Shifts in dominance and species reordering can occur in response to global change. However, it is not clear how altered precipitation and disturbance regimes interact to affect species composition and dominance.We explored community‐level diversity and compositional similarity responses, both across and within years, to a manipulated precipitation gradient and annual clipping in a mixed‐grass prairie in Oklahoma, USA. We imposed seven precipitation treatments (five water exclusion levels [−20%, −40%, −60%, −80%, and −100%], water addition [+50%], and control [0% change in precipitation]) year‐round from 2016 to 2018 using fixed interception shelters. These treatments were crossed with annual clipping to mimic hay harvest.We found that community‐level responses were influenced by precipitation across time. For instance, plant evenness was enhanced by extreme drought treatments, while plant richness was marginally promoted under increased precipitation.Clipping promoted species gain resulting in greater richness within each experimental year. Across years, clipping effects further reduced the precipitation effects on community‐level responses (richness and evenness) at both extreme drought and added precipitation treatments. Synthesis: Our results highlight the importance of studying interactive drivers of change both within versus across time. For instance, clipping attenuated community‐level responses to a gradient in precipitation, suggesting that management could buffer community‐level responses to drought. However, precipitation effects were mild and likely to accentuate over time to produce further community change., Shifts in dominance and species reordering can occur in response to global change. We manipulated an experimental precipitation gradient combined with clipping in a temperate mixed‐grass prairie to examine how both treatments can shape plant communities. We found that initial shifts in abundance were detected by examining species‐ to community‐level changes over time; drought increased evenness and this was related to the decline of the dominant species and increase in subdominants, while mesic conditions mildly promoted plant richness.

The impacts of coastal reclamation on carbon (C) and nitrogen (N) sinks of coastal wetlands remain unclearly understood. This study was conducted to investigate the alterations of soil organic C and N (SOC and SON) pools following conversion of Phragmites australis salt marsh into fishpond, wheat and rapeseed fields and town construction land through reclamation along Jiangsu coast in eastern China. Coastal reclamation significantly increased stocks of soil total, labile and recalcitrant organic C and N (SLOC, SLON, SROC, and SRON), and concentrations of water-soluble organic C (WSOC), microbial biomass C and N (SMBC and SMBN), cumulative CO2-C mineralization (MINC) following conversion of P. australis salt marsh into fishpond, wheat and rapeseed fields. However, coastal reclamation reduced SOC, SLOC, SROC, SRON, WSOC, SMBC, SMBN, and MINC following conversion of P. australis salt marsh into town construction land. Our results suggest that coastal reclamation affects C and N sinks of coastal wetlands by changing SOC and SON pools size, stability and dynamics changes following conversion of P. australis salt marsh into other land use types. This finding were primarily attributed to alterations in quantity and quality of exogenous materials returning the soil, and soil physiochemical properties as affected by coastal reclamation.

Ubiquitous thermal acclimation of leaf respiration could mitigate the respiration increase. However, whether species of different plant functional groups showing distinct or similar acclimation justifies the simple prediction of respiratory carbon (C) loss to a warming climate. In this study, leaf dark respiration (Rd) of illinois bundleflower (IB, legume), stiff goldenrod (GR, C₃ forbs), indian grass, little bluestem and king ranch bluestem (IG, LB and KB, C₄ grass) were measured with detached leaves sampled in a 17-year warming experiment. The results showed that Rd at 20°C and 22°C (R₂₀ and R₂₂) were significantly lower in the warming treatment for all the five species. Lower R₂₂ in warmed than R₂₀ in control in GR, KB, LB and IG imply acclimation homeostasis, but not in IB. The significant decline in temperature sensitivity of respiration (Q₁₀) of GR resulted in the marginal reduction of Q₁₀ across species. No significant changes in Q₁₀ of C₄ grasses suggest different acclimation types for C₃ forbs and C₄ grass. The magnitude of acclimation positively correlated with leaf C/N. Our results suggest that non-legume species had a relative high acclimation, although the acclimation type was different between C₃ forbs and C₄ grasses, and the legume species displayed no acclimation in Rd. Thus, the plant functional types should be taken into account in the grassland ecosystem C models.

