Chagas disease (CD) is one of the main neglected tropical diseases, diagnosed mainly by serological tests performed in centralized laboratories, which severely limits the clinical management of the disease in communities with scarce resources. Herein, an electrochemical impedance biosensor for the detection of CD was developed for the first time using a cruzipain-based sensor surface. The protein, highly immunogenic and isolated from Trypanosoma cruzi, was immobilized over the surface of gold disc electrodes modified with 11-mercaptoundecanoic (MUA) and 6-mercapto-1-hexanol (MCH) self-assembled monolayers (SAMs). Reproducible sensor surfaces, yielding 38 ± 3% coverage as measured by Surface Plasmon Resonance, were obtained by amide coupling of 120 μg/mL cruzipain onto 1/10 MUA/MCH SAMs for 30 min. Under optimized operational conditions, the impedimetric immunosensor recognized specific interactions for anti-T. cruzi antibodies in 1/800 diluted human serum patient samples. The charge transfer resistance of the biosensors increases by ∼18% in the presence of the positive samples, whereas the negative samples give rise to a negligible increase of around 6%. An excellent selectivity to clinical samples from patients infected with T. cruzi was obtained. The clear signal difference obtained for positive and negative clinical samples highlights the applicability of the sensors for the point-of-care diagnosis of CD.

Cancer companion diagnostics are incredibly important in helping to determine whether a patient will benefit from immune checkpoint inhibitor (ICI) treatment. Determining the chances of treatment success helps to inform clinicians to make the best treatment decisions for a particular patient. Many immune checkpoint related proteins show potential as biomarkers for ICI success, such as the checkpoint proteins themselves, cytokines, interleukins and other immune response related proteins. The most investigated checkpoint inhibitor protein is Programmed Death Ligand 1 (PD-L1), which is used as a biomarker in clinical diagnostic tests but, with some limitations. In the near future, tests for many different biomarkers will start becoming commercially available along with tests for multiple biomarkers simultaneously, giving an even better prediction of potential ICI success. Electrochemical sensors are a high sensitivity point of care diagnostic technique that can have the potential to achieve detection of multiple biomarkers at once. The main problem facing this field is improving their sensitivity to be able to detect the incredibly low concentrations of biomarkers found in liquid biopsy samples. Many methods such as enhancing an electrode surface with high conductivity materials or increasing the measured electrochemical signal via signal amplifying molecules have been investigated with promising results. This review investigates the potential biomarkers relevant to predicting ICI success, as well as the current electrochemical sensors that have been developed to determine the expression levels of these proteins.

Recent advances in robotics and affordable genomic sequencing technologies have made it possible to establish and quantitatively track the assembly of enrichment communities in high-throughput. By conducting community assembly experiments in up to thousands of synthetic habitats, where the extrinsic sources of variation among replicates can be controlled, we can now study the reproducibility and predictability of microbial community assembly at different levels of organization, and its relationship with nutrient composition and other ecological drivers. Through a dialog with mathematical models, high-throughput enrichment communities are bringing us closer to the goal of developing a quantitative predictive theory of microbial community assembly. In this short review, we present an overview of recent research on this growing field, highlighting the connection between theory and experiments and suggesting directions for future work.

Abstract Background Different pathologies, affecting the skeletal system, were reported to display altered bone and/or cartilage innervation profiles leading to the deregulation of the tissue homeostasis. The patterning of peripheral innervation is achieved through the tissue-specific expression of attractive or repulsive axonal guidance cues in specific space and time frames. During the last decade, emerging findings attributed to the extracellular vesicles (EV) trading a central role in peripheral tissue innervation. However, to date, the contribution of EV in controlling bone innervation is totally unknown. Results Here we show that sensory neurons outgrowth induced by the bone resorbing cells—osteoclasts—is promoted by osteoclast-derived EV. The EV induced axonal growth is achieved by targeting epidermal growth factor receptor (EGFR)/ErbB2 signaling/protein kinase C phosphorylation in sensory neurons. In addition, our data also indicate that osteoclasts promote sensory neurons electrophysiological activity reflecting a possible pathway in nerve sensitization in the bone microenvironment, however this effect is EV independent. Conclusions Overall, these results identify a new mechanism of sensory bone innervation regulation and shed the light on the role of osteoclast-derived EV in shaping/guiding bone sensory innervation. These findings provide opportunities for exploitation of osteoclast-derived EV based strategies to prevent and/or mitigate pathological uncontrolled bone innervation.

