Severe leaf spot disease was observed on Aloe vera plants in the winters of 2011 and 2012 during a survey of various nurseries of Gwalior, India. Irregular, sunken, dark creamish brown spots having reddish brown margin were noticed on both surfaces of the leaves. The causal organism was consistently isolated from symptomatic leaves on potato dextrose agar media (PDA). A total 59 isolates of fungi were recovered from diseased A. vera leaves, and 37 isolates were identified as belonging to the genus Fusarium. On the basis of morphological characteristics and internal transcribed spacer (ITS) region of rDNA amplified using the primers ITS4/ITS5 the pathogen was identified as Fusarium proliferatum (Matsushima) Nirenberg and pathogenicity of the isolate was confirmed by using Koch’s postulates. To the best of our knowledge, this is the first report of leaf spot disease caused by Fusarium proliferatum on A. vera plants in India.

The squash beetle Epilachna chrysomelina (F.) is an important insect pest which causes severe damage to cucurbit plants in Iraq. The aims of this study were to isolate and characterize an endogenous isolate of Myrothecium-like species from cucurbit plants and from soil in order to evaluate its pathogenicity to squash beetle. Paramyrothecium roridum (Tode) L. Lombard & Crous was isolated, its phenotypic characteristics were identified and ITS rDNA sequence analysis was done. The pathogenicity of P. roridum strain (MT019839) was evaluated at a concentration of 107 conidia · ml–1) water against larvae and adults of E. chrysomelina under laboratory conditions. The results revealed the pathogenicity of the isolate to larvae with variations between larvae instar responses. The highest mortality percentage was reported when the adults were placed in treated litter and it differed significantly from adults treated directly with the pathogen. Our results documented for the first time that P. roridum has potential as an insect pathogen.

Potato is an important crop in Shanxi province, located in north-central China. In 2019 to 2020, 319 potato leaf samples were collected from eight locations distributed in three major potato production areas in Shanxi. BioChip testing revealed the presence of several potato viruses, of which Potato virus Y (PVY) was the most common, reaching an incidence of 87.8% of all symptomatic samples. Immunocaptured multiplex reverse transcription (RT) PCR was used to identify strains for all 280 PVY-positive samples, unveiling 242 samples infected with a single strain of PVY (86.4%) and 38 (13.6%) with a mixed infection. Of samples with a single-strain infection, PVY-SYR-II accounted for 102 (42.1%), followed by PVYN-Wi (33, 13.6%), PVY-SYR-I (28, 11.6%), 261-4 (22, 9.1%), PVYNTNa (20, 8.3%), PVYNTNb (19, 7.9%), and PVY-SYR-III (18, 7.4%). Seven isolates representing different recombinants were selected for whole genome sequencing. Phylogenetic and recombination analyses confirmed the RT-PCR–based strain typing for all seven strains of PVY found in Shanxi. SXKL-12 is the first SYR-III strain from potato reported from China. However, unlike that in other known SYR-III isolates, the region positioned from 1,764 to 1,902 nt in SXKL-12 shared the highest sequence identity of 82.2% with an uncharacterized PVY isolate, JL-23, from China. Interestingly, PVYN-Wi isolate SXZY-40 also possessed a more divergent sequence for the region positioned from 6,156 to 6,276 nt than other N-Wi isolates known to date, sharing the highest identity of 86.6% with an uncharacterized Chinese PVY isolate, JL-11. Pathogenicity analysis of dominant strains PVY-SYR-II and PVYN-Wi in six local popular potato cultivars revealed that ‘Kexin 13’, ‘Helan 15’, and ‘Jizhangshu 12’ were susceptible to these two strains, with mild mottling or mosaic symptom expression, and three cultivars, ‘Jinshu 16’, ‘Qingshu 9’, and ‘Xisen 6’, were fully resistant.

Cronobacter species are a group of Gram-negative opportunistic pathogens, which cause meningitis, septicemia, and necrotizing enterocolitis in neonates and infants, with neurological sequelae in severe cases. Interest in Cronobacter has increased significantly in recent years due to its high virulence in children. In this review, we summarize the current understanding of the prevalence of Cronobacter species in several important food types. We discuss the response mechanisms enabling persistence in adverse growth conditions, as well as its pathogenicity. We emphasize the food safety concerns caused by Cronobacter and subsequent control methods, and clinical treatments.

Root rot is a common disease in soybean (Glycine max) and field pea (Pisum sativum), which restrain increased production in Canada. Sixty-seven isolates of Rhizoctonia were recovered from various diseased plants in Alberta, Canada along with three isolates from diseased soybean plants in Manitoba, Canada. According to their anastomosis behavior, 23 (32.9%) of the isolates were identified as anastomosis group (AG) 4 (AG4), 7 (10.0%) were AG2-1, 10 (14.3%) were AG2-2, 7 (10.0%) were AG5, 3 (4.3%) were AG-E and the AGs of the remaining 20 (28.6%) isolates could not be determined. Isolates belonging to AG4 produced typical symptoms of stem rot and root rot on seedlings of soybean and pea and were more aggressive than the AG2-1, AG2-2, AG5 and AG-E isolates. Selected isolates of AG4, AG2-1, AG2-2, AG5 and AG-E were to some degree able to infect common crops in Alberta, which included barley, canola, corn, faba bean, flax, lupin, lentil, pea, potato, soybean, and wheat. The genetic variability among these isolates was evaluated using phylogenetic analysis based on the rDNA ITS sequences and inter-simple sequence repeat (ISSR) markers. For the ITS sequence analysis, a neighbour-joining tree was constructed using the PAUP program, which clustered the Rhizoctonia isolates into five groups (Groups I to V). However, no correlation was observed between AGs, locations, aggressiveness or host origins. For the ISSR analysis, 54 polymorphic ISSR patterns were identified, indicating a high level of diversity among the isolates.

