Cell-free expression systems have drawn increasing attention as a tool to achieve complex biological functions outside of the cell. Several applications of the technology involve the delivery of functionality to challenging environments, such as field-forward diagnostics or point-of-need manufacturing of pharmaceuticals. To achieve these goals, cell-free reaction components are preserved using encapsulation or lyophilization methods, both of which often involve an embedding of components in porous matrices like paper or hydrogels. Previous work has shown a range of impacts of porous materials on cell-free expression reactions. Here, we explored a panel of 32 paperlike materials and 5 hydrogel materials for the impact on reaction performance. The screen included a tolerance to lyophilization for reaction systems based on both cell lysates and purified expression components. For paperlike materials, we found that (1) materials based on synthetic polymers were mostly incompatible with cell-free expression, (2) lysate-based reactions were largely insensitive to the matrix for cellulosic and microfiber materials, and (3) purified systems had an improved performance when lyophilized in cellulosic but not microfiber matrices. The impact of hydrogel materials ranged from completely inhibitory to a slight enhancement. The exploration of modulating the rehydration volume of lyophilized reactions yielded reaction speed increases using an enzymatic colorimetric reporter of up to twofold with an optimal ratio of 2:1 lyophilized reaction to rehydration volume for the lysate system and 1.5:1 for the purified system. The effect was independent of the matrices assessed. Testing with a fluorescent nonenzymatic reporter and no matrix showed similar improvements in both yields and reaction speeds for the lysate system and yields but not reaction speeds for the purified system. We finally used these observations to show an improved performance of two sensors that span reaction types, matrix, and reporters. In total, these results should enhance efforts to develop field-forward applications of cell-free expression systems.

Whatman No. 1 chromatography paper is widely used as a substrate for cellulose-based immunoassays. The immobilized proteins are used to capture target biomarkers for detection. However, alternative paper substrates may facilitate mass production of immunoassays as diagnostic tests. Here, we assessed the physical characteristics and protein immobilization capabilities of different commercial papers. Some substrates fulfilled our design criteria, including adequate flow rate and sufficient protein immobilization for efficient target capture. This study demonstrates that a variety of paper substrates can be bioactivated and used to capture target biomarkers, enabling development of affordable diagnostic tests from a range of starting materials.

Cotton is cheap, easily available and widely used as surgical material. Therefore, cotton would be good raw material to design a scaffold for tissue engineering applications. In this work, the gelatin-coated microfibrous paper scaffold was fabricated successfully by a papermaking process. Microfibers-based scaffold could overcome the limitations of nanofibrous material-based scaffold for tissue engineering application. The physicochemical and mechanical properties of the scaffolds were characterized. The results revealed that the gelatin contributed to the enhanced cell attachment and proliferation over the scaffolds, whereas cellulose as a strong backbone in the scaffold to support it for keeping its appearance. Their tensile strength and water absorption capacity were improved, but pore size and porosity were decreased after incorporation of gelatin. Hence, results suggested that fabricated scaffolds have huge prospective as a bioactive, well-designed and economical scaffold stand for tissue engineering application.

A cellulose paper-based biosensor was developed for the equipment-free and naked-eye detection of a transcription factor (TF) using dopamine as the signal-responsive material based on exonuclease III-assisted cycling amplification. Compared with other TF biosensors, the proposed biosensor demonstrates low-cost, portable, disposable, and naked-eye detection features.

