Background and Objective: Poverty, drug resistance and the advent of human immunodeficiency virus infection (HIV) have led to a recent upsurge in the incidence of tuberculosis including intracranial tuberculosis. In this article, we report 3 patients who had solitary brain tuberculomas and were otherwise healthy to underscore the importance of continuing vigilance for this disease., Case Reports: Three patients (57 years, female; 52 years, male; 7 years male) presented to our unit with features of intracranial tumours. They were all HIV negative with no previous history of tuberculosis. Cranial computed tomography scans demonstrated uniformly contrast enhancing falcine supratentorial masses in the adult patients and a cerebellar hemispheric lesion with peripheral contrast enhancement in the paediatric patient, Intervention: All the patients had gross total tumour excision. The histology confirmed a tuberculoma. They all had antituberculous therapy after histological confirmation. The outcome was good in all the patients., Conclusion: Intracranial tuberculoma can occur in otherwise healthy individuals and should always be considered in the differential diagnosis of solitary intracranial mass lesions in sub-Saharan Africans so that minimally invasive procedures can be used to establish the correct diagnosis.

The teratogenic effect of maternal cyanide consumption on the gross morphology of the post-natal phase of the developing rat cerebellum was studied. Twenty pregnant female rats weighing between 170 g and 190 g were separated into control and experimental groups. The control animals were fed a standard diet of mice cubes, while the experimental animals were fed 500 ppm potassium cyanide, mixed with the standard diet. The diets were fed to the animals and their litters in separate cages and water provided ad libitum during gestation and to the offspring after birth. After birth, the offspring (five per group) of days 1, 9, 14, 21, 28 and 50 were weighed, killed by cervical dislocation and the gross parameters studied. In the experimental animals, no significant differences were observed in the studied parameters between the control and experimental animals on day 1. A significant reduction in body weight was observed on day 14 (P < 0.05). The brain weight was significantly reduced on day 9 (P < 0.05). Similarly, the cerebellar weight was significantly reduced on days 14,21 and 28 (P < 0.05). The maximum vermal length was significantly reduced on day 50 (P < 0.05), and the maximum side-to-side dimension of the cerebellum was also reduced on day 28 (P < 0.05). There was no reduction in the thickness (anteroposterior dimension) of the cerebellum in the experimental group (P > 0.05). From the result, it is inferred that maternal consumption of 500 ppm cyanide causes reduction in the cerebellar weight, vermal length and side-to-side dimension of the developing cerebellum in postnatal life in rats.

Objective: To investigate the microscopic effect of maternal cyanide consumption on the developing cerebellum of Wistar rats., Materials and Methods: Twenty pregnant female rats weighing between 160 g and 180 g were used in this study. The rats were separated into two groups comprising ten control and ten experimental animals. The control animals were fed a standard diet of mice cubes, while the experimental animals were fed 500 ppm potassium cyanide, mixed with the standard diet. The diets were fed to the animals and their litters in separate cages and water provided ad libitum during pre and postnatal life. After birth, the offspring (five per group) of days 1,9,14,21,28 and 50 were weighed and killed by cervical dislocation. The cerebellar tissues were processed and microscopic parameters studied., Results: A thicker external granular layer (EGL) was seen in the control group on day 1(39+/-9.2microm) compared with the experimental group (29+/-5.8microm) and on day 9(83+/-7.1microm) compared with the experimental group (78+/-13microm). However, these were not significantly different statistically. A thicker and persistent EGL was observed in the experimental group on days 14 and 21. A significant (P<0.05) reduction in the thickness of molecular layer (ML) was observed on days 28 and 50 in the experimental group. The density and size of the Purkinje cells were the same in both the control and experimental groups (P>0.05)., Conclusion: Maternal consumption of 500 ppm cyanide in rats does not significantly affect light microscopic prenatal cerebellar development, but causes mild changes in the post-natal life. Maternal cyanide consumption causes delayed maturation of the cerebellum, as evidenced by the thicker EGL, and reduction in the ML in the experimental group which become noticeable only at about 28th day of postnatal life.

