In contrast to mammals, early B cell differentiation and diversification of the antibody repertoire in chickens do not take place in the bone marrow but in a specialized gut associated lymphoid tissue (GALT), the bursa of Fabricius. During embryonic development, B cell precursors migrate to the bursa anlage, where they proliferate and diversify their B cell receptor repertoire. Around hatch these diversified B cells start to emigrate from the bursa of Fabricius and populate peripheral lymphoid organs, but very little is known how the migratory processes are regulated. As CXCL12 (syn. SDF-1) and CXCR4 were shown to be essential for the control of B cell migration during the development of lymphoid tissues in mammals, we analyzed expression and function of this chemokine/chemokine-receptor pair in the chicken bursa. We found a strong variation of mRNA abundance of CXCL12 and CXCR4 in different stages of bursa development, with high abundance of CXCL12 mRNA in the bursa anlage at embryonic day 10 (ED10). In situ hybridization demonstrated disseminated CXCL12 expression in the early bursa anlage, which condensed in the developing follicles and was mainly restricted to the follicle cortex post-hatch. Flow cytometric analysis detected CXCR4 protein already on early B cell stages, increasing during bursal development. Post-hatch, a subpopulation with the hallmarks of emigrating B cells became detectable, which had lower CXCR4 expression, suggesting that downregulation of CXCR4 is necessary to leave the CXCL12-high bursal environment. In vivo blockade of CXCR4 using AMD3100 at the time of B cell precursor immigration strongly inhibited follicle development, demonstrating that CXCL12 attracts pre-bursal B cells into the bursal anlage. Glycyrrhizinate dipotassium (DG) is a widely used anti-inflammatory, antimicrobial and anti-viral agent isolated from the licorice root. This study aims to investigate the potential antiviral activity of DG and its underlying mechanisms of protection against infectious bursal disease virus (IBDV) in vivo. The international guiding principles for Biomedical Research Involving Animals were followed during animal experiment. Measured the indices of immune organs including spleen, thymus and bursa of Fabricius, level of anti-IBDV antibody (Ab), IFN-γ and IFN-β in serum through ELISA, lymphocyte proliferation and the ratio of CD4+/CD8+ in blood using MTT and flow cytometry respectively and the expression of IBDV VP1 and TLR3 host gene in bursa of Fabricius were also evaluated by qRT-PCR. The DG treatment groups showed increased immunological index of spleen, thymus and bursa of Fabricius, a much higher ratio of CD4+/CD8+, the levels of anti-IBDV Ab, IFN-γ and IFN-β in serum were up-regulated varyingly, inhibit the VP1 expression and also up-regulated the TLR3 expression in bursa of Fabricius. Background: The molecular mechanisms underlying stress-influenced immune function of chicken (Gallus Gallus) are not clear. The stress models can be established effectively by feeding chickens corticosterone (CORT) hormone. The bursa of Fabricius is a unique central immune organ of birds. RNA-Seq technology was used to investigate differences in the expression profiles of immune-related genes and associated pathways in the bursa of Fabricius to clarify molecular mechanisms. The aim of this study was to broaden the understanding of the stress-influenced immune function in chickens. Results: Differentially expressed genes (DEGs) in the bursa of Fabricius between experimental group (basal diet with added CORT 30 mg/kg; C_B group) and control group (basal diet; B_B group) were identified by using RNA-seq technology. In total, we found 1434 significant DEGs (SDEGs), which included 199 upregulated and 1235 downregulated genes in the C_B group compared with the B_B group. The immune system process GO term was the top significantly GO term, including MYD88, TLR4, IL15, VEGFA gene and so on. The cytokine-cytokine receptor interaction pathway and the Toll-like receptor signaling pathway were the key pathways affected by stress. The protein-protein interaction (PPI) analysis of the SDEGs showed that VEGFA, MyD88 and IL15 were hub genes and module analysis showed that MYD88, TLR4 and VEGFA play important roles in response to stress. Conclusion: This study showed that the VEGFA and ILs (such as IL15) via the cytokine-cytokine receptor interaction pathway, MYD88 and TLR4 via the Toll-like receptor signaling pathway may play important roles in the regulation of immune function under stress condition with CORT administration. Toll like receptor 4 (TLR4), eosinophils and mast cells play significant role in host immunity during several pathogenic infections. However in vivo tissue expression of TLR4 and distribution pattern of eosinophils and mast cells in chicken bursa of Fabricius (BF) during Salmonella enterica serovar Typhimurium (STm) infection is poorly studied. Therefore, herein, following immunostaining, we found localization of TLR4 in follicular cortex and medulla and its expression was statistical increased after 36 h and 72 h of STm stimulation. Chromotrope 2R staining revealed that eosinophils were mostly distributed in follicular cortex, inter-follicular spaces and in or around blood vessels and their number in BF were statistical increased after 72 h of STm stimulation. The presence of eosinophils was confirmed using immunostaining with anti-rabbit eosinophil cationic protein antibody. Toluidine blue stained mast cells were mostly distributed in connective tissues between inter-follicular spaces while some were also present in follicular cortex of BF. However, STm stimulation illustrated non-significant effect on the number of mast cells or their de-granulation, instead their number were gradually decreased in BF with advancement in age of chickens. The Harderian gland is a sparsely characterized immune tissue known to play an important role in local immunity. The function of the Harderian gland, however, is not clearly defined. Measuring the expression of all genes using RNA-seq enables the identification of genes, pathways, or networks of interest. Our relative RNA-seq expression analysis compared the chicken Harderian gland transcriptome to other important primary and secondary immune tissues including the bursa of Fabricius, thymus, and spleen of non-challenged birds. A total of 2,386 transcripts were identified as highly expressed in the Harderian gland. Gene set enrichment showed the importance of G-protein coupled receptor signaling and several immune pathways. Among the genes highly expressed in the Harderian gland were 48 miRNAs, a category of genetic elements involved in regulation of gene expression. Coronavirus infection delays the development of the cortico-medullary (CM) capillary network which results in retarded