ABSTRACT Soil attributes can provide indicators for the management and conservation of natural and agrarian ecosystems. This study was carried out before the rupture of the Córrego do Feijão dam and aimed to evaluate and quantify the stocks of carbon, nitrogen and the C/N ratio in soils around the Paraopeba-MG river basin in five regions corresponding to the high, middle, and low river course, considering three usage factors: forest, pasture, and Shrubbery forest, correlating them with the chemical and physical properties of the soil, aiming at the interpretation of past use, to obtain management and conservation strategies for ecosystems. The results indicate a significant variation in carbon and nitrogen stocks with an increase in the 20-40 cm layer. Concerning the type of use, the forest system was the most efficient in accumulating carbon. About to site, the regions of Brumadinho and Fortuna de Minas showed similarities in the incorporation of carbon in the soil, differing from the regions of Jeceaba, Florestal, and Três Marias. There was a close correlation between stocks and the chemical and physical properties of the soil, making it possible to assess the quality of the soils in the study regions in terms of structure and fertility.

Throughout history, the yeast Saccharomyces cerevisiae has played a central role in human society due to its use in food production and more recently as a major industrial and model microorganism, because of the many genetic and genomic tools available to probe its biology. However, S. cerevisiae has proven difficult to engineer to expand the carbon sources it can utilize, the products it can make, and the harsh conditions it can tolerate in industrial applications. Other yeasts that could solve many of these problems remain difficult to manipulate genetically. Here, we engineered the thermotolerant yeast Kluyveromyces marxianus to create a new synthetic biology platform. Using CRISPR-Cas9 (clustered regularly interspaced short palindromic repeats with Cas9)-mediated genome editing, we show that wild isolates of K. marxianus can be made heterothallic for sexual crossing. By breeding two of these mating-type engineered K. marxianus strains, we combined three complex traits-thermotolerance, lipid production, and facile transformation with exogenous DNA-into a single host. The ability to cross K. marxianus strains with relative ease, together with CRISPR-Cas9 genome editing, should enable engineering of K. marxianus isolates with promising lipid production at temperatures far exceeding those of other fungi under development for industrial applications. These results establish K. marxianus as a synthetic biology platform comparable to S. cerevisiae, with naturally more robust traits that hold potential for the industrial production of renewable chemicals.IMPORTANCE The yeast Kluyveromyces marxianus grows at high temperatures and on a wide range of carbon sources, making it a promising host for industrial biotechnology to produce renewable chemicals from plant biomass feedstocks. However, major genetic engineering limitations have kept this yeast from replacing the commonly used yeast Saccharomyces cerevisiae in industrial applications. Here, we describe genetic tools for genome editing and breeding K. marxianus strains, which we use to create a new thermotolerant strain with promising fatty acid production. These results open the door to using K. marxianus as a versatile synthetic biology platform organism for industrial applications.

Abstract Background Metastasis causes the most breast cancer-related deaths in women. Here, we investigated the antitumor effect of solid lipid nanoparticles (SLN-DTX) when used in the treatment of metastatic breast tumors using 4T1-bearing BALB/c mice. Results Solid lipid nanoparticles (SLNs) were produced using the high-energy method. Compritol 888 ATO was selected as the lipid matrix, and Pluronic F127 and Span 80 as the surfactants to stabilize nanoparticle dispersion. The particles had high stability for at least 120 days. The SLNs’ dispersion size was 128 nm, their polydispersity index (PDI) was 0.2, and they showed a negative zeta potential. SLNs had high docetaxel (DTX) entrapment efficiency (86%), 2% of drug loading and showed a controlled drug-release profile. The half-maximal inhibitory concentration (IC50) of SLN-DTX against 4T1 cells was more than 100 times lower than that of free DTX after 24 h treatment. In the cellular uptake test, SLN-DTX was taken into the cells significantly more than free DTX. The accumulation in the G2-M phase was significantly higher in cells treated with SLN-DTX (73.7%) than in cells treated with free DTX (23.0%), which induced subsequent apoptosis. TEM analysis revealed that SLN-DTX internalization is mediated by endocytosis, and fluorescence microscopy showed DTX induced microtubule damage. Economical biofuel production from plant biomass requires the conversion of both cellulose and hemicellulose in the plant cell wall. The best industrial fermentation organism, the yeast Saccharomyces cerevisiae, has been developed to utilize xylose by heterologously expressing either a xylose reductase/xylitol dehydrogenase (XR/XDH) pathway or a xylose isomerase (XI) pathway. Although it has been proposed that the optimal means for fermenting xylose into biofuels would use XI instead of the XR/XDH pathway, no clear comparison of the best publicly-available yeast strains engineered to use XR/XDH or XI has been published. We therefore compared two of the best-performing engineered yeast strains in the public domain—one using the XR/XDH pathway and another using XI—in anaerobic xylose fermentations. We find that, regardless of conditions, the strain using XR/XDH has substantially higher productivity compared to the XI strain. By contrast, the XI strain has better yields in nearly all conditions tested.

