European Snacks Association (ESA) data on acrylamide in potato crisps from 2002 to 2019 (99704 observations) were analysed. Acrylamide levels have plateaued since 2011, although the lowest mean so far was attained in 2018 at 353 ± 2.7 ng g-1: a 54% reduction since 2002. The 85th, 90th and 95th quantiles did show evidence of continued downward progress, the 90th quantile being lower than the 750 ng g-1 European Benchmark Level from 2017 to 2019. A smaller dataset from the European Food Safety Authority (2124 observations) for 2011-2018, was also analysed. The yearly means were higher than those of the ESA data, but showed a fall in average acrylamide from 715 ± 40.5 ng g-1 in 2015 to 505 ±28.5 ng g-1 in 2018, as well as steep falls in the 85th, 90th and 95th quantiles. Nevertheless, even the 85th quantile remained above the 750 ng g-1 Benchmark Level. The ESA data showed a reduction in the proportion of samples with acrylamide exceeding 750 ng g-1, from over 40 % in 2002 to 7.75 % in 2019. Seasonality was evident, with highest acrylamide levels from November to May. Crisp type had little effect except that thicker types had a higher proportion of samples containing >750 ng g-1 acrylamide. Analysis of the region of origin in Europe of the final product revealed improvements in the east and north. Geographical factors combined with seasonality continued to be problematic but was also an aspect in which progress was most evident. The findings show that improvements have been made in reducing the number of samples with very high levels of acrylamide, but do not suggest that mean acrylamide levels could be reduced substantially below where they have been since 2011, or that levels could be kept consistently below the current Benchmark Level.

The major components of wheat storage proteins are gliadins and glutenins, and as they contribute differently to baking quality, a balanced mixture of these components is essential. The application of foliar nitrogen (N) at anthesis is a common practice to improve protein concentration and composition. The aim of this study was to investigate the effects of a foliar N application at anthesis on storage protein gene expression during grain development and on the distribution of protein concentration and protein body size within the grain. In this experiment, an additional N application at anthesis stimulated the expression of genes of the majority of storage proteins when the N supply was low. Furthermore, it led to higher protein concentrations in the subaleurone layers, while in the center of the lobes, the protein concentrations were decreased. These changes will affect the protein recovery in white flours, as proportionally more protein might be lost during milling processes.

Colorful ornaments are hypothesized to have evolved in response to sexual selection for honest signals of individual quality that provide information about potential mates. Red carotenoid coloration is common in diverse groups, and in some vertebrate taxa, red coloration is a sexually selected trait whereby mates with the reddest ornaments are preferred. Despite being widespread among invertebrates, whether red carotenoid coloration is assessed during mate choice in these taxa is unclear. The marine copepod Tigriopus californicus displays red coloration from the accumulation of the carotenoid astaxanthin. Previous research on copepods has shown that astaxanthin provides protection from UV radiation and xenobiotic exposure and that carotenoid production is sensitive to external stressors. Because of the condition dependency of the red coloration, we hypothesized that Tigriopus would use it as a criterion during mate choice. To test this hypothesis, we conducted trials in which males chose between females that were wild‐type red (carotenoid‐rich algae diet) or white (carotenoid‐deficient yeast diet). To control for dietary differences and to isolate the effect of carotenoid coloration, we also presented males with restored‐red females fed a carotenoid‐supplemented yeast diet. We found that wild‐type red females were weakly preferred over white females. After controlling for diet, however, we found that restored‐red females were avoided. Our observations do not support the hypothesis that male copepods prefer the carotenoid coloration of females during mate choice. We hypothesize that algal‐derived compounds other than carotenoids play a role in mate choice. Red coloration in copepods appears to be a condition‐dependent trait that is not assessed during mating.

This perspective addresses recent advances in lipid transport across the Gram-negative inner and outer membranes. While we include a summary of previously existing literature regarding this topic, we focus on the maintenance of lipid asymmetry (Mla) pathway. Discovered in 2009 by the Silhavy group [J. C. Malinverni, T. J. Silhavy, Proc. Natl. Acad. Sci. U.S.A. 106, 8009–8014 (2009)], Mla has become increasingly appreciated for its role in bacterial cell envelope physiology. Through the work of many, we have gained an increasingly mechanistic understanding of the function of Mla via genetic, biochemical, and structural methods. Despite this, there is a degree of controversy surrounding the directionality in which Mla transports lipids. While the initial discovery and subsequent studies have posited that it mediated retrograde lipid transport (removing glycerophospholipids from the outer membrane and returning them to the inner membrane), others have asserted the opposite. This Perspective aims to lay out the evidence in an unbiased, yet critical, manner for Mla-mediated transport in addition to postulation of mechanisms for anterograde lipid transport from the inner to outer membranes.

Tertiary Lymphoid Structures (TLS) are commonly identified in human tumors with improved outcome, but how they are orchestrated remains elusive. We show that ablation of the genomic organizer SATB1 triggers the polarization of CD4+ T-cells into antigen-specific T Follicular Helper (TFH) cells. In contrast, SATB1 expression is required for conversion of CD4+ T-cells into induced regulatory T-cells (Treg), including T Follicular Regulatory (TFR) cells. Paradoxically, Tgf-β-driven Satb1 silencing causes Icos de-repression and prevents TFR differentiation, driving TFH polarization and increased isotype-switched B cell responses. Tumors in mice carrying Satb1-deficient CD4+ T-cells accumulate more tumor antigen-specific, Light/Cxcl13/Il-21-producing TFH cells, causing germinal center formation and spontaneous TLS assembly, which antagonizes tumor growth. Accordingly, TFH cell transfer induces large TLS within orthotopic ovarian tumors. Therefore, SATB1 repression in CD4+ T-cells licenses TFH cell differentiation while preventing Treg conversion, a central mechanism during TLS assembly that can be exploited to boost anti-tumor immunity.