Many animals exhibit an astonishing ability to form groups of large numbers of individuals. The dynamic properties of such groups have been the subject of intensive investigation. The actual grouping processes and underlying neural mechanisms, however, remain elusive. Here, we established a social clustering paradigm in Drosophila to investigate the principles governing social group formation. Fruit flies spontaneously assembled into a stable cluster mimicking a distributed network. Social clustering was exhibited as a highly dynamic process including all individuals, which participated in stochastic pair-wise encounters mediated by appendage touches. Depriving sensory inputs resulted in abnormal encounter responses and a high failure rate of cluster formation. Furthermore, the social distance of the emergent network was regulated by ppk-specific neurons, which were activated by contact-dependent social grouping. Taken together, these findings revealed the development of an orderly social structure from initially unorganised individuals via collective actions.

Succession, a central theme throughout the history of ecology, has been well studied predominantly in plant communities, but the general trends in soil microbial communities during succession remain unclear. Here, we compiled a comprehensive data set of 85 age sequences worldwide with the aims to (1) examine the trends in soil microbial composition, bioenergetics, and activity during secondary succession, and (2) explore their coordinating changes with soil properties. The results showed that the fungi to bacteria ratio ( fungi:bacteria ) increased, while the microbial respiration per unit biomass carbon ( R/C mic ) decreased as the succession proceeds. Secondary succession had the rising trends in microbial biomass carbon to soil carbon ratio ( C mic / C soil ) and microbial biomass nitrogen to soil nitrogen ratio ( N mic / N soil ). These successional trends in microbes were coincident with the macro-ecological succession theory in plants and animals. Specifically, early successional stages tended to be dominated by r -strategists (bacteria) that had higher R/C mic and lower C mic / C soil and N mic / N soil , whereas late successional stages tended to be dominated by K -strategists (fungi) that behaved oppositely. The soil C to N ratio ( C:N soil ) increased significantly with the successional stage, with a fast increasing C:N soil ratio being accompanied by a fast increase of fungi:bacteria , a slow decrease of R/C mic , and a slow increase of C mic /C soil . This result suggests that the stoichiometry theory may provide a feasible approach to explain the divergent successional trends in microbial communities. In conclusion, our global synthesis highlights the application of the existing macro-ecological theory to soil microbial ecology studies.

Anthropogenic nitrogen (N) deposition is expected to increase substantially and continuously in the future. Soil N availability regulates microbial communities and the decomposition and formation of soil organic matter, which have great impacts on global carbon (C) cycling. We conducted a meta-analysis based on 454 N-addition experiments in order to synthesize the patterns and mechanisms of responses by soil microbial communities to N addition in various biomes (i.e., boreal forest, temperate forest, tropical/subtropical forest, grassland, and desert). Results showed that the effects of N addition on the total microbial biomass varied depending on biome types, methodologies (fumigation–extraction technique vs. total phospholipid fatty acid), and N-addition rates. Nitrogen addition consistently decreased the microbial C:N and fungi to bacteria ratio (F:B), but increased Gram positive bacteria to Gram negative bacteria ratio (GP:GN) among biome types and N-addition rates. Nitrogen addition increased soil N availability and thereby resulted in soil acidification. Regression technique and principal component analyses showed that the shifts in the F:B and GP:GN mainly resulted from enhanced N availability due to N addition rather than soil acidification. When the N addition rate is lower than 100 kg N ha−1 year−1, about ten times higher than of global normal rate, the positive response of microbial growth was found. Overall, these findings revised the previous notion that N addition inhibited the microbial growth. Microbial species shifts might accentuate or mitigate the effects of alterations in microbial biomass at the ecosystem level, highlighting the critical role of microbial community composition in soil ecosystem functions under N deposition scenarios.

