Introdução: As próteses fixas implanto-suportadas são alternativas de tratamentos cada dia mais frequente nas clinicas odontológicas. Elas podem ser parafusadas ou cimentadas dependendo do planejamento cirúrgico. As próteses parafusadas são reversíveis, tem maior infiltrado bacteriano, estética prejudicada pelo orifício dos parafusos. As próteses cimentadas têm dificuldades de reversão, perda da prótese em caso de afrouxamento de parafuso e uma melhor estética por não ter orifício do parafuso. Objetivos: Nesse contexto, o presente estudo, pretender avaliar os níveis de resistência a tração dos copping´s cimentados por 2 diferentes tipos de cimentos para fixação de coroas implanto-suportadas cimentadas. Metodologia: Foram confeccionados três corpos de prova com implantes e coroas cimentadas com diferentes matérias, onde cada um desses corpos de prova foi submetido a um teste de tracionamento. Resultados: Os copping´s cimentados com cimento fosfato de zinco (CP1 e CP4) exigiram uma força de tração de 118.02 kgf e 213.63 kgf respectivamente, enquanto os copping´s cimentados com cimento resinoso (C2 e C3) exigiram uma força de tração de 258.93 kgf e 228.87 kgf respectivamente. Conclusão: Dentro das limitações das atuais condições in vitro empregadas neste estudo, a retenção de copping’s de coroa cimentada nos pilares de implantes com cimento resinoso e cimento fosfato de zinco foi significativamente afetada pelo tipo de cimento. O cimento resinoso exigiu uma força de tração significativamente maior que o cimento fosfato de zinco para a remoção dos copipng’s.

No. of bitstreams: 0 Previous issue date: 2020-04-29 Background: Sedum praealtum has been used for a long time in traditional medicine as an analgesic and antiinflammatory agent. Its beneficial effects have been known since ancient times, when Latinos used it to treat sore and swollen eyes. This research evaluated the antimicrobial potential, the cytotoxic and genotoxic effects, and some chromatographic profiles of the hydroethanolic extract of leaves, stems and roots of S. praealtum. Methods: The antimicrobial activities were carried out by broth microdilution and agar diffusion. In vitro cytotoxicity was evaluated by cell cultures of Aedes albopictus and the selectivity index (SI) was estimated: SI=CI50/MIC. Genotoxic and systemic toxic effects of S. praealtum leaves were analyzed by micronucleus assay in mice bone marrow. Chromatographic profiles and mass spectra were investigated by GC-MS. Results: Gram-positive (B. subtilis, B. cereus, M. luteus, E. faecalis and S. aureus) and gram-negative (E. coli, E. aerogenes, S. marcescens, P. aeruginosa, P. mirabilis and S. typhimurium) bacteria exhibited MICs ranging from 12.5–50 and 0–50 mg/ ml, respectively. Sedum praealtum showed no efficacy against M. tuberculosis and M. bovis. Cytotoxicity (CI50) of S. praealtum was 4.22 and 5.96 mg/ml for leaves and stems, respectively, while its roots showed no cytotoxicity. Micronucleated polychromatic erythrocytes (MNPCEs) analyzes showed no differences between treatment doses (0.5–2 g/kg) and negative control (NaCl), but the PCE/NCE ratio (polychromatic erythrocyte/normochromatic erythrocyte) showed significant differences. Phytochemical screening identified thirteen compounds in the leaves, stems and roots of S. praealtum potentially associated with their biological activities. Conclusions: This research comprises a first scientific study on genotoxicity, cytotoxicity and antimicrobial effects of S. praealtum (Balsam), and it provides an initial theoretical foundation for its comprehensive use. Results showed antibacterial action of S. praealtum against gram-positive bacteria and some gram-negative species (depending on the plant anatomical part), but ineffective antimycobacterial action. However, S. praealtum leaves and stems display potential cytotoxicity, contributing to the SI < 1 values. In addition, S. praealtum leaves exhibit no clastogenic and/or aneugenic effects, but it has systemic toxicity dose-independent. We compared respiratory mechanics between the positive end-expiratory pressure of minimal respiratory system elastance (PEEPminErs) and three levels of PEEP during low-tidal-volume (6 mL/kg) ventilation in rats. Twenty-four rats were anesthetized, paralyzed, and mechanically ventilated. Airway pressure (Paw), flow (F), and volume (V) were fitted by a linear single compartment model (LSCM) Paw(t) = Ers × V(t) + Rrs × F(t) + PEEP or a volume- and flow-dependent SCM (VFDSCM) Paw(t) = (E1 + E2 × V(t)) × V(t) + (K1 + K2 × |F(t)|) × F(t) + PEEP, where Ers and Rrs are respiratory system elastance and resistance, respectively; E1 and E2× V are volume-independent and volume-dependent Ers, respectively; and K1 and K2 × F are flow-independent and flow-dependent Rrs, respectively. Animals were ventilated for 1 h at PEEP 0 cmH2O (ZEEP); PEEPminErs; 2 cmH2O above PEEPminErs (PEEPminErs+2); or 4 cmH2O above PEEPminErs (PEEPminErs+4). Alveolar tidal recruitment/derecruitment and overdistension were assessed by the index %E2 = 100 × [(E2 × VT)/(E1 + |E2| × VT)], and alveolar stability by the slope of Ers(t). %E2 varied between 0 and 30% at PEEPminErs in most respiratory cycles. Alveolar Tidal recruitment/derecruitment (%E2 30) were predominant in the absence of PEEP and in PEEP levels higher than PEEPminErs, respectively. The slope of Ers(t) was different from zero in all groups besides PEEPminErs+4. PEEPminErs presented the best compromise between alveolar tidal recruitment/derecruitment and overdistension, during 1 h of low-VT mechanical ventilation.