Root elongation depends on the action of the gibberellin (GA) growth hormones, which promote cell production in the root meristem and cell expansion in the elongation zone. Sites of GA biosynthesis in the roots of 7-d-old Arabidopsis thaliana seedlings were investigated using tissue-specific GA inactivation in wild-type (Col-0) or rescue of GA-deficient dwarf mutants. Tissue-specific GA depletion was achieved by ectopic expression of the GA-inactivating enzyme AtGA2ox2, which is specific for C-GAs, and AtGA2ox7, which acts on C-GA precursors. In addition, tissue-specific rescue of ga20ox triple and ga3ox double mutants was shown. Furthermore, GUS reporter lines for major GA20ox, GA3ox and GA2ox genes were used to observe their expression domains in the root. The effects of expressing these constructs on the lengths of the root apical meristem and cortical cells in the elongation zone confirmed that roots are autonomous for GA biosynthesis, which occurs in multiple tissues, with the endodermis a major site of synthesis. The results are consistent with the early stages of GA biosynthesis within the root occurring in the meristematic region and indicate that the penultimate step of GA biosynthesis, GA 20-oxidation, is required in both the meristem and elongation zone., Biotechnology and Biological Sciences Research Council. The marine copepod, Tigriopus californicus, produces the red carotenoid pigment astaxanthin from yellow dietary precursors. This ‘bioconversion’ of yellow carotenoids to red is hypothesized to be linked to individual condition, possibly through shared metabolic pathways with mitochondrial oxidative phosphorylation. Experimental inter-population crosses of lab-reared T. californicus typically produces low-fitness hybrids is due in large part to the disruption of coadapted sets nuclear and mitochondrial genes within the parental populations. These hybrid incompatibilities can increase variability in life history traits and energy production among hybrid lines. Here, we tested if production of astaxanthin was compromised in hybrid copepods and if it was linked to mitochondrial metabolism and offspring development. We observed no clear mitonuclear dysfunction in hybrids fed a limited, carotenoid-deficient diet of nutritional yeast. However, when yellow carotenoids were restored to their diet, hybrid lines produced less astaxanthin than parental lines. We observed that lines fed a yeast diet produced less ATP and had slower offspring development compared to lines fed a more complete diet of algae, suggesting the yeast-only diet may have obscured effects of mitonuclear dysfunction. Astaxanthin production was not significantly associated with development among lines fed a yeast diet but was negatively related to development in early generation hybrids fed an algal diet. In lines fed yeast, astaxanthin was negatively related to ATP synthesis, but in lines fed algae, the relationship was reversed. Although the effects of the yeast diet may have obscured evidence of hybrid dysfunction, these results suggest that astaxanthin bioconversion may still be related to mitochondrial performance and reproductive success.

Amino acids are delivered into developing wheat grains to support the accumulation of storage proteins in the starchy endosperm, and transporters play important roles in regulating this process. RNA-seq, RT-qPCR, and promoter-GUS assays showed that three amino acid transporters are differentially expressed in the endosperm transfer cells (TaAAP2), starchy endosperm cells (TaAAP13), and aleurone cells and embryo of the developing grain (TaAAP21), respectively. Yeast complementation revealed that all three transporters can transport a broad spectrum of amino acids. RNAi-mediated suppression of TaAAP13 expression in the starchy endosperm did not reduce the total nitrogen content of the whole grain, but significantly altered the composition and distribution of metabolites in the starchy endosperm, with increasing concentrations of some amino acids (notably glutamine and glycine) from the outer to inner starchy endosperm cells compared with wild type. Overexpression of TaAAP13 under the endosperm-specific HMW-GS (high molecular weight glutenin subunit) promoter significantly increased grain size, grain nitrogen concentration, and thousand grain weight, indicating that the sink strength for nitrogen transport was increased by manipulation of amino acid transporters. However, the total grain number was reduced, suggesting that source nitrogen remobilized from leaves is a limiting factor for productivity. Therefore, simultaneously increasing loading of amino acids into the phloem and delivery to the spike would be required to increase protein content while maintaining grain yield. © 2021 Wan et al. The asymmetric outer membrane (OM) of Gram-negative bacteria functions as a selective permeability barrier to the environment. Perturbations to OM lipid asymmetry sensitize the cell to antibiotics. As such, mechanisms involved in lipid asymmetry are fundamental to our understanding of OM lipid homeostasis. One such mechanism, the Maintenance of lipid asymmetry (Mla) pathway has been proposed to extract mislocalized glycerophospholipids from the outer leaflet of the OM and return them to the inner membrane (IM). Work on this pathway inAcinetobacter baumanniisupport conflicting models for the directionality of the Mla system being retrograde (OM to IM) or anterograde (IM to OM). Here, we show conclusively thatA. baumannii mlamutants exhibit no defects in anterograde transport. Furthermore, we identify an allele of the GTPaseobgEthat is synthetically sick in the absence of Mla; providing another link between cell envelope homeostasis and stringent response.

Critical experiments using an insertion zone with FLiBe salt performed at the LR-0 reactor at Research Centre Řež (RC Řež) have been compared to application models for solid-fueled, fluoride-salt-cooled high-temperature reactor (FHR) and liquid-fueled molten salt reactor (MSR) concepts using sensitivity and uncertainty (S/U) analysis techniques. These experiments support FHR and MSR advanced reactor concepts by informing on neutron spectral effects and nuclear data uncertainties related to fluoride salts. The FLiBe salt in the LR-0 experiments is from the Oak Ridge National Laboratory Molten Salt Reactor Experiment and is enriched to greater than 99.99% 7Li. This work is part of a broader collaboration between the United States and the Czech Republic on MSR and FHR technology. Results from the S/U analyses for FHR and MSR models indicated the most significant potential source of eigenvalue bias due to nuclear data within the FLiBe salt is radiative capture in 7Li. Other smaller but potentially significant contributions come from 19F, 6Li, and other 7Li reactions. Similarity comparisons of the RC Řež LR-0 experiments and FHR and MSR application models indicate that the LR-0 FLiBe experiments could be useful as candidate benchmarks for low-enriched uranium–fueled application models. However, sensitivity differences observed included both spectral effects, due to the LR-0 reactor being moderated by water instead of graphite and fluoride salt, and sensitivity magnitude effects driven by the amount of FLiBe inserted and its location. New experiments with refined designs to increase the volume and importance of salt in the system could provide improved data. Results also demonstrate that salt systems using thorium and/or 233U fuel will require additional experiments with relevant driver fuels. Increased contributions from the graphite moderator and 19F and covariance between reactions such as 233U fission and 233U radiative capture indicate higher uncertainty contributions from the salt and significant differences in contributions from the fuel species due to underlying uncertainties in their nuclear data.

