The aim of this analysis was to determine whether pregnancy loss (PL) after embryo transfer (ET) in cattle was related to maternal progesterone (P4) concentrations during and shortly after ET, and maternal bovine pregnancy-associated glycoprotein-1 (bPAG-1) concentrations in plasma at days 25-35 of gestation. Embryos (n=260) were produced either in vivo after superovulation (n=115), or in vitro from oocytes (obtained with ovum pick-up) in co-culture (n=44) or cultured in a synthetic medium (n=101). Overall, PL was 56.9% (148) and no significant differences occurred in calving rate among the three embryo production groups. There was no difference in P4 concentrations on days 7-14 of gestation in the three groups, nor between ongoing and interrupted pregnancies. Between days 25 and 35 of pregnancy, bPAG-1 concentrations were unaffected by embryo production, but in cattle that had PL between days 26 and 120, four bPAG-1 profiles could be detected. Between days 25 and 32, bPAG-1 concentrations were influenced by PL, and concentrations were significantly lower in animals in which PL occurred between days 26 and 120 than in those animals that aborted later or calved at term. Early P4 concentrations suggested that maternal luteal factors were not responsible for PL which appeared to be caused by impaired conceptus development (regardless of embryo type) as reflected by low maternal bPAG-1 concentrations prior to embryonic death., (Copyright © 2012 Elsevier Ltd. Various parenteral treatment forms of oxytocin, as often used under praxis circumstances, may act differently on contractility of the uterus during the first days of the puerperium. Various patterns of such induced uterotonic responses may lead to alterations in the emptying characteristics of the uterine lumen, thus influencing, as a late consequence, the process of involution. Therefore, this study was designed to test whether two different parenteral administration forms of oxytocin induce changes in peripheral plasma concentrations of 15-ketodihydro-prostaglandin F(2α) (PGF(2α) metabolite) in early post-partum cows. Between 13 and 15 h after uncomplicated calving, healthy dairy cows without retained foetal membranes were treated with 50 IU oxytocin, either intramuscularly (OT-IM group; n = 15) or intravenously (OT-IV group; n = 16). Saline solution was administered intramuscularly as controls (CON group; n = 15). Jugular blood samples were taken at 10-min intervals from 1 h before to 2 h after treatment. Plasma PGF(2α) metabolite levels were measured by radioimmunoassay. No significant differences in peripheral plasma PGF(2α) metabolite concentrations occurred in the OT-IM and CON groups, but mean values significantly increased in the OT-IV group, peaking at 20 min after treatment and reaching pre-treatment baseline values again at 120 min. Although the source of prostaglandins was not investigated in this study, our results suggest that exogenous oxytocin may enhance secretion of prostaglandins by the uterus during the first day after normal calving. These prostaglandins might contribute, by an endocrine or paracrine route, to the stimulation of myometrial contractility when exogenous oxytocin is given during this early post-partum stage., (© 2011 Blackwell Verlag GmbH.)

The bovine cervix contains a large amount of smooth muscle cells distributed over an outer muscular layer and within a stromal layer. The stromal layer exhibits no electromyographic (EMG) activity at parturition. This leads to the question whether the stromal smooth muscle cells of the bovine cervix are prepared to contract with parturition, or whether they have another function. To this end, cervical biopsies were repeatedly taken from 10 pregnant cows at day-185 and -275 of gestation, at spontaneous, uncomplicated calving and at 30 days after calving. The smooth muscle bundles of the stroma were immunohistochemically analysed (n = 5) with regard to their integrity and cellular density, and the degree of staining for connexin-43, smooth muscle actin alpha (SMA), desmin and vimentin. Additionally, the mRNA expression for connexin-43, SMA, desmin and vimentin was determined with RT-PCR (n = 5). The smooth muscle tissue was arranged in bundles, also at parturition. However, the cellular density of these bundles and the SMA mRNA expression were decreased at parturition. Additionally, the SMA staining and connexin-43 expression and staining remained constant during pregnancy and at parturition. This might indicate that stromal smooth muscle cells are not prepared to contract with parturition, in contrast to the myometrial smooth muscle cells. The smooth muscle cells, stained for SMA, also expressed vimentin, and the proportion of co-expression was increased at day-275 of pregnancy. This suggests that the stromal smooth muscle cells predominantly have a secretory function in cows.

