Chromatin-modifying enzymes, especially protein arginine methyltransferases (PRMTs), have been identified as candidate targets for cancer. Cellular or animal-based evidence has suggested an association between coactivator-linked arginine methyltransferase 1 (CARM1) and cancer progression. However, the relationship between CARM1 and patient prognosis and immune infiltration in pancancer patients is unknown. On the basis of the GEO and TCGA databases, we first investigated the possible oncogenic functions of CARM1 in thirty-three tumor types. CARM1 expression was elevated in many types of tumors. In addition, there was a significant association between CARM1 expression and the survival rate of tumor patients. Uterine corpus endometrial carcinoma (UCES) samples had the highest CARM1 mutation frequency of all cancer types. In head and neck squamous cell carcinoma (HNSC) and lung squamous cell carcinoma (LUSC), CARM1 expression was associated with the level of CD8+ T cell infiltration, and cancer-associated fibroblast infiltration was also observed in other tumors including kidney renal papillary cell carcinoma (KIRC) and prostate adenocarcinoma (PRAD). CARM1 was involved in immune modulation and played an important role in the tumor microenvironment (TME). Furthermore, activities associated with RNA transport and its metabolism were included in the possible mechanisms of CARM1. Herein, our first pancancer research explores the oncogenic role of CARM1 in various tumors. CARM1 is associated with immune infiltrates and can be employed as a predictive biomarker in pancancer.

PurposeThe spread of the Internet has transformed the dating landscape. Given the increasing popularity of online dating and rising immigration to Canada, this study takes an intersectional lens to examine nativity and gender differentials in heterosexual online dating.Design/methodology/approachIn 2018, a random-digit-dial telephone survey was conducted in Canada. Logistic regression models were used to analyze original data from this survey (N = 1,373).FindingsResults show that immigrants are more likely than native-born people to have used online dating in Canada, possibly because international relocation makes it more difficult for immigrants to meet romantic partners in other ways. In online-to-offline transitions, both native-born and immigrant online daters follow gendered scripts where men ask women out for a first date. Finally, immigrant men, who likely have disadvantaged positions in offline dating markets, also experience the least success in finding a long-term partner online.Originality/valueExtending search theory of relationship formation to online dating, this study advances the understanding of change and continuity in gendered rituals and mate-selection processes in the digital and globalization era. Integrating search theory and intersectionality theory, this study highlights the efficiency of using the Internet to search for romantic partners and the socially constructed hierarchy of desirability as interrelated mechanisms that produce divergent online dating outcomes across social groups. Internet dating, instead of acting as an agent of social change, may reproduce normative dating practices and existing hierarchies of desirability.

Three-dimensional (3-D) freehand ultrasound (US) imaging has been applied to the investigation of spine deformity. However, it is a challenge for the current 3-D imaging reconstruction algorithms to achieve a balance between image quality and computation time. The objectives of this article are to implement a new fast reconstruction algorithm that can fulfill the request of immediate demonstration and processing for high-quality 3-D spine imaging, and to evaluate the reliability and accuracy of scoliotic curvature measurement when using the algorithm. The fast dot-projection (FDP) algorithm was applied for voxel-based nearest neighbor (VNN), multiple plane interpolation (MPI), and pixel nearest neighbor (PNN) protocols to reduce the reconstruction time. The 3-D image volume was reconstructed from the datasets acquired from scoliotic subjects. The computational cost, image characteristics, and statistical analyses of curve measurements were compared and evaluated among different reconstruction protocols. The results illustrated that the 3-D spine images using the FDP-MPI4 algorithm showed higher brightness (20%), contrast (14%), and signal-to-noise ratio (SNR) (26%) than FDP-VNN. The measurement performed by trainee rater exhibited significant improvement in measurement reliability and accuracy using FDP-MPI4 in comparison with FDP-VNN ( ${p} ), and the intraclass correlation coefficient (ICC) of interrater measurement increased from 0.88 to 0.96. The FDP-PNN method could acquire and reconstruct spine images simultaneously and present the results in 1–2 min, which showed the potential to provide the approximate real-time visualization for fast screening.

BACKGROUND Heterotopic cesarean scar pregnancy (HCSP) is very rare and has a high risk of massive uterine bleeding. Preservation of concurrent intrauterine pregnancy (IUP) is one of the great challenges in the management of HCSP. No universal treatment protocol has been established when IUP is desired to be preserved. CASE SUMMARY We report a case of HCSP at 8+ wk gestation in a 34-year-old woman with stable hemodynamics. A two-step intervention was applied. Selective embryo aspiration was performed first, and surgical removal of ectopic gestational tissue by suction and curettage was performed 2 d later. Both steps were performed under ultrasound guidance. The patient had an uneventful course, and a healthy baby was delivered at 34+6 wk gestation. CONCLUSION Selective embryo aspiration followed by suction and curettage was successful in the preservation of IUP in the management of HCSP. This approach is an alternative option for HCSP in the first trimester when the IUP is desired to be preserved.

The instrumental variables for insomnia were derived from the largest genome-wide association study of 1,331,010 participants, while the genetic instruments for migraine were available from the largest meta-analysis of migraine with 59,674 cases and 316,078 controls.Results: In case–control study, subjects with insomnia have significantly higher risk of migraine (OR=4.29, 95% CI: 3.21– 5.74, P< 0.001), compared with those without insomnia. The bidirectional two-sample MR analysis revealed that insomnia was significantly associated with higher risk of migraine (OR=1.24, 95% CI: 1.11– 1.38, P=1.01× 10-4), and the results were validated in the UK Biobank data. The results showed no indication for directional pleiotropy effects as assessed by the MR-Egger intercept (P> 0.05).Conclusion: Conclusively, our study highlighted that increased migraine risk was confined to subjects with a genetic pre-disposition to insomnia, and these findings had potential implications for improving the sleep quality to reduce the burden of migraine.Keywords: mendelian randomization, insomnia, migraine, case–control

What is already known about this topic? Attitudes of disapproval toward public health measures led to behaviors that could increase vulnerability to contracting coronavirus disease 2019 (COVID-19). What is added by this report? Chinese citizens rated the necessity of mitigation measures for combating COVID-19 higher than did Americans (4.81 vs. 4.03 on a 1–5 scale). The China–US difference was attributable to 1) Chinese citizens having more confidence in scientists than Americans and 2) Chinese citizens almost invariably accepting the necessity of COVID-19 mitigation measures, regardless of their confidence in scientists. What are the implications for public health practice? Building public support for population health measures and public trust in science is crucial for handling epidemic crises.

Highly efficient and stable blue organic lighting-emitting diodes (OLEDs) continue to be a challenge. The substitution strategy has been proven to be one of the most effective chemical approaches to improve the electroluminescence (EL) performance and the operational stability of blue OLEDs based on thermally activated delay fluorescent (TADF) emitters. Herein, the effects of tert-butyl and 9-phenyl-9-fluorenyl substituents on the physicochemical properties and EL performance are systematically studied for aromatic-imide-based TADF molecules. The robust 9-phenyl-9-fluorenyl units outperform the tert-butyl groups in improving the thermal and electrochemical stability. Consequently, sky blue OLEDs based on 4-DPFCzAIAd containing 9-phenyl-9-fluorenyl substituents exhibit excellent comprehensive EL performance with a maximum external quantum efficiency of 28.2% and a long LT50 (time to 50% of initial luminance) of 178 h at an initial luminance of 500 cd m−2, which is much better than that of the device with a tert-butyl substituted emitter. These results demonstrate that the 9-phenyl-9-fluorenyl substituent is superior to the tert-butyl group in constructing TADF emitters for efficient and stable blue OLEDs.