Research on molten salt reactor (MSR) neutronics continues in Research Centre Rez (Czech Republic) with experimental work being conducted using fluoride salt that was originally used in the Molten Salt Reactor Experiment (MSRE). Previous results identified significant variations in the neutron spectrum measured in LiF-NaF salt. These variations could originate from the fluorine description in current nuclear data sets. Subsequent experiments were performed to try to confirm this phenomenon. Therefore, another fluorine-rich compound, Teflon, was used for testing. Critical experiments showed slight discrepancies in C/E-1 for both compounds, Teflon and FLIBE, and systematic overestimation of criticality was observed in calculations. Different nuclear data libraries were used for data set testing. For Teflon, the overestimation is higher when using JENDL-3.3, JENDL-4, and RUSFOND-2010 libraries, all three of which share the same inelastic-to-elastic scattering cross section ratio. Calculations using other libraries (ENDF/B-VII.1, ENDF/B-VII.0, JEFF-3.2, JEFF-3.1, and CENDL-3.1) tend to be closer to the experimental value. Neutron spectrum measurement in both substances revealed structure similar to that seen in previous measurements using LiF-NaF salt, which indicates that the neutron spectrum seems to be strongly shaped by fluorine. Discrepancies between experimental and calculational results seem to be larger in the neutron energy range of 100–1300 keV than in higher energies. In the case of neutron spectrum calculation, none of the tested libraries gives overall better results than the others.

The fuel cycle performance and core design of the Transatomic Power liquid-fueled molten salt reactor concept is analyzed. This advanced reactor concept uses configurable zirconium hydride moderator rod assemblies to shift the neutron spectrum in the core from intermediate at beginning of life to thermal at end of life. With a harder spectrum during the early years of reactor operation, this spectral shift design drives captures in fertile 238U. The converted fissile plutonium makes up over 50% of the fissile material in the fuel salt over the last half (∼15 years) of reactor operation. A softer spectrum late in reactor life helps drive the fuel to a burnup of 90 GWd/MTU. Continuously changing physics necessitates time-dependent analyses resolved over long timescales (i.e., months to years), as this concept does not meet an equilibrium condition. The spectral shift and molten salt reactor material feeds and removals enable this concept to perform better in fuel cycle metrics, increasing resource utilization by more than 50% compared with a typical light water reactor (i.e., from ∼0.6% to ∼1%). These metrics are compared to similar fuel cycles using alternate technologies. Additional core design and analysis challenges associated with the spectral shift and use of molten salt reactor technology are identified and discussed.

Fully ceramic microencapsulated (FCM) fuel is a proposed fuel type for improved accident performance in LWRs (Light Water Reactors) that involves TRISO (TRistructural-ISOtropic) particles embedded in a nano-powder sintered silicon carbide (SiC) matrix. The TRISO particles contain a spherical fuel kernel ranging from 500 to 800 µm in diameter. The kernel and buffer layer are then coated with three layers, each of which is 30–40 µm thick, composed of dense inner pyrolytic carbon (IPyC), chemically vapor deposited silicon carbide (SiC) layer, and an outer pyrolytic carbon (OPyC) layer. These TRISO particles are then embedded in a fully dense sintered SiC matrix with an expected particle packing fraction of about 35–40% by volume. As is the case for gas reactor applications, the release of radioactivity into the coolant is dependent on the integrity of the silicon carbide layer of the TRISO particles, in addition to the SiC matrix. In this work, we report on fuel performance modeling of TRISO-bearing FCM fuel using the BISON code to simulate the thermo-mechanical behavior of this fuel in a prototypic LWR environment. This paper considers the effects of embedding a TRISO particle in the SiC pellet matrix and includes a discussion of the irradiation-induced dimensional change in the pyrolytic carbon (PyC) layers of the TRISO particle. Additionally, methods were developed to simulate a FCM pellet containing a large number of discrete and independent particles. Future work will report on developing an interface debonding model, a fracture model, and a radionuclide transport model.

Nitrogen supply and post-anthesis temperature affect the protein distribution gradient in the wheat grain endosperm, as measured using a novel image analysis technique for light microscopy sections., Gradients exist in the distribution of storage proteins in the wheat (Triticum aestivum) endosperm and determine the milling properties and protein recovery rate of the grain. A novel image analysis technique was developed to quantify both the gradients in protein concentration, and the size distribution of protein bodies within the endosperm of wheat plants grown under two different (20 or 28 °C) post-anthesis temperatures, and supplied with a nutrient solution with either high or low nitrogen content. Under all treatment combinations, protein concentration was greater in the endosperm cells closest to the aleurone layer and decreased towards the centre of the two lobes of the grain, i.e. a negative gradient. This was accompanied by a decrease in size of protein bodies from the outer to the inner endosperm layers in all but one of the treatments. Elevated post-anthesis temperature had the effect of increasing the magnitude of the negative gradients in both protein concentration and protein body size, whilst limiting nitrogen supply decreased the gradients.

Highlights • Widely differing free asparagine concentration in wheat from two harvest years. • Wide range of acrylamide-forming potential (AFP) in 73 commercial wheat varieties. • Eight varieties identified with consistently low AFP, seven of them soft types, but benefit of using a low AFP variety is lost if sulphur supply is inadequate. • Selecting varieties for low AFP solely on the basis of them being soft would be simplistic. • Information on free asparagine concentration should be available at variety launch., Free asparagine concentration, which is the determining factor for acrylamide-forming potential in cereals, was measured in grain from wheat grown in field trials in the United Kingdom in 2011–2012 and 2012–2013. There were 25 varieties in 2012 and 59 in 2013, with eleven present in both trials. The trials were split-plot, with half of each plot supplied with sulphur and the other half not. The varietal means (mmol per kg) for free asparagine in the sulphur-fed wheat ranged from 1.521 to 2.687 in 2011–2012 and 0.708 to 11.29 in 2012–2013. Eight varieties were identified as having consistently low free asparagine concentration. There was a differential response of varieties to sulphur, and much higher levels of free asparagine in 2012–2013 versus 2011–2012. Given the short commercial lifespan of some wheat varieties, it is concluded that information on free asparagine concentration should be made available when a variety is launched.

Asymmetry in the outer membrane has long defined the cell envelope of Gram-negative bacteria. This asymmetry, with lipopolysaccharide (LPS) or lipooligosaccharide (LOS) exclusively in the outer leaflet of the membrane, establishes an impermeable barrier that protects the cell from a number of stressors in the environment. Work done over the past 5 years has shown that Acinetobacter baumannii has the remarkable capability to survive with inactivated production of lipid A biosynthesis and the absence of LOS in its outer membrane. The implications of LOS-deficient A. baumannii are far-reaching – from impacts on cell envelope biogenesis and maintenance, bacterial physiology, antibiotic resistance, and virulence. This review examines recent work that has contributed to our understanding of LOS-deficiency and compares it to studies done on Neisseria meningitidis and Moraxella catarrhalis; the two other organisms with this capability.

