Anaesthesia combined with functional neuroimaging provides a powerful approach for understanding the brain mechanisms that change as consciousness fades. Although propofol is used ubiquitously in clinical interventions that reversibly suppress consciousness, its effect varies substantially between individuals, and the brain bases of this variability remain poorly understood. We asked whether three networks that are primary sites of propofol-induced sedation and key to conscious cognition — the dorsal attention (DAN), executive control (ECN), and default mode (DMN) network — underlie responsiveness variability under anaesthesia. Healthy participants (N=17) underwent propofol sedation inside the fMRI scanner at dosages of ‘moderate’ anaesthesia, and behavioural responsiveness was measured with a target detection task. To assess information processing, participants were scanned during an active engagement condition comprised of a suspenseful auditory narrative, in addition to the resting state. A behavioural investigation in a second group of non-anesthetized participants (N=25) qualified the attention demands of narrative understanding, which we then related to the brain activity of participants who underwent sedation. 30% of participants showed no delay in reaction times relative to wakefulness, whereas the others, showed significantly delayed and fragmented responses, or full omission of responses. These responsiveness differences did not relate to information processing differences. Rather, only the functional connectivity within the ECN during wakefulness differentiated the participants’ responsiveness level, with significantly stronger connectivity in the fast relative to slow responders. Consistent with this finding, fast responders had significantly higher grey matter volume in the frontal cortex aspect of the ECN. For the first time, these results show that responsiveness variability during propofol anaesthesia relates to inherent differences in brain function and structure within the executive control network, which can be predicted prior to sedation. These results shed light on the brain bases of responsiveness differences and highlight novel markers that may help to improve the accuracy of awareness monitoring during clinical anaesthesia.

Humans have structures dedicated to the processing of faces, which include cortical components (e.g., areas in occipital and temporal lobes) and subcortical components (e.g., superior colliculus and amygdala). Although faces are processed more quickly than stimuli from other categories, there is a lack of consensus regarding whether subcortical structures are responsible for rapid face processing. In order to probe this, we exploited the asymmetry in the strength of projections to subcortical structures between the nasal and temporal hemiretina. Participants detected faces from unrecognizable control stimuli and performed the same task for houses. In Experiments 1 and 3, at the fastest reaction times, participants detected faces more accurately than houses. However, there was no benefit of presenting to the subcortical pathway. In Experiment 2, we probed the coarseness of the rapid pathway, making the foil stimuli more similar to faces and houses. This eliminated the rapid detection advantage, suggesting that rapid face processing is limited to coarse representations. In Experiment 4, we sought to determine whether the natural difference between spatial frequencies of faces and houses were driving the effects seen in Experiments 1 and 3. We spatially filtered the faces and houses so that they were matched. Better rapid detection was again found for faces relative to houses, but we found no benefit of preferentially presenting to the subcortical pathway. Taken together, the results of our experiments suggest a coarse rapid detection mechanism, which was not dependent on spatial frequency, with no advantage for presenting preferentially to subcortical structures. Recent evidence suggests that visual short-term memory (VSTM) capacity estimated using simple objects, such as colours and oriented bars, may not generalise well to more naturalistic stimuli. More visual detail can be stored in VSTM when complex, recognisable objects are maintained compared to simple objects. It is not yet known if it is recognisability that enhances memory precision, nor whether maintenance of recognisable objects is achieved with the same network of brain regions supporting maintenance of simple objects.We used a novel stimulus generation method to parametrically warp photographic images along a continuum, allowing separate estimation of the precision of memory representations and the number of items retained. The stimulus generation method was also designed to create unrecognisable, though perceptually matched, stimuli, to investigate the impact of recognisability on VSTM. We adapted the widely-used change detection and continuous report paradigms for use with complex, photographic images.Across three functional magnetic resonance imaging (fMRI) experiments, we demonstrated greater precision for recognisable objects in VSTM compared to unrecognisable objects. This clear behavioural advantage was not the result of recruitment of additional brain regions, or of stronger mean activity within the core network. Representational similarity analysis revealed greater variability across item repetitions in the representations of recognisable, compared to unrecognisable complex objects. We therefore propose that a richer range of neural representations support VSTM for complex recognisable objects.

Cardiovascular health declines with age, increasing the risk of hypertension and elevated heart rate in middle and old age. Here, we used multivariate techniques to investigate the associations between cardiovascular health (diastolic blood pressure, systolic blood pressure, and heart rate) and white matter macrostructure (lesion volume and number) and microstructure (as measured by diffusion-weighted imaging) in the cross-sectional, population-based Cam-CAN cohort (N = 667, aged 18–88). We found that cardiovascular health and age made approximately similar contributions to white matter health and explained up to 56% of variance therein. Lower diastolic blood pressure, higher systolic blood pressure, and higher heart rate were each strongly, and independently, associated with white matter abnormalities on all indices. Body mass and exercise were associated with white matter health, both directly and indirectly via cardiovascular health. These results highlight the importance of cardiovascular risk factors for white matter health across the adult lifespan and suggest that systolic blood pressure, diastolic blood pressure, and heart rate affect white matter health via separate mechanisms., Highlights • Cardiovascular health is related to white matter lesion burden and diffusivity. • Low diastolic pressure, high systolic pressure, and higher heart rate contribute independently. • Cardiovascular health and age explain up to 56% of variance in white matter health. • The uncinate fasciculus, inferior fronto-occipital fasciculus, and forceps minor show most sensitivity. • Lower BMI and more exercise may have protective effects.