development of bursal follicles. The smaller size of the medulla in the coronavirus-infected birds may lead to a lower number of B lymphocytes and bursal secretory dendritic cells, which negatively affects the reactivity and efficacy of the immune system. Contrary to the wild-type infectious bronchitis virus (IBV) strain, infection induced by H120 vaccine virus exerts only a moderate influence on caveolin-1 expression of the CM capillary web and on follicular development compared to the untreated controls. The gross anatomical study of thymus and bursa of Fabricius was carried out on 30 Kadaknath birds of different age groups ranging from day old to 26 week. The birds were divided into five equal groups according to age as day old to 2 week, 2 to 4 weeks, 4 to 8 weeks, 8 to 16 weeks and 16 to 26 weeks, each consisted of six birds. The shape, colour and location were observed. The thymus gland in Kadaknath consisted of two long chains having 6-8 irregular shaped lobes. The shape and color of thymic lobes as well as length and width of thymus varied amongst groups. Grossly there was no regression of thymus after 26th week of age. The bursa of Fabricius was located on the dorsal wall of proctodeum of cloaca. The shape and color of bursa of Fabricius varied amongst age groups. Objective: The bursa of Fabricius (BF) is a central humoral immune organ belonging specifically to avians. Recent studies had suggested that miRNAs were active regulators involved in the immune processes. This study was to investigate the possible differences of the BF at miRNA level between two genetically disparate duck breeds. Methods: Using Illumina next-generation sequencing, the miRNAs libraries of ducks were established. Results: The results showed that there were 66 differentially expressed miRNAs and 28 novel miRNAs in bursa. A set of abundant miRNAs (i.e., let-7, miR-146a-5p, miR-21-5p, miR-17∼92) which are involved in immunity and disease were detected and the predicted target genes of the novel miRNAs were associated with duck high anti-adversity ability. By gene ontology analysis and enriching KEGG pathway, the targets of differential expressed miRNAs were mainly involved in immunity and disease, supporting that there were differences in the BF immune functions between the two duck breeds. In addition, the metabolic pathway had the maximum enriched target genes and some enriched pathways that were related to cell cycle, protein synthesis, cell proliferation and apoptosis. It indicted that the difference of metabolism may be one of the reasons leading the immune difference between the BF of two duck breeds. Conclusion: This data lists the main differences in the BF at miRNAs level between two genetically disparate duck breeds and lays a foundation to carry out molecular assisted breeding of poultry in the future. Embryonic tissues contain highly ramified stellate-shaped cells expressing CD45 and MHC II antigens but their origin and immunophenotype are unknown. Using staged avian embryos and cell-type-specific antibodies, we establish a detailed spatiotemporal ontogeny of cells that express CD45, the earliest marker of hematopoietic stem cells in the chick. CD45 immunostaining marks three distinct embryonic cell populations: round, ramified and amoeboid cells. The round and ramified CD45+ cells appear first in yolk-sac blood islands before the onset of circulation. A subpopulation of round cells co-expresses the thrombocyte-specific CD51/CD61 antigen. Amoeboid cells express macrophage-specific antigens and frequently occur in regions of apoptosis. Ramified cells are distributed uniformly in the embryonic mesenchyme, colonize lymphoid and non-lymphoid organs and later express MHC II. To study the origin of CD45+ cells, 2-day-old chick embryos were ablated from the yolk sac before the establishment of circulation and incubated for 2-5 days. Large numbers of CD45+MHC II+ ramified cells differentiated in the yolk sac. Yolk-sac chimeras were generated by grafting embryos into GFP-expressing de-embryonated yolk sacs. GFP/CD45 co-expressing ramified and amoeboid cells colonized all organ primordia in the donor embryo. We also recombined GFP+ yolk sac with the bursa of Fabricius and found ramified GFP+CD45+ cells in the bursa where they differentiated into dendritic cells. Thus, CD45 cells are first present in the yolk sac during primitive hematopoiesis and then migrate from the extra-embryonic yolk sac to give rise to cells throughout all organ primordia, including dendritic cells in the bursa of Fabricius. Background: The Toll-like receptor 4 (TLR4) pathway involves in the pathogen recognition and defense against infection in mammals. Considering that avian and mammalian TLR are differentially mediated, the action of a natural product on avian TLR4 pathway was unclear. High, medium and low doses of Astragalus polysaccharide (APS), were treated the chicken at 7-days-old age by gavage. The sIgA level in the intestinal fluid, the expression of chTLR4 mRNA/protein in bursa of Fabricius as well as the expression of downstream molecules of chTLR4 (chMyD88, chTRIF, chNF-κB, chIRF3, chIFN-β and chTNF-α) were measured on alternate days. Results: The content of sIgA and the chTLR4 mRNA expression/protein level were increased in non-dose-dependent manner after APS supplement. Also, the expressions of a subset of MyD88-independent pathway genes were more than MyD88-independent, in particular with low doses of APS supplement for 7 days. Conclusions: These suggest that administration of APS activates chTLR4 pathway in bursa of Fabricius in MyD88-independent pathway. We investigated lead (Pb)-induced oxidative stress and immune damage in the chicken bursa of Fabricius (BF) and the ameliorative effect of selenium (Se). Seven-day-old male chickens were randomly divided into four groups and were provided standard diet and drinking water, NaSeO added to the standard diet and drinking water, standard diet and (CHCOO)Pb added to drinking water, and NaSeO added to the standard diet and (CHCOO)Pb added to drinking water for 30, 60, and 90 days. The presence of Pb inhibited total antioxidant capacity (T-AOC), glutathione peroxidase (GPx), glutathione S-transferase (GST), superoxide dismutase (SOD), and catalase (CAT) activities; decreased glutathione (GSH) content; increased malondialdehyde (MDA) and hydrogen peroxide (HO) contents; inhibited interleukin (IL)-2 and interferon-γ (IFN-γ) messenger RNA (mRNA) expression; and increased IL-4, IL-6, IL-10, IL-12β, and IL-17 mRNA expression. The presence of Se relieved all of the above Pb-induced changes. There were close correlations among GSH, CAT, T-AOC, SOD, GPx, MDA, and HO and among IL-2, IL-4, IL-6, IL-12β, IL-17, and IFN-γ. The bursa of Fabricius, the central humoral immune organ unique to birds, plays an important role in B-lymphocyte