This study reports a SiN x channel optical waveguide on a SiO2-covered bulk Si platform. The waveguide shows a low propagation loss of ∼0.6 dB/cm at 1550 and 1310 nm wavelengths. The SiN x film is deposited by reactive sputtering and is free from N–H species, which cause optical absorption loss at ∼1520 nm. No post-deposition high-temperature annealing is required as an increase in sputtering temperature, from room temperature to a moderate temperature of 200 °C, is effective for low-loss propagation. Based on chemical and optical analyses, the quality of the sputtered film at 200 °C is comparable to that prepared by low-pressure chemical vapor deposition at a high temperature of 820 °C. Fabricated fundamental passive optical devices (a multi-mode interferometer splitter and a directional coupler) reveal reasonable device function. A ring resonator shows a thermally stable operation because of the small thermo-optic coefficient for the SiN x , on the order of 10−5 K−1. The results indicate that the SiN x film by reactive sputtering at 200 °C can be favorably applied for photonic integration concerning CMOS back-end processing and hybrid-integration processing with non-CMOS materials. A Si-on-insulator wafer is not necessarily used as the platform, but a low-cost bulk Si wafer is replaced depending on the application. The reduction in the SiO2 under-cladding thickness, as thin as 1.0 µm, is examined regarding the potential in multilayered interconnects.

Applications in biotechnology and synthetic biology often make use of soluble proteins, but there are many potential advantages to anchoring enzymes to a stable substrate, including stability and the possibility for substrate channeling. To avoid the necessity of protein purification and chemical immobilization, there has been growing interest in bio-assembly of protein-containing nanoparticles, exploiting the self-assembly of viral capsid proteins or other proteins that form polyhedral structures. But these nanoparticle are limited in size which constrains the packaging and the accessibility of the proteins. The axoneme, the insoluble protein core of the eukaryotic flagellum or cilium, is a highly ordered protein structure that can be several microns in length, orders of magnitude larger than other types of nanoparticles. We show that when proteins of interest are fused to specific axonemal proteins and expressed in living cells, they become incorporated into linear arrays which have the advantages of high protein loading capacity, high stability, and single-step purification with retention of biomass. The arrays can be isolated as membrane enclosed vesicle or as exposed protein arrays. The approach is demonstrated for both fluorescent proteins and enzymes, and in the latter case it is found that incorporation into axoneme arrays provides increased stability for the enzyme.

Hypercoagulability associated with malignant tumors causes thrombosis, termed Trousseau's syndrome, but is rarely associated with benign gynecological tumors, such as myoma and adenomyosis. We herein report a 47-year-old Japanese woman with uterine adenomyosis who developed multiple cerebral infarcts during menstruation. Edoxaban was initially used for prevention but failed to prevent recurrence of thrombosis. However, hysterectomy and bilateral salpingo-oophorectomy resulted in the successful prevention of recurrence of cerebral infarct for five years without antiplatelet or anticoagulant agents. In our patient, the surgical removal of adenomyosis was highly effective for preventing thrombosis in a patient with adenomyosis.

In this study, several nanometer-thick transition metal overlayers (M) were formed on an Fe–Cr–Al metal (SUS) foil by pulsed cathodic arc-plasma deposition. The thin-film materials prepared, exhibiting a full-coverage M overlayer (M/SUS), were applied as catalysts for NH3 oxidation in the presence of excess O2 (300 ppm NH3, 8% O2, and He balance). Under dry conditions (0% H2O in the gas feed), the observed NH3 oxidation activity decreased in the order of Ir > Pt > Co > Pd > Cu > Rh when the product selectivity was not taken into consideration. The activities of the materials composed of Co and Cu decreased significantly in the presence of 10% H2O, while those of the catalysts containing Pt and Ir remained nearly unchanged. Based on the kinetic analyses and X-ray photoelectron spectrometry experiments conducted, it was determined that the contrasting effect of H2O on the activity of M/SUS was related to the ease of oxidation of the non-precious metals studied. The resulting hydrophilic oxide surface was more susceptible to the competitive adsorption of H2O and thus inhibited the NH3 reaction than a metallic surface. The negative effect of H2O was evident by the decrease in the NH3 concentration in the gas feed. Notably, the metallic surface of the Pt overlayer was stable even under oxidizing reaction conditions; therefore, the oxidation of NH3 was less susceptible to the effects of high H2O concentrations.