Certain transformations must occur within the brain to allow rapid processing of familiar experiences. Complex objects are thought to become unitized, whereby multifeature conjunctions are retrieved as rapidly as a single feature. Behavioral studies strongly support unitization theory, but a compelling neural mechanism is lacking. Here, we examined how unitization transforms conjunctive representations to become more “feature-like” by recruiting posterior regions of the ventral visual stream (VVS) whose architecture is specialized for processing single features. We used functional magnetic resonance imaging to scan humans before and after visual training with novel objects. We implemented a novel multivoxel pattern analysis to measure a conjunctive code, which represented a conjunction of object features above and beyond the sum of the parts. Importantly, a multivoxel searchlight showed that the strength of conjunctive coding in posterior VVS increased posttraining. Furthermore, multidimensional scaling revealed representational separation at the level of individual features in parallel to the changes at the level of feature conjunctions. Finally, functional connectivity between anterior and posterior VVS was higher for novel objects than for trained objects, consistent with early involvement of anterior VVS in unitizing feature conjunctions in response to novelty. These data demonstrate that the brain implements unitization as a mechanism to refine complex object representations over the course of multiple learning experiences.

Assessing language development in the first postnatal year is difficult, as receptive and expressive skills are rudimentary. Although outward manifestations of change are limited, the auditory language system is thought to undergo critical development at this age, as the foundations are laid for the rapid onset of spoken language in the second and third years. We recruited 11 infants, 7 healthy controls (gestational age = 40.69 ± 0.56; range from 40 to 41.43) and preterm babies (gestational age = 28.04 ± 0.95; range from 27.43 to 29.43) who underwent a Magnetic Resonance Imaging study during the first postnatal year (age at scan = 194.18 ± 97.98). We assessed white matter tracts using diffusion-weighted magnetic resonance imaging with probabilistic tractography. Fractional anisotropy was found to be largely mature even at one month, although there was a little further increase during the first postnatal year in both the acoustic radiation and the direct brainstem-Heschl's pathway.

Testing infants in the laboratory is expensive in time and money; consequently, many studies are underpowered, reducing their reproducibility. We investigated whether the online platform, Amazon Mechanical Turk (MTurk), could be used as a resource to more easily recruit and measure the behavior of infant populations. Using a looking time paradigm, with users' webcams we recorded how long infants aged 5 to 8months attended while viewing children's television programs. We found that infants (N=57) were more reliably engaged by some movies than by others and that the most engaging movies could maintain attention for approximately 70% of a 10- to 13-min period. We then identified the cinematic features within the movies. Faces, singing-and-rhyming, and camera zooms were found to increase infant attention. Together, we established that MTurk can be used as a rapid tool for effectively recruiting and testing infants.

Neuroimaging is a valuable diagnostic tool for the early detection of neonatal brain injury, but equipment and radiologic staff are expensive and unavailable to most hospitals in developing countries. We evaluated an affordable, portable ultrasound machine as a quantitative and qualitative diagnostic tool and to establish whether a novice sonographer could effectively operate the equipment and obtain clinically important information. Cranial ultrasonography was performed on term healthy, pre-term and term asphyxiated neonates in Rwandan and Kenyan hospitals. To evaluate the detection of ventriculomegaly and compression injuries, we measured the size of the lateral ventricles and corpus callosum. The images were also assessed for the presence of other cerebral abnormalities. Measurements were reliable across images, and cases of clinically relevant ventriculomegaly were detected. A novice sonographer had good-to-excellent agreement with an expert. This study demonstrates that affordable equipment and cranial ultrasound protocols can be used in low-resource settings to assess the newborn brain.

Physical activity has positive effects on brain health and cognitive function throughout the life span. Thus far, few studies have examined the effects of physical activity on white matter microstructure and psychomotor speed within the same, population-based sample (critical if conclusions are to extend to the wider population). Here, using diffusion tensor imaging and a simple reaction time task within a relatively large population-derived sample (N = 399; 18–87 years) from the Cambridge Centre for Ageing and Neuroscience (Cam-CAN), we demonstrate that physical activity mediates the effect of age on white matter integrity, measured with fractional anisotropy. Higher self-reported daily physical activity was associated with greater preservation of white matter in several frontal tracts, including the genu of corpus callosum, uncinate fasciculus, external capsule, and anterior limb of the internal capsule. We also show that the age-related slowing is mediated by white matter integrity in the genu. Our findings contribute to a growing body of work, suggesting that a physically active lifestyle may protect against age-related structural disconnection and slowing.

Online testing of infants by recording video with a webcam has the potential to improve the replicability of developmental studies by facilitating larger sample sizes and by allowing methods (including recruitment) to be specified in code. However, the recorded video still needs to be manually scored. This labour-intensive process puts downward pressure on sample sizes and requires subjective judgements that may not be reproducible in a different laboratory. Here we present the first fully automatic pipeline, using a face analysis software-as-a-service and a discriminant-analysis classifier to score infant videos acquired online. We compare human and machine performance for looking time and preferential looking paradigms; machine performance demonstrates a promising proof of principle for looking time and is above chance in classifying preferential looking. Additionally, we studied the characteristics of the video and the child that influenced automated scoring, so that future studies can acquire data that maximises the performance of automatic gaze coding and/or focus on improving automatic coding for particularly challenging data. We believe this technology has great promise for developmental science.

