Objective Variants of concern (VOCs) associated with relatively high transmissibility appear to be spreading rapidly in Gabon. Therefore, it is imperative to understand the distribution of several variants of concern in the population, which could have implications for transmissibility and vaccine efficacy. Methods Between February and May 2021, SARS-CoV-2 genomes were sequenced using the Oxford nanopore MinION method and the respective genome diversity was elucidated. Phylogenetic analysis was performed and genomes were classified using pangolin lineages. Results The results highlight the increase (46%) of the alpha variant of concern (B.1.1.7) in the Gabonese population over the study period. In addition, an increase (31%) in the B.1.1.318 lineage, which is associated with high transmission and impaired vaccine efficacy (D614G+E484K+Y144del), was detected. Conclusion With the second wave ongoing, our findings highlight the need for surveillance of the SARS-CoV-2 genome in the Republic of Gabon and should provide useful guidance to policy makers in selecting an appropriate vaccine for the population.

To overcome these limitations, we established a high-throughput platform for the quantitative and qualitative analysis of human TCR repertoires that provides insights into the clonal and functional composition of human CD4(+) and CD8(+) αβ T cells at the molecular and cellular level. The strategy is a powerful tool to qualitatively assess differences between antigen receptors of phenotypically defined αβ T cell subsets, e.g. in immune responses to cancer, vaccination, or infection, and in autoimmune diseases.

Plants produce many secondary metabolites showing considerable inter- and intraspecific diversity of concentration and composition as a strategy to cope with environmental stresses. The evolution of plant defenses against herbivores and pathogens can be unraveled by understanding the mechanisms underlying chemical diversity. Pyrrolizidine alkaloids are a class of secondary metabolites with high diversity. We performed a qualitative and quantitative analysis of 80 pyrrolizidine alkaloids with liquid chromatography-tandem mass spectrometry of leaves from 17 Jacobaea species including one to three populations per species with 4–10 individuals per population grown under controlled conditions in a climate chamber. We observed large inter- and intraspecific variation in pyrrolizidine alkaloid concentration and composition, which were both species-specific. Furthermore, we sequenced 11 plastid and three nuclear regions to reconstruct the phylogeny of the 17 Jacobaea species. Ancestral state reconstruction at the species level showed mainly random distributions of individual pyrrolizidine alkaloids. We found little evidence for phylogenetic signals, as nine out of 80 pyrrolizidine alkaloids showed a significant phylogenetic signal for Pagel's λ statistics only, whereas no significance was detected for Blomberg's K measure. We speculate that this high pyrrolizidine alkaloid diversity is the result of the upregulation and downregulation of specific pyrrolizidine alkaloids depending on ecological needs rather than gains and losses of particular pyrrolizidine alkaloid biosynthesis genes during evolution.

Anthropogenic climate change is resulting in spatial redistributions of many species. We assessed the potential effects of climate change on an abundant and widely distributed group of diving birds, Eudyptes penguins, which are the main avian consumers in the Southern Ocean in terms of biomass consumption. Despite their abundance, several of these species have undergone population declines over the past century, potentially due to changing oceanography and prey availability over the important winter months. We used light-based geolocation tracking data for 485 individuals deployed between 2006 and 2020 across 10 of the major breeding locations for five taxa of Eudyptes penguins. We used boosted regression tree modelling to quantify post-moult habitat preference for southern rockhopper (E. chrysocome), eastern rockhopper (E. filholi), northern rockhopper (E. moseleyi) and macaroni/royal (E. chrysolophus and E. schlegeli) penguins. We then modelled their redistribution under two climate change scenarios, representative concentration pathways RCP4.5 and RCP8.5 (for the end of the century, 2071-2100). As climate forcings differ regionally, we quantified redistribution in the Atlantic, Central Indian, East Indian, West Pacific and East Pacific regions. We found sea surface temperature and sea surface height to be the most important predictors of current habitat for these penguins; physical features that are changing rapidly in the Southern Ocean. Our results indicated that the less severe RCP4.5 would lead to less habitat loss than the more severe RCP8.5. The five taxa of penguin may experience a general poleward redistribution of their preferred habitat, but with contrasting effects in the (i) change in total area of preferred habitat under climate change (ii) according to geographic region and (iii) the species (macaroni/royal vs. rockhopper populations). Our results provide further understanding on the regional impacts and vulnerability of species to climate change.

