Objectives. Resident macrophages are well known to be present in the cochlea, but the exact patterns thereof in spiral ligaments have not been discussed in previous studies. We sought to document the distribution of macrophages in intact cochleae using three-dimensional imaging.Methods. Cochleae were obtained from C-X3-C motif chemokine receptor 1+/GFP mice, and organ clearing was performed. Three-dimensional images of cleared intact cochleae were reconstructed using two-photon microscopy. The locations of individual macrophages were investigated using 100-μm stacked images to reduce bias. Cochlear inflammation was then induced by lipopolysaccharide (LPS) inoculation into the middle ear through the tympanic membrane. Four days after inoculation, three-dimensional images were obtained.Results. Macrophages were scarce in areas adjacent to the stria vascularis, particularly the area just beneath it even though many have suspected macrophages to be abundant in this area. This finding remained consistent upon LPS-induced cochlear inflammation, despite a significant increase in the number of macrophages, compared to non-treated cochlea.Conclusion. Resident macrophages in spiral ligaments are scarce in areas adjacent to the stria vascularis.

Background BMS-470539, a recently introduced selective agonist of the melanocortin 1 receptor, is known to have anti-inflammatory properties. In this study, we investigated the effects of BMS-470539 on lipopolysaccharide (LPS)-induced inflammatory responses and delayed apoptosis with its signaling pathways in human neutrophils. Methods Isolated human neutrophils were incubated with various concentrations of BMS-470539 (1, 10, and 100 µM) in the presence or absence of LPS (100 ng/ml), and the expression of pro-inflammatory cytokines, such as tumor necrosis factor alpha, interleukin (IL)-6, and IL-1β, were assessed. The effects of BMS-470539 on the expression of mitogen-activated protein kinases (MAPKs), such as p38, extracellular-signal-regulated kinase 1/2, and c-Jun N-terminal kinase, and the expression of nuclear factor kappa B (NF-κB) in LPS-stimulated human neutrophils, were evaluated by enzyme-linked immunosorbent assay. Neutrophil apoptosis was also measured by fluorescence-activated cell sorting (annexin V/propidium iodide) in LPS-stimulated neutrophils under treatment with BMS-470539. Results BMS-470539 attenuated LPS-induced expression of pro-inflammatory cytokines, and phosphorylation of MAPKs and NF-κB. LPS stimulation reduced neutrophil apoptosis compared to the controls; however, BMS-470539 significantly inhibited the reduction of neutrophil apoptosis. Conclusions BMS-470539 can suppress the inflammatory responses of LPS-stimulated neutrophils by inhibition of MAPK pathways or NF-κB pathway, and it can also inhibit LPS-delayed neutrophil apoptosis.

Background Fermented Rhus verniciflua Stokes extract (FRVE) reported an anti-hepatic lipidemic property mediated by the upregulation of AMP-activated protein kinase (AMPK) in cell and animal models. However, it remains unclear whether there is an effect of FRVE on liver disease-related parameters and serum lipid levels in humans. We investigated the effects of FRVE intake for 12 weeks on liver disease-related parameters and serum lipid profiles in Korean adults. Methods A randomized, double-blind, placebo-controlled study was conducted among 79 subjects. An aqueous extract of fermented Rhus verniciflua Stokes that was filtered and fermented was prepared. For 12 weeks, the test group (n = 39) consumed two capsules of FRVE (main components: fustin 129 mg and fisetin 59 mg) once daily. The control group (n = 40) consumed two placebo pills (main component: lactose 627.0 mg) once daily. A 1:1 randomization of control and test was performed using computer-generated randomization. Both before and after FRVE intake, anthropometric parameters, liver function-related parameters, and clinical laboratory parameters were measured. The effects between the test and control groups were compared using the Mann-Whitney U test and independent t-test, and the difference between baseline and follow-up values was compared using Wilcoxon rank-sum test and paired t-test. Results There was no significant difference when comparing the change values of liver disease-related parameters and serum lipid profiles in between groups. Conclusions In our study, we did not confirm the significance in liver function parameters and serum lipid profiles. Understanding the magnitude and causes of isotopic fractionation between organisms and their dietary resources is crucial for gaining knowledge on stable isotope ecology. However, little is known regarding the diet-tissue fractionation values of marine ciliates, which play a critical role in the reconstruction of microbial food webs. In the present study, we conducted experiments on two benthic (Pseudokeronopsis pararubra and Protocruzia labiata) and two pelagic (Strombidium sulcatum and Uronemella filificum) marine ciliates, where they were fed with isotopically constant foods (Chaetoceros calcitrans and Isochrysis galbana) under laboratory culture conditions to determine their carbon and nitrogen isotopic fractionation values (Δ13C and Δ15N). The stable isotope values (δ13C and δ15N) of ciliates for all experiments rapidly increased after the initial feeding, with half-lives ranging from 6.1 to 23.0h for δ13C and from 3.1 to 24.9h for δ15N. The Δ13C and Δ15N for all ciliates represented significantly positive enrichments, with overall mean fractionations of 0.6±0.2 and 1.2±0.4, respectively. Irrespective of the dietary type, both Δ13C and Δ15N were very similar for the same ciliate species. These results suggest that Δ13C and Δ15N for marine ciliates are similar to those found in common marine organisms with very little food-dependent variation. Overall, quantifying the specific isotopic fractionation of marine ciliates is expected to provide fundamental information on the trophic transfer of carbon, nitrogen, and energy flow through the microbial pathway in marine ecosystems.

Decellularized esophageal matrices are ideal scaffolds for esophageal tissue engineering. Unfortunately, in order to improve transplantation possibilities, they require modification to reduce their degradation rate and immunogenicity. To date, no modifying agent has been approved to overcome these limitations. The objective of this study was to evaluate the ability of silver nanoparticles (AgNPs) to improve the structural stability and biocompatibility of decellularized rat esophagi. AgNPs have the advantage over currently used agents in that they bind with collagen fibers in a highly ordered manner, via non-covalent binding mechanisms forming multiple binding sites, while other agents provide only two-point connections between collagen molecules. Rat esophagi were decellularized, loaded with 5 μg/mL of AgNPs (100 nm), and then treated with an immobilization-complex buffer composed of ethyl carbodiimide hydrochloride and N-hydroxysuccinimide (EDC/NHS). Then, they were evaluated in terms of ultra-structural morphology, water uptake, in vitro resistance to enzymatic and thermal degradation, indentation strength, in vitro anti-calcification, cytocompatibility with rat bone marrow derived stromal cells (rat-BMSCs), angiogenic properties, and in vivo biocompatibility, and compared to scaffolds modified using glutaraldehyde and EDC/NHS complex buffer alone. AgNP-modified scaffolds showed an improved ultrastructure, good water uptake, and considerable resistance against in vitro degradation and indentation, and a high resistance against in vitro calcification. Moreover, they were cytocompatible for allogeneic rat-BMSCs. Additionally, AgNPs did not alter the angiogenic properties of the modified scaffolds and decreased host immune responses after their subcutaneous implantation. The structural properties and biocompatibility of decellularized esophageal matrices could be improved by conjugation with AgNPs.