Ericoid mycorrhizal associations are a symbiotic relationship between soil fungi and ericaceous plants. Diversity of fungi associated with hair roots of ericaceous plants may vary as a result of frequent disturbances by human activities. The fungal diversity and communities associated with hair roots of Rhododendron ovatum were investigated along a human disturbance gradient in subtropical forests of China. Nine hundred fungal operational taxonomic units were determined by high-throughput sequencing, including different phyla such as Ascomycota, Basidiomycota, Zygomycota, Chytridiomycota, and Glomeromycota. The dominant phylum in Cunninghamia lanceolata plantations and old-growth forest was Ascomycota, while Basidiomycota was the dominant phylum in secondary forests. The indicator species analyses showed that more pathogenic indicator fungi appeared in the disturbed forests, whereas more putative ericoid mycorrhizal fungi existed in the old-growth forests. Principal component analysis also showed that the fungal communities in the hair roots of R. ovatum were distinct between natural forests and plantations, suggesting that the fungal communities associated with hair roots of R. ovatum after logging were resilient and could recover to predisturbance status. The results of envfit analysis showed that performance of host plants rather than accompanying plant community and soil parameters of plots was the key determinant of the root-associated fungal community of R. ovatum.

Food consumption is under the tight control of the brain, which integrates the physiological status, palatability, and nutritional contents of the food, and issues commands to start or stop feeding. Deciphering the processes underlying the decision-making of timely and moderate feeding carries major implications in our understanding of physiological and psychological disorders related to feeding control. Simple, quantitative, and robust methods are required to measure the food ingestion of animals after experimental manipulation, such as forcibly increasing the activities of certain target neurons. Here, we introduced dye-labeling-based feeding assays to facilitate the neurogenetic study of feeding control in adult fruit flies. We review available feeding assays, and then describe our methods step-by-step from setup to analysis, which combine thermogenetic and optogenetic manipulation of neurons controlling feeding motivation with dye-labeled food intake assay. We also discuss the advantages and limitations of our methods, compared with other feeding assays, to help readers choose an appropriate assay.

The effects of thermosonication (TS) and 100 ppm nisin-assisted TS (TS + nisin) on the inactivation of naturally occurring microorganisms, retention of nutritional quality and extension of shelf life of fresh apple juice were evaluated, with nisin and mild heat (nisin + MH) treatments as control. Fresh apple juice was addressed by nisin + MH, TS and TS + nisin at 37, 42, 47, and 52 °C for 5–40 min. After processing, microbial growth was evaluated during storage at 8 °C at every 5 days. Temperature played a vital role in the inactivation of aerobic bacteria and yeasts and molds by TS and TS + nisin, higher temperature up to 52 °C could cause a considerable inactivation of microbial cells in apple juice. As apple juice was subjected to TS and TS + nisin at 52 °C for 30 min, retention of original quality including 89% ascorbic acid, non-visible color change, no significant alteration in BD, pH, TA and TSS values of fresh apple juice, and extension shelf life to 15 d at 8 °C were obtained. Nisin exhibit additional inactivation effect of aerobic bacteria in apple juice while not obviously effect on yeast and molds. These results indicated a potential application of TS and TS + nisin (100 ppm) to produce fresh-like quality apple juice and/or to extend its shelf life.

The monosaccharide compositions, molecular weights, structure were investigated for the black fungus polysaccharides. Analysis of sugar components by gas chromatograph-mass spectrometer (GC-MS) showed that component ratios of neutral sugars in the polysaccharides were Glc:Man=82.97:12.93. Fourier transform infrared (FT-IR) analysis showed that the band at about 1649cm(-1) can indicate the presence of CO groups or CC groups vibration in structures. Pharmacological experiment showed that black fungus polysaccharides administration could significantly reduce malondialchehyche (MDA), lipofuscin (LF) contents and enhance superoxide dismutase (SOD) activities in skin. This indicated that black fungus polysaccharides may slow down skin aging.