ABSTRACT The humified organic matter presents bioactivity similar to the auxinic effect. As bokashi is produced by a special process of humification, information is needed about the bioactive potential of its humic acids. The objective of this work was studying the initial performance of corn-indicator plants in response to the application of different concentrations of humic acids isolated from bokashi. The corn seeds were treated for 16 hours with solutions containing 0, 10, 20, 30, 40 and 80 mmol L-1 of C in the form of humic acids. Then, the seeds were planted in pots of 1 dm3 containing corrected and fertilized soil, in greenhouse. Growth characteristics of shoot and root systems were evaluated. The results showed that the humic acids extracted from bokashi had positive effects on the initial performance of corn.

More than 783 million people worldwide are currently without access to clean and safe water. Approximately 1 in 5 cases of mortality due to waterborne diseases involve children, and over 1.5 million cases of waterborne disease occur every year. In the developing world, this makes waterborne diseases the second highest cause of mortality. Such cases of waterborne disease are thought to be caused by poor sanitation, water infrastructure, public knowledge, and lack of suitable water monitoring systems. Conventional laboratory-based techniques are inadequate for effective on-site water quality monitoring purposes. This is due to their need for excessive equipment, operational complexity, lack of affordability, and long sample collection to data analysis times. In this review, we discuss the conventional techniques used in modern-day water quality testing. We discuss the future challenges of water quality testing in the developing world and how conventional techniques fall short of these challenges. Finally, we discuss the development of electrochemical biosensors and current research on the integration of these devices with microfluidic components to develop truly integrated, portable, simple to use and cost-effective devices for use by local environmental agencies, NGOs, and local communities in low-resource settings.

The inability to diagnose numerous diseases rapidly is a significant cause of the disparity of deaths resulting from both communicable and non-communicable diseases in the developing world in comparison to the developed world. Existing diagnostic instrumentation usually requires sophisticated infrastructure, stable electrical power, expensive reagents, long assay times, and highly trained personnel which is not often available in limited resource settings. This review will critically survey and analyse the current lateral flow-based point-of-care (POC) technologies, which have made a major impact on diagnostic testing in developing countries over the last 50 years. The future of POC technologies including the applications of microfluidics, which allows miniaturisation and integration of complex functions that facilitate their usage in limited resource settings, is discussed The advantages offered by such systems, including low cost, ruggedness and the capacity to generate accurate and reliable results rapidly, are well suited to the clinical and social settings of the developing world.

The vegetative propagation of ornamental plants can be accelerated by applying plant growth regulators. Amongst them, the use of auxins, plant hormones with physiological effects on cell elongation and rooting have stood out. Alternatively, the application of humic acids, bioactive fraction of soil organic matter, also results in increases in rooting cuttings of ornamental plants. The objective of this work was to study the growth characteristics and the nutritional contents of croton and hibiscus plants during acclimation of seedlings in response to different concentrations of indolebutyric acid (IBA) and humic acid (HA) applied to cuttings for rooting. The experiment was conducted in greenhouse, and the apical stem cuttings were treated with solutions with concentrations of 0, 250, 500, 1000 and 2000 mg L-1of IBA and 0, 10, 20, 30 and 40 mg L-1 of C from HA. At 45 days of rooting in carbonized rice husk, they were individually transferred to plastic bags of 2.0 dm3 containing a mixture of soil: sand: manure (2: 1: 1) as substrate. At 90 days of acclimation, the plants were collected for measurement of growth and nutritional variables. The results showed that the application of the IBA stimulates the absorption of nutrients and growth of croton cuttings and transplanted hibiscus, contributing to formation of vigorous seedlings. A similar response occurred with the application of HA in hibiscus cuttings