Metastable vaterite blocks are expected to show high reactivity. However, they have not been reported to date. In this study, the feasibility of fabricating an artificial vaterite block was investigated by exposing a Ca(OH)2 compact to CO2 under several conditions. Under a 100% H2O atmosphere, a calcite block was formed within three days. Under a 100% methanol atmosphere, no carbonation was observed for seven days. Under 90% methanol without atmospheric flow, a vaterite block containing 5 mass% of unreacted Ca(OH)2 and 3 mass% of calcite was formed after 24 h. On day 7, it was transformed into a block with 70 mass% of calcite, 27 mass% of vaterite, and 3 mass% of unreacted Ca(OH)2. By contrast, under 90% methanol with atmospheric flow to remove the accumulated H2O in the compact, a pure CaCO3 block of 98 mass% vaterite and 2 mass% calcite was formed on day 3. Therefore, the regulation of the amount of H2O is important for fabricating vaterite blocks. Porosimetry measurements revealed that the calcite block fabricated under 90% methanol without atmospheric flow demonstrated a pore size profile closer to that of the vaterite block than that of the calcite block fabricated under 100% H2O atmosphere because the vaterite with small spherical crystals was formed initially before its transformation to calcite.

Carbonate apatite (CO3Ap) artificial bone has attracted much attention due to its high osteoconductivity and replacement to bone. CO3Ap is an inorganic component of bone, and has been fabricated by compositional transformation through a dissolution–precipitation reaction in Na2HPO4 solution using calcium carbonate as a precursor. In this study, the effects of the CO32− concentration in the Na2HPO4 solution on the CO3Ap fabrication reaction and the properties of the fabricated CO3Ap were investigated. The rate of the compositional transformation became faster, and more CO3 containing CO3Ap was fabricated using CO32− concentrated in Na2HPO4 solution. The crystal shape of CO3Ap changed from scale-like to hexagonal crystals. In addition, the crystallite size increased with increasing CO32− concentration in the Na2HPO4 solution. Several oxides containing group 14 elements were characterized to investigate catalytic activity factors and the reaction mechanism for oxidative coupling of methane (OCM). Lithium-based silicate Li2SiO3 and germanate Li2GeO3 showed much higher selectivities for C2 hydrocarbons than their Sn-based counterpart Li2SnO3. Our XPS study suggested that strong metal–oxygen bonds in silicates and germanates could be of importance to effectively suppress the deep oxidation of methane at the catalyst surface, resulting in enhanced C2 selectivities. In contrast, weakly bound surface oxygen of Sn-based oxides is subject to direct oxygenation of methane to form COx as by-products, emphasizing the importance of metal–oxygen bonding strength. To achieve high OCM catalytic activity, effective methane activation by strong basicity is also required, and the local environment around oxygen sites could be a key factor to enhance CH4 conversion. Based on the chemical/structural viewpoints, we found that silicate and germanate with higher Li contents, Li4SiO4 and Li4GeO4, are significantly OCM active, comparable to the previously reported catalyst Li2CaSiO4.

