Juniperus L. cinsi; yaprakları sivri ve uzun, meyveleri kozalak şeklinde parlak siyah renkte, boyu ortalama 8-9 metre uzunlukta, çok uzun yıllar yaşayabilen bir ağaçtır. Bu çalışmada geleneksel tıpta kullanımı çok eskiden beri bilinen, Juniperus excelsa subsp. excelsa M.Bieb. ve Juniperus foetidissima Willd. bitkilerinin doğal ortamından toplanan yapraklarından elde edilen uçucu yağlar antimikrobiyal etkileri yönüyle araştırılmıştır. Çalışma materyalini oluşturan uçucu yağlar Neo-Clevenger cihazında ekstrakte edilmiştir. Antimikrobiyal aktiviteleri, oyuk agar yöntemi kullanılarak gıda, kozmetik ve farmasötik ürünlerde tıbbi olarak önemli olan 6 patojen ve mikrobiyal kirleticisine karşı (Enterobacter aerogenes (ATCC 13048), Enterococcus faecalis (ATCC 29212), Salmonella enteritidis (ATCC 13075), Staphylococcus aureus subsp. aureus (ATCC 25923), Escherichia coli (ATCC 25922) ve Serratia marcescens (ATCC 13880)) değerlendirilmiştir. Her iki bitkinin uçucu yağlarının tüm patojenlere karşı antibakteriyel aktivite sergilediği görülmüştür (8.13±0.67-13.43±0.31 mm). Ancak Gram negatif bakterilerin J. excelsa subsp. excelsa ve J. foetidissima’ ya karşı daha hassas olduğu belirlenmiştir., Juniperus L. genus; It is a tree with pointed and long leaves, cone-shaped bright black fruits, an average of 8-9 meters in length, and can live for many years. In this study, essential oils obtained from the leaves of Juniperus. excelsa subsp. excelsa M.Bieb. and Juniperus foetidissima Willd.plants, which have been known for a long time in traditional medicine, were investigated in terms of their antimicrobial effects. The essential oils that make up the study material were extracted in the Neo-Clevenger device. Their antimicrobial activities were evaluated against 6 medically important pathogens (Enterobacter aerogenes (ATCC 13048), Enterococcus faecalis (ATCC 29212), Salmonella enteritidis (ATCC 13075), Staphylococcus aureus subsp. aureus (ATCC 25923), Escherichia coli (ATCC 25922) ve Serratia marcescens (ATCC 13880)) and microbial contaminants in food, cosmetics and pharmaceutical products using the hollow agar method. It was observed that the essential oils of both plants exhibited antibacterial activity against all pathogens (8.13±0.67-13.43±0.31 mm). However, it was determined that Gram-negative bacteria were more sensitive to Juniperus excelsa subsp. excelsa and Juniperus foetidissima.

The study aimed to karyologically analyse three Lallemantia species [L. peltata, L. iberica Mey. and L. canescens] grown in Turkey. Also, it was calculated various karyotype asymmetry and S/AI, CVCL and MC values in this study. Current study found that the chromosome numbers are 2n=14 and they possess median (m) and submedian (sm) centromere. In addition, this study suggested that three Lallemantia species are 2A group based on Stebbins classifications. Furthermore, present study showed Pearson correlation using karyotype asymmetry values and a scatter diagram was formed using A1 and A2. On the other hand, the results obtained from the study were compared with the results of karyotype analysis performed by different literatures and it was concluded that there may be differences according to locality.

