BACKGROUND: COVID-19 pandemic continues to pose a huge threat to public health. Mass vaccination is needed to achieve herd immunity against SARS-CoV-2. Currently, several vaccines are being inoculated on a large-scale. The willingness of COVID-19 vaccination had been well investigated in the pre-vaccination era, but no reported data in the post-vaccination era yet. METHODS: We conducted a large-scale survey among industrial workers during the vaccination campaign in China. Chi-square test and rank sum test were used to identify differences for various intentions regarding COVID-19 vaccination. Univariate analysis and multivariate regression models were utilized to analyze the relationship among demographic factors, related influencing factors and acceptance of COVID-19 vaccination. RESULTS: A total of 23,940 industrial workers were included, 66.0% were willing to take COVID-19 vaccine, 16.6% were unwilling, and 17.4% were unsure. Participants were more likely to get vaccinated if they were male, aged 45–65, being good educated, married, or being recommended by doctors or nurses. Participants with strong risk perception of COVID-19 infection, strong confidence in COVID-19 vaccine, high attention to COVID-19 vaccine, good health status, bad health habit, and a history of vaccination within three months were also more likely to be vaccinated. CONCLUSIONS: This study calls for more attention and health-related education among industrial workers to improve their acceptance of COVID-19 vaccination.

Background: Acute kidney injury (AKI) is a significant public health problem with high morbidity and mortality. Unfortunately, no definitive treatment is currently available for AKI. RNA interference (RNAi) provides a new and potent method for gene therapy to tackle this dilemma. Methods We engineered red blood cell-derived extracellular vesicles (REVs) with targeting peptides and therapeutic siRNAs to treat experimental AKI in a mouse model following renal ischemia/reperfusion (I/R) injury and unilateral ureteral obstruction (UUO). Phage display identified peptides that bind to the kidney injury molecule-1 (Kim-1). RNA-sequencing (RNA-seq) characterized the transcriptome of ischemic kidney to explore potential therapeutic targets. Results REVs targeted with Kim-1-binding LTH peptide (REVLTH) efficiently homed to and accumulated at the injured tubules in kidney following I/R injury. We identified transcription factors P65 and Snai1 that drive inflammation and fibrosis as potential therapeutic targets. Taking advantage of the established REVLTH, siRNAs targeting P65 and Snai1 were efficiently delivered to ischemic kidney and consequently blocked the expression of P-p65 and Snai1 in tubules. Moreover, dual suppression of P65 and Snai1 significantly improved I/R- and UUO-induced kidney injury by alleviating tubulointerstitial inflammation and fibrosis, and potently abrogated the transition to chronic kidney disease. Conclusions A red blood cell-derived extracellular vesicle platform targeted Kim-1 in acutely injured mouse kidney and delivered siRNAs for transcription factors P65 and Snai1, alleviating inflammation and fibrosis in the tubules.

Oxygen homeostasis disturbances play a critical role in the pathogenesis of acute kidney injury (AKI). The transcription factor hypoxia-inducible factor-1 (HIF-1) is a master regulator of adaptive responses to hypoxia. Aside from posttranslational hydroxylation, the mechanism of HIF-1 regulation in AKI remains largely unclear. In this study, the mechanism of HIF-α regulation in AKI was investigated. We found that tubular HIF-1α expression significantly increased at the transcriptional level in ischemia-reperfusion-, unilateral ureteral obstruction-, and sepsis-induced AKI models, which was closely associated with macrophage-dependent inflammation. Meanwhile, NF-κB, which plays a central role in the inflammation response, was involved in the increasing expression of HIF-1α in AKI, as evidenced by pharmacological modulation (NF-κB inhibitor BAY11-7082). Mechanistically, NF-κB directly bound to the HIF-1α promoter and enhanced its transcription, which occurred not only under hypoxic conditions but also under normoxic conditions. Moreover, the induced HIF-1α by inflammation protected against tubular injury in AKI. Thus, our findings not only provide novel insights into HIF-1 regulation in AKI but also offer to understand the pathophysiology of kidney diseases.NEW & NOTEWORTHY Here, the mechanism of hypoxia-inducible factor-α (HIF-α) regulation in acute kidney injury (AKI) was investigated. Mesenchymal stem cells-derived exosomes (MSC-exos) have attracted great interest as a cell-free therapy for acute kidney injury (AKI). However, the in vivo biodistribution of MSC-exos in ischemic AKI has not been established. The potential of MSC-exos in promoting tubular repair and the underlying mechanisms remain largely unknown. Methods: Transmission electron microscopy, nanoparticle tracking analysis, and western blotting were used to characterize the properties of human umbilical cord mesenchymal stem cells (hucMSCs) derived exosomes. The biodistribution of MSC-exos in murine ischemia/reperfusion (I/R) induced AKI was imaged by the IVIS spectrum imaging system. The therapeutic efficacy of MSC-exos was investigated in renal I/R injury. The cell cycle arrest, proliferation and apoptosis of tubular epithelial cells (TECs) were evaluated in vivo and in HK-2 cells. The exosomal miRNAs of MSC-exos were profiled by high-throughput miRNA sequencing. One of the most enriched miRNA in MSC-exos was knockdown by transfecting miRNA inhibitor to hucMSCs. Then we investigated whether this candidate miRNA was involved in MSC-exos-mediated tubular repair. Results: Ex vivo imaging showed that MSC-exos was efficiently homing to the ischemic kidney and predominantly accumulated in proximal tubules by virtue of the VLA-4 and LFA-1 on MSC-exos surface. MSC-exos alleviated murine ischemic AKI and decreased the renal tubules injury in a dose-dependent manner. Furthermore, MSC-exos significantly attenuated the cell cycle arrest and apoptosis of TECs both in vivo and in vitro. Mechanistically, miR-125b-5p, which was highly enriched in MSC-exos, repressed the protein expression of p53 in TECs, leading to not only the up-regulation of CDK1 and Cyclin B1 to rescue G2/M arrest, but also the modulation of Bcl-2 and Bax to inhibit TEC apoptosis. Finally, inhibiting miR-125b-5p could mitigate the protective effects of MSC-exos in I/R mice. Conclusion: MSC-exos exhibit preferential tropism to injured kidney and localize to proximal tubules in ischemic AKI. We demonstrate that MSC-exos ameliorate ischemic AKI and promote tubular repair by targeting the cell cycle arrest and apoptosis of TECs through miR-125b-5p/p53 pathway. This study provides a novel insight into the role of MSC-exos in renal tubule repair and highlights the potential of MSC-exos as a promising therapeutic strategy for AKI.

