The recent sequencing of the complete genome of the peach, together with the availability of new high-throughput genome, transcriptome, proteome, and metabolome analysis technologies, offers new possibilities for Prunus breeders in what has been described as the postgenomic era. In this context, new biological challenges and opportunities for the application of these technologies in the development of efficient marker-assisted selection strategies in Prunus breeding include genome resequencing using DNA-Seq, the study of RNA regulation at transcriptional and posttranscriptional levels using tilling microarray and RNA-Seq, protein and metabolite identification and annotation, and standardization of phenotype evaluation. Additional biological opportunities include the high level of synteny among Prunus genomes. Finally, the existence of biases presents another important biological challenge in attaining knowledge from these new high-throughput omics disciplines. On the other hand, from the philosophical point of view, we are facing a revolution in the use of new high-throughput analysis techniques that may mean a scientific paradigm shift in Prunus genetics and genomics theories. The evaluation of scientific progress is another important question in this postgenomic context. Finally, the incommensurability of omics theories in the new high-throughput analysis context presents an additional philosophical challenge.

Vegetable breeding companies invest a considerable amount of their resources in phenotyping. The advancement of computer vision technology has made it possible to digitalize these processes, leading to improved efficiency and quality. However, phenotyping activities often take place in outdoor fields or greenhouses, where the environmental/illumination conditions are constantly changing. This lack of standardization presents a problem for automatically isolating the relevant elements in the images, which is an important first step for phenotyping. Classical image analysis methods have shown not to be robust enough for that in these changing conditions. However, in the last years, deep learning models have demonstrated to be able to identify and learn meaningful features that are more robust and representative of the underlying patterns, enabling them to handle diverse and changeable conditions effectively.In this work, we propose a pepper instance segmentation solution based on deep learning after harvest under field conditions. We implement the method and validate it for three pepper varieties: Blocky Bell, Jalapeño and Lamuyo. We compare the performance of this new method for each variety with a previous solution based on classical image processing techniques, with the objective of measuring and demonstrating the superiority of deep learning-based instance segmentation over traditional methods as a first step for phenotyping.The instance segmentation deep learning based models outperform the results obtained by classical image processing algorithms for the three pepper varieties: in Blocky Bell mAP is increased from 0.63 to 0.97, in Jalapeño from 0.39 to 0.52 and in Lamuyo from 0.67 to 0.97.

The main objective of this study was to monitor apricot development and ripening through gene expression analysis of key candidate genes using the RT-qPCR technique. Eight apricot cultivars were selected to analyze phenological and genetic patterns from pre-ripening stages through to postharvest. In addition, 19 selected genes were analyzed in the contrasting cultivars ‘Cebas Red’ and ‘Rojo Pasión’ in different stages (two preharvest stages S1 and S2, one harvest stage S3, and two postharvest stages S4 and S5). This pool of genes included genes related to fruit growth and ripening, genes associated with fruit color, and genes linked to the fruit’s nutraceutical aspects. Among the studied genes, Polygalacturonase (PG), Pectin methylesterase (PME), Aminocyclopropane-1-carboxylate synthase (ACS), and Myo-inositol-1-phosphate synthase (INO1) were directly related to fruit maturation and quality. Significant differential expression was observed between the cultivars, which correlated with variations in firmness, shelf life, and sensory characteristics of the apricots. ‘Rojo Pasión’ displayed high levels of PG, associated with rapid maturation and shorter postharvest shelf life, whereas ‘Cebas Red’ exhibited lower levels of this gene, resulting in greater firmness and extended shelf life. Genes CCD4, CRTZ, and ZDS, related to carotenoids, showed varied expression patterns during growth and postharvest stages, with higher levels in ‘Rojo Pasión’. On the other hand, Sucrose synthase (SUSY) and Lipoxygenase (LOX2) were prominent during the postharvest and growth stages, respectively. Additionally, GDP-L-galactose phosphorylase (VTC2_5) was linked to better postharvest performance. This research provides valuable insights for future breeding initiatives aimed at enhancing the quality and sustainability of apricot cultivation.

