Most new species and records in a 1987 survey were from acid waters (pH < 4.0) of dune lakes on the west coast (42° S). Marked intra- and interhabitat differences in rotifer communities of lakes and ponds were demonstrated by cluster analysis and related to habitat heterogeneity.

Murray-Darling Freshwater Research CentreMDFRC item.The spatial and temporal dynamics of rotifers in the littoral ecotone of Lake Rotomanuka (37° 55 S, 175° 19 E) were studied from February to November 1994. Rotifers were sampled with artificial substrates at two or three weekly intervals from eight sites chosen with respect to macrophyte species distribution from near shore to deeper water. 58 rotifer species were found, a high diversity in comparison to that of New Zealand limnetic communities, which usually have less than ten species. Rotifers had peak abundances in summer within emergent and submerged vegetation, when shallow regions were dry. Lecane bulla (Gosse) and Testudinella parva (Ternetz) generally had the highest numerical densities. Three major temporal groupings of species were distinguished by cluster analysis and canonical correspondence analysis (CCA): summer–autumn (e.g. Lecane hornemanni (Ehrenberg), L. bulla), winter-spring (e.g. Mytilina mucronata (Müller), Trichocerca porcellus (Gosse)), and late autumn to mid spring (e.g. T. parva, Polyarthra vulgaris (Carlin)). Rotifer species composition appeared to depend on seasonal change of water level and the associated shift from heterogeneous to homogenous physical and chemical conditions across the ecotone. Temporal variability in the abundance of zindividual rotifer species was far greater than their spatial variability. CCA indicated that temperature and pH were the factors most strongly associated with temporal variation in abundances of rotifer species. Macrophytes appeared to play the major role in determining spatial distribution, both because of differences in physical structure between species (affecting microhabitat diversity) and by causing variations in physical and chemical conditions (e.g. oxygen and pH) by inhibiting mixing.

La Trobe University Faculty of Science, Technology and Engineering Murray Darling Freshwater Research CentreMDFRC item.The mandibles, first and second maxillae, and maxillipeds of Boeckella major, B. pseudochelae, and Hemiboeckella searli from temporary pools of the upper River Murray catchment were dissected off and examined by light microscopy for features associated with carnivorous feeding. In B. major adaptations for carnivory are enlarged ventral mandibular teeth, strong falciform medial setae on the second maxillae, falciform endopodal setae on the maxilliped, large body size and large mouthpart size in relation to body size. Its mandibular edge index is 772, in the upper omnivore range. B. major is predicted to be an omnivore with strong carnivorous tendencies and able to handle large prey items. Carnivorous features in H. searli are enlarged ventral mandibular teeth, a mandibular edge index of 1395, and strong unguiform and falciform setae on the second maxillae. It is predicted to be an omnivore with moderate carnivorous tendencies, but unable to handle prey as large as can B. major. In B. pseudochelae the only adaptations for carnivory are an edge index of 1080 and falciform setae on the maxilliped. It is predicted to be an omnivore with herbivorous tendencies. Examination of gut contents confirmed these predictions. B. major guts contained 19 animal taxa, mainly planktonic cladocerans, copepods and rotifers, H. searli 12 taxa, mainly copepods and rotifers, and B. pseudochelae 8 taxa, mainly rotifers. All had consumed algae. We suggest that predation by B. major may be an important factor affecting community structure in fishless Australian temporary pools.

Community size structures and species diversity are very variable and many ponds contain one or more of the large calanoid copepods, Boeckella major, Boeckella pseudochelae and Hemiboeckella searli. Gut content analysis showed that these species were omnivores with B, major being the most carnivorous and B. pseudochelae the least. B. major guts contained 21 animal taxa (cladocerans, calanoid and cyclopoid copepodids, nauplii and rotifers), H. searli contained 15 taxa (calanoid, cyclopoid and harpacticoid copepods, nauplii and rotifers) and B. pseudochelae 8 taxa (small cladocerans, nauplii and rotifers). Mean numbers of animals per gut differed significantly between the large calanoids and in B. major, B. pseudochelae and H. searli were, respectively, 2.9, 0.6 and 0.2. In B. major "Rotifers" were positively selected, "Copepodites" and "Others" neutrally selected and "Cladocera" and "Nauplii" negatively preferred. In H. searli "Copepodites", "Nauplii" and "Rotifers" were neutrally selected, and "Others" negatively preferred. B. pseudochelae showed negative preference for "Cladocera", "Nauplii" and "Others", and "Rotifers" were neutrally selected. At sites containing B. major, species diversity was significantly reduced and community size structure (measured as mean and median log-length, % of sizes >500 mu m) was significantly larger than it was at sites lacking large calanoids or with only B. pseudochelae and H. searli. It is hypothesised that predation by B. major may be an important force structuring the communities of these habitats.

Murray-Darling Freshwater Research CentreMDFRC item.Intra- and interspecific variation in the intersetular distance (i.s.d.) of some southern Australian cladocerans was examined. The seasonal range of i.s.d. inDaphnia carinata adults (2.34±0.07 mm) was 0.17–0.25 µm. I.s.d. increased with increasing body size forD. carinata andD. thomsoni. Variation in i.s.d. between nine adult cladoceran taxa (several of which coexisted) also was examined; significant differences did occur, but the size range was small (0.14 to 0.58 µm). The possibility that intersetular distances are an index of niche separation based upon the capture of different particle sizes is discussed.

La Trobe University Faculty of Science, Technology and Engineering Murray Darling Freshwater Research CentreMDFRC item.Density-size distributions of a river zooplankton community were examined about biweekly over a year in the upper tidal freshwater portion of a regulated Australian river, specifically the Hawkesbury-Nepean, Results were compared with those of similar studies in lakes (and reservoirs) to characterize the similarities and differences in the structure and grazing function of river and lake zooplankton communities. The density-size distributions of the river zooplankton community were similar to those of lake plankton communities, in terms of a marked temporal variation in shape and overall bimodal shape. They differed in terms of the truncation of the upper body size and the absence of a significant relationship between the slope (regression coefficient) of a log-linear density-size model and environmental variables. The river zooplankton community appeared to have a high average rate of biomass increase when measured against body-mass class and a low mass-specific grazing rate, compared with those of lake communities. Because of methodological differences between studies, intersystem comparisons of size spectra and grazing rates need cautious interpretations and generalizations.