Decellularization of a whole organ is an attractive process that has been used to create 3D scaffolds structurally and micro-architecturally similar to the native one. Currently used decellularization protocols exhibit disrupted extracellular matrix (ECM) structure and denatured ECM proteins. Therefore, maintaining a balance between ECM preservation and cellular removal is a major challenge. The aim of this study was to optimize a multistep Triton X-100 based protocol (either using Triton X-100/ammonium hydroxide mixture alone or after its modification with DNase, sodium dodecyl sulfate or trypsin) that could achieve maximum decellularization with minimal liver ECM destruction suitable for subsequent organ implantation without immune rejection. Based on our findings, Triton X-100 multistep protocol was insufficient for whole liver decellularization and needed to be modified with other detergents. Among all Triton X-100 modified protocols, a Triton X-100/DNase-based one was considered the most suitable. It maintains a gradual but sufficient removal of cells to generate decellularized biocompatible liver scaffolds without any significant alteration to ECM micro- and ultra-structure.

Zero-dimensional-two-dimensional (0D-2D) hybrid optoelectronic devices have demonstrated high sensitivity and high performance due to the high absorption coefficient of 0D materials with a tunable detection range and a high carrier transport property of 2D materials. However, the reported 0D-2D hybrid devices employ toxic nanomaterials as sensitizing layers, which can limit the practical applications. In this study, we first fabricated the 0D-2D hybrid photodetector using nontoxic InP quantum dots (QDs) as a light-absorbing layer and black phosphorus (BP) as a transport layer. The surface treatment using 1,2-ethanedithiol and thermal treatment were carried out to remove the surface long ligands of colloidal QDs, which can accelerate the charge injection of the photogenerated carriers through the interfaces between InP QDs and BP. The InP QDs/BP hybrid photodetector demonstrates a high responsivity of 1 × 109 A/W and detectivity of 4.5 × 1016 Jones at 0.05 μW cm-2 under 405 nm illumination. The results show that 0D-2D hybrid photodetectors based on III-V semiconducting QD materials can be optimized for high-performance photodetectors.

Background Overactivation of inflammatory cells, including macrophages and neutrophils, is associated with acute lung injury. BMS-470539 is a selective agonist of melanocortin 1 receptor, which triggers the inhibition of proinflammatory responses, suppressing neutrophil infiltration and protecting tissue. This study evaluated the effects of BMS-470539 on lipopolysaccharide-induced acute lung injury in a mouse model. Methods Mice received a subcutaneous injection of saline or BMS-470539 (18.47 mg/kg) 1 hour before an intratracheal injection of saline or lipopolysaccharide (20 μg). Mice were sacrificed to analyze the severity of pulmonary edema (lung wet-to-dry weight [W/D] ratio) and inflammatory responses (level of leukocytes, polymorphonuclear neutrophils [PMNs] and tumor necrosis factor alpha [TNF-α] in bronchoalveolar lavage fluid [BALF]), and neutrophil infiltration (myeloperoxidase activity). TNF-α activation was also measured in neutrophils from bone marrow. Survival was investigated in a second-hit sepsis mouse model. To investigate the effect of choline alfoscerate (CA) on hearing amplification in patients with age related hearing loss, we performed a prospective case-control observational study from March 2016 to September 2020. We assessed patients with bilateral word recognition score (WRS) p = 0.035). However, there was no significant between-group difference in the change in pure tone thresholds and aided speech intelligibility index calculated from REM. These findings suggest that the difference in WRS was relevant to central speech understanding rather than peripheral audibility. Therefore, administering oral CA could effectively enrich listening comprehension in older HA users.

The quantitative label-free detection of neurotransmitters provides critical clues in understanding neurological functions or disorders. However, the identification of neurotransmitters remains challenging for surface-enhanced Raman spectroscopy (SERS) due to the presence of noise. Here, we report spread spectrum SERS (ss-SERS) detection for the rapid quantification of neurotransmitters at the attomolar level by encoding excited light and decoding SERS signals with peak autocorrelation and near-zero cross-correlation. Compared to conventional SERS measurements, the experimental result of ss-SERS shows an exceptional improvement in the signal-to-noise ratio of more than three orders of magnitude, thus achieving a high temporal resolution of over one hundred times. The ss-SERS measurement further allows the attomolar SERS detection of dopamine, serotonin, acetylcholine, γ-aminobutyric acid, and glutamate without Raman reporters. This approach opens up opportunities not only for investigating the early diagnostics of neurological disorders or highly sensitive biomedical SERS applications but also for developing low-cost spectroscopic biosensing applications., Identification of neurotransmitters remains challenging for surface enhanced Raman spectroscopy (SERS) due to presence of noise. Introduction: Bilirubin is a product of the heme catabolism pathway, and it is excreted in bile and removed from the body through the urine. Bilirubin has potent antioxidant properties but also plays a role in anti-inflammation by protecting the body against endotoxin-induced lung inflammation, down-regulating the expression of adhesion molecules, and inhibiting the infiltration of inflammatory cells. Thus, bilirubin is a promising agent that could use in inflammation disease treatment. The application of bilirubin on the &ldquo, two-hit&rdquo, sepsis animal model has been, to date, unknown. Methods: we used lipopolysaccharide to induce initial insults in C57BL/6 mice. After 24 h, mice underwent cecal ligation and puncture to induce the &ldquo, sepsis model. Next, mice were administered 30 mg/kg bilirubin and we observed an improvement. Results: We observed that bilirubin inhibited the expression of pro-inflammatory cytokines, while the levels of anti-inflammatory cytokines were significantly augmented in the lung. Bilirubin improved the survival rate in the sepsis model. Furthermore, we suggest that bilirubin can modulate the accumulation of T-regulatory cells and myeloid-derived suppressor cells. Notably, bilirubin suppressed the activation and functions of T-cells. Conclusions: These results clarified that bilirubin might improve tissue injury in sepsis through anti-inflammatory mechanisms.

