There is a population of p63+/Krt5+ distal airway stem cells (DASCs) quiescently located in the airway basal epithelium of mammals, responding to injury and airway epithelial regeneration. They hold the ability to differentiate into multiple pulmonary cell types and can repopulate the epithelium after damage. The current study aims at gaining further insights into the behavior and characteristics of the DASCs isolated from the patient lung and exploring their clinical translational potential. Human DASCs were brushed off through the bronchoscopic procedure and expanded under the pharmaceutical-grade condition. Their phenotype stability in long-term cell culture was analyzed, followed by safety evaluation and tumorigenic analysis using multiple animal models including rodents and nonhuman primate. The chimerism of the human-mouse lung model indicated that DASC pedigrees could give rise to multiple epithelial types, including type I alveolar cells as well as bronchiolar secretory cells, to regenerate the distal lung. Taken together, the results suggested that DASC transplantation could be a promising therapeutic approach for unmet needs in respiratory medicine including the COVID-19-related diseases.

Introduction: Lipid disturbances are common in ESRD patients. In peritoneal dialysis (PD) patients, dyslipidemia is even more common. This study aimed to examine whether serum lipids were associated with prognosis of PD patients. Methods: Patients from a multicenter retrospective cohort were used for the present study. The primary endpoint was all-cause mortality. Cox regression was used to analyze the association between serum lipids including total cholesterol, low-density lipoprotein (LDL), high-density lipoprotein, and triglycerides and the prognosis. Results: The results showed that lower total cholesterol and LDL levels at the initiation of PD predicted higher all-cause mortality in PD patients. Multivariate analysis reveal that the association disappeared after adjusting for age, gender, albumin, prealbumin, protein catabolic rate normalized to body weight, C-reactive protein, and residual renal function. Further analysis showed that patients with lower total cholesterol/LDL had a higher mortality only during the first 24 months of follow-up. In the patients who survived >2 years after PD, lower total cholesterol/LDL was not associated with higher long-term all-cause mortality any more. Conclusion: Lower total cholesterol/LDL levels at the initiation of PD were associated with overall mortality in PD patients. The association could be potentially modified by malnutrition, inflammation, and residual renal function or disappeared after 24 months.

Background Diabetes mellitus has become an increasing global health burden with rapid growing prevalence. Patients with diabetes have higher susceptibility to acute kidney injury (AKI). Liver transplantation (LT) predisposes the kidney to injury. However, the association between diabetes and AKI in LT patients remains unclear. Methods We conducted a retrospective cohort study examining risk factors for AKI in patients undergone orthotopic LT. Potential risk factors including baseline estimated glomerular filtration rate (eGFR), the model for end-stage liver disease (MELD) score, diabetes, hypertension and intraoperative blood loss were screened. The primary endpoint was AKI occurrence. Multivariate logistic regression was used to analyze the association between potential risk factors and AKI. Results A total of 291 patients undergone orthotopic LT were included in the present study. Among them, 102 patients (35.05%) developed AKI within 5 days after LT. Diabetes was identified as an independent risk factor for AKI. Patients who developed AKI had worse graft function recovery and higher mortality within 14 days after LT compared to those who did not develop AKI. AKI patients with diabetes had a significant decline of eGFR within the first postoperative year, compared with patients who did not develop AKI and who developed AKI but without diabetes. Conclusions Diabetes is an independent risk factor for AKI after orthotopic LT. AKI is associated with delayed graft function recovery and higher mortality in short-term postoperative period. Diabetic patients who developed AKI after LT experience a faster decline of eGFR within the first year after surgery.

A whispering gallery mode (WGM) microre- sonator based on grapefruit microstructured optical fibers (MOFs) is proposed for in-situ DNA biosensing. The MOF microresonator has a porous structure that offers natural microfluidic channels to load microfluidic samples of volume down to $100~\mu \text{L}$ . Moreover, the WGM mechanism greatly enhances the interaction between WGM light and liquid sample kept inside the microcavity. The inner surface of the microcavity is functionalized with poly-L-lysine monolayer to achieve label-free binding of the single-strand probe DNA (ssDNA) through electrostatic force, and the probe immobilization as well as DNA hybridization processes are monitored in real time. The linear fitting of resonance wavelength shift versus DNA concentration in logarithmic scale indicates that DNA detection is available from 0.01 fmol/ $\mu \text{L}$ to 100 fmol/ $\mu \text{L}$ . The proposed WGM microresonator biosensor possesses such desirable merits as ease of operation, fast response, label free method, and high specificity, making it a promising candidate for in-situ DNA detection and related biosensing applications.

A 473-nm-laser-tuned whispering gallery mode (WGM) silica microbubble resonator integrated with iron oxide particles is demonstrated in this paper. Owing to the photo-induced thermal effect, the WGM resonance wavelength could be tuned by adjusting laser power density of the illuminating light absorbed by the iron oxide particles. A wavelength tuning sensitivity of 0.03 nm/(mW·mm−2) and a tuning range of 0.18 nm are experimentally achieved. Moreover, the influence of ambient temperature on the WGM spectral characteristics is experimentally studied, and a silica-microbubble-based reference scheme is demonstrated to compensate for the temperature-caused resonance wavelength variation. The proposed laser-tuned microresonator has great potential in optical modulation and high-precision optical filtering applications.

Depth of invasion (DOI) and pattern of invasion (POI) indicate tumor invasiveness of oral squamous cell carcinoma (OSCC). However, preoperative DOI evaluation is challenging, and the correlations between DOI and POI are unknown. We aimed to assess DOI and worst pattern of invasion (WPOI) in early-stage OSCC, and evaluate the preoperative predictive ability of biopsy pattern of invasion (BPOI) for WPOI and DOI. This retrospective study included n = 444 OSCC patients with pT1-2N0M0. The prognostic value of DOI, WPOI, and BPOI and the predictive prognostic option for WPOI and DOI by BPOI were assessed. WPOI (1-3 vs 4-5), but not BPOI, predicted the lowest survival rate and highest DOI. To evaluate the difference between WPOI and BPOI, we conducted a POI type-matching analysis of patients with BPOI1-4 and WPOI1-5. Based on each WPOI type, the false-prediction rates (FPR) of BPOI types 1 (n = 23), 2 (n = 89), 3 (n = 252), and 4 (n = 80) were 52.17%, 52.81%, 36.90%, and 0%, respectively. BPOI4 perfectly predict WPOI 4-5. As the false-predicted BPOI 1-2 was almost WPOI2-3 (79.7%), regardless of the existed FPR, patients with BPOI1-2 have longer survival and lower DOI than those with BPOI 4. However, this phenomenon was not observed in BPOI3, because all false-predicted BPOI3 were WPOI4-5 with a high DOI. We provide an alternative predictive prognostic option for WPOI and DOI by evaluating BPOI during OSCC surgical planning, with the recommendation of conservative treatment in patients with BPOI 1-2.

