In a detailed investigation of lichen diversity in different regions of Pakistan, utilizing molecular and morphological techniques, we identified and reported three species – Dimelaena altissima, Lecanora chlarotera, and Usnea lapponica – as new records for Pakistan. Furthermore, nrITS sequences for Evernia prunastri, Lecanora horiza, L. hybocarpa, Peltigera polydactylon, and P. praetextata were generated for the first time from Pakistan. Detailed morpho-anatomical descriptions, habitat information, distribution data, and results of biochemical analyses are also provided for a better understanding of the lichen diversity and ecology in this region.

Two species of Tricholoma , T. bonii and T. triste , are reported as new records for Pakistan. Phylogenetic analysis based on the nuclear ribosomal internal transcribed spacer (nrITS) region clustered the Pakistani collections with T. bonii and T. triste sequences from previous worldwide collections. Detailed descriptions of macro- and microcharacters, habitat, general distribution, and diagnostic features are presented.

Panaeolus punjabensis M. Asif, Q. Firdous, A. Izhar, Niazi & Khalid sp. nov. was collected from three different localities (Bahawalnagar, Kasur, and Lahore) in Punjab, Pakistan. Morphological observations and phylogenetic analyses based on nuclear encoded internal transcribed spacers (ITS1-5.8S-ITS2 = ITS) and D1/D2 domain of large subunit (28S) rDNA confirmed the taxonomic distinctness of this species. The new species is potentially hallucinogenic and characterized by a parabolic pileus with a light brown center, broadly fusiform basidiospores, presence of cheilocystidia, pileocystidia, and caulocystidia, and absence of pleurocystidia and clamp connections. The DNA sequences of the species clustered together in a well-supported distinct clade. We present a detailed description, photographs, and line drawings, and elucidate and discuss the phylogenetic position of the new species. Morphological comparisons with phylogenetically and morphologically allied species are discussed.

ABSTRACT Diploschistes pakistanicus sp. nov. is described from the Himalayan moist temperate forest, Pakistan. ITS sequences confirm its position within the genus Diploschistes and, together with its morphology and chemistry, suggest that it is separate from other species of this genus. The taxon is characterized by grey to greyish white pruinose thallus, perithecioid-type ascomata, small apothecia 0.1‒0.4 mm wide, hypothecium 20‒25µm thick, ascus of 85‒110 × 9‒17 µm in size, 3‒5 transverse and 2‒4 longitudinal septa in large ascospores 42‒55 × 18‒30; also differing from related species in ITS region.

A new species, Clitopilus cretoalbus A.Izhar, Zaman, M.Asif, H.Bashir, Niazi & Khalid sp. nov., is described herein based on several collections from Punjab, Pakistan. It is characterized by a clitocyboid to somewhat omphaloid stature combined with a white pileus, decurrent lamellae, an almost central to slightly eccentric whitish relatively long stipe, the occurrence of cheilocystidia, and basidiospores with 6 to 9 ridges in polar view. Molecular phylogenetic analyses of nrITS and nrLSU performed using the maximum likelihood method supported the novelty of this Pakistani species and its placement within the genus Clitopilus section Scyphoides. A comparison with other morpho-anatomically close species confirmed that the newly described species is distinct from others.

During a survey of mushrooms of Pakistan, Macrocybe gigantea was collected from University of the Punjab, Lahore, Pakistan under the Morus species. For the domestication of this wild species, its culturability and cultivation potential was assessed by using different synthetic culture media and substrates. Among these different media used, maximum cultural growth was observed on Potato Dextrose Agar (PDA) medium at 30 °C followed by Malt Extract Agar (MEA), Compost Extract Agar (CEA), Glucose Peptone Agar (GPA), and Saboraud Dextrose Agar (SDA). Strains on PDA medium were used for production of spawning material on wheat, sorghum and barleyngrains. Sorghum grains at 30 °C were the best combination for spawn production. A mixed substrate of wheat straw and Tea waste at 30 °C produced the highest yield. Mineral analysis of the wild and cultivated strain revealed that both forms enrich Potassium and Calcium. These findings show that this giant edible mushroom species could be domesticated on the number of media and substrates. Its domestication can provide nutritional, economical, medicinal and tasty food to the growing population that would otherwise be restricted to natural production at a specific time of the year.

ABSTRACT During our surveys of mushrooms in 2019 and 2020, an interesting species of the genus Lepiota was collected from three different areas of Punjab (Bahawalnagar, Sheikhupura, and Muzaffarghar), Pakistan. These areas vary in their altitude and somewhat climatic conditions, nevertheless, morpho-anatomical comparisons and molecular analyses proved that these collections represented the same taxon, described here as Lepiota brunneogranulosa. This new species is characterized by a pileus with a yellowish-brown central disc, with brown colored granules on a very pale brown background, dextrinoid, ellipsoid basidiospores, frequently bi-sporic basidia, and narrowly clavate cheilocystidia. Because of the hymeniform pileus covering, and the placement in the phylogeny, based on Internal Transcribed Spacer (nrITS) and Larger Subunit (LSU), it belongs to the Lepiota sect. Lilaceae.

ABSTRACT The newly described species, Lecidella iqbalii sp. nov., is so far only known from Azad Jammu and Kashmir, Pakistan. A comparative morpho-anatomical study combined with a phylogenetic analysis based on the marker ITS placed this species together with two accessions from southern South America in a distinct and highly supported group at the base of the Lecidella stigmatea clade. The taxon is characterized by a shiny thallus, strongly convex apothecia, a thin excipulum, a dark brown epihymenium and relatively small ascospores (10-) 11-12 (-13) × (6-) 6.5-8(-9) µm.

During mycological surveys of different areas of Punjab, Pakistan, we collected a new species from the genus Candolleomyces. This is the first report of any species of this genus from Pakistan. Candolleomyces asiaticus M.Asif, A.Izhar, Niazi & Khalid sp. nov. is characterized by ellipsoid to oblong-ellipsoid (8.2 × 5 µm) basidiospores with indistinct germ pores, lageniform to utriform cheilocystidia without adhering deposits at the apices, absence of pleuro-, pileo- and caulocystidia, pileipellis with spherical, subglobose to columnar cells, and presence of clamp connections. The morpho-anatomical and molecular phylogenetic analysis of nrITS revealed its distinct phylogenetic position, further supporting the recognition of a new species.
