Population growth and rapid urbanisation have increased the global demand for animal feed and protein sources. Therefore, traditional animal feed production should be increased through the use of alternative nutrient sources. Insects as feed are beginning to fill this need. One such insect is the black soldier fly, Hermetia illucens L. (Diptera: Stratiomyidae). However, to more effectively mass produce the black soldier fly, a better understanding of its thermal biology is needed. Thus, the aim of this study was to evaluate the impact of age, size, and sex on adult black soldier fly thermal preference. The thermal preference of adult black soldier flies was determined by exposing flies to a thermal gradient with a range of surface temperatures and monitoring their positions over time. An aluminium plate was used to create a linear thermal gradient where surface temperatures ranged from ~15-60 °C. Flies were distinguished by age (1-d-post-emergence vs 7-d-post-emergence), size (large vs small) and sex (male vs female) to assess whether thermal preference differed by specific life-history traits. Thermal preference for 7-d-post-emergence adults was significantly lower (19.2 °C) than 1-d-post-emergence adults (28.7 °C), respectively. Similarly, small adults selected significantly cooler (21.1 °C) temperatures than large adults (26.9 °C). No significant differences in thermal preferences were found between sex, regardless of age or size. In fact, males and females had similar thermal preference of 23.8 and 24.2 °C, respectively. This study reveals that multiple life-history traits of adult black soldier fly affect their thermal preference, and thus should be taken into consideration by mass rearing facilities to optimize production.

Pre-emergence (PRE) herbicides are commonly applied simultaneously with fertilizers to turfgrass; however, the influence of PRE herbicides on nitrogen (N) uptake and leaching from turfgrass remains unclear. The hypothesis of this study was that PRE herbicides applied simultaneously with N fertilizers increase N leaching from Tifway 419 bermudagrass [Cynodon dactylon (L.) Pers. × C. transvaalensis Burtt-Davy, 'Tifway'] above that from fallow soil. A nutrient leaching study was conducted from June 2017 through June 2019 in Fort Lauderdale, FL. Treatments consisted of indaziflam (25 g a.i. ha-1 ), prodiamine (540 g a.i. ha-1 ), and oxadiazon (4,480 g a.i. ha-1 ); a nontreated turfgrass control (turfgrass fertilized but not treated with PRE herbicides); and a fallow soil. Fertilizer (15-2-12) was applied every 60 d at 49 kg N ha-1 , and PRE herbicides were applied every 120 d. Pre-emergence herbicides resulted in a 3.6- and 5.5-fold increases in NO3 -N concentration compared with fallow soil during June 2017 and January 2018, respectively, whereas fallow soil resulted in increased NO3 -N concentration during 10 mo and ranged from 3.8- to 15-fold greater than that from turfgrass plots. Turfgrass plots resulted in reduced N leaching of ∼7% during 5 mo compared with fallow soil and did not result in increased N leaching during any month. Cumulative N leached from turfgrass plots ranged from 75 to 120 kg ha-1 and did not differ from fallow soil. Turfgrass growth rate and N uptake were not influenced by PRE herbicide. The results indicated that fertilizers applied with PRE herbicides does not result in increased N leaching or reduced N uptake.

Fungicides are routinely applied to golf course putting greens throughout the growing season. Gaining a better understanding of fungicide fate can improve fungicide use and stewardship. Therefore, optimizing fungicide applications with post-application management practices may enhance fungicide movement and limit potential off-target effects. Two field studies were initiated on a golf course putting green to evaluate the influence of post-fungicide application irrigation and mowing timing on fungicide movement into the soil profile and removal in turfgrass clippings. Plots were treated with a single application of either pyraclostrobin, triadimefon, or penthiopyrad and received 0.64 cm post-application irrigation immediately or 6 h after application or received no post-application irrigation. Clippings were collected 0, 1, and 3 d after treatment (DAT). Cores were harvested 0, 1, 3, 5, 7, and 14 DAT and dissected into the remaining aboveground vegetation (RAV; verdure/thatch; 0-to-2.5-, 2.5-to-5.1-, and 5.1-to-7.6-cm soil subsections). A small amount of fungicide (3.6%) was removed with clippings regardless of mowing and irrigation treatment. Post-application irrigation treatment influenced fungicide movement; however, 50% of fungicide remained restricted to the RAV for the first 3 DAT. Less fungicide remained restricted to the RAV, and more fungicide was detected in deeper soil depths when plots were irrigated immediately after application. Fungicide was only detected at the 5.1-to-7.6-cm depth when plots were irrigated immediately. Applying post-application irrigation immediately may result in more fungicide moving down to soilborne targets. Irrigating 6 h after application facilitated moderate fungicide movement compared with irrigating immediately but was better than no post-application irrigation.

Additional active ingredients are needed for use in aquatic systems to respond to new threats or treatment scenarios, enhance selectivity, reduce use rates, and mitigate the risk of herbicide resistance. Florpyrauxifen-benzyl is a new synthetic auxin developed for use as an aquatic herbicide. A study was conducted at North Carolina State University in which 10 µg L−1 of 25% radiolabeled florpyrauxifen-benzyl was applied to the isolated shoot tissue of 10 different aquatic plant species to elucidate absorption and translocation patterns in these species. Extremely high levels of shoot absorption were observed for all species, and uptake was rapid. Highest shoot absorptions were observed for crested floatingheart [Nymphoides cristata (Roxb.) Kuntze] (A192 = 20 µg g−1), dioecious hydrilla [Hydrilla verticillata (L. f.) Royle] (A192 = 25.3 µg g−1), variable watermilfoil (Myriophyllum heterophyllum Michx.) (A192 = 40.1 µg g−1), and Eurasian watermilfoil (Myriophyllum spicatum L.) (A192 = 25.3 µg g−1). Evidence of translocation was observed in all rooted species tested, with the greatest translocation observed in N. cristata (1.28 µg g−1 at 192 h after treatment). The results of this study add to the growing body of knowledge surrounding the behavior of this newly registered herbicide within aquatic plants.

