Mosquitoes are the vectors of numerous diseases such as malaria, Dengue and Zika. Controlling mosquito vectors is essential for preventing and managing diseases transmitted by mosquitoes. Larvicides target larvae within their breeding habitats before they mature into adult mosquitoes and disperse. In this study, using α-hexyl cinnamaldehyde (HCA) as a lead compound, three series of derivatives comprising a total of 78 compounds were designed and synthesized. The larvicidal activities of these compounds against Aedes albopictus were evaluated, resulting in the identification of ten promising compounds. Among them, furan-3-ylmethyl cinnamate (compound 2v) and (E)-allyl 3-(4-chlorophenyl)acrylate (compound 4c) exhibited the lowest LC50 values (8.0 ppm and 7.2 ppm respectively). In a 96-h acute toxicity assay conducted on zebrafish, 2v showed no or very low toxicity with an LC50 higher than 100 ppm. To elucidate their mode of action, morphological and histopathologic analyses were performed, and the effects of the compounds on the acetylcholinesterase (AChE) and phosphatase activities were evaluated. Results suggested that the active HCA derivatives induce injury to intestinal epithelial cells by inhibiting phosphatase activity, ultimately resulting in the mortality of mosquito larvae.

Abstract The low mechanical strength, water deficiency, and oxidative protection of organic membranes impede their use as food-grade packaging materials. Cinnamaldehyde (CIN) tends to lose its activity owing to its instability. In this study, CIN was added to flaxseed gum (FG)/chitosan (CS) films prepared in a “sandwich” structure. The influence of CIN dosage on the properties of the composite films was studied, and the film formation mechanism of the membrane was explored. The elongation at break, water vapor permeability, oxygen permeability, and light transmittance of the composite film with 1.5% CIN were lower than those of the FG/CS/FG film. Supplementation of the composite membrane with CIN generated new hydrogen bonds, electrostatic interactions, and C-O-C bonds, which converted the structure of the composite film into a sheet and increased its crystallinity without markedly affecting its thermal stability. Therefore, CIN is an extremely useful additive for improving the applicability of flaxseed gum/CS membranes as food-grade packaging films.

The use of cinnamaldehyde and Vitamin C can improve immunity and intestinal health. A two-way factorial design was employed to investigate the main and interactive effects of cinnamaldehyde and vitamin C on the growth, carcass, and intestinal health of broiler chickens. A total of 288 one-day-old female Arbor Acres broiler chicks were randomly distributed among four treatment groups, consisting of six replicate cages with 12 birds each. Four treatments were basal diet or control (CON), supplemental cinnamaldehyde (CA) 300 g/ton (g/t), vitamin C (VC) 300 g/t, and cinnamaldehyde 300 g/t, and vitamin C 300 g/t (CA + VC), respectively. The results showed that supplemental CA did not affect the growth performance or slaughter performance of broilers at 21 days (d), 42 days (d), and 1–42 days (d); however, it could improve intestinal barrier function at 42 d of age and reduce the mRNA expression of inflammatory factors in the intestine at 21 d and 42 d of age. Supplemental VC showed a trend towards increasing body weight gain (BWG) at 21 d (p = 0.094), increased breast muscle rate (at 21-d 5.33%, p < 0.05 and at 42-d 7.09%, p = 0.097), and decreased the abdominal fat (23.43%, p < 0.05) and drip loss (20.68%, p < 0.05) at 42-d. Moreover, VC improves intestinal morphology and intestinal barrier function and maintains a balanced immune response. The blend of CA and VC significantly upregulated the mRNA expression of myeloid differentiation factor 88 (MyD-88) in the intestine at 21 d of age, the mRNA expression of catalase (CAT), Occludin, Claudin-1, Mucin-2, nuclear factor-kappa B (NF-κB) and toll-like receptor 4 (TLR-4) in the intestine at 42 d of age (p < 0.01), and downregulated the mRNA expression of interleukin 10 (IL-10), interleukin 6 (IL-6), tumor necrosis factor-alpha (TNF-α) in the intestine at 21-d and 42-d of age, and interleukin-1 beta (IL-1β) mRNA in intestine at 42 d of age (p < 0.01). This study suggested that the combination of CA and VC had the potential to regulate intestinal health and result in better carcass character of broilers.