Objectives The investigation of cochlear hair cells and lateral wall is a time-consuming and labor-intensive process. However, it is a mandatory experiment in audiology research. Here we suggest a novel method for investigating the inner ear microstructures from intact cochleae using two-photon laser scanning microscopy (TPLSM). This technique guarantees fewer artifacts and technical simplicity. Methods Using TPLSM, we investigated the whole mount cochleae, decalcified cochleae, and cleared cochleae of wild type C57BL/6 mice. CX3CR1+/GFP mice were used to investigate the feasibility of visualizing cellular structures in the cochlear spiral ligament. All samples were investigated without staining. Results Endogenous fluorescence emission from the outer hair cells was strong enough to be distinguished from the other structures in all samples. From the single apical view, 50 and 90% of the whole hair cells of the decalcified cochleae and cleared cochleae, respectively, could be visualized without staining using TPLSM. Capillary structure of stria vascularis and spiral ligament could be visualized by endogenous fluorescence without staining. Conclusion We successfully investigated the hair cells and lateral wall of mouse cochleae using TPLSM without using staining or any destructive procedures. This method is easier, faster, and more reliable than conventional methods.

Whole kidney decellularization is a promising approach in regenerative medicine for engineering a functional organ. The reaction of the potential host depends on the biocompatibility of these decellularized constructs. Despite the proven ability of decellularized kidney scaffolds to guide cell attachment and growth, little is known about biocompatibility and hemocompatibility of these scaffolds. Our aim is to prepare decellularized kidneys of a clinically relevant size and evaluate its biocompatibility and hemocompatibility. Porcine kidneys were cannulated via the renal artery, and then perfused with 0.1% sodium dodecyl sulfate solution. Hematoxylin and eosin as well as DAPI staining confirmed cellular clearance from native kidneys in addition to preservation of the microstructure. SEM confirmed the absence of any cellular content within the scaffold, which is maintained in a well-organized 3D architecture. Decellularized kidneys retained the intact renal vasculature upon examination with contrast radiography. The essential structural extracellular matrix molecules were well-preserved. Scaffolds were susceptible to enzymatic degradation upon collagenase treatment. Scaffolds showed a good hemocompatibility when exposed to porcine blood. Decellularization was efficient to remove 97.7% of DNA from native kidneys in addition to the immunogenic and pathogenic antigens. Scaffolds did not induce the human immune response in vitro. Decellularized kidneys were non-cytotoxic to pig kidney cells (PKs). PKs were able to grow and proliferate within the decellularized renal scaffolds with maintaining a higher function than cells grown as monolayers. Thus, we have developed a rapid decellularization technique for generating biocompatible kidney scaffolds that represents a step toward development of a transplantable organ. © 2018 Wiley Periodicals, Inc. We report a new family of inorganic ligands, namely, transition metal-based thiometallates, for the surface functionalization of colloidal nanocrystals (NCs). We synthesized Pt-, Fe-, Co-, and Ni-based thiometallates, in which transition metal ions were complexed with polysulfides. These inorganic anions easily exchanged the surface organic ligands of various nanocrystals of metal, semiconductor, and oxide materials, without affecting the NCs’ primary structural and optical characteristics. Furthermore, upon heating, these complexes were decomposed and transformed into crystalline phases of metal sulfides or pure metals, accompanied by the evaporation of S. Based on this effect, we selectively synthesized homogeneously distributed atomic Pt clusters or Pt nanoparticles on Fe3O4 nanomaterials by heating thioplatinate-capped Fe3O4 NCs. Three cruises were undertaken along a meridional transect in the East/Japan Sea (EJS) in spring (May 2007), summer (July 2009), and fall (October 2012) to determine the geographic variations in phytoplankton biomass and community composition. This study revealed a gradient of surface temperature and a fluctuation of hydrographic conditions along the transect. Although a subpolar front (SPF) formed between the warm- and cold-water masses (37–40°N), no significant differences in phytoplankton biomass and community composition were detected between the southern and northern parts of the EJS. These results disprove our initial hypothesis that different water masses may contain differently structured phytoplankton communities. In the present study, isothermal layers (≤ 12 °C) fluctuated over a depth of 50 m in both warm- and cold-water masses, depending on the SPF. In contrast, the nitracline (i.e. 2.5 μM nitrate isopleth) depth was recorded within a limited range of 20–40 m in spring, 30–50 m in summer, and 40–60 m in fall. The chlorophyll a concentrations at the subsurface chlorophyll maxima (SCM) were significantly higher in spring and summer (356 ± 233 and 270 ± 182 ng L –1 , respectively) than in fall (117 ± 89 ng L –1 ). The relative contributions of individual phytoplankton groups to the depth-integrated chlorophyll a concentration conformed to the composition of the phytoplankton community in the SCM layer, showing a dominance of diatoms (58 ± 19% in spring, 48 ± 11% in summer, and 30 ± 20% in fall). Canonical correspondence analysis revealed that the geographic structures of phytoplankton communities were strongly associated with the vertical structures of water temperature and nutrient concentration in the water column rather than with horizontal gradients of hydrographic conditions. Finally, our findings suggest that water column stability and light–nutrient availability in the euphotic zone play a key role in determining geographical consistency of the biomass and the community structure of phytoplankton in the EJS.

