Proanthocyanin (PA) dimers isolated from Acacia mearnsii bark were enzymatically acylated with palmitic acid as the acyl donor by immobilized Candida antarctica lipase on acrylic resin (Novozym 435). The acylation reaction conditions were optimized by comparing the amount of enzyme, the temperature, the reaction solvents, initial water content, substrate molar ratios and reaction time. The highest acylation conversion of 96.53% was achieved under the follow conditions: PA dimers/palmitic acid at a molar ratio of 1: 10 in tert-amyl alcohol; initial water content of 5% at 60 ℃ for 12 h with 30 g/L enzyme dosage. Introducing palmitic acid into PA dimers significantly improved both the lipophilicity and antioxidant properties. The 1-octanol/water partition coefficient of the PA dimers and their derivatives showed that the lipophilicity of the derivatives were 2.4 times higher than that of the PA dimers. The derivatives exhibited strong antioxidant scavenging capacities, approximately 1.6 times greater than the original dimers. This work is of great significance to expand the application of natural PA dimers in cosmetic and food industries and also lay a foundation for the high value-added utilization of A. mearnsii.

Acacia cyclops, Acacia saligna and Acacia mearnsii are characterized as prolific invasive alien plants (IAPs) presenting a substantial ecological and economic burden on South Africa. While conventionally these species are perceived as weeds having little value, this study attempts to demonstrate their respective potential as a phytomedicinal resource in the treatment of type 2 diabetes – thereby incentivizing their eradication. Moreover, this study aimed to assess the antidiabetic and cytotoxic effects of extracts from these three invasive Australian acacias in vitro. The α-glucosidase and ɑ-amylase activities of ethanolic and aqueous extracts derived from aerial tissues of A. cyclops, A. saligna, and A. mearnsii were investigated, followed by a cytotoxicity assessment using Hoechst 33342-Propidium Iodide (PI) dual staining and quantitative fluorescence microscopy on the human colon (Caco-2) cell line. Of the extracts screened, A. saligna ethanolic bark demonstrated the highest inhibitory activity against α-amylase and α-glucosidase with an IC50 of 10.45 ± 3.79 μg/ml and 2.35 ± 0.61 μg/ml followed by leaf extract depicting strong α-glucosidase (IC50 of 3.64 ± 1.59 μg/ml) and moderate α-amylase (IC50 of 17.67 ± 3.84 μg/ml) inhibition. The IC50 values of all extracts were significantly (p ± 28.36 μg/ml) and Epigallocatechin gallate (IC50 of 68.2 ± 8.34 μg/ml). All extracts were found to be nontoxic at test concentrations on Caco-2 cells as confirmed by Hoescht 33342-PI dual staining. Overall, the findings presented in this study provide the first concurrent account of three invasive Acacias in South Africa reporting on their potential as an alternative therapeutic option for the treatment of type 2 diabetes.

Context Tannins are phenolic compounds derived from secondary plant metabolism that at moderate levels can limit the excessive degradation of the protein in the rumen and provide greater amino acids availability in the small intestine. Aims Two experiments were performed to determine the best level of inclusion of condensed tannins (CT) from Acacia mearnsii extract in the diet of goat kids. An experiment was performed with 32 castrated crossbreed Boer goat kids, with a mean age of 4 months and mean bodyweight (BW) of 19.7 ± 2.05 kg, to measure nutrient intake and performance, and another experiment was developed with 20 Boer goat kids, at 5 months of age and 29.0 ± 2.45 kg BW, in which the digestibility, nitrogen (N) balance, microbial protein synthesis, ingestive behaviour and blood urea N were determined. In both experiments, a completely randomised design was used, including CT levels at 0, 16, 32 and 48 g/kg DM. Results DM; crude protein; ether extract; neutral detergent fibre (NDF); non-fibrous carbohydrates and total digestible nutrients intakes, ether extract digestibility; time spent on ruminating, idling or chewing; number of chews per bolus; eating and ruminating rate of DM and NDF; N intake and retained, blood urea N and loin eye area were not affected (P > 0.05) by the addition of CT from A. mearnsii extract in the goat kid diets. However, the NDF g/kg BW and BW0.75 intakes and N faecal excretion showed a linear increase (P < 0.05). There was a linear increase trend (P < 0.1) in time spent eating, and a linear decrease on N urinary excretion by CT from A. mearnsii extract inclusion. The digestibility of DM, crude protein, NDF, non-fibrous carbohydrates and total digestible nutrients exhibited a linear decrease (P < 0.05) by CT from A. mearnsii extract inclusion. There was a quadratic increase (P < 0.05) in synthesis and microbial efficiency, final BW, average daily gain, total gain, gain:feed ratio, and hot and cold carcass weights and yields. Conclusion The inclusion of CT from A. mearnsii extract up to 17.7 g/kg DM is recommended, as it improves the synthesis and efficiency of microbial production, increasing the gain:feed ratio and performance of goat kids. Implications The use of condensed tannins from A. mearnsii does not affect the nutrient intakes on goat kids’ diet and also improves performance and carcass trait of these animals.

This study aimed at obtaining an extract enriched in phenolic compounds from the aerial parts of black wattle waste (Acacia mearnsii De Wild), characterization of its composition, and investigation of its antioxidant potential and preclinical toxicological profile. First, a phenolic compound enriched extract (PCEE) was obtained by sequential fractionation. Thereafter, the main classes of secondary metabolites present in PCEE were identified and quantified. The antioxidant, cytotoxic, and cytogenotoxic activities were evaluated using in vitro assays. Toxicological effects on locomotor function were assessed using in vivo open field and plus maze models. Cinnamic acid and coumarin were isolated from the PCEE and identified. In addition, high contents of phenolic compounds and flavonoids, responsible for the promising antioxidant activity, were quantified. The extract should a dose-dependent in vitro cytotoxicity in HaCaT cells. In the cytogenotoxicity bioassay, no differences in the mitotic index and micronuclei frequency were observed between PCEE and negative control groups. Also, there were no significant effects of the extract on the motor function. Taken together, these results show that the aerial parts of A. mearnsii can be used for obtaining bioactive compounds with the potential for promoting human and animal health.

