter binding and suggest a novel function for PA in modulating promoter binding.

Inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) related to Alzheimer’s disease as well as tyrosinase (TYR) relevant to Parkinson’s disease is an important approach to find novel drug candidates for these diseases. The extracts from fourteen plant species in various polarities were subjected to high-throughput screening against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase (TYR), the key enzymes related to Alzheimer’s and Parkinson’s diseases. The extracts were subjected to the microtiter enzyme inhibition assays at 100 μg mL-1. Antioxidant effect of the extracts was tested for their scavenging activity against DPPH, DMPD, and NO radicals as well as their ferric- (FRAP) and phosphomolibdenum-reducing power (PRAP) and metal-chelation capacity. Total phenol and flavonoid quantities in the extracts were determined spectrophotometrically. HPLC analysis was performed on Atriplex lasiantha, Conringia grandiflora, and Vaccaria hispanica. The active extracts inhibiting AChE over 50 % were Centaurium erythraea subsp. rhodense (51.33 ± 3.35 %) and Posidonia oceanica (61.88 ± 2.23 %), while BChE was inhibited most effectively by the root extract of P. oceanica (82.55 ± 2.14 %), followed by Origanum haussknechtii (66.88 ± 0.17 %), which also had the highest inhibition toward TYR (35.28 ± 1.90 %). The extracts from Zostera noltii, P. oceanica, and Ricotia carnosula possessed the best DPPH scavenging activity, whereas Z. noltii caused the highest NO scavenging activity (70.19 ± 0.43 %) and FRAP (1.326 ± 0.065). Atriplex lasiantha and Ecballium elaterium had the strongest effect in PRAP and metal-chelation assays, respectively. Besides, A. lasiantha was found to be a rich source of rutin. Among the screened plants, Centaurium erythraea subsp. rhodense and Origanum haussknechtii, and the roots of Posidonia oceanica seems to deserve further investigation for their neuroprotective potential.

Background/aims Intestinal bacteria induce endogenous signals that play a pathogenic role in hepatic insulin resistance and non-alcoholic fatty liver disease. Probiotics could modulate the gut flora and could influence the gut-liver axis. We aimed to investigate the preventive effect of two probiotic mixtures on the methionine choline-deficient diet-induced non-alcoholic steatohepatitis model in rats. Methods Two studies, short-term (2 weeks) and long-term (6 weeks), were carried out using 60 male Wistar rats. The 2-week study included six groups. Rats were fed with methionine choline-deficient diet or pair-fed control diet and were given a placebo or one of two probiotic mixtures (Pro-1 and Pro-2) by orogastric gavage. In the 6-week study, rats were allocated into four groups and were fed with methionine choline-deficient diet or pair-fed control diet and given a placebo or Pro-2. At the end of the 2- and 6-week periods, blood samples were obtained, the animals were sacrificed, and liver tissues were removed. Serum alanine aminotransferase activity was determined; histologic and immunohistochemical analysis was performed for steatosis, inflammation, protein expression of tumor necrosis factor-α, and apoptosis markers. Results In both studies, methionine choline-deficient diet caused an elevation of serum alanine aminotransferase activity, which was slightly reduced by Pro-1 and Pro-2. In the 2- and 6-week studies, feeding with methionine choline-deficient diet resulted in steatosis and inflammation, but not fibrosis, in all rats. In the 2-week study, in rats fed with methionine choline-deficient diet and given Pro-1, steatosis and inflammation were present in 2 of 6 rats. In rats fed with methionine choline-deficient diet and given Pro-2, steatosis was detected in 3 of 6 rats, while inflammation was present in 2 of 6 rats. In the 6-week study, in rats fed with methionine choline-deficient diet and given Pro-2, steatosis and inflammation were present in 3 of 6 rat livers. In both the 2- and 6-week studies, methionine choline-deficient diet resulted in tumor necrosis factor-α, proapoptotic Bax, caspase 3, caspase 8, and anti-apoptotic Bcl-2 expression in all rat livers. Pro-1 and Pro-2 treatment influenced protein expression involved in apoptosis and tumor necrosis factor-α in varying degrees. Conclusions Pro-1 and Pro-2 decrease methionine choline-deficient diet-induced steatohepatitis in rats. The preventive effect of probiotics may be due, in part, to modulation of apoptosis and their anti-inflammatory activity.