In this article, we propose two laser-controlled Fano resonance refractive index (RI) sensors based on eccentric hole fibers (EHFs) integrated with photo-sensitive azobenzene (azo) by exploiting whispering gallery mode (WGM) coupling between the eccentric hole and the EHF microresonators. Azo coating is coated over the external surface of the EHFs to achieve laser tuning of the RI sensing range. Fano resonance would emerge when the phase matching condition of the WGMs that respectively propagate through the azo coating and the eccentric hole is satisfied. Fano resonance sensing with tunable RI range by adjusting the laser power density is theoretically demonstrated. The EHF-based Fano resonance sensor having the silica-based eccentric hole infiltrated with liquid sample of RI between 1.61364 and 1.61436 achieves a RI sensitivity of 875.185 nm/RIU, while the RI sensitivity of the one with a higher index layer serving as the internal surface of the eccentric hole turns to be 1106.351 nm/RIU for a RI range of 1.33748 to 1.33805. More significantly, simulation results indicate that as 5 mW/mm2 laser is applied, the RI sensing ranges of the proposed Fano resonance sensors respectively show 101.39% and 101.75% increases compared with the cases when no laser illumination is applied. The EHF-based Fano resonance sensors provide promising candidates for applications in bio/chemical sensing with laser-controlled extended RI sensing range. And moreover, the proposed laser tuning method paves a new way toward developing tunable photonic devices and all-optical networking systems with desired operation wavelength ranges.

In this paper, a capillary-based microfluidic whispering gallery mode (WGM) microresonator is proposed to dynamically monitor the conformation change of G-quadruplex driven by K+ ion. We have theoretically analyzed the resonance wavelength shift mechanism for the DNA folding process, and the resonance mode with the largest wavelength sensitivity of 995.63 nm/RIU according to our simulation result is selected for monitoring of the DNA folding as well as the PGQS immobilization processes. The experimentally observed resonance wavelength shift caused by the immobilization of streptavidin is in accordance with our theoretical analysis on the WGM properties of the proposed microresonator. Moreover, experimental results indicate that PGQS could be folded with a KCl concentration down to $1~\mu \text{M}$ and a resonance wavelength shift rate of $18.85\times 10^{-5}$ nm/s, which decreases to $2.03\times 10^{-5}$ nm/s when the KCl concentration increases to 10 mM due to the reduction of available PGQS folding sites. Our proposed WGM-microresonator-based measurement platform possesses many desirable merits such as high sensitivity, high integration, microfluidic, label-free and real-time monitoring operation, which make it a promising candidate for DNA sensing, measurement of trivial amount biochemical analyte and related applications.

A novel coronavirus SARS-CoV-2 was identified in Wuhan, Hubei Province, China in December of 2019. According to WHO report, this new coronavirus has resulted in 76,392 confirmed infections and 2,348 deaths in China by 22 February, 2020, with additional patients being identified in a rapidly growing number internationally. SARS-CoV-2 was reported to share the same receptor, Angiotensin-converting enzyme 2 (ACE2), with SARS-CoV. Here based on the public database and the state-of-the-art single-cell RNA-Seq technique, we analyzed the ACE2 RNA expression profile in the normal human lungs. The result indicates that the ACE2 virus receptor expression is concentrated in a small population of type II alveolar cells (AT2). Surprisingly, we found that this population of ACE2-expressing AT2 also highly expressed many other genes that positively regulating viral entry, reproduction and transmission. This study provides a biological background for the epidemic investigation of the COVID-19, and could be informative for future anti-ACE2 therapeutic strategy development.

In this paper, a fault-tolerant control (FTC) scheme is presented to counteract actuator faults, model uncertainties and external disturbances for a class of three degrees of freedom (3-DOF) helicopter. A fixed-time observer is introduced to estimate composite disturbances which results in a better tracking performance compared with other sliding mode observers (SMO). Finally, a novel control strategy is proposed via back-stepping design technique to guarantee the stability of closed-loop nonlinear system. Experimental results are given to show the effectiveness and advantages of the proposed method.

3D printing, as a driving force of innovation over many areas, brings numerous manufacturing methods together from the macro to nano scales. New revolutionary materials (such as polymeric materials and natural biomaterials) can be produced into unique 3D printed nanostructures. The morphology and functionality of various 3D printing methods as well in vitro and in vivo results of their use towards regenerating bone are discussed in this review. This review further focuses nano scale 3D bioprinting technology for bone tissue engineering, mainly including recent progress in research on technical materials and methods, typical applications, and crucial achievements; explaining the scientific and technical challenges for bone tissue fabrication; and describing micro-nano scale 3D printing application prospects, development directions, and trends for the future for this field to realize its full potential.

Purpose The aim of the study was to investigate the incidence of acute kidney injury (AKI) occurring after high-dose methotrexate (HDMTX) administration and the role of type 2 diabetes (T2D) playing in the occurrence of AKI. Methods We assessed associations between T2D along with other confounding factors mainly including baseline estimated glomerular filtration rate (eGFR), methotrexate (MTX) elimination and urine pH, and AKI occurrence. Patients who were diagnosed as primary central nervous system lymphoma with treatment of HDMTX and with eGFR ≥60 mL/min/1.73 m2 were enrolled in this study. Results Of the 507 courses enrolled in this study, 132 courses have T2D. Lower baseline eGFR, delayed MTX elimination, lower urine pH, and higher incidence of AKI were observed in T2D group. Using univariate logistic regression, several confounding factors including baseline eGFR, hypertension, MTX elimination, and urine alkalinization statistically and clinically important were screened out. After adjusting for these factors, T2D remained an independent association with AKI occurrence. AKI outcome had no significant relationship with severe hematological toxicity or hepatotoxicity. AKI was associated with faster eGFR decline after a series of HDMTX treatment courses. Conclusions Patients with T2D have a higher sensitivity to AKI when administrated with HDMTX. This conclusion addresses safety concerns for making chemotherapy regimen for this population.

We introduce a novel approach for synthesizing realistic speeches for comics. Using a comic page as input, our approach synthesizes speeches for each comic character following the reading flow. It adopts a cascading strategy to synthesize speeches in two stages: Comic Visual Analysis and Comic Speech Synthesis. In the first stage, the input comic page is analyzed to identify the gender and age of the characters, as well as texts each character speaks and corresponding emotion. Guided by this analysis, in the second stage, our approach synthesizes realistic speeches for each character, which are consistent with the visual observations. Our experiments show that the proposed approach can synthesize realistic and lively speeches for different types of comics. Perceptual studies performed on the synthesis results of multiple sample comics validate the efficacy of our approach.

