There are 130 described species in the genus Mycodrosophila Oldenberg, 1914, distributed across all biogeographic regions. Most of these species show essential mycophagy. Currently, ten species are known from the Neotropical Region, nine of which are found in the Amazon, Atlantic Forest, Cerrado and Pampa biomes of Brazil. In this study, we describe the adult external morphology and structures of male and/or female terminalia for two new species from the Amazon Biome of Brazil. In addition, we propose a new species group, the Mycodrosophila neoprojectans group, encompassing the two new species described here, together with three previously described species from the Neotropics.

The aim of this study was to evaluate the relationship between temperament in Nellore bulls with carcass and meat quality traits. In total, 1,400 bulls were studied, and temperament was assessed using two measurements: movement score (MOV) and flight speed test (FS). Both MOV and FS were measured at two time points, with background (MOVb and FSb) temperament measured at yearling age, ~550 d after birth, and the preslaughter (MOVps and FSps) temperament measured at the end of the feedlot period. The change of temperament resulting in an increase or decrease in reactivity was also used to measure meat quality. The traits used to define carcass and meat quality included carcass bruises (BRU), hot carcass weight (HCW, kg), ribeye area (REA, cm2), backfat thickness (BFT, cm), marbling score (MS), meat pH after thawing (pH), presence or absence of dark cutters, color parameters of luminosity (L*), redness (a*) and yellowness (b*), cooking loss (CL, %), and Warner–Bratzler shear force (WBSF, kg). A principal component (PC) analysis was initially applied to the carcass and meat quality traits, followed by logistic regression models and linear mixed models to evaluate the effects of temperament on carcass and meat quality. The risks of carcass bruises and dark cutters did not differ as a function of any temperament trait (P > 0.05). In turn, animals classified as high MOVb (reactive) had lower PC3 values (P = 0.05), CL (P = 0.02), and tended to have lower MS (P = 0.08). In addition, animals classified as high FSb (faster and reactive cattle) produced carcasses with smaller REA (P < 0.01), higher meat pH (P < 0.01), lower color gradients (L*, P = 0.04; b*, P < 0.01), and lower PC1 and PC4 scores (P < 0.01) when compared with the low FSb class. For preslaughter temperament, high MOVps was related to lower color a* (P = 0.04), whereas high FSps was related to lower HCW, MS, and PC2 (P < 0.01) than the calmer ones (low FSps). The reduction in MOV was related to more tender meat, and the reduction in FS to heavier carcass and brighter meat. We conclude that excitable temperament in Nellore cattle may have negative effects in some of the carcass and meat quality attributes assessed, mainly those related to muscle deposition on carcass and color gradients. Measurement of temperament before the cattle entered the feedlot was a better predictor of carcass and meat quality traits, compared with temperament assessment at the end of the feeding period.

This medium is used for the growth of Erwinia/Pectobacterium species to ensure that virulence genes are turned ON. These bacteria turn on virulence (plant cell-wall degrading enzymes = PCWDEs) in response to various environmental signals. One of these signals is of plant origin, and it is thought to allow the bacterium to determine whether it has located a potential plant host. These plant signals are sensed by the bacterial sensor/regulator KdgR.

The severe acute respiratory syndrome coronavirus 2 responsible for COVID-19 remains a persistent threat to mankind, especially for the immunocompromised and elderly for which the vaccine may have limited effectiveness. Entry of SARS-CoV-2 requires a high affinity interaction of the viral spike protein with the cellular receptor angiotensin-converting enzyme 2. Novel mutations on the spike protein correlate with the high transmissibility of new variants of SARS-CoV-2, highlighting the need for small molecule inhibitors of virus entry into target cells. We report the identification of such inhibitors through a robust high-throughput screen testing 15,000 small molecules from unique libraries. Several leads were validated in a suite of mechanistic assays, including whole cell SARS-CoV-2 infectivity assays. The main lead compound, Calpeptin, was further characterized using SARS-CoV-1 and the novel SARS-CoV-2 variant entry assays, SARS-CoV-2 protease assays and molecular docking. This study reveals Calpeptin as a potent and specific inhibitor of SARS-CoV-2 and some variants.

Training was divided in two phases (P1 and P2): habituation to humans (P1), and to the milking parlor and human physical contact (P2). Flight distances (FD) were assessed during P1, and numbers of steps and kicks (as binary) were recorded during P2. The last two measurements and milking reactivity (scored from 1 = cow stands quietly to 8 = cow is very agitated) were recorded during the first four days of milking. Repeated behavioral measurements were used to evaluate individual changes fitting a linear regression for each animal, and the b-coefficients were used to classify heifers as habituated, neutral or sensitized. The percentages of assessment days that each heifer/cow kicked during P2, and during the first four days of milking were calculated. Pearson's correlation coefficients between b-coefficients were estimated. General linear models were fitted to assess the effects of interval classes (defined by the number of days between the last day in training and the first day in miking) and behavioral responses during P1 and P2 on milking reactivity and number of steps on the first day of milking and on b-coefficients of these variables. Individual differences in behavioral responses were observed. Most heifers (∼87 %) habituated during P1 and ∼ 44 % of them reduced number of steps during P2. Almost 52 % of the heifers did not step or exhibit only one step, and ∼93 % of them did not kick when fitting the milking cluster in the first day of milking. Around 56 % of them increased the number of steps along milking assessments, while ∼33 % reduced it. The b-coefficients of FD and for the number of steps in P2 were not associated to the b-coefficients of milking assessed variables. The b-coefficient and the number of steps on the first assessment day of P2 showed a significant effect (P < 0.05) on milking reactivity scores (F = 6.06 and 6.54, respectively) and number of steps (F = 5.73 and 7.88, respectively) assessed in the first day of milking. None of the other independent variables showed significant effect on b-coefficients of milking reactivity scores and number of steps during milking. We conclude that training reduces fear to humans and cows’ reactivity to milking, and that individual differences between heifers play an important role in the training results. Poor teat and udder structure, frequently associated with older cows, impact cow production and health as well as calf morbidity and mortality. However, producer culling, for reasons including age, production, feed availability, and beef markets, creates a bias in teat (TS) and udder scores (US) assessed and submitted to the Canadian Angus Association for genetic evaluations toward improved mammary structure. In addition, due to the infancy of the reporting program, repeated scores are rare. Prior to the adoption of genetic evaluations for TS and US in Canadian Angus cattle, it is imperative to verify that TS and US from young cows are the same traits as TS and US estimated on mature cows. Genetic parameters for TS and US from all cows (n = 4,192) and then from young cows (parities 1 and 2) and from mature cows (parity ≥ 4) were estimated using a single-trait animal model. Genetic correlations for the traits between the two cow age groups were estimated using a two-trait animal model. Estimates of heritability (posterior SD [PSD]) were 0.32 (0.07) and 0.45 (0.07) for young TS and US and 0.27 (0.07) and 0.31 (0.07) for mature TS and US, respectively. Genetic correlation (PSD) between the young and mature traits was 0.87 (0.13) for TS and 0.40 (0.17) for US. Genome-wide association studies were used to further explore the genetic and biological commonalities and differences between the two groups. Although there were no genes in common for the two USs, 12 genes overlapped for TS in the two cow age groups. Interestingly, there were also 23 genes in common between TS and US in mature cows. Based on these findings, it is recommended that producers collect TS and US on their cow herd annually.