ObjectiveFunctional connectivity magnetic resonance imaging (fcMRI) of neonates with perinatal brain injury could improve prediction of motor impairment before symptoms manifest, and establish how early brain organization relates to subsequent development. Methods: This cohort study is the first to describe and quantitatively assess functional brain networks and their relation to later motor skills in neonates with a diverse range of perinatal brain injuries. Infants (n=65, included in final analyses: n=53) were recruited from the neonatal intensive care unit (NICU) and were stratified based on their age at birth (premature vs. term), and on whether neuropathology was diagnosed from structural MRI. Functional brain networks and a measure of disruption to functional connectivity were obtained from 14 minutes of fcMRI acquired during natural sleep at term-equivalent age.ResultsDisruption to connectivity of the somatomotor and frontoparietal executive networks predicted motor impairment at 4 and 8 months. This disruption in functional connectivity was not found to be driven by differences between clinical groups, or by any of the specific measures we captured to describe the clinical course.ConclusionfcMRI was predictive over and above other clinical measures available at discharge from the NICU, including structural MRI. Motor learning was affected by disruption to somatomotor networks, but also frontoparietal executive networks, which supports the functional importance of these networks in early development. Disruption to these two networks might be best addressed by distinct intervention strategies.

We studied developmental plasticity using functional magnetic resonance imaging (fMRI) in a preterm infant with brain injury on structural MRI. fMRI showed preserved brain function and subsequent neurodevelopment was within the normal range. Multimodal neuroimaging including fMRI can improve understanding of neural plasticity after preterm birth and brain injury.

The brainstem, critical for motor function, autonomic regulation, and many neurocognitive functions, undergoes rapid development from the third trimester. Accordingly, we hypothesized it would be vulnerable to insult during this period, and that a difficult clinical course in the neonatal intensive care unit (NICU) would affect development, and be reflected through atypical shape. Our study population consisted of 66 neonates – all inpatients from the NICU at Victoria Hospital, London Health Sciences Centre, ON, Canada, of which 45 entered the final analysis. The cohort varied in gestational age (GA) and ranged from neurologically healthy to severely brain-injured. Structural MRI was used to quantify brainstem shape at term-equivalent age. From these images, brainstems were semi-automatically segmented and co-registered across subjects. The anterior-posterior dimensions on a sagittal maximum intensity projection were used as the basis for shape comparison. Factor analysis was used to summarize variation in shape and in clinical course to determine three shape factors and three clinical factors, and their relationship assessed using correlation. A factor driven by low GA and associated complications correlated with alterations in the posterior medulla, while a factor driven by complications independent of GA correlated with alterations in the midbrain. Additionally, single clinical measures most representative of their respective clinical factor (days in NICU; days on ventilation) predicted the changes. Thus, different clinical courses in the NICU may have different effects on the shape of the brainstem, and may mediate some of the distinct neurodevelopmental profiles observed in premature and brain-injured neonates., Highlights • Gestational age & complications correlated with posterior medulla alterations • Complications independent of gestational age correlated with midbrain alterations • Single clinical measures representative of their clinical factor predicted changes • Different NICU clinical courses found to have distinct effects on brainstem shape

An important component of perception, attention, and memory is the structuring of information into subsets (“objects”), which allows some parts to be considered together but kept separate from others. Portions of the posterior parietal lobe respond proportionally to the number of objects in the scope of attention and short-term memory, up to a capacity limit of around four, suggesting they have a role in this important process. This study investigates the relationship of discrete object representation to other parietal functions. Two experiments and two supplementary analyses were conducted to evaluate responsivity in parietal regions to the number of objects, the number of spatial locations, attention switching, and general task difficulty. Using transparent motion, it was found that a posterior and inferior parietal response to multiple objects persists even in the absence of a change in visual extent or the number of spatial locations. In a monitoring task, it was found that attention switching (or task difficulty) and object representation have distinct neural signatures, with the former showing greater recruitment of an anterior and lateral intraparietal sulcus (IPS) region, but the latter in a posterior and lateral region. A dissociation was also seen between selectivity for object load across tasks in the inferior IPS and feature or object-related memory load in the superior IPS.

Mental imagery is a critical cognitive function, clinically important, but poorly understood. When visual objects are perceived, many of their sensory, semantic and emotional properties are represented in occipitotemporal cortex. Visual imagery has been found to activate some of the same brain regions, but it was not known what properties are re-created in these regions during imagery. We therefore examined the representation during imagery for two stimuli in depth, by comparing the pattern of fMRI response to the patterns evoked by the perception of 200 diverse objects chosen to de-correlate their properties. Real-time, adaptive stimulus selection allowed efficient sampling of this broad stimulus space. Our experiments show that occipitotemporal cortex, which encoded sensory, semantic and emotional properties during perception, can robustly represent semantic and emotional properties during imagery, but that these representations depend on the object being imagined and on individual differences in style and reported vividness of imagery.

Multivariate pattern analysis is often assumed to rely on signals that directly reflect differences in the distribution of particular neural populations. The source of the signal used in these analyses remains unclear however, and an alternative model suggests that signal from larger draining veins may play a significant role. The current study was designed to investigate the vascular contribution to pattern analyses at 3T by comparing the results obtained from gradient and spin echo data. Classification analyses were carried out comparing line orientations in V1, tone frequencies in A1, and responses from different fingers in M1. In all cases, classification accuracy in the spin echo data was not significantly different from chance. In contrast, classification accuracies in the gradient echo data were significantly above chance, and significantly higher than the accuracies observed for the spin echo data. These results suggest that at the field strength and spatial resolution used for the majority of fMRI studies, a considerable proportion of the signal used by pattern analysis originates in the vasculature.