[Background and Aims] Multiple developments of squamous dysplasia and squamous cell carcinoma (SCC) in the upper aerodigestive tract have been explained by field cancerization phenomenon and were associated with alcohol and cigarette use. Second primary SCC development after curative treatment impairs patients’ quality of life and survival; however, how these consumption and cessation affect field cancerization is still unknown. [Methods] This is a multicenter cohort study including 331 patients with superficial esophageal SCC (ESCC) treated endoscopically and pooled data from 1022 healthy subjects for comparison. Physiological condition in the background esophageal mucosa was classified into 3 groups based on the number of Lugol-voiding lesions (LVLs) per endoscopic view: grade A, 0; grade B, 1–9; or grade C, ≥10 LVLs. Lifestyle surveys were conducted using a self-administered questionnaire. Patients were counseled on the need for alcohol and smoking cessation by physicians and were endoscopically surveyed every 6 months. [Results] LVL grades were positively associated with alcohol drinking intensity, flushing reactions, smoking, and high-temperature food and were negatively associated with eating green and yellow vegetables and fruit. Second primary ESCC and head/neck SCC were significantly more prevalent in the grade C LVL (cumulative 5-y incidences 47.1%, 95% confidence interval [CI] = 38.0–57.2 and 13.3%, 95% CI = 8.1–21.5, respectively). Alcohol and smoking cessation significantly reduced the development of second primary ESCC (adjusted hazard ratios 0.47, 95% = CI 0.26–0.85 and 0.49, 95% CI = 0.26–0.91, respectively). [Conclusion] Alcohol drinking, smoking, flushing reaction, and high-temperature food were closely associated with field cancerization, and cessation of alcohol and smoking significantly reduced the risk of development of second primary cancer. Dinaphtho[2,1-b:1′,2′-d]thiophene (DNT) is one of the “dynamic thiahelicenes” that is obtained as a racemate due to its low energy barrier for enantiomerization. Careful evaluation of both the metastable DNT film and the stable films of DNT/polymethyl metacrylate (PMMA) blends revealed that DNT has an extra-high refractive index of 1.808 at 633 nm, which is rarely achieved in organic small molecules. The fundamental factors responsible for the unusual observation of DNT were explored by comparing the properties of DNT with those of a flat-shaped dibenzothiophene (DBT) both in experiments and DFT calculations, and by analysing them with the Lorenz-Lorentz equation. It became clear that the unique properties of DNT were based on its flexible dynamic thiahelicene structure, leading to the metastable film-forming ability, the high compatibility with PMMA, and the exhibition of an extra-high refractive index caused by the dense packing in the amorphous film. Chemical modification of DNT provided new halogen-free vinyl monomers, which could be easily polymerized to form vinyl polymers with high refractive indices of 1.72-1.74, as well as film-forming ability, high thermal stability, and high transparency in the visible region. Thus, we could demonstrate that the DNT structure is a new promising candidate for organic polymeric high refractive index materials.

Scaffolds capable of inducing ectopic osteogenesis (i.e., osteoinduction) have the potential to treat congenital and massive bone defects. However, the mechanisms underlying osteoinduction remain elusive, largely because of rapid tissue changes occurring early in the process. A model with decelerated osteoinduction is, therefore, needed. Here, we developed carbonate apatite honeycomb scaffolds capable of osteoinduction in rabbit muscle, which inherently exhibits minimal osteoinduction. The key events observed were: (1) fibrin accumulation in scaffolds at week 1; (2) appearance of tartrate‐resistant acid phosphatase (TRAP)‐ and RAM11‐positive cells and formation of collagen fibers and blood vessels between weeks 2 and 3; (3) appearance of osteocalcin‐positive cells and decrease in the number of TRAP‐positive cells between weeks 3 and 4; (4) continuous proliferation of osteocalcin‐positive cells at postoperative week 4 and formation of osteoid and mineralized tissues at weeks 8 and 12, respectively. These findings suggest that the appearance of TRAP‐positive cells that are considered foreign body giant cells is essential for collagen formation, angiogenesis, and osteogenesis. These insights may aid in the development of scaffolds for bone tissue engineering.

