A new series of propionamide derivatives was developed as dual μ-opioid receptor agonists and σ1 receptor antagonists. Modification of a high-throughput screening hit originated a series of piperazinylcycloalkylmethyl propionamides, which were explored to overcome the challenge of achieving balanced dual activity and convenient drug-like properties. The lead compound identified, 18g, showed good analgesic effects in several animal models of both acute (paw pressure) and chronic (partial sciatic nerve ligation) pain, with reduced gastrointestinal effects in comparison with oxycodone.

The synthesis and pharmacological activity of a new series of 4-alkyl-1-oxa-4,9-diazaspiro[5.5]undecane derivatives as potent dual ligands for the σ1 receptor (σ1R) and the μ-opioid receptor (MOR) are reported. A lead optimization program over the initial 4-aryl analogues provided 4-alkyl derivatives with the desired functionality and good selectivity and ADME profiles. Compound 14u (EST73502) showed MOR agonism and σ1R antagonism and a potent analgesic activity, comparable to the MOR agonist oxycodone in animal models of acute and chronic pain after single and repeated administration. Contrary to oxycodone, 14u produces analgesic activity with reduced opioid-induced relevant adverse events, like intestinal transit inhibition and naloxone-precipitated behavioral signs of opiate withdrawal. These results provide evidence that dual MOR agonism and σ1R antagonism may be a useful strategy for obtaining potent and safer analgesics and were the basis for the selection of 14u as a clinical candidate for the treatment of pain.

In this study, we observe that the poromechanical parameters in human meniscus vary spatially throughout the tissue. The response is anisotropic and the porosity is functionally graded. To draw these conclusions, we measured the anisotropic permeability and the “aggregate modulus” of the tissue, i.e., the stiffness of the material at equilibrium, after the interstitial fluid has ceased flowing. We estimated those parameters within the central portion of the meniscus in three directions (i.e., vertical, radial and circumferential) by fitting an enhanced model on stress relation confined compression tests. We noticed that a classical biphasic model was not sufficient to reproduce the observed experimental behaviour. We propose a poroelastic model based on the assumption that the fluid flow inside the human meniscus is described by a fractional porous medium equation analogous to Darcy’s law, which involves fractional operators. The fluid flux is then time-dependent for a constant applied pressure gradient (in contrast with the classical Darcy’s law, which describes a time independent fluid flux relation). We show that a fractional poroelastic model is well-suited to describe the flow within the meniscus and to identify the associated parameters (i.e., the order of the time derivative and the permeability). The results indicate that mean values of λβ,β in the central body are λβ=5.5443×10−10m4Ns1−β, β=0.0434, while, in the posterior and anterior regions, are λβ=2.851×10−10m4Ns1−β, β=0.0326 and λβ=1.2636×10−10m4Ns1−β, β=0.0232, respectively. Furthermore, numerical simulations show that the fluid flux diffusion is facilitated in the central part of the meniscus and hindered in the posterior and anterior regions.

Co-crystal of tramadol–celecoxib (CTC) is a novel active pharmaceutical ingredient API–API co-crystal formed by an intrinsic 1:1 molecular ratio of rac-tramadol·HCl (TRM) and celecoxib (CXB) in late-stage development for the treatment of pain. In line with previous intrinsic dissolution rate studies, we report here that the kinetic dissolution profile of CTC in hydroxypropyl methyl cellulose and buffered solutions led to a supersaturation state of CXB and a release significantly faster from CTC than from the free combination, which in turn was quite similar to the release from CXB alone. Inversely, TRM was released much more slowly from CTC than from the free combination and TRM alone. Experimental and predicted solubility–pH curves showed that the thermodynamic solubility of CTC lies in between those of TRM and CXB within the physiologically meaningful pH range. Spheroids encapsulated within alginate capsules are emerging as suitable in vitro tools to investigate the impact of mechanical forces on tumor growth since the internal tumor pressure can be retrieved from the deformation of the capsule. Here we focus on the particular case of Cellular Capsule Technology (CCT). We show in this contribution that a modeling approach accounting for the triphasic nature of the spheroid (extracellular matrix, tumor cells and interstitial fluid) offers a new perspective of analysis revealing that the pressure retrieved experimentally cannot be interpreted as a direct picture of the pressure sustained by the tumor cells and, as such, cannot therefore be used to quantify the critical pressure which induces stress-induced phenotype switch in tumor cells. The proposed multiphase reactive poro-mechanical model was cross-validated. Parameter sensitivity analyses on the digital twin revealed that the main parameters determining the encapsulated growth configuration are different from those driving growth in free condition, confirming that radically different phenomena are at play. Results reported in this contribution support the idea that multiphase reactive poro-mechanics is an exceptional theoretical framework to attain an in-depth understanding of CCT experiments, to confirm their hypotheses and to further improve their design.

International audience; The kinetic energy release distribution (KERD) in the vibrational autodetachment (VAD) from sulfur hexafluoride anion SF6- has been measured in a velocity map imaging spectrometer for delays in the range of a few tens of microseconds. The experimental KERD is analyzed within the framework of the detailed-balance: first using the standard Langevin model and subsequently using a more refined and realistic model based on the experimental attachment cross section. A discussion on the processes involved in the attachment and the VAD is presented based on an empirical fit of the attachment cross section. The lifetime derived from the model is in good agreement with the experimental time window, strengthening this theoretical approach for this model system.