differentiation. In order to gain a better understanding of the molecular mechanism of critical biological processes like Bcell immigration, differentiation, and final emigration, the transcriptional changes during embryonic and posthatch development of this organ were investigated. We generated a cDNA library from total RNA isolated from 3 representative developmental stages (embryonic day [ED] 10, posthatch d 2 and d 21). We generated over 70 million high-quality reads from the cDNA library by using deep sequencing. The uniquely mapped sequences of ED 10, d 2 and d 21 were 71087280, 59167491 and 70263675 respectively. All of the differential expressed genes were involved in Vitamin A metabolism, regulation of actin cytoskeleton, Wnt signaling pathway, MAPK signaling pathway, Jak-STAT signaling pathway, Notch signaling pathway, Toll-like receptor signaling pathway. The RNA-seq analysis provides a powerful method for analyzing the transcriptome and investigating the transcriptional changes of different development stages of bursa of Fabricius. The bursa of Fabricius (BF) is a lymphoepithelial organ found only in birds. Differences in morphology of BF could play an important role in immune response. The objective of this study was to investigate the histological and histochemical characteristics of the bursa of Fabricius in the long-legged buzzard (Buteo rufi nus). The material for the study comprised bursa samples obtained from three long-legged buzzards with permission of the General Directorate of Nature Protection and National Parks (Ankara, Turkey). Briefl y, interfollicular epithelium (IFE) was shown to be columnar in shape and not to contain goblet cells. Reticular fi bers were located in interfollicular septae. Each lymphoid follicle in the bursa of Fabricius in the long-legged buzzard was remarkably linked to the follicle associated epithelium (FAE). Namely, FAE has been reported to stimulate antibody production by transferring antigens to the medulla and have a leading role in developing of local immune response. Among the others, the speciesspecifi c differences in bursa of Fabricius morphology of long-legged buzzard (Buteo rufi nus) also might support the continuity of this species in nature. Growth hormone (GH) is expressed in several extra-pituitary tissues, including the primary and secondary lymphoid organs of the immune system. In birds, GH mRNA and protein expression show a specific developmental distribution pattern in the bursa of Fabricius (BF), particularly in epithelial and B cells. Changes in the bursal concentration and distribution of locally produced GH during ontogeny suggest it is involved in B cell differentiation and maturation, as well as in a functional survival role in this organ, which may be mediated by paracrine/autocrine mechanisms. Here, we analyzed the anti-apoptotic effect of GH in BF and the intracellular signaling pathways involved in this activity. Also, we studied if this effect was exerted directly by GH or mediated indirectly by IGF-I. Bursal cell cultures showed an important loss of their viability after 4 h of incubation and a significant increase in apoptosis. However, treatment with 10 nM GH or 40 nM IGF-I significantly increased B cell viability (16.7 ± 0.67% and 13.4 ± 1.12%, respectively) when compared with the untreated controls. In addition, the presence of apoptotic bodies (TUNEL) dramatically decreased (5.5-fold) after GH and IGF-I treatments, whereas co-incubation with anti-GH or anti-IGF-I, respectively, blocked their anti-apoptotic effect. Likewise, both GH and IGF-I significantly inhibited caspase-3 activity (by 40 ± 2.0%) in these cultures. However, the use of anti-IGF-I could not reverse the GH anti-apoptotic effects, thus indicating that these were exerted directly. The addition of 100 nM wortmannin (a PI3K/Akt inhibitor) blocked the GH protective effects. Also, GH stimulated (3-fold) the phosphorylation of Akt in bursal cells, and adding wortmannin or an anti-GH antibody inhibited this effect. Furthermore, GH was capable to stimulate (7-fold) the expression of Bcl-2. Taken together, these results indicate that the direct anti-apoptotic activity of GH observed in the chicken bursal B cell cultures might be mediated through the PI3K/Akt pathway. IgMIgG B cells were detected, by immunofluorescence staining of single cells, in the bursa of Fabricius after hatching. To study the role of maternal IgG (MIgG) in this emergence of IgMIgG B cells, MIgG-free chicks were established from surgically bursectomized hens. Deprivation of MIgG in chicks completely prevented the appearance of IgMIgG B cells in the bursa 1 week after hatching. However, introduction of fluorescein-isothiocyanate-labeled MIgG to MIgG-free chick embryos on day 18 of incubation retrieved IgMIgG B cells in the bursa 1 week after hatching. Thus, IgMIgG B cells are induced by the binding of MIgG to IgM B cells in the bursa after hatching. Nevertheless, no binding of MIgG to IgM B cells was observed in the bursa of chick embryos in which B-cell proliferation and differentiation were independent of external antigens (Ags). Additionally, the binding of MIgG to IgM B cells after hatching was prevented by the isolation of the bursa from environmental stimuli by bursal duct ligation. Therefore, Ag stimulation from the external environment to the bursa is indispensable for the binding of MIgG to IgM B cells in the bursa. Several causes may induce change and atrophy in the bursa of Fabricius (BF). Databases on BF standards are available from published studies, however, updated references are needed to adjust the BF standards to present changes in highly specialized broiler genetic lines. The aim of this study was to evaluate BF-related measurements (weight and dimensions) under controlled conditions that would mimic field situations. Chickens were kept in isolation, thus avoiding exposure to disease agents by vaccination or field infections. This study was conducted using male Cobb 500 commercial broilers from the same hatch and source. Absence of disease was confirmed throughout the study. Despite the presence of individual variations, a minimum bursa-to-body weight ratio standard of 0.11 is proposed in broilers from 7 to 42 days of age. [ABSTRACT FROM AUTHOR]