A study of the mammals of the Gila region of New Mexico was conducted from 2012 through 2020, with 2,919 voucher specimens collected through fieldwork and collaborations with commercial trappers, in addition to data from camera traps, review of major holdings at 46 museums ( n = 12,505 georeferenced specimens), and literature review. Specimens cover a 170-year span, dating back to 1850 and were unevenly distributed spatially and temporally across the Gila region. Most areas were very poorly represented and when summed across all mammal species, ranged from 0.02 to 3.7 specimens per km 2 . The survey documented 108 species (104 now extant) for the region. High species richness, greater than that reported for 38 states in the United States, is likely due to the juxtaposition of multiple biomes in the Gila, including the Sonoran, Chihuahuan, and Great Basin deserts, the Rocky Mountains and Sierra Madre Occidental, and nearby “sky islands’’ of the Southwest. Two species, Leptonycteris yerbabuenae and Zapus luteus, are documented for the first time from the study area. Expansions of the known range of these species , and Sciurus arizonensis are described from specimen and camera data. Preliminary phylogeographic studies of four species ( Notiosorex crawfordi, Neotoma albigula, Perognathus flavus, and Thomomys bottae ) using the mitochondrial cytochrome- b gene reveal the dynamic biogeographic history of the region and reinforce how landscape complexity and climate change have jointly contributed to diversification and thus high mammalian diversity in the region.

The present study evaluated whether in vitro measures of aromatase inhibition as inputs into a quantitative adverse outcome pathway (qAOP) construct could effectively predict in vivo effects on 17β-estradiol (E2) and vitellogenin (VTG) concentrations in female fathead minnows. Five chemicals identified as aromatase inhibitors in mammalian-based ToxCast assays were screened for their ability to inhibit fathead minnow aromatase in vitro. Female fathead minnows were then exposed to 3 of those chemicals: letrozole, epoxiconazole, and imazalil in concentration-response (5 concentrations plus control) for 24 h. Consistent with AOP-based expectations, all 3 chemicals caused significant reductions in plasma E2 and hepatic VTG transcription. Characteristic compensatory upregulation of aromatase and follicle-stimulating hormone receptor (fshr) transcripts in the ovary were observed for letrozole but not for the other 2 compounds. Considering the overall patterns of concentration-response and temporal concordance among endpoints, data from the in vivo experiments strengthen confidence in the qualitative relationships outlined by the AOP. Quantitatively, the qAOP model provided predictions that fell within the standard error of measured data for letrozole but not for imazalil and epoxiconazole. However, the inclusion of measured plasma concentrations of the test chemicals as inputs improved model predictions, with all predictions falling within the range of measured values. Seed germination is one of the most important processes in plant biology and ecology because it determines the timing and magnitude of seedling emergence events every growing season influencing community dynamics. Our aim was to determine whether polyethylene glycol (PEG) solutions simulate soil water potential accurately and recreate germination responses to soil water availability. In this study, we compared seed germination of four plant species in PEG and four soils with different textures under six water potentials under controlled laboratory conditions. Total seed germination for all species significantly differed between soil and PEG under the same water potentials, as well as among soil water potentials for each of PEG and soil materials. Due to the inconsistent total germination associated with soil water potential, we evaluated unsaturated soil hydraulic conductivity (Kh) as a predictor of germination. The germination of all species followed the same response to Kh. Germination rate (GR50) was more directly related to water potential than total germination, but Kh provided a more robust description of GR50 across species and soils than PEG-osmotic potentials. Our findings showed that Kh is a more informative variable to predict both total seed germination and germination rate in soil, and caution must be used when considering results obtained using PEG solutions to infer germination behavior under field conditions.

Information regarding black soldier fly (Diptera: Stratiomyidae) adult biology is vital as this is the life stage that produces eggs and thus drives population size. The goal of this study was to determine key biological characteristics of adult black soldier flies as they age in relation to: (1) the thermal preferences (Tsel) of males and females; (2) the impact of temperature on heat shock protein expression in males and females; as well as (3) the sperm count; and (4) the sperm viability in males. Cotton (Gossypium hirsutum L.) production in Oklahoma has more than tripled in the last ten years. This increase in cotton acreage in Oklahoma, a region that traditionally produces winter wheat, has led to increased incidences of 2,4 D contamination in sprayers that are used to spray cotton crops. Cotton is extremely sensitive to 2,4 D, with losses of yield and ultimately profit to the cotton producer in cases of drift or tank carryover. In this study, six cotton cultivars (three 2,4 D susceptible and three 2,4 D tolerant) were grown in combination with four N treatments to determine the benefits or drawbacks of using a green manure as N source. Seedcotton, seed, and lint yield were all negatively impacted by 2,4 D in the susceptible cultivars. Additionally, water use was increased in the susceptible cultivars compared to the tolerant cultivars. The negative effects of 2,4 D on cotton growth were partially mitigated by grasspea, though the supply of too much nitrogen increased the negative effects of 2,4 D. Applying nitrogen to a susceptible cultivar of cotton contaminated by 2,4 D is not recommended for short season environments like Oklahoma.