Summary Monosaccharide anhydrides (MAs) are routinely analysed to track modern and past biomass burning. While liquid chromatography (LC) methods are promising, MA compounds (galactosan, levoglucosan and mannosan) are typically not well separated. This shortcoming results in a cumbersome and time-consuming isotopic characterization of MAs. Here we describe novel liquid chromatography-electrospray ionization-tandem mass spectrometry and liquid chromatography-isotope ratio mass spectrometry methods for rapid analysis of MAs. We also developed a simple protocol for MA extraction and workout from air and water filters. In addition to their simplicity and the rapid analysis of MAs, our new methods have several advantages. First, we extract and separate MA compounds only with ultrapure water, which enables compound specific isotopic analysis under ideal conditions. Second, we use small solvent volumes for MA extraction to limit sample dilution. Third, we achieve baseline separation of all MAs. Fourth, we achieve detection and quantification limits as small as 50–100 pg of injected MA compounds with post-column ammonia addition and MA analysis in multiple reaction monitoring mode. Therefore, our methodological developments further demonstrate that LC methods compare favourably with gas chromatography methods for the analysis of MAs in environmental samples.

Inhibition of mTOR is the standard of care for lymphangioleiomyomatosis (LAM). However, this therapy has variable tolerability and some patients show progressive decline of lung function despite treatment. LAM diagnosis and monitoring can also be challenging due to the heterogeneity of symptoms and insufficiency of non-invasive tests. Here, we propose monoamine-derived biomarkers that provide preclinical evidence for novel therapeutic approaches. The major histamine-derived metabolite methylimidazoleacetic acid (MIAA) is relatively more abundant in LAM plasma, and MIAA values are independent of VEGF-D. Higher levels of histamine are associated with poorer lung function and greater disease burden. Molecular and cellular analyses, and metabolic profiling confirmed active histamine signaling and metabolism. LAM tumorigenesis is reduced using approved drugs targeting monoamine oxidases A/B (clorgyline and rasagiline) or histamine H1 receptor (loratadine), and loratadine synergizes with rapamycin. Depletion of Maoa or Hrh1 expression, and administration of an L-histidine analog, or a low L-histidine diet, also reduce LAM tumorigenesis. In this paper, a multi-scale technique is presented to study the mechanical behavior of nano-composites by linking the atomistic information from lower scale to the continuum model in upper scale. Molecular dynamics simulations are employed to compute the material properties of graphene/epoxy nano-composites using the COMPASS interatomic force field. In order to obtain the atomistic stress surfaces used to evaluate the mechanical properties of material in upper scale, the biaxial loading is applied to different representative volume elements. On the continuum level, the hyperelastic strain energy functions are utilized to calculate the material parameters using the hyperelastic functions from atomistic data. The stress and elasticity tensors are obtained by computing the first and second order derivatives of hyperelastic functions with reference to the components of the right Cauchy–Green deformation tensor. The stress–strain surfaces of hyperelastic functions in lower scale are used to calculate the properties of nano-composite material in upper scale. The efficiency and applicability of the proposed technique is presented through various numerical examples. It is shown that the proposed multi-scale technique is able to solve large problems within acceptable computational time, which is not possible using conventional molecular dynamics approaches.

Gene duplication generates new functions and traits, enabling evolution. Human-specific duplicated genes in particular are primary sources of innovation during our evolution although they have very few known functions. Here we examine the brain function of one of these genes (CHRFAM7A) and its product (dupα7 subunit). This gene results from a partial duplication of the ancestral CHRNA7 gene encoding the α7 subunit that forms the homopentameric α7 nicotinic acetylcholine receptor (α7-nAChR). The functions of α7-nAChR in the brain are well defined, including the modulation of synaptic transmission and plasticity underlying normal attention, cognition, learning, and memory processes. However, the role of the dupα7 subunit remains unexplored at the neuronal level. Here, we characterize that role by combining immunoblotting, quantitative RT-PCR and FRET techniques with functional assays of α7-nAChR activity using human neuroblastoma SH-SY5Y cell variants with different dupα7 expression levels. Our findings reveal a physical interaction between dupα7 and α7 subunits in fluorescent protein-tagged dupα7/α7 transfected cells that negatively affects normal α7-nAChR activity. Specifically, in both single cells and cell populations, the [Ca2+]i signal and the exocytotic response induced by selective stimulation of α7-nAChR were either significantly inhibited by stable dupα7 overexpression or augmented after silencing dupα7 gene expression with specific siRNAs. These findings identify a new role for the dupα7 subunit as a negative regulator of α7-nAChR-mediated control of exocytotic neurotransmitter release. If this effect is excessive, it would result in an impaired synaptic transmission that could underlie the neurocognitive and neuropsychiatric disorders associated with α7-nAChR dysfunction. The transcription factor Eomesodermin (EOMES) promotes IL-10 production of CD4+T-cells, which has been linked to immunosuppressive and cytotoxic activities. We detected EOMES-expressing CD4+T-cells in lymph node samples of patients with chronic lymphocytic leukemia (CLL) or diffuse large B-cell lymphoma. This was in line with an observed expansion of EOMES-positive CD4+T-cells in leukemic Eµ-TCL1 mice, a well-established model of CLL, and upon adoptive transfer of TCL1 leukemia in mice. Transcriptome and flow cytometry analyses revealed that EOMES does not only drive the transcription of IL-10, but rather controls a unique differentiation program in CD4+T-cells. Moreover, EOMES was necessary for the accumulation of a specific CD4+T-cell subset that expresses IFNγ and IL-10, as well as inhibitory receptors, like PD-1 and LAG3. T-cell transfer studies in leukopenicRag2-/-mice showed that EOMES-deficient CD4+T-cells were inferior in controlling TCL1 leukemia development compared to wildtype T-cells, even though expansion ofEomes-/-CD4+T-cells was observed. We further showed that control of TCL1 leukemia was driven by IL-10 receptor-mediated signals, asIl10rb-deficient CD4+T-cells showed impaired anti-leukemia activity. Altogether, our data suggest that IL-10 producing PD-1+CD4+T-cells contribute to CLL control in an EOMES- and IL-10R-dependent manner.