Seasonal variability of sinking fluxes of total mass (TMF), particulate organic carbon and nitrogen (POC and PON) was examined using sinking particles collected from sediment traps during July 2011 to December 2011, and December 2012 to June 2013 at an offshore channel site; and from November 2013 to August 2014 at a nearshore slope site of the Ulleung Basin in the East/Japan Sea. δ13C and δ15N values of sinking particles were measured to elucidate the major export processes of POC and PON. Annual TMF (112–638 g m−2 yr−1) and fluxes of POC and PON (9.6–32.1 g C m–2 yr–1 and 1.2–4.5 g N m−2 yr−1, respectively) in the Ulleung Basin corresponded to the upper limit of values reported for other open seas and oceans in the world. No great seasonal variability in both quantitative (TMF, and fluxes and contents of POC and PON) and qualitative (C/N ratios, and δ13C and δ15N values) estimates of vertical fluxes was observed, reflecting a steady standing stock of chlorophyll a in the upper part of water column. Furthermore, high contents of POC and PON and nearly constant δ13C and δ15N values in sinking particles collected in the sediment traps, indicate that primary production in the euphotic zone may be a good predictor of TMF and export flux of organic matter. Ethanol organosolv fractionation combined with ball milling was conducted on three major agricultural residues: Rice husk (RH), rice straw (RS), and barley straw (BS). The highest lignin extraction yields of RH, RS, and BS were 55.2%, 53.1%, and 59.4% and the purity of lignin recovered was 99.5% for RH and RS, and 96.8% for BS, with similar chemical characteristics, i.e., low molecular weight distributions (1453&ndash, 1817 g/mol) and poly dispersity index (1.15&ndash, 1.28). However, considering the simultaneous production of hemicellulose-derived sugars, distinctive fractionation behaviors were shown for the three agricultural residues. The highest hemicellulose-derived sugar yield was 73.8% when RH was fractionated at 170 &deg, C for 30 min. Meanwhile, very low sugar yields of 31.9% and 35.7% were obtained from RS and BS, respectively. The highest glucan-to-glucose conversion yield from enzymatic hydrolysis of fractionated RH reached 85.2%. Meanwhile, the enzymatic digestibility of the fractionated RS and BS was 60.0% and 70.5%, respectively. Consequently, the fractionation efficiency for RH can be improved with fine refinement. For the case of RS, other fractionation process should be applied to achieve effective fractionation performance.

Acute liver failure (ALF) occurs due to severe liver damage that triggers rapid loss of normal liver function. Here, we investigate the usefulness of an injectable liver extracellular matrix (LECM)-rich hydrogel generated from an optimized decellularization protocol incorporated with silver nanoparticles (AgNPs) as a promising therapy for ALF. First, we optimized a non-destructive protocol for rat liver decellularization to obtain ECM-rich well-preserved scaffold. Then, LECM hydrogel generated from two commonly used decellularization protocols were compared by LECM hydrogel obtained from our optimized protocol. The ALF model was induced by an intraperitoneal (IP) thioacetamide (TAA) injection followed by the IP injection of LECM hydrogel, collagen-AgNP mixture, or LECM hydrogel-AgNP mixture. LECM-rich scaffold and hydrogel were successfully obtained using our optimized decellularization protocol. Use of the LECM hydrogel-AgNP mixture to treat TAA-induced ALF greatly improved liver injury and histological liver regeneration. Interleukin-6 and transforming growth factor-beta expressions were significantly reduced, while albumin, hepatocyte growth factor, and Ki67-positive cells were highly expressed. Moreover, aspartate transaminase and alanine transaminase plasma levels and liver homogenate nitric oxide level were significantly lowered. In conclusion, the LECM hydrogel-AgNP mixture has potential efficient therapeutic and regenerative effects on TAA-induced liver injury.

BackgroundThe objective of this study was to assess the efficacy of intra-articular injections of hyaluronic acid (HA) and a novel, on-site conjugate of HA with autologous fibrinogen in platelet-rich plasma (HA-PRP) in a canine model of osteoarthritis (OA)MethodsTwelve beagle dogs underwent a unilateral resection of the cranial cruciate ligament (CrCL) of the stifle joint. Clinical and radiographic signs of OA were confirmed in all dogs 8 weeks following CrCL resection and prior to treatment. The dogs were randomized into three groups: saline (n= 4), HA (n= 4), and HA-PRP (n= 4). Each dog received intra-articular injections of the respective substance into the affected joint at pre-determined time points. The dogs were assessed for adverse effects for 3 days after each injection and for lameness, pain, range of motion, kinetics, and radiographic OA severity prior to treatment and 3 months after injection. OA severity as determined by radiographic examination was not significantly different among the groups at any time point. The dogs were then humanely euthanatized and the stifle joint assessed by gross and histological examinations.ResultsDogs treated with four weekly injections of HA or two biweekly injections of HA-PRP were significantly (p< 0.05) better than dogs treated with four weekly injections of saline at 2-, 4-, and 12-week time points based on a comfortable range of motion (CROM) and clinical lameness score. Gait analysis measuring symmetry and weight distribution on pressure sensor walkway showed significantly (p< 0.05) improved limb function for dogs treated with HA and HA-PRP compared with dogs treated with saline yet with better clinical outcome for the HA-PRP-treated group at 12 and 20 weeks follow-up. Gross and histological analysis of synovium and articular cartilage demonstrated significant (p< 0.05) improvement by both treatments groups compared to controls. There was however significantly (p< 0.05) less damage to the cartilage in the HA-PRP group compared to the HA-treated group.ConclusionsThese data suggest that while injection of HA and HA-PRP may be sufficient for short-term amelioration of the symptoms associated with OA, treatment with HA-PRP conjugates may be superior, providing significantly better long-term cartilage preservation.

The parameters of the alkaline fractionation process were investigated and optimized using a statistical analysis method to simultaneously remove hemicellulose and ash from rice husk (RH) concomitantly. After the alkaline fractionation process, the residual solid contained high cellulose, and the recovery yield of hemicellulose was enhanced in the fractionated liquid hydrolyzate. The hemicellulosic sugar recovery yield (71.6%), de-ashing yield (&gt, 99%), and lignin removal (&gt, 80%) were obtained at the reaction conditions of 150 °C of temperature, 40 min of reaction time, and 6% (w/v) of NaOH concentration. Subsequently, nano-structured silica was synthesized using black liquor obtained as a by-product of this fractionation process. For the production of nano-structured silica, it was observed that the pH of a black liquor and the heat treatment temperature significantly influenced the textural properties of silica product. In addition, the two-stage bleaching of solid residue followed by colloid milling for the production of high value-added CNF with was attempted. As a result, in addition to 119 g of fermentable sugar, 143 g of high-purity (&gt, 98%) silica with a surface area of 328 m2g−1 and 273.1 g of high-functional CNF with cellulose content of 80.1% were simultaneously obtained from 1000 g of RH.

