Pneumonia, such as acute lung injury (ALI), has been a type of lethal disease that is generally caused by uncontrolled inflammatory response and excessive generation of reactive oxygen species (ROS). Herein, we report Fe-curcumin-based nanoparticles (Fe-Cur NPs) with nanozyme functionalities in guiding the intracellular ROS scavenging and meanwhile exhibiting anti-inflammation efficacy for curing ALI. The nanoparticles are noncytotoxic when directing these biological activities. Mechanism studies for the anti-inflammation aspects of Fe-Cur NPs were systematically carried out, in which the infected cells and tissues were alleviated through downregulating levels of several important inflammatory cytokines (such as TNF-α, IL-1β, and IL-6), decreasing the intracellular Ca2+ release, inhibiting NLRP3 inflammasomes, and suppressing NF-κB signaling pathways. In addition, we performed both the intratracheal and intravenous injection of Fe-Cur NPs in mice experiencing ALI and, importantly, found that the accumulation of such nanozymes was enhanced in lung tissue (better than free curcumin drugs), demonstrating its promising therapeutic efficiency in two different administration methods. We showed that the inflammation reduction of Fe-Cur NPs was effective in animal experiments and that ROS scavenging was also effectively achieved in lung tissue. Finally, we revealed that Fe-Cur NPs can decrease the level of macrophage cells (CD11bloF4/80hi) and CD3+CD45+ T cells in mice, which could help suppress the inflammation cytokine storm caused by ALI. Overall, this work has developed the strategy of using Fe-Cur NPs as nanozymes to scavenge intracellular ROS and as an anti-inflammation nanodrugs to synergistically cure ALI, which may serve as a promising therapeutic agent in the clinical treatment of this deadly disease. Background:Nuezhenide (NZD), an iridoid glycoside isolated from Ilex pubescens Hook. & Arn. var. kwangsiensis Hand.-Mazz., used as a traditional Chinese medicine for clearing Hook. & Arn. var. kwangsiensis Hand.-Mazz., used as a traditional Chinese medicine for clearing away heat and toxic materials, displays a variety of biological activities such as anti-tumor, antioxidant, and other life-protecting activities. However, a few studies involving anti-inflammatory activity and the mechanism of NZD have also been reported. In the present study, the anti-inflammatory and antioxidative effects of NZD are illustrated.Objective:This study aims to test the hypothesis that NZD suppresses LPS-induced inflammation by targeting the NF-κB pathway in RAW264.7 cells.Methods:LPS-stimulated RAW264.7 cells were employed to detect the effect of NZD on the release of cytokines by ELISA. Protein expression levels of related molecular markers were quantitated by western blot analysis. The levels of ROS, NO, and Ca2+were detected by flow cytometry. The changes in mitochondrial reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) were observed and verified by fluorescence microscopy. Using immunofluorescence assay, the translocation of NF-κB/p65 from the cytoplasm into the nucleus was determined by confocal microscopy.Results:NZD exhibited anti-inflammatory activity and reduced the release of inflammatory cytokines such as nitrite, TNF-α, and IL-6. NZD suppressed the expression of the phosphorylated proteins like IKKα/β, IκBα, and p65. Besides, the flow cytometry results indicated that NZD inhibited the levels of ROS, NO, and Ca2+in LPS-stimulated RAW264.7 cells. JC-1 assay data showed that NZD reversed LPS-induced MMP loss. Furthermore, NZD suppressed LPS-induced NF-B/p65 translocation from the cytoplasm into the nucleus.Conclusions:NZD exhibits anti-inflammatory effects through the NF-κB pathway on RAW264.7 cells.

Economical and efficient non-noble metal catalysts should be developed practically, instead of commercial Pt/C for fuel cells. In this paper, manganese, nitrogen co-doped porous carbon (Mn–N–C) was synthesized to catalyze oxygen reduction reaction (ORR) through the one-step carbonization of ZIF-8 in the Mn-containing (MnCl2) atmosphere. During the carbonization process, MnCl2 gas was captured with ZIF-8 and then transformed into uniform Mn–N active sites distributed in the porous carbon materials. The Mn–N–C catalyst exhibited plentiful porous structures, large specific surface areas, high graphitization and conductivity, which contributed to the transfer and transport of charge and exposed more active sites. The Mn–N–C catalyst exhibited superior catalytic ability in alkaline and acidic solutions. Half-wave potential of the Mn–N–C could reach 0.88 and 0.73 V in 0.1 M KOH and 0.5 M H2SO4, respectively. In addition, the Mn–N–C catalyst showed a prominent stability after the stability test of 18,000 s. Excellent electrochemical performance and endurance make the Mn–N–C expect to be an effective ORR catalyst to build high-performance fuel cells.

Oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) are the main reaction processes in metal-air batteries, fuel cells, and water electrolysis devices. It is of great significance to develop highly efficient and low-cost bifunctional catalysts for these two reactions. In this study a rapid deflagration method was reported by using Vulcan XC/72 carbon black, sublimated sulfur, potassium nitrate, urea and ammonium nitrate as raw materials to fast synthesize N and S co-doped and oxygen-functionalized carbon materials (D-NSOCs) in one step. The prepared D-NSOCs electrocatalyst exhibits excellent ORR and OER bifunctional catalytic reactivity with a small potential gap (ΔE) of 0.77 V between the corresponding potential when the OER current density reaches 10 mA cm−2 (Ej = 10) and the half-wave potential (E1/2), which is slightly better than 0.78 V for the commercial Pt/C + IrO2. This deflagration method is generally applicable to other carbon materials. It can dope nitrogen and sulfur elements into carbon materials and realize the functionalization of oxygen on the surface of carbon materials.

The development of bifunctional catalysts possessing high efficiency and excellent stability was still a challenge as regards overall water splitting. Herein, a reasonable space division strategy of Ni foam (NF) was proposed to design a bifunctional catalyst. Ni-P alloy was loaded on carbon nanotubes (CNTs) through Pd-free electroless plating (Ni-P/CNTs). Subsequently, the Ni-P/CNT catalysts mixed with melamine were annealed for N doping. The N-doped CNTs coated with N-doped Ni3P and Ni nanoparticles (N-Ni3P-Ni/N-CNTs) were obtained. Finally, the N-Ni3P-Ni/N-CNTs were used as a filler to divide the pore size of NF by electroplating Ni, obtaining a network-like hierarchical porous electrode (N-Ni3P-Ni/N-CNTs/NF). This hierarchical porous structure exposed abundant catalytically active sites, facilitated the mass transfer, and prevented the catalyst agglomeration and corrosion that promote electrolyte contact and favor facile electron transfer kinetics. On the basis of the rational design, a self-supported N-Ni3P-Ni/N-CNTs/NF electrode showed a high specific surface area and exhibited superior electrocatalytic performances. The electrode achieved low overpotential of 73 and 270 mV at 10 mA cm–2 for the hydrogen and oxygen evolution reaction, respectively. Besides, it required a cell voltage of only 1.57 V to achieve 20 mA cm–2 when assembled in a 1 M KOH electrolyte for overall water splitting. Hence, this study represented a rational design of self-supported bifunctional electrode for overall water splitting.