The current strategy to achieve high luminosity for high energy physics experiments is to store huge currents distributed in many bunches in circular machines such as BEPCII, so the longitudinal coupled bunch instabilities of the beam are inevitable. Therefore, a longitudinal feedback system is needed to suppress longitudinal oscillation. Otherwise, the beam energy spread will increase; the luminosity and the beam lifetime will decrease under a certain beam current. For filter design of the feedback system, the time domain least square fitting (TDLSF) method is compared to the selective wave filter method; digital signal processing electronics based on commercial board have been developed and tested on BEPCII. The final result shows that the self-developed longitudinal feedback system can suppress the longitudinal beam oscillation at 750 mA beam current and meet the requirements of BEPCII; the restrain depth to the sidebands reaches to 40.8 dB.

To establish a nuclear resonant scattering beamline at the High Energy Photon Source (HEPS), it is essential to develop tools for detection and cleaning of parasitic bunches, for meeting the stringent demands on bunch purity. To this end, a novel time-correlated single-photon counting system was implemented at the electron storage ring of the Beijing Electron–Positron Collider II (BEPCII). The purity deterioration process over a week-long operation was recorded by the system. In this study, the mechanism of impurity growth was analyzed by numerical methods and validated on measurements. The agreement between the experimental results and the calculation was fairly good. Two main sources of parasitic bunches, pre-accelerators and the Touschek scattering were confirmed. A bunch-cleaning technique, based on a sinusoidal signal mixed with a pseudo-square wave, was also developed and implemented, and its capability to improve the bunch purity to the level of 10–7 was experimentally demonstrated. We present the experimental setup, principle, and measurement results of a system for detection and cleaning of parasitic bunches.

Background: The production of intestinal gases and fecal short-chain fatty acids (SCFAs) by infant gut microbiota may have a significant impact on their health, but information about the composition and volume of intestinal gases and SCFA profiles in preterm infants is scarce.Objective: This study examined the change of the composition and volume of intestinal gases and SCFA profiles produced by preterm infant gut microbiota in vitro during the first 4 weeks of life.Methods: Fecal samples were obtained at five time points (within 3 days, 1 week, 2 weeks, 3 weeks, and 4 weeks) from 19 preterm infants hospitalized in the neonatal intensive care unit (NICU) of Shanghai Children's Hospital, Shanghai Jiao Tong University between May and July 2020. These samples were initially inoculated into four different media containing lactose (LAT), fructooligosaccharide (FOS), 2′-fucosyllactose (FL-2), and galactooligosaccharide (GOS) and thereafter fermented for 24 h under conditions mimicking those of the large intestine at 37.8°C under anaerobic conditions. The volume of total intestinal gases and the concentrations of individual carbon dioxide (CO2), hydrogen (H2), methane (CH4), and hydrogen sulfide (H2S) were measured by a gas analyzer. The concentrations of total SCFAs, individual acetic acid, propanoic acid, butyric acid, isobutyric acid, pentanoic acid, and valeric acid were measured by gas chromatography (GC).Results: The total volume of intestinal gases (ranging from 0.01 to 1.64 ml in medium with LAT; 0–1.42 ml with GOS; 0–0.91 ml with FOS; and 0–0.44 ml with FL-2) and the concentrations of CO2, H2, H2S, and all six fecal SCFAs increased with age (p-trends < 0.05). Among them, CO2 was usually the predominant intestinal gas, and acetic acid was usually the predominant SCFA. When stratified by birth weight (2 was more pronounced among infants whose weight was ≥1,500 g than among those whose weight was p-trends < 0.05).Conclusions: Our findings suggested that the intestinal gases and SCFAs produced by preterm infant gut microbiota in vitro increased with age during the first 4 weeks of life.

Mass vaccination against the COVID-19 pandemic is ongoing worldwide to achieve herd immunity among the general population. However, little is known about how the COVID-19 vaccination would affect mental health and preventive behaviors toward the COVID-19 pandemic. In this study, we conducted a cross-sectional survey to address this issue among 4244 individuals at several COVID-19 vaccination sites in Guangzhou, China. Using univariate analysis and multiple linear regression models, we found that major demographic characteristics, such as biological sex, age, education level, and family per capita income, are the dominant influencing factors associated with health beliefs, mental health, and preventive behaviors. After propensity score matching (PSM) treatment, we further assessed the changes in the scores of health belief, mental health, and preventive behaviors between the pre-vaccination group and the post-vaccination group. When compared to individuals in the pre-vaccination group, a moderate but statistically significant lower score was observed in the post-vaccination group (p = 0.010), implying possibly improved psychological conditions after COVID-19 vaccination. In addition, there was also a moderate but statistically higher score of preventive behaviors in the post-vaccination group than in the pre-vaccination group (p &lt, 0.001), suggesting a higher probability to take preventive measures after COVID-19 vaccination. These findings have implications for implementing non-pharmaceutical interventions combined with mass vaccination to control the rebound of COVID-19 outbreaks.

Objective: To report outcomes of balloon dilation Eustachian tuboplasty combined with tympanostomy tube insertion and middle ear pressure equalization therapy in treatment of recurrent secretory otitis media. Methods: Fifty one patients with recurrent secretory otitis media (62 ears) underwent balloon dilation of Eustachian tube and tympanic tube insertion under general anesthesia, followed by long term middle ear pressure equalization therapies. The Eustachian tube score (ETS) and Eustachian tube function questionnaire (ETDQ-7) were used for pre- and postoperative (up to 12 months) evaluation of Eustachian tube functions. Results: The mean ETS score was 2.34 ± 0.97 preoperatively, and 6.17 ± 1.54, 7.23 ± 1.62, 8.24 ± 1.97, and 7.63 ± 1.86 at 1, 3, 6 and 12 months postoperatively, respectively (P