Ferrite-austenite lightweight steels showing TRansformation-induced plasticity were developed by varying the aging temperature with or without prestraining, and their effects on tensile properties were investigated in relation with microstructural evolution of carbide formation. The aged steels contained austenite, pearlite, and martensite in the ferrite matrix, and the austenite volume fraction decreased with the increasing aging temperature because some austenite grains decomposed to pearlites. This austenite decomposition to pearlite was favorable for the improvement of yield strength, but negatively influenced overall tensile properties. The prestraining promoted the austenite decomposition by a diffusion-controlled phase transformation, and changed the morphology of the cementite from a long lamellar shape to a densely agglomerated particle shape. In order to obtain the large increase in yield strength as well as excellent combination of strength and ductility, the strain-induced aging treatment, i.e., prestraining followed by aging, is important like in the prestrained and 673 K (400 °C)-aged steel. This large increase in yield strength, in spite of a reduction of elongation (65 to 43 pct), was basically attributed to an appropriate amount of decomposition of austenite to pearlite (e.g., 4 vol pct), while having sufficient austenite to martensite transformation (e.g., 14.5 vol pct martensite).

Using first principles density functional theory (DFT) calculations we obtain thermodynamic and kinetic properties of U in an electrorefining process for spent nuclear fuels using a LiCl-KCl eutectic molten salt and Mo as a cathode. The thermodynamic stability of electrodeposited U from the molten salt onto the Mo(110) surface electrode is evaluated by activity coefficients as function of surface coverages of U and Cl. Additionally, ab-initio molecular dynamic simulations combined with the Stokes-Einstein-Sutherland relation enables us to calculate the viscosity of the LiCl-KCl eutectic molten salt. Our results well agree with previously reported experimental data endorsing the credibility. Based on our atomic-level mechanical understanding we propose that an accurate computational model system incorporating the electrochemical conditions of the electrorefining process essential for the purpose of establishing thermodynamic and kinetic database of U, otherwise critical deviations are inevitable. More interestingly, the effect of coadsorption of Cl with U on the Mo(110) surface plays a key role in stabilizing electrodeposited U on the cathode. Our approach can be useful for validating published experimental database and for identifying key factors guiding a rational design of highly efficient electrorefining system for spent nuclear fuels, and thus reducing high-level radioactive nuclear wastes.

Sufficient vitamin D levels are associated with reduced Helicobacter pylori infections, which can cause gastric carcinogenesis. We examined associations between vitamin D concentrations and gastric cancer (GC) prevalence in a Korean population. We analyzed data of 33,119 adults using serum 25-hydroxyvitamin D (25(OH)D) concentrations as a biomarker of vitamin D status. Participants were classified with GC if previously diagnosed as such by a physician. After controlling for age, sex and body mass index (model A), odds ratio (OR) for GC was 0.81 (95% confidence interval [CI]: 0.70, 0.95), with a 5-ng/mL increment in total 25(OH)D concentrations. In fully adjusted models (model B), the OR for GC was 0.84 (95% CI: 0.72, 0.98), with a 5-ng/mL increment in total vitamin D. Following the classification of vitamin D concentrations into three categories or for GC in model A was 0.52 (95% CI: 0.30, 0.92) comparing between higher (&ge, 20 ng/mL) and lower (&lt, 12 ng/mL) total 25(OH)D concentrations. In model B OR for GC was 0.57 (95% CI: 0.32, 1.00) comparing between higher and lower total 25(OH)D concentrations. Our results suggested that high vitamin D concentration was associated with lower ORs of GC in Korean adults.

Harmful algal blooms involving the dinoflagellate Cochlodinium polykrikoides occur every summer off the Korean peninsula's central southern coast. To determine whether Cochlodinium bloom-derived organic carbon is incorporated into the subtidal macrobenthic food web, we compared the δ13C and δ15N values of suspended particulate organic matter (SPOM) and sedimentary organic matter, and macrobenthic consumers between bloom and non-bloom seasons. Chemotaxonomic analysis revealed the presence of Cochlodinium blooms in summer and a predominance of diatoms in autumn. Both the δ13C and δ15N values of SPOM were higher in the bloom than in the non-bloom seasons. Such temporal shifts in the δ13C and δ15N values were also observed for most macrobenthic consumers collected in both seasons. Consistent temporal isotopic shifts in SPOM and macrobenthos revealed that the Cochlodinium bloom-derived carbon was incorporated into the coastal benthic food web, resulting from its increasing availability during blooms.

The development of biocompatible patches that can provide adequate mechanical properties and mimicking the physiological relevance still has enormous challenges in the field of tissue engineering. In this study, the aim is to engineer and develop tri-layered nanofiber patches to synchronous the mechanical properties and mimic the complex structure of living cardiovascular tissues. The developed patches consisting of blended silk fibrin (SF) and polyvinyl alcohol (PVA) composite as a hydrophilic middle layer. The upper and lower layers were composed of polycaprolactone polymer (PCL) and Poly (Lactic Acid) (PLA), respectively. Layer by layer of the developed patches were fabricated by electrospinning technique. The patches characteristics such as morphology, surface topography, and physiochemical, biodegradable and mechanical stability were investigated. Furthermore, a human endothelial cell line (EA. hy926) was exploited to assess the biological properties of the fabricated patches. The results show that the patches had high endothelial cells biocompatibility, mechanical elasticity, and unique nanofibers structure.

Objectives Neonatal hyperbilirubinemia is considered one of the most common causative factors of hearing loss. Preterm infants are more vulnerable to neuronal damage caused by hyperbilirubinemia. This study aimed to evaluate the effect of hyperbilirubinemia on hearing threshold and auditory pathway in preterm infants by serial auditory brainstem response (ABR). In addition, we evaluate the usefulness of the unconjugated bilirubin (UCB) level compared with total serum bilirubin (TSB) on bilirubin-induced hearing loss. Methods This study was conducted on 70 preterm infants with hyperbilirubinemia who failed universal newborn hearing screening by automated ABR. The diagnostic ABR was performed within 3 months after birth. Follow-up ABR was conducted in patients with abnormal results (30 cases). TSB and UCB concentration were compared according to hearing threshold by ABR. Results The initial and maximal measured UCB concentration for the preterm infants of diagnostic ABR ≥40 dB nHL group (n=30) were statistically higher compared with ABR ≤35 dB nHL group (n=40) (P=0.031 and P=0.003, respectively). In follow-up ABR examination, 13 of the ABR ≥40 dB nHL group showed complete recovery, but 17 had no change or worsened. There was no difference in bilirubin level between the recovery group and non-recovery group. Conclusion UCB is a better predictor of bilirubin-induced hearing loss than TSB in preterm infants as evaluated by serial ABR. Serial ABR testing can be a useful, noninvasive methods to evaluate early reversible bilirubin-induced hearing loss in preterm infants.