Introduction and Aim: Pirfenidone and nintedanib are anti-fibrotic approved treatments for idiopathic pulmonary fibrosis (IPF). Its efficacy and safety in other fibrotic interstitial lung diseases (ILD) is under study in clinical trials. The aim was to analyse the effect of these drugs in non-IPF progressive fibrotic patients that couldn’t be included in these trials. Methods: Multicenter retrospective study of 34 progressive PF non-IPF patients treated with anti-fibrotic drugs. Demographic, clinical data and treatment-related variables were evaluated. The reason to start off-label anti-fibrotic was disease progression with previous therapy and exclusion from clinical trials. Results: Patients with non-IPF progressive PF were: 38% unclassifiable PF (UPF), 20% familial pulmonary fibrosis (FPF), 18% CTD-ILD, 15% chronic hypersensitivity pneumonitis (CHP) and 9% pneumoconiosis. Mean age was 67±12 years, 74% were male and 47% ex-smokers. 21% of cases had usual interstitial pneumonia HRCT pattern. Mean FVC was 66%±23 and DLCO 45%±12. Co-medications were: low-dose prednisone (20), mycophenolate (5) and sirolimus (1). Twenty-four subjects (71%) received pirfenidone and ten (29%) nintedanib. After one year, a FVC decrease of 5-10% was observed in only 3 patients (1 CHP, 1 pneumoconiosis, 1 UPF), while others showed stability or improvement. Main reported adverse effects were: 20% hypertransaminasemia, 27% diarrhea and 15% weight loss, requiring 5 patients to stop therapy. Conclusion: After one year of anti-fibrotic therapy, most patients with non-IPF progressive PF showed functional stability. Clinical trials are mandatory to evaluate their potential efficacy, but these drugs are a safe option in progressive PF patients.

A chemical reaction model, consisting of two gas-phase and a surface reaction, for the deposition of copper from copper amidinate is investigated, by comparing results of an efficient, reduced order CFD model with experiments. The film deposition rate over a wide range of temperatures, 473K-623K, is accurately captured, focusing specifically on the reported drop of the deposition rate at higher temperatures, i.e above 553K that has not been widely explored in the literature. This investigation is facilitated by an efficient computational tool that merges equation-based analysis with data-driven reduced order modeling and artificial neural networks. The hybrid computer-aided approach is necessary in order to address, in a reasonable time-frame, the complex chemical and physical phenomena developed in a three-dimensional geometry that corresponds to the experimental set-up. It is through this comparison between the experiments and the derived simulation results, enabled by machine-learning algorithms that the prevalent theoretical hypothesis is tested and validated, illuminating the possible underlying dominant phenomena.

Many lung diseases lead to an increase in ventilation heterogeneity (VH). Two clinical practices for the measurement of patient VH are in vivo imaging, and the inert gas multiple breath washout (MBW). In this study computational modelling was used to compare the responses of MBW indices LCI and scond and MRI measured global and local ventilation indices, σr and σlocal, to constriction of airways in the conducting zone of the lungs. The simulations show that scond, LCI and σr behave quite similarly to each other, all being sensitive to increases in the severity of constriction, while exhibiting little sensitivity to the depth at which constriction occurs. In contrast, the local MRI index σlocal shows strong sensitivity to depth of constriction, but lowered sensitivity to constriction severity. We finish with an analysis of the sensitivity of MRI indices to grid sizes, showing that results should be interpreted with reference to the image resolution. Overall we conclude that the application of both local and global VH measures may help to classify different types of bronchoconstriction.

The replacement of acylamino by cyclic substituents in the position 4 of the pyrazolo[3,4-d]pyrimidine scaffold, led to highly active sigma-1 receptor (σ1R) ligands. Phenyl or pyrazolyl groups were the best in terms of affinity for the σ1R and the 4-(1-methylpyrazol-5-yl) derivative, 12f, was the most selective. Compound 12f is also one of the best σ1R ligands ever described in terms of lipophilic ligand efficiency, which translates into a good physicochemical and ADMET profile. In addition, 12f was identified as an antagonist of the σ1R in view of its potent antinociceptive profile in several pain models in mice.

In dam contexts, sluicing operations can be performed to reestablish sediments continuity, as proposed by the EU Water Framework Directive, as well as to preserve the reservoirs' water storage capacity. Such management permits the rapid release of high quantities of reservoir sediments through the opening of dam bottom valves. This work aims to study the impact of such operation on the evolution of environmental physicochemical conditions notably changes in dissolved metallic elements concentrations (Al, As, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Zn) through field and laboratory investigations. Results were interpreted in terms of concentrations and fluxes, and compared with data collected on an annual basis regarding both suspended matter and metallic elements. The release of high quantities of sediments (4,500tons dry weight in 24h), with concentrations representing up to 300 times the inter-annual mean suspended sediments discharge, significantly modified water parameters, notably solid/liquid (S/L) ratio, pH and redox conditions. Despite the fact that they are mainly trapped in stable phases, a clear increase of the solubilized metals content was measured, representing up to 60 times the maximum values of current exploitation. This solubilization is related to desorption phenomena from sediments through changes in chemical equilibriums as highlighted by laboratory characterizations and experiments. These chemical modifications are mainly attributed to S/L ratio variations. Indeed, the low S/L ratios (≤1.3g·L(-1)) measured in situ are typically the ones for which metals solubilization is the highest, as shown by laboratory experiments. Additional thermodynamic modeling highlighted that the decrease in pH measured during the operation favors the release of the free forms of metallic elements (Al and Cu), and decreases the OM complexation influence. These changes, either in term of physical conditions or speciation, increasing metals long term bioavailability notably during redeposition phase, may have adverse effects on aquatic biota.