The spleen and bursa of Fabricius in birds are organs that play an important role in fighting parasite infections. The size of these organs can be used by ecologists as a measure of immune investment, with larger size implying greater investment. The bursa only occurs in juvenile birds during the development of the B cell repertoire, whereas the spleen, which is the main site of lymphocyte differentiation and proliferation, is present in both juveniles and adults. We investigated spleen and bursa mass in relation to parasite measures for 541 Rock Ptarmigan Lagopus muta collected in northeast Iceland during October from 2007 to 2012. Of these 541birds, 540 carried at least one parasite species. Juveniles had heavier spleens than adults, and adult females had heavier spleens than adult males, but there were no sex differences in juveniles. Spleen mass increased from 2007 to 2009, then decreased up to 2011, before slightly increasing again in 2012. Spleen and bursa mass in juveniles increased with improved body condition, but decreased in adults, and this effect differed significantly among years. Spleen mass in juveniles was positively associated with parasite species richness and abundance, in particular endoparasite abundance, with coccidian parasites being the main predictors. Bursa mass was negatively associated with elevated ectoparasite abundance, with two chewing lice being the main predictors. These two immune defense organs appeared to relate to different stimuli. Mean annual spleen mass of juveniles changed in synchrony with Ptarmigan body condition and population density over the years of this study. The only parasite measure that showed any relation to density was coccidian prevalence in juvenile birds, with an approximately 2-year time-lag, suggesting that factors other than parasites are probably more important in triggering changes in spleen mass. After examination of 150 female quails, four of them showed the dedifferentiate tumor of the bursa of Fabricius. To the periphery, it consists of the dedifferentiate hemangiosarcoma and liposarcoma, the predominant area being constituted from lymph nodes of variable sizes. Among the lymphocytes rows the extensions of the reticular cells appeared positive for MHC II antibody. The lymphocytes showed the negativity to p53. The Bcl-2 marker manifested a limited positive reaction only in certain follicles. IgA and IgM markers showed the relatively low level of positivity in lymph nodes. Only the dense cords of the pleomorphic melanocytes manifested the positivity to Bcl-2. At the junctions between ventriculus and duodenum; ileum and cecum, especially in serosa, there were identified lymphocytes positive to MHC II, Bcl-2 and p53 markers surrounded by clusters of lipoid cells located inside the intestine wall. The presence of Cryptosporidium spp. was detected in broiler chickens of 32-40 days of age, colonizing the bursa of Fabricius. The histologic study with haematoxylin and eosin staining was done on specimens of trachea, complete intestinal tract and bursa. Samples of intestinal content were also studied using Kinyoun staining technique. All birds with signs of illness were positive to the presence of parasite, showing a hypertrophic bursa of Fabricius, filled with caseous content. The bursal epithelial cells were full of different developmental stages of Cryptosporidium spp. This note describing the presence of Cryptosporidium spp. in industrial flocks of broiler chickens, is the first report of this parasite in Uruguay. The aim of this work was to investigate developmental changes in cell proliferation and apoptosis in normal duck bursa of Fabricius using flow cytometry and iimnunohistochemistry. Studies were carried out on Tianfu ducks on days 24 and 27 of embryogenesis (E24 and E27) along with days 20,70, and 200 of postnatal development (P20, P70, and P200). Results showed that the percentage of G0/G1 bursa cells significantly increased between E24 and P200 while the percentage of cells in the S phase or G2 + M phase as well as the proliferating index obviously decreased during the same period. Proliferation cell nuclear antigen was detected in lymphocyte and interfollicular epithelium. The proliferative lymphocyte density tended to decrease from E24 to P200. Apoptotic bodies in macrophages, free apoptotic bodies, or nuclei with condensed chromatin in lymphocytes in follicles were identified by transferase-mediated dUTP nick-end labeling. Both flow cytometry and microscopic analysis reveal that the proportion of apoptotic cells and apoptotic lymphocyte density increased from E24 to P20, fell on P70, then rose again on P200. Background Avian leukosis virus subgroup J (ALV-J) causes tumours and immunosuppression in chickens. The host-ALV interactions at the transcriptional level are unknown. In this study, gene expression profiling was performed to analyse the bursa response induced by ALV-J strain JS09GY3 in chickens. Results A total of 594 gene transcripts displaying differential expression during ALV-J infection were identified. These differentially expressed genes are involved in binding, biological regulation, metabolic processes (MYF6 and FABP3), response to stimulus (F13A1 and CNGA3) and immune system processes (LY86, CATHL2, CCL4, and OASL), and several differentially expressed genes (e.g., ETV7, MMP9, and NOV) are involved in tumourigenesis. Eight differentially expressed genes were confirmed by quantitative reverse transcription-PCR (qRT-PCR). Based on pathway analysis, the notable signalling pathways mainly included cytokine-cytokine receptor interaction, the JAK-STAT signalling pathway and the RIG-1-like receptor signalling pathway. Bisphenol A (BPA) is a chemical estrogen widely used in the food packaging industry, especially in baby bottles. Its toxicity for the fetus has become a great concern in recent years. In the present study, the effects of BPA on the development of central immune organs in chick embryos were investigated. A total of 30 specific-pathogen-free (SPF) chick embryos were divided into BPA, control, and vehicle group. Chick embryos were exposed to BPA (250 μg per egg), saline (control), or corn oil (vehicle) on embryonic day 9 (ED9) by injection into the allantoic cavity. Thymuses and bursae of Fabricius were collected on ED22. The microscopic examination of tissue structure and ultrastructure was carried out for histopathological changes of thymus and the bursa of Fabricius morphology under light and scanning electron microscopes (SEM). In the BPA group, the weight index of the bursae of Fabricius was significantly reduced (p < 0.01); the number of lymphatic follicles in the bursae of Fabricius was remarkably decreased (p < 0.01); and the thickness of the thymus cortex and medulla was reduced (p < 0.01). Light microscope and SEM examinations further showed that the lymphatic follicles and epithelial cells of the bursa of Fabricius and thymus were damaged by BPA. Our study confirms a direct toxicity of BPA at a very low-dose level on the development of the central immune organs of SPF chick embryos. Forsythiaside, a phenylethanoside product isolated from air-dried fruits of Forsythia suspensa, has been demonstrated to exhibit antioxidant, antibacterial and anti-inflammatory activities in vitro. However, its mechanism and the effects of lipopolysaccharide (LPS)-induced injury on the bursa of Fabricius (BF) of chickens are poorly understood. The present study aimed to investigate the anti-inflammatory effects of forsythiaside on LPS-induced acute inflammation. In addition, the potential molecular mechanisms of forsythiaside were analyzed in the BF, a special immune organ in chickens. Forty 15-day-old chickens were randomly divided into control, LPS and LPS plus forsythiaside (30 or 60 mg/kg) groups (n=10 for each group). In the LPS plus forsythiaside (30 or 60 mg/kg) groups, the chickens were orally administered with forsythiaside at doses of 30 and 60 mg/kg for seven days. At 21 days old, the chickens were intravenously injected with 200 µg/kg body weight LPS. Chickens in the control and LPS groups were only administered with vehicle or LPS, respectively, at day 21. At 3 h post-injection, the body temperature and nitric oxide (NO) levels were analyzed. In addition, the levels and mRNA expression of pro-inflammatory cytokines, including tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and IL-1β, and the mRNA expression of nuclear factor-κB (NF-κB), cyclooxygenase-2 (COX-2) and inducible NO synthase (iNOS), were examined in the BFs isolated from the chickens. The results revealed that forsythiaside was able to attenuate the LPS-induced inflammatory responses in the BFs of the chickens. The mechanisms by which forsythiaside exerted its anti-inflammatory effect were found to correlate with the inhibition of IL-6, IL-1β, TNF-α and COX-2 production, via the inactivation of NF-κB, indicating that the NF-κB-iNOS-NO signaling pathway may be important in this process. The bursa of Fabricius of the chicken is known to be both a primary lymphoid organ and a secondary lymphoid tissue. Bursal follicles are equipped with antigen-trapping follicle-associated epithelium. However, bioactive antigens such as protein and bacteria have not been detected in the bursal parenchyma. By immunoperoxidase staining with a polyspecific antibody (Ab) against Escherichia coli, we detected aggregated E. coli antigens in the medulla of bursal follicles after hatching. The distribution of aggregated E. coli antigens is restricted to the medulla of bursal follicles. The antigens are not found in the spleen or the parenchyma of the caecal tonsil. The bursa is thus a trapping site for E. coli antigens from the external environment. Furthermore, two-color immunostaining clarified that these antigens form immune complexes with maternal IgG (MIgG) and are retained by reticular cells. Additionally, immune complexes in the bursa were shown to induce the rapid development of serum IgM Ab for indigenous E. coli. Our results suggest that immune complexes of MIgG and environmental antigens in the medulla of bursal follicles exert positive effects on B-cell differentiation in the bursa in situ. Abstract: Toll-like receptors (TLRs) are an evolutionarily conserved group of pattern recognition receptors that play an important role in mediating host-responses to pathogens. Several TLRs have been identified in chickens and their expression has been detected in many immune cell subsets including in B cells. However, the mechanisms through which TLRs modulate B cell responses have not been well characterized in chickens. The aim of the present study was to elucidate the responses mounted by cells of the bursa of Fabricius to treatment with the TLR 3, 4 and 21 ligands, poly I:C, lipopolysaccharide (LPS) and CpG oligodeoxynucleotides (ODN), respectively. The relative expression of several immune system genes was quantified at 1, 3, 8 and 18h post-treatment. The results show that all three ligands induced the up-regulation of interferon (IFN)-γ and interleukin (IL)-10 transcripts and promoted the up-regulation of transcripts associated with antigen presentation, namely CD80 and major histocompatibility complex (MHC) class II. Furthermore, the results indicated that LPS and poly I:C induced the greatest IFN-γ and IL-10 responses, respectively, while CpG ODN was the most efficacious inducer of CD80 and MHC-II expression. Future studies may be aimed at elucidating the mechanisms of TLR-mediated activation of chicken B cells. Antibiotics are commonly used to prevent and treat poultry microbial infections, but certain antibiotic families depress humoral immunity, such as antibody production. Poultry humoral immunity depends on the normal functioning of the bursa of Fabricius and the B lymphocytes that mature in that gland. In this study, recommended therapeutic doses of enrofloxacin, florfenicol, or ceftiofur were administered to 2-d-old chicks. On d 7 post-hatch, bursae were sampled for histological, immunohistochemical, and flow cytometric determination of Bu-1-positive (Bu-1+) cell number, percentage, and distribution. The bursa of Fabricius from all treatment and control groups had normal morphology. The administration of antibiotics significantly decreased the number of Bu-1+ cells in the bursal medulla, with a simultaneous increase of these cells in the cortex. Flow cytometry revealed a significant decrease in the percentage of bursal Bu-1+ cells from all of the studied antibiotics: enrofloxacin (93.91 ± 3.27), florfenicol (87.84 ± 7.14), and ceftiofur (89.16 ± 5.68) compared with that of the control (96.48 ± 2.60). Abstract: Glucocorticoids (GCs) affect the development of both the immune and nervous systems. To do so, GCs bind to intracellular receptors, mineralocorticoid receptors (MR) and glucocorticoid receptors (GR). In addition, GCs bind to membrane-associated corticosteroid receptors (mCR). Two well-known GCs are corticosterone and cortisol. Whereas corticosterone is the primary GC in zebra finch plasma, cortisol is the primary GC in zebra finch lymphoid organs and is also present in the brain and plasma during development. Here, we characterized binding sites for corticosterone and cortisol in plasma, liver, lymphoid organs, and brain of developing zebra finches. In tissues, we examined both intracellular and membrane-associated binding sites. For intracellular receptors, there were MR-like sites and GR-like sites, which differentially bound corticosterone and cortisol in a tissue-specific manner. For mCR, we found little evidence for membrane-associated receptors in immune organs, but this could be due to the small size of immune organs. Interestingly, cortisol, but not corticosterone, showed a low amount of specific binding to bursa of Fabricius membranes. For neural membranes, corticosterone bound to one site with low affinity but a relatively high B max, and in contrast, cortisol bound to one site with high affinity but a lower B max. Our results indicate that intracellular and membrane-associated receptors differentially bind corticosterone and cortisol suggesting that corticosterone and cortisol might have different roles in immune and nervous system development. Abstract: Growth hormone (GH) is expressed in the chicken bursa of Fabricius (BF), an organ that undergoes three distinct developmental stages: rapid growth (late embryogenesis until 6–8weeks of age [w]), plateaued growth (between 10 and 15w), and involution (after 18–20w). The distribution and abundance of GH-immunoreactivity (GH-IR) and GH mRNA expression in stromal and non-stromal bursal cells during development, as well as the potential anti-apoptotic effect of GH in bursal cell survival were the focus of this study. GH mRNA expression was mainly in the epithelial layer and in epithelial buds at embryonic day (ED) 15; at 2w it was widely distributed within the follicle and in the interfollicular epithelium (IFE); at 10w it clearly diminished in the epithelium; whereas at 20w it occurred in only a few cortical cells and in the connective tissue. Parallel changes in the relative proportion of GH mRNA expression (12, 21, 13, 1%) and GH-IR (19, 18, 11, <3%) were observed at ED 15, 2w, 10w, and 20w, respectively. During embryogenesis, GH-IR co-localized considerably with IgM-IR, but scarcely with IgG-IR, whereas the opposite was observed after hatching. Significant differences in bursal cell death occurred during development, with 9.3% of cells being apoptotic at ED 15, 0.4% at 2w, 0.23% at 10w, and 21.1% at 20w. Addition of GH increased cultured cell survival by a mechanism that involved suppression (up to 41%) of caspase-3 activity. Results suggest that autocrine/paracrine actions of bursal GH are involved in the differentiation and proliferation of B lymphocytes and in BF growth and cell survival in embryonic and neonatal chicks, whereas diminished GH expression in adults may result in bursal involution. Abstract: Stem cell antigen 2 (SCA2) is a Ly6 family member whose function is largely unknown. To characterize biological properties and tissue distribution of chicken SCA2, SCA2 was expressed in E. coli, purified, and a polyclonal antibody developed. Utilizing the polyclonal antibody, SCA2 is a 13kDa cell surface protein anchored by a glycosyl-phosphatidylinositol (GPI) moiety. SCA2 is expressed in connective tissues of thymus and bursa based on immunohistochemistry, immunoprecipitation, and western blots. In bursal follicles, SCA2 is specifically expressed on the cortical-medullary epithelial cells (CMEC) surrounded by MHC class II presenting cells. Expression profiles of bursal cells induced by contact with SCA2-expressing cells shows down-regulation of numerous genes including CD79B, B cell linker (BLNK), spleen tyrosine kinase (SYK), and gamma 2-phospholipase C (PLCG2) that are involved in the B cell receptor (BCR) and immune response signaling pathways. Abstract: Bursa of Fabricius is a primary lymphoid organ in the birds and its microenvironment is responsible for the B cell maturation. The epithelial anlage develops through epithelial–mesenchymal interaction into which –as a third party – hemopoietic cells immigrate. Chemical bursectomy can be made by dipping or injecting eggs by testosterone solution. Both methods of treatments compromise the development of the whole embryo. Heparin beads of 70–150 µm in diameter were soaked in 5% testosterone solution and implanted into the tail bud of 4 day old chicken embryos before the bursal anlage began to develop at embryonic day 5. The locally applied testosterone inhibited the differentiation of the bursal secretory dendritic cell (BSDC) precursors, which were the inducer of the follicular epithelial bud formation. Testosterone treatment did not blocked the entering of B cells and other hemopoietic cells into the bursal mesenchyme, but in the absence of BSDC precursor cells the dendroepithelial microenvironment is not established, subsequently B cell precursors are lodged only in the bursal mesenchyme. Abstract: The structure and function of the lower intestinal tract of Rhea americana were characterized to evaluate the evolutionary relationship to other struthioniform and avian species. In 5 rheas the gross anatomy and the light and transmission electron microscopy were studied in parallel to in vitro electrophysiological measurements of ion transport. The mucosa in the colon was amplified with villi, often branched, and in the coprodeum with folds. In both tissues the epithelium was a monolayer composed of columnar absorptive cells, goblet cells and mitochondria-rich cells. Colon and coprodeum appeared to produce large amounts of mucus. The proctodeal diverticulum was rich in lymphoid tissue arranged into lobuli bursales, and it was concluded that this structure is a modified bursa of Fabricius. The sparse interlobular epithelium of the diverticulum resembled that of colon and coprodeum. Baseline short circuit currents (ISC) averaged 114.5±13.8 µA/cm2 in colon, 193.1±30.3 µA/cm2 in coprodeum and 60.4±9.1 µA/cm2 in the diverticulum. Amiloride sensitive Na+-transport amounted to 31, 88 and 38% of the baseline ISC in these three tissues, respectively. In all tissues, there was also a modest, theophylline activated chloride secretion response, and ouabain, the Na+/K+-ATPase inhibitor, abolished most of the ISC. The transepithelial resistance (TER) of the diverticulum was much higher than the other tissues. Upon dissection, urate from ureteral urine was observed in the lower third of the colon and to a lesser extent in the proctodeal diverticulum, indicating retrograde peristalsis of the urine. Thus, unlike the ostrich, there is no sphincter separating colon and coprodeum. On the other hand, a thick mucus layer was seen overlying the mucosa in both colon and coprodeum, as in the ostrich. This may help to prevent osmotic water loss, despite the presence of hyperosmotic urine (up to 800 mOsm) in the lower intestine. Both morphological and electrophysiological data from the rhea support the hypothesis that the rhea lower intestine contributes to post-renal modification of ureteral urine and to the regulation of osmotic balance, as also seen in domestic fowl and other avian species. Abstract: The bursa of Fabricius serves as an important tissue in the process of Marek''s disease virus (MDV) pathogenesis, since B cells of the bursa harbor the cytolytic phase of MDV replication cycle. In the present study, host responses associated with MDV infection in the bursa of Fabricius of chickens were investigated. The expression of MDV phosphoprotein (pp)38 antigen, MDV glycoprotein (gB) and MDV viral interleukin (vIL)-8 transcripts was at the highest at 4 days post-infection (d.p.i.) and then showed a declining trend. On the contrary, the expression of meq (MDV EcoRI Q) gene as well as the viral genome load increased gradually until day 14 post-infection. The changes in viral parameters were associated with significantly higher infiltration of macrophages and T cell subsets, particularly CD4+ T cells