Abstract: In order to ascertain the role of plant transglutaminases (TGase) in growth and abiotic stress response, the AtPng1 knock out (KO) line of A. thaliana has been analyzed during plant development and under heat and wound stress. Comparing wild type (WT) and KO lines a 58-kDa band was immunodetected by anti-AtPng1p antibody in the cell wall and chloroplasts only in the WT line. A residual TGase activity, not showing correlation with development nor stress response, was still present in the KO line. The KO line was less developed, with a juvenile phenotype characterized by fewer, smaller and less differentiated cells. Chloroplast TGase activity was insensitive to mutation. Data on stressed plants showed that (i) KO plants under heat stress were more juvenile compared to WT, (ii) different responses between WT and KO lines after wounding took place. TGase activity was not completely absent in the KO line, presenting high activity in the plastidial fraction. In general, the mutation affected A. thaliana growth and development, causing less differentiated cytological and anatomical features.

Abstract: Allogeneic hematopoietic stem cell transplantation is the treatment of choice for autosomal recessive osteopetrosis caused by defects in the TCIRG1 gene. Despite recent progress in conditioning, an important number of patients are not eligible for allogeneic stem cell transplantation because of the severity of the disease and significant transplant-related morbidity. We exploited peripheral CD34(+) cells, known to circulate at high frequency in the peripheral blood of TCIRG1-deficient patients, as a novel cell source for autologous transplantation of gene corrected cells. Detailed phenotypical analysis showed that circulating CD34(+). cells have a cellular composition that resembles bone marrow (BM), supporting their use in gene therapy protocols. Transcriptomic profile revealed enrichment in genes expressed by hematopoietic stem and progenitor cells (HSPC). To overcome the limit of BM harvest/HSPC mobilization and serial blood drawings in TCIRG1 patients, we applied UM171-based ex vivo expansion of HSPC coupled with lentiviral gene transfer. Circulating CD34(+). cells from TCIRG1-defective patients were transduced with a clinically-optimized lentiviral vector expressing TCIRG1 under the control of phosphoglycerate promoter and expanded ex vivo. Expanded cells maintained long-term engraftment capacity and multi-lineage repopulating potential when transplanted in vivo both in primary and secondary NOD scid gamma common chain (NSG) recipients. Moreover, when CD34(+) cells were differentiated in vitro, genetically corrected osteoclasts resorbed the bone efficiently. Overall, we provide evidence that expansion of circulating HSPC coupled to gene therapy can overcome the limit of stem cell harvest in osteopetrotic patients, thus opening the way to future gene-based treatment of skeletal diseases caused by BM fibrosis.

Abstract: Consumption of edible insects has been widely suggested as an environmentally sustainable substitute for meat to reduce greenhouse gas emissions. However, the novel research field for edible insects relies on the content of bioactive ingredients and on the ability to induce a functional effect in humans. The goal of this manuscript is to review the available body of evidence on the properties of edible insects in modulating oxidative and inflammatory stress, platelet aggregation, lipid and glucose metabolism and weight control. A search for literature investigating the functional role of edible insects was carried out in the PubMed database using specific keywords. A total of 55 studies, meeting inclusion criteria after screening, were divided on the basis of the experimental approach: in vitro studies, cellular models/ex vivo studies or in vivo studies. In the majority of the studies, insects demonstrated the ability to reduce oxidative stress, modulate antioxidant status, restore the impaired activity of antioxidant enzymes and reduce markers of oxidative damage. Edible insects displayed anti-inflammatory activity reducing cytokines and modulating specific transcription factors. Results from animal studies suggest that edible insects can modulate lipid and glucose metabolism. The limited number of studies focused on the assessment of anti-coagulation activity of edible insects makes it difficult to draw conclusions. More evidence from dietary intervention studies in humans is needed to support the promising evidence from in vitro and animal models about the functional role of edible insect consumption.

4. Phenotypic variability and developmental instability in rodents from different agricultural farming systems: organic vs. conventional

Abstract: Contrary to conventional agriculture, organic farming, where agrochemicals are entirely avoided, is considered more environmentally friendly. Numerous studies undertaken in agroecosystems have found that the intensification of agriculture has a negative impact on some small mammals species. In this study, we used two morphological indicators to evaluate the impact of agriculture intensification. Phenotypic variability and fluctuating asymmetry (FA- development instability proxy) have been widely used as morphological indicators of developmental stress. We implemented geometric morphometric methods to assess the influence of different agricultural farming systems on three rodent species. We hypothesize that conventional farming produces more stressful conditions than organic farming affecting species susceptible to agricultural intensification. We predicted that Akodon azarae, negatively affected by landscape simplification and more dependent on habitat quality, will show higher levels of phenotypic variation and greater FA in conventional farming. Whereas, Calomys musculinus and C. laucha, unaffected by agricultural intensification, will not show differences between farming systems. Akodon azarae exhibited higher phenotypic variability in conventional farms, while C. musculinus had no difference between farming systems. Contrarily, C. laucha exhibited higher values in conventional farms. Regarding FA, both Calomys species showed no differences between farming systems. Females of A. azarae tend to have higher FA values in conventional farming. Our results suggest that the effect of agricultural farming systems would vary according to the species, where the species most dependent on habitat quality would be more affected by intensive agriculture.