In this retrospective study we assessed the efficacy and safety of tocilizumab in patients with critical or severe coronavirus disease 2019 (COVID-19). We enrolled 181 patients admitted to Huoshenshan Hospital (Wuhan, China) with confirmed COVID-19 between January 2020 and February 2020. Ninety-two patients were treated with tocilizumab, and 89 patients were treated conventionally. We analyzed the clinical manifestations, changes in CT scan images, and laboratory tests before and after tocilizumab treatment, and compared these results with the conventionally treated group. A significant reduction in the level of C-reactive protein was observed 1 week after tocilizumab administration. In some cases this meant the end of the IL-6-related cytokine storm. In addition, tocilizumab relieved fever, cough, and shortness of breath with no reported adverse drug reactions. These findings suggest tocilizumab improves clinical outcomes and is effective for treatment of patients with critical or severe COVID-19. However, future clinical trials are needed to better understand the impact of tocilizumab interference with IL-6 and provide a therapeutic strategy for treatment of COVID-19.

Prior studies of assortative mating have shown that people tend to marry someone of the same educational level, but why individuals value a mate’s education and the process of mate selection itself remain a black box in predominantly quantitative studies. With online dating’s growing popularity, research needs to examine how online daters navigate dating markets given educational preferences they hold and “freedom of choice” offered by technologies. This study aims to investigate individuals’ educational preferences and how educational preferences shape mate selection processes in online dating. In-depth interviews were conducted with 29 university-educated, heterosexual online daters (13 men, 16 women) in Shanghai. Data were analyzed through a combination of abductive and inductive coding strategies. Results showed that both educational levels and university prestige were primary mate selection criteria in online dating. Both genders considered educational sorting essential for achieving cultural matching, but only men emphasized the importance of spouse’s education for their future children’s education. Furthermore, guided by their educational preferences, online daters deliberately chose dating platforms and screened dating candidates. We argue that online daters’ emphasis on university prestige is rooted in China’s hierarchical higher education system, and gendered rationales for educational preferences stem from ingrained gender roles in Chinese families. Seemingly “personal” preferences are therefore shaped by cultural norms and institutional contexts. Moreover, results suggest that online dating may reinforce social closure among China’s educational elites.

Background This study aimed to assess the independent prognostic value of tumor size compared with other clinical and pathologic features of primary invasive cutaneous melanoma (CM). Methods This study included 28,593 patients with primary invasive CM in Surveillance, Epidemiology, and End Results Program database diagnosed from 2004 through 2016. Tumor size was divided into five subgroups (≤6, 7‐12, 13‐30, 31‐42, and >42 mm). The primary endpoint was melanoma‐specific survival (MSS). Results The relationship between tumor size and survival was piecewise. After adjusting for age, sex, primary site, histopathologic cell type, Breslow thickness, ulceration, mitotic rate, regional metastasis, and distant metastasis, the hazard ratio (HR) of MSS increased with increasing tumor size until a peak at 31‐42 mm (HRs, 1.33, 1.59, 2.41, respectively; all P 42 mm subgroup had the shortest MSS in T4. In addition, tumor size with a cutoff value of 12 mm showed stronger prognostic value for MSS (HR, 2.32; 95% CI, 1.80‐2.98; P, Tumor size was an important independent prognostic factor for MSS in patients with primary invasive CM. Tumor size larger than 30 mm would provide additional important prognostic information in each T subcategory of localized CM.Tumor size with a cutoff value of 12 mm has great potential in improving the accuracy of melanoma T1 substaging.

Nano-modified bitumen binders can be tailored to exhibit excellent properties for potential application in pavement engineering. However, a homogeneous dispersion of nano-materials in bitumen is still a challenge, which obscures the maximum property improvement of bitumen that can be achieved. In this study, a CNT sponge, which is a macroscopic assembly of nanotubes and represents one of the most promising lightweight carbon nanomaterials with homogeneous three-dimensional network and a high porosity, was used to reinforce bitumen. Bitumen can autonomously penetrate into the porous CNT sponge, and fill up all the pores, forming a bitumen/CNT nanocomposite with homogeneous CNT dispersion quality. We found that, the addition of CNTs significantly improves the performance of bitumen at elevated temperatures, as indicated by the much improved properties of stiffness, rutting resistance, creep and recovery behavior as well as high-temperature stability. Such effective modification of bitumen by CNT are ascribed to the highly uniform dispersion of CNTs, as well as the percolation structure formed throughout bitumen matrix. This study highlights the importance of CNT dispersion quality, and sheds light on the bitumen property improvement that could be achieved.

Context: Oxidative imbalance-induced cognitive impairment is among the most urgent clinical concerns. Isoflurane has been demonstrated to impair cognitive function via an increase in oxidative stress. GSP has strong antioxidant capacities, suggesting potential cognitive benefits. Objective: This study investigates whether GSP pre-treatment can alleviate isoflurane-induced cognitive dysfunction in mice. Materials and methods: C57BL/6J mice were pre-treated with either GSP 25–100 mg/kg/d for seven days or GSP 100–400 mg/kg as a single dose before the 6 h isoflurane anaesthesia. Cognitive functioning was examined using the fear conditioning tests. The levels of SOD, p-NR2B and p-CREB in the hippocampus were also analysed. Results: Pre-treatment with either a dose of GSP 50 mg/kg/d for seven days or a single dose of GSP 200 mg/kg significantly increased the % freezing time in contextual tests on the 1st (72.18 ± 12.39% vs. 37.60 ± 8.93%; 78.27 ± 8.46% vs. 52.72 ± 2.64%), 3rd (93.80 ± 7.62% vs. 52.94 ± 14.10%; 87.65 ± 10.86% vs. 52.89 ± 1.73%) and 7th (91.36 ± 5.31% vs. 64.09 ± 14.46%; 93.78 ± 3.92% vs. 79.17 ± 1.79%) day after anaesthesia. In the hippocampus of mice exposed to isoflurane, GSP 200 mg/kg increased the total SOD activity on the 1st and 3rd day and reversed the decreased activity of the NR2B/CREB pathway. Discussion and conclusions: These findings suggest that GSP improves isoflurane-induced cognitive dysfunction by protecting against perturbing antioxidant enzyme activities and NR2B/CREB pathway. Therefore, GSP may possess a potential prophylactic role in isoflurane-induced and other oxidative stress-related cognitive decline.