Murray-Darling Freshwater Research CentreMDFRC item.Grazing rates by a zooplankton community were measured in situ by a radiotracer cellmethod at depths of 1 m and 4 m at the upper tidal freshwater portion of a regulated river over a year.The objectives were to evaluate the likely grazing impact on the river phytoplankton community and toproduce predictive models by regressing the measured grazing rates against zooplankton biomass,temperature and food concentrations (represented by chlorophyll a). Grazing attained rates (overallaverage 0.2 day-', range 0.01-0.59 day-', expressed as instantaneous mortality rates of algal cells)comparable to those reported for lentic zooplankton communities. The measured community grazingrates were predictable largely as a function of total biomass or rotifer biomass and surface temperaturefor 1 m depth, and as a function of total biomass or juvenile copepod biomass and surface temperaturefor 4 m depth, with all-positive regression coefficients in the models. Owing to the predominance ofmicrozooplankton in the river, the impact of zooplankton community grazing appears likely to be linkedto a small-size fraction of the phytoplankton community all year. Management strategies for river waterquality may need to take account of possible functional demarcation of grazing by river zooplankton.

In mammalian embryos, proper zygotic genome activation (ZGA) underlies totipotent development. Double homeobox (DUX)-family factors participate in ZGA, and mouse Dux is required for forming cultured two-cell (2C)-like cells. Remarkably, in mouse embryonic stem cells, Dux is activated by the tumor suppressor p53, and Dux expression promotes differentiation into expanded-fate cell types. Long-read sequencing and assembly of the mouse Dux locus reveals its complex chromatin regulation including putative positive and negative feedback loops. We show that the p53–DUX/DUX4 regulatory axis is conserved in humans. Furthermore, we demonstrate that cells derived from patients with facioscapulohumeral muscular dystrophy (FSHD) activate human DUX4 during p53 signaling via a p53-binding site in a primate-specific subtelomeric long terminal repeat (LTR)10C element. In summary, our work shows that p53 activation convergently evolved to couple p53 to Dux/DUX4 activation in embryonic stem cells, embryos and cells from patients with FSHD, potentially uniting the developmental and disease regulation of DUX-family factors and identifying evidence-based therapeutic opportunities for FSHD. p53 activates Dux in mouse embryos and embryonic stem cells, as well as DUX4 in human facioscapulohumeral muscular dystrophy cell models.

It has often been suggested that the productivity of an ecosystem affects the number of species that it can support. Despite decades of study, the nature, extent, and underlying mechanisms of this relationship are unclear. One suggested mechanism is the “more individuals” hypothesis (MIH). This proposes that productivity controls the number of individuals in the ecosystem, and that more individuals can be divided into a greater number of species before their population size is sufficiently small for each to be at substantial risk of extinction. Here, we test this hypothesis using REvoSim: an individual‐based eco‐evolutionary system that simulates the evolution and speciation of populations over geological time, allowing phenomena occurring over timescales that cannot be easily observed in the real world to be evaluated. The individual‐based nature of this system allows us to remove assumptions about the nature of speciation and extinction that previous models have had to make. Many of the predictions of the MIH are supported in our simulations: Rare species are more likely to undergo extinction than common species, and species richness scales with productivity. However, we also find support for relationships that contradict the predictions of the strict MIH: species population size scales with productivity, and species extinction risk is better predicted by relative than absolute species population size, apparently due to increased competition when total community abundance is higher. Furthermore, we show that the scaling of species richness with productivity depends upon the ability of species to partition niche space. Consequently, we suggest that the MIH is applicable only to ecosystems in which niche partitioning has not been halted by species saturation. Some hypotheses regarding patterns of biodiversity implicitly or explicitly overlook niche theory in favor of neutral explanations, as has historically been the case with the MIH. Our simulations demonstrate that niche theory exerts a control on the applicability of the MIH and thus needs to be accounted for in macroecology., We use simulation experiments to test the predictions of the more‐individuals hypothesis of biodiversity. We find that many of these predictions are supported, but that interspecific competition and limits to niche diversity can provide additional complexity not predicted by the more‐individuals hypothesis.

Genus Fusarium (Ascomycota, Hypocreales, Nectriaceae) includes many economically important plant pathogens that cause devastating diseases of a wide range of crops and trees. Interestingly, there is increasing evidence that some Fusarium species also live as endophytes and benefit plant growth and stress tolerance. In this work, we sequence the whole genomes of endophytic F. culmorum and F. pseudograminearum, isolated from a coastal dunegrass (Leymus mollis), using long-read single-molecule real-time sequencing technology. Their genomes are assembled into four chromosomes and a mitochondrial genome with a total assembly size of 40.05 and 42.90 M, respectively. This resource should not only facilitate functional studies designed to better understand what makes the two Fusarium species such successful plant-beneficial fungi but should also reveal their genome evolution and adaptation. Antimicrobial resistance has rendered many conventional therapeutic measures, such as antibiotics, ineffective. This makes the treatment of infections from pathogenic micro-organisms a major growing health, social, and economic challenge. Recently, nanomaterials, including two-dimensional (2D) materials, have attracted scientific interest as potential antimicrobial agents. Many of these studies, however, rely on the input of activation energy and lack real-world utility. In this work, we present the broad-spectrum antimicrobial activity of few-layered black phosphorus (BP) at nanogram concentrations. This property arises from the unique ability of layered BP to produce reactive oxygen species, which we harness to create this unique functionality. BP is shown to be highly antimicrobial toward susceptible and resistant bacteria and fungal species. To establish cytotoxicity with mammalian cells, we showed that both L929 mouse and BJ-5TA human fibroblasts were metabolically unaffected by the presence of BP. Finally, we demonstrate the practical utility of this approach, whereby medically relevant surfaces are imparted with antimicrobial properties via functionalization with few-layer BP. Given the self-degrading properties of BP, this study demonstrates a viable and practical pathway for the deployment of novel low-dimensional materials as antimicrobial agents without compromising the composition or nature of the coated substrate.