Background: The relationship between major adverse cardiac events (MACE) and serum polyunsaturated fatty acid (PUFA) parameters has not been well documented in patients who have undergone percutaneous coronary intervention (PCI). The aim of the present study was to investigate this relationship. Methods and Results: A total of 284 consecutive patients who underwent elective PCI were enrolled and stratified according to median serum levels of n-6 PUFAs (arachidonic acid [AA]), n-3 PUFAs (eicosapentaenoic acid [EPA] and docosahexaenoic acid [DHA]), and serum EPA/AA and DHA/AA ratios. The relationship between these PUFA parameters and the incidence of MACE including cardiac death, acute coronary syndrome, PCI for de novo lesions, and coronary artery bypass grafting, was analyzed. Multivariate analysis showed that among the PUFA parameters, only a high serum EPA/AA ratio was significantly associated with a low incidence of MACE in all the models tested (model A, without adjusted variables: hazard ratio [HR], 0.52; 95% confidence interval [CI]: 0.27-0.99, P=0.048; model B, adjusted for age and diabetes: HR, 0.51; 95%CI: 0.26-0.98, P=0.043; model C, adjusted for age, sex, diabetes, hypertension, smoking, and low-density lipoprotein cholesterol: HR, 0.49; 95%CI: 0.25-0.94, P=0.033). Conclusions: The incidence of MACE in patients who have undergone PCI is significantly associated with serum EPA/AA ratio. (Circ J 2012; 76: 423-429)

Doxorubicin (DOX) is a chemotherapeutic agent, and is widely used in cancer treatment. The most common side effect of DOX was indicated on cardiovascular system by experimental studies. There are some studies suggesting oxidative stress-induced toxic changes on liver related to DOX administration. The aim of the present study was to evaluate whether antioxidant N-acetylcysteine (NAC) relieves oxidative stress in DOX- induced liver injury in rat. Twenty-four male rats were equally divided into three groups. First group was used as a control. Second group received single dose of DOX. NAC for 10 days was given to constituting the third group after giving one dose of DOX. After 10 days of the experiment, liver tissues were taken from all animals. Lipid peroxidation (LP) levels were higher in the DOX group than in control whereas LP levels were lower in the DOX+NAC group than in control. Vitamin C and vitamin E levels were lower in the DOX group than in control whereas vitamin C and vitamin E levels were higher in the DOX+NAC group than in the DOX group. Reduced glutathione levels were higher in the DOX+NAC group than in control and DOX group. Glutathione peroxidase, vitamin A and β-carotene values were not changed in the three groups by DOX and NAC administrations. In histopathological evaluation of DOX group, there were mononuclear cell infiltrations, vacuolar degeneration, hepatocytes with basophilic nucleus and sinusoidal dilatations. The findings were totally recovered by NAC administration. In conclusion, N-acetylcysteine induced modulator effects on the doxorubicin-induced hepatoxicity by inhibiting free radical production and supporting the antioxidant vitamin levels.

We have used low-temperature (77K) resonance Raman (RR) spectroscopy as a probe of the electronic and molecular structure to investigate weak pi-pi interactions between the metal ion-coordinated His imidazoles and aromatic side chains in the second coordination sphere of blue copper proteins. For this purpose, the RR spectra of Met16 mutants of Achromobacter cycloclastes pseudoazurin (AcPAz) with aromatic (Met16Tyr, Met16Trp, and Met16Phe) and aliphatic (Met16Ala, Met16Val, Met16Leu, and Met16Ile) amino acid side chains have been obtained and analyzed over the 100-500cm(-1) spectral region. Subtle strengthening of the Cu(II)-S(Cys) interaction on replacing Met16 with Tyr, Trp, and Phe is indicated by the upshifted (0.3-0.8cm(-1)) RR bands involving nu(Cu-S)(Cys) stretching modes. In contrast, the RR spectra of Met16 mutants with aliphatic amino acids revealed larger (0.2-1.8cm(-1)) shifts of the nu(Cu-S)(Cys) stretching modes to a lower frequency region, which indicate a weakening of the Cu(II)-S(Cys) bond. Comparisons of the predominantly nu(Cu-S)(Cys) stretching RR peaks of the Met16X=Tyr, Trp, and Phe variants, with the molar absorptivity ratio epsilon(1)/epsilon(2) of sigma( approximately 455nm)/pi( approximately 595nm) (Cys)S-->Cu(II) charge-transfer bands in the optical spectrum and the axial/rhombic EPR signals, revealed a slightly more trigonal disposition of ligands about the copper(II) ion. In contrast, the RR spectra of Met16Z=Ala, Val, Leu, and Ile variants with aliphatic amino acid side chains show a more tetrahedral perturbation of the copper active site, as judged by the lower frequencies of the nu(Cu-S)(Cys) stretching modes, much larger values of the epsilon(1)/epsilon(2) ratio, and the increased rhombicity of the EPR spectra.

