Alzheimer's Disease is a complex progressive neurodegenerative disorder characterized by neurofibrillary tangles and senile plaques in various parts of the brain particularly cerebral cortex affecting memory and cognition. Nuclear receptors such as Peroxisome proliferator-activated receptor γ [PPAR-γ] is reported to have a role in lipid and glucose homeostasis in the brain, reduces the synthesis of Aβ (beta-amyloid plaques) and also regulates mitochondrial biogenesis and inhibit the neuro-inflammation, which contributes for the improvement in the cognitive function in AD. Hence PPAR-γ is one of the newer targets for the researchers to understand the pathology of AD and to evolve the novel strategy to retard/reverse the progression of AD. PPAR-γ agonists such as Rosiglitazone and Pioglitazone have shown promising results in AD by decreasing neuro-inflammation and restoring glucose dysmetabolism leading to a reduction in neuronal deterioration. These agonists possess poor blood-brain permeability and are poor candidates for clinical use in AD. Therefore, search, design, and development for new PPAR- γ agonists with improved BBB penetration ability are imperative. The present work deals with the use of computational tools and techniques such as molecular docking, molecular dynamics to discover PPAR-γ agonists from the unexplored Seaweed Metabolite Database and predicts it's toxicological and physiochemical profile, thereby saving time and resources. Out of 1,110 seaweed compounds, the hit molecule BS052 displayed a strong binding affinity towards PPAR-γ, which possessed better lipid solubility indicating the potential to be considered as a PPAR-γ agonist, which may be useful in the management of AD.Communicated by Ramaswamy H. A series of 2-(naphthalen-1-yl)-methyl-6-arylimidazo[2,1-b][1,3,4]thiadiazole derivatives was prepared and studied for cytotoxicity against murine leukemia L1210, human cervix carcinoma HeLa, and human T-lymphocyte CEM cell lines. The preliminary study showed that compounds 5g, 6g, 7a-c, 7e, and 8e were more potent among the tested compounds. The pharmacokinetic properties of all compounds were then investigated with FAF-Drugs, a tool for prediction of ADME and toxicity. Finally, in order to support in vitro studies, molecular docking studies were performed by using AutoDock Vina with a Lamarckian genetic algorithm to determine whether or not the synthesized compounds could be used as inhibitors for the protein structure 1m17 (EGFR). The docking scores of many compounds were found to be higher than [6,7-bis(2-methoxy-ethoxy)quinazoline-4-yl]-(3-ethynyl phenyl)amine, an inhibitor of the 1m17 EGFR receptor. Among the selected compounds 7b, 7c, 7e, 7f, 7g, and 8g showed better stability in the molecular dynamics simulation study.

Halogenated organic chemicals form one of the most important groups of natural chemicals. Their misuse in industry and agriculture represent the intrusion of those chemicals into the environment, leading to widespread distribution and often to undesirable conditions, i.e., environmental pollution. It is important to accept the beneficial properties of halogenated organic compounds, and their potential impact on the environment. Decrease of dehalogenation that is the main known strategy for biodegradation is certainly a significant toxic chemical including exceptionally hexachlorobenzene (HCB), polychlorinated biphenyls (PCBs), pentachlorophenol (PCP), and tetrachloroethene (PCE). These halogen mutations can increase the hydrophobicity of organic chemicals, increasing their tendency to be collected by food chains and sorb to soil. Finally, halogen compounds can contribute to the harmful biological effects of living compounds, increasing their toxicity, mutagenicity, and other harmful forces. Objectives: This study aimed to establish the pharmacognostic profile of aerial parts of Euphorbia tirucalli (ET) as per World Health Organization guidelines for ensuring the quality and identification of adulteration. Materials and Methods: Standardization parameters such as macroscopic and microscopic characteristics of the study plant were evaluated. Hydroalcoholic and ethyl acetate extracts were prepared and subjected to preliminary phytochemical screening. Further, the extracts were used to analyze total phenol and flavonoid contents, and their antioxidant activities were estimated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and nitric oxide assay. Results: Shape, size, color, odor, surface characteristics, and microscopic images exhibited useful diagnostic characteristics of aerial parts of plants. Total ash, water-soluble, and acid-insoluble ash were found to be 16.65' ± 1.050', 5.623' ± 1.11', and 2.56' ± 0.706, respectively. The loss on drying was 9.6', and water and alcoholic extractive values were 4.0' and 26', respectively. Phytochemical screening revealed the presence of saponins, steroidal triterpenoids, phenols, and flavonoids. Total flavanoid and phenol content in hydroalcoholic and ethyl acetate extracts of ET was found to be 246 mg rut/g, 120 mg rut/g, 81.36 mg gallic acid equivalent (GAE)/g, and 279.58 mg GAE/g respectively. 2,2-diphenyl-1-picrylhydrazyl and nitric oxide scavenging assay revealed the IC50 values of hydroalcoholic and ethyl acetate extracts as 69.599 μg/ml, 20.454 μg/ml, 17.017 μg/ml, and 17.562, respectively. Conclusion: The findings obtained from the present study help to authenticate and establish the pharmacopeia standards for the ET plant.