Accumulating evidence indicates that inhalation anesthetics induce or increase the risk of cognitive impairment. GLYX-13 (rapastinel) acts on the glycine site of N-methyl-D-aspartate receptors (NMDARs) and has been shown to enhance hippocampus-dependent learning and memory function. However, the mechanisms by which GLYX-13 affects learning and memory function are still unclear. In this study, we investigated these mechanisms in a mouse model of long-term anesthesia exposure. Mice were intravenously administered 1 mg/kg GLYX-13 at 2 hours before isoflurane exposure (1.5% for 6 hours). Cognitive function was assessed using the contextual fear conditioning test and the novel object recognition test. The mRNA expression and phosphorylated protein levels of NMDAR pathway components, N-methyl-D-aspartate receptor subunit 2B(NR2B)-Ca2+/calmodulin dependent protein kinase II (CaMKII)-cyclic adenosine monophosphate response element binding protein (CREB), in the hippocampus were evaluated by quantitative RT-PCR and western blot assay. Pretreatment with GLYX-13 ameliorated isoflurane exposure-induced cognitive impairment and restored NR2B, CaMKII and CREB mRNA and phosphorylated protein levels. Intracerebroventricular injection of KN93, a selective CaMKII inhibitor, significantly diminished the effect of GLYX-13 on cognitive function and NR2B, CaMKII and CREB levels in the hippocampus. Taken together, our findings suggest that GLYX-13 pretreatment alleviates isoflurane-induced cognitive dysfunction by protecting against perturbation of the NR2B/CaMKII/CREB signaling pathway in the hippocampus. Therefore, GLYX-13 may have therapeutic potential for the treatment of anesthesia-induced cognitive dysfunction. This study was designed to explore correlations between SIRT7 and KIRC characteristics using the TCGA database.Methods: Relationships between SIRT7 expression and KIRC patient clinicopathological characteristics were assessed through Kruskal–Wallis tests, Wilcoxon signed-rank tests, and logistic regression analyses. Area under the ROC curve (AUC) values were used to assess the prognostic value of SIRT7 as a means of classifying clear cell renal cell carcinoma patients. The functional role of SIRT7 in this cancer type was assessed through GO/KEGG enrichment analyses and immune cell infiltration analyses.Results: In KIRC patients, higher levels of SIRT7 expression were associated with Race, M stage, T stage (all P < 0.05). SIRT7 offered significant diagnostic value in ROC curve analyses (AUC = 0.912), and elevated SIRT7 levels were linked to worse patient overall survival (OS; P < 0.001). The expression of SIRT7 was independently related with KIRC patient OS (HR: 1.827; 95% CI: 1.346– 2.481; P< 0.001). In GO/KEGG analyses, SIRT7 was found to be associated with ubiquitin-mediated proteolysis and nucleotide excision repair. Higher SIRT7 expression was related to the enhanced infiltration of certain immune cells.Conclusion: Increased SIRT7 expression was associated with a worse KIRC patient prognosis, and immune infiltrates, suggesting it may offer value as a prognostic biomarker for this cancer type.Keywords: SIRT7, biomarker, KIRC, prognosis, tumor-immune infiltration

Background Owing to the crucial role the endometrium plays in embryo implantation, the four main endometrial preparation protocols have become important factors in the study of pregnancy outcomes in the FET cycles. Previous studies have shown that the best of these four protocols remains controversial for women undergoing FET. Methods A total of 10333 FET cycles from January 2018 to December 2018 were analyzed in this study. They were categorized into four groups according to endometrial preparation regimen: natural cycles (Group NC,n = 815), hormone replacement therapy cycles (Group HRT ,n = 6434), GnRH agonist artificial cycles (Group GAC,n = 1392) and ovarian stimulation cycles (Group OC, n = 1692). All patients were followed up for at least 1 year. Pregnancy outcomes were compared between the four groups and multiple logistic regression models were used to adjust for the effects of confounding factors. Results The ectopic pregnancy rate (P = 0.627) and miscarriage rate (P = 0.164) were not statistically significant between the four groups. Moreover, biochemical pregnancy rate, clinical pregnancy rate and live birth rate in the NC group were not statistically significant compared to the other three groups. After adjusting for covariates, multiple logistic regression analysis showed no statistical significance in pregnancy outcomes in the HRT, GAC and OC groups compared to the NC group.And the adjusted OR for live births was 0.988 (95 % CI0.847-1.152) for the HRT group, 0.955 (95 % CI0.795-1.146) for the GAC group,0.898 (95 % CI0.754-1.070) for the OC group. Conclusions Our study showed that natural cycles have similar pregnancy outcomes in terms of clinical pregnancy and live birth to the other three endometrial preparation options. As it has other advantages, the natural cycle protocol can therefore be the recommended option for endometrial preparation in the FET population.

This paper addresses the problem of generating dense point clouds from given sparse point clouds to model the underlying geometric structures of objects/scenes. To tackle this challenging issue, we propose a novel end-to-end learning-based framework. Specifically, by taking advantage of the linear approximation theorem, we first formulate the problem explicitly, which boils down to determining the interpolation weights and high-order approximation errors. Then, we design a lightweight neural network to adaptively learn unified and sorted interpolation weights as well as the high-order refinements, by analyzing the local geometry of the input point cloud. The proposed method can be interpreted by the explicit formulation, and thus is more memory-efficient than existing ones. In sharp contrast to the existing methods that work only for a pre-defined and fixed upsampling factor, the proposed framework only requires a single neural network with one-time training to handle various upsampling factors within a typical range, which is highly desired in real-world applications. In addition, we propose a simple yet effective training strategy to drive such a flexible ability. In addition, our method can handle non-uniformly distributed and noisy data well. Extensive experiments on both synthetic and real-world data demonstrate the superiority of the proposed method over state-of-the-art methods both quantitatively and qualitatively., 15 pages, 16 figures, 6 tables

General anesthetics can induce cognitive impairments and increase the risk of Alzheimer’s disease (AD). However, the underlying mechanisms are still unknown. Our previous studies shown that long-term isoflurane exposure induced peripheral and central insulin resistance (IR) in adult mice and aggravated IR in type 2 diabetes mellitus (T2DM) mice. Clinical and preclinical studies revealed an association between impaired insulin signaling and tau pathology in AD and other tauopathies. We investigated if alleviation of hippocampal IR by the antidiabetic agent metformin could reduce tau hyperphosphorylation and cognitive decline induced by isoflurane in mice. The effects of prolonged (6 h) isoflurane anesthesia on hippocampal IR, hippocampal tau hyperphosphorylation, and hippocampus-dependent cognitive function were evaluated in wild type (WT) adult mice and the high-fat diet plus streptozotocin (HFD/STZ) mouse model of T2DM. Here we shown that isoflurane and HFD/STZ dramatically and synergistically induced hippocampal IR and fear memory impairment. Metformin pretreatment strongly ameliorated hippocampal IR and cognitive dysfunction caused by isoflurane in WT mice, but was less effective in T2DM mice. Isoflurane also induced hippocampal tau hyperphosphorylation and metformin reversed this effect. In addition, isoflurane significantly increased blood glucose levels in both adult and T2DM mice, and metformin reversed this effect as well. Administration of 25% glucose to metformin-pretreated mice induced hyperglycemia, but surprisingly did not reverse the benefits of metformin on hippocampal insulin signaling and fear memory following isoflurane anesthesia. Our findings show hippocampal IR and tau hyperphosphorylation contribute to acute isoflurane-induced cognitive dysfunction. Brief metformin treatment can mitigate these effects through a mechanism independent of glycemic control. Future studies are needed to investigate whether long-term metformin treatment can also prevent T2DM-induced hippocampal IR and cognitive decline.

