Transport of active species (i.e., ions) leaching from pigment particles incorporated in a polymer matrix is the main mechanism behind the anticorrosive performance of protective coatings. Understanding this mechanism is necessary for the effective design of the systems utilizing pigments less toxic than the most efficient chromate salts. It was demonstrated that anticorrosive pigment particles can themselves facilitate the transport of active species via the pathways formed after pigment leaching from a coating. It was also suggested that other paint components, e.g., certain additives, pigments, and fillers can be involved in the formation of transport pathways. Investigation of the possible influence of inert pigment (TiO2) on creating the pathways for chromate ion transport in polymer coatings was the primary objective of this work. In an experiment mimicking the transport of pigment species (i.e., chromate ions), a model epoxy coating containing particles of a single pigment (TiO2) was exposed to a chromate solution (aqueous, or with the addition of acetone as a polymer swelling agent). It was shown that the chromate ions can be transported in the epoxy film preferentially via the TiO2 particles/polymer matrix interface.

A set of guiding principles for the development of a standard for FAIR management of spectroscopic data are outlined and discussed. The principles form the basis for future recommendations of IUPAC Project 2019-031-1-024 specifying a detailed data model and metadata schema for describing the contents of an “IUPAC FAIRData Collection” and the organization of digital objects within that collection. Foremost among the recommendations will be a specification for an “IUPAC FAIRData Finding Aid” that describes the collection in such a way as to optimize the findability, accessibility, interoperability, and reusability of its contents. Results of an analysis of data provided by an American Chemical Society Publications pilot study are discussed in relation to potential workflows that might be used in implementing the “IUPAC FAIRSpec” standard based on these principles.

A synthetic material of silicone rubber was used to construct an artificial lens capsule (ALC) in order to replicate the biomechanical behaviour of human lens capsule. The silicone rubber was characterised by monotonic and cyclic mechanical tests to reveal its hyper-elastic behaviour under uniaxial tension and simple shear as well as the rate independence. A hyper-elastic constitutive model was calibrated by the testing data and incorporated into finite element analysis (FEA). An experimental setup to simulate eye focusing (accommodation) of ALC was performed to validate the FEA model by evaluating the shape change and reaction force. The characterisation and modelling approach provided an insight into the intrinsic behaviour of materials, addressing the inflating pressure and effective stretch of ALC under the focusing process. The proposed methodology offers a virtual testing environment mimicking human capsules for the variability of dimension and stiffness, which will facilitate the verification of new ophthalmic prototype such as accommodating intraocular lenses (AIOLs).

A transport mechanism of anticorrosive pigment species (i.e. chromates) in polymer films was investigated. This mechanism plays a crucial role in the performance of anticorrosive polymer coatings which relays on leaching of active anticorrosive species from sparingly soluble pigment particles distributed in the coating. Except for anticorrosive pigments other particles are included in coatings such as pigments with different functions or fillers.The goal of this work was to investigate how filler particles with no anticorrosive function (i.e. BaSO4) could be involved in formation of the pathways for chromate ion transport . Polymer films containing BaSO4 particles with different pigment volume concentration (PVC) were exposed to chromate solution allowing migration of chromate species.

Detection and quantification of low molecular weight components in polymericsamples via nuclear magnetic resonance (NMR) spectroscopy can be difficultdue to overlapping signal caused by line broadening characteristics ofpolymers. A way of overcoming this problem could be the exploitation of thedifference in relaxation between small molecules and macromolecular species,such as the application of a T2 filter by using the Carr–Purcell–Meiboom–Gill(CPMG) spin-echo pulse sequence. This technique, largely exploited in metabolomicsstudies, is applied here to material sciences. A Design of Experimentsapproach was used for evaluating the effect of different acquisitionparameters (relaxation delay, echo time and number of cycles) and sample-relatedones (concentration and polymer molecular weight) on selectedresponses, with a particular interest in performing a reliable quantitative analysis.Polymeric samples containing small molecules were analysed by NMRwith and without the application of the filter, and analysis of variance wasused to identify the most influential parameters. Results indicated that increasingthe polymer concentration, hence sample viscosity, further attenuates polymersignals in CPMG experiments because the T2 of those signals tends todecrease with increasing viscosity. The signal-to-noise ratio measured for smallmolecules can undergo a minimum loss when specific parameters are chosenin relation to the polymer molecular weight. Furthermore, the difference indynamics between aliphatic and aromatic nuclei, as well as between mobileand stiff polymers, translates into different results in terms of polymer signal reduction, suggesting that the relaxation filter can also be used for obtaininginformation on the polymer structure.

Presbyopia is a common eye disorder among aged people which is attributed to the loss of accommodation of the crystalline lens due to the increasing stiffness. One of the potential techniques to correct presbyopia involves removing the lens substance inside the capsule and replacing it with an artificial lens. The development of such devices, e.g., accommodating intraocular lenses (AIOLs), relies on the understanding of the biomechanical behaviour of the lens capsule and the essential design verification ex vivo. To mimic the eye’s dynamic focusing ability (accommodation), an artificial lens capsule (ALC), from silicone rubber accompanied by a lens radial stretching system (LRSS) was developed. The ALC was manufactured to offer a dimension and deforming behaviour replicating the human lens capsule. The LRSS was calibrated to provide a radial stretch simulating the change of diameter of capsules during accommodating process. The biomechanical function of the ALC was addressed by studying its evolution behaviour and reaction force under multiaxial stretch from the LRSS. The study highlighted the convenience of this application by performing preliminary tests on prototypes of ophthalmic devices (e.g., AIOLs) to restore accommodation.