5. CULTIVO CONSORCIADO DE BATATA-DOCE E MILHO PIPOCA: CRESCIMENTO E PRODUÇÃO DAS CULTURAS

Abstract: O objetivo foi avaliar o crescimento das plantas, produção de raízes de batata-doce (Ipomoea batatas) e grãos de milho pipoca (Zea mays) em sistema de cultivo solteiro e consorciado. Foram avaliadas quatro cultivares de batata-doce e uma de pipoca em blocos casualizados com três repetições em condições de campo. No cultivo solteiro, a pipoca e batata-doce foram espaçadas 75 cm entre linhas. No consorciado, o plantio da batata-doce foi no centro das entre linhas da pipoca. A adubação foi realizada com cama de frango. Aos 30, 60 e 90 dias foi avaliado o número de folhas, altura das plantas, diâmetro do colmo/baraços e comprimento de baraços. No florescimento foi determinada a matéria seca das plantas, na maturação fisiológica a produção de grãos da pipoca e aos 180, as raízes da batata-doce. Observou-se que a pipoca apresentou a maior produção de matéria seca, número de folhas, diâmetro de colmos e altura de plantas quando solteiro, as cultivares de batata-doce quando consorciadas menor produção de matéria seca e raízes, a produção de grãos de pipoca não apresentou diferença entres os sistemas de produção. O sistema consorciado de pipoca e batata-doce promove maior produção de matéria seca e produção de alimentos por área. Intercropped cultivation of sweet potatoes and popcorn: growth and production of crops ABSTRACT: The objective was to evaluate plant growth, sweet potato productivity (Ipomoea batatas) and grains of popcorn (Zea mays) in single and intercropped systems. Four sweet potato cultivars and one popcorn in randomized blocks were evaluated with three replications in field conditions. In single cultivation, popcorn and sweet potatoes were spaced 75 cm between rows. In the intercropped, the sweet potato was in the center of between the lines of popcorn. Fertilization was carried out with chicken manure. At 30, 60 and 90 days, the number of leaves, plant height, stem/tiller diameter and tether length were evaluated. At flowering, the dry matter of the plants was determined, at physiological maturity the grain yield of the popcorn and at 180, the roots of the sweet potato. It was observed that the popcorn showed the highest production of dry matter, number of leaves, stem diameter and plant height when single, sweet potato cultivars when intercropped, lower production of dry matter and roots, the productivity of popcorn grains there was no difference between the production systems. The intercroped system of popcorn and sweet potatoes promotes greater production of dry matter and food productivity per area. Abstract: The European physical journal / C 81(11), 973 (2021). doi:10.1140/epjc/s10052-021-09544-4, The OSIRIS detector is a subsystem of the liquid scintillator filling chain of the JUNO reactor neutrino experiment. Its purpose is to validate the radiopurity of the scintillator to assure that all components of the JUNO scintillator system work to specifications and only neutrino-grade scintillator is filled into the JUNO Central Detector. The aspired sensitivity level of $10^{��16}$ g/g of $^{238}$U and $^{232}$Th requires a large (��20 m$^3$) detection volume and ultralow background levels. Abstract: Severe intra-abdominal infection commonly requires intensive care. Mortality is high and is mainly determined by disease-specific characteristics, i.e. setting of infection onset, anatomical barrier disruption, and severity of disease expression. Recent observations revealed that antimicrobial resistance appears equally common in community-acquired and late-onset hospital-acquired infection. This challenges basic principles in anti-infective therapy guidelines, including the paradigm that pathogens involved in community-acquired infection are covered by standard empiric antimicrobial regimens, and second, the concept of nosocomial acquisition as the main driver for resistance involvement. In this study, we report on resistance profiles of Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Enterococcus faecalis and Enterococcus faecium in distinct European geographic regions based on an observational cohort study on intra-abdominal infections in intensive care unit (ICU) patients. Resistance against aminopenicillins, fluoroquinolones, and third-generation cephalosporins in E. coli, K. pneumoniae and P. aeruginosa is problematic, as is carbapenem-resistance in the latter pathogen. For E. coli and K. pneumoniae, resistance is mainly an issue in Central Europe, Eastern and South-East Europe, and Southern Europe, while resistance in P. aeruginosa is additionally problematic in Western Europe. Vancomycin-resistance in E. faecalis is of lesser concern but requires vigilance in E. faecium in Central and Eastern and South-East Europe. In the subcohort of patients with secondary peritonitis presenting with either sepsis or septic shock, the appropriateness of empiric antimicrobial therapy was not associated with mortality. In contrast, failure of source control was strongly associated with mortality. The relevance of these new insights for future recommendations regarding empiric antimicrobial therapy in intra-abdominal infections is discussed.

9. Propagation by Stem Cuttings of Tropaeolum pentaphyllum Lam. (Crem): An Alternative for Production of Seed Tubers

Abstract: The herbaceous vine Tropaeolum pentaphyllum Lam. (Tropaeolaceae), popularly known as crem, has great potential for use, especially its tubers. However, there is still a lack of studies regarding this species. The objective of this study was to test the viability of propagation by stem cuttings for the production of T. pentaphyllum seed tubers. Two experiments were carried out in a greenhouse under controlled conditions. In both experiments, experimental design was completely randomized. In the first one, five treatments were evaluated (stem cuttings with 10, 15, 20, 25, and 30 cm) with three replicates, each one with 10 stem cuttings. In the second experiment, three treatments were evaluated (stem cuttings positions: apical, middle and basal), with four replications, each one with 10 stem cuttings. Data were submitted to analysis of variance (ANOVA) and the means were compared by Tukey’s test (p < 0.05). Stem cuttings with 15 cm length had the highest rate of tuber formation (20.0%) and produced tubers with highest mass (3.2 g). Cuttings from the middle portion of the stem had the highest rooting (52.5%) and tuber formation (45.0%) rates. Thus, cuttings with 15 cm length from the middle portion of the stem are indicated for propagation by cuttings of T. pentaphyllum. Considering the potential for the use of this species in the future, the results found in this study are very important, since the propagation by cuttings would enable the production of a large amount of seed tubers in a short period of time.

Abstract: Fungal biofilms, such as Candida albicans biofilms, are capable of surviving in hostile environments owing to their remarkable ability to adhere to surfaces and their tolerance to chemical interventions. Currently, therapeutic treatment options are few, making these biofilm-based infections problematic particularly due to their great tolerance to conventional antimicrobial drugs, thus causing serious health and economic problems. Therefore, the development of new drugs and antibiofilm specific therapies for the prevention and treatment of antifungal to eradicate biofilms are needed. This study was aimed at carrying out a patent review analysis to identify the innovation trends, and to explore the latest antifungal drugs and the specific therapeutic strategies available for the treatment of fungal biofilms. The present patent review was carried out using the Espacenet database, using the key words "biofilm and antifungal," from 2002 to December 2019. Through this review, it was possible to identify that most of the patent contents refer to new synthetic drugs derived from natural products and associations thereof with existing antifungal drugs. Methods and biomaterials for the treatment and prevention of fungal biofilms, mainly for C. albicans biofilms, which is the most isolated and studied fungal species, were also disclosed. The lack of scientific and technical information on the biofilm eradication subject is remarkable and further confirmed by the small number of patents identified in this survey.

Abstract: Polydnaviruses (PDV) are viral symbionts associated with ichneumonid and braconid wasps parasitizing moth larvae, which are able to disrupt the host immune response and development, as well as a number of other physiological pathways. The immunosuppressive role of PDV has been more intensely investigated, while very little is known about the PDV-encoded factors disrupting host development. Here we address this research issue by further expanding the functional analysis of ankyrin genes encoded by the bracovirus associated with Toxoneuron nigriceps (Hymenoptera, Braconidae). In a previous study, using Drosophila melanogaster as experimental model system, we demonstrated the negative impact of TnBVank1 impairing the ecdysone biosynthesis by altering endocytic traffic in prothoracic gland cells. With a similar approach here we demonstrate that another member of the viral ank gene family , TnBVank3, does also contribute to the disruption of ecdysone biosynthesis, but with a completely different mechanism. We show that its expression in Drosophila prothoracic gland (PG) blocks the larval-pupal transition by impairing the expression of steroidogenic genes. Furthermore, we found that TnBVank3 affects the expression of genes involved in the insulin/TOR signaling and the constitutive activation of the insulin pathway in the PG rescues the pupariation impairment. Collectively, our data demonstrate that Tn BVANK3 acts as a virulence factor by exerting a synergistic and non-overlapping function with TnBVANK1 to disrupt the ecdysone biosynthesis.