Mas-related G-protein-coupled receptor subtype C (MrgC) has been shown to play an important role in the development of bone cancer pain. Ubiquitination is reported to participate in pain. However, whether MrgC ubiquitination plays a role in bone cancer pain remains unclear. To answer this question, we designed and performed this study. Osteosarcoma cells were implanted into the intramedullary space of the right femurs of C3H/HeJ mice to induce progressive bone cancer pain. MrgC agonist bovine adrenal medulla 8-22 (BAM 8-22) or MrgC antagonist anti-MrgC antibody were injected intrathecally on day 14 after bone cancer pain was successfully induced. The pain behaviors, the MrgC ubiquitination levels and intracellular calcium concentration in spinal neurons were measured before and after injection, respectively. With comparison to normal and sham group, mice in tumor group exhibited serious bone cancer pain on day 14, and the level of MrgC ubiquitination and intracellular calcium concentration in spinal neurons was significantly higher. Intrathecal injection of BAM 8-22 significantly alleviated bone cancer pain, increased the MrgC ubiquitination level and decreased intracellular calcium concentration in spinal neurons; however, these effects were reversed by administration of anti-MrgC antibody. Our study reveals that MrgC ubiquitination participates in the production and maintenance of bone cancer pain in mice, possibly through the regulation of intracellular calcium concentration in mice spinal neurons.

Keeping notes can be seen as the process of the extraction and re-creation of fine-grained knowledges, i.e., knowledge innovation. It is an important learning method for MOOC learners, while there are little efforts in favoring their note-taking behavior upon watching course videos. In this paper, we summarized the limitations of existing note services provided by popular MOOC platforms and proposed a prototype of a cloud-based dynamic video note service for diverse MOOC Platforms. The new service utilized the idea of bulletin comments for personalized real-time note rendering upon MOOC videos, based its architecture on cloud microservices for ubiquitous video note management, lied the basis on the knowledge collaboration theory for socialized knowledge innovation, and performed big-data analysis for learning process evaluation. Evaluations based on running instances and a questionnaire survey proved the effectiveness of the protype.

With the development of artificial intelligence and big data technology, people are paying more and more attention to its application in eco-tourism marketing. To make better use of big data for eco-tourism marketing, and analyze and meet the psychological needs of tourists, this article uses some artificial intelligence algorithms, including analytic hierarchy process and fuzzy comprehensive evaluation model, to evaluate rural eco-tourism options and put forward suggestions for constructing firefly eco-tourism.

Background: Although renal denervation (RDN) may theoretically be effective on type 2 diabetes mellitus (T2DM), most clinical data failed to demonstrate such a concept. We investigated the safety and primary effects of catheter-based endovascular denervation (EDN) at new sites of the celiac artery (CA), and the abdominal aorta around CA using a novel multi-electrode device on glycemic control in T2DM patients. Methods: With a novel six-electrode catheter system, EDN was conducted in T2DM patients at new sites of CA and aorta between CA and superior mesenteric artery (SMA) in purpose of targeting on the postganglionic efferent nerves innervating the liver, islet, and other metabolic organs. The major inclusion criteria were T2DM patients diagnosed over 5 years, with hemoglobin A1c (HbA1c) over 7.5%. The primary outcomes were evaluated by the safety, HbA1c, homeostasis-model assessment of insulin resistance (HOMA-IR), fasting plasma glucose (FPG), 2-hour postprandial plasma glucose (2hPG), and oral glucose tolerance test (OGTT) based 3-hour C-peptide test. The antidiabetic medication, lifestyle, physical condition, blood pressure (BP), plasma norepinephrine, angiotensin II, liver biochemistry, and plasma lipids were also recorded.Results: A total of 11 patients were included for analysis. The technical success was 100% and no severe treatment-related adverse events or major complications were observed. Both HbA1c and HOMA-IR significantly reduced at 6 months (9.9 vs. 8.0, P = 0.005; 13.3 vs. 6.0, P = 0.016, respectively). Decreases of FPG and 2hPG were observed (15.4 vs.10.1 mmol/L, P < 0.001; 17.9 vs. 11.4 mmol/L, P = 0.001, respectively). OGTT based C-peptide test indicated improved beta-cell function ([AUC] 0.23 vs. 0.28 pmol/mL, P =0.046). A reduction of daily insulin injection (P = 0.02) and improvement of liver function (ALT, P = 0.014; GGT, P = 0.021) were also observed without changes in lifestyle, physical conditions, and other metabolic measurements.Conclusions: The 6-month analysis of this trial showed that EDN at new sites of CA and the abdominal aorta around celiac artery using the novel six-electrode catheter system elicits a clinically significant improvement in glycemic control and insulin resistance in patients with T2DM, with good tolerability. S-1-based and capecitabine-based adjuvant treatments are proved efficacious for patients with gastric cancer, but conventional meta-analyses of the direct comparisons between two alternative adjuvant regimens to resection of GC have not been attempted. The aim of this review was to compare the disease-free survival, overall survival and adverse events in patients receiving the S-1- and capecitabine-based adjuvant chemotherapies for treatment of gastric cancer (GC) patients undergoing resection. A search of the academic literature was performed in PUBMED, SCOPUS, CENTRAL and EMBASE databases along with manual search in relevant journals for studies in English, to identify comparative studies comparing the effect of S-1-based chemotherapy and capecitabine-based adjuvant chemotherapy (AC), used in combination with surgical resection for treatment of gastric cancer. Both qualitative and quantitative analyses was carried out for all the included studies. The hazard ratios (HR) of disease-free survival (DFS) and overall survival (OS) were pooled using generic inverse variance method. The included studies were assessed for risk of bias using ROBINS-E (risk of bias in non-randomized studies of exposures) tool. Seven retrospective cohort studies, two prospective cohort studies and one randomized clinical trial were included. Both S-1- and capecitabine-based adjuvant chemotherapy for treatment of stage 2 or 3 gastric cancer had similar effects on the 3-year and 5-year DFS rates, overall survival and adverse events in the included studies. There was no difference in the adjusted hazard ratios (HR) of OS and DFS (0.86 95% CI (0.68, 1.09); p = 0.21 and 0.96 95% CI (0.75, 1.24), respectively). Oral mucositis was increasingly associated with S-1-based AC, while incidences of adverse events such as neutropenia, anaemia and thrombocytopenia were similar to those of capecitabine-based regimen. The quality of the included studies was found to be low to moderate. S-1- and capecitabine-based adjuvant chemotherapies can be used interchangeably as an adjuvant chemotherapeutic regimen postradical gastrectomy with D2 lymph node dissection.

This study constructs the hierarchical theoretical model of image design to realize more accurate aesthetic feature identification for females. The study mainly takes the scale, material and hollowed-out technique of Suzhou Sandalwood Fans as the carrier for the perception of products’ aesthetic image, then retrieves words of female perceptual image with factor analysis in SPSS and other computer-aided information analysis methods, and evaluates the weights of different aesthetic image factors to screen out case models with higher adaptability, thus improving the market acceptance of subsequent design schemes. Based on relevant Suzhou cultural elements, it carries out the detailed optimization design practice to verify the feasibility of this method.