Various applications of human-computer interaction are based on the estimation of head pose, which is challenging due to different facial appearance, inhomogeneous illumination, partial occlusion, etc. In this paper, we propose a deep neural network following the Coarse-to-Fine strategy to estimate head poses. The scheme includes two branches: Coarse classification phase classifying the input image into four categories, and Fine Regression phase estimating the accurate pose parameters. The two sub-networks are trained jointly. To tackle the problem of insufficient annotated data in training process, we design a rendering pipeline to synthesize realistic head images and generate an annotated dataset with a collection of 310k head poses. The results on benchmark datasets and synthetic dataset validate the effectiveness of our approach, as well as the results on images with diverse illumination, occlusion, and motion blur. Moreover, our method can be easily extended to estimate head poses on depth images.

We studied serum levels of CD147, DcR3, and IL33 in different stages of KD to explore the value of CD147, DcR3, and IL33 in the pathophysiology of KD.Methods: We measured serum levels of CD147, DcR3, and IL33 by enzyme-linked immunosorbent assay (ELISA) at different stages with 71 KD patients and 66 healthy control children. We apply for network tools GeneMANIA and Cytoscape APP to analyze the functions of these pro-inflammatory factors at the gene and protein level.Results: Serum levels of CD147, DcR3, and IL33 were significantly increased in KD patients before IVIG treatment. Serum levels of CD147, DcR3, and IL33 gradually decreased over time after the treatment of IVIG. Eight cases were IVIG non-responders, while nine KD patients got CALs, but they did not overlap. And there were no statistical differences between group IVIG responders and IVIG non-responders or between groups without CALs and with CALs. We explored the functions of CD147, DcR3, and IL33 from GeneMANIA and Cytoscape APP and found these third pro-inflammatory factors were coexpressed, physical interactions, genetic interactions with other KD-related factors.Conclusion: CD147, DcR3, and IL33 are involved in the pathophysiology of KD, which provides novel evidence for diagnosing and treating KD with their inhibitors.Keywords: Kawasaki disease, coronary artery lesions, CD147, DcR3, IL33, PI3K/AKT pathway

Bronchopulmonary dysplasia (BPD) is a chronic lung disease most commonly occurring in premature infants, and its pathological manifestations are alveolar hypoplasia and dysregulation of pulmonary vasculature development. The effective treatment for BPD has not yet been established. Non-coding RNAs, including microRNAs and long non-coding RNAs do not encode proteins, but can perform its biological functions at the RNA level. Non-coding RNAs play an important role in the incidence and development of BPD by regulating the expression of genes related to proliferation, apoptosis, angiogenesis, inflammation and other cell activities of alveolar epithelial cells and vascular endothelial cells. Here we summarize the role of non-coding RNAs in BPD, which provides possible molecular marker and therapeutic target for the diagnosis and treatment of BPD.

Previous studies have reported abnormal muscle morphology and functions in patients with adolescent idiopathic scoliosis (AIS). To answer whether such abnormalities could be reflected in their circulation and their clinical implication for predicting curve progression to the surgical threshold, this preliminary study explored the presence of baseline muscle-related proteins and their association with curve progression. Plasma samples were collected at the first clinical visit for AIS, with patients divided into non-progressive or progressive groups (N = four and four) according to their Cobb angle in six-year follow-ups, with age- and sex-matched healthy subjects (N = 50). Then, the samples were subjected to isobaric tags for relative and absolute quantitation (iTRAQ) for global comparison of untargeted protein expression. Seventy-one differentially expressed proteins (DEPs) were found elevated in progressive AIS. Functional analysis showed that 18 of these are expressed in muscles and play an essential role in muscle activities. Among the muscle-related DEPs, α-actin had the highest fold change in progressive/non-progressive groups. This preliminary study firstly suggested higher circulating levels of muscle structural proteins in progressive AIS, indicating the likelihood of structural damage at the microscopic level and its association with progression to the surgical threshold. Further studies with larger sample sizes are warranted to validate these novel candidates for early diagnosis and predicting progression.

Enhanced external counterpulsation is a Food and Drug Administration–approved, non-invasive, assisted circulation therapy for ischemic cardiovascular and cerebrovascular diseases. Previous studies have confirmed that EECP stimulation induces largely different cerebral hemodynamic responses in patients with ischemic stroke and healthy controls. However, the underlying mechanisms remain uncertain. We hypothesize that different blood redistributions at the carotid bifurcation may play a key role. Ten subjects were enrolled in this study, namely, five patients with neurological disorders and five young healthy volunteers as controls. Magnetic resonance angiography (MRA) was performed on the carotid artery. All the subjects received a single session of EECP treatment, with external cuff pressures ranging from 20 to 40 kPa. Vascular ultrasound measurements were taken in the common carotid artery (CCA), external carotid artery (ECA) and internal carotid artery (ICA). Three-dimensional patient-specific numerical models were developed to calculate the WSS-derived hemodynamic factors. The results indicated that EECP increased CCA and ECA blood flow in both groups. The ICA blood flow in the patient group exhibited a mean increase of 6.67% during EECP treatment compared with the pre-EECP condition; a mean decrease of 9.2% was observed in the healthy control group. EECP increased the averaged wall shear stress (AWSS) throughout the carotid bifurcation in the patient group; the ICA AWSS of the healthy group decreased during EECP. In both groups, the oscillatory shear index (OSI) in the ICA increased proportionally with external cuff pressure. In addition, the relative resident time (RRT) was constant or slightly decreased in the CCA and ECA in both groups but increased in the ICA. We suggest that the benefits of EECP to patients with neurological disorders may partly result from blood flow promotion in the ICA and increase in WSS at the carotid bifurcation. In the healthy subjects, the ICA blood flow remained constant during EECP, although the CCA blood flow increased significantly. A relatively low external cuff pressure (20 kPa) is recommended as the optimal treatment pressure for better hemodynamic effects. This study may play an important role in the translation of physiological benefits of EECP treatment in populations with or without neurological disorders.

Background interference of impurity in strong water is the key factor restricting the detection distance and target characteristics of the weak target. Aiming at the problems of less echo characteristics and difficult identification of the characteristics of the underwater weak target laser detection, the paper studies the problem of the broadening of the laser echo pulse of the weak target under the background of strong water interference. A system of underwater weak target laser measurement and analysis is designed to suppress the near-field water scattering interference. The system can achieve matching adjustment among laser emission energy, laser beam angle and laser APD receiving gain. The detection performance of different underwater weak targets (small-scale metal, black rubber, low reflectivity reef, water grass, transparent plastic) was tested under different laser emission energy, different detection distance and different laser echo receiving gain. Through statistical analysis of data, the pulse width and material of the target of different underwater weak targets were obtained The relationship between the power of pulse laser, the distance between laser emission and target. The results show that the pulse broadening characteristics of the laser echo signal can be used to identify the weak target laser detection characteristics under the background of strong water impurity interference.