The prevailing approach of total clearing to contain the spread of invasive plants (IP) in South Africa is generating enormous lignocellulosic wastes. This study examined the possibility of utilizing these wastes in the production of geopolymer composites for use in outdoor environments. Untreated wood particles from Acacia mearnsii and A. longifolia, as well as sugarcane bagasse residues, were incorporated into a geopolymer matrix developed from a binary precursor system of 75% fly ash and 25% metakaolin. The variables considered included precursor-activator ratio (PA), curing pattern (CP), lignocellulosic material (LM), and alkali concentration (Mcon). The production process was established using a mixed factorial experimental design. PA and CP were considered at 2 levels, while LM and MCon were considered at 3 levels. The density of the boards exceeded 1 g cm−3 and are classified as high-density boards. The boards have comparable sorption properties to the cement-bonded particleboard according to the EN 632-2: 2007 standard. However, only A. longifolia boards produced with 12 M NaOH and PA ratio of 2:1 met the mechanical strength requirements. Thermogravimetric analysis revealed that the boards are thermally stable. These results have shown that South African woody IPs are suitable for geopolymer wood composites, but there is still concern about their durability in the alkaline matrix. Scanning electron microscopy micrographs indicated mineralization of the particles and a partial degradation of hemicellulose was confirmed by Fourier transform infrared spectroscopy. Although the degraded components did not prevent geopolymer setting, there is need to further investigate the extent and means of preventing degradation as this can derail the intended use of the product.

The aim of this study is to assess the curing behaviour and thermal properties of phenol–formaldehyde (PF) adhesives with different degree of tannin substitution using several thermal analysis techniques. Tannin from Acacia mearnsii bark was copolymerized with PF to form a bonding agent for plywood. The resin mixture was heated at 50 °C prior to adding 40% w/w tannin solution, followed by a continuous stirring for 2 h. The amounts of tannin substitution were 20%, 30% and 40% (w/w of PF solids) and 100% PF were used as control. The study shows that the addition of tannin quickened the gel time of the resin notably, parallel with the increase in viscosity of the resin. The dry shear bond strength of the tannin phenol–formaldehyde (TPF) co-polymer ranged between 1.71 and 2.58 MPa as compared to 3.41 MPa for PF. At higher addition of tannin, the shear bond strength reduced significantly predominantly the 40% tannin substitution. Formulation containing 20% tannin was found comparable to that of neat PF. Thermal test revealed that TPF starts to degrade at a lower temperature than PF resin. All TPF resins started to cure at about 125 °C and peaked at 160 °C compared to PF that began to cure at 145 °. The DMA of the resin showed an increase in the resin stiffness with an addition of tannin. These results imply that even though TPF starts to degrade at lower temperature, the cured TPF is relatively stronger than pure PF.

Tannin has gained wider acceptance as a dietary supplement in contemporary animal nutrition investigations because of its potential to reduce enteric methane emission. However, a major drawback to dietary tannin intake is the bitter taste and instability in the gastrointestinal tract (GIT). The utilization of fats as coating materials will ensure appropriate masking of the tannin’s aversive taste and its delivery to the target site. The aims of this study were to encapsulate mimosa tannin with palm oil or sunflower oil, and to assess the microcapsules in terms of encapsulation efficiency, morphology, density, and in vitro release of tannin in media simulating the rumen (pH 5.6), abomasum (pH 2.9) and small intestine (pH 7.4). The microencapsulation of mimosa tannin in palm or sunflower oils was accomplished using a double emulsion technique. The results revealed that encapsulated mimosa tannins in palm oil (EMTP) and sunflower oil (EMTS) had high yields (59% vs. 58%) and encapsulation efficiencies (70% vs. 68%), respectively. Compared to unencapsulated mimosa tannin (UMT), the morphology showed that the encapsulated tannins were smaller in size and spherical in shape. The UMT had (p &lt, 0.01) higher particle density (1.44 g/cm3) compared to 1.22 g/cm3 and 1.21 g/cm3 for the EMTS and EMTP, respectively. The proportion of tannins released by the UMT after 24 h in the rumen (94%), abomasum (92%) and small intestine (96%) simulated buffers, reduced (p &lt, 0.01) to 24%, 21% and 19% for the EMTS and 18%, 20% and 16% for the EMTP in similar media and timeframe. The release kinetics for the encapsulated tannins was slow and steady, thus, best fitted by the Higuchi model while the UMT dissolved quickly, hence, only fitted to a First order model. Sequential tannin release also indicated that the EMTS and EMTP were stable across the GIT. It was concluded that the microencapsulation of mimosa tannin in palm or sunflower oils stabilized tannins release in the GIT simulated buffers with the potential to modify rumen fermentation. Further studies should be conducted on the palm and sunflower oils microcapsules’ lipid stability, fatty acid transfer rate in the GIT and antioxidant properties of the encapsulated tannins.

Abstract In the present study, the tannins from stem and root barks of Tessmannia burttii Harms (Caesalpiniaceae), a plant species abundantly growing in Tanzania and other parts of Africa, were investigated for their suitability in hides tanning. Tannin powder was extracted at selected temperatures (30, 50 and 80 °C) and the influence of each temperature on the crosslinking capacity was evaluated. The interaction mechanism between hide powder collagen and the tannins was studied by Differential Scanning Calorimetry (DSC), trinitrobenzensulfonic (TNBS) acid assay and amino acid hydrolysis methods. Extraction temperatures showed low influence on crosslinking capacity of the tannins. However, extract obtained at 50 °C exhibited best performance in terms of gap size between Tonset and Tpeak. The stem bark extract yield was higher than that from the root bark, but both were within the recommended ranges. The tannin content (61%) of T. burttii stem bark extract was above recommended value (10%), whereas its total phenolic content and total flavonoic content were found to be above that of commercial Acacia mearnsii tannin. The study of cross-linking parameters as a function of pH showed cross-linking to occur via a covalent mechanism at the basic amino groups. However, the bonds were not resistant to acid hydrolysis. The observed interaction mechanism indicated that tannins from stem and root barks of T. burttii belong to the condensed tannin, similar to A. mearnsii (black wattle), a commercial tannin source that was used in this study as a reference. Findings from this study depict that T. burttii extracts are auspicious eco-friendly alternative source of vegetable tannins to overcome the use of chromium salts in the leather industry. Quantity of water is a major concern, but the quality is of more urgent concern due to heavy metal pollution. In this study, the focus was to develop adsorbents (tannin resin (TR) and iron-doped tannin resin (Fe-TR)) from invasive Acacia mearnsii for Pb2+ and Ni2+ in solution. The developed TR and Fe-TR were prepared directly by crosslinking and iron-grafting; respectively with thermal stable at ≤ 350 °C. Surface functionality analysis revealed the presence of chelating groups at ≈ 1688 and 3661 cm−1. The crystallite sizes of the adsorbents were smaller compared to tannin; however, the % carbon and nitrogen were significantly higher. Also, the Brunauer–Emmet–Teller study revealed that iron-grafting of tannin after crosslinking increases the surface area, pore volume and pore size. Furthermore, the surface morphology of adsorbents indicated a well-defined structure with the formation of a whitish deposit and rough lump-like after ion uptake. Both adsorbents showed a type II isotherm with adsorption capacities of 40.650 mg/g and 13.763 mg/g for Pb2+and 120.63 mg/g and 110.74 for Ni2+ ion by TR and Fe-TR; respectively. The absorbate-adsorbent interactions were observed to be endothermic with negative and positive Gibb's free energy values for the adsorption of Pb2+ and Ni2+; respectively. The adsorption kinetics of Ni2+ is governed by several models; however, Pb2+ interaction is regulated by Pseudo-second-order model. Furthermore, the data generated offered the possibility of both chemisorption and physisorption processes; highlighting the flexibility and multifunctionality of tannin traps for the erasure of ion impurities in wastewater.