The planetary system around the naked-eye star $\nu^2$ Lupi (HD 136352; TOI-2011) is composed of three exoplanets with masses of 4.7, 11.2, and 8.6 Earth masses. The TESS and CHEOPS missions revealed that all three planets are transiting and have radii straddling the radius gap separating volatile-rich and volatile-poor super-earths. Only a partial transit of planet d had been covered so we re-observed an inferior conjunction of the long-period 8.6 Earth-mass exoplanet $\nu^2$ Lup d with the CHEOPS space telescope. We confirmed its transiting nature by covering its whole 9.1 h transit for the first time. We refined the planet transit ephemeris to P = 107.1361 (+0.0019/-0.0022) days and Tc = 2,459,009.7759 (+0.0101/-0.0096) BJD_TDB, improving by ~40 times on the previously reported transit timing uncertainty. This refined ephemeris will enable further follow-up of this outstanding long-period transiting planet to search for atmospheric signatures or explore the planet's Hill sphere in search for an exomoon. In fact, the CHEOPS observations also cover the transit of a large fraction of the planet's Hill sphere, which is as large as the Earth's, opening the tantalising possibility of catching transiting exomoons. We conducted a search for exomoon signals in this single-epoch light curve but found no conclusive photometric signature of additional transiting bodies larger than Mars. Yet, only a sustained follow-up of $\nu^2$ Lup d transits will warrant a comprehensive search for a moon around this outstanding exoplanet., Comment: 16 pages, 9 figures. Context. Tidal orbital decay is suspected to occur for hot Jupiters in particular, with the only observationally confirmed case of this being WASP-12 b. By examining this effect, information on the properties of the host star can be obtained using the so-called stellar modified tidal quality factor Q′∗, which describes the efficiency with which the kinetic energy of the planet is dissipated within the star. This can provide information about the interior of the star. Aims. In this study, we aim to improve constraints on the tidal decay of the KELT-9, KELT-16, and WASP-4 systems in order to find evidence for or against the presence of tidal orbital decay. With this, we want to constrain the Q′∗ value for each star. In addition, we aim to test the existence of the transit timing variations (TTVs) in the HD 97658 system, which previously favoured a quadratic trend with increasing orbital period. Methods. Making use of newly acquired photometric observations from CHEOPS (CHaracterising ExOplanet Satellite) and TESS (Transiting Exoplanet Survey Satellite), combined with archival transit and occultation data, we use Markov chain Monte Carlo (MCMC) algorithms to fit three models to the data, namely a constant-period model, an orbital-decay model, and an apsidal-precession model. Results. We find that the KELT-9 system is best described by an apsidal-precession model for now, with an orbital decay trend at over 2 σ being a possible solution as well. A Keplerian orbit model with a constant orbital period provides the best fit to the transit timings of KELT-16 b because of the scatter and scale of their error bars. The WASP-4 system is best represented by an orbital decay model at a 5 σ significance, although apsidal precession cannot be ruled out with the present data. Aims. DE Boo is a unique system, with an edge-on view through the debris disk around the star. The disk, which is analogous to the Kuiper belt in the Solar System, was reported to extend from 74 to 84 AU from the central star. The high photometric precision of the Characterising Exoplanet Satellite (CHEOPS) provided an exceptional opportunity to observe small variations in the light curve due to transiting material in the disk. This is a unique chance to investigate processes in the debris disk. Methods. Photometric observations of DE Boo of a total of four days were carried out with CHEOPS. Photometric variations due to spots on the stellar surface were subtracted from the light curves by applying a two-spot model and a fourth-order polynomial. The photometric observations were accompanied by spectroscopic measurements with the 1m RCC telescope at Piszkésteto and with the SOPHIE spectrograph in order to refine the astrophysical parameters of DE Boo. Results. We present a detailed analysis of the photometric observation of DE Boo. We report the presence of nonperiodic transient features in the residual light curves with a transit duration of 0.3-0.8 days. We calculated the maximum distance of the material responsible for these variations to be 2.47 AU from the central star, much closer than most of the mass of the debris disk. Furthermore, we report the first observation of flaring events in this system. Conclusions. We interpreted the transient features as the result of scattering in an inner debris disk around DE Boo. HD$\,$172555 is a young ($\sim$20$\,$Myr) A7V star surrounded by a 10$\,$au wide debris disk suspected to be replenished partly by collisions between large planetesimals. Small evaporating transiting bodies, exocomets, have also been detected in this system by spectroscopy. After $\beta\,$Pictoris, this is another example of a system possibly witnessing a phase of heavy bombardment of planetesimals. In such system, small bodies trace dynamical evolution processes. We aim at constraining their dust content by using transit photometry. We performed a 2-day-long photometric monitoring of HD$\,$172555 with the CHEOPS space telescope in order to detect shallow transits of exocomets with a typical expected duration of a few hours. The large oscillations in the lightcurve indicate that HD$\,$172555 is a $\delta\,$Scuti pulsating star. Once removing those dominating oscillations, we find a hint for a transient absorption. If fitted with an exocomet transit model, it corresponds to an evaporating body passing near the star at a distance of $6.8\pm1.4\,$R$_\star$ (or $0.05\pm 0.01\,$au) with a radius of 2.5 km. These properties are comparable to those of the exocomets already found in this system using spectroscopy, as well as those found in the $\beta\,$Pic system. The nuclei of solar system's Jupiter family comets, with radii of 2-6$\,$km, are also comparable in size. Context. Measurements of the occultation of an exoplanet at visible wavelengths allow us to determine the reflective properties of a planetary atmosphere. The observed occultation depth can be translated into a geometric albedo. This in turn aids in characterising the structure and composition of an atmosphere by providing additional information on the wavelength-dependent reflective qualities of the aerosols in the atmosphere. Aims. Our aim is to provide a precise measurement of the geometric albedo of the gas giant HD 189733b by measuring the occultation depth in the broad optical bandpass of CHEOPS (350 - 1100 nm). Methods. We analysed 13 observations of the occultation of HD 189733b performed by CHEOPS utilising the Python package PyCHEOPS. The resulting occultation depth is then used to infer the geometric albedo accounting for the contribution of thermal emission from the planet. We also aid the analysis by refining the transit parameters combining observations made by the TESS and CHEOPS space telescopes. Results. We report the detection of an $24.7 \pm 4.5$ ppm occultation in the CHEOPS observations. This occultation depth corresponds to a geometric albedo of $0.076 \pm 0.016$. Our measurement is consistent with models assuming the atmosphere of the planet to be cloud-free at the scattering level and absorption in the CHEOPS band to be dominated by the resonant Na doublet. Taking into account previous optical-light occultation observations obtained with the Hubble Space Telescope, both measurements combined are consistent with a super-stellar Na elemental abundance in the dayside atmosphere of HD 189733b. Sooty Albatrosses (Phoebetria fusca; Endangered) are biennially breeding birds with successful breeders typically spending at least 15 months at-sea (‘sabbatical’) before returning to their breeding grounds on sub-Antarctic islands. Stable isotope analysis of feathers suggests that non-breeding adult Sooty Albatrosses moult in sub-tropical waters, north of the Sub-Tropical Front (STF). The Prince Edward Islands (Marion and Prince Edward) provide nesting grounds for ca 24% of the world’s Sooty Albatrosses. We tracked 20 adult Sooty Albatrosses from Marion Island with geolocators (GLS loggers) and satellite transmitters (PTT) during their non-breeding sabbaticals between 2008 and 2014. Stable isotope analysis also was performed on feathers collected from GLS-tracked birds upon device retrieval. Adult birds mostly remained within international waters in the southern Indian Ocean during their sabbatical period, splitting their time between sub-tropical and sub-Antarctic waters. Sooty Albatrosses were more active during the day on average but spent similar time in flight during full moon periods. Periods of reduced flight activity, measured by time on water, suggest that moulting occurs mainly around the STF. Breeding success influenced moult phenology, with unsuccessful birds moulting in late summer, immediately following a failed breeding attempt (Feb–Mar), whilst successful breeders moulted early the following summer (Oct–Dec). Failed breeders spent more time flying between breeding attempts than successful breeders, particularly whilst moulting. Our study identifies key areas utilised by non-breeding Sooty Albatrosses, which is critical to implement appropriate management strategies that may help population recovery of this endangered species.

Reefs of the flat oyster (Ostrea angasi) were once common along the southern coasts of Australia. Historical and current literature relating to O. angasi was used to identify bays and estuaries where this species once existed. In many estuaries of Western Australia, current populations are significantly lower than historical levels, including in Princess Royal Harbour and Oyster Harbour, near Albany. The main causes of the declines included overfishing, combined with the use of destructive fishing methods, such as dredging. Other factors, such as sedimentation, increased nutrient input and loss of seagrass, may have contributed to the loss of oyster reefs, and may have inhibited effective recovery. The possible impact of the protozoan pathogen Bonamia exitiosa is uncertain, although it is known to have severely affected flat oyster populations in other parts of the world. The fact that O. angasi reefs in Oyster Harbour did not recover after the fishery ceased suggests that restoration activities, aimed at restarting the ecosystem services that the oyster reefs once provided, should be undertaken. This paper suggests that the historical presence of O. angasi could be an effective starting point for prioritising potential restoration sites and details the prioritisation protocol that was used in recent restoration activities.

