The microvasculature facilitates gas exchange, provides nutrients to cells, and regulates blood flow in response to stimuli. Vascular abnormalities are an indicator of pathology for various conditions, such as compromised vessel integrity in small vessel disease and angiogenesis in tumors. Traditional immunohistochemistry enables the visualization of tissue cross-sections containing exogenously labeled vasculature. Although this approach can be utilized to quantify vascular changes within small fields of view, it is not a practical way to study the vasculature on the scale of whole organs. Three-dimensional (3D) imaging presents a more appropriate method to visualize the vascular architecture in tissue. Here we describe the complete protocol that we use to characterize the vasculature of different organs in mice encompassing the methods to fluorescently label vessels, optically clear tissue, collect 3D vascular images, and quantify these vascular images with a semi-automated approach. To validate the automated segmentation of vascular images, one user manually segmented one hundred random regions of interest across different vascular images. The automated segmentation results had an average sensitivity of 83±11% and an average specificity of 91±6% when compared to manual segmentation. Applying this procedure of image analysis presents a method to reliably quantify and characterize vascular networks in a timely fashion. The dialysis catheter indwelling in human bodies has a high risk of inducing thrombus and stenosis. Biomechanical research showed that such physiological complications are triggered by the wall shear stress of the vascular vessel. This study aimed to assess the impact of CVC implantation on central venous haemodynamics and the potential alterations in the haemodynamic environment related to thrombus development. The SVC structure was built from the images from computed tomography. The blood flow was calculated using the Carreau model, and the fluid domain was determined by CFD. The vascular wall and the CVC were computed using FEA. The elastic interaction between the vessel wall and the flow field was considered using FSI simulation. With consideration of the effect of coupling, it was shown that the catheter vibrated in the vascular systems due to the periodic variation of blood pressure, with an amplitude of up to 10% of the vessel width. Spiral flow was observed along the catheter after CVC indwelling, and recirculation flow appeared near the catheter tip. High OSI and WSS regions occurred at the catheter tip and the vascular junction. The arterial lumen tip had a larger effect on the WSS and OSI values on the vascular wall. Considering FSI simulation, the movement of the catheter inside the blood flow was simulated in the deformable vessel. Endothelial cells (ECs) line the wall of blood vessels and regulate arterial contractility to tune regional organ blood flow and systemic pressure. Chloride (Cl-) is the most abundant anion in ECs and the Cl- sensitive With-No-Lysine (WNK) kinase is expressed in this cell type. Whether intracellular Cl- signaling and WNK kinase regulate EC function to alter arterial contractility is unclear. Here, we tested the hypothesis that intracellular Cl- signaling in ECs regulates arterial contractility and examined the signaling mechanisms involved, including the participation of WNK kinase. Our data obtained using two-photon microscopy and cell-specific inducible knockout mice indicated that acetylcholine, a prototypical vasodilator, stimulated a rapid reduction in intracellular Cl- concentration ([Cl-]i) due to the activation of TMEM16A, a Cl- channel, in ECs of resistance-size arteries. TMEM16A channel-mediated Cl- signaling activated WNK kinase, which phosphorylated its substrate proteins SPAK and OSR1 in ECs. OSR1 potentiated transient receptor potential vanilloid 4 (TRPV4) currents in a kinase-dependent manner and required a conserved binding motif located in the channel C terminus. Intracellular Ca2+ signaling was measured in four dimensions in ECs using a high-speed lightsheet microscope. WNK kinase-dependent activation of TRPV4 channels increased local intracellular Ca2+ signaling in ECs and produced vasodilation. In summary, we show that TMEM16A channel activation reduces [Cl-]i, which activates WNK kinase in ECs. WNK kinase phosphorylates OSR1 which then stimulates TRPV4 channels to produce vasodilation. The deformability of blood vessels in one‐dimensional blood flow models is typically described through a pressure‐area relation, known as the tube law. The most used tube laws take into account the elastic and viscous components of the tension of the vessel wall. Accurately parametrizing the tube laws is vital for replicating pressure and flow wave propagation phenomena. Here, we present a novel mathematical‐property‐preserving approach for the estimation of the parameters of the elastic and viscoelastic tube laws. Our goal was to estimate the parameters by using ovine and human in vitro data, while constraining them to meet prescribed mathematical properties. Results show that both elastic and viscoelastic tube laws accurately describe experimental pressure‐area data concerning both quantitative and qualitative aspects. Additionally, the viscoelastic tube law can provide a qualitative explanation for the observed hysteresis cycles. The two models were evaluated using two approaches: (i) allowing all parameters to freely vary within their respective ranges and (ii) fixing some of the parameters. Microvasculature analysis is an important task in the medical field due to its various applications. It has been used for the diagnosis and threat of diseases in fields such as ophthalmology, dermatology, and neurology by measuring relative blood flow or blood vessel morphological properties. However, light scattering at the periphery of the blood vessel causes a decrease in contrast around the vessel borders and an increase in the noise of the image, making the localization of blood vessels a challenging task. Therefore, this work proposes integrating known information from the experimental setup into a deep learning architecture with multiple inputs to improve the generalization of a computational model for the segmentation of blood vessels and depth estimation in a single inference step. The proposed R-UNET + ET + LA obtained an intersection over union of 0.944 ± 0.065 and 0.812 ± 0.080 in the classification task for validation (in vitro) and test sets (in vivo), respectively, and a root mean squared error of 0.0085 ± 0.0275 μ m in the depth estimation. This approach improves the generalization of current solutions by pre-training with in vitro data and adding information from the experimental setup. A detailed study of the effect of coarctation on the blood flow of the thoracic aorta was