The potential of capillary electrophoresis (CE) with ultraviolet (UV)-excited fluorescence detection for sensitive chiral analysis of amino acids (AAs) was investigated. dl-AAs were derivatized with 9-fluorenylmethoxycarbonyl chloride (FMOC)-Cl to allow their fluorescence detection and enhance enantioseparation. Fluorescence detection was achieved employing optical fibers, leading UV excitation light (< 300 nm) from a Xe-Hg lamp to the capillary window, and fluorescence emission to a spectrograph equipped with a charge-coupled device (CCD). Signal averaging over time and emission wavelength intervals was carried out to improve the signal-to-noise ratio of the FMOC-AAs. A background electrolyte (BGE) of 40 mM sodium tetraborate (pH 9.5), containing 15% isopropanol (v/v), 30 mM sodium dodecyl sulfate (SDS), and 30 mM β-cyclodextrin (β-CD), was found optimal for AA chemo- and enantioseparation. Enantioresolutions of 1.0 or higher were achieved for 16 proteinogenic dl-AAs. Limits of detection (LODs) were in the 10–100-nM range (injected concentration) for the d-AA enantiomers, except for FMOC-d-tryptophan (536 nM) which showed intramolecular fluorescence quenching. Linearity (R2 > 0.997) and repeatability for peak height (relative standard deviations (RSDs) < 7.0%; n = 5) and electrophoretic mobility (RSDs < 0.6%; n = 5) of individual AA enantiomers were established for chiral analysis of dl-AA mixtures. The applicability of the method was investigated by the analysis of cerebrospinal fluid (CSF). Next to l-AAs, endogenous levels of d-glutamine and d-aspartic acid could be measured in CSF revealing enantiomeric ratios of 0.35 and 19.6%, respectively. This indicates the method’s potential for the analysis of low concentrations of d-AAs in presence of abundant l-AAs.

Developing tools for the study of protein carbohydrate interactions is an important goal in glycobiology. Cholera toxin inhibition is an interesting target in this context, as its inhibition may help the fight against cholera. For the study of novel ligands an affinity capillary electrophoresis (ACE) method was optimised and applied. The method uses unlabeled cholera toxin B-subunit (CTB) and unlabeled carbohydrate ligands based on ganglioside GM1-oligosaccharides (GM1os). In an optimized method at pH 4, adsorption of the protein to the capillary walls was prevented by a polybrene-dextran sulfate-polybrene (PB-DS-PB) coating. Different concentrations of the ligands were added to the background electrolyte. CTB binding was observed by a mobility shift that could be used for dissociation constant (Kd) determination. The Kd values of two GM1 derivatives differed by close to an order of magnitude (600 ± 20 nM and 90 ± 50 nM) which was in good agreement with the differences in their reported nanomolar IC50 values of an ELISA-type assay. The conserved RNA-binding protein, Hfq, has multiple regulatory roles within the prokaryotic cell, including promoting stable duplex formation between small RNAs and mRNAs, and thus hfq deletion mutants have pleiotropic phenotypes. Previous proteome and transcriptome studies of Neisseria meningitidis have generated limited insight into differential gene expression due to Hfq loss. In this study, reversed-phase liquid chromatography combined with data-independent alternate scanning mass spectrometry (LC-MSE) was utilized for rapid high-resolution quantitative proteomic analysis to further elucidate the differentially expressed proteome of a meningococcal hfq deletion mutant. Whole-cell lysates of N. meningitidis serogroup B H44/76 wild-type (wt) and H44/76 Delta hfq (Delta hfq) grown in liquid growth medium were subjected to tryptic digestion. High carbon dioxide in reservoirs limits successful exploration in many petroliferous basins, particularly in Southeast Asia. High reservoir CO2 in the offshore Malay Basin represents a significant exploration challenge. Some fields contain >80% CO2, which makes them unattractive targets for development. Various hypotheses on the origin of CO2 have been proposed but remain controversial. This paper shows that geochemistry and advanced petroleum system modeling help to resolve the origins of reservoir CO2 and allow quantitative estimates of CO2 in prospective reservoir targets prior to drilling. A novel workflow estimates the CO2 content in reservoirs based on knowledge of the chemical mechanisms for the origin of the CO2 and numerical simulation of geologic burial history. Heat flow, deposition of overburden rock, and the kinetics of specific reaction mechanisms control the timing of CO2 generation and the relative contributions of CO2 from different sources. In this study, stable carbon isotope ratios of CO2 and methane (δ13CCO2 and δ13CCH4, ‰) were used to identify the source of the CO2 in Malay Basin gas samples. For example, Figure 3 shows δ13CCO2 and δ13CCH4 for samples from various depths in the nearby field. The isotope data indicate that the samples contain mixed CO2 derived by different mechanisms from two sources. Partial least squares (PLS) regression of δ13CCO2 and δ13CCH4 and depth for 61 samples from the nearby field, where %CO2 was set as the dependent variable, resulted in a systematic correlation between predicted and measured %CO2. Alternate least squares (ALS) confirms that the data can be explained by mixing of gases from two endmembers: (1) shallower samples show lower %CO2 that is isotopically depleted in δ13CCH4 and δ13CCO2, and (2) deeper samples show higher %CO2 that is isotopically enriched in δ13CCH4 and δ13CCO2. The relative proportion of each endmember in the mixture can be calculated for each gas. Examples of near endmember gases in the nearby field (Figure 3) are: (1) shallow thermogenic CO2 derived by cracking of kerogen, e.g., 1681 m, 5% CO2, δ13CCH4 = -60‰, δ13CCO2 = -13‰, (100:0 mix); and (2) deep CO2 from carbonate decomposition, e.g., 2918 m, 74% CO2, δ13CCH4 = -32‰, δ13CCO2 = -3‰ (15:85 mix). These results are consistent with the general observation that tested Miocene traps in the Malay Basin and show a general trend of higher concentrations of CO2 in the deeper traps that are nearer carbonate basement. Biogenic CO2 may represent a third endmember in other parts of the basin.

d-Amino acids (AAs) are increasingly being recognized as essential molecules in biological systems. Enantioselective analysis of proteinogenic AAs in biological samples was accomplished by CE-MS employing β-CD as chiral selector and ESI via sheath-liquid (SL) interfacing. Prior to analysis, AAs were fully derivatized with FMOC, improving AA-enantiomer separation and ESI efficiency. In order to optimize the separation and MS detection of FMOC-AAs, the effects of type and concentration of CD in the BGE, the composition of the SL, and MS-interfacing parameters were evaluated. Using a BGE of 10 mM β-CD in 50 mM ammonium bicarbonate (pH 8) containing 15% v/v isopropanol, a SL of isopropanol-water-1 M ammonium bicarbonate (50:50:1, v/v/v) at a flow rate of 3 μL/min, and a nebulizer gas pressure of 2 psi, 15 proteinogenic AAs could be detected with enantioresolutions up to 3.5 and detection limits down to 0.9 μM (equivalent to less than 3 pg AA injected). The selectivity of the method was demonstrated by the analysis of spiked cerebrospinal fluid, allowing specific detection of d-AAs. Repeatability and linearity obtained for cerebrospinal fluid were similar to standard solutions, with peak area and migration-time RSDs (n = 5) below 16.2 and 1.6%, respectively, and a linear response (R(2) ≥ 0.977) in the 3-90 μM range.

