Among several potential animal models that can be used for adipogenic studies, Wagyu cattle is the one that presents unique molecular mechanisms underlying the deposit of substantial amounts of intramuscular fat. As such, this review is focused on current knowledge of such mechanisms related to adipose tissue deposition using Wagyu cattle as model. So abundant is the lipid accumulation in the skeletal muscles of these animals that in many cases, the muscle cross-sectional area appears more white (adipose tissue) than red (muscle fibers). This enhanced marbling accumulation is morphologically similar to that seen in numerous skeletal muscle dysfunctions, disease states and myopathies; this might indicate cross-similar mechanisms between such dysfunctions and fat deposition in Wagyu breed. Animal models can be used not only for a better understanding of fat deposition in livestock, but also as models to an increased comprehension on molecular mechanisms behind human conditions. This revision underlies some of the complex molecular processes of fat deposition in animals.

One hundred-sixty Holstein growing-finishing steers (initial BW of 185 kg) were blocked by BW to determine the effectiveness of long-term bovine somatotropin (bST) administration on lean, skeletal, and carcass measurements. Steers were randomly allocated to 4 treatments (10 steers/treatment) within a block (n = 4 blocks). Treatments were control, no bST (C-C); bST from d 0 to 182 (bST-C); bST from d 183 to slaughter (C-bST); and bST from d 0 to slaughter (bST-bST). Steers received a s.c. injection of placebo or bST at 14-d intervals. Doses were 320 mg of bST/injection from d 0 to 112 and 640 mg of bST/injection from d 113 to slaughter. The last treatment was administered 31 d before slaughter. Steers received a 14% CP (DM basis) diet from d 0 to 182 and 11.5% CP from d 183 to slaughter that consisted of dry, whole-shelled corn and a pelleted protein-mineral supplement. Steers were slaughtered when BW per block averaged 615 kg (d 325, 353, 367, and 381 for the 4 blocks, respectively). Thirty steers were removed from the study because of poor performance with respect to their pen mates, illness, lameness, death, incomplete castration, and incorrect treatment. Serum IGF-I concentrations increased 151% (P < 0.01) from d 7 through 35 in bST-treated steers compared with control steers. During the first 182 d, bST-C and bST-bST steers were heavier (P < 0.01) and had greater (P < 0.01) ADG, G:F, hip height, and hip height gain compared with C-C and C-bST steers. From d 183 to slaughter, C-bST steers had reduced (P < 0.05) daily DMI and greater G:F than bST-C steers. At final slaughter, C-bST and bST-bST steers had greater (P < 0.05) hip height than C-C steers. Noncarcass weight was increased and dressing percent reduced (P < 0.05) in C-bST and bST-bST steers compared with C-C steers. Quality grade was least (P < 0.05) in bST-bST carcasses compared with C-C, whereas bST-C and C-bST carcasses were intermediate. At final slaughter, steers receiving bST had greater (P < 0.05) carcass protein and water composition and lower (P < 0.05) carcass lipid and lipid accretion than C-C steers. Bovine somatotropin was effective in reducing carcass fat and increasing edible lean. Administering bST to young, lightweight steers increased skeletal growth and noncarcass weight without an increase in total carcass weight, but decreased carcass quality.

Forty-one Holstein cows were injected with 0, 5, or 14 mg/d of bST for the last 46 +/- 6 d before parturition. Compared with data for controls, the 5- and 14-mg doses of bST increased apparent protein synthesis about 16% before parturition. Exogenous bST before parturition increased apparent protein degradation 30% during wk 1 after parturition. During wk 1 of lactation, 14 mg of bST also increased milk protein yield 33%. No treatment differences were present in concentration of serum NEFA, body condition score, or thickness of subcutaneous fat. Therefore, administration of bST before parturition did not alter metabolism of subcutaneous fat. Prepartum treatment with 5 and 14 mg of bST increased and maintained serum somatotropin at 6.5 and 22.7 ng/ml, respectively, compared with 1.6 ng/ml in controls. Concentrations of serum IGF-I were initially increased but were not maintained as parturition approached. On d -23, IGF binding protein 3 was increased 65% but was not different among groups by d -7. For groups administered the 5 and 14 mg/d of bST, IGF binding protein 2 was decreased 40%. Administration of bST before parturition increased protein reserves and stimulated milk protein yield for 1 wk but did not alter metabolism of subcutaneous fat. Furthermore, energy balance appeared to be a major regulator of concentrations of IGF binding protein 3 and responsiveness of IGF-I to exogenous somatotropin before parturition.