Abstract: Objectives Procalcitonin (PCT) has been proposed for differentiating viral vs. bacterial infections. In COVID-19, some preliminary results have shown that PCT testing could act as a predictor of bacterial co-infection and be a useful marker for assessment of disease severity. Methods We studied 83 COVID-19 hospitalized patients in whom PCT was specifically ordered by attending physicians. PCT results were evaluated according to the ability to accurately predict bacterial co-infections and death in comparison with other known biomarkers of infection and with major laboratory predictors of COVID-19 severity. Results Thirty-three (39.8%) patients suffered an in-hospital bacterial co-infection and 44 (53.0%) patients died. In predicting bacterial co-infection, PCT showed a relatively low accuracy (area under receiver-operating characteristic [ROC] curve [AUC]: 0.757; 95% confidence interval [CI]: 0.651–0.845), with a strength for detecting the outcome not significantly different from that of white blood cell count and C-reactive protein (CRP). In predicting patient death, PCT showed an AUC of 0.815 (CI: 0.714–0.892), not better than those of other more common laboratory tests, such as blood lymphocyte percentage (AUC: 0.874, p=0.19), serum lactate dehydrogenase (AUC: 0.860, p=0.47), blood neutrophil count (AUC: 0.845, p=0.59), and serum albumin (AUC: 0.839, p=0.73). Conclusions Procalcitonin (PCT) testing, even when appropriately ordered, did not provide a significant added value in COVID-19 patients when compared with more consolidated biomarkers of infection and poor clinical outcome. Abstract: The aim of this work was to evaluate the effects of increasing doses of mineral and organic fertilizers on the root chemotaxis of Avena strigosa and avoidance of Folsomia candida. Five soil samples were used for the mineral fertilization with NPK and other five soil samples were used for the organic fertilization with poultry litter-based organic fertilizer. The experiment included control soils (without applications) and soils containing 4-, 8-, 16- and 32-ton fertilizer per ha. A. strigosa was planted and cultivated in the soils for 14, 21, 28 and 35 days and avoidance behavior of F. candida. The root amounts in the soils containing mineral fertilizer were higher than those of the soils containing poultry litter-based organic fertilizer. The avoidance behavior of F. candida from the soils containing poultry litter-based organic fertilizer increased from 8- to 32-ton ha-1. Applications of high poultry litter-based organic fertilizer doses in soil increase the toxic effects for A. strigosa and F. candida.

Abstract: We compared the effects of a nitrogen-containing bisphosphonate (N-BP), zoledronic acid (ZA), and an anti-mouse RANKL antibody (anti-mRANKL Ab) on the bone tissue pathology of a transgenic mouse model of human fibrous dysplasia (FD). For comparison, we also reviewed the histological samples of a child with McCune–Albright syndrome (MAS) treated with Pamidronate for 3 years. EF1α-GsαR201C mice with FD-like lesions in the tail vertebrae were treated with either 0.2 mg/kg of ZA at day 0, 7, and 14 or with 300 μg/mouse of anti-mRANKL Ab at day 0 and 21. All mice were monitored by Faxitron and histological analysis was performed at day 42. ZA did not affect the progression of the radiographic phenotype in EF1α-GsαR201C mice. FD-like lesions in the ZA group showed the persistence of osteoclasts, easily detectable osteoclast apoptotic activity and numerous “giant osteoclasts”. In contrast, in the anti-mRANKL Ab-treated mice, osteoclasts were markedly reduced/absent, the radiographic phenotype reverted and the FD-like lesions were extensively replaced by newly formed bone. Numerous “giant osteoclasts” were also detected in the samples of the child with MAS. This study supports the hypothesis that osteoclasts per se, independently of their resorptive activity, are essential for development and expansion of FD lesions.

Abstract: Medulloblastoma (MB) is the most common malignant brain tumor in children. It is currently classified in four main molecular subgroups with different clinical outcomes: sonic hedgehog, wingless, group 3, and group 4 (MBSHH, MBWNT, MBGRP3, or MBGRP4). Presently, a 22-gene expression panel has been efficiently applied for molecular subgrouping using nCounter technology. In this study, formalin-fixed, paraffin-embedded samples from 164 Brazilian medulloblastomas were evaluated, applying the 22-gene panel, and subclassified into the low and high expression of nine key medulloblastoma-related genes. In addition, TP53 mutation status was assessed using TruSight Tumor 15 Panel, and its correlation with expression and prognostic impact was evaluated. Samples from 149 of 164 patients (90%) were classified into MBSHH (47.7%), MBWNT (16.1%), MBGRP3 (15.4%), and MBGRP4 (20.8%). GNAS presented the highest expression levels, with higher expression in MBSHH. TP53, MYCN, SOX2, and MET were also up-regulated in MBSHH, whereas PTEN was up-regulated in MBGRP4. GNAS, TP53, and PTEN low expression was associated with the unfavorable patient outcome only for MBSHH (P = 0.04, P = 0.01, and P = 0.02, respectively). TP53 mutations were detected in 28.57% of MBSHH cases and exhibited association with lower expression and worse clinical outcome, although not statistically significant. The 22-gene panel for molecular classification of medulloblastoma associated with the expression of GNAS, TP53, and PTEN improves the patient prognostication in MBSHH subgroup and can be easily incorporated in the 22-gene panel without any additional costs.

Abstract: Leukocyte- and platelet-rich fibrin (L-PRF) is an autologous platelet concentrate rich in growth factors and plasma proteins, obtained by centrifugation of patient whole blood, and widely used in oral surgery. This report describes a case of alveolar ridge preservation with L-PRF membranes. Postextractive alveolar healing was then assessed through a histologic and histomorphometric analysis. A patient requiring tooth extraction and subsequent implant rehabilitation was treated with simple extraction and socket filling with L-PRF membranes. Implant placement was performed at 3 months, and a bone biopsy was obtained for histomorphometric analysis. Histological examination of the grafted sites showed that the use of L-PRF could achieve good results in terms of bone dimension and quality and soft tissue healing. The results of this study support the use of L-PRF membranes to preserve hard and soft tissues after tooth extraction.

Abstract: Objectives Recently, defects in the protein kinase mTOR (mammalian target of rapamycin) and its associated pathway have been correlated with hemimegalencephaly (HME). mTOR acts as a central regulator of important physiological cellular functions such as growth and proliferation, metabolism, autophagy, death, and survival. This study was aimed at identifying specific variants in mTOR signaling pathway genes in patients diagnosed with HME. Methods Using amplicon and whole exome sequencing (WES) of resected brain and paired blood samples from five HME patients, we were able to identify pathogenic mosaic variants in the mTOR pathway genes MTOR, PIK3CA, and DEPDC5. Results These results strengthen the hypothesis that somatic variants in PI3K‐Akt‐mTOR pathway genes contribute to HME. We also describe one patient presenting with a pathogenic variant on DEPDC5 gene, which reinforces the role of DEPDC5 on cortical structural changes due to mTORC1 hyperactivation. These findings also provide insights into when in brain development these variants occurred. An early developmental variant is expected to affect a larger number of cells and to result in a larger malformation, whereas the same variant occurring later in development would cause a minor malformation. Significance In the future, numerous somatic variants in known or new genes will undoubtedly be revealed in resected brain samples, making it possible to draw correlations between genotypes and phenotypes and allow for a genetic clinical diagnosis that may help to predict a given patient's outcome.