Sediments play an important role on the quality of aquatic ecosystems, notably in the reservoir areas where they can either be a sink or a source of contaminants, depending on the management and hydrological conditions. The physicochemical properties of 25 surface sediments samples of a reservoir catchment (Vaussaire, Cantal, France) were studied. Results show a strong influence of dam presence, notably on the grain size and organic matter (OM) contents. The concentrations of trace metals and metalloids (As, Cd, Cr, Cu, Ni, Pb and Zn) were also measured and compared with worldwide reservoir concentrations and international sediment quality guideline levels in order to assess the intensity of the metallic contamination. Cr and Ni are the trace elements presenting the significantly highest values at the catchment scale. Enrichment Factors (EF), calculated using both local and national backgrounds, show that metals have mainly a natural origin, explaining especially the Cr and Ni values, linked with the composition of parental rocks. Unexpectedly, all the observed metal concentrations are lower in the reservoir than upstream and downstream, which might be related to the high fresh OM inputs in the reservoir, diluting the global metallic contamination. Multivariate statistical analyses, carried out in order to identify the relationship between the studied metals and sediment characteristics, tend to support this hypothesis, confirming the unusually low influence of such poorly-degraded OM on trace element accumulation in the reservoir.

Small extracellular vesicles (sEVs) are nanoparticles responsible for cell-to-cell communication released by healthy and cancer cells. Different roles have been described for sEVs in physiological and pathological contexts, including acceleration of tissue regeneration, modulation of tumor microenvironment, or premetastatic niche formation, and they are discussed as promising biomarkers for diagnosis and prognosis in body fluids. Although efforts have been made to standardize techniques for isolation and characterization of sEVs, current protocols often result in co-isolation of soluble protein or lipid complexes and of other extracellular vesicles. The risk of contaminated preparations is particularly high when isolating sEVs from tissues. As a consequence, the interpretation of data aiming at understanding the functional role of sEVs remains challenging and inconsistent. Here, we report an optimized protocol for isolation of sEVs from human and murine lymphoid tissues. sEVs from freshly resected human lymph nodes and murine spleens were isolated comparing two different approaches&mdash, (1) ultracentrifugation on a sucrose density cushion and (2) combined ultracentrifugation with size-exclusion chromatography. The purity of sEV preparations was analyzed using state-of-the-art techniques, including immunoblots, nanoparticle tracking analysis, and electron microscopy. Our results clearly demonstrate the superiority of size-exclusion chromatography, which resulted in a higher yield and purity of sEVs, and we show that their functionality alters significantly between the two isolation protocols.

We herein report the design, synthesis, and functional impact of an okadaic acid (OA) small analogue, ITH12680, which restores the activity of phosphoprotein phosphatase 2A (PP2A), whose deficient activity has been implicated in nicotine-mediated tumor progression and chemoresistance in non-small cell lung cancer (NSCLC). For its design, we paid attention to the structure of the PP2A-OA complex, where the C16-C38 OA fragment confers PP2A affinity and selectivity, but it is not involved in the inhibitory effect. Confirming this hypothesis, PP2A activity was not inhibited by ITH12680. By contrast, the compound partially restored OA-exerted PP2A inhibition in vitro. Moreover, flow cytometry and immunoblotting experiments revealed that ITH12680 reversed nicotine-induced cisplatin resistance in NSCLC cells, as it prevented nicotine-induced reduction of Bax expression and inhibited nicotine-mediated activation of cell survival and proliferation kinases, Akt and ERK1/2. The effect of 6-benzylaminopurine (6-BA) alone or in combination with naphthaleneacetic acid or indoleacetic acid on the morphogenetic response of cotyledon explants of Citrullus colocynthis (L.) Schrad. was tested. The best results were obtained with a medium containing 25 µm 6-BA, which yielded organogenic calli at a frequency of 81.8%. When these organogenic calli were transferred to elongation medium (basal medium supplemented with 0.5 µm 6-BA), 80% produced well-developed shoots. These shoots rooted normally when cultured on rooting medium containing indolebutyric acid at 2.5 or 5.0 µm. Plants grew to maturity under greenhouse conditions and gave normal fruits. Cotyledon explants were transformed by cocultivation with Agrobacterium tumefaciens LBA4404 carrying the binary vector pBI121 which bears the reporter gene β-glucuronidase (gus) and the marker gene neomycin phosphotransferase (nptII). Transformants were selected for growth capacity on medium with 100 mgl–1 of kanamycin. On the basis of β-glucuronidase expression, the transformation frequency was 14.2%. Molecular characterization by polymerase chain reaction confirmed the presence of the two genes transferred (gus, nptII) in the transgenic plants. Sexual transmission of both genes was also confirmed by studying their expression in progenies from several transgenic plants.

International audience; Four macrophytes commonly used to clean up Cu contaminated effluents, i.e. Arundo donax L., Cyperus eragrostis Lam., Iris pseudacorus L. and Phalaris arundinacea L., were assessed to produce Cu-rich plant biomass intended for ecofriendly catalyst preparation. 7-month-old plants were exposed to a Cu gradient (0.08, 2, 10, 20 and 40 mu M Cu) in batch conditions during 2 months. Copper exposure affected the root DW yield of C. eragrostis from 2 mu M Cu, whereas I. pseudacorus and A. donax developed well. Maximum Cu concentration in the biomass of C. eragrostis and P. arundinacea (i.e. 255 and 838 mg Cu kg(-1) DW respectively) did not reach the 1000 mg Cu kg(-1) DW threshold value needed to produce Cu-ecocatalysts. Copper concentrations in the roots of I. pseudacorus and A. donax exceeded this threshold value at 40 mu M and over 10 mu M Cu, i.e. 1099 and 1809 mg Cu kg(-1) DW, respectively, making them relevant candidates for producing Cu-ecocatalysts.