LTR-retrotransposons are structurally similar to retroviruses, as they possess the enzymes reverse transcriptase, Ribonuclease H, integrase, proteinase, and the gag gene and are flanked by long terminal repeats (LTRs). The 412/mdg1 lineage, belonging to the Ty3/Gypsy group, consists of the TEs 412, mdg1, stalker, pilgrim, and blood. The 412/mdg1 lineage is distinguished from the others in the gypsy group in that it has small ORFs at the beginning of the TE and is highly similar to the pol ORF among the TEs that make up the lineage. In this study, our aim was to elucidate the evolutionary history of the 412/mdg1 lineage in the 127 dipteran genomes available to date, and the characteristics of the sequences in each genome. We used the canonical TE 412 probe described in Drosophila melanogaster as the query. We found sequences homologous to the 412/mdg1 lineage restricted to the suborder Brachycera. These sequences are widely distributed in drosophilids but are also present in other groups of flies. We note the presence of the 412/mg1 lineage in tsetse flies (Glossina). Furthermore, our results showed an elaborate evolutionary history for the pol ORF in the 412/mdg1 lineage of the LTR-Retrotransposon.

Metabolomic phenotypes are influenced by combination of genetic and environmental effects allowing for metabolome-genome-wide association studies (mGWAS) as a powerful tool to investigate the relationship between these phenotypes and genetic variants. The objectives of this study were to estimate genomic heritability and perform mGWAS andin silicofunctional enrichment analyses for a set of plasma metabolites in Canadian crossbred beef cattle. Thirty-three plasma metabolites and 45,266 single nucleotide polymorphisms (SNPs) were available for 475 animals. Genomic heritability for all metabolites was estimated using genomic best linear unbiased prediction (GBLUP) including genomic breed composition as covariates in the model. A single-step GBLUP implemented in BLUPF90 programs was used to determine SNPPvalues and the proportion of genetic variance explained by SNP windows containing 10 consecutive SNPs. The top 10 SNP windows that explained the largest genetic variation for each metabolite were identified and mapped to detect corresponding candidate genes. Functional enrichment analyses were performed on metabolites and their candidate genes using the Ingenuity Pathway Analysis software. Eleven metabolites showed low to moderate heritability that ranged from 0.09 +/- 0.15 to 0.36 +/- 0.15, while heritability estimates for 22 metabolites were zero or negligible. This result indicates that while variations in 11 metabolites were due to genetic variants, the majority are largely influenced by environment. Functional and enduring mammary structure is pivotal for producer profitability, and animal health and welfare in beef production. Genetic evaluations for teat and udder score in Canadian Angus cattle have previously been developed. The aim of this study was to identify genomic regions associated with teat and udder structure in Canadian Angus cows thereby enhancing knowledge of the biological architecture of these traits. Thus, we performed a weighted single-step genome wide association study (WssGWAS) to identify candidate genes for teat and udder score in 1,582 Canadian Angus cows typed with the GeneSeek® Genomic Profiler Bovine 130K SNP array. Genomically enhanced estimated breeding values (GEBVs) were converted to SNP marker effects using unequal variances for markers to calculate weights for each SNP over three iterations. At the genome wide level, we detected windows of 20 consecutive SNPs that explained more than 0.5% of the variance observed in these traits. A total of 35 and 28 windows were identified for teat and udder score, respectively, with two SNP windows in common for both traits. Using Ensembl, the SNP windows were used to search for candidate genes and quantitative trait loci (QTL). A total of 94 and 71 characterized genes were identified in the regions for teat and udder score, respectively. Of these, 7 genes were common for both traits. Gene network and enrichment analysis, using Ingenuity Pathway Analysis (IPA), signified key pathways unique to each trait. Genes of interest were associated with immune response and wound healing, adipose tissue development and morphology, and epithelial and vascular development and morphology. Genetic architecture from this GWAS confirms that teat and udder score are distinct, polygenic traits involving varying and complex biological pathways, and that genetic selection for improved teat and udder score is possible.

Stimulating brown adipose tissue (BAT) activity represents a promising therapy for overcoming metabolic diseases. mTORC2 is important for regulating BAT metabolism, but its downstream targets have not been fully characterized. In this study, we apply proteomics and phosphoproteomics to investigate the downstream effectors of mTORC2 in brown adipocytes. We compare wild-type controls to isogenic cells with an induced knockout of the mTORC2 subunit RICTOR (Rictor-iKO) by stimulating each with insulin for a 30-min time course. In Rictor-iKO cells, we identify decreases to the abundance of glycolytic and de novo lipogenesis enzymes, and increases to mitochondrial proteins as well as a set of proteins known to increase upon interferon stimulation. We also observe significant differences to basal phosphorylation because of chronic RICTOR loss including decreased phosphorylation of the lipid droplet protein perilipin-1 in Rictor-iKO cells, suggesting that RICTOR could be involved with regulating basal lipolysis or droplet dynamics. Finally, we observe mild dampening of acute insulin signaling response in Rictor-iKO cells, and a subset of AKT substrates exhibiting statistically significant dependence on RICTOR.

In Arabidopsis thaliana, chromosomal rearrangements are commonly associated with compound T-DNA insertions due to endogenous double-stranded break and repair mechanisms. During the production of a double homozygous mutant for the CCCH zinc finger proteins, AtC3H6 and AtC3H68, we uncovered evidence for a reciprocal I-V chromosomal translocation in SALK_093620. Mapping the T-DNA borders in this insertion line confirmed an insertion site at the AtC3H6 locus on chromosome I and identified a second insertion in an intergenic region at the end of chromosome V. Our investigation of the progeny distribution and gamete transmission from reciprocal crosses indicated that male and female gametes are differentially affected by this predicted translocation. Male gametes with the haploid genotype AtC3H6(+);AtC3H68(+) had significantly higher transmission than female gametes with the same genotype. Our work highlights the importance of detailing T-DNA insertion events in transgenetic lines produced through Agrobacterium-mediated transformation, which can uncover novel chromosomal anomalies and imbalanced genome compositions.

Rhinoleucophenga Hendel comprises an unusual Drosophilidae (Diptera) genus with predaceous larvae, currently compounded by 29 nominal species with New World distribution. In the present study, Rhinoleucophenga brasiliensis (Costa Lima) and R. fluminensis (Costa Lima) are redescribed. These two species are commonly misidentified in Drosophilidae species inventories, mainly by the few morphological character details presented in the original taxonomic description. Thus, by the morphological review performed here, lectotype and paralectotypes designed to R. brasiliensis and R. fluminensis, as well as new morphological characters, drawings and photos (for the first time) are presented in order to avoid further taxonomic mistakes with those referred sibling species of Rhinoleucophenga.

The DNA methyltransferase 2 (DNMT2) protein is the most conserved member of the DNA methyltransferase family. Nevertheless, its substrate specificity is still controversial and elusive. The genomic role and determinants of DNA methylation are poorly understood in invertebrates, and several mechanisms and associations are suggested. In Drosophila, the only known DNMT gene is Dnmt2. Here we present our findings from a wide search for Dnmt2 homologs in 68 species of Drosophilidae. We investigated its molecular evolution, and in our phylogenetic analyses the main clades of Drosophilidae species were recovered. We tested whether the Dnmt2 has evolved neutrally or under positive selection along the subgenera Drosophila and Sophophora and investigated positive selection in relation to several physicochemical properties. Despite of a major selective constraint on Dnmt2, we detected six sites under positive selection. Regarding the DNMT2 protein, 12 sites under positive-destabilizing selection were found, which suggests a selection that favors structural and functional shifts in the protein. The search for new potential protein partners with DNMT2 revealed 15 proteins with high evolutionary rate covariation (ERC), indicating a plurality of DNMT2 functions in different pathways. These events might represent signs of molecular adaptation, with molecular peculiarities arising from the diversity of evolutionary histories experienced by drosophilids.