Alkylmethylbis(imidazolyl)borate ligands, [B(Im N-Me ) 2 (alkyl)-M e ] - (L alkyl ; alkyl = methyl and n-butyl), have been synthesized by nucleophilic substitution from the chloride to the corresponding alkyl derivative on the boron center of the chloroborate precursor. In homoleptic complexes of nickel(II) with L alkyl , [Ni II (L alkyl ) 2 ] (1 alkyl ) one of the two alkyl groups on the boron atom faces the nickel(II) center to form a C-H...M interaction. In the analogous homoleptic nikel(II) complexes of the hydride, phenyl, and methoxy derivatives 1 x (X = H, Ph, OMe), the boron-attached CH 3 moieties do not turn towards the metal center. Steric repulsion between the methyl substituent on the imidazolyl ligands and the boron-attached methyl group directs the orientation of the alkyl groups on L alkyl . In the solution state the molecular structures of 1 alkyl observed in the solid state are retained. Mixed-ligand complexes, [Ni II (L alkyl )(Tp iPr2 )] (2 alkyl ). can be synthesized by reaction of [Ni II (Y)(Tp iPr2 )] (Y = Cl, NO 3 ) with L alkyl through selective ligand metathesis. In 2 Me , one methyl group of L Me approaches the nickel center retaining the bent conformation of the two imidazolyl groups. Conversely, the carbon atoms of the boron-attached nBu and Me groups of L Bu are located away from the nickel center, and the orientation of the two imidazolyl groups is almost coplanar in 2 Bu . The structural characteristics of these nickel(II) complexes demonstrate the flexibility of the dialkybis(imidazolyl)borate scaffold.

The acetoxy-functionalized bis(imidazolyl)borate [B(ImN-Me)2(OC( O)Me)Me]− (=LOAc) is synthesized by the reaction of the alkoxy precursor [B(ImN-Me)2(OPri)Me]− (=LOiPr) with acetic acid. In the presence of weak Bronstead acid, migration of nickel-bound acetate anion to the boron center giving LOAc occurs. The boron–acetoxy linkage survives upon the treatment of the nickel complexes with OH−, although the acetoxy group on LOAc does not coordinate to the nickel center.

Osteopontin is critically involved in rheumatoid arthritis; however, the molecular cross-talk between osteopontin and joint cell components that leads to the inflammatory joint destruction is largely unknown. We found that not only osteopontin but also tenascin-C and their common receptor, α9 integrin, are expressed at arthritic joints. The local production of osteopontin and tenascin-C is mainly due to synovial fibroblasts and, to a lesser extent, synovial macrophages. Synovial fibroblasts and macrophages express α9 integrin, and autocrine and paracrine interactions of α9 integrin on synovial fibroblasts and macrophages and its ligands contribute differently to the production of proinflammatory cytokines and chemokines. α9 integrin is also involved in the recruitment and accumulation of inflammatory cells. Inhibition of α9 integrin function with an anti-α9 integrin Ab significantly reduces the production of arthrogenic cytokines and chemokines and ameliorates ongoing arthritis. Thus, we identified α9 integrin as a critical intrinsic regulator that controls the development of autoimmune arthritis.

To enhance the photocatalytic performance of titanium dioxide, the structures of both bulk and surface were modified. Doping of sulfur atoms to be substituted for lattice oxygen atoms of titanium dioxide was carried out to extend the light absorption by atmosphere-controlled pulsed laser deposition, which allows direct preparation of impurity-included thin film such as sulfur-doped titanium dioxide. On the other hand, to enhance the surface catalytic reaction, nickel nanoparticles were deposited at the thin film substrate by chemical vapor reductive deposition method, which is a novel preparation technique of metallic nanoparticles on the substrate surface. Obtained sulfur-doped titanium dioxide was found to possess sensitivity to visible light with the wavelength up to 550 nm, indicating the photocatalytic activity in visible region. Sulfur doping induced the dye degradation activity under visible light irradiation. When nickel nanoparticles were deposited, a remarkable enhancement of the hydrogen evolution activity through ethanol decomposition of more than 20 times as much as unmodified titanium dioxide thin film was accomplished. In addition, the stability of sulfur atom doped into titanium dioxide structure was investigated.