Objective To determine if long interspersed element-1 (L1) retrotransposons convey risk for idiopathic temporal lobe epilepsy (TLE). Methods Surgically resected temporal cortex from individuals with TLE (N = 33) and postmortem temporal cortex from individuals with no known neurological disease (N = 33) were analyzed for L1 content by Restriction Enzyme Based Enriched L1Hs sequencing (REBELseq). Expression of three KCNIP4 splice variants was assessed by droplet digital PCR (ddPCR). Protein ANalysis THrough Evolutionary Relationships (PANTHER) was used to determine ontologies and pathways for lists of genes harboring L1 insertions. Results We identified novel L1 insertions specific to individuals with TLE, and others specific to controls. Although there were no statistically significant differences between cases and controls in the numbers of known and novel L1 insertions, PANTHER analyses of intragenic L1 insertions showed statistically significant enrichments for epilepsy-relevant gene ontologies in both cases and controls. Gene ontologies "neuron projection development" and "calcium ion transmembrane transport" were among those found only in individuals with TLE. We confirmed novel L1 insertions in several genes associated with seizures/epilepsy, including a de novo somatic L1 retrotransposition in KCNIP4 that occurred after neural crest formation in one patient. However, ddPCR results suggest this de novo L1 did not alter KCNIP4 mRNA expression. Significance Given current data from this small cohort, we conclude that L1 elements, either rare heritable germline insertions or de novo somatic retrotranspositions, may contribute only minimally to overall genetic risk for idiopathic TLE. We suggest that further studies in additional patients and additional brain regions are warranted.

The polyketide synthase (PKS)‐like protein TerB, consisting of inactive dehydratase, inactive C‐methyltransferase, and functional ketoreductase domains collaborates with the iterative non reducing PKS TerA to produce 6‐hydroxymellein, a key pathway intermediate during the biosynthesis of various fungal natural products. The catalytically inactive dehydratase domain of TerB appears to mediate productive interactions with TerA, demonstrating a new mode of trans‐interaction between iterative PKS components., A new architecture for iterative Type I polyketide synthases (PKS) from fungi has been investigated. TerA is a canonical non‐reducing PKS which interacts with TerB, a fragment of a highly reducing PKS containing a functional ketoreductase. Interaction appears to be mediated between a non‐functional dehydratase domain of TerB and the TerA product template domain. Working together, TerAB synthesises 6‐hydroxymellein, a precursor of terrein and other fungal metabolites.

Over the past two decades, the designs of redox polymers have become critical to the field of mediated bioelectrocatalysis and are used in commercial glucose biosensors, as well as other bioelectrochemical applications (e.g., energy harvesting). These polymers are specifically used to immobilize redox mediators on electrode surfaces, allowing for self-exchange-based conduction of electrons from enzymes far from the electrode to the electrode surface. However, the synthesis of redox polymers is challenging and results in large batch-to-batch variability. Herein, we report a rapid entrapment of mediators for NAD+-dependent bioelectrocatalysis within reverse ionically condensed polyelectrolytes. A high ionic strength aqueous solution of oppositely charged polyelectrolytes, composed of cationic polyguanidinium (PG) chloride and anionic sodium hexametaphosphate (P6), undergoes phase inversion into a solid microporous polyelectrolyte complex (PEC) when introduced into a low ionic strength aqueous solution. The ionic strength-triggered phase inversion of PGP6 solutions was investigated as a means to entrap mediators on the surface of electrodes for mediated bioelectrocatalysis. Compared to the traditional cross-linked immobilizations using redox polymers, this phase inversion takes place within seconds and requires up to 60 min for complete stabilization. In this work, redox mediator phenazine ethosulfate (PES) was entrapped within PGP6 on electrode surfaces for nicotinamide adenine dinucleotide (NAD+)-dependent bioelectrocatalysis. In the bulk solution, NAD+-dependent dehydrogenase enzymes catalyze the oxidation of the substrate while reducing NAD to reduced nicotinamide adenine dinucleotide (NADH). The resulting NADH is reoxidized to NAD+ by the entrapped PES that gets reduced on the electrode, completing the NAD+-regeneration-based bioelectrocatalysis. To show the use of these new materials in an application, biofuel cells were evaluated using four different anodic enzyme systems (alcohol dehydrogenase, lactate hydrogenase, glycerol dehydrogenase, and glucose dehydrogenase).

Apostolepis is a diverse Dipsadid snake genus, with 35 known species, inhabiting most of South America at east of the Andes. In this study, we redescribe Apostolepis assimilis Reinhardt, 1861, and evaluate the taxonomic status of a controversial taxon, Apostolepis tertulianobeui, described based on a single specimen from an unknown locality of Minas Gerais state, southeastern Brazil. A careful reanalysis of the type specimen and additional material provides compelling evidence for the synonymy of A. tertulianobeui with A. assimilis. We also provide comments on the taxonomy of Apostolepis, osteological and hemipenial descriptions, and a species distribution modelling for A. assimilis.

A suite of processes drive variation in coral populations in space and time, yet our understanding of how variation in coral density affects coral performance is limited. Theory predicts that reductions in density can send coral populations into a predator pit, where concentrated corallivory maintains corals at low densities. In reality, how variation in coral density alters corallivory rates is poorly resolved. Here, we experimentally quantified the effects of corallivory and coral density on growth and survival of small colonies of the staghorn coral Acropora pulchra. Our findings suggest that coral density and corallivory have strong but independent effects on coral performance. In the presence of corallivores, corals suffered high but density-independent mortality. When corallivores were excluded, however, vertical extension rates of colonies increased with increasing densities. While we found no evidence for a predator pit, our results suggest that spatio-temporal variation in corallivore and coral densities can fundamentally alter population dynamics via strong effects on juvenile corals.