The acacia bark extract derived from Acacia mearnsii De Wild is rich in proanthocyanidins, whose constituent units are robinetinidol, fisetinidol, catechin, and gallocatechin. In this study, we examined the effect of proanthocyanidins on obesity and diabetes using KKAy mice, a type 2 diabetes model. KKAy mice were fed either a low-fat diet, a high-fat diet, or a high-fat diet mixed with an acacia bark extract, a proanthocyanidins fraction, and other fraction for 7 weeks. Monitoring the changes in the body weight revealed that acacia bark extract and proanthocyanidins fraction could prevent excessive weight gain resulting from a high-fat diet. In addition, increases in the fasting blood glucose level due to high-fat diet intake were found to be suppressed by acacia bark extract and proanthocyanidins fraction. Furthermore, proanthocyanidins derived from acacia bark were found to increase the expression of adiponectin in white adipose tissue, which enhances the action of insulin. In addition, acacia bark-derived proanthocyanidins suppressed gluconeogenesis and fatty acid synthesis in the liver, as well as suppressing the decrease in energy production under pathological conditions in skeletal muscle. In addition, acacia bark-derived proanthocyanidins showed AMPK activation and DPP-4 inhibitory action. Therefore, it was suggested that acacia bark-derived proanthocyanidins lowered fasting blood glucose levels through the above mechanism. These results suggest that proanthocyanidins derived from acacia bark are the active ingredients of the anti-obesity and anti-diabetic effects of acacia bark extract.

No. of bitstreams: 0 Previous issue date: 2021-02-07 Clarification of sugarcane juice is one of the most important steps in the sugar manufacturing process, considering that an efficient clarification may result in highly beneficial effects on the quality and yield of the final product, crystal or refined sugar. Currently, depending on the industrial plant and intended product, the use of calcium hydroxide, phosphoric acid and sulfur dioxide as auxiliary agents for clarification has been consolidated, the latter being questioned for causing adverse health reactions. In this context, the present study evaluated the performance of the natural black Acacia (Acacia mearnsii) tannin extract as an alternative for the sugarcane juice clarification. The results showed that tannin dosages between 300 and 500 ppm along with liming applied at pH 7.3 and 3 ppm of anionic flocculant provided the best performance, resulting in 32-34% of color removal and 89-93% of turbidity removal. Under these conditions, starch and dextran removal increased, 90 and 98%, as compared to 69% and 96%, obtained with conventional treatment. The performance of the tannin also reduced the calcium hardness of clarified juice by up to 18% compared to the conventional treatment, which in turn may prevent fouling in the subsequent steps. Due to their natural origin and broad availability, industrial tannin extracts are extending their application field from the classical, and still principal, application in tannery, to oenology, pharmacy, adhesives and material science. Despite the growing interest, not much is known about the thermal valorization of these biomolecules. In this article we present the thermal properties of the eight most common commercial tannin extracts and their elemental chemical composition to shed light on the end-life of the bio-materials based on these polyphenols. Results have shown that hydrolysable tannins can be managed similarly to wood, while condensed tannins have higher heating values but also generally higher chlorine content. Sumac and gambier leaf extracts registered contained heating values and significant amounts of ashes rich in calcium and sulfur. The high chlorine content observed for the extracts of mimosa (Acacia Mearnsii) does not hinder the combustion of its derived products which can still be thermally valorized. Mimosa tannin-based materials can also be added up to 2% to wood pellets and briquettes without significantly affecting the overall quality of the biofuels according to the in-force standards.

Tannins are compounds able to form complexes with proteins limiting their ruminal degradation and thus the synthesis of some odour-active compounds may be inhibited. Tannins are broadly divided in condensed tannins (CT) and hydrolysable tannins (HT). The study aimed to assess the influence of dietary inclusion of three commercial tannin extracts, namely mimosa (Acacia mearnsii; CT), chestnut (Castanea sativa; HT) or tara (Caesalpinia spinosa; HT) on volatile profile and flavour of meat and kidney fat from lambs. Comisana male lambs were divided into four groups (n = 9 each) and fed for 75 days with a concentrate-based diet (CON) or CON supplemented with 4% of one of the tannin extracts. Tannins reduced “pastoral” odour in perirenal fat of lambs the meat of which was characterized by a very low perception of this attribute. It may be assumed that p-cresol and 8-methylnonanoic acid mostly contributed to “pastoral” odour expression in the diet without condensed or hydrolysable tannins.