Objective The addition of residual oils such as palm fibre oil (PFO) and sludge palm oil (SPO) to crude palm oil (CPO) can be problematic within supply chains. PFO is thought to aggravate the accumulation of monochloropropanediols (MCPDs) in CPO, whilst SPO is an acidic by-product of CPO milling and is not fit for human consumption. Traditional targeted techniques to detect such additives are costly, time-consuming and require highly trained operators. Therefore, we seek to assess the use of gas chromatography–ion mobility spectrometry (GC–IMS) for rapid, cost-effective screening of CPO for the presence of characteristic PFO and SPO volatile organic compound (VOC) fingerprints. Results Lab-pressed CPO and commercial dispatch tank (DT) CPO were spiked with PFO and SPO, respectively. Both additives were detectable at concentrations of 1% and 10% (w/w) in spiked lab-pressed CPO, via seven PFO-associated VOCs and 21 SPO-associated VOCs. DT controls could not be distinguished from PFO-spiked DT CPO, suggesting these samples may have already contained low levels of PFO. DT controls were free of SPO. SPO was detected in all SPO-spiked dispatch tank samples by all 21 of the previously distinguished VOCs and had a significant fingerprint consisting of four spectral regions. The distributions of nitrate, nitrite and ammonium at various monitoring sites across the Humber basin (area 24 000 km2) were examined within a Geographical Information System (GIS) framework. This basin contains diverse characteristics, from areas of high population and industry to rural and arable regions. The Humber River is a major provider of and nutrient fluxes to the North Sea from the UK. Within the GIS analysis, the distributions of mean and mean flow weighted concentrations, flux and flux per unit area, were investigated. Empirical relationships between land characteristics and water quality for the whole catchment draining to each water quality monitoring site were established. Thirty-eight catchments were chosen for this analysis, with areas ranging from 46 km2 to 8225 km2. These catchments are distributed across the Humber, encompassing the different conditions across the basin, thus allowing relationships between water quality and catchment characteristics to be used to estimate the nitrogen concentrations and flux throughout the basin river network. The main water quality data source was the Land Ocean Interaction Study (LOIS) dataset. The Environment Agency of England and Wales water quality datasets were used to infill areas of sparse LOIS monitoring network density within the Humber. The work shows the feasibility of estimating nitrate and, to a lesser extent, nitrite and ammonium concentrations and fluxes across the river network based on land characteristics, using a GIS methodology. The estimations work particularly well for the main river channels. However, there are local anomalies which are more difficult to predict. Maps showing concentration variations at 500 m intervals along the Humber basin river networks are presented; these are of particular value for environmental managers and socio-economists. Contains fulltext : 161386.pdf (Publisher’s version ) (Open Access) Historically, advances in the field of ion mobility spectrometry have been hindered by the variation in measured signals between instruments developed by different research laboratories or manufacturers. This has triggered the development and application of chemometric techniques able to reveal and analyze precious information content of ion mobility spectra. Recent advances in multidimensional coupling of ion mobility spectrometry to chromatography and mass spectrometry has created new, unique challenges for data processing, yielding high-dimensional, megavariate datasets. In this paper, a complete overview of available chemometric techniques used in the analysis of ion mobility spectrometry data is given. We describe the current state-of-the-art of ion mobility spectrometry data analysis comprising datasets with different complexities and two different scopes of data analysis, i.e. targeted and non-targeted analyte analyses. Two main steps of data analysis are considered: data preprocessing and pattern recognition. A detailed description of recent advances in chemometric techniques is provided for these steps, together with a list of interesting applications. We demonstrate that chemometric techniques have a significant contribution to the recent and great expansion of ion mobility spectrometry technology into different application fields. We conclude that well-thought out, comprehensive data analysis strategies are currently emerging, including several chemometric techniques and addressing different data challenges. In our opinion, this trend will continue in the near future, stimulating developments in ion mobility spectrometry instrumentation even further.

This work reports the analysis of the time-resolved photoluminescence behaviour on the nanosecond and microsecond time scale of fourteen historical and contemporary titanium white pigments. The pigments were produced with different production methods and post-production treatments, giving rise to a remarkable variability of titanium dioxide powders and, in some cases, to the formation of a complex surface of the crystal agglomerates. The pigments have been further characterized by Raman spectroscopy, scanning transmission electron microscopy coupled with energy dispersive X-ray spectroscopy and inductively coupled plasma atomic emission spectrometry. Our study provides a clear view of the main features of the photoluminescence (PL) emission of anatase- and rutile-based pigments. For both the polymorphs of titanium dioxide the room-temperature photoluminescence emission is complex and involves different relaxation paths, related to shallow levels close to the conduction bands and mid-gap trap states. The PL behaviour appears to be little affected by post-production treatments such as organic and inorganic coatings. Instead, the presence of niobium impurities in the TiO2 crystal lattice, as residues of the sulphate synthesis process, induce a remarkable quenching of the visible emission of anatase-based pigments. We confirm that rutile-based and anatase-based pigments are significantly different in terms of photoluminescence behaviour. This clear distinction is a valuable point for non-invasive pigment identification by in-situ photoluminescence spectroscopy. In particular, while many organic binding media emit in the visible region, the near-infrared emission of rutile is specific and can likely be used to identify the pigment in more complex materials as paints. This research paves the way to future studies of the photo-physical properties of titanium white pigments, which is imperative to understand the risk of degradation induced by the well-known photocatalytic activity of this widely used 20th century pigment.