MIMO technology is a key technology of 5G, which is widely used in next-generation scenarios such as heterogeneous networks, millimeter-wave networks, and automotive networks. How to build a large-scale MIMO system security situation assessment model for 5G has become the main topic of current concern. This paper analyzes the situational awareness theory of 5g-oriented MIMO system security. Firstly, based on the theory of MIMO system, the influence of MIMO system on 5G network security and the theory of situation awareness technology, the security situation awareness system model of 5g-oriented large-scale MIMO system is constructed. In the security situation assessment section of the MIMO system, according to the rules of evidence reasoning, of different active attack and passive eavesdropping two kinds of empowerment behavior under different attribute data, and then put the empowerment process of average power and large scale attenuation coefficient, interrupt probability, series, and maintain confidentiality gain data as model inputs, such as implementing MIMO system level of situation assessment. In the security situation prediction part of MIMO system, based on the actual situation level of the system as the criterion, 30 sets of experimental simulation data are used to predict the next moment situation level of the MIMO system. In order to verify the stability and validity of the model, MATLAB is used to simulate the experiment. The results show that the mean square error RMSE of different iterations is kept below 0.02, and the TSQ values of both sides are kept below 0.16. The overall prediction effect of the model was good, indicating that the system could provide effective decision support for 5G MIMO security situation prediction

Background and Aims In recent years, it has been found that targeted mRNA detection of sediment cells in urine can be used as novel biomarkers for the diagnosis of diabetic nephropathy (DN). However, its value in predicting the progression of DN is not clear. The purpose of this study is to seek for urinary mRNA markers that evaluate the prognosis of DN through systems biological screening, clinical verification and prospective studies. Method GEO database and “Nephroseq” platform were searched, and the transcriptome data of DN glomeruli and tubules and their clinical information were obtained. Weighted gene co-expression network analysis (WGCNA) combined with gene ontology (GO) annotation and KEGG pathway enrichment analysis were used to screen the hub genes negatively related to eGFR, and the hub genes were used as candidate markers for mRNA detection in urine sediment in DN patients. A total of 91 patients with DN diagnosed by renal biopsy were included, and 60 patients with type 2 diabetes and 61 healthy people were selected as the control groups. The mRNA expression of candidate molecules was detected by Taqman probe quantitative PCR, and the correlation between mRNA expression and eGFR and urinary protein levels were analyzed. Patients with DN were followed up for a median time of 21 months, and the primary end point was defied as end stage renal disease or eGFR decreasing by more than 50%. Multivariate Cox regression was used to evaluate the value of mRNA in predicting DN progression. Results GSE30528 and GSE30529 datasets were selected for analysis, including mRNA expression data of 9 cases of DN and 13 cases of normal glomeruli; and 10 cases of DN and 12 cases of normal tubules respectively. The clinical data of the patients in this study, including gender, race, age and eGFR, were searched on the Nephroseq platform. The gene modules negatively related to eGFR were screened by WGCNA. GO and KEGG analysis showed that the main function of the gene modules in both datasets were related to the activation of inflammatory cells and chemokines pathway. Through the screening of hub genes and the comparison of expression levels, CCL5, CXCL1, CXCL6 and CXCL12 were finally obtained as candidate genes. Quantitative PCR showed that the levels of CCL5 and CXCL1 was significantly increased in DN group, CCL5 was negatively correlated with eGFR and positively correlated with urinary protein level, while CXCL1 was negatively correlated with eGFR, but had no significant correlation with urinary protein level. Multivariate Cox regression showed that eGFR, urinary protein level, degree of renal fibrosis and urinary CCL5 were independent risk factors of primary end point. Conclusion The activation of chemokine signal pathway in renal tissue is involved in the progression of DN. Urinary CCL5 mRNA can independently predict the prognosis of DN and may be served as a novel biomarker for the progression of DN.

Background and Aims Hypoxia inducible factor-prolyl hydroxylase inhibitor (HIF-PHI) is a novel small molecule inhibitor in clinical treatment for renal anemia. Several studies have shown that sustained HIF activation may also have deleterious effects, such as tubulointerstitial fibrosis. Here, the potential effects of different treatment frequency of HIF-PHI (FG-4592) on CKD (chronic kidney disease) mice were investigated. Method Male C57BL/6J mice were constructed by subtotal nephrectomy of 5/6 to serve as a model for CKD. We then compared four different strategies based on the frequency of FG-4592(intragastric administration, 30mg/kg) over 9 weeks: once a week (qw), twice a week (biw), three times a week (tiw), and every day (qd). In vitro, HK-2 cells were divided into three groups according to the different administration methods of FG-4592(30μM): FG-4592 for 20h (FG0); FG-4592 for 8h, followed by washing with medium for 12h(FG1); twice FG-4592 for 8h, followed by washing twice with medium for2h(FG2). Results Hemoglobin in the biw group, tiw group or qd group was significantly higher than that in vehicle group. EPO, HIF-1α and HIF-2α of tiw group, biw group and qd group were significantly higher than those in vehicle group. Makers of fibrosis and inflammation in qd group were higher than that in vehicle group, while there was no statistical difference in the changes of the above indicators in tiw group and biw group. HIF-1α and HIF-2α were increased in intermittent administration, but the expression of VEGF, makers of fibrosis or inflammation were not affected. In HK2, HIF-1α and HIF-2α were activated by continuous administration of FG-4592. At the same time, the expression of VEGF, inflammatory and fibrosis indicators were increased significantly during continuous administration. In HepG2, EPO was increased in intermittent administration in HepG2 cells without VEGF increasing. Conclusion Taken together, our studies demonstrated that FG-4592 administration 2-3 times a week can effectively improve renal anemia without side effects on inflammatory and fibrosis.

Despite the small share in total energy investments, energy investments in research and development (R&D) incubate early-stage energy technology, improve energy efficiency in the manufacturing processes, and facilitate the expansion of green energy systems. This chapter distinguishes R&D investments in energy efficiency from other energy R&D investments. Our research formulates the way in which R&D investments in energy efficiency enhance energy production through efficiency knowledge accumulation. Our research explores how R&D investments in energy efficiency affect the other types of energy R&D investments, total energy R&D investments, and the share of energy R&D investments in the capital goods sector. Also, the study examines the impact of R&D investments in energy efficiency on economic welfare, economic structure, carbon taxes, energy transition, carbon emissions, and climate change. The study simulates three emissions abatement scenarios: an optimal abatement scenario, a 2 °C policy scenario, and a 1.5 °C policy scenario. Our study provides a reference level for the amount of R&D investment in energy efficiency. Policy makers may overinvest in other energy R&D investments under all abatement scenarios if they overlook R&D investments in energy efficiency. R&D investments in energy efficiency also bring considerable economic gains, however, its climate impact is not significant.