Paper-based microfluidics devices can create a new healthcare model. Cellulose is carbohydrate polymer biocompatible and hydrophilic. These characteristics enhance the development of user-friendly diagnostic devices, but the link between paper manufacturing process and performance of the devices is still unclear. Previous studies focused on either commercial papers or lab papers from wood-cellulose fibers, with different basis-weight. This work introduces the effect of refining process and lab paper from nonwood-cellulose fibers, focusing on sisal fibers to overcome the aforementioned challenge. Structural characteristics of paper, such as basis-weight and degree of refining, are optimized and correlated with blood typing test resolution. Unrefined sisal paper of 50 g/m2 and 100 g/m2 basis-weight exhibit a higher gray intensity level than refined paper, and also maximal capillary rise and a pore size suitable for blood grouping tests. Two different blood types were evaluated with results consistent with the traditional methods, testifying the usefulness of this methodology. © 2018 Wiley Periodicals, Inc. This paper addresses two key challenges toward an integrated forward error-correcting biosensor based on our previously reported self-assembled quick-response (QR) code. The first challenge involves the choice of the paper substrate for printing and self-assembling the QR code. We have compared four different substrates that includes regular printing paper, Whatman filter paper, nitrocellulose membrane and lab synthesized bacterial cellulose. We report that out of the four substrates bacterial cellulose outperforms the others in terms of probe (gold nanorods) and ink retention capability. The second challenge involves remote activation of the analyte sampling and the QR code self-assembly process. In this paper, we use light as a trigger signal and a graphite layer as a light-absorbing material. The resulting change in temperature due to infrared absorption leads to a temperature gradient that then exerts a diffusive force driving the analyte toward the regions of self-assembly. The working principle has been verified in this paper using assembled biosensor prototypes where we demonstrate higher sample flow rate due to light induced thermal gradients.

A visual colorimetric microfluidic paper-based analytical device (μPAD) was constructed following the direct synthesis of enzyme-inorganic hybrid nanomaterials on the paper matrix. An inorganic solution of MnSO4 and KH2PO4 containing a diluted enzyme (glucose oxidase, GOx) was subsequently pipetted onto cellulose paper for the in situ growth of GOx@Mn3(PO4)2 hybrid functional materials. The characterization of the morphology and chemical composition validated the presence of hybrid materials roots in the paper fiber, while the Mn3(PO4)2 of the hybrid provided both a surface for enzyme anchoring and a higher peroxidase-like catalytic activity as compared to the Mn3(PO4)2 crystal that was synthesized without enzyme modulation. This new approach for the in situ growth of an enzyme-inorganic hybrid on a paper matrix eliminates centrifugation and the dry process by casting the solution on paper. The sensing material loading was highly reproducible because of the accuracy and stability of pipetting, which eventually contributed to the reliability of the μPAD. The self-assembled natural and artificial enzyme hybrid on the μPADs specifically detected glucose from a group of interferences, which shows great specificity using this method. Moreover, the colorimetric signal exhibited detection limitation for glucose is 0.01mM, which lies in the physiological range of glucose in biological samples.

Background Until now, environmental sustainability issues are almost entirely unsolved for packaging materials. With the final aim of finding materials with a single recycling channel, cellulose fiber/poly(vinyl)alcohol composites were investigated. Methods After extrusion and injection molding, samples of composite with different cellulose fiber content (30%, 50% and 70% w/w) were tested. Results Tensile mechanical tests exhibited an improvement in composite stiffness when the reinforcement content was increased together with a decrease in composite elongation. Solubility tests performed at room temperature and 45°C showed different behavior depending on the water-resistant film applied on the composite (50% cellulose fiber content). In particular, the uncoated composite showed complete solubility after 2 hours, whereas at the same time point, no solubility occurred when a non-water-soluble varnish was used. Conclusions The proposed composites, named Poly-paper, appear to warrant further investigation as highly sustainable packaging.

In this study, we demonstrate a three-dimensional surface-modified origami-paper-based analytical device (3D-soPAD) for immunoassay applications. The platform enables the sequential steps of immunoassays to be easily performed using a folded, sliding paper design featuring multiple pre-stored reagents, allowing us to take advantage of the vertical diffusion of the analyte through the different paper layers. The cellulose substrate is composed of carboxymethyl cellulose modified with N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride and N-hydroxysuccinimide, which provide covalent bonding sites for bio-recognition molecules. After the optimization of the operation parameters, we determined the detection limit of the 3D-soPAD for human immunoglobulin G (HIgG) which can be as low as 0.01 ng mL-1, with a total turnaround time of 7 min. In order to study the long-term storage of the platform, anti-HIgG horseradish peroxidase (aHIgG-HRP) conjugates were stored by freeze-drying in sugar matrices composed of 10% sucrose/10% trehalose (w/w%) on the paper device, retaining 80% of their activity after 75 days of storage at 4 °C. To evaluate the performance of the paper device using real samples, we demonstrated the detection of protein A (a biomarker for Staphylococcus aureus infection) in highly viscous human synovial fluid. These results show that the proposed 3D-soPAD platform can provide sensitive, high-throughput, and on-site prognosis of infection in resource-limited settings.