Kola nut (from Cola nitida) is popular in Nigeria and West Africa and is commonly consumed by pregnant women during the first trimester to alleviate morning sickness and dizziness. There is, however, a dearth of information on its effects on the developing brain. This study, therefore, investigated the potential effects of kola nut on the structure of the developing neonatal and juvenile cerebellum in the rat. Pregnant Wistar rats were administered water (as control) or crude (aqueous) kola nut extract at 400, 600, and 800 mg/kg body weight orally, from pregnancy to day 21 after birth. On postnatal days 1, 7, 14, 21 and 28, the pups were weighed, anaesthetised, sacrificed and perfused with neutral buffered formalin. Their brains were dissected out, weighed and the cerebellum preserved in 10% buffered formalin. Paraffin sections of the cerebellum were stained with haematoxylin and eosin for cerebellar cytoarchitecture, cresyl violet stain for Purkinje cell count, Glial Fibrillary Acidic Protein (GFAP) immunohistochemistry (IHC) for estimation of gliosis, and B-cell lymphoma 2 (Bcl-2) IHC for apoptosis induction. The kola nut-treated rats exhibited initial reduction in body and brain weights, persistent external granular layer, increased molecular layer thickness, and loss of Bergmann glia. Their Purkinje cells showed reduction in density, loss of dendrites and multiple layering, and their white matter showed neurodegeneration (spongiosis) and GFAP and Bcl-2 over-expression, with evidence of reactive astrogliosis. This study, therefore, demonstrates that kola nut, administered repeatedly at certain doses to pregnant dams, could disrupt normal postnatal cerebellar development in their pups. The findings suggest potential deleterious effects of excessive kola nut consumption on human brain and thus warrant further studies to understand the wider implications for human brain development. Radiation side effects have been reported to induce oxidative stress by free radical generation. The protective effect of Carica papaya (CP) fruit extract, vitamins C and E against gamma radiation-induced oxidative damage on postnatal developing rat cerebellum was studied. Forty-two female Wistar rats were mated and divided into six groups (n=7). Group A served as control and received water, Group B received a dose of 2.5 Gy irradiation only, Group C received 200 mg/kg aqueous extract of CP only, Group D received 200 mg/kg aqueous extract of CP + 2.5 Gy irradiation, Group E received 500mg/kg Vitamin E + 2.5 Gy irradiation and Group F received 200 mg/kg Vitamin C + 2.5 Gy irradiation. The pregnant rats were exposed to irradiation with radiation from Cobalt 60 delivered by an AECL machine in prone position on 7th day of pregnancy, while aqueous extract of CP, Vitamin E and Vitamin C were administered orally from day 7 of gestation to postnatal day 28. Behavioural and haematological assessments of pups were done on day 21, and killed. Some cerebella of the pups of days 1, 7 14 21 and 28 were fixed in 10% formol-saline for histological and histomorphometric studies, while other cerebella of day 21 were preserved in phosphate buffered saline at 4oC and pH 7.4 for biochemical assays. Data were presented as Mean ± Standard Deviation and analysed using ANOVA at p < 0.05. Results showed in the irradiated rats, a significant reduction in body weight, increased brain weight, decreased time spent on the forelimb grip, decreased haemoglobin (Hb) concentration and increased lipid peroxidation (LPO) and H2O2 levels in the cerebellum compared with the control, CP, vitamins C and E rats at p<0.05. Histologically, there was persistent external granular layer (EGL) as well as reduction in molecular layer (ML) thickness in the irradiated group compared with the control and other treated groups on postnatal day 21. In conclusion, 2.5 Gy whole body irradiation of pregnant rats induced oxidative stress in the developing rat cerebellum. Treatment, with aqueous extract of CP reduced the damage caused by irradiation when compared with vitamins C and E. [ABSTRACT FROM AUTHOR]