CINAMMON is a phase IV, open-label, single-arm, pilot study assessing maraviroc (MVC) in the central nervous system (CNS) when added to darunavir/ritonavir monotherapy (DRV/r) in virologically suppressed HIV-infected subjects. CCR5 tropic participants on DRV/r were recruited. Participants remained on DRV/r for 12 week (w) (control phase). MVC 150 mg qd was added w12-w36 (intervention phase). Lumbar puncture (LP) and neurocognitive function (Cogstate) examinations scheduled at baseline, w12 and w36; MRI before w12, again at w36. Primary endpoint was CSF inflammatory marker changes during intervention phase. Secondary endpoints included changes in NC function and MRI parameters. CSF/plasma DRV/r concentrations measured at w12 and w36, MVC at w36. Nineteen patients recruited, 15 completed (17M, 2F). Dropouts: headache (2), knee problem (could not attend, 1), personal reasons (1). Mean age (range) 45.4 years (27.2-65.1), 13/19 white, 10/19 MSM. No changes in selected CSF markers were seen w12-w36. Overall NC function did not improve w12-w36: total age adjusted z score improved by 0.27 (weighted paired t test; p = 0.11); for executive function only, age adjusted z score improved by 0.54 (p = 0.03). MRI brain parameters unchanged. DRV plasma:CSF concentration ratio unchanged between w12 (132) and w36 (112; p = 0.577, Wilcoxon signed-rank). MVC plasma:CSF concentration ratio was 35 at w36. No changes in neuroinflammatory markers seen. In this small study, addition of 24w MVC 150 mg qd to stable DRV/r monotherapy showed possible improvement in executive function with no global NC effect. Learning effect cannot be excluded. This effect should be further evaluated.

Glycine levels are inversely associated with branched-chain amino acids (BCAAs) and cardiometabolic disease phenotypes, but biochemical mechanisms that explain these relationships remain uncharted. Metabolites and genes related to BCAA metabolism and nitrogen handling were strongly associated with glycine in correlation analyses. Stable isotope labeling in Zucker fatty rats (ZFRs) shows that glycine acts as a carbon donor for the pyruvate-alanine cycle in a BCAA-regulated manner. Inhibition of the BCAA transaminase (BCAT) enzymes depletes plasma pools of alanine and raises glycine levels. In high-fat-fed ZFRs, dietary glycine supplementation raises urinary acyl-glycine content and lowers circulating triglycerides but also results in accumulation of long-chain acyl-coenzyme As (acyl-CoAs), lower 5' adenosine monophosphate-activated protein kinase (AMPK) phosphorylation in muscle, and no improvement in glucose tolerance. Collectively, these studies frame a mechanism for explaining obesity-related glycine depletion and also provide insight into the impact of glycine supplementation on systemic glucose, lipid, and amino acid metabolism.

The chemical nature and composition of the growth medium of monocrystalline (MC) diamonds is still a matter of debate, partially because carbonate-bearing high-density fluids (HDFs) that are common in fibrous diamonds have not been found in MC diamonds. Here we report the first finding of HDF microinclusions in a MC octahedral diamond from Finsch, South Africa and in the MC octahedral core of a coated diamond from Kankan, Guinea; both diamonds carry nitrogen in B-centers. Numerous microinclusions in diamond Finsch_2a_cap1 are restricted to two thin layers parallel to the (1 1 1) face, ∼20 and 200 μm from the diamond rim. Low-Mg carbonatitic HDFs are found along the inner layer while the outer layer trapped saline compositions. The major and trace element compositions of the inclusions and their infrared spectra are highly similar to those of microinclusions found in fibrous diamonds. A few isolated microinclusions of saline compositions are scattered around a sulfide inclusion in the center of the octahedral core of diamond ON-KAN-383. This evidence for the involvement of oxidized fluids in the formation of MC diamonds adds to previous reports on the antiquity of HDFs in fibrous diamonds, the presence of carbonate and halide phases in inclusions in MC diamonds and the similarity of trace element pattern of a MC diamond to those of low-Mg carbonatitic HDF in fibrous diamonds. In addition, we show that the interaction of HDFs with depleted garnets can produce sinusoidal REE patterns which are one of the primary features of lherzolitic and harzburgitic garnet inclusions in MC diamonds. Together, these observations suggest that HDFs are involved in the formation of many types of diamonds from the Archaean to the Phanerozoic. HDFs are trapped in large quantities during rapid, fibrous growth, but must also be present during the growth of many MC diamonds.

Peptides have a dominant role in biology; yet the study of their physical properties is at best sporadic. Peptide quantitative structure-activity relationship (QSAR) lags far behind the QSAR analysis of drug-like organic small molecules. Traditionally, QSAR has focussed on experimentally determined partition coefficients as the main descriptor of hydrophobicity. A partition coefficient () is the ratio between the concentrations of an uncharged chemical substance in two immiscible phases: most typically water and an organic solvent, usually 1-octanol. A distribution coefficient () is the equivalent ratio for charged molecules. We report here a compilation of partition and distribution coefficients for linear peptides compiled from literature reports, suitable for the development and benchmarking of peptide and prediction algorithms.

The temporal co-ordination of ribulose 1·5-bisphosphate carboxylase/oxygenase (Rubisco) and phosphoenolpyruvate carboxylase (PEPc) activities by Mesembryanthemum crystallinum L. in C(3) and crassulacean acid metabolism (CAM) modes was investigated under conventional light-dark (LD) and continuous light (LL) conditions. When C(3) , net CO(2) assimilation rate increased during each subjective night under LL with maximum carboxylation unrelated to Rubisco activation state. The CAM circadian rhythm of CO(2) uptake was more pronounced, with CO(2) assimilation rate maximal towards the end of each subjective night. In vivo and in vitro techniques were integrated to map carboxylase enzyme regulation to the framework provided by CAM LL gas exchange activity. Rubisco was activated in vitro throughout each subjective dark period and consistently deactivated at each subjective dawn, similar to that observed at true dawn in constitutive CAM species. Instantaneous carbon isotope discrimination showed in vivo carboxylase co-dominance during the CAM subjective night, initially by Rubisco and latterly C(4) (PEPc), despite both enzymes seemingly activated in vitro. The circadian rhythm in titratable acidity accumulation was progressively damped over successive subjective nights, but maintenance of PEPc carboxylation capacity ensures that CAM plants do not become progressively more 'C(3) -like' with time under LL.