Decellularization of tissues can significantly improve regenerative medicine and tissue engineering by producing natural, less immunogenic, three-dimensional, acellular matrices with high biological activity for transplantation. Decellularized matrices retain specific critical components of native tissues such as stem cell niche, various growth factors, and the ability to regenerate in vivo. However, recellularization and functionalization of these matrices remain limited, highlighting the need to improve the characteristics of decellularized matrices. Incorporating nanoparticles into decellularized tissues can overcome these limitations because nanoparticles possess unique properties such as multifunctionality and can modify the surface of decellularized matrices with additional growth factors, which can be loaded onto the nanoparticles. Therefore, in this minireview, we highlight the various approaches used to improve decellularized matrices with incorporation of nanoparticles and the challenges present in these applications.

We report the synthesis of 2D nanosheets of GeS and GeSe by facile solution based approaches. The synthesized nanosheets are single-crystalline in nature with lateral dimensions in micrometers. Band structures calculated from DFT calculations predicted a direct bandgap value of 1.67 and 1.37 eV for GeS and GeSe, respectively. The experimental bandgap values (GeS, Eg = 1.6 eV and GeSe, Eg = 1.2 eV) determined from optical measurements are slightly smaller than the predicted ones. Photoresponse measurements of GeS and GeSe nanosheets revealed that the nanosheets are extremely photoresponsive toward the incident light and exhibit a high photoresponsivity of up to 173 and 870 A W−1 under a 405 nm laser diode, respectively. These values are several orders of magnitude higher than those of previous reports for graphene and many other metal chalcogenide nanosheet photodetectors. In addition, the photodetectors show a fast photoresponse time and a specific detectivity on the order of 1013 Jones. These results show that both the GeS and GeSe nanosheets are promising narrow bandgap semiconductors for high performance photodetectors.

An empirical ignition delay model has been developed for the fuel blends of isooctane, n-heptane, toluene and ethanol based on their molar fractions in the stoichiometric condition. The model employs traditional Arrhenius type correlation and cool-flame temperature rise correlations to describe the negative temperature coefficient (NTC) region for the fuel blends. The overall ignition delay of a fuel blend is correlated with individual ignition delay information based on the molar fraction of the fuel component in the mixture. The proposed model is successfully validated against the published experimental ignition delay data using various binary, ternary and quaternary fuel blends of isooctane, n-heptane, toluene and ethanol.

The effect of consumption of two probiotic strains, Lactobacillus curvatus HY7601 and Lactobacillus plantarum KY1032, on weight loss, body adiposity and lipoprotein-associated phospholipase A2 (Lp-PLA2) activities in overweight subjects was examined. After 12-week probiotic treatment, the probiotic group presented reductions in body weight (−0.65 ± 0.23 kg), body fat percentage (−0.57 ± 0.19%) and body fat mass (−616 ± 161 g) measured using DEXA, and L1 subcutaneous fat area (−2.68 ± 1.31 cm2) measured using CT, compared to baseline. The kinetic analysis method using non-isothermal technique was proposed to determine the kinetic parameters for the transesterification reaction of waste pig fat in supercritical alcohols. To investigate the transesterification of waste pig fat, the waste pig fat to alcohol ratio (w/w) was varied from 1:1.5 to 1:2.5 between the temperatures 220 and 290 °C at an interval of 10 °C in a 25 mL batch reactor. The products were analyzed by gas chromatography mass spectrometry. To verify the effectiveness of the proposed kinetic analysis method, the experimental values were compared with the values calculated using the kinetic parameters obtained from this work. It was found that the proposed kinetic analysis method gave reliable kinetic parameters for the transesterification of waste pig fat in supercritical alcohols. Further, it was found that the apparent activation energy for supercritical ethanol was lower than the value for supercritical methanol.

Previous studies have suggested that stearoyl lysophosphatidlycholine (LPC) protects against lethal experimental sepsis by inhibiting lipopolysaccharide (LPS)-induced extracellular release of high-mobility group box 1 (HMGB1). However, limited information exists on the mechanism by which stearoyl-LPC suppresses the extracellular release of HMGB1 in monocyte/macrophages stimulated with LPS. In this study, we found that stearoyl-LPC increased the phosphorylation of AMP-activated protein kinase (AMPK) in macrophages. Exposure of LPS-stimulated macrophages to stearoyl-LPC decreased the extracellular release of HMGB1 in peritoneal macrophages, which were inhibited by the AMPK inhibitor, compound C. In addition, stearoyl-LPC-mediated suppression of HMGB1 release was abolished by siRNA-mediated knock-down of AMPKα1. Stearoyl-LPC increased the phosphorylation of acetyl-CoA carboxylase (ACC), a downstream target of activated AMPK, in mice lungs and decreased HMGB1 levels in bronchoalveolar lavage fluids in mice administered LPS. These results reveal a novel mechanism by which stearoyl-LPC regulates LPS-mediated cellular translocation of HMGB1.

Background: Flecainide acetate is a drug used primarily for cardiac arrhythmia. Some studies also imply that flecainide acetate has the potential to regulate inflammatory-immune responses; however, its mechanism of action is contended. We determined the effects of flecainide acetate on lipopolysaccharide (LPS)-stimulated human neutrophils in vitro and on mortality in a septic rat model. Methods: Neutrophils from human blood were cultured with varying concentrations of flecainide acetate (1 μM, 10 μM, or 100 μM) with or without LPS (100 ng/ml). To assess neutrophil activation, the protein levels of tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-6 and IL-8 were measured after a 4-hour culture period. To assess the intracellular signaling pathways, the levels of phosphorylation of p38 mitogen-activated protein kinase (p38), extracellular signal-regulated kinase (ERK) 1/2, and c-Jun N-terminal kinase (JNK) were measured after a 30-minute culture period, and the nuclear translocation of nuclear factor (NF)-κB was measured after a 1-hour culture period. Additionally, the survival rate was investigated in a rat sepsis model. Results: Flecainide acetate down-regulated the activation of proinflammatory cytokines, including TNF-α and IL-6 and IL-8, and intracellular signaling pathways including ERK 1/2 and NF-κB. Flecainide acetate also improved the survival rate in the rat sepsis model. Conclusions: Collectively, these findings indicate that flecainide acetate can improve survival in a rat sepsis model by attenuating LPS-induced neutrophil responses. We therefore suggest that flecainide acetate plays an important role in modulating inflammatory-immune responses.