Impaired calcium uptake resulting from reduced expression and activity of the cardiac sarco-endoplasmic reticulum Ca2+ ATPase (SERCA2a) is a hallmark of heart failure (HF). Recently, new mechanisms of SERCA2a regulation, including post-translational modifications (PTMs), have emerged. Our latest analysis of SERCA2a PTMs has identified lysine acetylation as another PTM which might play a significant role in regulating SERCA2a activity. SERCA2a is acetylated, and that acetylation is more prominent in failing human hearts. In this study, we confirmed that p300 interacts with and acetylates SERCA2a in cardiac tissues. Several lysine residues in SERCA2a modulated by p300 were identified using in vitro acetylation assay. Analysis of in vitro acetylated SERCA2a revealed several lysine residues in SERCA2a susceptible to acetylation by p300. Among them, SERCA2a Lys514 (K514) was confirmed to be essential for SERCA2a activity and stability using an acetylated mimicking mutant. Finally, the reintroduction of an acetyl-mimicking mutant of SERCA2a (K514Q) into SERCA2 knockout cardiomyocytes resulted in deteriorated cardiomyocyte function. Taken together, our data demonstrated that p300-mediated acetylation of SERCA2a is a critical PTM that decreases the pump's function and contributes to cardiac impairment in HF. Streptococcus gordonii, one of the early colonizers of oral biofilms, is involved in the development of dental caries, periodontal disease, and infective endocarditis. The Hsa adhesin of S. gordonii DL1 has the ability to bind strongly to the terminal sialic acid groups of host glycoproteins via the binding region, NR2, which is important for the pathogenicity of S. gordonii DL1. Low similarity with the NR2 of Hsa homologs among other streptococcal species has been reported. However, the reports have been limited to certain strains. This study attempted to assess frequency of the expression on the bacterial cell surface and to analyze the diversity of Hsa homologs among different wild strains of oral streptococci. We isolated 186 wild-type strains of oral streptococci from healthy volunteers and analyzed their hemagglutinating activity on human erythrocytes and their Hsa homologs and NR2 homologous regions by dot immunoblotting using anti-Hsa and anti-NR2 antisera, respectively. We found 30 strains reacted with anti-NR2 antiserum (NR2-positive) and determined the sequence of the NR2 regions. Many strains with high hemagglutinating activity were also NR2-positive, suggesting that the NR2 region may be associated with hemagglutinating activity. Among the NR2-positive strains, four different amino acid sequence patterns were observed, demonstrating diversity in the NR2 region. Notably, S. gordonii strains frequently possessed Hsa homologs and NR2-like antigens compared to other streptococci. It is speculated that the possessing frequency of Hsa homologs and the amino acid sequence of NR2 region may vary among streptococcal species. This article is protected by copyright. Carbonate apatite (CO3 Ap), an inorganic component of human bone, has been clinically applied as an artificial bone substitute. In this study, the effects of the CO3 content in CO3 Ap on the replacement by new bone were studied by fabricating CO3 Ap granules containing 0.9-8.3 wt% of CO3 . The dissolution rate of CO3 Ap in a weak acidic solution, mimicking the Howship's lacunae, was rapid for the CO3 Ap granules containing a larger amount of CO3 . Histological analyses demonstrated the rapid resorption in CO3 Ap and replacement by natural bone tissue when the CO3 content was increased. Therefore, the CO3 content in CO3 Ap is a key factor that influences the replacement of the bone tissue. This article is protected by copyright. All rights reserved.

Low-temperature plasma (LTP) is a partially ionized gas that contains electrons, ions, radicals, light, etc. Recently, the bio-medical application of LTP has become a hot topic in plasma science and biological science. Cancer treatment with plasma is the most challenging topic in plasma bio-medical applications. Many in vitro and in vivo experiments have been conducted to investigate the anti-tumor effects of LTP. Extracellular reactive oxygen and nitrogen species (RONS) in plasma-activated solutions are key factors for the anti-tumor effects, and amino acid modifications by LTP may affect cellular responses. Intracellular RONS are also key factors for the anti-tumor effects. Various signaling pathways, such as p53 signaling pathways, survival and proliferation signaling pathways, and oxidative stress-dependent signaling pathways are activated by LTP.