The objective of this study was to determine the optimum level of condensed tannins (CTs), delivered as Acacia mearnsii extract, for inclusion in the diet of goats for enhancing growth, commercial cuts, the physicochemical composition and fatty acid (FA) profile of the meat and meat sensory attributes. Thirty-two castrated crossbreed Boer goats with a mean age of four months and a mean body weight (BW) of 19.7 ± 2.05 kg were distributed in a completely randomized design. The diets varied in level of CTs [0, 16, 32 and 48 g/kg dry matter (DM)] from an Acacia mearnsii extract. Final BW; acid detergent fiber (ADF); shoulder, neck and rib weight; and loin yield increased quadratically (P ≤ 0.05) with the amount of CTs in the diet. Except for a trend of a quadratic reduction in luminosity color (P = 0.07) and a linear reduction in collagen deposition (P = 0.03) with increasing level of dietary CTs, the meat physicochemical parameters were unaffected by CTs inclusion. With increasing amount of CTs from Acacia mearnsii extract in the diet, the concentrations of C17:1 and C18:1 cis-9 in the longissimus lumborum of goats linearly decreased, whereas those of C18:0, C18:2 cis-9, cis-12 and C18:3 cis-9, cis-12, cis-15 (P ≤ 0.05) linearly increased. In addition, the values of total saturated FA (ΣSFA), polyunsaturated FA (ΣPUFA), ratio of ΣPUFA:ΣSFA and ΣPUFA:monounsaturated FA (ΣMUFA) increased (P ≤ 0.05) with increasing CTs level; in contrast, ΣMUFA and the ΣMUFA/ΣSFA ratio decreased. Other FA sums and ratios were not affected by the inclusion of CTs in the diet. The inclusion of dietary CTs tended to unfavorably reduce the FA index and enzymatic activity of Δ9-desaturase C16 (P ≤ 0.10). However, there was a linear decrease (P ≤ 0.05) in Δ9-desaturase C18 and elongase enzymatic activities and the hypocholesterolemic to hypercholesterolemic FA ratio (h:H) index with increasing level of dietary CTs, and the atherogenicity index (AI) tended to increase as the CTs level increased (P = 0.06). The dietary inclusion of the Acacia mearnsii extract improved the flavor, aroma, softness and overall acceptance of goat kid meat (P ≤ 0.05). The aim of this study was to evaluate the effect of the dietary inclusion of Acacia mearnsii tannin extract (TA) on nutrients intake and digestibility, and nitrogen (N) retention by sheep given a tropical grass-based diet. The trial was conducted with six castrated male sheep in a cross-over design in two 21-days experimental periods. The sheep were housed in metabolic cages and offered Tifton 85 hay (Cynodon dactylon) ad libitum plus concentrate at a rate of 12 g of dry matter (DM)/kg body weight (BW). Acacia mearnsii gum is not commercially exploited, being characterized as residue from A. mearnsii cultivation. This work investigated the A. mearnsii gum polysaccharide composition, its cytotoxicity and the technological effect as a stabilizer in ice cream. A. mearnsii gum showed a similar chemical structure to commercial gum Arabic and did not decrease the viability and proliferation of fibroblast cells (Balb/3T3) and hepatocarcinoma (HepG2). Rheological tests showed that the ice cream stabilized by the A. mearnsii gum had a more structured system (more interactions between the mixture components) and the same melting characteristics as the ice cream samples made with commercial gum Arabic. The results showed that A. mearnsii gum, which is actually an agro-industrial residue from tannin production for industry, is a potential stabilizing gum for the food industry, contributing to the economic development of the exploitation chain of A. mearnsii products and by-products.

The objective of this study was to evaluate the effect of inclusion of condensed tannins (CT) from black wattle (Acacia mearnsii) on feed intake, ruminal protozoa population, ruminal fermentation, and nutrient digestibility in Jersey steers. Five ruminally-cannulated steers were used in a 5x5 Latin square design, with five periods of 20 days each (14 days for diet adaptation and six days for sample collection per period). Treatments were composed of dietary inclusion levels of condensed tannins at 0, 5, 10, 15, and 20 g/kg of diet dry matter. Intakes of dry matter, organic matter, ether extract, crude protein, neutral detergent fiber, and total digestible nutrients were not affected by condensed tannins. The ruminal pH was reduced linearly with tannin levels. Ruminal ammonia nitrogen concentration was not affected by tannins. Tannins reduced the molar proportion of acetate and did not affect the ruminal protozoal population, which might be related to the low doses used. Digestibilities of dry matter, organic matter, and neutral detergent fiber were not altered, however, there was a linear reduction in crude protein digestibility. Based on these results, CT extracts from black wattle are not recommended for improving nutrient utilization in steers at the tested levels.

The objective of this study was to evaluate the effects of five levels of condensed tannins (CT) from black wattle (Acacia mearnsii) in the diets of lactating dairy cows on intake, nutrient digestibility, ruminal microbial protein synthesis, milk production, composition, oxidative profile, and blood metabolites. Five Holstein cows (88 &plusmn, 26.8 days in milk) were allocated in a 5 x 5 Latin square design for a period of 20 days (14 days of diet adaptation and six for sampling). Treatments were the inclusion levels of CT at 0, 5, 10, 15 and 20 g/kg of dry matter (DM) in the diet. There was no effect of CT on DM intake. The digestibility of DM and neutral detergent fiber changed quadratically, with the maximum values at 12.2 and 11.4 g/kg of DM, respectively. There was no effect on ruminal microbial protein synthesis and milk production, however, milk casein concentration was reduced linearly. There was no effect on the milk oxidative profile. Inclusion of CT at levels up to 20 g/kg of DM did not affect intake or microbial protein synthesis, however, added CT depressed the production of energy corrected milk and milk casein concentration.

Previously, we fully investigated the chemical and structural characteristics of two acacia gum samples. Now, the gums' physical chemical properties were investigated using zeta potential, fluorescence spectroscopy, potentiometric titration, viscometry and SAXS, and correlated to the gums's structural and self-association particularities. Since acacia gums are widely used in many food products, our aim was to elucidate the origin of the Acacia mearnsii gum (GN) properties in comparison with a commercial Acacia gum (GA). Both gums are negatively charged polyelectrolytes, nevertheless, the uronic acids in GN are stronger in view of the pKa of the carboxyl groups are ∼10–25% lower than in GA. Also, the acid dissociation constant (pKa) for both gums, can be related to the gums aggregation behavior. The pKa variability is more pronounced at lower gum concentrations and an inflexion point is noted close to the critical adsorption concentration. GN presents a more pronounced self-association behavior and a higher intrinsic viscosity (0.23 dL g−1 vs 0.13 dL g−1), indicating a less compact structure. The SAXS scattering profile of GA revealed two size levels, consistent with a thin ellipsoid morphology with 2.0 nm (thickness) and 12.0 nm (radius). GN, though, presented just one broad bulge, suggesting a higher dispersity level. It was demonstrated that specific variations in the gums chemical structure reflect directly on their self-assembly feature which, in turn, plays an important role on the gums intrinsic physical chemistry properties, whatever the probed length scale.

