Morphological and agronomical describers are traditionally used in plant characterization. However, the usage of these describers have some limitations such as susceptibility to abiotic and biotic stress and environmental factors. Furthermore, the describers are not stable over time and many can only be evaluated during the adult phase of the plants, which requires time and physical space. Molecular markers offer numerous advantages compared to the conventional alternatives based on phenotype: they are stable and detectable in all vegetable tissues, and are independent of the environment and development phase. One of the main advantages of the use of molecular markers is the time reduction in the identification of genetic diversity among the studied subjects, as the genotypes may even be described for the seed or seedling phase. Many countries have already adopted molecular markers to identify olive cultivars more accurately. The aim of this study was to evaluate the genetic identity of eight olive accessions supposedly belonging to cultivar Arbequina by using microsatellite (SSR) and Sequence Characterized Amplified Region (SCAR) markers. One accession corresponding to the cultivar was also incorporated into the analysis as a reference genotype. The molecular marker data were analyzed on the software GENALEX6. The markers generated an accumulated PI and PE of 1.26 x 10-6 and 0.949, respectively. The results supported the hypothesis that all accessions belong to the cultivar Arbequina, and the markers can therefore be applied to other varieties of olive species. Descritores morfológicos e agronômicos são tradicionalmente utilizados na caracterização de plantas. Apesar de recomendado, o emprego destes descritores apresenta algumas limitações como a influência a estresses abióticos e bióticos e aos efeitos do ambiente. Além disso, não são estáveis ao longo do tempo e muitos só podem ser avaliados durante a fase adulta das plantas, o que requer tempo e espaço físico para as avaliações. Os marcadores moleculares oferecem numerosas vantagens relativamente às alternativas convencionais baseadas no fenótipo, pois são estáveis e detectáveis em todos os tecidos vegetais, independente do ambiente e fase de desenvolvimento e uma das principais vantagens da utilização destes é proporcionar a redução do tempo na identificação da diversidade genética entre os indivíduos trabalhados, podendo ser avaliadas genótipos ainda na fase de semente ou de plântula. O objetivo deste estudo foi avaliar a identidade genética de oito acessos de oliveira supostamente pertencentes a cultivar Arbequina usando microssatélites (SSR) e Sequence Characterized Amplified Region (SCAR) marcadores. Um acesso correspondente a cultivar também foi incorporado na análise como o genótipo de referência. Os dados de marcadores moleculares foram analisados com o software GENALEX 6. Como resultado, os marcadores SSR geraram um PI acumulada e PE de 1,26 x 10- 6 e 0,949, respectivamente. The production of extra virgin olive oil (EVOO) in Brazil developed quite recently, and information on commercial Brazilian EVOO’s typical features is very scarce. In just one of the previously published works on Brazilian olive oil, the assessed samples were commercially available. In this study, a comprehensive characterization of EVOO samples acquired at local stores (at Rio de Janeiro and Rio Grande do Sul, from the two most prevalent cultivars, Arbequina and Koroneiki) was carried out considering the most relevant quality parameters, antioxidant capacity, oxidative stability, total phenolic content, fatty acid composition, and minor component metabolic profiling. The latter included: (1) the determination of individual phenolic compounds (belonging to four diverse chemical classes) and triterpenic acids by means of a powerful multi-class reversed-phase LC-MS method; (2) the quantitative profiling of tocopherols, phytosterols, and pigments by normal-phase LC-DAD/fluorescence; and (3) the quantitative appraisal of the volatile pattern of the oils by solid-phase microextraction (SPME)-gas chromatography (GC)-MS. By applying these methods, the concentrations of approximately 70 minor compounds were determined in commercial EVOOs from Brazil. To the best of our knowledge, the content of a very large number of phenolic compounds of those determined in the current report (mainly secoiridoids), the three triterpenic acids (maslinic, betulinic, and