Contents Compared with other parts of the reproductive cycle, the parturition process of domestic animals has received relatively little attention from researchers during the last two decades. Yet, interventions during parturition, either by the owner or the veterinarian, are frequently judged necessary and perinatal losses in most of the domestic species are (still) rather high. The present paper focuses on the birth process in three domestic species, the dog, pig and cow, mainly because most of the experimental and research work of the authors themselves was dealing with these three species. Against the background of specific clinical problems, breeding circumstances and economic performance, selected topics and questions are addressed to identify areas for future research. These are, among others, related to the mechanisms that trigger the birth process (are the foetuses involved in the dog and sow?), the role of hormones and cytokines during ripening of the cervix (activation and/or de-activation of specific matrix metallo-proteinases and their local inhibitors), the functional significance of fetal body movements to prepare the foetus for expulsion, and the exact time course of events involved in the transportation of an individual foetus from its intrauterine location, via the pelvic canal, to the outside world. Research on some of these topics, such as the ripening of the cervix in cows, might also contribute to resolve clinical problems in human obstetrics.

Concentrations of bovine placental lactogen (bPL) were determined in fetal plasma samples by twelve double-antibody competitive radioimmunoassay systems (RIA I-XII) based on either recombinant bPL (non-glycosylated) or native bPL (glycosylated). Both preparations were used as standard and tracer, and for primary antisera production. The minimum detection limit measured by these RIA varied from 0.02 to 0.6 ng bPL mL(-1). The coefficients of correlation of different bPL RIA systems were up to 90% (P < 0.0001) when each RIA was tested against the average values of all twelve RIA systems. All developed RIA were used to investigate the incidence of different bPL isoforms in bovine fetal serum samples (n = 71). Fetal concentrations ranged from 11.8 to 35.7 ng mL(-1) at the third month and from 1.1 to 13.5 ng mL(-1) at the ninth month of gestation. They tended to decrease with advancing gestation. In general, those RIA systems that used recombinant bPL as the standard measured higher values than those using the native bPL preparation. These differences decreased toward the end of gestation (P < 0.05), suggesting a lower rate of glycosylation. Our results provide evidence of different glycosylated isoforms of bPL in fetal serum at different gestation periods.

Calves derived from IVP embryos may suffer from the large offspring syndrome that has been related to effects of in vitro culture on the intrinsic quality of the embryo. Limited information is available on the role of the placenta in such cases. In this study, bovine pregnancy-associated glycoprotein (bPAG) was used as a marker to test whether placental function is influenced by the route of embryo production. Therefore, from day 7 until day 119 of ongoing gestations, resulting from transfer of MOET (n = 53), IVP-co-culture (n = 21) and IVP-SOF (n = 38) embryos, bPAG levels were compared in peripheral plasma of recipients. Plasma progesterone levels were compared as well. From day 25 of gestation onwards, bPAG could be detected in all recipients and the levels were significantly influenced by the day of gestation. Although IVP calves were significantly heavier than the in vivo produced calves, this difference was not reflected in the bPAG profiles of the embryo production groups. Yet, the mean bPAG level of the three last sampling moments (days 105-119) tended to be positively related to the birth weight of the calves, irrespective of the embryo production technique. Progesterone concentrations were not influenced by route of embryo production, but were significantly affected by parity of the recipient and day of gestation.

The Large Offspring Syndrome has frequently been reported for in vitro produced calves. The objective of this study was to determine whether any differences in body dimensions (biparietal diameter of the cranium (BPD), cross-section of the abdomen at the insertion of the umbilical cord (CAU)) and heart rate (FHR) can be detected during the first 108 days of gestation between bovine foetuses derived from different methods of embryo production. Three groups of pregnancies with calvings at term resulted from non-surgical transfers of three types of embryos: recipients carrying an embryo obtained by standard MOET procedures (n = 25); recipients carrying an embryo produced in vitro from OPU-derived oocytes, using co-culture-medium (n = 14) or SOF-medium (n = 22). Transrectal ultrasonographic examinations were performed weekly. Ultrasound images were recorded and during off-line analysis FHR, BPD and CAU were determined. For each foetus a curve was fitted and the estimates on fixed time intervals were used as dependent variables in an analysis of variance to detect differences between the three pregnancy groups. Neither gestation length nor birth weight differed significantly between the three pregnancy groups, nor could any differences with respect to BPD, CAU or FHR be detected between Days 35 and 108 of gestation. It is concluded that no differences exist between the early development of bovine foetuses, derived from MOET, IVP-co-culture or IVP-SOF embryos, and resulting in calves with normal birth weights.