Abstract: Here, we describe differences in the establishment of the leaf limb area and morphology of peach tree attributed to different seasonal conditions at a high-altitude tropical climate. The cultivars ‘Tropic Beauty’, ‘BRS Kampai’ and ‘BRS Rubimel’ were cultivated in a production area located in Ervália – MG, Brazil. A series of experiments is being carried out in this area since 2016, and the data collection for the present report was carried out in the 2018 productive cycle, when the plants were 4 years old. Fully expanded leaves were collected from ‘brindilla’ type (with same flushes vegetative growth) branches according to the period of the year. Leaves were collected for two seasons in year, August (first flushes) and December (second flushes) 2018. Three hundred leaves from each cultivar at each season were collected randomly as representative of the canopy in its middle portion. They were separated into ten replicates and evaluated individually for length, width and leaf area. All cultivars exhibited cyclic and significant differences in length, width and leaf area attributed to the environmental conditions, where and when they were developed. Further investigation is needed regarding this differential leaf growth and its impact on the production of peach trees grown in tropical highland regions.

Abstract: Aim The aim of this study was to analyze, by the aid of microbiological analysis and the field emission scanning electron microscopical (FE-SEM) analysis, the role of high-density polytetrafluoroethylene (d-PTFE) membranes in avoiding the microbial colonization of a nanocrystalline hydroxyapatite (nc-HA) bone graft and the involvement of this colonization in the healing process. Materials and methods Six patients underwent extraction of unrecoverable teeth, and a socket preservation technique was carried out with nc-HA synthetic bone graft and then covered with a d-PTFE membrane. After 28 days from surgery, FE-SEM analysis and BioTimer assay technique to assess the microbiological count of streptococci species were carried out. Data were collected and analyzed by the Student's t test (confidence interval: 95%). Results The mean amount of bacteria measured on the upper side of the membrane was 6.52 ± 0.50 CFU, while on the lower side, it was 6.59 ± 0.40 CFU. Significant differences were not found between the two sides of the membrane or between the different sectors (p > 0.05). The FE-SEM analysis revealed structured biofilms on both sides of the membrane: species of cocci, bacilli, and fusobacteria were recognizable in occasional settled vegetations. Conclusion Since the amount of bacteria found was low, the improved impermeability of the d-PTFE membrane permitted the healing process to proceed uneventful and without signs of infection or inflammation. Clinical relevance The infection of the graft site could lead to a failure of the socket preservation technique which could delay or compromise the rehabilitation following procedures. The use of d-PTFE can improve the bone regeneration thanks to its antimicrobial properties.

Abstract: Objectives People with schizophrenia (SCZ) present serious and generalised deficits in social cognition (SC), which affect negatively patients' functioning and treatment outcomes. The genetic background of SC has been investigated in disorders other than SCZ providing weak and sparse results. Thus, our aim was to explore possible genetic correlates of SC dysfunctions in SCZ patients with a genome-wide study (GWAS) approach. Methods We performed a GWAS meta-analysis of data coming from two cohorts made of 242 and 160 SCZ patients, respectively. SC was assessed with different tools in order to cover its different domains. Results We found GWAS significant association between the TMEM74 gene and the patients' ability in social inference as assessed by The Awareness of Social Inference Test; this association was confirmed by both SNP-based analysis (lead SNP rs3019332 p-value = 5.24 × 10-9) and gene-based analysis (p-value = 1.09 × 10-7). Moreover, suggestive associations of other genes with different dimensions of SC were also found. Conclusions Our study shows for the first time GWAS significant or suggestive associations of some gene variants with SC domains in people with SCZ. These findings should stimulate further studies to characterise the genetic underpinning of SC dysfunctions in SCZ.

Abstract: Morus nigra L., popularly known as blackberry, mulberry or blackberry bush, is a plant species widely used by folk medicine. Although there are few scientific studies that denote its therapeutic features, an important highlight of black mulberry is the recurrent use of tea leaves in popular environment for treatment of climacteric because it is considered a source of substances with estrogenic activity, which, if deficient, decreases osteoblastic action. Thus, the objective of the present study was to analyze the effect of the infusion of the leaves of M. nigra L. on bone repair experimental models. Twenty female Wistar rats (Rattus norvegicus), ∼ 100 days old, were submitted to fracture with bone loss in the left fibula. After that, they ingested tea leaves of M. nigra L. for 30 days without interruption. The animals suffered euthanasia after the treatment period. The results showed a significant healing of fractures in all animals that ingested the tea, when compared with other groups. In this way, it is possible to suggest that M. nigra L. tea is able to trigger a positive response in bone repair mechanism; however, it is not possible to state at what level this positive interference occurs, since mechanisms of its action have not yet been elucidated, thus requiring more specific studies on the phytotherapeutic effects of this plant.

Abstract: Acute leukemia relapsing after chemotherapy plus allogeneic hematopoietic stem cell transplantation can be treated with donor-derived T cells, but this is hampered by the need for donor/recipient MHC-matching and often results in graft-versus-host disease, prompting the search for new donor-unrestricted strategies targeting malignant cells. Leukemia blasts express CD1c antigen-presenting molecules, which are identical in all individuals and expressed only by mature leukocytes, and are recognized by T cell clones specific for the CD1c-restricted leukemia-associated methyl-lysophosphatidic acid (mLPA) lipid antigen. Here, we show that human T cells engineered to express an mLPA-specific TCR, target diverse CD1c-expressing leukemia blasts in vitro and significantly delay the progression of three models of leukemia xenograft in NSG mice, an effect that is boosted by mLPA-cellular immunization. These results highlight a strategy to redirect T cells against leukemia via transfer of a lipid-specific TCR that could be used across MHC barriers with reduced risk of graft-versus-host disease., Leukaemia therapy may benefit from the use of antigens that are less restricted to individual donors. Here the authors engineered T cells with a TCR specific for a CD1c restricted lipid leukaemia antigen and show that they can protect against disease progression in mouse leukaemia xenograft models.