The Xenarthra are one of the least studied mammalian groups in Honduras. Their ecology, natural history, and distribution in the country are poorly known. Here we compile and discuss, for the first time, the available information about the seven species of Xenarthra that occur in Honduras. We also present new distribution records and comment on their main threats. The lack of specific scientific studies, poaching, illegal traffic, cultural beliefs, and deforestation are among the main threats to xenarthrans in Honduras. Xenarthra es uno de los grupos de mamíferos menos estudiados en Honduras, y de los que menos se sabe sobre su ecología, historia natural y distribución en el país. Aquí recopilamos y discutimos, por primera vez, la información disponible de cada una de las siete especies de Xenarthra que ocurren en Honduras. También presentamos nuevos registros de distribución y comentamos sobre las principales amenazas. La falta de estudios científicos, la cacería y el tráfico ilegal, creencias culturales y la deforestación están entre algunas de las principales amenazas para los xenartros en Honduras. A growing body of research suggests female- and male-headed households in low- and middle-income countries differ in terms of crop choices, access to resources for growing different crops, and values placed on crops for home consumption versus market sale. To better understand relationships between gender of the household head, household resources, individual values, and crop choices, we draw on original survey data collected from 1,001 rural households in Kenya, Tanzania, and Uganda. Bivariate and multivariate analyses suggest that female-headed households are less likely to grow cash crops, reflecting a combina­tion of resource constraints and social norms. However, on average, female-headed households plant more diverse food crops per hectare of land to which they have access, consistent with past findings suggesting crop diversity is a strategy em­ployed by resource-constrained female-headed households to meet household food security needs. We also find that women surveyed on behalf of their households place a higher value on crops for food security, while men more frequently empha­size income potential. These results provide novel cross-country evidence on how female- and male-headed households, and women and men farmers within households, may prefer different crops and also face different levels of access to resources needed for market-oriented agriculture. Such findings support recent calls for development practitioners to carefully consider how market-oriented programs and policies may differentially affect female- and male-headed households and individuals residing within them. We also under­score the importance of collecting gender-disaggre­gated data to capture meaningful differences in preferences and constraints across women and men at the inter- and intra-household level. In recent years, there has been increased use of dicamba due to the introduction of dicamba-resistant cotton and soybean in the United States. Therefore, there is a potential increase in off-target movement of dicamba and injury to sensitive crops. Flue-cured tobacco is extremely sensitive to auxin herbicides, particularly dicamba. In addition to yield loss, residue from drift or equipment contamination can have severe repercussions for the marketability of the crop. Studies were conducted in 2016, 2017, and 2018 in North Carolina to evaluate spray-tank cleanout efficiency of dicamba using various cleaning procedures. No difference in dicamba recovery was observed regardless of dicamba formulation and cleaning agent. Dicamba residue decreased with the number of rinses. There was no difference in dicamba residue recovered from the third rinse compared with residue from the tank after being refilled for subsequent tank use. Recovery ranged from 2% to 19% of the original concentration rate among the three rinses. Field studies were also conducted in 2018 to evaluate flue-cured tobacco response to reduced rates of dicamba ranging, from 1/5 to 1/10,000 of a labeled rate. Injury and yield reductions varied by environment and application timing. When exposed to 1/500 of a labeled rate at 7 and 11 wk after transplanting, tobacco injury ranged from 39% to 53% and 10% to 16% 24 days after application, respectively. The maximum yield reduction was 62%, with a 55% reduction in value when exposed to 112 g ha–1 of dicamba. Correlations showed significant relationships between crop injury assessment and yield and value reductions, with Pearson values ranging from 0.24 to 0.63. As a result of genetic selection, the modern broiler is more efficient, higher yielding, and faster growing than the bird of the 1950s. Unfortunately, as a result of improvement in growth rate, the modern broiler has the potential to struggle under heat stress conditions. The present study evaluates 3 different random bred populations and a common ancestor under both a thermal neutral and heat stress conditions after a 54-D grow-out period. The lines used in this study included the Athens Canadian Random Bred (ACRB), a 1995 Random Bred (95RAN), a 2015 Random Bred (MRB), and a Junglefowl (JF). Male chicks (n = 150/line) were placed by line in environmentally controlled chambers. An 8-h daily cyclic heat stress (36°C) was applied to half of the chambers beginning on day 28 (HS) and lasting until processing at day 55, while the remaining chambers remained thermal neutral (TN) at 26°C. Dock weights and carcass weights were lower in the HS-95RAN and HS-MRB, compared to their TN counterparts, while the ACRB and JF had no difference in dock and carcass weights regardless of environmental condition. The MRB line had the highest breast yield (27.79%) while the JF (12.79%) and ACRB (12.42%) had the lowest. The 95RAN line had the highest abdominal fat percentage (2.83%) while the MRB line had the lowest moisture uptake during chill. The HS exposure lowered overall breast yield and breast pH at 15 min and 4 h postmortem but did not have an impact on color (L∗) or 24 h breast drip loss. The MRB was scored for both woody breast and white striping. The TN-MRB group had a higher incidence of moderate and severe woody breast and white striping than the HS-MRB group. Based on the results of this study, it appears that HS has a greater negative impact on the higher yielding lines (MRB and 95RAN) than the ACRB and JF and that clear line differences exist between the random bred lines and their common ancestor.

Predictive approaches to assessing the toxicity of contaminant mixtures have been largely limited to chemicals that exert effects through the same biological molecular initiating event. However, by understanding specific pathways through which chemicals exert effects, it may be possible to identify shared "downstream" nodes as the basis for forecasting interactive effects of chemicals with different molecular initiating events. Adverse outcome pathway (AOP) networks conceptually support this type of analysis. We assessed the utility of a simple AOP network for predicting the effects of mixtures of an aromatase inhibitor (fadrozole) and an androgen receptor agonist (17β-trenbolone) on aspects of reproductive endocrine function in female fathead minnows. The fish were exposed to multiple concentrations of fadrozole and 17β-trenbolone individually or in combination for 48 or 96 h. Effects on 2 shared nodes in the AOP network, plasma 17β-estradiol (E2) concentration and vitellogenin (VTG) production (measured as hepatic vtg transcripts) responded as anticipated to fadrozole alone but were minimally impacted by 17β-trenbolone alone. Overall, there were indications that 17β-trenbolone enhanced decreases in E2 and vtg in fadrozole-exposed fish, as anticipated, but the results often were not statistically significant. Failure to consistently observe hypothesized interactions between fadrozole and 17β-trenbolone could be due to several factors, including lack of impact of 17β-trenbolone, inherent biological variability in the endpoints assessed, and/or an incomplete understanding of interactions (including feedback) between different pathways within the hypothalamic-pituitary-gonadal axis. Sorgoleone-358 is an important allelochemical of the oily droplets exuded from root hairs of various species in the Sorghum genus. Due to its hydrophobic nature, sorgoleone-358 can be strongly adsorbed onto soil organic matter, resulting in increased sorgoleone soil persistence. Because of the herbicidal activity of sorgoleone on many small-seeded weeds, concerns have been raised that sorghum residues may have a detrimental effect on emergence of wheat used as a double crop in the southeastern United States. Laboratory experiments were conducted to evaluate root exudate production and its sorgoleone-358 content for 36 cultivated sorghum cultivars as well as eight shattercane [Sorghum bicolor (L.) Moench ssp. arundinaceum (Desv.) de Wet & Harlan] accessions and one johnsongrass [S. halepense (L.) Pers.] accession. Using a capillary growing mat system, root exudate was extracted with dichloromethane and subjected to chromatography analysis to determine sorgoleone-358 content. Root biomass of 7- to 12-d-old seedlings averaged 18.8 mg g-1 seed, and root exudate production ranged from 0.2 and 4.8 mg g-1 root fresh weight (RFW). The amount of sorgoleone produced varied greatly among sorghum accessions. Sorgoleone-358 amount in the root exudate averaged 0.5 mg g-1 RFW and varied from 0.13 to 1.05 mg g-1 for shattercane cultivar S7 and cultivated sorghum cultivar 992123, respectively. Regarding volume of root biomass, sorgoleone-358 levels averaged 0.49 mg g-1 (range, 0.06-1.46 mg g-1 ) for sorghum cultivar AAS3479 and shattercane cultivar S2, respectively. Segregation of commercial sorghum cultivars according to their maturity group did not show any difference in root biomass and dry extract production, but early-maturing cultivars produced on average 18% less sorgoleone-358 compared with medium- and late-maturing cultivars. These results suggest that sorgoleone production may be genetically constitutive because sorghum growing conditions were identical across cultivars.