Effect of geraniin extracted from sugar maple (Acer saccharum) leaves on the viability of the phytopathogen Xanthomonas campestris pv. vitians was evaluated with the SYTOX Green nucleic acid stain, penetrating only compromised membranes, and plate counts. In parallel, structural changes of treated bacteria were examined in transmission electron microscopy (TEM). Based on SYTOX Green and plate counts, geraniin at the minimum bactericidal concentration (3.125 mg/ml) increased mortality after 45 min by 37% and 62%, respectively, when compared with controls. According to observations in TEM, geraniin caused morphological alterations of these rod-shaped bacteria, including degradation of their envelopes, as also suggested by the incorporation of SYTOX. These alterations were often accompanied by cytoplasm leakage and the formation of more pronounced whitish areas in the cytoplasm similar to vacuolization. Moreover, multi-membranous and/or –wall systems were at times formed in the treated bacteria. The presence of some extracellular electron-dense material was frequently noted around the treated bacteria. The matrix surrounding control bacteria tended to disappear after geraniin treatment. This study highlights for the first time the effect of geraniin on bacterial ultrastructure, thus contributing to a better understanding of the mechanism by which this molecule exerts antibacterial activity.

The non-canonical fungal α-humulene synthase was investigated through isotopic labelling experiments for its stereochemical course regarding inversion or retention at C-1, the face selectivity at C-11, and the stereoselectivity of the final deprotonation. A new and convenient desymmetrisation strategy was developed to enable a full stereochemical analysis of the catalysed steps to the achiral α-humulene product from stereoselectively labelled farnesyl diphosphate.

In this study, we investigated the feasibility of anaerobic sewage treatment at extremely low temperatures (4 °C) using two reactor setups: upflow anaerobic sludge blanket reactors (UASB) without and with (AnMBRUASB(UF)) a membrane. Both reactors were inoculated with seeds derived from sediments that were putatively acclimatized to low temperatures. A preliminary batch trial showed that treatment is feasible with the removal of carbon coupled to methane and sulphide production. The reactors operated for 180 days at a hydraulic retention time of 3 days. After 40 days acclimation, both systems met the EU chemical oxygen demand (COD) effluent standard (

Fusarochromene isolated from the plant pathogenic fungus, Fusarium sacchari is closely related to a group of mycotoxins including fusarochromanone previously isolated from various Fusaria spp. Despite their assumed polyketide biogenesis, incorporation studies with 13C-labelled acetate, glycerol and tryptophans show that fusarochromene is unexpectedly derived via oxidative cleavage of the aromatic amino acid tryptophan. A putative biosynthetic gene cluster has been identified.

Resolution of relationships at lower taxonomic levels is crucial for answering many evolutionary questions, and as such, sufficiently varied species representation is vital. This latter goal is not always achievable with relatively fresh samples. To alleviate the difficulties in procuring rarer taxa, we have seen increasing utilization of historical specimens in building molecular phylogenies using high throughput sequencing. This effort, however, has mainly focused on large‐bodied or well‐studied groups, with small‐bodied and under‐studied taxa under‐prioritized. Here, we utilize both historical and contemporary specimens, to increase the resolution of phylogenetic relationships among a group of under‐studied and small‐bodied metazoans, namely, cheilostome bryozoans. In this study, we pioneer the sequencing of air‐dried cheilostomes, utilizing a recently developed library preparation method for low DNA input. We evaluate a de novo mitogenome assembly and two iterative methods, using the sequenced target specimen as a reference for mapping, for our sequences. In doing so, we present mitochondrial and ribosomal RNA sequences of 43 cheilostomes representing 37 species, including 14 from historical samples ranging from 50 to 149 years old. The inferred phylogenetic relationships of these samples, analyzed together with publicly available sequence data, are shown in a statistically well‐supported 65 taxa and 17 genes cheilostome tree, which is also the most broadly sampled and largest to date. The robust phylogenetic placement of historical samples whose contemporary conspecifics and/or congenerics have been sequenced verifies the appropriateness of our workflow and gives confidence in the phylogenetic placement of those historical samples for which there are no close relatives sequenced. The success of our workflow is highlighted by the circularization of a total of 27 mitogenomes, seven from historical cheilostome samples. Our study highlights the potential of utilizing DNA from micro‐invertebrate specimens stored in natural history collections for resolving phylogenetic relationships among species., Here, we present a workflow that utilizes historical and contemporary specimens, to increase the resolution of phylogenetic relationships among understudied and small‐bodied metazoans, namely, cheilostome bryozoans. The complete mitochondrial genomes of Brachionus manjavacas German strain were 10,721 bp (mitochondrial DNA I) and 12,274 bp (mitochondrial DNA II) in size, while the complete mitochondrial genomes of B. manjavacas Australian strain were 10,889 bp (mitochondrial DNA I) and 12,443 bp (mitochondrial DNA II) in size. Of 12 protein-coding genes (PCGs), 99.6% of amino acid sequences were identical between the two strains. Of 12 PCGs of both B. manjavacas strains, three genes (ND1, ATP6, and ND5) had incomplete stop codon T. Furthermore, ATA was the start codon for ND4, ND5, and CO3 genes, whereas that for other PCGs was ATG in both strains. The base compositions of 12 PCGs in the B. manjavacas strains were similar, indicating that the mitochondrial genome of the two strains was structurally conserved over evolution. The gene structure and its orientation of 12 PGCs of B. manjavacas strains were identical, as shown in other marine Brachionus rotifers and the freshwater Brachionus rotifers, while the freshwater rotifer B. calyciflorus had an additional cytochrome b gene in the mitochondrial DNA I.