Motivated by the intuition that one can transform two aligned point clouds to each other more easily and meaningfully than a misaligned pair, we propose CorrNet3D -- the first unsupervised and end-to-end deep learning-based framework -- to drive the learning of dense correspondence between 3D shapes by means of deformation-like reconstruction to overcome the need for annotated data. Specifically, CorrNet3D consists of a deep feature embedding module and two novel modules called correspondence indicator and symmetric deformer. Feeding a pair of raw point clouds, our model first learns the pointwise features and passes them into the indicator to generate a learnable correspondence matrix used to permute the input pair. The symmetric deformer, with an additional regularized loss, transforms the two permuted point clouds to each other to drive the unsupervised learning of the correspondence. The extensive experiments on both synthetic and real-world datasets of rigid and non-rigid 3D shapes show our CorrNet3D outperforms state-of-the-art methods to a large extent, including those taking meshes as input. CorrNet3D is a flexible framework in that it can be easily adapted to supervised learning if annotated data are available. Going beyond a focus on individual‐level employment outcomes, we investigate couples' changing work patterns in the United Kingdom (UK) and the United States (US) during the COVID‐19 pandemic. Analyzing longitudinal panels of 2186 couples from the Understanding Society COVID‐19 Survey (UK) and 2718 couples from the Current Population Survey (US), we assess whether the pandemic has elevated the importance of human capital vis‐à‐vis traditional gender specialization in shaping couples' work patterns. The UK witnessed a notable increase in sole‐worker families with the better‐educated partner working, irrespective of gender. The impact of the pandemic was similar but weaker in the US. In both countries, couples at the bottom 25% of the prepandemic family income distribution experienced the greatest increase in neither partner working but the least growth in sole‐worker arrangements. Through a couple‐level analysis of changing employment patterns, this study highlights the importance of human capital in shaping couples' paid‐work organization during the pandemic, and it reveals the socioeconomic gradient in such organization.

Postoperative cognitive dysfunction (POCD) is a common clinical phenomenon characterized by cognitive deficits in patients after anesthesia and surgery. Advanced age is a significant independent risk factor for POCD. We previously reported that in young mice, sleep-wake rhythm is involved in the isoflurane-induced memory impairment. In present study, we sought to determine whether advanced age increased the risk of POCD through aggravated and prolonged post-anesthetic circadian disruption in the elderly. We constructed POCD model by submitting the mice to 5-h 1.3% isoflurane anesthesia from Zeitgeber Time (ZT) 14 to ZT19. Under novel object recognition assay (NOR) and Morris water maze (MWM) test, We found 5-h isoflurane anesthesia impaired the cognition of young mice for early 3 days after anesthesia but damaged the aged for at least 1 week. With Mini-Mitter continuously monitoring, a 3.22 ± 0.75 h gross motor activity acrophase delay was manifested in young mice on D1, while in the aged mice, the gross motor activity phase shift lasted for 3 days, consistent with the body temperature rhythm trends of change. Melatonin has been considered as an effective remedy for circadian rhythm shift. In aged mice, melatonin was pretreated intragastrically at the dose of 10 mg/kg daily for 7 consecutive days before anesthesia. We found that melatonin prevented isoflurane-induced cognitive impairments by restoring the locomotor activity and temperature circadian rhythm via clock gene resynchronization. Overall, these results indicated that Long-term isoflurane anesthesia induced more aggravated and prolonged memory deficits and circadian rhythms disruption in aged mice. Melatonin could prevent isoflurane-induced cognitive impairments by circadian rhythm resynchronization.

Transcranial electric motor evoked potentials (TCeMEPs) play an important role in reducing the risk of iatrogenic paraplegia. TCeMEPs could be obviously suppressed by neuromuscular blockade (NMB). The aims of this study were to examine the effects of NMB on TCeMEPs and to determine an appropriate level of partial neuromuscular blockade (pNMB) for TCeMEPs during surgical correction of idiopathic scoliosis under total intravenous anesthesia (TIVA). All patients were maintained with TIVA. The pNMB levels were classified into five phases: one or two train-of-four (TOF) counts (TOF1); three TOF counts, or T4/T1 (TOFR, T1,4, first or four twitch height of TOF) ≤ 15% (TOF2); TOFR at 16–25% (TOF3); TOFR at 26–50% (TOF4); and TOFR at 51–75% (TOF5). No neuromuscular blockade (nNMB) was achieved when TOFR was more than 75%. The absolute and relative latency, amplitude and area under curve (AUC), efficacy of TCeMEPs and rate of unexpected movement were compared among these phases. Neither the amplitude and AUC nor the efficacy of TCeMEPs were affected at TOF4−5 of abductor halluces muscles TCeMEPs (AH-TCeMEPs) or at TOF3−5 of tibialis anterior muscles TCeMEPs (TA-TCeMEPs) compared with nNMB. However, the rate of unexpected movement was increased significantly at TOF5 and nNMB compared with TOF1 and TOF4. The application of pNMB with TOFR aimed at 26–50% for AH-TCeMEPs or 16–50% for TA-TCeMEPs seems to be an appropriate regimen for TCeMEPs during surgical correction for idiopathic scoliosis under TIVA.

With China’s rise in the global economy, more couples participate in financial investing. Using the 2011 China Household Finance Survey, we examined factors influencing stock and fixed-income investments in the cities. Couples with urban residency were more likely to invest than couples without urban residency. Compared to traditional couples with highly-educated husbands only, couples with only highly-educated wives invested similarly, whereas couples with two highly- (less-) educated spouses were more (less) likely to invest. Further, we investigated how these relationships were mediated by household income and wealth, financial literacy, information acquisition, and risk tolerance. Overall, our findings suggest that household investing is shaped by both family structure (i.e., spouses’ educational pairing) and institutional advantage (indicated by urban residency).

Background The association between intraoperative hypotension (IOH) and postoperative outcomes is not fully understood. We performed a meta-analysis to determine whether IOH is associated with increased risk of 30-day mortality, major adverse cardiac events (MACEs) and acute kidney injury (AKI) after non-cardiac surgery. Methods We searched PubMed and Embase through May 2016 to identify cohort studies that investigated the association between IOH and risk of 30-day mortality, MACEs, or AKI in adult patients after non-cardiac surgery. Ascertainment of IOH and assessment of outcomes were defined by the individual study. Considering the level of clinical heterogeneity, adjusted odds ratios (ORs) with 95% confidence interval (CIs) were pooled using a random-effects model. This meta-analysis is registered on PROSPERO (CRD42016049405). Results We included 14 cohort studies that were heterogeneous in terms of definition of IOH. IOH alone was associated with increased risk of 30-day mortality (OR 1.29 [95% CI, 1.19–1.41]), MACEs (OR 1.59 [95% CI, 1.23–2.05]), especially myocardial injury (OR 1.67 [95% CI, 1.31–2.13]), and AKI (OR 1.39 [95% CI, 1.09–1.77]). Triple low (IOH coincident with low bispectral index and low minimum alveolar concentration) also predicts increased risk of 30-day mortality (OR 1.32 [95% CI, 1.03–1.68]). Conclusions IOH alone significantly increases the risk of postoperative 30-day mortality, MACEs, especially myocardial injury, and AKI in adult patients after non-cardiac surgery. Triple low also predicts increased risk of 30-day mortality after non-cardiac surgery. These findings provide evidence that IOH should be recognized as an independent risk factor for postoperative adverse outcomes after non-cardiac surgery.