Engineering demonstration reactors are nuclear reactors built to establish proof of concept for technology options that have never been built. Examples of engineering demonstration reactors include Peach Bottom 1 for high temperature gas-cooled reactors and the Experimental Breeder Reactor-II for sodium-cooled fast reactors. Engineering demonstrations have historically played a vital role in advancing the technology readiness level of reactor concepts. This paper details a preconceptual design for a fluoride salt-cooled engineering demonstration reactor. The fluoride salt-cooled high-temperature reactor (FHR) demonstration reactor (DR) is a concept for a salt-cooled reactor with 100 megawatts of thermal output. It would use tristructural-isotropic (TRISO) particle fuel in compacts within prismatic graphite blocks. FLiBe (2 7LiF-BeF2) is the reference primary coolant. The FHR DR is designed to be small, simple, and affordable. Development of the FHR DR is an intermediate step to enable near-term commercial FHRs. The design philosophy of the FHR DR was focused on safety, near-term deployment, and flexibility. Lower risk technologies are purposely included in the initial FHR DR design to ensure that the reactor can be built, licensed, and operated as an engineering demonstration with minimal risk and cost. These technologies include TRISO particle fuel, replaceable core structures, and consistent structural material selection for core structures and the primary and intermediate loops, and tube-and-shell primary-to-intermediate heat exchangers. Important capabilities to be demonstrated by building and operating the FHR DR include: • core design methodologies, • heat exchanger performance (including passive decay heat removal), • pump performance, • reactivity control, • salt chemistry control to maximize plant life, • salt procurement, handling, maintenance and ultimate disposal, and • tritium management. Non-nuclear separate and integral test efforts (e.g., heated salt loops or loops using simulant fluids) are necessary to develop the technologies that will be demonstrated in the FHR DR.

Aim: With the advent of rapid metabolic profiling techniques and of portable mass spectrometers we examined whether cells distinguished by their cytology and persistence of human papillomavirus infection, could be easily differentiated by their metabolite profile. Materials & methods: Direct injection electrospray mass spectrometry was used in a nontargeted double-blind experiment. Samples were collected from women diagnosed with one of two grades of cervical cytology and exhibiting either human papilloma virus persistence or clearance. Cell extracts were prepared using a DNA extraction procedure and the resulting supernatant, normally discarded, was analyzed. Data were interpreted using principal component analysis. Results: The results indicate strongly that a simple metabolite profiling method could be used to rapidly identify women at increased risk of cervical cancer.

Highlights • AX was determined in wheat grain after commercial and laboratory alcohol production. • Samples from laboratory scale production were similar to those from commercial processing. • The concentration, solubility and dynamic viscosity of AX increased during processing. • AX in DDGS was less variable in composition and properties than that in whole grain. • DDGS has potential to provide a source of AX for novel products., Laboratory produced DDGS samples were compared with commercial samples from a distillery and a biofuel plant. Changes in structure, solubility and content of arabinoxylan (AX) was determined. The distillation process results in a relative increase of AX content compared to the starting material. The heating and drying processes involved in the production of DDGS lead to an increased solubility and viscosity of water-extractable AX. Production of DDGS results in structural changes to the AX. There is a decrease in 2- and 3-linked arabinose oligosaccharides, that contributes to around a 50% reduction in arabinosylation in DDGS compared with the starting grains. The current study shows that laboratory-scale DDGS provide an accurate representation of the commercial scale and that the AX composition of DDGS is consistently uniform irrespective of starting material. The uniformity of DDGS and thin stillage makes them a good potential source of AX for production of prebiotics or other novel products.

Purpose To test the prebiotic activity of wheat arabinogalactan-peptide (AGP), which is a soluble dietary fibre composed of arabinogalactan polysaccharide linked to a 15-residue peptide, which accounts for up to 0.4% of the dry weight of wheat flour. Methods The prebiotic activity of AGP prepared from white wheat flour was tested using in vitro fermentation by colonic bacteria in automated pH-controlled anaerobic stirred batch cultures and compared to fructooligosaccharide (FOS) and wheat flour arabinoxylan (AX). Bacterial populations were measured using fluorescence in situ hybridisation (flow-FISH) and short chain fatty acid (SCFA) concentrations were measured using HPLC. Results Fermentation of AGP resulted in a significant bifidogenic activity and increased concentrations of SCFAs, mainly acetate after 24 h of fermentation. Conclusions These results were comparable to those obtained with AX and confirm the prebiotic potential of AGP. Furthermore, fermentation of a mixture of AGP and AX was faster compared to the single substrates and more similar to FOS, indicating that combinations of fermentable carbohydrates with different structures are potentially more effective as prebiotics than single substrates. When subcooled from the melt to any temperature between about 90 to 110 °C, polybutene-1 transforms at a readily measurable rate to a crystalline phase denoted “form 2,” which is typically spherulitic. The kinetics of this process have been measured by optical and dilatometric techniques. The results are consistent with the concepts that the initiation is heterogeneous, that the initiation is followed by spherical growth (Avrami n=3), and that the growth rate is nucleation controlled. Values arc obtained for the surface free energies and related quantities that are consistent with crystallization by chain folding: σ=7.2 erg/cm(2) or 7.2 mJ/m(2), σ(e)=15.5 erg/cm(2) or 15.5 mJ/m(2), and work of chain folding q=1.7 × 10(−13) erg/fold. (For comparison, q is 2.1 × 10(−13) erg/fold for polyethylene.) The work of chain folding of polybutene-1 is also compared with that of polychlorotrifluoroethylene, and it is shown that an increase of q is connected with an increase of equilibrium melting temperature. Crystal form 2 subsequently converts slowly near room temperature to a crystalline phase denoted “form 1” of different helicity and density, and the rate of this crystal-crystal transformation is also studied. The Avrami parameter for the form 2→form 1 process is n = 2. The process appears to be nucleation controlled. It is shown that form 1 is, everywhere below its melting point, the stable form. The equilibrium melting temperature for form 2 is estimated to be about 128 °C, and for form 1 about 141 °C. A degree of crystallinity scale based on specific volume is established. The degree of crystallinity of form 2 is about 52 percent, and after conversion to form 1, the crystallinity is about 77 percent.

Modification of structures of lipooligosaccharides (LOS) represents one prevalent mechanism by which Gram-negative bacteria can become resistant to key antibiotics. Owing to the significant complexity of LOS, the structural characterization of these amphipathic lipids has largely focused on elucidation of the lipid A substructures. Analysis of intact LOS enables detection of core oligosaccharide modifications and gives insight into the heterogeneity that results from combinations of lipid A and oligosaccharide substructures. Top-down analysis of intact LOS also provides the opportunity to determine unknown oligosaccharide structures, which is particularly advantageous in the context of glycoconjugate vaccine development. Advances in mass spectrometry technologies, including the development of MSn capabilities and alternative ion activation techniques, have made top-down analysis an indispensable tool for structural characterization of complex biomolecules. Here we combine online chromatographic separations with MS3 utilizing ultraviolet photodissociation (UVPD) and higher-energy collisional dissociation (HCD). HCD generally provides information about the presence of labile modifications via neutral loss fragments in addition to the saccharide linkage arrangement, whereas UVPD gives more detailed insight about saccharide branching and the positions of nonstoichiometric modifications. This integrated approach was used to characterize LOS from Acinetobacter baumannii 1205 and 5075. Notably, MS3 analysis of A. baumannii 1205, an antibiotic-resistant strain, confirmed phosphoethanolamine and hexosamine modification of the lipid A substructure and further enabled derivation of a core oligosaccharide structure.