O entendimento dos fenômenos de trocas de carbono entre solo, água e atmosfera é fundamental para o manejo e a conservação de ecossistemas naturais e agrários. Os objetivos deste trabalho foram determinar os estoques de carbono orgânico, as suas frações e a fertilidade do solo sob os manejos florestal (mata remanescente), integrando agricultura e florestas (eucalipto e cutieira) e em monoculturas agrícola (sob pivô central) e pastoril (braquiária), na Bacia do Rio Paraopeba, em Florestal-MG, visando a propor indicadores para a avaliação de manejo e conservação do solo. As amostras de solo foram coletadas em remanescente de mata nativa (Mata), em área agrícola cultivada sob irrigação de pivô central (Pivô) e em áreas de cultivo de Corymbia citriodora (Corymbia citriodora (Hook.) K.D. Hill & L.A.S. Johnson) (Eucalipto), Joannesia princeps Vell. (Cutieira) e Brachiaria decumbens Stapf (Pasto), nas profundidades de 0 a 20 e 20 a 40 cm. Os resultados indicaram que as estimativas dos estoques e de estabilidade do carbono orgânico aumentaram com a fertilidade do solo. Os sistemas florestais, principalmente a Mata, acompanhada pelos sistemas de manejo do solo integrando florestas à pastagem, em comparação com o cultivo contínuo ou a monocultura de pastagem, apresentaram estoques de carbono maiores, mais estáveis e menos solúveis, com formas mais aromáticas e hidrofóbicas (maior relação AH/AF), indicando menor potencial de lixiviação de carbono para o sistema aquático adjacente.

Qual é a estrutura molecular das substâncias húmicas? Ainda não foi possível encontrar a resposta. Há muita controvérsia e pouco consenso sobre as estruturas dessas substâncias. Destacam-se dois pontos principais: o acúmulo de informações e os desafios metodológicos. Esta revisão objetivou reunir informações sobre a natureza dos ácidos húmicos e suas funções nos ecossistemas naturais e agrários, visando ao manejo e à conservação, bem como às oportunidades tecnológicas para o desenvolvimento de novos insumos para a agricultura tropical.

Besides fixing N2, some diazotrophic bacteria or diazotrophs, also synthesize organic acids and are able to solubilize rock phosphates, increasing the availability of P for plants. The application of these bacteria to pineapple leaf axils in combination with rock phosphate could increase N and P availability for the crop, due to the bacterial activity of biological nitrogen fixation and phosphate solubilization. The objectives of this study were: (i) to select and characterize diazotrophs able to solubilize phosphates in vitro and (ii) evaluate the initial performance of the pineapple cultivars Imperial and Pérola in response to inoculation with selected bacteria in combination with rock phosphate. The experiments were conducted at Universidade Estadual do Norte Fluminense Darcy Ribeiro, in 2009. In the treatments with bacteria the leaf contents of N, P and K were higher than those of the controls, followed by an increase in plant growth. These results indicate that the combined application of diazotrophic phosphate-solubilizing bacteria Burkholderia together with Araxá rock phosphate can be used to improve the initial performance of pineapple slips.

The discovery of the phenomena known as localized surface plasmon resonance (LSPR) has provided the basis for many research areas, ranging from materials science to biosensing. LSPR has since been viewed as a transduction platform that could yield affordable, portable devices for a multitude of applications. This review aims to outline the potential applications within developing countries and the challenges that are likely to be faced before the technology can be effectively employed.

Gladiolus is an ornamental species produced for cut flowers and propagated by corms. The early flowering and increase in the number of flower buds, besides the production of commercial corms are constant challenges to be addressed in the crop improvement. Commercial production of ornamentals is technologically accelerated by means of growth regulators. Among them, the auxins stand out for their key role in the adventitious rooting and cell elongation. Alternatively, the humic substances present in the organic matter also have biostimulating effect, which is very similar to the auxinic effect. Therefore, this work aimed to study the growth and development of gladiolus in response to application of different concentrations of humic acids (HA) isolated from vermicompost. Corms were soaked for 24 hours in solutions containing 0, 10, 20, 30 and 40 mmol L-1 of C from HA. The corms were planted in 10-dm³ plastic bags filled with substrate and kept in a greenhouse. Growth of shoots and roots was evaluated. The results showed that the use of HA accelerates growth, and anticipates and increases flowering of Gladiolus.