Core cerebrospinal fluid (CSF) biomarkers (Aβ42, T-tau, P-tau) were included as supporting diagnostic criteria for Alzheimer's Disease (AD), but they lack the power to predict AD progression. On the other hand, a new biomarker CSF Neurogranin (Ng) has been shown to predict cognitive decline. This systematic review aims to synthesise the prognostic utility of CSF Ng in predicting cognitive decline in the AD continuum. Seven databases were searched systematically from inception to 30 September 2020. Participants were 55 years or older, who had baseline and at least one follow-up cognitive assessments. Risk of bias was assessed using the Quality in Prognosis Studies tool. Meta-analysis was conducted by pooling standardised beta coefficients and adjusted hazard ratios. Thirteen studies were included and high-quality evidence suggests that CSF Ng predicts Mini-Mental State Examination (MMSE) decline in Aβ+ mild cognitive impairment (MCI). Moderate quality evidence showed that CSF Ng could predict the decline of memory and executive function in MCI. Narrative synthesis found that CSF Ng/Aβ42 was also likely to predict cognitive decline. More studies are required to validate the use of CSF Ng as an AD prognostic marker and its application in future development of drug treatment and diagnosis.

PURPOSE To investigate the safety and efficacy of catheter-based endovascular denervation (EDN) at the celiac artery and abdominal aorta around the celiac artery on glycemic control in patients with type 2 diabetes mellitus (T2DM). MATERIALS AND METHODS With a novel catheter system, EDN was conducted at the celiac artery along with the abdominal aorta around the celiac artery in patients with T2DM whose glycosylated hemoglobin (HbA1c) level was >7.5%. The primary outcome was HbA1c level at 6 months. Other outcomes included safety, oral glucose tolerance test, homeostasis model assessment of insulin resistance (HOMA-IR), fasting plasma glucose (FPG) level, 2-hour postprandial plasma glucose (2hPG) level, and C-peptide test. RESULTS A total of 11 subjects were included for analysis. The technical success was 100%, and no severe treatment-related adverse events or major complications were observed. Both HbA1c level and HOMA-IR were significantly reduced at 6 months (9.9% vs 8.0%, P = .005; 13.3 vs 6.0, P = .016). Decreases in FPG and 2hPG levels were observed (227.2 vs 181.8 mg/dL, P < .001; 322.2 vs 205.2 mg/dL, P = .001). The C-peptide test indicated improved β-cell function (area under the curve, 0.23 vs 0.28 pmol/mL, P = .046). A reduction of daily insulin injection (P = .02) and improvement of liver function (alanine aminotransferase, P = .014; γ-glutamyl transpeptidase, P = .021) were also observed. CONCLUSIONS EDN in the celiac artery and abdominal aorta around the celiac artery elicited a clinically significant improvement in glycemic control and insulin resistance in patients with T2DM, with good tolerability as demonstrated by 6-month follow-up.

The buckling behavior of rod strings in wellbore is one of the most phenomena in petroleum engineering. The slender rod strings in wellbore were selected as research objects. Based on the energy method, the critical load of sinusoidal and helical buckling were derived for the string with the wellbore pressure at the bottom. According to the sinusoidal and helical buckling's geometry of the pressurized string, the contact friction state of post-buckling between rod strings and the wellbore was considered. In this paper, Lagrange multiplier method was adopted to describe the sliding displacement boundary conditions of wellbore and the gravitational potential energy and friction resistance dissipated energy were introduced in the energy method. The contact forces and friction resistances between rod string and wellbore were derived in the inclined and vertical wells. And the post buckling experimental apparatus was also developed in this paper. The results indicate that the sinusoidal and helical buckling critical loads and the friction resistances were derived under different loads. The experimental results are consistent with the theoretical results. Therefore, this article would provide an effective method for buckling string, such as drilling, coiled tubing.

Immigrant workers are a growing share of the U.S. labor force and are overrepresented in certain occupations. This much is well documented, yet few studies have examined the consequences of this division of labor between foreign-born and native-born workers. This research focuses on one of the consequences of occupational segregation—worker health. We merge data from the 2004–2014 National Health Interview Surveys with occupational-level data from the Occupational Information Network 20.1 database and the American Community Surveys to examine the relationship between occupational segregation and health. First, logistic regression models show that working in an occupation with a higher share of immigrants is associated with higher odds of poor physical and psychological health. This relationship is more pronounced among native-born workers than among foreign-born workers. Second, we propose two explanations for the association between occupational segregation and health: (1) workers with less human capital are typically sorted into culturally devalued occupations with a higher concentration of immigrants, and (2) occupations with a higher percentage of immigrants generally have relatively poor work environments. We find sorting variables play a major role, whereas the smaller contribution of occupational environments to the segregation-health link is partly because of the heterogeneous (i.e., both positive and negative) indirect effects of different exposure measures. With the sustained high levels of immigration to the United States, the implications of integrated or segregated experiences in the labor market and their impact on workers are important avenues for health policies and future research.

Whether it be for countries to improve the ability to undertake independent innovation or for enterprises to enhance their international competitiveness, tracing historical progression and forecasting future trends of technology evolution is essential for formulating technology strategies and policies. In this paper, we apply co-classification analysis to reveal the technical evolution process of a certain technical field, use co-word analysis to extract implicit or unknown patterns and topics, and employ main path analysis to discover significant clues about technology hotspots and development prospects. We illustrate this hybrid approach with 3D printing, referring to various technologies and processes used to synthesize a three-dimensional object. Results show how our method offers technical insights and traces technology evolution pathways, and then helps decision-makers guide technology development.