The aim of this study was to evaluate the effects of caffeic acid phenethyl ester (CAPE) on osteoblast-like cell cultures (SAOS-2).SAOS-2 were exposed to CAPE at 1 nM, 10 nM, 100 nM, 1 μM, and 10 μM. Non-exposed cultures were used as control. The following parameters were assayed: 1) cell viability at 1, 3, and 7 days; 2) alkaline phosphatase (ALP) activity at 5 and 10 days; 3) matrix mineralization at 14 days; and 4) Runt-related transcription factor 2 (RUNX2), ALP, osteopontin (SPP1), and osteocalcin (BGLAP) gene expression at 5 and 10 days. The data were analyzed by ANOVA two-way or Kruskal-Wallis (α = 5%).At day 1, cell viability was similar among all groups (p0.05). At days 3 and 7, cultures exposed to CAPE at 10 μM exhibited a significant reduction in cell viability compared with the others groups (p0.05). At day 5, ALP activity was similar among all experimental groups; at day 10, however, the stain intensity was higher in cultures exposed to CAPE at 100 nM and 10 nM in comparison with the other groups (p0.05). At days 5 and 10, RUNX2, ALP, SPP1, and BGLAP gene expression was greater in cultures exposed to CAPE in comparison with the control (p0.05). At day 14, matrix mineralization was similar in cultures exposed to CAPE at 1 nM and 10 nM (p0.05), but superior to those ones observed in the other experimental groups (p0.05).CAPE at low concentrations can positively module the osteogenesis in vitro.

The magnitude of the pre-salt projects, most located in Brazil ultra-deep waters, coupled with the enormous potential already auctioned, leads to the following questions: what will be Brazil's share in the world's deep-water development? How should suppliers prepare themselves to assist Brazil in the task of making these opportunities viable? Since 2013, the country awarded over R$ 43 billion in signing bonuses. Despite the pessimism associated to the oil price drop in 2014, Brazil has gone forward with the development of pre-salt areas and the resume of the bidding rounds, reaching the top 10th position in world's biggest oil producers (BP 2019). This paper forecasts the oil potential for these areas, as well as the Brazilian oil production for the next 15 years and the demand for facilities to reach that potential. Those forecasts clarify the increasing importance of Brazilian deep-water oil in the global scenario.

An experimental and numerical assessment of multilayer active magnetic regenerators (AMR) composed of gadolinium (Gd) and gadolinium–yttrium (Gd–Y) alloys (Gd[Formula: see text]Y[Formula: see text], Gd[Formula: see text]Y[Formula: see text] and Gd[Formula: see text]Y[Formula: see text]) is presented. First, by calculating the adiabatic temperature change and the isothermal entropy change from the experimental data for the above materials, we show that, with reasonable accuracy for engineering design purposes, these properties can be determined by shifting the properties of pure Gd to the Curie temperature of the Gd–Y alloy — a common but not yet validated assumption in the design of Gd–Y AMRs with a low Y content. Next, we show that the optimal Gd–Y layer fraction in multilayer AMRs can be determined using the figure of merit known as the material refrigerant capacity (RC), which agrees well with the results from a more complex one-dimensional thermal non-equilibrium porous medium AMR model. Finally, the performance of the latter model is verified against the experimental cooling power data for two- and three-layer Gd–Y regenerators at temperature spans of 25, 30 and 35[Formula: see text]K.