The inoculation with plant growth-promoting bacteria can be a technological approach useful for increasing the production of maize. The objective of this study was to evaluate the initial performance of maize in response to application of doses of NPK combined with the inoculation of the diazotrophic bacteria Herbaspirillum seropedicae in an greenhouse experiment. The experiment consisted of six fertilizer levels: 0, 25, 50, 75, 100 and 200% of the recommended dose of NPK applied to maize inoculated and non-inoculated with H. seropedicae. At 30 days after the treatment application, the growth characteristics and nutritional status of the plants were evaluated. Plant development was influenced by fertilization, but it was enhanced by combination with the bacteria, which resulted in significant increases in the dry mass of shoots (7%) and leaf area (9%) when compared with non-inoculated plants. The results showed increases in the concentration of N (11%), P (30%) and K (17%) of maize plants in response to bacterial inoculation together with NPK compared with plants that were applied fertilize only. The greater consistency and stability response of the host plant to bacterization in the presence of chemical fertilizer indicate a promissory biotechnological approach for improving the initial growth and adaptation of maize to the cultivation environment.A inoculação de bactérias promotoras de crescimento de plantas pode ser uma abordagem tecnológica útil para aumentar a produção de milho. O objetivo deste trabalho foi avaliar o desempenho inicial de plantas de milho, em resposta à aplicação de doses de NPK combinadas com a inoculação da bactéria diazotrófica Herbaspirillum seropedicae, em experimento em casa de vegetação. A matriz experimental consistiu em seis níveis de adubação: 0, 25, 50, 75, 100 e 200 % da dose de NPK recomendada, aplicados em plantas de milho inoculadas e não inoculadas com H. seropedicae. Aos 30 dias após a aplicação dos tratamentos, foram avaliadas as características de crescimento e nutricionais das plantas. O crescimento do milho foi influenciado pela adubação, mas foi reforçado pela combinação com a bactéria, que resultou em aumentos significativos na matéria seca da parte aérea (7 %) e área foliar (9 %), quando comparado com as das plantas não inoculadas. Os resultados indicaram incrementos no acúmulo de N (11 %), P (30 %) e K (17 %), em plantas de milho, em resposta à inoculação bacteriana em conjunto com NPK, quando comparado com os das plantas que apenas receberam adubação. A estabilidade e maior consistência da resposta da planta hospedeira para bacterização, na presença de fertilização química, apontam para uma ferramenta biotecnológica promissora para melhorar o crescimento inicial e adaptação de plantas de milho ao ambiente de cultivo.

Os tumores espontâneos da glândula mamária representam, nas cadelas, a neoplasia mais observada na clínica de pequenos animais, justificando o grande interesse dos pesquisadores no estudo dessa lesão. Este trabalho teve por objetivo determinar a frequência dos tumores mamários em cadelas atendidas no HOSPMEV/UFBA, no município de Salvador, no período de janeiro de 2006 a abril de 2008, bem como realizar a caracterização clínica, a classificação histopatológica e o georreferenciamento dessa patologia. Os resultados revelaram alta prevalência de neoplasias mamárias em cadelas, em sua maioria tumores, em estádios mais avançados, sendo os carcinomas em tumores mistos o tipo histológico predominante. Observou-se ainda a correlação das características socioeconômicas da população estudada com fatores sabidamente prognósticos para neoplasias mamárias em cadelas. Esses resultados refletem a procura tardia da assistência Médica Veterinária. Por isso, é urgente educar a população quanto prevenção e detecção da neoplasia mamária nas cadelas, buscando o diagnóstico precoce e a terapêutica adequada.

Adventitious rooting of ornamental plants can be accelerated by the application of growth regulators, such as auxin. Humic acids, organic matter in soil and organic compounds also have a biostimulant effect. This work evaluated the rooting in cuttings of croton (Codianeum variegatum L. Rumph) and hibiscus (Hibiscus rosa-sinensis L) in response to the application of different concentrations of indolbutyric acid (IBA) and humic acid (HA). The experiment was carried out in a greenhouse. Apical stem cuttings were treated with solutions at concentrations of: 0, 250, 500, 1000, 2000 mg L-1 IBA and 0, 10, 20, 30, 40 mmol L-1 HA carbon isolated from vermicomposting. Forty-five days after the applications, the cuttings were removed from the pots containing carbonized rice hull and the following variables were measured: rooting number, length and width of leaves, fresh and dry matter of root and aerial part and root area. The results were subjected to analysis of variance and the qualitative and quantitative effects of the treatments were compared by contrast and regression, respectively. Regression equations were used to determine the maximum efficiency level of root dry matter according to IBA and HA. Higher accumulation of root dry matter was recorded for the treatments with the doses 579 mg L-1 IBA and 14 mmol L-1 HA and 970 mg L-1 IBA and 50 mmol L-1 HA for root cuttings of croton and hibiscus, respectively. It was found that the application of eiher IBA or HA at the indicated doses accelerates rooting in cuttings of croton and hibiscus and contributes to the formation of vigorous plants.