Over the past decades, researchers have been seeking attractive substrate materials to keep microfluidics improving to outbalance the drawbacks and issues. Cellulose substrates, including thread, paper and hydrogels are alternatives due to their distinct structural and mechanical properties for a number of applications. Thread have gained considerable attention and become promising powerful tool due to its advantages over paper-based systems thus finds numerous applications in the development of diagnostic systems, smart bandages and tissue engineering. To the best of our knowledge, no comprehensive review articles on the topic of thread-based microfluidics have been published and it is of significance for many scientific communities working on Microfluidics, Biosensors and Lab-on-Chip. This review gives an overview of the advances of thread-based microfluidic diagnostic devices in a variety of applications. It begins with an overall introduction of the fabrication followed by an in-depth review on the detection techniques in such devices and various applications with respect to effort and performance to date. A few perspective directions of thread-based microfluidics in its development are also discussed. Thread-based microfluidics are still at an early development stage and further improvements in terms of fabrication, analytical strategies, and function to become low-cost, low-volume and easy-to-use point-of-care (POC) diagnostic devices that can be adapted or commercialized for real world applications., Highlights • Thread microfluidics is a powerful tool in making diagnostic systems. • This review focus on the advances of thread-based microfluidic diagnostic devices. • Thread microfluidic devices have numerous advantages over traditional and paper-based microfluidics. • A perspective on the future trends for the thread microfluidics is also given.

This work reports on the development of a 3D microfluidic paper-based device (3D µPAD) for glucose detection using organic-inorganic hybrid nanoflower technology to immobilize the bi-enzymatic system (glucose oxidase and horseradish peroxidase). The system is based on nanoflowerssupported on cellulose paper (the microreactor zone) coupled to 3,3',5,5'-tetramethylbenzidine (TMB) as the colorimetric probe in the detection zone. We used a digital camera for the quantitative analysis of glucose with the S coordinate of the HSV color space as the analytical parameter. Under optimal operational conditions, linearity was observed for glucose concentrations up to 300 μM, with a detection limit of 15.6 µM. The biosensor is reusable and remains stable for 75 days in conventional storage conditions.

Background:Paper is one of the important inputs for the printing industry, and the most important leading parameter in the printing process is its brightness. Brightness can be brought to paper using coatings and sizing. Desired surface properties and, most importantly, surface roughness can be achieved by changing the contents of the coating and sizing of the materials it contains. The use of biomaterials is becoming more important in the paper industry, as they represent substances with a lower carbon footprint. Fish scales are already used as a filling material, cosmetic material and fish food, as well as for determining the age of fish.Methods:Fish scales were brought to different sizes by a milling process. Paper formulations including different amounts of fish scales were prepared with fish scales, and coatings on raw paper were subjected to test printings in IGT-C1, with formulations and physical characteristics of coatings such as brightness, lightfastness, strength, adhesion etc. being determined.Results:Regarding the value of yellowness, mixtures of 2.5%-10% can be used. The maximum value of brightness was obtained from a mixture of 10%. Aging visibly changed the colors.Conclusions:The coatings obtained were brighter than the initial coating compositions. The top quality formulation was the coating with 5% medium-sized fish scale particles.

Paper-based microfluidics is a rapidly progressing inter-disciplinary technology driven by the need for low-cost alternatives to conventional point-of-care diagnostic tools. For transport of reagents/analytes, such devices often consist of interconnected hydrophilic fluid-flow channels that are demarcated by hydrophobic barrier walls that extend through the thickness of the paper. Here, we present a laser-based fabrication procedure that uses polymerisation of a photopolymer to produce the required fluidic channels in paper. Experimental results showed that the structures successfully guide the flow of fluids and allow containment of fluids in wells, and hence the technique is suitable for fabrication of paper-based microfluidic devices. The minimum width for the hydrophobic barriers that successfully prevented fluid leakage was ~120 μm and the minimum width for the fluidic channels that can be formed was ~80 μm, the smallest reported so far for paper-based fluidic patterns.