CRISPR (short for “Clustered Regularly Interspaced Short Palindromic Repeats”) is a technology that research scientists use to selectively modify the DNA of living organisms. CRISPR was adapted for use in the laboratory from the naturally occurring genome-editing systems found in bacteria. In this work, we reviewed the methods used to introduce CRISPR/Cas-mediated genome editing into fruit species, as well as the impacts of the application of this technology to activate and knock out target genes in different fruit tree species, including on tree development, yield, fruit quality, and tolerance to biotic and abiotic stresses. The application of this gene-editing technology could allow the development of new generations of fruit crops with improved traits by targeting different genetic segments or even could facilitate the introduction of traits into elite cultivars without changing other traits. However, currently, the scarcity of efficient regeneration and transformation protocols in some species, the fact that many of those procedures are genotype-dependent, and the convenience of segregating the transgenic parts of the CRISPR system represent the main handicaps limiting the potential of genetic editing techniques for fruit trees. Finally, the latest news on the legislation and regulations about the use of plants modified using CRISPR/Cas systems has been also discussed.

No natural sources of resistance to Plum pox virus (PPV, sharka disease) have been identified in peach. However, previous studies have demonstrated that grafting a “Garrigues” almond scion onto “GF305” peach rootstock seedlings heavily infected with PPV can progressively reduce disease symptoms and virus accumulation. Furthermore, grafting a “Garrigues” scion onto the “GF305” rootstock has been shown to completely prevent virus infection. This study aims to analyse the rewiring of gene expression associated with this resistance to PPV transmitted by grafting through the phloem using RNA-Seq and RT-qPCR analysis. A total of 18 candidate genes were differentially expressed after grafting “Garrigues” almond onto healthy “GF305” peach. Among the up-regulated genes, a HEN1 homolog stands out, which, together with the differential expression of RDR- and DCL2-homologs, suggests that the RNA silencing machinery is activated by PPV infection and can contribute to the resistance induced by “Garrigues” almond. Glucan endo-1,3-beta D-glucosidase could be also relevant for the “Garrigues”-induced response, since its expression is much higher in “Garrigues” than in “GF305”. We also discuss the potential relevance of the following in PPV infection and “Garrigues”-induced resistance: several pathogenesis-related proteins; no apical meristem proteins; the transcription initiation factor, TFIIB; the speckle-type POZ protein; in addition to a number of proteins involved in phytohormone signalling.

Kiwifruit (Actinidia chinensis var. deliciosa (A Chev) Liang et Ferguson) is a sub-tropical vine species from the Actinidiaceae family native to China. This species has an allohexaploid genome (from diploid and autotetraploid parents), contained in 174 chromosomes producing a climacteric and fleshy fruit called kiwifruit. Currently, only a small body of transcriptomic and proteomic data are available for A. chinensis var. deliciosa. In this low molecular knowledge context, the main goal of this study is to construct a tissue-specific de novo transcriptome assembly, generating differential expression analysis among these specific tissues, to obtain new useful transcriptomic information for a better knowledge of vegetative, floral and fruit growth in this species. In this study, we have analyzed different whole transcriptomes from shoot, leaf, flower bud, flower and fruit at four development stages (7, 50, 120 and 160 days after flowering; DAF) in kiwifruit obtained through RNA-seq sequencing. The first version of the developed A. chinensis var. deliciosa de novo transcriptome contained 142,025 contigs (x¯ = 1044 bp, N50 = 1133 bp). Annotation was performed with BLASTX against the TAIR10 protein database, and we found an annotation proportion of 35.6% (50,508), leaving 64.4% (91,517) of the contigs without annotation. These results represent a reference transcriptome for allohexaploid kiwifruit generating a database of A. chinensis var. deliciosa genes related to leaf, flower and fruit development. These results provided highly valuable information identifying over 20,000 exclusive genes including all tissue comparisons, which were associated with the proteins involved in different biological processes and molecular functions.