To investigate the ultrastructural changes, X-ray microanalysis and leakages of protein and lipids from bacteria treated with the ethanolic extract of Acacia mearnsii. The ultrastructural changes were investigated using scanning electron microscope (SEM). The X-ray microanalyses were determined using electron dispersive spectroscopy (EDS) while the protein and lipid leakages were determined spectrophotometrically. The scanning electron microscopy indicated varied ultrastructural changes in the morphology of bacterial cells treated with the ethanol extract. The X-ray microanalysis showed significant differences between the elemental contents of extract-treated and broth-cultured and untreated bacteria strains. Lipids and proteins were leaked to a greater extent from the extract-treated bacterial strains in comparison with the broth-cultured and untreated ones used as controls. The possible antibacterial effects of the ethanol extract of A. mearnsii could be inhibition of a significant step in peptidoglycan assembly, inhibition of metabolic processes as well as the cell wall and cell membrane disruptions resulting in the efflux of lipid and protein in all the bacteria tested. This is a report identifying elemental components and associating their disintegrations in bacterial isolates with the antibacterial activities of medicinal plants. While ethanol extract of A. mearnsii caused ultrastructural changes, breakdown of elemental components into different elements and leaked lipid and protein from treated bacteria, the physiological damages to the cell wall further justify the bactericidal ability of the extract.

Tannin extracts from the bark of Acacia mearnsii and wood of Schinopsis balansae, commonly known as Quebracho, were employed. These were modified at laboratory sale via the Mannich aminomethylation with formaldehyde and dimethylamine hydrochloride. Some reaction conditions were varied, namely the formaldehyde dosage and reaction time, while keeping the Mannich solution activation time constant, and their influence on the shear viscosity of the created bio-coagulants was evaluated. The effect of the final pH of the products on their shear viscosity was also analyzed. Up-scaling of the Mannich reaction for tannin from South Africa was performed and the procedure developed at 1-L scale was reproducible in upscaled conditions. One example of a modified South Africa tannin and the modified Quebracho tannin was subsequently selected for the treatment of an industrial wastewater and tested for color and turbidity reduction in jar tests. The effluent treatment was carried out in a single and dual system with cationic synthetic flocculation agents of different charge degree. Good turbidity and decoloration results (93 and 89% reduction, respectively) were obtained with the simultaneous introduction of a cationic, 40% charged polyacrylamide, with minimal dosage (5 ppm) of the latter additive. The tannin-based coagulant from Acacia mearnsii was successfully applied in dual system with cationic polyacrylamide flocculant for industrial wastewater treatment at pilot plant scale. It was shown to satisfactorily treat the water and generate less sludge.

The objective of this study was to evaluate the effect of supplemented condensed tannins (CT) from the bark of the Black Wattle tree (Acacia mearnsii) on in vivo nutrient digestibility and energy values measured with sheep under standardized conditions. A commercial A. mearnsii extract (containing 0.203 g CT/g dry matter (DM)) was mixed into a pelleted concentrate. Four treatments consisting of grass hay and concentrate were investigated, containing different concentrations of the CT-rich extract (CON, without CT; CT1, CT3 and CT5, with 1, 3 and 5% CT-rich extract in ration DM), resulting in CT concentrations of 0, 2.03, 6.19 and 10.2 g/kg DM, respectively. A new, low cost and alternative adsorbent was prepared from black acacia bark wastes by acetosolv treatment, which consists in the use of organic solvents and inorganic acids. Both materials, black acacia bark (BAB) and acetosolv treated black acacia bark (BAB–AT) were characterized in detail and used to remove crystal violet dye (CV) from aqueous media. Adsorption parameters like pH, adsorbent dosage, kinetic, equilibrium, thermodynamics and ionic strength were evaluated to determine the efficacy and, the best conditions for CV removal. The characterization showed than the most important changes occurred in lignin portion of the material, resulting in many functional groups on the BAB–AT surface. BAB–AT presented superior potential in relation BAB, reaching removal percentages of 95%, with adsorbent dosage of 750 mg L−1 and pH of 10. Homogeneous solid diffusion model (HSDM) was able to explain the kinetic profile. The equilibrium was attained within 2 h and the intraparticle diffusion coefficients were from 10−12 to 10−8 cm2 s−1. The equilibrium was well represented by the Freundlich model. Maximum adsorption capacity was 280 mg g−1. The adsorption process was spontaneous favorable and endothermic. In summary, the acetosolv treatment is a promising alternative to transform black acacia bark wastes in and efficient adsorbent (BAB–AT), thus aggregating value to this material. BAB–AT in turn is a low cost and efficient material which can be used to treat colored effluents.

The present study was designed to characterize the polyphenols isolated from Acacia mearnsii bark crude extract (B) and fractions (B1-B7) obtained by high-speed counter-current chromatography (HSCCC) and evaluate their anti-inflammatory and carbolytic enzymes (α-glucosidase and α-amylase) inhibitory activities. Fractions B4, B5, B6, B7 (total phenolics 850.3, 983.0, 843.9, and 572.5 mg·g-1, respectively; proanthocyanidins 75.7, 90.5, 95.0, and 44.8 mg·g-1, respectively) showed significant activities against reactive oxygen species (ROS), nitric oxide (NO) production, and expression of pro-inflammatory genes interleukin-1β (IL-1β) and inducible nitric oxide synthase (iNOS) in a lipopolysaccharide (LPS)-stimulated mouse macrophage cell line RAW 264.7. All the extracts suppressed α-glucosidase and α-amylase activities, two primary enzymes responsible for carbohydrate digestion. A. mearnsii bark samples possessed significantly stronger inhibitory effects against α-glucosidase enzyme (IC50 of 0.4-1.4 μg·mL-1) than the pharmaceutical acarbose (IC50 141.8 μg·mL-1). B6 and B7 (IC50 17.6 and 11.7 μg·mL-1, respectively) exhibited α-amylase inhibitory activity as efficacious as acarbose (IC50 15.4 μg·mL-1). Moreover, B extract, at 25 µg·mL-1, significantly decreased the non-mitochondrial oxidative burst that is often associated with inflammatory response in human monocytic macrophages.

Mimosa (Black wattle) tannin extract is one of the few industrially available natural sources of polyphenols. Even if its composition is rather known, limited attempts for its purification were done until now. A Soxhlet extraction, using various solvents, with single and sequential processes was performed, and the separated fractions were analytically investigated. Yield, molecular mass, phenolic and condensed tannin contents, antioxidant activity and FT-IR spectroscopy have shown that the ethyl acetate fraction strongly contains antioxidants and low molecular mass tannins (also hydrolyzable), while the alcoholic fractions contain purified flavonoids with higher molecular mass; the final residues resulted enriched in carbohydrates and ashes. Sequential extractions optimize the class of compounds extracted according to the specific application. Conforming to statistical analysis, significant correlations have been observed between phenolic content and the antioxidant activity, as well as between molecular masses and ashes content.