Background Mammary gland tumors are the most common tumors in sexually intact female dogs; however, they are rare in male dogs. This study was conducted to investigate the relationship between sexual hormones and mammary gland tumors in a male dog. Case presentation A 13-year-old, intact male Cocker Spaniel presented to the Veterinary Teaching Hospital of Kangwon National University, Republic of Korea, with an acute right ruptured caudal abdominal mass. Physical examination revealed a 14 × 14 cm ruptured mass in the right caudal abdomen, as well as a 1.5 × 1.5 cm mass in the first right mammary gland. The estrogen and progesterone concentrations in serum were within normal levels. Total mastectomy was done on the right side mammary glands. Following surgery, the site was fully recovered; however, a mass that had grown to 2 × 2 cm was found in the left fifth mammary gland and a testis tumor was also found over the period of 4 months. Mastectomy was performed on the left caudal mammary gland and castration was also performed. After the final surgery, the dog fully recovered. Histopathological examination of all three masses revealed high grade mammary adenocarcinoma in the mammary gland and the testis was diagnosed as Leydig cell adenoma. Immunohistochemical analysis revealed that the estrogen and progesterone receptors were expressed on limited cells in mammary and testis tumors. Conclusion The results of this study suggest that mammary tumors and testes tumors can occur in male dogs without relationship to female sexual hormone.

N 값은 각각 평균 −18.9(±1.7)‰과 11.6(±0.7)‰로 나타났는데, 이와 같은비값들은 CPOM 보다는 FPOM의 월별 변동과 비슷한 경향을 보여 이들이 CPOM에 비하여 상대적으로 FPOM에더 높은 영양 의존도를 가진다는 것을 강하게 시사해 주었다. 그러나 미더덕의 먹이원으로서 CPOM과 FPOM의 상대적인 중요성은 시기에 따른 각 요소의 가용성(availability)에 크게 의존하는 것으로 나타났는데, 이와 같은 결과는부유입자물질의 크기에 관계없이 미더덕에 의한 비선택적 먹이 섭식을 잘 반영하는 듯 하였다. 본 연구 결과는 미더덕의 지속적인 양식생산을 유지하기 위해 가용 먹이원으로서 식물플랑크톤과 함께 미소 부유입자유기물의 유용성을고려해야함을 잘 나타내 주었다.

Hydroxyapatite/cellulose acetate composite webs were fabricated by an electro-spinning process. This electro-spinning process makes it possible to fabricate complex three-dimensional shapes. Nano fibrous web consisting of cellulose acetate and hydroxyapatite was produced from their mixture solution by using an electro-spinning process under high voltage. The surface of the electro-spun fiber was modified by a plasma and alkaline solution in order to increase its bioactivity. The structure, morphology and properties of the electro-spun fibers were investigated and an in-vitro bioactivity test was evaluated in simulated body fluid (SBF). Bioactivity of the electro-spun web was enhanced with the filler concentration and surface treatment. The surface changes of electro-spun fibers modified by plasma and alkaline solution were investigated by FT-IR (Fourier Transform Infrared Spectroscopy) and XPS (X-ray Photoelectron Spectroscopy).

We have isolated two isomeric solids 1 and 2 of N,N′-bis(3,5-dichlorosalicylidene)-2,2′-ethylenedianiline and characterized by IR, UV/Vis, X-ray powder diffraction, thermogravimetric analysis/differential thermal analysis, and X-ray crystallography. Although the solids are same formulas, each shows different colors and crystal structures. Orange solid (1) shows endo conformation while yellow solid (2) exhibits exo form depending on packing modes. UV/Vis spectra of 1 and 2 appear very similar patterns in the solid state; however, the bands of 1 are slightly red-shifted compared with those of 2. 1 displays a strong fluorescent emission band at ~582 nm while 2 shows an intense fluorescent signal at ~563 nm. The charge density populations of 1 and 2 have been studied by computational simulations using density functional theory at pbe1pbe/6-311G** level. The calculated highest occupied molecular orbital and lowest unoccupied molecular orbital energies of 1 and 2 confirm that charge transfer occurs within the organic molecules. The energy difference of HOMO-LUMO in 1 is smaller slightly than that of 2 about 0.05 eV (~17 nm). Copyright © 2014 John Wiley & Sons, Ltd.

By using first principles DFT calculations, we reveal oxygen reduction reaction mechanisms in N-doped graphene (N-Gr). Considering both the morphology and the concentration of dopant N atoms in bulk and edge N-Gr forms, we calculate the energies of a large number of N-Gr model systems to cover a wide range of possible N-Gr structures and determine the most stable N-Gr forms. In agreement with experiments, our DFT calculations suggest that doping levels in stable N-Gr forms are limited to less than approximately 30 at. % N, above which the hexagonal graphene framework is broken. The ground state structures of bulk and edge N-Gr forms are found to differ depending on the doping level and poisoning of the edge bonds. Oxygen reduction reaction mechanisms are evaluated by using Gibbs free-energy diagrams with and without water solvation. Our results indicate that N doping significantly alters the catalytic properties of pure graphene and that dilutely doped bulk N-Gr forms are the most active.

The relative importance of different organic matter sources to macrobenthic consumers was investigated at two vegetational habitats (Phragmites australis and Suaeda japonica) and at a bare intertidal flat in Suncheon Bay in May and August 2010, using stable carbon and nitrogen isotope tracers. The δ13C of consumers was much narrower in range than that of potential food sources (microphytobenthos, marsh plants, and riverine and marine particulate organic matter), and values were similar between sites. However, cluster analysis based on the consumer’s isotope values showed that they are divided according to functional groups rather than habitat type and taxonomical group, suggesting that the exploitation of different dietary sources depends on feeding strategy. Except for two suspension feeders and nonselective deposit feeders at the Phragmites bed, most consumers had similar δ13C to those of microphytobenthos. An isotopic mixing model calculation revealed the consistent importance of microphytobenthos to the nutrition of benthic invertebrates among wetland habitats in different seasons. In contrast, very little contribution of marsh plant-derived organic matter to consumer nutrition was detected at all sites. Furthermore, our results suggest that the summer monsoon event does not lead to a seasonal shift in dietary sources of consumers in this coastal wetland system.