Osteoarthritis (OA) is a degenerative joint disorder that is distinguished by inflammation and chronic cartilage damage. Interleukin-1β (IL-1β) is a proinflammatory cytokine that plays an important role in the catabolic processes that underlie the pathogenesis of OA. In this study, we investigate the therapeutic efficacy of exosomes derived from untreated bone-marrow-derived mesenchymal stem cells (BMMSC-Exo) and those treated with cinnamaldehyde (BMMSC-CA-Exo) for preventing the in vitro catabolic effects of IL-1β on chondrocytes. We stimulated chondrocytes with IL-1β to mimic the inflammatory microenvironment of OA. We then treated these chondrocytes with BMMSC-Exo and BMMSC-CA-Exo isolated via an aqueous two-phase system and evaluated their effects on the key cellular processes using molecular techniques. Our findings revealed that treatment with BMMSC-Exo reduces the catabolic effects of IL-1β on chondrocytes and alleviates inflammation. However, further studies directly comparing treatments with BMMSC-Exo and BMMSC-CA-Exo are needed to determine if CA preconditioning can provide additional anti-inflammatory benefits to the exosomes beyond those of CA preconditioning or treatment with regular BMMSC-Exo. Through a comprehensive molecular analysis, we elucidated the regulatory mechanisms underlying this protective effect. We found a significant downregulation of proinflammatory signaling pathways in exosome-infected chondrocytes, suggesting the potential modulation of the NF-κB and MAPK signaling cascades. Furthermore, our study identified the molecular cargo of BMMSC-Exo and BMMSC-CA-Exo, determining the key molecules, such as anti-inflammatory cytokines and cartilage-associated factors, that may contribute to their acquisition of chondroprotective properties. In summary, BMMSC-Exo and BMMSC-CA-Exo exhibit the potential as therapeutic agents for OA by antagonizing the in vitro catabolic effects of IL-1β on chondrocytes. The regulation of the proinflammatory signaling pathways and bioactive molecules delivered by the exosomes suggests a multifaceted mechanism of action. These findings highlight the need for further investigation into exosome-based therapies for OA and joint-related diseases.

Most reported breast cancer-associated deaths are directly correlated with metastatic disease. Additionally, the primary goal of treating metastatic breast cancer is to prolong life. Thus, there remains the need for more effective and safer strategies to treat metastatic breast cancer. Recently, more attention has been given to natural products (or phytochemicals) as potential anticancer treatments. This study aimed to investigate the synergistic effects of the combination of the phytochemicals chlorogenic acid and cinnamaldehyde (CGA and CA) toward inhibiting metastasis. The hypothesis was that CGA and CA in combination decrease the metastatic potential of breast cancer cells by inhibiting their invasive and migratory abilities as well as the induction of apoptosis via the downregulation of the Akt, disrupting its signal transduction pathway. To test this, wound-healing and Transwell™ Matrigel™ assays were conducted to assess changes in the migration and invasion properties of the cells; apoptosis was analyzed by fluorescence microscopy for Annexin V/propidium iodide; and immunoblotting and FACSort were performed on markers for the epithelial-to-mesenchymal transition status. The results show that CGA and CA significantly downregulated Akt activation by inhibiting phosphorylation. Consequently, increased caspase 3 and decreased Bcl2-α levels were observed, and apoptosis was confirmed. The inhibition of metastatic behavior was demonstrated by the attenuation of N-cadherin, fibronectin, vimentin, and MMP-9 expressions with concomitant increased expressions of E-cadherin and EpCAM. In summary, the present study demonstrated that CGA and CA in combination downregulated Akt activation, inhibited the metastatic potential, and induced apoptosis in different breast cancer cell lines.

In this study, clove essential oil (CL), eugenol (EU), and cinnamaldehyde (CI) were immobilized in starch sodium octenyl succinate (SSO) using an emulsification method. The main characteristics, stability, and biological properties of the prepared emulsions were established. Particle size analyses using dynamic light scattering showed that the smaller droplets were characteristic of emulsions containing CI (205–218 nm) and EU (181–236 nm), while the largest droplets were determined for CL emulsions (293–348 nm). Moreover, the highest antioxidant activity was determined for CL (79–83%) and EU (80–88%) emulsions, while CI emulsions showed the greatest antibacterial activity. The obtained emulsions were applied to the paper sheets and the bioactive properties of coated paper were studied. Evaluation of antioxidant properties revealed that high antioxidant activity reaching 76–92% and 87–91% was characteristic of coatings containing CL and EU, respectively. Meanwhile, coatings containing CI showed quite low antioxidant activity (4–9%) but demonstrated the greatest antimicrobial effect on Gram-negative and Gram-positive bacteria as well as yeasts. Hence, CL and EU emulsions could be used as effective natural antioxidants, whereas CI emulsions could be applied as an antimicrobial agent on cellulose-based substrates for a wide range of human health protection applications.

Porphyromonas gingivalis (Pg), a Gram-negative oral pathogen, promotes and accelerates periodontitis-associated gut disorders. Intestinal epithelial barrier dysfunction is crucial in the pathogenesis of intestinal and systemic diseases. In this study, we sought to elucidate the protective role of cinnamaldehyde (CNM, an activator of Nrf2) against P. gingivalis (W83) and Pg-derived lipopolysaccharide (Pg-LPS) induced intestinal epithelial barrier dysfunction via antioxidative mechanisms in IEC-6 cells. IEC-6 (ATCC, CRL-1592) cells were pretreated with or without CNM (100 µM), in the presence or absence of P. gingivalis (strain W83, 109 MOI) or Pg-LPS (1, 10, and 100 µg/mL), respectively, between 0–72 h time points by adopting a co-culture method. Intestinal barrier function, cytokine secretion, and intestinal oxidative stress protein markers were analyzed. P. gingivalis or Pg-LPS significantly (p < 0.05) increased reactive oxygen species (ROS) and malondialdehyde (MDA) levels expressing oxidative stress damage. Pg-LPS, as well as Pg alone, induces inflammatory cytokines via TLR-4 signaling. Furthermore, infection reduced Nrf2 and NAD(P)H quinone dehydrogenase 1 (NQO1). Interestingly, inducible nitric oxide synthase (iNOS) protein expression significantly (p < 0.05) increased with Pg-LPS or Pg infection, with elevated levels of nitric oxide (NO). CNM treatment suppressed both Pg- and Pg-LPS-induced intestinal oxidative stress damage by reducing ROS, MDA, and NO production. Furthermore, CNM treatment significantly upregulated the expression of tight junction proteins via increasing the phosphorylation levels of PI3K/Akt/Nrf2 suppressing inflammatory cytokines. CNM protected against Pg infection-induced intestinal epithelial barrier dysfunction by activating the PI3K/Akt-mediated Nrf2 signaling pathway in IEC-6 cells.