Skeletal muscle protein degradation, measured by urinary N tau-methylhistidine excretion, and circulating concentrations of growth hormone (GH), insulin (INS), and cortisol (CT) were monitored in steers before and after implantation with estradiol-17 beta (E2; 24 mg) and trenbolone acetate (TBA; 300 mg). Yearling crossbred steers (n = 43) were randomly assigned to four treatment groups in a 2 x 2 factorial arrangement: nonimplanted controls (C); TBA; E2; and TBA plus E2 (TBA+E2). A subgroup (Block 1) of 16 steers was bled on d -12, 31, and 72 after implanting. Deposition of skeletal muscle protein was markedly increased (P less than .001) by E2 and TBA+E2 treatment. This response occurred mainly within the first 40 d after implantation and declined (P less than .001) in concert with decreasing (P less than .01) concentration of serum E2. Anabolic steroid treatment did not affect the rate of skeletal muscle protein breakdown. There was no apparent relationship between reduced serum CT concentration (linear effect; P less than .01) in TBA-treated steers and skeletal muscle protein degradation rate. Blood concentration and pulse activity of INS were not affected by anabolic steroid administration. Both TBA- and TBA+E2-implanted steers displayed a linear decrease (P less than .05) in serum GH concentration over time, which was similar to C. Lowered mean GH concentration resulted from a reduction (TBA main effect; P less than .05) in pulse amplitude of GH. Unlike TBA, TBA+E2, and C, only E2 maintained serum GH concentrations over time. Although increased muscle protein deposition was evident in TBA+E2-treated steers, an obvious causal relationship between this response and circulating GH, INS, and CT was not revealed. These results do not support the concept that combined androgenic agent and estrogen administration effectively reduce bovine muscle protein degradation by static modulation of circulating endogenous anabolic and antianabolic hormones.

Two experiments were conducted to ascertain lysine required for maintenance and milk production in lactating dairy cows. Multiparous Holstein cows fitted with abomasal cannulas were utilized in a replicated 3 x 3 extra-period Latin square design and infused with 0, 45, or 90 g of L-lysine-HCl daily (Experiment 1). In Experiment 2, 12 cows were utilized in a 5 x 5 Latin square design (with 2 replacement cows) and infused with 0, 22.5, 45, 90, and 180 g of L-lysine-HCl daily. Cows were housed in comfort stalls, milked at 0600 and 1700 h, exposed to a 16:8 h light:darkness cycle, and fed for ad libitum intake a 15.7% CP diet formulated to contain approximately 75% of dietary protein from corn-based feed ingredients. Milk yield was not affected in Experiment 1, but milk production increased linearly with lysine infusion level in Experiment 2. A linear response in daily yield of milk protein to lysine infusion was observed in both experiments. As lysine infusion increased, coccygeal plasma lysine concentrations tended to increase in Experiment 1, but in Experiment 2 free lysine increased linearly and quadratically in coccygeal plasma and increased linearly in plasma from the subcutaneous abdominal vein. The quadratic response in plasma free lysine (Experiment 2) suggested that lysine was the limiting AA for milk production from the composite protein flow to the small intestine. Using break-point analysis of plasma free lysine, it was estimated that the additional amount of abomasal lysine required to satisfy requirements was 64 g/d in cows producing about 30 kg of milk daily. Although additional lysine above microbial and undegraded intake protein lysine flow stimulated milk protein production, conversion efficiency to milk protein lysine was low.