Chiral analysis of dl-amino acids was achieved by micellar electrokinetic chromatography coupled with UV-excited fluorescence detection. The fluorescent reagent (+)-1-(9-fluorenyl)ethyl chloroformate was employed as chiral amino acid derivatizing agent and sodium dodecyl sulfate served as pseudo-stationary phase for separating the formed amino acid diastereomers. Sensitive analysis of (+)-1-(9-fluorenyl)ethyl chloroformate-amino acids was achieved applying a xenon-mercury lamp for ultraviolet excitation, and a spectrograph and charge-coupled device for wavelength-resolved emission detection. Applying signal integration over a 30 nm emission wavelength interval, signal-to-noise ratios for derivatized amino acids were up to 23 times higher as obtained using a standard photomultiplier for detection. The background electrolyte composition (electrolyte, pH, sodium dodecyl sulfate concentration, and organic solvent) was studied in order to attain optimal chemo- and enantioseparation. Enantioseparation of 12 proteinogenic dl-amino acids was achieved with chiral resolutions between 1.2 and 7.9, and detection limits for most derivatized amino acids in the 13–60 nM range (injected concentration). Linearity (coefficients of determination > 0.985) and peak-area and migration-time repeatabilities (relative standard deviations lower than 2.6 and 1.9%, respectively) were satisfactory. The employed fluorescence detection system provided up to 100-times better signal-to-noise ratios for (+)-1-(9-fluorenyl)ethyl chloroformate-amino acids than ultraviolet absorbance detection, showing good potential for d-amino acid analysis.

Surface plasmon resonance (SPR) currently is the major platform to study protein–protein interactions, but it lacks the selectivity to distinguish between binding components within one sample. Capillary electrophoresis (CE) can provide efficient separation of intact proteins under near-physiological conditions. We have hyphenated CE with SPR to achieve affinity assessment of mixture components. A microfluidic flow cell allowing straightforward coupling of CE and SPR was developed. Initial testing with non-interacting dyes showed good performance using a flow-cell channel volume of 100 nL until the detection point. Appropriate closing of the CE electric circuit was achieved using the SPR gold-sensor as grounding electrode. Division of the (bio)sensor into an electrode part (providing grounding) and a detection part (bearing the affinity surface) was crucial to avoid disturbance of the SPR signal by the CE voltage. This approach permitted CE separation and binding assessment for separation voltages up to 30 kV. Human serum albumin (HSA) or aprotinin were immobilized on carboxymethyldextran hydrogel-coated gold sensors and target proteins (anti-HSA, and trypsin and α-chymotrypsin, respectively) were analyzed. Efficient CE separation of the intact protein analytes was accomplished under native conditions by employing neutral and positively-charged capillary coatings. Selective binding of separated proteins to the target surface could be monitored by SPR down to 2 ng of injected protein. Regeneration of the biosensor surface was achieved by an on-line rising, allowing repeatable CE-SPR analyses of proteins with RSDs below 1% and 5% for migration time and signal intensity, respectively.

The on-line coupling of micellar electrokinetic chromatography and mass spectrometry (MEKC-MS) is often hampered by incompatibility problems leading to reduced separation performance and unfavorable limits of detection (LODs). Here we propose a new selective and highly sensitive MEKC-MS/MS method employing a sheathless porous-tip interface in combination with a micellar phase comprised of semi-volatile surfactant molecules. Carbamate pesticides (CRBs) were selected as representative model compounds being neutral toxic pollutants potentially present at trace levels in environmental water samples. A background electrolyte of 75 mM perfluorooctanoic acid adjusted to pH 9.0 with ammonium hydroxide allowed efficient separation of 15 CRBs and appeared fully compatible with electrospray ionization (ESI)-MS. Interfacing parameters, such as the distance between the capillary tip and mass-spectrometer inlet, ESI voltage, and dry gas temperature and flow were optimized in order to attain good spray stability and high analyte signal-to-noise ratios. For CRBs the LODs ranged from 0.2 to 3.9 ng L −1 (13 nL injected, i.e., 2% of capillary volume), representing an improvement for certain CRBs of more than 300-fold when compared with conventional sheath-liquid interfacing. Good linearity (R 2 > 0.99) and satisfactory reproducibility were obtained for all CRBs with interday RSD values for peak area and migration time of 4.0–11.3% and below 1.5%, respectively. Analysis of spiked mineral water showed that the new MEKC-MS/MS method allows selective and quantitative determination of CRB concentrations below the maximum residue limit of 100 ng L −1 without the need for sample preconcentration.

Capillary electrophoresis-mass spectrometry (CE-MS) is increasingly used for the targeted and untargeted analysis of metabolites in biological samples. CE-MS is particularly useful for the profiling of highly polar metabolites without the need for derivatization and/or extensive sample preparation. This overview covers reported studies in which CE or CE-MS was used with direct MS, gas chromatography (GC)-MS, liquid chromatography (LC)-MS, and/or nuclear magnetic resonance (NMR) spectroscopy for the analysis of metabolites. We include an extensive overview of comparative metabolic profiling studies involving multiple analytical techniques, including CE. Tables set out the particular conditions and the characteristics of each study. Furthermore, we discuss and compare the results of these studies using selected examples. Based on the outcomes of the comparative studies, we conclude that CE is highly complementary to the other analytical technologies and can add essential information on the metabolic profile of biological samples.

Hydrophilic interaction chromatography-mass spectrometry (HILIC-MS) was used for anionic metabolic profiling of urine from antibiotic-treated rats to study microbial-host co-metabolism. Rats were treated with the antibiotics penicillin G and streptomycin sulfate for four or eight days and compared to a control group. Urine samples were collected at day zero, four and eight, and analyzed by HILIC-MS. Multivariate data analysis was applied to the urinary metabolic profiles to identify biochemical variation between the treatment groups. Principal component analysis found a clear distinction between those animals receiving antibiotics and the control animals, with twenty-nine discriminatory compounds of which twenty were down-regulated and nine up-regulated upon treatment. In the treatment group receiving antibiotics for four days, a recovery effect was observed for seven compounds after cessation of antibiotic administration. Thirteen discriminatory compounds could be putatively identified based on their accurate mass, including aconitic acid, benzenediol sulfate, ferulic acid sulfate, hippuric acid, indoxyl sulfate, penicillin G, phenol and vanillin 4-sulfate. The rat urine samples had previously been analyzed by capillary electrophoresis (CE) with MS detection and proton nuclear magnetic resonance (1H NMR) spectroscopy. Using CE-MS and 1H NMR spectroscopy seventeen and twenty-five discriminatory compounds were found, respectively. Both hippuric acid and indoxyl sulfate were detected across all three platforms. Additionally, eight compounds were observed with both HILIC-MS and CE-MS. Overall, HILIC-MS appears to be highly complementary to CE-MS and 1H NMR spectroscopy, identifying additional compounds that discriminate the urine samples from antibiotic-treated and control rats.