Ion mobility Spectrometry is used to detect volatile analytes within human breath directly. Many volatile organic compounds (VOC) show significant day-to-day variation in the signal height related to the concentration of the analyte, although the breath collection had been performed under the same conditions with respect to similar sampling procedure, similar dead volume, similar measurement time, and measurement conditions. Variations of 8 different analytes are investigated over a time period of 11 months in the exhaled breath of the same person in the same room environment. The individual variability is reported for Benzothiazole; D-Limonene; Eucalyptol; Decamethylcyclopentasiloxane; Decanal; 1-Hexanol, 2-ethyl-; Cyclohexanone, 5-methyl-2-(1-methylethyl) and Nonanal. The paper shows, that the individual variability must be taken into consideration to relate the findings to medical questions. Therefore, the room air concentration of VOCs must be taken into account, so that the difference between exhaled and inhaled air has to be used as indicator. Finally, starting with individual variabilities, the normal variation related to the specific analyte should be considered in addition.

The engineering of ion mobility spectrometers has made significant advances in recent years with new levels of sensitivity being achieved which facilitate their use, for example, in the medical analysis of trace components in human breath. In the study reported here direct measurements of 10 mL of exhaled air were carried out using ion mobility spectrometry (IMS) and the influence of the ambient air composition on the validation of potential biomarkers carefully considered. Changes in the IMS-signals observed within a room used for clinical studies were analysed in the absence of clinical staff. A time series of one measurement every hour of ambient air was referenced against clean bottled synthetic air for comparison using two different IMS instruments equipped with multi-capillary columns (MCC). 26 different peaks were identified and selected for the signal intensity monitoring during 1 week. Some peaks show no changes, while certain peaks varied with time in a seemlingly random manner. Correlated peak intensity changes were observed as well as changes apparently relating to variations of the ventilation system during day and night cycling. It is important to fully understand these variation if incorrect conclusions are to be avoided during, for example, the important hunt for validatable biomarkers. These results will be applicable not only to IMS studies but to all experimental design strategies in this fields regardless of the selected instrumentation. As an example it is possible to cite a particular time series where a particular peak observed within the IMS-Chromatogram shows an analyte intensity decrease within the room air between the evening and the next morning of 75%. On a subsequent day the increase between the morning and the evening was observed by a factor of about 3. During 4 days, setting the mean value to 100%, an increase of up to 173% and the decrease of up to 21% were observed. In case of measurements near to the detection limit of the method of investigation, the influence of the surrounding inhaled air must be considered carefully. The daily changes of the concentrations of specific analytes in the ambient air should be taken into account in addition to the measurement of the analyte concentrations taken from the exhaled air of a patient. Recommendations will be made covering more robust validation strategies which include near-simultaneous background ambient air measurements alongside synthetic air measurements in clinical studies. Care should be taken to avoid unnecessary changes to the ambient room air during measurements such as caused by doors opening during the entire measurement time. Ambient air measurements should be included with sampling on the patient and the surrounding air, before and after measurement of the patient breath.

Motivation: There is much interest in reducing the complexity inherent in the representation of the 20 standard amino acids within bioinformatics algorithms by developing a so-called reduced alphabet. Although there is no universally applicable residue grouping, there are numerous physiochemical criteria upon which one can base groupings. Local descriptors are a form of alignment-free analysis, the efficiency of which is dependent upon the correct selection of amino acid groupings. Results: Within the context of G-protein coupled receptor (GPCR) classification, an optimization algorithm was developed, which was able to identify the most efficient grouping when used to generate local descriptors. The algorithm was inspired by the relatively new computational intelligence paradigm of artificial immune systems. A number of amino acid groupings produced by this algorithm were evaluated with respect to their ability to generate local descriptors capable of providing an accurate classification algorithm for GPCRs. Epitopes mediated by T cells lie at the heart of the adaptive immune response and form the essential nucleus of anti-tumour peptide or epitope-based vaccines. Antigenic T cell epitopes are mediated by major histocompatibility complex (MHC) molecules, which present them to T cell receptors. Calculating the affinity between a given MHC molecule and an antigenic peptide using experimental approaches is both difficult and time consuming, thus various computational methods have been developed for this purpose. A server has been developed to allow a structural approach to the problem by generating specific MHC:peptide complex structures and providing configuration files to run molecular modelling simulations upon them. A system has been produced which allows the automated construction of MHC:peptide structure files and the corresponding configuration files required to execute a molecular dynamics simulation using NAMD. The system has been made available through a web-based front end and stand-alone scripts. Previous attempts at structural prediction of MHC:peptide affinity have been limited due to the paucity of structures and the computational expense in running large scale molecular dynamics simulations. The MHCsim server (http://igrid-ext.cryst.bbk.ac.uk/MHCsim) allows the user to rapidly generate any desired MHC:peptide complex and will facilitate molecular modelling simulation of MHC complexes on an unprecedented scale.

Current challenges of clinical breath analysis include large data size and non-clinically relevant variations observed in exhaled breath measurements, which should be urgently addressed with competent scientific data tools. In this study, three different baseline correction methods are evaluated within a previously developed data size reduction strategy for multi capillary column - ion mobility spectrometry (MCC-IMS) datasets. Introduced for the first time in breath data analysis, the Top-hat method is presented as the optimum baseline correction method. A refined data size reduction strategy is employed in the analysis of a large breathomic dataset on a healthy and respiratory disease population. New insights into MCC-IMS spectra differences associated with respiratory diseases are provided, demonstrating the additional value of the refined data analysis strategy in clinical breath analysis.

