Abstract

The unprecedented coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is a serious threat to global public health. Development of effective therapies against SARS-CoV-2 is urgently needed. Here, we evaluated the antiviral activity of a remdesivir parent nucleotide analog, GS441524, which targets the coronavirus RNA-dependent RNA polymerase enzyme, and a feline coronavirus prodrug, GC376, which targets its main protease, using a mouse-adapted SARS-CoV-2 infected mouse model. Our results showed that GS441524 effectively blocked the proliferation of SARS-CoV-2 in the mouse upper and lower respiratory tracts via combined intranasal (i.n.) and intramuscular (i.m.) treatment. However, the ability of high-dose GC376 (i.m. or i.n. and i.m.) was weaker than GS441524. Notably, low-dose combined application of GS441524 with GC376 could effectively protect mice against SARS-CoV-2 infection via i.n. or i.n. and i.m. treatment. Moreover, we found that the pharmacokinetic properties of GS441524 is better than GC376, and combined application of GC376 and GS441524 had a synergistic effect. Our findings support the further evaluation of the combined application of GC376 and GS441524 in future clinical studies.ImportanceSevere acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causes coronavirus disease 2019 (COVID-19), which has seriously threatened global public health and economic development. Currently, effective therapies to treat COVID-19 are urgently needed. In this study, we assessed the efficacy of the preclinical inhibitors GC376 and GS441524 using a mouse-adapted SARS-CoV-2 infected mouse model for the first time. Our results showed that low-dose combined application of GC376 and GS441524 could effectively protect mice from HRB26M infection in the upper and lower respiratory tracts. Hence, the combined application should be developed and considered for future clinic practice.

Abstract

This paper presents an analytical study on evaluation of support loss in micromechanical resonators undergoing in-plane flexural vibrations. Two-dimensional elastic wave theory is used to determine the energy transmission from the vibrating resonator to the support. Fourier transform and Green's function technique are adopted to solve the problem of wave motions on the surface of the support excited by the forces transmitted by the resonator onto the support. Analytical expressions of support loss in terms of quality factor, taking into account distributed normal stress and shear stress in the attachment region, and coupling between the normal stress and shear stress as well as material disparity between the support and the resonator, have been derived. Effects of geometry of micro-beam resonators, and material dissimilarity between support and resonator on support loss are examined. Numerical results show that ‘harder resonator’ and ‘softer support’ combination leads to larger support loss. In addition, the Perfectly Matched Layer (PML) numerical simulation technique is employed for validation of the proposed analytical model. Comparing with results of quality factor obtained by PML technique, we find that the present model agrees well with the results of PML technique and the pure-shear model overestimates support loss noticeably, especially for resonators with small aspect ratio and large material dissimilarity between the support and resonator.

Abstract

In this paper, the crushing responses of tubes with different shapes of windows as well as the simple tube are investigated under axial and oblique loading conditions. Numerical model was constructed with FE code LS-DYNA and was validated by experiment. The model was then employed for the dynamic crush simulation of each tube at different load angles. The initial peak, crush force efficiency, energy absorption and specific energy absorption of windowed and simple tubes were compared and their overall crushing performances were evaluated by TOPSIS method. The results have proven the effectiveness of introducing windows to improve the tube's crushing performance and have showed that square and rectangle are the best window shapes.

Abstract

Extracting buildings from high resolution remotely sensed images is very practical, which can be applied to urban modeling and so on. The development of computer vision has become better, and the accuracy of recognition of convolutional neural networks has exceeded the accuracy of recognition of human eyes. In this paper, we used a deep convolutional neural network in remote sensing to achieve building extraction. The method in this paper is not based on semantic segmentation, but instance segmentation, which considered each building as an independent individual to achieve building extraction. The results showed that the proposed method is able to extract buildings with high accuracy.

