The cultivated tomato Solanum lycopersicum suffered a severe attack by the whitefly Bemisia tabaci (Gennadius), causing damage to leaves by feeding as well as transmitting the tomato yellow leaf curl virus (TYLCV), while the wild tomato S. habrochaites is considerably less appealing to this insect species. It is reported that B. tabaci shows innate avoidance to R-curcumene, which is produced naturally by S. habrochaites. However, the mechanisms involved in the avoidance behavior of B. tabaci in response to this chiral compound are still unclear yet. In this study, the functional and binding characterization of odorant-binding protein 1 of B. tabaci (BtOBP1) were examined in vivo and in vitro against R-curcumene. The obtained results showed that BtOBP1 exhibits specific binding activity to R-curcumene, which acts as repellents to B. tabaci. By using a fluorescence-based binding assay, the difference of binding-affinity for R-curcumene between wild type BtOBP1 and the mutant BtOBP1 to R-curcumene was performed, which resulted in a single amino acid mutation (ASN108 > SER); moreover, BtOBP1-N108 displays significantly decreased binding affinities to R-curcumene. Most interestingly, a knock-down experiment with the BtOBP1 showed that the whitefly responses to R-curcumene are impaired. This study illustrated that BtOBP1 is a crucial protein involved in the perception and discrimination of R-curcumene. Our findings may provide an excellent chance of finding a suitable antagonist of eco-friendly features that can block the perception of chemosensory signals in insects, preventing behaviors like food-finding.

Spodoptera litura is an important pest that causes significant economic damage to numerous crops worldwide. Sex pheromones (SPs) mediate sexual communication in S. litura and show a characteristic degree of rhythmic activity, occurring mainly during the scotophase; however, the specific regulatory mechanisms remain unclear. Here, we employed a genome-wide analysis to identify eight candidate circadian clock genes in S. litura. Sequence characteristics and expression patterns were analyzed. Our results demonstrated that some circadian clock genes might regulate the biosynthesis and perception of SPs by regulating the rhythmic expression of SP biosynthesis-related genes and SP perception-related genes. Interestingly, all potential genes exhibited peak expression in the scotophase, consistent with the SP could mediate courtship and mating behavior in S. litura. Our findings are helpful in elucidating the molecular mechanism by which circadian clock genes regulate sexual communication in S. litura.

The tobacco cutworm Spodoptera litura (Lepidoptera: Noctuidae) is a polyphagous pest with a highly selective and sensitive chemosensory system involved in complex physiological behaviors such as searching for food sources, feeding, courtship, and oviposition. However, effective management strategies for controlling the insect pest populations under threshold levels are lacking. Therefore, there is an urgent need to formulate eco-friendly pest control strategies based on the disruption of the insect chemosensory system. In this study, we identified 158 putative chemosensory genes based on transcriptomic and genomic data for S. litura, including 45 odorant-binding proteins (OBPs, nine were new), 23 chemosensory proteins (CSPs), 60 odorant receptors (ORs, three were new), and 30 gustatory receptors (GRs, three were new), a number higher than those reported by previous transcriptome studies. Subsequently, we constructed phylogenetic trees based on these genes in moths and analyzed the dynamic expression of various genes in head capsules across larval instars using quantitative real-time polymerase chain reaction. Nine genes–SlitOBP8, SlitOBP9, SlitOBP25, SlitCSP1, SlitCSP7, SlitCSP18, SlitOR34, SlitGR240, and SlitGR242–were highly expressed in the heads of 3- to 5-day-old S. litura larvae. The genes differentially expressed in olfactory organs during larval development might play crucial roles in the chemosensory system of S. litura larvae. Our findings substantially expand the gene inventory for S. litura and present potential target genes for further studies on larval feeding in S. litura.