SummaryAcylcarnitine metabolites have gained attention as biomarkers of nutrient stress, but their physiological relevance and metabolic purpose remain poorly understood. Short-chain carnitine conjugates, including acetylcarnitine, derive from their corresponding acyl-CoA precursors via the action of carnitine acetyltransferase (CrAT), a bidirectional mitochondrial matrix enzyme. We show here that contractile activity reverses acetylcarnitine flux in muscle, from net production and efflux at rest to net uptake and consumption during exercise. Disruption of this switch in mice with muscle-specific CrAT deficiency resulted in acetyl-CoA deficit, perturbed energy charge, and diminished exercise tolerance, whereas acetylcarnitine supplementation produced opposite outcomes in a CrAT-dependent manner. Likewise, in exercise-trained compared to untrained humans, post-exercise phosphocreatine recovery rates were positively associated with CrAT activity and coincided with dramatic shifts in muscle acetylcarnitine dynamics. These findings show acetylcarnitine serves as a critical acetyl buffer for working muscles and provide insight into potential therapeutic strategies for combatting exercise intolerance.

Antigenic peptide is presented to a T-cell receptor through the formation of a stable complex with a Major Histocompatibility Complex (MHC) molecule. Various predictive algorithms have been developed to estimate a peptide’s capacity to form a stable complex with a given MHC Class II allele, a technique integral to the strategy of vaccine design. These have previously incorporated such computational techniques as quantitative matrices and neural networks. We have developed a novel predictive technique that uses molecular modeling of predetermined crystal structures to estimate the stability of an MHC Class II peptide complex. This is the 1st structure-based technique, as previous methods have been based on binding data. ROC curves are used to quantify the accuracy of the molecular modeling technique. The novel predictive technique is found to be comparable with the best predictive software currently available.

A liquid metal ion source (LMIS) based upon a gold/germanium eutectic has been developed. The LMIS emits a variety of ions including monatomic gold and gold clusters. Gold ions have been utilised for SIMS analysis of the polypeptide gramicidin and the polymer poly(ethylene-terepthalate) (PET). It has been found that monatomic gold (Au + ) increases secondary ion yields up to a factor of four compared to gallium, for both gramicidin and PET. The Au 3 + cluster produces a strong non-linear increase in yield over monatomic gold, for both gramicidin and PET. This effect is greatest at high mass, the yield for the gramicidin molecular ion increasing by a factor of over 60. No evidence has been found to suggest increased fragmentation as a result of cluster ion bombardment. The LMIS also exhibits good static SIMS imaging capacity.

Ion mobility spectrometry combined with multicapillary column separation (MCC-IMS) is a well-known technology for detecting volatile organic compounds (VOCs) in gaseous samples. Due to their large data size, processing of MCC-IMS spectra is still the main bottleneck of data analysis, and there is an increasing need for data analysis strategies in which the size of MCC-IMS data is reduced to enable further analysis. In our study, the first untargeted chemometric strategy is developed and employed in the analysis of MCC-IMS spectra from 264 breath and ambient air samples. This strategy does not comprise identification of compounds as a primary step but includes several preprocessing steps and a discriminant analysis. Data size is significantly reduced in three steps. Wavelet transform, mask construction, and sparse-partial least squares-discriminant analysis (s-PLS-DA) allow data size reduction with down to 50 variables relevant to the goal of analysis. The influence and compatibility of the data reduction tools are studied by applying different settings of the developed strategy. Loss of information after preprocessing is evaluated, e.g., by comparing the performance of classification models for different classes of samples. Finally, the interpretability of the classification models is evaluated, and regions of spectra that are related to the identification of potential analytical biomarkers are successfully determined. This work will greatly enable the standardization of analytical procedures across different instrumentation types promoting the adoption of MCC-IMS technology in a wide range of diverse application fields.

The interpretation of the results of pyrolysis studies of humic substances is fraught with difficulties if only a single analytical technique is employed for substructure analyses. In this paper we have carried out investigations into humic substances utilising the additional information from the combination of gas chromatography/infrared spectroscopy and gas chromatography/mass spectrometry detection following Curie-point pyrolysis at five different temperatures. The results were compared to those obtained from conventional flash pyrolysis. The aim was to ensure the validation of the results of the compounds identified by at least one other spectroscopic technique. Some 30 substances have been positively identified by infrared spectroscopy, although their appearance is dependent on pyrolysis method and temperature. Most assignments were validated by mass spectrometry or the substance class confirmed.

Infrared attenuated total reflectance spectroscopy has been assessed for the analysis of extra virgin olive oil samples from various Mediterranean sites and their adulteration by sunflower oil. In this study two different silver halide fiber-optic probes were separately tested for the mid-infrared spectroscopic measurement of pure olive oil samples and these same oils adulterated with sunflower oil. One fiber-optic probe contained an exchangeable U-shaped section of the silver halide fiber, whereas the second probe consisted of a fiber-coupled diamond crystal, which performed slightly less well than the whole fiber probe. The optimum standard error of prediction for the sunflower oil concentrations in spiked olive oil samples, obtained by optimized partial least-squares (PLS) calibration models and leave-one-out cross-validation, was 1.2% by weight with the use of a special variable selection strategy based on a pairwise consideration of significant respective minima and maxima of the optimum PLS regression vector, calculated for broad spectral intervals. Calibration robustness was proven by also using packages of 10 randomly selected samples within a further cross-validation for calculating independent prediction values. The implications for product monitoring are discussed.