Background Chemotherapy toxicity is a serious problem from which non-small cell lung cancer (NSCLC) patients suffer. The mismatch repair (MMR) system is associated with platinum-based chemotherapy toxicity in NSCLC patients. In this study, we aimed to investigate the relationship between genetic polymorphisms in the MMR pathway and platinum-based chemotherapy toxicity in NSCLC patients. Methods A total of 220 Chinese lung cancer patients who received at least two cycles of platinum-based chemotherapy were recruited for this study. Toxicity was evaluated in each patient after two cycles of chemotherapy. A total of 44 single nucleotide polymorphisms were selected to investigate their associations with platinum-based chemotherapy toxicity. Results MutS homolog 2 (MSH2) rs6544991 [odds ratio (OR) 2.98, 95% confidence interval (CI) 1.20–7.40, P = 0.019] was associated with gastrointestinal toxicity in the dominant model; MSH3 rs6151627 (OR 2.38, 95% CI 1.23–4.60, P = 0.010), rs6151670 (OR 2.05, 95% CI 1.07–3.93, P = 0.031), and rs7709909 (OR 2.38, 95% CI 1.23–4.64, P = 0.010) were associated with hematologic toxicity in the dominant model. Additionally, MSH5 rs805304 was significantly associated with overall toxicity (OR 2.21, 95% CI 1.19–4.09, P = 0.012), and MSH5 rs707939 was significantly associated with both overall toxicity (OR 0.42, 95% CI 0.23–0.76, P = 0.004) and gastrointestinal toxicity (OR 0.44, 95% CI 0.20–0.96, P = 0.038) in the dominant model. Conclusion Genetic polymorphisms in the MMR pathway are potential clinical markers for predicting chemotherapy toxicity in NSCLC patients. Background: Prostate cancer is one of the most prevalent cancers in men. Exposure to heavy metals and their association with prostate cancer risk has been studied extensively, but combined effects remain largely inconclusive. Objectives: To elucidate the association between serum concentrations of heavy metals and prostate cancer risk. Methods: Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the concentrations of a panel of 10 heavy metals (Mn, Cu, Zn, As, Se, Sb, Co, Cu, Cd and Pb) in serum samples of 141 cases and 114 controls in the Singapore Prostate Cancer Study. Linear probit regression models were used to estimate risk differences (RDs) and 95% confidence intervals (CIs) for the associations between log-centered serum metal concentrations and prostate cancer risk with adjustment for potential confounders. Bayesian kernel machine regression (BKMR) models were used to account for nonlinear, interactive, and joint metal effects. Results: Using probit regression, four heavy metals (As, Zn, Mn, Sb) were significantly and positively associated with prostate cancer risk in the unadjusted models. Using BKMR analysis, both As and Zn had positive risk differences on prostate cancer risk when all other metals were held fixed at the 25th and 50th percentiles (RD, 25th percentile: As: 0.15, Zn: 0.19, RD, 50th percentile: As: 0.45, Zn: 0.37). In addition, the overall mixture risk difference was positive and the 95% credible intervals did not include 0 when all metals in the mixture were jointly above their 55th percentile, as compared to when all metals were below their median values. Conclusions: In summary, we found positive associations between the serum levels of As and Zn and prostate cancer risk on the risk difference scale using BKMR models. The overall mixture effect was also associated with increased prostate cancer risk. Future studies are warranted to validate these findings in prospective studies. Exposure to fine particulate matter (PM2.5) is associated with cardiovascular disease risk. To date, there are few studies on short-term PM2.5 exposure in different microenvironments and its impact on immediate health effects, particularly in the Southeast Asia region. This study assessed PM2.5 concentrations in different microenvironments in a densely populated city in the tropics using low-cost personal PM2.5 sensors as well as their associations with acute cardiovascular health outcomes. A total of 49 adult participants affiliated with the National University of Singapore (NUS) community were recruited. Personal low-cost sensors were used to measure PM2.5 concentrations between September 2017 and March 2019. Demographic information and time-activity patterns were collected using questionnaires. Wilcoxon pairwise comparisons were used to determine statistical differences between PM2.5 exposures at 18 different microenvironments. Generalized Estimating Equations (GEE) models were used to assess the association between PM2.5 exposure and blood pressure as well as heart rate. All models were adjusted for age, sex, body mass index, physical activity, temperature, duration of exposure, and baseline cardiovascular parameters. Significant differences in PM2.5 concentrations were observed across different microenvironments. Air-conditioned offices and tertiary teaching spaces had the lowest (median = 13.1 μg/m3) and hawker centres had the highest (median = 32.0 μg/m3) PM2.5 concentrations. Significant positive associations between PM2.5 exposure and heart rate (β = 0.40, p = 4.6 × 10−5) as well as diastolic blood pressure (β = 0.16, p = 0.0077) were also observed. Short-term exposure to PM2.5 was significantly associated with higher heart rate and blood pressure. Further work is needed to investigate the variations within each type of microenvironment and expand the study to other sub-populations such as the elderly and children.

Wrinkles, as a form of mechanical instability, can be employed in many different areas, including self-cleaning coating, flexible electronics, smart adhesion, channel for microfluidic devices, and mechanical property measurements. After two decades of research, the wavelength and amplitude of wrinkles can be well controlled by proper design of physical properties of thin films and substrates. However, it is still very difficult to change their orientations and complexity in a single piece of thin film, which is critically important for many practical applications. In previous studies, the orientation of all the wrinkles are either along the same direction or can only be controlled to some extent by the patterning of relief structures which rely on the complicated lithography techniques and cannot be adjusted for a given substrate. Here, we employed the shape memory polymer (SMP) as a smart substrate, combined with local/selective heating to control the boundary conditions for the strain field, and realized the gradual change in the orientation of wrinkles in a single piece of thin film. The wrinkles with a gradual change in orientations exhibited angle-dependent colors. Furthermore, by changing the sequence of thin film coating and partial triggering of an SMP, complex surface features with a sharp interface can be obtained. Finite element simulation investigations uncover the basic principles and requirements that need to be satisfied, as well as the limitations of our method, to generate wrinkles with controlled orientations in a single piece of thin film.

Aiming at the technical challenge of spinning forming used for ultra long tubular parts, a new spinning technology based on floating die and synchronous rotation of both ends of the spinning blank was put forward. That is die ring spinning, which is divided into die ring forward spinning and die ring backward spinning. The die ring spinning utilizes to improve the stress condition of deformed metal parts and enhances the fluidity of crystal phase structure along the slip surface. It can not only eliminate the phenomenon of material bulging before spinning wheels, but also improve the straightness and reduce the ellipticity of pipes at the same time.

Numerous clinical investigations have revealed the circadian rhythm changes in the perception of chronic pain, and most clinical chronic pain types peak in the night. However, it is still undiscovered whether circadian rhythm of pain exists in rodents and the specific mechanism that may underlie it. Our study was conducted to investigate the rhythmic changes of hyperalgesia behavior in a chronic constrictive injury (CCI) model of rodents and to explore the role of the N-methyl-d-aspartate receptor 2B (NR2B)-cAMP response element binding protein (CREB)-CREB-regulated transcription coactivator 1 (CRTC1) signaling pathway in this pain rhythm.A CCI operation was performed to mimic clinical chronic pain. Paw mechanical withdrawal threshold and paw withdrawal thermal latency were used to test pain behavior in rats; a von Frey cilia test was used to test mechanical hyperalgesia in mice at Zeitgeber time (ZT) 4, ZT10, ZT16, and ZT22 for 14 contiguous days. The relative mRNA and protein expression of NR2B, CREB and CRTC1 in the suprachiasmatic nuclei and the dorsal horn were measured by real-time polymerase chain reaction and Western blot. CRTC1 and CREB interference adenovirus vectors were injected intrathecally at 2 time points, respectively (ZT12 and ZT0), to further explore the proper time point for pain treatment.During the period of chronic pain state, the pain behavior of CCI rodents showed a circadian rhythm with the peak at ZT4 or ZT10 daily. The pain thresholds were significantly different between the activity period and the rest period. The expressions of NR2B, CRTC1, and CREB at the spinal level were consistent with the pain rhythm. The intrathecal treatment with CRTC1 or CREB interference adenovirus from day 7 to day 9 after CCI surgery markedly improved pain behaviors. Nevertheless, when given at ZT0, they were both more effective at relieving peak pain than drugs given at ZT12.Pain behavior in the chronic pain of CCI displayed circadian rhythm and was associated with circadian secretion of pain-related receptors. The NR2B-CREB-CRTC1 signaling pathway may play a crucial role in this rhythm. Moreover, our results suggest that measures to relieve pain should be taken before pain reaches its peak.