into the bursa of Fabricius. Of the genes examined, the expression of interferon (IFN)-α, IFN-γ genes and inducible nitric oxide synthase (iNOS) was significantly up-regulated in response to MDV infection in the bursa of Fabricius. Sex steroid hormones have been reported to be modulators that augment or suppress immune functions. Applying estrogen to chick embryos has been reported to influence antibody production after hatching, suggesting that estrogen acts on B cell differentiation and proliferation in the bursa of chick embryos. We previously reported the presence of estrogen receptor α (ERα) in the bursa during the late period of embryogenesis. In the present study we examined the time course of ERα expression in the bursa of chick embryos at the late period of embryogenesis by ERα-messenger RNA (mRNA) expression analysis by reverse-transcription-polymerase chain reaction (RT-PCR) using primers for chicken ERα, and immunohistochemistry using an anti-ER antibody. The quantity of ERα-mRNA expressed, estimated from the relative densities of the ERα RT-PCR products to those of β-actin, changed with time during the late period of embryogenesis (day 10 to day 21). ERα-mRNA expression was observed at all ages examined in the present experiment. The expression increased between day 10 and day 15 of embryogenesis and then the value was decreased between day 15 and day 21 of embryogenesis. The numbers of ER-positive cells in the bursa also changed with time during the late period of embryogenesis (day 14 to day 18). ER-positive cells showed the highest level on day 14 of embryogenesis, and then the value declined. ER-positive cells were observed in lymphoid follicular cells, stromal cells and epithelial cells, and the density of ER-positive cells was highest in epithelial cells among the three cell components of the bursa. The high level of ER expression in the bursa of Fabricius (BF) of chick embryos at the late period of embryogenesis suggests that this stage of embryogenesis is critical in B cell differentiation in the bursa in connection with estrogen effects on antibody production after hatching. The aim of this study was to investigate histomorphometrical characteristics of the thymus, bursa of Fabricius and spleen in the chickens vaccinated with a vaccine against Marek's disease. For this purpose, we used newly hatched chickens of the light hybrid line, obtained from a local hatchery. The chickens were vaccinated on the 5th day after hatching with a bivalent cell-associated Marek's disease vaccine (PFU-2000 per dose). On day 13 both vaccinated chickens and unvaccinated controls were sacrificed, and thymus, bursa and spleen were removed and processed for light microscopy. The serial tissue sections, hematoxylin-eosine stained, were used for histomorphometric analysis. Vaccination against Marek's disease decreased the relative mass of the lymphoid organs, and caused significant damage of the thymus and spleen in experimental chicken. In addition, vaccination, similar to Marek's disease virus, induced morphometric changes in the lymphoid organs. Namely, it significantly decreased the diameter and volume of lymphoid follicles, volume of follicular medulla and number of cells in the follicular cortex in the bursa of Fabricius. In the thymus, vaccination reduced the thymus volume and the absolute number of thymocytes. However, vaccination against Marek's disease caused an increase in the diameter, number and volume of lymphoid follicles in the spleen. The present data suggest that vaccination against Marek's disease was able to induce the immune response in processed organs, although it reduced the mass and number of lymphocytes in the major lymphoid organs. The rate of behavioural innovation, such as opportunistic feeding innovation, may facilitate adaptation to novel environments. Because parasites may affect how their hosts adopt novel means of resource acquisition, or because opportunistic behaviours may involve the risk of being exposed to a large parasite fauna, we hypothesize an evolutionary link between the rate of feeding innovations and parasitism. We investigated the phylogenetic relationship between relative frequency of feeding innovations (adjusted for research effort and population size) and relative size of immune defense organs (as a relative measure of parasite-mediated selection) and the prevalence of blood parasites in birds. Using generalized least squares models, we found that species with relatively large bursa of Fabricius, thymus, and spleen had higher rates of feeding innovations than species with small immune defense organs. Similarly, there was a positive interspecific association between feeding innovation and haematozoa prevalence. These relationships were not confounded by migration, relative brain size, geographical distribution, and male plumage brightness. Analyses of causality relying on evolutionary modelling of discrete variables and path analysis suggest that increasing rate of feeding innovation may place species under intense selection due to parasitism. The aim of the present study was to demonstrate estrogen receptor (ER)α (ER-α) and ER-β expression in the bursa of Fabricius and cultured bursal epithelial cells (CBEC) by reverse transcription (RT)-polymerase chain reaction (PCR) using a primer set design based on the sequences of chicken ER-α complementary DNA (cDNA) and chicken ER-β-cDNA. In the case of ER-α, RT-PCR products of the expected size (698 base pairs (bp)) were amplified from cDNA derived from the bursa and CBEC, and from the oviduct, which was used as a positive control. All of the products were cleaved into two fragments of 476 bp and 222 bp by treating the products with restriction nuclease Hind III, indicating that these RT-PCR products from the bursa, CBEC and genital organs were the same. In contrast, in the case of ER-β, PCR products of the expected size (303 bp) were not detected in the bursa of Fabricius or CBEC, or in the oviduct and ovary. These results indicate that ER-α is expressed in the bursa of Fabricius and in CBEC, but ER-β is not expressed in the bursa or CBEC, or in the female genital organs of chickens. Bursal anti-steroidogenic peptide (BASP), purified from the chicken bursa of Fabricius (BF), has been previously demonstrated to be a potent and efficacious inhibitor of steroid hormone biosynthesis from chicken ovarian, and both mammalian and avian adrenal cells in vitro. Other studies have demonstrated that BASP can markedly reduce avian and mammalian mitogen-stimulated lymphocyte proliferation. Recent studies have indicated that BASP has a structural and functional relationship with histone H1. Immunohistochemical studies using a monoclonal antibody, which is known to recognize a common histone H1 epitope from several plant and animal species identified the