Prunus species are important crops in temperate regions. In these regions, drought periods are predicted to occur more frequently due to climate change. In this sense, to reduce the impact of climate warming, obtaining new tolerant/resistant cultivars and rootstocks is a mandatory goal in Prunus breeding. Therefore, the current study assembled three Prunus species including almond, (P. dulcis Mill D.A. Webb), apricot (P. armeniaca L.) and peach (P. persica L.) to model the temporal effects of drought. A hybrid peach × almond and a wild almond-relative species Prunus webbii were also included in the study. Physiological traits associated with photosynthetic activity, leaf water status, and chlorophyll content were assessed under three watering treatments. Results showed that effects of time, genotype, and treatment interact significantly in all traits. In addition, results confirmed that P. webbii have a greater tolerance to drought than commercial rootstocks. However, “Real Fino” apricot showed the fastest recovery after re-irrigation while being one of the most affected cultivars. In addition, from the better response to these watering treatments by the almond genotypes, two different trends were observed after re-irrigation treatment that clearly differentiate the response of the almond cultivar “Garrigue” from the rest of Prunus genotypes. A better characterization of the short-term drought response in Prunus, an accurate and more efficient evaluation of the genotype effect was obtained from the use of mixed models considering appropriate variance–covariance structures. Although the advantages of these approaches are rarely used in Prunus breeding, these methodologies should be undertaken in the future by breeders to increase efficiency in developing new breeding materials.

In this study, the control of eco- and endo-dormancy release led to the modulation of the flowering time in almond (Prunus dulcis (Mill.) D.A. Webb). The study was performed in almond cultivars with contrasting flowering times: the extra-early flowering cultivar Desmayo Largueta and the ultra-late cultivar Tardona. Temperature control in the “Autumn”, “Winter” and “Spring” chambers successfully delayed the flowering time in Desmayo Largueta. Advance flowering in the cultivar Tardona was limited, however, even with the application of sufficient chill in the Winter chamber. The ecodormancy period and the heat accumulation for flowering were not stable among cultivars, even though the heat accumulation was generally high, in accordance with that accumulated in field conditions. The heat requirements of the early cultivar Desmayo Largueta were lower than those of the ultra-late cultivar Tardona. We observed a decreasing pattern in ecodormancy along treatments that was probably related to the temperature in the Spring chamber. Finally, flowering and fruit set were highly variable, and these parameters were more dependent on the cultivar assayed than on the treatment applied. Although the ripening time under our experimental conditions was earlier than the phenological dates observed in the field, the flowering time delayed the ripening time in the case of the extra-early cultivar Desmayo Largueta. The fruit weight increased in the last treatments, whereas the kernel/fruit ratio decreased, as the kernel weight did not vary significantly along treatments. The results obtained show that flowering time can be modulated by temperature control and that other uncontrolled factors, such as photoperiod, can be involved in the control of endodormancy release and flowering time, especially in late flowering cultivars.

Genotyping by sequencing (GBS) provides a large quantity of useful data suitable for the identification of single nucleotide polymorphisms (SNPs), facilitating accurate genomic studies in plant species. In this study, GBS-based SNPs were used to characterise 11 Japanese plum cultivars and to explore their natural allelic diversity in relation to the most important phenology events (flowering date, ripening date and fruit development period) and fruit quality traits (weight, shape, skin and flesh colour, over colour, skin and flesh chlorophyll index, flesh firmness and soluble solids concentration). GBS-based SNPs were shown to be a powerful tool for genetic diversity and other genomic studies where SNP markers were related to several traits, particularly for flowering date, ripening date, fruit development period, skin chlorophyll degradation, flesh chlorophyll degradation and flesh colour. Forster's Terns ( Sterna forsteri) breed mainly in central Canada and the north-central USA, and along both coasts of North America. In Mexico, only five nesting colonies were known prior to this study and all are coastal, three from Baja California, one from Colima on the Pacific coast, and one from Tamaulipas on the Gulf of Mexico. In 2014, a new inland colony was found at the Lake of Texcoco, east of Mexico City, Mexico. Eight nests that were located on small islets 400 m away from shore were monitored from April to June 2014. Almost 90% of the eggs ( n = 16) were lost by flooding, and only two chicks hatched. One chick died and the other survived to fledging, and at 25 days old it dispersed more than 3 km from the colony. This nesting colony at the Lake of Texcoco represents a new breeding site of Forster's Tern in Mexico, the first breeding record in an interior wetland of Mexico, and a southward expansion of the known breeding range for this species. These observations reinforce the importance of the Lake of Texcoco for breeding waterbirds in the Valley of Mexico. [ABSTRACT FROM AUTHOR]