In the present study three different procedures have been compared for the determination of the lipophilicity of the unionized species (log Po/w) of neutral, acidic, basic, amphoteric, and zwitterionic drugs. Shake-flask, potentiometric and chromatographic approaches have been assayed in a set of 66 representative compounds in different phases of advanced development. An excellent equivalence has been found between log Po/w values obtained by shake-flask and potentiometry, while the chromatographic approach is less accurate but very convenient for screening purposes when a high-throughput is required. In the case of zwitterionic and amphoteric compounds, either for shake-flask and chromatographic methods, the pH has to be accurately selected in order to ensure the compound to be in its neutral form.

International audience; Chromium-coated zirconium alloys are being studied as Enhanced Accident Tolerant Fuel Cladding for Light Water Reactors (LWRs). Those materials are especially studied to improve the oxidation resistance of LWRs current fuel claddings in nominal and at High Temperature (HT) for hypothetical accidental conditions such as LOss of Coolant Accident. Beyond their HT behavior, it is essential to assess the materials behavior under irradiation. A first generation chromium/Zircaloy-4 interface was thus irradiated with 20 MeV Kr$^{8+}$ ions at 400°C up to 10 dpa. High-Resolution Transmission Electron Microscopy and chemical analysis (EDS) were conducted at the Cr/Zr interface. The atomic structure of the interface reveals the presence of Zr(Fe, Cr)$_2$ Laves phase, displaying both C14 and C15 structure. After irradiation, only the C14 structure was observed and atomic row matching was preserved across the different interfaces , thus ensuring a good adhesion of the coating after irradiation.

Compared to most bulk materials, open-cell aluminium (Al) foams (OCAFs) are light-weight and can absorb a significant amount of energy in compression, e.g. during impact. When coated with nickel (Ni), OCAFs can absorb even more energy, making them more appropriate for impacts at higher velocities than uncoated OCAFs. When Ni-coated OCAFs experience low-velocity impact however, the stopping distance during the impact is small compared to that of uncoated OCAFs and hence, deceleration occurs fast. This exposes devices (and possibly human beings) protected by OCAFs to large internal forces leading to internal damage. An OCAF that combines the properties of uncoated and coated OCAFs can absorb energy during both low-velocity and high-velocity impact scenarios. This contribution introduces two of such OCAFs which are created by partially and gradually coating OCAFs. The general mechanics of the two OCAFs are revealed using experimental and numerical observation methods. The Oxide Dispersion Strengthened (ODS) materials are potential candidates as cladding tubes for Sodium-cooled Fast Reactors. The nano-oxides are finely dispersed within the grains and confer excellent mechanical properties to these alloys. Hence, assessing nano-particle stability under irradiation remains crucial to guarantee safe use of these materials. Although neutron irradiation remains a binding and challenging experimental study to conduct, difficulties can be overcome by ion beam processing. Ion beam processing of the ODS material allows to identify the radiation-induced Ostwald ripening as the mechanism governing the nano-particle response under irradiation. The result is the increase in size and a decrease in density of the finely dispersed Y2Ti2O7 nano-particles. Under neutron irradiation, radiation-induced Ostwald ripening appears to be less effective since a slight growth of nano-particles is observed. Further, our approach shows that nanoparticle growth kinetics should scale as φ1/3, φ being the radiation flux. This suggests that the low irradiation flux is at the origin of the slower growth kinetics of the neutron irradiated particles. Both neutron and ion irradiation induce a modification of the nanoparticles/matrix interfaces which are generally flat and sharp prior to irradiation and present steps after irradiation. This could alter the nano-particle coarsening during irradiation.

The α7 nicotinic receptor subunit and its partially duplicated human-specific dupα7 isoform are coexpressed in neuronal and non-neuronal cells. In these cells, α7 subunits form homopentameric α7 nicotinic acetylcholine receptors (α7-nAChRs) implicated in numerous pathologies. In immune cells, α7-nAChRs are essential for vagal control of inflammatory response in sepsis. Recent studies show that the dupα7 subunit is a dominant-negative regulator of α7-nAChR activity in Xenopus oocytes. However, its biological significance in mammalian cells, particularly immune cells, remains unexplored, as the duplicated form is indistinguishable from the original subunit in standard tests. Here, using immunocytochemistry, confocal microscopy, coimmunoprecipitation, FRET, flow cytometry, and ELISA, we addressed this challenge in GH4C1 rat pituitary cells and RAW264.7 murine macrophages transfected with epitope- and fluorescent protein-tagged α7 or dupα7. We used quantitative RT-PCR of dupα7 gene expression levels in peripheral blood mononuclear cells (PBMCs) from patients with sepsis to analyze its relationship with PBMC α7 mRNA levels and with serum concentrations of inflammatory markers. We found that a physical interaction between dupα7 and α7 subunits in both cell lines generates heteromeric nAChRs that remain mainly trapped in the endoplasmic reticulum. The dupα7 sequestration of α7 subunits reduced membrane expression of functional α7-nAChRs, attenuating their anti-inflammatory capacity in lipopolysaccharide-stimulated macrophages. Moreover, the PBMC's dupα7 levels correlated inversely with their α7 levels and directly with the magnitude of the patients' inflammatory state. Objective— SNRK (sucrose nonfermenting 1-related kinase) is a novel member of the AMPK (adenosine monophosphate-activated protein kinase)-related superfamily that is activated in the process of angiogenesis. Currently, little is known about the function of SNRK in angiogenesis in the physiological and pathological conditions. Approach and Results— In this study, in Snrk global heterozygous knockout mice, retina angiogenesis and neovessel formation after hindlimb ischemia were suppressed. Consistently, mice with endothelial cell (EC)-specific Snrk deletion exhibited impaired retina angiogenesis, and delayed perfusion recovery and exacerbated muscle apoptosis in ischemic hindlimbs, compared with those of littermate wide-type mice. Endothelial SNRK expression was increased in the extremity vessel samples from nonischemic human. In ECs cultured in hypoxic conditions, HIF1α (hypoxia inducible factor 1α) bound to the SNRK promoter to upregulate SNRK expression. In the nuclei of hypoxic ECs, SNRK complexed with SP1 (specificity protein 1), and together, they bound to an SP1-binding motif in the ITGB1 (β1 integrin) promoter, resulting in enhanced ITGB1 expression and promoted EC migration. Furthermore, SNRK or SP1 deficiency in ECs ameliorated hypoxia-induced ITGB1 expression and, consequently, inhibited EC migration and angiogenesis. Conclusions— Taken together, our data have revealed that SNRK/SP1-ITGB1 signaling axis promotes angiogenesis in vivo.