The willistoni species subgroup has been the subject of several studies since the latter half of the past century and is considered a Neotropical model for evolutionary studies, given the many levels of reproductive isolation and different evolutionary stages occurring within them. Here we present for the first time a phylogenetic reconstruction combining morphological characters and molecular data obtained from 8 gene fragments (COI, COII, Cytb, Adh, Ddc, Hb, kl-3 and per). Some relationships were incongruent when comparing morphological and molecular data. Also, morphological data presented some unresolved polytomies, which could reflect the very recent divergence of the subgroup. The total evidence phylogenetic reconstruction presented well-supported relationships and summarized the results of all analyses. The diversification of the willistoni subgroup began about 7.3 Ma with the split of D. insularis while D.paulistorum complex has a much more recent diversification history, which began about 2.1 Ma and apparently has not completed the speciation process, since the average time to sister species separation is one million years, and some entities of the D. paulistorum complex diverge between 0.3 and 1 Ma. Based on the obtained data, we propose the categorization of the former "semispecies" of D. paulistorum as a subspecies and describe the subspecies D. paulistorum amazonian, D. paulistorum andeanbrazilian, D. paulistorum centroamerican, D. paulistorum interior, D. paulistorum orinocan and D. paulistorum transitional.

Temperament was evaluated using the movement (MOV), tension (TEN) and crush (CS) scores (measured with animals inside the squeeze chute) as well as the flight speed (FS) and temperament score (TS). Reproductive traits included i) heifer rebreeding (HR), which evaluates heifers’ ability to become pregnant, given that they had calved once; and ii) stayability (STAY), which measures cows’ ability to calve at least 3 offspring before reaching 65 months of age. We used Bayesian inference and Gibbs sampling in a two-trait analysis to estimate genetic parameters applying a linear model for FS and threshold models for MOV, TEN, CS, TS, HR and STAY. The animal model included contemporary group as a fixed effect, direct additive genetic and residual effects as random effects, and animal age at yearling as a covariate (with linear and quadratic effects). Heritability estimates for MOV, TEN, CS, FS, TS, HR and STAY were 0.14 ± 0.04, 0.11 ± 0.03, 0.09 ± 0.03, 0.22 ± 0.02, 0.19 ± 0.04, 0.13 ± 0.02 and 0.13 ± 0.02, respectively. The genetic correlation estimates were low to moderate and the highest values (in magnitude) were − 0.19 ± 0.21 (HR-CS), − 0.21 ± 0.15 (STAY-TEN) and − 0.24 ± 0.16 (STAY-CS), indicating that the selection to improve cattle temperament does not negatively affect HR and STAY. These results indicate that all traits had sufficient genetic variability to respond to direct selection; however, given the low estimated heritability, we expect to see only long-term genetic changes. Genetic correlations showed that there is no antagonism of temperament with fertility and longevity; however, we recommend including these traits as selection criteria in Nellore breeding programs to obtain satisfactory genetic changes. During this procedure the newly calved cows usually become agitated and, in some cases, exhibit aggressive behavior toward humans, increasing the risk of accidents for both, humans and animals. In order to understand the dynamic relationships between cow, calf, and cowhand's behavior expressed in this particular situation, a conceptual model was built by using the system dynamics methodology. Three dynamic hypotheses were formulated regarding the cause-effect relationship between: i) livestock-handling quality and maternal aggressiveness; ii) livestock-handling quality and newborn calf reactivity, and; iii) newborn calf reactivity and maternal aggressiveness. The first two hypotheses indicated that the improvement (increase) in handling quality will cause an effect in the opposite direction, decreasing cow's aggressiveness and calf's reactivity towards the cowhands. The third hypothesis indicated that the greater the calf's reactivity, the greater will be the cow's aggressiveness, interconnecting cow-calf behaviors. Moreover, the cow, calf, and cowhand subsystems are also comprised of other variables such as human and animal welfare and the occurrence of labor accidents. The combination of these three subsystems constituted a complex general dynamic model highlighting the connection among all agents involved in newborn handling procedure. Despite the complexity it is possible to identify components that promote, simultaneously, human and animal welfare when carrying out newborn calves handling procedures, offering opportunities to develop and adopt system management strategies. In this way, to reduce the risks of labor accidents and the impoverishment of overall welfare it is necessary to focus on three different factors, as follow: i) human behavior, ii) animal behavior, and iii) handling facilities. Thus, it is recommended to implement human capacity-building courses, the culling of aggressive cows and the use of appropriate facilities for the handling of newborn calves. Biodiversity is believed to be low in regions with vegetation that has adapted to water stress. Additionally, there is little interest by authorities in establishing and expanding conservation units in these areas. In Brazil, the Caatinga and the Cerrado biomes comprise this xerophilous vegetation. In both, climate is tropical and the dry season is long and well-defined. The Caatinga is the Brazilian biome with the smallest area protected by conservation units. This study evaluates the efficacy of conservation units in the Caatinga biome based on abundance and richness of drosophilids. Flies were collected inside and outside these areas. In total, approximately 23,000 flies of 32 species were collected in six conservation and six non-conservation sites. Two non-described species occurred exclusively inside protected areas, underlining the importance of conservation efforts in the maintenance of biodiversity. Other species were recorded exclusively outside conservation areas, which emphasizes the importance of establishing and expanding conservation units in the Caatinga. Native species were significantly more abundant inside conservation units, though the richness was similar in protected and non-protected areas. Abundance of exotic species outside conservation areas was statistically different in comparison with that of native ones.

Rhinoleucophenga Hendel is an endemic genus of the New World with most species recorded in Brazil. Rhinoleucophenga obesa (Loew) seemed to be the most widespread species, being recorded in the United States of America, Mexico and Brazil. In the Neotropical region, identifications of R. obesa were commonly based on the description of non-type specimens determined by Costa Lima (1935) and Malogolowkin (1946), although their identities were doubtful. However, the recent redescription of R. obesa from the type-series from Texas, USA, confirmed a long period of misidentifications and a new species, R. cantareira Vilela & Bächli, was proposed based on Brazilian specimens. Thus, review of morphological and molecular traits of specimens previously identified as R. obesa in Neotropical biomes are necessary to check the identity and distribution of a group of sibling species: R. obesa, R. cantareira, R. gigantea (Thomson), R. pallida Hendel and R. pampeana Poppe et al. In the present paper, specimens previously determined as R. obesa, sampled in different Brazilian localities, were compared by morphological and molecular traits. Plates of female terminalia of R. cantareira and R. gigantea are presented for the first time. The spermathecal capsules (as well as the male epandrium) revealed useful characteristics to differentiate those species; these morphological differences were corroborated by a sequence fragment of COI. The specimens on which were based the descriptions of Rhinoleucophenga obesa sensu Costa Lima (1935) and sensu Malogolowkin (1946) were reviewed and confirmed as R. cantareira. Other Brazilian specimens from different localities, previously misidentified as R. obesa, were determined as R. gigantea or R. cantareira, with new records for both species. Therefore, the present study corresponds to the advance of identity definition and distribution of sibling species of Rhinoleucophenga commonly sampled in Neotropical inventory studies.