Coughs and allergies are often treated at home with cinnamon (Cinnamomum verum). COVID-19 patients were also benefited by the herb. Aspirating C. verum powder caused pulmonary toxicity, hypercapnia, and respiratory failure in humans and animals. The toxicity of C. verum during fetal lung development is generally unknown. C. verum's effects on lung development were studied in zebrafish. Zebrafish lack lungs but have a swim bladder that functions like mammalian lungs. This study examined C. verum's role in embryonic lung formation using zebrafish swim bladder development. C. verum bark was extracted in methanol, chloroform, ethyl acetate, and hexane. Zebrafish embryos received serial dilutions of these extracts. Methanol extracts of C. verum were not harmful, while hexane, chloroform, and ethyl acetate extracts prevented swim bladder formation and caused neurotoxicity in zebrafish embryos. Organogenesis and lung tumorigenesis share biology. The anti-cancer effect of C. verum against lung cancer is unclear, hence an invitro cell viability study was performed utilizing three human lung cancer cell lines. All extracts decreased lung cancer cell viability, but hexane extract was most effective, inhibiting growth at IC 50 concentrations below 50 µg/ml. The LD50 dose of hexane extract in zebrafish embryonic toxicity exceeds 100 µg/ml, indicating more activity in cancer than normal cells. The extracts also exhibited significant level of ameliorative activity against CuSO4 induced oxidative stress in live zebrafish larvae. Hexane, ethyl acetate, and chloroform fractions have high cinnamaldehyde levels according to GC–MS analyses. Thus, cinnamaldehyde may be the key element in C. verum's lung toxicity and anticancer properties. This study suggested that C. verum crude extract or pure cinnamaldehyde could treat lung cancer. The dose in pregnant women must be carefully monitored to avoid teratogenic effects on fetus lung development.

Pt/Co0.5Fe0.5Al2O4 catalysts with the average Pt particle size of 1.6–3.4 nm were prepared and applied to the selective hydrogenation of cinnamaldehyde (CAL). Not only the TOF was promoted from 0.19 to 2.70 s−1, but also the selectivity to cinnamyl alcohol (COL) was improved from 68% to 94%, with the increase of Pt particle size. Stronger interaction occurred between Co0.5Fe0.5Al2O4 and smaller-sized Pt particles, resulting in electron-deficient Pt particles, so that CO and CC bond could be activated simultaneously. However, electron-rich Pt species existed on Pt/Co0.5Fe0.5Al2O4 with larger Pt size, which is beneficial for the preferential activation of CO bond.

Plasmodium falciparum malaria contributes to significant global diseases. Computer-aided drug design, screening, and discovery were used to analyze a novel series of Methyl Eugenol Benzaldehyde Thiosemicarbazone (MEBThi) and Methyl Eugenol Cinnamaldehyde Thiosemicarbazone (MECThi) derivatives for malaria falciparum inhibition. This present study showed that 16 molecules from 28 of MEBThi and MECThi have affinities and interaction with active-site residues of cysteine protease, a key player in erythrocyte proliferation P. falciparum. 13-MECThi demonstrates the best binding affinity at ˗8.0 kcal/mol while co-drug ˗5.6 kcal/mol. Physicochemical and pharmacokinetic assays of 13-MECThi have also revealed this potent compound. Toxicity analysis shows that 13-MECThi does not have mutagenicity and carcinogenicity characters, whereas co-drug has mutagenicity probability. The molecular dynamic evaluation illustrated that the 13-MECThi complex has higher Root Mean Square Deviation (RMSD) values that indicate its structure is more flexible than the chloroquine complex. Root Mean Square Fluctuation (RMSF) complex of receptor and 13-MECThi has no fundamental differences with chloroquine complex. These compounds that have been designed should be considered as falciparum antimalarial drugs.

Since efficient hydrogenation was valued for manufacture of pharmaceutical intermediates and fragrances in recent years, the development of easily accessible heterogeneous catalysts was preferably made. Here we designed a facile one-pot synthesis of Ni modified COF-derived catalyst for selective catalytic hydrogenation of cinnamaldehyde. The nickel-based COFs catalyst reduced at 600 °C exhibited better catalytic activity for the selective production of hydrocinnamyl with an excellent yield of 98%, and the nickel-based COFs reduced at 500 °C and 700 °C preferred to selectively produce benzenepropanal. Overall, this work proves cost-effective and sustainable access to the wide application of selective hydrogenation reaction.