During the period of June to October in 2018, a widespread decline was observed on kiwifruit vines in the vineyards located in the Altınordu, Fatsa, and Perşembe districts of Ordu province in Turkey. The symptoms were associated with reddish-brown rots expanding from the root to the collar with sparse off-color foliage. Based on the percentage of the total infected samples across 18 vineyards, the most common oomycete species were Globisporangium intermedium (37.1%), Phytopythium vexans (34.3%), Globisporangium sylvaticum (14.3%), Globisporangium heterothallicum (11.4%), and Pythium dissotocum (2.9%). The morphological identification of isolates was confirmed based on partial DNA sequences containing the nuclear rDNA internal transcribed spacer region gene and the mitochondrial cytochrome c oxidase subunit II gene. The optimum growth temperature and the optimum pH values of the five species ranged from 22.98 to 28.25°C and 5.67 to 8.51, respectively. Pathogenicity tests on the seedlings of kiwifruit cv. Hayward revealed significant differences in virulence among isolates. Phytopythium vexans and Globisporangium sylvaticum isolates caused severe root and collar rot resulting in seedling death, while Globisporangium heterothallicum and Globisporangium intermedium isolates had relatively lower virulence. All Globisporangium spp. and Phytopythium vexans isolates significantly decreased plant growth parameters (plant height, shoot and root dry weights, and root length); however, Pythium dissotocum caused very mild symptoms and did not affect these parameters of growth. To our knowledge, this is the first study reporting Globisporangium sylvaticum, Globisporangium heterothallicum, and Globisporangium intermedium causing root and collar rot on kiwifruit not only in Turkey but also in the world.

ibrio parahaemolyticus is the main cause of foodborne gastroenteritis, which is widely distributed in shellfish and other seafood. Most V. parahaemolyticus are nonpathogenic, and only a few types, such as serotype O3:K6, are pathogenic, which is also the most prevalent strain in Asia. However, the relationship between this serotype and pathogenicity has yet to be established. The waaf gene is located in the O antigen synthesis gene cluster. Thus, we constructed a waaf gene deletion mutant (i.e., △waaf) of wild-type (WT) which isolated from shellfish serotype O3:K6 via chitin-mediated transformation technology. We then constructed the △waaf complementary strain (i.e., C-Δwaaf) via the Escherichia coli S17 λpir strain by conjugation. The basic physiological characteristics, adhesion to Caco2 cells, and pathogenicity of the WT, Δwaaf, and C-Δwaaf strains were compared. Growth curves showed no remarkable differences between the WT and Δwaaf strains. However, the Δwaaf strain non-reactive to O3 antisera and other 12 O-group antisera of V. parahaemolyticus. Moreover, the number of flagella and extracellular polysaccharides decreased, the adhesion decreased, and the pathogenicity weakened. These characteristics of the C-Δwaaf strain were similar to those of the WT strain. These results indicated that the waaf gene is vital to the serotype in V. parahaemolyticus, and changes in O antigen could affect the pathogenicity of this bacterium. This study will be helpful to understand the pathogenic mechanism of V. parahaemolyticus.

Mulberry crinkle leaf virus (MCLV) is a novel geminivirus identified from mulberry. The pathogenicity and natural vector transmission of MCLV remain unknown. Here, infectious clones consisting of the complete tandem dimeric genome of MCLV in a binary vector were constructed and agroinoculated into young mulberry plants. The results showed that the infectious clones of MCLV were systemically infectious in mulberry, but the infected mulberry plants did not show any virus infection-like symptoms. The natural transmission vectors of MCLV were also identified from possible vector insects occurring on the MCLV-infected mulberry plants. The vector ability of Tautoneura mori was identified through an inoculation assay. Three of 21 (14.3%) plants inoculated with T. mori collected from MCLV-infected mulberry plants grown naturally were found to be MCLV-positive 50 days postinoculation. These MCLV-positive mulberry plants did not show any virus infection-like symptoms. Collectively, these results suggest that MCLV is infectious to mulberry plants but, by itself, does not induce infection symptoms. The leafhopper T. mori was experimentally determined to be a transmission vector of MCLV for the first time.

The analysis of available scientific literature sources on the modern aspects of pulmonary mycobacteriosis is described in the article. The incidence of mycobacteriosis is 2.3 per 100 thousand of population, and pulmonary manifestation is observed in 94 % of cases. Analytical review has shown that in the clinic of internal and infectious diseases, there are increasingly diagnosed different variants of mycobacteriosis that indicates an increase of atypical mycobacteria role in the formation of respiratory patho­logy in humans. It is ascertained that microbiological study of the basic characteristics of nontuberculosis mycobacteria are not paid enough attention. It was concluded that most often mycobacterioses have tuberculosis-like clinical pattern and are difficultly diagnosed, often form mixed infection with tuberculosis; atypical mycobacteria are characterized by a wide range of resistance and potential pathogenicity for humans and animals. Risk factors of pulmonary mycobacteriosis, variants of clinical course, modern methods of the diagnosis and treatment approaches depending on the course are analyzed. It is shown that treatment of pulmonary mycobacteriosis is long and lasts for 12 months after receiving a negative sputum culture (conversion often takes place in 3–6 months).

Botryosphaeria dothidea causes white rot, which is among the most devastating diseases affecting apple crops globally. In this study, we assessed B. dothidea resistance to carbendazim by collecting samples from warts on the infected branches of apple trees or from fruits exhibiting evidence of white rot. All samples were collected from different orchards in nine provinces of China in 2018 and 2019. In total, 440 B. dothidea isolates were evaluated, of which 19 isolates from three provinces were found to exhibit carbendazim resistance. We additionally explored the fitness and resistance stability of these isolates, revealing that they were no less fit than carbendazim-sensitive isolates in terms of pathogenicity, sporulation, and mycelial growth and that the observed carbendazim resistance was stable. Sequencing of the β-tubulin gene in carbendazim-resistant isolates showed the presence of a substitution at codon 198 (GAG to GCG) that results in an alanine substitution in place of glutamic acid (E198A) in all 19 resistant isolates. A loop-mediated isothermal amplification (LAMP) method was then developed to rapidly and specifically identify this E198A mutation. This LAMP method offers value as a tool for rapidly detecting carbendazim-resistant isolates bearing this E198A mutation and can thus be used for the widespread monitoring of apple crops to detect and control the development of such resistance.

Rice false smut (RFS) is a destructive disease of rice worldwide caused by Ustilaginoidea virens. Nevertheless, there is a lack of efficient and stable artificial inoculation method to simulate the natural infection of U. virens, which is an important factor limiting further research on the pathogen. The purpose of this study was to establish an artificial inoculation method, which can simulate the natural infection process of U. virens without destroying the panicle sheath structure of rice. In this research, rice plants were inoculated by soaking roots at the seedling stage, spraying at the tillering stage, injecting at the booting stage, and again spraying at the flowering stage to determine the appropriate artificial inoculation time. Meanwhile, the panicle sheath instillation method and the injection inoculation method were compared. The results show that stages 6 to 8 of young panicle differentiation are an important period for U. virens infection. There were no significant differences in the mean rates of infected panicles, mean rates of infected grains, and maximum infected grains per panicle between the two inoculation methods. However, the frequency of RFS ball occurrence at the upper part of the panicles was significantly higher on the spikelets inoculated by the injection method than that of spikelets inoculated by natural infection and panicle sheath instillation. Therefore, panicle sheath instillation method was more similar to the natural infection of U. virens in the field. This research exhibited an innovative artificial inoculation method for identification of U. virens pathogenicity and evaluation of rice resistance against RFS.