Ecological functions of bioturbation in ecosystems have received increasing attention over the recent decades, and crab burrowing has been considered as one of the major bioturbations affecting the physical and chemical processes in salt marshes. This study assessed the integrated effects of crab excavating and burrow mimic trapping on sediment turnover and vertical C and N distributions in a Chinese salt marsh in the Yangtze River estuary. Crab burrowing increased soil water content and the turnover of carbon and nitrogen and decreased bulk soil density. Vertical movement of materials, nutrient cycling and reuse driven by crab burrowing might be obstructed by vegetation (Phragmites australis and Spartina alterniflora communities). The amount of soil excavated by crab burrowing was higher than that deposited into burrow mimics. In Phragmites marshes, Spartina marshes and unvegetated mudflats, net transport of soil to the marsh surface was 171.73, 109.54, and 374.95 g m−2 d−1, respectively; and the corresponding estimated soil turnover time was 2.89, 4.07 and 1.83 years, respectively. Crab burrowing in salt marshes can mix surface and deeper soil over a period of years, accelerating litter decomposition and promoting the efficient reuse of nutrients by plants. Therefore, bioturbation affects soil physical processes and functioning of ecosystems, and needs to be addressed in ecosystem management.

Biomass allocation is an important plant trait that responds plastically to environmental heterogeneities. However, the effects on this trait of pollutants owing to human activities remain largely unknown. In this study, we investigated the response of biomass allocation of Phragmites australis to petroleum pollution by a 13 CO 2 pulse-labelling technique. Our data show that plant biomass significantly decreased under petroleum pollution, but the root–shoot ratio for both plant biomass and 13 C increased with increasing petroleum concentration, suggesting that plants could increase biomass allocation to roots in petroleum-polluted soil. Furthermore, assimilated 13 C was found to be significantly higher in soil, microbial biomass and soil respiration after soils were polluted by petroleum. These results suggested that the carbon released from roots is rapidly turned over by soil microbes under petroleum pollution. This study found that plants can modulate biomass allocation in response to petroleum pollution.

Age-dependent ectopic fat accumulation (EFA) in animals contributes to the progression of tissue aging and diseases such as obesity, diabetes, and cancer. However, the primary causes of age-dependent EFA remain largely elusive. Here, we characterize the occurrence of age-dependent EFA in Drosophila and identify HDAC6, a cytosolic histone deacetylase, as a suppressor of EFA. Loss of HDAC6 leads to significant age-dependent EFA, lipid composition imbalance, and reduced animal longevity on a high-fat diet. The EFA and longevity phenotypes are ameliorated by a reduction of the lipid-droplet-resident protein PLIN2. We show that HDAC6 is associated physically with the chaperone protein dHsc4/Hsc70 to maintain the proteostasis of PLIN2. These findings indicate that proteostasis collapse serves as an intrinsic cue to cause age-dependent EFA. Our study suggests that manipulation of proteostasis could be an alternative approach to the treatment of age-related metabolic diseases such as obesity and diabetes.

The Yangtze River estuary is an important ecoregion. However, Spartina alterniflora, native to North America, was introduced to the estuary in the 1990s through both natural dispersal and humans and now it is a dominant species in the estuarine ecosystems, with its invasions leading to multiple consequences to the estuary. S. alterniflora had great competitive effects on native species, including Scirpus mariqueter and Phragmites australis, and could potentially exclude the natives locally. The presence of S. alterniflora had little influence on the total density of soil nematodes and macrobenthonic invertebrates, but significantly altered the structure of trophic functional groups of nematode and macrobenthonic invertebrate communities. The conversion of mudflats to Spartina meadows had significant effects on birds of Charadriidae and Scolopacidae, which might be attributable to the reduction of food resources and the physical alterations of habitats for shorebirds. S. alterniflora invasions increased the primary productivity of the invaded ecosystems, and altered carbon and nitrogen cycling processes. Our studies focused mainly on the effects of S. alterniflora invasions on the structure of native ecosystems; thus further studies are clearly needed to investigate how ecosystem functioning is affected by the modification of the structure of estuarine ecosystems by S. alterniflora invasions.