Alcohol consumption is the third leading risk factor for disease and mortality in Europe. As evaluated by the International Agency for Research on Cancer (IARC) Monographs, a causal relationship is established for consumption of alcoholic beverages and cancers of the oral cavity, pharynx, larynx, oesophagus, liver, colorectum and female breast, even at low and moderate alcohol intakes. The higher the amount of alcohol consumed, the higher the risk of developing cancer. In Europe, an estimated 10% (95% CI: 7%–13%) of all cancer cases in men and 3% (95% CI: 1%–5%) of all cancer cases in women are attributable to alcohol consumption. Several biological mechanisms explain the carcinogenicity of alcohol; among them, ethanol and its genotoxic metabolite, acetaldehyde, play a major role. Taking all this evidence into account, a recommendation of the 4th edition of European Code against Cancer is: “If you drink alcohol of any type, limit your intake. Not drinking alcohol is better for cancer prevention.”

To test the hypothesis that the plant stress related elicitor cis-jasmone (cJ) provides protection in soybean pods against the seed-sucking stink bug pest, Euschistus heros, the growth of E. heros on cJ-treated pods was investigated using three soybean cultivars differing in insect susceptibility, i.e. BRS 134 (susceptible), IAC 100 (resistant) and Dowling (resistant). E. heros showed reduced weight gain when fed cJ-treated Dowling, whereas no effect on weight gain was observed when fed other treated cultivars. Using analysis of variance, a three factor (cultivar x treatment x time) interaction was observed with concentrations of the flavonoid glycosides daidzin and genistin, and their corresponding aglycones, daidzein and genistein. There were increases in genistein and genistin concentrations in cJ-treated Dowling at 144 and 120 h post treatment, respectively. Higher concentrations of malonyldaidzin and malonylgenistin in Dowling, compared to BRS 134 and IAC 100, were observed independently of time, the highest concentrations being observed in cJ-treated seeds. Levels of glycitin and malonylglycitin were higher in BRS 134 and IAC 100 compared to Dowling. Canonical variate analysis indicated daidzein (in the first two canonical variates) and genistein (in the first only) as important discriminatory variables. With the increasing occurrence of antibiotic resistance among Acinetobacter sp., the race is on for researchers to not only isolate resistant isolates but also utilize basic and applied microbiological techniques to study mechanisms of resistance. For many antibiotics, the limit of efficacy against Gram-negative bacteria is dependent on its ability to permeate the outer membrane and access its target. As such, it is critical that researchers be able to isolate and analyze the lipid components of the cell envelope from any number of Acinetobacter sp. that are either resistant or sensitive to antibiotics of interest. The following chapter provides in-depth protocols to confirm the presence or absence of lipooligosaccharide (LOS) in Acinetobacter sp., isolate lipid A, and glycerophospholipids and analyze them using qualitative (mass spectrometry) and semiquantitative (thin-layer chromatography) methods.

The outer membrane of Gram-negative bacteria is a critical barrier that prevents entry of noxious compounds. Integral to this functionality is the presence of lipopolysaccharide (LPS) or lipooligosaccharide (LOS), a molecule that is located exclusively in the outer leaflet of the outer membrane. Its lipid anchor, lipid A, is a glycolipid whose hydrophobicity and net negative charge are primarily responsible for the robustness of the membrane. Because of this, lipid A is a hallmark of Gram-negative physiology and is generally essential for survival. Rare exceptions have been described, including Acinetobacter baumannii, which can survive in the absence of lipid A, albeit with significant growth and membrane permeability defects. Here, we show by an evolution experiment that LOS-deficient A. baumannii can rapidly improve fitness over the course of only 120 generations. We identified two factors which negatively contribute to fitness in the absence of LOS, Mla and PldA. These proteins are involved in glycerophospholipid transport (Mla) and lipid degradation (PldA); both are active only on mislocalized, surface-exposed glycerophospholipids. Elimination of these two mechanisms was sufficient to cause a drastic fitness improvement in LOS-deficient A. baumannii. The LOS-deficient double mutant grows as robustly as LOS-positive wild-type bacteria while remaining resistant to the last-resort polymyxin antibiotics. These data provide strong biological evidence for the directionality of Mla-mediated glycerophospholipid transport in Gram-negative bacteria and furthers our knowledge of asymmetry-maintenance mechanisms in the context of the outer membrane barrier.

Malaria parasites (Plasmodium) can change the attractiveness of their vertebrate hosts to Anopheles vectors, leading to a greater number of vector-host contacts and increased transmission. Indeed, naturally Plasmodium-infected children have been shown to attract more mosquitoes than parasite-free children. Here, we demonstrate Plasmodium-induced increases in the attractiveness of skin odor in Kenyan children, and reveal quantitative differences in the production of specific odor components in infected, versus parasite-free, individuals. We found the aldehydes heptanal, octanal and nonanal to be produced in greater amounts by infected individuals, and detected by mosquito antennae. In behavioral experiments, we demonstrated that these, and other, Plasmodium-induced aldehydes enhanced the attractiveness of a synthetic odor blend mimicking ‘healthy’ human odor. Heptanal alone increased the attractiveness of ‘parasite-free’ natural human odor. Should the increased production of these aldehydes by Plasmodium-infected humans lead to increased mosquito biting in a natural setting, this would likely affect the transmission of malaria.

This paper presents analyses of possible reactor representations of a nuclear fuel cycle with continuous recycling of thorium and produced uranium (mostly U-233) with thorium-only feed. The analysis was performed in the context of a U.S. Department of Energy effort to develop a compendium of informative nuclear fuel cycle performance data. The objective of this paper is to determine whether intermediate spectrum systems, having a majority of fission events occurring with incident neutron energies between 1 eV and 105 eV, perform as well as fast spectrum systems in this fuel cycle. The intermediate spectrum options analyzed include tight lattice heavy or light water-cooled reactors, continuously refueled molten salt reactors, and a sodium-cooled reactor with hydride fuel. All options were modeled in reactor physics codes to calculate their lattice physics, spectrum characteristics, and fuel compositions over time. Based on these results, detailed metrics were calculated to compare the fuel cycle performance. These metrics include waste management and resource utilization, and are binned to accommodate uncertainties. The performance of the intermediate systems for this self-sustaining thorium fuel cycle was similar to a representative fast spectrum system. However, the number of fission neutrons emitted per neutron absorbed limits performance in intermediate spectrum systems.