A key challenge of object recognition is achieving a balance between selectivity for relevant features and invariance to irrelevant ones. Computational and cognitive models predict that optimal selectivity for features will differ by object, and here we investigate whether this is reflected in visual representations in the human ventral stream. We describe a new real‐time neuroimaging method, dynamically adaptive imaging (DAI), that enabled measurement of neural selectivity along multiple feature dimensions in the neighborhood of single referent objects. The neural response evoked by a referent was compared to that evoked by 91 naturalistic objects using multi‐voxel pattern analysis. Iteratively, the objects evoking the most similar responses were selected and presented again, to converge upon a subset that characterizes the referent's “neural neighborhood.” This was used to derive the feature selectivity of the response. For three different referents, we found strikingly different selectivity, both in individual features and in the balance of tuning to sensory versus semantic features. Additional analyses placed a lower bound on the number of distinct activation patterns present. The results suggest that either the degree of specificity available for object representation in the ventral stream varies by class, or that different objects evoke different processing strategies. Three experiments studied auditory streaming using sequences of alternating "ABA" triplets, where "A" and "B" were 50-ms tones differing in frequency by Δf semitones and separated by 75-ms gaps. Experiment 1 showed that detection of a short increase in the gap between a B tone and the preceding A tone, imposed on one ABA triplet, was better when the delay occurred early versus late in the sequence, and for Δf = 4 vs. Δf = 8. The results of this experiment were consistent with those of a subjective streaming judgment task. Experiment 2 showed that the detection of a delay 12.5 s into a 13.5-s sequence could be improved by requiring participants to perform a task on competing stimuli presented to the other ear for the first 10 s of that sequence. Hence, adding an additional task demand could improve performance via its effect on the perceptual organization of a sound sequence. The results demonstrate that attention affects streaming in an objective task and that the effects of build-up are not completely under voluntary control. In particular, even though build-up can impair performance in an objective task, participants are unable to prevent this from happening.

Critical to perceiving an object is the ability to bind its constituent features into a cohesive representation, yet the manner by which the visual system integrates object features to yield a unified percept remains unknown. Here, we present a novel application of multivoxel pattern analysis of neuroimaging data that allows a direct investigation of whether neural representations integrate object features into a whole that is different from the sum of its parts. We found that patterns of activity throughout the ventral visual stream (VVS), extending anteriorly into the perirhinal cortex (PRC), discriminated between the same features combined into different objects. Despite this sensitivity to the unique conjunctions of features comprising objects, activity in regions of the VVS, again extending into the PRC, was invariant to the viewpoints from which the conjunctions were presented. These results suggest that the manner in which our visual system processes complex objects depends on the explicit coding of the conjunctions of features comprising them.

The presence of the head in an MRI scanner leads to inhomogeneities in the magnetic field. These cause the 'susceptibility artifacts' of image distortion and signal dropout. In this paper, we evaluate a technique called passive shimming, which has the potential to reduce field inhomogeneities and the resultant artifacts. A piece of a magnetically active material (pyrolytic graphite) is held on the roof of the participant's mouth by a plastic mouth mould. We evaluate the effects in several different ways. We show that the presence of a shim reduces field inhomogeneity across much of the brain. From field maps, we generate simulations of EPI image intensity and BOLD sensitivity. Both of these are mainly improved by the presence of shim, although there were small reductions in some regions. Measured EPI image intensity also mostly increased. Finally, we ran a reward-punishment task in our subjects, and found that the presence of a shim increased functional sensitivity in the orbitofrontal cortex. Using the BOLD sensitivity measure, we provide estimates of the improvement to be expected in functional studies for a range of neural structures. Passive shims are quick to make and reasonably comfortable to wear, and have substantial potential for researchers investigating inferior frontal brain regions using MRI.

Often, the sound arriving at the ears is a mixture from many different sources, but only 1 is of interest. To assist with selection, the auditory system structures the incoming input into streams, each of which ideally corresponds to a single source. Some authors have argued that this process of streaming is automatic and invariant, but recent evidence suggests it is affected by attention. In Experiments 1 and 2, it is shown that the effect of attention is not a general suppression of streaming on an unattended side of the ascending auditory pathway or in unattended frequency regions. Experiments 3 and 4 investigate the effect on streaming of physical gaps in the sequence and of brief switches in attention away from a sequence. The results demonstrate that after even short gaps or brief switches in attention, streaming is reset. The implications are discussed, and a hierarchical decomposition model is proposed.