Gastric cancer (GC) organoids are frequently used to examine cell proliferation and death as well as cancer development. Invasion/migration assay, xenotransplantation, and reactive oxygen species (ROS) production were used to examine the effects of antioxidant drugs, including perillaldehyde (PEA), cinnamaldehyde (CA), and sulforaphane (SFN), on GC. PEA and CA repressed the proliferation of human GC organoids, whereas SFN enhanced it. Caspase 3 activities were also repressed on treatment with PEA and CA. Furthermore, the tumor formation and invasive activities were repressed on treatment with PEA and CA, whereas they were enhanced on treatment with SFN. These results in three-dimensional (3D)-GC organoids showed the different cancer development of phase II enzyme ligands in 2D-GC cells. ROS production and the expression of TP53, nuclear factor erythroid 2-related factor (NRF2), and Jun dimerization protein 2 were also downregulated on treatment with PEA and CA, but not SFN. NRF2 knockdown reversed the effects of these antioxidant drugs on the invasive activities of the 3D-GC organoids. Moreover, ROS production was also inhibited by treatment with PEA and CA, but not SFN. Thus, NRF2 plays a key role in the differential effects of these antioxidant drugs on cancer progression in 3D-GC organoids. PEA and CA can potentially be new antitumorigenic therapeutics for GC.

In recent years, infections caused by multidrug-resistant (MDR) bacteria have greatly threatened human health and imposed a burden on global public health. To overcome this crisis, there is an urgent need to seek effective alternatives to single antibiotic therapy to circumvent drug resistance and prevent MDR bacteria. According to previous reports, cinnamaldehyde exerts antibacterial activity against drug-resistant Salmonella spp. This study was conducted to investigate whether cinnamaldehyde has a synergistic effect on antibiotics when used in combination, we found that cinnamaldehyde enhanced the antibacterial activity of ceftriaxone sodium against MDR Salmonella in vitro by significantly reduced the expression of extended-spectrum beta-lactamase, inhibiting the development of drug resistance under ceftriaxone selective pressure in vitro, damaging the cell membrane, and affecting its basic metabolism. In addition, it restored the activity of ceftriaxone sodium against MDR Salmonella in vivo and inhibited peritonitis caused by ceftriaxone resistant strain of Salmonella in mice. Collectively, these results revealed that cinnamaldehyde can be used as a novel ceftriaxone adjuvant to prevent and treat infections caused by MDR Salmonella, mitigating the possibility of producing further mutant strains.

Cinnamon is a natural spice with a wide range of pharmacological functions, including anti-microbial, antioxidant, and anti-tumor activities. The aim of this study is to investigate the effects of cinnamaldehyde-rich cinnamon extract (CRCE) on the colorectal cancer cell lines HCT 116 and HT-29. The gas chromatography mass spectrometry analysis of a lipophilic extract of cinnamon revealed the dominance of trans-cinnamaldehyde. Cells treated with CRCE (10–60 µg/mL) showed significantly decreased cell viability in a time- and dose-dependent manner. We also observed that cell proliferation and migration capacity were inhibited in CRCE-treated cells. In addition, a remarkable increase in the number of sub-G1-phase cells was observed with arrest at the G2 phase by CRCE treatment. CRCE also induced mitochondrial stress, and finally, CRCE treatment resulted in activation of apoptotic proteins Caspase-3, -9, and PARP and decreased levels of mu-2-related death-inducing gene protein expression with BH3-interacting domain death agonist (BID) activation.

Cinnamon is one of the most commonly used spices worldwide. In some Arab countries, cinnamon is used with other ingredients to relieve bronchospasm and treatment of airways-related disorders. In the current study, GC, GC-MS and tracheal relaxant effect comparison were performed using the three available types in Saudi Arabia, Cinnamomum verum (Ceylon cinnamon), C. cassia (Chinese cinnamon) and C. loureiroi (Vietnamese cinnamon). The essential oil of C. verum was the most potent in the relaxation of guinea pig isolated tracheal muscles against carbachol (CCh, 1 uM)-evoked bronchospasm at the concentration range from 0.03 to 3 mg/mL followed by C. bureiroi at 0.03 to 5 mg/mL; whereas, C. cassia was the least potent oil. Cinnamaldehyde (1), isolated as the main component of the three oils induced complete relaxation of low K+ (25 mM)-evoked contractions, with mild effect on the contractions evoked by high K+ (80 mM). Pre-incubation of the tracheal tissues with glibenclamide (10 μM) significantly opposed the relaxation of low K+ by cinnamaldehyde. The standard drug, cromakalim also inserted glibenclamide-sensitive inhibition of low K+ without relaxing high K+. These results indicate that cinnamaldehyde acts predominantly by ATP-specific K+ channel opening followed by weak Ca++ antagonistic effects. The obtained results justify the medicinal value of cinnamon oil in respiratory disorders.