Invasive plants strongly affect physical and biotic environments of native ecosystems. Insects and other arthropods as one of the major components of many ecosystems are very sensitive to subtle changes in abiotic and biotic environments. We examined the effects of exotic Spartina alterniflora invasion on community structure and diets of arthropods in a saltmarsh previously dominated by native Phragmites australis in Yangtze River estuary through net sweeping and plant harvesting methods and stable isotope analysis. Our results showed that diversity indices were not significantly different between exotic and native plant communities, but the total abundance of insects estimated through plant harvesting method was found to be lower in Spartina monoculture than that in Phragmites monoculture. Community structure of insects in Spartina monoculture was dissimilar to that in Phragmites monoculture and Phragmites–Spartina mixture. Moreover, stable carbon isotope patterns of arthropods were significantly different between Phragmites and Spartina monocultures. Although some native arthropods (perhaps generalists) shifted their diets, many native taxa did prefer Phragmites to Spartina even in Spartina monoculture. Spartina invasions resulted in reduced abundances of some arthropds, and increased dominance of others feeding preferably on Spartina. This study provides evidence that invasive plants can change the community structure and diets of native arthropods, which will eventually alter the arthropod food web, and affect the integrity and functioning of native ecosystems within a nature reserve that has been set aside for conserving the native biodiversity and maintaining the ecosystem integrity. In this sense, Spartina invasions in the Yangtze River estuary need to be managed appropriately.

Whether plant invasion increases ecosystem carbon (C) stocks is controversial largely due to the lack of knowledge about differences in ecophysiological properties between invasive and native species. We conducted a field experiment in which we measured ecophysiological properties to explore the response of the ecosystem C stocks to the invasion of Spartina alterniflora (Spartina) in wetlands dominated by native Scirpus mariqueter (Scirpus) and Phragmites australis (Phragmites) in the Yangtze Estuary, China. We measured growing season length, leaf area index (LAI), net photosynthetic rate (Pn), root biomass, net primary production (NPP), litter quality and litter decomposition, plant and soil C and nitrogen (N) stocks in ecosystems dominated by the three species. Our results showed that Spartina had a longer growing season, higher LAI, higher Pn, and greater root biomass than Scirpus and Phragmites. Net primary production (NPP) was 2.16 kg C m−2 y−1 in Spartina ecosystems, which was, on average, 1.44 and 0.47 kg C m−2 y−1 greater than that in Scirpus and Phragmites ecosystems, respectively. The litter decomposition rate, particularly the belowground decomposition rate, was lower for Spartina than Scirpus and Phragmites due to the lower litter quality of Spartina. The ecosystem C stock (20.94 kg m−2) for Spartina was greater than that for Scirpus (17.07 kg m−2), Phragmites (19.51 kg m−2) and the mudflats (15.12 kg m−2). Additionally, Spartina ecosystems had a significantly greater N stock (698.8 g m−2) than Scirpus (597.1 g m−2), Phragmites ecosystems (578.2 g m−2) and the mudflats (375.1 g m−2). Our results suggest that Spartina invasion altered ecophysiological processes, resulted in changes in NPP and litter decomposition, and ultimately led to enhanced ecosystem C and N stocks in the invaded ecosystems in comparison to the ecosystems with native species.

Ericoid mycorrhiza (ERM) are expected to facilitate establishment of ericaceous plants in harsh habitats. However, diversity and driving factors of the root-associated fungi of ericaceous plants are poorly understood. In this study, hair-root samples of Vaccinium carlesii were taken from four forest types: old growth forests (OGF), secondary forests with once or twice cutting (SEC I and SEC II) and Cunninghamia lanceolata plantation (PLF). Fungal communities were determined using high-throughput sequencing and impacts of human disturbances and the intra- and inter-annual variability of root-associated fungal community were evaluated. Diverse fungal taxa were observed and our results showed that (1) Intra- and inter-annual changes in root-associated fungal community were found and the Basidiomycota to Ascomycota ratio was related to mean temperature of the sampling month; (2) Human disturbances significantly affected structure of root-associated fungal community of V. carlesii and two secondary forest types were similar in root-associated fungal community and were closer to that of the old growth forest; (3) Plant community composition, edaphic parameters and geographic factors significantly affected root-associated fungal communities of V. carlesii. These results may be helpful in better understanding the maintenance mechanisms of fungal diversity associated with hair roots of ERM plants under human disturbances.