Capillary electrophoresis (CE) played an important role in developments in the life sciences. The technique is nowadays used for the analysis of both large and small molecules in applications where it performs better than or is complementary to liquid chromatographic techniques. In this review, principles of different electromigration techniques, especially capillary isoelectric focusing (CIEF), capillary gel (CGE) and capillary zone electrophoresis (CZE), are described and recent developments in instrumentation, with an emphasis on mass spectrometry (MS) coupling and microchip CE, are discussed. The role of CE in the life sciences is shown with applications in which it had a high impact over the past few decades. In this context, current practice for the characterization of biopharmaceuticals (therapeutic proteins) is shown with CIEF, CGE and CZE using different detection techniques, including MS. Subsequently, the application of CGE and CZE, in combination with laser induced fluorescence detection and CZE-MS are demonstrated for the analysis of protein-released glycans in the characterization of biopharmaceuticals and glycan biomarker discovery in biological samples. Special attention is paid to developments in capillary coatings and derivatization strategies for glycans. Finally, routine CE analysis in clinical chemistry and latest developments in metabolomics approaches for the profiling of small molecules in biological samples are discussed. The large number of CE applications published for these topics in recent years clearly demonstrates the established role of CE in life sciences.

SummaryBlood dilution after transfusion fluids leads to diminished coagulant activity monitored by rotational thromboelastometry, assessing elastic fibrin clot formation, or by thrombin generation testing. We aimed to determine the contributions of blood cells (platelets, red blood cells) and plasma factors (fibrinogen, prothrombin complex concentrate) to fibrin clot formation under conditions of haemodilution in vitro or in vivo. Whole blood or plasma diluted in vitro was supplemented with platelets, red cells, fibrinogen or prothrombin complex concentrate (PCC). Thromboelastometry was measured in whole blood as well as plasma; thrombin generation was determined in parallel. Similar tests were performed with blood from 48 patients, obtained before and after massive fluid infusion during cardiothoracic surgery. Addition of platelets or fibrinogen, in additive and independent ways, reversed the impaired fibrin clot formation (thromboelastometry) in diluted whole blood. In contrast, supplementation of red blood cells or prothrombin complex concentrate was ineffective. Platelets and fibrinogen independently restored clot formation in diluted plasma, resulting in thromboelastometry curves approaching those in whole blood. In whole blood from patients undergoing dilution during surgery, elastic clot formation was determined by both the platelet count and the fibrinogen level. Thrombin generation in diluted (patient) plasma was not changed by fibrinogen, but improved markedly by prothrombin complex concentrate. In conclusion, in dilutional coagulopathy, platelets and fibrinogen, but not red blood cells or vitamin K-dependent coagulation factors, independently determine thromboelastometry parameters measured in whole blood and plasma. Clinical decisions for transfusion based on thromboelastometry should take into account the platelet concentration.

A new MEKC-ESI-MS method for the analysis of amino acids (AAs) in human urine was developed employing ammonium perfluorooctanoate (APFO) as volatile surfactant. The influence of APFO on the MS signal of AAs was evaluated by infusion experiments, which showed that APFO hardly affects analyte responses and presents significantly less ion suppression than equal concentrations of ammonium acetate. In order to obtain efficient separation of AAs, MEKC parameters such as the pH and APFO concentration of the BGE, were optimized. Optimum AA resolution, including baseline separation of leucine and isoleucine, was obtained using 150 mM APFO (pH 9.0) as BGE, representing a considerable selectivity improvement over CE using 50 mM ammonium acetate (pH 9.0). Optimization of CE-MS parameters, such as sheath liquid composition and flow rate, and ESI and MS settings, led to LODs ranging from 9 to 26 ng/mL for the 20 tested AAs, which is highly favorable for an MEKC-MS method. This review treats the coupling of capillary electrophoresis (CE) with fluorescence detection (Flu) for the analysis of natively fluorescent biomolecular and pharmaceutical compounds. CE-Flu combines the excellent separation efficiency of CE with the high selectivity and sensitivity of Flu. In CE-Flu, an appropriate design of the fluorescence detection cell is very important in order to achieve efficient analyte excitation in and emission light collection from the small cylindrically-shaped detection volume. Therefore, due attention is paid to the various optical detection designs used for CE-Flu, including the applied excitation sources and emission light detectors. Special attention is devoted to wavelength-resolved Flu and to sensitivity issues. Furthermore, he specific requirements for fluorescence detection in microfluidic systems (i.e. chip-based electrophoresis) are discussed. Subsequently, an overview of described applications of CE-Flu for the analysis of natively fluorescent biomolecules and drugs is presented in extensive tables, treating amino acids, peptides, proteins, bioactive compounds, flavins, pharmaceuticals and also single cell analysis. The tables provide information on analyte nature, sample matrix, optical detection aspects, CE mode and limits of detection. A selection of descriptive applications is discussed in detail to illustrate the potential of native fluorescence detection in CE. It is concluded that CE-Flu is a powerful tool for biomolecular and pharmaceutical analysis, and provides good opportunities for use in lab-on-chip devices.

A recently developed capillary electrophoresis (CE)-negative-ionisation mass spectrometry (MS) method was used to profile anionic metabolites in a microbial-host co-metabolism study. Urine samples from rats receiving antibiotics (penicillin G and streptomycin sulfate) for 0, 4, or 8 days were analysed. A quality control sample was measured repeatedly to monitor the performance of the applied CE-MS method. After peak alignment, relative standard deviations (RSDs) for migration time of five representative compounds were below 0.4 %, whereas RSDs for peak area were 7.9-13.5 %. Using univariate and principal component analysis of obtained urinary metabolic profiles, groups of rats receiving different antibiotic treatment could be distinguished based on 17 discriminatory compounds, of which 15 were downregulated and 2 were upregulated upon treatment. Eleven compounds remained down- or upregulated after discontinuation of the antibiotics administration, whereas a recovery effect was observed for others. Based on accurate mass, nine compounds were putatively identified; these included the microbial-mammalian co-metabolites hippuric acid and indoxyl sulfate. Some discriminatory compounds were also observed by other analytical techniques, but CE-MS uniquely revealed ten metabolites modulated by antibiotic exposure, including aconitic acid and an oxocholic acid. This clearly demonstrates the added value of CE-MS for nontargeted profiling of small anionic metabolites in biological samples.