Abstract: Significant release of rare earth elements (REEs) into the environment is mainly due to active or abandoned mining sites, but their presence is globally increasing due to their use in several industrial sectors. The effects on primary producers as Raphidocelis subcapitata are still limited. This research focused on La and Ce as the two most widespread REEs that can be currently found up to hundreds of μg/L in water and wastewater. Microalgae were exposed to La and Ce for 3 days (pH = 7.8) (short-term exposure) to derive the effective concentrations inhibiting the growth on 10% (EC10) of the exposed population. EC10 values (0.5 mg/L of La and 0.4 mg/L of Ce) were used for the 28 days long-term exposure (renewal test) to observe after 7, 14, 21, and 28 days on a multi-endpoint basis microalgae growth inhibition (GI), biomarkers of stress (reactive oxygen species (ROS), superoxide dismutase (SOD), and catalase (CAT)), and bioconcentration. Results evidenced that La and Ce EC10 increased GI (day 28) up to 38% and 28%, respectively. ROS, CAT, and SOD activities showed differential responses from day 7 to day 14, 21, and 28, suggesting, in most of the cases, that La and Ce effects were counteracted (i.e., being the values at day 28 not significantly different, p > 0.05, from the relative negative controls), except for La-related ROS activities. La and Ce significantly bioconcentrated in microalgae populations up to 2- and 5-fold (i.e., at day 28 compared to day 7), in that order. Bioconcentrated La and Ce were up to 3157 and 1232 μg/g dry weight (day 28), respectively. These results suggested that low La and Ce concentrations can be slightly toxic to R. subcapitata having the potential to be bioaccumulated and potentially transferred along the food web.

In this case study, we used camera traps to assess the seasonal abundance variations of wild boar and roe deer in a 560-hectares hunting-ban beech forest (95.48%) in southern Tuscany managed for timber production. Camera-trapping sessions were performed in both early summer and autumn. The roe deer abundance index is higher in early summer, although statistically not significant, potentially affecting seedling survival. Inversely, wild boars significantly (F = 79.125; p = 0.001) increase their abundance at the local scale in autumn when probably, according to the ecology of fear, they shift their home range toward refuge areas temporary. In autumn, the high density of wild boars can reduce seed availability at the local scale. Further analyses assessing the correlation between seasonal wild ungulate abundance and forest regeneration rates are in progress, based on data recorded within and around three fenced sample plots.

Abstract: Awd, the Drosophila homologue of NME1/2 metastasis suppressors, plays key roles in many signaling pathways. Mosaic analysis of the null awdJ2A4 allele showed that loss of awd gene function blocks Notch signaling and the expression of its target genes including the Wingless (Wg/Wnt1) morphogen. We also showed that RNA interference (RNAi)-mediated awd silencing (awdi) in larval wing disc leads to chromosomal instability (CIN) and to Jun amino-terminal kinases (JNK)-mediated cell death. Here we show that this cell death is independent of p53 activity. Based on our previous finding showing that forced survival of awdi-CIN cells leads to aneuploidy without the hyperproliferative effect, we investigated the Wg expression in awdi wing disc cells. Interestingly, the Wg protein is expressed in its correct dorso-ventral domain but shows an altered cellular distribution which impairs its signaling. Further, we show that RNAi-mediated knock down of awd in wing discs does not affect Notch signaling. Thus, our analysis of the hypomorphic phenotype arising from awd downregulation uncovers a dose-dependent effect of Awd in Notch and Wg signaling.

Abstract: Despite extensive research and development of new treatments, acute myeloid leukemia (AML)-backbone therapy has remained essentially unchanged over the last decades and is frequently associated with poor outcomes. Eradicating the leukemic stem cells (LSCs) is the ultimate challenge in the treatment of AML. Emerging evidence suggests that AML remodels the bone marrow (BM) niche into a leukemia-permissive microenvironment while suppressing normal hematopoiesis. The mechanism of stromal-mediated protection of leukemic cells in the BM is complex and involves many adhesion molecules, chemokines, and cytokines. Targeting these factors may represent a valuable approach to complement existing therapies and overcome microenvironment-mediated drug resistance. Some strategies for dislodging LSCs and leukemic blasts from their protective niche have already been tested in patients and are in different phases of the process of clinical development. Other strategies, such as targeting the stromal cells remodeling processes, remain at pre-clinical stages. Development of humanized xenograft mouse models, which overcome the mismatch between human leukemia cells and the mouse BM niche, is required to generate physiologically relevant, patient-specific human niches in mice that can be used to unravel the role of human AML microenvironment and to carry out preclinical studies for the development of new targeted therapies.

Abstract: Despite great efforts, it is not known which oligomeric population of amyloid beta (Aß) peptides is the main neurotoxic mediator in Alzheimer's disease. In vitro and in vivo experiments are challenging, mainly because of the high aggregation tendency of Aß (in particular of Aß 1–42 peptide), as well as because of the dynamic and non covalent nature of the prefibrillar aggregates. As a step forward in these studies, an analytical platform is here proposed for the identification and characterization of Aß 1–42 oligomeric populations resulting from three different sample preparation protocols. To preserve the transient nature of aggregates, capillary electrophoresis is employed for monitoring the oligomerization process in solution until fibril precipitation, which is probed by transmission electron microscopy. Based on characterization studies by ultrafiltration and SDS-PAGE/Western Blot, we find that low molecular weight oligomers build up over time and form bigger aggregates (> dodecamers) and that the kinetics strongly depends on sample preparations. The use of phosphate buffer results to be more aggregating, since trimers are the smallest species found in solution, whereas monomers and dimers are obtained by solubilizing Aß 1–42 in a basic mixture. For the first time, attenuated total reflection-Fourier transform infrared spectroscopy is used to assign secondary structure to the separated oligomers. Random coil and/or α-helix are most abundant in smaller species, whereas ß-sheet is the predominant conformation in bigger aggregates, which in turn are demonstrated to be responsible for Aß 1–42 toxicity.

Abstract: Flow cytometry procedures can be used for evaluation of both spermatogenic efficiency and diagnose disorders of stallion spermatogenesis. Aims of this study were to compare two testicular sample acquisition techniques (needle aspirate-N and tissue wedge-T) and results when using flow cytometry and histology procedures. Testicular cell types were stained with acridine orange, and nine regions (R2 to R10) were identified and enumerated following acquisition by either N or T. Testes were also grouped and analyzed by size and sexual maturity (Small [immature] compared with Large [mature]) and used to determine if flow cytometry procedures could be used to detect differences. For both N and T, percentages of 2n cell types were greater in the Small than Large testes, whereas percentages of 1n cell types in N were greater in the Large than Small testes (P.05). Testicular cell types in N regions were correlated to similar T regions (r between 0.51 and 0.99; P.05) in both groups. Flow cytometry and histology scores were correlated in both groups (r between -0.95 and 0.93, P.05). There were small differences in number of testicular cell types from N and T. With both sample acquisition methods, there was discrimination between the Small and Large testes, therefore, evaluation of testicular cell types using flow cytometry procedures might have clinical applications. Results with comparison of flow cytometry to histology procedures indicate that flow cytometry can be applied clinically to identify changes in testicular cell types of stallions using a needle aspirate.

