Studies suggested that exposure to air pollutants, with endocrine disrupting (ED) properties, have a key role in breast cancer (BC) development. Although the population is exposed simultaneously to a mixture of multiple pollutants and ED pollutants may act via common biological mechanisms leading to synergic effects, epidemiological studies generally evaluate the effect of each pollutant separately. We aimed to assess the complex effect of exposure to a mixture of four xenoestrogen air pollutants (benzo-[a]-pyrene (BaP), cadmium, dioxin (2,3,7,8-Tétrachlorodibenzo-p-dioxin TCDD)), and polychlorinated biphenyl 153 (PCB153)) on the risk of BC, using three recent statistical methods, namely weighted quantile sum (WQS), quantile g-computation (QGC) and Bayesian kernel machine regression (BKMR). The study was conducted on 5222 cases and 5222 matched controls nested within the French prospective E3N cohort initiated in 1990. Annual average exposure estimates to the pollutants were assessed using a chemistry transport model, at the participants' residence address between 1990 and 2011. We found a positive association between the WQS index of the joint effect and the risk of overall BC (adjusted odds ratio (OR) = 1.10, 95% confidence intervals (CI): 1.03-1.19). Similar results were found for QGC (OR = 1.11, 95%CI: 1.03-1.19). Despite the association did not reach statistical significance in the BKMR model, we observed an increasing trend between the joint effect of the four pollutants and the risk of BC, when fixing other chemicals at their median concentrations. BaP, cadmium and PCB153 also showed positive trends in the multi-pollutant mixture, while dioxin showed a modest inverse trend. Despite we found a clear evidence of a positive association between the joint exposure to pollutants and BC risk only from WQS and QGC regression, we observed a similar suggestive trend using BKMR. Cadmium (Cd) is a toxic heavy metal widespread in the environment leading to human exposure in particular through diet (when smoking is excluded), as documented by recent human biomonitoring (HBM) surveys. Exposure to Cd at environmental low-exposure levels has been associated with adverse effects such as renal toxicity and more recently bone effects. The implication, even if limited, of Cd in the etiology of osteoporosis can be of high importance at the population level given the significant prevalence of osteoporosis and the ubiquitous and life-long exposure to Cd. Therefore, the osteoporosis cases attributable to Cd exposure was estimated in three European countries (Belgium, France and Spain), based on measured urinary Cd levels from HBM studies conducted in these countries. The targeted population was women over 55 years old, for which risk levels associated with environmental Cd exposure were available. Around 23% of the cases were attributed to Cd exposure. Moreover, in a prospective simulation approach of lifelong urinary Cd concentrations assuming different intakes scenarios, future osteoporosis attributable cases were calculated, based on urinary Cd levels measured in women aged under 55. Between 6 and 34% of the considered populations under 55 years were at risk for osteoporosis. Finally, the costs associated to the burden of osteoporosis-related fractures attributable to Cd for each country targeted in this paper were assessed, standing for a major contributing role of Cd exposure in the overall social costs related to osteoporosis. Absolute costs ranged between 0.12 (low estimate in Belgium) and 2.6 billion Euros (high estimate in France) in women currently over 55 years old and at risk for fractures. In situ measurements provide data that are the highly representative of the natural environment. In this paper, laboratory-determined biomarkers of Cd stress that were previously identified for the green alga Chlamydomonas reinhardtii, were tested in two French rivers: a contaminated site on the Riou Mort River and an “uncontaminated” reference site on the Lot River. Transcript abundance levels were determined by real time qPCR for biomarkers thought to be Cd sensitive. Transcript levels were significantly higher (>5 fold) for organisms exposed to the contaminated site as compared to those exposed at the uncontaminated site. Biomarker mRNA levels were best correlated to free Cd (Cd2+) rather than intracellular Cd, suggesting that they may be useful indicators of in situ stress. The paper shows that biomarker expression levels increased with time, were sensitive to metal levels and metal speciation and were higher in the “contaminated” as opposed to the “reference” site. National-scale soil datasets exhibit variation over widely disparate spatial scales. Geological variation is an important source at the coarsest scale, and one in which exhaustive information is commonly available, in geological maps. Superimposed on this is continuous spatial variation caused by factors such as relief, vegetation and