Due to the high energy requirement and demand for non-renewable resources for the production of chemical fertilizers, added also to the environmental impact caused by the use of such products, it is important to intensify research on bio-based agricultural inputs. The use of nitrogen-fixing endophytic and phosphate solubilizing bacteria can provide these nutrients to the plants from the air and poorly soluble phosphorus sources, such as phosphate rock. The objective of this study was to evaluate the nutrition and initial growth of maize (Zea mays L.) in response to the inoculation of nitrogen-fixing and rock phosphate solubilizing endophytic bacteria, in single or mixed formulation, applied with vermicompost. The treatments containing bacteria, both diazotrophic and phosphate solubilizing, when compared to controls, showed higher levels of leaf nitrogen and phosphorus in maize, as well as higher growth characteristics. The application of vermicompost showed synergistic effect when combined with endophytic bacteria. Thus, the innovation of the combination of the studied factors may contribute to the early development of maize.

Humic substances isolated from soil organic matter had been used as stimulators of plant metabolism. Arabidopsis thaliana (L.) Heynh. with only five chromosomes, short cycle and size, is an important model to evaluate the physiological effects of these substances, which are qualitatively and quantitatively influenced by morphogenesis, mineralogy and chemistry of soils. The objective of this study was to evaluate the ambience effects on bioactivity of humic acids. A and B horizons of four typical soils of the North Fluminense were sampled. After isolation and purification, humic acids were applied to plants in increasing concentrations. The number and length of lateral roots and main root length were evaluated and, subsequently, the concentrations of maximum stimulation were determined by dose-response curves and regression equations. The results showed that more stable humic acids isolated from soil in less advanced stages of weathering, high activity clay and high base saturation resulted in better physiological stimulants for Arabidopsis.

Rock phosphates have low solubility in water, but good solubility in acid. The use of organic compounds together with these phosphorus sources applied to the basal leaf axils of pineapple can increase the solubility of this phosfate source and increase the P availability to the crop. A greenhouse experiment was conducted using Araxá rock phosphate (10 g) in combination or not with solutions containing increasing concentrations of humic acids (0 to 40 mmol L-1 of carbon), with or without citric acid (0.005 mmol L-1), applied to basal leaf axils of pineapple cv. Pérola. Growth and nutritional characteristics of aerial plant parts were assessed. Growth rates of aerial parts and N, P, K, Ca and Mg contents increased curvilinearly with increasing concentration of carbon in the form of humic acids. Maximum values were found for the concentration of 9.3 mmol L-1 of carbon combined with 0.005 mmol L-1 of citric acid and natural phosphate.

Abstract Background The basidiomycete fungus Moniliophthora perniciosa is the causal agent of Witches' Broom Disease (WBD) in cacao (Theobroma cacao). It is a hemibiotrophic pathogen that colonizes the apoplast of cacao's meristematic tissues as a biotrophic pathogen, switching to a saprotrophic lifestyle during later stages of infection. M. perniciosa, together with the related species M. roreri, are pathogens of aerial parts of the plant, an uncommon characteristic in the order Agaricales. A genome survey (1.9× coverage) of M. perniciosa was analyzed to evaluate the overall gene content of this phytopathogen. Results Genes encoding proteins involved in retrotransposition, reactive oxygen species (ROS) resistance, drug efflux transport and cell wall degradation were identified. The great number of genes encoding cytochrome P450 monooxygenases (1.15% of gene models) indicates that M. perniciosa has a great potential for detoxification, production of toxins and hormones; which may confer a high adaptive ability to the fungus. We have also discovered new genes encoding putative secreted polypeptides rich in cysteine, as well as genes related to methylotrophy and plant hormone biosynthesis (gibberellin and auxin). Analysis of gene families indicated that M. perniciosa have similar amounts of carboxylesterases and repertoires of plant cell wall degrading enzymes as other hemibiotrophic fungi. In addition, an approach for normalization of gene family data using incomplete genome data was developed and applied in M. perniciosa genome survey. Conclusion This genome survey gives an overview of the M. perniciosa genome, and reveals that a significant portion is involved in stress adaptation and plant necrosis, two necessary characteristics for a hemibiotrophic fungus to fulfill its infection cycle. Our analysis provides new evidence revealing potential adaptive traits that may play major roles in the mechanisms of pathogenicity in the M. perniciosa/cacao pathosystem.