BACKGROUND Carboxyl/cholinesterases (CCEs) are thought to play a pivotal role in the degradation of sex pheromones and plant-derived odorants in insects, but their exact biochemistry and physiological functions remain unclear. RESULTS In this study, two paralogous antennae-enriched CCEs from Plutella xylostella (PxylCCE16a and 16c) were identified and functionally characterized. High-purity protein preparations of active recombinant PxylCCE16a and 16c have been obtained from Sf9 insect cells by Ni2+ affinity purification. Our results revealed that the purified recombinant PxylCCE016c is able to degrade two sex pheromone components Z9-14:Ac and Z11-16:Ac at 27.64 ± 0.79% and 24.40 ± 3.07%, respectively, while PxylCCE016a presented relatively lower activity. Additionally, a similar difference in activity was measured in plant-derived odorants. Furthermore, both CCEs displayed obvious preferences for the two sex pheromone components, especially on Z11-16:Ac (Km values are in the range 7.82-45.06 μmol L-1 ) which much lower than plant odorants (Km values are in the range 1290-4030 μmol L-1 ). Furthermore, the activity of the two newly identified CCEs is pH-dependent. The activity at pH 6.5 is obviously higher than that at pH 5.0. Interestingly, only PxylCCE016c can be inhibited by a common esterase inhibitor triphenyl phosphate (TPP) with LC50 of 1570 ± 520 μmol L-1 . CONCLUSION PxylCCE16c plays a more essential role in odorant degradation than PxylCCE16a. Moreover, the current study provides novel potential pesticide targets for the notorious moth Plutella xylostella. © 2021 Society of Chemical Industry.

The German cockroach Blattella germanica (L.) is an important pest in medical, veterinary, and public health. Studies on the olfaction mechanism of hemimetabolous insects have rarely been reported, especially in cockroaches. Pheromone-binding proteins (PBPs) play a vital role in insect sex pheromone recognition, which solubilize and carry the hydrophobic pheromonal compounds through the antennal lymph to receptors. In this study, two potential PBPs (BgerOBP26 and BgerOBP40) were identified on the basis of their biased expression in male antennae using tissue transcriptome data and verified by the quantitative real-time polymerase chain reaction approach. We then expressed and purified the two identified odorant-binding proteins (OBPs) using the Escherichia coli expression system and affinity purification. In vitro binding studies showed that the two OBPs display stronger binding affinities to the female volatile sex pheromone blattellaquinone than to its analogues and contact sex pheromone components. Finally, three-dimensional modeling of the two OBPs and dock conformation with sex pheromone molecules showed BgerOBP26 has a larger odorant cavity and more conservative active amino acid residues than BgerOBP40. These results illuminated the binding characteristics of potential PBPs of B. germanica, which could lay the groundwork for improved understanding of many aspects of the chemical ecology of B. germanica. Moreover, this information complements the understanding of the olfactory molecular mechanism in cockroaches and provides potential gene targets for B. germanica control.

Background Chemosensory proteins (CSPs) are thought to play essential roles in insect chemical communication, but their exact physiological functions remain unclear. Results In this study, we investigated the functions of the CSP2 gene in the whitefly Bemisia tabaci using protein expression and the binding affinity spectrum of CSP2 to different types of odor molecules. Moreover, the evolutionary characteristics of the CSP2 gene were studied. The data obtained using binding assay showed that the CSP2 protein can bind to a broad range of plant volatiles including the homoterpene (E)-3,8-dimethyl-1,4,7-nonatriene (DMNT) and its analogs. In addition, using a behavioral experimental approach we identified that DMNT can repel the selection and oviposition of B. tabaci. Furthermore, protein structure modeling, molecular docking analyses and a functional mutation experiment were carried out resulting in the final identification of key amino acid residue Y11, which displayed important roles in the binding of CSP2 to DMNT. The results also showed that Y11 is located in the pocket region where CSP2 has a pi-alkyl interaction with DMNT. Meanwhile, comparative and evolutionary analyses indicated that CSP2 shared a high sequence similarity with CSPs of other insect family members such as Sternorrhyncha and Auchenorrhyncha including aphids, whiteflies and planthoppers. Conclusion These results suggested that CSP2 likely contributes to mediating responses of B. tabaci to plant volatiles, which may play a pivotal role in its feeding and oviposition preferences. Moreover, these findings could provide key information for exploring efficiency monitoring and integrated pest management strategies of B. tabaci.