Methane, a powerful greenhouse gas, is considered to be a significant loss of productive potential in ruminants. The objective was to evaluate the effect of monensin and Acacia mearnsii tannins on ruminal fermentation efficiency in cattle. Six rumen-cannulated cows were distributed three diets, each of which differed in the additive used, in a replicated 3×3 Latin square experimental design. Treatments were the Control, Monensin (300 mg per animal and day or about 18 mg/kg of dry matter - DM) and a tannin-rich extract from Acacia mearnsii (100 g per animal and day or about 0.6% of DM). Each experimental period consisted of 21 days; the first 15 days were used for diet adaptation and the last 5 days for data collection. On experimental day 21, ruminal pH was analyzed by a continuous measurement probe. In order to quantify short-chain fatty acids (SCFA), methane (CH 4 ), NH 3 -N (ammonia nitrogen) production, protozoa and the ruminal dynamics, rumen contents were sampled prior to and 3, 6, 9 and 12 h after morning feeding. The fermentation technique consisted of incubation of liquid and solid rumen contents in bottles in a water bath (39 °C) for 30 min. Subsequent measurement of methane production was made using gas chromatography and used for the final estimation of Relative Energy Loss (REL). Vegetable tannins are complex polyphenols, which occur widely in nature. Traditionally, natural tannins have been used for tanning leather. In Brazil, the main tannin-based products are obtained from Acacia mearnsii, which is a leguminous tree native to Southeastern of Australia, and the first seeds were brought to Brazil in 1928. The main cultivation of acacia was established in Rio Grande do Sul state due to the possibility of raw material for charcoal, adhesives to fuel and for tanning leather. The leather tanning processes based on plant tannins are thought to be less harmful than chromium-based tanning, and it has been used as a sustainable alternative. However, there is scarce information about the environmental impact of the leather tanning processes, with most studies reporting inhibitory effects against microorganisms at high doses and stimulatory and positive health effects at low concentrations. The aim of this study was to evaluate the toxicity of tannin preparations extracted from A. mearnsii in Saccharomyces cerevisiae. Recently, Acacia crassicarpa has been planted in peatland areas with acidic soil in Indonesia for use in pulp and paper materials. Its bark is not suitable to produce bleached pulp; hence, it is discarded as waste. Meanwhile, in South Africa and other countries, Acacia mearnsii has been planted for a long time, and its bark extracts have been used as a leather tanning agent. First, the structure of condensed tannin from the bark waste of A. crassicarpa is characterized. The yield of the extracts obtained from A. crassicarpa using a 70% acetone aqueous solution (7% based on bark weight) is less than that obtained from A. mearnsii (34%). A novel flavan dimer from the condensed tannin, specific to A. crassicarpa, is isolated from the bark extracts. To the best of our knowledge, this dimer is a new compound as evidenced from pyrolysis–gas chromatography–mass spectrometry and nuclear magnetic resonance analyses; it corresponds to a gallocatechin–catechin flavan dimer with the absence of one oxygen atom at the 3C of the pyran ring. In addition, 2,4,6-trimethoxybenzoic acid methyl ester is identified as a novel pyrolysis product obtained from the cleavage of the pyran ring.

The biosynthesis of silver nanoparticles (Ag-NPs) using the hydrosol extract of the dry stem bark of Acacia mearnsii as reducing and capping agents, and their antinociceptive properties are hereby reported. By varying the temperature and reaction time, the temporal evolution of the optical and morphological properties of the as-synthesized material was investigated. The NPs were characterized by UV–visible absorption spectroscopy, transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy-dispersive x-ray spectroscopy (EDS) and x-ray diffractometry (XRD) The optical analyses show that the position of the maximum surface plasmon resonance (SPR) peak is red-shifted as the reaction temperature decreased. The TEM micrographs show that the as-synthesized Ag-NPs are spherical while the X-ray diffraction shows that the material is highly crystalline with face-centered cubic structures. The anti-inflammatory efficacy, analyzed by the formalin model, indicates that the as-synthesized Ag-NPs are very effective, with an inhibition rate of about 76%.

The effects of dietary supplements for modifying cattle digestion and metabolism are typically measured after one or more weeks of adaptation. Consequently, how quickly the effects occur remains unknown. The long-term efficacy of Acacia mearnssii bark tannins (Acacia) on methane mitigation has been previously demonstrated. The present study, therefore, investigated the time it took for the extract to affect methane emissions and milk fatty acid profiles. Twenty lactating Brown Swiss dairy cows, categorized as 10 low- and 10 high methane emitters (average difference in methane yield: 0.10 of total), were housed in respiration chambers for 4 days. A control diet consisting of a mixed ration supplemented with grass pellets and concentrate pellets was fed initially (Day 0). The original pellets were then replaced with pellets containing 141 g Acacia/kg, providing 30 g Acacia/kg of dietary dry matter (DM) (Days 1–3). Methane emissions were measured every 10 min and gas chromatography was used to analyze individual fatty acids from daily milk samples. A significant decline in methane production was detected 20 min after starting supplementation, with methane production (g/day) and methane yield (g/kg DM intake) decreasing in a linear fashion from Day 0 to Day 3 by up to proportionately 0.18 and 0.16, respectively. Additionally, changes in proportions of various milk fatty acids occurred within 3 days of Acacia feeding. Using stepwise multiple regression analysis, several milk fatty acids were identified as being related to methane emissions. Applicable equations from the literature also showed relationships to methane emissions of high and low emitters as well as to the Acacia diet effect on methane emissions. The equations with close relationships all included minor and non-dietary milk fatty acids like odd-chain fatty acids. These specific fatty acids originate from rumen microbial activity. In conclusion, A. mearnsii extract is an immediately acting methane mitigating supplement. Certain milk fatty acids are related to methane emission in dairy cows and may assist in determining whether and when tanniferous supplements will act against enteric methane formation.

The chemical composition of Acacia mearnsii wood extract is described in this paper for the first time. This wood is cultivated in Brazil and has been used to complement the demand for hardwood in the pulp industry. In this study, we performed extractions with acetone (total extracts) and dichloromethane (lipophilic extracts responsible for the pitch formation), with the obtained percentages being 1.68 and 0.68%, respectively. The lipophilic extracts were derivatized and analyzed by Fourier transform infrared spectroscopy (FTIR) and gas chromatography-mass spectrometry (GC-MS) before and after alkaline hydrolysis. The results showed that 57 compounds were detected in the lipophilic extracts and these are mainly constituted by fatty acids ca. 32.8 mg kg-1 and sterols 26.9 mg kg-1. These chemical classes have always been present in pitch deposits in the pulp industry. Therefore, these results may be used by pulp mills to establish strategies for pitch control and represent an important advance in the knowledge of A. mearnsii.