The anomalous gate-edge leakage current in n-channel metal-oxide-semiconductor field-effect transistors (nMOSFETs), which is caused by the encroached growth of nickel silicide across the p-n junction, is first reported. Furthermore, this encroached growth, which is caused by the isotropic and rapid diffusion of Ni atoms during the silicidation annealing, is successfully suppressed by the advanced Si+ ion-implantation (Si-I.I.) technique. Using the Si-I.I. technique, both the anisotropic silicidation to the perpendicular direction and the phase transition from Ni2Si to NiSi are enhanced by the introduction of damaged layers into Si substrates, such as vacancy and amorphous Si layers, and as a result, the silicidation region is confined at the source and drain regions. In addition, we propose a new evaluation method for the quantitative analysis of the encroached growth based on its growth properties, namely, the variability of encroached growths, which is three standard deviations of the roughness at silicide edges. The usefulness of this simple analysis for a large number of nMOSFETs is also demonstrated.

Osteopontin (OPN) contains at least two major integrin recognition domains, Arg159-Gly-Asp161 (RGD) and Ser162-Val-Val-Tyr-Gly-Leu-Arg168 (SVVYGLR), recognized by alphavbeta3 and alpha5beta1 and alpha4 and alpha9 integrins, respectively. OPN is specifically cleaved by thrombin and matrix metalloproteinase (MMP)-3 or MMP-7 at a position of Arg168/Ser169 (R/S) and Gly166/Leu167 (G/L), respectively. We in this study examined the requirement of residues within SVVYGLR for the alpha4 and alpha9 integrin recognition and how MMP-cleavage influences the integrin recognition. The residues, Val164, Tyr165, and Leu167 are critical for alpha4 and alpha9 integrin recognition in both cell adhesion and cell migration. The residue Arg168 is additionally required for alpha9 integrin recognition in cell adhesion and this explains why alpha9 integrin binds to only thrombin cleaved form of OPN. alpha4 integrin is able to bind to SVVYG (MMP-cleaved form of RAA OPN-N half), while alpha9 integrin is not, supporting the above notion that Arg168 is additionally required for alpha9 integrin-mediated cell adhesion. The residue Val163 is important for alpha4, but not for alpha9 integrin recognition in cell migration. Importantly, we found that the replacement of Arg168 by Ala (R168A mutant) induces the augmentation of cell migration via alpha4 and alpha9 integrins.

The effect of employing diborane (B2H6) instead of silane (SiH4) in a tungsten (W) atomic layer deposition (ALD) nucleation layer on contact resistance is studied. A low resistance of 50–70 Ω in a 32-nm-node contact with a diameter of about 50 nm is achieved with optimized barrier and nucleation processes. This result indicates that the resistance required for 32 nm complementary metal–oxide–semiconductor (CMOS) devices can successfully be satisfied without employing a copper plug process. It is revealed that a W film with a B2H6-reduced nucleation layer has a larger grain size and a lower fluorine concentration in nucleation W and barrier metal layers, resulting in a lower resistivity than that with a SiH4-reduced nucleation W layer. On the other hand, the B2H6-reduced nucleation process still shows a poorer crystallization and a higher fluorine concentration in the filling W layer than the SiH4-reduced nucleation process. We revealed that a further decrease in the resistivity of a W film with the B2H6 nucleation process is achieved by improving these issues.

We demonstrated highly reliable Cu interconnects using a high-quality silicon nitride film grown at temperatures below 300 °C. The low-temperature silicon nitride (LT-SiN) film, which was used as a Cu-diffusion barrier layer and a final passivation layer, was deposited at 275 °C by plasma-enhanced chemical vapor deposition at a low SiH4 flow ratio. The low SiH4 flow ratio was due to the use of a highly dilute nitrogen flow, leading to the generation of many nitrogen radicals or ions in the plasma. These radicals or ions might reduce the hydrogen concentration and defect density of the film. As a result, a stoichiometric silicon nitride (Si3N4) film with a low hydrogen concentration was successfully obtained. By applying this LT-SiN film in 130-nm-node Cu interconnects for magnetoresistive random access memory, highly reliable via-hole electromigration (Via-EM) and line-to-line time-dependent dielectric breakdown (TDDB) characteristics were obtained.

As the cell size of flash memories is scaled down, the reading error due to threshold voltage (Vth) interference has become a more serious problem, particularly in the case of multi-level-cell (MLC) flash memories, it is necessary for the Vth distribution to be narrower than that of the single-level-cell (SLC). In this work, we propose an advanced air gap structure and process to reduce the interference due to the capacitance between floating gate and floating gate. By applying an air gap between neighboring floating gates as a lowest-dielectric-constant material, we can suppress Vth interference markedly. In addition, we clarified the correlation between the air gap forming process and the memory cell reliability. Hydrogen included in the SiO2 film, which is deposited by plasma-enhanced chemical vapor deposition (CVD) during air gap formation causes the degradation of memory cell endurance and the de-trapping characteristics. We were able to achieve a high-reliability memory cell by reducing the hydrogen concentration in the SiO2 by optimizing the deposition process for air gap formation.