carried out by modeling the interaction between blood and the aortic wall. Fluid-structure interaction (FSI) simulations were performed on the thoracic aorta geometries of healthy and diseased patients obtained from medical images. The pulsating flow of non-Newtonian blood was set by the boundary condition for the velocity in the form of a periodic physiological wave depending on time. As a result, the parameters of blood flow and vessel deformation were evaluated. It is shown that the velocity value at the site of aortic narrowing increases and falls after the narrowing. In addition, it can be noted that the value of pressure in the healthy aorta was higher than in the aorta with CoA, but at the same time, the pressure in the CoA model increased after the coarctation zone. It was noted that the shear stresses for the two models showed little difference and WSS was overestimated in the diseased aorta. As can be seen from the obtained data, using numerical simulation, important characteristics were determined to assess the influence of the fluid-structure interaction during blood flow in the flexible stenotic thoracic aorta. The endovascular neural interface provides an appealing minimally invasive alternative to invasive brain electrodes for recording and stimulation. However, stents placed in blood vessels have long been known to affect blood flow (haemodynamics) and lead to neointimal growth within the blood vessel. Both the stent elements (struts and electrodes) and blood vessel wall geometries can affect the mechanical environment on the blood vessel wall, which could lead to unfavourable vascular remodelling after stent placement. With increasing applications of stents and stent-like neural interfaces in venous blood vessels in the brain, it is necessary to understand how stents affect blood flow and tissue growth in veins. We explored the haemodynamics of a stent-mounted neural interface in a blood vessel model. Results indicated that blood vessel deformation and tapering caused a substantial change to the lumen geometry and the haemodynamics. The neointimal proliferation was evaluated in sheep implanted with an endovascular neural interface. Analysis showed a negative correlation with the mean Wall Shear Stress pattern. Objectives: Intrapulpal calcifications can occur in the dental pulp of patients with diabetes. We focused on the association between ectopic calcifications in the dental pulp and advanced glycation end products (AGEs) in Spontaneously Diabetic Torii (SDT)‐fatty rats, an obese type 2 diabetic rat model, to determine the mechanism of calcification with pulp stone in the dental pulp. Materials and Methods: Pathologic calcification in the dental pulp of SDT‐fatty rats was observed using electron microscopy and immunohistochemical analysis. Moreover, mechanical analysis of periapical region of molar tooth against occlusal force was performed. Results: In SDT‐fatty rats, pathogenic pulpal calcifications occurred during blood glucose elevation after 6 weeks, and granular calcification was observed in the dental pulp after 11 weeks. Pentosidine, a major AGE, and the receptor for AGEs were strongly expressed in the dental pulp of SDT‐fatty rats. S100A8, TNF‐α, and IL‐6 also showed positive response in the dental pulp of the SDT‐fatty rat, which indicated pulpal inflammation. Blood flow disorder and hypoxic dental pulp cells were also observed. In silico simulation, strain from occlusal force concentrates on the root apex. Conclusions: Glycation makes blood vessels fragile, and occlusal forces damage the vessels mechanically. These are factors for intrapulpal calcification of diabetes. [ABSTRACT FROM AUTHOR]

Endothelial cells line at the most inner layer of blood vessels. They act to control hemostasis, arterial tone/reactivity, wound healing, tissue oxygen, and nutrient supply. With age, endothelial cells become senescent, characterized by reduced regeneration capacity, inflammation, and abnormal secretory profile. Endothelial senescence represents one of the earliest features of arterial ageing and contributes to many age-related diseases. Compared to those in arteries and veins, endothelial cells of the microcirculation exhibit a greater extent of heterogeneity. Microcirculatory endothelial senescence leads to a declined capillary density, reduced angiogenic potentials, decreased blood flow, impaired barrier properties, and hypoperfusion in a tissue or organ-dependent manner. The heterogeneous phenotypes of microvascular endothelial cells in a particular vascular bed and across different tissues remain largely unknown. Accordingly, the mechanisms underlying macro- and micro-vascular endothelial senescence vary in different pathophysiological conditions, thus offering specific target(s) for therapeutic development of senolytic drugs. The reconstruction of blood velocity in a vessel from contrast enhanced x‐ray computed tomography projections is a complex inverse problem. It can be formulated as reconstruction problem with a partial differential equation constraint. A solution can be estimated with the a variational adjoint method and proper orthogonal decomposition (POD) basis. In this work, we investigate new inversion approaches based on PODs coupled with deep learning methods. The effectiveness of the reconstruction methods is shown with simulated realistic stationary blood flows in a vessel. Endothelial cells of mammalian blood vessels have multiple levels of heterogeneity along the vascular tree and among different organs. Further heterogeneity results from blood flow turbulence and variations in shear stress. In the aorta, vascular endothelial protein tyrosine phosphatase (VE-PTP), which dephosphorylates tyrosine kinase receptor Tie2 in the plasma membrane, undergoes downstream polarization and endocytosis in endothelial cells exposed to laminar flow and high shear stress. VE-PTP sequestration promotes Tie2 phosphorylation at tyrosine992 and endothelial barrier tightening. The present study characterized the heterogeneity of VE-PTP polarization, Tie2-pY992 and total Tie2, and claudin-5 in anatomically defined regions of endothelial cells in the mouse descending thoracic aorta, where laminar flow is variable and IgG extravasation is patchy. We discovered that VE-PTP and Tie2-pY992 had mosaic patterns, unlike the uniform distribution of total Tie2. Claudin-5 at tight junctions also had a mosaic pattern, whereas VE-cadherin at adherens junctions bordered all endothelial cells. Importantly, the amounts of Tie2-pY992 and claudin-5 in aortic endothelial cells correlated with downstream polarization of VE-PTP. VE-PTP and Tie2-pY992 also had mosaic patterns in the vena cava, but claudin-5 was nearly absent and extravasated IgG was ubiquitous. Correlation of Tie2-pY992 and claudin-5 with VE-PTP polarization supports their collective