Abstract: Inhibitors of indoleamine 2,3-dioxygenase 1 (IDO1) are considered a promising strategy in cancer immunotherapy as they are able to boost the immune response and to work in synergy with other immunotherapeutic agents. Despite the fact that no IDO1 inhibitor has been approved so far, recent studies have shed light on the additional roles that IDO1 mediates beyond its catalytic activity, conferring new life to the field. Here we present a novel class of compounds originated from a structure-based virtual screening made on IDO1 active site. The starting hit compound is a novel chemotype based on a [1,2,4]triazolo[4,3-a]pyridine scaffold, so far underexploited among the heme binding moieties. Thanks to the rational and in silico-guided design of analogues, an improvement of the potency to sub-micromolar levels has been achieved, with excellent in vitro metabolic stability and exquisite selectivity with respect to other heme-containing enzymes.

Abstract: There is currently an increased demand for foodstuffs that are classified as gluten-free including beer. Beer produced using gluten-free grains has a distinct flavor profile that differs greatly from that of beer produced from gluten-containing grains. The chemical difference between beers made from these two different grain sources has been explored and some key differences have been identified. Here malt sources containing gluten (barley) and malt without gluten (sorghum) were used to determine which compounds are statistically different based upon their concentrations. A total of 14 (7 barley and 7 sorghum) small-batch beers were made from malt extract. The aroma profile was sampled using SPME with chemical separation and identification and quantification using GC-MS. As expected, the differences were not the result of unique compounds but compounds present in differing amounts. A total of 17 compounds were found to be present in beer brewed from both extracts but in amounts that were highly significantly different.

Abstract: Objective A major challenge that limits understanding and treatment of epileptic events from mesial temporal structures comes from our inability to detect and map interictal networks reproducibly using scalp electrodes. Here, we developed a novel approach to map interictal spike networks and demonstrate their relationships to seizure onset and lesions in patients with foramen ovale electrode implantations. Methods We applied the direct Directed Transfer Function to reveal interictal spike propagation from bilateral foramen ovale electrodes on 10 consecutive patients and co-registered spatially with both seizure onset zones and temporal lobe lesions. Results Highly reproducible, yet unique interictal spike networks were seen for each patient (correlation: 0.93 ± 0.13). Interictal spikes spread in both anterior and posterior directions within each temporal lobe, often reverberating between sites. Spikes propagated to the opposite temporal lobe predominantly through posterior pathways. Patients with structural lesions (N = 4), including tumors and sclerosis, developed reproducible spike networks adjacent to their lesions that were highly lateralized compared to patients without lesions. Only 5% of mesial temporal lobe spikes were time-locked with scalp electrode spikes. Our preliminary observation on two lesional patients suggested that along with lesion location, Interictal spike networks also partially co-registered with seizure onset zones suggesting interrelationship between seizure onset and a subset of spike networks. Conclusions This is the first demonstration of patient-specific, reproducible interictal spike networks in mesial temporal structures that are closely linked to both temporal lobe lesions and seizure onset zones. Significance Interictal spike connectivity is a novel approach to map epileptic networks that could help advance invasive and non-invasive epilepsy treatments.

Abstract: Base Editors are emerging as an innovative technology to introduce point mutations in complex genomes. So far, the requirement of an NGG Protospacer Adjacent Motif (PAM) at a suitable position often limits the editing possibility to model human pathological mutations in animals. Here we show that, using the CBE4max-SpRY variant recognizing the NRN PAM sequence, we could introduce point mutations for the first time in an animal model and achieved up to 100% efficiency, thus drastically increasing the base editing possibilities. With this near PAM-less base editor we could simultaneously mutate several genes and developed a co-selection method to identify the most edited embryos based on a simple visual screening. Finally, we applied our method to create a new zebrafish model for melanoma predisposition based on the simultaneous editing of multiple genes. Altogether, our results considerably expand the Base Editor application to introduce human disease-causing mutations in zebrafish.

Abstract: Bacterial nutrition is a fundamental aspect of pathogenesis. While the host environment is in principle nutrient-rich, hosts have evolved strategies to interfere with nutrient acquisition by pathogens. In turn, pathogens have developed mechanisms to circumvent these restrictions. Changing the availability of bioavailable metal ions is a common strategy used by hosts to limit bacterial replication. Macrophages and neutrophils withhold iron, manganese, and zinc ions to starve bacteria. Alternatively, they can release manganese, zinc, and copper ions to intoxicate microorganisms. Metals are essential micronutrients and participate in catalysis, macromolecular structure, and signalling. This review summarises our current understanding of how central carbon metabolism in pathogens adapts to local fluctuations in free metal ion concentrations. We focus on the transcriptomics and proteomics data produced in studies of the iron-sparing response in Mycobacterium tuberculosis , the etiological agent of tuberculosis, and consequently generate a hypothetical model linking trehalose accumulation, succinate secretion and substrate-level phosphorylation in iron-starved M. tuberculosis . This review also aims to highlight a large gap in our knowledge of pathogen physiology: the interplay between metal homeostasis and central carbon metabolism, two cellular processes which are usually studied separately. Integrating metabolism and metal biology would allow the discovery of new weaknesses in bacterial physiology, leading to the development of novel and improved antibacterial therapies.

Abstract: Eradicating the malignant stem cell is the ultimate challenge in the treatment of leukaemia. Leukaemic stem cells (LSC) hijack the normal haemopoietic niche, where they are mainly protected from cytotoxic drugs. The anti-leukaemic effect of L-asparaginase (ASNase) has been extensively investigated in acute lymphoblastic leukaemia, but only partially in acute myeloid leukaemia (AML). We explored the susceptibility of AML-LSC to ASNase as well as the role of the two major cell types that constitute the bone marrow (BM) microenvironment, i.e., mesenchymal stromal cells (MSC) and monocytes/macrophages. Whilst ASNase was effective on both CD34+ CD38+ and CD34+ CD38- LSC fractions, MSC and monocytes/macrophages partially counteracted the effect of the drug. Indeed, the production of cathepsin B, a lysosomal cysteine protease, by BM monocytic cells and by AML cells classified as French-American-British M5 is related to the inactivation of ASNase. Our work demonstrates that, while MSC and monocytes/macrophages may provide a protective niche for AML cells, ASNase has a cytotoxic effect on AML blasts and, importantly, LSC subpopulations. Thus, these features should be considered in the design of future clinical studies aimed at testing ASNase efficacy in AML patients.