Males of Acanthoscelides obtectus (Coleoptera: Chrysomelidae, Bruchinae) emit methyl (E,R)-2,4,5-tetradecatrienoate that attracts females for mating. This study identified further roles for this compound in the sexual behavior of A. obtectus. Earlier observations revealed that males touched females with their antennae while tandem-running with them and initiated mounting and copulation, whereas they showed no such behavior toward other males. A series of subsequent laboratory choice tests were set up to establish if certain cuticular compounds aid contact sex recognition in A. obtectus. Males chose virgin females over other males. The activity toward females could be eliminated by rinsing with hexane, but was regained by application of female extract onto previously rinsed females. Gas chromatographic (GC) comparison of hexane extracts revealed the presence of two male-specific compounds, methyl (E,R)-2,4,5-tetradecatrienoate and octadecanal, which were absent from the behaviorally active female samples. Of the two compounds, methyl (E,R)-2,4,5-tetradecatrienoate was found to be responsible for the inhibition of male sexual behavior, similar to that observed with crude male extracts applied to virgin females. Furthermore, males preferred virgin over mated females. GC analyses revealed the presence of methyl (E,R)-2,4,5-tetradecatrienoate in mated females in amounts sufficient to curtail mating attempts. It appears that methyl (E,R)-2,4,5-tetradecatrienoate, besides being a male-produced sex pheromone, acts as a male-recognition signal in A. obtectus. Males also transfer it onto females during mating, resulting in mated females being avoided by courting males.

Since starch is by far the major component of the mature wheat grain, it has been assumed that variation in the capacity for starch synthesis during grain filling can influence final grain weight. We investigated this assumption by studying a total of 54 wheat genotypes including elite varieties and landraces that were grown in two successive years in fields in the east of England. The weight, water content, sugars, starch, and maximum catalytic activities of two enzymes of starch biosynthesis, ADP-glucose pyrophosphorylase and soluble starch synthase, were measured during grain filling. The relationships between these variables and the weights and starch contents of mature grains were analysed. Final grain weight showed few or no significant correlations with enzyme activities, sugar levels, or starch content during grain filling, or with starch content at maturity. We conclude that neither sugar availability nor enzymatic capacity for starch synthesis during grain filling significantly influenced final grain weight in our field conditions. We suggest that final grain weight may be largely determined by developmental processes prior to grain filling. Starch accumulation then fills the grain to a physical limit set by developmental processes. A study analyzed the neutronics of alternate cladding materials in a pressurized water reactor (PWR) environment. Austenitic type 310 (310SS) and 304 stainless steels, ferritic Fe-20Cr-5Al (FeCrAl) and APMT™ alloys, and silicon carbide (SiC)-based materials were considered and compared with Zircaloy-4. SCALE 6.1 was used to analyze the associated neutronics penalty/advantage, changes in reactivity coefficients, and spectral variations once a transition in the cladding was made. In the cases examined, materials containing higher absorbing isotopes invoked a reduction in reactivity due to an increase in neutron absorption in the cladding. Higher absorbing materials produced a harder neutron spectrum in the fuel pellet, leading to a slight increase in plutonium production. A parametric study determined the geometric conditions required to match cycle length requirements for each alternate cladding material in a PWR. A method for estimating the end of cycle reactivity was implemented to compare each model to that of standard Zircaloy-4 cladding. By using a thinner cladding of 350 μm and keeping a constant outer diameter, austenitic stainless steels require an increase of no more than 0.5 wt% enriched 235U to match fuel cycle requirements, while the required increase for FeCrAl was about 0.1%. When modeling SiC (with slightly lowermore » thermal absorption properties than that of Zircaloy), a standard cladding thickness could be implemented with marginally less enriched uranium (~0.1%). Moderator temperature and void coefficients were calculated throughout the depletion cycle. Nearly identical reactivity responses were found when coolant temperature and void properties were perturbed for each cladding material. By splitting the pellet into 10 equal areal sections, relative fission power as a function of radius was found to be similar for each cladding material. FeCrAl and 310SS cladding have a slightly higher fission power near the pellet’s periphery due to the harder neutron spectrum in the system, causing more 239Pu breeding. An economic assessment calculated the change in fuel pellet production costs for use of each cladding. Furthermore, implementing FeCrAl alloys would increase fuel pellet production costs about 15% because of increased 235U enrichment and the additional UO2 pellet volume enabled by using thinner cladding.« less

Hypoglycemia is the most common complication of diabetes, causing morbidity and death. Recurrent hypoglycemia alters the cascade of physiological and behavioral responses that maintain euglycemia. The extent to which these responses are normally triggered by decreased whole-brain cerebral glucose metabolism (CMRglc) has not been resolved by previous studies. We measured plasma counterregulatory hormonal responses and whole-brain CMRglc (along with blood-to-brain glucose transport rates and brain glucose concentrations) with 1-[11C]-d-glucose positron emission tomography during hyperinsulinemic glucose clamps at nominal plasma glucose concentrations of 90, 75, 60, and 45 mg/dL (5.0, 4.2, 3.3, and 2.5 mmol/L) in 18 healthy young adults. Clear evidence of hypoglycemic physiological counterregulation was first demonstrated between 75 mg/dL (4.2 mmol/L) and 60 mg/dL (3.3 mmol/L) with increases in both plasma epinephrine (P = 0.01) and glucagon (P = 0.01). In contrast, there was no statistically significant change in CMRglc (P = 1.0) between 75 mg/dL (4.2 mmol/L) and 60 mg/dL (3.3 mmol/L), whereas CMRglc significantly decreased (P = 0.02) between 60 mg/dL (3.3 mmol/L) and 45 mg/dL (2.5 mmol/L). Therefore, the increased epinephrine and glucagon secretion with declining plasma glucose concentrations is not in response to a decrease in whole-brain CMRglc.