To enlarge the linear detection range without sacrificing the sensitivity is one of the urgent problems in the development of high-performance piezoresistive flexible sensors. Inspired by a multilayer corrugated board, this study develops a new multilayer microspherical sensor in which conductive core-shell SiO2/Polyaniline (PANI) (PS) microspheres serve as active particles, while insulated silk fibroin (SF)/poly lactic-co-glycolic acid (PLGA) (SP) fibers are used as the support. The size of conductive microspheres attached to the insulated layer is controllable. The multiple layers of assembly endow the flexible sensor with a high sensitivity (0.071 kPa−1) and a wide linear detection (from 10 Pa to 380 kPa) simultaneously. This corrugated sensor also have a fast response time (145 ms) and an excellent durability (over 2000 cycles), and it can be used to detect human joint pressure signals and transmit encrypted information. Moreover, flexible keyboard, safety protection of machinery, as well as object position tracking can be achieved based on this sensor. Most importantly, the sensor encapsulated by biological polysaccharide kappa-carrageenan (KC) is skin-friendly and breathable, and it can be decomposed in 90 °C hot water. In conclusion, this multilayer microspherical sensor presents great potential for flexible wearable devices.

Common pursuits of developing nanometric logic and neuromorphic applications have motivated intensive research studies into low-dimensional resistive random-access memory (RRAM) materials. However, fabricating resistive switching medium with inherent stability and homogeneity still remains a bottleneck. Herein, we report a self-assembled uniform biphasic system, comprising low-resistance 3 nm-wide (Bi0.4,La0.6)FeO3-δ nanosheets coherently embedded in a high-resistance (Bi0.2,La0.8)FeO3-δ matrix, which were spinodally decomposed from an overall stoichiometry of the (Bi0.24,La0.76)FeO3-δ parent phase, as a promising nanocomposite to be a stable and endurable RRAM medium. The Bi-rich nanosheets accommodating high concentration of oxygen vacancies as corroborated by X-ray photoelectron spectroscopy and electron energy loss spectroscopy function as fast carrier channels, thus enabling an intrinsic electroforming-free character. Surficial electrical state and resistive switching properties are investigated using multimodal scanning probe microscopy techniques and macroscopic I-V measurements, showing high on/off ratio (∼103) and good endurance (up to 1.6 × 104 cycles). The established spinodal decomposition-driven phase-coexistence BLFO system demonstrates the merits of stability, uniformity, and endurability, which is promising for further application in RRAM devices.

Objective: To explore the efficacy comparison between epidermal growth factor receptor–tyrosine kinase inhibitors (EGFR-TKIs) combined with traditional Chinese medicine (TCM) and single EGFR-TKIs for advanced non-small cell lung cancer (NSCLC).Methods: A total of 91 NSCLC patients with EGFR mutation were divided into an experimental group and a control group (in a ratio of 2:1) to receive TCM and EGFR-TKIs (61 cases) or single EGFR-TKIs (30 cases). Patients in the control group took EGFR-TKIs and those in the experimental group took EGFR-TKIs plus TCM. We analyzed the progression-free survival (PFS), overall survival (OS), disease control rate (DCR), and treatment-related adverse events of two groups.Results: The mPFS of the experimental group and the control group was 12.3 and 8.9 months (P = 0.02), respectively, and the mOS of the experimental group and the control group was 28.2 and 24.2 months (P = 0.02), respectively. Subgroup analysis showed that for the patients with exon 19 deletion mutation (19DEL), mPFS between experimental group and control group was 12.7 and 10.1 months, respectively (P = 0.12). For exon 21 deletion mutation (L858R), the PFS of two groups was 10.8 vs. 8.2 months, respectively (P = 0.03). The subgroup analysis also showed that, for the patients with exon 19 deletion mutation, mOS between the experimental group and the control group was 30.3 and 28.7 months, respectively (P = 0.19). For exon 21 deletion mutation, the mOS of two groups was 25.5 vs. 21.3 months, respectively (P = 0.01). The DCR of the experimental group and the control group was 93.3% and 80.1%, respectively (P = 0.77). Grade 3–4 treatment-related adverse events were less common with the experimental group (11.48%) than the control group (26.67%).Conclusion: For NSCLC patients with EGFR mutation, EGFR-TKIs combined with TCM had a certain effect to prolong mPFS and mOS, compared with the use of EGFR-TKIs alone, especially for the patients with L858R. This conclusion has a significant effect on improving the survival of NSCLC patients after EGFR-TKIs resistance. It deserves further study.

Since thermal power generation is still one of the main sources of carbon emissions in China, the economic benefits and productivity of the thermal power generation industry have been seriously affected in recent years with the increasingly strict environmental regulations and restrictions on carbon emissions, as well as by the sharp fluctuations of coal prices. Therefore, it has been an important issue to improve the productivity performance of the thermal power industry. Due to the regional heterogeneity among different regions of China, we introduced a meta-frontier framework into the energy cost productivity model to develop a meta-energy cost productivity model. The energy cost gap between the group-specific and meta-frontiers was also utilized to assess the convergence rate of the group-specific frontier to the meta-frontier. The estimated results present that the energy cost efficiency of the eastern region outperformed that of the other two regions, and the cost Malmquist (CM) productivity of these three regions all showed positive growth, in which the progress of allocative efficiency and price effect were the main driving factors. Additionally, the central and western regions displayed the convergence of group-specific CM productivity towards the meta-frontier.

Background and Aims The characteristics of valvular calcification (VC) in early stage are extracellular matrix (ECM) accumulation and muti-cells activation. In our previous work, we found high-phosphorus (HP) diet aggravated ECM accumulation in both aortic valve and mitral valve in rats with chronic kidney disease (CKD). However, the underlying mechanism of HP contribution in ECM accumulation of CKD-induced VC is still unknown. Method canine valvular interstitial cells (VICs), valvular endothelial cells (VECs) and human umbilical vein endothelial cells (HUVECs) were used in this study. CKD mice (C57b and Tek-EGFP-PolyA) was build by 0.2% adenine-diet for 6 weeks and HP diet/NP diet for 10 weeks. Results As for VICs, HP induced qVICs transfer into aVICs, not oVICs, which was characterized with upregulated level of smoothelin and viemitin. There was no calcium accumulation was observed, suggesting that VICs do not have the ability to synthesize calcium crystals under pure HP intervention. As for VECs, aVICs activated by HP induced VECs EndMT in a transwell-assay, which was characterized with decreasing protein levels of endothelial markers (CD31, vWF, VE-cadherin) and increasing protein levels of mesenchymal makers (α-SMA, FSP1, N-cadherin). Then, IL-8 was found as the main factor releasing from VICs to induce VECs EndMT. In vitro, the concentration of IL-8 in the lower chamber could reach 2-4ng/ml. Reparixin was used to inhibit IL-8 receptor of VECs, which could relive aVICs-induced EndMT. In vivo, the expression of valve CXCL-2 (the mouse IL8 functional homolog) was increased in HP-diet compared with NP-diet, though the serum level of CXCL-2 was similar between two groups. AAV9-sm22a-CXCL-2 and Reparixin could inhibit VECs EndMT by inhibiting VICs relasing CXCL-2 and inhibiting VECs IL-8 receptor in CKD mice of Tek-EGFP-PolyA respectively. Then, IL-8 was found to induced VECs EndMT by miR-214-3p/PTEN/Akt pathway. Inhibiting EndMT by blocking IL-8/miR-214-3p could alleviate ECM accumulation. Conclusion HP could induce qVICs transfer into aVICs, and aVICs could cause VECs EndMT via IL-8/miR-214-3p/PTEN/Akt pathway. Both take part in ECM accumulation in CKD-induced VC.