interaction in the regulation of endothelial barrier function in the aorta, yet differences between the aorta and vena cava indicate additional flow-related determinants of permeability. Pericytes (PCs) are located surrounding the walls of small blood vessels, particularly capillaries and microvessels. In addition to their functions in maintaining vascular integrity, participating in angiogenesis, and regulating blood flow, PCs also serve as a reservoir for multi-potent stem/progenitor cells in white, brown, beige, and bone marrow adipose tissues. Due to the complex nature of this cell population, the identification and characterization of PCs has been challenging. A comprehensive understanding of the heterogeneity of PCs may enhance their potential as therapeutic targets for metabolic syndromes or bone-related diseases. This mini-review summarizes multiple PC markers commonly employed in lineage-tracing studies, with an emphasis on their contribution to adipogenesis and functions in different adipose depots under diverse metabolic conditions. The reactive hyperaemia index (RHI) assesses endothelial function, with a proposed cut-off of <1.67 for prevalent endothelial dysfunction (ED). However, uncertainties remain about whether this cut-off is age-dependent and applicable in healthy individuals. We aimed to explore ED in relation to age within a large population-based cohort of young to middle-aged, healthy individuals. Within the Malmö Offspring Study, a total of 1812 subjects (50.9% women, mean age 48 ± 11 years) were included. Post-occlusion/pre-occlusion ratio of the pulsatile signal amplitudes in the non-dominant upper arm was used to calculate RHI by EndoPat®. ED was defined as RHI < 1.67. Multivariable regression models were used to explore associations between ED and age. Prevalent ED was found in 534 (29.5%) participants. In subjects aged ≤30 years, ED was present in 47.4% compared to 27.6% in subjects ≥30 years (p < 0.001). In multivariable logistic regression analyses, ED was associated with younger age (p < 0.001), higher BMI (p < 0.001) and current smoking (p < 0.001). No sex differences were observed. In a large healthy population, RHI < 1.67, an early marker of endothelial dysfunction, was more prevalent in younger individuals, implying that RHI might not be a suitable measure of endothelial function in individuals under 30 years of age. Our findings suggest that low RHI in young, healthy individuals may not necessarily indicate true ED but rather an artefact of the limited ability of young and healthy arteries to dilate post-occlusion. Therefore, the term "pseudo-ED" may be applicable to young individuals with low RHI values. What is the context? In this paper, we developed a mathematical model to simulate virus transport through a viscous background flow driven by the natural pumping mechanism. Two types of respiratory pathogens viruses (SARS-Cov-2 and Influenza-A) are considered in this model. The Eulerian–Lagrangian approach is adopted to examine the virus spread in axial and transverse directions. The Basset–Boussinesq–Oseen equation is considered to study the effects of gravity, virtual mass, Basset force, and drag forces on the viruses transport velocity. The results indicate that forces acting on the spherical and non-spherical particles during the motion play a significant role in the transmission process of the viruses. It is observed that high viscosity is responsible for slowing the virus transport dynamics. Small sizes of viruses are found to be highly dangerous and propagate rapidly through the blood vessels. Endothelial cells lining blood vessels are essential for maintaining vascular homeostasis and mediate several pathological and physiological processes. Mechanical stresses generated by blood flow and other biomechanical factors significantly affect endothelial cell activity. Here, we review how mechanical stresses, both in situ and in vitro, affect endothelial cells. We review the basic principles underlying the cellular response to mechanical stresses. Prediction of blood cell flow is known as the difficult research by reason of the complexity of blood vessel. In this study, considering the complex structure of blood vessels, a mechanical model for red blood cell (RBC) based on unstructured grid has been established to study the flow characteristics of RBCs in complex blood vessels. In the model, the strain‐energy function by Skalak is employed to model the shear elasticity and surface‐area conservation of the membrane, and the hinge spring is used to describe the forces originating from local bending of the membrane. The immersed boundary method is utilized to couple the interphase force. Using the model, the stretching test of RBC is compared with the experiment data, and the good agreement verified the validation of the present model. The morphology of red blood cell and the blood viscosity in micro‐vessel are studied. RBCs move with a symmetric shape (parachute shape) in small blood vessels, and the buckling instability is observed when the RBC flow slowly through a micro‐vessel or a converging–diverging capillary. When the vessel diameter is around 10 μm, the reverse Fahraeus‐Lindqvist effect is presented. The blood apparent viscosity shows linear increase with the blood hematocrit. Vasculature within the human body is arranged in a hierarchical manner, beginning with large macrovessels that taper into smaller mesovessels, and finally into the smallest microvessels. Replicating this hierarchical vasculature within engineered tissue constructs is a critical hurdle for the clinical translation of engineered tissues and organs. A major limitation to the development of hierarchical vasculature are the distinct and diverse fabrication strategies required for creating vasculature at each scale. New advances in fabrication strategies, particularly 3D bioprinting, are enabling the engineering of a multiscale vascular hierarchy within a single 3D tissue construct. Multidisciplinary collaboration and cooperation between experts in academia, industry, and government will be essential to address this grand challenge and further propel hierarchical vasculature toward clinical and commercial success. In human vascular anatomy, blood flows from the heart to organs and tissues through a hierarchical vascular tree, comprising large arteries that branch into arterioles and further into capillaries, where gas and nutrient exchange occur. Engineering a complete, integrated vascular hierarchy with vessels large enough to suture, strong enough to withstand hemodynamic forces, and a branching structure to permit immediate perfusion of a fluidic circuit across scales would be transformative for regenerative medicine (RM), enabling the translation of engineered tissues of clinically relevant size, and perhaps whole organs. In