Transposable elements (TEs) have the main role in shaping the evolution of genomes and host species, contributing to the creation of new genes and promoting rearrangements frequently associated with new regulatory networks. Support for these hypotheses frequently results from studies with model species, and Drosophila provides a great model organism to the study of TEs. Micropia belongs to the Ty3/Gypsy group of long terminal repeats (LTR) retroelements and comprises one of the least studied Drosophila transposable elements. In this study, we assessed the evolutionary history of Micropia within Drosophilidae, while trying to assist in the classification of this TE. At first, we performed searches of Micropia presence in the genome of natural populations from several species. Then, based on searches within online genomic databases, we retrieved Micropia-like sequences from the genomes of distinct Drosophilidae species. We expanded the knowledge of Micropia distribution within Drosophila species. The Micropia retroelements we detected consist of an array of divergent sequences, which we subdivided into 20 subfamilies. Even so, a patchy distribution of Micropia sequences within the Drosophilidae phylogeny could be identified, with incongruences between the species phylogeny and the Micropia phylogeny. Comparing the pairwise synonymous distance (dS) values between Micropia and three host nuclear sequences, we found several cases of unexpectedly high levels of similarity between Micropia sequences in divergent species. All these findings provide a hypothesis to the evolution of Micropia within Drosophilidae, which include several events of vertical and horizontal transposon transmission, associated with ancestral polymorphisms and recurrent Micropia sequences diversification.

Leigh syndrome is a fatal neurometabolic disorder caused by defects in mitochondrial function. mTOR inhibition with rapamycin attenuates disease progression in a mouse model of Leigh syndrome (Ndufs4 KO mouse); however, the mechanism of rescue is unknown. Here we identify PKC downregulation as a key event mediating the beneficial effects of rapamycin treatment of Ndufs4 KO mice. Assessing the impact of rapamycin on the brain proteome and phosphoproteome of Ndufs4 KO mice we find that rapamycin restores mitochondrial protein levels, inhibits signaling through both mTOR complexes, and reduces the abundance and activity of multiple protein kinase C (PKC) isoforms. Administration of PKC inhibitors increases survival, delays neurological deficits, prevents hair loss, and decreases inflammation in Ndufs4 KO mice. Thus, PKC may be a viable therapeutic target for treating severe mitochondrial disease. Despite their heritability and influence on female productivity, there are currently no genetic evaluations for teat and udder structure in Canadian Angus cattle. The objective of this study was to develop optimal genetic evaluations for these traits in the Canadian Angus population. Guidelines recommended by Beef Improvement Federation (BIF) were used to score teat and udder structure in 1,735 Canadian Angus cows from 10 representative herds. Cows scored ranged in parity from 1 to 13; however, >70% of cows were parity ≤4. Scores ranged from 1 (large, bottle shaped) to 9 (very small) for teats and from 1 (very pendulous) to 9 (very tight) for udders. Consistent with parity distribution, >70% of teat and udder scores were ≥6. Teat and udder scores (TS9 and US9, respectively) were modeled using a multiple trait animal model with random effects of contemporary group (herd-year-season) and additive genetic effect, and fixed effects of breed, parity group, and days between calving and scoring. To test good versus poor structure, a binary classification of 1 or 2 (TS2, US2) [comprised of scores 1 to 5 = 1 (poor structure) and scores 6 to 9 = 2 (good structure)] was created. Further, to assess the impact of grouping less frequently observed poor scores, a 1 to 7 scale (TS7, US7) was created by combining teat and udder scores 1 to 3. Analyses for teat and udder scores on scales TS9, US9, TS7, US7, and TS2, US2 were compared. In addition, both threshold and linear animal models were used to estimate variance components for the traits. Data treatment and models were evaluated based on correlation of resulting estimated breeding value (EBV) with corrected phenotypes, Spearman’s rank correlation coefficient, average EBV accuracies (r), and deviance information criteria (DIC). TS9, US9 scales for teat and udder scores and linear models performed best. Estimates of heritability (SE) for teat and udder score were 0.32 (0.06) and 0.15 (0.04), respectively, indicating these traits were moderately heritable and that genetic improvement for teat and udder scores was possible. Estimates of phenotypic and genotypic correlations for teat and udder score were 0.46 (0.02) and 0.71 (0.09), respectively. Estimates of genotypic correlations with birth weight (BW), weaning weight (WW), and yearling weight (YW), ranged from −0.04 (0.10) to −0.20 (0.12), verifying the importance of selecting for improved teat and udder score as individual traits, alongside performance traits.

The Pampas biome is a subtropical–temperate grassland region that presents striking climatic characteristics, such as temperatures below zero in the winters and up to 40°C in the summers. Hence, long sampling periods are necessary to better understand the structure of Drosophilidae assemblages through current climatic conditions in the heterogeneous landscape of the Pampas. Samples of drosophilids were taken in natural and urban areas during 12 climatic periods. The environmental influence on the structure of assemblages was temporally and spatially analyzed through nonmetric multidimensional scaling, indicator value, and permutational multivariate analysis of variance. The following hypotheses were tested: 1) the structure of drosophilids assemblages vary through different habitats from the same area, and 2) the presence and abundance of Drosophilidae species in a particular environment is regulated by the interaction between the phytophysionomy and climatic traits of each locality. The Pampas assemblages were dominated by a few species, although it is a highly diverse biome with regards to Drosophilidae. The environmental heterogeneity was confirmed as a determinant factor of the structure of assemblages, and it explained 8.86% of the assemblage composition. Nonetheless, the interaction between temperature and humidity was revealed as the most influential environmental component of the Drosophilidae assemblages, and it explained 37.28% of all the recorded variation. Together, environment type and the climatic elements were able to explain 56.45% of the assemblages’ variation. Open and close environments, such as grassland and forest patches, respectively, presented a peculiar composition of Drosophilidae assemblages and should be equally considered in the Pampas preservation strategies. Resumo O bioma Pampa e uma regiao campestre subtropical–temperada que apresenta caracteristicas climaticas marcantes, invernos com temperaturas negativas e veroes com registros de mais de 40°C. Longos periodos de amostragem sao necessarios para o melhor entendimento da estrutura das assembleias de Drosophilidae na paisagem heterogenea do Pampa. Coletas de drosofilideos foram realizadas em areas naturais e urbanas do Pampa durante 12 periodos climaticos. A influencia ambiental sobre a estrutura das assembleias foi temporal e espacialmente analisada por meio de nMDS, IndVal e PERMANOVA. As seguintes hipoteses foram testadas: 1) a estrutura das assembleias de drosofilideos variam atraves de diferentes habitats de uma mesma area e, 2) a presenca e a abundância das especies de Drosophilidae em um ambiente sao reguladas pela interacao da fitofisionomia e caracteristicas climaticas de cada local. As assembleias do Pampa foram dominadas por poucas especies, embora este seja um bioma muito diverso com relacao a fauna de Drosophilidae. A heterogeneidade ambiental foi um fator determinante na estrutura das assembleias, explicando 8,86% da composicao das mesmas. No entanto, a interacao entre temperatura e os niveis de umidade foi indicada como o componente ambiental mais influente sobre as assembleias de Drosophilidae, explicando 37,28% de toda a variacao percebida. O tipo de ambiente amostrado e os componentes climaticos juntos explicaram 56,45% da variacao nas assembleias. Ambientes abertos e fechados, campos e matas, respectivamente, apresentaram uma composicao propria para as assembleias de Drosophilidae e, devem ser igualmente considerados em estrategias de preservacao do Pampa.

In this study, semi-arid environments were tested to see if they support insect diversity. This was evaluated through the structure of the composition of assemblies of drosophilids in three conservation units placed in three different ecoregions in the dryland forests, Caatinga. This is a unique biome in northeast Brazil, comprising approximately 10% of the country. Species richness was investigated over 2 years during a prolonged drought, considered the worst affliction the Caatinga ecosystem had experienced in the last 50 years. Alpha diversity indices and the ecological similarity between the samples were calculated to determine how the environments drive the composition of Drosophilidae in such semi-arid places. A total of 7352 specimens were sampled. They were classified into 20 species belonging to four genera: Drosophila, Rhinoleucophenga, Scaptodrosophila, and Zaprionus. Drosophila nebulosa Sturtevant (44.5%) and Drosophila cardini Sturtevant (12.5%) were the most abundant species. The occurrences and abundances of all the species differed greatly between sites. These results and other ecological analyses indicate that although placed in the same biome, there are great variability in the drosophilid species and abundance among the three protected and conserved dryland environments.