Lassa fever (LF) is a deadly viral hemorrhagic disease that is endemic to West Africa. The causative agent of LF is Lassa virus (LASV), which causes approximately 300,000 infections and 5,000 deaths annually. There are currently no approved therapeutics or FDA-approved vaccines against LASV. The high genetic variability between LASV strains and immune evasion mediated by the virus complicate the development of effective therapeutics and vaccines. Here, we aim to provide a comprehensive review of the basic biology of LASV and its mechanisms of disease pathogenesis and virulence in various animal models, as well as an update on prospective vaccines, therapeutics, and diagnostics for LF. Until effective vaccines and/or therapeutics are available for use to prevent or treat LF, a better level of understanding of the basic biology of LASV, its natural genetic variations and immune evasion mechanisms as potential pathogenicity factors, and of the rodent reservoir-vector populations and their geographical distributions, is necessary for the development of accurate diagnostics and effective therapeutics and vaccines against this deadly human viral pathogen.

Aspergillus flavus (A. flavus) is one of the most important model environmental fungi which can produce a potent toxin and carcinogen known as aflatoxin. Aflatoxin contamination causes massive agricultural economic loss and a critical human health issue each year. Although a functional vacuole has been highlighted for its fundamental importance in fungal virulence, the molecular mechanisms of the vacuole in regulating the virulence of A. flavus remain largely unknown. Here, we identified a novel vacuole-related protein in A. flavus, the ortholog of phosphatidylinositol-3-phosphate-5-kinase (Fab1) in Saccharomyces cerevisiae. This kinase was located at the vacuolar membrane, and loss of fab1 function was found to affect the growth, conidia and sclerotial development, cellular acidification and metal ion homeostasis, aflatoxin production and pathogenicity of A. flavus. Further functional analysis revealed that Fab1 was required to maintain the vacuole size and cell morphology. Additional quantitative proteomic analysis suggested that Fab1 was likely to play an important role in maintaining vacuolar/cellular homeostasis, with vacuolar dysregulation upon fab1 deletion leading to impaired aflatoxin synthesis in this fungus. Together, these results provide insight into the molecular mechanisms by which this pathogen produces aflatoxin and mediates its pathogenicity, and may facilitate dissection of the vacuole-mediated regulatory network in A. flavus.

A recent olive trunk disease survey performed in the Western Cape Province, South Africa, identified several fungi associated with olive trunk disease symptoms, including species of Basidiomycota, Botryosphaeriaceae, Coniochaetaceae, Calosphaeriaceae, Diaporthaceae, Diatrypaceae, Phaeomoniellaceae, Phaeosphaeriaceae, Symbiotaphrinaceae, Togniniaceae, and Valsaceae. Many of the species recovered had not yet been reported from olive trees; therefore, the aim of this study was to determine their pathogenicity toward this host. Pathogenicity tests were first conducted on detached shoots to select virulent isolates, which were then used in field trials. During field trials, 2-year-old olive branches of 15-year-old trees were inoculated by inserting colonized agar plugs into artificially wounded tissue. Measurements were made of the internal lesions after 8 months. In total, 58 isolates were selected for the field trials. Species that formed lesions significantly larger than the control could be considered as olive trunk pathogens. These included Biscogniauxia rosacearum, Celerioriella umnquma, Coniochaeta velutina, Coniothyrium ferrarisianum, isolates of the Cytospora pruinosa complex, Didymocyrtis banksiae, Diaporthe foeniculina, Eutypa lata, Fomitiporella viticola, Neofusicoccum stellenboschiana, Neofusicoccum vitifusiforme, Neophaeomoniella niveniae, Phaeoacremonium africanum, Phaeoacremonium minimum, Phaeoacremonium oleae, Phaeoacremonium parasiticum, Phaeoacremonium prunicola, Phaeoacremonium scolyti, Phaeoacremonium spadicum, Pleurostoma richardsiae, Pseudophaeomoniella globosa, Punctularia atropurpurascens, Vredendaliella oleae, an undescribed Cytospora sp., Geosmithia sp., two undescribed Neofusicoccum spp., and four Xenocylindrosporium spp. Pseudophaeomoniella globosa can be regarded as one of the main olive trunk pathogens in South Africa because of its high incidence from olive trunk disease symptoms in established orchards and its high virulence in pathogenicity trials.

Bacteria of the genus Legionella are natural pathogens of amoebae that can cause a severe pneumonia in humans called Legionnaires' Disease. Human disease results from inhalation of Legionella-contaminated aerosols and subsequent bacterial replication within alveolar macrophages. Legionella pathogenicity in humans has resulted from extensive co-evolution with diverse genera of amoebae. To replicate intracellularly, Legionella generates a replication-permissive compartment called the Legionella-containing vacuole (LCV) through the concerted action of hundreds of Dot/Icm-translocated effector proteins. In this review, we present a collective overview of Legionella pathogenicity including infection mechanisms, secretion systems, and translocated effector function. We also discuss innate and adaptive immune responses to L. pneumophila, the implications of Legionella genome diversity and future avenues for the field.

Soybean is threatened by many pathogens that negatively affect this crop’s yield and quality, such as various Fusarium species that cause wilting and root rot diseases. Fusarium root rot (FRR) in soybean can be caused by F. graminearum and other Fusarium spp. that are associated with Fusarium head blight (FHB) in cereals. Therefore, it was important to inquire whether Fusarium pathogens from soybean can cause disease in wheat and vice versa. Here, we investigated the FRR complex in Manitoba (Canada) from symptomatic plants, using both culture- and molecular-based methods. We developed a molecular diagnostic toolkit to detect and differentiate between several Fusarium spp. involved in FHB and FRR, then we evaluated cross-pathogenicity of selected Fusarium isolates collected from soybean and wheat, and the results indicate that isolates recovered from one host can infect the other host. Trichothecene production by selected Fusarium spp. was also analyzed chemically via liquid chromatography mass spectrometry in both soybean (root) and wheat (spike) tissues. Trichothecenes were also analyzed in soybean seeds from plants with FRR to check the potentiality of trichothecene translocation from infected roots to the seeds. All of the tested Fusarium isolates were capable of producing trichothecenes in wheat spikes and soybean roots, but no trichothecenes were detected in soybean seeds. This study provided evidence, for the first time, that trichothecenes were produced by several Fusarium spp. (F. cerealis, F. culmorum, and F. sporotrichioides) during FRR development in soybean.