Capillary electrophoresis (CE) with wavelength-resolved fluorescence detection (wrFlu) was evaluated for the study of protein unfolding using non-reduced and reduced β-lactoglobulin B (β-LGB) as model compounds. Protein unfolding was achieved by incubation in sodium phosphate (pH 3.0) containing increasing concentrations of urea (0–7.1 M). CE-wrFlu was performed using the incubation media as background electrolytes (BGEs). At low urea concentrations (0–3.1 M), CE-wrFlu analysis of non-reduced β-LGB showed a single peak with a maximum emission wavelength (λmax) of 333 nm, indicating the protein was in its folded state. When β-LGB was exposed to 3.6 and 4.1 M urea, CE-wrFlu revealed an additional peak with a λmax of 353 nm and a reduced migration time, suggesting the formation of fully unfolded species. Upon further raising the urea concentration up to 6.5 M, the peak intensity of the unfolded protein increased. At the same time, the later-migrating peak became wider and lower, showing a decrease of migration time and a shift of λmax (333–353 nm), indicating gradual unfolding. Construction of a λmax-based transition curve for the later-migrating β-LGB species provided a denaturant-concentration midpoint of unfolding (cm) of 5.39 M, which was similar to the cm determined by plotting the corrected effective electrophoretic mobility (μeff,c) vs. urea concentration. Stand-alone fluorescence spectroscopy of the same β-LGB solutions revealed a lower cm (4.97 M), most probably because the respective β-LGB species were not separated, yielding ensemble average data. For reduced β-LGB, at all tested urea concentrations one protein peak was observed, whereas λmax and μeff,c indicated protein unfolding at much lower urea concentrations (cm of 1.2 M). We conclude that CE-wrFlu can distinguish protein conformational states and thus may provide useful additional information on unfolding pathways.

We propose a new method to search for hypothetical scalar particles that have feeble interactions with Standard-Model particles. In the presence of massive bodies, these interactions produce a non-zero Yukawa-type scalar-field magnitude. Using radio-frequency spectroscopy data of atomic dysprosium, as well as atomic clock spectroscopy data, we constrain the Yukawa-type interactions of a scalar field with the photon, electron, and nucleons for a range of scalar-particle masses corresponding to length scales $ > 10$ cm. In the limit as the scalar-particle mass $m_\phi \to 0$, our derived limits on the Yukawa-type interaction parameters are: $\Lambda_\gamma \gtrsim 8 \times 10^{19}$ GeV, $\Lambda_e \gtrsim 1.3 \times 10^{19}$ GeV, and $\Lambda_N \gtrsim 6 \times 10^{20}$ GeV. Our measurements also constrain combinations of interaction parameters, which cannot otherwise be probed with traditional anomalous-force measurements. Since its introduction in 1987, CE-MS has become an increasingly important technique for the analysis of biomolecules. Since our previous update on CE-MS methods within the field of intact protein analysis (Electrophoresis 2011, 32, 66-82), a variety of interesting methodological improvements and applications have been reported in literature. Therefore, this article presents an overview of the development and application of CE-MS for intact protein analysis as published between June 2010 and June 2012. The article is divided in sections that treat CE coupled to MS through ESI, MALDI, and ICP ionization, respectively. In the section about CE-ESI-MS, technological developments with respect to CE-MS interfacing, prevention of protein adsorption, and chip-based CE-MS are treated in more detail. Novel interfacing strategies and the development of improved capillary coating strategies appeared to be the major developments. Furthermore, in all sections, the applicability of CE-MS for intact protein analysis is demonstrated by representative examples, including important developments in the fields of biopharmaceutical characterization and the analysis of proteins in biological samples. High volumes of copper hydroxide, cuprous oxide and copper oxychloride were sprayed under natural conditions onto mature orange trees to compare the retention on citrus leaves and fruit over a period of 56 days by means of copper residue analyses and a spray deposition assessment protocol using fluorometry, photomacrography and digital image analyses. Rainfall and increase in fruit size were also recorded to determine if it had an influence on weathering of copper residues. Initial retention following application of the different fungicides differed on Valencia leaves and fruit: applications with cuprous oxide retained significantly more copper residue and fluorescent pigment, while copper hydroxide retained higher copper and pigment levels on Navel leaves and fruit. Nonetheless, persistence of copper residues deposited by the three copper fungicide formulations was similar and decreased at the same tempo during both seasons; initially a fast reduction (48 and 60% for year one and two respectively) in residue during the first 14 days followed by a more gradual decline (41 and 24% for year one and two respectively) from 14 to 56 days. The loss of copper residues was attributed to weathering (days after treatment), fruit growth and cumulative rainfall as these factors were inversely correlated with copper residue levels (Pearson's r = −0.840, −0.722 and −0.733 respectively). A 76% and 90% correlation was observed between the copper residue analyzed and the quantitative fluorescent pigment measurements on mature leaves and fruit, respectively; showing that fruit is more reliable for fluorometry analyses and that this technique proved to be an effective tool for spray deposition and persistence assessment of copper fungicides. All copper formulations tested at these registered rates at 35-day spray intervals were effective in controlling Guignardia citricarpa .

The potential of CE with native fluorescence detection (Flu) for the profiling of the therapeutic protein erythropoietin (EPO) was studied. EPO is a highly heterogeneous glycoprotein comprising a large number of isoforms. CE was applied to induce separation among the various glycoforms. Native Flu of EPO provided high detection selectivity yielding good signal-to-noise ratios and stable baselines, particularly when compared to conventional UV absorbance detection. In order to enhance EPO isoform resolution, CE was performed using a capillary with a neutral coating in combination with a simple BGE of 2.0 M acetic acid (pH 2.1). CE-Flu analysis of the EPO biological reference preparation of the European Pharmacopeia resulted in a highly detailed glycoform profile. Migration time RSDs for selected EPO isoforms were less than 0.22% and 0.80% for intraday and interday repeatability, respectively. RSDs for relative peak intensity of the major EPO isoforms were less than 3%. The achieved resolution, migration time stability, and sensitivity allowed discrimination of different EPO products (EPO-α and EPO-β) based on the recorded glycoform pattern. The developed CE-Flu method is relatively straightforward, and shows potential for quality control in biopharmaceutical production.