The recent rise of antibiotic resistance amongst Staphylococcus aureus (S. aureus) populations has made treating Staph-based infections a global medical challenge. Therapies that specifically target the peptidoglycan layer of S. aureus have emerged as new treatment avenues, towards which bacteria are less likely to develop resistance. While the majority of antibacterial polymers/oligomers have the ability to disrupt bacterial membranes, the design parameters for the enhanced disruption of peptidoglycan outer layer of Gram-positive bacteria remain unclear. Here, the design of oligomeric structures with favorable conformational characteristics for improved disruption of the peptidoglycan outer layer of Gram-positive bacteria is reported. Molecular dynamics simulations were employed to inform the structure design and composition of cationic oligomers displaying collapsed and expanded conformations. The most promising diblock and triblock cationic oligomers were synthesized by photo-induced atom transfer radical polymerization (photo ATRP). Following synthesis, the diblock and triblock oligomers displayed average antibacterial activity of ~99% and ~98% for S. aureus and methicillin-resistant S. aureus (MRSA), respectively, at the highest concentrations tested. Importantly, triblock oligomers with extended conformations showed significantly higher disruption of the peptidoglycan outer layer of S. aureus compared to diblock oligomers with more collapsed conformation, as evidenced by a number of characterization techniques including scanning electron, confocal and atomic force microscopy. This work provides new insight into the structure/property relationship of antibacterial materials and advances the design of functional materials for combating the rise of drug-resistant bacteria.

Water-level fluctuations can have significant effects on lake biological communities. Thirlmere Lakes are a group of five interconnected lakes located near Sydney. Water levels in Thirlmere Lakes have fluctuated over time, but there has been a recent decline that is of significant concern. In this study, we examined over one year the species composition and richness of zooplankton (Rotifera, Cladocera and Copepoda) and abiotic conditions in Lakes Nerrigorang and Werri Berri, two of the five Thirlmere lakes, with reference to lake water level. We recorded a total of 66 taxa of zooplankton, with the first report of the rotifer Notommata saccigera from Australia, and the first report of the rotifers Keratella javana, Lecane rhytida and Rousseletia corniculata from New South Wales. There was a marked difference in abiotic conditions between the two lakes, with more variable conditions in Lake Nerrigorang. There was a significant positive correlation between zooplankton species richness and lake water level but only for Lake Nerrigorang. Although the two lakes are closely situated and thought to be potentially connected at high water levels, they show distinct ecological characters and the effect of water-level fluctuations on zooplankton species richness seems to differ between the lakes.

Pimelea poisoning of cattle, historically known as St. George Disease or Marree Disease, is a prevailing issue in arid grazing regions of inland Australia. Ingestion of the toxic native Pimelea species that contain the secondary metabolite simplexin, a diterpene orthoester with potent protein kinase C activity, induces diarrhoea, characteristic oedema and potentially fatal right-sided heart failure in cattle. Outbreaks of toxic Pimelea in the grazing field depend on seasonal prevalence. However, all stages of the plant carry the toxin, from seeds, juvenile plants to dead plant material. Livestock generally avoid consuming green Pimelea plants and only consume toxic Pimelea when pasture is minimal or where Pimelea growing through grass tussocks results in inadvertent ingestion. Our knowledge base of Pimelea poisoning has greatly improved with past research, yet the health hazards for livestock grazing in Pimelea affected pastures remains a significant issue whilst the ongoing search to develop effective strategies to mitigate poisoning continues. The goal of this review is to collate historical and recent research giving an overview of the current understandings of Pimelea poisoning, the toxin, its toxic effects and progress made towards remedies to alleviate the effects of Pimelea intoxication.

Methanolic extracts of liana of Caesalpinia welwitschiana and leaves of C. bonduc were found to possess moderate antifeedant and ovicidal activities against Tuta absoluta. Bioassay-guided isolation of constituents from the most active fraction of C. welwitschiana led to the identification of four known compounds [isobonducellin 1a and bonducellin 1 b, intricatinol 2, (-)-epigallocatechin-3-O-gallate 4] and one new constituent [welwitschianic acid 3]. The most active fraction of C. bonduc afforded two known constituents neocaesalpin L 5 and neocaesalpin A 6. The isolated structures were elucidated on the basis of their MS, UV, IR and 1 & 2 D NMR spectra and by comparison with literature data. Compounds 2, 4-6 were showed antifeedant and ovicidal properties against T. absoluta, some comparable to that of azadirachtin at 50, 100 and 200 ng/µl. Overall, the present study, conclude that the two species of the plant could be a promising source of eco-friendly botanical constituents.

Antibiotic resistance is a global human health threat, causing routine treatments of bacterial infections to become increasingly difficult. The problem is exacerbated by biofilm formation by bacterial pathogens on the surfaces of indwelling medical and dental devices that facilitate high levels of tolerance to antibiotics. The development of new antibacterial nanostructured surfaces shows excellent prospects for application in medicine as next-generation biomaterials. The physico-mechanical interactions between these nanostructured surfaces and bacteria lead to bacterial killing or prevention of bacterial attachment and subsequent biofilm formation, and thus are promising in circumventing bacterial infections. This Review explores the impact of surface roughness on the nanoscale in preventing bacterial colonization of synthetic materials and categorizes the different mechanisms by which various surface nanopatterns exert the necessary physico-mechanical forces on the bacterial cell membrane that will ultimately result in cell death.