Bone apatite is not hydroxyapatite (HAp), it is carbonate apatite (CO3Ap), which contains 6–9 mass% carbonate in an apatitic structure. The CO3Ap block cannot be fabricated by sintering because of its thermal decomposition at the sintering temperature. Chemically pure (100%) CO3Ap artificial bone was recently fabricated through a dissolution–precipitation reaction in an aqueous solution using a precursor, such as a calcium carbonate block. In this paper, methods of fabricating CO3Ap artificial bone are reviewed along with their clinical and animal results. CO3Ap artificial bone is resorbed by osteoclasts and upregulates the differentiation of osteoblasts. As a result, CO3Ap demonstrates much higher osteoconductivity than HAp and is replaced by new bone via bone remodeling. Granular-type CO3Ap artificial bone was approved for clinical use in Japan in 2017. Honeycomb-type CO3Ap artificial bone is fabricated using an extruder and a CaCO3 honeycomb block as a precursor. Honeycomb CO3Ap artificial bone allows vertical bone augmentation. A CO3Ap-coated titanium plate has also been fabricated using a CaCO3-coated titanium plate as a precursor. Quasicrystals (QCs), first discovered in 1984, generally do not exhibit long-range magnetic order. Here, we report on long-range magnetic order in the real icosahedral quasicrystals (i QCs) Au–Ga–Gd and Au–Ga–Tb. The Au65Ga20Gd15i QC exhibits a ferromagnetic transition at TC = 23 K, manifested as a sharp anomaly in both magnetic susceptibility and specific heat measurements, along with an appearance of magnetic Bragg peak below TC. This is the first observation of long-range magnetic order in a real quasicrystal, in contrast to the spin-glass-like behaviors observed for the other magnetic quasicrystals found to date. Moreover, when Gd is replaced by Tb, i.e., for the Au65Ga20Tb15i QC, a ferromagnetic behavior is still retained with TC = 16 K. Although the sharp anomaly in the specific heat observed for the Au65Ga20Gd15i QC becomes broadened upon Tb substitution, neutron diffraction experiments clearly show marked development of magnetic Bragg peaks just below TC, indicating long-range magnetic order for the Au65Ga20Tb15i QC also. Our findings can contribute to the further investigation of exotic magnetic orders formed on real quasiperiodic lattices with unprecedented highest global symmetry, i.e., icosahedral symmetry.

The living cationic polymerizations of sulfide-containing vinyl ethers, namely, alkyl-sulfide-containing 2-(ethylthio)ethyl vinyl ether (ESEVE) and arylsulfide-containing 2-(phenylthio)ethyl and 2-[4-(methylthio)phenoxy]ethyl vinyl ethers (PSEVE and MSPEVE, respectively), were examined using HCl/ZnCl2, 1-(isobutoxy)ethyl acetate (IBEA)/Et1.5AlCl1.5, and IBEA /Et1.5AlCl1.5/SnCl4 as initiators. The polymerization of ESEVE terminated at a 35% monomer conversion, and a high-molecular-weight polymer was not obtained; instead, the nucleophilicity of the sulfur moiety in ESEVE led to cyclization, forming five-membered ring sulfonium species. In contrast, the side reactions were suppressed when PSEVE was used, which has a less-nucleophilic sulfur moiety on its benzene ring, and polymerization proceeded to an 88% conversion, affording a high-molecular-weight polymer with a narrow molecular weight distribution (Mw = 12,500, Mw/Mn = 1.18). Furthermore, cyclic sulfonium was not produced during the polymerization of MSPEVE, in which the sulfur atom was farther from the generated carbocation; the polymerization proceeded quantitatively producing a high-molecular-weight polymer with a narrow molecular weight distribution, despite the short lifetime of the growing species. In particular, a high-molecular-weight polymer with the narrowest distribution (Mw = 13,500, Mw/Mn = 1.08) was obtained when SnCl4 was used as the Lewis acid.