Abstract: During the breeding programs for Plum pox virus (PPV, Sharka) resistance in Prunus, the evaluation of the new releases through symptoms observation on leaves has been contradictory and represents one of the main handicaps in these programs. In order to increase the accuracy of this traditional evaluation method, we here analyze an alternative method based on the study of the ability of a genotype to allow the long-distance movement of the virus through its vascular vessels. Two different plant models have been assayed: (a) in Model I, the inoculation was performed in the ‘GF305’ rootstock with a later grafting of the genotype under evaluation and a scion of healthy control ‘GF305’, to evaluate the long-distance movement through the studied genotype from the rootstock to the scion (xylem transport), and (b) in Model II, the inoculation with ‘GF305’ diseased scions was performed by grafting these diseased scions onto the studied genotypes, which were grafted onto healthy ‘GF305’ peach seedlings, to evaluate the long-distance movement through the studied genotype from the scion to the rootstock (phloem transport). The results show that, regardless of the presence of symptoms, susceptible genotypes allowed the movement of the virus through their vascular vessels in both directions studied. However, the resistant apricot ‘Stark Early Orange’ did not allow this movement. We propose the study of the ability of a genotype to allow the long-distance movement of the virus as an alternative and more accurate method for the evaluation of PPV resistance. However, this protocol is much more tedious than the traditional one and could be used mainly in the evaluation of a reduced number of more interesting genotypes. [Copyright &y& Elsevier]

This work describes, for the first time, the changes taking place in the antioxidative system of the leaf apoplast in response to plum pox virus (PPV) in different Prunus species showing different susceptibilities to PPV. The presence of p-hydroxymercuribenzoic acid (pHMB)-sensitive ascorbate peroxidase (APX) (class I APX) and pHMB-insensitive APX (class III APX), superoxide dismutase (SOD), peroxidase (POX), NADH-POX, and polyphenoloxidase (PPO) was described in the apoplast from both peach and apricot leaves. PPV infection produced different changes in the antioxidant system of the leaf apoplast from the Prunus species, depending on their susceptibility to the virus. In leaves of the very susceptible peach cultivar GF305, PPV brought about an increase in class I APX, POX, NADH-POX, and PPO activities. In the susceptible apricot cultivar Real Fino, PPV infection produced a decrease in apoplastic POX and SOD activities, whereas a strong increase in PPO was observed. However, in the resistant apricot cultivar Stark Early Orange, a rise in class I APX as well as a strong increase in POX and SOD activities was noticed in the apoplastic compartment. Long-term PPV infection produced an oxidative stress in the apoplastic space from apricot and peach plants, as observed by the increase in H2O2 contents in this compartment. However, this increase was much higher in the PPV-susceptible plants than in the resistant apricot cultivar. Only in the PPV-susceptible apricot and peach plants was the increase in apoplastic H2O2 levels accompanied by an increase in electrolyte leakage. No changes in the electrolyte leakage were observed in the PPV-inoculated resistant apricot leaves, although a 42% increase in the apoplastic H2O2 levels was produced. Two-dimensional electrophoresis analyses revealed that the majority of the polypeptides in the apoplastic fluid had isoelectric points in the range of pI 4–6. The identification of proteins using MALDI-TOF (matrix-assisted laser desorption/ionization-time of flight) and peptide mass fingerprinting analyses showed the induction of a thaumatin-like protein as well as the decrease of mandelonitrile lyase in peach apoplast due to PPV infection. However, most of the selected polypeptides showed no homology with known proteins. This fact emphasizes that, at least in Prunus, most of the functions of the apoplastic space remain unknown. It is concluded that long-term PPV infection produced an oxidative stress in the leaf apoplast, contributing to the deleterious effects produced by PPV infection in leaves of inoculated, susceptible Prunus plants. The article presents a letter detailing a record of a Common Buzzard or Buteo buteo nesting in an abandoned building in the vicinity of Mengibar in southeastern Spain. It informs that the choice of the site for nesting was exceptional as there were no previous record of such a nesting site and as the nest lacked protection from weather or predators like rabbits. The article also describes that buzzards are sedentary, medium-sized raptors that are abundant on the Iberian Peninsula and usually nest in deciduous trees.