Circulating cancer-derived small extracellular vesicles (EVs) are nanoscale membranous vesicles shed from cancer cells that are released into surrounding body fluids. Small EVs contain biomolecules associated with cancer such as DNA and proteins for cell-to-cell communication. Therefore, small EVs have been regarded as important cancer biomarkers for liquid biopsy-based cancer diagnosis and drug treatment monitoring. However, because of the high heterogeneity and low level of small EVs in body fluids, there is a high demand for sensitive detection and characterization of such vesicles at a molecular level. In this study, we have developed a sensitive and effective approach to simultaneously profile multiple protein biomarkers expressed on cancer-derived small EVs using surface-enhanced Raman spectroscopy (SERS) nanotags in a single test, without complex isolation steps. Rapid and multiplexed phenotypic profiling of small EVs is achieved by mixing specific detection antibody-coated SERS nanotags, filtered conditioned EV-suspended medium (conditioned EVs), and capture antibody (CD63)-conjugated magnetic beads to form a sandwich immunoassay. As a proof-of-concept demonstration, we applied this approach to characterize pancreatic cancer-derived EVs by simultaneously detecting three specific EV surface receptors including Glypican-1, epithelial cell adhesion molecules (EpCAMs), and CD44 variant isoform 6 (CD44V6). The sensitivity of this method was measured down to 2.3 × 106 particles/mL, which is more sensitive and shows higher multiplexing capability than most other reported EV profiling techniques, such as western blot, enzyme-linked immunosorbent assay, and flow cytometry. Furthermore, phenotypic profiling of small EVs from colorectal cancer and bladder cancer cell lines (SW480 and C3) was conducted and compared to those derived from pancreatic cancer (Panc-1), highlighting the significant difference in EV phenotypes for various cancer cell types suspended in both phosphate-buffered saline and plasma. Thus, we believe that this technology enables a comprehensive evaluation of small secreted EV heterogeneity with high sensitivity, offering strong potential for accurate noninvasive cancer diagnosis and monitoring of drug treatment. In addition, this assay provides point-of-care use because of the easy sample preparation and portable nature of the Raman spectrometer.

Different extracts prepared from wastes (barks, branches, needles, or leaves) of different trees (grey alder, balsam fir, American larch, red maple, sugar maple, white spruce, black spruce, jack pine, white pine, quaking aspen, sweet cherry, and northern red oak) were investigated for their potential use as antibacterial agents for the management of lettuce varnish spot and bacterial leaf spot caused by Pseudomonas cichorii (Swingle) Stapp and Xanthomonas campestris pv. vitians (Brown) Dye, respectively. Extracts were first screened for their antibacterial activities against P. cichorii and X. campestris pv. vitians using the in vitro disk diffusion assay. Based on the diameter of the inhibition zone, ethanol (95%) extract prepared from sugar maple autumn-shed leaves (SMASL) and aqueous ethanol (50%, v/v) extracts prepared from SMASL and from sugar maple green leaves showed the strongest antibacterial activities. Ethanol (95%) SMASL extract was further investigated for its efficacy to manage bacterial diseases when applied on lettuce plants grown in the greenhouse. Foliar application of ethanol (95%) SMASL extract at a concentration of 3.2 g L−1 was shown to significantly (P ≤ 0.05) reduce bacterial leaf spot severity compared with the control without causing phytotoxicity symptoms that could prevent the commercial marketing of the lettuce. Ethanol (95%) SMASL extract (1.6 and 3.2 g L−1) was also shown to significantly reduce varnish spot severity in one experiment out of two. This study identifies for the first time the possibility of exploiting SMASL to manage bacterial diseases affecting horticultural crops.

The great gerbil (Rhombomys opimus) is a social rodent living in permanent, complex burrow systems distributed throughout Central Asia, where it serves as the main host of several important vector-borne infectious pathogens including the well-known plague bacterium (Yersinia pestis). Here, we present a continuous annotated genome assembly of the great gerbil, covering over 96% of the estimated 2.47-Gb genome. Taking advantage of the recent genome assemblies of the sand rat (Psammomys obesus) and the Mongolian gerbil (Meriones unguiculatus), comparative immunogenomic analyses reveal shared gene losses within TLR gene families (i.e., TLR8, TLR10, and the entire TLR11-subfamily) for Gerbillinae, accompanied with signs of diversifying selection of TLR7 and TLR9. Most notably, we find a great gerbil-specific duplication of the MHCII DRB locus. In silico analyses suggest that the duplicated gene provides high peptide binding affinity for Yersiniae epitopes as well as Leishmania and Leptospira epitopes, putatively leading to increased capability to withstand infections by these pathogens. Our study demonstrates the power of whole-genome sequencing combined with comparative genomic analyses to gain deeper insight into the immunogenomic landscape of the great gerbil and its close relatives.

Antibiotic resistance has made the treatment of biofilm-related infections challenging. As such, the quest for next-generation antimicrobial technologies must focus on targeted therapies to which pathogenic bacteria cannot develop resistance. Stimuli-responsive therapies represent an alternative technological focus due to their capability of delivering targeted treatment. This study provides a proof-of-concept investigation into the use of magneto-responsive gallium-based liquid metal (LM) droplets as antibacterial materials, which can physically damage, disintegrate, and kill pathogens within a mature biofilm. Once exposed to a low-intensity rotating magnetic field, the LM droplets become physically actuated and transform their shape, developing sharp edges. When placed in contact with a bacterial biofilm, the movement of the particles resulting from the magnetic field, coupled with the presence of nanosharp edges, physically ruptures the bacterial cells and the dense biofilm matrix is broken down. The antibacterial efficacy of the magnetically activated LM particles was assessed against both Gram-positive and Gram-negative bacterial biofilms. After 90 min over 99% of both bacterial species became nonviable, and the destruction of the biofilms was observed. These results will impact the design of next-generation, LM-based biofilm treatments.

The factors leading to reintroduction success are rarely determined in species translocation programmes attempting to prevent extirpations. Examining interactions between translocated fish and ecological aspects will provide information to increase success of future reintroduction efforts. The zooplankton are a key food source for small-bodied fishes; therefore, the examination of prey availability may contribute useful insight. Fish that are ecological specialists may have preference for invertebrates associated with specific habitat conditions. Food availability for ecological specialists also may be influenced by the presence of alien fishes, through competition for prey and habitat exclusion, including invasive Eastern Gambusia (Gambusia holbrooki) which is linked to the decline of numerous small-bodied fishes. The Murray–Darling Basin in south-eastern Australia is a highly regulated floodplain system where native fishes have declined over recent decades, especially ecological specialists adapted to ephemeral wetlands with natural flow regimes. Yarra Pygmy Perch (Nannoperca obscura) is an ecological specialist that was extirpated in 2008 during a prolonged drought. The objective of this study is to examine dietary aspects of reintroduced Yarra Pygmy Perch as related to zooplankton prey availability and the potential for competition with Eastern Gambusia. The study demonstrated that prey was being consumed by Yarra Pygmy Perch within 24 h of release and provides evidence of overlap in diet with cohabiting Eastern Gambusia. The findings provide direction for further study and a new understanding regarding reintroduction ecology of threatened small-bodied wetland fishes to aid species recovery and persistence.