SUMMARY By applying a rigorous computational procedure combining first principles density functional theory (DFT) calculations and statistical mechanics, we acquire thermochemical properties of materials for a pyroprocessing system recycling spent nuclear fuels. Cluster expansions to DFT obtained energies parameterize atomic interaction potentials of Cl-Cl and Cl-U adsorbed on W(110) surface from a molten salt (KCL-LiCl). Using these databases of the long-range and multibody interactions, Monte Carlo simulations identify thermodynamically stable configurations of the adsorbates on the W(110) surface in grand canonical open system at T = 773 K. Our results indicate that Cl atoms adsorbed at the interface of the molten salt and W(110) surface substantially drive electrochemical deposition of U ions at low chemical potential of Cl. This behavior, however, stops after approximately 1/3 ML coverage of U because the atomic sites on W(110) surface are mostly blocked by adsorbed Cl, which implies that the attractive interactions of Cl-W are stronger than Cl-U as well as the repulsive interactions between U atoms are effective at these coverage ranges. We also predict the solubility limit of U ion in the molten LiCl-KCl phases at T = 773 K should be about 5 atomic percent, which well agrees with previous reports by experimental measurements. This study indicates that accurate characterization of the stable Cl structure at the interface is vital for understanding the fundamental mechanisms of recycling spent nuclear fuels and for screening high functional electrode materials in the pyroprocessing system. Using first principles DFT (density functional theory), we have examined the thermochemical mechanism of electrorefining spent uranium (U) from a LiCl–KCl molten salt on a tungsten (W) surface. We calculated 197 different U/W(110) surfaces to identify the most thermodynamically and electrochemically stable structures as a function of U and Cl coverages. The results indicate that local structures of the double-layer interface between the W(110) surface and the LiCl–KCl salt are the key factors governing the electrorefining performance. The results also provide important thermodynamic properties for the design of efficient recycling systems for spent nuclear fuels, such as pyroprocessing technologies, and may be applicable as well to general electrochemical applications involving strong redox reactions of transition metals exposed to non-aqueous solutions.

Using first principles density functional theory calculations, we systematically studied oxygen reduction reactions (ORRs) on N-doped graphene (N-Gr) with and without a Cu metallic support (N-Gr/Cu(111) surface). Our ab-initio calculated free energy diagrams surprisingly show that oxygen molecules are dissociated into two oxygen atoms on the N-Gr/Cu(111) surface, which alters the well-known associative ORR mechanisms on pure graphene and N-Gr. Our results, however, indicate that the mechanistic shift does not directly lead to enhancement of ORR activity once water molecules solvate the N-Gr/Cu(111) surface via substantially stabilized intermediates such as O, OH, and OOH. Our results suggest that transition metal supports can be a promising way to control the ORR mechanism on doped graphene and that the evaluation of ORR activity requires understandings of not only the electronic structures of doped graphene but also the chemical interactions between the intermediates and solvating water molecules.

To examine the influence of habitat alteration by tideflat reclamation on the physiological ecology of Ruditapes philippinarum, seasonal variations in its condition, gross biochemical composition, and reproductive cycle were compared for a yearly cycle between a reclaimed flat (Gomsohang, GS) and a natural flat (Hajun, HJ) in Gomso Bay, Korea. Concentrations of chlorophyll a and seston (as well as its energy value as food available to the clam) were consistently higher at HJ than at GS. Condition, dry tissue weight, and energy reserves (proteins and carbohydrates) of the clams were much higher at HJ than at GS during spring-summer, when fast growth and gametogenic development occur. Furthermore, their spring gametogenic development and spawning were advanced at HJ compared with GS. Our results suggest that the Manila clam encounters nutritionally stressful environmental conditions in altered habitats after tideflat reclamation that lead to lowered nutrient accumulation and a changing reproductive cycle.

Previous studies have shown that sauchinone modulates the expression of inflammatory mediators through mitogen-activated protein kinase (MAPK) pathways in various cell types. However, little information exists about the effect of sauchinone on neutrophils, which play a crucial role in inflammatory process such as acute lung injury (ALI). We found that sauchinone decreased the phosphorylation of p38 MAPK in lipopolysaccharide (LPS)-stimulated murine bone marrow neutrophils, but not ERK1/2 and JNK. Exposure of LPS-stimulated neutrophils to sauchinone or SB203580, a p38 inhibitor, diminished production of tumor necrosis factor (TNF)-α and macrophage inflammatory protein (MIP)-2 compared to neutrophils cultured with LPS. Treatment with sauchinone decreased the level of phosphorylated ribosomal protein S6 (rpS6) in LPS-stimulated neutrophils. Systemic administration of sauchinone to mice led to reduced levels of phosphorylation of p38 and rpS6 in mice lungs given LPS, decreased TNF-α and MIP-2 production in bronchoalveolar lavage fluid, and also diminished the severity of LPS-induced lung injury, as determined by reduced neutrophil accumulation in the lungs, wet/dry weight ratio, and histological analysis. These results suggest that sauchinone diminishes LPS-induced neutrophil activation and ALI.

The effects of Al content on cracking phenomena occurring during cold rolling of ferritic lightweight steels were investigated in relation to microstructural modification including κ-carbides. Three steels were fabricated, varying the Al content between 4 and 6 wt.%, and the center and edge areas of steel sheets containing 6 wt.% Al were cracked during cold rolling. The three steels were composed of ferrite grains and κ-carbides in a banded shape, and the overall volume fraction of κ-carbide increased with increasing Al content. The shape of lamellar κ-carbides inside κ-carbide bands was changed from short and thin to long and thick. Microstructural observation of the deformed region of fractured tensile specimens revealed that deformation bands were homogeneously formed in wide areas of ferrite matrix in the steels containing 4–5 wt.% Al, and κ-carbide bands and boundary κ-carbides were hardly cracked. In the steel containing 6 wt.% Al, however, microcracks were initiated at grain boundary κ-carbides and long lamellar κ-carbides inside κ-carbide bands. They led to the center or edge cracking during cold rolling. To prevent or minimize cracking, it was necessary to avoid the lengthening or thickening of lamellar κ-carbides. Therefore, it was recommended that the steels should be rapidly cooled from the finish rolling temperature to the coiling temperature through the formation temperature of κ-carbide.