It is a challenging task for selective hydrogenation of cinnamaldehyde (CAL) to hydrocinnamaldehyde (HCAL) without additional by-product formation. In this work, a La2O3 supported high Ni content nanoparticle catalyst was prepared for CAL selective hydrogenation. Meanwhile, Co-La2O3 catalysts were used as a reference catalyst. XRD, TEM, STEM-HAADF, XPS, and H2-TPR measurements were used to investigate the physicochemical properties of Ni-La2O3 catalysts. The experimental results confirmed that the CAL conversion and HCAL selectivity were effectively promoted with the increase of Ni loading amounts. At a Ni/La molar ratio of four, a high HCAL selectivity of 87.4% was obtained at a CAL conversion of 88.1% under mild reaction conditions. The catalyst was recycled five times without activity loss. Combined with various characterizations, it could be inferred that the good hydrogen adsorption and dissociation capacity of Ni and the presence of a certain amount of oxygen vacancies on the La2O3 support have a positive effect on the improvement of HCAL selectivity. This work provided an effective path to design transition-metal-based supported oxide catalyst for the cinnamaldehyde hydrogenation to hydrocinnamaldehyde.

The effect of the mobile interconversion of [RuHCl(CO)(mtppms-Na)3] 1, and [RuH(H2O)(CO)(mtppms-Na)3]+ 2, was studied in hydrogenation of phenylacetylene and cinnamaldehyde in aqueous–organic biphasic systems, as a function of the chloride concentration and the pH of the aqueous phase. Catalytic activity of the two complexes was also determined in homogeneous organic solvents without any additives. In the biphasic system, the rate of selective hydrogenation of phenylacetylene to styrene was strongly increased upon addition of NaCl, while the reaction of cinnamaldehyde slowed, with no change in product distribution. Both reactions responded with a rate decrease upon increasing the pH of the aqueous phase. It was concluded that hydrogenation of phenylacetylene was catalyzed by 1 with no chloride dissociation, while in the reduction of cinnamaldehyde, the aquo-complex 2 was the active catalytic species. Catalytic cycles were suggested to rationalize these findings.

Eugenol, cinnamaldehyde and D-limonene, the main components of natural essential oils, are endowed with antioxidant and anti-inflammatory properties which allow them to induce beneficial effects on intestinal, cardiac and neuronal levels. In order to characterize their pharmacokinetic profiles and aptitude to permeate in the central nervous system after intravenous and oral administration to rats, new analytical procedures, easily achievable with HPLC-UV techniques, were developed. The terminal half-lives of these compounds range from 12.4 ± 0.9 (D-limonene) and 23.1 ± 1.6 min (cinnamaldehyde); their oral bioavailability appears relatively poor, ranging from 4.25 ± 0.11% (eugenol) to 7.33 ± 0.37% (cinnamaldehyde). Eugenol evidences a marked aptitude to permeate in the cerebrospinal fluid (CSF) of rats following both intravenous and oral administrations, whereas cinnamaldehyde appears able to reach the CSF only after intravenous administration; limonene is totally unable to permeate in the CSF. Eugenol was therefore recruited for in vitro studies of viability and time-/dose-dependent dopamine release in neuronal differentiated PC12 cells (a recognized cellular model mimicking dopaminergic neurons), evidencing its ability to increase cell viability and to induce dopamine release according to a U-shaped time-course curve. Moreover, concentration-response data suggest that eugenol may induce beneficial effects against Parkinson’s disease after oral administration.

A colorimetric anion sensor of methyl-3-(2-hydroxy-5-nitrophenyl amino)-3-phenylpropanoate bearing –OH and –NH groups as binding sites and nitrophenyl as a signaling unit, has been successfully accomplished. The compound functioned as a colorimetric chemosensor for H2PO4– and AcO–, in particular, the sensor showed significant naked-eye detectable color change from colorless to light yellow. In contrast, no color change was detected upon addition of other anions such as SO42–, NO3–, and CIO4–. The anion sensing ability of the sensor was further investigated by UV-Vis absorption spectroscopy in acetone. Characteristic UV-Vis spectra changes were revealed upon addition of H2PO4– and AcO–.