Background Osteoarthritis of the knee is a common degenerative joint disorder in our ageing population. A combination of thermal therapy with a self-management exercise have shown a positive effect in the management of osteoarthritis of the knee. This study aimed to compare the effectiveness of topical heat pack versus focal application of heat therapy at the acupressure points in the treatment of osteoarthritis of the knee. Methods A randomized controlled trial was conducted in 76 patients with osteoarthritis of the knee, diagnosed by an experienced orthopedic surgeon. Following inclusion and exclusion selection, patients were randomly allocated to group 1 (Heat pack) or group 2 (Thermal gun). All patients received 30 min of treatment in each session, twice a week for 4 weeks. They also received an education program and taught home knee exercises. Outcome measurements were the visual analog scale (VAS) for pain intensity, muscle power, knee ROM, WOMAC and SF-12v2. Results In the Thermal gun group, function and total scores (WOMAC) and Physical Composite Scale (SF-12v2) were significantly improved after 8 sessions. Quadriceps strength was significantly improved after 8 weeks (from 4.42 to 4.63; p = 0.02). In the Heat pack group, flexion was significantly improved after 8 sessions (p = 0.02). Mean VAS scores after Heat pack treatment was consistently better (lower) than mean VAS scores after Thermal gun treatment. Conclusion The combination of focal thermal therapy at acupressure points is a viable conservative treatment in osteoarthritis of the knee. The pressure at the acupressure points has a synergistic benefit than topical thermal therapy alone. Background. This study is aimed at characterizing the human distal airway stem cells (DASCs) and assessing their therapeutic potential in patients with chronic, degenerative lung diseases. These findings will provide a comprehensive understanding for further clinical applications utilizing autologous airway stem cells as therapeutic intervention in respiratory diseases. Methods. DASCs were isolated from healthy subjects or patients diagnosed with bronchiectasis, chronic obstructive pulmonary diseases (COPD), or interstitial lung disease (ILD). Differentiation capacity, a key property of the stem cells, was studied using a novel monolayer differentiation system. The differentiated cells were evaluated for alveolar and bronchial cell marker expression, and the quantified expression level of differentiated cells was further examined for their relationship with age and pulmonary function of the subjects. Results and Conclusions. Differentiation of DASCs and tracheal stem cells (TSCs) yielded an alveolus-like structure and a tube-shaped structure, respectively, with distinct marker gene expression. Additionally, single-cell-derived clones showed diverse differentiation fates, even if the clones arise from identical or different individuals. More importantly, the alveolar differentiation potency was higher in DASCs derived from patients than from healthy people. The differentiation efficiency of DASCs also correlates with age in patients with bronchiectasis and ILD.

Cutaneous melanoma is one of the most common malignant skin tumors and advanced melanoma is usually associated with a poor prognosis. In the current study, we demonstrated the tumor suppressing role of epithelial membrane protein-2 (EMP2) by inducing apoptosis in a A375 human melanoma cell line. Mechanistically, the low expression of EMP2 in melanoma is partially due to autophagic protein degradation mediated by the mTOR pathway. These results suggest there is regulation of autophagy as well as EMP2 levels might be an interesting novel targeted therapeutic strategy for melanoma. Although the further investigation is needed to deeply understand the regulatory mechanisms of EMP2 in melanoma progression and metastasis, our results clarify the functions and mechanisms of autophagy in melanoma, and shed new light on novel targeted therapeutics for melanoma.

Objectives Chronic obstructive pulmonary disease (COPD) is characterized by irreversible lung tissue damage including chronic bronchitis and emphysema, which could further develop into respiratory failure. Many studies have revealed a potential regenerative function of the distal airway stem/progenitor cells (DASCs) after lung injury. Materials and Methods Mouse and human DASCs were expanded, analysed, and engrafted into injured mouse lungs. Single‐cell analyses were performed to reveal the differentiation path of the engrafted cells. Finally, human DASCs were transplanted into COPD mice induced by porcine pancreatic elastase (PPE) and lipopolysaccharide (LPS) administration. Results We showed that isolated mouse and human DASCs could be indefinitely expanded and were able to further differentiate into mature alveolar structures in vitro. Single‐cell analysis indicated that the engrafted cells expressed typical cellular markers of type I alveolar cells as well as the specific secreted proteins. Interestingly, transplantation of human DASCs derived from COPD patients into the lungs of NOD‐SCID mice with COPD injury repaired the tissue damage and improved the pulmonary function. Conclusions The findings demonstrated that functional lung structure could be reconstituted by intrapulmonary transplantation of DASCs, suggesting a potential therapeutic role of DASCs transplantation in treatment for chronic obstructive pulmonary disease., Mouse and human DASCs could be indefinitely expanded and were able to further differentiated into mature alveolar structures during in vitro cultures. Coronavirus Disease 2019 (COVID-19) has become one of the most urgent public health events worldwide due to its high infectivity and mortality. Computed tomography (CT) is a significant screening tool for COVID-19 infection, and automatic segmentation of lung infection in COVID-19 CT images can assist diagnosis and health care of patients. However, accurate and automatic segmentation of COVID-19 lung infections is faced with a few challenges, including blurred edges of infection and relatively low sensitivity. To address the issues above, a novel dilated dual attention U-Net based on the dual attention strategy and hybrid dilated convolutions, namely D2A U-Net, is proposed for COVID-19 lesion segmentation in CT slices. In our D2A U-Net, the dual attention strategy composed of two attention modules is utilized to refine feature maps and reduce the semantic gap between different levels of feature maps. Moreover, the hybrid dilated convolutions are introduced to the model decoder to achieve larger receptive fields, which refines the decoding process. The proposed method is evaluated on an open-source dataset and achieves a Dice score of 0.7298 and recall score of 0.7071, which outperforms the popular cutting-edge methods in the semantic segmentation. The proposed network is expected to be a potential AI-based approach used for the diagnosis and prognosis of COVID-19 patients.