Hydrogen ions drifting into underlying layers during HDP-CVD were successfully suppressed by the insertion of plasma deposited silicon oxynitride ( p -SiO x N y H z ) film, and the hydrogen-trapping mechanism was clarified. The hydrogen ions are trapped in bonding states, not in interstitial ones. After HDP-CVD undoped silicate glass (HDP-USG) film deposition on the p -SiO x N y H z film, the decrease of the dangling bonds in the p -SiO x N y H z film measured by ESR was much lower than the increase of the desorbed hydrogen concentration measured by TDS. These results suggest that new hydrogen-trapping sites are mainly generated from ESR-inactive bonds by drifted hydrogen ions and atomic hydrogen during HDP-CVD.

We report the photochemical reaction of volatile organic compounds (VOC's), such as acetaldehyde and toluene, over nitrogen doped TiO2(TiO-N) under visible light irradiation, using liquid chromatography (LC), gas chromatography (GC) and gas chromatograph mass spectrometer (GC-MS) for gases analysis, and diffuse reflectance fourier transform infrared spectroscopy (DRIFTS) and ion chromatography (IC) for adsorbed organics. We evaluated the photo degradation rates and intermediates for the above gases. As a result, the degradation rates of acetaldehyde gas on TiO-N were 1.2-3 times greater than those on TiO2, and those of toluene gas on TiO-N were 4-49 times greater than those on TiO2. Low concentrations of acetone and acetic acid were produced during the photo degradation of acetaldehyde by the TiO-N. From toluene, very low concentrations of formaldehyde, acetaldehyde, acetone and benzaldehyde gases were produced. We also revealed the decomposition processes of toluene. Results obtained indicate that toluene, weakly adsorbed on the photocatalyst surface, is initially photooxidized to benzaldehyde which adsorbs onto the TiO-N surface more strongly, leading to the formation of ring-opening products such as carboxylic acids and aldehydes. These intermediate products were gradually photodegraded to CO2 and H2O under visible light irradiation.

Background Most mobile phones emit electromagnetic radiation at 900 MHz or 1800 MHz. An electromagnetic field has some biological effects on the behavior of the cell population of bone. The aim of this work is to evaluate the effects of the radiation emitted by mobile phones on bone mineral density (BMD). The effects of caffeic acid phenethyl ester (CAPE) on the radiation-induced changes were also investigated. Methods In the study, 48 Sprague Dawley rats were used. Rats were divided into five groups as follows: control, irradiated with 900 MHz, irradiated with 900 MHz and treatment, irradiated with 1800 MHz, irradiated with 1800 MHz and treatment groups. The rats in the control group (first group) were left within the experimental setup during 30 min/day for 28 days without radiation exposure. Nine hundred-MHz radiation group was exposed to irradiate both second and third groups for 28 days (30 min/day); 1800-MHz radiation group was exposed to irradiate both fourth and fifth groups for 28 days (30 min/day). Third and fifth groups were also treated by CAPE for 28 days. Treatment groups received 10 μml/kg/day CAPE i.p. before the irradiation. Bone mineral densities were determined in all groups. Results BMD was found to be decreased in the irradiated groups and to be increased in the treatment groups. Conclusions The changes were not significant (p >0.05).

The RNA-dependent RNA polymerase of influenza virus is a heterotrimer formed by the PB1, PB2, and PA subunits. Although PA is known to be required for polymerase activity, its precise role is still unclear. Here, we investigated the function of the N-terminal region of PA. Protease digestion of purified recombinant influenza virus A/PR/8/34 PA initially suggested that its N-terminal region is folded into a 25-kDa domain. We then systematically introduced point mutations into evolutionarily conserved amino acids in the N-terminal region of influenza virus A/WSN/33. Most alanine-scanning mutations between residues L109 and F117 caused PA degradation, mediated by a proteasome-ubiquitin pathway, and as a consequence interfered with polymerase activity. Three further PA mutations, K102A, D108A, and K134A, were investigated in detail. Mutation K102A caused a general decrease both in transcription and replication in vivo, whereas mutations D108A and K134A selectively inhibited transcription. Both the D108A and K134A mutations completely inhibited endonuclease activity in vitro, explaining their selective defect in transcription. K102A, on the other hand, resulted in a significant decrease in both cap binding and viral RNA promoter-binding activity and consequently inhibited both transcription and replication. These results suggest that the N-terminal region of PA is involved in multiple functions of the polymerase, including protein stability, endonuclease activity, cap binding, and promoter binding.