Inoculation with Actinomyces pyogenes and administration of prostaglandin (PG)F(2alpha) were used to induce late embryonic mortality (LEM) in heifers (n=8) on Days 30-38 of pregnancy in order to compare the profile for bovine pregnancy associated glycoprotein 1 (PAG1), progesterone and 15-keto-13,14-dihydro-PGF(2alpha) (PGFM). Two pregnant heifers were used as controls. Inoculation into the uterine body caused LEM, as established by ultrasonography in each heifer within 24h of treatment. When the inoculum was injected into the first part of the cervix, LEM occurred in one of two heifers (Heifer A) between 48 and 72 h after treatment. Similarly, PGF(2alpha) treatment caused LEM in three of four heifers. In six of eight heifers, PAG1 started to decrease steadily when it was accompanied by the subsequent death of the embryo. Inoculation through the cervix caused luteolysis in three of four heifers within 6-10 days after induction. After induction of LEM, PGFM concentrations showed a two to 3.8 fold increase in three of four heifers during the following six days, and from that time changed within normal ranges. The results of this study indicate that a PAG1 assay may provide an alternative method to ultrasonography for determining LEM in the cow.

In the cow the foetal endocrine signals that initiate the calving process result in prepartum luteolysis. Withdrawal of progesterone (P4) action is a prerequisite for a normal calving. The rather abrupt declining influence of P4 is followed by a cascade of physiological processes in the myometrium and cervix. This contribution will focus on some of these events. Like in many other species, the myometrium in cows is not completely inactivated during pregnancy. So-called contractures have been registered during the final weeks of gestation and their EMG-characteristics in cows show a low frequency (on average: 13.6 per day) and long duration (on average 12.1 min). They are not evenly spread over the day because they occur less frequently when the cows are disturbed for feeding or cleaning their stables. Contractures affect several foetal functions. In the cow these contractures disappear during a period of about 8-9h when maternal plasma P4 levels are rapidly declining before calving. There is experimental evidence that this temporary inhibition is associated with prepartal luteal regression. The cause of this inhibition is still unknown. Because nitrous oxide inhibits smooth muscle cells and evidence in laboratory animals indicates that expression of the inducible form of nitrous oxide (iNOS) is downregulated in myometrium, but upregulated in the cervix around the onset of parturition, we started to investigate the role of this enzyme in bovine tissues around calving. By means of a RT-PCR technique, we obtained a first indication that iNOS is hardly expressed in the myometrium during calving, while expression was clearly detected at day 4 after calving. Analysis of prepartum en periparturient biopsies from myometrium and cervix with quantitative PCR is still underway. In six pregnant cows, provided with uterine EMG-electrodes and with ultrasonic crystals implanted on the caudal cervical rim to measure cervical dilatation, calving was induced with an injection of prostaglandin (PG) F2alpha. While maternal plasma P4 levels had significantly declined within 8h after PG treatment, the myometrium escaped from temporary inhibition with the development of a parturient contractility pattern on average at 13.5h after injection. However, it was only at 28 h after PG treatment that the first sustained increase of the opening of the vaginal ostium of the cervix was measured.

One of the major contributors to poor fertility of a dairy herd is ineffective detection of estrus. Recently, it has become evident that cow factors contribute largely to low detection rates. Until now, standing behavior has been the symptom used to determine the right moment for insemination. However, standing behavior is not observed in over 50% of the cows in estrus in a number of herds, and no data are available to relate the moment of ovulation to other estrous symptoms. Therefore, in the present study, cows were observed for a number of visual signs of estrus, twice per day for 30 min, and the moment of ovulation was estimated by transrectal ultrasonographic scanning of the ovaries. In this study, 100 cows were detected in estrus visually, using a scoring system. Standing behavior was observed in 50% of those cows, although there was more than one cow in estrus at a time in most (85%) cases. There was no correlation between follicular size and ovulation time or estrous behavior score. Levels of 305-d milk yield and parity were also not correlated with estrous behavior scores. However, a significant correlation (0.31) was found between detection score and day of ovulation. A retrospective analysis revealed that cows that ovulated 0 to 24 h after artificial insemination (AD) scored almost three times the number of estrous behavior points compared to the ones that ovulated 24 to 48 h after AI. Ovulation more than 48 h after AI resulted in pregnancy in only 15% of the cows.