Gate reliability and its degradation mechanism were studied for the normally off high-electron-mobility transistors (HEMTs) with regrown p-GaN gate and AlN/SiN x stack passivation. Through comparing the forward leakage current in two designed structures, the conduction mechanism is determined to be Fowler-Nordheim tunneling whose current is independent of temperature when the Pd/p-GaN Schottky junction is under high electric field. Even for the regrown p-GaN gates, this Schottky junction fails at first, resulting in an abrupt increase in gate current. The maximum gate operation voltage with a failure rate of 1% for 10-year lifetime is estimated to be about 6.87 and 6.07 V at room temperature by adopting power law and exponential law as extrapolation fitting, respectively. Degradation process monitoring reveals that the net acceptor concentration $N_{\mathrm {A}}$ extracted through $C$ – $V$ fitting presents an apparent decreasing trend from $3.8\times 10^{19}$ to $1.1\times 10^{19}$ cm−3 with the stress time increased from 30 to 2600 s. This is assumed to be related with the defects generation in the Schottky depletion region under high tunneling current and high electric field. These analyses show the feasibility of the normally off HEMTs with a regrown p-GaN gate and AlN/SiN x stack passivation for practical applications, giving directions for further improving the gate breakdown voltage and lifetime.

Acute lung injury (ALI) is a serious clinical disease. Rotundic acid (RA), a natural ingredient isolated from Ilex rotunda Thunb, exhibits multiple pharmacological activities. However, RA's therapeutic effect and mechanism on ALI remain to be elucidated. The present study aimed to further clarify its regulating effects on inflammation in vitro and in vivo. Our results indicated that RA significantly inhibited the overproduction of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), cyclooxygenase-2 (COX-2), and inducible nitric oxide synthase (iNOS). RA decreased ROS production and calcium influx. In addition, RA inhibited the activation of PI3K, MAPK, and NF-κB pathways and enhanced the activity of nuclear factor E2-related factor 2 (Nrf2) signaling. The cellular thermal shift assay and docking results indicated that RA bind to TLR4 to block TLR4 dimerization. Furthermore, RA pretreatment effectively inhibited ear edema induced by xylene and LPS-induced endotoxin death and had a protective effect on LPS-induced ALI. Our findings collectively indicated that RA has anti-inflammatory effects, which may serve as a potential therapeutic option for pulmonary inflammation.

A high-performance quasi-vertical GaN Schottky barrier diode (SBD) was successfully fabricated by using a high-quality n−-GaN drift layer with a precisely-controlled n-type doping. A high current on/off ratio of 1010, an ideality factor of 1.03, a low specific on-resistance of 1.41 $\text{m}\Omega \cdot $ cm2, and a relatively high breakdown voltage (BV) of 250 V have been achieved for the SBD without edge termination. Furthermore, with an Argon-implanted termination, the as-fabricated GaN-on-Si SBD shows a record high BV of 405 V, yielding a critical electric field of ~ 2 MV/cm, while the forward conduction characteristics are well maintained.

A Mn coordinated with N atoms aligned hierarchical porous carbon catalyst is prepared through an inorganic metal salt sublimation doping strategy. Gelatin is served as a carbon source and N source, Ca2+ is acted as templates to establish aligned porous structure during carbonization. MnCl2 sublimates into gas to serve as Mn source after reaching the melting point. This method can effectively avoid the agglomeration of Mn atoms, which is beneficial to form Mn-Nx active sites. The prepared optimal catalyst exhibits a large specific surface area with an aligned hierarchical porous structure. XAFs result demonstrates that Mn coordinates with N atoms to form Mn-Nx configuration in the carbon structure. Notably, it exhibits outstanding catalytic ORR performance with a positive half-wave potential (0.86 V vs. RHE) and excellent durability, superior to Pt/C (20 wt%) catalyst under alkaline medium. Meanwhile, enhanced catalytic ORR performance and stability in an acidic medium are also achieved.

With the serious intensification of energy shortage and greenhouse effect, people begin to look for the sustainable energy sources to replace fossil energy sources. Herein, self-supporting expanded graphite sheet (SSEGS) was developed as an ideal catalyst support through electrochemically intercalating flexible graphite sheet in alkaline solution. Electroless deposition was employed to synthesize Ni–Cu–P alloy on SSEGS and then an amorphous NiFe hydroxide/Ni–Cu–P/SSEGS (NiFe–OH/Ni–Cu–P/SSEGS) composite catalyst was further constructed through electrodeposition. Benefitting from the unique structural advantage of SSEGS and the synergistic effect between two amorphous Ni-based materials (Ni–Cu–P alloy and NiFe–OH), the resulting electrode exhibited superior bifunctional electrocatalytic performance in 1 M KOH. For H2 evolution reaction and O2 evolution reaction, the NiFe–OH/Ni–Cu–P/SSEGS composite catalyst could reach 10 mA cm−2 at low overpotentials of 75 and 240 mV, respectively. Remarkably, the two-electrode system driven by NiFe–OH/Ni–Cu–P/SSEGS as the anode and cathode could afford 10 mA cm−2 at a low cell voltage of 1.56 V vs. RHE. And after the 12 h stability test, the cell voltage at 10 mA cm−2 increased by only 7 mV, indicating that the two-electrode system had excellent stability. The preparation of NiFe–OH/Ni–Cu–P/SSEGS material with superior bifunctional electrocatalytic performance has a significance influence to the development and expansion of hydrogen production technology.

Lung cancer is the leading cause of cancer death and the most common malignant tumor, the long-term survival of which has stagnated in the past several decades. Pileostegia tomentella Hand. Mazz is a traditional Chinese medicine called "Zhongliuteng" (ZLT) in the pharmacopeia, which has been proved to possess a potent anti-tumor effect on various cancers. In this study, the effects of ZLT N-butanol extraction (ZLTN) and ZLT ethyl acetate extraction (ZLTE) on the viability of non-small cell lung cancer cell (NSCLC) lines H1299 and A549 were evaluated. Here, we firstly reported that ZLTE significantly inhibited H1299 cells growth without affecting the release of lactate dehydrogenase (LDH). In addition, ZLTE induced caspase-dependent apoptosis in a concentration-dependent manner and increased the expression cleaved-PARP and decreased pro-caspase-3, pro-caspase-7, pro-caspase-8, and pro-caspase-9. Moreover, ZLTE increased the level of cellular reactive oxygen species (ROS) in H1299 cells to lead to apoptosis, which was reversed by N-acetyl-cysteine (NAC). Taken together, our results revealed that ZLTE induced caspase-dependent apoptosis via ROS generation, suggesting that ZLTE is a promising herbal medicine for the treatment of NSCLC.