The aim of the present study was to evaluate the prevalence of serogroup A of Pasteurella multocida in Syrian Awassi sheep. Of 1630 samples collected from nasal swabs of healthy and pneumonic sheep (125 herds) and pneumonic sheep lungs, a total of 228 (13.9%) strains were isolated and identified as P. multocida subsp. multocida by phenotypic and biochemical characterisation. However, of them only 117 (51.3%) were identified as serogroup A of P. multocida when PCR assay with specific primers for serogroup A strains was applied. The highest rate of serogroup A isolation was from apparently healthy sheep (49.6%) with consideration that all lung isolates (23 isolates) belonged to serogroup A. Geographical and seasonal distribution showed that about 60% of positively isolated bacteria originated from Syrian desert (29 isolates) and central parts of semi-arid step zone (41 isolates). A significant increase (P≤0.05) in the rate of positive isolates was observed in winter as compared to spring. Pathogenicity tests of 10 isolates with 50 or 10 LD50 values showed that 5 isolates were able to induce symptoms of fowl cholera in challenge-exposed chickens indicating that migratory Awassi sheep might serve as a carrier for serogroup A of P. multocida and that ovine isolates may be virulent for local breed of chickens.

International audience; The invertebrate Galleria mellonella has increasingly and widely been used in the last few years to study complex host-microbe interactions. Aspergillus fumigatus is one of the most pathogenic fungi causing life-threatening diseases in humans and animals. Galleria mellonella larvae has been proven as a reliable model for the analysis of pathogenesis and virulence factors, enable to screen a large number of A. fumigatus strains. This review describes the different uses of G. mellonella to study A. fumigatus and provides a comparison of the different protocols to trace fungal pathogenicity. The review also includes a summary of the diverse mutants tested in G. mellonella, and their respective contribution to A. fumigatus virulence. Previous investigations indicated that G. mellonella should be considered as an interesting tool even though a mammalian model may be required to complete and verify initial data.

Severe Helicobacter pylori-linked gastric disorders are especially prevalent in the East Asia region. The ability of H. pylori to cause different clinical outcomes is thought to be associated with unique sets of its genetic features. However, only few genetic features have been definitively linked to specific gastrointestinal pathologies. Genome heterogeneity of clinical H. pylori strains from patients with four different gastric disorders was studied to explore the population structure and molecular genomic features and their association with pathogenicity. Population analysis showed that 92.9% of the Shanghai H. pylori isolates were clustered in the East Asia group. Among 2,866 genes detected in all genomes, 1,146 genes formed the core genome, whereas 209 unique genes were detected in individual disease groups. The unique genes of peptic ulcer and gastric cancer groups represented the inorganic ion transport and metabolism function gene clusters. Sixteen virulence genes were detected with statistically different detection rates among the four disease groups. Furthermore, 127 clustered regularly interspaced short palindromic repeats were found with significantly different rates in the four disease groups. A total of 337 putative genomic islands were identified, and three genomic islands were individually found in more than 10% of strains. The genomic islands included several metabolism-associated genes and many genes with unknown function. In total, 88 sequence types were detected among the 112 Shanghai H. pylori isolates. Our study provides an essential milestone in the mapping of specific genomic features and their functions to identify factors needed to induce specific gastric disorders in H. pylori.

The genus Rickettsia is the causative agent of several rickettsial diseases that are primarily transmitted by hard ticks. The occurrence of Rickettsia in chigger mites, which are vectors of scrub typhus in the western Pacific region, has been infrequently investigated. We identified Rickettsia spp. in chiggers collected from small mammals in six counties of Taiwan. Moreover, by capitalising on parallel Rickettsia detections on small mammals and their infested ticks and fleas, we were able to identify Rickettsia spp. that suggested more intimate associations with chigger mites. Rickettsia detection rates in 318 pools of chiggers were 21.7% and 22.3% when based on the ompB and gltA gene, respectively. Overall, we identified six (based on the ompB gene) and eight (gltA gene) Rickettsia species. Approximately half of the sequenced species were most similar to Rickettsia sp. clone MB74-1 (ompB gene) and Rickettsia sp. TwKM02 (gltA gene). Furthermore, both species were either infrequently or never identified in small mammals, ticks and fleas, which suggests that chigger mites might be the primary host of both rickettsiae. Whether both species are pathogenic to humans remains to be studied. They may also be microbial endosymbionts of chigger mites, with their potential effects on the pathogenicity of the aetiologic agent of scrub typhus deserving further investigations.

A Candida spp. possui elevada patogenicidade nos seres humanos imunocomprometidos, acometendo órgãos genitais, mucosa do trato respiratório e oral, entre outras partes do corpo, tornando-se cada vez mais resistente aos antifúngicos. Uma das opções para seu tratamento está relacionado ao uso de plantas medicinais as quais possuem ação antibacteriana e antifúngica. O presente estudo buscou avaliar a atividade antifúngica dos extratos in natura do Rosmarinus officinalis frente as cinco cepas de Candida spp. por meio de uma pesquisa experimental na qual foram testados dois extratos diferentes do alecrim in natura, sendo um do caule e outro das folhas da planta, frente as cepas de C. albicans, C. tropicalis, C. krusei, C. parapsilosis e C. glabrata. O alecrim apresentou atividade inibitória frente as cepas com variações de inibição relacionadas à diferentes concentrações, com inibição de crescimento em todas as concentrações dos extratos testados contra C. albicans e C. tropicalis. As cepas de C. krusei, C. parapsilosis e C. glabrata testadas demonstraram resultados distintos entre si relacionados a inibição do crescimento conforme as diluições propostas. Chilli (Capsicum annum L.) is a common spice crop grown in India. During 2018, twig blight symptoms were first observed in experimental plots of chilli in ICAR-IIHR-Central Horticultural Experimental Station, Odisha, India. High incidence of twig blight to the tune of 23% was documented during 2018 Kharif crop of chilli. There was no occurrence of twig blight in rabi and summer crops. This study reported the detailed cultural and micro-morphological characteristics of Choanephora cucurbitarum infecting chilli and pathogen identity was confirmed by molecular phylogenetic analyses. Pathogenicity tests were repeated twice with same results. To best of our knowledge, C. cucurbitarum infecting chilli is the first report in the state of Odisha, India.