oleanolic acids), and the individual chlorophyll derivatives had not been previously evaluated in Brazilian EVOOs. © The Author(s)., The health benefits of extra virgin olive oil (EVOO) are related to its chemical composition and the presence of bioactive compounds with antioxidant properties. The aim of this study was to evaluate antioxidant compounds (pigments, coenzyme Q10 (CoQ10) and phenolic compounds) and antioxidant properties of EVOO from the same region comparing different cultivars (Hojiblanca and Arbequina), harvest year and crop stage. Antioxidant properties of oils were studied before and after a gastrointestinal digestion process, by in vitro assays (DPPH, ABTS and FRAP) and antioxidant markers in Caco-2 cells (reactive oxygen species production). The content of bioactive compounds measured was significantly affected by cultivar and harvest year (except for carotenoids) and by the crop stage (except for coenzyme Q10). Higher amounts of coenzyme Q10 were observed in Hojiblanca than in Arbequina EVOO. Total phenol content and antioxidant properties were also different depending on cultivar and harvest year and the in vitro digestion process strongly improved antioxidant marker values. Antioxidant potential in bioaccessible fractions was mainly related to the content of coenzyme Q10 and phenolic compounds in EVOO. Chemometric analysis showed that the oils were clearly classified by cultivars, harvest and crop stage, according to the chemical composition and antioxidant activity analyzed in the present study

The phenolic compound contents and antioxidant activities of the leaf extracts of nine olive genotypes were determined, and the obtained data were analysed using chemometric techniques. In the crude extracts, 12 compounds belonging to the secoiridoids, phenylethanoids, and flavonoids were identified. Oleuropein was the primary component for all genotypes, exhibiting a content of 21.0 to 98.0 mg/g extract. Hydroxytyrosol, verbascoside, luteolin 7-O-glucoside, and luteolin 4&prime, O-glucoside were also present in noticeable quantities. Genotypes differed to the greatest extent in the content of verbascoside (0.45&ndash, 21.07 mg/g extract). The content of hydroxytyrosol ranged from 1.33 to 4.03 mg/g extract, and the aforementioned luteolin glucosides were present at 1.58&ndash, 8.67 mg/g extract. The total phenolic content (TPC), DPPH&bull, and ABTS&bull, + scavenging activities, ferric reducing antioxidant power (FRAP), and ability to inhibit the oxidation of -carotene-linoleic acid emulsion also varied significantly among genotypes. A hierarchical cluster analysis enabled the division of genotypes into three clusters with similarity above 60% in each group. GGE biplot analysis showed olive genotypes variability with respect to phenolic compound contents and antioxidant activities. Significant correlations among TPC, FRAP, the values of both radical scavenging assays, and the content of oleuropein were found. The contents of 7-O-glucoside and 4&prime, O-glucoside correlated with TPC, TEAC, FRAP, and the results of the emulsion oxidation assay.

Production of virgin olive oil is beginning in Brazil. This paper analyzes the characteristics of the EVOO Arbequina from Brazil in comparison with Spanish Arbequina from different regions. Quality parameters, oxidative stability, pigments, colour and fatty acid profile were assessed, and relationships with geographic and climatic conditions were studied. All the samples presented good quality and met EU standards for extra-virgin olive oil, but there were significant differences between regions and countries for many of the parameters evaluated. Major differences between Brazilian and Spanish samples were observed for free acidity and colour of the oils, as well as minor variations in the fatty acid profile. The colour differences were related to rainfall, whereas the fatty acid content was strongly influenced by altitude and temperature. These results highlight the fact that geographic area and environmental factors influence the characteristics of Arbequina oil and play an important role in newly introduced cultivars. A oliveira (Olea europaea L.) é uma planta característica da região Mediterrânea que teve seu cultivo disseminado para diversos países do mundo, devido ao grande interesse na produção de olivas e de seu azeite, ricos em ácidos graxos essenciais. As folhas da oliveira podem ser um importante subproduto gerado pela