Mediator complex subunit 16 (MED16) is a component of the mediator complex and functions as a coactivator in transcriptional events at almost all RNA polymerase II–dependent genes. In this study, we report that the expression of MED16 is markedly decreased in papillary thyroid cancer (PTC) tumors compared with normal thyroid tissues. In vitro, MED16 overexpression in PTC cells significantly inhibited cell migration, enhanced sodium/iodide symporter expression and iodine uptake, and decreased resistance to radioactive (131)I (RAI). Conversely, PTC cells in which MED16 had been further knocked down (MED16(KD)) exhibited enhanced cell migration, epithelial–mesenchymal transition, and RAI resistance, accompanied by decreased sodium/iodide symporter levels. Moreover, cell signaling through transforming growth factor β (TGF-β) was highly activated after the MED16 knockdown. Similar results were obtained in MED12(KD) PTC cells, and a co-immunoprecipitation experiment verified interactions between MED16 and MED12 and between MED16 and TGF-βR2. Of note, the application of LY2157299, a potent inhibitor of TGF-β signaling, significantly attenuated MED16(KD)-induced RAI resistance both in vitro and in vivo. In conclusion, our findings indicate that MED16 reduction in PTC contributes to tumor progression and RAI resistance via the activation of the TGF-β pathway.

Background Cryptotanshinone (CPT), as a major component of Salvia miltiorrhiza Bunge (Danshen), displays many pharmacological activities including anti-inflammatory effects. However, the exact cellular and molecular mechanisms of the anti-inflammatory activities of CPT remain to be elucidated. The present study was aimed to clarify its mechanisms on lipopolysaccharide (LPS)-induced inflammatory responses in mouse macrophages, RAW264.7 cells. Methods In the current study, the anti-inflammatory properties of CPT were evaluated using LPS-stimulated RAW264.7 cell model. MTT assay was used to determine the viability of RAW264.7 cells. The anti-inflammatory effects of CPT were measured based on the detection of nitric oxide (NO) production (Griess and flow cytometry assay), and tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) release (ELISA). Cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) enzyme expressions were also determined by western blotting. Besides, by using flow cytometry, we also evaluated the effect of CPT on LPS-induced calcium influx. Finally, the underlying anti-inflammatory mechanisms of CPT were investigated using western blotting to assess the protein levels of toll-like receptor 4 (TLR4), myeloid differentiation factor 88 (MyD88), phosphatidylinositol 3-kinase (PI3K)/AKT, nuclear factor erythroid 2 related factor 2 (Nrf2), mitogen-activated protein kinase (MAPK), and nuclear factor-kappa B (NF-κB) pathways. Results Our data showed that CPT inhibited LPS-induced pro-inflammatory cytokine release like IL-6, and TNF-α, as well as NO production. It displayed a significant inhibitory effect on the protein expressions such as iNOS, COX-2, NF-κB pathway like inhibitor of kappa B kinase (IKK)α/β, inhibitor of kappa B (IκB)-α and NF-κB/p65, PI3K/AKT pathway like PI3K and AKT, and MAPK pathway like c-Jun N-terminal kinase (JNK)1/2, extracellular signal-regulated kinase (ERK)1/2, and p38, in LPS-stimulated RAW264.7 macrophages. Moreover, the immunofluorescence results indicated that CPT suppressed NF-κB/p65 translocation from the cytoplasm into the nucleus. Further investigations showed that CPT treatment increased NAD(P)H quinone oxidoreductase-1 (NQO1) and heme oxygenase-1 (HO-1) expressions together with its upstream mediator, Nrf2. In addition, CPT inhibited LPS-induced toll-like receptor 4 (TLR4) and MyD88 expressions in RAW264.7 macrophages. Conclusions Collectively, we suggested that CPT exerted significant anti-inflammatory effects via modulating TLR4-MyD88/PI3K/Nrf2 and TLR4-MyD88/NF-κB/MAPK pathways.

The preparation of low-cost, high-efficiency and stable oxygen reduction reaction (ORR) catalysts is highly anticipated but remains a challenge. Herein, a novel strategy of CaCl2 solution inducing gelatin crosslinking and acting as aligned pore template was put forward for preparation of Fe-N doped aligned hierarchical porous carbon. Furthermore, the microtopography and length of aligned pore can be controlled by adding NaCl solution. The prepared catalyst exhibited short-range aligned honeycomb hierarchical porous structure with large specific surface area and highly homogeneous distribution of Fe-Nx active sites (Fe-NHHPC-900). Notably, in addition to excellent durability in the alkaline electrolyte, Fe-NHHPC-900 possessed excellent ORR performance with positive half-wave potential of 0.86 V, superior to commercial Pt/C catalysts. As electrode materials of supercapacitors, Fe-NHHPC-900 also displayed a high specific capacitance, excellent rate performance and excellent stability. This work also proved that the unique aligned porous structure can effectively optimize catalyst activity and improve supercapacitor performance.

Polymeric barrier materials are critical in contemporary industries for food, medicine, and chemical packaging. However, these materials, such as PET films, are impeded by the optimization of barrier properties by virtue of molecular design. Herein, a new methyl methacrylate-methyl acrylate-diallyl maleate-maleic acid (MMA-MAc-DAM-MA) was synthesized to tailor the surface properties of PET films for maximizing oxygen barrier properties. During the MMA-MAc-DAM-MA coating and curing process, the chemical structure evolutions of MMA-MAc-DAM-MA coatings were characterized, indicating that the cross-linking conversion and proportion of –COOH groups are critical for the oxygen barrier properties of coatings. The inherent –COOH groups are transformed into designed structures, including intramolecular anhydride, inter-chain anhydride and retained carboxylic acid. Therein, the inter-chain anhydride restraining the activity of coated polymer chain mainly contributes to enhanced barrier properties. The thermal properties of novel coatings were analyzed, revealing that the curing behavior is strongly dependent on the curing temperatures. The impacts of viscosity of the coating solution, coating velocity, and coating thickness on the oxygen permeability (Po2) of the coatings were investigated using a gas permeability tester to explore the optimum operating parameters during practical applications, which can reduce the Po2 of PET film by 47.8%. This work provides new insights on advanced coating materials for excellent barrier performance.

We demonstrate slot waveguides on ultra-silicon-rich nitride designed for optimal high group index birefringence $( > 1)$ . High speed characterization using 30Gb/s non-return-to-zero and 56Gb/s pulsed amplitude modulation 4 data are performed on fabricated waveguides showing error-free performance.