diffuse ‘background’ pollution. Further variation is caused by locally distinct factors such as point pollution from industrial sites or occasional geological anomalies. In this paper, we propose a single statistical model to encompass all of these effects which we describe as ‘geological variation’, ‘continuous spatial variation’ and ‘local anomalies'. In our model, the geological and continuous spatial variation are described, respectively, by the fixed and random effects of a linear mixed model (LMM) and the local anomalies lead to observations which are spatial outliers with respect to the LMM. We fit the model to a survey of 1887 observations of cadmium concentration in soil (Cd) collected on an incomplete regular grid across the French metropolitan territory (550 000 km2) and use it to predict Cd across France. We find that (i) it is not possible to fit a valid model—in terms of cross-validation statistics—of Cd variation unless the effects of local anomalies are identified and separated from the larger-scale processes; (ii) the LMM is not valid if the outliers are merely discarded but a valid model does result if the outliers are winsorized. Simple Summary: Testicular cancer is the most common cancer among men below 40 years old, and its causes remain largely unknown. Marked geographic differences in the occurrence along with the relatively young age at diagnosis suggest a possible role of environmental and occupational exposures that occur early in life, probably already during embryonic development. Using data from a French case–control study, we investigated associations between paternal occupational exposure to five heavy metals and welding fumes before and at birth and testicular cancer risk in sons. We estimated exposures from job titles and considered other potential risk factors in the statistical analyses. We found no association. Thus, further research is necessary to identify potentially modifiable risk factors. Testicular cancer is the most common cancer in young men. Its causes are largely unknown, although prenatal occupational and environmental exposures have been suggested. We investigated paternal occupational exposure to heavy metals and welding fumes and the risk of testicular germ cell tumors (TGCT) in their offspring. A total of 454 cases and 670 controls were included from a French nationwide case–control study. The INTEROCC job exposure matrix was used to assign occupational exposures (cadmium, chromium, iron, nickel, lead, and welding fumes) to the fathers' jobs. Odds ratios (ORs) for TGCT were estimated using conditional logistic regression models for frequency-matched sets. Three complementary analytical approaches were used: (1) single-agent analysis, (2) analysis by groups, and (3) principal component analysis (PCA). The proportion of paternal exposure to different heavy metals and welding fumes ranged from 0.7% (cadmium) to 11.3% (lead). Based on PCA, three principal components explained 93.5% of the cumulative variance. No associations were found between heavy metals or welding fumes and TGCT. European and French populations are overexposed to cadmium (Cd) through their foods. The risk of increased cadmium exposure for consumers needs to be limited by reduced maximum limits (ML) for novel foodstuffs such as edible seaweed in France. The objective was to derive a low and protective cadmium concentration in edible seaweeds to limit cadmium overexposure in consumers. To do so, we applied a probabilistic approach to the data collected on French seaweed consumers, taking into account other sources of exposure for cadmium. This approach led to the identification of a cadmium concentration which should ensure that the seaweed-consuming population does not exceed the tolerable daily intake (TDI) of cadmium according to a probability of cases, when simultaneously exposed to other cadmium dietary sources. Considering the 5% of the population exceeding TDI, the estimated ML is equal to 0.35 mg Cd kg−1 dry matter of seaweed as an unprocessed food with a 95% confidence interval of [0.18,1.09]. The proposed approach is generic and could be applied to other relevant food/substance pairs when considering the setting of MLs in the regulatory system. The ability of tobacco (Nicotiana tabacum L. cv. Badischer Geudertheimer) for phytomanaging and remediating soil ecological functions at a contaminated site was assessed with a potted soil series made by fading an uncontaminated sandy soil with a contaminated sandy soil from the Borifer brownfield site, Bordeaux, SW France, at the 0%, 25%, 50%, 75%, and 100% addition rates. Activities of sandblasting and painting with metal-based paints occurred for decades at this urban brownfield, polluting the soil with metal(loid)s and organic contaminants, e.g., polycyclic aromatic hydrocarbons, in addition to past backfilling. Total topsoil metal(loid)s (e.g., 54,700 mg Zn and 5060 mg Cu kg−1) exceeded by seven- to tenfold