4. Effects of Fennel and Cumin Extracts on Flax Seed Germination Parameters and Mitotic Activity in the Root Tip Cells

This study was carried out to determine the effects of cumin and fennel seeds extracts, on the germination, seedling development and mitotic activity in root end meristem cells of flax seed. The experiment was established in laboratory according to randomized plot factorial experiment design with three replications in incubator having 23℃ temperature. At the end of 14 days of germination period some germination parameters were measured. According to the control, it was observed that fennel extract applications positively affected the germination and seedling development of flax seed except 100 g/l dose. While germination is seen at 100 g/l, seedling growth is completely prevented. It was observed that all cumin extract applications had a negative effect on the germination and seedling development of flax seed compared to the control. This negativity has shown more effect in increasing doses. While the germination rate is gradually decreasing in increasing doses, it is observed that the growth of seedlings is completely prevented except 12.5 and 25 g/lcumin extract. Increased doses at doses other than 100 g/lincreased the mitotic index in fennel extract application and decreased the dose increase in cumin extract application. The percentage of chromosomal abnormality decreased in parallel with the increase in doses other than 100 g/l in fennel, but percentage of chromosomal abnormality increase with increasing doses in cumin extract. During mitotic examinations; mitotic abnormalities such as failure to collect the metaphase plate, bridge in anaphase and telophase were observed and their pictures were recorded., Bu çalışma kimyon ve rezene tohum ekstraktlarının keten tohumunun çimlenme, fide gelişimi ve kök ucu meristem hücreleri üzerindeki mitotik aktivitelerini belirlemek amacıyla yapılmıştır. Deney, laboratuvar koşullarında 23℃’deki inkübatörde tesadüf parsellerinde faktöriyel deneme desenine göre üç tekerrürlü olarak yapılmıştır. İki farklı öğütülmüş tohum türünden 4 farklı çözelti hazırlanmıştır. 14 günlük çimlenme süresinin sonunda, bazı çimlenme parametreleri ölçülmüştür. Kontrol uygulamasına göre, rezene ekstraktı uygulamalarının 100 g/l dozu hariç keten tohumunun çimlenme ve fide gelişimini olumlu yönde etkilediği görülmüştür. 100 g/l dozunda çimlenme görülürken, fide büyümesi tamamen engellenmiştir. Tüm kimyon ekstraktı uygulamalarının, kontrol uygulamasına kıyasla keten tohumunun çimlenme ve fide gelişimi üzerinde olumsuz bir etkisi olduğu belirlenmiştir. Bu olumsuzluk artan dozlarda daha fazla etki göstermiştir. Çimlenme oranı artan dozlarda kademeli olarak düşerken, 12.5 ve 25 g/l kimyon ekstresi hariç fidelerin büyümesinin tamamen önlendiği görülmektedir. 100 g/l dışındaki dozlar, rezene ekstraktı uygulamasında mitotik indeksi artırmış ve kimyon ekstraktı uygulamasında doz artışı mitotik indeksi azaltmıştır. Kromozomal anormallik yüzdesi, rezenelerde 100 g/l dışındaki dozlardaki artışa paralel olarak azalmış, ancak kimyon ekstraktında artan dozlarla kromozomal anormallik yüzdesi artmıştır. Mitotik incelemelerde; metafaz plağında toplanamama, anafazda ve telofazda köprü gibi mitotik anormallikler gözlenmiş ve resimleri kaydedilmiştir. Tüm bu bilgiler ışığında, rezene ekstraktının keten tohumunda çimlenmeyi teşvik ettiği görülmüştür. Bu etkinin, yapılacak daha ayrıntılı çalışmalar sayesinde farklı tohumlardaki dormansiyi kırmak için kullanılabileceği düşünülmektedir. Bunun yanında kimyon ekstraktının bazı tohumlarda çimlenmeyi önlemek üzere kullanılabileceği değerlendirilmektedir.

WOS: 000390834400003, In this study, the chromosome number and detailed morphometric properties of six endemic taxa belonging to five dicot families which grow naturally in Turkey were examined. The somatic chromosome number and karyotype formula of the taxa of Asteraceae family were determined to be 2n=18 and 8m+1sm in Tanacetum argenteum ssp. argenteum and 2n=18 and 7m+2sm+1B in Tanacetum densum ssp. sivasicum, respectively. The somatic chromosome number and karyotype formula were determined as 2n=32 and 10m+6sm in Fumana trisperma belonging to Cistaceae, 2n=14 and 5m+2sm in Onobrychis alblflora belonging to Fabaceae, 2n=14 and 2m+5sm in Salvia blepharochlaena belonging to Lamiaceae, and 2n=14 and 2m+5sm in Glaucium acutidentatum belonging to Papaveraceae family. The somatic chromosome numbers of six taxa collected from different localities in Turkey were counted for the first time, except for T. argenteum var. argenteum and Salvia blepharocleana.

In this study, morphological, morphometrical, karyological and detailed pollen morphology of Ebenus haussknechtii endemic to Turkey was studied for the first time. In the morphologic study, some morphological and morphometrical features of the plant were observed and compared with the published findings in the Flora of Turkey. Karyogram and idiograms of the plant were also prepared. The chromosome number of E. haussknechtii was determined as 2 n = 14 and haploid karyotype formula 5 m + 1 sm + 1 M. Metaphase chromosome length ranged from 2.16 to 1.93 μm and the total haploid chromosome length was 17.90 μm. The results of the light microscope investigation of the pollen revealed that the pollen grains were radially symmetrical, isopolar, tricolpate and prolate. Measurement of the reticulum heterobrachate indicated their ornamentation to be reticulate.

The chromosome number and chromosome morphology of endemic species Centaurea fenzlii and Origanum acutidens naturally growing in Turkey were investigated using karyological techniques. The specimens were collected from natural habitats in the years 2013. The basic chromosome number of Centaurea fenzlii is: x= 9 and diploid chromosome numbers were found to be 2n=18, species of Origanum acutidens basic chromosome number is: x= 15 and diploid chromosome numbers were found to be 2n=30. Chromosomes had median region (m) and submedian (sm) centromers. Karyotype analysis of Origanum acutidens endemic species has been conducted by us for the first time. © Afyon Kocatepe Üniversitesi, Türkiye’ de doğal olarak yetişen Centaurea fenzlii Reichardt. ve Origanum acutidens (Hand.-Mazz.) Ietswaart. endemik türlerinin kromozom sayısı ve kromozom morfolojisi karyolojik teknikler kullanılarak araştırıldı. Bitki örnekleri 2013 yılında doğal habitatlarından toplandı.