When a head is placed in an MRI scanner, differences between the magnetic susceptibility of tissue, bone, and air distort the magnetic field. While some of the resulting inhomogeneity can be corrected by the shimming process, much of it cannot, and this causes distortion (sometimes referred to as geometric distortion) of echo-planar images (EPIs). One strategy for the correction of distortion is to acquire a map of the magnetic field achieved in each subject and then to use this to undistort their EPIs after reconstruction (Jezzard and Balaban, 1995). Here, we present five experiments to evaluate the application of such a strategy on data from a 3-T scanner. We show that after undistortion, the shape of EPIs is more similar to the true shape of the brain, and we investigate the effect of head movement on the efficacy of undistortion. If undistortion was applied first, it was found that less nonlinear warping was required to transform EPIs into a standard space, particularly in the phaseencode direction. We show that if SPM 99 normalization is used to perform a nonlinear warp to standard space, the prior application of undistortion increases the statistical power of group studies with motor and auditory tasks. We show that this increase in power is due to an increase in the overlap of activation of different subjects. Finally, we evaluate where in the brain undistorting EPIs might be expected to have the greatest effect, in terms both of mislocalization of activation and of a reduction in power. Overall, undistorting EPIs using field maps has proved extremely successful, improving the anatomical localization of activation and increasing statistical power. © 2002 Elsevier Science (USA)

To isolate the neural mechanisms associated with recognizing objects from those processing basic visual properties, control stimuli are required that contain the same perceptual properties as the objects but are unrecognizable. We demonstrate that conventional methods for generating control stimuli (phase scrambling, box scrambling, texture scrambling) yield poor controls because they dramatically distort the basic visual properties (e.g., spatial frequency, perceptual organization) to which even the earliest stages of visual processing are sensitive. We developed a new scrambling method, using a diffeomorphic transformation that preserves the basic perceptual properties of the image while removing meaning. We acquired perceptual ratings to determine the least amount of scrambling necessary to remove recognition. We hypothesized that our "diffeomorphic" images would produce neural activity at the earliest stages of the visual system that more closely matched activity in response to intact images relative to the other scrambling methods. To test this hypothesis, we used the HMAX computational model of object recognition and compared the simulated neural activity at the earliest stages of the visual system (layers S1, C1, and S2) between a set of 149 images scrambled using each distortion method to their intact version. We found that scrambled "diffeomorphed" images were indistinguishable to intact images in each layer of the model, but all of the other distortion methods yielded quite different patterns. Our results indicate that "diffeomorphed" images serve as more appropriate control stimuli in neuroimaging studies that aim to disentangle the representations of perceptual and semantic object properties.

Differences in the timbre of sounds in a sequence can affect their perceptual organization. Using a performance measure, Hartmann and Johnson (1991) concluded that streaming could be predicted primarily by the extent to which sounds were passed by different peripheral channels. However, results from a rating task by Dannenbring and Bregman (1976) suggested that sounds in the same spectral region (passed by the same peripheral channels) can be allocated to different streams. In Experiment 1, it was found, using an interleaved melody task, that target sounds could be selected from distractors in the same spectral region more easily when they differed in timbre. This finding might result from primitive stream segregation or schema-driven selection, but not from peripheral channeling. In Experiment 2, a rhythm discrimination task was used, requiring the sounds to be integrated for good performance. Differences in timbre impaired performance, indicating the occurrence of primitive stream segregation.

Recent years have seen neuroimaging data sets becoming richer, with larger cohorts of participants, a greater variety of acquisition techniques, and increasingly complex analyses. These advances have made data analysis pipelines complicated to set up and run (increasing the risk of human error) and time consuming to execute (restricting what analyses are attempted). Here we present an open-source framework, automatic analysis (aa), to address these concerns. Human efficiency is increased by making code modular and reusable, and managing its execution with a processing engine that tracks what has been completed and what needs to be (re)done. Analysis is accelerated by optional parallel processing of independent tasks on cluster or cloud computing resources. A pipeline comprises a series of modules that each perform a specific task. The processing engine keeps track of the data, calculating a map of upstream and downstream dependencies for each module. Existing modules are available for many analysis tasks, such as SPM-based fMRI preprocessing, individual and group level statistics, voxel-based morphometry, tractography, and multi-voxel pattern analyses (MVPA). However, aa also allows for full customization, and encourages efficient management of code: new modules may be written with only a small code overhead. aa has been used by more than 50 researchers in hundreds of neuroimaging studies comprising thousands of subjects. It has been found to be robust, fast, and efficient, for simple-single subject studies up to multimodal pipelines on hundreds of subjects. It is attractive to both novice and experienced users. aa can reduce the amount of time neuroimaging laboratories spend performing analyses and reduce errors, expanding the range of scientific questions it is practical to address.

Processing multiple complex features to create cohesive representations of objects is an essential aspect of both the visual and auditory systems. It is currently unclear whether these processes are entirely modality specific or whether there are amodal processes that contribute to complex object processing in both vision and audition. We investigated this using a dual-stream target detection task in which two concurrent streams of novel visual or auditory stimuli were presented. We manipulated the degree to which each stream taxed processing conjunctions of complex features. In two experiments, we found that concurrent visual tasks that both taxed conjunctive processing strongly interfered with each other but that concurrent auditory and visual tasks that both taxed conjunctive processing did not. These results suggest that resources for processing conjunctions of complex features within vision and audition are modality specific.

It is now well established that some patients who are diagnosed as being in a vegetative state or a minimally conscious state show reliable signs of volition that may only be detected by measuring neural responses. A pertinent question is whether these patients are also capable of logical thought. Here, we validate an fMRI paradigm that can detect the neural fingerprint of reasoning processes and moreover, can confirm whether a participant derives logical answers. We demonstrate the efficacy of this approach in a physically non-communicative patient who had been shown to engage in mental imagery in response to simple auditory instructions. Our results demonstrate that this individual retains a remarkable capacity for higher cognition, engaging in the reasoning task and deducing logical answers. We suggest that this approach is suitable for detecting residual reasoning ability using neural responses and could readily be adapted to assess other aspects of cognition., Highlights ► Hampshire et al. develop a paradigm for assessing reasoning ability using fMRI alone. ► Inferior frontal sulcus activation is used as a marker of engagement in reasoning. ► Visual and pre-motor activation is used to detect logically correct answers. ► An overtly non-responsive patient is able to solve grammatical reasoning problems. ► Frontal lobe sub-regions activate at distinct stages of the reasoning task.