the background values for French sandy soils, but the soil pH was 7.9, and overall, the 1M NH4NO3 extractable soil fractions of metals were relatively low. Leaf area, water content of shoots, and total chlorophyll (Chl) progressively decreased with the soil contamination, but the Chl fluorescence remained constant near its optimum value. Foliar Cu and Zn concentrations varied from 17.8 ± 4.2 (0%) to 27 ± 5 mg Cu kg−1 (100%) and from 60 ± 15 (0%) to 454 ± 53 mg Zn kg−1 (100%), respectively. Foliar Cd concentration peaked up to 1.74 ± 0.09 mg Cd kg−1, and its bioconcentration factor had the highest value (0.2) among those of the metal(loid)s. Few nutrient concentrations in the aboveground plant parts decreased with the soil contamination, e.g., foliar P concentration from 5972 ± 1026 (0%) to 2861 ± 334 mg kg−1 (100%). Vulnerability to drought-induced embolism (P50) did not differ for the tobacco stems across the soil series, whereas their hydraulic efficiency (Ks) declined significantly with increasing soil contamination. Overall, this tobacco cultivar grew relatively well even in the Borifer soil (100%), keeping its photosynthetic system healthy under stress, and contaminant exposure did not increase the vulnerability of the vascular system to drought. This tobacco had a relevant potential to annually phytoextract a part of the bioavailable soil Zn and Cd, i.e., shoot removals representing here 8.8% for Zn and 43.3% for Cd of their 1M NH4NO3 extractable amount in the potted Borifer soil. The French waterway system consists of large navigable rivers and canals connecting many regions. Maintaining a safe navigation channel, in the Nord-Pas De Calais (NPDC) region in France, requires the regular removal of accumulated sediments which are often contaminated with zinc, lead, cadmium, and mercury. New legislations, such as the European Water Framework Directive (DCA, 2000), ensure that disposal should be reduced to minimal level and the waterways sediment should be treated. Such measures increase the research interest to find the potential costumers and create the need for establishing an efficient waterway sediment network. This paper discusses the design of a multi-modal supply chain network (SCN) in order to satisfy the demand of the treated sediments and to respect environmental, social and economic requirements. First, we reviewed models for strategic supply chain network design. After that, we proposed a multi-modal, single period, supply chain network design (SCND) model for waterway sediments problem using a mixed integer linear programming (MILP) approach. Then, results obtained solving a real case SCND model of French waterways (Voies Navigables de France: VNF) in NPDC region are presented. There is huge variability among populations of the hyperaccumulator Noccaea caerulescens (formerly Thlaspi caerulescens) in their capacity to tolerate and accumulate cadmium. To gain new insights into the mechanisms underlying this variability, we estimated cadmium fluxes and further characterized the N. caerulescens heavy metal ATPase 4 (NcHMA4) gene in three populations (two calamine, Saint-Félix-de-Pallières, France and Prayon, Belgium; one serpentine, Puente Basadre, Spain) presenting contrasting levels of tolerance and accumulation. Cadmium uptake and translocation varied among populations in the same way as accumulation; the population with the highest cadmium concentration in shoots (Saint Félix-de-Pallières) presented the highest capacity for uptake and translocation. We demonstrated that the four NcHMA4 copies identified in a previous study are not fixed at the species level, and that the copy truncated in the C-terminal part encodes a functional protein. NcHMA4 expression and gene copy number was lower in the serpentine population, which was the least efficient in cadmium translocation compared to the calamine populations. NcHMA4 expression was associated with the vascular tissue in all organs, with a maximum at the crown. Overall, our results indicate that differences in cadmium translocation ability of the studied populations appear to be controlled, at least partially, by NcHMA4, while the overexpression of NcHMA4 in the two calamine populations may result from convergent evolution. Margaritifera margaritifera is a critically endangered species in Europe. Among the causes explaining its decline, metal pollution had never been deeply studied. Thus, an ecotoxicological investigation was developed on this species which comes from the Dronne River (South-West of France). Cadmium (Cd) exposure of mussels at 2 and 5 μg/L for 7 days was conducted to test their vulnerability to this metal, and also the potential endocrine disruption power of Cd. Morphometric analyses, gonad histological observations, metal bioaccumulation, metallothionein (MTs) production, measures of malondialdehyde (MDA), and finally quantitative relative expression analysis of genes involved in various metabolic functions