this paper, we study the blood flow through blood vessels of various radii (including the case of variable cross section as well as modeling the blood flow through venae and arteries). Two approaches are discussed in order to mimic the dependence of blood viscosity on red blood cells aggregation, which changes with the shear rate and position inside the vessel: Two microstructural parameters together with empirical constitutive equations as a characteristic of aggregation are proposed, namely the microinertia as well as the volume fraction of blood particles (erythrocytes, platelets and leukocytes). Consequently, the Navier–Stokes system of equations for an incompressible fluid is supplemented by a constitutive equation for the moment of inertia in one case and for the volume fraction in another. The problems are solved numerically by the finite volume method for vessels of various geometries in spatial description. A comparison with experimental data for a narrow capillary shows the efficiency of the proposed constitutive equations for describing blood flow. Also, velocity profiles are obtained on the basis of compiled empirical formula for various sections of a blood vessel of variable radius. The heart is widely acknowledged as the unique driver of blood circulation. Recently, we discovered a flow-driving mechanism that can operate without imposed pressure, using infrared (IR) energy to propel flow. We considered the possibility that, by exploiting this mechanism, blood vessels, themselves, could propel flow. We verified the existence of this driving mechanism by using a three-day-old chick-embryo model. When the heart was stopped, blood continued to flow for approximately 50 minutes, albeit at a lower velocity. When IR was introduced, the postmortem flow increased from ~41.1 ± 25.6 μm/s to ~153.0 ± 59.5 μm/s (n = 6). When IR energy was diminished under otherwise physiological conditions, blood failed to flow. The purpose of this study was to study the blood circulation in large blood vessels and in the abdominal organs during inflammation of the abdominal cavity. Using the method of rheograph and laser ultrasonic dopplerography, the blood flow velocity in the abdominal aorta and inferior vena cava was studied. The circulatory system maintains the stability of the body in violation of internal organs and tissues.In large blood vessels and organs of the abdominal cavity, inflammation leads to disruption of blood supply in the microvasculature of the vessel. The rate of blood flow in the main vessel in acute inflammation of the abdominal cavity decreased compared to the control group. The resistance index in the main vessel RI decreased by 38.3%, which is associated with an increase in the diameter of the main vessel. With inflammation of the abdominal cavity, an increase in the velocity of blood flow in the portal vein and abdominal aorta was recorded.As a result of a decrease in the tone of blood vessels after a violation of liver function, the blood supply to the abdominal cavity decreased. The decrease in the functional properties of arteries and veins led to a decrease in blood flow in the abdominal cavity, which, in turn, manifests itself as a result of the preservation of microcirculatory disorders. The pulmonary circulation is a low-pressure, low-resistance circuit whose primary function is to deliver deoxygenated blood to, and oxygenated blood from, the pulmonary capillary bed enabling gas exchange. The distribution of pulmonary blood flow is regulated by several factors including effects of vascular branching structure, large-scale forces related to gravity, and finer scale factors related to local control. Hypoxic pulmonary vasoconstriction is one such important regulatory mechanism. In the face of local hypoxia, vascular smooth muscle constriction of precapillary arterioles increases local resistance by up to 250%. This has the effect of diverting blood toward better oxygenated regions of the lung and optimizing ventilation–perfusion matching. However, in the face of global hypoxia, the net effect is an increase in pulmonary arterial pressure and vascular resistance. Pulmonary vascular resistance describes the flow-resistive properties of the pulmonary circulation and arises from both precapillary and postcapillary resistances. The pulmonary circulation is also distensible in response to an increase in transmural pressure and this distention, in addition to recruitment, moderates pulmonary arterial pressure and vascular resistance. Stroke occur mainly due to arterial thrombosis and rupture of cerebral blood vessels. Previous studies showed that blood flow-induced wall shear stress is an essential bio marker for estimating atherogenesis. It is a common practice to use computational fluid dynamics (CFD) simulations to calculate wall shear stress and to quantify blood flow. Reliability of predicted CFD results greatly depends on the accuracy of applied boundary conditions. Previously, the boundary conditions were estimated by varying values so that they matched the clinical data. It is applicable upon the availability of clinical data. Meanwhile, in most cases all that can be accessed are arterial geometry and inflow rate. Consequently, there is a need to devise a tool to estimate boundary values such as resistance and compliance of arteries. In this article, the hemodynamics of nanofluid flow through the modelled stenosis-aneurysm models in the presence of the catheter has been studied. The eight stenosis-aneurysm models are developed to mimic biological observations and thus make the model more realistic. The mathematical understanding helps in treating the stenosis in the blood vessel by targeting the unhealthy region to the drug, which is coated on nanoparticles. The catheter achieves the active drug release to the aimed organs by coating on the catheter surface, which adds additional benefits. In the present hemodynamic study, the blood is modeled as a couple stress fluid; as a result, the highly non-linear momentum, temperature, and concentration equations were obtained. The fluid flow equations' complexity is further increased by incorporating the variable viscosity effects that arose due to the suspension of nanoparticles. The resultant mathematical model is solved by using the homotopy perturbation method. The convergence of the perturbed solutions is studied and depicted the degree of deformation in the case of temperature and concentration. The effects of the porous nature of the stenosis, no-slip at the catheter surface, and the free slip at the blood vessel boundary in the non-stenotic region are also considered in the model. The essential physiological property like surface shear stress is computed, and various parameters' influence on shear stress is analyzed. The present analysis can be helpful in understanding the