The objective of this study was to investigate the temporal changes in dilatation of the caudal cervix during induced calvings (n = 5). We used ultrasound cervimetry, allowing the continuous recording of the distance between a transmitting and receiving ultrasound crystal, which were implanted opposite to each other on the caudal rim of the cervix. We started recording between 19 and 21 h after injecting a prostaglandin analogue (PG) on day 272 of gestation. A fluid-filled catheter had been introduced transcervically between the fetal membranes and the uterine wall for measurements of intra-uterine pressure (IUP). While the characteristics of calving varied widely between the five animals, it appeared possible to divide the process of dilatation into four phases. During the latent phase, which lasted until 25-43 h after PG, no net gain in dilatation occurred. We found an acceleration phase (4.3-6.8 h), in which the dilatation rate speeds up (0.49-0.84 cm/h) in three of the cows. During the phase of maximum slope (lasting 0.5-4.8 h), we measured an even higher rate (1.47-8.48 cm/h), decreasing again during the deceleration phase (rate 0.24-2.28 cm/h) in four cows. The quality of the IUP measurements precluded us from continuously investigating the relationship between cervical dilatation and uterine contractions. However, short term simultaneous recordings revealed that the cervical opening changed momentarily in the absence of IUP during the latent phase, while during the phase of maximum slope, temporary changes of dilatation coincided with uterine contractions. We concluded that the method of ultrasound cervimetry used in this study provides a valuable way to study the process of cervical dilatation in parturient cows in vivo.

An immunohistochemical study of the expression of oestrogen (ER) and progesterone receptors (PR) in different regions along the longitudinal and vertical axes of the cervix of non-pregnant cows was performed. Animals were separated into two groups depending on the presence or absence of a functional corpus luteum in their ovaries, as indicated by blood progesterone concentrations. The high progesterone group (HP4) had serum progesterone concentrations > 2.0 ng mL(-1) (n = 6) and the low progesterone group (LP4) had serum progesterone concentrations < or = 0.5 ng mL(-1) (n = 4). Significantly higher concentrations of oestrogen were found in the cervical tissue of animals in the LP4 group than those in the HP4 group (473 +/- 53 v.149 +/- 46 pg g(-1) wet weight; P < 0.01). Furthermore, there was a significant effect of tissue layer (epithelium to deep stroma) on the number of ER (P < 0.01) and PR (P < 0.05) immunoreactive nuclei per 1000 cells. For both ER and PR the proportion of cells expressing the receptor increased from epithelium to subepithelial stroma (P < 0.01) and from subepithelium to deep stroma (ER P < 0.05; PR P =0.061). When the number of receptor-positive cells were expressed per mm2 tissue, differences between the subepithelial stroma and the deep stroma became even more marked. In addition, the vaginal part of the cervix had significantly more (P < 0.01) ER and PR immunoreactive nuclei per 1000 cells than the uterine part, but these differences were no longer apparent when a correction was made for cell density. There was no relationship between progesterone status of the animals, nor local tissue oestrogen concentrations and ER or PR immunoreactivity in the cervix of these non-pregnant cows. Instead, a strong relationship between both longitudinal and vertical positioning of tissue in the cervix and expression of both receptor types was shown. In addition, a strong correlation between ER and PR expression in the subepithelial stroma (R = 0.85, P < 0.01) and the deep stroma (R = 0.83 P < 0.01) was evident. In conclusion, these results demonstrate that in studies of steroid hormone receptor expression in the cervix, careful description of sampling site and depth are necessary if the results are to be interpreted meaningfully.

At Days 26 to 58 after AI, 138 Holstein-Friesian dairy cows were repeatedly examined by ultrasonography, using a 7.5 MHz linear-array rectal transducer. The total calving rate was 37.6% (52/138), and late embryonic mortality occurred 8.6% of the cows (12/138). On the days of ultrasound scanning, blood samples were drawn from the jugular vein for measuring the concentration of bovine pregnancy-specific protein B (bPSPB) and bovine pregnancy-associated glycoprotein 1 (bPAG 1). When compared with calving results, there were no significant differences in accurate diagnosis of pregnant cows were found between the 3 methods. However, when recognition of an embryo proper with a beating heart was used as the criterion for positive ultrasonographic diagnosis significantly fewer (P < 0.001) pregnant cows were correctly identified than by the other 2 tests. When compared with the noncalving cows, significantly fewer (P < 0.001) false positive diagnoses were made by the 2 ultrasonographic tests than by the PSPB and bPAG 1 tests, while significantly fewer (P < 0.001) false positive diagnoses were made by the bPSPB test than by the bPAG 1 test. The accuracy of detecting nonpregnant animals by both protein tests was limited by the relatively long half-life of these proteins after calving and by early embryonic mortality.