PURPOSE To identify the clinical factors resulting in the failure of dutasteride add-on treatment to alpha-adrenergic antagonist for patients with lower urinary tract symptoms and benign prostatic enlargement (BPE). METHODS We retrospectively surveyed the patient cohort who had been enrolled in the study of dutasteride add-on treatment to alpha-adrenergic antagonist from December 2009 to November 2011. Treatment failure was defined as receiving surgery for BPE or requiring intermittent catheterization or permanent bladder catheter for urinary retention or huge postvoid residual urine. Clinical parameters before dutasteride treatment were compared between the successful and failed group. RESULTS Of 92 patients, 23 (25%) were defined as treatment failure at 7-109 months (mean: 38 months) after dutasteride add-on treatment. In the failed group, the patient' age was younger (71.6 ± 6.8 vs 75.4 ± 8.4, p = 0.033), prostatic volume (PV) was larger (76 ± 41 vs 49 ± 26 ml, p = 0.005), voiding efficiency was lower (54 ± 27 vs 68 ± 24%, p = 0.045) and bladder outlet obstruction index was higher (73 ± 30 vs 48 ± 30, p = 0.015). The cox proportional-hazards model indicated that only intravesical prostatic protrusion (IPP) was associated with treatment failure. Visualization of the interfacial electrostatic complementarity (VIINEC) is a recently developed method for analyzing protein–protein interactions using electrostatic potential (ESP) calculated via the ab initio fragment molecular orbital method. In this Letter, the molecular interactions of the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein with human angiotensin-converting enzyme 2 (ACE2) and B38 neutralizing antibody were examined as an illustrative application of VIINEC. The results of VIINEC revealed that the E484 of RBD has a role in making a local electrostatic complementary with ACE2 at the protein–protein interface, while it causes a considerable repulsive electrostatic interaction. Furthermore, the calculated ESP map at the interface of the RBD/B38 complex was significantly different from that of the RBD/ACE2 complex, which is discussed herein in association with the mechanism of the specificity of the antibody binding to the target protein.

Whether free fatty acids (FFAs), which are generators of reactive oxygen species and substrates of cytotoxic lipid peroxidation products in proximal tubules of the kidney, can be a predictor of worsening renal function (WRF) is not fully elucidated. A total of 110 patients with ST-segment elevation myocardial infarction (STEMI) who underwent primary percutaneous coronary intervention within 24 h after symptom onset were included. The exclusion criteria were out-of-hospital cardiac arrest, vasospastic angina, hemodialysis, and/or lack of data. FFAs and serum cystatin C were measured on admission, and urinary liver-type fatty acid-binding protein (L-FABP) was measured 3 h after admission. WRF, defined as an increase in serum creatinine by ≥ 0.3 mg/dL for 2-year follow-up, was observed in 16 patients (15%). A multivariate logistic regression analysis (a stepwise algorithm) revealed that the FFA level was an independent predictor of WRF (P = 0.024). The FFA level was associated with WRF adjusted after serum cystatin C (odds ratio [OR]: 1.378 per 1 mEq/L, P = 0.017), L-FABP (OR: 1.370 per 1 mEq/L, P = 0.016), or the Mehran contrast-induced nephropathy (CIN) risk score (OR: 1.362 per 1 mEq/L, P = 0.021). The FFA level was inversely associated with the change in estimated glomerular filtration rate level for 2 years (R2 = 0.051, P = 0.018). The FFA level on admission was associated with the mid-term WRF in patients with STEMI.

Iron pyrite [cubic FeS2 (cFeS2)] is considered as an earth-abundant and low-cost thin-film photovoltaic material. However, the conversion efficiency of cFeS2-based solar cells remains below 3%. To elucidate this limitation, we evaluate the optical and electrical characteristics of cFeS2 single crystals that are grown using the flux method, thus providing us an understanding of the electron transport behavior of cFeS2 single crystals. The oxide layer on the surface of cFeS2, which can possibly have an influence on the electrical characteristics of cFeS2, is removed prior to characterization via optical spectroscopy and electrical transport measurement. The optical property of cFeS2 was found to have both indirect and direct transitions. We also observed the presence of a band tail below the conduction band. The obtained electrical transport behavior indicates that cFeS2 bulk exhibits a high defect density and a disordered phase, thus leading to the hopping conduction mechanism. Our results will pave the way for the development of photovoltaic applications with iron pyrite.