International audience; In a hydroelectric reservoir, sediments are subject to remobilization events, water-level fluctuations and physicochemical changes. Depending on their associated metallic content, surficial oxic to suboxic sediments could constitute a major source of metals. To identify the key parameters that control metallic elements in terms of their mobility and sensitivity to reservoir management, sediments were subject to resuspension and drying/wetting cycle experiments over a wide range of pH values, solid/liquid ratios (S/L) and redox (Eh) conditions. During these tests, special attention was also paid to the influence of pretreatments on samples, i.e., drying, aeration and the leachate composition (ultrapure water vs. natural water); on the preservation of the sediment characteristics; and especially on metallic element release. The results of this study show that the pH, S/L ratio and Eh parameters are key variables in metal solubilization; the pH influences metal mobility primarily through sorption-desorption phenomena as well as the dissolution of metallic-bearing phases, the S/L ratio modifies the sorption-desorption equilibria, and the Eh primarily affects the reducible sensitive phases and associated metallic elements through dissolution-precipitation processes. Under environmental conditions, evolution of these parameters can lead to a > 20% solubilization of the most mobile elements, i.e., As and Cd. These results are influenced by the sample pretreatment and experimental conditions. In fact, even if the solubilization patterns show no significant differences between dry and wet sediment depending on the physicochemical conditions, the magnitude of their release is significantly affected. Drying pretreatment induces changes in metal speciation, notably altering the distribution of the most weakly bound elements; there is almost half the amount of metallic elements associated with the exchangeable fraction in dry compared to wet sediments. The solubilization percentages were higher in the ultrapure phase than in reservoir water primarily due to the low pH, which influenced the sorption equilibria.

International audience; A detailed-balance model for thermionic emission from polyanions has been developed and applied to fullerene dianions. The specificity of this delayed decay process is electron tunneling through the repulsive Coulomb barrier (RCB). An analytical expression of the RCB is derived from electrostatic modeling of the fullerene cage. The reverse process, namely, electron attachment to the singly charged anion, is described by a hard sphere cross section weighted by the Wentzel-Kramers-Brillouin tunneling probability. This simple expression leads to a very good agreement with a measured time-resolved kinetic energy distribution of C$_{84}$$^{2−}$. Electron binding energy is reduced when the fullerene cage size decreases, leading to an almost zero one for C$_{70}$$^{2−}$ and a negative one for C$_{60}$$^{2−}$. Extension of the model to these systems of interest is discussed, and model outputs are compared with the experimental data from the literature.

BACKGROUND Accurate allergen quantification is needed to document the consistency of allergen extracts used for immunotherapy. Herein, we characterize the epitope specificities of two monoclonal antibodies used in an ELISA for the quantification of the major birch pollen allergen Bet v 1, established as a reference by the BSP090 European project. METHODS The ability of mAbs 5B4 and 6H4 to recognize Bet v 1 isoforms was addressed by immunochromatography. The capacity of each mAb to compete with patients' IgE for binding to Bet v 1 was measured by ELISA inhibition. Epitope mapping was performed by pepscan analysis, site-directed mutagenesis, and hydrogen/deuterium exchange-mass spectrometry. RESULTS The 5B4 epitope corresponds to a peptide sequence (I56-K68) overlapping with the binding sites of patients' serum IgEs. Mutation of residues P59, E60, and K65 abolishes 5B4 binding to Bet v 1 and reduces the level of IgE recognition. In contrast, 6H4 recognizes a conformational epitope lying opposite to the 5B4 binding site, involving residues located in segments I44-K55 and R70-F79. Substitution of E45 reduces the binding capacity of 6H4, confirming that it is critical for the interaction. Both mAbs interact with >90% of Bet v 1 content present in the birch pollen extract, while displaying a weak cross-reactivity with other allergens of the PR-10 family. CONCLUSIONS MAbs 5B4 and 6H4 recognize structurally distinct epitopes present in the vast majority of Bet v 1 isoforms. These results support the relevance as a reference method of the Bet v 1-specific quantitative ELISA adopted by the European Pharmacopoeia.

A SAR study identified the 4-(1-methylpyrazol-5-yl) derivative, 12f, as a selective σ1R antagonist with a good ADMET profile and potent antinociception.

The R(+) and R(-) chicken lines have been divergently selected for high (R(+)) or low (R(-)) residual feed intake. For the same body weight and egg production, the R(+) chickens consume 40% more food than their counterparts R(-) lines. In the present study we sought to determine the hypothalamic expression profile of feeding-related neuropeptides in these lines maintained under fed or food-deprived conditions. In the fed condition, the suppressor of cytokine signaling 3 (SOCS3) was 17-fold lower (P0.05) and the ghrelin receptor was 7-fold higher (P0.05) in R(+) compared to R(-) chicken lines. The hypothalamic expression of the other studied genes remained unchanged between the two lines. In the fasted state, orexigenic neuropeptide Y and agouti-related peptide were more responsive, with higher significant levels in the R(+) compared to R(-) chickens, while no significant differences were seen for the anorexigenic neuropeptides pro-opiomelanocortin and corticotropin releasing hormone. Interestingly, C-reactive protein, adiponectin receptor 1 and ghrelin receptor gene expression were significantly higher (12-, 2- and 3-folds, respectively), however ghrelin and melanocortin 5 receptor mRNA levels were lower (4- and 2-folds, P=0.05 and P=0.03, respectively) in R(+) compared to R(-) animals. We identified several key feeding-related genes that are differently expressed in the hypothalamus of R(+) and R(-) chickens and that might explain the difference in feed intake observed between the two lines.

