Recent observations suggest a lower incidence of malignancies in patients infected with HIV during treatment with Highly Active Anti-Retroviral Therapy (HAART) utilizing protease inhibitors. We investigated the effects of ritonavir, a FDA approved HIV protease inhibitor, on proliferation of pancreatic ductal adeno-carcinoma (PDAC) cell lines. Human PDAC cell lines BxPC-3, MIA PaCa-2, and PANC-1 were propagated under standard conditions and treated with serial dilutions of ritonavir. Ritonavir inhibited cell growth in a dose-dependent manner as well as activated the intrinsic apoptotic pathway in human pancreatic ductal adenocarcinoma (PDAC) cell lines. We observed down-modulation of cell-cycle promoting and up-regulation of cell-cycle inhibitory genes; enhanced interaction of retinoblastoma protein (RB) with E2F-1 transcription factor; inhibition of phosphorylation of RB, resulting in sequestration of E2F-1 and subsequent down-regulation of S phase genes; decreased interaction of E2F-1 with its consensus binding sites; inhibition of cell motility and invasiveness; and inhibition of the AKT pathway. Our results demonstrate a potential use of ritonavir as part of combination chemotherapy for PDAC. Since ritonavir is FDA approved for HIV, drug repositioning for PDAC would limit the costs and reduce risks.

This report details the validation methods used to analyze consumption at each of these homes. It includes a detailed end-use examination of consumptions from the following categories: 1) Heating, 2) Cooling, 3) Lights, Appliances, and Miscellaneous Electric Loads (LAMELS) along with Domestic Hot Water Use, 4) Ventilation, and 5) PV generation. A utility bill disaggregation method, which allows a crude estimation of space conditioning loads based on outdoor air temperature, was also performed and the results compared to the actual measured data.

The arch, as a structural element, has been used since ancient times, originally in stone or timber, more recently in reinforced concrete or steel. However, the more modern materials involving steel and concrete are highly energy and carbon intensive. Hardwood timber, although renewable, takes between 30 and 50 years to grow when used structurally. But a highly sustainable tropical material, bamboo, a woody grass, requires only 4–5 years of growth before its considerable compressive and tensile strength can be used for developing structural arches for load-bearing applications. After progressively incorporating ENERGY STAR(R) for Homes Versions 1, 2, and 3 into its standard practices over the years, this builder, Brookside Development, was seeking to build an even more sustainable product that would further increase energy efficiency, while also addressing indoor air quality, water conservation, renewable-ready, and resiliency. These objectives align with the framework of the DOE Challenge Home program, which 'builds upon the comprehensive building science requirements of ENERGY STAR for Homes Version 3, along with proven Building America innovations and best practices. Other special attribute programs are incorporated to help builders reach unparalleled levels of performance with homes designed to last hundreds of years.' CARB partnered with Brookside Development on the design optimization and construction of the first home in a small development of seven planned new homes being built on the old Singer Estate in Derby, CT.

While previous versions of the International Energy Conservation Code (IECC) have included provisions to improve the air tightness of dwellings, for the first time, the 2012 IECC mandates compliance verification through blower door testing. Simply completing the Air Barrier and Insulation Installation checklist through visual inspection is no longer sufficient by itself. In addition, the 2012 IECC mandates a significantly stricter air sealing requirement. In Climate Zones 3 through 8, air leakage may not exceed 3 ACH50, which is a significant reduction from the 2009 IECC requirement of 7 ACH50. This requirement is for all residential buildings, which includes low-rise multifamily dwellings. While this air leakage rate requirement is an important component to achieving an efficient building thermal envelope, currently, the code language doesn't explicitly address differences between single family and multifamily applications. In addition, the 2012 IECC does not provide an option to sample dwellings for larger multifamily buildings, so compliance would have to be verified on every unit. With compliance with the 2012 IECC air leakage requirements on the horizon, several of Consortium for Advanced Residential Building's (CARB’s) multifamily builder partners are evaluating how best to comply with this requirement. Over recent years, heat pump water heaters (HPWHs) have become more readily available and more widely adopted in the marketplace. For a 6-month period, the Building America team Consortium for Advanced Residential Buildings monitored the performance of a GE Geospring HPWH in Windermere, Florida. The study found that the HPWH performed 144% more efficiently than a traditional electric resistance water heater, saving approximately 64% on water heating annually. The monitoring showed that the domestic hot water draw was a primary factor affecting the system's operating efficiency.

Steven Winter Associates, Inc. (SWA) monitored several advanced mechanical systems within a 2012 deep energy retrofitted home in the small Orlando suburb of Windermere, FL. This report provides performance results of one of the home's heat pump water heaters (HPWH) and the whole-house dehumidifier (WHD) over a six month period. In addition to assessing the energy performance of these systems, this study sought to quantify potential comfort improvements over traditional systems. This information is applicable to researchers, designers, plumbers, and HVAC contractors. Though builders and homeowners can find useful information within this report, the corresponding case studies are a likely better reference for this audience.

Load-bearing brick-masonry multifamily buildings are prevalent in urban areas across much of the Northeast and mid-Atlantic. In most instances, these buildings are uninsulated unless they have been renovated within the past two decades. Affordable housing capital budgets typically limit what can be spent and energy improvements often take a back seat to basic capital improvements such as interior finish upgrades and basic repairs. The Consortium for Advanced Residential Buildings is researching cost-effective solution packages for significant energy efficiency and indoor air-quality improvements in these urban buildings.

In 2009, the state of Nevada received nearly forty million dollars in Neighborhood Stabilization Funds from the Department of Housing and Urban Development. The purpose of this funding was to stabilize communities that have suffered from foreclosures and abandonment. In an effort to provide guidance to local officials and maximize how effectively this NSP funding is utilized in retrofitting homes, CARB provided design specifications, energy modeling, and technical support for the Building America Retrofit Alliance (BARA) team and its local partners - Better Building Performance, Nevada Energy Star Partners Green Alliance, and Home Free Nevada - for two retrofit test homes. One home was to demonstrate a modest retrofit and the other a deep energy retrofit. Through this project, CARB has provided two robust solution packages for retrofitting homes built in this region between the 1980s and early 1990s without substantially inconveniencing the occupants. The two test homes, the Carmen and Sierra Hills, demonstrate how cost-effectively energy efficient upgrades can be implemented in the hot, dry climate of the Southwest. In addition, the homes were used as an educational experience for home performance professionals, building trades, remodelers, and the general public. In-field trainings on air-sealing, HVAC upgrades, and insulating weremore » provided to local contractors during the retrofit and BARA documented these retrofits through a series of video presentations, beginning with a site survey and concluding with the finished remodel and test out.« less