Malaria is still one of the most prevalent tropical and parasitic diseases throughout the world representing a global health concern. Even with all efforts to restrict disease transmission and national malaria control programs, malaria infection continues to cause considerable morbidity and mortality in resource-poor countries. Malaria infection occurs in two stages, the Exoerythrocytic and Erythrocytic stage. The exact biology of malaria parasite in human hosts is likely relatively similar, with the main differences being attributable to the human immune response, the number of previous infections and the exposure profile. The disease severity can be determined by the balance between the pro-inflammatory and anti-inflammatory cytokines. Although, research about the clinical characterization and histology of malaria has shown some information regarding the pathogenesis, the actual mechanisms by which malaria parasites produce severe disease, the immunity defends against infection is remained unknown. Studies in animal models can reveal details about the processes of severe malaria infection and human defense mechanisms. Because of its similarities to the Plasmodium species that cause human malaria, Plasmodium berghei is used as a model organism for the experimental research. In addition, of affecting the central nervous system, Plasmodium berghei infection in the Swiss Webster mouse causes systemic damage, and affects numerous organs including the liver and lymphoid organs. The infected spleen demonstrations includethe splenomegaly, re-modelling and other basic changes consist of the red pulp’s expansion, marginal zone’s slight damage, enlarged vasculature and the barrier cells activities. Moreover, the liver shows hyperplastic Kupffer cells, fatty change, portal tract inflammation, cholestasis, liver cell necrosis, sequestration of Parasitized Red Blood Cells (PRBCs) and deposition of hemozoin pigments.

Autophagy is responsible for maintaining fundamental cellular homeostasis and is, therefore, essential for diverse development processes. This study reported that PoElp3, the putative catalytic subunit of Elongator complex, is involved in the maintenance of autophagy homeostasis to facilitate asexual development and pathogenicity in the rice blast fungus Pyricularia oryzae. It was found that the ΔPoelp3 strains were defective in vegetative growth, conidiation, stress response, and pathogenicity. The mutants exhibited hyper-activated autophagy in the vegetative hyphae under both nutrient-rich and nutrient-deficient conditions. The hyper-activation of autophagy possibly suppressed the production of vegetative hyphae in the ΔPoelp3 strains. Moreover, the ΔPoelp3 strains were found to be more sensitive to rapamycin during vegetative- and invasive-hyphal growth but have no effect on Target-of-Rapamycin (TOR) signaling inhibition. Taken together, these results demonstrated that PoElp3 is involved in asexual development and pathogenicity by regulating autophagy in the rice blast fungus.

White mold of pea caused by Sclerotinia sclerotiorum is a common disease in China. However, we discovered that the diverse Sclerotinia species could cause white mold on pea plants in Chongqing and Sichuan of China during recent disease surveys. Thus, the objective of this study was to confirm the causal agents from diseased pea plants. The obtained isolates of white mold from Chongqing and Sichuan were identified by morphological characters and molecular characterization to determine the pathogen species, and their pathogenicity was confirmed on pea through completing Koch's postulates. Fungal isolates of Sclerotinia-like were obtained from diseased plants or sclerotia. Based on morphological characteristics and molecular characterization, 30 isolates were identified to three species, six isolates as S. minor, seven as S. sclerotiorum, and 17 as S. trifoliorum. In pathogenicity tests on pea cultivars Zhongwan 4 and Longwan 1, all 30 isolates caused typical symptoms of white mold on the inoculated plants, and the inoculated pathogens were re-isolated from the diseased plants. This study confirmed that white mold of pea was caused by three Sclerotinia species, S. sclerotiorum, S. minor and S. trifoliorum in Chongqing and Sichuan. It is the first report that S. minor and S. trifoliorum cause white mold of pea in Southwest China.

Pestalotiopsis and related genera, including Neopestalotiopsis and Pseudopestalotiopsis have damaged many plants for many decades; however, there is little available information about the fungi on tropical fruit in Thailand. This study isolated and characterized pestalotioid fungi on tropical fruit, investigated host specificity, and screened whether plant extracts could control the fungi. In total, 53 diseased fruit samples were sampled from eight types of fruit trees (jackfruit, rose apple, mangosteen, plum, snake fruit, rambutan, strawberry, and avocado). Based on morphological characteristics, 44 isolates were classified as belonging to pestalotioid taxa. Of these isolates, seven with distinct characteristics were selected for identification using molecular analysis, and six isolates were identified as Neopestalotiopsis and one as Pseudopestalotiopsis. In the cross-inoculation experiment, the isolates exhibited nonhost specificity and could infect at least two host plants. The isolates were used to screen for a potential biocontrol resource using six crude plant extracts (clove, ginger, lemongrass, mangosteen, roselle, and turmeric). All crude extracts except mangosteen could inhibit the growth of Neopestalotiopsis and Pseudopestalotiopsis. Application of crude plant extracts could be a potential treatment to control these diseases on tropical fruit.

Background Pathogenicity of the entomopathogenic fungi (EPF), isolated from soil samples collected from Ordu Province, Turkey, was evaluated on the second-instar larvae of the cotton leaf worm Spodoptera littoralis (Boisd.) (Lepidoptera: Noctuidae) under laboratory conditions. Results Firstly, single-concentration response tests were conducted in order to determine the efficacy of the 64 isolates on S. littoralis larvae at the concentration of 1 × 108 conidia/ml. The five isolates displaying the highest mortality rates in single-concentration response tests, ORU-50, ORM-40, ORP-13, ORP-27 and ORM-48 (which included Beauveria bassiana, Metarhizium brunneum and Clonostachys rogersoniana), were subjected to concentration–response tests at the concentrations of 1 × 105–1 × 109 conidia/ml. The lowest LC50 and LC90 values were recorded at ORP-27 with 1.68 × 107 and 4.60 × 108 conidia/ml, respectively, followed by ORP-13 and ORM-40. Conclusions Accordingly, it was found that M. brunneum isolates were more effective than B. bassiana and C. rogersoniana against S. littoralis larvae. ORP-27, ORP-13 and ORM-40 of M. brunneum isolates can be a potential biological control agent used against S. littoralis larvae.