were performed. The main results showed Cd accumulation increasing in a dose-dependent manner, especially in the gills. The same trend was observed for gene expression relative to oxidative stress. Histological analysis of the gonads highlighted a predominance of hermaphrodite individuals, but after 7 days of exposure to Cd, the percentage of female was largely increased compared with controls, from 17 to 33%. These results demonstrate the endocrine disruption effect of Cd on freshwater pearl mussels. The pearl mussel Margaritifera margaritifera is sensitive to cadmium since the metallothioneins are poorly induced, gene expression reveals oxidative stress, and gonads tend to be feminized. We investigated the influence of the acanthocephalan parasite Polymorphus minutus on the mortality of its intermediate host, Gammarus roeseli, exposed to cadmium, by the measure of LC values as well as the bioaccumulation of cadmium both in the host and in its parasite. LC results revealed that infected G. roeseli males died less under cadmium stress than uninfected ones; while the converse has been observed in females. Cadmium resistance of infected males could be explained by a weaker bioconcentration factor (BCF) than in females. The lower BCF in infected individuals was closely related with an uptake of cadmium by P. minutus in its host. Nevertheless, although infected females had both weaker BCF and cadmium concentration in their body, the presence of P. minutus did not induce lower mortality than uninfected females. On the contrary, their sensitivity to cadmium was increased by the presence of P. minutus. We discuss the hypothesis that differences of mortality between uninfected and infected gammarids could be explained by a difference of cadmium bioconcentration in host, and by the cadmium bioaccumulation in the parasite. Indeed, results suggested that P. minutus could help G. roeseli to face with stress, what contributed to keep the host alive and favour the parasite transmission. [ABSTRACT FROM AUTHOR]

Cadmium (Cd) is a highly toxic element and its presence in the environment needs to be closely monitored. Recent systematic surveys in French soils have revealed the existence of areas in eastern and central France, which show systematically high cadmium concentrations. It has been suggested that at least part of these anomalous levels are of natural origin. For the Lower Burgundy area in particular, a direct heritage from the Jurassic limestone bedrock is highly suspected. This potential relationship has been studied in several localities around Avallon and this study reports new evidence for a direct link between anomalously elevated cadmium contents of Bajocian and Oxfordian limestone and high cadmium concentrations in deriving soils. Soils in this area show cadmium concentrations generally above the average national population values, with contents frequently higher than the 'upper whisker' value of 0.8 μg g determined by statistical evaluation. In parallel, limestone rocks studied in the same area exhibit cadmium concentrations frequently exceeding the mean value of 0.030-0.065 μg g previously given for similar rocks by one order of magnitude, with a maximum of 2.6 μg g. Mean ratios between the cadmium concentrations of limestone bedrock and deriving soils (Cd/Cd), calculated for different areas, range from 4.6 to 5.7. Calculations based on the analyses of both soils from a restricted area and fragments of bedrock sampled in the immediate vicinity of high-concentration soils are around 5.5-5.7. Cd/Cd is useful in determining the potential of soils in Lower Burgundy to reflect and exacerbate the high concentrations of cadmium present in parent bedrocks. [ABSTRACT FROM AUTHOR]

Recycling sewage sludges as fertilisers on soils for crop production has several potential benefits such as providing large amounts of phosphorous and organic matter. However, the spreading of urban sewage sludge is a constant cause of controversy because it is known to introduce potentially toxic trace metals into the soil, particularly cadmium. In order to clarify this debate, this review article presents a synthesis of the results of several studies carried out in France on the impact of sewage-sludge spreading. This article reports mainly Cd results but also some results on other trace metals such as Cr, Cu, Hg, Ni, Pb and Zn. Two kinds of data are presented: (1) soil data including total metal contents and data from partial extraction to evaluate the phytoavailable fractions, and (2) plant data including metal content of wheat, a major agricultural crop. The field experiments involved very different amounts of applied sewage sludges and Cd. Indeed, three categories of experiments stand out, the first and second involving high amounts of applied Cd, and the third involving low amounts of applied Cd: (1) during the 1970s and '80s, sludges with a high trace metal content, especially Cd, were spread at the