In recent years, the brain-computer interface (BCI) has made great progress, especially in steady-state visual evoked potentials (SSVEP) because of its high recognition rate and information transfer rate (ITR), which has received extensive research attention. This paper proposes a feature recognition based on extended canonical correlation analysis (eCCA) method, and adds the phase characteristic of canonical correlation analysis sine and cosine signal to electroencephalogram(EEG) signal for reference. Compared with task-related component analysis and eCCA methods, research shows that this method can strengthen the property of BCI based on SSVEP.

Using extracellular vesicles for targeted delivery of IL-10 is an efficient therapeutic strategy against ischemic AKI., Recently, extracellular vesicles (EVs) have been attracting strong research interest for use as natural drug delivery systems. We report an approach to manufacturing interleukin-10 (IL-10)–loaded EVs (IL-10+ EVs) by engineering macrophages for treating ischemic acute kidney injury (AKI). Delivery of IL-10 via EVs enhanced not only the stability of IL-10, but also its targeting to the kidney due to the adhesive components on the EV surface. Treatment with IL-10+ EVs significantly ameliorated renal tubular injury and inflammation caused by ischemia/reperfusion injury, and potently prevented the transition to chronic kidney disease. Mechanistically, IL-10+ EVs targeted tubular epithelial cells, and suppressed mammalian target of rapamycin signaling, thereby promoting mitophagy to maintain mitochondrial fitness. Moreover, IL-10+ EVs efficiently drove M2 macrophage polarization by targeting macrophages in the tubulointerstitium. Our study demonstrates that EVs can serve as a promising delivery platform to manipulate IL-10 for the effective treatment of ischemic AKI.

Background and Aims The fact that activation of the innate immune system and chronic inflammation are closely involved in the pathogenesis of diabetic Kidney disease (DKD). Recent studies have suggested the inflammatory process plays a crucial role in the progression of DKD. Identifying novel inflammatory molecules closely related to the decline of renal function is of significance in diagnosing and predicting the progression of DKD. The weighted gene co-expression network analysis (WGCNA) algorithm represents a novel systems biology method that provide the approach of association between gene modules and clinical traits to find the genes involvement into the certain phenotypic trait. The goal of this study was to identify hub genes and their roles in DKD from the gene sets associated with the decline of renal function by WGCNA. Method The Gene Expression Omnibus (GEO) database and “Nephroseq” website were searched and transcriptome study from DN biopsies with well-established clinical phenotypic data were selected for analysis. Next, we constructed a weighted gene co-expression network and identified modules negatively correlated with eGFR by WGCNA in the data of glomerular tissue. Functional annotations of the genes in modules negatively correlated with eGFR were analyzed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. Through protein-protein interaction (PPI) analysis and hub gene screening, the hub genes were obtained. Furthermore, we compared the expression level of hub genes between DKD and normal control and drew ROC curves to determine the diagnosis value to DKD of these genes. Results The microarray-based expression datasets GSE30528 were screened out for analysis, which included glomeruli tissue of 9 cases of DKD and 13 cases of control. This microarray platform represented the transcriptome profile of 12411 well-characterized genes. Using WGCNA, a total of 19 gene modules were identified. Then module eigengene were analyzed for correlation with clinical traits of age, sex, ethnicity and eGFR and the “MEhoneydew1” module showed negative associated with eGFR (r=-0.58). GO functional annotation showed that these 551 genes in the “MEhoneydew1” module mainly enriched in the T cell activation. KEGG annotation showed mainly enriched in chemokine signaling pathway. Except for C3, top 10 hub genes, CCR5, CXCR4, CCR7, CCL5, CXCL8, CCR2, CCR1, CX3CR1, C3AR1 and C3, are all members of chemokines or chemokine receptors. Furthermore, we compared the expression level of these 9 genes between DKD and control, and found that all of these 9 genes increased in the DKD group, and the differences of 6 genes, CCR5, CCR7, CCL5, CCR2, CCR1, C3AR1, were of statistical significance. Linear correlation analysis showed that the expression of these 6 genes was negatively correlated with eGFR, and the ROC curve showed that the area under the curve could reach 0.812∼1.0. Conclusion We identified a panel of 6 hub genes focused on chemokines and chemokine receptors critical for decline of renal function of DKD using WGCNA. These genes may serve as biomarkers for diagnosis/prognosis and as putative novel therapeutic targets for DKD.

Identifying novel molecules that are involved in the pathogenesis of DN has both diagnostic and therapeutic implications. The gene co-expression network analysis (WGCNA) algorithm represents a novel systems biology approach that provide the approach of association between gene modules and clinical traits to find the module involvement into the certain phenotypic trait. The goal of this study was to identify hub genes and their roles in DN from the aspect of whole gene transcripts analysis. Method Various types of chronic kidney diseases (CKD), including DN, microarray-based mRNA gene expression data, listed in the Gene Expression Omnibus (GEO) database, were analyzed. Next, we constructed a weighted gene co-expression network and identified modules distinguishing DN from normal or other types of CKD by WGCNA. Functional annotations of the genes in modules specialized for DN were analyzed by Gene Ontology (GO) enrichment analysis. Through protein-protein interaction (PPI) analysis and hub gene screening, the hub genes specific for DN were obtained. Furthermore, we drew ROC curves to determine the diagnosis and differential diagnosis value to DN of hub genes. Finally, another study of microarray in the GEO database was selected to verify the expression level of hub genes and in the “Nephroseq” database, the correlation between the gene expression level and eGFR was analyzed. Results “GSE99339”, glomerular tissue microarray in 187 patients with a total of 10947 genes, was selected for analysis. After excluding the inappropriate cases, a total of 179 specimens were analyzed, including 14 cases of DN, 22 cases of focal segmental glomerulosclerosis (FSGS), 15 cases of hypertensive nephropathy (HT), 26 cases of IgA nephropathy (IgAN), 13 cases of minimal change disease (MCD), 21 cases of membranous nephropathy (MGN), 23 cases of rapidly progressive glomerulonephritis (RPGN), 30 cases of lupus nephritis (LN) and 14 cases of kidney tissue adjacent to tumor. Co-expression network analysis by WGCNA identified 23 distinct gene modules of the total 10947 genes and revealed “MEsaddlegreen” module was strongly correlated with DN (r=0,54), but not with other groups. GO functional annotation showed that these 64 genes in the “MEsaddlegreen” module mainly enriched in the deposition of extracellular matrix, which represents the specific and diagnostic pathophysiological process of DN. Further PPI and hub gene screening analysis revealed that LUM, ELN, FBLN1, MMP2, FBLN5 and FMOD can be served as hub genes, which had been proved to play an important role in the deposition of extracellular matrix. Furthermore, we found that the expression of hub genes was the highest in DN group and for the diagnosis value of DN by each gene, the area under the ROC curve is about 0.75∼0.95. The external verification of another study showed that compared with the normal control group, the expression of these hub genes was the highest in the DN group, and their expression level was negatively correlated with eGFR. Conclusion Using WGCNA and further bioinformatics approach, we identified six hub genes that appear to be identical to DN development. As such, they may represent potential diagnostic biomarkers as well as therapeutic targets with clinical utility.