Natural plant essential oils cannot be applied on a large scale due to their high volatility, easy deactivation, etc. This study provides a new method to prepare a long-lasting, slow-release essential oil product by taking advantage of solid lipid nanoparticles, which will provide a scientific guideline for the future essential oil industry. In this article, solid lipid cinnamaldehyde nanoparticles were prepared using an ultrahigh-pressure homogenization method. SLN-CA with a particle size of 74 ± 5 nm, PDI of 0.153 ± 0.032, and zeta potential of −44.36 ± 2.2 mV was screened using an additional amount of cinnamaldehyde, the ratio of oil phase components, and the homogenization pressure and number of times as factors. Differential thermal analysis and spectroscopy demonstrated that cinnamaldehyde was successfully encapsulated inside the nanoparticles. The change in particle size of nanoparticles under different conditions and times was used as an indicator of stability. The stability of the finished nanoparticles was evaluated. The retention and slow-release ability of cinnamaldehyde were investigated using the concentration of cinnamaldehyde in nanoparticles as an indicator. For the effective clinical antibacterial application of biomaterials, such as for wound management and tissue repair, the biomaterials need to show proper antibacterial capability as well as non-cytotoxicity. Furthermore, the material needs to have suitable mechanical characteristics for further medical use. Chitosan hydrogel is a potential candidate for various antibacterial biomedical applications due to its amine functionalities that lead to antimicrobial characteristics. Nevertheless, its antimicrobial capability is dependent upon the degree of protonation of amine groups caused by the pH value. Moreover, its mechanical compressive strength may not be high enough for clinical use if not chemically or physically crosslinked. This study utilized a novel chemical crosslinker, mercaptosuccinic acid, to improve its mechanical characteristics. The natural antibacterial agent, cinnamaldehyde, was grafted onto the crosslinked chitosan to improve its antimicrobial capability. Meanwhile, to take advantage of the thiol functionality in the mercaptosuccinic acid, the bactericidal silver nanoparticles were incorporated through silver-thiol covalent bounding. NMR analyses indicated the chitosan was successfully mercaptosuccinic acid-crosslinked and grafted with cinnamaldehyde at different ratios. Combined the results from the mechanical assessment, swelling experiments, antimicrobial assessment, and cytotoxicity assay, the chitosan hydrogel with the highest crosslinked degree and grafted with cinnamaldehyde and silver nanoparticles is of great promise for further clinical uses.

Bacterial biofilm formation is a major cause of drug resistance and bacterial persistence; thus, controlling pathogenic biofilms is an important component of strategies targeting infectious bacterial diseases. Cinnamaldehyde (CNMA) has broad-spectrum antimicrobial and antibiofilm activities. In this study, we investigated the antibiofilm effects of ten CNMA derivatives and trans-CNMA against Gram-negative uropathogenic Escherichia coli (UPEC) and Gram-positive Staphylococcus aureus. Among the CNMA analogs tested, 4-nitrocinnamaldehyde (4-nitroCNMA) showed antibacterial and antibiofilm activities against UPEC and S. aureus with minimum inhibitory concentrations (MICs) for cell growth of 100 µg/mL, which were much more active than those of trans-CNMA. 4-NitroCNMA inhibited UPEC swimming motility, and both trans-CNMA and 4-nitroCNMA reduced extracellular polymeric substance production by UPEC. Furthermore, 4-nitroCNMA inhibited the formation of mixed UPEC/S. aureus biofilms. Collectively, our observations indicate that trans-CNMA and 4-nitroCNMA potently inhibit biofilm formation by UPEC and S. aureus. We suggest efforts be made to determine the therapeutic scope of CNMA analogs, as our results suggest CNMA derivatives have potential therapeutic use for biofilm-associated diseases.

Cinnamaldehyde contained in cinnamon oil is useful as an antidiabetic; however, it has very low oral bioavailability. One effort to increase oral bioavailability of cinnamaldehyde is by encapsulation using chitosan. Encapsulation of cinnamaldehyde in chitosan has been successfully made in the form of powder. The aim of this study was to obtain data of stability, mucoadhesive and cinnamaldehyde release from cinnamaldehyde encapsulated chitosan. Stability tests were performed physically and chemically. Mucoadhesive tests were carried out in vitro in two steps, which were granule fabrication and then mucoadhesive test. The results showed that the levels of cinnamaldehyde decreased within 4 weeks, from the first to the fourth week 100%; 99.87%; 98.85%; 97.33%, respectively. Cinnamaldehyde powder did not change significantly in specific gravity, pH, centrifugation and organoleptic after a month. The release precentage of cinnamaldehyde in acidic media pH 1.2 for 180 minutes was 83.4%, whereas in alkaline media pH 7.4 for 360 minutes was 61%. The cinnamaldehyde encapsulated chitosan nanoparticle powder performed mucoadhesive capacity in the gastric mucosa and in the intestinal mucosa of 91.5% and 84.61%, respectively.

β-Carotene (vitamin A precursor) and α-tocopherol, the utmost energetic form of vitamin E (VE), are known to be fat-soluble vitamins (FSVs) and essential nutrients needed to enhance the growth and metabolic functions of the human body. Their deficiencies are linked to numerous chronic disorders. Loading of FSVs within nanoemulsions could increase their oxidative stability and solubility. In this research, VE and β-Carotene (BC) were successfully co-entrapped within oil-in-water nanoemulsions of carrier oils, including tuna fish oil (TFO) and medium-chain triglycerides (MCTs), stabilized by modified starch and Tween-80. These nanoemulsions and free carrier oils loaded with vitamins were stored for over one month to investigate the impact of storage circumstances on their physiochemical characteristics. Entrapped bioactive compounds inside the nanoemulsions and bare oil systems showed a diverse behavior in terms of oxidation. A more deficiency of FSVs was found at higher temperatures that were more noticeable in the case of BC. VE behaved like an antioxidant to protect BC in MCT-based nanoemulsions, whereas it could not protect BC perfectly inside the TFO-loaded nanoemulsions. However, cinnamaldehyde (CIN) loading significantly enhanced the oxidative stability and FSVs retention in each nanoemulsion. Purity gum ultra (PGU)-based nanoemulsions comprising FSVs and CIN presented a greater BC retention (42.3%) and VE retention (90.1%) over one-month storage at 40 °C than Twee 80. The superior stability of PGU is accredited to the OSA-MS capabilities to produce denser interfacial coatings that can protect the entrapped compounds from the aqueous phase. This study delivers valuable evidence about the simultaneous loading of lipophilic bioactive compounds to enrich functional foods.