Studies of improved seed adoption in developing countries are almost always based on household surveys and are premised on the assumption that farmers can accurately self‐report their use of improved seed varieties. However, recent studies suggest that farmers’ reports of seed varieties planted, or even whether the seed is local or improved, are sometimes inconsistent with the DNA fingerprinting results of those crops. We use household survey data from Tanzania to test the alignment between farmer‐reported and DNA‐identified maize seed types planted. In the sample, 70% of maize seed observations are correctly reported as local or improved, while 16% are type I errors (falsely reported as improved) and 14% are type II errors (falsely reported as local). Type I errors are more likely to have been sourced from other farmers, rather than formal channels. An analysis of input use, including seed, fertiliser, and labour allocations, reveals that farmers tend to treat improved maize differently, depending on whether they correctly perceive it as improved. This suggests that errors in farmers’ seed type awareness may translate into suboptimal management practices. The average yield of seed that is correctly identified as improved is almost 700 kg per hectare greater than that of type I errors. This indicates that investments in farmers’ access to information, seed labelling, and seed system oversight are needed to complement investments in seed variety development.

Biogeochemical analysis of a chronosequence of secondary forest succession in lowland Central Africa suggests that calcium becomes an increasingly scarce and potentially limiting resource with stand age and ecosystem calcium storage shifts from soil to woody biomass. Secondary forests constitute an increasingly important component of tropical forests worldwide. Although cycling of essential nutrients affects recovery trajectories of secondary forests, the effect of nutrient limitation on forest regrowth is poorly constrained. Here we use three lines of evidence from secondary forest succession sequences in central Africa to identify potential nutrient limitation in regrowing forests. First, we show that atmospheric phosphorus supply exceeds demand along forest succession, whereas forests rely on soil stocks to meet their base cation demands. Second, soil nutrient metrics indicate that available phosphorus increases along the succession, whereas available cations decrease. Finally, fine root, foliar and litter stoichiometry show that tissue calcium concentrations decline relative to those of nitrogen and phosphorus during succession. Taken together, these observations suggest that calcium becomes an increasingly scarce resource in central African forests during secondary succession. Furthermore, ecosystem calcium storage shifts from soil to woody biomass over succession, making it a vulnerable nutrient in the wake of land-use change scenarios that involve woody biomass export. Our results thus call for a broadened focus on elements other than nitrogen and phosphorus regarding tropical forest biogeochemical cycles and identify calcium as a scarce and potentially limiting nutrient in an increasingly disturbed and dynamic tropical forest landscape.

Take-all root rot is a disease of ultradwarf bermudagrass putting greens caused by Gaeumannomyces graminis (Gg), Gaeumannomyces sp. (Gx), Gaeumannomyces graminicola (Ggram), Candidacolonium cynodontis (Cc), and Magnaporthiopsis cynodontis (Mc). Many etiological and epidemiological components of this disease remain unknown. Improving pathogen identification and our understanding of the aggressiveness of these pathogens along with growth at different temperatures will advance our knowledge of disease development to optimize management strategies. Take-all root rot pathogens were isolated from symptomatic bermudagrass root and stolon pieces from 16 different golf courses. Isolates of Gg, Gx, Ggram, Cc, and Mc were used to inoculate ‘Champion’ bermudagrass in an in planta aggressiveness assay. Each pathogen was also evaluated at 10, 15, 20, 25, 30, and 35°C to determine growth temperature optima. Infected plant tissue was used to develop a real-time PCR high-resolution melt assay for pathogen detection. This assay was able to differentiate each pathogen directly from infected plant tissue using a single primer pair. In general, Ggram, Gg, and Gx were the most aggressive while Cc and Mc exhibited moderate aggressiveness. Pathogens were more aggressive when incubated at 30°C compared with 20°C. While they grew optimally between 24.4 and 27.8°C, pathogens exhibited limited growth at 35°C and no growth at 10°C. These data provide important information on this disease and its causal agents that may improve take-all root rot management.

Neonates are at an increased risk of an infectious disease. This is consistent with an increased abundance of myeloid-derived suppressor cells (MDSCs) compared with older children and adults. Using a murine model of neonatal bacterial sepsis, we demonstrate that MDSCs modulate their activity during an infection to enhance immune suppressive functions. A gene expression analysis shows that MDSCs increased NOS2, Arg-1 and IL-27p28 expression in vitro and in vivo in response to Escherichia coli O1:K1:H7 and this is regulated at the level of the gene expression. Changes in the effector gene expression are consistent with increased enzymatic activity and cytokine secretion. The neonatal MDSCs express toll-like receptor (TLR) 2, 4 and 5 capable of recognizing pathogen-associated molecular patterns (PAMPS) on E. coli. However, a variable level of effector expression was achieved in response to LPS, peptidoglycan or flagellin. Individual bacterial PAMPs did not stimulate the expression of Arg-l and IL-27p28 equivalently to E. coli. However, the upregulation of NOS2 was achieved in response to LPS, peptidoglycan and flagella. The increased immune suppressive profile translated to an enhanced suppression of CD4+ T cell proliferation. Collectively, these findings increase our understanding of the dynamic nature of MDSC activity and suggest that these cells abundant in early life can acquire activity during an infection that suppresses protective immunity.

This study examined Culex pipiens pallens responses to different combinations of colors and chemicals employed via a mosquito trap under semifield conditions. Our results indicated that Cx. p. pallens has color and chemical concentration preferences. Culex p. pallens had a 38.0% greater response to white than black color treated traps. Further, Cx. p. pallens showed differences in olfactory attraction depending on the chemical and concentration. Culex p. pallens was 107.6% more attracted to traps employing 500 ppm ammonia than control (i.e., unscented). Similarly, Cx. p. pallens was 117.5%, 128.8%, and 140.3% more attracted to traps employing, respectively, 1,000, 10,000, and 20,000 ppm of ammonia hydrogen carbonate compared to controls. And the response to lactic acid showed that Cx. p. pallens was most attracted to concentrations of 100 and 500 ppm (135.7% and 142.9%, respectively) compared to controls.