A ternary nanocomposite containing ZnO, ZnS and MoS2 (ZnMoNCs) has been firstly synthesized by Keggin-type polyoxomolybdate and ZIF-8 as starting materials to provide Mo and Zn sources simultaneously. As compared to the mono- or bi-component complex, ZnMoNCs present more sensitive and stable surface-enhanced Raman scattering (SERS) activity. When rhodamine 6G is used as a probing molecule, the detection limit of 10−9 mol L−1 can be achieved on the ZnMoNC substrate with the enhancement factor (EF) of 1.4 × 108, which is the highest sensitivity among the known semiconductor SERS substrates. The increased adsorption sites and the synergistically improved charge transfer are conducive to the large enhancement of the SERS activity. It was also proved that the ZnMoNC substrate possesses high stability and self-cleaning properties, and can be reused in SERS detection without losing its activity even after 5 cycles. This work paves a new way to the design of new semiconductor SERS substrates by the utility of a multicomponent complex.

Objective The aim of this study was to characterize the patterns and treatment of disease recurrence in patients achieving a pathological complete response (pCR) following neoadjuvant chemoradiation for advanced pancreatic ductal adenocarcinoma (PDAC). Summary of background data A pCR is an independent predictor for improved survival in PDAC. However, disease recurrence is still observed in these patients. Methods Patients with advanced PDAC who were treated with neoadjuvant therapy and had a pCR were identified between 2009 and 2017. Overall survival (OS) was determined from the initiation of neoadjuvant, disease-free survival (DFS) from the date of surgery, and post-recurrence survival (PRS) from the date of recurrence. Factors associated with recurrence were analyzed using a Cox-regression model. Results Of 331 patients with borderline resectable or locally advanced PDAC, 30 achieved a pCR following neoadjuvant treatment and pancreatectomy. The median DFS for pCR patients was 29 months and OS 76 months. Recurrence was observed in 14 patients. No clinicopathologic or treatment characteristics were associated with survival. The median PRS following recurrence was 25 months. Treatment following recurrence included chemotherapy, radiation or ablation, and surgical resection. Hepatectomy or completion pancreatectomy was accomplished in 2 patients that remain alive 13 and 62 months, respectively, following metastasectomy. Conclusions A pCR following neoadjuvant therapy in patients with advanced PDAC is associated with remarkable survival, although recurrence occurs in about half of patients. Nevertheless, patients with pCR and recurrence respond well to treatment and survival remains encouraging. Advanced molecular characterization and longitudinal liquid biopsy may offer additional assistance with understanding tumor biologic behavior after achieving a pCR.

Radical antegrade modular pancreatosplenectomy (RAMPS) has been adopted by some surgeons in the treatment of left-sided pancreatic cancer (PDAC). Low disease incidence and heterogenous disease biology make robust prospective comparison of RAMPS and standard distal pancreatosplenectomy (DPS) difficult.Consecutive cases of chemo-naïve patients undergoing open RAMPS and DPS for PDAC between 2010-2017 at two international high-volume pancreatectomy centers were compared. Cox proportional hazard modeling was utilized for multivariate analysis.We identified 193 DPS and 253 RAMPS during the study period. DPS was associated with higher rates of median estimated blood loss (500 vs. 300 cc, P0.001), median total harvested lymph nodes (18 vs. 12, P0.001) and R0 resection (94.3% vs. 88.9%, P=0.013). There were no differences in rates of postoperative pancreatic fistula (16.5% vs. 17.8%, P=1) or postoperative hemorrhage (5.9% vs. 3.6%, P=0.385) (DPS vs. RAMPS). After controlling for significant clinical pathological parameters, RAMPS was associated with non-superior recurrence-free survival (RFS) (HR 0.29; 95% CI, 0.07-1.27, P=0.101) and overall-survival (HR 1.03; 95% CI, 0.71-1.49, P=0.895) compared with DPS. Similar results were observed in node-positive patients.RAMPS is safe and effective in the treatment of PDAC, but is not associated with an improvement in either RFS or overall-survival over DPS.

Herpes simplex virus type 1 (HSV-1) is a leading cause of infectious blindness. Current treatments for HSV-1 do not eliminate the virus and are incapable of modulating the virus reservoir. Here, we target HSV-1 genome directly using mRNA-carrying lentiviral particle (mLP) that simultaneously delivers spCas9 mRNA and two viral genes-targeting gRNAs (designated HSV-1-erasing lentiviral particles, HELP). We showed HELP efficiently blocked HSV-1 replication in both acute and recurrent infection models, and prevented occurrence of herpetic stromal keratitis (HSK). We further showed retrograde transportation of HELP from corneas to trigeminal ganglia (TG) where HSV-1 established latency and found evidence of HELP modulating herpes reservoir. Additionally, the potent antiviral activity of HELP was also replicable in human-derived corneas. These results strongly support clinical development of HELP as a new antiviral therapy and may accelerate mRNA-based CRISPR therapeutics.

In the operating room (OR), ventilation and thermal plumes can affect the distribution of bacteria-carrying particles (BCPs). Especially in the operating microenvironment, the development of human thermal plumes cannot be ignored. Therefore, in this study, six different air supply velocities were used to study human thermal plumes and the diffusion of BCPs. The numerical simulation based on computational fluid dynamics was verified by experiments, and the realizable k-e turbulence model and Lagrangian particle tracking model were used. The results show that the interaction of the thermal plumes and ventilation significantly affected the diffusion of BCPs in the operating area, which cannot be ignored. The maximum thermal plumes in the ORs are markedly higher than those in other rooms. The strong thermal plumes caused BCPs to reach the operating area and infect the wound of the patient. For most ORs with a vertical unidirectional air supply, ventilation and thermal plumes achieve a good offset when the air supply velocity reaches 0.25 m/s. In addition, increasing the air supply velocity alone cannot effectively control the effect of thermal plumes on BCPs. This study can provide a practical guideline for controlling the diffusion and distribution of BCPs in ORs.