The objective was to assess the effects short-term use of monensin and Acacia mearnsii tannins as feed additives on nutrient intake, digestibility, and CH4 production in cattle. Six rumen-cannulated Holstein cows were distributed in two 3×3 Latin square experimental design, and each experimental period lasted 21 days. The basal diet was composed of corn silage and concentrate in a 50:50 dry matter (DM) basis proportion. Treatments were control, monensin (18 mg kg?1 of DM), and tannin-rich extract from Acacia mearnsii (total tannins equivalent to 6 g kg?1 of DM). Nutrient intake and apparent digestibility coefficients were not affected by the addition of monensin or tannins to diets. However, tannins showed a tendency to reduce crude protein digestibility. Monensin decreased CH4 emission by 25.6% (g kg?1 of body weight) compared with the control treatment. This study was carried out to evaluate the effects of tannin supplementation on ruminal microbiota of sixteen lambs infected and non-infected with Haemonchus contortus and Trichostrongylus colubriformis. This study was carried out to evaluate the impact of including Acacia mearnsii tannin extract (TA) as a feed additive on nutrition and productive performance of dairy cows grazing a high-quality temperate pasture and receiving supplementation with a concentrate feedstuff. Fourteen multiparous Holstein cows were assigned to either of the following treatments: concentrate without or with 20 g TA/kg dry matter (DM). Concentrate intake accounted for 32% of the total DM intake. Tannin addition increased the herbage DM intake by 22% (p < .05). There was no effect of TA inclusion on milk yield, milk composition, milk nitrogen (N) excretion, milk and plasma urea-N concentration, urinary excretion of total N, urea-N, and purine derivatives. However, TA inclusion increased the N intake and retention, total N excretion in manure, fecal N to urine N ratio, and decreased the dietary N efficiency for milk production and the percentage of ingested N excreted in urine (p < .05). In conclusion, supplementing dairy cows grazing a high-quality temperate pasture with a concentrate containing 20 g TA/kg DM showed the potential of decreasing the proportion of ingested N excreted in urine without affecting the productive performance.

This study investigated the effect of urea or calcium nitrate with or without the inclusion of Acacia tannin extract on dry matter intake, nutrient digestibility, growth performance, and methane emission in growing lambs. Forty South African Mutton Merino lambs (95 days old, average body weight of 34.7 kg) were blocked by weight and sex and randomly allocated to four groups and fed diets supplemented with: urea (control); nitrate; urea + tannin; and nitrate + tannin. The concentration of urea and nitrate source in the diet was 10 g/kg and 32 g/kg, respectively, while the tannin was an extract of Acacia mearnsii added at 42 g/kg DM. Lambs were gradually adapted to the diets for 21 days after which growth performance was monitored for 60 days. BackgroundRecent studies demonstrated that warming and elevated carbon dioxide (CO2) indirectly affect the soil microbial community structure via plant root exudates. However, there is no direct evidence for how the root exudates affect soil microbes and how the compositions of root exudates respond to climate change.ResultsThe results showed that warming directly decreased biomass of soil-borne bacteria and fungi forAcacia mearnsiiDe Willd but it did not impact soil microbial community forEucalyptus urophylla S.T. Blake. In contrast, elevated CO2had strong direct effect on increasing soil microbial biomass for both plant species. However, plant roots could significantly increase the secretion of antibacterial chemicals (most probable organic acids), which inhibited the growth of bacteria and fungi in elevated CO2environment. This inhibitory effect neutralized the facilitation from increasing CO2concentration on microbial growth.ConclusionsWe concluded that climate change can directly affect microorganisms, and indirectly affect the soil microbial community structure by changes in composition and content of plant root exudates.

Tannins have become important phytochemicals in ruminant production, due to their wide range of biological activities. The use of a crude extract often comes with limitations, such as reduced feed intake and fibre digestibility, which could be overcome by the use of encapsulated tannin extract. In this study, four rumen-cannulated Merino wethers were used in a 4 &times, 4 Latin square design to determine the effect of encapsulating Acacia mearnsii tannin extract on intake, nutrient digestibility, and methane emission. The animals were placed on one of the following diets: control diet only, diet + silvafeed (Silvafeed ByPro, 10 g/kg feed), diet + Acacia tannin extract (ATE), 40 g/kg feed), and, diet + lipid-encapsulated-ATE (palm oil encapsulated ATE, 50 g/kg feed) in 4 cycles. Wethers were offered an Eragrotis and Lucerne hay-based total mixed ration diet above maintenance requirement with forage: concentrate ratio 50:50. Silvafeed, a commercial tannin additive, was used as a positive control. Nutrient intake was not different across the treatments, but nutrient digestibility was affected by dietary additives (p &lt, 0.05). Compared to the control, and unlike the crude extract, encapsulated-ATE and silvafeed did not reduce dry matter, organic matter, and neutral detergent fibre digestibility. While the overall N-retention and total N-excretion (g/d) were not affected by dietary additives, ATE and encapsulated-ATE diets reduced urine-N excretion (g/d) and only a slight reduction was observed in silvafeed diet. The faecal-N proportion was highest in the ATE diet (388 g/kg N-intake), followed by encapsulated-ATE (317 g/kg), and silvafeed (267 g/kg), with the control diet having the lowest proportion (230 g/kg). The acetate:propionate (A:P) ratio reduced as a result of the inclusion of dietary additives with crude ATE and silvafeed having lower A:P ratio compared to the control diet. Methane production expressed in g/kg dry matter (DM) intake was reduced by 12%, 30% and 19% in the silvafeed, crude ATE and encapsulated-ATE diets, respectively (p &lt, 0.05). The reduced methane production with higher neutral detergent fibre (NDF) digestibility in the encapsulated-ATE, compared to the crude-ATE, confirms that encapsulated-tannin can be used as an additive in ruminant diets.

One of the highlighted areas in the development of new materials is the generation of micro- and nanoparticles as drug carriers which allow the progress in formulations with the ability to release active agents in a controlled way. The proanthocyanidins (PAC) extracted from the bark of the Black Wattle have stood out for their biological activities. However, most polyflavonoids have some features which limit their application in the pharmaceutical field, such as light fastness, low bioavailability of active agents, and unpleasant taste. In this context, this study aims to present the synthesis and characterization of PAC-loaded lactic-co-glycolic acid (PLGA) microparticles obtained by the multiple emulsion method. The incorporation of PAC into PLGA was successfully achieved with PAC encapsulation efficiency around 73%. Spherical microparticles were obtained with a size distribution in the range of 0.6 to 2.4 μm. The presence of PAC modified the thermal properties of the PLGA matrix. The results of in vitro assays with Vero and T24 lineage celss showed that PLGA/PAC microparticles did not promote any effect on cell proliferation by MTT assay after 24 h. The novel Acacia mearnsii proanthocyanidin-loaded PLGA microparticles have potential for application in biological systems.