Electrolysis of seawater can be a promising technology, but chloride ions in seawater can lead to adverse side reactions and the corrosion of electrodes. A new transition metal boride-based self-supported electrocatalyst was prepared for efficient seawater electrolysis by directly soaking nickel foam (NF) in a mixture of phenolic resin (PR) and boron carbide (B4 C), followed by an 800 °C annealing. During PR carbonization process, the reaction of B4 C and NF generated nickel boride (Nix B) with high catalytic activity, while PR-derived carbon coating was doped with boron atoms from B4 C (B-CPR ). The B-CPR coating fixed Nix B/B4 C particles in the frames and holes to improve the space utilization of NF. Meanwhile, the B-CPR coating effectively protected the catalyst from the corrosion by seawater and facilitates the transport of electrons. The optimal Nix B/B4 C/B-CPR /NF required 1.50 and 1.58 V to deliver 100 and 500 mA cm-2 , respectively, in alkaline natural seawater for the oxygen evolution reaction.

Infrared (IR)-emitting RE doped materials have been extensively used to fabricate active components of integrated optical devices in various fields, such as fiber amplifiers, telecommunications, optoelectronics, and waveguides. Among various RE elements, trivalent erbium ions (Er 3+) are of great interest since their emissive behavior span the low loss telecommunication window of 1300–1650 nm. In this paper, we report two types of polymeric waveguide amplifiers. 8 cm long, lithographically patterned spiral waveguides provide 8 dB of gain using a 980 nm pump power of 95 mW. Gain is observed from 1530 to 1590 nm. We further report the first demonstration of polymeric waveguide amplifiers fabricated using 3D printing methods based on two-photon lithography, paving the way for rapid prototyping of active 3D printed devices and active photonic devices which may transcend planar limitations.

Due to the high price and low reserve of platinum (Pt) base precious catalysts, and non-noble metal catalysts are unable to catch up with the overpotential of Pt-based electrocatalysts, more and more attention has been paid to study low-noble metal catalysts. In this work, we report a highly active and steady electrocatalyst with ruthenium and cobalt bimetal encapsulated in nitrogen-doped carbon material derived of ZIF-67. ZIF-67 was firstly calcined under vacuum to form nitrogen-doped carbon containing the cobalt particles, and then made Ru atoms partly replaced some Co atoms to prepare the ruthenium and cobalt bimetal catalyst. The electrocatalyst is prepared with low loading Ru (3.5 wt%) and has an excellent hydrogen evolution reaction (HER) catalytic performance in acidic and alkaline medium. It displays the overpotential of only 23 mV at 10 mA cm−2 in alkaline solution, which even exceeds commercial 20 wt% Pt/C catalyst. Besides, the current density of the electrocatalyst is only reduced by less than 10% after continuous operation of 10,000 s, indicating excellent durability.

A simple method similar to frying was used to pyrolysis of renewable biomass carbon milk powder in NaCl/Na2CO3 molten salt at 700 °C under the air atmosphere to prepare nitrogen-doped hierarchical porous carbon. This preparation method displayed a simple preparation process with the low requirements for equipment and low-cost raw materials. The prepared optimal nitrogen-doped porous carbon material exhibited a hierarchical porous with macropores, mesopores, and micropores and had a high specific surface area of 1095 m2 g−1. As supercapacitors electrodes, The prepared optimal sample displayed a high specific capacitance of 391.5 F g−1 at 0.5 A g−1 and an excellent rate performance. It also exhibited excellent cycle stability (only loss of 4.4% capacitance) at 4 A g−1 over 5000 cycles. Moreover, In a symmetrical two-electrode system of 1 M Na2SO4, the assembled supercapacitor displayed a high energy density of 33.0 Wh kg−1 at 0.25 A g−1, maintained a high energy density of 22.3 Wh kg−1 at 20 A g−1 and exhibited excellent stability at 5 A g−1 over 5000 cycles. The assembled symmetrical PVA/Na2SO4 solid supercapacitor can successfully light up the light emitting diode (LED). This work provided a sample and low consumption way to prepare N doped hierarchical porous carbon electrode material for high performance supercapacitors.

Inflammation is a complex physiological process that poses a serious threat to people’s health. However, the potential molecular mechanisms of inflammation are still not clear. Moreover, there is lack of effective anti-inflammatory drugs that meet the clinical requirement. Procyanidin A1 (PCA1) is a monomer component isolated from Procyanidin and shows various pharmacological activities. This study further demonstrated the regulatory role of PCA1 on lipopolysaccharide (LPS)-stimulated inflammatory response and oxidative stress in RAW264.7 cells. Our data showed that PCA1 dramatically attenuated the production of pro-inflammatory cytokines such as NO, iNOS, IL-6, and TNF-α in RAW264.7 cells administrated with LPS. PCA1 blocked IκB-α degradation, inhibited IKKα/β and IκBα phosphorylation, and suppressed nuclear translocation of p65 in RAW264.7 cells induced by LPS. PCA1 also suppressed the phosphorylation of JNK1/2, p38, and ERK1/2 in LPS-stimulated RAW264.7 cells. In addition, PCA1 increased the expression of HO-1, reduced the expression of Keap1, and promoted Nrf2 into the nuclear in LPS-stimulated RAW264.7 cells. Cellular thermal shift assay indicated that PCA1 bond to TLR4. Meanwhile, PCA1 inhibited the production of intracellular ROS and alleviated the depletion of mitochondrial membrane potential in vitro. Collectively, our data indicated that PCA1 exhibited a significant anti-inflammatory effect, suggesting that it is a potential agent for the treatment of inflammatory diseases.

Normally-off HEMTs have been fabricated with regrown p-GaN gate and SiNx passivation by Low-pressure chemical vapor deposition (LPCVD) using an AlN pre-layer, featuring a high-temperature passivation-first technique with no incompatibility issues. Through careful surface treatments, high-quality interfaces (regrown p-GaN/AlGaN and SiNx/AlGaN), as well as a stable p-GaN contact, can be achieved, enabling a robust gate operation and improved dynamic performance. The as-fabricated device exhibits a low reverse gate leakage of 2 nA/mm @ VGD = −200 V, a stable forward gate current of $10~\mu \text{A}$ /mm @ VGS = 5 V, a positive threshold voltage of +1.7 V @ ${I}_{\text {DS}} = {10}\,\,\mu \text{A}$ /mm with a high saturation current of 435 mA/mm, a high Ion/Ioff ratio of ${5}\times {10}^{10}$ , and a low Ron,d/Ron,s ratio of 1.5. The as-developed technique with the LPCVD SiNx passivation and p-GaN gate regrowth may provide a way to enhance the performance of p-GaN E-HEMTs.