Drosophila willistoni is a geographically widespread Neotropical species. The genome of strain Gd-H4-1 from Guadeloupe Island (Caribbean) was sequenced in 2007 as part of the 12 Drosophila Genomes Project. The assembled scaffolds were joined based on conserved linkage and assigned to polytene chromosomes based on a handful of genetic and physical markers. This paucity of markers was particularly striking in the metacentric chromosome II, comprised two similarly sized arms, IIL and IIR, traditionally considered homologous to Muller elements C and B, respectively. In this paper we present the cytological mapping of 22 new gene markers to increase the number of markers mapped by in situ hybridization and to test the assignment of scaffolds to the polytene chromosome II arms. For this purpose, we generated, by polymerase chain reaction amplification, one or two gene probes from each scaffold assigned to the chromosome II arms and mapped these probes to the Gd-H4-1 strain’s polytene chromosomes by nonfluorescent in situ hybridization. Our findings show that chromosome arms IIL and IIR correspond to Muller elements B and C, respectively, directly contrasting the current homology assignments in D. willistoni and constituting a major reassignment of the scaffolds to chromosome II arms.

In several arthropod groups, male genitalia is the most important feature for species identification, especially in cryptic species. Cryptic species are very common in the Drosophila genus, and the Neotropical Drosophila willistoni species group is a good example. This group currently includes 24 species divided into three subgroups: alagitans, bocainensis and willistoni. There are six sibling species in the willistoni subgroup – D. willistoni, D. insularis, D. tropicalis, D. equinoxialis, D. pavlovskiana and D. paulistorum, which is a species complex composed of six semispecies – Amazonian, Andean-Brazilian, Centroamerican, Interior, Orinocan and Transitional. The objective of this study was to characterize male genitalia of the willistoni subgroup, including the D. paulistorum species complex, using scanning electron microscopy and light microscopy. We also tried to contribute to the identification of these cryptic species and to add some comments about evolutionary history, based on male genitalia characters. Despite being cryptic species, some differences were found among the siblings, including the Drosophila paulistorum semispecies. Genetic parameters for sensory traits and their associations with pH and intramuscular fat were estimated in pork chops. A univariate animal model was used to estimate the heritability of the sensory traits in ASReml using pedigree information, while phenotypic and genetic correlations were estimated using a bivariate animal model. Low heritability (h2 < 0.2) was found for all measured sensory traits, and genetic correlations estimated between sensory traits and pH were smaller than that estimated between intramuscular fat and sensory traits. Positive genetic correlations of sensory measurements with pH and intramuscular fat were found, indicating that selection to increase intramuscular pH and fat can help to increase sensory scores. However, as the genetic correlations were low to moderate, increase in pork sensory scores through selection for loin pH and intramuscular fat content would be slow, and because of the small sample size, the results should be interpreted with caution. Based on previous studies using Bos Taurus cattle, it is expected that reactivity measured at weaning could be used as a predictor of feedlot performance, and that cattle with greater flight speed (FS) would have reduced average daily gain (ADG). Aims: The aims of this study were to (1) determine if FS (m/s) measured before entry into the feedlot could be used as a predictor of cattle performance, (2) evaluate the consistency of FS over time, and (3) determine when FS should be measured to best predict animal suitability to the feedlot. Methods: A total of 54 Nellore and 40 Guzerat calves were assessed by using FS, which was collected at the time of (FS0) and 1 week after weaning (FS1), as well as on Day 28 (FS28), 56 (FS56) and 168 (FS168) after entry into the feedlot, and a FS average was calculated using FS28, FS56 and FS168 (FS28-168). Performance using ADG (kg/calf.day) was calculated for each animal over three periods: ADG1-28, ADG28-168 and ADG1-168. Bunk attendance (min/calf.day) was recorded over a 173-day feeding period. Key results: The ADG28-168 was reduced by 0.14 kg/calf.day for each unit increase in FS0 (P < 0.05). Both ADG28-168 and ADG1-168 were reduced by 0.02 kg/calf.day for each unit increase in FS1 (P ≤ 0.01), but neither FS0 nor FS1 was associated with ADG1-28 (P > 0.10). A significant increase was observed between FS1 and FS28 (P < 0.01), followed by a reduction in FS56 (P < 0.01) and FS168 (P < 0.01). Conclusion: FS measured at or within 1 week of weaning has the potential to be used as an indicator of feedlot growth performance in Nellore and Guzerat cattle. Implications: Although our results do not indicate the efficacy of FS as a predictor of early suitability of cattle to the feedlot, it does have the potential to support its recommendation as an indicator of overall feedlot growth performance in Nellore and Guzerat breeds. Damage-associated molecular patterns (DAMPs) are molecules exposed or released by dead cells that trigger or modulate immunity and tissue repair. In vertebrates, the cytoskeletal component F-actin is a DAMP specifically recognised by DNGR-1, an innate immune receptor. Previously we suggested that actin is also a DAMP in Drosophila melanogaster by inducing STAT-dependent genes (Srinivasan et al., 2016). Here, we revise that conclusion and report that α-actinin is far more potent than actin at inducing the same STAT response and can be found in trace amounts in actin preparations. Recombinant expression of actin or α-actinin in bacteria demonstrated that only α-actinin could drive the expression of STAT target genes in Drosophila. The response to injected α-actinin required the same signalling cascade that we had identified in our previous work using actin preparations. Taken together, these data indicate that α-actinin rather than actin drives STAT activation when injected into Drosophila.

Animals live together with diverse bacteria that can impact their biology. In Drosophila melanogaster, gut-associated bacterial communities are relatively simple in composition but also have a strong impact on host development and physiology. It is generally assumed that gut bacteria in D. melanogaster are transient and their constant ingestion with food is required to maintain their presence in the gut. Here, we identify bacterial species from wild-caught D. melanogaster that stably associate with the host independently of continuous inoculation. Moreover, we show that specific Acetobacter wild isolates can proliferate in the gut. We further demonstrate that the interaction between D. melanogaster and the wild isolated Acetobacter thailandicus is mutually beneficial and that the stability of the gut association is key to this mutualism. The stable population in the gut of D. melanogaster allows continuous bacterial spreading into the environment, which is advantageous to the bacterium itself. The bacterial dissemination is in turn advantageous to the host because the next generation of flies develops in the presence of this particularly beneficial bacterium. A. thailandicus leads to a faster host development and higher fertility of emerging adults when compared to other bacteria isolated from wild-caught flies. Furthermore, A. thailandicus is sufficient and advantageous when D. melanogaster develops in axenic or freshly collected figs, respectively. This isolate of A. thailandicus colonizes several genotypes of D. melanogaster but not the closely related D. simulans, indicating that the stable association is host specific. This work establishes a new conceptual model to understand D. melanogaster–gut microbiota interactions in an ecological context; stable interactions can be mutualistic through microbial farming, a common strategy in insects. Moreover, these results develop the use of D. melanogaster as a model to study gut microbiota proliferation and colonization., Author summary Animals, including humans, live together with complex bacterial communities in their gut that influence their physiology and health. The fruit fly Drosophila melanogaster is an excellent model organism to study host–microbe interactions and harbors a relatively simple gut bacterial community. The dominating hypothesis in the field is that gut bacteria in D. melanogaster are unstable and their constant ingestion with food is required to maintain their levels in the gut. Here, however, we show in D. melanogaster collected from a natural population, that stable gut bacteria do exist. We isolated specific species that can proliferate in the gut and form a stable association that is beneficial for both the bacteria and the flies. For the bacteria, they benefit from being constantly disseminated by the flies as they move around. For the flies, this is a form of farming, as the next generation of flies benefits from the association with these particular bacteria during development. Flies associated with these bacteria during the larval stages become adults faster and are more fertile than if they develop with other bacteria encountered in nature. Our findings show that D. melanogaster has stable colonizing bacteria in the gut, which are important for host development, establishing a new framework to study host–gut bacteria interactions.