Pesticides can move off-target resulting in contamination of sensitive water bodies and causing adverse effects on inhabiting species. Through best management practices, such as the implementation of vegetative buffer strips, off-target movement of pesticides can be decreased, and compound degradation can be increased via phytoremediation. In this study, blueflag iris (Iris versicolor), broomsedge (Andropogon virginicus) and switchgrass (Panicum virgatum) were planted in soil treated with one of three commonly used pesticides. At 28, 56 and 112 days after treatment (DAT), plants were destructively harvested and analyzed for pesticide residue in soil and above-ground and below-ground vegetation using high-performance liquid chromatography (HPLC). Relative to the amount of pesticide found in planted pots compared to non-planted pots, I. versicolor was found to reduce greater atrazine in soil compared to non-planted pots at 112 DAT by 58.7%. I. versicolor was also the most capable of reducing azoxystrobin, by 86.9% compared to non-planted pots, from the soil at 112 DAT. At the same sampling time, I. versicolor and P. virgatum reduced greater imidacloprid from soil by 62.5% and 64.3% compared to non-planted pots, respectively. This information supports the recommendation for establishment of diverse plant species for optimization of phytoremediation capacities. Novelty statement While research has found that plants can absorb and remediate synthetic chemicals, this practice is only sustainable if used with native plants that require low maintenance and are tolerant to the applied substances. Various previous studies observe plants that are fast-growing, tolerant to environmental conditions, require low-maintenance, and are hardy. However, these plant species are not always suitable for any location and are often considered invasive and/or weed-like. The present research initiates a list of plant species which can be used within the southeastern United States and similar areas to phytoremediate commonly used pesticides atrazine, azoxystrobin, and imidacloprid and prevent off-target movement.

Epigenetic mechanisms such as histone modifications play critical roles in adaptive tuning of chromatin structures. Profiling of various histone modifications at the genome scale using tissues from animal and human samples is an important step for functional studies of epigenomes and epigenomics-based precision medicine. Because the profile of a histone mark is highly specific to a cell type, cell isolation from tissues is often necessary to generate a homogeneous cell population, and such operations tend to yield a low number of cells. In addition, high-throughput processing is often desirable because of the multiplexity of histone marks of interest and the large quantity of samples in a hospital setting. In this protocol, we provide detailed instructions for device fabrication, setup, and operation of microfluidic oscillatory washing-based chromatin immunoprecipitation followed by sequencing (MOWChIP-seq) for profiling of histone modifications using as few as 100 cells per assay with a throughput as high as eight assays in one run. MOWChIP-seq operation involves flowing of chromatin fragments through a packed bed of antibody-coated beads, followed by vigorous microfluidic oscillatory washing. Our process is semi-automated to reduce labor and improve reproducibility. Using one eight-unit device, it takes 2 d to produce eight sequencing libraries from chromatin samples. The technology is scalable. We used the protocol to study a number of histone modifications in various types of mouse and human tissues. The protocol can be conducted by a user who is familiar with molecular biology procedures and has basic engineering skills.

Management of many crown and root diseases of turfgrasses includes the use of fungicides. The physicochemical properties of the fungicides used vary greatly, but most have low mobility and are not phloem mobile, which results in little active ingredient present in the basal and underground structures of turfgrass plants. Two studies were conducted in a laboratory setting to determine the effects of post-application irrigation amounts (0, 0.3, 0.6, 1.3, and 2.5 cm) on the distribution of 14C myclobutanil and 14C tebuconazole in a soil profile of 90% sand and 10% peat moss by volume. In addition, growth chamber experiments were conducted to examine the effect of post-application irrigation amount on azoxystrobin efficacy against summer patch (Magnaporthiopsis poae) in ‘Penn A-4’ creeping bentgrass (Agrostis stolonifera L.). The creeping bentgrass was treated with azoxystrobin and immediately irrigated with 0, 0.25, 0.3, or 0.6 cm of irrigation. Lastly, an in vitro fungicide sensitivity assay was conducted on three M. poae isolates to determine their sensitivities to two succinate dehydrogenase inhibitors (SDHIs), three demethylation inhibitors (DMIs), and four strobilurins (QoIs) to determine if fungicide concentrations in the soil profile reached levels high enough to suppress fungal growth. In both 14C experiments, more than 54% of the 14C was retained in the top 5 cm of soil for all irrigation treatments. For the 14C myclobutanil experiment, lysimeters treated with 2.5 cm of post-application irrigation resulted in 3.9% of 14C recovered in the 7.6–10.2 cm sampling depth, which was higher than all other irrigation treatments. Post-application irrigation at 2.5 cm in the 14C tebuconazole experiment yielded 6.3% of the 14C at the 7.6–10.2 cm sampling depth and 2.3% at the 10.2–12.7 cm sampling depth—recoveries at both depths were higher with 2.5 cm of irrigation than all other irrigation treatments. No 14C was detected below 12.7 cm for either experiment. Less disease was observed when azoxystrobin received post-application irrigation. Both 0.25 and 0.3 cm of post-application irrigation increased turf quality compared to no irrigation; moreover, 0.25 and 0.6 cm of post-application irrigation increased root length compared to no irrigation. In the in vitro fungicide sensitivity assay, isolates of M. poae were sensitive to all fungicides with only minor sensitivity to the SDHIs. In general, isolates were most sensitive to the QoIs with some variability in isolate sensitivities noted. Isolates of M. poae are sensitive to commonly used fungicides and efficacy is enhanced by post-application irrigation due to improved fungicide distribution into the soil profile.

The Hutton's shearwater Puffinus huttoni is an endangered seabird endemic to Kaikōura, New Zealand, but the spatial and temporal aspects of its at‐sea foraging behavior are not well known. To identify foraging areas and estimate trip durations, we deployed Global Positioning Systems (GPS) devices and Time‐Depth Recorders (TDR) on 26 adult Hutton's shearwaters during the chick‐rearing period in 2017 and 2018. We found Hutton's shearwaters traveled much further from their breeding grounds at Kaikōura than previously considered, with most individuals foraging in coastal and oceanic areas 125–365 km south and near Banks Peninsula. Trip durations varied from 1 to 15 days (mean = 5 days), and total track lengths varied from 264 to 2,157 km (mean = 1092.9 km). Although some diving occurred in near‐shore waters near the breeding colony, most foraging was concentrated in four regions south of Kaikōura. Dive durations averaged 23.2 s (range 8.1 to 71.3 s) and dive depths averaged 7.1 m (range 1.5 to 30 m). Foraging locations had higher chlorophyll a levels and shallower water depths than nonforaging locations. Birds did not feed at night, but tended to raft in areas with deeper water than foraging locations. Mapping the spatial and temporal distribution of Hutton's shearwaters at sea will be fundamental to their conservation, as it can reveal potential areas of overlap with fisheries and other industrial users of the marine environment.

