• This is the first study to report anatomy and endogenous hormonal profiles in the early seedling stage of kohlrabi. • The de novo formation of supplementary vascular bundles and phloem ring are the key features of kohlrabi stem. • IAA, IAAsp, GA and tZ profiles were significantly varied in the stems while the ABA profile was different in the leaves. • The stunted stem and vascular activity of cambium are positively correlated with endogenous GA and cytokinin in kohlrabi stem. To unveil the mechanism of tuberous stem formation at early growth stages in kohlrabi, the comparative stem morphology, anatomy, and endogenous plant hormone profiles were studied at different days after germination (10 to 30 DAG). Stem anatomy revealed that the notable feature of the kohlrabi stem during the transition from primary to secondary growth was the formation of vascular bundle circles, with many phloems formed separately and arranged in a circular pattern along with the cambium layer, resulting in an expanded ring of primary and secondary phloem. In addition, medullary supplemental vascular bundles were differentiated in the central region of pith and successively supplied small-sized meristematic parenchymatous cells which contribute to the tuber initiation in kohlrabi. Six phytohormones were quantified in the stems and leaves of both genotypes. The absence of Gibberellin A4 (GA4) in kohlrabi stem might be the cause of the stunted stem of kohlrabi. The total endogenous cytokinin in stem of kohlrabi was more than twice of that of broccoli, where trans-zeatin was the most abundant bioactive form. This can be explained by the meristematic nature of the cambium of kohlrabi stem. Profiles for Indole-3-acetic acid, Indole-3-acetylaspartic acid, and Abscisic acid of the stems and leaves of kohlrabi and broccoli were significantly different at several time points tested. Thus, the key features of kohlrabi during the initiation of tuberous stem were the formation of the phloem ring along with the cambium and the supplemental vascular bundles. This is the first paper showing the hormonal profile in the early seedling stage of kohlrabi. These findings provide useful information to elucidate the mechanism of tuberous stem formation of kohlrabi and other tuber crops. [ABSTRACT FROM AUTHOR]

The paper characterises selected vegetable species of the Brassica genus: white headed cabbage (Brassica oleracea L. var. capitata f. alba), savoy cabbage (Brassica oleracea L. var. sabauda L.), napa cabbage ((Brassica rapa L. subsp. pekinensis), broccoli (Brassica oleracea L. var. italica), kale (Brassica oleracea L. var. sabellica L.), cauliflower (Brassica oleracea L. var. botrytis), kohlrabi (Brassica oleracea L. var. gongylodes L.), Brussels sprout (Brassica oleracea L. var. gemmifera). It describes their medicinal properties and economic importance, as well as presents their value as a resource for the food and pharmaceutical industries. Brassicas are characterised by high vitamin and mineral content and have various medicinal properties. They also have a high nutritional value, high antioxidative activity and health benefits. From the point of view of nutrition, brassicas are perceived as valuable in cancer prevention. It is noteworthy that kale (Brassica oleracea L. var. gongylodes L.) is characterised by significant nutritional and health benefits, yet it is not popular in Poland. [ABSTRACT FROM AUTHOR]

4. Mediation of host selection and oviposition behavior in the diamondback moth Plutella xylostella and its predator Chrysoperla carnea by chemical cues from cole crops

Host plant-mediated orientation and oviposition by diamondback moth (DBM) Plutella xylostella (L.) (Lepidoptera: Yponomeutidae) and its predator Chrysoperla carnea Stephens (Neuroptera: Chrysopidae) were studied in response to four different brassica host plants: cabbage, (Brassica oleracea L. subsp. capitata), cauliflower (B. oleracea L. subsp. botrytis), kohlrabi (B. oleracea L. subsp. gongylodes), and broccoli (B. oleracea L. subsp. italica). Results from laboratory wind tunnel studies indicated that orientation of female DBM and C. carnea females towards cabbage and cauliflower was significantly greater than towards either broccoli or kohlrabi plants. However, DBM and C. carnea males did not orient towards any of the host plants. In no-choice tests, oviposition by DBM did not differ significantly among the test plants, while C. carnea layed significantly more eggs on cabbage, cauliflower, and broccoli than on kohlrabi. However, in free-choice tests, oviposition by DBM was significantly greater on cabbage, followed by cauliflower, broccoli, and kohlrabi, while C. carnea preferred to oviposit on cabbage and cauliflower, followed by broccoli and kohlrabi. The predation rates of DBM by C. carnea on kohlrabi and broccoli were not significantly different from one another, but were significantly higher than that on cabbage and cauliflower. When two types of plant, intact and injured, were available to adult DBM, female oviposition was significantly greater on injured plant leaves than on intact plants leaves. Similarly, C. carnea oviposition was significantly greater on injured plant leaves than on intact leaves. Implications regarding the possible role of green leaf volatiles in host selection/preference, as well as in tritropic interactions, are discussed. [Copyright &y& Elsevier]