Ralstonia solanacearum RSc0454 is predicted as a FAD-linked oxidase based on protein homologies, while it contains distinct domains of lactate dehydrogenase and succinate dehydrogenase. A previous study demonstrated that RSc0454 exhibits lactate dehydrogenase activity using pyruvate and NADH as substrates, and is essential for pathogenicity of R. solanacearum. Here, we genetically characterized involvement of RSc0454 on bacterial growth and expression of genes for the type III secretion system (T3SS, a pathogenicity determinant) in R. solanacearum. The RSc0454 mutant grew normally in rich medium but grew faintly in host plants, and failed to grow in minimal medium. Supplementary succinate but not lactate could substantially restore some phenotypes of RSc0454 mutants, including faint growth in host plants, diminished growth in the minimal medium, and lost pathogenicity toward host plants. Expression of T3SS genes is directly controlled by a master regulator, HrpB, and hrpB expression is positively regulated by HrpG and PrhG in parallel ways. Deletion of RSc0454 substantially reduced expression levels of hrpB and T3SS both in vitro and in planta. Moreover, RSc0454 is revealed to be required for the T3SS expression via HrpG and PrhG, although through some novel pathway, and impaired expression of these genes was not due to growth deficiency of RSc0454 mutants. RSc0454 is suggested to be important for redox balance inside cells, and supplementary NADH partially restored diminished growth of the RSc0454 mutant in the minimal medium only in the presence of succinate at some moderate concentrations, indicating that the unbalanced redox in the RSc0454 mutant might be responsible for its diminished growth in the minimal medium. Taken together, these results provide novel insights into the understanding of various biological functions of this FAD-linked oxidase RSc0454 and involvement of the redox balance on expression of the T3SS in R. solanacearum. Ralstonia solanacearum species complex (RSSC) contains a group of destructive plant pathogenic bacteria, causing bacterial wilt of >200 species of crops and trees, such as Casuarina equisetifolia, worldwide. RSSC can survive in the soil environment for a long time and start infection after activation by host plants. This study conducted a transcriptome analysis on the expression pattern of the pathogenicity-related genes of a new isolated RSSC strain YQ (Ralstonia pseudosolanacearum phylotype I-16) in response to C. equisetifolia cladophyll (a branch of a stem that resembles and functions as a leaf) and root debris under in vitro culture. The cladophyll debris induced more genes up-regulated than the root debris, including pathogenicity-related genes involved in motility, effectors, type III secretion systems, quorum sensing, and plant cell wall degradation. Besides, many differentially expressed genes were related to transcriptional regulator such as cyclic dimeric guanosine monophosphate. Moreover, the cultures with cladophyll debris induced a faster wilting in bioassays, and the cell swimming was enhanced by cladophyll exudate. C. equisetifolia cladophylls could activate the expression of pathogenicity-related genes of strain YQ and accelerate infection. Our findings suggest that litterfall management in C. equisetifolia forests, or even other plantations, should receive attention to prevent the induction of bacterial wilt disease caused by RSSC.

Stenotrophomonas maltophilia is ubiquitous in diverse environmental habitats. It merits significant concern because of its increasing incidence of nosocomial and community-acquired infection in immunocompromised patients and multiple drug resistance. It is rarely reported as a phytopathogen except in causing white stripe disease of rice in India and postharvest fruit rot of Lanzhou lily. For this study, Dickeya zeae and S. maltophilia strains were simultaneously isolated from soft rot leaves of Clivia miniata in Guangzhou, China, and were both demonstrated to be pathogenic to the host. Compared with the D. zeae strains, S. maltophilia strains propagated faster for greater growth in lysogeny broth medium and produced no cellulases or polygalacturonases, but did produce more proteases and fewer extracellular polysaccharides. Furthermore, S. maltophilia strains swam and swarmed dramatically less on semisolid media, but formed a great many more biofilms. Both D. zeae and S. maltophilia strains isolated from clivia caused rot symptoms on other monocot hosts, but not on dicots. Similar to previously reported S. maltophilia strains isolated from other sources, the strain JZL8 survived under many antibiotic stresses. The complete genome sequence of S. maltophilia strain JZL8 consists of a chromosome of 4,635,432 bp without a plasmid. Pan-genome analysis of JZL8 and 180 other S. maltophilia strains identified 50 genes that are unique to JZL8, seven of which implicate JZL8 as the potential pathogen contributor in plants. JZL8 also contains three copies of Type I Secretion System machinery; this is likely responsible for its greater production of proteases. Findings from this study extend our knowledge on the host range of S. maltophilia and provide insight into the phenotypic and genetic features underlying the plant pathogenicity of JZL8.

To identify apple canker casual agents and evaluate their pathogenicity and virulence in apple production hubs including West Azarbaijan, Isfahan and Tehran provinces; samples were collected from symptomatic apple trees. Pathogenic isolates on the detached branches were identified as Cytospora cincta, Diplodia bulgarica, Neoscytalidium dimidiatum and Eutypa cf. lata. E. cf. lata was reported as a potential apple canker causal agent in Iran for the first time based on the pathogenicity test on the detached branches, whereas it caused no canker symptoms in apple trees until 6 months after inoculation. Currently, E. cf. lata seems to be adapted to a single city. C. cincta, D. bulgarica and N. dimidiatum caused canker symptoms in apple trees. “C. cincta” and also “C. cincta and N. dimidiatum” were the most widespread and aggressive apple canker species, respectively, associated with apple canker in Iran. Therefore, they are considered to be the main threat to apple production in Iran and should be carefully monitored. Disease progress curve, area under the disease progress curve and optimum temperatures were determined for mentioned species. It is concluded that the establishment of each species occurs in appropriate areas and times in terms of the optimum temperature for their growth.

Macracanthorhynchus hirudinaceus is a zoonotic parasite affecting suids worldwide which are the definitive hosts for this helminth species. Macracanthorhynchus hirudinaceus is of significant economic and management concern due to its pathogenicity, causing intestinal obstruction and perforation in the definitive hosts. Current study is the preliminary investigation from Sardinia, Italy, reporting the pathomorphological findings and molecular characterization of M. hirudinaceus in the wild boars (Sus scrofa meridionalis). A total of 59 wild boars were examined showing acanthocephalan infection in 8 (13.6%) animals. In total, 49 parasites were collected with a mean intensity of 6.1. Comparatively higher infection levels were observed for males (16.7%) and young boars (14.3%); however, these epidemiological differences were statistically non-significant. Histopathological examination revealed the presence of a variable number of nodules (∼5 mm) in the intestine of M. hirudinaceus infested animals surrounded by a hyperemic-hemorrhagic halo. Several parasites were recovered from the intestinal lumen attached by the means of characteristic hooks showing necrosis in muscle layers. A moderate number of plump reactive fibroblasts and lesser numbers of fibrocytes were embedded with and at the borders of the inflammatory nodules in a moderate amount of homogeneous intensely eosinophilic fibrillary material rupturing the cell membrane. For molecular characterization, six isolated worms were amplified for the partial mitochondrial cox1 gene showing distinct interindividual variations. This first pathological and molecular description from southern Europe provided new knowledge about the diffusion of M. hirudinaceus in wild boars, furthering the research into the origin and transmission status of M. hirudinaceus in endemic localities.