Thromboelastometry (ROTEM) is a popular point-of-care test. It generates results quickly and may benefit individualised guided haemostatic therapy. However, processing of specimens by non-technicians might decrease the quality and reproducibility of results. Centralised laboratory equipment receiving specimens through a pneumatic tube system (PTS) could avoid this. This study aimed to evaluate the influence of PTS transport on ROTEM results and its contribution to contact activation assessed by thrombin generation (TG). Methods: Specimens from 44 patients were drawn immediately after arterial puncture. Two were anti-coagulated by citrate and two by citrate/corn trypsin inhibitor, a Factor XIIa pathway inhibitor. Both types of samples were transported by walking and PTS. Subsequently, analysis was performed: ROTEM on citrated blood, and TG on citrated and corn trypsin inhibitor (CTI) blood using either 0 or 1 pM tissue factor (TF). Results: In ROTEM analysis the NATEM assay showed significant differences. The EXTEM assay revealed small significant differences for clot formation time: 65 seconds (SD +/- 20) versus 67 seconds (SD +/- 17), and alpha angle 79 degrees (SD +/- 3) versus 77 degrees (SD +/- 3). The results remained within reference range. TG was not significantly affected by the type of tube transport, independent of the amount of TF. Conclusion: PTS for ROTEM analysis is feasible except for NATEM assays. The amount of contact activation via Factor XIIa in terms of TG is independent of transport type. However, due to the different characteristics of pneumatic systems, hospitals should check its impact on the results before introducing this route of transport. Crown

A thinlayer of carbon nanofibers (CNFs) was synthesized on a thin polycrystalline rhodium (Rh) metal layer by decomposing ethylene in the presence of hydrogen. Interaction of Rh crystals with carbon results in fragmentation and formation of Rh-nanoparticles, facilitating CNF growth. CNFs are immobilized on the surface by an apparently amorphous intermediate layer containing both Rh and C. Maximum CNF growth was achieved at intermediate hydrogen concentrations. A CNF growth mechanism by (sub-) surface diffusion of carbon on Rh is suggested. Carbon nanofibers/tubes (CNF/Ts) are finding applications in e.g. electronic devices, electrochemical capacitors, catalyst support layers in microreactors, additives to polymers and super hydrophobic coatings. Controlled growth of these structures is important to realize application in nanoscale devices. Synthesis of CNF/Ts by catalytic chemical vapor deposition method offers high rate of carbongrowth and easy control of the reaction conditions [1]. Fe, Co, Ni are widely studied metals for CNF/T growth, however, the presence of residual ferro-magnetic catalyst particles in the CNF/Ts may be an obstacle for research on the magnetic properties of CNF/Ts, as well as for applications in electronic devices, electrochemical capacitors and catalysts. In addition to ferro-magnetic metals, also Pd, Pt, Ag, Au, Ru, Re, Rh, Ir and Os enable CNF/T growth. So far, exclusively Pd was studied for growing CNF/Ts on thin metal films. In the present work, we report on synthesis CNFs on thin Rh layers, exploring the effect of the conditions on CNF growth. A 100 nm polycrystalline rhodiumlayer was deposited at room temperature on a single crystalline silicon wafer (Ø 100 mm) using DC-sputtering technique. Prior to rhodium deposition, a 15 nm thick titanium adhesion layer was sputter deposited on the silicon wafer. CNF formation was carried out at atmospheric pressure in a quartz reactor. The fresh wafers were heated in nitrogen (100 ml/min) from room temperature to 600 °C (6 °C/min), followed by reduction in H2 (5 vol.%) for 15 min. After flushing with N2 for 5 min, ethylene was introduced, growing CNFs during 30 min. The concentration of ethylene (10 vol.%) in the feed was kept constant in all experiments, whereas the hydrogen concentration was varied between 0 and 20 vol.% (balance N2, total flowrate 100 ml/min). Pretreatment of fresh wafers with H2 results in significant sintering of Rh grains from ∼10 nm to ∼85 nm, as can be seen in Fig. 1. XRD analysis of the wafers confirmed this as the intensity of the Rh(1 1 1) peak-width decreased significantly (Table 1). The only phase in the layer was metallic Rh according to XRD. Carbon deposition on the Rh surface resulted in the formation of CNFs with metal particles in the tips (Fig. 2), containing exclusively Rh and carbon, according TEM–EDX. This confirms that Rh particles are responsible for the formation of CNFs and no other metals contribute to the growth

The enormous interest in proteomics research in recent years has inspired many developments in peptide chromatography. Different strategies have been developed to cope with the vast complexity of proteomics samples, trying to provide sufficient degree of separation to be able to exploit fully the potential of protein identification by mass spectrometry (MS). As reversed-phase liquid chromatography (RPLC) coupled to MS is still the method of choice for the analysis of protein digests, many efforts focus on the development of high-efficiency RP methods (e.g., monolithic columns and ultra-high-performance LC). This can also increase the speed and the sensitivity of the analysis of protein digests. As RPLC-MS alone is unlikely to provide sufficient resolution to unravel the composition of highly complex samples comprehensively, multidimensional methods will remain essential in proteome research. In this area, hydrophilic interaction chromatography (HILIC) seems to be a promising alternative to the traditional strong cation-exchange-based methods. Also, HILIC has found application in the analysis of post-translational modifications (e.g., phosphorylation and glycosylation). This review describes recent developments in LC methods for proteomics research, focusing on advances in column technology and the application of novel column materials. Illustrative examples show the possibilities of the new columns in proteomics research.

The strong regulation of plant carbon allocation and growth by trehalose metabolism is important for our understanding of the mechanisms that determine growth and yield, with obvious applications in crop improvement. To gain further insight on the growth arrest by trehalose feeding, we first established that starch-deficient seedlings of the plastidic phosphoglucomutase1 mutant were similarly affected as the wild type on trehalose. Starch accumulation in the source cotyledons, therefore, did not cause starvation and consequent growth arrest in the growing zones. We then screened the FOX collection of Arabidopsis (Arabidopsis thaliana) expressing full-length cDNAs for seedling resistance to 100 mm trehalose. Three independent transgenic lines were identified with dominant segregation of the trehalose resistance trait that overexpress the bZIP11 (for basic region/leucine zipper motif) transcription factor. The resistance of these lines to trehalose could not be explained simply through enhanced trehalase activity or through inhibition of bZIP11 translation. Instead, trehalose-6-phosphate (T6P) accumulation was much increased in bZIP11-overexpressing lines, suggesting that these lines may be insensitive to the effects of T6P. T6P is known to inhibit the central stress-integrating kinase SnRK1 (KIN10) activity. We confirmed that this holds true in extracts from seedlings grown on trehalose, then showed that two independent transgenic lines overexpressing KIN10 were insensitive to trehalose. Moreover, the expression of marker genes known to be jointly controlled by SnRK1 activity and bZIP11 was consistent with low SnRK1 or bZIP11 activity in seedlings on trehalose. These results reveal an astonishing case of primary metabolite control over growth by way of the SnRK1 signaling pathway involving T6P, SnRK1, and bZIP11.