A fungal biofilm refers to the agglomeration of fungal cells surrounded by a polymeric extracellular matrix (ECM). The ECM is composed primarily of polysaccharides that facilitate strong surface adhesion, proliferation, and cellular protection from the surrounding environment. Biofilms represent the majority of known microbial communities, are ubiquitous, and are found on a multitude of natural and synthetic surfaces. The compositions, and in-turn nanomechanical properties, of fungal biofilms remain poorly understood, because these systems are complex, composed of anisotropic cellular and extracellular material, and importantly are species and environment dependent. Therefore, genomic variation, and/or mutations, as well as environmental and growth factors can change the composition of a fungal cell's biofilm. In this work, we probe the physico-mechanical and biochemical properties of two fungal species, Candida albicans (C. albicans) and Cryptococcus neoformans (C. neoformans), as well as two antifungal resistant sub-species of C. neoformans, fluconazole-resistant C. neoformans (FlucRC. neoformans) and amphotericin B-resistant C. neoformans (AmBRC. neoformans). A new experimental methodology of characterization is proposed, employing a combination of atomic force microscopy (AFM), instrumented nanoindentation, and Synchrotron ATR-FTIR measurements. This allowed the nano-mechanical and chemical characterisation of each fungal biofilm.

Sorbicillinoids are a large family of fungal secondary metabolites with a diverse range of structures and numerous bioactivites, some of which have pharmaceutical potential. The flavin-dependent monooxygenase SorD from Penicillium chrysogenum (PcSorD) utilizes sorbicillinol to catalyze a broad scope of reactions: formation of oxosorbicillinol and epoxysorbicillinol; intermolecular Diels-Alder and Michael-addition dimerization reactions; and dimerization of a sorbicillinol derivative with oxosorbicillinol. PcSorD shares only 18.3% sequence identity with SorD from Trichoderma reesei (TrSorD) and yet unexpectedly catalyzes many of the same reactions, however, the formation of oxosorbicillinol and bisvertinolone by PcSorD extends the range of reactions catalyzed by a single enzyme. Phylogenetic analysis indicates that PcSorD and TrSorD bind the flavin cofactor covalently but via different residues and point mutations confirm these residues are essential for activity.

The biosynthetic gene cluster of the antifungal metabolite sporothriolide 1 was identified from three producing ascomycetes: Hypomontagnella monticulosa MUCL 54604, H. spongiphila CLL 205 and H. submonticulosa DAOMC 242471. A transformation protocol was established, and genes encoding a fatty acid synthase subunit and a citrate synthase were simultaneously knocked out which led to loss of sporothriolide and sporochartine production. In vitro reactions showed that the sporochartines are derived from non-enzymatic Diels–Alder cycloaddition of 1 and trienylfuranol A 7 during the fermentation and extraction process. Heterologous expression of the spo genes in Aspergillus oryzae then led to the production of intermediates and shunts and delineation of a new fungal biosynthetic pathway originating in fatty acid biosynthesis. Finally, a hydrolase was revealed by in vitro studies likely contributing towards self-resistance of the producer organism., A new family of fungal biosynthetic pathways is elucidated based on the use of fatty acid and citrate-like intermediates.

In Climate Leviathan, Mann and Wainwright address the political implications of climate change by theorizing four possible planetary futures: Climate Leviathan as capitalist planetary sovereignty, Climate Mao as non-capitalist planetary sovereignty, Climate Behemoth as capitalist non-planetary sovereignty, and Climate X as non-capitalist non-planetary sovereignty. The authors of the present article agree that the depth and scale of destabilizations induced by climate change cannot be navigated justly from within the present social-political-economic system. We disagree, however, on which of the non-capitalist orientations is better suited for generating viable alternatives to the worst dystopian futures. The article thus stages a debate to elucidate the theoretical and political divergence between Climate X and Climate Mao (renamed Climate Jacobin).

Glyceryl trinitrate (GTN) is one of the earliest known treatments for angina with a fascinating history that bridges three centuries. However, despite its central role in the nitric oxide (NO) story as a NO-donating compound, establishing the precise mechanism of how GTN exerts its medicinal benefit has proven to be far more difficult. This review brings together the explosive and vasodilatory nature of this three-carbon molecule while providing an update on the likely in vivo pathways through which GTN, and the rest of the organic nitrate family, release NO, nitrite, or a combination of both, while also trying to explain nitrate tolerance. Over the last 20 years the alcohol detoxification enzyme, aldehyde dehydrogenase (ALDH), has undoubtedly emerged as the front runner to explaining GTN’s bioactivation. This is best illustrated by reduced GTN efficacy in subjects carrying the single point mutation (Glu504Lys) in ALDH, which is also responsible for alcohol intolerance, as characterized by flushing. While these findings are significant for anyone following the GTN story, they appear particularly relevant for healthcare professionals, and especially so, if administering GTN to patients as an emergency treatment. In short, although the GTN puzzle has not been fully solved, clinical study data continue to cement the importance of ALDH, as uncovered in 2002, as a key GTN activator.

Most patients with rare diseases do not receive a molecular diagnosis and the aetiological variants and causative genes for more than half such disorders remain to be discovered1. Here we used whole-genome sequencing (WGS) in a national health system to streamline diagnosis and to discover unknown aetiological variants in the coding and non-coding regions of the genome. We generated WGS data for 13,037 participants, of whom 9,802 had a rare disease, and provided a genetic diagnosis to 1,138 of the 7,065 extensively phenotyped participants. We identified 95 Mendelian associations between genes and rare diseases, of which 11 have been discovered since 2015 and at least 79 are confirmed to be aetiological. By generating WGS data of UK Biobank participants2, we found that rare alleles can explain the presence of some individuals in the tails of a quantitative trait for red blood cells. Finally, we identified four novel non-coding variants that cause disease through the disruption of transcription of ARPC1B, GATA1, LRBA and MPL. Our study demonstrates a synergy by using WGS for diagnosis and aetiological discovery in routine healthcare.