Six lactating Holstein cows, fitted with T-type cannulas in the proximal duodenum, were used in a replicated 3 x 3 Latin square design to determine influence of supplemental protein on amino acid profiles of duodenal chyme and plasma. Protein sources were blood meal, corn gluten meal, and cottonseed meal, which furnished approximately 46% of the total protein in corn-grain corn silage diets. Markers were 15N to estimate rumen bacteria and chromic oxide to estimate nutrient flow. Dry matter intake was lowest on blood meal. Duodenal flow of N exceeded N intake 23% on blood meal and corn gluten meal. Percentages of organic matter and protein digested in the rumen were 56.5, 41.8; 61.2, 53.4; and 50.6, 56.2 for the respective diets. Variation in patterns of amino acids flowing to the duodenum, but not in coccygeal blood, closely reflected dietary differences with six of eight essential amino acids highest for the same treatment in both diet and duodenal chyme. Essential amino acids in least abundance for milk production, measured by mammary extraction coefficients, differed among diets. We conclude that supplemental protein source influences greatly the quantity and pattern of amino acids available for digestion in lactating dairy cows.

Digesta containing microbial protein, undegraded (bypass) feed protein, some free amino acids and oligopeptides, and microbial nucleic acids passes from the rumen-reticulum through the omasum to the lower gut (abomasum and small intestine). The rate of neutralization of abomasal digesta flow is slower in ruminants than in nonruminants, and activation and peak activity of the pancreatic proteases is delayed to the mid jejunum. The overall apparent digestibility in the small intestine of proteins and microbial or exogenous nuclei acids is 60-70% and 75-85% respectively. Absorption mechanisms in ruminants for protein and nucleic acid digestion end products appear similar to those described in nonruminants, but the highest rate of amino acid uptake was found in the mid to lower ileum. Essential amino acids (Met, THr, Val, Ile, Leu, Phe, Lys, His, Arg) are absorbed faster than nonessential amino acids (Gly, Glu, Asp, Ser, Pro, Cys, Tyr). After feeding, plasma amino acids remain unchanged or decline in a mature ruminant, while in nursing lambs, plasma free amino acids levels increase after a meal.

Tissue free amino acid pools account for only a small fraction of the total amino acid content in the body. Blood of plasma free amino acid pools represent only a very small proportion of tissue free amino pools. Plasma free amino acid profiles have been used as a tool to study nutritional and metabolic aspects of protein status. Static measurements of plasma amino acid levels have limited value as sensitive indicators of protein status because such measurements do not reflect the magnitude of amino acid fluxes in and out free amino acid pools. Use of plasma amino acid profiles, under well defined experimental conditions, in assessing limiting amino acids and essential amino acid requirements in ruminants is discussed.

Isolated porcine and bovine muscle strips were incubated in Krebs Ringer bicarbonate buffer to determine in vitro protein synthesis (PS) and protein degradation (PD) rates to validate the in vitro system for use with livestock species. The addition of 5X plasma concentrations of amino acids to the medium stimulated PS 30%. Addition of 3.5 mM leucine to a leucine-deficient buffer supplemented with amino acids decreased PD 37% and stimulated PS 24%. The addition of .1 U/ml insulin reduced PD 28% and increased PS 30%. Protein degradation was elevated in longitudinally split rat soleus and extensor digitorum longus muscles compared to their contralateral intact muscles. Muscle strips must be removed within 15 min of exsanguination because PD rates become greatly elevated thereafter. ATP concentrations declined during incubation, but the addition of ATP or creatine had no effect on either PD or PS. Neither PD nor PS was affected by the addition of transferrin, fetuin, ascorbate, dexamethasone or indomethacin to the incubation medium. However, muscle strips were sensitive to the addition of triiodothyronine (T3), PD was increased up to 75% as T3 concentration was increased, and PS rates doubled compared to controls. Serum from mature barrows or gilts had no effect on protein turnover, but the addition of 10% and 15% serum from boars increased both PD and PS. With fasted pigs a continual decline in PS occurred over 5 d, whereas PD was elevated at 3 d and then declined to rates comparable to the fed state after 5 d. These data suggest that the in vitro system has application for assessing relative changes that occur in vivo following nutritional, physiological and endocrinological manipulation.

