Our objectives were to determine the effects of early ovulation on fertility and uterine health of dairy cows. Four hundred and forty-five Holstein cows (185 primiparous and 260 multiparous) from five herds were used. Blood samples were collected at 21, 35 and 49 days in milk (DIM) and cows were considered to be cyclic at 21 DIM (Cyc21) if serum progesterone (P4) concentration was above 1 ng/ml, cyclic by 49 DIM (Cyc49) if P4 concentration was above 1 ng/ml at 35 or 49 DIM, or not cyclic (NotCyc) if P4 concentration was below 1 ng/ml at all sample times. Endometrial cytology for diagnosis of subclinical endometritis was examined at 49 DIM in a subset of 414 cows. Cows in the group Cyc21 had increased hazard of insemination, for the first service, compared with cows in Cyc49 [hazard ratio (HR) = 1.40; 95% CI = 1.10-1.79; p = 0.006] and NotCyc (HR = 2.07; 95% CI = 1.52-2.82; p < 0.001). Cows in the Cyc49 group also had increased hazard of insemination compared with cows in the NotCyc group (HR = 1.47; 95% CI = 1.13-1.93; p = 0.005). Median days to insemination were, respectively, 71, 76 and 96 for cows in Cyc21, Cyc49 and NotCyc groups. Cows in Cyc21 had greater first service pregnancy per AI than Cyc49 [38.6 vs 28.1%; adjusted odds ratio (AOR) = 1.67; 95% CI = 1.01-2.75; p = 0.04] and NotCyc (38.6 vs 23.6%; AOR = 2.08; 95% CI = 1.08-4.00; p = 0.03). Pregnancy per AI was similar in Cyc49 and NotCyc cows (28.1 vs 23.6%; AOR = 1.25; 95% CI = 0.70-2.24; p = 0.45). Cows in Cyc21 had increased hazard of pregnancy up to 300 DIM compared with Cyc49 (HR = 1.52; 95% CI = 1.17-1.96; p = 0.002) and NotCyc (HR = 1.98; 95% CI = 1.41-2.78; p < 0.001). Cows in Cyc49 tended to have increased hazard of pregnancy compared with NotCyc (HR = 1.31; 95% CI = 0.96-1.77; p = 0.09). Median days to pregnancy were, respectively, 103, 147 and 173 for cows in Cyc21, Cyc49 and NotCyc groups. Cows in the Cyc21 group had decreased prevalence of subclinical endometritis compared with cows in the NotCyc group (29.9 vs 43.7%; AOR = 0.53; 95% CI = 0.29-0.97; p = 0.04); however, the prevalence did not differ from the Cyc49 group (29.9 vs 39.1%; AOR = 0.68; 95% CI = 0.41-1.14; p = 0.15). Cyc49 cows had similar prevalence of subclinical endometritis compared with NotCyc cows (AOR = 0.77; 95% CI = 0.46-1.29; p = 0.32). The objective was to compare the reproductive efficacy of Ultrasynch, a synchronization program based on functionality of the corpus luteum as determined by ultrasonography, with an Ovsynch protocol. A randomized field trial was conducted on a commercial dairy in Cayuga County, New York, during scheduled weekly pregnancy examinations. Cows (n=745) determined nonpregnant 28 to 34 d after artificial insemination (AI) were randomly assigned to Ultrasynch or Ovsynch protocols. Cows assigned to the Ultrasynch management program (n=366) were treated based on corpus luteum (CL) diameter: cows with a CL > 23 mm received an injection of PGF(2alpha) and were bred via AI following detection of estrus (Ultra-PGF), whereas cows with a CL < or = 23 mm received injections and were bred on an Ovsynch protocol. Cows assigned to the Ovsynch management program (n=379) were placed on an Ovsynch protocol regardless of CL diameter. Pregnancy status was rechecked 28 to 34 d after AI; cows determined nonpregnant after initial enrollment were maintained in their assigned management group and received treatments based on CL diameter if in the Ultrasynch group and Ovsynch treatments if in the Ovsynch group. Hazard of pregnancy was similar between Ultrasynch and Ovsynch (hazard ratio=1.10, 95% confidence interval=0.88-1.36). Median days to conception were 98 and 87 for Ultrasynch and Ovsynch, respectively. The detection of estrus rate of cows in the Ultra-PGF group was 49%; better performance of an Ultrasynch management program may be achievable in a herd with a higher rate of estrus detection., (2010 American Dairy Science Association. The objectives were to evaluate the accuracy of detecting a functional CL by transrectal palpation and ultrasonography, and to optimize the accuracy of detecting a functional CL by ultrasonography in Holstein cows. In Experiment 1, four veterinarians performed transrectal palpation in 1250 cows at 37 d in milk (DIM), two veterinarians repeated transrectal palpation in 823 cows at 58 DIM, and one veterinarian performed 206 ultrasonographic examinations at 37 DIM. In Experiment 2, 987 and 983 ultrasonographic examinations were performed at 21 and 24 d after AI by one veterinarian for detection and measurement of CL. Cows with a blood progesterone concentration > or =1ng/mL were assumed to have a functional CL. Sensitivity and specificity were optimized using receiver operating characteristic analysis. In Experiment 1, sensitivity of transrectal palpation for diagnosing a functional CL ranged from 33.3 to 59.9% at 37 DIM and from 48.3 to 68.4% at 58 DIM, whereas specificity ranged from 76.7 to 93.2% at 37 DIM and from 73.3 to 86.7% at 58 DIM. Sensitivity and specificity for ultrasonography were 89.4 and 45.7%, respectively. In Experiment 2, the sensitivity and specificity of ultrasonography were 97.3 and 38.1% at 21 d after AI, and were 97.9 and 51.0% at 24 d after AI. Sensitivity and specificity were optimized using a cutoff diameter of 23mm at 21 d and 22mm at 24 d, which resulted in sensitivity and specificity of 87.2 and 83.0% at 21 d, and 89.5 and 89.4% at 24 d after AI, respectively. Sensitivity was low and specificity was high for transrectal palpation, whereas ultrasonography resulted in high sensitivity and low specificity. Using a cutoff diameter during ultrasonography improved accuracy of detection of a functional CL compared with either ultrasonography without cutoff or transrectal palpation.