A key question in cognitive neuroscience is how the brain combines low-level features processed in remote sensory cortices to represent meaningful multisensory objects in our everyday environment. Models of visual object processing typically assume a feedforward cascade through the hierarchically organized ventral stream. We contrasted this feedforward view with an alternate hypothesis in which object processing is viewed as an interactive, feedforward and feedback process. We found that higher-order regions in anterior temporal (AT) and inferior prefrontal cortex (IPC) performed audio-visual (AV) integration 100 ms earlier than a sensory-driven region in the posterior occipital (pO) cortex, and were modulated by semantic variables (congruency), from as early as 50-100 ms. We propose that the brain represents familiar and complex multisensory objects through early interactivity between higher-order and sensory-driven regions. This interactivity may underpin the enhanced behavioral performance reported for semantically congruent AV objects.

An electroencephalographic (EEG) marker of the limited contents of human visual short-term memory (VSTM) has previously been described. Termed contralateral delay activity (CDA), this consists of a sustained, posterior, negative potential that correlates with memory load and is greatest contralateral to the remembered hemifield. The current investigation replicates this finding and uses magnetoencephalography (MEG) to characterise its magnetic counterparts and their neural generators as they evolve throughout the memory delay. A parametric manipulation of memory load, within and beyond capacity limits, allows separation of signals that asymptote with behavioural VSTM performance from additional responses that contribute to a linear increase with set-size. Both EEG and MEG yielded bilateral signals that track the number of objects held in memory, and contralateral signals that are independent of memory load. In MEG, unlike EEG, the contralateral interaction between hemisphere and item load is much weaker, suggesting that bilateral and contralateral markers of memory load reflect distinct sources to which EEG and MEG are differentially sensitive. Nonetheless, source estimation allowed both the bilateral and the weaker contralateral capacity-limited responses to be localised, along with a load-independent contralateral signal. Sources of global and hemisphere-specific signals all localised to the posterior intraparietal sulcus during the early delay. However the bilateral load response peaked earlier and its generators shifted later in the delay. Therefore the hemifield-specific response may be more closely tied to memory maintenance while the global load response may be involved in initial processing of a limited number of attended objects, such as their individuation or consolidation into memory.

Auditory object analysis requires two fundamental perceptual processes: the definition of the boundaries between objects, and the abstraction and maintenance of an object's characteristic features. Although it is intuitive to assume that the detection of the discontinuities at an object's boundaries precedes the subsequent precise representation of the object, the specific underlying cortical mechanisms for segregating and representing auditory objects within the auditory scene are unknown. We investigated the cortical bases of these two processes for one type of auditory object, an “acoustic texture,” composed of multiple frequency-modulated ramps. In these stimuli, we independently manipulated the statistical rules governing (1) the frequency–time space within individual textures (comprising ramps with a given spectrotemporal coherence) and (2) the boundaries between textures (adjacent textures with different spectrotemporal coherences). Using functional magnetic resonance imaging, we show mechanisms defining boundaries between textures with different coherences in primary and association auditory cortices, whereas texture coherence is represented only in association cortex. Furthermore, participants' superior detection of boundaries across which texture coherence increased (as opposed to decreased) was reflected in a greater neural response in auditory association cortex at these boundaries. The results suggest a hierarchical mechanism for processing acoustic textures that is relevant to auditory object analysis: boundaries between objects are first detected as a change in statistical rules over frequency–time space, before a representation that corresponds to the characteristics of the perceived object is formed.

Recent studies suggested that fMRI voxel patterns can convey information represented in columnar-scale neuronal population codes, even when spatial resolution is insufficient to directly image the patterns of columnar selectivity (Kamitani and Tong, 2005; Haynes and Rees, 2005). Sensitivity to subvoxel-scale pattern information, or "fMRI hyperacuity," would greatly enhance the power of fMRI when combined with pattern information analysis techniques (Kriegeskorte and Bandettini, 2007). An individual voxel might weakly reflect columnar-level information if the columns within its boundaries constituted a slightly unbalanced sample of columnar selectivities (Kamitani and Tong, 2005), providing a possible mechanism for fMRI hyperacuity. However, Op de Beeck (2009) suggests that a coarse-scale neuronal organization rather than fMRI hyperacuity may explain the presence of the information in the fMRI patterns. Here we argue (a) that the present evidence does not rule out fMRI hyperacuity, (b) that the mechanism originally suggested for fMRI hyperacuity (biased sampling by averaging within each voxel's boundaries; Kamitani and Tong, 2005) will only produce very weak sensitivity to fine-grained pattern information, and (c) that an alternative mechanism (voxel as complex spatiotemporal filter) is physiologically more accurate and promises stronger sensitivity to fine-grained pattern information: We know that each voxel samples the neuronal activity pattern through a unique fine-grained structure of venous vessels that supply its blood oxygen level-dependent signal. At the simplest level, the drainage domain of a venous vessel may sample the neuronal pattern with a selectivity bias (Gardner, 2009; Shmuel et al., 2009). Beyond biased drainage domains, we illustrate with a simple simulation how temporal properties of the hemodynamics (e.g., the speed of the blood in the capillary bed) can shape spatial properties of a voxel's filter (e.g., how finely structured it is). This suggests that a voxel, together with its signal-supplying vasculature, may best be thought of as a complex spatiotemporal filter. Such a filter may well have greater sensitivity to high spatial frequencies than the Gaussian or averaging-box kernels typically invoked to characterize voxel sampling (compact kernels, both of which would act like anti-aliasing filters that minimize such sensitivity). Importantly, the complex-spatiotemporal-filter hypothesis of fMRI hyperacuity can account for the observed robustness to slight shifts of the voxel grid caused by head motion: Because the fine-grained components of the filter are vascular, they will remain in a constant relationship to the neuronal patterns sampled as the voxel grid is slightly shifted.