U ovom radu su ispitani hemijski sastav i biološka aktivnost vrsta Brassica oleracea i Brassica rapa poreklom iz organskog i konvencionalnog sistema proizvodnje. Ispitivanja hemijskog sastava su obuhvatila određivanje volatilnih komponenata primenom headspace GC-MS tehnike, kvalitativnu analizu sokova ispitivanih vrsta pomoću LC-DAD-MS/MS tehnike, kvantifikaciju odabranih fenolnih jedinjenjapomoću LC-MS/MS tehnike i spektrofotometrijsko određivanje sadržaja ukupnih glukozinolata, fenola i flavonoida i sadržaja ukupnih monomernih antocijana. Biološka aktivnost povrća je ispitana određivanjem antioksidantnog potencijal, antimikrobne i antitumorske aktivnosti sokova. Dobijeni rezultati pokazuju da su glavne volatilne komponente ispitivanih vrsta degradacioni proizvodi masnih kiselina i organosumpornih jedinjenja od kojih neke od identifikovanih komponenata predstavljaju ključne mirisne komponente. Na osnovu njihove distribucije nije bilo moguće utvrditi uticaj sistema proizvodnje na miris i ukus ispitivanih vrsta. Pomoću LC-DAD-MS/MS i LC-MS/MS tehnike je utvrđeno prisustvo primarnih metabolita, slobodnih hidroksibenzoevih i hidroksicimetnih kiselina, derivate hinske kiselinesa hidroksicimetnim kiselinama, glikozida flavonola, biflavonoida, i kumarina u sokovima spitivanih vrsta. Pored toga u svim sokovima subili prisutni kondenzacioni proizvodi degradacionih proizvoda indolnih glukozinolata sa askorbinskom kiselinom- askorbigen i metoksiaskorbigen. U sokovima crvenog kupusa su bili prisutni antocijani. Najveći sadržaj ukupnih glukozinolata je određen u brokoliju, dok je crveni kupus sadržao najveći sadržaj ukupnih fenola ukupnih flavonoida kao i hinske kiseline. Crveni kupus je bio varijetet koji je ispoljio najviši antioksidantni kapacitet u svim primenjenim testovima, brokoli i karfiol su ispoljili najjaču antimikrobnu aktivnost, dok je kelj bio varijetet sa najjačim antitumorskim delovanjem. Na osnovu analize hemijskog sastava i biološke aktivnosti vrsta B. oleracea i B. rapa može se zaključiti da u zavisnosti od primenjenog tipaproizvodnje mogu biti poboljšane neke od karakteristika ispitivanih vrsta i da je uticaj sistema proizvodnje značajan za pojedine varijetete, a nikako za celu vrstu. U konačnom obliku može se zaključiti da bi potencijalna kombinacija sokova crvenog kupusa, brokolija, karfiola i kelja mogla ostvariti značajno hemopreventivno i hemoterapeutsko delovanje, ispoljavajući istovremeno antioksidantnu, antimikrobnu i antitumorsku aktivnost., Chemical composition and biological activity of Brassica oleracea and B. rapa species were investigated in the present work. Investigation of chemical composition encompassed determination of volatile compounds by headspace GC-MS technique, qualitative analysis of juices of investigated vegetables by LC-DAD-MS/MS technique, quantification of selected phenolic compounds by LC-MS/MS technique and spectrophotometric determination of total content of glucosinolates, phenolic compounds and flavonoids and total content of monomeric anthocyanes. Biological activity of vegetables was investigated by determination of antioxidant capacity, antimicrobial and antitumorigenic effect of juices. Obtained results showed that main volatile compounds were degradation products of fatty acids and organo-sulphur compounds among which some of the identified components were the key aroma compounds. Based on their distribution it was not possible to evaluate influence of cropping system on aroma and taste of investigated species. By LC-DAD-MS/MS and LC-MS/MS techniques, it was determined the presence of primary metabolites, freehydroxybenzoic and hydroxycinnamic acids, derivatives of quinic acid and hidroxicinnamic acids, flavonol glucosides, biflavnoids and a cumarin in juices of the investigated species. Furthermore, condensation products of degradation products of indole glucosinolates with ascorbic acid- ascorbigen and metoxiascorbigen were present in all juices. Anthocyanes were present in juices of red cabbage. The highest total content of glucosinolates was determined in broccoli, while red cabbage had the highest total phenolic content, total flavonoid content, as well as content of qunic acid. Red cabbage was a variety that showed the highest antioxidant capacity measured by different assays. Broccoli and cauliflower expressed the highest antimicrobial potential, while Savoy cabbage was a variety with the highest antitumor effect. Investigation of chemical composition and biological activity showed that applied cropping conditions may be useful for improving some of the characteristics of investigated species and significance of cropping system only for particular varieties, but not for the whole species in general. As the conclusion, potential combination of juices of red cabbage, broccoli, cauliflower and Savoy cabbage could be potent chemopreventive and chemotherapeutic agent, expressing simultaneously antioxidant, antimicrobial and antitumor activity.