Bacterial communities can sense their neighbors, regulating group behaviors in response to cell density and environmental changes. The diversity of signaling networks in a single species has been postulated to allow custom responses to different stimuli; however, little is known about how multiple signals are integrated and the implications of this integration in different ecological contexts. In the plant pathogen Pectobacterium wasabiae (formerly Erwinia carotovora), two signaling networks—the N-acyl homoserine lactone (AHL) quorum-sensing system and the Gac/Rsm signal transduction pathway—control the expression of secreted plant cell wall-degrading enzymes, its major virulence determinants. We show that the AHL system controls the Gac/Rsm system by affecting the expression of the regulatory RNA RsmB. This regulation is mediated by ExpR2, the quorum-sensing receptor that responds to the P. wasabiae cognate AHL but also to AHLs produced by other bacterial species. As a consequence, this level of regulation allows P. wasabiae to bypass the Gac-dependent regulation of RsmB in the presence of exogenous AHLs or AHL-producing bacteria. We provide in vivo evidence that this pivotal role of RsmB in signal transduction is important for the ability of P. wasabiae to induce virulence in response to other AHL-producing bacteria in multispecies plant lesions. Our results suggest that the signaling architecture in P. wasabiae was coopted to prime the bacteria to eavesdrop on other bacteria and quickly join the efforts of other species, which are already exploiting host resources., IMPORTANCE Quorum-sensing mechanisms enable bacteria to communicate through small signal molecules and coordinate group behaviors. Often, bacteria have various quorum-sensing receptors and integrate information with other signal transduction pathways, presumably allowing them to respond to different ecological contexts. The plant pathogen Pectobacterium wasabiae has two N-acyl homoserine lactone receptors with apparently the same regulatory functions. Our work revealed that the receptor with the broadest signal specificity is also responsible for establishing the link between the main signaling pathways regulating virulence in P. wasabiae. This link is essential to provide P. wasabiae with the ability to induce virulence earlier in response to higher densities of other bacterial species. We further present in vivo evidence that this novel regulatory link enables P. wasabiae to join related bacteria in the effort to degrade host tissue in multispecies plant lesions. Our work provides support for the hypothesis that interspecies interactions are among the major factors influencing the network architectures observed in bacterial quorum-sensing pathways.

The distribution of franciscana Pontoporia blainvillei Gervais & d'Orbigny, 1884, is restricted to the estuarine and coastal waters of the southwestern Atlantic. The diet of this dolphin is composed of fishes, squids, and shrimps, many of which harbor helminths that may infect franciscana. Larval forms of the trematode Synthesium pontoporiae and the acanthocephalan Bolbosoma turbinella have been recorded in franciscana; however, they have not yet been identified in any of the prey species of this cetacean. We evaluated 3 components of the diet of franciscana as possible transmission sources of parasitiasis. Specimens of Sao Paulo squid Doryteuthis sanpaulensis (n = 50), banded croaker Paralonchurus brasiliensis (n = 43), and rough scad Trachurus lathami (n = 50) were necropsied. Organs were washed and examined under a stereomicroscope. Helminths were collected and mounted on slides. None of the species analyzed showed infection by metacercariae of S. pontoporiae. Helminths found in Sao Paulo squid have not been recorded in franciscana. Cistacanths of Corynosoma australe were found in the coelomic cavity and mesentery of croaker (prevalence [P] = 53.49%; mean infection intensity [MII] = 6.74) and scad (P = 4%; MII = 1.50). Cistacanths of B. turbinella were also found in the same sites in scad (P = 14%; MII = 2.14). Banded croaker and rough scad are recorded in this study as new paratenic hosts for C. australe, while scad is a new paratenic host for B. turbinella.

Detailed chromosome photomaps are the first step to develop further chromosomal analysis to study the evolution of the genetic architecture in any set of species, considering that chromosomal rearrangements, such as inversions, are common features of genome evolution. In this report, we analyzed inversion polymorphisms in 25 different populations belonging to six neotropical species in the cardini group: Drosophila cardini, D. cardinoides, D. neocardini, D. neomorpha, D. parthenogenetica and D. polymorpha. Furthermore, we present the first reference photomaps for the Neotropical D. cardini and D. parthenogenetica and improved photomaps for D. cardinoides, D. neocardini and D. polymorpha. We found 19 new inversions for these species. An exhaustive pairwise comparison of the polytene chromosomes was conducted for the six species in order to understand evolutionary patterns of their chromosomes.

Comparative genomics in Drosophila began in 1940, when Muller stated that the ancestral haploid karyotype of this genus is constituted by five acrocentric chromosomes and one dot chromosome, named A to F elements. In some species of the willistoni group such as Drosophila willistoni and D. insularis, the F element, instead of a dot chromosome, has been incorporated into the E element, forming chromosome III (E + F fusion). The aim of this study was to investigate the scope of the E + F fusion in the willistoni group, evaluating six other species. Fluorescent in situ hybridization was used to locate two genes of the F element previously studied—cubitus interruptus (ci) and eyeless (ey)—in species of the willistoni and bocainensis subgroups. Moreover, polytene chromosome photomaps corresponding to the F element (basal portion of chromosome III) were constructed for each species studied. In D. willistoni, D. paulistorum and D. equinoxialis, the ci gene was located in subSectction 78B and the ey gene in 78C. In D. tropicalis, ci was located in subSection 76B and ey in 76C. In species of the bocainensis subgroup, ci and ey were localized, respectively, at subsections 76B and 76C in D. nebulosa and D. capricorni, and 76A and 76C in D. fumipennis. Despite the differences in the subsection numbers, all species showed the same position for ci and ey. The results confirm the synteny of E + F fusion in willistoni and bocainensis subgroups, and allow estimating the occurrence of this event at 15 Mya, at least.

This paper presents the first report of the occurrence of Drosophila suzukii (Diptera: Drosophilidae) in South America. Several samples were collected during various seasons in 2012 and 2013, and a significant number of individuals were obtained and identified. Based on the data recorded during each sampling campaign, including the locality, temperature, time of year, and number of individuals collected, several conclusions were drawn about the development of this invasion in southern Brazil.

The hAT superfamily comprises a large and diverse array of DNA transposons found in all supergroups of eukaryotes. Here we characterized the Drosophila buzzatii BuT2 element and found that it harbors a five-exon gene encoding a 643-aa putatively functional transposase. A phylogeny built with 85 hAT transposases yielded, in addition to the two major groups already described, Ac and Buster, a third one comprising 20 sequences that includes BuT2, Tip100, hAT-4_BM, and RP-hAT1. This third group is here named Tip. In addition, we studied the phylogenetic distribution and evolution of BuT2 by in silico searches and molecular approaches. Our data revealed BuT2 was, most often, vertically transmitted during the evolution of genus Drosophila being lost independently in several species. Nevertheless, we propose the occurrence of three horizontal transfer events to explain its distribution and conservation among species. Another aspect of BuT2 evolution and life cycle is the presence of short related sequences, which contain similar 5' and 3' regions, including the terminal inverted repeats. These sequences that can be considered as miniature inverted repeat transposable elements probably originated by internal deletion of complete copies and show evidences of recent mobilization.