Abstract

Building extraction is a hotspot of research. Buildings in remote sensing images are numerous, and buildings have different distribution patterns in different regions, which means it is difficult to extract buildings by one traditional method for all areas. Deep learning plays more and more important part of computer version and image processing, which has made some progress in target detection and identification, especially in medical graphic recognition. In this paper, we compared three common datasets, and selected the most suitable one. Mask R- CNN is applied to this dataset. We provided a better method and this method can be used in different areas when dataset is rich enough for training. The method in this paper just need high resolution aerial images to extract buildings automatically without any other steps, because identification and extraction are achieved in one network. The research showed: the dataset we selected is suitable; Mask R-CNN can be used in high resolution remotely images; Mask R-CNN is good at building extraction, and greatly reduce the loss of classification and leakage. Therefore, this paper provided a suitable method for automatic building extraction.

Abstract

Background: Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy.Methods: RESTART was a prospective, randomised, open-label, blinded-endpoint, parallel-group trial at 122 hospitals in the UK that assessed whether starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. For this prespecified subgroup analysis, consultant neuroradiologists masked to treatment allocation reviewed brain CT or MRI scans performed before randomisation to confirm participant eligibility and rate features of the intracerebral haemorrhage and surrounding brain. We followed participants for primary (recurrent symptomatic intracerebral haemorrhage) and secondary (ischaemic stroke) outcomes for up to 5 years (reported elsewhere). For this report, we analysed eligible participants with intracerebral haemorrhage according to their treatment allocation in primary subgroup analyses of cerebral microbleeds on MRI and in exploratory subgroup analyses of other features on CT or MRI. The trial is registered with the ISRCTN registry, number ISRCTN71907627.Findings: Between May 22, 2013, and May 31, 2018, 537 participants were enrolled, of whom 525 (98%) had intracerebral haemorrhage: 507 (97%) were diagnosed on CT (252 assigned to start antiplatelet therapy and 255 assigned to avoid antiplatelet therapy, of whom one withdrew and was not analysed) and 254 (48%) underwent the required brain MRI protocol (122 in the start antiplatelet therapy group and 132 in the avoid antiplatelet therapy group). There were no clinically or statistically significant hazards of antiplatelet therapy on recurrent intracerebral haemorrhage in primary subgroup analyses of cerebral microbleed presence (2 or more) versus absence (0 or 1) (adjusted hazard ratio [HR] 0·30 [95% CI 0·08–1·13] vs 0·77 [0·13–4·61]; pinteraction=0·41), cerebral microbleed number 0–1 versus 2–4 versus 5 or more (HR 0·77 [0·13–4·62] vs 0·32 [0·03–3·66] vs 0·33 [0·07–1·60]; pinteraction=0·75), or cerebral microbleed strictly lobar versus other location (HR 0·52 [0·004–6·79] vs 0·37 [0·09–1·28]; pinteraction=0·85). There was no evidence of heterogeneity in the effects of antiplatelet therapy in any exploratory subgroup analyses (all pinteraction>0·05).Interpretation: Our findings exclude all but a very modest harmful effect of antiplatelet therapy on recurrent intracerebral haemorrhage in the presence of cerebral microbleeds. Further randomised trials are needed to replicate these findings and investigate them with greater precision.Funding: British Heart Foundation.

Abstract

Background: Results of small trials indicate that fluoxetine might improve functional outcomes after stroke. The FOCUS trial aimed to provide a precise estimate of these effects.Methods: FOCUS was a pragmatic, multicentre, parallel group, double-blind, randomised, placebo-controlled trial done at 103 hospitals in the UK. Patients were eligible if they were aged 18 years or older, had a clinical stroke diagnosis, were enrolled and randomly assigned between 2 days and 15 days after onset, and had focal neurological deficits. Patients were randomly allocated fluoxetine 20 mg or matching placebo orally once daily for 6 months via a web-based system by use of a minimisation algorithm. The primary outcome was functional status, measured with the modified Rankin Scale (mRS), at 6 months. Patients, carers, health-care staff, and the trial team were masked to treatment allocation. Functional status was assessed at 6 months and 12 months after randomisation. Patients were analysed according to their treatment allocation. This trial is registered with the ISRCTN registry, number ISRCTN83290762.Findings: Between Sept 10, 2012, and March 31, 2017, 3127 patients were recruited. 1564 patients were allocated fluoxetine and 1563 allocated placebo. mRS data at 6 months were available for 1553 (99·3%) patients in each treatment group. The distribution across mRS categories at 6 months was similar in the fluoxetine and placebo groups (common odds ratio adjusted for minimisation variables 0·951 [95% CI 0·839–1·079]; p=0·439). Patients allocated fluoxetine were less likely than those allocated placebo to develop new depression by 6 months (210 [13·43%] patients vs 269 [17·21%]; difference 3·78% [95% CI 1·26–6·30]; p=0·0033), but they had more bone fractures (45 [2·88%] vs 23 [1·47%]; difference 1·41% [95% CI 0·38–2·43]; p=0·0070). There were no significant differences in any other event at 6 or 12 months.Interpretation: Fluoxetine 20 mg given daily for 6 months after acute stroke does not seem to improve functional outcomes. Although the treatment reduced the occurrence of depression, it increased the frequency of bone fractures. These results do not support the routine use of fluoxetine either for the prevention of post-stroke depression or to promote recovery of function.Funding: UK Stroke Association and NIHR Health Technology Assessment Programme.