Aims: Most chemotherapeutic agents exploit apoptotic signaling to trigger cancer cell death, which frequently results in drug resistance. Necroptosis, a nonapoptotic form of regulated cell death, offers an alternative strategy to eradicate apoptosis-resistant cancer cells. We previously reported a natural necroptosis inducer 2-methoxy-6-acetyl-7-methyljuglone (MAM) in A549 lung cancer cells. The current study is designed to investigate the detailed necroptotic signaling and its cytotoxicity on drug-resistant cancer cells. Furthermore, in vivo anticancer effects were also evaluated in nude mice model. Results: MAM directly targets receptor-interacting protein 1 (RIP1) kinase in A549 and H1299 cells, which is responsible for reactive oxygen species (ROS, mainly hydrogen peroxide) generation. A positive feedback loop between calcium (Ca2+) and c-Jun N-terminal kinase (JNK) occurred following ROS generation, leading to lysosomal membrane permeabilization and mitochondrial dysfunction. MAM showed similar cytotoxic potency toward cisplatin-resistant A549 (A549/Cis) cells by inducing necroptosis as confirmed by the protective effect of 7-Cl-O-Nec-1 (Nec-1s) and by the morphological characteristics obtained via transmission electron microscopy. Interestingly, tumor necrosis factor alpha (TNFα) was not involved in this process. Intraperitoneal injection of MAM significantly suppressed tumor growth in A549 tumor xenograft without significant body weight loss and multiorgan toxicities. Innovation and Conclusion: Our findings demonstrate that MAM induces necroptosis in A549 and H1299 lung cancer cells by targeting RIP1 kinase and ROS in a TNFα-independent manner. MAM kills A549/Cis cells with similar potency through induction of necroptosis. MAM shows anticancer effect in animal model. The present study raises the therapeutic possibility and strategy to combat cancer by the induction of necroptosis.

Developing metal-free catalysts possessing high oxygen reduction reaction (ORR) catalytic activity and splendid stability has become one of the most attractive topics in contemporary society. In this study, Triarrhena sacchariflora panicles were hydrothermally treated in KOH aqueous solution. The KOH activation was simplified. After carbonization and nitrogen doping process, the N-doped carbon material was obtained. The material presents a 3D interconnected hierarchical porous structure and an excellent ORR catalytic activity (E1/2 = 0.83 V), along with a high stability (6% loss of current density after 10,000 s) in alkaline media. Compared to the commercial Pt/C catalyst (20 wt%), the catalyst exhibits a similar ORR catalytic activity and much higher stability in alkaline media. Taking this method can not only dispose of the biowaste well but also reduce the amount of KOH compared with the traditional KOH activation method, resulting in low cost and little environmental pollution.

Graphical abstract, Traditional Chinese herbal compound prescription in Xuanfei Baidu Tang (XBT) has obvious effects in the treatment of COVID-19. However, its effective compounds and targets for the treatment of COVID-19 remain unclear. Computer-Aided Drug Design is used to virtually screen out the anti-inflammatory or anti-viral compounds in XBT, and predict the potential targets by Discovery Studio 2020. Then, we searched for COVID-19 targets using Genecards databases and Protein Data Bank (PDB) databases and compared them to identify targets that were common to both. Finally, the target we screened out is: TP53 (Tumor Protein P53). This article also shows that XBT in the treatment of COVID-19 works in a multi-link and overall synergistic manner. Our results will help to design the new drugs for COVID-19.

Pyroptosis, a type of programmed cell death (PCD), is characterized by cell swelling with bubbles, and the release of inflammatory cell cytokines. Cucurbitacin B (CuB), extracted from muskmelon pedicel, is a natural bioactive product that could effectively exert anti-tumor activities in lung cancer. However, the exact molecular mechanisms and the direct targets of CuB in non-small cell lung cancer (NSCLC) remain to be discovered. Here, we firstly found that CuB exerted an anti-tumor effect via pyroptosis in NSCLC cells and NSCLC mice models. Next, based on the molecular docking and cellular thermal shift assay (CETSA), we identified that CuB directly bound to Toll-like receptor 4 (TLR4) to activate the NLRP3 inflammasome, which further caused the separation of N- and C-terminals of Gasdermin D (GSDMD) to execute pyroptosis. Moreover, CuB enhanced the mitochondrial reactive oxygen species (ROS), mitochondrial membrane protein Tom20 accumulation, and cytosolic calcium (Ca2+) release, leading to pyroptosis in NSCLC cells. Silencing of TLR4 inhibited CuB-induced pyroptosis and decreased the level of ROS and Ca2+ in A549 cells. In vivo study showed that CuB treatment suppressed lung tumor growth in mice via pyroptosis without dose-dependent manner, and CuB at 0.75 mg/kg had a better anti-tumor effect compared to the Gefitinib group. Taken together, our findings revealed the mechanisms and targets of CuB triggering pyroptosis in NSCLC, thus supporting the notion of developing CuB as a promising therapeutic agent for NSCLC.

Background Tanshinone I (TI) is a primary component of Salvia miltiorrhiza Bunge (Danshen), which confers a favorable role in a variety of pharmacological activities including cardiovascular protection. However, the exact mechanism of the cardiovascular protection activity of TI remains to be illustrated. In this study, the cardiovascular protective effect and its mechanism of TI were investigated. Methods In this study, tert-butyl hydroperoxide (t-BHP)-stimulated H9c2 cells model was employed to investigate the protective effect in vitro. The cell viability was determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay and lactate dehydrogenase (LDH) kit. The reactive-oxygen-species (ROS) level and mitochondrial membrane potential (MMP) were investigated by the flow cytometry and JC-1 assay, respectively. While in vivo experiment, the cardiovascular protective effect of TI was determined by using myocardial ischemia–reperfusion (MI/R) model including hematoxylin–eosin (H&E) staining assay and determination of superoxide dismutase (SOD) and malondialdehyde (MDA). Tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) release were detected by Enzyme-linked immunosorbent assay (ELISA). Receptor interacting protein kinase 1 (RIP1), receptor interacting protein kinase 3 (RIP3), receptor interacting protein kinase 3 (MLKL), protein kinase B (Akt), Nuclear factor erythroid 2 related factor 2 (Nrf2), Heme oxygenase-1 (HO-1) and NAD(P)H: quinone oxidoreductase-1 (NQO-1) were determined by western blotting. Results Our data demonstrated that TI pretreatment attenuated t-BHP and MI/R injury-induced necroptosis by inhibiting the expression of p-RIP1, p-RIP3, and p-MLKL. TI activated the Akt/Nrf2 pathway to promote the expression of antioxidant-related proteins such as phosphorylation of Akt, nuclear factor erythroid 2 related factor 2 (Nrf2), quinone oxidoreductase-1 (NQO-1) and heme oxygenase-1 (HO-1) expression in t-BHP-stimulated H9c2 cells. TI relieved oxidative stress by mitigating ROS generation and reversing MMP loss. In vivo experiment, TI made electrocardiograph (ECG) recovery better and lessened the degree of myocardial tissue damage. The counts of white blood cell (WBC), neutrophil (Neu), lymphocyte (Lym), and the release of TNF-α and IL-6 were reversed by TI treatment. SOD level was increased, while MDA level was decreased by TI treatment. Conclusion Collectively, our findings indicated that TI exerted cardiovascular protective activities in vitro and in vivo through suppressing RIP1/RIP3/MLKL and activating Akt/Nrf2 signaling pathways, which could be developed into a cardiovascular protective agent.