Cadmium is one of the most toxic pollutants in environment. Cadmium accumulation in blood affects the renal cortex and causes renal failure. In this study, we aimed to evaluate the effects of cadmium on rat liver tissue. Eighteen male albino rats aged ten weeks old were used in the study. 15 ppm of cadmium was administered to rats via consumption water daily. At the end of the 30th study day, the animals were killed under ether anesthesia. After the liver tissue samples were taken, histopathological and biochemical examinations were performed. Histopathologic changes have included vacuolar and granular degenerations in hepatocytes, heterochromatic nucleuses and sinusoidal and portal widenings. Central vein diameters were normal in cadmium exposed group. Whereas, there was statistically significant difference between two groups by means of sinusoidal (p< 0.001) and portal triad diameters (p< 0.01). Malondialdehyde (MDA) is an indicator of lipid peroxidation. In this study, MDA was used as a marker of oxidative stress-induced liver impairment in cadmium exposed rats. Superoxide dismutase (SOD) and catalase (CAT) activities were also measured to evaluate the changes in antioxidative system in liver tissues. Current findings showed that MDA levels were increased and SOD and CAT activities were decreased in cadmium exposed group compared to control group. The difference between two groups was statistically significant (pvalues: MDA,p< 0.01; CAT,p< 0.01 and SOD,p< 0.05). In conclusion, these findings suggest the role of oxidative mechanisms in cadmium-induced liver tissue damage

A unicellular marine green alga, Chlamydomonas perigranulata, was demonstrated to synthesize starch through photosynthesis, store it in a cell, and ferment it under anaerobic conditions in the dark to produce ethanol, 2,3-butanediol (butanediol), acetic acid, and carbon dioxide (CO2). Previous fermentation data of an algal biomass cultivated outdoors in a 50-L tubular photo-bioreactor showed good carbon (C) recovery in the fermentation balance, with a higher ratio to alcohols and, therefore, lower ratio to CO2 in the C distribution of products than what would be expected from the embden-Myerhof-Parnas pathway. These findings led to a proposed concept for a CO2-ethanol conversion system (CDECS). The above data were evaluated in terms of hydrogen (H) recovery with the following results: C recovery at 105% was well balanced, although H recovery was as high as 139%, meaning an additional gain of H through fermentation. This finding was reproduced wholly in a set of experiments carried out in the same month of the following year, October, whereas another set of experiments was carried out in the following June provided ordinary fermentation results in terms of C and H recoveries with poor growth. Further analyses of these data revealed that butanediol is equal to ethanol as a product from a putative conversion system from CO2 to the detected fermentation products, leading to the revision of the CDECS concept to a CO2-alcohol conversion system (CDACS). The relevance of the CDACS will be discussed in relation to the cultivation conditions employed by chance.

Caffeic acid phenethyl ester (CAPE), a flavonoid like compound, is one of the major components of honeybee propolis. It has been used in folk medicine for many years in Middle East countries. It was found to be a potent free radical scavenger and antioxidant recently. The aim of this study was to examine long-term applied 900 MHz emitting mobile phone-induced oxidative stress that promotes production of reactive oxygen species (ROS) and, was to investigate the role of CAPE on kidney tissue against the possible electromagnetic radiation (EMR)-induced renal impairment in rats. In particular, the ROS such as superoxide and nitric oxide (NO) may contribute to the pathophysiology of EMR-induced renal impairment. Malondialdehyde (MDA, an index of lipid peroxidation) levels, urinary N-acetyl-β-d-glucosaminidase (NAG, a marker of renal tubular injury) and nitric oxide (NO, an oxidant product) levels were used as markers of oxidative stress-induced renal impairment and the success of CAPE treatment. The activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) in renal tissue were determined to evaluate the changes of antioxidant status. The rats used in the study were randomly grouped (10 each) as follows: i) Control group (without stress and EMR), ii) Sham-operated rats stayed without exposure to EMR (exposure device off), iii) Rats exposed to 900 MHz EMR (EMR group), and iv) A 900 MHz EMR exposed + CAPE treated group (EMR + CAPE group). In the EMR exposed group, while tissue MDA, NO levels and urinary NAG levels increased (p < 0.0001), the activities of SOD, CAT, and GSH-Px in renal tissue were reduced (p < 0.001). CAPE treatment reversed these effects as well (p < 0.0001, p < 0.001 respectively). In conclusion, the increase in NO and MDA levels of renal tissue, and in urinary NAG with the decrease in renal SOD, CAT, GSH-Px activities demonstrate the role of oxidative mechanisms in 900 MHz mobile phone-induced renal tissue damage, and CAPE, via its free radical scavenging and antioxidant properties, ameliorates oxidative renal damage. These results strongly suggest that CAPE exhibits a protective effect on mobile phone-induced and free radical mediated oxidative renal impairment in rats.