Binding and unbinding of transcription factors to DNA are kinetically controlled to regulate the transcriptional outcome. Control of the release of the transcription factor NF-κB from DNA is achieved through accelerated dissociation by the inhibitor protein IκBα. Using single-molecule FRET, we observe a continuum of conformations of NF-κB in free and DNA-bound states interconverting on the subseconds to minutes timescale, comparable to in vivo binding on the seconds timescale, suggesting that structural dynamics directly control binding kinetics. Much of the DNA-bound NF-κB is partially bound, allowing IκBα invasion to facilitate DNA dissociation. IκBα induces a locked conformation where the DNA-binding domains of NF-κB are too far apart to bind DNA, whereas the loss-of-function IκBα mutant retains the NF-κB conformational ensemble. Overall, our results suggest a novel mechanism with a continuum of binding modes for controlling association and dissociation of transcription factors.

BACKGROUND Neutrophilic asthma (NA) is a clinically important asthma phenotype, the cellular and molecular basis of which is not completely understood. Airway macrophages are long-lived immune cells that exert important homeostatic and inflammatory functions which are dysregulated in asthma. Unique transcriptomic programmes reflect varied macrophage phenotypes in vitro. We aimed to determine whether airway macrophages are transcriptomically altered in NA. METHODS We performed RNASeq analysis on flow cytometry-isolated sputum macrophages comparing NA (n = 7) and non-neutrophilic asthma (NNA, n = 13). qPCR validation of RNASeq results was performed (NA n = 13, NNA n = 23). Pathway analysis (PANTHER, STRING) of differentially expressed genes (DEGs) was performed. Gene set variation analysis (GSVA) was used to test for enrichment of NA macrophage transcriptomic signatures in whole sputum microarray (cohort 1 - controls n = 16, NA n = 29, NNA n = 37; cohort 2 U-BIOPRED - controls n = 16, NA n = 47, NNA n = 57). SARS-CoV-2 is an endemic positive-sense RNA virus naturally transmissible between numerous species with notable infectivity and associated mortality. It is characterized by a poly-adenylated structure capping the genomic terminus. This poly(A) tail is crucial to a cascade of viral replicative activity occurring both extra- and intra-cellular during infection. As a route to proposing potential chemotherapy, this study suggests simple biplanar adenine quadruplexes (A4s) which may fold in specific sequences of the viral genome. To the best of our knowledge, uniquely biplanar A4s have not been previously described in any context. Using molecular modeling techniques and molecular dynamics simulations, some of these non-canonical structures show reasonable stability in a biological context. Notably, mRNA configured as a biplanar A4, shows less dynamic activity than DNA equivalents. This observation may be especially relevant in a physiological context. Furthermore, in contrast to well-characterized guanine quadruplexes, co-ordination with cations appears not to impact on stability. Our molecular dynamics simulations and analyses demonstrate that some A4s are stable in biologically relevant terms. These conclusions may apply to SARS-CoV-2, its variants and other pathogenic RNA viruses. © 2021 World Scientific Publishing Company.

BACKGROUND: Chorioamnionitis, an intrauterine infection of the placenta and fetal membranes, is a common risk factor for adverse pulmonary outcomes in premature infants including BPD, which is characterized by an arrest in alveolar development. As endogenous epithelial stem/progenitor cells are crucial for organogenesis and tissue repair, we examined whether intrauterine inflammation negatively affects these essential progenitor pools.METHODS: In an ovine chorioamnionitis model, fetuses were intra-amniotically exposed to LPS, 2d or 7d (acute inflammation) before preterm delivery at 125d of gestation, or to intra-amniotic Ureaplasma parvum for 42d (chronic inflammation). Lung function, pulmonary endogenous epithelial stem/progenitor pools, and downstream functional markers were studied.RESULTS: Lung function was improved in the 7d LPS and 42d Ureaplasma groups. However, intrauterine inflammation caused a loss of P63+ basal cells in proximal airways and reduced SOX-9 expression and TTF-1+ Club cells in distal airways. Attenuated type-2 cell numbers were associated with lower proliferation and reduced type-1 cell marker Aqp5 expression, indicative for impaired progenitor function. Chronic Ureaplasma infection only affected distal airways, whereas acute inflammation affected stem/progenitor populations throughout the lungs.CONCLUSIONS: Acute and chronic prenatal inflammation improve lung function at the expense of stem/progenitor alterations that potentially disrupt normal lung development, thereby predisposing to adverse postnatal outcomes.IMPACT: In this study, prenatal inflammation improved lung function at the expense of stem/progenitor alterations that potentially disrupt normal lung development, thereby predisposing to adverse postnatal outcomes.Importantly, we demonstrate that these essential alterations can already be initiated before birth. Within this context, our data suggest that interventions, which promote function or repair of endogenous stem cells in the lungs, hold great promise.

There are currently fewer than 10 antifungal drugs in clinical development, but new fungal strains that are resistant to most current antifungals are spreading rapidly across the world. To prevent a second resistance crisis, new classes of antifungal drugs are urgently needed. Metal complexes have proven to be promising candidates for novel antibiotics, but so far, few compounds have been explored for their potential application as antifungal agents. In this work, we report the evaluation of 1039 metal-containing compounds that were screened by the Community for Open Antimicrobial Drug Discovery (CO-ADD). We show that 20.9% of all metal compounds tested have antimicrobial activity against two representative Candida and Cryptococcus strains compared with only 1.1% of the >300,000 purely organic molecules tested through CO-ADD. We identified 90 metal compounds (8.7%) that show antifungal activity while not displaying any cytotoxicity against mammalian cell lines or hemolytic properties at similar concentrations. The structures of 21 metal complexes that display high antifungal activity (MIC ≤1.25 μM) are discussed and evaluated further against a broad panel of yeasts. Most of these have not been previously tested for antifungal activity. Eleven of these metal complexes were tested for toxicity in the Galleria mellonella moth larva model, revealing that only one compound showed signs of toxicity at the highest injected concentration. Lastly, we demonstrated that the organo-Pt(II) cyclooctadiene complex $\textbf{Pt1}$ significantly reduces fungal load in an in vivoG. mellonella infection model. These findings showcase that the structural and chemical diversity of metal-based compounds can be an invaluable tool in the development of new drugs against infectious diseases.