Bus has become an integral traffic mode for urban residents along with the development of public transport system; and as a kind of public service, urban public transport is the basic infrastructure closely related to people, and is also required to constantly improve its service levels to better serve people. Therefore, how to evaluate its service level has become one of the important projects that need to be studied. Based on establishing the mathematical model of fuzzy comprehensive evaluation and illustrated by the case of Xi’an, this paper verifies the model rationality, assesses the bus development level in Xi’an, and further puts forward countermeasures for its bus services according to the results.

Research into detection of dense communities has recently attracted increasing attention within network science, various metrics for detection of such communities have been proposed. The most popular metric -- Modularity -- is based on the so-called rule that the links within communities are denser than external links among communities, has become the default. However, this default metric suffers from ambiguity, and worse, all augmentations of modularity and based on a narrow intuition of what it means to form a "community". We argue that in specific, but quite common systems, links within a community are not necessarily more common than links between communities. Instead we propose that the defining characteristic of a community is that links are more predictable within a community rather than between communities. In this paper, based on the effect of communities on link prediction, we propose a novel metric for the community detection based directly on this feature. We find that our metric is more robustness than traditional modularity. Consequently, we can achieve an evaluation of algorithm stability for the same detection algorithm in different networks. Research Purpose: To explore the role of Internet platform in clinical microbiology laboratory practice teaching reform, so as to improve students' comprehensive quality and be competent for future clinical laboratory work. Proposed Methods: Students were randomly divided into two groups, control group and reform group. On the basis of traditional teaching methods, the reform group added the network-based hybrid teaching method, while the control group adopted the traditional teaching method. The theoretical and clinical skills of the two groups were compared. Results: The score of clinical microbiological examination skill and theoretical examination in the reform group were higher than those in the control group, and the difference was statistically significant. Conclusion: In the era of "Internet +", integrating network based blended teaching mode into clinical microbiology examination teaching practice is not only conducive to students to expand and enrich their theoretical knowledge, but also greatly improve their clinical practice skills, laying a good foundation for their smooth work in the future.

Background: Recently, cinacalcet (CINA) has been shown to be effective for attenuating bone loss in the treatment of secondary hyperparathyroidism (SHPT) in patients with chronic kidney disease (CKD), which might be associated with the reduction in serum parathyroid hormone (PTH) levels. However, the exact mechanism is largely unclear. Emerging studies have revealed that an increased number of bone marrow adipocytes (BMAs) are involved in bone loss and the endothelial-to-adipocyte transition via the endothelial-to-mesenchymal transition (EndMT) might play a key role in this pathological process. Here, we assessed whether CINA could attenuate bone loss via inhibiting endothelial-to-adipocyte transition in CKD rats. Methods: A rat model of CKD was induced by adenine and a high phosphorus diet. CINA was orally administrated to CKD animals (10 mg/kg once a day). Dual energy X-ray absorptiometry, micro-computed tomography, bone histomorphometry, and bone mechanical tests were used to determine the skeletal changes. The bone marrow expression of EndMT markers was also examined. The effect of elevated PTH levels on the endothelial-to-adipocyte transition was studied in endothelial cells (ECs). Results: Elevation of serum PTH levels, remarkable bone loss and increased numbers of BMAs were observed in rats with CKD compared with the controls, and these changes were attenuated after treatment with CINA. Furthermore, the CINA treatment abolished the upregulation of mesenchymal markers (FSP1 and α-SMA) and the downregulation of an endothelial marker (CD31) in bone tissues from rats with CKD. The serum PTH concentrations were correlated with the bone marrow protein levels of these EndMT-related proteins. An in vitro treatment in ECs demonstrated that PTH induced the EndMT in a concentration- and time-dependent manner. Accordingly, ECs treated with PTH exhibited adipogenic potential following growth in adipogenic culture medium. Conclusions: Our study indicated CINA treatment attenuated bone loss in CKD rats, which might be associated with inhibiting PTH-induced endothelial-to-adipocyte transition in CKD rats.

To investigate the long-term clinical effect of balloon dilation eustachian tuboplasty (BET) combined with tympanic tube insertion (TTI) in the treatment of chronic recurrent secretory otitis media (CRSOM). A retrospective study of 30 cases of CRSOM treated with BET combined with TTI under general anesthesia between August 2014 and September 2016. Thirty cases of CRSOM treated with TTI in the same period were taken as the control group. All cases were followed over 24 months. The scores of eustachian tube (ET) function preoperation, 1 month, 6 months, 12 months, and 24 months postoperation were collected and analyzed, respectively. A satisfaction questionnaire was used to evaluate the therapy at 24-months postoperation. The symptoms were significantly improved and the ET score was obviously increased postsurgery in most cases treated with BET plus TTI compared with those treated with TTI alone. The highest ET score was obtained at 6 months post BET. Five (14%) cases (6 ears) of CRSOM recurred. The 24-month postoperation follow-up questionnaire showed that 84.6% of the patients were satisfied with the treatment, while ten cases (25%) in the TTI group recurred. BET combined with TTI surgery is an effective therapy for patients with CRSOM.

In this study, Au deposited TiO2 (Au-TiO2) nanocomposites were prepared by photocatalytic reduction of Au species (HAuCl4) on TiO2 nanoparticles (NPs) and served as efficient SERS-active substrates. The observed considerable SERS enhancement of 4-MBA molecules on Au-TiO2 NPs compared with that on pure TiO2 NPs can be attributed to the synergistic contribution of semiconductors and metals by the charge-transfer mechanism from the substrate to the molecule. In the preparation of Au-TiO2 nanocomposites, the photo-reduction time and concentration of HAuCl4 have an important influence on the SERS enhancement effect of the composite substrate. Moreover, for the recycle of substrates, the photocatalytic experiment of Au-TiO2 nanocomposites was carried out. The results show that these nanocomposite substrates are characterized by a self-cleaning property and can be repeatedly used in SERS experiments without losing their activity and with high stability.