Whiteflies are Hemipterans that typically feed on the undersides of plant leaves. They cause severe damage by direct feeding as well as transmitting plant viruses to a wide range of plants. However, it remains largely unknown which genes play a key role in development and host selection. In this study, weighted gene co-expression network analysis was applied to construct gene co-expression networks in whitefly. Nineteen gene co-expression modules were detected from 15560 expressed genes of whitefly. Combined with the transcriptome data of salivary glands and midgut, we identified three gene co-expression modules related to host plant selection. These three modules contain genes related to host-plant recognition, such as detoxification genes, chemosensory genes and some salivary gland-associated genes. Results of Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses elucidated the following pathways involved in these modules: lysosome, metabolic and detoxification pathways. The modules related to the development contain two co-expression modules; moreover, the genes were annotated to the development of chitin-based cuticle. This analysis provides a basis for future functional analysis of genes involved in host-plant recognition.

The whitefly Bemisia tabaci (Gennadius) causes severe damage to cultivated tomato in many regions of the world through direct feeding and indirectly through transmission of plant viruses. Field observations show that B. tabaci is rarely infested the non-host plants such as the wild tomato Solanum habrochaites; however, the molecular mechanism involved in the recognition of wild plant odors is still unclear. In this study, we assessed the effects of S. habrochaites on the survival, fecundity, and egg hatchability of the Mediterranean (MED) species of B. tabaci. Expression and splicing of stress-response genes in whitefly exposed to S. habrochaites was analyzed using RNA-sequencing data and alternative splicing analysis. These results indicated that the S. habrochaites treatment can induce the expression of environmental stress genes in B. tabaci. This study may help us to a better understanding of the molecular mechanisms involved in the olfactory recognition of non-host volatiles particularly wild tomato relative. Furthermore, the findings of this study may provide excellent chances of finding a suitable antagonist of eco-friendly properties which can block the perception of chemosensory signals. Thereby, the feeding behavior and food preferences of B. tabaci can be manipulated and thus insect populations can eventually be controlled.

Background The German cockroach, Blattella germanica, is one of the most severe pests of urban and rural areas. High-throughput genetic screening approaches indicate that the olfactory system of this pest is extremely powerful because it has an extensive array of olfactory receptor genes compared with many other insect species. Several of these genes have been identified previously, but their functions have not yet been characterized. Results This study describes the sequence of five transcriptomes of B. germanica adult male antennae, female antennae, maxillary palps, legs, and fifth-instar nymph antennae to investigate expression patterns of odorant receptors (ORs). Approximately 90% of ORs were found to be the most highly expressed genes in adult or nymph antennae. Additionally, every OR requires an odorant co-receptor (Orco) to become fully functional, and this was selected and successfully inhibited by injection of the corresponding double-stranded (ds)RNA targeting the Orco. A strong RNA interference (RNAi) effect was observed in which > 75% of Orco messenger RNA (mRNA) was clearly suppressed after 72 h of treatment. Olfactory behavioral assays showed that Orco-impaired B. germanica respond more slowly and show less attraction to one volatile sex pheromone and food resources compared with a control group. Conclusion The results show that Orco plays a pivotal role in both sex pheromone and food-seeking olfactory processes, and provide an alternative genetic technique for controlling this urban pest species by olfactory disruption. © 2020 Society of Chemical Industry.