A synthetic route to prepare a poly(ethylene glycol) with a single cinnamaldehyde acetal unit in the polymer chain, was successfully established using a newly synthesized cinnamaldehyde acetal diethylene glycol (CADEG) as initiator. This HO-PEG(ca)-OH is non-toxic and would be degraded into a cinnamaldehyde and two PEG diols in acid environment. A whole polyethylene glycol based hydrogel was easily fabricated by thiol-ene “click” reaction in alkalescence aqueous solution using acrylate-PEG(ca)-acrylate and 4-arm PEG-SH as raw materials at room temperature. The gel time was dependent on the pH of the solution and its alkalinity can promote gel. The hydrogel can be degradable in acidic conditions and the stronger the acidity, the faster the degradation. This HO-PEG(ca)-OH also can be used in synthesis of cinnamaldehyde containing PEG derivatives, block copolymers or other acid degradable materials.

The selective oxidation of alcohols to aldehydes has attracted a lot of attention because of its potential use in agrochemicals, fragrances, and fine chemicals. However, due to homogenous catalysis, low yield, low selectivity, and hazardous oxidants, traditional approaches have lost their efficiency. The co-precipitation method was used to synthesize the silver-cobalt bimetallic catalyst supported on functionalized multi-walled carbon nanotubes (Ag-Co/S). Brunauer Emmet Teller (BET), scanning electron microscopy (SEM), energy dispersive X-ray analysis (EDX), and X-ray diffraction (XRD) were used to characterize the catalyst. For the oxidation of cinnamyl alcohol (CA) with O2 as an oxidant, the catalyst’s selectivity and activity were investigated. The impacts of several parameters on catalyst’s selectivity and activity, such as time, temperature, solvents, catalyst dosage, and stirring speed, were comprehensively studied. The results revealed that in the presence of Ag-Co/S as a catalyst, O2 could be employed as an effective oxidant for the catalytic oxidation of cinnamyl alcohol to cinnamaldehyde (CD) with 99% selectivity and 90% conversion. In terms of cost effectiveness, catalytic activity, selectivity, and recyclability, Ag-Co/S outperforms the competition. As a result, under the green chemistry methodology, it can be utilized as an effective catalyst for the conversion of CA to CD.

Nanhai No. 1, a shipwreck in the Southern Song Dynasty, China, has a history of more than 800 years. It was salvaged in 2007 and is now on display in the Guangdong Maritime Silk Road Museum. Due to the fact that the hull is a wooden cultural relic and exposed to the air, the biological corrosion and biodegradation caused by microorganisms are key problems of hull protection. At present, the antimicrobial agent Euxyl® K100 (isothiazolinone) has a significant antimicrobial effect in the field, but it has a certain negative impact on the environment and archeologists. In order to reduce the use of chemical antimicrobial agents, we evaluated the inhibitory effects of cinnamaldehyde on the main destructive microorganisms of Nanhai No. 1. Cinnamaldehyde is the main active component of cinnamon, and has broad-spectrum antimicrobial properties. The paper diffusion method, gas diffusion method and minimum inhibitory concentration experiment were used to detect the inhibitory effects of cinnamaldehyde on the main microorganisms of Nanhai No. 1. We found that cinnamaldehyde had significant inhibitory effects on Bacillus tequilensis NK-NH5, Bacillus megaterium NK-NH10, Bacillus velezensis NK-NH11, Bacillus sp. NK-NH15, Bacillus sp. NK-NH16, Bacillus sp. NK-NH17, Fusariumsolani NK-NH1 and Scedosporiumapiospermum NK.W1-3. At the same time, cinnamaldehyde had more inhibitory effects on fungi than bacteria. Finally, we verified that cinnamaldehyde can effectively inhibit the growth of microorganisms in water, for storing the scattered wood blocks of the Nanhai No. 1 hull through laboratory simulation experiments. Cinnamaldehyde, as an environment-friendly antimicrobial agent, is of great significance to protecting water-saturated wooden relics from microbial corrosion and degradation in the future.

