Bradykinin (BK) isolated from plasma of the African lungfish, Protopterus annectens, contains four amino acid substitutions compared with BK from mammals (Arg(1)-->Tyr, Pro(2)-->Gly, Pro(7)-->Ala, Phe(8)-->Pro). Bolus intra-arterial injections of synthetic lungfish BK (1-1000 pmol/kg body wt.) into unanaesthetised, juvenile lungfish (n=5) produced a dose-dependent increase in arterial blood pressure and pulse pressure. The maximum pressor response occurred 2-3 min after injection and persisted for up to 15 min. The threshold dose producing a significant (P<0.01) rise in pressure was 50 pmol/kg and the maximum increase, following injection of 300 pmol/kg, was 9.3 +/- 2.3 mmHg. Injection of the higher doses of lungfish BK produced a significant (P<0.05) increase in heart rate (2.8 +/- 0.8 beats/min at 100 pmol/kg). In contrast, bolus intra-arterial injections of mammalian BK, in doses up to 1000 pmol/kg, produced no significant cardiovascular effects in the lungfish. The data support the existence of a functioning kallikrein-kinin system in the lungfish and demonstrate that the ligand-binding properties of the receptor(s) mediating the cardiovascular actions of lungfish BK are appreciably different from mammalian B1 and B2 receptors.

Incubation of heat-denatured plasma from six species occupying different evolutionary positions within the Sarcopterygian lineage [the dipnoan, Protopterus annectens (African lungfish); the urodele, Amphiuma tridactylum (three-toed amphiuma); the colubrid snakes, Pituophis melanoleucus sayi (bullsnake) and Masticophis flagellum (coachwhip); and the lizards Heloderma suspectum (Gila monster) and Varanus Grayi (Gray's monitor)] with trypsin generated bradykinin-related peptides that were detected by radioimmunoassay using an antiserum raised against mammalian bradykinin (BK). The peptides were purified by HPLC and their primary structures were established as lungfish [Tyr1,Gly2,Ala7,Pro8]BK, amphiuma [Phe1,Ile2, Leu5]BK, bullsnake and coachwhip [Val1,Thr6]BK, Gila monster [Leu2, Thr6]BK, and Gray's monitor [Thr6]BK. Monitor BK is identical to the peptide generated in turtle and alligator plasma and coachwhip/bullsnake BK shows one amino acid substitution (Ala1 --> Val) compared with the peptide generated in the plasma of the python. The data provide further evidence for the widespread occurrence of a kallikrein-kininogen system in nonmammalian vertebrates but indicate that the primary structure of BK has been poorly conserved during evolution., (Copyright 1998 Academic Press.)

The Harderian gland of the terrapin Pseudemys scripta has four types of acinar cells. Type IV cells are very similar to the salt secreting cells of the salt secretory glands of various marine vertebrates. The presence and localization of the Ile5-Angiotensin II, Atrial Natriuretic Peptide, and Bradykinin has been investigated by immunohistochemical methods. Immunoreactivity is confined to the type IV cells. Changes in the environmental salinity resulted in different patterns in the immunoreactivity especially after incubation with Ab-Angiotensin II and Ab-Atrial Natriuretic Peptide. Immunoreactive Angiotensin II cells are more numerous in animals maintained in distilled water, when reabsorption of sodium is needed. In contrast, immunoreactive Angiotensin II cells are very few in animals maintained in seawater. On the contrary, the number of immunoreactive cells for Atrial Natriuretic Peptide is high in seawater maintained animals, and weaker in animals in distilled water. The type IV cell may be considered a candidate for ion regulation in the terrapin Harderian gland.