Between days 27 and 59 after artificial insemination (AI), 189 ultrasonographic pregnancy diagnoses were made in 56 dairy cows using a 7.5 MHz linear-array rectal transducer. Blood samples were withdrawn from a jugular vein on the day of AI, on day 21, and after each ultrasonographic examination between days 27 and 31, days 34 and 38, days 41 and 45 and days 55 and 59 after AI. Plasma concentrations of bovine pregnancy-associated glycoprotein 1 (bPAG-1) were measured by radioimmunoassay. The results showed that before day 31, ultrasonographic scanning was not very sensitive because six of the 30 calving cows were incorrectly diagnosed as non-pregnant. In five of these animals, the uterus was located far cranial to the pelvic inlet. Five of the cows examined between days 27 and 31 were pregnant on the basis of plasma bPAG-1 levels on the same day, using 0.5 ng/ml as the cut-off point. Plasma levels of bPAG-1 and progesterone proved that four of the cows which had early positive ultrasonographic diagnoses but did not produce a calf, were pregnant when they were examined.

A 5-MHz sector transducer (experiment 1) and a 7.5 MHz linear-array transducer (experiment 2) were used for early pregnancy diagnosis in two Hungarian dairy herds under field conditions. In the first experiment, ultrasound scanning was conducted three times at 3 or 4-day intervals between 24 and 33 days after artificial insemination (AI), while in the second experiment, it was performed two times at 7-day intervals between 27 and 38 days post-insemination. Significantly more incorrect non-pregnancy diagnoses were made between 24 and 33 days (first experiment) and between 27 and 38 days (second experiment) after AI in cows in which the uterus was located far cranial to the pelvic inlet than in animals in which the uterus was located within or close to the pelvic inlet at the first ultrasonographic examination.

The time from birth to attaining sternal recumbency (T-SR) was recorded for 219 newborn calves in order to evaluate its use as a parameter for diagnosing vitality. Calves were defined as vital if they received routine care without medical treatment and survived seven days from birth without any symptom of illness (n = 192). Those which did not fulfil these conditions were categorised as non-vital (n = 27). Ten calves were born spontaneously and the others were delivered either by caesarean section (n = 105), normal extraction (n = 78) or forced extraction (n = 26). The mean (+/- sd) T-SR values of the vital calves in these delivery groups were: 4.0 +/- 2.2, 4.5 +/- 3.1, 5.4 +/- 3.3 and 9.0 +/- 3.3 minutes, respectively. Jugular blood gas and pH measurements were performed at several fixed times after birth until pH and base excess had reached values of over 7.2 and more than -5.0 mmol/litre, respectively. Mean pH and base excess values of vital calves 10 minutes post partum were significantly higher and lower, respectively, than those of non-vital calves. Forcefully extracted calves had longer T-SR, more serious acidosis, recovered more slowly from acidosis, died more, and exhibited trauma more frequently. A T-SR of at least 15 minutes had a predictive value of 84 per cent for non-vitality, while 10 minute pH values of less than 6.9 had a lower predictive value, namely 68 per cent, for non-vitality. Correlations between T-SR values and 10 minute pH and base excess values were moderate, whereas correlations between T-SR values and PCO2 values were weaker still and much more dependent on the type of delivery. It is concluded that T-SR determination is a valuable, practicable and objective diagnostic tool for estimating the condition of newborn calves during the first 15 minutes of life. The diagnostic value of a single pH measurement on the jugular blood of a neonatal calf is doubtful.