A simple approach to the evaluation of blood coagulation using a microfluidic paper-based lateral flow assay (LFA) device for point-of-care (POC) and self-monitoring screening is reported. The device utilizes whole blood, without the need for prior separation of plasma from red blood cells (RBC). Experiments were performed using animal (rabbit) blood treated with trisodium citrate to prevent coagulation. CaCl2 solutions of varying concentrations are added to citrated blood, producing Ca(2+) ions to re-establish the coagulation cascade and mimic different blood coagulation abilities in vitro. Blood samples are dispensed into a paper-based LFA device consisting of sample pad, analytical membrane and wicking pad. The porous nature of the cellulose membrane separates the aqueous plasma component from the large blood cells. Since the viscosity of blood changes with its coagulation ability, the distance RBCs travel in the membrane in a given time can be related to the blood clotting time. The distance of the RBC front is found to decrease linearly with increasing CaCl2 concentration, with a travel rate decreasing from 3.25 mm min(-1) for no added CaCl2 to 2.2 mm min(-1) for 500 mM solution. Compared to conventional plasma clotting analyzers, the LFA device is much simpler and it provides a significantly larger linear range of measurement. Using the red colour of RBCs as a visible marker, this approach can be utilized to produce a simple and clear indicator of whether the blood condition is within the appropriate range for the patient's condition.

We have reported the fabrication of flexible graphene-paper electrode (GPE) with a flat surface, whose internal structure has been formed with gradient porous build-up (from the surface to the 2-hydroxyethyl cellulose (HC)-coated paper). HC solution was used as a binder to form the gradient porous graphene layer, enabling it to create an anchoring force between the porous graphene layer and the filter paper. The morphology of GPE was investigated using a scanning electron microscope, and the surface resistance of the GPE as a function of graphene content was determined using four-probe method. The electrochemical performance of the GPE was evaluated using a three-electrode test cell by cyclic voltammetry. The gravimetric capacitance of GPE was found to be 120 F per gram of graphene, and the capacitance retention was within ca. 96% for over 500 cycles. This could be attributed to both the low surface resistance resulting from the flat surface and the high electrochemical activity caused by the gradient porous structure. This unique structure not only offers an enhanced conductivity and good electrical contact between the electrode and electrolyte but also helps GPE to maintain good cyclic stability, proving its potential for use in various rechargeable and portable energy-storage devices.

Dental root canal treatment is required when dental caries progress to infection of the dental pulp. A major goal of this treatment is to provide complete decontamination of the dental root canal system. However, the morphology of dental root canal systems is complex, and many human dental roots have inaccessible areas. In addition, dental reinfection is fairly common. In conventional treatment, a cotton pellet and paper point made from plant cellulose is used to dry and sterilize the dental root canal. Such sterilization requires a treatment material with high absorbency to remove any residue, the ability to improve the efficacy of intracanal medication and high biocompatibility. Bacterial cellulose (BC) is produced by certain strains of bacteria. In this study, we developed BC in a pointed form and evaluated its applicability as a novel material for dental canal treatment with regard to solution absorption, expansion, tensile strength, drug release and biocompatibility. We found that BC has excellent material and biological characteristics compared with conventional materials, such as paper points (plant cellulose). BC showed noticeably higher absorption and expansion than paper points, and maintained a high tensile strength even when wet. The cumulative release of a model drug was significantly greater from BC than from paper points, and BC showed greater compatibility than paper points. Taken together, BC has great potential for use in dental root canal treatment.