Damage-associated molecular patterns (DAMPs) are molecules released by dead cells that trigger sterile inflammation and, in vertebrates, adaptive immunity. Actin is a DAMP detected in mammals by the receptor, DNGR-1, expressed by dendritic cells (DCs). DNGR-1 is phosphorylated by Src-family kinases and recruits the tyrosine kinase Syk to promote DC cross-presentation of dead cell-associated antigens. Here we report that actin is also a DAMP in invertebrates that lack DCs and adaptive immunity. Administration of actin to Drosophila melanogaster triggers a response characterised by selective induction of STAT target genes in the fat body through the cytokine Upd3 and its JAK/STAT-coupled receptor, Domeless. Notably, this response requires signalling via Shark, the Drosophila orthologue of Syk, and Src42A, a Drosophila Src-family kinase, and is dependent on Nox activity. Thus, extracellular actin detection via a Src-family kinase-dependent cascade is an ancient means of detecting cell injury that precedes the evolution of adaptive immunity. To do this, the body triggers a process called inflammation at the site of injury to remove dead and damaged cells, keep the area free from infection and trigger repair. However, if an area becomes excessively inflamed, or remains inflamed for a long period of time, it can contribute to diseases like cancer and Alzheimer’s disease. Inflammation starts when the body detects molecules that are released when cells die or are damaged to the extent that they become leaky. Actin, which is a protein that usually provides structural support to the cell, is one molecule that is sensed by the immune system in mammals when released from dying cells. However, it is not clear whether released actin can also trigger reactions in simpler animals like fruit flies. To address this question, Srinivasan, Gordon et al. injected fruit flies with actin. These animals developed a widespread reaction reminiscent of inflammation seen in mammals. Further experiments showed that actin switches on a signalling pathway called JAK/STAT, which is known to become active when flies experience other types of stress. The JAK/STAT proteins activate a signalling pathway that leads to changes in gene activity. Srinivasan, Gordon et al. also showed that part of a cascade of signals triggered by released actin in mammals is shared in fruit flies. These findings suggest that this important response to actin evolved a long time ago. A future challenge is to find out how the body detects and mops up released actin, which may help us to understand how actin can contribute to various inflammatory diseases. Diptera is a megadiverse order, reaching its peak of diversity in Neotropics, although our knowledge of dipteran fauna from this region is grossly lacking. This applies even to the most studied families, such as Drosophilidae. Despite its prominence, most aspects of the biology of these insects are still poorly understood, especially those linked to natural communities. Field studies on drosophilids are highly biased towards fruit-breeding species. Flower-breeding drosophilids, however, are worldwide distributed, especially in tropical regions, although being mostly neglected. The present paper shows the results of a biodiversity inventory of flower-breeding drosophilids carried out in several localities in Brazil, based on samples of 125 plant species, from 47 families. Drosophilids were found in flowers of 56 plant species, from 18 families. The fauna discovered turned out to be mostly unknown, comprising 28 species, with 12 of them (>40%) still undescribed. Not taking into account opportunistic species, two-thirds of the flower-exclusive diversity was undescribed. The Drosophila bromeliae species group was the most representative taxon, with eight species (six undescribed), including four polyphagous and four Solanum-specialized species. This specialization on Solanum is reported for the first time for Drosophilidae. Other taxa of restricted flower-breeding drosophilids were the Drosophila lutzii species group and two species of the genus Zygothrica Wiedemann. Some specimens of the genera Cladochaeta Coquillett, Rhinoleucophenga Hendel and Scaptomyza Hardy were found, but their relations to flowers are unclear. Additionally, ten species of broad niche were found using flowers opportunistically. Localities and host plants were recorded for all species collected. The binding protein (BiP) has been demonstrated to participate in innate immunity and attenuate endoplasmic reticulum- and osmotic stress-induced cell death. Here, we employed transgenic plants with manipulated levels of BiP to assess whether BiP also controlled developmental and hypersensitive programmed cell death (PCD). Under normal conditions, the BiP-induced transcriptome revealed a robust down-regulation of developmental PCD genes and an up-regulation of the genes involved in hypersensitive PCD triggered by nonhost-pathogen interactions. Accordingly, the BiP-overexpressing line displayed delayed leaf senescence under normal conditions and accelerated hypersensitive response triggered by Pseudomonas syringae pv tomato in soybean (Glycine max) and tobacco (Nicotiana tabacum), as monitored by measuring hallmarks of PCD in plants. The BiP-mediated delay of leaf senescence correlated with the attenuation of N-rich protein (NRP)-mediated cell death signaling and the inhibition of the senescence-associated activation of the unfolded protein response (UPR). By contrast, under biological activation of salicylic acid (SA) signaling and hypersensitive PCD, BiP overexpression further induced NRP-mediated cell death signaling and antagonistically inhibited the UPR. Thus, the SA-mediated induction of NRP cell death signaling occurs via a pathway distinct from UPR. Our data indicate that during the hypersensitive PCD, BiP positively regulates the NRP cell death signaling through a yet undefined mechanism that is activated by SA signaling and related to ER functioning. By contrast, BiP’s negative regulation of leaf senescence may be linked to its capacity to attenuate the UPR activation and NRP cell death signaling. Therefore, BiP can function either as a negative or positive modulator of PCD events.

Infections by the endosymbiotic bacterium Wolbachia developed a rapid global expansion within Old World Drosophila species, ultimately infecting also Neotropical species. In this sense, screenings are necessary to characterize new variants of Wolbachia or new hosts, and also in order to map the dynamics of already known infections. In this paper, we performed a double screening approach that combined Dot-blot and PCR techniques in order to reevaluate the infection status by Wolbachia in species from the willistoni subgroup of Drosophila. Genomic DNA from isofemale lines descendent from females collected in the Amazonian Rainforest (n=91) were submitted to Dot-blot, and were positive for Wolbachia, producing a gradient of hybridization signals, suggesting different infection levels, which was further confirmed through quantitative PCR. Samples with a strong signal in the Dot-blot easily amplified in the wsp-PCR, unlike most of the samples with a medium to weak signal. It was possible to molecularly characterize three Drosophila equinoxialis isofemale lines that were found to be infected in a low density by a wMel-like Wolbachia strain, which was also verified in a laboratory line of Drosophila paulistorum Amazonian. We also found Drosophila tropicalis to be infected with the wAu strain and a Drosophila paulistorum Andean-Brazilian semispecies laboratory line to be infected with a wAu-like Wolbachia. Moreover, we observed that all Drosophila willistoni samples tested with the VNTR-141 marker harbor the same Wolbachia variant, wWil, either in populations from the South or the North of Brazil. Horizontal transfer events involving species of Old World immigrants and Neotropical species of the willistoni subgroup are discussed.

Environmental variables such as temperature and rainfall can directly affect the community structure of dipterans. Seasonal oscillations in the abundance of species of Drosophilidae reflect differences in how tolerant populations are to climatic conditions. Over a period of 14 months, we collected samples in two habitats in the Pampa biome in the municipality of Sao Luiz Gonzaga, state of Rio Grande do Sul, Brazil (28°24′28″S, 54°57′39″W). The influence of environmental variables on populations of Drosophilidae was evaluated for both collecting sites by using correlation analysis. The results suggested a negative correlation between the abundances of Drosophila cardinoides Dobzhansky & Pavan, Drosophila maculifrons Duda, Drosophila melanogaster Meigen, Drosophila nigricruria Patterson & Mainland, and Zygothrica vittimaculosa Burla with temperature, which is reflected in the distribution of these species within Brazil. Our findings are important for characterizing and preserving biodiversity in this almost-unknown biome in southern Brazil given the current climate change scenario.