The relatively young field of ion mobility spectrometry has now advanced to the stage where the need to reliably exchange the spectroscopic data obtained worldwide by this technique has become extremely urgent. To assist in the validation of the various new spectrometer designs and to assist in inter-comparisons between different laboratories reference data collections are being established for which an internationally recognized electronic data exchange format is essential. To make the data exchange between users and system administration possible, it is important to define a file format specially made for the requirements of ion mobility spectrometry. The format should be computer readable and flexible enough for extensive comments to be included. In this document, we define a data exchange format, agreed on by a working group of the International Society for Ion Mobility Spectrometry at Hilton Head Island, USA (1998) and Buxton, UK (1999). This definition of this format is based on the IUPAC JCAMP-DX protocols, which were developed for the exchange of infrared spectra [1] and extended to chemical structures [2], nuclear magnetic resonance data [3], and mass spectra [4]. This standard of the Joint Committee on Atomic and Molecular Physical Data is of a flexible design. The International Union of Pure and Applied Chemistry have taken over the support and development of these standards and recently brought out an extension to cover year 2000 compatible date strings and good laboratory practice [5]. The aim of this paper is to adapt JCAMP-DX to the special requirements of ion mobility spectra [6].

The electrochemical oxidation of N,N-dimethylphenylenediamine at glassy carbon electrodes in aqueous solution in the pH range 3-6 is shown to lead to currents which are considerably amplified in the presence of dissolved hydrogen sulfide. The mechanism of the electrode process is investigated and the reaction is shown to provide an attractive basis for the amperometric sensing of sulfide with an accessible range of ca. 3 to 150 muM (0.1 to 4.8 ppm).

A tailor-designed acoustic trap has been built and tested as a sample handling device for use in laser spectroscopy. It is based on a piezoelectric vibrator that sets up a standing acoustic wave with several nodes into which liquid or solid samples may be placed and held in a levitated position. The usefulness of such a system to hold samples without the need for sample cells was demonstrated for various experiments involving direct spectroscopic measurements on the suspended sample. Changes in the UV spectrum of bromocresol green dye during a simple pH titration on a drop of the levitated solution were followed by dual dye laser spectrometry. A chemical actinometry spectrochemistry experiment was conducted on Aberchrome 540 fulgide dissolved in DMSO in the levitated state by following changes in the molecular structure of the fulgide by Raman spectroscopy. Finally, crystallization experiments were carried out on droplets and bubbles of tris-(hydroxymethyl)-amino-methane and l-cystein.

Determination of the authenticity of extra virgin olive oils has become more important in recent years following some infamous adulteration and contamination scandals. There is significant economic advantage to be gained by falsely marketing lower quality oils as top-quality products or by adulterating the end product with cheaper alternative oil. The analysis of olive oils is complicated by their complex nature. The conventional analytical techniques involve complicated analyses of individual components in the oil in order to determine their authenticity; however, molecular spectroscopy combined with modern chemometric techniques offers a rapid analytical solution to this complex problem. This paper describes the initial results from the experimental application of Fourier transform Raman spectroscopy in this area.

The aims of this study were, to use agonists selective for the 3 mGlu receptor groups to identify developmental changes in their effects, and to assess the usefulness of proposed selective antagonists as pharmacological tools. Hippocampal slices (400 μm) were prepared from neonate (9–14 days) and young adult (5–7 weeks) Sprague-Dawley rats. Field excitatory postsynaptic potentials (fEPSP) were recorded from CA1. DHPG (100 μM), a group I agonist, produced a slowly developing enhancement of fEPSP slope in slices from adults. In slices from neonates, DHPG (75 μM) depressed fEPSP slope. DCG-IV (500 nM), a group II agonist, did not affect the fEPSP recorded from slices from adults whereas perfusion in neonate slices produced a sustained depression. The group III agonist L-AP4 (50 μM) was ineffective in adult slices but depressed fEPSP slope in slices prepared from neonates. DHPG-induced depression of fEPSP slope was inhibited by 4-CPG (400 μM), a group I antagonist, but was unaffected by MCCG (500 μM) and MAP4 (500 μM), group II and III receptor antagonists respectively. MCCG but not MAP4 antagonized the effects of DCG-IV with 4-CPG producing variable effects. The effect of L-AP4 was unaffected by MCCG, blocked by MAP4, and enhanced by 4-CPG. The results show that the effects of the agonists for all groups of mGlu receptors are developmentally regulated. Furthermore, MCCG and MAP4 behave as effective and selective antagonists for group II and group III mGlu receptors respectively, whereas the usefulness of 4-CPG as a group I antagonist may be limited. Porphyromonas gingivalis, a black-pigmenting anaerobe implicated in the aetiology of periodontal disease, contains two loci, rgpA and rgpB, encoding the extracellular Arg-X specific proteases (RGPs, Arg-gingipains), and kgp, which encodes a Lys-X specific protease (KGP, Lys-gingipain). The rgpA and kgp genes encode polyproteins comprising pro-peptide and catalytic domain with large N- and C-terminal extensions which require proteolytic processing at several Arg and Lys residues to generate mature enzymes. The product of rgpB contains only a pro-peptide and the catalytic domain which requires processing at an Arg residue to generate active enzyme. An rgpA rgpB double mutant (E8) of P. gingivalis was constructed to study the role of RGPs in the processing of KGP. A kgp mutant (K1A) was also studied to investigate the role of KGP in the generation of RGPs. E8 was stable in the absence of the antibiotics tetracycline and clindamycin (selection markers for rgpA and rgpB, respectively) and exhibited the same pigmentation, colony morphology and identical growth rates to the parent W50 strain in the absence of antibiotics, in both complex and chemically defined media. The KGP activity of E8, grown in the absence of tetracycline, in whole cultures and in culture supernatants (up to 6 d) was identical to levels in W50. However, in the presence of tetracycline in the growth medium, the level of KGP was reduced to 50% of levels present in whole cultures of W50. Since tetracycline had no effect on RGP or KGP activity when incorporated into assay buffer, this effect is most likely to be on the synthesis of Kgp polypeptide. K1A was also stable in the absence of antibiotics but was unable to pigment, and remained straw-coloured throughout growth. RGP activity in whole cultures of K1A was identical to levels in W50, but RGP activity in 6 d culture supernatants was reduced to 50% of levels present in W50. Thus, although KGP is not required for generation of RGP activity from RgpA and RgpB polypeptides, its absence affects the release/transport of RGP into culture supernatant.