Abstract: Background Recent studies suggest that microRNAs (miRNAs) can participate in depression pathogenesis by altering a host of genes that are critical in corticolimbic functioning. The present study focuses on examining whether alterations in the miRNA network in the amygdala are associated with susceptibility or resiliency to develop depression-like behavior in rats. Methods Amygdala-specific altered miRNA transcriptomics were determined in a rat depression model following next-generation sequencing method. Target prediction analyses (cis- and trans) and qPCR-based assays were performed to decipher the functional role of altered miRNAs. miRNA-specific target interaction was determined using in vitro transfection assay in neuroblastoma cell line. miRNA-specific findings from the rat in vivo model were further replicated in postmortem amygdala of major depressive disorder (MDD) subjects. Results Changes in miRNome identified 17 significantly upregulated and 8 significantly downregulated miRNAs in amygdala of learned helpless (LH) compared with nonlearned helpless rats. Prediction analysis showed that the majority of the upregulated miRNAs had target genes enriched for the Wnt signaling pathway. Among altered miRNAs, upregulated miR-128-3p was identified as a top hit based on statistical significance and magnitude of change in LH rats. Target validation showed significant downregulation of Wnt signaling genes in amygdala of LH rats. A discernable increase in expression of amygdalar miR-128-3p along with significant downregulation of key target genes from Wnt signaling (WNT5B, DVL, and LEF1) was noted in MDD subjects. Overexpression of miR-128-3p in a cellular model lead to a marked decrease in the expression of Dvl1 and Lef1 genes, confirming them as validated targets of miR-128-3p. Additional evidence suggested that the amygdala-specific diminished expression of transcriptional repressor Snai1 could be potentially linked to induced miR-128-2 expression in LH rats. Furthermore, an amygdala-specific posttranscriptional switching mechanism could be active between miR-128-3p and RNA binding protein Arpp21 to gain control over their target genes such as Lef1. Conclusion Our study suggests that in amygdala a specific set of miRNAs may play an important role in depression susceptibility, which could potentially be mediated through Wnt signaling.

Abstract: Summary Bacterial nutrition is an essential aspect of host–pathogen interaction. For the intracellular pathogen Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis in humans, fatty acids derived from lipid droplets are considered the major carbon source. However, many other soluble nutrients are available inside host cells and may be used as alternative carbon sources. Lactate and pyruvate are abundant in human cells and fluids, particularly during inflammation. In this work, we study Mtb metabolism of lactate and pyruvate combining classic microbial physiology with a ‘multi‐omics’ approach consisting of transposon‐directed insertion site sequencing (TraDIS), RNA‐seq transcriptomics, proteomics and stable isotopic labelling coupled with mass spectrometry‐based metabolomics. We discovered that Mtb is well adapted to use both lactate and pyruvate and that their metabolism requires gluconeogenesis, valine metabolism, the Krebs cycle, the GABA shunt, the glyoxylate shunt and the methylcitrate cycle. The last two pathways are traditionally associated with fatty acid metabolism and, unexpectedly, we found that in Mtb the methylcitrate cycle operates in reverse, to allow optimal metabolism of lactate and pyruvate. Our findings reveal a novel function for the methylcitrate cycle as a direct route for the biosynthesis of propionyl‐CoA, the essential precursor for the biosynthesis of the odd‐chain fatty acids., Mycobacterium tuberculosis assimilates lactate and pyruvate utilising glyoxylate shunt and methylcitrate cycle, two pathways traditionally associated with fatty acid metabolism in bacteria. Surprisingly, we found that M. tuberculosis synthesises propionyl‐CoA by a reverse methylcitrate cycle and another partially characterised (in this study) pathway. Our results point to a re‐interpretation of the mechanistic role and utilisation of these pathways by Mtb during infection, i.e., their potential reversibility and importance during lactate and pyruvate metabolism in vivo.

Abstract: espanolEl trigo es uno de los cereales de mayor importancia en el mundo para el consumo humano, por lo que sus enfermedades fungicas y su control representan un gran desafio. Buscando alternativas de control biologico,se realizo un experimento con el objetivo deevaluar los efectos de diferentes dosis del formulado comercial a base de Trichoderma harzianum, sobre la calidad fisiologica y sanitaria de las semillas. Se utilizaron dos metodos de siembra: Blotter test y siembra en almacigo. Se aplico un diseno completamente al azar, con cinco tratamientos y ocho repeticiones de 50 semillas, totalizando 400 semillas, y con tres diferentes dosis del producto (100, 200, 300 ml/100 kg de semilla), mas el testigo absoluto y el quimico (Carbendazin + Thiram). Los resultadosfueron sometidos al ANAVA y al test de Tukey (α= 0,05). Los taxones de hongos identificados en las semillas de trigo fueron Rhizopusspp. y Aspergillus flavus; en la muestra testigoprevalecieron hongos del genero Rhizopus, mientras que las aplicaciones de diferentes dosis de T. harzianumpermitieron la disminucion de sus colonias. Segun el nivel de control, fueron satisfactorias las tres dosis del formulado comercial utilizado, ademas de influenciar positivamente el porcentaje de germinacion de las semillas con relacion al testigo. El mayor indice de velocidad de emergencia fue observado con la aplicacion de T. harzianuma una dosis de 300 ml/100 kg de semilla. The effects of different doses of the Trichoderma harzianum-based commercially formulated product on physiological and sanitary quality of the wheatseeds were evaluated. Two different planting methods were employed: Blotter test and sowing in seedbed. A completely random design was applied, with five treatments with eight repetitions of 50 seeds, reaching a total of 400 seeds, and with three different doses of the product (100, 200, 300 ml/100 kg seeds), in addition to the absolute control and the chemical (Carbendazin + Thiram). The results were subjected to the ANAVA and the Tukey tests (α=0.05). The fungal taxa identified in the wheat seeds were Rhizopusspp. and Aspergillus flavus; fungi if the genus Rhizopusprevailed in the control sample, while the application of different doses of T. harzianumresulted in a decrease of their colonies. Depending on the control level, the three doses of the commercial formulation applied were satisfactory, in addition to positively influencing the germination percentage of the seeds in relation to the control. Abstract: The objective of this study was to evaluate whether the antiparasitic drug eprinomectin causes hepatic oxidative stress and impairment of hepatic energetic metabolism in the silver catfish (Rhamdia quelen), in conditions of experimental aquatic contamination. The fish were exposed for 24 and 48 h at various concentrations of eprinomectin (0.0, 1.124, 1.809 and 3.976 μg L−1), followed by 48 h of recovery in eprinomectin-free water. We measured levels of reactive oxygen species (ROS) and lipoperoxidation (LOOH) in hepatic tissue, as well as levels of antioxidant capacity against peroxyl radicals (ACAP), and activities of the antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx) and glutathione S-transferase (GST), in addition to the activities of enzymes involved in energy metabolism, adenylate kinase (AK) and pyruvate kinase (PK). After 24 h, hepatic ROS levels increased in fish exposed to all concentrations of eprinomectin, and the same occurred with LOOH at the two highest concentrations of eprinomectin. ACAP levels decreased after 24 h of exposure to the highest concentration, also occurring with SOD activity at the two highest concentrations. Glutathione enzymes activities (GPx and GST) decreased after 48 h of exposure to the highest concentration. Fish transfer to eprinomectin-free water, with re-analysis after 48 h post-recovery, was unable to reverse ROS and LOOH increases, likewise with the reductions of ACAP, SOD, GPx and GST, especially at the two highest concentrations. AK and PK activities decreased after 48 h of exposure, AK decreased at the two highest concentrations and PK decreased at the highest concentration. AK activity remained lower after 48 h in eprinomectin-free water. PK activity returned to normal after 48 h in eprinomectin-free water. In summary, silver catfish exposed to aquatic contamination with the antiparasitic drug eprinomectin showed hepatic oxidative stress, possibly related to impairment of the phosphotransfer network and energy metabolism.