A goal in proteomics is the analysis of proteins by LC-MS. The proteins are enzymatically digested and the resulting peptides are chromatographically separated and introduced into a tandem MS. The obtained MS data are used for a search in sequence databases, providing identification scores for the proteins. A method to improve that score is to increase the chromatographic separation and peak capacity. In this study, the chromatographic conditions were optimized for a relatively large gradient time by varying the flow rate and gradient composition. The influence of the monolithic column length (15 and 64 cm) and particle diameter (1.8 μm; 15-cm length) on the sample peak capacity, productivity and identification score was studied. For comparison of gradient systems, a scaling factor was introduced to normalize the properties/performance of columns for material, diameter and length. As model proteins/digests, a simple (myoglobin) and a larger (BSA) protein were used. The smallest peak width, highest identification scores (54 and 89% for BSA and myoglobin, respectively) and productivity (5.0 and 4.0, respectively) were obtained for the 15-cm particulate column. The study also demonstrates that a further increase in the chromatographic performance is beneficial for BSA but hardly increases the identification score for the relatively small myoglobin.

Trehalose 6-phosphate (T6P) is a sugar signal that regulates metabolism, growth, and development and inhibits the central regulatory SNF1-related protein kinase1 (SnRK1; AKIN10/AKIN11). To better understand the mechanism in wheat (Triticum aestivum) grain, we analyze T6P content and SnRK1 activities. T6P levels changed 178-fold 1 to 45 d after anthesis (DAA), correlating with sucrose content. T6P ranged from 78 nmol g−1 fresh weight (FW) pregrain filling, around 100-fold higher than previously reported in plants, to 0.4 nmol g−1 FW during the desiccation stage. In contrast, maximum SnRK1 activity changed only 3-fold but was inhibited strongly by T6P in vitro. To assess SnRK1 activity in vivo, homologs of SnRK1 marker genes in the wheat transcriptome were identified using Wheat Estimated Transcript Server. SnRK1-induced and -repressed marker genes were expressed differently pregrain filling compared to grain filling consistent with changes in T6P. To investigate this further maternal and filial tissues were compared pre- (7 DAA) and during grain filling (17 DAA). Strikingly, in vitro SnRK1 activity was similar in all tissues in contrast to large changes in tissue distribution of T6P. At 7 DAA T6P was 49 to 119 nmol g−1 FW in filial and maternal tissues sufficient to inhibit SnRK1; at 17 DAA T6P accumulation was almost exclusively endospermal (43 nmol g−1 FW) with 0.6 to 0.8 nmol T6P g−1 FW in embryo and pericarp. The data show a correlation between T6P and sucrose overall that belies a marked effect of tissue type and developmental stage on T6P content, consistent with tissue-specific regulation of SnRK1 by T6P in wheat grain.

CE coupled to MS has proven to be a powerful analytical tool for the characterization of intact proteins, as it combines the high separation efficiency of CE with the selectivity of MS. This review provides an overview of the development and application of CE-MS methods within the field of intact protein analysis as published between January 2007 and June 2010. Ongoing technological developments with respect to CE-MS interfacing, capillary coatings for CE-MS, coupling of CIEF with MS and chip-based CE-MS are treated. Furthermore, CE-MS of intact proteins involving ESI, MALDI and ICP ionization is outlined and overviews of the use of the various CE-MS methods are provided by tables. Representative examples illustrate the applicability of CE-MS for the characterization of proteins, including glycoproteins, biopharmaceuticals, protein-ligand complexes, biomarkers and dietary proteins. It is concluded that CE-MS is a valuable technique with high potential for intact protein analysis, providing useful information on protein identity and purity, including modifications and degradation products.

Two polystyrene-based capillary monolithic columns of different length (50 and 250 mm) were used to evaluate the effects of column length on gradient separation of protein digests. A tryptic digest of a 9-protein mixture was used as a test sample. Peak capacities were determined from selected extracted ion chromatograms, and tandem mass spectrometry data were used for database matching using the MASCOT search engine. Peak capacities and protein identification scores were higher for the long column with all gradients. Peak capacities appear to approach a plateau for longer gradient times; maximum peak capacity was estimated to be 294 for the short column and 370 for the long column. Analyses with similar gradient slope produced a ratio of the peak capacities of 3.36 for the long and the short column, which is slightly higher than the expected value of the square root of the column length ratio. The use of a longer monolith improves peptide separation, as reflected by higher peak capacity, and also increases protein identification, as observed from higher identification scores and a larger number of identified peptides. Attention has also been paid to the peak production rate (PPR, peak capacity per unit time). For short analysis times, the short column produces a higher PPR, while for analysis times longer than 40 min, the PPR of the 250-mm column is higher. A capillary electrophoresis–mass spectrometry (CE–MS) method using sheath liquid electrospray ionization interfacing was studied and optimized for the analysis of intact basic proteins. To prevent protein adsorption, capillaries with a noncovalent positively charged coating were utilized. Capillaries were coated by subsequent rinsing with solutions of Polybrene, dextran sulfate and Polybrene. The coating proved to be fully compatible with MS detection, causing no background signals and ionization suppression. The composition of the sheath liquid and BGE was optimized using the model proteins α-chymotrypsinogen A, ribonuclease A, lysozyme and cytochrome c . A sheath liquid of isopropanol–water–acetic acid (75:25:0.1, v/v/v) at 2 μL min −1 resulted in optimal signal intensities for most proteins, but caused dissociation of the heme group of cytochrome c . Optimum protein responses were obtained with a BGE of 50 mM acetic acid (pH 3.0), which allowed a baseline separation of the test protein mixture. Several minor impurities present in the mixture could be detected and provisionally identified using accurate mass and a protein modification database. The selectivity of the CE–MS system was investigated by the analysis of acetylated lysozyme. Eight highly related species, identified as non-acetylated lysozyme and lysozyme acetylated in various degrees, could be distinguished. The CE–MS system showed good reproducibility yielding interday (three weeks period) RSDs for migration time and peak area within 2% and 10%, respectively. With the CE–MS system, determination coefficients ( R 2 ) for protein concentration and peak area were higher than 0.996, whereas detection limits were between 11 and 19 nM.

A lamp-based fluorescence detection (Flu) system for CE was extended with a wavelength-resolved (WR) detector to allow recording of full protein emission spectra. WRFlu was achieved using a fluorescence cell that employs optical fibres to lead excitation light from a Xe-Hg lamp to the capillary window and protein fluorescence emission to a spectrograph equipped with a CCD. A 280 nm band pass filter etc. together with a 300 nm short pass cut-off filter was used for excitation. A capillary cartridge was modified to hold the detection cell in a commercial CE instrument enabling WRFlu in routine CE. The performance of the WRFlu detection was evaluated and optimised using lysozyme as model protein. Based on reference spectral data, a signal-intensity adjustment was introduced to correct for transmission losses in the detector optics that occurred for lower protein emission wavelengths. CE-WRFlu of lysozyme was performed using BGEs of 50 mM sodium phosphate (pH 6.5 or 3.0) and a charged-polymer coated capillary. Using the 3-D data set, signal averaging over time and emission-wavelength intervals was carried out to improve the S/N of emission spectra and electropherograms. The detection limit for lysozyme was 21 nM, providing sufficient sensitivity to obtain spectral information on protein impurities.