The Indian halibut (Psettodes erumei) is an important flatfish species in tropical and subtropical waters of the northern Indian Ocean. However, inappropriate aging methods used in previous studies of Indian halibut limited the use of age and growth data from those studies. This study highlights the importance of correct aging methods and identification of sexually dimorphic growth, particularly if growth characteristics are used to estimate mortality rates. Counts of opaque zones in whole otoliths of Indian halibut were almost always fewer than those in their corresponding sections, particularly for older individuals. Multiple validation methods demonstrate opaque zones form annually in otoliths of Indian halibut. Although the frequency of females increased with size, due to sexually dimorphic growth, males attained a greater maximum age (16 versus 11 years). Because of the greater ages and rapid early growth of Indian halibut described in this study, natural mortality estimates calculated by using Pauly’s equation, particularly for males, were higher than estimates derived from the same equation in previous studies, which employed growth parameter estimates based on ages derived from whole otoliths, vertebrae, or monthly length–frequency trends and, typically, did not account for sexually dimorphic growth. In northwestern Australia, spawning in Indian halibut commences when water temperature and day length begin to increase and occurs between early spring and late summer.

A series of (NHC)Au(I)Cl monocarbene complexes and their gold(III) analogues (NHC)Au(III)Cl3 were prepared and investigated as antibacterial agents and inhibitors of bacterial TrxR. The complexes showed stronger antibacterial effects against the Gram-positive MRSA and E. faecium strains than against several Gram-negative bacteria. All complexes were efficient inhibitors of bacterial thioredoxin reductase, indicating that inhibition of this enzyme might be involved in their mechanism of action. The efficacy of gold(I) and gold(III) analogues was comparable in most of the assays. The cytotoxicity of the gold NHC compounds against cancer and human cells was overall weaker than the activity against the Gram-positive bacteria, suggesting that their optimization as antibacterials warrants further investigation.

Purpose Development and validation of an animal model of labral healing would facilitate translation of novel surgical and biological strategies to improve glenolabral healing. The purpose of this study was to characterize the anatomic and histological properties of the shoulder labrum in rat, rabbit, dog, pig, goat, and humans. Given the demonstrated similarities in size and structural morphology in other joints, it was hypothesized that the goat glenoid with surrounding capsulolabral complex would most closely resemble that of humans in terms of dimensions and structure, as observed grossly and histologically. Methods Cadaveric glenohumeral joints from rats (n = 8), New Zealand white rabbits (n = 13), Mongrel dogs (n = 9), Spanish goats (n = 10), Yorkshire pigs (n = 10), and humans (n = 9) were freshly harvested. Photographs were taken of the glenoid with its surrounding capsulolabral complex. Linear dimensions of the glenoid articular surface were measured. It was determined where the capsulolabral complex was continuous with, or recessed from, the articular glenoid surface. The glenoid was divided into 6 equal segments radiating out toward 12, 2, 4, 6, 8, and 10 o’clock positions. Samples were sectioned and stained with Safranin O/Fast green and Mallory Trichrome. Insertion of the capsulolabral tissue onto the glenoid was qualitatively assessed and compared with gross morphology. Results Dimensions of the goat glenoid most closely paralleled dimensions of the human glenoid. A capsulolabral complex was continuous with the glenoid surface from ~ 9 to 12 o’clock in the rats, 7 to 12 o’clock in rabbits, 5 to 12 o’clock in the dogs, and 9 to 12 o’clock in goats, 6 to 12 o’clock in pigs, and 2 to 8 o’clock in humans. In contrast to humans, no other species demonstrated an organized fibrocartilaginous labrum either macroscopically or histologically. Conclusion The animals in the present study did not possess a discrete fibrocartilaginous labrum by gross or histological evaluation, as directly compared to humans. While models using these animals may be acceptable for examining other shoulder pathologies, they are not adequate to evaluate labral pathology. BACKGROUND: Primary cytomegalovirus (CMV) infection during pregnancy carries a risk of congenital infection and possible severe sequelae. There is no established intervention for preventing congenital CMV infection. METHODS: In this multicenter, double-blind trial, pregnant women with primary CMV infection diagnosed before 24 weeks’ gestation were randomly assigned to receive a monthly infusion of CMV hyperimmune globulin (at a dose of 100 mg per kilogram of body weight) or matching placebo until delivery. The primary outcome was a composite of congenital CMV infection or fetal or neonatal death if CMV testing of the fetus or neonate was not performed. RESULTS: From 2012 to 2018, a total of 206,082 pregnant women were screened for primary CMV infection before 23 weeks of gestation; of the 712 participants (0.35%) who tested positive, 399 (56%) underwent randomization. The trial was stopped early for futility. Data on the primary outcome were available for 394 participants; a primary outcome event occurred in the fetus or neonate of 46 of 203 women (22.7%) in the group that received hyperimmune globulin and of 37 of 191 women (19.4%) in the placebo group (relative risk, 1.17; 95% confidence interval [CI], 0.80 to 1.72; P = 0.42). Death occurred in 4.9% of fetuses or neonates in the hyperimmune globulin group and in 2.6% in the placebo group (relative risk, 1.88; 95% CI, 0.66 to 5.41), preterm birth occurred in 12.2% and 8.3%, respectively (relative risk, 1.47; 95% CI, 0.81 to 2.67), and birth weight below the 5th percentile occurred in 10.3% and 5.4% (relative risk, 1.92; 95% CI, 0.92 to 3.99). One participant in the hyperimmune globulin group had a severe allergic reaction to the first infusion. Participants who received hyperimmune globulin had a higher incidence of headaches and shaking chills while receiving infusions than participants who received placebo. CONCLUSIONS: Among pregnant women, administration of CMV hyperimmune globulin starting before 24 weeks’ gestation did not result in a lower incidence of a composite of congenital CMV infection or perinatal death than placebo. Quantitative functional anatomy of amniote thoracic and abdominal regions is crucial to understanding constraints on and adaptations for facilitating simultaneous breathing and locomotion. Crocodilians have diverse locomotor modes and variable breathing mechanics facilitated by basal and derived (accessory) muscles. However, the inherent flexibility of these systems is not well studied, and the functional specialisation of the crocodilian trunk is yet to be investigated. Increases in body size and trunk stiffness would be expected to cause a disproportionate increase in muscle force demands and therefore constrain the basal costal aspiration mechanism, necessitating changes in respiratory mechanics. Here, we describe the anatomy of the trunk muscles, their properties that determine muscle performance (mass, length and physiological cross‐sectional area [PCSA]) and investigate their scaling in juvenile Alligator mississippiensis spanning an order of magnitude in body mass (359 g–5.5 kg). Comparatively, the expiratory muscles (transversus abdominis, rectus abdominis, iliocostalis), which compress the trunk, have greater relative PCSA being specialised for greater force‐generating capacity, while the inspiratory muscles (diaphragmaticus, truncocaudalis ischiotruncus, ischiopubis), which create negative internal pressure, have greater relative fascicle lengths, being adapted for greater working range and contraction velocity. Fascicle lengths of the accessory diaphragmaticus scaled with positive allometry in the alligators examined, enhancing contractile capacity, in line with this muscle's ability to modulate both tidal volume and breathing frequency in response to energetic demand during terrestrial locomotion. The iliocostalis, an accessory expiratory muscle, also demonstrated positive allometry in fascicle lengths and mass. All accessory muscles of the infrapubic abdominal wall demonstrated positive allometry in PCSA, which would enhance their force‐generating capacity. Conversely, the basal tetrapod expiratory pump (transversus abdominis) scaled isometrically, which may indicate a decreased reliance on this muscle with ontogeny. Collectively, these findings would support existing anecdotal evidence that crocodilians shift their breathing mechanics as they increase in size. Furthermore, the functional specialisation of the diaphragmaticus and compliance of the body wall in the lumbar region against which it works may contribute to low‐cost breathing in crocodilians., The figure shows the head and upper torso of an American alligator.