enhancement in mass dispersion and heat transfer in unhealthy blood vessels, which could be used for drug delivery in the treatment of stenotic conditions. Activation of Gq-type G protein-coupled receptors (GPCRs) gives rise to large cytosolic Ca2+ elevations in astrocytes. Previous in vitro and in vivo studies have indicated that astrocytic Ca2+ elevations are closely associated with diameter changes in the nearby blood vessels, which astrocytes enwrap with their endfeet. However, the causal relationship between astrocytic Ca2+ elevations and blood vessel diameter changes has been questioned, as mice with diminished astrocytic Ca2+ signaling show normal sensory hyperemia. We addressed this controversy by imaging cortical vasculature while optogenetically elevating astrocyte Ca2+ in a novel transgenic mouse line, expressing Opto-Gq-type GPCR Optoα1AR (Astro-Optoα1AR) in astrocytes. Blue light illumination on the surface of the somatosensory cortex induced Ca2+ elevations in cortical astrocytes and their endfeet in mice under anesthesia. Blood vessel diameter did not change significantly with Optoα1AR-induced Ca2+ elevations in astrocytes, while it was increased by forelimb stimulation. Next, we labeled blood plasma with red fluorescence using AAV8-P3-Alb-mScarlet in Astro-Optoα1AR mice. We were able to identify arterioles that display diameter changes in superficial areas of the somatosensory cortex through the thinned skull. Photo-stimulation of astrocytes in the cortical area did not result in noticeable changes in the arteriole diameters compared with their background strain C57BL/6. Together, compelling evidence for astrocytic Gq pathway-induced vasodiameter changes was not observed. Lampreys are blood-sucking vampires in marine environments. From a survival perspective, it is expected that the lamprey buccal gland exhibits a repository of pharmacologically active components to modulate the host's homeostasis, inflammatory and immune responses. By analyzing the metabolic profiles of 14 different lamprey tissues, we show that two groups of metabolites in the buccal gland of lampreys, prostaglandins and the kynurenine pathway metabolites, can be injected into the host fish to assist lamprey blood feeding. Prostaglandins are well-known blood-sucking-associated metabolites that act as vasodilators and anticoagulants to maintain vascular homeostasis and are involved in inflammatory responses. The vasomotor reactivity test on catfish aortic ring showed that kynurenine can also relax the blood vessels of the host fish, thus improving the blood flow of the host fish at the bite site. Finally, a lamprey spatial metabolomics database (https://www.lampreydb.com) was constructed to assist studies using lampreys as animal model. A spatial metabolomics study of the lampreys leads to the identification of the kynurenine pathway metabolites and prostaglandins in their buccal gland secretions (saliva), which are likely involved in promoting blood flow when feeding on host fishes. Fourteen healthy men (age 24 ± 2 years) underwent four 4‐min exercises at 80–90% of maximal workload (Wmax) interspaced by 3‐min active rest at 50–60% Wmax. Transcranial Doppler measured middle cerebral artery blood velocity (CBV). Systemic haemodynamics (Modelflow) and aortic pressure (AoP, general transfer function) were estimated from an invasively recorded brachial arterial pressure waveform. Using transfer function analysis, gain and phase between AoP and CBV (0.39–10.0 Hz) were calculated. Stroke volume, aortic pulse pressure and pulsatile CBV increased during exercise (time effect: P < 0.0001 for all), but a time‐domain index of aortic–cerebral pulsatile transition (pulsatile CBV/pulsatile AoP) decreased throughout the exercise bouts (time effect: P < 0.0001). Furthermore, transfer function gain reduced, and phase increased throughout the exercise bouts (time effect: P < 0.0001 for both), suggesting the attenuation and delay of pulsatile transition. The cerebral vascular conductance index (mean CBV/mean arterial pressure; time effect: P = 0.296), an inverse index of cerebral vascular tone, did not change even though systemic vascular conductance increased during exercise (time effect: P < 0.0001). The use of a stent to coil an aneurysm can alter the position of the main blood vessel and affect blood flow within the sac. This study thoroughly examines the impact of stent-induced changes on the risk of MCA aneurysm rupture. The research aims to assess the effects of coiling and vessel deformation on blood flow dynamics by comparing the OSI, WSS, and blood structure of two distinct MCA aneurysms to identify high-risk areas for hemorrhage. Computational fluid dynamics is used to model blood flow. Plaque reduces the conductivity of the blood vessel and its shape is more important than its quantity. We show that, for given quantity, the conductivity is maximal if the plaque forms a uniform layer next to the vessel wall and leaves a circular hole in the middle. Introduction: We aimed to describe physiological changes in endometrial blood flow (minute arterioles running through the endometrium) from ovulation to the mid‐luteal phase using superb microvascular imaging. Material and methods: The study involved 17 women (median age, 32.5 years; first to third interquartile range, 29.8–40.0 years) with regular menstrual cycles who were managed in our institute from 2020 to 2021. The uterus was delineated at the sagittal section using transvaginal ultrasonography incorporated with superb microvascular imaging. For each participant, a total of 28 cycles were observed; 17 cycles observed within one day of ovulation and the implantation period, 5–7 days (D5–7) after ovulation in the same cycle, and nine cycles in which only ovulation was observed, and two cycles in which only D5–7 was observed. Therefore, 26 and 19 images at ovulation and D5–7, respectively, were acquired. Endometrial blood flow was evaluated by depth of the vascular signal in the endometrium and categorized as follows: signals only in the basal layer of the endometrium (grade 1), reaching up to half the endometrium (grade 2), and covering the whole endometrium (grade 3). Changes in the grade of endometrial blood flow from ovulation to D5–7 after ovulation, and the relationship between the grade of endometrial blood flow and the endometrial thickness on ovulation and D5–7 after ovulation, were analyzed. Statistical significance was set at p < 0.05. Results: The endometrial blood flow from ovulation to D5–7 after ovulation during the same menstrual period showed a downgrade in 14 of 17 cycles (82.3%) and no change in the remaining three cycles (17.6%), indicating a decrease in the endometrial blood flow from ovulation to D5–7 after ovulation (p = 0.001). There were differences between the grade of endometrial blood flow