Abstract Sharka, a disease caused by Plum pox virus (PPV), mainly affects some Prunus species, including apricot, peach and plum, and to a lesser degree, sweet cherry and sour cherry. In almond, different PPV isolates have been transmitted experimentally to the ‘Aï’ cultivar. In this study, the resistance of 10 almond cultivars to a Dideron PPV isolate was evaluated in controlled conditions by grafting the cultivars on to inoculated GF305 peach rootstocks. The results demonstrated a high level of resistance to PPV in all the almond cultivars assayed. They did not show any symptoms and were ELISA and RT-PCR negative, despite the strong symptoms observed in their GF305 rootstocks. The implications of these results for the dispersion of PPV, and the potential role of almond as a source of resistance to PPV in other Prunus species such as peach, are also discussed. Abstract Prunus mandshurica [(Maxim.) Koehne] from Central Asia is a species related to apricot and grown in China and Mongolia. This species has been used in apricot breeding as a source of frost resistance. In addition, P. mandshurica has been suggested as the possible origin of some North American apricot cultivars resistant to sharka (Plum pox virus, PPV). The aim of this work was to transmit the resistance to PPV from P. mandshurica to the Spanish apricot cultivar ‘Currot’ by traditional crossing. The resistance to a Dideron PPV isolate of the descendants of P. mandshurica × ‘Currot’ and their progenitors was evaluated under controlled conditions in a greenhouse. The results showed the susceptibility of both, the progenitors and the offspring to PPV, as being much higher than in other apricot seedlings. The results showed that the P. mandshurica accession studied is not a good progenitor in breeding apricot for PPV resistance, but just the opposite. The possible role of P. mandshurica as a source of resistance in apricot resistant cultivars is questioned. [ABSTRACT FROM AUTHOR]

Abstract: The resistance/susceptibility level of 213 descendants from three different crosses between the French apricot cultivar ‘Polonais’ (susceptible to Plum pox virus, sharka), and the North American cultivar ‘Stark Early Orange’ (resistant) was evaluated during four cycles of study under controlled greenhouse conditions. Resistant:susceptible ratios were 83:17 in the case of the ‘Stark Early Orange’ open-pollination descendants, 62:38 in ‘Polonais’בStark Early Orange’ descendants, and 28:72 in ‘Polonais’בPolonais’. These ratios were checked against the expected ratios of different genetic control hypotheses, combining one, two or three genes involved in the expression of this trait and with the resistance being a dominant (Hypothesis A) or recessive trait (Hypothesis B). In addition, two other, more complex hypotheses were proposed: that the resistance is controlled by 2 independent genes, the resistance being recessive for one and dominant for the other (Hypothesis C), and another hypothesis considering the resistance to be controlled by one dominant gene and admitting a 25% error in the evaluation process (Hypothesis D). The χ 2-test was applied to compare the goodness-of-fit of the proposed hypotheses. Hypothesis D seems to be the most consistent in the three apricot crosses studied. [Copyright &y& Elsevier]

This study aimed at the monitoring of the apricot (Prunus armeniaca L.) ripening progression through the expression analysis of 25 genes related to fruit quality traits in nine cultivars with great differences in fruit color and ripening date. The level of pigment compounds, such as anthocyanins and carotenoids, is a key factor in food taste, and is responsible for the reddish blush color or orange skin and flesh color in apricot fruit, which are desirable quality traits in apricot breeding programs. The construction of multiple linear regression models to predict anthocyanins and carotenoids content from gene expression allows us to evaluate which genes have the strongest influence over fruit color, as these candidate genes are key during biosynthetic pathways or gene expression regulation, and are responsible for the final fruit phenotype. We propose the gene CHS as the main predictor for anthocyanins content, CCD4 and ZDS for carotenoids content, and LOX2 and MADS-box for the beginning and end of the ripening process in apricot fruit. All these genes could be applied as RNA markers to monitoring the ripening stage and estimate the anthocyanins and carotenoids content in apricot fruit during the ripening process.

DNA methylation and histone post-translational modifications have been described as epigenetic regulation mechanisms involved in developmental transitions in plants, including seasonal changes in fruit trees. In species like almond (Prunus dulcis (Mill.) D.A: Webb), prolonged exposure to cold temperatures is required for dormancy release and flowering. Aiming to identify genomic regions with differential methylation states in response to chill accumulation, we carried out Illumina reduced-representation genome sequencing on bisulfite-treated DNA from floral buds. To do this, we analyzed almond genotypes with different chilling requirements and flowering times both before and after dormancy release for two consecutive years. The study was performed using epi-Genotyping by Sequencing (epi-GBS). A total of 7317 fragments were sequenced and the samples compared. Out of these fragments, 677 were identified as differentially methylated between the almond genotypes. Mapping these fragments using the Prunus persica (L.) Batsch v.2 genome as reference provided information about coding regions linked to early and late flowering methylation markers. Additionally, the methylation state of ten gene-coding sequences was found to be linked to the dormancy release process.