Aberrant DNA methylation of multiple promoter associated CpG islands is a frequent phenomenon in acute lymphocytic leukemia (ALL). Recently, methylation of a cell cycle control pathway composed of P73, P15 and P57KIP2 has been shown to confer poor prognosis to adult patients with ALL. Using bisulfite PCR methods, we have explored the prevalence of methylation of this pathway in a cohort of children with ALL (N=20), and compared these results with those observed in a group of adult patients (N=53). P73 was methylated in 4 (20%) pediatric patients, P15 in 3 (15%), and P57KIP2 in 2 (10%). These compared to 14 (26%), p=0.5, 16 (30%), p=0.04 and 20 (37%), p=0.04, respectively in adult patients. Methylation of two or more genes was not observed in any pediatric patient, but in 15 (28%) adult patients (p=0.003). Poor survival of adult patients was associated with methylation of > or =2 genes (p=0.003). These results indicate that differences in DNA methylation of specific molecular pathways may contribute to the prognostic differences known to occur between pediatric and adult patients with ALL.

Melatonin and caffeic acid phenethyl ester (CAPE), a component of honeybee propolis, were recently found to be potent free radical scavengers and antioxidants. There are a number of reports on the effects induced by electromagnetic radiation (EMR) in various cellular systems. Mechanisms of adverse effects of EMR indicate that reactive oxygen species may play a role in the biological effects of this radiation. The present study was carried out to compare the protective effects of melatonin and CAPE against 900 MHz EMR emitted mobile phone-induced renal tubular injury. Melatonin was administered whereas CAPE was given for 10 days before the exposure. Urinary N-acetyl-beta-D-glucosaminidase (NAG, a marker of renal tubular injury) and malondialdehyde (MDA, an index of lipid peroxidation), were used as markers of oxidative stress-induced renal impairment in rats exposed to EMR. Superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities were studied to evaluate the changes of antioxidant status in renal tissue. Urinary NAG and renal MDA were increased in EMR exposed rats while both melatonin and CAPE caused a significant reduction in the levels of these parameters. Likewise, renal SOD and GSH-Px activities were decreased in EMR exposed animals while melatonin caused a significant increase in the activities of these antioxidant enzymes but CAPE did not. Melatonin caused a significant decrease in urinary NAG activity and MDA levels which were increased because of EMR exposure. CAPE also reduced elevated MDA levels in EMR exposed renal tissue, but the effect of melatonin was more potent than that of CAPE. Furthermore, treatment of EMR exposed rats with melatonin increased activities of SOD and GSH-Px to higher levels than those of control rats. In conclusion, melatonin and CAPE prevent renal tubular injury by reducing oxidative stress and protect the kidney from oxidative damage induced by 900 MHz mobile phone. Nevertheless, melatonin seems to be a more potent antioxidant compared with CAPE in kidney.

Although iron (Fe), plays an important role in different oxidative steps during the metabolism of the human body, it can cause free radical damage. Iron ions seem to play a major role in initiation and promotion reactions of intracellular lipid peroxidation. The aim of this study was to investigate if vitamin E has a protective effect on oxidative changes in erythrocytes induced by Fe treatment. Thirty male New Zealand white rabbits weighing 14009 / 50 g were used in the study. The animals were divided into three groups. The first group (n:10) was given 500 mg/kg iron-dextran through intraperitoneal (ip) injection. The second group was given 500 mg/kg iron-dextran+100 mg/kg vitamin E(ip). The third group constituted the control group and received a saline solution injection. The activities of erythrocyte antioxidant enzymes; Superoxide Dismutase (SOD), Glutatione peroxidase (GSH-Px), Catalase (CAT) and Malondialdehyde (MDA) level, an indicator of lipid peroxidation, were determined. Erythrocyte SOD, GSH-Px and CAT activities were decreased and MDA level was increased in iron-dextran treated animals compared to the control group (PB / 0.05). The activities of the three antioxidant enzymes were increased and MDA level was decreased in iron-dextran and vitamin E treated group compared to the control group (PB / 0.05). Our data indicate that lipid peroxidation occurs after iron overload in the blood. In the light of our findings, vitamin E administration can prevent the toxic oxidative effects induced by iron-dependent free radical damage in erythrocytes.