and median endometrial thickness on ovulation (grade 1: 5.9 mm, grade 2: 9.1 mm, and grade 3: 11.2 mm); however, no differences in the endometrial thickness were found between the grades on D5–7 after ovulation. Conclusions: In the normal menstrual cycle, endometrial blood flow decreased from ovulation to the mid‐luteal phase, and the endometrial thickness in the ovulatory phase was related to the endometrial perfusion. Volumetric imaging of a mouse brain in vivo with one-photon and two-photon ultralong anti-diffracting (UAD) beam illumination was performed. The three-dimensional (3D) structure of blood vessels in the mouse brain were mapped to a two-dimensional (2D) image. The speed of volumetric imaging was significantly improved due to the long focal length of the UAD beam. Comparing one-photon and two-photon UAD beam volumetric imaging, we found that the imaging depth of two-photon volumetric imaging (80 μm) is better than that of one-photon volumetric imaging (60 μm), and the signal-to-background ratio (SBR) of two-photon volumetric imaging is two times that of one-photon volumetric imaging. Networks of neurons are the primary substrate of information processing. Conversely, blood vessels in the brain are generally viewed to have physiological functions unrelated to information processing, such as the timely supply of oxygen, and other nutrients to the neural tissue. However, recent studies have shown that cerebral microvessels, like neurons, exhibit tuned responses to sensory stimuli. Tuned neural responses to sensory stimuli may be enhanced with experience‐dependent Hebbian plasticity and other forms of learning. Hence, it is possible that the microvascular network might also be subject to some form of competitive learning rules during early postnatal development such that its fine‐scale structure becomes optimized for metabolic delivery to a given neural micro‐architecture. To explore the possibility of adaptive lateral interactions and tuned responses in cerebral microvessels, we modelled the cortical neurovascular network by interconnecting two laterally connected self‐organizing networks. The afferent and lateral connections of the neural and vascular networks were defined by trainable weights. By varying the topology of lateral connectivity in the vascular network layer, we observed that the partial correspondence of feature selectivity between neural and hemodynamic responses could be explained by lateral coupling across local blood vessels such that the central domain receives an excitatory drive of more blood flow and a distal surrounding region where blood flow is reduced. Critically, our simulations suggest a new role for feedback from the vascular to the neural network because the radius of vascular perfusion determines whether the cortical neural map develops into a clustered vs. salt‐and‐pepper organization. Takayasu arteritis (TAK) is a chronic granulomatous vasculitis with an unknown etiology that often manifests over time. It has a dismal prognosis and is currently clinically refractory. The theory of poisonous evil has drawn more attention in recent years in the treatment of autoimmune disorders. Poisonous evil tends to accumulate and transform into heat, which causes a variety of symptoms to gradually emerge. Our team believes that the main cause of TAK is the prolonged stagnation of poisonous evil, the transformation of heat into fire, and the damage to the vessels and collaterals. Blood stasis is the basic pathogenesis. This disease is located in the blood vessels, which is also closely related to the spleen and kidney. TAK also belongs to a deficiency in origin and excess in superficislity. Kidney and spleen deficiencies, and a lack of essence and blood, cause only partially filled blood vessels and restricted blood flow. The collection of evil qi and the transformation of heat-toxin culminates in the accumulation of heat-toxin and congestion of blood vessels. The illness persists and the evil qi remains and penetrates the internal organs, causing a lack of vital qi, and the viscera loses its nourishment. Unblocking the blood vessels and boosting blood circulation is the cornerstone of clinical treatment. (I) Introduction: The use of Doppler ultrasound allows us to indirectly assess the effect of increased intrarenal pressure on renal blood flow during retrograde intrarenal surgery (RIRS). On the basis of vascular flow spectra from selected blood vessels in the kidney, it is possible to determine Doppler parameters that reflect the renal perfusion status, which indirectly shows the degree of vasoconstriction and reflects the resistance of kidney tissue. (II) Materials and methods: A total of 56 patients were included in the study. The study assessed the changes of three Doppler parameters of intrarenal blood flow: resistive index-RI, pulsatility index-PI, and acceleration time-AT in the ipsilateral and contralateral kidneys during RIRS. The effects of mean stone volume, energy used, and pre-stenting were examined as predictors and calculated at two time intervals. (III) Results: The mean value of RI and PI was significantly higher in the ipsilateral kidney than in the contralateral kidney just after RIRS. The mean value of the acceleration time was not significantly different before and after RIRS. The values of all three parameters 24 h after the procedure were comparable to their values immediately after the RIRS. The size of the stone exposed to laser lithotripsy, the value of the energy used, and pre-stenting are not factors that significantly influence Doppler parameters during RIRS. (IV) Conclusions: The significant increase in RI and PI after RIRS in the ipsilateral kidney suggests a vasoconstriction of the interlobar arteries generated by increased intrarenal pressure during the procedure. The uterine artery pulsatility index (PI) assessed by Doppler ultrasound reflects the impedance to the blood flow in the vessel distal to the sampling point. We aimed to assess the accuracy of the uterine artery PI for the diagnosis of puberty in girls. A PRISMA-ScR-compliant scoping review was performed in the MEDLINE and Embase databases with the search terms "puberty" and "Doppler ultrasonography". Studies that included girls aged 0–18 years who underwent pelvic Doppler ultrasound with calculation of uterine artery PI were eligible. Ten studies comprising 1385 girls aged 1.2–18 years were included. The selected studies included participants from Italy, Brazil, Iran, Belgium and Denmark, and were published between 1996 and 2021. Six studies selected girls who were referred for evaluation of pubertal disorders, while four studies included only healthy girls. Nine studies found a significant difference in Doppler signal pattern and PI according to pubertal stage, with PI cutoff points ranging from 2.5 to 4.6 for the diagnosis of puberty, with a sensitivity of 77%–94%, specificity of 85%–100%, and accuracy of 79%–98%. Doppler assessment of the uterine arteries with PI calculation is a useful noninvasive tool in the diagnosis of pubertal onset in girls. [ABSTRACT FROM AUTHOR]