Bloom's syndrome is an autosomal recessive genome-instability disorder associated with a predisposition to cancer, premature aging and developmental abnormalities. It is caused by mutations that inactivate the DNA helicase activity of the BLM protein or nullify protein expression. The BLM helicase has been implicated in the alternative lengthening of telomeres (ALT) pathway, which is essential for the limitless replication of some cancer cells. This pathway is used by 10–15% of cancers, where inhibitors of BLM are expected to facilitate telomere shortening, leading to apoptosis or senescence. Here, the crystal structure of the human BLM helicase in complex with ADP and a 3′-overhang DNA duplex is reported. In addition to the helicase core, the BLM construct used for crystallization (residues 640–1298) includes the RecQ C-terminal (RQC) and the helicase and ribonuclease D C-terminal (HRDC) domains. Analysis of the structure provides detailed information on the interactions of the protein with DNA and helps to explain the mechanism coupling ATP hydrolysis and DNA unwinding. In addition, mapping of the missense mutations onto the structure provides insights into the molecular basis of Bloom's syndrome.

Angiotensin II (Ang II), the key effector of the renin-angiotensin system (RAS) promotes the development of Dilated Cardiomyopathy (DCM), which ultimately leads to heart failure (HF). Previously, our lab has reported that a serine threonine kinase, Sucrose nonfermenting 1-related kinase (SNRK), a member of AMP-activated protein kinase (AMPK) family is critical for cardiac metabolism, and specific loss of SNRK in cardiomyocytes results in DCM. Thus, we hypothesize that, SNRK is involved in maintaining cardiac homeostasis and Ang II exacerbates cardiac dysfunction in cardiomyocyte-specific SNRK knockout mice (SNRK-CKO). To investigate this hypothesis, we performed ECHO on 4 months adult SNRK-CKO mice with and without Ang II, and found that most cardiac function parameters were compromised, followed by death in 14 days. These cardiac function defects were not observed in Ang II-infused SNRK-endothelial (EC) KO mice. We next investigated signaling pathways previously implicated in cardiac dysfunction. Indeed, upon stimulation with Ang II, the heart tissue shows downregulation of pERK and pJNK proteins and upregulation of pAKT protein. In contrast, in SNRK-ECKO, pERK and pJNK proteins were upregulated and pAKT protein was downregulated. Because pAKT, is known to increase inflammatory signaling pathways, in particular IKK-NF-κB, we investigated the IKK-NF-κB signaling in heart lysates from SNRK-CKO and SNRK-ECKO mice. Interestingly, we found upregulation of proinflammatory markers such as IL-6, TNFα and p65 NF-κB and downregulation of anti-inflammatory IL-10 in SNRK-CKO hearts only. This increase in inflammatory signaling is often followed by fibrosis, which was observed via Sirus Red staining for collagen accumulation in Ang II-infused SNRK-CKO hearts. At present, we are investigating the inflammatory and signaling pathways associated with lentivirus-mediated knockdown of SNRK in HL-1 cardiomyocytes in vitro. Overall, these results suggest that SNRK could be a potential target in regulating inflammatory mechanisms, which exerts cardioprotective effect in the heart.

International audience; Manganeseoxides areknown tobe efficient sorbents in soils, and thus potentially useful in soil remediation. A novel synthetic amorphous manganese oxide (AMO) was prepared using the first step of the procedure generally used for birnessite synthesis (i.e., omitting the heating step of the gel). Its long-term (90 days) stability was evaluated using a pot experiment in contrasting soil samples (pH = 4.2, 5.4, and 7.3). Up to 113 mgMn/L was released into porewater at the beginning of the experiment in more acidic soils, indicating a partial dissolution/alteration of AMO surfaces. Manganese release into the pore water stabilized after 15 days, in agreement with the mass loss measurements. Mass loss decreased again after 60 days of incubation for the neutral soil due to the massive formation of secondary rhodochrosite (MnCO3) and/or kutnahorite [(Ca,Mn)CO3] detected on the AMO surfaces by both X-ray diffraction analysis (XRD) and scanning electron microscopy (SEM). Mineralogical transformations and aggregation of fine grained AMO particles probably lead to the decrease of binding surfaces. Despite the fact that 10-18% of AMO dissolved during the 90 days of the experiment, the sorbent was found to be relatively stable, primarily in near neutral soils; consequently, its in situ applications to polluted soils need to be evaluated further.

International audience; We determined the acidity constants of tebuconazole and penconazole, two fungicides from the group of 1,2,4-triazoles. Potentiometric titrations were performed in a 20 % (v/v) acetonitrile/water mixture at 25 °C and at a fixed ionic strength (KNO3, 0.1 mol*dm−3). The pKa values (representing thermodynamic constants) were determined to be 5.0 ± 0.1 and 5.2 ± 0.1 for tebuconazole and penconazole, respectively. These values could be used in pure water solutions to consider the protonated or deprotonated forms when studying the field behavior of these fungicides. Molecular modeling calculations allowed identifying the N4 atom as the protonation site.

An original workflow allowing inline FAIMS separation, electrospray ionization, mass analysis and ion spectroscopy (IRMPD: InfraRed Multiple Photon Dissociation) is presented for multidimensional molecular analysis. This new instrument consists of an ultraFAIMS (Owlstone) device interfaced to a linear ion trap (LTQ XL Thermo Scientific) which was modified for IRMPD spectroscopy. Two modes of operation are demonstrated on an isomeric mixture of paracetamol and 2-phenylglycine. In the first mode a FAIMS (high-Field Asymmetric waveform Ion Mobility Spectrometry) separation of the isomers is performed with a static compensation field for mass- and isomer- selective ion spectroscopy. In the second mode, the compensation field is scanned while the ions are irradiated at a fixed wavenumber. The advantages of this workflow as compared to traditional FAIMS-MS and IRMPD spectroscopy are described. The potential of the two modes for molecular spectroscopy and analytical applications, in particular the new “omics” are discussed.