Stillbirth is defined as calf mortality shortly before, during, or shortly after parturition. Economic losses to the dairy industry in the U.S. just due to the loss of replacement heifers were estimated to be $125 million per year. Our objective in this study was to evaluate the effect of stillbirth on the dam's subsequent lactation. Data were collected from 20 farms in the U.S.A. (19 from NY, one from IN) from May 2005 until September 2006. Calving-ease scores (scale of 1-5; no problem and extreme difficulty, respectively) and stillbirths were recorded by farm personnel. We used 32,299 calvings for the final analysis where 6.5% of the calvings were stillbirths. The incidence of stillbirths for primiparous cows was 10.7% (of 11,891) and 4% (of 20,408) for multiparous cows. Stillbirth parturition significantly reduced milk production (-1.1 kg/day). The losses in milk production were evaluated in a generalized mixed linear model analysis. The daily milk production for cows with stillbirths was 34.2 kg, compared to 35.3 kg for the cows that had a live calf. Milk-production reduction was greatest early in lactation and declined afterwards. Economic losses from stillbirths include not just the lost calf, the reduced survival, and increased days open but also the decreased milk yield.

The objectives of this study were to evaluate the effect of stillbirth on survival and reproductive performance of lactating dairy cows. Data were collected from 2 different regions of the US calving-ease scores (CES) were recorded by farm personnel on a scale of 1 (no problem) to 5 (extreme difficulty). Stillbirths were recorded by farm personnel. The final analysis included 13,608 calvings of which 93.4% were live calves and 6.6% stillbirths. An increasing or decreasing trend in the incidence of stillbirth by parity and by CES was analyzed by Cochran-Armitage trend tests. A significant decreasing trend in the incidence of stillbirth by parity group was detected. The incidence of stillbirth increased as the CES increased. The incidence of stillbirths was 3.6, 11.2, 25.9, and 60.1% for CES score 1, 2, 3, and 4, respectively. Dam survival in the herd and reproductive performance were analyzed by the Cox proportional hazards model. Variables that decreased dam survival time were stillbirths, primiparity, and CES of 3 and 4. The variables that reduced reproductive performance were stillbirths, multiparity, male calves, and CES of 3 and 4. Cows that had stillbirths had significantly increased risk of culling/death throughout the lactation and increased median days open by 88 d compared with cows that had live calves. In conclusion, losses from stillbirths are far greater than just the value of the calf.