Sulfamethoxazole/trimethoprim causes hyperkalemia; however, the effect of sulfamethoxazole/trimethoprim dose and co-administered glucocorticoids on hyperkalemia has not been clarified.This single-center, retrospective, observational cohort, chart review study involving patients (20 years) who were treated with sulfamethoxazole/trimethoprim was conducted at Tokyo Women's Medical University, Medical Center East from June 2015 to May 2019. Multivariate Cox proportional hazard model was used to identify risk factors for hyperkalemia (serum potassium level 5.5 mEq/L). Additionally, Kaplan-Meier curve analyzed the cumulative incidence of hyperkalemia focusing on sulfamethoxazole/trimethoprim dose and concomitant use of glucocorticoids with mineralocorticoid activity.Among 333 patients, 44 (13%) patients developed hyperkalemia associated with sulfamethoxazole/trimethoprim use for over 49 (interquartile range; 17-233) days. We found associations between the time to hyperkalemia development and sulfamethoxazole/trimethoprim dose (hazard ratio 1.238, 95% confidence interval 1.147-1.338, p 0.001) and glucocorticoid use (hazard ratio 0.678, 95% confidence interval 0.524-0.877, p = 0.003). Interestingly, the Kaplan-Meier curves revealed that the concomitant use of glucocorticoids did not attenuate the risk of hyperkalemia in patients receiving high-dose sulfamethoxazole/trimethoprim (p = 0.747), whereas concomitant use of glucocorticoids significantly reduced the risk of hyperkalemia in patients receiving non-high dose sulfamethoxazole/trimethoprim (p 0.001).High-dose sulfamethoxazole/trimethoprim is a significant predictor of hyperkalemia. The effect of glucocorticoids on hyperkalemia varies depending on the sulfamethoxazole/trimethoprim dose.

4-Nonylphenol (4-NP) is an endocrine-disrupting and persistent chemical and is partially degraded in conventional wastewater treatment processes. Ferrate(VI) can be used as an environment-friendly oxidizing agent to mediate 4-NP degradation. Thus, this paper evaluates the biodegradability of 4-NP and its degradation products after the addition of ferrate(VI). The biodegradability was examined using NP labeled with 14C as a tracer and activated sludge microorganisms as an inoculum. The addition of ferrate(VI) to the 4-NP solution spiked with the tracer resulted in no remarkable decrease in the concentration of 14C, indicating incomplete mineralization of 4-NP and formation of degradation products. The degradation products from 4-NP with Fe(VI) were estimated based on mass spectra, which detected a unique peak at m/z 223 at low intensity. Four hydrogen atoms might have been added to 4-NP by degradation with Fe(VI). In addition, the effect of ferrate(VI) concentration on the estrogenic activity of 4-NP in an aqueous solution was investigated using a yeast bioassay. The results show that estrogenic activity was significantly decreased at a mass ratio of Fe(VI) to 4-NP greater than or equal to 2.5.

The nonenzymatic peptide bond cleavage at the C-terminal side of Asn residues is a protein post-translational modification that occurs under physiological conditions. This reaction proceeds much slower than the deamidation of the Asn side chain and causes denaturation and hypofunction of proteins. The peptide bond cleavage of Asn is detected primarily in crystallins and aquaporin 0 in the eye lens. Therefore, cleavage is thought to be involved in age-related cataracts. In this study, to clarify the mechanism underlying succinimide formation for the peptide bond cleavage of the Asn residue, we performed quantum chemical calculations on the model compound Ace-Asn-Gly-Nme (Ace = acetyl and Nme = methylamino). The density functional theory with the B3LYP/6-31+G(d,p) level of theory was used to obtain optimized geometries. The results suggested that the reaction proceeds through two steps, cyclization and C-terminal fragment release, and the required proton transfers can be mediated by H2PO4– and HCO3– ions. The conformational change of the main chain on the N-terminal side of Asn was needed for the C-terminal fragmentation step, and a separate conformational change at the C-terminal side was required for the cyclization step. Furthermore, the calculated activation barriers of the reactions catalyzed by the H2PO4– ion (130 kJ mol–1) and the HCO3– ion (123 kJ mol–1) were sufficiently low for the reactions to occur under normal physiological conditions.