Acute kidney injury (AKI) is a common critical illness that involves multiple systems and multiple organs with a rapid decline in kidney function over short period. It has a high mortality rate and presents a great treatment challenge for physicians. Oleuropein, the main active constituent of Ilex pubescens Hook. et Arn. var. kwangsiensis Hand.-Mazz. displays significant anti-inflammatory activity, although oleuropein’s therapeutic effect and mechanism of action in AKI remain to be elucidated. The present study aimed to further clarify the mechanism by which oleuropein exerts effects on inflammation in vitro and in vivo. In vitro, the inflammatory effect and mechanism were investigated through ELISA, Western blotting, the thermal shift assay, co-immunoprecipitation, and immunofluorescence staining. Lipopolysaccharide (LPS) induced acute kidney injury was employed in an animal model to investigate oleuropein’s therapeutic effect on AKI and mechanism in vivo. The underlying mechanisms were investigated by Western blot analysis of kidney tissue. In LPS-stimulated macrophages, our data demonstrated that oleuropein significantly reduced the expression of inflammatory mediators like NO, IL-6, TNF-α, iNOS, and COX-2. Moreover, oleuropein inhibited NF-κB/p65 translocation, and had a negative regulatory effect on key proteins in the NF-κB and MAPK pathways. In addition, the thermal shift and co-immunoprecipitation assays revealed that oleuropein played an essential role in binding to the active sites of TLR4, as well as inhibiting TLR4 dimerization and suppressing the binding of TLR4 to MyD88. Oleuropein markedly alleviated LPS induced acute kidney injury, decreased serum creatinine and blood urea nitrogen (BUN) levels and proinflammatory cytokines. More importantly, the TLR4-MyD88-NF-κB/MAPK pathways were confirmed to play an important role in the oleuropein treatment of AKI. In this study, oleuropein exhibited excellent anti-inflammatory effects by regulating TLR4-MyD88-NF-κB/MAPK axis in vitro and in vivo, suggesting oleuropein as a candidate molecule for treating AKI.

This paper documents the first attempt to apply the differential game theory for investigating some properties of a low-carbon supply chain coordination by employing the cost-sharing mechanism. The manufacturer is the brand owner and the corresponding low-carbon goodwill is increased with respect to the reference emission reduction effect and consumption promotion. Three dynamic games are well-studied by considering the multiple effects of price and non-price factors on the market. It can be observed that the manufacturer always prefers to employ the coordination mechanism. When the manufacturer opts for a cost-sharing program, the manufacturer and retailer under Model-D are always economically better off, and therefore a cost-sharing program is always profit-Pareto-improving.

Nonlinear integrated optics leveraging platforms with high nonlinear figure of merit offer energy efficient optical signal processing capabilities. Over the last five years, CMOS-compatible ultra-silico-rich nitride (USRN) has emerged as a promising platform on which to implement various nonlinear optics functions. Bandgap engineered to maximize the Kerr nonlinearity while maintaining two-photon absorption free behavior at telecommunications wavelengths, USRN possesses a nonlinear refractive index that is 100× larger than that in stoichiometric silicon nitride. In this article, we review the recent progress made in USRN-based nonlinear integrated optics devices. The impacts it has made spanning from high gain optical parametric amplification and observations of soliton effects in photonic waveguides and photonic crystal waveguide structures will be discussed. Finally, we assess the future outlook of CMOS-compatible USRN-based nonlinear optics.

Ilicis Rotundae Cortex (IRC) consists of the bark of Ilex rotunda Thunb, and its chemical constituents mainly include flavonoid glycosides, phenols, and triterpenoid saponins. In this study, a preliminary analysis was performed to identify and obtain the chemical components from IRC to better control the quality of the medicinal materials and provide a chemical basis for the study of the efficacy of the active components. Simple and efficient sample pretreatment and ultrasonic-assisted extraction methods were used to analyze the mass spectrum fragments and fracture modes in the anion mode by UPLC-Q-TOF-MS/MS. Using a two-step strategy, the neutral loss, diagnostic ions, and characteristic fragments were studied to screen diverse skeletons and substitutions, and the possible compounds were identified by comparison with databases. The representative compounds were compared with the standard, and the mass spectrogram was found to match perfectly. Thus, our findings reveal that this method is feasible and reliable and can be used to analyze the chemical components of IRC. We identified 105 compounds, including 22 triterpenoid saponins, 15 chlorogenic acids, 33 phenylpropanoids and phenylpropanosides, 3 iridoids, 1 flavonoid, 10 lignans, 12 glycosides, and 9 other compounds. This method lays the foundation for further elucidating the pharmacodynamics of IRC and provides a practical method for the identification of IRC.

Acute lung injury (ALI) is an acute inflammatory process in the lung parenchyma. Anemoside B4 (B4) was isolated from Pulsatilla, a plant-based drug against inflammation and commonly applied in traditional Chinese medicine. However, the anti-inflammatory effect and the mechanisms of B4 are not clear. In this study, we explored the potential mechanisms and anti-inflammatory activity of B4 both in vitro and in vivo. The results indicated that B4 suppressed the expression of iNOS, COX-2, NLRP3, caspase-1, and IL-1β. The ELISA assay results showed that B4 significantly restrained the release of inflammatory cytokines like TNF-α, IL-6, and IL-1β in macrophage cells. In addition, B4 rescued mitochondrial membrane potential (MMP) loss in (lipopolysaccharide) LPS plus ATP stimulated macrophage cells. Co-IP and molecular docking results illustrated that B4 disrupted the dimerization of TLR4. For in vivo results, B4 exhibited a protective effect on LPS and bleomycin- (BLM-) induced ALI in mice through suppressing the lesions of lung tissues, the release of inflammatory cytokines, and the levels of white blood cells, neutrophils, and lymphoid cells in the blood. Collectively, B4 has a protective effect on ALI via blocking TLR4 dimerization and NLRP3 inflammasome activation, suggesting that B4 is a potential agent for the treatment of ALI.