The objective of the study was to determine the effects of treatment with a controlled internal drug-release (CIDR) insert containing progesterone in a PGF2alpha-based presynchronization protocol on pregnancy rates at first service in lactating Holstein cows. A total of 1,318 (656 treatment and 662 control) cows from 5 farms were used in the analysis. Cows received a CIDR insert as part of the presynchronization protocol of 2 PGF2alpha injections given 14 d apart. The CIDR insert was applied during 7 d before the second PGF2alpha injection, whereas control cows received no CIDR insert. Serum progesterone concentrations were measured in samples collected at 37 +/- 3 d in milk (DIM; 7 d after the first PGF2alpha injection) and at 58 +/- 3 DIM, just before initiation of the Ovsynch protocol. According to serum concentrations of progesterone, cows were classified as having either high (> or = 1 ng/mL) or low (< 1 ng/mL) progesterone. The proportion of cows with low progesterone at 37 +/- 3 DIM was similar for cows treated later with the CIDR insert (60.7%; n = 654) and for control cows (59.2%; n = 657). In contrast, use of the CIDR insert resulted in fewer low-progesterone cows (17.4%; n = 402) compared with control cows (30.6%; n = 399) at 58 +/- 3 DIM. No significant effect of the CIDR insert was detected on overall pregnancy rates. Pregnancy rates, as measured by the percentage of cows pregnant at 37 +/- 3 d post timed artificial insemination, for control cows having high or low progesterone at 58 +/- 3 DIM were 46.6 and 22.1%, respectively. For the CIDR group, pregnancy rates were 40.4 and 11.4%, respectively, for high- and low-progesterone cows at 58 +/- 3 DIM. Overall pregnancy rates were 36.4 and 34.5% for control cows and cows receiving the CIDR insert, respectively. A significant decreasing trend was observed in the proportion of cows having low progesterone as the body condition score increased, at 37 +/- 3 and 58 +/- 3 DIM. A significant increasing trend in the pregnancy rate was observed as body condition score increased. In conclusion, incorporation of CIDR inserts into a presynchronization protocol reduced the proportion of cows having low progesterone; however, the pregnancy rate did not differ between control cows and those receiving the CIDR insert. Earlier expression of estrus after the second PGF2alpha injection, and consequently improper timing of initiation of the Ovsynch protocol, could have negatively affected fertility in the CIDR-treated cows.

Objective: To determine whether a commercially available water hardness test kit could be used to measure total serum calcium concentration and diagnose hypocalcemia in dairy cows., Design: Prospective study., Animals: 30 dairy cows from 19 commercial herds., Procedure: Serum calcium concentration was determined using a water hardness test kit and a standard, laboratory-based method. Simple linear regression was used to determine whether there was a linear relationship between results of the 2 methods, and Spearman's rank correlation was used to calculate correlation between measurements. Sensitivity, specificity, and predictive values of using test kit-derived values for diagnosis of hypocalcemia (laboratory value < 8 mg/dl) were calculated., Results: There was a high correlation and significant linear relationship between results of the 2 methods. Sensitivity, specificity, predictive value of a positive test result, and predictive value of a negative test result were 100, 73, 86, and 100%, respectively. Accuracy was improved by using a test kit-derived calcium concentration of 7 mg/dl as the cut-off for determining hypocalcemia., Clinical Implications: Results indicate that a commercially available water hardness test kit can be used as a rapid, inexpensive method of estimating serum calcium concentrations and diagnosing hypocalcemia in dairy cattle. However, the test is not practical for cow-side use, because blood samples must be centrifuged to obtain serum for use in the test kit.