Carotenoid coloration is widely recognized as a signal of individual condition in various animals, but despite decades of study, the mechanisms that link carotenoid coloration to condition remain unresolved. Most birds with red feathers convert yellow dietary carotenoids to red carotenoids in an oxidation process requiring the gene encoding the putative cytochrome P450 enzyme CYP2J19. Here, we tested the hypothesis that the process of carotenoid oxidation and feather pigmentation is functionally linked to mitochondrial performance. Consistent with this hypothesis, we observed high levels of red ketolated carotenoids associated with the hepatic mitochondria of molting wild house finches (Haemorhous mexicanus), and upon fractionation, we found the highest concentration of ketolated carotenoids in the inner mitochondrial membrane. We further found that the redness of growing feathers was positively related to the performance of liver mitochondria. Structural modeling of CYP2J19 supports a direct role of this protein in carotenoid ketolation that may be functionally linked to cellular respiration. These observations suggest that feather coloration serves as a signal of core functionality through inexorable links to cellular respiration in the mitochondria.

Free amino acids and reducing sugars participate in the Maillard reaction during high-temperature cooking and processing. This results not only in the formation of colour, aroma and flavour compounds, but also undesirable contaminants, including acrylamide, which forms when the amino acid that participates in the reaction is asparagine. In this study, tubers of 13 varieties of potato (Solanum tuberosum), which had been produced in a field trial in 2010 and sampled immediately after harvest or after storage for 6 months, were analysed to show the relationship between the concentrations of free asparagine, other free amino acids, sugars and acrylamide-forming potential. The varieties comprised five that are normally used for crisping, seven that are used for French fry production and one that is used for boiling. Acrylamide formation was measured in heated flour, and correlated with glucose and fructose concentration. In French fry varieties, which contain higher concentrations of sugars, acrylamide formation also correlated with free asparagine concentration, demonstrating the complex relationship between precursor concentration and acrylamide-forming potential in potato. Storage of the potatoes for 6 months at 9°C had a significant, variety-dependent impact on sugar and amino acid concentrations and acrylamide-forming potential.

The expression of genes encoding components of ABA and metabolic signalling pathways in developing barley endosperm and embryo was investigated. The genes included HvRCAR35_47387 and HvRCAR35_2538 (encoding ABA receptors), HvABI1d (protein phosphatase 2C), HvSnRK2.4, HvSnRK2.6 and HvPKABA1 (SnRK2-type protein kinases) and HvABI5 (ABA response element binding protein; AREBP), as well as two genes encoding SnRK1-type protein kinases. Both SnRK1 and SnRK2 phosphorylate AREBPs, but SnRK2 is activated by ABA whereas SnRK1 may be broken down. Multiple cereal AREBPs with two conserved SnRK1/2 target sites and another class of BZIP transcription factors with SnRK1/2 binding sites, including HvBLZ1, were identified. Barley grain (cv. Triumph) was sampled at 15, 20, 25 and 30 days post-anthesis (dpa). HvRCAR35_47387, HvABI1d, HvSnRK2.4 and HvABI5 were expressed highly in the endosperm but at much lower levels in the embryo. Conversely, HvPKABA1 and HvRCAR35_2538 were expressed at higher levels in the embryo than the endosperm, while HvSnRK2.6 was expressed at similar levels in both. HvRCAR35_47387, HvABI1d, HvSnRK2.4 and HvABI5 all peaked in expression in the endosperm at 20 dpa. A model is proposed in which ABA brings about a transition from a SnRK1-dominated state in the endosperm during grain filling to a SnRK2-dominated state during maturation., Highlights • Components of ABA signalling pathway differ between endosperm and embryo in barley. • ABA signalling pathway components are highly expressed in barley endosperm at 20 dpa. • ABA may bring about change from SnRK1-dominated state to SnRK2-dominated state. • This may play key role in transition from grain filling to maturation in barley. • BLZ1/OHP1 class of bZIP transcription factors contain multiple SnRK1/2 target sites.

The starchy endosperm is the major storage tissue in the mature wheat grain and exhibits quantitative and qualitative gradients in composition, with the outermost cell layers being rich in protein, mainly gliadins, and the inner cells being low in protein but enriched in high-molecular-weight (HMW) subunits of glutenin. We have used sequential pearling to produce flour fractions enriched in particular cell layers to determine the protein gradients in four different cultivars grown at two nitrogen levels. The results show that the steepness of the protein gradient is determined by both genetic and nutritional factors, with three high-protein breadmaking cultivars being more responsive to the N treatment than a low-protein cultivar suitable for livestock feed. Nitrogen also affected the relative abundances of the three main classes of wheat prolamins: the sulfur-poor ω-gliadins showed the greatest response to nitrogen and increased evenly across the grain; the HMW subunits also increased in response to nitrogen but proportionally more in the outer layers of the starchy endosperm than near the core, while the sulfur-rich prolamins showed the opposite trend.

Acrylamide is a probable human carcinogen that forms in plant-derived foods when free asparagine and reducing sugars react at high temperatures. The identification of rye varieties with low acrylamide-forming potential or agronomic conditions that produce raw material with low acrylamide precursor concentrations would reduce the acrylamide formed in baked rye foods without the need for additives or potentially costly changes to processes. This work compared five commercial rye varieties grown under a range of fertilisation regimes to investigate the effects of genotype and nutrient (nitrogen and sulphur) availability on the accumulation of acrylamide precursors. A strong correlation was established between the free asparagine concentration of grain and the acrylamide formed upon heating. The five rye varieties accumulated different concentrations of free asparagine in the grain, indicating that there is genetic control of this trait and that variety selection could be useful in reducing acrylamide levels in rye products. High levels of nitrogen fertilisation were found to increase the accumulation of free asparagine, showing that excessive nitrogen application should be avoided in order not to exacerbate the problem of acrylamide formation. This effect of nitrogen was mitigated in two of the varieties by the application of sulphur., Highlights ► Acrylamide-forming potential is a key quality parameter for rye grain. ► Free asparagine level is main determinant of acrylamide-forming potential in rye. ► Rye varieties differ significantly in free asparagine concentration in the grain. ► High levels of nitrogen (N) increase the acrylamide-forming potential of rye. ► Sulphur reduces the effect of N on acrylamide-forming potential in some varieties.

Microglia are the intrinsic immune sentinels of the central nervous system. Their activation restricts tissue injury and pathogen spread, but in some settings, including viral infection, this response can contribute to cell death and disease. Identifying mechanisms that control microglial responses is therefore an important objective. Using replication-incompetent adenovirus 5 (Ad5)-based vectors as a model, we investigated the mechanisms through which microglia recognize and respond to viral uptake. Transgenic, immunohistochemical, molecular-genetic, and fluorescence imaging approaches revealed that phosphatidylserine (PtdSer) exposure on the outer leaflet of transduced cells triggers their engulfment by microglia through TAM receptor-dependent mechanisms. We show that inhibition of phospholipid scramblase 1 (PLSCR1) activity reduces intracellular calcium dysregulation, prevents PtdSer externalization, and enables months-long protection of vector-transduced, transgene-expressing cells from microglial phagocytosis. Our study identifies PLSCR1 as a potent target through which the innate immune response to viral vectors, and potentially other stimuli, may be controlled.