The purpose of this study was to compare cardiovascular and respiratory effects of two volumes of bupivacaine 0.25% (0.2 mL kg-1—treatment BUP02—and 0.4 mL kg-1 –treatment BUP04) administered epidurally at the lumbosacral intervertebral space in dogs anesthetized with isoflurane. This experimental prospective randomized crossover design trial used six mixed breed adult dogs, four neutered males and two spayed females. Each dog was anesthetized on three different occasions: the first for isoflurane minimum alveolar concentration (MAC) measurement, and the following two assigned treatments (BUP02 or BUP04). On the two treatment days, anesthesia was induced and maintained with isoflurane at 1.3 MAC during the experiments. Cardiovascular and respiratory measurements were recorded before (T0) and 5, 15, 30, 60 and 90 minutes after the epidural administration of bupivacaine. Comparisons between and within groups were performed by a mixed-model ANOVA and Friedman's test when appropriate followed by Bonferroni post-hoc test or Dunnet's test to compare time points within each treatment with T0 (p < 0.05). Mean arterial pressure decreased significantly from 15 to 90 minutes after the administration of BUP02 and from 5 to 60 minutes in BUP04, with lower values in BUP04 than in BUP02 lasting up to 30 minutes after bupivacaine administration. No significant changes in cardiac output and systemic vascular resistance were observed in either treatment. Hypoventilation was only detected in BUP04. Hemoglobin concentration and arterial oxygen content decreased after both treatment of bupivacaine with no significant decrease in oxygen delivery. Two dogs in BUP04 developed Horner's syndrome. The epidural administration of 0.4 mL.kg-1 of bupivacaine to dogs in sternal recumbency anesthetized with isoflurane 1.3 MAC caused more cardiovascular and respiratory depression than 0.2 mL.kg-1. Bladder cancer is one of the leading causes of cancer-related death in men, however, there was only limited effective treatment for invasive bladder cancer. DAPK1 has been shown to play important role in apoptosis and autophagy to suppress cancer progression. Previous results have shown that DAPK1 promoter was hypermethylated in the majority of bladder cancer specimens, however, the prognostic significance of DAPK1 in bladder cancer has yet to be demonstrated. In the present study, we found that DAPK1 expression was negatively associated with tumor stage and a low level expression of DAPK1 in bladder cancer specimens were associated with shorter survival in bladder cancer patients in 3 independent bladder cancer datasets (n = 462). Further investigation showed that FGFR3 knockdown resulted in downregulation of DAPK1 in bladder cancer cell line, suggesting that FGFR3 may be an upstream factor of DAPK1. Further analysis of the 3 independent bladder cancer datasets have identified ACOX1, UPK2, TRAK1, PLEKHG6 and MT1X genes had their expression significantly correlated with that of DAPK1. Knockdown of DAPK1 in bladder cancer T24 cells resulted in downregulation of ACOX1, UPK2 and TRAK1. Interestingly, TRAK1, by itself, was a favorable prognostic marker in the 3 independent bladder cancer datasets. Importantly, by using connectivity mapping with DAPK1-associated gene signature, we found that vemurafenib and trametinib could possibly reverse DAPK1-associated gene signature, suggesting that inhibition of Raf/MEK pathway may be a potential therapeutic approach for bladder cancer. Indeed, treatment of vemurafenib in T24 bladder cancer cells resulted in upregulation of DAPK1 confirming our connectivity mapping, while knockdown of DAPK1 resulted in reduced sensitivity towards inhibition of Braf signaling by vemurafenib. Together, our results suggest that DAPK1 is an important prognostic marker and therapeutic target for bladder cancer and have identified possible therapeutic agents for future testing in bladder cancer models with low DAPK1 expression.

Cell division is a tightly-regulated process that involves the contribution of a large number of proteins. Before they are able to undergo mitosis, cells must first synthesize new DNA, effectively and accurately duplicating their genome. This occurs during what is called the S-phase and requires a fine control in order to avoid replication errors. The synthesis of new DNA takes place in the origin, specific locations in the genome where the double strands of DNA are unwound and separated, allowing for the binding of proteins and complexes that will build new strands of the genomic material, using the existing ones as molds, in what is referred to as semi-conservative process. While the overall flow of the DNA synthesis process has been elucidated, its regulation and the exact role of its contributors are not yet entirely understood. It is believed that the Minichromosome Maintenance (MCM) proteins occupy a central role in DNA synthesis. Given their contribution to a central aspect in the conservation of life, further studies have been launched to understand how the MCM proteins may affect or be affected by pathologies involving cell division, such as neoplasia. In this review, we aim to give an overview on the members of the MCM family, what their functions are in a healthy environment and how they are altered in cancer.