Abstract: SUMMARY In order to evaluate the effects of a symbiotic on 1–42-d-old broiler chicks, performance and carcass yield (CY), as well as litter concentrations of phosphorus and nitrogen were measured in a trial using 640 male chicks (Cobb 500). Birds were randomly distributed into 4 treatments with 8 replicates (pens) of 20 birds each. The treatments were different levels of the symbiotic (probiotic bacteria, exogenous enzymes, and autolyzed yeast) in the diets: 0.0, 0.5, 1.0, and 1.5 kg/t, and regression analysis using orthogonal polynomial contrasts was performed. As a challenge, it simulated conditions of commercial broilers sheds. The symbiotic provides beneficial effects for the animal's performance, especially weight gain, which increased 4.3%, in the treatment with 1.5 kg/t inclusion compared with the diet without symbiotic. Inclusion of the symbiotic in the diets provided improvements in CY and breast yield. The data suggested that the tested symbiotic is a good tool for enhancing poultry performance through nutrition.

Abstract: The objective of this study was to evaluate whether antiparasitic eprinomectin may be an environmental contaminant in water compartment in low concentrations, negatively affecting neurotransmission and, consequently, the natural behavior of the jundia (Rhamdia quelen). Fish were randomly allocated in tanks and exposed for 24 and 48 h to eprinomectin concentrations in water [0.0 (Control), 1.124 (T1), 1.809 (T2) and 3.976 (T3) μg L−1], followed by 48 h of recovery in eprinomectin-free water, in order to evaluate the behavioral parameters, levels of reactive oxygen species (ROS) in the brain, as well as cerebral enzymatic activities of acetylcholinesterase (AChE) and of the sodium-potassium ATPase pump (Na+/K+-ATPase). Especially at the two highest concentrations of eprinomectin (T2 and T3), the fish showed alterations in natural behavior, particularly hyperlocomotion and longer time on the surface. Furthermore, at these same concentrations, cerebral ROS levels increased and cerebral AChE activity decreased. At the highest concentration (T3) cerebral Na+/K+-ATPase activity was reduced. Increased ROS and impairment of AChE and Na+/K+-ATPase enzymes in the brain may have contributed directly to behavioral changes, due to neuronal damage and synapse impairment. Even after 48 h in water without eprinomectin, behavioral changes and neurotoxic effects were observed in fish, suggesting residual effects of the antiparasitic. In conclusion, eprinomectin even in low concentrations may be a hazardous environmental contaminant for aquatic organisms, as it causes brain damage and affects the natural behavior of fish.

Abstract: Influenza A virus (IAV) infection is a recognized cause of acute respiratory disease in pigs that can culminate in the decline of performance due to increasing feed conversion and costs of antimicrobial drugs to control secondary infections. Biosecurity practices are the key to prevent transmission of highly contagious agents. The aim of this study was to assess the effect of biosecurity practices on IAV seroprevalence through a cross-sectional study carried out in 404 sows from 21 herds. An indirect ELISA was used to detect antibodies against a nucleoprotein of IAV. To evaluate IAV subtypes (H1N1pdm09, H1N2 and H3N2), all samples positive by ELISA were tested using the hemagglutination inhibition assay (HI). Prevalence ratios (PR) estimates were calculated using multivariate Poisson regression accounted with survey weights. Sixty-four percent (261/404) of sows were positive in the rNP-ELISA and the estimated prevalence was 63.9% (95% CI 55%–73%). All farms had at least one seropositive sow; the frequency of IAV subtypes found in seropositive sows was 51.9% for H1N1pdm09, 38.1% for codetection H1N1pdm09 and H1N2, 8.6% for H1N2, and 0.6% for codetection H1N1pdm09 and H3N2, and 19 herds presented coinfection of H1N1 pdm09 and H1N2. Variables significantly associated with IAV seroprevalence found in the final model were ‘bird-proof net’ (PR = 0.75; 95% CI: 0.65–0.86) and ‘gilt acclimatization unit’ (PR = 0.57, 95% CI: 0.50–0.66), showing a protective effect against IAV seroprevalence, and ‘external replacement’, which had a positive effect on IAV seroprevalence (PR = 1.38, 95% CI: 1.17–1.64). This study suggests that preventing contact among wild species and swine and using an adaptation area for animals before entry into the herd can be strategies to control the influenza virus in breeding herds.

Abstract: Purpose Primary breast and prostate cancers can be cured, but metastatic disease cannot. Identifying cell factors that predict metastatic potential could guide both prognosis and treatment. Methods We used Cell-SELEX to screen an RNA aptamer library for differential binding to prostate cancer cell lines with high vs. low metastatic potential. Mass spectroscopy, immunoblot, and immunohistochemistry were used to identify and validate aptamer targets. Aptamer properties were tested in vitro, in xenograft models, and in clinical biopsies. Gene expression datasets were queried for target associations in cancer. Results We identified a novel aptamer (Apt63) that binds to the beta subunit of F1Fo ATP synthase (ATP5B), present on the plasma membrane of certain normal and cancer cells. Apt63 bound to plasma membranes of multiple aggressive breast and prostate cell lines, but not to normal breast and prostate epithelial cells, and weakly or not at all to non-metastasizing cancer cells; binding led to rapid cell death. A single intravenous injection of Apt63 induced rapid, tumor cell-selective binding and cytotoxicity in MDA-MB-231 xenograft tumors, associated with endonuclease G nuclear translocation and DNA fragmentation. Abstract: The European distribution of Echinococcus multilocularis has been reported to be expanding east and north, whereas its southern limits are deemed stable. During a study on Echinococcus granulosus s.l. infections in wolves and dogs in the Italian Maritime Alps, we unexpectedly detected the presence of E. multilocularis eggs in four fecal samples from at least two shepherd dogs, and in five wolf fecal samples. This finding, in an area about 130 km south of the southernmost E. multilocularis report in the Alps, may suggest a rapid expansion southward. While infections in foxes are currently being investigated, these data seem to indicate the potential for a new E. multilocularis endemic area. If this will be confirmed, the implementation of surveillance programs in wild and domestic canids and preventative measures will become a priority., Graphical abstract Image 1, Highlights • We detected Em infections in wolves and shepherd dogs 130 km south of the southern-most case reported so far in the European Union. • Em infections in wolves and dogs in this area may indicate a range expansion and the establishment of a new endemic area. • This area is along an ecological corridor connecting the Alps to the Apennines, with the potential for further expansion. • It is priority to implement surveillance on the local fox population, and inform practitioners of the increased risk.