The objective of this study was to evaluate the natural levels of the plant defence compound DIMBOA (2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one) in young leaves of eight hexaploid Brazilian wheat genotypes and the impact of the genotypes upon development of cereal aphids, Rhopalosiphum padi and Sitobion avenae. HPLC analysis revealed that the DIMBOA levels varied from 5.376 (in BRS Guabiju) to 30.651 mmol kg FW−1 (in BRS Timbauva) with two genotypes outperforming Solstice, a UK variety used as reference. Bioassays were conducted to evaluate the development and fecundity of both aphids when grown on the wheat genotypes. Although BRS Guabiju and BRS Timbauva were among the genotypes showing the highest susceptibility and resistance, respectively, against both aphid species, no correlation could be found between DIMBOA levels and antibiosis effects. The cultivar BRS 327 was among the genotypes showing lower intrinsic rate of population increase for the two aphid species. Elucidating the role of secondary metabolites in plant resistance to aphids and the characterization of the genotypes that allowed reduced development of R. padi and S. avenae are important steps to achieve a better natural resistance in hexaploid Brazilian wheat.

Western blotting, ELISA and 1H-NMR spectroscopy showed that RNAi down-regulation of the wheat Gsp-1 gene resulted in reduced contents of both arabinogalactan peptide (AGP) and grain softness protein (GSP-1) in mature wheat grains confirming that these components are encoded by the same gene. A small increase in grain hardness and decrease in the viscosity of aqueous extracts of the transgenic lines also indicated small effects on functional properties. Immunolocalisation using a novel wheat AGP monoclonal antibody in conjunction with confocal microscopy showed that the major form of AGP which was eliminated in knockout lines is located within the cell, probably in the vacuole, and not in the plasma membrane or cell wall. However, clear localisation of the AGP epitope to the plasma membrane was observed in both control and transgenic lines and probably resulted from the presence of one or more separate forms of arabinogalactan protein. The existence of such additional form(s) was also indicated by 1H-NMR spectroscopy which showed that the ratio of arabinose to galactose differed between the control and transgenic lines.

European manufacturers’ data on acrylamide in potato crisps from 2002 to 2016 were analysed. A previous study showed a 53 % reduction in mean acrylamide levels from 763 ng g-1 in 2002 to 358 ng g-1 in 2011. Analysis of data from the longer period showed that since 2011 there has been a levelling off, with the mean level for 2016 being 412 ng g-1 (still a 46 % reduction from 2002), suggesting that the most effective acrylamide reduction measures had been devised and implemented by 2011. There were similar trends in the 90th and 95th quantile values, with the 90th quantile values being below 1000 ng g-1 (the European Commission’s current ‘Indicative Value’ for acrylamide in potato crisps) since 2010. The proportion of samples with acrylamide above 2000 ng g-1 fell from 4.8 % in 2002 to 0.6 % in 2016. Acrylamide levels showed marked seasonal variability, being highest in the first half of the year when potatoes were being used from storage, and lowest from July to September when potatoes were being harvested. Acrylamide levels were higher in thicker types of crisp in the early years of the study, but this difference disappeared in the later years, suggesting that manufacturers had acted to reduce acrylamide formation in these products. Higher values for acrylamide were recorded in north and east Europe than in the south and west up to 2013. Levels in the north and east declined in recent years, but remained higher in the north than in the other regions. The manufacturers’ data were compared with a much smaller dataset provided by the European Food Safety Authority (EFSA). Levels of acrylamide in the EFSA dataset were consistently higher than in the manufacturers’ data, possibly due to uneven sampling through the year and the seasonality of acrylamide levels.

Three experiments were conducted to compare the attraction and repulsion of second-stage juveniles (J2) of the root-knot nematode, Meloidogyne graminicola, to rice plants supplied with different forms of nitrogen. The rice plants were hydroponically grown in a full nutrient solution containing different concentrations (0.1 or 10.0 mM) of nitrate (NO3−) or forms of nitrogen supply (2.85 mM calcium nitrate (Ca(NO3)2), ammonium nitrate (NH4NO3) or ammonium chloride (NH4Cl)) for 2 weeks. Five rice plants were placed with their roots in one corner of a Perspex X or Y-chamber partly filled with agar and J2 were inoculated onto the chambers. The data show that J2 of M. graminicola were significantly attracted towards the roots of rice plants grown in hydroponics containing 0.1 mM NO3− and 2.85 mM Ca(NO3)2, but repelled by 10.0 mM NO3−, 2.85 mM NH4NO3 and NH4Cl. The results suggest that the application of ammonia-based nitrogen fertiliser to the rice nursery bed may interfere with nematode attraction and thus reduce invasion, and the application of chemical nitrification inhibitors to rice nursery beds may decrease nematode invasion.

Acrylamide forms from free asparagine and reducing sugars during frying, baking, roasting, or high-temperature processing, and cereal products are major contributors to dietary acrylamide intake. Free asparagine concentration is the determining factor for acrylamide-forming potential in cereals, and this study investigated the effect of fungicide application on free asparagine accumulation in wheat grain. Free amino acid concentrations were measured in flour from 47 varieties of wheat grown in a field trial in 2011–2012. The wheat had been supplied with nitrogen and sulfur and treated with growth regulators and fungicides. Acrylamide formation was measured after the flour had been heated at 180 °C for 20 min. Flour was also analyzed from 24 (of the 47) varieties grown in adjacent plots that were treated in identical fashion except that no fungicide was applied, resulting in visible infection by Septoria tritici, yellow rust, and brown rust. Free asparagine concentration in the fungicide-treated wheat ranged from 1.596 to 3.987 mmol kg–1, with a significant (p < 0.001 to p = 0.006, F test) effect of variety for not only free asparagine but all of the free amino acids apart from cysteine and ornithine. There was also a significant (p < 0.001, F test) effect of variety on acrylamide formation, which ranged from 134 to 992 μg kg–1. There was a significant (p < 0.001, F test) correlation between free asparagine concentration and acrylamide formation. Both free asparagine concentration and acrylamide formation increased in response to a lack of fungicide treatment, the increases in acrylamide ranging from 2.7 to 370%. Free aspartic acid concentration also increased, whereas free glutamic acid concentration increased in some varieties but decreased in others, and free proline concentration decreased. The study showed disease control by fungicide application to be an important crop management measure for mitigating the problem of acrylamide formation in wheat products.