AFLS (Adaptive front lighting system) is being applied to improve safety in driving automotive at night. Safe embedded system design for controlling head-lamps is required to improve noise robust ECU hardware and software simultaneously by considering safety requirement of hardware-dependent software under severe environ-mental noise. In this paper, we propose an adaptive headlight controller with a newly-designed symmetric angle sensor compensator, especially based on the proposed steering-swivel angle lookup table to determine whether the current controlling target is safe. The proposed system includes an additional backup hardware to compare the system status and provides safe swivel-angle management using a controlling algorithm based on the pre-defined lookup table (LUT), which is a symmetric mapping relationship between the requested steering angle and expected swivel angle target. The implemented system model shows that the proposed architecture effectively detects abnormal situations and restores safe status of controlling the light-angle in AFLS operations under severe noisy environment.

Serine/threonine kinase 33 (STK33) is a novel protein that has been the focus of an increasing number of studies in recent years; however, the role of STK33 in tumorigenesis remains controversial. Previous studies have demonstrated that STK33 is overexpressed in several human cancers and exerts a pro-tumorigenic effect through the promotion of cell proliferation. However, the role of STK33 in colorectal cancer (CRC), which is one of the most aggressive human malignancies, remains unclear. The aim of the current study was to investigate the methylation status of STK33 in CRC and to determine its clinical significance. The results demonstrated that STK33 was hypermethylated in CRC cell lines and promoted the proliferation of CRC cells. In addition, the methylation status and expression of STK33 in 94 pairs of cancer and noncancerous tissues obtained from patients with CRC was investigated. The increased flow cytometry enumeration of peripheral blood circulating CD34+ cells in patients with acute leukemia has been found in our previous work. In this study, we also demonstrated that acute promyelocytic leukemia (APL) patients not only had elevated CD34+ cell count, but also had some clinical features.Fifty APL patients and 19 healthy volunteers were included in the study. The enumeration of circulating CD34+ cells, cytogenetic subgroup, immunophenotype analysis, and leukemic-related gene mutation detection were performed.Some APL patients with higher count of CD34+ cells (≤10 × 10(6)/l) usually possessed one or more poor prognostic factors (higher WBCs count, PML/RARa gene complex fusion, chemotherapy-related APL, normal karyotype/complex karyotype abnormalities, CD56/CD34 antigen positive expression, FLT3-ITD positive mutation, myelofibrosis, and marrow necrosis). A cut-off value of 10 × 10(6)/l CD34+ cells may have the power to distinguish APL patients with above adverse clinical prognostic factor from other APL subjects.The circulating CD34+ cell count appears to increase in some APL patients and a higher CD34+ cell count may be indicative of inferior survival and serve as an adverse biomarker for APL.

The copyrolysis and cocombustion behaviors of Pingshuo coal and the biomasses (sawdust and rice straw) have been investigated using a thermogravimetric analyzer. The experimental results indicate that there exist synergetic effects between the biomasses and Pingshuo coal during their coconversion process. The initial temperature of volatile emission from Pingshuo coal and the temperature corresponding to the maximum conversion rate during the copyrolysis change with the biomass mixture ratio. Moreover, it can be deduced from the comparison between the experimental and the calculated DTG curves that the copyrolysis process is not the sum of Pingshuo coal and the biomass conversion. Background and Objective: Radiation usually results in paranasal sinusitis in patients with nasopharyngeal carcinoma (NPC), which influences patients' quality of life. This study aimed to determine the relationships between dose distribution in the nasal cavity and nasal mucous injury in patients with NPC treated by intensity-modulated radiation therapy (IMRT), and to find the tolerable radiation dose for the nasal mucous. Methods: Sixty-six patients with NPC treated by IMRT between October 2006 and November 2008 were enrolled. The irradiation dose in the nasal cavity was determined by the computer with the IMRT work platform. Mucociliary transport rate (MTR) was detected by modified saccharine test before IMRT, at the end of IMRT, and at 3, 6, and 12 months after IMRT. Results: The data were available for 129 nasal cavities. The cavities receiving a mean dose below or equal to 37 Gy showed substantial preservation of nasal mucous after IMRT. The MRT decreased to (62.82±38.59)%, (56.78±37.79)%, (64.05±39.37)%, and (71.13±39.55)% of pre-IMRT value at 4 time points after IMRT, with significant differences among the data (P＜0.05). In contrast, when the cavities received a mean dose higher than 37Gy, no significant differences in MTR among the time points were observed. At 3 months after IMRT, the MTR was the lowest (38.27% of pre-RT value). Conclusions: A mean radiation dose of 37 Gy for the nasal cavity is an optimal dose to protect the nasal cavity function.

Objective— Increasing evidence shows that autoimmune response contributes importantly to pathogenesis of abdominal aortic aneurysm (AAA). This work was aimed to assess the possibly altered function of peripheral CD4 + CD25 + T regulatory cells (Tregs) that might breakdown immunologic self-tolerance in AAA patients. Methods and Results— Peripheral blood from 22 AAA patients, 11 patients with abdominal aortic atherosclerotic occlusive disease (AOD), and 32 healthy controls (HCs) was analyzed to determine the percentage of CD4 + CD25 + Tregs in the total CD4 + T-cell population and FOXP3 expression by means of flow cytometry. The frequencies of the CD4 + CD25 + Treg population were not significantly different between groups (AAA, 5.69±0.99%; AOD, 5.52±1.13%; HC, 5.88±1.55%; P >0.05). However, the frequency of CD4 + CD25 + FOXP3 + T cells in AAA patients (2.45±0.57%) was significantly lower than that in AOD group (3.41±0.72%; P P + CD25 + Tregs from AAA group than either of other 2 groups ( P + CD25 + Tregs from patients with AAA exhibited significantly less suppressive activity than those from AOD patients or HCs ( P + CD25 + T cells and CD4 + CD25 − T cells from AAA patients and HCs demonstrated that the primary regulatory defect is due to a dysfunction of CD4 + CD25 + Tregs, and not a resistance of CD4 + CD25 − responder T cells to suppression in AAA patients. Conclusion— Our data demonstrate a reduced level of FOXP3 expression in peripheral CD4 + CD25 + Tregs and decreased frequency of CD4 + CD25 + FOXP3 + T cells in a cohort of AAA patients enrolled in the study, which leads to a functional deficiency of CD4 + CD25 + Tregs as a whole. This indicates an impaired immunoregulation by Tregs that may contribute to AAA pathogenesis.