Background The peach fruit fly, Bactrocera zonata (Saunders) (Diptera: Tephritidae), is a serious polyphagous pest of fruits and vegetables. Chemical management of B. zonata in fruits results in toxic residues that have adverse health effects on consumers resulting in increased demand for eco-friendly approaches. Laboratory bioassay was conducted to determine the pathogenicity of fungal and bacterial biopesticides against B. zonata. Results The pathogenicity of all tested bioinsecticides revealed that the maximum concentration (1 × 108 CFU ml−1) of Metarhizium anisopliae, Beauveria bassiana, Lecanicillium lecanii and Bacillus thuringiensis caused 95.8–100%, 95.9–97.9%, 32.7–39.6%, and 20.0–22.4% mortality in B. zonata, respectively at 7 d post-application interval (PAI) as compared to mortality demonstrated by the same concentration at 5 d PAI. All tested microbial insecticides induced statistically similar mortality in both male and female B. zonata at each concentration for the same PAI. Correlation coefficient (r) values reveal that concentrations of each microbial insecticide had a high positive correlation with mortalities of male and female B. zonata. Regression parameters reveal that concentrations of tested microbial insecticides had significant linear relationship with and explained significant variability in B. zonata mortality (P M. anisopliae was proved more pathogenic to males and females of B. zonata demonstrating the least LC50 values (5.48 × 103, and 6.17 × 103 CFU ml−1, respectively) 7 d post-application intervals, followed by B. bassiana which explained LC50 value of 1.14 × 104 CFU and 1.15 × 105 CFU ml−1 for B. zonata males and females, respectively, at the same period of application, but less than that of L. lecanii (2.77 × 109 and 1.43 × 109 CFU ml−1) and then B. thuringiensis (3.40 × 1010 and 1.39 × 1010 CFU ml−1) for the males and females, respectively, at 7 d PAIs. Conclusion Metarhizium anisopliae incorporated adult diet was proved more effective against B. zonata, followed by B. bassiana, L. lecanii, and B. thuringiensis. Hence, M. anisopliae can be recommended for incorporating in bait-traps to develop attract-and-kill technology for B. zonata.

Henipaviruses, including Nipah virus, are regarded as pathogens of notable epidemic potential because of their high pathogenicity and the paucity of specific medical countermeasures to control infections in humans. We review the evidence of medical countermeasures against henipaviruses and project their cost in a post-COVID-19 era. Given the sporadic and unpredictable nature of henipavirus outbreaks, innovative strategies will be needed to circumvent the infeasibility of traditional phase 3 clinical trial regulatory pathways. Stronger partnerships with scientific institutions and regulatory authorities in low-income and middle-income countries can inform coordination of appropriate investments and development of strategies and normative guidelines for the deployment and equitable use of multiple medical countermeasures. Accessible measures should include global, regional, and endemic in-country stockpiles of reasonably priced small molecules, monoclonal antibodies, and vaccines as part of a combined collection of products that could help to control henipavirus outbreaks and prevent future pandemics.

Sclerotinia sclerotiorum, the notorious necrotrophic phytopathogenic fungus with wide distribution, is responsible for sclerotium disease in more than 600 plant species, including many economic crops such as soybean, oilseed rape, and sunflower. The compound appressorium is a crucial multicellular infection structure that is a prerequisite for infecting healthy tissues. Previously, the Forkhead‐box family transcription factors (FOX TFs) SsFoxE2 and SsFKH1 were shown to play a key regulatory role in the hyphae growth, sexual reproduction, and pathogenicity of S. sclerotiorum. However, little is known about the roles of SsFoxE3 regulating growth and development and pathogenicity. Here, we report SsFoxE3 contributes to sclerotium formation and deletion of SsFoxE3 leads to reduced formation of compound appressoria and developmental delays. Transcripts of SsFoxE3 were greatly increased during the initial stage of infection and SsFoxE3 deficiency reduced virulence on the host, while stabbing inoculation could partially restore pathogenicity. The SsFoxE3 mutant showed sensitivity to H2O2, and the expression of reactive oxygen species detoxification and autophagy‐related genes were reduced. Moreover, expression of SsAtg8 was also decreased during the infection process of the SsFoxE3 mutant. Yeast 1‐hybrid tests suggested that SsFoxE3 interacted with the promoter of SsAtg8. Disruption of SsAtg8 resulted in a phenotype similar to that of the SsFoxE3 mutant. Comparative analysis of the level of autophagy in the wild type and SsFoxE3 mutant showed that N starvation‐induced autophagy was reduced in the SsFoxE3 mutant. Taken together, our findings indicate that SsFoxE3 plays an important role in compound appressorium formation and is involved in transcriptional activation of SsAtg8 during infection by S. sclerotiorum., Transcription factor SsFoxE3 plays an important role in compound appressorium formation and is involved in transcriptional activation of SsAtg8 during infection by Sclerotinia sclerotiorum.

Purpose of review Here, we review recent progress made on the genetic characterization of Giardia duodenalis assemblages and their relationship with virulence. We also discuss the implications of virulence factors in the pathogenesis of giardiasis, and advances in the development of vaccines and drugs based on knowledge of virulence markers. Recent findings The use of transcriptomic and proteomic technologies as well as whole genome sequencing (WGS) from single cysts has allowed the assembly of the draft genome sequences for assemblages C and D of G. duodenalis. These findings, along with the published genomes for assemblages A, B, and E, have allowed comparative genomic investigations. In addition, the use of these methodologies for the characterization of the secretomes of trophozoite-epithelial cell interactions for assemblages A/B has led to the identification of virulence markers including energy metabolism enzymes, proteinases, high-cysteine membrane proteins (HCMPs), and variant surface proteins (VSPs). Recently, some drugs and vaccines, targeting virulence factors have been developed, offering possible alternatives to current treatment and prevention options against giardiasis. Summary Among the nine recognized species of Giardia, G. duodenalis stands out because of its broad spectrum of hosts and its socio-economic importance. This species comprises eight genetic assemblages (A to H), of which A and B are zoonotic, and the other assemblages have narrow host specificities. Assemblages A and B may be considered as the most virulent ones, but the existence of asymptomatic carriers and considerable genetic variability within and among these assemblages hampers the definition of common virulence factors. The attachment of Giardia trophozoites to epithelial cells and structural cytoskeleton components of the adhesive disk, such as giardins or tubulins, is proposed to play key roles, but toxins have not yet been precisely defined. However, recent transcriptomic and proteomic analyses of the secretomes of trophozoites representing assemblages A and B and interacting with particular epithelial cell lines have defined a series of virulence factors, including glycolytic (e.g., enolase) and arginolytic (e.g., arginine deiminase) enzymes, cysteine proteases (e.g., giardipain-1) and VSPs (e.g., VSP9B10A). Other factors, such as HCMPs and tenascins, have been consistently found to be excreted/secreted, but their role(s) in the pathogenesis of giardiasis has not yet been elucidated. Interestingly, recent investigations of single cysts representing assemblages C and D using advanced sequencing and informatic methods have suggested that the transcription/expression profiles of virulence factors vary both within and between assemblages, thus assemblage-specific molecules might allow adaptation to the microenvironment within the host. Importantly, some drugs active against cysteine-rich proteins of Giardia, including giardipain-1, VSPs and arginine deiminase, have been shown to be targeted by cysteine-modifying compounds as disulfiram, L-canavanin and allicin. On the other hand, VSPs are presently considered as key vaccine candidates because they induce protection against Giardia in rodents and dogs. Overall, this review reveals that much more work is needed to identify, characterize, and understand the roles of virulence factors in Giardia and to assess their validity as drug and vaccine targets. Clear, advanced omics and informatic tools should assist in this future endeavor, with a focus on targeting virulence factors that are common and/or unique to distinct assemblages to develop new and effective interventions against Giardia.