This study compared three programs for reproductive management of the postpartum period for reproductive performance and net economic benefit within three dairy herds (n = 1624 cows). Cows on one program received PGF alpha injection at 25 to 32 d postpartum for reproductive therapy, and cows on a second program received additional PGF2 alpha at 39 to 46 d postpartum for synchronization of estrus. These programs were compared with a postpartum program of rectal palpation based on veterinary intervention. Survival analysis indicated that cows receiving PGF2 alpha for reproductive therapy and synchronization of estrus had an 11% higher rate of first AI and 10% higher rate of pregnancy than did cows receiving the rectal palpation. No differences existed between the cows receiving rectal palpation and those receiving the PGF2 alpha. Because overall conception rates and conception rates at first AI did not differ among programs, the improved reproductive performance of cows receiving PGF2 alpha for both therapy and synchronization may be attributed to greater synchronization of estrus, which resulted in improved estrus detection. A partial budget indicated that the PGF2 alpha programs were less expensive than the rectal palpation program. When PGF2 alpha was used for postpartum reproductive therapy and synchronization of estrus, reproductive performance and net economic benefit were increased compared with those of the other programs.

The objective of this article was to review the current literature of the influence of gonadotropin releasing hormone (GnRH) and GnRH agonists on conception rate in dairy cattle. The application of GnRH or agonists at artificial insemination (first and subsequent) and between days 7 and 34 after parturition were considered. The variations between studies were discussed as well as different mechanisms concerning the influence of GnRH for establishing pregnancy. From a critical point of view the routine use of GnRH or agonists at the time of first or subsequent breeding or during the postpartum period cannot be supported.

Two hundred and fifty-one cows fresh at least 37 days with normal reproductive tracts and palpable ovarian structures which clinicians believed to be corpora lutea and which were presumed to be producing progesterone were treated with 500 micrograms of cloprostenol. The cows were stratified into three groups based on milk progesterone concentrations in whole milk samples taken at the time of examination. The low group had milk progesterone concentrations less than or equal to 1 ng/ml, the intermediate group had milk progesterone concentrations between 1 and 3 ng/ml, and the high group had milk progesterone concentrations greater than or equal to 3 ng/ml. The proportion of cows in each group inseminated within 5 days of treatment and the fertility at that breeding were compared. There were no significant differences among the groups with respect to the proportion of cows that came into estrus within 5 days. However, the conception rate of the high group (54%) was significantly greater than that of the intermediate group (26%). To study the relationship between practitioner experience and diagnostic accuracy the proportion of cows with low, intermediate and high concentrations of progesterone selected by clinicians with more than 3 years experience was compared to that for clinicians with 3 years of experience or less. Less experienced clinicians were significantly more likely to make a false positive diagnosis of functional luteal tissue in cows with palpable ovarian structures than were more experienced clinicians.

Sulfadiazine (SDZ)/trimethoprim (TMP; 30 mg of SDZ/TMP/kg of body weight) was given IV to the same 6 male calves at 1, 7, and 42 days of age and to 2 additional calves at 7 days of age. Serum concentrations of SDZ and TMP were best represented by a 2-compartment open model, but in 42-day-old calves, CSF concentrations of both drugs were best represented by a 1-compartment open model with first-order input. Between 1 and 42 days of age, the elimination half-life (t1/2(beta)) of SDZ decreased from 5.7 to 3.6 hours, and total body clearance (CLtot) increased from 1.43 to 1.88 ml/min/kg; the area under the curve (AUC0----infinity) decreased from 291.5 to 225.4 mg/L.h. The distribution coefficient (Vd(area)/kg of body weight) decreased with age, changing from 0.72 to 0.59 L/kg, between 1 and 42 days of age. Therapeutic concentrations of SDZ in serum (greater than 2 micrograms/ml) were maintained for 24 hours in 1-day-old calves and for about 15 hours in 7- and 42-day-old calves. The elimination rate of TMP increased about 9-fold; t1/2(beta) was 8.4, 2.1, and 0.9 hours, respectively, at 1, 7, and 42 days of age. Other values also reflected an increase in TMP elimination rate with age: CLtot increased from 2.8 to 12 to 28.9 ml/min/kg, k13 increased from 0.336 to 0.654 to 1.664/h and AUC0----infinity decreased from 32.8 to 7.9 to 3.1 mg/L.h, respectively. Therapeutic concentrations (greater than 0.1 microgram/ml) were maintained for 15 hours, 8 hours, and about 6 hours in 1-, 7-, and 42-day-old calves, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Path analysis was used to model the direct and indirect relationships among age, previous lactation yield or estimated transmitting ability for milk, body weight, days dry, reproductive disorders, milk fever, mastitis, reproductive performance, current milk yield, and culling. Prospective data were from 784 primiparous and 2,066 multiparous Holstein lactations from 33 herds. Heifers that were older, of lighter weight, or who had lower estimated transmitting ability for milk had more problems, less milk, and poorer survival. Dystocia in heifers had several detrimental consequences including 2.9 to 4 times more retained placenta, metritis, and culling and +7.4 d more to first service. Cystic ovaries were associated directly with 376 kg greater milk yield and with a 16.5-d delay in first service. Failure to conceive at first service and mastitis increased risk of culling 5.2 to 10 times. In multiparous cows, milk fever increased risk of reproductive disorders by 1.6 to 4.2 times and indirectly contributed to poor breeding performance and increased culling. Risk of culling was increased 2.1 to 3.7 times directly by mastitis and dystocia and by poor breeding performance.