N tau-methylhistidine (N tau MH) was used as an index for muscle protein degradation and this index was utilized to evaluate degradation rates in young growing cattle. Initially, two Charolais crossbred heifers, 12 months of age, were used to measure the recovery of radioactivity in the urine for a 120-hour period after intravenous injection of [14C]N tau MH. Of the radioactivity injected into the animals, 89.7% was recovered after 120 hours. With rate and amount of clearance as the criteria, the excretion of N tau MH in urine appears to be a valid index of muscle protein degradation in cattle. Eight steers of two genetic types were used to evaluate the effect of frame size on turnover rates of muscle proteins with N tau MH as an index. Large frame cattle (LG) excreted more N tau MH per day (P less than 0.03) throughout the trial. Total daily creatinine excretion was less for small frame (SM) cattle (P less than 0.02) showing an increase with time (P less than 0.01) in LG and SM cattle. N tau MH-to-creatinine ratios showed a decline with time. Fractional breakdown rates (FBR) and fractional synthesis rates (FSR) appeared to parallel each other with rates tending to decrease with age. No differences were observed between LG and SM cattle for FBR, FSR or fractional growth rate (FGR).

Feeding for 150 days of 25 g per day of methionine hydroxy analog was tested on 100 cows (52 treated and 48 controls) in the Brigham Young University herd. Effect of days after parturition of initiating methionine hydroxy analog feeding also was observed. Milk yields were not affected by methionine hydroxy analog, but fat percent and fat yields were increased 21 and 17% by the additive. Abnormally low milk fat by control cows (2.84%) magnified the response to methionine hydroxy analog feeding. Feeding methionine hydroxy analog beginning 0 to 12 days postpartum elicited much larger increases of milk fat than started later (17 to 102 days). Blood from the coccygeal vein of 20 cows from each treatment had 10% more triglycerides from cows fed methionine hydroxy analog than from control cows. Increases of arterio-venous differences across the mammary gland of triglycerides and lipoproteins of blood serum suggested that increases of milk fat could have resulted from greater uptake of performed fat by the udder. Feeding methionine hydroxy analog increased methionine, isoleucine, and leucine in serum of coccygeal vein, but methionine was the only amino acid with significantly higher arteriovenous differences across the mammary gland.

Two experiments were conducted to evaluate the effects of dietary CP level on rate, efficiency and composition of gain of growing beef bulls. In Exp. 1, 59 bulls (333 +/- 15.8 kg) were used. Eleven bulls were slaughtered on d 0 to provide an estimate of initial carcass composition (9-10-11 rib section chemical analyses), and remaining bulls were assigned to treatment diets containing 10, 12 or 14% dietary CP. Bulls fed the 10% CP diet grew slower (P less than .05) than bulls fed the 12 or 14% CP diets, although dry matter intake and feed-to-gain ratio did not differ. Bulls fed the 12% CP diet had fatter carcasses (P less than .05) than bulls fed the 10 or 14% CP diets and had greater daily fat accretion than bulls fed the 10% CP diet. In Exp. 2, 60 bulls (318 +/- 9.0 kg) were used. Bulls were assigned to initial slaughter (n = 6) or to one of three dietary treatments, 10, 12 or 14% CP, and were slaughtered after feeding for 66, 136 or 202 d (n = 6 . treatment -1 . slaughter time -1). Bulls fed 10% CP diets had lower (P less than .05) rates of carcass protein accretion during d 0 to 136 and d 0 to 202. Carcass fat gain was similar among treatments over the entire experiment, although bulls fed the 14% CP diet gained more fat during d 0 to 136 than bulls fed the other treatments.(ABSTRACT TRUNCATED AT 250 WORDS)