Background. Sepsis is a major cause of morbidity and mortality worldwide. Sepsis with acute kidney injury (AKI) is associated with higher mortality risk when compared with those with sepsis and without AKI. Therefore, it is necessary to detect the predictors of sepsis-associated acute kidney injury (SA-AKI) in order to timely prevent, diagnose and treat this complication. Methods. From July 2016 to December 2019, 419 patients with sepsis admitted to the intensive care unit (ICU) were randomly divided into two groups: training group (n= 302) and validation group (n = 117). A least absolute shrinkage and selection operator (LASSO) regression was constructed to select variables within 24 h of admission, and then were included in a logistic regression model to find the independent risk factors of AKI. Hence, a nomogram for predicting SA-AKI with statistically significant covariates was constructed. Discrimination, calibration, and clinical utility of the nomogram performance were assessed and then validated.Results. The risk factors yielded by logistic regression were hypertension(HT), diabetes mellitus(DM), C-reactive protein(CRP), procalcitonin(PCT), activated partial thromboplastin time(APTT), platelet(PLT), and then were incorporated into the nomogram. The areas under the ROC curve of the nomogram in the training and validation groups were 0.856 and 0.885, respectively. The calibration curves demonstrated favorable consistency between the predictions of nomogram and the actual observations in both training as well as validation groups. Decision curve analysis (DCA) showed clinical usefulness of the proposed nomogram model.Conclusions. A risk prediction model by integrating variables can assist in identifying patients who are at high risk of developing SA-AKI. The nomogram had excellent predictive ability and might have significant clinical implications for early detection of AKI in patients undergoing sepsis.

PurposeStandardised Training of Paediatric Resident (STPR) plays an essential role in training qualified paediatricians. Until now, China had no paediatric resident competency index system to effectively guide and evaluate the competence of paediatric residents. This study aimed to establish a competency index system for paediatric residents in China to provide a reference for improving the training system and quality of STPR.Study design and settingThis study conducted two rounds of Delphi expert consultation survey among paediatric medical experts (n=16), followed by screening, revising and supplementing indicators using the boundary value method. Next, the analytic hierarchy process was used to determine the weight of indicators and finally establish a competency index system for paediatric residents.ResultsThe results of the statistical analysis revealed a positive coefficient of 100% for both rounds of expert consultation. The expert authority coefficient values were 0.82 and 0.83, and the expert coordination coefficient test was pConclusionsIn this study, a competency index system for paediatric residents was constructed following the characteristics and quality requirements for paediatric residents in China and is expected to significantly improve the overall level of paediatricians’ medical service quality and supply.

Objective: To analyze the expression and clinical significance of retinoic acid-induced protein 14 ( RAI14) in gastric cancer and its relationship with immune cell infiltration by mining databases such as Oncomine, TIMER, UALCAN, and Kaplan Meier Plotter. Methods: RAI14 expression in various cancer types was analyzed using the Oncomine and TIMER databases. We used the Kaplan-Meier Plotter and UALCAN databases to evaluate the impact of RAI14 on clinicopathological parameters in gastric cancer. The correlation between RAI14 expression and immune cell invasion was studied using TIMER. TIMER was also used to analyze the correlation between RAI14 expression and marker levels of tumor-infiltrating immune cells. Results: High RAI14 expression in gastric cancer was significantly associated with poor overall survival (OS; hazard ratio [HR] = 1.82, 95% confidence interval [CI] = 1.53–2.15, P < 0.001) and poor progression-free survival (PFS; HR = 2.16, 95% CI = 1.77–2.65, P < 0.001). Furthermore, high RAI14 expression was significantly associated with poor prognosis of patients with stage 2–4 gastric cancer, but not with OS and PFS of stage 1 patients (OS P = 0.17; PFS P = 0.09), and patients with stage N0 PFS had nothing to do (PFS P = 0.238). RAI14 expression was positively correlated with the infiltration levels of monocytes, tumor-associated macrophages, macrophages, neutrophils, and Treg cells in gastric cancer. Besides, RAI14 expression was closely related to various marker genes in immune cells. Conclusion: RAI14 is highly expressed in gastric cancer, and its expression level is correlated with the prognosis of patients with gastric cancer. RAI14 plays also an important role in the recruitment and regulation of infiltrating immune cells and is, thus, expected to become a target for the optimal treatment of gastric cancer.

Ephb6 gene knockout causes hypertension in castrated mice. EPHB6 controls catecholamine secretion by adrenal gland chromaffin cells (AGCCs) in a testosterone-dependent way. Nicotinic acetylcholine receptor (nAChR) is a ligand-gated Ca2+/Na+ channel, and its opening is the first signaling event leading to catecholamine secretion by AGCCs. There is a possibility that nAChR might be involved in EPHB6 signaling, and thus sequence variants of its subunit genes are associated with hypertension risks. CHRNA3 is the major subunit of nAChR used in human and mouse AGCCs. We conducted a human genetic study to assess the association of CHRNA3 variants with hypertension risks in hypogonadic males. The study cohort included 1,500 hypogonadic Chinese males with (750 patients) or without (750 patients) hypertension. The result revealed that SNV rs3743076 in the fourth intron of CHRNA3 was significantly associated with hypertension risks in the hypogonadic males. We further showed that EPHB6 physically interacted with CHRNA3 in AGCCs, providing a molecular basis for nAChR being in the EPHB6 signaling pathway.

In this paper, we propose a novel optimization framework to synthesize an aesthetic pose for the virtual character with respect to the presented user's pose. Our approach applies aesthetic evaluation that exploits fully connected neural networks trained on example images. The aesthetic pose of the virtual character is obtained by optimizing a cost function that guides the rotation of each body joint angles. In our experiments, we demonstrate the proposed approach can synthesize poses for virtual characters according to user pose inputs. We also conducted objective and subjective experiments of the synthesized results to validate the efficacy of our approach.

In this paper, we introduce an interactive background music synthesis algorithm guided by visual content. We leverage a cascading strategy to synthesize background music in two stages: Scene Visual Analysis and Background Music Synthesis. First, seeking a deep learning-based solution, we leverage neural networks to analyze the sentiment of the input scene. Second, real-time background music is synthesized by optimizing a cost function that guides the selection and transition of music clips to maximize the emotion consistency between visual and auditory criteria, and music continuity. In our experiments, we demonstrate the proposed approach can synthesize dynamic background music for different types of scenarios. We also conducted quantitative and qualitative analysis on the synthesized results of multiple example scenes to validate the efficacy of our approach.