OBJECTIVES Adult GH deficiency (GHD) is associated with profound alterations in body composition, lipid profiles and quality of life which frequently improve after GH therapy. However, the beneficial effects of treatment are not derived by all and consequently some scepticism persists with regard to the use of GH therapy in adults. We assessed whether a 3-month therapeutic assessment with GH therapy could be used to determine which GHD adults should be treated over the longer term. We also assessed the continued prescription of GH by general practitioners (GPs) following the initial therapeutic assessment. DESIGN A three month open therapeutic trial of GH in GHD adults. Patients were treated with GH at an initial dose of 0.01 iU/kg/d, increased after 1 month to 0.015 iU/kg/d for males and 0.02 iU/kg/d for females. After completion of the three months the continued prescription of GH by the GPs was assessed. PATIENTS All adult GHD patients were considered for GH therapy. Thirty-nine GHD adults wanted GH therapy (group 1) and their baseline characteristics such as age, duration of GHD, and IGF-1 concentration were compared with 24 subjects who declined to receive GH (group 2). MEASUREMENTS Measurements of body composition using bioelectrical impedance analysis, lipids and quality of life measured using a dedicated questionnaire were made before and after GH therapy. The response of the general practitioners to continued GH therapy after the initial therapeutic assessment was also noted. _The rapid development of the Internet as a major communication tool between scientists has led to the need for a co-ordinated IUPAC presence. Many diverse groups have already initiated distribution of IUPAC related material via their Web sites. These guidelines will provide the structure on which the official IUPAC Internet site maintained through the Secretariat will be based. Rules governing the interaction between this central site and various sites operated by other IUPAC bodies are published here as well as guidelines for the operation of sites maintained by other bodies which contain IUPAC related information. Version 5.00 of the JCAMP-DX specifications were published for NMR and Mass Spectrometry file formats in Appl. Spectrosc.47, 1093-1099, (1993) and Appl. Spectrosc.48, 1545-1552, (1994). Since publication of these protocols developments in spectroscopy have led to a large number of requests for additions for applications not originally covered. Following careful consideration, it has become apparent that a few minor modifications will significantly increase the range of possible applications.In addition, new data labels have been introduced to ensure that files are year 2000 compliant and allow for conformity with good laboratory practices (GLP). These modifications are detailed in this publication as well as examples of the official NTUPLE JCAMP-DX definition as applied to NMR data.

Analytical spectroscopists must be computer literate to effectively carry out the tasks assigned to them. This has often been resisted within organizations with insufficient funds to equip their staff properly, a lack of desire to deliver the essential training and a basic resistance amongst staff to learn the new techniques required for computer assisted analysis. In the past these problems were compounded by seriously flawed software which was being sold for spectroscopic applications. Owing to the limited market for such complex products the analytical spectroscopist often was faced with buying incomplete and unstable tools if the price was to remain reasonable. Long product lead times meant spectrometer manufacturers often ended up offering systems running under outdated and sometimes obscure operating systems. Not only did this mean special staff training for each instrument where the knowledge gained on one system could not be transferred to the neighbouring system but these spectrometers were often only capable of running in a stand-alone mode, cut-off from the rest of the laboratory environment. Fortunately a number of developments in recent years have substantially changed this depressing picture. A true multi-tasking operating system with a simple graphical user interface, Microsoft Windows NT4, has now been widely introduced into the spectroscopic computing environment which has provided a desktop operating system which has proved to be more stable and robust as well as requiring better programming techniques of software vendors. The opening up of the Internet has provided an easy way to access new tools for data handling and has forced a substantial re-think about results delivery (for example Chemical MIME types, IUPAC spectroscopic data exchange standards). Improved computing power and cheaper hardware now allows large spectroscopic data sets to be handled without too many problems. This includes the ability to carry out chemometric operations in minutes rather than hours. Fast networks now enable data analysis of even multi-dimensional spectroscopic data sets remote from the measuring instrument. A strong tendency to opt for a more unified graphical user interface which is substantially more user friendly allows even inexperienced users to rapidly get acquainted with even the complex mathematical analyses. Some examples of new spectroscopic software products will be given to demonstrate the aforesaid points and highlight the ease of integration into a modern analytical spectroscopy workplace.

Automated NMR spectra interpretation packages are becoming increasingly important especially now that high-throughput synthesis often vastly increases the amount of data to be analyzed in a given time frame. It is vitally important for these interpretation packages to be robust, and this means that it essential to know how the quality of NMR data varies is real life experiments. A practical test concerning the reproducibility of chemical shift information from recorded 13C NMR spectra was carried out using eight test substances. Two hundred seventy spectra were contributed from both Academia and Industry and have been subjected to a standardized interpretation. For these spectra the confidence limits of their chemical shifts have been calculated. The standard deviations found vary from 0.2 up to about 2.5 ppm, and the range of values for the shifts reported vary from ±0.7 up to more than ±7 ppm around the respective average shift. We have previously shown that coapplication of arachidonic acid (10 microM) and (1S,3R)-1-aminocyclopentane-1,3-dicarboxylic acid (ACPD, 50 microM) evokes an enhancement of synaptic transmission in the CA1 region of the rat hippocampal slice. Here we have investigated whether the metabolites of arachidonic acid are implicated in this potentiation. Inclusion of the cyclo-oxygenase inhibitor indomethacin (10 microM) did not block the potentiation induced by coapplication of arachidonic acid and ACPD. However, the presence of either the cyclo-, lipo- and epoxygenase inhibitor 5,8,11,14-eicosatetraynoic acid (ETYA, 20 microM), or the lipoxygenase inhibitor nordihydroguaiaretic acid (10 microM), prevented the long-lasting enhancement. The results suggest that the lipoxygenase and epoxygenase metabolites of arachidonic acid may be involved in the induction of this form of synaptic potentiation.