A therapeutic potential of the TRPA1 channel agonist cinnamaldehyde for use in inflammatory bowel disease is emerging, but the mechanisms are unclear. Semi-quantitative qPCR of various parts of the porcine gastrointestinal tract showed that mRNA for TRPA1 was highest in the colonic mucosa. In Ussing chambers, 1 mmol·L−1 cinnamaldehyde induced increases in short circuit current (ΔIsc) and conductance (ΔGt) across the colon that were higher than those across the jejunum or after 1 mmol·L−1 thymol. Lidocaine, amiloride or bumetanide did not change the response. The application of 1 mmol·L−1 quinidine or the bilateral replacement of 120 Na+, 120 Cl− or 25 HCO3− reduced ΔGt, while the removal of Ca2+ enhanced ΔGt with ΔIsc numerically higher. ΔIsc decreased after 0.5 NPPB, 0.01 indometacin and the bilateral replacement of 120 Na+ or 25 HCO3−. The removal of 120 Cl− had no effect. Cinnamaldehyde also activates TRPV3, but comparative measurements involving patch clamp experiments on overexpressing cells demonstrated that much higher concentrations are required. We suggest that cinnamaldehyde stimulates the secretion of HCO3− via apical CFTR and basolateral Na+-HCO3− cotransport, preventing acidosis and damage to the epithelium and the colonic microbiome. Signaling may involve the opening of TRPA1, depolarization of the epithelium and a rise in PGE2 following a lower uptake of prostaglandins via OATP2A1.

trans-Cinnamaldehyde is a major component of Cinnamomum cassia and has been reported to give rise to antimicrobial properties in cinnamon spice. In order to better understand the relationship between the structure of trans-cinnamaldehyde and its antimicrobial role, fifteen structurally different trans-cinnamaldehyde derivatives were selected for study based on their predicted electrophilicities. Both synthesized and commercial trans-cinnamaldehyde derivatives were evaluated for their antibacterial activity, with modest mM activity levels found against both E. coli and S. aureus. An initial mechanism of action study, suggesting that the electrophilicity of the trans-cinnamaldehydes affects the antibacterial activity, was conducted as well as an exploration of their ability to alter bacterial cell wall integrity. In addition, in vivo toxicity levels were determined using the larvae of the greater wax moth, Galleria mellonella, with all derivatives tested showing low toxicity.

To obtain selective hydrogenation catalysts with low noble metal content, two carbon-supported Mo-Pt bimetallic catalysts have been synthesized from two different molybdenum precursors, i.e., Na2MoO4 and (NH4)6Mo7O24. The results obtained by X-ray photoelectron spectroscopy (XPS) and transmission electron microscopy (TEM) combined with the presence and strength of acid sites clarified the different catalytic behavior toward cinnamaldehyde hydrogenation. After impregnating the carbon support with Mo precursors, each sample was used either as is or treated at 400 °C in N2 flow, as support for Pt nanoparticles (NPs). The heating treatment before Pt deposition had a positive effect on the catalytic performance. Indeed, TEM analyses showed very homogeneously dispersed Pt NPs only when they were deposited on the heat-treated Mo/C supports, and XPS analyses revealed an increase in both the exposure and reduction of Pt, which was probably tuned by different MoO3/MoO2 ratios. Moreover, the different acid properties of the catalysts resulted in different selectivity.

Disinfection is crucial to control and prevent microbial pathogens on surfaces. Nonetheless, disinfectants misuse in routine disinfection has increased the concern on their impact on bacterial resistance and cross-resistance. This work aims to develop a formulation for surface disinfection based on the combination of a natural product, cinnamaldehyde, and a widely used biocide, cetyltrimethylammonium bromide. The wiping method was based on the Wiperator test (ASTM E2967−15) and the efficacy evaluation of surface disinfection wipes test (EN 16615:2015). After formulation optimization, the wiping of a contaminated surface with 6.24 log10 colony-forming units (CFU) of Escherichia coli or 7.10 log10 CFU of Staphylococcus aureus led to a reduction of 4.35 log10 CFU and 4.27 log10 CFU when the wipe was impregnated with the formulation in comparison with 2.45 log10 CFU and 1.50 log10 CFU as a result of mechanical action only for E. coli and S. aureus, respectively. Furthermore, the formulation prevented the transfer of bacteria to clean surfaces. The work presented highlights the potential of a combinatorial approach of a classic biocide with a phytochemical for the development of disinfectant formulations, with the advantage of reducing the concentration of synthetic biocides, which reduces the potentially negative environmental and public health impacts from their routine use.

Lung cancer is the largest cause of cancer-induced deaths. Non-small cell lung cancer (NSCLC) is the most frequently observed subtype of lung cancer. Although recent studies have provided many therapeutic options, there is still a need for effective and safe treatments. This paper reports the combined effects of cinnamaldehyde (CNM), a flavonoid from cinnamon, together with hyperthermia, a therapeutic option for cancer treatment, on the A549 NSCLC cell line. A hyperthermia treatment of 43 °C potentiated the cytotoxicity of CNM in A549 cells. This was attributed to an increase in the apoptosis markers and suppression of the survival/protective factors, as confirmed by Western blot assays. Flow cytometry supported this result because the apoptotic profile, cell health profile, and cell cycle profile were regulated by CNM and hyperthermia combination therapy. The changes in reactive oxygen species (ROS) and its downstream target pathway, mitogen-activated protein kinases (MAPK), were evaluated. The CNM and hyperthermia combination increased the generation of ROS and MAPK phosphorylation. N-acetylcysteine (NAC), a ROS inhibitor, abolished the apoptotic events caused by CNM and hyperthermia co-treatment, suggesting that the cytotoxic effect was dependent of ROS signaling. Therefore, we suggest CNM and hyperthermia combination as an effective therapeutic option for the NSCLC treatment.