In the last several decades, apoptosis interference has been considered clinically irrelevant in the context of renal injury. Recent discovery of programmed necrotic cell death, including necroptosis, ferroptosis, and pyroptosis refreshed our understanding of the role of cell death in kidney disease. Pyroptosis is characterized by a lytic pro- inflammatory type of cell death resulting from gasdermin-induced membrane permeabilization via activation of inflammatory caspases and inflammasomes. The danger-associated molecular patterns (DAMPs), alarmins and pro-inflammatory cytokines are released from pyroptotic cells in an uncontrolled manner, which provoke inflammation, resulting in secondary organ or tissue injuries. The caspases and inflammasome activation-related proteins and pore-forming effector proteins known as GSDMD and GSDME have been implicated in a variety of acute and chronic microbial and non-microbial kidney diseases. Here, we review the recent advances in pathological mechanisms of pyroptosis in kidney disease and highlight the potential therapeutic strategies in future.

Objective Emerging evidence suggest abnormal bone metabolism and defective bone qualities are associated to etipathogenesis of Adolescent Idiopathic Scoliosis (AIS). Systemic low bone mass is important prognosticator to predict risk of curve progression in AIS. The underlying mechanism is still unclear. We hypothesize that aberrant bone turnover correlates with bone qualities in AIS and associates to risk of curve progression. Subjects and methods Two cohorts were included in this study. The case-control study recruited 161 AIS girls and 161 ethnic/age-matched healthy girls. The longitudinal cohort recruited 128 AIS girls with two-year follow-up. Areal bone mineral density (BMD) at femoral necks were measured with dual-energy x-ray absorptiometry (DXA), and bone qualities of distal radius by high-resolution peripheral quantitative computed tomography (HR-pQCT). Time-lapse analysis of registered HR-pQCT images estimated local bone remodeling quantitatively. Serum levels of CTX and P1NP were measured with ELISA kits. Results AIS presented significantly higher serum level of P1NP. In both AIS and control, the negative correlations were consistently observed between serum CTX/P1NP levels and most cortical bone quality parameters after adjustment to age. Significant correlation between serum bone turnover markers and trabecular bone parameters have been observed only in control. Progressive AIS has significant increase of serum P1NP level at first clinic visit. Time lapse register analysis showed high bone resorption and low net bone gain was associated with risk of progression in AIS. Conclusions Our study characterized AIS with higher serum bone turnover markers, which may contribute to defective bone qualities in AIS. For the first time, we showed that progressive AIS had higher systemic bone turnover markers level and local bone remodeling. This fresh evidence indicated association between disrupted bone turnover and risk of progression of AIS, which set the foundation of new prognostic method and of novel treatment target to curve progression. This study demonstrated the importance of bone metabolism in developing disease management of AIS to achieve goal of early prediction and non-surgical modulation.

Background. Rosacea is a common inflammatory skin disorder. Several studies, but not all, have suggested a high prevalence of cardiovascular diseases (CVDs) in rosacea patients. This study is aimed at investigating the association between rosacea and CVDs and related risk factors. Methods. We performed a literature search through PubMed, Embase, and Web of Science databases, from their respective inception to December 21, 2019. Two reviewers independently screened the articles, extracted data, and performed analysis, following the PRISMA guidelines. Odds ratios (OR) or standardized mean differences (SMD) and 95% confidence intervals (CI) were calculated for outcomes. The included studies’ quality was evaluated using the Newcastle Ottawa Scale (NOS). Results. The final meta-analysis included ten studies. The pooled analysis found no association between rosacea prevalence and the incidence of CVDs (OR 0.97; 95% CI 0.86-1.10). Rosacea was found to be significantly associated with several risk factors for CVDs (OR 1.17; 95% CI 1.05-1.31), including hypertension (OR 1.17; 95% CI 1.02-1.35), dyslipidemia (OR 1.34; 95% CI 1.00-1.79), and metabolic syndrome (OR 1.72; 95% CI 1.09-2.72). However, no association was found between rosacea and diabetes mellitus (OR 0.98; 95% CI 0.82-1.16). Among the biological parameters, a significant association was found between rosacea and total cholesterol (SMD=0.40; 95% CI=−0.00, 0.81; p<0.05), low-density lipoprotein cholesterol (SMD=0.28; 95% CI=0.01, 0.56; p<0.05), and C-reactive protein (CRP) (SMD=0.25; 95% CI=0.10, 0.41; p<0.05). We found no association between rosacea and high-density lipoprotein cholesterol (SMD=0.00; 95% CI=−0.18, 0.18; p=0.968) or triglycerides (SMD=0.10; 95% CI=−0.04, 0.24; p=0.171). Conclusions. Although no significant association was found between rosacea and CVDs, rosacea was found to be associated with several of related risk factors. Patients with rosacea should pay more attention to identifiable CVD risk factors, especially those related to inflammatory and metabolic disorders.

Background To promote the integration of medical resources, Beijing has built medical alliances since 2012, but this reform has not changed the state of disordered medical treatment. Patients are still willing to go to high-level hospitals for medical treatment. What causes patients to prefer high-level hospitals? To explore the reasons behind this preference for high-level medical treatment among patients and to guide patients to make visits in an orderly manner, we conducted this study and compared patients’ satisfaction with hospitals of different levels in the context of medical resource integration. Methods This study conducted a questionnaire survey among 1250 patients who were selected in 18 medical alliances in Beijing from October to December 2016. The study type is a comparative study based on cross-sectional data. Patient satisfaction was the main outcome, and descriptive analysis, chi-square tests, nonparametric tests and binary logistic regression analysis were used. The level of statistical significance was set at p Results The overall satisfaction score of the medical alliances was 3.375, and the satisfaction scores of core hospitals and cooperative hospitals were 2.77 and 3.07, respectively. The overall patient satisfaction rate was 44.62%, and the satisfaction rates of core hospitals and cooperative hospitals were 34.34 and 50.43%, respectively. The type of hospital and understanding of medical alliance policy were the factors associated with patient satisfaction with the medical alliance. Conclusions The patients’ satisfaction with cooperative hospitals was higher than their satisfaction with core hospitals. Although the patients were more satisfied with the service attitude of the cooperative hospitals, they still preferred core hospitals due to their higher expectations for their medical treatment and greater trust in their medical technology. It is necessary to explore the establishment of closed medical alliances under the unified management of human and financial resources to enable medical alliances to become a community of common interests and provide integrated medical services for patients.

Significant advancements in the understanding of signaling pathways associated with IL-37 have been made in recent years. IL-37 binds to IL-18R and recruits IL-1R8 to form the IL-37/IL-1R8/IL-18Rα complex. Capase-1 plays a key role in the nuclear transduction of IL-37 signal, processing precursor IL-37 into the mature isoform, and interacting with Smad3. IL-37 exerts its role by activating anti-inflammation pathways including AMPK, PTEN, Mer, STAT3 and p62, and promoting tolerogenic dendritic cells and Tregs. In addition, IL-37 inhibits pro-inflammatory cytokines such as IL-1, IL-6, IL-8, IL-17, IL-23, TNF-α, and IFN-γ, and suppresses Fyn, MAPK, TAK1, NFκB, and mTOR signaling. The final effects of IL-37 depend on the interaction among IL-18R, IL-1R8, IL-37 and IL-18BP. Previous studies have deciphered the role of IL-37 in the development and pathogenesis of autoimmune diseases, chronic infections and cancer. In this review, we discuss the role of IL-37 in psoriasis, atopic dermatitis, Behcet’s diseases, systemic lupus erythematosus, and other skin and connective tissue diseases.