The aim of this study was to evaluate the effects of the novel free radical scavenger caffeic acid phenethyl ester (CAPE) on extracorporeal shock wave lithotripsy (ESWL) induced renal impairment. The study was performed using 30 rabbits which were divided into two groups, each exposed to 3,000 shock waves at 18 kV: (1) control group, (2) ESWL+CAPE treated group. Malodialdehyde (MDA), urine N-acetyl-beta-glucosaminidase (NAG) activity, uric acid and white cell counts were used as markers of oxidative stress. Following shock wave exposure there was a significant rise in MDA, NAG and uric acid and white cell counts. CAPE reduced the rise in MDA, NAG, uric acid and white cell counts. Thus CAPE treatment to a great extent prevented the induction of these renal changes. Our results suggest that the antioxidant capacity of the kidney tissue was reduced after ESWL treatment and that the tissue was exposed to oxidant stress. We conclude that CAPE treatment provided significant protection against ESWL induced free radical damage.

The aim of this study was to investigate effects of an organophosphate insecticide, diazinon (DI), on pancreas, and possible ameliorating role of vitamins E and C. We examined both in vivo and in vitro effects of DI on serum activities of alkaline phosphatase (ALP), γ-glutamyltransferase (GGT), amylase, and lipase enzymes. We also evaluated possible ameliorating effects of vitamins E and C combination against DI toxicity and blood levels of thiobarbituric acid reactive substances (TBARS) only in vivo. In vivo experimental groups were: control group, DI-treated group, and DI + vitamins E plus C-treated group. In vitro study groups were: control group and DI-treated group. The biochemical analyses were determined in in vitro experiments at both hour 0 and 24 while in in vivo experiments were determined only at hour 24. Lipase activity and TBARS level were found increased by DI in in vivo experiments while lipase activity was found decreased in in vitro experiments. Amylase and ALP activities were found decreased by DI in both in vivo and in vitro experiments. Also, the combination of vitamins E and C was found to partially improve these disorders. These results suggest that DI treatment causes pancreas damage via increasing oxidative stress in rats, and a combination vitamins E and C can reduce this lipoperoxidative effect.

Microwaves (MW) from cellular phones may affect biological systems by increasing free radicals, which may enhance lipid peroxidation (LP), and by changing the antioxidative activities of the liver, thus leading to oxidative damage. Caffeic acid phenethyl ester (CAPE), an active component of propolis extract, exhibits antioxidant properties and several studies suggest that supplementation with antioxidant can influence MW induced hepatotoxicity. The present study was designed to determine the effects of MW on the liver oxidant/antioxidant system, and the possible protective effects of CAPE on liver toxicity induced by MW. Twenty nine male Spraque- Dawley rats were divided into three groups: control (n = 9), 1800 MHz MW (n = 10) and 1800 MHz MW + CAPE (n = 10). The preparation of phenyltris[1-adamantyl(thiomethyl)]borate [PhTtAd], which incorporates the large 1-adamantyl thioether substituent, is described. This derivative was sought to permit the preparation of four-coordinate nickel(I) complexes. Following a synthetic scheme for the related tert-butyl ligand, [PhTtBut], the following complexes were prepared and characterized fully, [PhTtAd]Ni(CO), [PhTtAd]Ni(PMe3) and [κ2-PhTtAd]Ni(η2-CH2SAd). The last species is a degradation product resulting from attempted reduction of the nickel(II) precursor, [PhTtAd]NiCl, in the absence of a ligand suitable for trapping the nickel(I) fragment.

The biochemical properties of hepatitis C virus (HCV) RNA-dependent RNA polymerase (RdRp) truncated with C-terminal 21 amino acids and expressed in insect cells were analyzed. The enzyme carried copy-back and de novo RNA synthesis activity but not terminal nucleotidyl transferase activity. k(pol) and K(m) for de novo RNA synthesis were calculated as 10.0 pmol/microg/h and 2.5 microM under 0.5 mM GTP and 2.0 pmol/microg/h and 3.5 microM under 50 microM GTP, respectively. Those for copy-back RNA synthesis were similar under both conditions (k(pol), 1.8 pmol/microg/h; K(m), 3.0 microM). De novo RNA synthesis was activated by 0.5 mM GTP. However, the ratio of GTP to three other NTPs was important for activation. Our HCV RdRp showed high activity for the complementary sequence of the HCV internal ribosomal entry site and a synergistic effect of Mg(2+) to Mn(2+).
