Abstract

A study was conducted to characterize the cosolute-induced folding of an acidic 30-residue peptide A to a coiled coil. A 433A peptide synthesizer ws utilized to determine peptide A while CD measurements were carried out on a Jasco-715 spectropolarimeter. Results showed how salt can override electrostatic repulsion in a coiled coil. Findings also indicated the fragile relationship between electrostatic and hydrophobic effects in folding folding.

Abstract

A study was conducted to analyze the energetics and kinetics of model leucine zippers, with emphasis on a structure that cannot establish interhelical electrostatic bonds. The study also examined how hydrophobic effects and electrostatic repulsion influence the rate of folding and thermodynamic stability of a dimeric leucine zipper. Results indicated that the protonation of Glu increased the folding rate and stability of the leucine zipper during the absence of any interhelical electrostatic interactions.