In mammalian embryos, proper zygotic genome activation (ZGA) underlies totipotent development. Double homeobox (DUX)-family factors participate in ZGA, and mouse Dux is required for forming cultured two-cell (2C)-like cells. Remarkably, in mouse embryonic stem cells, Dux is activated by the tumor suppressor p53, and Dux expression promotes differentiation into expanded-fate cell types. Long-read sequencing and assembly of the mouse Dux locus reveals its complex chromatin regulation including putative positive and negative feedback loops. We show that the p53–DUX/DUX4 regulatory axis is conserved in humans. Furthermore, we demonstrate that cells derived from patients with facioscapulohumeral muscular dystrophy (FSHD) activate human DUX4 during p53 signaling via a p53-binding site in a primate-specific subtelomeric long terminal repeat (LTR)10C element. In summary, our work shows that p53 activation convergently evolved to couple p53 to Dux/DUX4 activation in embryonic stem cells, embryos and cells from patients with FSHD, potentially uniting the developmental and disease regulation of DUX-family factors and identifying evidence-based therapeutic opportunities for FSHD. p53 activates Dux in mouse embryos and embryonic stem cells, as well as DUX4 in human facioscapulohumeral muscular dystrophy cell models.

Suillus is among the best-known examples of an ectomycorrhizal (ECM) fungal genus that demonstrates a high degree of host specificity. Currently recognized host genera of Suillus include Larix, Pinus, and Pseudotsuga, which all belong to the pinoid clade of the family Pinaceae. Intriguingly, Suillus sporocarps have been sporadically collected in forests in which known hosts from these genera are locally absent. To determine the capacity of Suillus to associate with alternative hosts in both the pinoid and abietoid clades of Pinaceae, we examined the host associations of two Suillus species (S. punctatipes and S. glandulosus) through field-based root tip sampling and seedling bioassays. Root tip collections underneath Suillus sporocarps were molecularly identified (fungi: nuc rDNA internal transcribed spacer region ITS1-5.8S-ITS2 [ITS barcode]; plant: trnL) to assess the association with multiple hosts. The bioassays contained both single- and two-species treatments, including a primary (Larix or Pseudotsuga) and a secondary (Picea, Pinus, or Abies) host. For the S. punctatipes bioassay, an additional treatment in which the primary host was removed after 8 mo was included to assess the effect of primary host presence on longer-term ECM colonization. The field-based results confirmed that Suillus fungi were able to associate with Abies and Tsuga hosts, representing novel host genera for this genus. In the bioassays, colonization on the primary hosts was detected in both single- and two-species treatments, but no colonization was present when Picea and Abies hosts were grown alone. Removal of a primary host had no effect on percent ECM colonization, suggesting that primary hosts are not necessary for sustaining Suillus colonization once they are successfully established on secondary hosts. Collectively, our results indicate that host specificity is more flexible in this genus than previously acknowledged and help to explain the presence of Suillus in forests where recognized hosts are not present.

Background Mice with a loss of function mutation in Wdpcp were described previously to display severe birth defects in the developing heart, neural tube, and limb buds. Further characterization of the skeletal phenotype of Wdpcp null mice was limited by perinatal lethality. Results We utilized Prx1-Cre mice to generate limb bud mesenchyme specific deletion of Wdpcp. These mice recapitulated the appendicular skeletal phenotype of the Wdpcp null mice including polydactyl and limb bud signaling defects. Examination of late stages of limb development demonstrated decreased size of cartilage anlagen, delayed calcification, and abnormal growth plates. Utilizing in vitro assays, we demonstrated that loss of Wdpcp in skeletal progenitors lead to loss of hedgehog signaling responsiveness and associated proliferative response. In vitro chondrogenesis assays showed this loss of hedgehog and proliferative response was associated with decreased expression of early chondrogenic marker N-Cadherin. E14.5 forelimbs demonstrated delayed ossification and expression of osteoblast markers Runx2 and Sp7. P0 growth plates demonstrated loss of hedgehog signaling markers and expansion of the hypertrophic zones of the growth plate. In vitro osteogenesis assays demonstrated decreased osteogenic differentiation of Wdpcp null mesenchymal progenitors in response to hedgehog stimulation. Conclusions These findings demonstrate how Wdpcp and associated regulation of the hedgehog signaling pathway plays an important role at multiple stages of skeletal development. Wdpcp is necessary for positive regulation of hedgehog signaling and associated proliferation is key to the initiation of chondrogenesis. At later stages, Wdpcp facilitates the robust hedgehog response necessary for chondrocyte hypertrophy and osteogenic differentiation.

Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis (OA). In this study, we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs (DMOADs). Specifically, human bone marrow-derived mesenchymal stromal cells (MSCs) were expanded in vitro up to passage 10 (P10-MSCs). Following their senescent phenotype formation, P10-MSCs were subjected to pellet culture in chondrogenic medium. Results from qRT-PCR, histology, and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents, when compared to that from normal passage 4 (P4)-MSCs. Interestingly, the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples, as demonstrated by RNA Sequencing data and other analysis methods. Lastly, the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics. The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage. The P4- and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans.