We present the observation of a checkerboard-like interference pattern in transverse momentum distributions measured for near-threshold photoionization of hydrogen atoms in a DC electric field. We analyze the pattern in terms of constructive and destructive interference between electron trajectories that directly leave the vicinity of the ion and indirect trajectories that remain in the vicinity of the ion for one or more orbital periods, and show that the interference pattern can be discussed in terms of ionization time delays between these two classes of trajectories.

Spent catalysts are generated in large quantities as solid waste on a yearly basis. Owing to their hazardous nature, environmental regulations for discarded spent catalysts are very strict. Consequently, from the ecological and economical viewpoint, metal recovery from spent catalysts is very important. Molybdenum (Mo) is one of the elements present in spent catalysts at high concentrations. The rapidly growing demands for Mo and its products create a need to develop novel recovery processes from secondary resources, i.e. spent catalysts, as there is a projected shortage of the primary resources for this element. This chapter overviews: (i) the general characteristics of Mo along with the research conducted in recent years concerning Mo chemical leaching, i.e. with strong inorganic acids, sodium hydroxide, hydrogen peroxide and acid mixtures, (ii) Mo bioleaching, i.e. with bacteria Acidithiobacillus ferrooxidans, Acidithiobacillus thiooxidans and Acidianus brierleyi or fungi Penicillium simplicissimum and Aspergillus niger and (iii) Mo recovery, i.e. precipitation, adsorption, ion exchange and solvent extraction, from spent catalysts. Chemical leaching offers Mo leaching yields exceeding 90%. Sulphuric acid seems to be the best leachant owing to its high oxidising power. Bioleaching offers more cost-efficient, simpler and more environmentally friendly processes. However, bioleaching has relatively long leaching cycles (usually around 20 days) and extraction efficiencies of Mo are below 70%. Among the recovery techniques, solvent extraction is well-established in hydrometallurgy for commercial production of high purity Mo.

The Malliavin calculus is an extension of the classical calculus of variations from deterministic functions to stochastic processes. In this paper we aim to show in a practical and didactic way how to calculate the Malliavin derivative, the derivative of the expectation of a quantity of interest of a model with respect to its underlying stochastic parameters, for four problems found in mechanics. The non-intrusive approach uses the Malliavin Weight Sampling (MWS) method in conjunction with a standard Monte Carlo method. The models are expressed as ODEs or PDEs and discretised using the finite difference or finite element methods. Specifically, we consider stochastic extensions of; a 1D Kelvin-Voigt viscoelastic model discretised with finite differences, a 1D linear elastic bar, a hyperelastic bar undergoing buckling, and incompressible Navier-Stokes flow around a cylinder, all discretised with finite elements. A further contribution of this paper is an extension of the MWS method to the more difficult case of non-Gaussian random variables and the calculation of second-order derivatives. We provide open-source code for the numerical examples in this paper.

International audience; Allergy to the short ragweed (Ambrosia artemisiifolia) pollen is a major health problem. The ragweed allergen repertoire has been recently expanded with the identification of Amb a 11, a new major allergen belonging to the cysteine protease family. To better characterize Amb a 11, a recombinant proform of the molecule with a preserved active site was produced in Escherichia coli, refolded, and processed in vitro into a mature enzyme. The enzymatic activity is revealed by maturation following an autocatalytic processing resulting in the cleavage of both N- and C-terminal propeptides. The 2.05-A resolution crystal structure of pro-Amb a 11 shows an overall typical C1A cysteine protease fold with a network of molecular interactions between the N-terminal propeptide and the catalytic triad of the enzyme. The allergenicity of Amb a 11 was confirmed in a murine sensitization model, resulting in airway inflammation, production of serum IgEs, and induction of Th2 immune responses. Of note, inflammatory responses were higher with the mature form, demonstrating that the cysteine protease activity critically contributes to the allergenicity of the molecule. Collectively, our results clearly demonstrate that Amb a 11 is a bona fide cysteine protease exhibiting a strong allergenicity. As such, it should be considered as an important molecule for diagnosis and immunotherapy of ragweed pollen allergy.

This work investigates the ability of EPS extracted by cationic exchange resin (CER) from 3 different anaerobic granular sludges to bind Pb or Cd as a function of pH. EPS display a better binding ability for Pb than Cd as it has been often shown in literature. The experimental sorption data show the presence of a pH sorption edge. The experimental data are well fitted by the Kurbatov model. Nevertheless, the molar ratio of proton exchanged with metal ions underlines that other mechanisms are also involved in metal ion sorption by EPS from anaerobic granular sludges. Moreover, the mineral fraction present in extracted EPS and trapped by the organic fraction or under particle form seems to be involved in the metal ions sorption process.

The third accelerator of the multi-ion irradiation platform JANNUS (Joint Accelerators for Nanosciences and NUclear Simulation), a 6SDH-2 Pelletron from National Electrostatic Corporation, Middleton was installed at Saclay in October 2009. The first triple beam irradiation combining Fe, He and H ion beams has been performed in March 2010. In the first part of this paper, we give a technical description of the triple beam facility, its performances and experimental capabilities. Typically, damage dose up to 100 dpa can be reached in 10 h irradiation with heavy ion beams, with or without simultaneous bombardment by protons, helium-4 ions or any other heavy ion beam. In the second part of this paper, we illustrate some IBA results obtained after irradiation and implantation experiments.