Investigation of globulin fractions by serum protein electrophoresis (SPE) is the first step towards evaluation of the proteome in the southern white rhinoceros (Ceratotherium simum simum). Furthermore, identification of changes in globulins in animals with poaching and other injuries can guide discovery of potentially useful biomarkers of inflammation. The aim of this study was to develop reference intervals for agarose gel SPE in healthy white rhinoceros and to compare these serum protein electrophoresis results to those from animals with tissue trauma. Reference intervals for total serum protein and agarose gel electrophoretic albumin and globulin fractions were generated using serum samples from 49 healthy free-ranging adult white rhinoceros. A standardised gating system together with identification of specific proteins by mass spectrometry aided in fraction identification. Six globulin fractions were identified: α1a, α1b, α2, β1, β2 and γ. Reference intervals were generated for total serum protein (76-111 g/L), albumin (10-27 g/L) and globulin fractions (α1a: 1.6-3.2 g/L; α1b: 1.7-3.6 g/L; α2: 16.1-26.6 g/L; β1: 6.6-18.2 g/L; β2: 11.8-30.4 g/L; γ: 10.4-23.1 g/L; albumin: globulin ratio: 0.12-0.39). Results were compared to those from 30 animals with various degrees and chronicities of tissue trauma. Wounded animals had lower concentrations of total serum protein, albumin, total globulin, α and β1 globulins, lower percentages of α2 and β1 globulins, and higher percentages of β2 and γ globulins. These protein changes are similar to those seen in human patients with wounds rather than classic acute phase or chronic inflammatory responses.

The use of pseudostationary phases (PSPs) in capillary electrophoresis provides powerful separation systems of high efficiency, selectivity and flexibility. Such electrokinetic chromatographic (EKC) systems are particularly useful for chiral analysis or for the analysis of samples containing a broad range of compounds. As the availability of mass and/or structural data on (unknown) sample constituents is increasingly important, the on-line coupling of EKC and mass spectrometry (MS) has gained attention. However, commonly used PSPs, such as micelles and cyclodextrines, may strongly interfere with electrospray ionization (ESI), making on-line EKC-MS quite a challenging task. This review covers the various approaches that have been proposed and developed to combine EKC and MS. A distinction is made between methodologies that prevent the PSP from entering the MS system, and methodologies that allow introduction of PSPs into the ion source. Various approaches such as partial filling of the separation capillary with PSP, use of reverse-migrating PSPs, employment of volatile PSPs, and alternative ionization modes, are outlined. Specific applications are described and overview tables are provided. It is concluded that there is no general solution for EKC-MS available yet, but new ionization techniques like atmospheric pressure photoionization may offer attractive perspectives for achieving full compatibility.

The influence of the BGE composition, including the addition of a single-isomer sulfated beta-CD derivative, on the ionization performance of the model compound carvedilol in NACE-ESI-MS was studied using an alternative infusion method. This approach employs voltage-induced infusion of the BGE containing the analyte, and takes into account the effects of variations in EOF and effective analyte mobility on the ESI-MS intensity. First, the optimal composition of the sheath liquid for CE-MS in terms of signal abundance and stability was determined. The BGE ammonium formate, acetate, and camphorsulfonate were found to have similar effects on analyte ionization. Addition of single-isomer sulfated beta-CD derivatives (available as sodium salt) to the BGE revealed that the anionic CD derivatives did not give rise to the same ionization suppression effect. This result can be attributed to differences in the dissociation state of these sodium salts. Finally, it is shown that information about chiral selectivity can also be obtained with the applied infusion method.

Background In chronic studies, the classical benzodiazepine chlordiazepoxide (CDP) is often the preferred drug because, unlike other benzodiazepines, it is soluble in water. However, rapid CDP hydrolysis in solution has been described. This would diminish plasma levels in chronic minipump studies and introduce the corelease of active compounds. Methods Therefore, the present study aimed to explore the putative hydrolysis of CDP in aqueous solution over time and to identify the hydrolysis products. Moreover, we aimed to characterize the hydrolysis products for their in vitro (3H-flunitrazepam binding and oocyte electrophysiology) and in vivo (stress-induced hyperthermia paradigm) GABAA receptor potency. Results CDP in solution hydrolyzed to the ketone structure demoxepam which was confirmed using mass spectrometry. The hydrolysis was concentration dependent (first-order kinetics) and temperature dependent. CDP exerted greater potency compared to demoxepam in vitro (increased activity at GABAA receptors containing α1 subunits) and in vivo (stress-induced hyperthermia), although 3H-flunitrazepam binding was comparable. Conclusions The classical benzodiazepine CDP is rapidly hydrolyzed in solution to the active compound demoxepam which possesses a reduced activity at the GABAA receptor. Chronic studies that use CDP in aqueous solution should thus be interpreted with caution. It is therefore important to consider drug stability in chronic minipump applications.

Protein adsorption to silica surfaces is a notorious problem in analytical separations. Evanescent-wave cavity ringdown spectroscopy (EW-CRDS) and capillary electrophoresis (CE) were employed to investigate the capability of positively charged polymer coatings to minimize the adsorption of basic proteins. Adsorption of cytochrome c (cyt c) to silica coated with a single layer of polybrene (PB), or a triple layer of PB, dextran sulfate (DS), and PB, was studied and compared to bare silica. Direct analysis of silica surfaces by EW-CRDS revealed that both coatings effectively reduce irreversible protein adsorption. Significant adsorption was observed only for protein concentrations above 400 μM, whereas the PB-DS-PB coating was shown to be most effective and stable. CE analyses of cyt c were performed with and without the respective coatings applied to the fused-silica capillary wall. Monitoring of the electroosmotic flow and protein peak areas indicated a strong reduction of irreversible protein adsorption by the positively charged coatings. Determination of the electrophoretic mobility and peak width of cyt c revealed reversible protein adsorption to the PB coating. It is concluded that the combination of results from EW-CRDS and CE provides highly useful information on the adsorptive characteristics of bare and coated silica surfaces toward basic proteins. © 2009 American Chemical Society.