Plantago virginica L. has invaded many lawn ecosystems in the Eastern part of China. The invasion has incurred an economic cost to remove them. In order to prevent the invasion, it is critical to understand the invasive mechanisms of this species. However, few studies have been conducted on the allelopathic mechanisms of its invasion. In this study, we examined allelopathic effects of P. virginica on germination of seeds and growth of seedlings of four widely used lawn species. We found extensive allelopathic potential of P. virginica on other lawn species, which varied with species and developmental stage. While most effects of the extracts of P. virginica were inhibitory, some variables in some species were promoted by the addition of the extracts. The extracts of P. virginica significantly inhibited seed germination of Agrostis matsumurae. While the overall differences in seed germination rate of Poa annua were significant among treatments, difference between control and any of the treatments was not significant. The height of seedlings of A. matsumurae and Cynodon dactylon was significantly lower under the treatments of adding extracts of P. virginica. In contrast, growth of seedlings of Festuca elata and P. annua did not show significant differences among treatments. The root length of A. matsumurae, C. dactylon and P. annua was suppressed by the extracts of P. virginica whereas root length of F. elata was not affected. Aboveground biomass of A. matsumurae and F. elata was significantly higher than control, except for F. elata at the concentration of 50mg/mL, whereas aboveground biomass of C. dactylon and P. annua was reduced at higher concentrations of the extracts. Except for A. matsumurae, root biomass of the other three lawn species declined under the treatments with the extracts of P. virginica. Our results revealed that P. virginica had allelopathic potential on four lawn species and supported the theory of “novel weapons hypothesis”. Invasion by P. virginica in lawn can be moderated by selecting those species that are not affected or promotionally affected by it.

The potential capacity of soil to sequester carbon in response to global warming is strongly regulated by the ratio of rhizosphere respiration to respiration by soil microbial decomposers, because of their different temperature sensitivities. To quantify relative contributions of rhizosphere respiration to total soil respiration as influenced by forest stand development, we conducted a trenching study in two larch (Larix gmelini (Rupr.) Rupr.) plantations, aged 17 and 31 years, in northeastern China. Four plots in each plantation were randomly selected and trenched in early May 2001. Soil surface CO2 effluxes both inside and outside the plots were measured from May 2001 to August 2002. Soil respiration (i.e., the CO2 effluxes outside the trenched plots) varied similarly in the two plantations from 0.8 micromol m(-2) s(-1) in winter to 6.0 micromol m(-2) s(-1) in summer. Rhizosphere respiration (i.e., CO2 efflux outside the trenched plots minus that inside the plots) varied from 0.2 to 2.0 micromol m(-2) s(-1) in the old forest and from 0.3 to 4.0 micromol m(-2) s(-1) in the young forest over the seasons. Rhizosphere respiration, on average, accounted for 25% of soil respiration in the old forest and 65% in the young forest. Rhizosphere and soil respiration were significantly correlated with soil temperature but not with soil water content. We conclude that the role forests play in regulating climate change may depend on their age.

Identifying binding sites recognized by transcription factors (TFs) is one of major challenges to decipher complex genetic regulatory networks encoded in a genome. A set of binding sites recognized by the same TF, called a motif, can be accurately represented by a position frequency matrix (PFM) or a position-specific scoring matrix (PSSM). Very often, we need to compare motifs when searching for similar motifs in a motif database for a query motif, or clustering motifs possibly recognized by the same TF. In this paper, we have designed a novel metric, called SPIC (Similarity between Positions with Information Contents), for quantifying the similarity between two motifs using their PFMs, PSSMs, and column information contents, and demonstrated that this metric outperforms the other state-of-the-art methods for clustering motifs of the same TF and differentiating motifs of different TFs.