Coronary artery-to-pulmonary artery fistulae are a not uncommon finding in patients with Tetralogy of Fallot (TOF) and collateral-dependent pulmonary blood flow. Management for these fistulae is often primary surgical ligation or unifocalization at the time of complete repair, dependent on the presence of dual blood flow to the involved areas. We present the case of a 32-week premature boy weighing 1.79 kg with TOF, confluent branch pulmonary arteries, major aortopulmonary collaterals, and right coronary artery to main pulmonary artery fistula. The patient demonstrated evidence of coronary steal into the pulmonary vasculature with an elevation in the troponin level without hemodynamic instability, and subsequently underwent successful transcatheter occlusion of the fistula via right common carotid access using a Medtronic 3Q microvascular plug. Blood flow reflects the eye's health and is disrupted in many diseases. Many pathological processes take place at the cellular level like as microcirculation of blood in vessels, and the processing of medical images is a difficult recognition task. Existing techniques for measuring blood flow are limited due to the complex assumptions, equipment and calculations requirements. In this paper, we propose a method for determining the blood flow characteristics in eye conjunctiva vessels, such as linear and volumetric blood speed and topological characteristics of the vascular net. The method preprocesses the video to improve the conditions of analysis and then builds an integral optical flow for definition of flow dynamical characteristic of eye vessels. These characteristics make it possible to determine changes in blood flow in eye vessels. We show the efficiency of our method in natural eye vessel scenes. The altered blood flow in the foot is an important indicator of early diabetic foot complications. However, it is challenging to measure the blood flow at the whole foot scale. This study presents an approach for estimating the foot arterial blood flow using the temperature distribution and an artificial neural network. To quantify the relationship between the blood flow and the temperature distribution, a bioheat transfer model of a voxel-meshed foot tissue with discrete blood vessels is established based on the computed tomography (CT) sequential images and the anatomical information of the vascular structure. In our model, the heat transfer from blood vessels and tissue and the inter-domain heat exchange between them are considered thoroughly, and the computed temperatures are consistent with the experimental results. Analytical data are then used to train a neural network to determine the foot arterial blood flow. The trained network is able to estimate the objective blood flow for various degrees of stenosis in multiple blood vessels with an accuracy rate of more than 90%. Compared with the Pennes bioheat transfer equation, this model fully describes intra- and inter-domain heat transfer in blood vessels and tissue, closely approximating physiological conditions. OBJECTIVE: To explore the value of vascular architecture detected by micro vascular imaging (MVI) in preoperative diagnosis of focal liver lesions (FLLs). METHODS: In this retrospective study, patients with surgery and histopathologically proved or radiologically confirmed FLLs were included. Vascular architecture of FLLs were acquired by color Doppler flow imaging (CDFI) and MVI on LOGIQ™ E20 ultrasound machine (C1–6 convex array probes). Alder semiquantitative analysis (grade 0–3) and morphologic features of blood vessels (pattern a–f) were used to assess the blood flow within the FLLs. Interobserver agreement for evaluating blood flow of FLLs was analyzed. Using Adler's grading or morphologic patterns as diagnostic criteria for malignant FLLs, the diagnostic efficiency was analyzed and compared. RESULTS: From October 2021 and February 2022, 50 patients diagnosed with 40 malignant FLLs and 10 benign FLLs were finally included. The Kappa value within two observers for evaluating the blood flow of FLLs was 0.78 for MVI and 0.55 for CDFI. According to Alder semiquantitative analysis, more high-level blood flow signals (grade 2–3) were detected by MVI than CDFI (P < 0.05). Based on high-level blood flow signals (grade 2–3) and hypervascular supply patterns (pattern e and f), the diagnostic accuracy for malignant FLLs were 76% and 68% for MVI, 56% and 38% for CDFI, respectively. CONCLUSION: MVI is superior to CDFI in evaluating vascular architecture of FLLs. The modular inner branched stent-graft (MIBSG), a novel interventional therapy, has demonstrated good effects in the endovascular treatment of aortic arch pathologies, especially those involving the supra-aortic branches. Nevertheless, the long-term efficacy of the MIBSG and in-depth quantitative evaluation of postoperative outcomes remain to be examined. Moreover, the regularity of postoperative vascular remodeling induced by MIBSG implantation has yet to be explored. To address these questions, we constructed four models (normal, preoperative, 1 week postoperative, and 6 months postoperative) based on a single patient case to perform computational fluid dynamics simulations. The morphological and hemodynamic characteristics, including the velocity profile, flow rate distribution, and hemodynamic parameter distribution (wall shear stress and its derivative parameters), were investigated. After MIBSG implantation, the morphology of the supra-aortic branches changed significantly, and the branch point moved forward to the proximal ascending aorta. Moreover, the curvature radius of the aortic arch axis continued to change. These changes in morphology altered the characteristics of the flow field and wall shear stress distribution. As a result, the local forces exerted on the vessel wall by the blood led to vessel remodeling. This study provides insight into the vascular remodeling process after MIBSG implantation, which occurs as a result of the interplay between vascular morphological characteristics and blood flow characteristics. Brown adipose tissue (BAT) is a fat tissue specialized in heat production (non-shivering thermogenesis) and used by mammals to defend core body temperature when exposed to cold. Several studies have shown that during non-shivering thermogenesis the increase in BAT oxygen demand is met by a local and specific increase in tissue's blood flow. While the vasculature of BAT has been extensively studied postmortem in rodents using histology, optical and CT imaging techniques, vasculature changes during stimulation