Insect antennae play a fundamental role in perceiving and recognizing a broad spectrum of conventional semiochemicals and host plant-derived odors. As such, genes that are tightly associated with the antennae are thought to have olfactory-related roles related to signal transduction mechanisms. Several mechanisms suggest that enzymatic inactivation could contribute to the signal termination process, such as odorant-degrading enzymes (ODEs). To date, a few ODEs have been identified and characterized in detail in insect herbivores, but little is known about aldehyde oxidases (AOXs); moreover, direct in vivo experimental evidence is needed. AOXs are a major family of metabolic enzymes that oxidize a variety of aromatic aldehydes, and they may also play a significant role in detoxification and degradation of environmental chemical cues. Here, we report on the identification and characterization of a novel cDNA encoding the putative odorant-degrading enzyme, PxylAOX3, from the antennae of the diamondback moth, (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae). The purified recombinant protein showed a wide-range of substrate zymography oxidizing both sex pheromone compounds as well as plant-derived aldehydes with distinct activities. Our data suggest PxylAOX3 might be involved in the degradation of many structurally diverse aldehyde odorants. Furthermore, PxylAOX3 could participate in olfactory neuron protection by inactivation of redundant odorants and xenobiotic detoxification, making it a potential target for pesticide development as well.

The sycamore lace bug, Corythucha ciliata (Say) is an invasive pest infesting trees of the genus Platanus. Both adults and nymphs damage the foliage of sycamore trees. Nymphs cannot survive in low temperatures; however, the sycamore lace bug overwinters as adults. In this study, we analyzed the metabolite profiles of this pest to determine significantly regulated metabolites during paurometabolous development from nymphs to adults. The identification of metabolites is essential to convert analytical data into meaningful biological knowledge. A total of 62 metabolites were identified using GC-MS. Among them, 29 different metabolites showed differences in content among nymphs, adult females (AF), and adult males (AM). Five of the 29 metabolites, including caffeic acid, D-glucose, D-mannose, glycerol and aminooxyacetic acid, were significantly increased and nine of them were significantly decreased during the developmental stages from nymph to adult. In addition, we identified three novel aldo-keto reductase (AKR) genes that may play a significant role in the control of glycerol biosynthesis. Moreover, the characteristics and expression levels of these genes were analyzed. This study will provide us with the necessary information to improve our understanding of the changes in metabolites in C. ciliata during paurometabolous development.

The small brown planthopper (SBPH), Laodelphax striatellus is one of the major insect pests of rice, but little is known about the molecular-level means by which it locates its hosts. SBPH host-seeking behavior heavily relies on chemosensory receptors (CRs). In this study, we utilized genome analysis of the SBPH to identify 169 CRs, including: 133 odorant receptors (ORs), 13 gustatory receptors (GRs) and 23 ionotropic receptors (IRs). The phylogenetic relationships of OR genes from three rice planthoppers and other insect species revealed that the odorant co-receptor (Orco) clade is the most conserved group. Among the candidate GRs, two sugar receptors and five fructose receptors have been identified but no carbon dioxide receptors investigated. Furthermore, we identified homologs of the three highly conserved IR co-receptors. The obtained results will provide us with precious information needed to better understand the interaction between insect pests and crop plants required for effective crop protection.

The whitefly, Bemisia tabaci, is an important invasive economic pest of agricultural crops worldwide. &beta, ionone has a significant oviposition repellent effect against B. tabaci, but the olfactory molecular mechanism of this insect for recognizing &beta, ionone is unclear. To clarify the binding properties of odorant-binding proteins (OBPs) with &beta, ionone, we performed gene cloning, evolution analysis, bacterial expression, fluorescence competitive binding assay, and molecular docking to study the binding function of OBP1 and OBP4 on &beta, ionone. The results showed that after the OBP1 and OBP4 proteins were recombined, the compound &beta, ionone exhibited a reduction in the fluorescence binding affinity to &lt, 50%, with a dissociation constant of 5.15 and 3.62 &mu, M for OBP1 and OBP4, respectively. Our data indicate that &beta, ionone has high affinity for OBP1 and OBP4, which play a crucial role in the identification of oviposition sites in B. tabaci. The findings of this study suggest that whiteflies employ &beta, ionone compound in the selection of the suitable egg-laying sites on host plants during the oviposition behavior.