A arte da Cantaria usando o quartzito foi utilizada na construção dos monumentos históricos de Ouro Preto/MG. Após mais de 300 anos de existência, parte de algum desses monumentos encontra-se em avançado estágio de deterioração. Na restauração desses monumentos, alguns restauradores empregam a resina poliéster ortoftálica. Nesse trabalho, o quartzito itacolomi não colado e colado com diversas resinas foi submetido a ensaio de resistência em flexão e constatou-se que a resina suportou carga maior que o quartzito. Todos os corpos-de-prova romperam fora da região da aplicação da resina, confirmando a eficácia das resinas para colar a rocha. The art of carving stone using itacolomi quartzite was employed in the monuments of Ouro Preto city in Minas Gerais. More than 350 years after being built, some of these monuments are in an advanced stage of deterioration. In the restoration of these buildings, some restaurateurs employ orthophthalic polyester resin. In the present study, both non-glued and glued (using different types of resins) itacolomi quartzite were submitted to a flexion strength test. After some trials, it was verified that the resins supported greater loads than the quartzite. All specimens broke outside the resin’s application area, thus confirming the effectiveness of resin adherence to rock.

Sorafenib is a chemotherapeutic agent approved for the treatment of hepatocellular carcinoma (HCC) in China. Digitoxin is a cardiotonic drug, which has been demonstrated to exhibit anticancer effects in a number of cancers, but not in HCC. The aim of the present study was to evaluate the combinational effect of sorafenib and digitoxin on the treatment of HCC and to investigate the relevant molecular mechanisms of action that underlie these effects. The proliferation, cell death and migration of HCC cell lines, HepG2 and BEL‑7402, were examined using MTT, acridine orange/ethidium bromide staining and scratch wound healing assays, respectively. In addition, alterations in the expression of phosphorylated-extracellular signal-regulated kinase (ERK), hypoxia‑inducible factor 1‑α (HIF‑1α), hypoxia‑inducible factor 2‑α (HIF‑2α) and vascular endothelial growth factor (VEGF) were measured prior to and following drug application using western blot analysis. Digitoxin and sorafenib synergistically inhibited cell viability, but did not inhibit migration, which was potentially mediated by suppression of ERK and hypoxia signaling. In downstream signaling pathways, the activity of ERK was synergistically suppressed by combinatorial treatment of HepG2 and BEL‑7402 cells with sorafenib and digitoxin. In addition, the expression of HIF‑1α, HIF‑2α and VEGF was synergistically downregulated by combinational treatment.

Results are reported on the preparation of bulk parts of La(Fe,Si)13Hy via the Hydrogen-Decrepitation-Sintering-Hydrogenation (HDSH) process. Net shape parts for application in room-temperature magnetic refrigeration have been produced in only 8 h of heat treatment which is considerably faster than the conventional ingot homogenization heat treatment of 7 days. The samples produced by HDSH showed higher amounts of hydrogen than the parts hydrogenated by the conventional method of thermal homogenization (20 h at 1423 K), milling to fine powder and subsequent hydrogenation. Hydrogenation parameters play an important role for the stability of the desired La(Fe,Si)13 phase during the process. Hydrogen desorption was seen to occur at two temperature ranges as a result of internal gradients. Dissimilar amounts of α-Fe were precipitated for different hydrogenation times. As a result, parts produced via HDSH with 2 and 4 h of hydrogenation exhibited different magnetocaloric behaviours. For a hydrogenation step of 4 h, parts with a demagnetization factor of 0.49 showed an adiabatic temperature change ( Δ T ad ) higher than 1 K for a temperature range of 40 K with a maximum value of 1.57 K for an applied magnetic field of 1.75 T. As the duration of the hydrogenation step of the HDSH process decreased to 2 h, Δ T ad was larger than 1 K for a temperature range of 24 K. However the maximum value of Δ T ad at 328 K was 2.2 K, which is 37.5% larger than the maximum value for a hydrogenation period of 4 h.

Background Insulin is a hormone that regulates blood glucose homeostasis and is a central protein in a medical condition termed insulin injection amyloidosis. It is intimately associated with glycaemia and is vulnerable to glycation by glucose and other highly reactive carbonyls like methylglyoxal, especially in diabetic conditions. Protein glycation is involved in structure and stability changes that impair protein functionality, and is associated with several human diseases, such as diabetes and neurodegenerative diseases like Alzheimer's disease, Parkinson's disease and Familiar Amyloidotic Polyneuropathy. In the present work, methylglyoxal was investigated for their effects on the structure, stability and fibril formation of insulin. Results Methylglyoxal was found to induce the formation of insulin native-like aggregates and reduce protein fibrillation by blocking the formation of the seeding nuclei. Equilibrium-unfolding experiments using chaotropic agents showed that glycated insulin has a small conformational stability and a weaker dependence on denaturant concentration (smaller m-value). Our observations suggest that methylglyoxal modification of insulin leads to a less compact and less stable structure that may be associated to an increased protein dynamics. Conclusions We propose that higher dynamics in glycated insulin could prevent the formation of the rigid cross-β core structure found in amyloid fibrils, thereby contributing to the reduction in the ability to form fibrils and to the population of different aggregation pathways like the formation of native-like aggregates.