of non-shivering thermogenesis have never been directly detected in vivo. Here, by using computed tomography (CT) angiography with gold nanoparticles we investigate, non-invasively, changes in BAT vasculature during adrenergic stimulation of non-shivering thermogenesis by norepinephrine, a vasoconstrictor known to mediate brown fat heat production, and by CL 316,243, a specific β3-adrenergic agonist also known to elicit BAT thermogenesis in rodents. We found that while CL 316,243 causes local vasodilation in BAT, with little impact on the rest of the vasculature throughout the body, norepinephrine leads to local vasodilation in addition to peripheral vasoconstriction. As a result, a significantly greater relative increase in BAT perfusion is observed following the injection of NE compared to CL. This study demonstrates the use of in vivo CT angiography as an effective tool in assessing vascular reactivity in BAT both qualitatively and quantitatively in preclinical studies. The assembly of a mature vascular network involves coordinated endothelial cell (EC) shape changes, including the process of EC elongation. How EC elongation is dynamically regulated in vivo is not fully understood. Here, we have generated a zebrafish mutant that is deficient for the integrin adaptor protein Talin 1 (Tln1). Using a new focal adhesion (FA) marker line expressing endothelial VinculinbeGFP, we demonstrate that EC FAs function dynamically and are lost in our tln1 mutants, allowing us to uncouple the primary roles of FAs in EC morphogenesis from the secondary effects that occur due to systemic vessel failure or loss of blood flow. Tln1 loss led to compromised F-actin rearrangements, perturbed EC elongation and disrupted cell-cell junction linearisation in vessel remodelling. Finally, chemical induction of actin polymerisation restored actin dynamics and EC elongation during vascular morphogenesis. Together, we identify that FAs are essential for EC elongation and junction linearisation in flow-pressured vessels and that they influence actin polymerisation in cellular morphogenesis. In order to analyze the mechanical properties of a new type of retrievable vena cava filter, the Denali and Aeginsy retrievable filters are selected, and the in vivo and in vitro mechanical properties of the three types of retrievable vena cava filters are compared. The nonlinear deformation process of the filter which is pressed into the transport sheath, flicked and released into the vascular vessel is simulated by using the finite element method, and the mechanical behaviors of the filter and the blood vessel wall are analyzed. During the crimping process, the elastic strain peak value of the filter wall increases with increasing amount of grip. The maximum elastic strain peak value of the three filters is less than 8%, and the safety factor is greater than 2, which meets the strength requirement. Equivalent stress and strain on the vascular wall decreases along the direction of blood flow after the filter is opened. The peak value of equivalent stress and strain on the vascular wall corresponding to Aegisy filter is the largest, and the impact on the vascular wall is the largest. We aimed to quantitatively compare the choroid blood flow and choroid thickness at the periphery among eyes with central serous chorioretinopathy (CSC), fellow eyes and healthy eyes using ultra-widefield swept-source optical coherence tomography angiography (UWF SS-OCTA). Retrospective analysis of 49 patients with CSC (98 eyes, including unaffected fellow eyes) and 49 age and sex matched controls were included. We obtained 3-dimensional data of vertical 20 mm × horizontal 24 mm × scan depth 6 mm, comprising 9 subfields (superotemporal, upper, superonasal, temporal, central, nasal, inferotemporal, lower, inferonasal regions). CSC eyes presented with greater density of large-vessel choroidal layer in all the 9 subfields compared with controls. Compared with normal eyes, CSC eyes had greater choroidal thickness (superotemporal, upper, superonasal, temporal, central, nasal, inferotemporal, and inferonasal subfields) and choroidal volume (superotemporal, upper, superonasal, temporal, central, and nasal subfields). Compared with control eyes, the choriocapillaris density in the superotemporal, inferotemporal and inferonasal subfields was greater in patients with CSC. Our study may provide further evidence for the congestion of vortex vein in the pathogenesis of CSC. Background: Creation of new capillary blood vessels from pre-existing vessels is known as angiogenesis. The restoration of normal blood flow and, by extension, the outflow of gases, nutrients, and growth hormones makes angiogenesis crucial in a wide range of physiological and pathological situations, including wound healing and bone repair and regeneration. Tissue engineering and regenerative medicine rely heavily on angiogenesis because it controls the survival, proliferation, and differentiation of newly generated tissue structures. On the other hand, tumor cells with an angiogenic character have more proangiogenic pathways than downregulating mechanisms. Because of this, endothelial cells experience a period of rapid proliferation that fosters the development of a tumor microenvironment that is abundant in oxygen and nutrients and promotes the tumor's metastasis to new locations. In order to treat persistent diseases or cancerous tumors, doctors have begun using antiangiogenesis-based therapies, which involve giving patients compounds that block the development of new blood vessels. Objective: To identify the anti-angiogenic activity of Zeaxanthin and Melatonin and their combination in ex vivo rat aorta antiangiogenic assay and in vivo chorioallantoic membrane (CAM) animals' study. Methods: 12-14 weeks-old Albino male rats were used for the study. The tested substances zeaxanthin and melatonin were serially diluted. An ex vivo rat aorta ring experiment has been used to examine zeaxanthin potential antiangiogenic properties; the zeaxanthin-induced zone of blood vessel inhibition was measured in CAM assay in vivo chorioallantoic membrane. Results: The result of this study showed that there is a significant antiangiogenic activity of zeaxanthin and melatonin and there is an additive antiangiogenic effect of Z and M combination in ex vivo rat aorta ring assay Each Z and M showed significant anti-angiogenesis activity in the in vivo assay and for the combination, it was discovered that melatonin and zeaxanthin each produced a significant inhibition zone of blood vessels in the CAM assay. Conclusion: Zeaxanthin and melatonin and their combination exerted significant antiangiogenic effect on ex vivo rat aortic ring, Zeaxanthin and melatonin and their combination showed significant antiangiogenic effect on in vivo chick chorioallantoic membrane model.