The generation of bradykinin (BK; Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg) in blood and kallidin (Lys-BK) in tissues by the action of the kallikrein-kinin system has received little attention in non-mammalian vertebrates. In mammals, kallidin can be generated by the coronary endothelium and myocytes in response to ischemia, mediating cardioprotective events. The plasma of birds lacks two key components of the kallikrein-kinin system: the low molecular weight kininogen and a prekallikrein activator analogous to mammalian factor XII, but treatment with bovine plasma kallikrein generates ornitho-kinin [Thr6,Leu8]-BK. The possible cardioprotective effect of ornitho-kinin infusion was investigated in an anesthetized, open-chest chicken model of acute coronary occlusion. A branch of the left main coronary artery was reversibly ligated to produce ischemia followed by reperfusion, after which the degree of myocardial necrosis (infarct size as a percent of area at risk) was assessed by tetrazolium staining. The iv injection of a low dose of ornitho-kinin (4 µg/kg) reduced mean arterial pressure from 88 ± 12 to 42 ± 7 mmHg and increased heart rate from 335 ± 38 to 402 ± 45 bpm (N = 5). The size of the infarct was reduced by pretreatment with ornitho-kinin (500 µg/kg infused over a period of 5 min) from 35 ± 3 to 10 ± 2% of the area at risk. These results suggest that the physiological role of the kallikrein-kinin system is preserved in this animal model in spite of the absence of two key components, i.e., low molecular weight kininogen and factor XII. Capillary electrophoresis/mass spectrometry (CE/MS) is predominantly carried out using electrospray ionization (ESI). Recently, atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization (APPI) have become available for CE/MS. With the VUV lamp turned off, the APPI source may also be used for CE/MS by thermospray ionization (TSI). In the present study the suitability of ESI, APCI, APPI and TSI for drug impurity profiling by CE/MS in the positive ion mode is evaluated. The drugs carbachol, lidocaine and proguanil and their potential impurities were used as test compounds, representing different molecular polarities. A background electrolyte of 100 mM acetic acid (pH 4.5) provided baseline separation of nearly all impurities from the respective drugs. APPI yielded both even- and odd-electron ions, whereas the other ionization techniques produced even-electron ions only. In-source fragmentation was more pronounced with APCI and APPI than with ESI and TSI, which was most obvious for proguanil and its impurities. In general, ESI and TSI appeared the most efficient ionization techniques for impurities that are charged in solution achieving detection limits of 100 ng/mL (full-scan mode). APPI and APCI showed a lower efficiency, but allowed ionization of low and high polarity analytes, although quaternary ammonium compounds (e.g. carbachol) could not be detected. Largely neutral compounds, such as the lidocaine impurity 2,6-dimethylaniline, could not be detected by TSI, and yielded similar detection limits (500 ng/mL) for ESI, APPI and APCI. In many cases, impurity detection at the 0.1% (w/w) level was possible when 1 mg/mL of parent drug was injected with at least one of the CE/MS systems. Overall, the tested CE/MS systems provide complementary information as illustrated by the detection and identification of an unknown impurity in carbachol.

The feasibility of atmospheric pressure chemical ionization (APCI) as an alternative ionization technique for capillary electrophoresis-mass spectrometry (CE-MS) was investigated using a grounded sheath-flow CE-MS sprayer and an orthogonal APCI source. Infusion experiments indicated that highest analyte signals were achieved when the sprayer tip was in close vicinity of the vaporizer entrance. The APCI-MS set-up enabled detection of basic, neutral, and acidic compounds, whereas apolar and ionic compounds could not be detected. In the positive ion mode, analytes could be detected in the entire transfer voltage range (0-5 kV), whereas highest signal intensities were observed when the corona discharge current was between 1000 and 2000 nA. In the negative ion mode, the transfer voltage typically was 500 V and the optimum corona discharge current was 6000 nA. Analyte signals were raised with increasing nebulizing gas pressure, but the pressure was limited to 25 psi to avoid siphoning and current drops. Signal intensities appeared to be optimal and constant over a wide range of sheath liquid flow rate (5-25 microL/min) and vaporizer temperature (200-350 degrees C). APCI-MS signals were unaffected by the composition of the background electrolyte (BGE), even when it contained sodium phosphate and sodium dodecyl sulfate (SDS). Consequently, BGE composition, sheath-liquid flow rate, and vaporizer temperature can be optimized with respect to the CE separation without affecting the APCI-MS response. The analysis of a mixture of basic compounds and a steroid using volatile and nonvolatile BGEs further demonstrates the feasibility of CE-APCI-MS. Detection limits (S/N = 3) were 1.6-10 microM injected concentrations.

A method for the detection of trehalose-6-phosphate (T6P) in tissue of the model plant Arabidopsis thaliana is presented. Liquid-liquid extraction (LLE) and mixed mode solid-phase extraction (SPE) were used for sample pretreatment followed by anion exchange chromatography (AEC) coupled with electrospray ionization mass spectrometry (MS) for highly selective quantitative analysis. LLE of plant material was performed with chloroform/acetonitrile/water (3:7:16, v/v/v) followed by SPE with Oasis MAX material, which significantly reduced the complexity of the extracts. On-line coupling of MS with gradient AEC using a sodium hydroxide eluent was accomplished with a postcolumn ion suppressor. The method allows specific quantification of T6P with good linearity for spiked plant extracts, from 80 nM to 1.3 microM (r(2)>0.98). The limit of detection in plant extracts was 40 nM. The recovery of the method was above 80% for relevant T6P levels. The method was applied to the determination of T6P in seedlings from four mutant A. thaliana lines (TRR1-4) resisting growth arrest caused by external supply of trehalose. Results reveal that T6P accumulation differed substantially in the four mutant lines and wild type (WT). It is concluded that the mutants circumvent the growth arrest observed in WT seedlings on 100mM trehalose by different mechanisms.

This paper describes the hyphenation of cIEF and MALDI-TOF MS via a fractionation or spotting device. After focusing in cIEF the compounds are hydrodynamically mobilized and deposited on a MALDI target plate using a sheath liquid interface, which provides the catholyte solution and the electrical ground. From previous experiments, sample conditions that resulted in a high resolution in cIEF and acceptable protein signal intensity in MS were selected [Silvertand et al., Electrophoresis, 2008, 29, 1985-1996]. Besides the mixture of test proteins, the sample solution contains 1% Pharmalyte, 0.3% hydroxyethyl cellulose and 0.1% Tween 20 and is used for both optimization as well as characterization of the cIEF-MALDI-TOF MS system. Hyphenation problems encountered are mainly due to transfer of the liquid from the needle to the MALDI target plate and are solved by choosing the proper sheath catholyte (200 mM NH4OH in 50% methanol with 0.1% Tween20). MS electropherograms were reconstructed by plotting the intensities of the m/z values corresponding to the proteins versus migration time (related to spot number). Reproducibility, peak width and signal intensity for different focusing and spotting (fractionation) times were calculated using these reconstructed MS electropherograms as well as the UV electropherograms. The best results were obtained with focusing time of 75 min (no under- or overfocusing) and a spotting time of 5 s (highest protein signal intensity in MS). The applicability of the system is demonstrated by the analysis of a biopharmaceutical (glucagon) and its deamidation product.