Steering angle-based adaptive headlight controller for safe operation in emergency situations is proposed by adopting the compensation method of the symmetric two angle sensor data. Adding the intelligence in automotive applications results in increased chance of unsafe operations, requiring the integration of additional safety functions. In this paper, we introduce our efforts to integrate the safety observer with the adaptive headlight controller, still providing safe angle control in an abnormal situations by the steering angle. If the symmetric behavior of two angles is broken in the unknown interruption, the backup angles are restored into the active angle control value.

Pulse-based charging method for battery cells has been recognized as a fast and efficient way to overcome the shortcoming of slow charging time in distributed battery cells. The pulse frequency for controlling the battery charge will change within a certain range. The optimal frequency is determined to achieve the minimized battery impedance. The adaptation of a duty cycle of the pulse by decreasing the concentrated polarization guarantees a safe charging operation of all the distributed cells within a temperature range of 5°C to 45°C. The proposed charging process can be completed to about 80% of its maximum capacity in less than 20 minutes without damaging the battery characteristics. The newly designed charging results show that about two times faster than the conventional constant-current constant-voltage (CC-CV) charging method and 52% more efficient than the constant current fast charging method. The implemented high fidelity electrical model provides an approach looking into the effect of the pulse on the internal electrochemical reaction process. In addition, cell terminal voltage and temperature are simultaneously monitored in real time so as to adjust the frequency and duty of the proposed charging pulse method preventing battery degradation.

Applications and networking are the key functions for connected vehicle. And applications of In-Vehicle Infotainment (IVI) system are one of the important characteristics in the automotive industry. Because of the fast evolution of electronic technologies, automotive industry cannot adopt new technology easily. IVI platform standard was developed by automotive OEM and suppliers but the problems still exist. In this paper, we propose an implementation technique of applications that are based on HTML5 and a simple way to access the mobile network. XML-based configuration is used to represent the capability of the vehicle, IVN signals of the production vehicle is used to validate the interaction between IVI system and vehicle. The application works well with native application APIs and HTML5 APIs as well as interact with vehicle and mobile network.

A secure microcontroller architecture has been proposed to protect the chip-level intrusion by injecting invalid firmware. The safe execution of the embedded firmware in microcontrollers is becoming important for applications requiring safety-critical operations, such as a safety-critical automotive controller, human-interactive interface. The chip-level instruction injection to bypass the original execution flow is a powerful method to acquire the privileged authority of firmware-controlled embedded systems. A code memory integrity validation accelerator for firmware authentication is proposed to provide fast hierarchical identification. The proposed validator, which is based on sector-segmented padding information, is integrated as a co-processor with the main CPU. The cyclic-redundancy code (CRC) as a fingerprint of the original code is inserted into the on-chip fab-programmed secure region to provide the sector-segmented code validation in long-range address space. The error-correcting code to identify invalid region in detail are applied to 64KB flash memory. The customized 8051-based microcontroller with the proposed accelerator is synthesized with 25,000 2-input NAND gates on 0.18um CMOS embedded-flash process.

Renal fibrosis is a histological outcome of chronic kidney disease (CKD) progression. However, the noninvasive detection of renal fibrosis remains a challenge. Here we constructed a renal fibrosis target mRNA array and used it to detect urinary mRNAs of CKD patients for investigating potential noninvasive biomarkers of renal fibrosis. We collected urine samples from 39 biopsy-proven CKD patients and 11 healthy controls in the training set. Urinary mRNA profiles of 86 genes showed a total of 21 mRNAs that were differentially expressed between CKD patients and controls ( P < 0.05), and vimentin (VIM) mRNA demonstrated the highest change fold of 9.99 in CKD vs. controls with robust correlations with decline of renal function and severity of tubulointerstitial fibrosis. Additionally, VIM mRNA further differentiated patients with moderate-to-severe fibrosis from none-to-mild fibrosis group with an area of the curve of 0.796 ( P = 0.008). A verification of VIM mRNA in the urine of an additional 96 patients and 20 controls showed that VIM is not only well correlated with renal function parameters but also correlated with proteinuria and renal fibrosis scores. Multiple logistic regression and receiver-operating characteristics analysis further showed that urine VIM mRNA is the best predictive parameter of renal fibrosis compared with estimated glomerular filtration rate, serum creatinine, and blood urea nitrogen. In addition, there is no improved predictive performance for the composite biomarkers to predict renal fibrosis severity compared with a single gene of VIM. Overall, urinary VIM mRNA might serve as a novel independent noninvasive biomarker to monitor the progression of kidney fibrosis.

Focused crawler is the core of the focused search engine, and the POI-oriented user need is a kind of new focused object which has not been well solved in previous studies. In this paper, we design and realize a POI-oriented focused crawler. The proposed focused crawler adopts classifiers to make relevant judgment and considers both current page’s relevance and the URL link information to make the URLs’ priority judgment. Experiments were conducted with two kinds of classification algorithms of Naive Bayes (NB) and Support Vector Machines (SVMs) on four sites, respectively. Experimental results show that the focused crawler with NB classifier obtains the average harvest of 95.97%, higher than the one with SVMs by 45.53%, but the focused crawler with SVMs attains the higher recall.

The pulse-based charging method for battery cells has been recognized as a fast and efficient way to overcome the shortcoming of a slow charging time in distributed battery cells, which is regarded as a connection of cells such as the Internet of Things (IoT). The pulse frequency for controlling the battery charge duration is dynamically controlled within a certain range in order to inject the maximum charge current into the battery cells. The optimal frequency is determined in order to minimize battery impedance. The adaptation of the proposed pulse duty and frequency decreases the concentration of the polarization by sensing the runtime characteristics of battery cells so that it guarantees a certain level of safety in charging the distributed battery cells within the operating temperature range of 5-45 degrees C. The sensed terminal voltage and temperature of battery cells are dynamically monitored while the battery is charging so as to adjust the frequency and duty of the proposed charging pulse method, thereby preventing battery degradation. The evaluation results show that a newly designed charging algorithm for the implemented charger system is about 18.6% faster than the conventional constant-current (CC) charging method with the temperature rise within a reasonable range. The implemented charger system, which is based on the proposed dynamic frequency and duty control by considering the cell polarization, charges to about 80% of its maximum capacity in less than 56 min and involves a 13 degrees C maximum temperature rise without damaging the battery.