The binding of the herbicides to aqueous humic substances was investigated using the herbicide mecoprop and a reference humic substance 'Hohlohsee'. Initial results from 2D fluorescence spectrometry, infra-red spectroscopy and Curie-point pyrolysis gas chromatography/infra-red spectroscopy tentatively characterise the nature of the interaction.

Lipid metabolism is tightly controlled by the nutritional state of the organism. Nutrient-rich conditions increase lipogenesis whereas nutrient deprivation promotes fat oxidation. In this study, we identify the mitochondrial sirtuin, SIRT4, as a novel regulator of lipid homeostasis. SIRT4 is active in nutrient-replete conditions to repress fatty acid oxidation while promoting lipid anabolism. SIRT4 deacetylates and inhibits malonyl CoA decarboxylase (MCD), an enzyme that produces acetyl CoA from malonyl CoA. Malonyl CoA provides the carbon skeleton for lipogenesis and also inhibits fat oxidation. Mice lacking SIRT4 display elevated MCD activity and decreased malonyl CoA in skeletal muscle and white adipose tissue. Consequently, SIRT4 KO mice display deregulated lipid metabolism leading to increased exercise tolerance and protection against diet-induced obesity. In sum, this work elucidates SIRT4 as an important regulator of lipid homeostasis, identifies MCD as a novel SIRT4 target, and deepens our understanding of the malonyl CoA regulatory axis.

The NPL published a forward to the ITS-90 text as follows:- "The purpose of the ITS is to define procedures by which certain specified practical thermometers of the required quality can be calibrated in such a way that the values of temperature obtained from them can be precise and reproducible, while at the same time closely approximating the corresponding thermodynamic values." [1]. The paper investigates the properties of thirty four lots of 6N pure metal used to make cells conforming to ITS-90 from mercury through silver over a period of twenty years. Three hundred individual cells are analysed by the impurities listed and supplied with each lot, melt and freeze curve slopes are also summarised for each lot and depressions calculated. These are then compared to the slopes and depressions suggested in the Supplementary Information for the ITS-90 and in CCT/2000-13 "Optimal Realizations". Results are summarised, tabulated and discussed. Three lots of the thirty four were found to produce cells outside 6N expectations; however the remaining thirty one lots no matter how well or badly the accompanying certification was presented produced cells that conformed to 6N expectations as suggested in Supplementary Information to ITS-90 and CCT/2000-13.

Dinocap is the generic name for a mixture of 2,6-dinitro-4-(1-methylheptyl)-phenylcrotonate, 2,4-dinitro-6-(1-methylheptyl)-phenylcrotonate and some other alkyl substituted isomers [1]. The mixture is applied as a non-specific acaricide and contact fungicide, recommended for the control of all kinds of mildew on fruit, vegetables, potatoes, tobacco and seeds and is used all over the world. Studies were carried out into the identification of this pesticide and its by-products using infrared spectroscopy and mass spectrometry following chromatographic separation.

The determination of Dinocap, one of the most often used acaricides and contact fungicides, is complicated by the fact that it consists of several isomeric alkyldinitro-phenyl-crotonates. In a first step, GC/IR and GC/MS has been used to separate the mixture, to determine the chromatographic behaviour and to fully characterize the six different isomeric compounds. The structures of additional impurities in the commerical preparation were shown to be the correlating phenols. An extraction/preconcentration method has been devised based on Carbopack B, yielding recoveries between 75 and 98%. A qualified estimation of the total Dinocap content of drinking water can be made by this approach for concentrations well below the limits set by the European drinking water regulations. The development of a complete analytical procedure based on these results appears to be feasible.

JCAMP-DX has, for several years, been the standard form for the exchange of infrared spectral data. More recently JCAMP-DX protocols have been published for chemical structure data and for nuclear magnetic resonance spectroscopy. This publication presents a new JCAMP-DX data exchange protocol for mass spectrometry, covering the transport of single spectra, spectral series, and raw data files. The protocol can be implemented on any computer system and storage media. It is completely manufacturer independent. As with previous publications in this series, the aim is to provide reliable data transfer without loss of information regardless of the hardware or software involved. A comparison to the work on a binary protocol currently being carried out by the Analytical Instrument Association is also presented.

The non-linear Raman spectrum of 4-cyano-4′-heptylbiphenyl is reported. Measurements of depolarization ratios of the vibrational bands at 2224 cm−1 (CN stretching), 1606 cm−1 (aromatic ring stretching) and 1185 cm−1 (CH aromatic in-plane deformation) are used to evaluate the order parameters (〈P2〉 and 〈P4〉) using a ‘line model’ for the molecules. The results are internally consistent and also show the same general features (including some discrepancies in 〈P4〉) reported by other workers. Nevertheless, the results demonstrate that the technique is capable of wide application to the study of the anisotropic properties of liquid crystals and provides a valuable probe into the study of the biaxial nature of these sytems.

Substructure components of various aquatic humic substances were investigated by a coupled pyrolysis — gas chromatography — Fourier-transform infrared spectroscopy (Py-GC-FTIR) procedure. The humic substances studied gave similar pyrolysis products, but in varying proportions. Many of the pyrolysis products (e.g. methanol, acetone, alkylbenzenes, cyclopentane, aliphatic and aromatic organic acids, acetamide, pyrrole and phenols) could be identified by their FTIR spectra using a digital library for automatic comparison. Some of the compounds are related to lignin fragments which form a large part of the humic substances investigated. Other products give hints to the involvement of tetrapyrroles, fatty acids, furanoses and amino compounds in the structure of humic macromolecules.