Visual imagery is mediated via top-down activation of visual cortex. Similar to stimulus-driven perception, the neural configurations associated with visual imagery are differentiated according to content. For example, imagining faces or places differentially activates visual areas associated with perception of actual face or place stimuli. However, while top-down activation of topographically specific visual areas during visual imagery is well established, the extent to which internally generated visual activity resembles the fine-scale population coding responsible for stimulus-driven perception remains unknown. Here, we sought to determine whether top-down mechanisms can selectively activate perceptual representations coded across spatially overlapping neural populations. We explored the precision of top-down activation of perceptual representations using neural pattern classification to identify activation patterns associated with imagery of distinct letter stimuli. Pattern analysis of the neural population observed within high-level visual cortex, including lateral occipital complex, revealed that imagery activates the same neural representations that are activated by corresponding visual stimulation. We conclude that visual imagery is mediated via top-down activation of functionally distinct, yet spatially overlapping population codes for high-level visual representations.

A novel method is presented for creating a probability map from histologically defined cytoarchitectonic data, customised for the anatomy of individual fMRI volunteers. Postmortem structural and cytoarchitectonic information from a published dataset is combined with high resolution structural MR images using deformable registration of a region of interest. In this paper, we have targeted the three subareas of the primary auditory cortex (located on Heschl's gyrus); however, the method could be applied to any other cytoarchitectonic region. The resulting probability maps show a significantly higher overlap than previously generated maps using the same cytoarchitectonic data, and more accurately span the macroanatomical structure of the auditory cortex. This improvement indicates a high potential for spatially accurate fMRI analysis, allowing more reliable correlation between anatomical structure and function. We validate the approach using fMRI data from nine individuals, taken from a published dataset. We compare activation for stimuli evoking a pitch percept to activation for acoustically matched noise, and demonstrate that the primary auditory cortex (Te1.0) and the lateral region Te1.2 are sensitive to pitch, whereas Te1.1 is not.

PUBLISHED, Action errors can occur when routine responses are triggered inappropriately by familiar cues. Here, EEG was recorded as volunteers performed a ?go/no-go? task of long duration that occasionally and unexpectedly required them to withhold a frequent, routine response. EEG components locked to the onset of relevant go trials were sorted according to whether participants erroneously responded to immediately subsequent no-go trials or correctly withheld their responses. Errors were associated with a significant relative reduction in the amplitude of the preceding P300, that is, a judgement could be made bout whether a responseinhibition error was likely before it had actually occurred. Furthermore, fluctuations in P300 amplitude across the task formed a reliable associate of individual error propensity, supporting its use as a marker of sustained control over action., MRC, NIH

Phase contrast magnetic resonance velocity imaging is a powerful technique for quantitative in vivo blood flow measurement. Current practice normally involves restricting the sensitivity of the technique so as to avoid the problem of the measured phase being ‘wrapped’ onto the range − π to + π . However, as a result, dynamic range and signal-to-noise ratio are sacrificed. Alternatively, the true phase values can be estimated by a phase unwrapping process which consists of adding integral multiples of 2 π to the measured wrapped phase values. In the presence of noise and data undersampling, the phase unwrapping problem becomes non-trivial. In this paper, we investigate the performance of three different phase unwrapping algorithms when applied to three-dimensional (two spatial axes and one time axis) phase contrast datasets. A simple one-dimensional temporal unwrapping algorithm, a more complex and robust three-dimensional unwrapping algorithm and a novel velocity encoding unwrapping algorithm which involves unwrapping along a fourth dimension (the ‘velocity encoding’ direction) are discussed, and results from the three are presented and compared. It is shown that compared to the traditional approach, both dynamic range and signal-to-noise ratio can be increased by a factor of up to five times, which demonstrates considerable promise for a possible eventual clinical implementation. The results are also of direct relevance to users of any other technique delivering time-varying two-dimensional phase images, such as dynamic speckle interferometry and synthetic aperture radar.