Insects recognize odorous compounds using sensory neurons organized in olfactory sensilla. The process odor detection in insects requires an ensemble of proteins, including odorant binding proteins, olfactory receptors, and odor degrading enzymes, each of them are encoded by multigene families. Most functional proteins seem to be broadly tuned, responding to multiple chemical compounds with different, but mostly quite similar structures. Based on the hypothesis that insects recognize host volatiles by means of general odorant binding proteins (GOBPs), the current study aimed to characterize GOBPs of the yellow peach moth, Conogethes punctiferalis (Guen&eacute, e). In oviposition preference tests, it was found that the yellow peach moth preferred volatiles from Prunus persica (peach) in finding their host plant. Exposure of the moth to volatiles from peaches affected the expression level of GOBP genes. Binding affinity of GOBPs from yellow peach moth was assessed for 16 host plant volatiles and 2 sex pheromones. The fluorescence ligand-binding assays revealed highest affinities for hexadecanal, farnesol, and limonene with KD values of 0.55 &plusmn, 0.08, 0.35 &plusmn, 0.04, and 1.54 &plusmn, 0.39, respectively. The binding sites of GOBPs from yellow peach moth were predicted using homology modeling and characterized using molecular docking approaches. The results indicated the best binding affinity of both GOBP1 and GOBP2 for farnesol, with scores of &minus, 7.4 and &minus, 8.5 kcal/mol. Thus, GOBPs may play an important role in the process of finding host plants.

The sycamore lace bug, Corythucha ciliata (Say) is a highly invasive pest insect that feeds on sycamore trees (Platanus spp.) worldwide. The interaction between Platanus species and this insect pest has not yet been studied at the molecular level. Therefore, a recent study was conducted to compare the gene expression and metabolite profiles of Platanus acerifolia leaves in response to C. ciliata feeding damage after 24 and 48 h. We employed high throughput RNA sequencing (RNA- seq) to identify a total of 2,828 significantly differentially expressed genes (DEGs) after C. ciliata feeding. In addition, 303 unigenes were found to be up-regulated at both time points. Moreover, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis showed that monoterpenoid biosynthesis, the linoleic acid metabolism pathway, and alpha- linolenic acid metabolism were the most prominent pathways among the DEGs. Further analysis of the metabolite profiles showed that nine metabolites were significantly different before and after C. ciliata damage. In addition, we analyzed DEGs detected in the P. acerifolia and C. ciliata interaction using Mapman. The terpene synthase gene family was also identified. We suggest that the results obtained from DEGs and metabolite analysis can provide important information for the identification of genes involved in the P. acerifolia&ndash, C. ciliata interaction, which might be necessary for controlling C. ciliata efficiently.