Twenty-one Simmental crossbred bulls (311 +/- 11 kg, 9 mo of age) were used to determine the effect of feeding 10, 12 or 14% CP on concentration of hormones in blood and the relationship of these hormones to composition of gain. Six bulls were slaughtered on d 0 to provide an estimate of initial carcass composition (9-11 rib section). Remaining bulls were assigned to dietary treatments. Blood samples were collected every 30 min from 0800 to 2000 on d 0, 66, 136 and 202 of treatment; bulls were slaughtered on d 203. Across all treatments, growth hormone (GH) declined (P less than .05) from d 0 to d 202. Free insulin-like growth factor I (IGF-I) was lowest (P less than .05) on d 0. In four randomly selected bulls, IGF-I fluctuated during the 12-h sampling periods. Within each treatment group, insulin was greatest on d 202 (P less than .05). Testosterone (T) increased from d 0 to d 66, then declined. Cortisol (C) was lowest on d 66. Thyroid hormones increased (P less than .05) after d 0. Growth hormone and IGF-I were correlated negatively with carcass fat percentage, fat accretion rate and fat thickness. IGF-I concentrations were correlated positively with percentage of carcass protein. Testosterone:cortisol ratio was not related to composition, but high T coupled with low C may be related to carcass leanness (mean carcass fat = 24.4%). These data suggest that GH and IGF-I are the hormones most related to composition of gain in growing beef bulls.

The effects of tallow supplementation [0% (NT) vs 7.5% (T)] and crude protein level [8.5% (LP) vs 12.0% (HP)] on ruminal fermentation, microbial protein (MCP) synthesis, digesta passage and site of digestion were estimated using yearling Angus X Simmental steers (390 kg) fitted with ruminal and T-type duodenal cannulae. Chromium-EDTA and ytterbium (Yb) chloride were used as markers of the liquid and solid phases. Passage and site of digestion data were estimated from the concentrations of Yb in the duodenal digesta and feces. Dry matter (DM) intakes were 6.8, 6.5, 6.3 and 6.6 kg/d for the NT-LP, NT-HP and T-HP diets, respectively. Ruminal ammonia concentrations (mg/100 ml) for the NT-LP, NT-HP, T-LP and T-HP diets were 1.22, 4.75, 1.05 and 3.41, respectively. Tallow decreased (P less than .05) acetate (mol/100 mol), increased (P less than .01) propionate (mol/100 mol) and decreased the total volatile fatty acid concentration. Tallow depressed apparent ruminal DM and organic matter (OM) digestibilities only on the HP diet. High protein increased ruminal DM, OM and fiber digestibilities. Tallow and LP tended to shift the site of OM digestion to the lower tract. The liquid and solid dilution rates for the NT-LP, NT-HP, T-LP and T-HP diets were 9.53, 3.37; 5.63, 3.28; 6.66, 5.10 and 6.79, 5.34%/h, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of photoperiod on growth, carcass composition and serum concentrations of prolactin, growth hormone and cortisol were determined in prepubertal and postpubertal Holstein heifers. Forty-two prepubertal (avg body wt 84 +/- 3.0 kg) and 42 postpubertal (avg body wt 303 +/- 7.1 kg) Holstein heifers were utilized. Ten prepubertal and 10 postpubertal heifers were slaughtered before treatment began to obtain initial carcass data. The remaining 32 prepubertal and 32 postpubertal heifers were paired by body weight and randomly assigned to short-day (8 h of light: 16 h of dark) or long-day (16 h of light: 8 h of dark) photoperiods. After exposure to treatments for an average of 139 d, 10 prepubertal and 10 postpubertal heifers from each photoperiod treatment were slaughtered. In prepubertal heifers, photoperiod did not affect (P greater than .10) average daily body weight gain, carcass weight, carcass composition, accretion of carcass fat and carcass protein, or serum concentrations of prolactin, growth hormone or cortisol. However, prepubertal heifers exposed to long-day photoperiods had reduced (P less than .01) urinary N tau-methylhistidine excretion compared with heifers given short-day photoperiods. Postpubertal heifers exposed to short-day photoperiods had greater average body weight daily gain than animals exposed to long-day photoperiods. Although there was no effect of photoperiod (P greater than .10) on carcass or fat depot weights, postpubertal heifers exposed to short days had greater (P = .06) percentages of fat and reduced (P = .07) percentages of protein in the soft tissue of the 9-10-11 rib sections. Fat accretion was greater (P less than .05) in carcasses of postpubertal heifers exposed to short days than heifers given long-day photoperiods, but there was no effect (P greater than .10) of photoperiod on protein accretion. Photoperiod did not affect serum concentrations of growth hormone and cortisol, but serum prolactin tended (P less than .10) to be greater in postpubertal heifers exposed to long days. Under the conditions of this experiment, we conclude that exposure to short-day photoperiods stimulated body weight gain and fat accretion in postpubertal but not prepubertal Holstein heifers.