Age-related changes in the absorption and distribution patterns of trimethoprim/sulfadiazine were studied following oral or subcutaneous administration of 15 mg/kg of the drug combination in calves. Following oral administration, the time course of trimethoprim/sulfadiazine appearance and dissipation in serum, synovial fluid and urine was followed for periods up to 48 hours in calves one day, one week and six weeks of age. The profiles of drug appearance-disappearance in these body fluids were also determined after subcutaneous administration in seven week old calves. The peak serum and synovial fluid levels of trimethoprim/sulfadiazine achieved following oral administration were substantially lower with increasing maturation. In ruminating (six and seven week old) calves, subcutaneous or oral administration of the combination led to high serum levels of sulfadiazine but little or no serum trimethoprim was detected in animals at this age. The data indicate that, while therapeutic concentrations and optimum ratios of the drugs may be achieved for extended time periods in neonatal life, this dosage is unable to produce optimum serum and synovial fluid concentrations as the calves mature.

Thirty milligrams per kilogram of sulfadiazine/trimethoprim (SDZ/TMP, Tribrissen) was given orally and subcutaneously (s.c.) to two groups of male, Holstein calves. One group was fed milk-replacer throughout the 13-week period of the study while the second group was weaned onto a chopped grain-fiber mixture when 5 weeks old. Serum and urine were assayed for concentrations of unchanged drug. Trimethoprim bioavailability, following oral administration at 1, 6 and 12 weeks of age, is higher in milk-fed calves (non-ruminants) than in grain-fiber-fed calves (ruminants); bioavailability decreases with increasing age in both groups of calves. Serum concentrations above 0.1 micrograms/ml (the level of sensitivity of the assay) could not be obtained in ruminating calves. The rate of SDZ absorption following oral administration, as determined by the Wagner-Nelson method, was very slow in all the calves in this study with average half-life values ranging from 8.2-12.67 h; absorption was slightly faster in ruminating calves. Absorption of SDZ is rate-limiting and determines the biological half-life of the drug; SDZ serum concentrations above 2 micrograms/ml were maintained in all calves for at least 24 h. Following s.c. administration of Tribrissen to 7-and 13-week-old calves, urinary excretion patterns indicated that TMP was slowly released from the injection site; serum concentrations were below 0.1 micrograms/ml. In contrast, absorption of SDZ was very rapid; values for tmax were 1.5-1.8 h. The pharmacokinetic parameters for SDZ were calculated according to a one-compartment open model; neither diet nor age had a significant effect on SDZ disposition following s.c. injection. Subcutaneous administration of 30 mg/kg Tribrissen, b.i.d., may be the best therapeutic regimen; even though measureable concentrations of TMP cannot be achieved in the serum following a single s.c. dose, TMP concentrations should accumulate and, because of its sustained release, provide almost continual potentiation of SDZ.

Abomasal fluid was collected from adult cows using a specially designed tube device. The device was placed in the abomasum through a rumen fistula. A filter on the end of the tube filtered out the larger particles so the liquid could be aspirated up a smaller tube placed inside a larger one. The smaller tube reduced the amount of dead space created by the distance from the abomasum to the rumen fistula.