No. of bitstreams: 0 Previous issue date: 2016-06-01 Introduction Cattle temperament is an important factor that affects the profitability of beef cattle enterprises, due to its relationship with productivity traits, animal welfare and labor safety. Temperament is a complex phenotype often assessed by measuring a series of behavioral traits, which result from the effects of multiple environmental and genetic factors, and their interactions. The aims of this study were to perform a genome-wide association study and detect genomic regions, potential candidate genes and their biological mechanisms underlying temperament, measured by flight speed (FS) test in Nellore cattle. Materials and Methods The genome-wide association study (GWAS) was performed using a single-step procedure (ssGBLUP) which combined simultaneously all 16,600 phenotypes from genotyped and non-genotyped animals, full pedigree information of 162,645 animals and 1,384 genotyped animals in one step. The animals were genotyped with High Density Bovine SNP BeadChip which contains 777,962 SNP markers. After quality control (QC) a total of 455,374 SNPs remained. Results Heritability estimated for FS was 0.21 ± 0.02. Consecutive SNPs explaining 1% or more of the total additive genetic variance were considered as windows associated with FS. Nine candidate regions located on eight different Bos taurus chromosomes (BTA) (1 at 73 Mb, 2 at 65 Mb, 5 at 22 Mb and 119 Mb, 9 at 98 Mb, 11 at 67 Mb, 15 at 16 Mb, 17 at 63 Kb, and 26 at 47 Mb) were identified. The candidate genes identified in these regions were NCKAP5 (BTA2), PARK2 (BTA9), ANTXR1 (BTA11), GUCY1A2 (BTA15), CPE (BTA17) and DOCK1 (BTA26). Among these genes PARK2, GUCY1A2, CPE and DOCK1 are related to dopaminergic system, memory formation, biosynthesis of peptide hormone and neurotransmitter and brain development, respectively. Conclusions Our findings allowed us to identify nine genomic regions (SNP windows) associated with beef cattle temperament, measured by FS test. Within these windows, six promising candidate genes and their biological functions were identified. Pectobacterium wasabiae (previously known as Erwinia carotovora ) is an important plant pathogen that regulates the production of plant cell wall-degrading enzymes through an N -acyl homoserine lactone-based quorum sensing system and through the GacS/GacA two-component system (also known as ExpS/ExpA). At high cell density, activation of GacS/GacA induces the expression of RsmB, a noncoding RNA that is essential for the activation of virulence in this bacterium. A genetic screen to identify regulators of RsmB revealed that mutants defective in components of a putative Trk potassium transporter ( trkH and trkA ) had decreased rsmB expression. Further analysis of these mutants showed that changes in potassium concentration influenced rsmB expression and consequent tissue damage in potato tubers and that this regulation required an intact Trk system. Regulation of rsmB expression by potassium via the Trk system occurred even in the absence of the GacS/GacA system, demonstrating that these systems act independently and are both required for full activation of RsmB and for the downstream induction of virulence in potato infection assays. Overall, our results identified potassium as an essential environmental factor regulating the Rsm system, and the consequent induction of virulence, in the plant pathogen P. wasabiae . IMPORTANCE Crop losses from bacterial diseases caused by pectolytic bacteria are a major problem in agriculture. By studying the regulatory pathways involved in controlling the expression of plant cell wall-degrading enzymes in Pectobacterium wasabiae , we showed that the Trk potassium transport system plays an important role in the regulation of these pathways. The data presented further identify potassium as an important environmental factor in the regulation of virulence in this plant pathogen. We showed that a reduction in virulence can be achieved by increasing the extracellular concentration of potassium. Therefore, this work highlights how elucidation of the mechanisms involved in regulating virulence can lead to the identification of environmental factors that can influence the outcome of infection.

O Pampa brasileiro (extremo sul do país) está, atualmente, vastamente modificado devido ao aumento das atividades agrícolas. Em muitos lugares, apenas pequenos fragmentos de campo permanecem em uma paisagem agrícola. Drosophilidae (Diptera) tem sido amplamente utilizadas como bioindicadores para monitorar os efeitos das mudanças antropogênicas em ambientes naturais. Porém, a fauna de Drosophilidae no Bioma Pampa de ambientes naturais ou perturbados, ainda permanece amplamente desconhecida. O presente estudo é uma das primeiras tentativas de preencher esta lacuna, apresentando resultados de coletas mensais no município de São Luiz Gonzaga (28º24'28"S, 54º57'39"W), no Pampa brasileiro. Um inventário de espécies foi conduzido em dois ambientes contrastantes, uma zona urbana e um remanescente de floresta (zona rural). Em ambos os locais, armadilhas com banana fermentada foram usadas para capturar drosofilideos adultos. A identificação foi feita através da morfologia externa e da terminália dos machos. No total, 13,379 drosofilideos foram analisados (zona rural: N = 8,812 and Sobs = 25; zona urbana: N = 4,567 and Sobs = 16). No presente estudo, 16 (60%) das 26 espécies coletadas foram encontradas exclusivamente ou preferencialmente no fragmento de mata. O período de maior riqueza foi entre os meses de junho a novembro (inverno-primavera), e o período de menor riqueza foi de dezembro a maio (verão-outono). Uma análise de cluster pelo Coeficiente de Jaccard mostrou que a composição da assembléia muda ligeiramente quando o período do ano com temperaturas mais elevadas (janeiro-maio) é comparado com o período de temperaturas menos elevadas (junho-outubro). A abundância das espécies foi também altamente afetada pela sazonalidade, como revelou o Índice de Morisita, onde as amostras foram agrupadas em períodos consecutivos dentro de uma mesma estação, mostrando a preferência sazonal de algumas espécies. O componente tempo foi determinante na diversidade da assembléia, superando o efeito espacial. A forte redução na diversidade na área urbana quando comparada com o pequeno fragmento de floresta, torna evidente a importância do ambiente natural para a preservação da diversidade no bioma Pampa, atualmente com sua paisagem altamente alterada. The Brazilian Pampa (the southernmost end of the country) is currently a highly modified environment because of increasing agricultural activities. In many places, only small parts of grasslands remain inside an agricultural landscape. Drosophilidae (Diptera) have been widely used as a potential bioindicators to monitor the effects of anthropogenic changes in natural environments. However, the fauna of Drosophilidae in the Pampa Biome from natural and disturbed environments, still remains largely unknown. The present study represents one of the first attempts to fill this gap, showing results from monthly collections in the municipality of São Luiz Gonzaga (28º24'28"S, 54º57'39"W), in the Brazilian Pampa. A species inventory was carried out in two contrasting environments, an urban zone and a forest remnant (rural zone). In both areas banana-baited traps were used to capture adult drosophilids. The identification was made using external morphology and male terminalia. In total, 13,379 drosophilids were analyzed (rural zone: N = 8,812 and Sobs = 25; urban zone: N = 4,567 and Sobs = 16). In the present study, 16 (60%) out of 26 species were found exclusively or preferentially in the forest. The period of highest richness was between the months of June to November (roughly winter and spring), and the period of lowest richness was from December to May (roughly summer and autumn). An analysis of cluster by the Coefficient of Jaccard showed that species composition slightly changes when the period of the year with higher temperatures (from January to May) is compared with the period with lower temperatures (from June to October). The species abundances were also highly affected by seasonality, as revealed by the Morisita Index, since the samples clustered into similar groups in consecutive periods and in the same season, showing the seasonal preference of some species. The time component was a determinant in the diversity of the assemblage, surpassing the spatial effect. The strong reduction in diversity in the urban area when compared to a small forest patch is evidence of the importance of the natural environments in maintaining the diversity in the Pampa biome, currently a highly disturbed landscape.