The COVID-19 pandemic has transformed every facet of society. In addition to directly affecting population health, the economic impact of this social shock has begun to be palpable at the individual level. Situated in this context, this research note draws on data collected from Mainland China in March–April, 2020 to examine the individual-level economic toll of the COVID-19 outbreak. We investigate how individuals’ income (when surveyed) has changed relative to before the outbreak, and pay particular attention to the potentially unequal distribution of economic vulnerability based on structural (dis)advantages and COVID-19 related conditions. We show that education, family economic status, Communist Party membership, state-sector employment, and urban hukou—all long-standing status markers in China—mitigate the adverse effects of the COVID-19 outbreak on individuals’ income losses. In addition, people who live in families or regions that were hit harder by COVID-19 are more likely to experience income losses. Taken together, this study shows that the COVID-19 pandemic not only exacerbates pre-existing social inequalities but also creates new forms of disparities. Therefore, instead of acting as a great equalizer, the pandemic may well magnify the social distribution of economic vulnerability. To alleviate inequality and aid post-pandemic recovery, public policies need to be oriented toward vulnerable and marginalized populations.

Purpose/Aim of the study In this study, we sought to observe the effects of Ca2+/calmodulin-dependent protein kinase II (CaMKII) on neuropathic pain and fear memory in a rat model of chronic constriction injury (CCI). Materials and methods Rats were randomly divided into the Sham, Control, CCI and m-AIP groups. In the m-AIP group, an intrathecal injection of m-AIP, the specific antagonist of CaMKII, was given either pretreatment or posttreatment in rats. Mechanical allodynia and thermal hyperalgesia tests were used to test pain behavior, and the passive avoidance test was used to measure fear memory in rats. Results The right side of hippocampus tissues were taken at varying time points. The expression levels of CaMKII-α, pCaMKII-α, CaMKII-β, pCaMKII-β, NR2A, pNR2A, NR2B and pNR2B were detected by Western blot analysis. Significant pain behaviors and impaired cognitive function were shown after CCI surgery, accompanied by the upregulation of proteins in the hippocampus. Pretreatment with m-AIP appeared to provide a temporary improvement in pain and fear memory and decreased the expression of the above proteins in the hippocampus seven days after surgery. Furthermore, postoperative treatment with m-AIP provided relief for pain behavior and protein expression but did not affect fear memory. Conclusions These data suggested that CaMKII played an important role in the crosstalk between neuropathic pain and fear memory, indicating that CaMKII may be a potential therapeutic target for neuropathic pain treatment.

Our previous study identified that it was correlated with platinum response in lung cancer. The current study aims to test the hypothesis that eIF3a may affect the drug response and prognosis of ovarian cancer patients receiving platinum-based chemotherapy by regulating xeroderma pigmentosum complementation group C (XPC) and p27 Kip1 . Immunohistochemistry and western blot was used to determine the expression of eIF3a in 126 human ovarian cancer tissues followed by association analysis of eIF3a expression with patient’s response and survival. Ectopic over-expression and RNA interference knockdown of eIF3a were carried out in A2780/cisplatin (DDP) and its parental A2780 cells, respectively, to determine the effect of altered eIF3a expression on cellular response to cisplatin by employing MTT assay. Western Blot analyses were also carried out to determine the regulation of eIF3a on XPC and p27 Kip1 . eIF3a expression was associated with response of ovarian cancer patients to DDP-based chemotherapy and their survival. Overexpression and knockdown of eIF3a increased and decreased the cellular response to cisplatin in A2780/DDP and A2780 cells, respectively. In addition, XPC and p27 Kip1 were down regulated by eIF3a. eIF3a improves ovarian cancer patients’ response to DDP-based chemotherapy via down regulating XPC and p27 Kip1 .

Eukaryotic translation initiation factor 3 subunit A (eIF3a) plays critical roles in regulating the initiation of protein translation, and eIF3a is highly expressed in lung cancer. In this study, we investigated the association of the positively selected SNPs of eIF3a with the response to and toxicity of platinum-based chemotherapy in Chinese patients with non-small cell lung cancer (NSCLC). SNP data for eIF3a locus were downloaded from HapMap database. For each SNP, haplotype, LD profile and population differentiation were analyzed. The long-range haplotype (LRH) test was employed to identify positively selected SNPs of eIF3a. A total of 325 NSCLC patients were enrolled and genotyped for these SNPs. Five positively selected (rs1409314, rs4752219, rs4752220, rs7091672 and rs10510050) and 5 non-positively selected SNPs (rs10886342, rs11198804, rs2275112, rs10787899 and rs4752269) were identified in the LRH test. However, none of them was correlated with the platinum-based chemotherapy response. In contrast, 4 of the positively selected SNPs (rs1409314, rs4752219, rs4752220 and rs7091672) were significantly correlated with the toxicities tested (neutropenia, anemia, thrombocytopenia, emesis and hepatotoxicity). In addition, rs10510050 was significantly correlated with thrombocytopenia, emesis and hepatotoxicity. None of the 5 non-positively selected SNPs was correlated with the 5 toxicities. The positively selected SNPs of eIF3a are significantly correlated with platinum-based chemotherapy toxicities in Chinese NSCLC patients.

To investigate the influence of ABCB1 polymorphisms on prognostic outcomes in Chinese patients with de novo intermediate-risk acute myeloid leukemia (AML) and to examine the gene expression level in relation to the genetic variation.In total, 263 Chinese intermediate-risk AML patients treated with anthracycline and cytarabine were enrolled. G2677T, C1236T, and C3435T of the ABCB1 gene were analyzed by the allele-specific matrix-assisted laser desorption. Expression of ABCB1 messenger RNA (mRNA) was tested in 101 patients of known genotype and haplotype for ABCB1 polymorphisms. Basic clinical characteristics of these patients were collected from medical records.Survival analysis showed that patients with AML (TTT haplotype) had a longer overall survival (OS) (p 0.001, 29.2 months, 95 % confidence interval [CI], 26.9-31.5 months) and relapse-free survival (RFS) (p = 0.005, 21.8 months, 95 % CI, 19.5-24.0 months) compared with those without TTT haplotype (21.9 months, 95 % CI, 19.6-24.2 months; 16.5 months, 95 % CI, 14.6-18.5 months). After adjusting for age; gender; leukocyte count; hemoglobin level; platelet levels; French, American, and British classification; lactate dehydrogenase levels; Eastern Cooperative Oncology Group performance status; nucleophosmin gene; and fms-related tyrosine kinase 3 gene, the multivariate survival analysis showed that the TTT haplotype appeared to be a predicting factor for OS (p = 0.001, hazard ratio = 1.854, 95 % CI, 1.301-2.641) and RFS (p = 0.009, hazard ratio = 1.755, 95 % CI, 1.153-2.671). Moreover, a significant association between the TTT haplotype and relapse in AML patients was observed in this study (p = 0.002, odds ratio = 0.410, 95 % CI, 0.235-0.715). Gene expression level was significantly lower in patients with the TTT haplotype than in the patients with the other haplotypes (p = 0.004).The findings suggested the TTT haplotype was possibly related to the OS, RFS, and relapse in Chinese patients with AML.
