This article describes a selective reduction of functionalized amides, including N-acyl amino esters and dipeptides, to the corresponding amines using simple [Rh(acac)(cod)]. Two irons for a smoother finish: An NAD(P)H model was regenerated readily in situ by iron‐catalyzed reduction with molecular hydrogen. The subsequent biomimetic reduction of α‐keto‐/ α‐iminoesters proceeded smoothly in the presence of an iron‐based Lewis acid (LA) to provide α‐hydroxyesters and amino acid esters in good to excellent yields (see scheme; NAD(P)+=nicotinamide adenine dinucleotide (phosphate), TM=transition metal). Polar solvents are often chosen for transition metal catalyzed C-N coupling reactions. In this work, it was found that the use of alkane solvents (e.g. cyclohexane, heptane) allows for highly efficient Ni-catalyzed C-N coupling of aryl pseudohalides with different types of amines in the presence of NHC ligands. Excellent functional group tolerance was achieved. Stronger hydrogen bonding effect between amine NH moiety and polar OTs/OTf in alkane solvents might be responsible for the high activity. Meanwhile, some of the obtained products showed in vitro antiproliferative activities. In this work, it was found that the use of alkane solvents allows for highly efficient Ni-catalyzed C-N coupling of aryl pseudohalides with different types of amines in the presence of NHC ligands. An efficient and halogen-free catalytic methodology for the synthesis of β-amino alcohols from aromatic amines and biomass-derived carbohydrates is demonstrated for the first time. The activation of C5/C6 sugars by a ruthenium catalyst selectively generates the C2 alkylating reagent glycolaldehyde. The transformation involves metal-catalyzed hydrogen borrowing for the reduction of the imine intermediate.