Background: Flavonolignans like silybin, hydnocarpin, and siliandrin are a group of natural compounds combining the structural moieties of flavonoid and phenylpropanoid (lignan). Hydnocarpin and silandrin have been less explored because of their trace occurrence in nature. Objective: The present study aimed at chemical conversion of silybin to hydnocarpin and siliandrin. Another objective was to synthesize a series of amide derivatives and biologically evaluate them with regard to their anti-cancer effects. Methods: In order to selectively convert silybin to 23-iodo silybin, 23-iodo hydnocarpin D and 23- iodo isosilandrin, the ratio of Ph3P, imidazole and molecular iodine was meticulously adjusted. These three iodide compounds were converted into amide compounds by chemical transformation. MTT method was applied to evaluate their anti-cancer potency. The binding affinity to related proteins was calculated by molecular docking. Results: A total of 45 new amido-derivatives were synthesized and structurally characterized by NMR and HRMS. Some of them showed moderate to good antiproliferative potency against cancer cells. The activity of compound 10j was further testified by colony formation assay and molecular docking. Conclusion: The synthesis of 23-iodo silybin, 23-iodo hydnocarpin D and 23-iodo isosilandrin from silybin was successfully accomplished by one simple iodination reaction. Some of the amide derivatives of sylibin/hydnocarpin D /silandrin exhibited a remarkable inhibitory effect of proliferation on cancer cells compared to silybin. These results would pave the way for further investigation on the derivatives of flavonolignans for the treatment of cancer.

As a biosynthetic precursor of the antimalarial drug artemisinin, artemisinic acid (AA) is abundant in Artemisia annua L. with a content of 8-10-fold higher than artemisinin, but less effective. In this study, the biotransformation of AA was carried out with an endophytic fungus Penicillium oxalicum B4 to extend its utility. After 10-day-culture of the endophyte with AA at 2 mg/mL, eight biotransformation metabolites were isolated from the culture broth, including five undescribed metabolites, namely 3α,14-dihydroxyartemisinic acid, 14-hydroxy-3-oxo-artemisinic acid, 15-hydroxy-3-oxo-artemisinic acid, 12, 15-artemisindioic acid and 1,2,3,6-tetradehydro-12, 15-artemisindioic acid. The fungal enzymes possess the selective capacity to hydroxylate, carbonylate and ketonize the allyl group of AA. The major biotransformation metabolite was the hydroxylated product 3-α-hydroxyartemisinic acid (33.3%) in the cultures of early stage (day 1–6), whereas most of the other biotransformation products were synthesized in the later stage (day 8–10). Compared with AA, some metabolites (3α,14-dihydroxyartemisinic acid, 15-hydroxy-3-oxo-artemisinic acid and 1,2,3,6-tetradehydro-12, 15-artemisindioic acid) possessed stronger cytotoxic activity to the human colon carcinoma cell line (LS174T) and promyelocytic leukemia cell line (HL-60). The metabolites 12, 15-artemisindioic acid and 3-α-hydroxyartemisinic acid exhibited significant inhibitory activity to the lipopolysaccharide-induced nitrite production of RAW 264.7 cells at 10.00 μM and 2.50 μM, respectively. The results demonstrated the potential of fungal endophytes on biotransforming AA to its bioactive derivatives.

Coriander, Coriandrum Sativum L., is one of the commonest food and medicinal plants in many countries, but its chemical ingredients and pivotal role in anti-inflammatory activity have not been fully explored. The present study aimed to identify new compounds in the fruits of coriander and explore their anti-inflammatory activity. The compounds were isolated by chromatographic seperations and identified using spectroscopic and spectrometric methods. RAW264.7 macrophage cells were used to detect the anti-inflammatory activity of the compounds via Griess assay, western blotting, ELISA, and flow cytometry methods. The study resulted in the discovery of four new compounds, which were identified as: 4α-(furo[2,3-d]pyrimidin-6′-ylmethyl)-9α-propylnonolactone (1), 4-(formyloxy)-4-(6′-methylcyclohex-1-en-1-yl)butanoate(2), (7α,8α)-3α-hydroxyl-12,13α-dimethyl-5(6)-en-bicyclo[5,3,0]caprolactone (3), 7-methoxy-4-methyl-5,6-dihydro-7H-(2-hydroxypropan-2-yl)furo[2,3-f] coumarin (4). Compound 3 showed the highest anti-inflammatory activity with IC50 of 6.25 μM for an inhibitory effect on nitrite oxide (NO) level. In addition, compound 3 decreased the lipopolysaccharides-stimulated generations of ROS and the inflammatory cytokines (IL-6 and TNF-α). Mechanism exploration indicated that compound 3 suppressed inflammatory mediators’ expression, like iNOS and COX-2. Furthermore, the NF-κB and MAPK pathways were involved in the anti-inflammatory process of compound 3.

Procyandin A2 (PCA2) is a polyphenolic compound which is isolated from grape seeds. It has been reported that PCA2 exhibits antioxidative and anti-inflammatory effects, but its molecular mechanism is still poorly understood. This study tests the hypothesis that PCA2 suppresses lipopolysaccharide (LPS)-induced inflammation and oxidative stress through targeting the nuclear factor-κB (NF-κB), mitogen-activated protein kinase (MAPK), and NF-E2-related factor 2 (Nrf2) pathways in RAW264.7 cells. PCA2 (20, 40, 80 μM) exhibited no significant cytotoxicity in RAW264.7 cells and showed an inhibitory effect on an LPS-induced nitrite level. Pro-inflammatory cytokines like tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), prostaglandin E2 (PGE2), nitric oxide (NO), and reactive oxygen species (ROS) were suppressed by PCA2 with a concentration range of 0-80 μM. The mRNA levels of TNF-α and IL-6 were inhibited by PCA2 (80 μM). The hallmark-protein expression of the NF-κB (p-IKKα/β, p-IκBα, and p-p65) and MAPK (p-p38, p-JNK, and p-ERK) pathways were decreased by PCA2 in LPS-stimulated RAW264.7 cells. In addition, immunofluorescence results indicated that PCA2 (80 μM) promoted the translocation of NF-κB/p65 from the cytoplasm into the nucleus. PCA2 upregulated the expressions of Nrf2 and HO-1 and downregulated the expression of Keap-1. Simultaneously, PCA2 (80 μM) reversed LPS-induced Nrf2 translocation from the nucleus into the cytoplasm. Collectively, PCA2 protect cells against the damage from inflammation and oxidative injury, which suggest a potential therapeutic strategy for inflammatory and oxidative stress through targeting NF-κB, MAPK, and Nrf2 pathways in RAW264.7 cells.