This review shows the recent advances and state of the art in paper-based analytical devices (PADs) through the analysis of their integration with microfluidics and LOC micro- and nanotechnologies, electrochemical/optical detection and electronic devices as the convergence of various knowledge areas. The important role of the paper design/architecture in the improvement of the performance of sensor devices is discussed. The discussion is fundamentally based on μPADs as the new generation of paper-based (bio)sensors. Data about the scientific publication ranking of PADs, illustrating their increase as an experimental research topic in the past years, are supplied. In addition, an analysis of the simultaneous evolution of PADs in academic lab research and industrial commercialization highlighting the parallelism of the technological transfer from academia to industry is displayed. A general overview of the market behaviour, the leading industries in the sector and their commercialized devices is given. Finally, personal opinions of the authors about future perspectives and tendencies in the design and fabrication technology of PADs are disclosed., A. M. Strong cellulose nanofibrils derived from bacteria or wood, can form ductile or tough networks, that are suitable as functional materials. Here, we show that freeze-dried bacterial cellulose nanofibril aerogels can be used as templates for making lightweight porous magnetic aerogels, which can be compacted into a stiff magnetic nanopaper. The 20–70-nm-thick cellulose nanofibrils act as templates for the non-agglomerated growth of ferromagnetic cobalt ferrite nanoparticles (diameter, 40–120 nm). Unlike solvent-swollen gels and ferrogels our magnetic aerogel is dry, lightweight, porous (98%), flexible, and can be actuated by a small household magnet. Moreover, it can absorb water and release it upon compression. In this paper, a cellulose-based Electro-Active Paper (EAPap) energy scavenging transducer is presented. Cellulose is proven as a smart material, and exhibits piezoelectric effect. Specimens were prepared by coating gold electrodes on both sides of cellulose film. The fabricated specimens were tested by a base excited aluminum cantilever beam at resonant frequency. Different tests were performed with single and multiple parallel connected electrodes coated on the cellulose film. A maximum of 131 mV output voltage was measured, when three electrodes were connected in parallel. It was observed that voltage output increases significantly with the area of electrodes. From these results, it can be concluded that the piezoelectricity of cellulose-based EAPap can be used in energy transduction application.

Paper-based analytical devices (PAD) emerge in the scientific community since 2007 as low-cost, wearable and disposable devices for point-of-care diagnostic due to the widespread availability, long-time knowledge and easy manufacturing of cellulose. Rapidly, electrodes were introduced in PAD for electrochemical measurements. Together with biological components, a new generation of electrochemical biosensors was born. This review aims to take an inventory of existing electrochemical paper-based biosensors and biofuel cells and to identify, at the light of newly acquired data, suitable methodologies and crucial parameters in this field. Paper selection, electrode material, hydrophobization of cellulose, dedicated electrochemical devices and electrode configuration in biosensors and biofuel cells will be discussed.

Herein, we introduce a new strategy for green, in-situ generation of silver nanoparticles using flexible and transparent bacterial cellulose nanopapers. In this method, adsorbed silver ions on bacterial cellulose nanopaper are reduced by the hydroxyl groups of cellulose nanofibers, acting as the reducing agent producing a bionanocomposite "embedded silver nanoparticles in transparent nanopaper" (ESNPs). The fabricated ESNPs were investigated and characterized by field emission scanning electron microscopy (FE-SEM), UV-visible spectroscopy (UV-vis), Fourier-transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA) and energy-dispersive X-ray spectroscopy (EDX). The important parameters affecting the ESNPs were optimized during the fabrication of specimens. The resulting ESNPs were used as a novel and sensitive probe for the optical sensing of cyanide ion (CN(-)) and 2-mercaptobenzothiazole (MBT) in water samples with satisfactory results. The change in surface plasmon resonance absorption intensity of ESNPs was linearly proportional to the concentration in the range of 0.2-2.5 µg mL(-1) and 2-110 µg mL(-1) with a detection limit of 0.012 µg mL(-1) and 1.37 µg mL(-1) for CN(-) and MBT, respectively.

The systematic modification of the surface charge of lignocellulose fibers was performed with a polyelectrolyte layer-by-layer (LbL) nanocoating process to produce negatively and positively charged fibers. The fibers were coated with 20–50 nm thick polymer surface layers which subsequently increased interaction between the fibers during paper formation. The modified fibers were added to standard fibers at varying proportions to produce paper with corresponding variation in properties such as strength and electrical conductivity. Paper strength was doubled by manipulating the surface charge and coating thickness of the LbL-treated pulp fibers. It is demonstrated that the LbL coating process increased the fiber interactions and that these interactions enhanced the paper properties. This process, when applied to a simulated sample of recycle grade of fibers, produce paper with an increase in tear strength as compared with untreated fiber paper. Nanocoating fibers with polythiophene/polyallylamine multilayers produced marginally conductive pulp and paper. Paper electrical conductivity was proportional to the number of the bilayers deposited.