Oil and natural gas development in the western United States has increased substantially in recent decades as technological advances like horizontal drilling and hydraulic fracturing have made extraction more commercially viable. Oil and gas pads are often developed for production, and then capped, reclaimed, and left to recover when no longer productive. Understanding the rates, controls, and degree of recovery of these reclaimed well sites to a state similar to pre-development conditions is critical for energy development and land management decision processes. Here we use a multi-decadal time series of satellite imagery (Landsat 5, 1984–2011) to assess vegetation regrowth on 365 abandoned well pads located across the Colorado Plateau in Utah, Colorado, and New Mexico. We developed high-frequency time series of the Soil-Adjusted Total Vegetation Index (SATVI) for each well pad using the Google Earth Engine cloud computing platform. BFAST time-series models were used to fit temporal trends, identifying when vegetation was cleared from the site and the magnitudes and rates of vegetation change after abandonment. The time series metrics are used to calculate the relative fractional vegetation cover (RFVC) of each pad, a measure of post-abandonment vegetation cover relative to pre-drilling condition. Mean and median RFVC were 36% (s.d. 33%) and 26%, respectively, five years after abandonment, with one third of well pads having RFVC greater than 50%. Statistical analyses suggest that much of the high vegetation cover is associated with weedy invasive annual species such as cheatgrass (Bromus tectorum) and Russian thistle (Salsola spp.). Climate conditions and the year of abandonment also play a role, with increased cover in later years associated with a wetter period. Non-linear change at many pads suggests longer recovery times than would be estimated by linear extrapolation. New techniques implemented here address a complex response of cover change to soils, management, and climate over time, and can be extended to the operational monitoring of energy development across large areas.

In many East African countries, women and men have different levels of access to formal markets for agricultural inputs, including seed, reflecting a combination of gender norms and resource constraints. As a result, women and men may have different levels of participation in—and reliance upon—informal seed systems for sourcing preferred planting material and accessing new crop varieties over time. We use network analysis to explore differences in seed networks accessed by women and men for three major food security crops—beans, finger millet, and sorghum—in Kenya, Tanzania, and Uganda. Drawing on data from an original survey of 1001 rural farm households across five study sites, we find that women, on average, have fewer connections to experts and farmers’ groups than men but are relatively better connected in farmer-to-farmer social networks across different farming systems. We further find women’s and men’s networks are clustered by gender (i.e., women’s networks include more women, and men’s networks include more men)—and that men’s networks are more likely to exchange improved seed. Women’s networks, though sometimes larger, are less likely to exchange improved varieties that might help farmers adapt to climate change. Women farmers across contexts may also be more reliant on farmer-to-farmer networks than men due to their relative isolation from other seed and information sources. Findings emphasize the need for careful attention to the different implications of seed policies, market interventions, and other seed system reforms to support gender-equitable food security options for women and men in sub-Saharan Africa.

Heat stress (HS) has been reported to disrupt nutrient digestion and absorption in broilers. These effects may be more prominent in fast-growing chickens due to their high metabolic activity. However, the underlying molecular mechanisms are not yet fully elucidated. Hence, the current study aimed to evaluate the effect of chronic HS on jejunal nutrient transport in slow- (Athens Canadian Random Bred, ACRB from 1950), moderate- (The 1995 random bred, 95RAN), rapid- (modern broilers, modern random bred, MRB) growing birds and their ancestor wild jungle fowl (JF). One-day male chicks (n = 150/line) were placed by line in environmentally controlled chambers and kept under the same industry-standard environmental conditions until d28. On d29, an 8-h daily cyclic HS (36°C) was applied to half of the chambers, which lasted until d55, while keeping the rest under thermal neutral (TN, 24°C) conditions. Jejunum tissues were collected for morphology assessment and molecular analysis of carbohydrate-, amino acid-, and fatty acid-transporters. MRB exhibited the highest body weight (BW) followed by 95RAN under both conditions. HS decreased feed intake (FI) in MRB and 95RAN, which resulted in lower BW compared to their TN counterparts; however, no effect was observed in ACRB and JF. MRB showed a greater villus height (VH) to crypt depth (CD) ratio under both environmental conditions. Molecular analyses showed that glucose transporter (GLUT) 2, 5, 10, and 11 were upregulated in MRB compared to some of the other populations under TN conditions. HS downregulated GLUT2, 10, 11, and 12 in MRB while it increased the expression of GLUT1, 5, 10, and 11 in JF. GLUT2 protein expression was higher in JF compared to ACRB and MRB under TN conditions. It also showed an increase in ACRB but no effect on 95RAN and MRB under HS conditions. ACRB exhibited greater expression of the EAAT3 gene as compared to the rest of the populations maintained under TN conditions. HS exposure did not alter the gene expression of amino acid transporters in MRB. Gene expression of CD36 and FABP2 was upregulated in HS JF birds. Protein expression of CD36 was downregulated in HS JF while no effect was observed in ACRB, 95RAN, and MRB. Taken together, these data are the first to show the effect of HS on jejunal expression of nutrient transporters in three broiler populations known to represent 70 years of genetic progress in the poultry industry and a Red Jungle Fowl population representative of the primary ancestor of domestic chickens.

Cyperus esculentus and Cyperus rotundus are common weeds of agricultural and horticultural cropping systems throughout tropical, subtropical, and temperate areas of the world. A network of rhizomes, tubers, and basal bulbs are the primary mechanism by which both species proliferate. Both species are favored by high light intensity and tolerate a wide range of soil types, but the most important factors affecting their prevalence are temperature, soil moisture, and photoperiod. High growth rates and quick emergence of these species create competition during early crop development that can result in yield reductions leading to significant economic losses. Crop quality, weed control strategies, and harvest operations are often compromised. No single control tactic provides season-long control of Cyperus species; therefore an integrated approach that involves biological, cultural, and chemical options will be most effective.