INRA trials at Couhins experimental farm on sandy soils and in the Vexin area on silty topsoils. The quantities of applied Cd were very high, ranging from 3600 g to 641 000 g per ha. Here, the results show a notable impact on total Cd contents of topsoil and cereal grains. (2) Sludges containing high levels of industrial cadmium were spread on acid soils in the Limousin region for more than twenty years up to 1998. Topsoil Cd contents were measured in fields where the cadmium input was highest, of 300 to 600 g Cd per ha. Here, a clear increase in the Cd content of cereal grains was found. (3) During the 1990s and 2000s, numerous experiments with sewage-sludge applications compatible with the new French regulations of 1998 were implemented. The amounts of applied Cd were therefore much lower, from 0.6 to 270 g/ha. Here, no impact was detected on the composition of cereal grains. This review article concludes that the application of huge quantities of sewage sludges in the 1970s and '80s had a clear and long-lasting effect on both soil and grain Cd compositions. A microcosm study was undertaken to examine the effects of dissolved cadmium at various concentrations (0, 10, and 100 μg · L−1) on biofilm accumulation and diatom assemblages. A natural biofilm sampled from the Riou-Mort River (Southwest France) was inoculated into three experimental systems, where biofilm settled on glass slides. Samples collected after 1, 2, 4, and 6 weeks of colonization were analyzed for metal accumulation (total metal content and intracellular metal content in the biofilm), biomass (as measured through dry weight and ash-free dry matter), and quantitative as well as qualitative analysis of diatom assemblages. There was a positive correlation between cadmium accumulation and dissolved cadmium concentrations and duration of exposure: a linear relationship was found between concentration factors (CFs) of growing biofilms and time (CFs/day = 0.25 and 0.38 under contaminations of 10 and 100 μgCd · L−1, respectively). Biofilm settlement, more than photosynthetic activity, was affected by high cadmium concentrations: we observed for all stages of settlement a drastic and significant ( p < 0.05) reduction in biofilm biomass and in diatom densities in the highest cadmium contamination, compared to control and low cadmium concentration units. The Seine estuary (France) is currently one of the world’s most contaminated estuaries, due in particular to its high cadmium and lead content. Proper understanding of contaminant transfer, transformation and retention mechanisms throughout the estuary and up to the adjacent marine zone require a range of studies involving data collections, experiments, and modeling tool. A multivariable transport model (SiAM-3D) was used to simulate dissolved and particulate transport and it is applied to several calculation grids; a speciation model (MOCO) was used to select key species and obtain a schematic system representation. The coupled model for Cd and Pb was compared with field measurements. The complementary character of various tools (model applied to annual time scale, in situ measurements and experiments) allowed to explore and quantify various hypotheses on the high dissolved cadmium concentrations observed during low river flow. The target is to achieve a compromise between acceptable computing times and adequate result accuracy. Although particle and contaminant behaviour is globally well-reproduced by the coarse grid, calculation errors relating to bayward fluxes and stocks deposited inside the estuary were highlighted after comparison with the fine grid. The presence of large amounts of trace metals creates a hazard in some soils that is strongly related to the speciation of these metals. The aim of this work was to apply an operationally defined fractionation based on kinetics, which could distinguish “labile” (quickly extracted) and “non-labile” (or slowly labile i.e. slowly extracted) cations by using the extraction kinetics of trace metals by EDTA. This kinetic fractionation was applied to 10 unpolluted soil samples from Burgundy (France) in order to determine their labile concentrations of lead and cadmium. According to this method, cadmium was found to be more mobile than lead, with respect to both labile concentration and the kinetic constants. Moreover, it was also found that the labile concentration of cadmium was related ( R 2=0.6) to its concentration in wheat shoots; however, no such correlation was found for lead. The article presents a study which determines exoenzymatic and respiratory activities of a microbial community and to determine and compare the impact of lead and cadmium from bacterial cultures. It states that fluorimetry was sued to determine glucosidase activity and that the study site involves two rivers in Grenoble, France. It concludes that dehydrogenase activity in aerobic conditions prove to be better adapted than exoenzymatic activities.