The fibrotic encapsulation of implants involves the mechanical activation of myofibroblasts and of pro-fibrotic transforming growth factor beta 1 (TGF-β1). Here, we show that both softening of the implant surfaces and inhibition of the activation of TGF-β1 reduce the fibrotic encapsulation of subcutaneous silicone implants in mice. Conventionally stiff silicones (elastic modulus, ~2 MPa) coated with a soft silicone layer (elastic modulus, ~2 kPa) reduced collagen deposition as well as myofibroblast activation without affecting the numbers of macrophages and their polarization states. Instead, fibroblasts around stiff implants exhibited enhanced intracellular stress, increased the recruitment of αv and β1 integrins, and activated TGF-β1 signalling. In vitro, the recruitment of αv integrin to focal adhesions and the activation of β1 integrin and of TGF-β were higher in myofibroblasts grown on latency-associated peptide (LAP)-coated stiff silicones than on soft silicones. Antagonizing αv integrin binding to LAP through the small-molecule inhibitor CWHM-12 suppressed active TGF-β signalling, myofibroblast activation and the fibrotic encapsulation of stiff subcutaneous implants in mice. The fibrotic encapsulation of subcutaneous silicone implants in mice can be suppressed by softening the implant surface or inhibiting the integrin-mediated activation of transforming growth factor beta 1.

Cryptorchidism causes spermatogenic failure and reduced serum androgen levels, as well as testicular oedema and fibrosis, which are hallmarks of inflammation. However, the role of inflammation and the effects of cryptorchidism on Sertoli cell and Leydig cell function at the molecular level remain ill-defined. Bilateral cryptorchidism was surgically induced in adult rats for 7 and 14 weeks. Testis weights decreased to 40% of normal within 7 weeks, due to loss of all developing spermatogenic cells except spermatogonia, but did not decrease further at 14 weeks. Serum FSH and LH were increased at both time points, consistent with a loss of feedback by inhibin and testosterone. This damage was accompanied by progressive accumulation of interstitial fluid and peritubular fibrosis, and a progressive decline of several critical Sertoli cell genes (Sox9, Inha (inhbin α-subunit), Cldn11 (claudin 11), Gja1 (connexin 43), and Il1a (interleukin-1α)) and the Leydig cell steroidogenic enzymes, Cyp11a1, Hsd3b1, and Hs17b3. Activin B and the activin-binding protein, follistatin, also declined, but the intratesticular concentration of activin A, which is a regulator of inflammatory responses, was largely unaffected at either time point. Expression of genes involved in inflammation (Tnf, Il10, Il1b, Mcp1) and fibrosis (Acta2, Col1a1) were considerably elevated at both time points. These data indicate that induction of experimental cryptorchidism, which causes complete failure of spermatogenesis in the adult rat, also induces chronic testicular inflammation, manifesting in oedema and fibrosis, and a progressive decline of Sertoli and Leydig cell gene expression and function.

Chironomids are a useful group for investigating body size responses to warming due to their high local abundance and sensitivity to environmental change. We collected specimens of six species of chironomids every 2 weeks over a 2-year period (2017–2018) from mesocosm experiments using five ponds at ambient temperature and five ponds at 4°C higher than ambient temperature. We investigated (1) wing length responses to temperature within species and between sexes using a regression analysis, (2) interspecific body size responses to test whether the body size of species influences sensitivity to warming, and (3) the correlation between emergence date and wing length. We found a significantly shorter wing length with increasing temperature in both sexes of Procladius crassinervis and Tanytarsus nemorosus, in males of Polypedilum sordens, but no significant relationship in the other three species studied. The average body size of a species affects the magnitude of the temperature-size responses in both sexes, with larger species shrinking disproportionately more with increasing temperature. There was a significant decline in wing length with emergence date across most species studied (excluding Polypedilum nubeculosum and P. sordens), indicating that individuals emerging later in the season tend to be smaller.

Objective To report the prevalence of polyparasitism during pregnancy in the Lambarene region of Gabon and its association with newborn birth weight. Method Pregnant women in their third trimester were recruited in a prospective study between November 2011 and March 2015. Parasite infection status was assessed microscopically in stool, urine and blood samples. Maternal demographic and obstetrical characteristics and newborns anthropometric data were collected. Multivariable logistic regression was used to assess the association between low birth weight and polyparasitism. Results 678 of 927 pregnant women were included for analysis with mean age (SD) of 25 (6.8) years. The analysis showed that 69% (468/678) were infected with at least one parasite (Plasmodium spp., Schistosoma spp., soil-transmitted helminths, filarial infections). This comprised of 38% with monoparasitism and 31% polyparasitism. The proportion of newborn babies with a weight below 2500 g (LBW) in our study was 21% (142/678). Compared to pregnant women without infection, women with monoparasitic infection had adjusted Odds Ratio confidence interval 95% CI (aOR [95%CI]) of 1.6 [0.95-2.73], those with two parasites had aOR 95%CI of 2.63 [1.51-4.62], and those with more than two parasites had aOR of 5.08 [2.5-10.38] for delivering a newborn with low birth weight. Conclusion In Lambarene, an endemic area for multiple parasite infections, there is a high prevalence of polyparasitism in pregnant women. Polyparasitism is associated with low birth weight. Therefore, there is an urgent need for active screening and treatment of parasite infections in pregnant women to assess the potential public health benefit of such interventions.

Recent studies indicate that aluminum (Al) could play an important role in the ocean carbon cycle by increasing phytoplankton carbon fixation and reducing organic carbon decomposition. However, how Al may influence the decomposition of organic carbon has not yet been explicitly examined. Here we report the effects of Al on carbon fixation by marine diatoms and their subsequent decomposition. By using radiocarbon as a tracer, the carbon fixation and decomposition of three model marine diatoms were examined in Aquil* media at different concentrations (0, 40, 200, and 2000 nM) of dissolved Al. Addition of Al enhanced net carbon fixation by the diatoms in the declining growth phase (by 9%–29% for Thalassiosira pseudonana, 15%–20% for T. oceanica, 15%–23% for T. weissflogii). Under axenic conditions the decomposition rates (d⁻¹) of the diatom-produced particulate organic carbon (POC) significantly decreased (by 21%–57% for T. pseudonana, 0%–41% for T. oceanica, 29%–58% for T. weissflogii) in the Al-enriched treatments. In the presence of bacteria, the decomposition rates of T. weissflogii-produced POC were still 37%–38% lower in Al-enriched treatments compared to the control. Significant increases in cell size, cellular carbon content (pmol C/cell) and cellular carbon density (pmol C/μm³) of T. weissflogii were also observed in the Al-enriched treatments compared to the control. The Al-related increase in net carbon fixation and cell size, and the decrease in POC decomposition rate may facilitate carbon export to ocean depths. The study provides new evidence for the iron–aluminum hypothesis, which suggests that Al could increase phytoplankton uptake of atmospheric CO₂ and influence climate change.