Primary tropical forests generally exhibit large gaseous nitrogen (N) losses, occurring as nitric oxide (NO), nitrous oxide (N2O) or elemental nitrogen (N2). The release of N2O is of particular concern due to its high global warming potential and destruction of stratospheric ozone. Tropical forest soils are predicted to be among the largest natural sources of N2O; however, despite being the world’s second-largest rainforest, measurements of gaseous N-losses from forest soils of the Congo Basin are scarce. In addition, long-term studies investigating N2O fluxes from different forest ecosystem types (lowland and montane forests) are scarce. In this study we show that fluxes measured in the Congo Basin were lower than fluxes measured in the Neotropics, and in the tropical forests of Australia and South East Asia. In addition, we show that despite different climatic conditions, average annual N2O fluxes in the Congo Basin’s lowland forests (0.97 ± 0.53 kg N ha−1 year−1) were comparable to those in its montane forest (0.88 ± 0.97 kg N ha−1 year−1). Measurements of soil pore air N2O isotope data at multiple depths suggests that a microbial reduction of N2O to N2 within the soil may account for the observed low surface N2O fluxes and low soil pore N2O concentrations. The potential for microbial reduction is corroborated by a significant abundance and expression of the gene nosZ in soil samples from both study sites. Although isotopic and functional gene analyses indicate an enzymatic potential for complete denitrification, combined gaseous N-losses (N2O, N2) are unlikely to account for the missing N-sink in these forests. Other N-losses such as NO, N2 via Feammox or hydrological particulate organic nitrogen export could play an important role in soils of the Congo Basin and should be the focus of future research., The ISME Journal, 15, ISSN:1751-7362, ISSN:1751-7370

(1) Background: Arthrofibrosis is a common cause of patient debility and dissatisfaction after total knee arthroplasty (TKA). The diversity of molecular pathways involved in arthrofibrosis disease progression suggest that effective treatments for arthrofibrosis may require a multimodal approach to counter the complex cellular mechanisms that direct disease pathogenesis. In this study, we leveraged RNA-seq data to define genes that are suppressed in arthrofibrosis patients and identified adiponectin (ADIPOQ) as a potential candidate. We hypothesized that signaling pathways activated by ADIPOQ and the cognate receptors ADIPOR1 and ADIPOR2 may prevent fibrosis-related events that contribute to arthrofibrosis. (2) Methods: Therefore, ADIPOR1 and ADIPOR2 were analyzed in a TGF&beta, 1 inducible cell model for human myofibroblastogenesis by both loss- and gain-of-function experiments. (3) Results: Treatment with AdipoRon, which is a small molecule agonist of ADIPOR1 and ADIPOR2, decreased expression of collagens (COL1A1, COL3A1, and COL6A1) and the myofibroblast marker smooth muscle &alpha, actin (ACTA2) at both mRNA and protein levels in basal and TGF&beta, 1-induced cells. (4) Conclusions: Thus, ADIPOR1 and ADIPOR2 represent potential drug targets that may attenuate the pathogenesis of arthrofibrosis by suppressing TGF&beta, dependent induction of myofibroblasts. These findings also suggest that AdipoRon therapy may reduce the development of arthrofibrosis by mediating anti-fibrotic effects in joint capsular tissues.

Epigenome constitutes an important layer that regulates gene expression and dynamics during development and diseases. Extensive efforts have been made to develop epigenome profiling methods using a low number of cells and with high throughput. Chromatin immunoprecipitation (ChIP) is the most important approach for profiling genome-wide epigenetic changes such as histone modifications. In this report, we demonstrate microfluidic ChIPmentation (mu-CM), a microfluidic technology that enables profiling cell samples that individually do not generate enough ChIP DNA for sequencing library preparation. We used a simple microfluidic device to allow eight samples to be processed simultaneously. The samples were indexed differently using a tagmentation-based approach (ChIPmentation) and then merged for library preparation. A histone modification profile for each individual sample was obtained by demultiplexing the sequencing reads based on the indexes. Our technology allowed profiling 20 cells and is well suited for cell-type-specific studies using low-abundance tissues.

Understanding how organisms respond to environmental change is one of the most pressing grand challenges of organismal biology. In the vast oceans that cover 71% of Earth’s surface, remote sensing technologies have created unprecedented opportunities to create new knowledge and deliver integrated understandings of marine organism-environment interactions via long-term monitoring. Using historic whaling records and >15 years of satellite-derived data, we show that movement parameters associated with long-distance humpback whale migrations, including utilization of a south-southeast directed migratory corridor, migration path straightness, direction, timing, and velocity, have not significantly changed during a period of dynamic oceanographic and geomagnetic conditions. These findings reveal an apparent paradox: humpback whale migrations do not change in a changing ocean. Geophysical analyses of the same humpback whale movements demonstrate that these whales maintained prolonged migratory fidelity to a limited suite of spatiotemporal trajectories through gravitational coordinates, raising the possibility that migratory decisions are relatively insensitive to changing oceanographic and geomagnetic conditions. Our findings highlight the importance of filling the knowledge gaps that currently limit our ability to understand and anticipate organismal responses to rapidly changing Earth system conditions.

In the present work, we hypothesized that increased levels of IL-27 predispose neonatal mice to more severe infection during Gram-negative sepsis. Peripheral tissue analysis revealed systemic IL-27 expression, while myeloid cell profiling identified Gr-1- and F4/80-expressing cells as the most abundant producers of IL-27 during infection. Increased IL-27 levels were consistent with increased mortality that was improved in IL-27 receptor α (IL-27Rα)−/− mice that lack a functional IL-27 receptor. Infected IL-27Rα−/− pups also exhibited improved weight gain and reduced morbidity. This was consistent with reduced bacterial burdens and more efficient bacterial killing by Ly6B.2+ myeloid cells and macrophages compared to WT neonates. Live animal imaging further supported a more severe and disseminated infection in WT neonates. This is the first report to describe the impact of elevated early-life IL-27 on the host response in a neonatal infection model while also defining the cell and tissue sources of cytokine. IL-27 is frequently associated with suppressed inflammation. In contrast, our findings demonstrate that IL-27 indirectly promotes an inflammatory cytokine response during neonatal sepsis by directly compromising control of bacteria that drive the inflammatory response. Collectively, our results suggest that IL-27 represents a therapeutic target to limit susceptibility and improve infectious outcomes in neonatal sepsis.