RESUMO A alface é uma das hortaliças folhosas mais consumidas no Brasil, gerando grande demanda por seu cultivo. É cultura exigente em nutrientes e requer solo rico em matéria orgânica para obtenção de produtividade elevada, com necessidade de aplicação de grandes doses de compostos orgânicos, onerando os custos da produção. Ácidos húmicos (AH) e bactérias diazotróficas (B) apresentam hormônios vegetais, como auxinas, que promovem alongamento celular, aumento das raízes e da absorção de nutrientes, levando ao crescimento e desenvolvimento das plantas. Por essas características, podem ser utilizados como bioestimulantes na promoção da produtividade da alface. O objetivo deste trabalho foi avaliar a produtividade da alface em resposta à aplicação de ácidos húmicos e bactérias diazotróficas, em condições de campo. Os ácidos húmicos foram isolados de esterco bovino e as bactérias usadas foram provenientes da coleção de bactérias do Setor de Floricultura da Universidade Federal de Viçosa - Campus Florestal, extraídas de orquídeas Cymbidium sp.. O experimento foi realizado em condições de campo, com o cv. Vanda e os tratamentos usados foram controle (cultivo convencional do produtor), aplicação isolada de ácido húmico e bactéria e aplicação combinada de ácido húmico+bactéria. O delineamento adotado foi em blocos casualizados, com cinco repetições. A aplicação dos bioestimulantes foi realizada logo após o transplantio. Foram avaliados atributos fitotécnicos da parte aérea das plantas. Concluiu-se que os tratamentos utilizados, principalmente a combinação AH+B, aumentaram a produtividade da alface.

RESUMO A matéria orgânica humificada apresenta bioatividade semelhante à das auxinas. Contudo, são necessárias informações sobre a capacidade de estímulo vegetal das substâncias húmicas extraídas de solos manejados com sistemas integrando lavoura, pecuárias e florestas. O objetivo deste trabalho foi avaliar o desempenho inicial do milho em resposta à aplicação de concentrações de substâncias húmicas isoladas em solos manejados com sistema de integração lavoura, pecuária e florestas, aplicadas por via da semente. Sementes de milho variedade AG 1051 foram tratadas por 16 horas, em soluções contendo 0, 10, 20, 30, 40 e 80 mmol L-1 de carbono na forma de substâncias húmicas. Em seguida, foram plantadas em vasos de 1 dm3, com solo corrigido e fertilizado, em casa de vegetação. Foram avaliadas características de crescimento da parte aérea e do sistema radicular. Observou-se que as substâncias húmicas extraídas de solos submetidos a integração lavoura, pecuária e florestas apresentaram efeitos positivos no desempenho inicial do milho, incrementando o acúmulo de matéria seca das plantas, em comparação com o efeito das substâncias extraídas de solos do controle, na área degradada inicial (133%), do cerrado (29%), bem como do eucalipto (30%) e da pastagem plantada solteira (56%).

ABSTRACT The economic and environmental costs of nitrogen fertilization have intensified the search for technologies that reduce mineral fertilization, for example atmospheric nitrogen-fixing (diazotrophic) bacteria inoculation. In this context, the present study addressed the isolation and quantification of diazotrophic bacteria in the sludge from treated wastewater of a poultry slaughterhouse; a description of the bacteria, based on cell and colony morphology; and an assessment of growth and N content of maize plants in response to inoculation. Sixteen morphotypes of bacteria were isolated in six N-free culture media (JMV, JMVL, NFb, JNFb, LGI, and LGI-P). The bacteria stained gram-positive, with 10 rod- and six coccoid-shaped isolates. To evaluate the potential of bacteria to promote plant growth, maize seeds were inoculated. The experiment consisted of 17 treatments (control plus 16 bacterial isolates) and was carried out in a completely randomized design with six replicates. The experimental units consisted of one pot containing two maize plants in a greenhouse. Forty-five days after planting, the variables plant height, leaf number, stem diameter, root and shoot fresh and dry weight, and N content were measured. The highest values were obtained with isolate UFV L-162, which produced 0.68 g total dry matter per plant and increased N content to 22.14 mg/plant, representing increments of 74 and 133%, respectively, compared with the control. Diazotrophs inhabit sludge from treated wastewater of poultry slaughterhouses and can potentially be used to stimulate plant development and enrich inoculants.