The two species of yellow-nosed albatross, Atlantic (Thalassarche chlororhynchos) and Indian (T. carteri), are morphologically similar, but they differ in breeding behaviour and distribution. Both species are listed as endangered by the IUCN due to the limited number of breeding sites, threats from introduced predators and diseases, and impact of commercial fishing. We quantified genetic variation between and within the two species. Using nuclear (microsatellites and two nuclear sequences) and mitochondrial (control region) markers, we analysed 354 samples from four breeding islands (Atlantic: Nightingale, Inaccessible, and Gough; Indian: Amsterdam) and bycatch samples from South Africa and New Zealand. In addition to all markers separating the two species, nuclear markers showed Atlantic yellow-nosed albatrosses from Gough Island are genetically distinct from those breeding at Nightingale and Inaccessible Islands in the Tristan da Cunha archipelago. Nuclear markers confirmed that all bycatch samples were Indian yellow-nosed albatrosses, however, the bycatch birds from South Africa and New Zealand were distinct from each other and from birds breeding on Amsterdam Island, suggesting colony specific dispersal at sea. Our study supports the current recognition of two yellow-nosed albatross species and recognises genetically distinct groups of both Atlantic and Indian yellow-nosed albatross breeding on different islands, which is important for their conservation and management.

Populations of the macaroni penguin Eudyptes chrysolophus and the eastern rockhopper penguin E. filholi breeding at Marion Island (Prince Edward Islands) in the sub-Antarctic Indian Ocean decreased from 1994 to 2018. We examined their diets when rearing chicks during this period. There was substantial overlap in the diets of the two species, with crustaceans, dominated by the euphausiids Thysanoessa vicina and Euphausia vallentini, making up >80% of the diets by number and >60% by mass over the study period. The lanternfishes Krefftichthys anderssoni and Protomyctophum tenisoni were the most-commonly consumed fish in the diet of breeding macaroni penguins and dominated in their diet in three of the 25 years (1994, 1998 and 2014); they were also the most commonly consumed fish by eastern rockhopper penguins. Macaroni penguins consumed more of the amphipod crustacean Themisto gaudichaudii and the lanternfish Electrona carlsbergi as well as a greater diversity of and larger-sized fish than did eastern rockhopper penguins. The horsefish Zanclorhynchus spinifer was found in substantial amounts in the diet of eastern rockhopper penguins in 1996 and 1997. Despite annual variations in relative prey contributions to the diets, there were no significant long-term changes in the diet of either penguin species over the study period or when compared with an earlier assessment in 1982. We conclude that changes in the relative proportions of prey in the diets of these penguin species during breeding are unlikely to account for the recent declines in these penguin populations.

Cryptococcal antigen (CrAg) detection could direct the timely initiation of antifungal therapy. We searched MEDLINE and Embase for studies where CrAg detection in serum/cerebrospinal fluid (CSF) and CSF fungal culture were done on adults living with human immunodeficiency virus (HIV) who had suspected cryptococcal meningitis (CM). With Quality Assessment of Diagnostic Accuracy Studies 2 (QUADAS-2), we evaluated the risk of bias in 11 included studies with 3600 participants, and used a random-effects meta-analysis to obtain summary sensitivity and specificity of serum and CSF CrAg, as well as agreement between CSF CrAg and CSF culture. Summary sensitivity and specificity of serum CrAg were 99.7% (97.4–100) and 94.1% (88.3–98.1), respectively, and summary sensitivity and specificity of CSF CrAg were 98.8% (96.2–99.6) and 99.3% (96.7–99.9), respectively. Agreement between CSF CrAg and CSF culture was 98% (97–99). In adults living with HIV who have CM symptoms, serum CrAg negativity may rule out CM, while positivity should prompt induction antifungal therapy if lumbar puncture is not feasible. In a first episode of CM, CSF CrAg positivity is diagnostic.

Based on the hypothesis that the variation of the metabolomes of latex is a response to selective pressure and should thus be affected differently from other organs, their variation could provide an insight into the defensive chemical selection of plants. Metabolic profiling was used to compare tissues of three Euphorbia species collected in diverse regions. The metabolic variation of latexes was much more limited than that of other organs. In all the species, the levels of polyisoprenes and terpenes were found to be much higher in latexes than in leaves and roots of the corresponding plants. Polyisoprenes were observed to physically delay the contact of pathogens with plant tissues and their growth. A secondary barrier composed of terpenes in latex and in particular, 24-methylenecycloartanol, exhibited antifungal activity. These results added to the well-known role of enzymes also present in latexes, show that these are part of a cooperative defense system comprising biochemical and physical elements. Estimation of realized fecundity (Freal, number of viable oocytes produced) is an essential, yet seldom-achieved element in the understanding of marine animal production and population dynamics. We used the Neutral Red (NR) vital stain to determine oocyte viability in spawns and gonad strippings of four species of commercially-important bivalves: Cerastoderma edule (L), Crassostrea gigas Thunberg, Mytilus edulis L, and Tapes philippinarum (Adams and Reeve). The utility of Trypan Blue as a complementary mortal stain was also assessed, and found to be unnecessary. Normal, live oocytes stained with NR, and were either spherical (mature oocytes) or pedunculated (immature oocytes); atresic/abnormal oocytes also stained with NR, allowing their ready recognition due to their abnormal shapes and cytoplasmic retraction. Dead oocytes did not stain with NR. Across the species studied, a considerable and highly variable proportion of spawned or stripped oocytes was either dead or non-viable; quantitative counts were performed for Cerastoderma edule, the only species for which > 5 spawns occurred. The high level and variability (34–85%) of dead or non-viable oocytes is consistent with a reproductive Red Queen dilemma, in which greater oocyte numbers do not translate to commensurately greater real fecundities, and also with a Sweepstakes Reproductive Success strategy, in which a large range of Freal confronts the considerable variability of intertidal environmental conditions. Neutral Red vital staining is a promising tool for the elucidation and optimization of crucial yet previously intractable aspects of bivalve hatchery production, genetic improvement, restocking, stock management, and conservation.