poda das árvores, pois apresentam quantidade significativa de compostos fenólicos e ácidos graxos benéficos à saúde. Tendo em vista a importância da constituição química das folhas de oliveira e o recente cultivo no sul do Brasil, este trabalho teve como objetivo fazer um estudo exploratório sobre a composição centesimal, compostos fenólicos totais, ácidos graxos e minerais presentes nas folhas da cultivar 'Arbequina', cultivada em Caçapava do Sul, RS. Os resultados encontrados mostraram que as folhas possuem 8,14±0,24% de lipídios totais, sendo que os ácidos graxos poli-insaturados representaram mais da metade do teor total de ácidos graxos, com elevada concentração de ácido linolênico (18:3n3, 46,7%) que está presente no azeite, normalmente em concentrações próximas a 1%. A concentração de compostos fenólicos totais foi de 30,21±0,31mg GAE g-1 e os minerais predominantes nas folhas foram Ca, K, Mg, P e S (entre 1153 e 11448µg g-1). A concentração de sódio encontrada foi baixa (65,4±5,20µg g-1). Esses resultados demonstram que as folhas de oliveira cultivadas na região de Caçapava do Sul apresentam grande potencial de uso para suplementação da dieta tanto humana quanto de animais, por serem ricas em nutrientes The olive tree (Olea europaea L.) is a plant characteristic of the Mediterranean region that is cultivated in various countries of the world due to the great interest in the production of olives and their oil, which is rich in essential fatty acids. The leaves of the olive tree are an important by-product generated by pruning of trees that have significant amount of phenolic and fatty acids important to health. Given the importance of compounds present in olive leaves and the recent crop in southern Brazil, this study aimed to perform an exploratory study on the proximate composition, total phenolic compounds, fatty acids and minerals present in the leaves of the 'Arbequina' variety from Caçapava do Sul, RS, Brazil. The results showed that the leaves had 8.14±0.24% of total lipids, with an amount of polyunsaturated fatty acids more than half of the total content of fatty acids, in special linolenic acid (18:3n3, 46,7%), which is present in the olive oil in general close to the concentration of 1%. The concentration of total phenolics was 30.21±0.31mg GAE g-1 and the predominant minerals in the leaves were Ca, K, Mg, P and S (between 1153 and 11448mg g-1). The sodium concentration was considered low (65.4±5.20mg g-1). These results demonstrate that the leaves from olive trees cultivated in the region of Caçapava do Sul have great potential for use in dietary supplementation both human and animal because are rich in nutrients

The chemical composition of virgin olive oils (95 samples) from Aragón (Spain) from two successive crop seasons (1997/98 and 1998/99) and its relationship with quality and oxidative stability is examined. The main characteristics were: free acidity 0.5 (% oleic acid), peroxide value 11.8 meq/kg, K232 1.92, K270 0.13, oxidative stability 51.4 h (Rancimat method), and antioxidant phenolic substances 168.5 mg/kg (as caffeic acid). From the distribution of fatty acids, a high percentage of linoleic acid (9.46 %) and a low level of stearic (2.10 %) and arachidic (0.36 %) acids, is worth noting. The effect of sample origin (7 productive regions from Aragón), olive variety (Empeltre and Arbequina), and the different steps through processing are also investigated. However, the major factor affecting results was the plague of olive flies during 1997/98 crop season. Se ha estudiado la composición química de 95 muestras de aceite de oliva virgen de Aragón procedente de dos campañas sucesivas (1997/98 y 1998/99) y su relación con la calidad y la estabilidad oxidativa. Las principales características fueron una acidez de 0,5 (% ácido oleico), índice de peróxidos 11,8 meq/kg, K232 1,92, K270 0,13, estabilidad a la oxidación 51,4 h (método Rancimat) y contenido en polifenoles antioxidantes 168,5 mg/kg (como ácido caféico). Del perfil de ácidos grasos, destaca el elevado contenido en linoleico (9,46 %) y las bajas tasas de esteárico (2,10 %) y aráquico (0,36 %). Se analiza el efecto de la procedencia de las muestras (7 comarcas productoras de Aragón), de la variedad de la aceituna (Empeltre o Arbequina) y de las distintas fases del procesado del aceite. Se destaca la gran influencia sobre la calidad del aceite de la plaga de mosca sufrida durante la campaña de 1997/98.