A highly sensitive analytical method for evaluation of poly(L-lactide) (PLA), poly(epsilon-caprolactone) (PCL), poly(beta-hydroxybutyrate) (PHB), and poly(butylene succinate) (PBS) degradability was developed using coated cellulose paper, prepared by penetration and adhesion of these plastics into/onto the cellulose paper. Enzymatic degradability of the obtained plastic coated papers was evaluated using various commercial proteases and lipases. PLA coated paper was highly susceptible to subtilisin and mammalian enzymes, alpha-chymotrypsin, elastase and trypsin. To our knowledge, this is the first report on the degradation of PLA coated paper using subtilisin and mammalian enzymes. Almost all lipase preparations degraded PCL and PHB coated papers but not PBS coated paper. The biodegradability of plastic coated paper was greater than that of plastic powder. The penetration of plastic into cellulose paper by coating improved the plastic degradability, and can be regulated easily.

Combining microfluidic paper-based analytical device (μ-PAD) and the molecular imprinting technique, a visible light photoelectrochemical (PEC) sensing platform for the detection of pentachlorophenol (PCP) was established on gold nanoparticles (AuNPs) decorated paper working electrode using polypyrrole-functionalized ZnO nanoparticles. Ascorbic acid (AA) was exploited as an efficient and nontoxic electron donor for scavenging photogenerated holes under mild solution medium and facilitating the generation of stable photocurrent. The microfluidic molecular imprinted polymer-based PEC analytical origami device is developed for the detection of PCP in the linear range from 0.01 ng mL −1 to 100 ng mL −1 with a low detection limit of 4 pg mL −1 . This disposable microfluidic PEC origami device would provide a new platform for sensitive, specific, and multiplex assay in public health, environmental monitoring, and the developing world.

Plant cell walls consist largely of cellulose, non-cellulosic polysaccharides and lignin. Concerted attempts are underway to convert wall polysaccharides from crop plant residues into renewable transport fuels and other valuable products, and to exploit the dietary benefits of cereal grain wall polysaccharides in human health. Attempts to improve plant performance for these applications have involved the manipulation of the levels and structures of wall components. Some successes in altering non-cellulosic polysaccharides has been achieved, but it would appear that drastic changes in cellulose are more difficult to engineer. Nevertheless, future prospects for both genetically modified (GM) and non-GM technologies to modify plant cell wall composition and structure remain bright, and will undoubtedly find applications beyond the current focus on human health and biofuel production.

The detection of pesticides has attracted considerable attention in numerous fields, such as environmental monitoring and food safety. Although traditional sensors for pesticides have been widely explored due to their high sensitivity and specificity, it is still challenging to develop a low-cost, portable, fast, and easy-to-use detection method for the public use at home or in the field. To address these challenges, herein, we report a novel paper-based molecular imprinted polymer (MIP)-grafted multi-disk micro-disk plate (P-MIP-MMP) for sensitive and specific chemiluminescence (CL) detection of pesticides through an indirect competitive assay using 2,4-dichlorophenoxyacetic acid (2,4-D) as a proof-of-concept analyte. The MIP-grafted paper disks were prepared by a simple in situ polymerization of MIP layer on the surface of cellulose fibers in paper. The quantification mechanism of this P-MIP-MMP is based on a competition between free 2,4-D and tobacco peroxidase (TOP) labeled 2,4-D and the enzyme catalyzed CL emission from the luminol-TOP-H2O2 CL system. At optimal conditions, this P-MIP-MMP can detect 2,4-D at the concentration of femtomolar level. This approach provided a powerful protocol for simple, low-cost, rapid, and high-throughput detection of pesticides in real samples with satisfactory results for use in areas such as food inspection and environmental monitoring.

In order to understand the electro-mechanical behavior of piezoelectric electro active paper (EAPap), the converse and direct piezoelectric characterization of cellulose EAPap was studied and compared. A delay between the electrical field and the induced strain of EAPap was observed due to the inner nano-voids or the localized amorphous regions in layer-by-layered structure to capture or hold the electrical charges and remnant ions. The linear relation between electric field and induced strain is also observed. The electro-mechanical performance of EAPap is discussed in detail in this paper.