Large mammalian herbivores are experiencing population reductions and range declines. However, we lack regional knowledge of population status for many herbivores, particularly in developing countries. Addressing this knowledge gap is key to implementing tailored conservation strategies for species whose population declines are highly variable across their range. White-lipped peccaries (Tayassu pecari) are important ecosystem engineers in Neotropical forests and are highly sensitive to human disturbance. Despite maintaining a wide distributional range, white-lipped peccaries are experiencing substantial population declines in some portions of their range. We examined the regional distribution and population status of the species in Mesoamerica. We used a combination of techniques, including expert-based mapping and assessment of population status, and data-driven distribution modelling techniques to determine the status and range limits of white-lipped peccaries. Our analysis revealed declining and highly isolated populations of peccaries across Mesoamerica, with a range reduction of 87% from historic distribution and 63% from current IUCN range estimates for the region. White-lipped peccary distribution is affected by indices of human influence and forest cover, and more restricted than other sympatric large herbivores, with their largest populations confined to transboundary reserves. To conserve white-lipped peccaries in Mesoamerica, transboundary efforts will be needed that focus on both forest conservation and hunting management, increased cross-border coordination, and reconsideration of country and regional conservation priorities. Our methodology to detail regional white-lipped peccary status could be employed on other poorly-known large mammals. Lower-extremity musculoskeletal injuries are common in sports such as basketball and soccer. Athletes competing in sports of this nature must maneuver in response to the actions of their teammates, opponents, etc. This limits their ability to preplan movements. The purpose of this study was to compare impact accelerations during preplanned versus unplanned lateral cutting. A total of 30 subjects (15 males and 15 females) performed preplanned and unplanned cuts while the authors analyzed impact accelerations using an accelerometer secured to their tibia. For the preplanned condition, subjects were aware of the movement to perform before initiating a trial. For the unplanned condition, subjects initiated their movement and then reacted to the illumination of one of 3 visual stimuli which dictated whether they would cut, land, or land-and-jump. A mixed-model analysis of variance with a between factor of sex (male and female) and a within factor of condition (preplanned and unplanned) was used to analyze the magnitude and variability of the impact accelerations for the cutting trials. Both males and females demonstrated higher impact accelerations (P = .01) and a trend toward greater intertrial variability (P = .07) for the unplanned cutting trials (vs preplanned cuts). Unplanned cutting may place greater demands on the musculoskeletal system.

2,4-dimethylamine salt (2,4-D) is a synthetic auxin herbicide used extensively in turfgrass for selective broadleaf weed control. Previous research has shown that 2,4-D can dislodge from treated turf, notably in the presence of canopy moisture. Practitioners commonly apply 2,4-D in combination with various commercially available surfactants to increase efficacy. Field research was completed to evaluate the effect of surfactant inclusion and sample collection time within a day on dislodgeable 2,4-D residue from perennial ryegrass. Research was initiated May 24, 2016 in Raleigh, NC and repeated in time to quantify dislodgeable 2,4-D following application (2.1 kg ae ha–1) either alone or with a nonionic surfactant (0.5% vol/vol). Sample collection occurred 1, 2, 3, 6, 12 or 24 d after treatment (DAT) at AM [7:00 AM Eastern Standard Time (EST)] and PM (2:00 PM EST) sample timings within a day. 2,4-D applied with surfactant (0.4% to 25.4% of applied) reduced dislodgeable foliar residue compared to 2,4-D applied alone (0.5% to 31.2%) from 1 through 6 DAT, whereas dislodgeable 2,4-D was not detected at 12 and 24 DAT. Regardless of surfactant inclusion or absence, samples collected in the AM resulted in a 5- to 10-fold increase in dislodgeable 2,4-D compared to samples collected in the PM from 1 through 6 DAT, suggesting that 2,4-D dislodgeability may be influenced by conditions favoring canopy moisture development. This research will improve turfgrass management practices and research designed to minimize human 2,4-D exposure.

The hypothalamic-pituitary-thyroid (HPT) axis is known to play a crucial role in the development of teleost fish. However, knowledge of endogenous transcription profiles of thyroid-related genes in developing teleosts remains fragmented. We selected two model teleost species, the fathead minnow ( Pimephales promelas ) and the zebrafish ( Danio rerio ), to compare the gene transcription ontogeny of the HPT axis. Control organisms were sampled at several time points during embryonic and larval development until 33 days post-fertilization. Total RNA was extracted from pooled, whole fish, and thyroid-related mRNA expression was evaluated using quantitative polymerase chain reaction. Gene transcripts examined included: thyrotropin-releasing hormone receptor ( trhr ), thyroid-stimulating hormone receptor ( tshr ), sodium-iodide symporter ( nis ), thyroid peroxidase ( tpo ), thyroglobulin ( tg ), transthyretin ( ttr ), deiodinases 1, 2, 3a, and 3b ( dio1, dio2, dio3a and 3b ), and thyroid hormone receptors alpha and beta ( thrα and β ). A loess regression method was successful in identifying maxima and minima of transcriptional expression during early development of both species. Overall, we observed great similarities between the species, including maternal transfer, at least to some extent, of almost all transcripts (confirmed in unfertilized eggs), increasing expression of most transcripts during hatching and embryo-larval transition, and indications of a fully functional HPT axis in larvae. These data will aid in the development of hypotheses on the role of certain genes and pathways during development. Furthermore, this provides a background reference dataset for designing and interpreting targeted transcriptional expression studies both for fundamental research and for applications such as toxicology.

Nitrate pollution is a global issue threatening the health and function of many lowland freshwater ecosystems. Quantifying nitrate loads and instream attenuation associated with land use is a critical requirement for improving freshwater management. One often overlooked nitrate source in catchments is invasive N-fixing trees such as Ulex europaeus (European gorse). This study compared nitrate concentrations in conjunction with stable isotope analyses (nitrate δ15N and δ18O) to investigate the effects of catchment gorse cover on stream nitrate export relative to three other land uses. These were regenerating native forest, low-intensity (dry-stock) and high-intensity (dairying) agriculture. We tested two hypotheses: (1) gorse is a regionally significant nitrate source; (2) instream nitrate attenuation is land-use dependent. The study was conducted in 24 reaches across six small, mixed land-use coastal catchments located on Banks Peninsula, New Zealand. Our results demonstrated that gorse-dominated stream reaches had significantly higher nitrate concentrations than all other land uses. Within the gorse-dominated reaches, nitrate concentration was significantly correlated with upstream catchment gorse cover. Furthermore, nitrate oxygen and nitrogen stable isotope compositions demonstrated that elevated nitrate concentrations in gorse streams were associated with decomposition of dead gorse foliage. The isotope data revealed sub-catchment-scale land-use-specific patterns of nitrate attenuation within streams. All three anthropogenic land uses (gorse, dry stock and dairy) showed distinctly different N-cycling from native-forested reaches where nitrate was efficiently cycled with evidence for highly localised nitrification. Stable isotope data demonstrated that overall nitrate attenuation became less efficient with higher nitrate loads. Our research demonstrates the significant impact N-fixing plants have on nitrate concentrations and instream attenuation. Quantifying the effects of N-fixing plants on water quality is an important step in achieving globally significant goals of sustaining ecosystem health and (sub)catchment-scale nutrient management.