Diets excessive in crude protein (CP) are unfavourable in terms of metabolic and environmental load. Dietary phenols, often binding to dietary proteins, may alleviate these problems. In an experiment with 60 lambs (3.2 ± 1.6 months of age; 29.7 ± 5.1 kg body weight), kept in pairs, five diets were tested. A diet with 157 g CP/kg dry matter (DM) served as negative control. Four diets with on average 229 (225-233) g CP/kg DM remained either non-supplemented or were supplemented with 13 g/kg DM of Acacia mearnsii extract, grapeseed extract, or a combination of both (26 g extract/kg DM). The analysed concentrations of total extractable phenols were 7.1, 8.1, 14.3, 16.6 and 25.4 g/kg DM for low (CP‒) and high CP (CP+), and high CP with acacia (CP+A), grapeseed (CP+G) and acacia plus grapeseed (CP+AG), respectively. Diets were fed for 10 weeks, and for 6 d faeces and urine were collected and subsequently stored as complete manure for 8 weeks. In blood plasma, phenol concentrations and activities of enzymes indicating liver and kidney stress were analysed. The CP+ diet increased apparent digestibility of N and its removal with the urine, with the expected increase in gaseous N emissions from the manure (13.5 vs 6.5 g/lamb per day during 8 weeks) compared to CP‒. However, no clear signs of metabolic stress were detected. Supplementing the extracts did not impair intake, growth performance and digestibility. Only the supplementation with both extracts decreased urinary N proportion of manure N, and the concomitant weak decline in gaseous emission from the manure was not significant. At least part of the phenols of both extracts seem to be bioavailable as their supplementation elevated blood plasma phenol concentrations by 15% to 40% compared to CP+. A combination of both extracts did not result in a further increase. Further studies have to identify the minimally effective dosage for reducing N emissions, which, at the same time, does not cause adverse side effects in performance.

The development of more sustainable antioxidants alternatives for the improvement of biodiesel oxidation stability is gaining importance. Vegetable tannins are known for their antioxidant properties. In this work, acacia (Acacia mearnsii), chestnut (Castanea sativa) and quebracho (Schinopsis lorentzii) tannins extracts were used in mixture with triethanolamine as antioxidants for soybean biodiesel. Tannins were characterized by Folin-Ciocalteau method for the determination of total phenolic content (TPC) and against DPPH (1,1-diphenyl-2-picryl-hydracyl) radical for scavenging capacity. When applied to the biodiesel at a concentration of 350 mg L−1 in mixture with 1.59 %wt of triethanolamine, tannins were highly effective in enhancing oxidative stability with superior performance than synthetic antioxidant tert-Butylhydroquinone (TBHQ). These findings indicate that vegetal tannins can be used as a renewable alternative to enhance biodiesel oxidation stability.

The nitrogen (N) excreted by grazing animals is the main source of nitrous oxide (N2O) emissions in livestock systems, and feeding strategies to mitigate N excretion should be further studied. The objective of this study was to evaluate the effects of ground corn and tannin extract supplementation on mitigating N2O emissions from sheep excreta. The emissions factor (EF,% of N excreted, which was emitted as N2O N) under the studied conditions were also measured. Two experiments were conducted. In the first one, six Lacaune lambs, with an average BW of 27 ± 3 kg, were distributed in a replicated 3 × 3 Latin square experimental design. The treatments included ad libitum access to ryegrass (Lolium multiflorum Lam.) silage, which contained 21.1 g N/kg DM and was supplemented with 60 g of soybean meal (S), 140 g of ground corn + 60 g of soybean meal (SC) or 140 g of ground corn + 60 g of soybean meal + 40 g/kg Acacia mearnsii tannin extract concentrate (SCT). The daily production of faeces and urine was measured, and samples were collected each day. In the second experiment, urine and faeces were applied to static chambers in the field to determine their N2O emissions. Three replicates of type of excreta from each treatment were included. The nitrogen intake was similar between the treatments, with an average of 18 g/day. The faecal N excretion obtained with tannin extract supplementation was 16% higher (P N/100 g N. The EF of the urine samples averaged 4.8 g N2O N/100 g N. Corn grain supplementation decreased urinary N excretion and N2O emissions from sheep excreta, whereas Acacia mearnsii tannin extract did not effectively improve this response.

Tannins from Acacia mearnsii were encapsulated using four different sol-gel methods acid (SGAR), basic (SGBR), silicate (SGSR) and non-hydrolytic (SGNHR) routes. The hybrid materials were analyzed using a set of techniques to characterize their structure, texture and morphology. The antimicrobial performance of the encapsulated materials was evaluated against different microorganisms (Staphylococcus aureus, Escherichia coli, Aspergillus niger and Candida sp.). The data showed that the encapsulation route significantly affects the characteristics of the resulting hybrid materials. Lettuce is often involved in foodborne outbreaks caused by pathogenic Escherichia coli. Current control strategies have often proved ineffective to ensure safe food production. For that reason, the present study compared the efficacy of tannin extracts and chlorine treatments on the reduction of E. coli ATCC 25922 adhered to lettuce leaves. E. coli was inoculated artificially on leaf surfaces of fresh crisp lettuce. Effectiveness of water, chlorine (200 mg/L), and three commercial available tannin extracts from Acacia mearnsii De Wild. (tannin AQ (2 %, w/v), tannin SG (1 %, v/v) and tannin SM (1 %, v/v)) treatments was evaluated using the viable plate count method and scanning electron microscopy (SEM). SEM results revealed that bacterial cells are attached as individual cells and in clusters to the leaf surface after 2 h of incubation. Biofilm formation was observed after 24 h of incubation. The tannin SM treatment was able to reduce counts in approximately 2 log CFU/cm2 on leaf segments. However, treatment was less effective in the reduction of E. coli counts after 24 h of incubation when compared to 2 h incubation of the same extract. The results suggest that the tannin SM extract diminishes E. coli counts adhered to and under biofilm formation on lettuce leaves and its effect is similar to the use of chlorine solutions.