Although intravenous immunoglobulin (IVIG) can effectively treat Kawasaki disease (KD), 10–20% of KD patients show no beneficial clinical response. Developing reliable criteria to discriminate non-responders is important for early planning of appropriate regimens. To predict the non-responders before IVIG treatment, gene expression dataset of 110 responders and 61 non-responders was obtained from Gene Expression Omnibus. After weighted gene co-expression network analysis, we found that modules positively correlated with the non-responders were mainly associated with myeloid cell activation. Transcripts up-regulated in the non-responders, IL1R2, GK, HK3, C5orf32, CXCL16, NAMPT and EMILIN2, were proven to play key roles via interaction with other transcripts in co-expression network. The crucial transcripts may affect the clinical response to IVIG treatment in acute KD. And these transcripts may serve as biomarkers and therapeutic targets for precise diagnosis and treatment of the non-responders.

The long text in our study involves a complete discourse structure with a title and approximately 1500 Chinese characters within. Long text sentiment analysis has faced a lot of difficulties on the grounds that a long text often comprises multiple sentiments and diverse focuses. This paper proposes to analyze the sentiment of long texts based on the combination of title and topic sentence. Based on the fine-labeled texts, the analysis extracted the important features of topic sentences, such as location, feature words, degree of topic relevance and emotional words. Then two topic sentences were extracted from each text that can best represent the topic through weighted calculation of these multi-dimensional features. Next this paper combined the two topic sentences with the title to complete the sentiment analysis of the whole document. The topic sentence extraction method has achieved good results in the task of extracting and judging key emotional sentences of news in the 6th COAE (Chinese Opinion Analysis Evaluation) (2014), which shows that the method is effective. This method has been put into practice in the National Language Public Opinion Monitoring system, with an accuracy of 0.82.

In the past few years, although annealing effects have been widely used to restore the power features of fiber amplifiers, their noise feature has not yet been sufficiently investigated. Thanks to the recovery of partial color centers (especially the ${{\rm Yb}^{2 + }}$Yb2+ center) through photobleaching, the absorption of seed signal light due to the presence of color centers is weakened, and the sensitized path of the irradiated erbium-ytterbium co-doped fiber amplifier (EYDFA) is partially restored, which helps to suppress amplified spontaneous emission as well as increases the gain of the EYDFA. The thermal annealing process is performed by the first amplification stage of the EYDFA, and the increase in the optical signal-to-noise ratio of the signal light indicates that our proposed scheme could be also exploited to further improve the output performance of the EYDFA. This work is anticipated to open up new prospects for future optical communications systems based on pre-amplification technology.

To address the problem that the traditional fault feature extraction method cannot be applied to the field of weak fault detection of autonomous underwater vehicles, a multidimensional feature extraction method based on wavelet packets and the Fast Fourier Transform (FFT) is presented. The method in this paper first reduces noise in the controller and sensor data based on the wavelet packets. Then, the empirical mode decomposition (EMD) and FFT methods are used to extract the energy features and the improved frequency domain feature of the noise-reduced data, respectively. The experimental results show that this method provide identification accuracies that are reliable and high, and it is suitable for the identification of weak faults for autonomous underwater vehicles.

Severe pulmonary infection is a major threat to human health accompanied by substantial medical costs, prolonged inpatient requirements, and high mortality rates. New antimicrobial therapeutic strategies are urgently required to address the emergence of antibiotic resistance and persistent bacterial infections. In this study, we show that the constitutive expression of a native antimicrobial peptide LL‐37 in transgenic mice aids in clearing Pseudomonas aeruginosa (PAO1), a major pathogen of clinical pulmonary infection. Orthotopic transplantation of adult mouse distal airway stem cells (DASCs), genetically engineered to express LL‐37, into injured mouse lung foci enabled large‐scale incorporation of cells and long‐term release of the host defense peptide, protecting the mice from bacterial pneumonia and hypoxemia. Further, correlates of DASCs in adult humans were isolated, expanded, and genetically engineered to demonstrate successful construction of an anti‐infective artificial lung. Together, our stem cell‐based gene delivery therapeutic platform proposes a new strategy for addressing recurrent pulmonary infections with future translational opportunities., Using adult distal airway stem cells (DASCs) as a natural vehicle to deliver the antimicrobial peptide LL‐37 into damaged lung, this study reports a technology to restore epithelium barrier and pulmonary innate immunity, which could have applications for the treatment of infectious lung diseases.

Adolescent idiopathic scoliosis (AIS) is a prevalent spinal deformity occurring during peripubertal growth period that affects 1-4% of adolescents globally without clear etiopathogenetic mechanism. Low bone mineral density is an independent and significant prognostic factor for curve progression. Currently, the cause underlying low bone mass in AIS remains elusive. Osteocytes play an important role in bone metabolism and mineral homeostasis, but its role in AIS has not been studied. In the present study, iliac bone tissues were harvested from 21 patients with AIS (mean age of 14.3 ± 2.20 yr old) with a mean Cobb angle of 55.6 ± 10.61° and 13 non-AIS controls (mean age of 16.5 ± 4.79 yr old) intraoperatively. Acid-etched scanning electron microscopy (SEM) images of AIS demonstrated abnormal osteocytes that were more rounded and cobblestone-like in shape and were aligned in irregular clusters with shorter and disorganized canaliculi. Further quantitative analysis with FITC-Imaris technique showed a significant reduction in the canalicular number and length as well as an increase in lacunar volume and area in AIS. SEM with energy-dispersive X-ray spectroscopy analysis demonstrated a lower calcium-to-phosphorus ratio at the perilacunar/canalicular region. Moreover, microindentaion results revealed lower values of Vickers hardness and elastic modulus in AIS when compared with controls. In addition, in the parallel study of 99 AIS (27 with severe Cobb angle of 65.8 ± 14.1° and 72 with mild Cobb angle of 26.6 ± 9.1°) with different curve severity, the serum osteocalcin level was found to be significantly and negatively associated with the Cobb angle. In summary, the findings in this series of studies demonstrated the potential link of abnormal osteocyte lacuno-canalicular network structure and function to the observed abnormal bone mineralization in AIS, which may shed light on etiopathogenesis of AIS.-Chen, H., Zhang, J., Wang, Y., Cheuk, K.-Y., Hung, A.