There are several types of experiment in which it is useful to have subjects speak overtly in a magnetic resonance imaging (MRI) scanner, including those studying the articulatory apparatus and the neural basis of speech production, and fMRI experiments in which speech is used as a response modality. Although it is relatively easy to record sound from the bore, it can be difficult to hear the speech over the very loud acoustic noise from the scanner. This is particularly a problem during echo‐planar imaging, which is usually used for fMRI. We present a post‐hoc sound cancellation algorithm, and describe a Windows‐based tool that implements it. The tool is fast and operates with minimal user intervention. We evaluate cancellation performance in terms of the improvement in signal‐to‐noise ratio, and investigate the effect of the recording medium. A substantial improvement in audibility was obtained. Carlyon et al (2001 Journal of Experimental Psychology: Human Perception and Performance27 115–127) have reported that the buildup of auditory streaming is reduced when attention is diverted to a competing auditory stimulus. Here, we demonstrate that a reduction in streaming can also be obtained by attention to a visual task or by the requirement to count backwards in threes. In all conditions participants heard a 13 s sequence of tones, and, during the first 10 s saw a sequence of visual stimuli containing three, four, or five targets. The tone sequence consisted of twenty repeating triplets in an ABA–ABA … order, where A and B represent tones of two different frequencies. In each sequence, three, four, or five tones were amplitude modulated. During the first 10 s of the sequence, participants either counted the number of visual targets, counted the number of (modulated) auditory targets, or counted backwards in threes from a specified number. They then made an auditory-streaming judgment about the last 3 s of the tone sequence: whether one or two streams were heard. The results showed more streaming when participants counted the auditory targets (and hence were attending to the tones throughout) than in either the ‘visual’ or ‘counting-backwards’ conditions.

The phase, as well as the magnitude, of MRI images can carry useful information. It may be used to encode flow or temperature, or to map the magnetic field for the undistorting of EPIs and automated shimming. In all cases, we measure the extra spin given to nuclei. Unfortunately, we can only measure the final phase of the spins: the rotation is wrapped into the range [−π, +π], and to obtain a measure of the parameter of interest the missing multiples of 2π must be replaced—a process known as phase unwrapping. While simple in principle, standard phase unwrapping algorithms fail catastrophically in the presence of even small amounts of noise. Here we present a new algorithm for robust three-dimensional phase unwrapping, in which unwrapping is guided, so that it initially works on less noisy regions. We test the algorithm on simulated phase data, and on maps of magnetic field, which were then used to successfully undistort EPI images. The unwrapping algorithm could be directly applied to other kinds of phase data.

Where a shift in midline was found, we measured the hearing threshold of each ear independently to rule out low-level perceptual effects. We also calculated a measure of spatial discrimination performance overall and on each side, to rule out an effect of asymmetric performance. We presented sounds using headphones to allow tight control of the stimuli and to discourage use of an external visual reference. We always presented the sound at the same level to both ears, to avoid an interaction of any possible hearing loss with the level of the sounds in each ear, and to prevent the use of monaural cues in the threeinterval task. We then altered the perceived location of sounds by adding a very small lag to the sound in one ear (an interaural time delay, ITD).

Anatomical tracing in animal models has shown that the thalamus projects to regions in the temporal lobe around primary auditory cortex, and many regions anterior to this. During development, post-mortem studies in humans have shown that myelination in the temporal lobes follows a posterior-to-anterior progression. Our aim was to investigate auditory development, and specifically the thalamo-temporal white matter connexions, in infants during the first postnatal year. We hypothesised that the connectivity will strengthen from birth through the first year, particularly in anterior temporal areas. We recruited 4 healthy controls (1 month, 3 months, 9 months and 11 months). We assessed white matter tracts using diffusion-weighted MRI with probabilistic tractography. A highly accelerated multiband EPI sequence (monopolar acquisition, acceleration factor=4, iPAT=0) with 128 non-collinear diffusion weighting directions was acquired (TR/TE=1980/71, voxel size 2 × 2 × 2 mm 3 , b=1500 s/mm 2 ). We found an increment in the strength of connectivity from 1 to 11 months between the thalamus and cortex (thalamo-temporal 60% and temporo-thalamic 37%). Moreover, the pathway’s fractional anisotropy increased by 26% and its mean diffusivity dropped by 90%. Furthermore, the pattern of development followed a posterior-to-anterior progression, with an extension of connections to more anterior temporal regions at 9–11 months. In conclusion, thalamic connections to the temporal lobe were found to strengthen in the first postnatal year, around primary auditory cortex, and especially in anterior temporal regions. The enhanced anterior temporal connectivity may reflect the development of the posterior-to-anterior cortical processing stream that in adults processes complex sounds such as language.

The entropy metric derived from information theory provides a means to quantify the amount of information transmitted in acoustic streams like speech or music. By systematically varying the entropy of pitch sequences, we sought brain areas where neural activity and energetic demands increase as a function of entropy. Such a relationship is predicted to occur in an efficient encoding mechanism that uses less computational resource when less information is present in the signal: we specifically tested the hypothesis that such a relationship is present in the planum temporale (PT). In two convergent functional MRI studies, we demonstrated this relationship in PT for encoding, while furthermore showing that a distributed fronto-parietal network for retrieval of acoustic information is independent of entropy. The results establish PT as an efficient neural engine that demands less computational resource to encode redundant signals than those with high information content., Author Summary Understanding how the brain makes sense of our acoustic environment remains a major challenge. One way to describe the complexity of our acoustic environment is in terms of information entropy: acoustic signals with high entropy convey large amounts of information, whereas low entropy signifies redundancy. To investigate how the brain processes this information, we controlled the amount of entropy in the signal by using pitch sequences. Participants listened to pitch sequences with varying amounts of entropy while we measured their brain activity using functional magnetic resonance imaging (fMRI). We show that the planum temporale (PT), a region of auditory association cortex, is sensitive to the entropy in pitch sequences. In two convergent fMRI studies, activity in PT increases as the entropy in the pitch sequence increases. The results establish PT as an important “computational hub” that requires less resource to encode redundant signals than it does to encode signals with high information content., A part of the auditory cortex (planum temporale) encodes the information content of pitch sequences.