Simple Summary Donacia provosti is one of the major pests of aquatic crops. It has been widely distributed in the world causing extensive damage to lotus and rice plants. The larvae generally live-in water; however, little is known about the evolution and molecular mechanisms underlying the adaptation. Here, we generated the first larval transcriptome of D. provosti in order to identify potential genetic mechanisms of aquatic adaptation. About 5036 orthologous clusters were identified among four species and 494 unique clusters were identified from D. provosti larvae including the visual perception. Moreover, 93 orthologous gene pairs were found evolving under positive selection. Our results also showed that 4 gene pairs out of the 93 gene pairs were associated with the “mTOR signaling pathway”, which are predicted to be involved in the molecular mechanism of D. provosti adaptation to the underwater environment. In the light of the increasing availability of transcriptomic information for beetle underwater habitat and evolutionary analyses, it is expected that this paper will provide us with some novel insights into aquatic adaptation in beetles and serves as a foundation for future studies aiming to identify candidate genes underlying the genetic basis of aquatic adaptation in beetles. Abstract Donacia provosti (Fairmaire, 1885) is a major pest of aquatic crops. It has been widely distributed in the world causing extensive damage to lotus and rice plants. Changes in gene regulation may play an important role in adaptive evolution, particularly during adaptation to feeding and living habits. However, little is known about the evolution and molecular mechanisms underlying the adaptation of D. provosti to its lifestyle and living habits. To address this question, we generated the first larval transcriptome of D. provosti. A total of 20,692 unigenes were annotated from the seven public databases and around 18,536 protein-coding genes have been predicted from the analysis of D. provosti transcriptome. About 5036 orthologous cutlers were identified among four species and 494 unique clusters were identified from D. provosti larvae including the visual perception. Furthermore, to reveal the molecular difference between D. provosti and the Colorado potato beetle Leptinotarsa decemlineata, a comparison between CDS of the two beetles was conducted and 6627 orthologous gene pairs were identified. Based on the ratio of nonsynonymous and synonymous substitutions, 93 orthologous gene pairs were found evolving under positive selection. Interestingly, our results also show that there are 4 orthologous gene pairs of the 93 gene pairs were associated with the “mTOR signaling pathway”, which are predicted to be involved in the molecular mechanism of D. provosti adaptation to the underwater environment. This study will provide us with an important scientific basis for building effective prevention and control system of the aquatic leaf beetle Donacia provosti.

The fruit fly Drosophila suzukii is a fruit crop pest that causes a severe economic threat to soft summer fruit worldwide. The male sex pheromone, cis-vaccenyl acetate (cVA) has multiple functions in intra-species communication in Drosophila melanogaster, which is required in male to suppress male-male courtship. D. suzukii males do not produce cVA; however, the odorant receptor for cVA (Or67d) is still functional. The lack of cVA in D. suzukii casts the question of whether this pheromone might have been replaced by another compound similar to cVA that disrupts mating in D. suzukii. In order to address this question, we cloned two D. suzukii adult antenna-specific odorant-binding proteins (OBPs) DsOBP69a and DsOBP76a and aligned with their D. melanogaster orthologues. Moreover, we examined the binding properties of the newly identified recombinant proteins against 26 potential ligands including cVA, using the fluorescence-based ligand binding assay. The alignment showed that DsOBP69a and DsOBP76a, have six conserved cysteines and belong to the classic OBP family. Furthermore, our results revealed that cVA did not bind to DsOBP69a or DsOBP76a proteins. Interestingly, the floral odorant β-ionone and the bitter substance berberine chloride and coumarin displayed high binding ability. It is also worth noting that DsOBP69a and DsOBP76a have different affinities to (Z)-7-Tricosene that may reflect different functional roles. International audience; Insecticides have long been used as the main method in limiting agricultural pests, but their widespread use has resulted in environmental pollution, development of resistances, and biodiversity reduction. The effects of insecticides at low residual doses on both the targeted crop pest species and beneficial insects have become a major concern. In particular, these low doses can induce unexpected positive (hormetic) effects on pest insects, such as surges in population growth exceeding what would have been observed without pesticide application. Methomyl and chlorpyrifos are two insecticides commonly used to control the population levels of the cotton leafworm Spodoptera littoralis, a major pest moth. The aim of the present study was to examine the effects of sublethal doses of these two pesticides, known to present a residual activity and persistence in the environment, on the moth physiology. Using a metabolomic approach, we showed that sublethal doses of methomyl and chlorpyrifos have a systemic effect on the treated insects. We also demonstrated a behavioral disruption of S. littoralis larvae exposed to sublethal doses of methomyl, whereas no effects were observed for the same doses of chlorpyrifos. Interestingly, we highlighted that sublethal doses of both pesticides did not induce a change in acetylcholinesterase activity in head of exposed larvae