protein within the cytoplasm and nucleus of distinct cells within both the cortex and medulla of all BF follicles. Additionally, epithelial cells within the BF expressed the protein strongly in the cytoplasm with reduced nuclear staining. In contrast, the same antibody did not recognize the protein in thymus of the same animals. The differential expression of histone H1 immunoreactivity within selected cells of the BF may support a previous proposed role of histone H1 in extranuclear and extracellular signaling in chickens and possibly other species. B lymphocyte development can occur in a variety of anatomical sites. While typically considered to be a process that occurs in the bone marrow throughout life, it is becoming clear that gut associates sites of B cell development are critically important in many species of veterinary importance. Among these sites, the bursa of Fabricius in chickens and the ileal Peyer’s patches of sheep are among the best studied. In these organs, it has become clear that many of the properties associated with B cell development in rodent and primate bone marrow do not apply. Thus while bone marrow B cell development typically involves an ongoing maturation of mature B cells from immature B lineage precursors that lack the expression of a surface immunoglobulin complex, gut associated lymphoid tissues (GALTs) may be colonized by a single wave of precursor cells during embryo development. Nonetheless, molecular analysis of the requirements for B lymphocyte development in GALTs reveals some striking parallels with requirements identified for B cell development in bone marrow. This article will discuss differences between B cell development in the bone marrow and GALTs and recent evidence emerging that yields insights into how these processes are regulated. The immunocytochemical study of the K-1 monoclonal antibody indicates that the epithelial components of the bursa of Fabricius of the chicken and guinea fowl express the K-1 positive molecule. During embryogenesis, the K-1 antigen expression appears together with the bud-formation. As the number of B cells increases in the developing follicle, the K-1 expression gradually diminishes in the medullary reticular epithelial cells and completely ceases by hatching, which suggests that the molecule is developmentally regulated. After hatching, the expression of the molecule is restricted to the sealing off zone of the lymphoepithelial or medullary region of the follicle: i.e. to the cortico-medullary (CM) epithelial cells and the follicle associated epithelium (FAE) supporting cells in guinea fowl and to the latter ones in the chicken. The expression of the K-1 antigen by these epithelial components may support their structural identity. After hatching, the K-1 molecule is restricted to the CM epithelial cells and/or FAE supporting cells, which suggests that the function of the embryonic epithelial bud is taken over by the CM epithelial cells. The K-1 positive CM epithelial cells form arches, which encompass blast-like cells. The thymus in vertebrates and the bursa of Fabricius in birds regress before reproduction, while the immunological information of these organs is maintained as cell memory. Regression at a certain age presupposes that individuals have achieved exposure to a large fraction of parasites in the environment. Here we present a new scenario for regression of immune defence organs, based on optimality reasoning. This scenario links early involution of immune defence organs with (1) effects of exposure to parasites on adaptive immune responses to these parasites, (2) exposure to local parasite communities during natal dispersal and migration as a means of “vaccination” against local parasites, and (3) the function of visits to future breeding sites by juveniles as a means of exposure to local parasites. This scenario provides explanations for why natal dispersal is longer than breeding dispersal, for sex differences in dispersal, and for why the bursa of Fabricius regresses relatively early in life among bird species with delayed start of reproduction. Changes from hatching to the involutive stage in the thymus and bursa of Fabricius of Gallus domesticus were studied. Pro-opiomelanocortin (POMC)-derived peptides and cytokines were also tested by immunocytochemical procedures. Thymic histological modifications appeared at 3 months and involved an increase in connectival argyrophilic reticular fibres, a proliferation of non-epithelial cells in the reticulum network, the presence of mucous cells and small mucous cysts, and an increase in the number of eosinophilic and myoid cells. A clear distinction between the cortex and the medulla was lost. Immunoreactive POMC-related molecules and cytokines were demonstrated in thymic cells from 4-day-old chicken, and their number increased with ageing. These molecules were expressed in a few single epithelial cells and in interdigitating cells. With ageing, the number of immunoreactive interdigitating cells also increased, and these appeared in an activated phase. Histological modifications in the bursa of Fabricius appeared at 2 months and concerned a folding of the interfollicular surface epithelium covering the bursal plicae and a reduction in lymphoid follicle-associated epithelium. Fibrous tissue gradually increased, and large mucoid cysts were evident. The expression of POMC-derived peptide-and cytokine-like molecules differed during the development and involution phases of the organ. Cells of follicle associated epithelium and dendritic reticular cells of lymphoid follicles were immunoreactive to β-endorphin between 4 days and 2 months, while ACTH-, α-MSH- and cytokine-like molecules were observed in follicles after 2 months. Antibody diversification in the bursa of Fabricius occurs by gene conversion: pseudogene-derived sequences replace homologous sequences in rearranged immunoglobulin genes. Bursal cells expressing a truncated immunoglobulin μ heavy chain, introduced by retroviral gene transfer, bypass normal requirements for endogenous surface immunoglobulin expression. Immunoglobulin light chain rearrangements in such cells undergo gene conversion under conditions where the products are not selected based on their ability to encode a functional protein. The efficiency with which gene conversion maintains a productive reading frame exceeds 97% under such non-selective conditions. By analysis of donor pseudogene usage we demonstrate that bursal cell development is not driven by a restricted set of antigenic specificities. We further demonstrate that gene conversion can restore a productive reading frame to out-of-frame VJL junctions, providing a rationale for the elimination of cells containing non-productive VJL rearrangements prior to the onset of gene conversion in normal bursal cell development.