For fruit tree (Prunus) species, flower bud dormancy completion determines the quality of bud break and the flowering time. In the present climate change and global warming context, the relationship between dormancy and flowering processes is a fundamental goal in molecular biology of these species. In almond [P. dulcis (Miller) Webb], flowering time is a trait of great interest in the development of new cultivars adapted to different climatic areas. Late flowering is related to a long dormancy period due to high chilling requirements of the cultivar. It is considered a quantitative and highly heritable character but a dominant gene (Late bloom, Lb) was also described. A major QTL (quantitative trait loci) in the linkage group (LG) 4 was associated with Lb, together with other three QTLs in LG1 and LG7. In addition, DAM (Dormancy-Associated MADS-Box) genes located in LG1 have been largely described as a gene family involved in bud dormancy in different Prunus species including peach [P. persica (L.) Batsch] and Japanese apricot (P. mume Sieb. et Zucc.). In this work, a DAM transcript was cloned and its expression was analysed by qPCR (quantitative Polymerase Chain Reaction) in almond flower buds during the dormancy release. For this purpose two almond cultivars (‘Desmayo Largueta’ and ‘Penta’) with different chilling requirements and flowering time were used, and the study was performed along two years. The complete coding sequence, designated PdDAM6 (Prunus dulcis DAM6), was subjected to a phylogenetic analysis with homologous sequences from other Prunus species. Finally, expression dynamics analysed by using qPCR showed a continuous decrease in transcript levels for both cultivars and years during the period analysed. Monitoring almond flower bud dormancy through DAM expression should be used to improve almond production in different climate conditions.

Plum pox virus (PPV, sharka) is a limiting factor for peach production, and no natural sources of resistance have been described. Recent studies, however, have demonstrated that grafting the almond cultivar “Garrigues” onto the “GF305” peach infected with Dideron-type (PPV-D) isolates progressively reduces disease symptoms and virus accumulation. Furthermore, grafting “Garrigues” onto “GF305” prior to PPV-D inoculation has been found to completely prevent virus infection, showing that resistance is constitutive and not induced by the virus. To unravel the phytohormone signaling of this mechanism, we analyzed the following phytohormones belonging to the principal hormone classes: the growth-related phytohormones cytokinin trans-zeatin (tZ) and the gibberellins GA3 and GA4; and the stress-related phytohormones ethylene acid precursor 1-aminocyclopropane-1-carboxylic acid (ACC), abscisic acid (ABA), salicylic acid (SA), and jasmonic acid (JA). PPV inoculation produced a significant increase in GA3 and ABA in peach, and these imbalances were related to the presence of chlorosis symptoms. However, grafting “Garrigues” almond onto the PPV-inoculated “GF305” peach produced the opposite effect, reducing GA3 and ABA contents in parallel to the elimination of symptoms. Our results showed the significant implication of SA in this induced resistance in peach with an additional effect on tZ and JA concentrations. This SA-induced resistance based in the decrease in symptoms seems to be different from Systemic Acquired Resistance (SAR) and Induced Systemic Resistance (ISR), which are based in other reactions producing necrosis. Further studies are necessary, however, to validate these results against PPV-D isolates in the more aggressive Marcus-type (PPV-M) isolates.

South American histricognath rodents Thrichomys apereoides laurentius and Thrichomys pachyurus are natural hosts of Trypanosoma cruzi, agent of Chagas disease. We established breeding colonies of these species to serve as experimental models in various parasitological studies. Both species of Thrichomys have all the requirements necessary to become excellent laboratory models: they can be easily maintained in the standard laboratory conditions and breed throughout the year and they do not have any special dietary demands and can be fed by standard food pellets designed for laboratory mice. Both species produce precocious offspring that have their eyes and ears open, teeth erupted, fur well developed, and can eat solid food in the first week of life. T. a. laurentius has larger litter sizes and lower body masses at birth and weaning than T. pachyurus. Moreover, females of T. a. laurentius reach puberty earlier and with lower body mass than T. pachyurus.