Abstract

Architected materials consisting of periodic unit cells are desirable for many engineering applications. Characterizing the elastic isotropy is of great significance for the mechanical design of architected materials. However, prevailing experimental and numerical approaches are normally too costly and time-consuming to screen out isotropic architected materials in the large design space. Here, a deep learning-based approach is developed as a highly efficient and portable tool to identify the elastic isotropy of architected materials directly from images of their unit cells with arbitrary component distributions. The measure of elastic isotropy for heterogeneous architected materials is derived firstly in this paper to construct a database with associated images of unit cells. Then a convolutional neural network is fully trained with the database, performing well on the isotropy identification with about 90% accuracy and milliseconds processing time per sample. Meanwhile, it exhibits enough robustness to maintain its performance under the fluctuating material properties in test sets. Moreover, the transfer learning of the convolutional neural network is successfully implemented among architected materials with different numbers of material components, which further promotes the efficiency of the deep learning-based approach without scarifying its identification performance. This study gives new inspirations on the rapid mechanical characterization of architectured materials, which holds promising applications in the big-data driven topological design and nondestructive testing of architected materials.

Abstract

Selective laser melting (SLM) has received increasing attention in recent years as an innovative manufacturing technique. The present SLM process only uses a single laser beam to melt and consolidate the powder, which may result in excessive evaporation. In this paper, a double beam scanning strategy is investigated in which the first laser beam preheats the powder just below the sintering point while the second laser beam completely melts the powder. An analytical solution on the temperature distribution heated by a moving laser beam in the powder-bulk domain is derived and is used to determine the critical radius of the first laser beam. The single and double beam scanning strategies are compared numerically and it is found that double beam scanning can effectively reduce material evaporation and increase the amount of powder melted in the SLM process.

Abstract

The Bleustein–Gulyaev (B–G) wave in piezoelectric materials of orthorhombic mm2 crystal class is reported to be a promising candidate for application to liquid sensing. In this work we present a rigorous quantitative investigation of the propagation of B–G wave in mm2 crystals in contact with a viscous liquid. An inversion algorithm is formulated to determine the liquid viscosity from the wave speed and attenuation data. Numerical results and discussions are given for potassium niobate (KNbO3). The inversion results demonstrate that the liquid viscosity can be successfully determined from wave propagation characteristics.

Abstract

The progressive collapse of tubular structures under axial loading is a challenging problem in mechanics. Due to the nonlinearities in large plastic deformation, such a problem can only be solved case by case under the assumption of an appropriate collapse mechanism. In this paper, a relationship between the progressive collapse of an axially loaded tube and the initial buckling of its windowed counterpart is presented. Numerical investigation was performed on the axial crushing of triangular, square and pentagonal tubes and the initial buckling modes of the corresponding windowed tubes. Results show that at the critical symmetric buckling mode, the theoretical mean crushing force of the angle-shaped column in the windowed tube matches very well with the actual mean crushing force of the conventional one. This relationship is crucial to the development of a generalized method for progressive collapse without assuming collapse mechanism. Based on it, an empirical equation on the mean crushing force of axially loaded square tubes is presented. The mean crushing forces predicted by this equation are in good agreement with the experimental results and theoretical values.