Background: Female childhood cancer survivors (CCSs) carry a risk of therapy-related gonadal dysfunction. Alkylating agents (AA) are well-established risk factors, yet inter-individual variability in ovarian function is observed. Polymorphisms in CYP450 enzymes may explain this variability in AA-induced ovarian damage. We aimed to evaluate associations between previously identified genetic polymorphisms in CYP450 enzymes and AA-related ovarian function among adult CCSs. Methods: Anti-Müllerian hormone (AMH) levels served as a proxy for ovarian function in a discovery cohort of adult female CCSs, from the pan-European PanCareLIFE cohort (n = 743, age (years): median 25.8, interquartile range (IQR) 22.1–30.6). Using two additive genetic models in linear and logistic regression, nine genetic variants in three CYP450 enzymes were analyzed in relation to cyclophosphamide equivalent dose (CED) score and their impact on AMH levels. The main model evaluated the effect of the variant on AMH and the interaction model evaluated the modifying effect of the variant on the impact of CED score on log-transformed AMH levels. Results were validated, and meta-analysis performed, using the USA-based St. Jude Lifetime Cohort (n = 391, age (years): median 31.3, IQR 26.6–37.4). Results: CYP3A4*3 was significantly associated with AMH levels in the discovery and replication cohort. Meta-analysis revealed a significant main deleterious effect (Beta (95% CI): −0.706 (−1.11–−0.298), p-value = 7 × 10−4) of CYP3A4*3 (rs4986910) on log-transformed AMH levels. CYP2B6*2 (rs8192709) showed a significant protective interaction effect (Beta (95% CI): 0.527 (0.126–0.928), p-value = 0.01) on log-transformed AMH levels in CCSs receiving more than 8000 mg/m2 CED. Conclusions: Female CCSs CYP3A4*3 carriers had significantly lower AMH levels, and CYP2B6*2 may have a protective effect on AMH levels. Identification of risk-contributing variants may improve individualized counselling regarding the treatment-related risk of infertility and fertility preservation options.

Marine fungi remain poorly covered in global genome sequencing campaigns; the 1000 fungal genomes (1KFG) project attempts to shed light on the diversity, ecology and potential industrial use of overlooked and poorly resolved fungal taxa. This study characterizes the genomes of three marine fungi:Emericellopsissp. TS7, wood-associatedAmylocarpus encephaloidesand algae-associatedCalycina marina.These species were genome sequenced to study their genomic features, biosynthetic potential and phylogenetic placement using multilocus data.Amylocarpus encephaloidesandC. marinawere placed in theHelotiaceaeandPezizellaceae (Helotiales), respectively, based on a 15-gene phylogenetic analysis. These two genomes had fewer biosynthetic gene clusters (BGCs) and carbohydrate active enzymes (CAZymes) thanEmericellopsissp. TS7 isolate.Emericellopsissp. TS7 (Hypocreales,Ascomycota) was isolated from the spongeStelletta normani. A six-gene phylogenetic analysis placed the isolate in the marineEmericellopsisclade and morphological examination confirmed that the isolate represents a new species, which is described here asE. atlantica. Analysis of its CAZyme repertoire and a culturing experiment on three marine and one terrestrial substrates indicated thatE. atlanticais a psychrotrophic generalist fungus that is able to degrade several types of marine biomass. FungiSMASH analysis revealed the presence of 35 BGCs including, eight non-ribosomal peptide synthases (NRPSs), six NRPS-like, six polyketide synthases, nine terpenes and six hybrid, mixed or other clusters. Of these BGCs, only five were homologous with characterized BGCs. The presence of unknown BGCs sets and large CAZyme repertoire set stage for further investigations ofE. atlantica. ThePezizellaceaegenome and the genome of the monotypicAmylocarpusgenus represent the first published genomes of filamentous fungi that are restricted in their occurrence to the marine habitat and form thus a valuable resource for the community that can be used in studying ecological adaptions of fungi using comparative genomics.

The leucine-rich repeat-containing family 8 member A (LRRC8A) is an essential subunit of the volume-regulated anion channel (VRAC). VRAC is critical for cell volume control, but its broader physiological functions remain under investigation. Recent studies in the field indicate that Lrrc8a disruption in the brain astrocytes reduces neuronal excitability, impairs synaptic plasticity and memory, and protects against cerebral ischemia. In the present work, we generated brain-wide conditional LRRC8A knockout mice (LRRC8A bKO) using NestinCre -driven Lrrc8aflox/flox excision in neurons, astrocytes, and oligodendroglia. LRRC8A bKO animals were born close to the expected Mendelian ratio and developed without overt histological abnormalities, but, surprisingly, all died between 5 and 9 weeks of age with a seizure phenotype, which was confirmed by video and EEG recordings. Brain slice electrophysiology detected changes in the excitability of pyramidal cells and modified GABAergic inputs in the hippocampal CA1 region of LRRC8A bKO. LRRC8A-null hippocampi showed increased immunoreactivity of the astrocytic marker GFAP, indicating reactive astrogliosis. We also found decreased whole-brain protein levels of the GABA transporter GAT-1, the glutamate transporter GLT-1, and the astrocytic enzyme glutamine synthetase. Complementary HPLC assays identified reduction in the tissue levels of the glutamate and GABA precursor glutamine. Together, these findings suggest that VRAC provides vital control of brain excitability in mouse adolescence. VRAC deletion leads to a lethal phenotype involving progressive astrogliosis and dysregulation of astrocytic uptake and supply of amino acid neurotransmitters and their precursors.