The influence of luteolysis on myometrial activity in late pregnant cows was studied by measuring electromyographic (EMG) activity and concentrations of progesterone and 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) in maternal arterial plasma of five cows. Application of two ear implants containing progestagen at day 261 of gestation was followed by an injection of a luteolytic dose of a prostaglandin F2 alpha (PGF2 alpha) analogue (PG) on day 264. Calving was initiated by removal of the implants and a single injection of 5 mg flumethasone on day 270. All calves were born alive at a mean interval of 36 h. After 4 h of hyperactivity immediately following the injection of PG, myometrial activity was almost completely absent for 20 h; before luteolysis the total duration of EMG activity was 7.21 +/- 0.31 min h-1 (mean +/- SEM), during the 4 h after PG 23.61 +/- 3.40 min h-1, and during inhibition 2.70 +/- 0.87 min h-1. After the last period, EMG activity recurred and reached values of 14.52 +/- 3.07 and 11.17 +/- 2.87 min h-1, on days 265 and 269, respectively. Concentrations of progesterone in maternal plasma decreased from 4.17 to 1.14 ng ml-1 within 12 h after PG, whereas concentrations of PGFM in plasma remained low, varying from 15 to 30 pg ml-1. After removal of the implants and injection of flumethasone on day 270, EMG activity increased without an inhibitory phase and reached maximum values around the time of expulsion of the calves.(ABSTRACT TRUNCATED AT 250 WORDS)

Five cows in the last month of gestation, provided with uterine electrodes and in which catheters had been chronically installed in the fetal aorta, were used to study patterns of fetal heart rate (FHR) and the influence of periods of myometrial electrical activity during gestation (contractures) on FHR. The FHR was calculated by counting the number of blood pressure pulses on the tracings during alternate periods of 12 seconds. Three 1-hour recordings without contractures and 10 recordings during the time of a contracture were randomly selected for each cow. The calculated data points were plotted on a graph to display FHR patterns. In 41 periods associated with single contractures, FHR data points were taken every 72 seconds. Changes in absolute and relative FHR in these periods were determined to analyze a possible effect of contractures on FHR. Three types of variation in FHR patterns could be distinguished: a short-term, low-amplitude variation of basal FHR; a second type in which the duration was < 4 minutes and the amplitude was > or = 15 beats/min; and prolonged periods with increased or decreased FHR values (> 4 minutes and > or = 15 beats/min). The relationship between these types of variation and fetal activity states remains to be established for cows. During the 60 hours of recordings that were analyzed, a period of several minutes during which FHR values were extremely high (> 180 beats/min) was found 3 times. There were no significant differences in absolute or relative FHR before, during, or after a contracture.

Concentration of the acute-phase protein serum amyloid-A (SAA) was measured by means of an indirect ELISA method, in plasma of 3 pregnant cows and in plasma taken by cannula from another 4 pregnant cows and, by separate cannula, from their fetuses. Blood samples were taken daily from approximately 72 hours before until 72 hours after parturition. After parturition, SAA concentration increased significantly (P < 0.05) in maternal plasma. In fetal plasma, only a nonsignificant increase was found at time of delivery. The concentration of maternal SAA started to increase within the first 24 hours after delivery, reaching a peak value between 24 and 48 hours after delivery. In the aforementioned plasma samples from the 4 pregnant cows and their fetuses, the concentration of maternally derived cortisol increased nonsignificantly after parturition. The concentration of fetally derived cortisol was significantly (P < 0.05) increased at parturition (t = 0), compared with the initial fetal cortisol concentration at 120 hours before delivery. Peripartum concentration of maternal SAA increased and maternal cortisol remained low, whereas fetal SAA concentration remained low and fetal cortisol concentration increased. These findings indicate possible suppressive action of fetal cortisol on fetal SAA production. However, it might be argued that the main cause of the difference in SAA concentration is the difference in tissue damage between cows and fetuses at parturition.

To investigate whether calving could be controlled by the withdrawal of progestogen implants from cows treated near term with prostaglandin three cows (group 1) received an intramuscular injection of 5 mg flumethasone on day 270 of pregnancy, and four cows (group 2) and three cows (group 3) received implants containing 3 mg norgestomet in both ears on day 262 of gestation and were treated with a prostaglandin F2 alpha analogue on day 264. On day 270 the implants were removed and at the same time the cows of group 3 were treated with 5 mg flumethasone. Jugular blood samples were taken daily to estimate progesterone concentrations in the plasma. Luteolysis was achieved by the injection of prostaglandin, as judged by the decrease in plasma progesterone concentration in the cows of groups 2 and 3 on day 264. Pregnancy was maintained in these cows until after the removal of the norgestomet implants. The interval from the removal of the implants and, or, the injection of flumethasone on day 270 until the onset of second stage labour ranged from 36 to 47 hours and the mean intervals for the three groups were not significantly different. In all the cows except one from group 3 the dilatation of the cervix and vagina and the softening of the pelvic ligaments appeared normal at calving. It is concluded that calving near term can be synchronised by a progestogen in the absence of a corpus luteum.