Abstract

The collapse of thin-walled tubes under axial and oblique loading is frequently encountered in real crash events. The windowing and multi-cell methods are effective in improving tubes' energy absorbing performance. In this paper, a comparative study on the performance of windowed and multi-cell square tubes of the same weight under axial and oblique loading is conducted numerically. The results show that the multi-cell tube can achieve higher mean crushing force than the windowed tube but the windowed tube has lower initial peak force. The effectiveness of both methods reduces as the load angle increases. Moreover, the multi-cell and windowed tubes may have worse performance than the conventional tube if the former two collapse in global bending and the later in axial mode.

Machine vision-based PCB defect inspection system is designed to meet high speed and high precision requirement in PCB manufacture industry field, which is the combination of software and hardware. This paper firstly introduced the whole system structure and the principle of vision detection, while described the relevant key technologies used during the PCB defect inspection, finally implemented one set of test system with the key technologies mentioned. The experimental results show that the defect of PCB can be effectively inspected, located and recognized with the key technologies.

Abstract

According to the PCB image feature in a PCB defect detection project, we studied a new image enhancement algorithm: double sigmoid transformation and also deduced the transformation arithmetic operator. Practical project experiments show that the new algorithm can effectively sharpen PCB image edges, at same time it can filter out noise in the image. Even if the original PCB image is not distinct enough and uniform brightness, the algorithm is able to precisely locate circuit edge, which can make the edge extracting and edge recognition easier.

Abstract

Because there are some defects with the detection of PCB (Printed Circuit Board) location hole with traditional methods, the paper presents a fast and accurate positioning algorithm of location hole based on potential function, which is not interfered by the bright spots within the pad image and also be adaptable to the fuzzy pad image and the deformed pad due to poor quality of circuit board corrosion. The algorithm was implemented and experiments show that the new method provides with stable performance, high positioning accuracy and good efficiency, which can be better meet the engineering real-time requirements.

Firstly, we deduced a new RGB threshold formula by limiting the hue change range. Secondly, we provided a fast method to detect license plate position according to the characteristics of colors of license plate. Finally, we performed a detection experiment with 416 images containing license plate and the result showed that almost all license plates could be discovered quickly with our method from the images with even quit complicated background.

Abstract

With the development and updating of the artificial intelligence techniques, some theories concerning artificial intelligence, especially in agent techniques, are applied to remote teaching system to open the new development space. The paper designs a remote Internet teaching system model on the basis of multi-agent techniques. And then describes each module functions and methods in the model and presents some technique basic and implementing steps for the system. Finally make a basic experiment, which show that because the new remote teaching system bases on the Agent, the model is more humane and the teaching effect are superior to the traditional remote teaching system.

Abstract

The essential functions in the management system of experimental consumables for colleges are introduced. In the system three roles such as experimental college governor, experimental department manager and laboratory assistant are involved. After traditional workflow networks are recalled, Workflow network of temporal constraints oriented role is defined in order to build the interface among roles. It can effectively describe transitions according to different roles in complicated workflow processes constrained by time. Schedule properties based on the management system of experimental consumables are analyzed. Transition series both in a role and among roles are generated after a cooperative task is completed. Finally system implements are partly illustrated.

Abstract

In this paper, the concept of pseudo-3D visualization in mobile GIS is firstly defined, then a pseudo-3D visualization model is proposed and a simplified pseudo-3D projection transformation algorithm of building pseudo-3D map based on existed 2D vector data is designed. Furthermore the spatial data organizing and processing principles regard to limited hardware and software resources of mobile devices are introduced. To show the stereo vision effect of geographical scenes smoothly, preprocessing measures of pseudo-3D visualization and necessary 3D scene enhancement technology are applied. Finally, an agisNavigator project is implemented with the pseudo-3D visualization technology based on 2D vector spatial data in PocketPC and some experiment results are presented and analyzed.

Abstract

This paper defines Mobile GIS of off-line system architecture and describes its main features. Because intelligent mobile terminals do not have powerful and professional spatial database system supported, one kind of practical spatial database structure of off-line mobile GIS is proposed. Algorithms of some kernel technologies are improved according to the features of intelligent mobile terminals. In order to verify the reasonableness of spatial data organization and the validity of the improved algorithms, mNav project is implemented and we show how the applied technologies influence the whole system performance.