Background Adnexal skin tumors are diagnostically challenging with few known molecular signatures. Recently, however, YAP1-MAML2 and YAP1-NUTM1 fusions were identified in poroid adnexal skin tumors. Methods Herein, we subjected eight poroid adnexal skin tumors (three poromas and five porocarcinomas) to fusion gene analysis by whole transcriptome sequencing and next-generation DNA sequencing analysis. Results YAP1 fusions were identified in six cases. YAP1-NUTM1 fusions were identified in two poromas and three porocarcinomas. A single case of porocarcinoma harbored a YAP1-MAML2 fusion. Two cases were negative for gene fusion. All cases that harbored YAP1-NUTM1 fusions showed nuclear protein in testis (NUT) expression by immunohistochemistry, with NUT being negative in the YAP1-MAML2-positive case. In this case series, we provide a detailed histopathologic description of six YAP1-fused poroid skin tumors, which we show harbor reproducible histopathologic features, to include broad, bulbous tumor tongues with admixtures of basaloid, poroid cells punctuated by squamatized cuticles and ductules, with uniform tumor nuclei featuring frequent grooves and pseudonuclear inclusions. Conclusions Awareness of the characteristic histopathologic features of YAP1-fused poroid adnexal skin tumor is a step toward a more reproducible classification of adnexal skin tumors as well as a step toward targeted therapy for metastatic and/or unresectable examples of this poroid group of neoplasms.

Functionalization of metal-organic frameworks (MOFs) is critical in exploring their structural and chemical diversity for numerous potential applications. Herein, we report multiple approaches for the tandem postsynthetic modification (PSM) of various MOFs derived from Zr(IV), Al(III), and Zn(II). Our current work is based on our efforts to develop a wide range of MOF platforms with a dynamic functional nature that can be chemically switched via thermally triggered reversible Diels-Alder (DA) and hetero-Diels-Alder (HDA) ligations. Furan-tagged MOFs (furan-UiO-66-Zr) were conjugated with maleimide groups bearing dienophiles to prepare MOFs with a chemically switchable nature. As HDA pairs, phosphoryl dithioester-based moieties and cyclopentadiene (Cp)-grafted MOF (Cp-MIL-53-Al) were utilized to demonstrate the cleavage and rebonding of the linkages as a function of temperature. In addition to these strategies, the Michael addition reaction was also applied for the tandem PSM of IRMOF-3-Zn. Maleimide groups were postsynthetically introduced in the MOF lattice, which were further ligated with cysteine-based biomolecules via the thiol-maleimide Michael addition reaction. On the basis of the versatility of the herein presented chemistry, we expect that these approaches will help in designing a variety of sophisticated functional MOF materials addressing diverse applications.

Amur Falcons Falco amurensis undergo one of the most extreme migrations of any raptor, crossing the Indian Ocean between their Asian breeding grounds and non-breeding areas in southern Africa. Adults are thought to replace all their flight feathers on the wintering grounds, but juveniles only replace some tail feathers before migrating. We compare the extent and symmetry of flight feather moult in a large sample of Amur Falcons killed at communal roosts during two hailstorms in KwaZulu-Natal, South Africa in March 2019, shortly before their northward migration. Most adults had completed replacing their remiges, with only a few still growing 1–3 feathers (mainly secondaries), but most were still growing their tail feathers. Juveniles only replaced tail feathers. Moult typically was distal from the central rectrices, but 25% of adults and 1% of juveniles replaced the outer tail first, and a few individuals exhibited other moult patterns (simultaneous moult across the tail, or among the inner and outer feathers). These different moult strategies were independent of sex. Adults that replaced the outer tail first typically had replaced a greater proportion of the rectrices (mean ± SD; 0.81 ± 0.19) than adults starting from the central tail (0.17 ± 0.08). Proportionally fewer distal moulting adults were killed on 9 March than 21 March, resulting in the average proportion of rectrices replaced by adults decreasing between the two storm events from 0.52 ± 0.26 to 0.43 ± 0.23. By comparison, juvenile tail moult increased from 9 March (0.34 ± 0.18) to 21 March (0.40 ± 0.15). Overall, the probability of replacement for T1 was similar for adults (0.82) and juveniles (0.83), but adults were more likely to have replaced T2–6 (0.40–0.45) than juveniles (0.18 for T2 and 0.04–0.07 for T3–6). Asymmetry in tail moult was greater at T1 for adults (15%) than juveniles (10%), but asymmetry for T2 to T6 was greater in juveniles (3–10%) than adults (1–4%), especially given the greater probability of feather replacement in adults. Despite these differences, the degree of asymmetry was less than expected by random replacement across all rectrices in both age classes. Interestingly, moult tended to be more advanced on the left than right side of the tail. The extent of tail moult was correlated with body condition in adults and juveniles, suggesting that moult pattern might be used as an indicator of fitness in falcons.

TOI-1055 is a Sun-like star known to host a transiting Neptune-sized planet on a 17.5-day orbit (TOI-1055 b). Radial velocity (RV) analyses carried out by two independent groups using nearly the same set of HARPS spectra have provided measurements of planetary masses that differ by $\sim$ 2$\sigma$. Our aim in this work is to solve the inconsistency in the published planetary masses by significantly extending the set of HARPS RV measurements and employing a new analysis tool that is able to account and correct for stellar activity. Our further aim was to improve the precision on measurements of the planetary radius by observing two transits of the planet with the CHEOPS space telescope. We fit a skew normal (SN) function to each cross correlation function extracted from the HARPS spectra to obtain RV measurements and hyperparameters to be used for the detrending. We evaluated the correlation changes of the hyperparameters along the RV time series using the breakpoint technique. We performed a joint photometric and RV analysis using a Markov chain Monte Carlo (MCMC) scheme to simultaneously detrend the light curves and the RV time series. We firmly detected the Keplerian signal of TOI-1055 b, deriving a planetary mass of $M_b=20.4_{-2.5}^{+2.6} M_{\oplus}$ ($\sim$12%). This value is in agreement with one of the two estimates in the literature, but it is significantly more precise. Thanks to the TESS transit light curves combined with exquisite CHEOPS photometry, we also derived a planetary radius of $R_b=3.490_{-0.064}^{+0.070} R_{\oplus}$ ($\sim$1.9%). Our mass and radius measurements imply a mean density of $\rho_b=2.65_{-0.35}^{+0.37}$ g cm$^{-3}$ ($\sim$14%). We further inferred the planetary structure and found that TOI-1055 b is very likely to host a substantial gas envelope with a mass of $0.41^{+0.34}_{-0.20}$ M$_\oplus$ and a thickness of $1.05^{+0.30}_{-0.29}$ R$_\oplus$., Comment: 13 pages, 6 figures, 5 tables.