The temporal relationship among changes of the concentrations of the 13,14-dihydro-15-keto metabolite of prostaglandin F2 alpha (PGFM), estrone (E1) and estrone sulphate (E1S) in maternal arterial plasma (MP) and amniotic fluid (AF), the prepartum progesterone (P4) decline in MP, and the evolution of uterine electromyographic (EMG) activity was investigated in 6 cows. Calving was induced by a single i.m. injection of 5 mg flumethason on Day 270 of gestation. The period under investigation was subdivided into four consecutive periods: Period 1 covered the last 2 days before flumethason treatment; Period 2 (mean +/- SEM duration: 16.1 +/- 2.5 h), Period 3 (8.8 +/- 1.1 h), and Period 4 (13.0 +/- 1.5 h) together included the interval between injection and the onset of the expulsive stage of induced parturition. Each was defined by its pattern of uterine EMG activity. During Periods 1 and 2, this activity occurred in long episodes (2-20 min; contractures) at a similar mean (+/- SEM) frequency (0.51 +/- 0.14/h and 0.42 +/- 0.07/h, respectively). No significant differences in hormonal concentrations in MP and AF between these two periods were detected. During Period 3, contractures nearly disappeared (freq: 0.09 +/- 0.05/h), and in MP mean P4 levels were significantly lower and PGFM levels were significantly higher than before. Mean PGFM concentrations in AF were not significantly changed during Period 3. Finally, during Period 4, EMG activity reappeared and a parturient EMG pattern gradually evolved in the presence of a further significant decline of P4 levels and significant increase of PGFM concentrations in MP.(ABSTRACT TRUNCATED AT 250 WORDS)

The acid-base status of 58 calves (all in normal anterior presentation) was determined from venous blood samples before the onset of traction and immediately after vaginal delivery. Calves were assigned to one of three groups according to their blood pH value: group 1 - normal, pH greater than 7.2; group 2 - acidotic, pH 7.2 to 7.0; group 3 - severely acidotic, pH less than 7.0. Before the onset of traction (i.e. during the 30 minutes following the appearance and rupture of the membranes), 43 calves (74.1%) had normal acid-base values, 14 (24.1%) had slight acidosis, and only 1 (1.7%) had severe acidosis. At birth the three groups of calves showed the following distribution: 23 (39.7%) were normal, 29 (50%) had slight acidosis, and 6 (10.3%) had severe acidosis. Seven calves (12.1%) died during birth or within 48 hours after birth; 2 were normal shortly before birth, 4 were acidotic shortly before birth, and 1 was severely acidotic even before the onset of traction. Traction was significantly longer for cows that delivered severely acidotic calves compared to cows in the other two groups.

Pregnancy diagnoses were performed under farm conditions in 201 cows (Hungarian Red Pied and crossbreds with Holstein) between 21 and 70 days after last insemination date, using a linear-array real-time ultrasound scanner with a 3.0 MHz rectal transducer. Identification of (a) sharply demarcated black area(s) and/or an embryo within the uterus were used as criterions for pregnancy. Pregnancy was confirmed by rectal examination at 6 and/or 8 weeks after A.I. From the total number of correct positive diagnoses (n = 129), incorrect positive diagnoses (n = 3, made on days 36, 40 and 44 after A I), correct negative diagnoses (n = 62) and incorrect negative diagnoses (n = 7, made on days 25, 28, 28, 29, 30, 31, and 33 after A I), a sensitivity of 94.8%, a specificity of 95,3%, a positive predictive value of 97.7% and a negative predictive value of 89.8% were calculated. From these preliminary results it was concluded that real-time ultrasound scanning is a useful and reliable technique for early pregnancy diagnosis in cows. It is suggested that a combination of milk/plasma progesterone estimation between days 21 and 24 and real-time ultrasound scanning between days 25 and 45 would allow the confirmation and quantification of (late) embryonic mortality in the cow.