10 Pags., 5 Tabls., 3 Figs., Phenolic compounds have a high importance in olive oil because of their effect on shelf life and sensory properties. This study reports on the HPLC profiles of the phenolic compounds of virgin olive oils obtained from Arbequina olives from the harvesting in a super-intensive orchard under a linear irrigation system. In addition, phenolic content, carotenoid and chlorophyllic pigments, and oxidative stability were analyzed. Total phenol content and 3,4-DHPEA-EDA increased up to a maximum throughout the ripening process. The simple phenols tyrosol and hydroxytyrosol acetate increased throughout the ripening process, however, there was not found a clear trend in hydroxytyrosol content. Minor constituents such as vanillic acid and p-coumaric acid increased up to a maximum and then decreased, since vanillin decreased progressively throughout the time of harvest. 3,4-DHPEA-EDA and lignans were present in considerable amounts in the studied samples, while oleuropein aglycone was present in a low amount. Total phenol content and oil stability followed the same trend throughout the study, so a very good correlation was established between them. Total secoiridoids and, specifically, 3,4-DHPEA-EDA seemed to be responsible for oil stability. The new olive cultivar 'Sikitita' was obtained from a cross between the 'Picual' and 'Arbequina' varieties. 'Sikitita' was selected for its features, making it particularly suited to high-density olive hedgerow orchards. From the standpoint of chloroplast pigment metabolism, the fruits of the 'Picual' and 'Arbequina' varieties have significant differences. It is therefore extremely interesting to analyze the descendants of both cultivars. With regard to chlorophyll catabolism, 'Sikitita' has proven to be a cultivar with low pigmentation and low levels of chlorophyllase activity. This is contrary to the findings obtained to date, where varieties with low pigmentation are a consequence of high chlorophyllase activity ('Arbequina') and highly pigmented fruits are due to low chlorophyllase activity ('Picual'). 'Arbequina' was, until recently, the only cultivar described that had developed a carotenogenic process, despite its anthocyanic ripening. However, from its father ('Arbequina'), the 'Sikitita' cultivar has inherited the pool of enzymes necessary to esterify xanthophylls at the chromoplast level. Olive tree varieties that were cultivated only in the Mediterranean basin a few decades ago are now planted in the Southern Hemisphere as well. The chemical composition of the oils produced in countries as far distant as Spain and Chile are affected by differences in latitude and climate. In this work, seven monovarietal virgin olive oils from Chile (Arbequina, Barnea, Frantoio, Koroneiki, Leccino, Manzanilla and Picual) have been characterized by the chemical compounds responsible for taste (phenols) and aroma (volatiles). The oils were produced in five regions of Chile, and the concentration values of some chemical compounds were related to the geographical location of the olive tree orchards. Virgin olive oils from the major cultivars, Arbequina and Picual, were characterized in comparison with the same monovarietal oils produced in Spain. The concentration values of fourteen volatile compounds showed significant differences (p < 0.05) between the oils produced in Spain and Chile. This work analyzes the effect of soil salinity on olive oil quality. The chemical composition of oils extracted from young ‘Arbequina' olives subject in the field to three soil salinity levels (low-L, medium-M and high-H) was determined throughout a period of three years. A total of 30 parameters were analyzed (moisture, oil content, acidity, peroxides, ultraviolet absorbances, poliphenols, aliphatic alcohols, sterols, waxes, and fatty acid composition). Soil salinity was measured on fourteen different dates with an electromagnetic sensor. Overall, salinity increased oil content (+15 % in H with respect to L) and decreased aliphatic alcohols (-29 %) and palmitoleic acid (-32 %) (values significant at P0,05). In summary, olive oil quality was practically independent of soil salinity and, according to the prevailing legislation, all the oils were classified as “extra virgin”. Este trabajo analiza el efecto de la salinidad edáfica sobre la calidad del aceite de oliva. Durante tres campañas se determinó la composición química de aceites provenientes de olivos jóvenes ‘Arbequina' sujetos en campo a tres niveles de salinidad (baja-B, media-M y alta-A). En total se analizaron 30 parámetros (humedad, grasa, acidez, peróxidos, lecturas en el ultravioleta, polifenoles, alcoholes alifáticos, esteroles, ceras y composición de ácidos grasos). La salinidad edáfica se midió en catorce fechas con un sensor electromagnético. La salinidad produjo un aumento del porcentaje de grasa (+15 % en A respecto a B), y un descenso de los alcoholes alifáticos (-29 %) y del ácido palmitoleico (-32 %) (valores significativos a P0,05). En conclusión, la calidad del aceite de oliva fue prácticamente independiente de la salinidad, clasificándose en todos los casos como “virgen extra” según la normativa vigente.