Flowering time is an important agronomic trait in almond since it is decisive to avoid the late frosts that affect production in early flowering cultivars. Evaluation of this complex trait is a long process because of the prolonged juvenile period of trees and the influence of environmental conditions affecting gene expression year by year. Consequently, flowering time has to be studied for several years to have statistical significant results. This trait is the result of the interaction between chilling and heat requirements. Flowering time is a polygenic trait with high heritability, although a major gene Late blooming (Lb) was described in ‘Tardy Nonpareil’. Molecular studies at DNA level confirmed this polygenic nature identifying several genome regions (Quantitative Trait Loci, QTL) involved. Studies about regulation of gene expression are scarcer although several transcription factors have been described as responsible for flowering time. From the metabolomic point of view, the integrated analysis of the mechanisms of accumulation of cyanogenic glucosides and flowering regulation through transcription factors open new possibilities in the analysis of this complex trait in almond and in other Prunus species (apricot, cherry, peach, plum). Abstract: Sixteen nuclear and 10 chloroplast SSR markers were evaluated for 40 almond genotypes including cultivated almond, 18 related species and 5 interspecific-hybrid populations. Results establish the value of SSR (nuclear and chloroplast) markers for distinguishing different genetic lineages and characterize an extensive gene pool available to almond genetic improvement. Hierarchical analysis using integrated nuclear and chloroplast DNA markers support Prunus fenzliana, a species native to the northeast Iran, as a probable ancestor of the cultivated almond. Results also established the importance of interspecific hybridization and subsequent genetic introgression in the development of cultivated almond and demonstrate continuing value of an interspecific gene pool for modern cultivar improvement. Molecular results implicate a dissemination of the cultivated almond from Asia to the Eastern Mediterranean and subsequently the Western Mediterranean and the New World is supported by the molecular analysis of regional germplasm. Abstract: In this study we compare the evaluation of Plum pox virus (Sharka) resistance of 29 apricot genotypes in controlled greenhouse conditions by grafting onto infected ‘GF305’ peach seedlings growing in pots, and in natural conditions by grafting onto infected 5-year-old apricot trees growing in the orchard. Apricot genotypes evaluated were initially classified into three groups: susceptible to PPV (presence of PPV symptoms and ELISA positive in greenhouse and field assays), resistant (absence of PPV symptoms and ELISA negative in both assays) and undetermined (evaluated differently in both assays). Results showed a similar behavior against Plum pox virus in both assays in 20 out of the 29 apricot genotypes studied (69%). However, in the other nine genotypes results were different (31%). Evaluation in field was more accurate, detecting a higher number of susceptible genotypes, probably due to the higher inoculation pressure in the old trees in comparison with the GF305 rootstocks in pots. However, greenhouse evaluation let to work in controlled isolated conditions with a higher number of genotypes. This situation is greatly required in areas where Sharka is not widely spread. Greenhouse evaluation could be then the first screening method to evaluate Plum pox virus resistance of apricot genotypes, and could be complemented with the evaluation onto infected trees in natural conditions in insect-proof quarantine shelter. Abstract: Seventeen peach simple sequence repeat (SSR) markers have been used in the molecular characterization of 8 apricot (Prunus armeniaca L.) cultivars from Spain, North America, France, and Greece; a new breeding line from the apricot breeding program of INRA (Avignon, France); and 13 breeding lines and 3 new releases from the apricot breeding program of CEBAS-CSIC (Murcia, Spain). DNA fingerprints have been developed establishing the genetic relatedness among cultivars, new releases, and breeding lines. Amplification of SSR loci was obtained for all 17 primer pairs and 14 of them produced polymorphic amplification. The number of presumed alleles revealed by the SSR analysis ranged from one to nine, although most primers revealed three alleles or more. The mean number of alleles per locus was 4.1. Results allowed the molecular identification of all the apricot genotypes assayed. Apricot genotypes clustered into seven principal groups in accordance with their origin and pedigree. The implications of these results for apricot breeding programs in terms of protection of new release and design of new crosses are also discussed. [Copyright &y& Elsevier]