Eighty-four Holstein cows in early lactation fed diets varying in protein percent (11, 14, and 17% crude protein) and degradability in the rumen (normal and heated soybean meal) were sampled for effects on amino acids and growth hormone in blood plasma, and amino acid profiles in the duodenal digesta. Essential and branched chain amino acids in plasma increased as protein and amount of protected protein in the diet increased. These changes were accompanied by decreased concentrations of nonessential amino acids. Plasma growth hormone was highest at low protein (11.3%) probably because of decreased intakes on this diet. Molar percentages of amino acids in duodenal contents were not affected by diet suggesting that our heat-treated soybean meal influenced quantity more than quality of amino acids available for absorption in the small intestines.

Experiments were conducted to evaluate the effects of dietary energy density or genetic background on protein and fat gain of growing cattle. In Exp. 1, 24 Limousin steers were used in a growing-finishing trial. A 2 X 2 factorial arrangement was used with steers randomly allotted to four treatment combinations and fed the following diets: 80% concentrate, high moisture corn-corn silage diet (HI) or a corn silage diet (LO) during both the growing (GRO) and(or) the finishing (FIN) phases. Body composition for both experiments was determined by a deuterium oxide dilution technique. Empty body weight gains were greater (P less than .05) for HI during GRO, FIN and the total trial. Daily protein gains (DPG) were greater (P less than .05) for HI during GRO and FIN, while cattle receiving HI during at least FIN had the greatest (P less than .05) overall DPG. Daily fat gains (DFG) followed the pattern of DPG, being more rapid (P less than .05) for cattle fed HI during either GRO or FIN. Cattle fed the HI diet also tended to be more energetically efficient. In Exp. 2, large frame (LG) and small frame (SM) cattle were used for the evaluation of frame size effects on protein and fat deposition. Steers were individually fed an 80% concentrate, corn-based diet during the entire trial. Average daily gains and daily dry matter intake (P less than .05, P less than .01) were greater for LG, while feed efficiency was similar for both cattle types. Large cattle had greater (P less than .05) DPG than SM cattle, however, DFG were not different. Small frame steers were energetically more efficient (P less than .05), apparently due to composition of gain difference.

The effects of crossbreeding, cattle type and dietary energy level on semitendinosus muscle (ST) development, feedlot performance, daily carcass protein and fat gain and serum anabolic hormone concentrations were studied. Over 3 consecutive years, 176 feedlot steers representing four cattle types - unselected Hereford (UH), selected Hereford (SH), Angus x Hereford x Charolais (AHC) and Angus x Hereford x Holstein (AHH) - were fed either an all-corn silage (HS) or a high grain (HG) diet. Steers were slaughtered on day 1 and at the end of the feedlot trial, and ST muscles were removed rapidly. During years 2 and 3, single blood samples were obtained from steers on days 1, 29, 57, 113 and 169, and analyzed for insulin and growth hormone (GH). Steers fed HG had a higher (P less than .005) average daily gain (ADG) than steers fed HS, and cattle type had an effect (P less than .005) on ADG. Cattle type and HG affected (P less than .005) daily carcass protein and fat gain. Weight of ST muscle and total muscle RNA, DNA and protein content increased with frame size, and HS steers had heavier (P less than .05) ST muscles than the HG steers. Steers fed HG had higher (P less than .01) serum insulin concentrations than steers fed HS, but there were no consistent cattle type effects. Serum GH concentrations were not affected by cattle type or diet. Serum insulin concentrations, combined across diet and cattle types, were correlated (P less than .01) with ADG; however, serum GH, assessed on the same basis, was not related to ADG. Average daily protein and fat gain were positively related to serum insulin and were negatively related to serum GH.