The fungal pathogen Setosphaeria turcica causes leaf blight on maize, which leads to considerable crop losses. However, how S. turcica establishes sustained systemic infection is largely unknown. Here, we report several novel factors contributing to S. turcica pathogenicity, identified using a genomic and transcriptional screen at different stages of S. turcica appressorium development. We identified two cytoskeleton regulators, SLM1 and SLM2, that are crucial for hypha and appressorium development. The SLM1 and SLM2 transcripts accumulated during germling stage but their levels were notably reduced at the appressorium stage. Deletion of SLM2 dramatically affected cell morphology, penetration ability, and pathogenicity. We also identified three different types of S. turcica glycosyl hydrolases that are critical for plant cell wall degradation. Their transcripts accumulated during the appressorium infection stage induced by cellophane and maize leaf. Most importantly, we characterized a novel and specific S. turcica effector, appressorium‐coupled effector 1 (StACE1), whose expression is coupled to appressorium formation in S. turcica. This protein is required for maize infection and induces cell death on expression in Nicotiana benthamiana. These observations suggest that the phytopathogen S. turcica is primed in advance with multiple strategies for maize infection, which are coupled to appressorium formation at the early infection stages., The phytopathogen Setosphaeria turcica is primed in advance with multiple strategies for maize infection, which are coupled to appressorium formation at the early infection stages.

Listeria monocytogenes is a saprophytic gram-positive bacterium, and an opportunistic foodborne pathogen that can produce listeriosis in humans and animals. It has evolved an exceptional ability to adapt to stress conditions encountered in different environments, resulting in a ubiquitous distribution. Because some food preservation methods and disinfection protocols in food-processing environments cannot efficiently prevent contaminations, L. monocytogenes constitutes a threat to human health and a challenge to food safety. In the host, Listeria colonizes the gastrointestinal tract, crosses the intestinal barrier, and disseminates through the blood to target organs. In immunocompromised individuals, the elderly, and pregnant women, the pathogen can cross the blood-brain and placental barriers, leading to neurolisteriosis and materno-fetal listeriosis. Molecular and cell biology studies of infection have proven L. monocytogenes to be a versatile pathogen that deploys unique strategies to invade different cell types, survive and move inside the eukaryotic host cell, and spread from cell to cell. Here, we present the multifaceted Listeria life cycle from a comprehensive perspective. We discuss genetic features of pathogenic Listeria species, analyze factors involved in food contamination, and review bacterial strategies to tolerate stresses encountered both during food processing and along the host’s gastrointestinal tract. Then we dissect host–pathogen interactions underlying listerial pathogenesis in mammals from a cell biology and systemic point of view. Finally, we summarize the epidemiology, pathophysiology, and clinical features of listeriosis in humans and animals. A member of the family Rhizinaceae, in the class Pezizomycetes, P. omnivora is a soilborne ascomycete fungus that is difficult to maintain in culture, currently genetically intractable, and for which there are no publicly available genomic resources. We have generated draft genome sequences of four P. omnivora isolates obtained from cotton and alfalfa, growing in Texas and Oklahoma. These genome sequences will provide new insights into the biology of the fungus, including the factors responsible for its broad host range and pathogenicity.

Pseudomonas cichorii secretes effectors that suppress defense mechanisms in host plants. However, the function of these effectors, including avirulence protein E1 (AvrE1), in the pathogenicity of P. cichorii, remains unexplored. In this study, to investigate the function of avrE1 in P. cichorii JBC1 (PcJBC1), we created an avrE1-deficient mutant (JBC1ΔavrE1) using CRISPR/Cas9. The disease severity caused by JBC1ΔavrE1 in tomato plants significantly decreased by reducing water soaking during early infection stage, as evidenced by the electrolyte leakage in infected leaves. The disease symptoms caused by JBC1ΔavrE1 in the cabbage midrib were light-brown spots compared to the dark-colored ones caused by PcJBC1, which indicates the role of AvrE1 in cell lysis. The avrE1-deficient mutant failed to elicit cell death in non-host tobacco plants. Disease severity and cell death caused by JBC1ΔavrE1 in host and non-host plants were restored through heterologous complementation with avrE1 from Pseudomonas syringae pv. tomato DC3000 (PstDC3000). Overall, our results indicate that avrE1 contributes to cell death during early infection, which consequently increases disease development in host plants. The roles of PcJBC1 AvrE1 in host cells remain to be elucidated.

Several weed species are known as alternative hosts of the Fusarium graminearum species complex (FGSC), and their epidemiological significance in Fusarium head blight (FHB) has been investigated; however, scant information is available regarding FGSC occurrence in weeds near Chinese wheat fields. To evaluate the potential role of gramineous weeds surrounding wheat fields in FHB, 306 FGSC isolates were obtained from 210 gramineous weed samples in 2018 in Jiangsu Province. Among them, 289 were Fusarium asiaticum, and the remainder were F. graminearum. Trichothecene genotype and mycotoxin analyses revealed that 74.3% of the F. asiaticum isolates were the 3-acetyldeoxynivalenol (3ADON) chemotype, and the remainder were the nivalenol (NIV) chemotype. Additionally, 82.4% of F. graminearum isolates were the 15-acetyldeoxynivalenol (15ADON) chemotype, and the remainder were the NIV chemotype. FHB severity and trichothecene analysis indicated that F. asiaticum isolates with the 3ADON chemotype were more aggressive than those with the NIV chemotype in wheat. 3ADON and NIV chemotypes of F. asiaticum isolated from weeds and wheat showed no significant differences in pathogenicity in wheat. All selected F. asiaticum isolates produced perithecia, with little difference between the 3ADON and NIV chemotypes. These results highlight the epidemiology of the FGSC isolated from weeds near wheat fields, with implications for reducing FHB inoculum in China.