Brassica vegetables are known to contain glucosinolates that are precursors for bioactive compounds like isothiocyanates that have been shown to play an important role in human health. This study reports the results of a screening of 11 Brassica oleracea crops consisting of 42 cultivars (6 white cabbage, 5 red cabbage, 7 Brussels sprouts, 2 kale, 1 tronchuda, 3 oxheart cabbage, 2 kohlrabi, 6 broccoli, 5 cauliflower, 3 romanesco and 2 Savoy cabbage). All these cultivars were cultivated under the same conditions on a single location in the same season. The variation found in the level of glucosinolates is expected to be mainly due to the genetic variation. A large variation was observed in the level and profile of glucosinolates. Total glucosinolates varied from 14 to 625 μmol/100 g fresh weight. Glucoraphanin, the precursor of the isothiocyanate sulforophane, varied from 0 to 141 μmol/100 g fresh weight. Within broccoli glucoraphanin varied from 27 to 141 μmol/100 g fresh weight. Glucoiberin that is structurally related to glucoraphanin varied from 6 to 397 μmol/100 g fresh weight. Within broccoli glucoiberin varied from 21 to 397 μmol/100 g fresh weight.

Development of Expressed Sequence Tag-Simple Sequence Repeat (EST-SSR) markers derived from public database is known to be more efficient, faster and low cost. The objective of this study was to generate a new set of EST-SSR markers for broccoli and its related species and their usefulness for assessing their genetic diversity. A total of 202 Brassica oleracea ESTs were retrieved from NCBI and then assembled into 172 unigenes by means of CAP3 program. Identification of SSRs was carried out using web-based tool, RepeatMasker software. Afterwards, EST-SSR markers were developed using Primer3 program. Among the identified SSRs, trinucleotide repeats were the most common repeat types, which accounted for about 50%. A total of eight primer pairs were successfully designed and yielded amplification products. Among them, five markers were polymorphic and displayed a total of 30 alleles with an average number of six alleles per locus. The polymorphic markers were subsequently used for analyzing genetic diversity of 36 B. oleracea cultivars including 22 broccoli, five cauliflower and nine kohlrabi cultivars based on genetic similarity matrix as implemented in NTSYS program. At similarity coefficient of 61%, a UPGMA clustering dendrogram effectively separated 36 genotypes into three main groups, where 30 out of 36 genotypes were clearly discriminated. The result obtained in the present study would help breeders in selecting parental lines for crossing. Moreover, the novel EST-SSR markers developed in the study could be a valuable tool for differentiating cultivars of broccoli and related species.