Ethnopharmacological relevance According to the theory of traditional Chinese medicine, the main pathogenesis of severe hand, foot and mouth disease (HFMD) is that the heat and wet poisons are deeply trapped in the viscera, which causes the deficiency of Qi and Yin in the patient's body. Ginsenoside Rb1 (Rb1) is the most abundant triterpenoid saponin in Panax quinquefolius L., which has the function of Qi-invigorating and Yin-nourishing. Enterovirus 71 (EV71) is one of the causative pathogens of HFMD, especially the form associated with some lethal complications. Therefore, the therapeutic effect of Rb1 on this disease caused by EV71 infection is worth exploring. Aim of the study We explored the effective antiviral activities of Rb1 against EV71 in vitro and in vivo and investigated its preliminary antiviral mechanisms. Material and methods EV71-infected two-day-old suckling mice model was employed to detect the antiviral effects of Rb1 in vivo. To detect the antiviral effects of Rb1 in vitro, cytopathic effect (CPE) reduction assay was performed in EV71-infected Rhabdomyosarcoma (RD) cells. Interferon (IFN)-β interference experiment was employed to detect the antiviral mechanism of Rb1. Results In this paper, we first found that Rb1 exhibited strong antiviral activities in EV71-infected suckling mice when compared to those of ribavirin. Administration of Rb1 reduced the CPE of EV71-infected RD cells in a dose-dependent manner. Moreover, EV71-induced viral protein-1 (VP-1) expression was significantly reduced by Rb1 administration in vitro and in vivo. Furthermore, Rb1 treatment could induce high cellular and humoral immune responses in vivo. Meanwhile, Rb1 contributed to the enhanced Type I IFN responses and IFN-β knockdown reversed the antiviral activity of Rb1 in vitro. Conclusion In summary, our findings suggest that Rb1 is an immune-stimulatory agent and provide an insight into therapeutic potentials of Rb1 for the treatment of EV71 infection.

Bi-layer organization of dissimilar two-dimensional (2D) nanosheets has attracted great attention because of their unique physicochemical properties, fascinating functions, and potential applications in portable and wearable electronic devices. In this study we showed that several types of 2D nanomaterials are capable of being exfoliated by graphene oxide (GO). Under optimal conditions, a concentration up to 1.2 mg mL−1 and an exfoliation yield as high as 24% were achieved for MoS2. The exfoliated transition metal dichalcogenide (TMD) nanosheets anchored in situ onto the surfaces of GO sheets, and formed heterojunction nanosheets due to strong interactions between GO and TMDs based on theoretical calculations. After chemically reducing GO, these heterojunction nanosheets could also be patterned onto biocompatible films of cellulose nanofibrils to construct a flexible planar supercapacitor, with exceptional flexibility, biodegradability, flame retardancy and remarkable energy density (52.7 mW h cm−2). More importantly, the capacitor exhibited intriguing photo-sensitivity, enabling it to be a promising photo-assisted flexible photodetector. Thus the preparation of heterojunction nanosheets not only sheds light on bi-layer organization of 2D nanomaterials, but also promises future applications in wearable, photo-sensitive and flame-retardant electronic devices.

Context: Lipopolysaccharide (LPS) is often used to induce immunoinflammatory reactions. TLR4/NFκB and NLRP3 signalling are major factors for inflammation. Dexamethasone (DXM) has an anti-immunoinflammatory effect. Objective: To investigate the inflammatory reaction in pathological changes of organs and the expression of inflammatory signalling during LPS infection. Materials and methods: ICR mice were divided into control group (n = 9), LPS group (n = 15) and LPS + DXM group (n = 14). LPS (10 mg/kg) was injected intravenously in LPS group and LPS + DXM group, normal saline was injected to the control group; DXM (0.5 mg/kg) was given by intragastric administration. 12 h after LPS, the blood was collected and the organs were isolated for biochemical analysis, protein expression, and morphological examination. Results: The results showed that BUN, Cre, ALT, AST in the LPS group increased distinctly by 81.42, 67.84, 40.53 and 36.05%, respectively, and CK, ALP, TP and ALB decreased by 71.37, 60.6, 12.57 and 19.73%, respectively, compared with the control group. In the morphologic observation, local necrosis in the liver, arterial vasodilation in the heart and kidney, alveolar secretions and pulmonary interstitial in the lungs, and mucosal shedding in the small and large intestines, the expression of TLR4-NFκB signalling were up-regulated distinctly whereas NLRP3 signalling was less broadly affected. DXM can decrease BUN and Cre, downregulate the expression of TLR4-NFκB signalling, but has no effect on the organ damage based on morphology. Conclusion: Acute injuries induced by LPS are extensive. The inflammatory damage in small and large intestines, liver and kidney was more severe than other organs. TLR4-NFκB signalling was the major response to LPS stress.

Direct bandgap III-V semiconductor lasers grown on silicon (Si) are highly desired for monolithic integration with Si photonics. Fabrication of semiconductor lasers with a Fabry–Pérot cavity usually includes facet cleavage, however, that is not compatible with on-chip photonic integration. Etching as an alternative approach holds a great advantage in preparing cavity mirrors with no need of breaking wafer into bars. However, gallium nitride (GaN) sidewalls prepared by dry etching often have a large roughness and etching damages, which would cause mirror loss due to optical scattering and carrier injection loss because of surface non-radiative recombination. A wet chemical polishing process of GaN sidewall facets formed by dry etching was studied in detail to remove the etching damages and smooth the vertical sidewalls. The wet chemical polishing technique combined with dry etching was successfully applied to the on-wafer fabrication of cavity mirrors, which enabled the realization of room temperature electrically injected InGaN-based laser diodes grown on Si.

Alzheimer's disease (AD) is a neurodegenerative disorder. Substrate-specific Acetylcholinesterase (AChE) plays a vital role in the AD treatment. Flavonoids with AChE inhibitory activities and low toxicity are used to developing new anti-AD agents. In this study, the best 3D QSAR pharmacophore model Hypo1 was generated by HypoGen program in Discovery Studio2016 based on the training set of flavonoids. We performed a virtual screening from Traditional Chinese Medicine (TCM), Druglike and MiniMaybridge databases using Hypo1. From docking analyses, we got the top 10 AChE inhibitors which were further evaluated by 8 different scoring functions. De Novo Evolution designed the top 10 derivatives, and three potential AChE inhibitor candidates were obtained eventually.

The aim of this study was to determine the effect of Astragalus polysaccharide (APS) on the expression of vascular endothelial growth factor (VEGF) and epidermal growth factor receptor (EGFR) in mice with Lewis transplantable lung cancer. It was an experimental study carried out from June 2017 to January 2018 at the College of Pharmacy, Guangxi University of Chinese Medicine, Nanning, China. Forty male SPF-level C57BL/6J mice were selected and inoculated with Lewis lung cancer cell suspension in the right axilla of the mice to establish a lung-cancer mouse model. The mice were randomly divided into a model control group and APS groups with high, middle and low dosages, respectively, 10 in each group. After inoculation of Lewis tumor cell suspension for 2 days, mice in the model control group were injected intraperitoneally with 50 mL/kg of 0.9% sodium chloride solution, whereas, mice in the APS groups with high, middle and low dosages were intraperitoneally injected with APS at 100, 50, 25 mg/kg, respectively. The research results showed that APS can effectively inhibit the growth and metastasis of Lewis lung cancer in mice, improve immune organ function, inhibit the protein expression of VEGF and EGFR in tumor tissues, and have a concentration-effect relationships.