Human T cell leukemia virus type 1 (HTLV-1) is responsible for adult T cell leukemia (ATL); however, molecular and cellular mechanisms underlying HTLV-1-induced leukemogenesis are unclear. BCL6 oncogene is involved in cancer progression and a preferred target of anti-cancer treatments. Here, we aimed to evaluate BCL6 expression and the effects of BCL6 inhibitor (FX1) on HTLV-1-infected T cell lines. BCL6 expression was higher in HTLV-1-infected T cell lines than that in uninfected T cell lines. BCL6 was localized mostly in the nucleus. The virus oncoprotein Tax induced BCL6 mRNA expression in T cells, whereas BCL6 knockdown reduced HTLV-1-infected T cell proliferation; thus, confirmed the association of BCL6 with cancer progression. Further, FX1 efficiently inhibited the cell growth and survival of HTLV-1-infected T cell lines in a dose- and time-dependent manner. The decreased levels of cell cycle regulatory proteins (phosphorylated retinoblastoma protein, cyclin-dependent kinase 4, cyclin D2 and c-Myc) and the increased levels of BCL6 target proteins (p21, p27 and p53) showed that FX1 arrested cell cycle at the G1 phase. Apoptosis was induced concomitantly with Bak upregulation and downregulation of survivin, Bcl-xL and Mcl-1, as well as with the activation of caspase-3, -8, -9 and poly(ADP-ribose) polymerase. FX1 also inhibited NF-κB and Akt signaling pathways. These events were because of the induction of the activity of cell cycle checkpoint proteins and relief of direct repression of the targets of cell cycle checkpoint proteins. Thus, BCL6 might be considered a novel target for ATL treatment.

Cs is absorbed by plants via the roots and is then distributed to different plant parts. In pot experiments with rice, Cs concentration in the above-ground parts changed dramatically from the heading stage until maturation. The aim of this study was to clarify the dynamics of Cs distribution in field-grown rice plants throughout the cultivation period. Field experiment was conducted using Indica cultivar ‘Habataki’ (higher Cs accumulation) and Japonica cultivar ‘Koshihikari’ (lower Cs accumulation) under different K application levels for 2 years. Changes in Cs and K concentration in each plant part and exchangeable Cs and K concentrations in soil were analyzed throughout the cultivation. Laser ablation-inductively coupled mass spectrometry was conducted to reveal tissue-specific accumulation of Cs and K in internodes. Cs concentrations in each part were at similar levels in the heading stage irrespective of the cultivars but greatly changed toward the maturation stage. Particularly, Cs concentration in internodes showed the largest increase. Cs accumulated to the same level in the large vascular bundles and parenchyma in the internodes. Habataki displayed a higher accumulation level of Cs in the internodes and distribution ratio of Cs to brown rice than Koshihikari. The effect of K application on Cs distribution was lower than that in the previous pot experiment. Cs accumulation in the internode tissues, along with its distribution to brown rice, was greatly increased during ripening period in field conditions. Habataki showed higher Cs accumulation in internode and distribution ratio to brown rice than Koshihikari.

Telomere binding protein Stn1 forms the CST (Cdc13/CTC1-STN1-TEN1) complex in budding yeast and mammals. Likewise, fission yeast Stn1 and Ten1 form a complex indispensable for telomere protection. We have previously reported that stn1-1, a high-temperature sensitive mutant, rapidly loses telomere DNA at the restrictive temperature due to frequent failure of replication fork progression at telomeres and subtelomeres, both containing repetitive sequences. It is unclear, however, whether Stn1 is required for maintaining other repetitive DNAs such as ribosomal DNA. In this study, we have demonstrated that stn1-1 cells, even when grown at the permissive temperature, exhibited dynamic rearrangements in the telomere-proximal regions of subtelomere and ribosomal DNA repeats. Furthermore, Rad52 and γH2A accumulation was observed at ribosomal DNA repeats in the stn1-1 mutant. The phenotypes exhibited by the stn1-1 allele were largely suppressed in the absence of Reb1, a replication fork barrier-forming protein, suggesting that Stn1 is involved in the maintenance of the arrested replication forks. Collectively, we propose that Stn1 maintains the stability of repetitive DNAs at subtelomeres and rDNA regions.