ABSTRACT Once it is stabilized in the soil, organic matter minimizes limitations of Brazilian Oxisols, such as low cation exchange capacity, low nutrient availability, toxicity due to high aluminum content, and phosphate adsorption. Moreover, humified organic matter fractions are bioactive. It is, therefore, important to evaluate the biostimulant ability of compounds present in soil carbon stocks to develop sustainable technologies for tropical agriculture based on renewable natural resources. The objective of this research was to correlate some soil quality indicators, redox properties, and bioactivity of humic acids isolated from integrated farming, livestock, and forestry systems aiming to understand the mechanisms involved in plant stimulation by humified organic matter. Carbon stocks and their stability were determined from oxidation by dichromatometry and iodometry, respectively. Bioactivity was assessed using yield data of corn indicator plants. The results indicated that when native-like forests were reintroduced instead of pastureland, soil carbon stocks and their stability increased along with overall improvements in soil fertility, chemical and physical properties, and soil biodiversity. The bioactivity of humic substances isolated from soils used in integrated crop, livestock, and forestry management was higher than that of soils derived from pastures or eucalyptus alone.

ABSTRACT Owing to numerous health benefits, the cultivation of medicinal plants has significantly increased. Achillea millefolium L. (common names: milefoil, yarrow) is attributed important medicinal properties. Consumers of herbal medicinal products demand for production systems based on ecological farming principles, thus an interesting technology would be application biostimulant based on humic acids. The objective of this study was to evaluate the rooting and initial performance of Achillea millefolium in response to the use of humic acids (HA) isolated from composted cattle manure and poultry litter and applied in different concentrations (0, 10, 20, 30, and 40 mmolL-1 of HA) at the propagation phase. Rooting and plant biomass accumulation were analyzed. Humic acids derived from poultry litter promoted root development and increase of fresh biomass and total dry mass, proving their biostimulant effect. The optimal concentration of humic acids estimated for the initial development of A. millefolium was 22.25 mmolL-1.

Cytokines are soluble proteins secreted by immune cells that act as molecular messengers relaying instructions and mediating various functions performed by the cellular counterparts of the immune system, by means of a synchronized cascade of signaling pathways. Aberrant expression of cytokines can be indicative of anomalous behavior of the immunoregulatory system, as seen in various illnesses and conditions, such as cancer, autoimmunity, neurodegeneration and other physiological disorders. Cancer and autoimmune diseases are particularly adept at developing mechanisms to escape and modulate the immune system checkpoints, reflected by an altered cytokine profile. Cytokine profiling can provide valuable information for diagnosing such diseases and monitoring their progression, as well as assessing the efficacy of immunotherapeutic regiments. Toward this goal, there has been immense interest in the development of ultrasensitive quantitative detection techniques for cytokines, which involves technologies from various scientific disciplines, such as immunology, electrochemistry, photometry, nanotechnology and electronics. This review focusses on one aspect of this collective effort: electrochemical biosensors. Among the various types of biosensors available, electrochemical biosensors are one of the most reliable, user-friendly, easy to manufacture, cost-effective and versatile technologies that can yield results within a short period of time, making it extremely promising for routine clinical testing.

ABSTRACT Seed treatment with inoculants based on plant growth-promoting bacteria (PGPB) or the application of humic acids (HA) may increase the productivity of plants of agricultural interest. The hypothesis of this work is that it is possible to combine the effect of plant growth promoting characteristic of HA with the inoculation of PGPB selected strains in the treatment of maize seeds. Thus, providing superior responses than in single applications of both in the initial maize development. To meet this purpose, we conducted isolated application of HA or PGPB inoculation of Burkholderia gladioli and Rhizobium cellulosilyticum, and the combined application of PGPB and HA for treatment of maize seeds. At the end of the experiment (45 days after germination), the plants were evaluated biometrically, nutritionally and a bacteria count was performed in plants using the Most Probable Number technique. The results showed that it is possible to combine the effects of HA with the inoculation of selected strains of PGPB, obtaining superior responses to the isolated application of both. Thus, the use of HA-based bio-stimulants in combination with PGPB is positive and complementary compared to inputs generally used in the treatment of maize seeds.
